Isostichopus badionotus and Isostichopus sp. are two holothuroids exploited in the Caribbean region of Colombia. Until recently, they were considered a single species. During one year, 222 individuals of Isostichopus sp. and 114 of I. badionotus were collected in two bays of the Santa Marta region to study their reproductive biology and collect information on their size, weight and habitat. Both sea cucumber morphotypes showed an annual reproductive cycle, with a reproductive season from September to November, closely related to the increase in water temperature and rainfall. In both sea cucumbers the population structure exhibited a unimodal distribution composed of mature individuals and a sex ratio of 1:1. Isostichopus sp. had an average size and weight (193 ± 52 mm and 178 ± 69 g) and size and weight at first maturity (175 mm and 155 g) that was much lower than I. badionotus (respectively, 324 ± 70 mm and 628 ± 179 g; 220 mm and 348 g). While 98% of Isostichopus sp. individuals were collected in the upper 2.5 m, on rocky bottoms between cracks, 73% of I. badionotus individuals were found between 3 and 7.8 m depth, exposed on sandy bottoms. Since the size of newly hatched larval fish is directly related to egg size, small differences in initial egg size can be critical to survival and further development of offspring. Underlying processes causing size variation in fish offspring are still not entirely understood. In this study we investigated whether the spatial position of an individual egg within a clutch affects size variation in two benthic spawning coral reef fishes, the clownfishes Amphiprion ocellaris and A. frenatus. To evaluate the effects of within-clutch position on embryonic development, egg growth metrics and protein content were analysed on day 2, 5 and 8 after deposition (adp). Additionally the activities of the key metabolic enzymes citrate synthase (CS) and lactate dehydrogenase (LDH) were investigated to evaluate the physiological status of the embryos. Central eggs of A. frenatus were significantly longer and heavier than peripheral eggs only on day 2 and 5 adp (2.07 mg, 2.59 mm vs. 1.84 mg, 2.49 mm). No significant differences were observed in A. ocellaris between eggs originating from a central or peripheral (5 mm from edge) position (1.33 mg, 2.26 mm vs. 1.15 mg, 2,18 mm). Diameter of the eyes did not differ between the two fish species nor between different positions, for any age group. The protein content of eggs (7.5% of wet weight) was independent of age, position and species. Enzymatic activity increased from 2 adp until peak activity was observed for both enzymes on day 8 adp, independent from position. The range of CS- and LDH-activity was 0.3–13.0 and 0.2–71.7 U g-1 wet weight, respectively. Significant differences in enzymatic activity were observed between age groups in both species, which in connection with significantly larger eggs of A. frenatus at day 2 and 5 adp could hint at a better O2 supply of central eggs. The Aerobic Scope (AS), which reflects the functional capacity for biological fitness, is a highly relevant proxy to determine thermal tolerance in various taxa. Despite the importance of this method, its implementation is often hindered, due to lacking techniques to accurately measure standard- (SMR) and maximal- (MMR) metabolic rates, especially in sluggish marine invertebrates with low oxygen consumption rates, such as sea cucumbers. In this study the AS concept was modified to define a Temperature-induced Aerobic Scope (TAS), based on metabolic rate changes due to temperature adjustments rather than traditionally used physical activity patterns. Consequentially, temperature dependent peak and bottom O2 consumption rates, defined as Temperature-induced Maximal- (TMMR) and Standard Metabolic Rates (TSMR), respectively, served as MMR and SMR alternatives for the sea cucumber Holothuria scabra. TMMR and TSMR were induced through acute temperature change (2°C per hour; 17–41°C) until critical warm (WTcrit) and cold (CTcrit) temperatures were reached, respectively. In addition, Hsp70 gene expression linked to respiration rates served as synergistic markers to confirm critical threshold temperatures. O2 consumption of H. scabra peaked distinctly at WTcrit of 38°C (TMMR = 33.2 ± 4.7 μgO2 g-1 h-1). A clear metabolic bottom line was reached at CTcrit of 22°C (TSMR = 2.2 ± 1.4 μgO2 g-1 h-1). Within the thermal window of 22–38°C H. scabra sustained positive aerobic capacity, with assumed optimal performance range between 29–31.5°C (13.85–18.7 μgO2 g-1 h-1). Between 39–41°C H. scabra decreased respiration progressively, while gene expression levels of Hsp70 increased significantly at 41°C, indicating prioritization of heat shock response (HSR) and homeostatic disruption. At the cold end (17–22°C) homeostatic disruption was visible through incrementally increasing energetic expenses to fuel basal maintenance costs, but no Hsp70 overexpression occurred. TMMR, TSMR and TAS proved to be reliable metrics, similar to the traditional energetic key parameters MMR, SMR and AS, to determine a specific aerobic performance window for the sluggish bottom dwelling species H. scabra. In addition, the linkage between respiration physiology and molecular defense mechanisms showed valuable analytical synergies in terms of mechanistic prioritization as response to thermal stress. In nature, a multitude of both abiotic and biotic stressors influence organisms with regard to their overall fitness. Stress responses that finally impair normal biological functions may ultimately result in consequences for whole populations. This study focused on the metabolic response of the intertidal rock pool fish Gobius paganellus towards simulated predation risk. Individuals were exposed to a mixture of skin extracts from conspecifics and chemical alarm cues from a top predator, Octopus vulgaris. Oxygen consumption rates of single fish were measured to establish standard (SMR) and routine metabolic rates (RMR) of G. paganellus, and to address the direct response towards simulated predation risk, compared to handling and light stress. The SMR of G. paganellus (0.0301 ± 0.0081 mg O2 h-1 g-1 WW) was significantly lower than the RMR (0.0409 ± 0.0078 mg O2 h-1 g-1 WW). In contrast to increased respiration due to handling and light stress, the exposure to chemical predation cues induced a significant reduction in oxygen consumption rates (0.0297 ± 0.0077 mg O2 h-1 g-1 WW). This metabolic suppression was interpreted as a result of the stereotypic freezing behaviour as antipredator response of gobiid fish. Results underline the importance of biotic interactions in environmental stress assessments and predation as a biotic factor that will provide more realistic scenarios when addressing stress impacts in tidal rock pool organisms. Coral reefs are facing major global and local threats due to climate change-induced increases in dissolved inorganic carbon (DIC) and because of land-derived increases in organic and inorganic nutrients. Recent research revealed that high availability of labile dissolved organic carbon (DOC) negatively affects scleractinian corals. Studies on the interplay of these factors, however, are lacking, but urgently needed to understand coral reef functioning under present and near future conditions. This experimental study investigated the individual and combined effects of ambient and high DIC (pCO2 403 μatm/ pHTotal 8.2 and 996 μatm/pHTotal 7.8) and DOC (added as Glucose 0 and 294 μmol L-1, background DOC concentration of 83 μmol L-1) availability on the physiology (net and gross photosynthesis, respiration, dark and light calcification, and growth) of the scleractinian coral Acropora millepora (Ehrenberg, 1834) from the Great Barrier Reef over a 16 day interval. High DIC availability did not affect photosynthesis, respiration and light calcification, but significantly reduced dark calcification and growth by 50 and 23%, respectively. High DOC availability reduced net and gross photosynthesis by 51% and 39%, respectively, but did not affect respiration. DOC addition did not influence calcification, but significantly increased growth by 42%. Combination of high DIC and high DOC availability did not affect photosynthesis, light calcification, respiration or growth, but significantly decreased dark calcification when compared to both controls and DIC treatments. On the ecosystem level, high DIC concentrations may lead to reduced accretion and growth of reefs dominated by Acropora that under elevated DOC concentrations will likely exhibit reduced primary production rates, ultimately leading to loss of hard substrate and reef erosion. It is therefore important to consider the potential impacts of elevated DOC and DIC simultaneously to assess real world scenarios, as multiple rather than single factors influence key physiological processes in coral reefs. Knowledge on interactive effects of global (e.g. ocean warming) and local stressors (e.g. pollution) is needed to develop appropriate management strategies for coral reefs. Surfactants and diesel are common coastal pollutants, but knowledge of their effects on hard corals as key reef ecosystem engineers is scarce. This study thus investigated the physiological reaction of Pocillopora verrucosa from Lombok, Indonesia, to exposure with a) the water-soluble fraction of diesel (determined by total polycyclic aromatic hydrocarbons (PAH); 0.69 ± 0.14 mg L-1), b) the surfactant linear alkylbenzene sulfonate (LAS; 0.95 ± 0.02 mg L-1) and c) combinations of each pollutant with high temperature (+3°C). To determine effects on metabolism, respiration, photosynthetic efficiency and coral tissue health were measured. Findings revealed no significant effects of diesel, while LAS resulted in severe coral tissue losses (16–95% after 84 h). High temperature led to an increase in photosynthetic yield of corals after 48 h compared to the control treatment, but no difference was detected thereafter. In combination, diesel and high temperature significantly increased coral dark respiration, whereas LAS and high temperature caused higher tissue losses (81–100% after 84 h) and indicated a severe decline in maximum quantum yield. These results confirm the hypothesized combined effects of high temperature with either of the two investigated pollutants. Worldwide, coral reefs are challenged by multiple stressors due to growing urbanization, industrialization and coastal development. Coral reefs along the Thousand Islands off Jakarta, one of the largest megacities worldwide, have degraded dramatically over recent decades. The shift and decline in coral cover and composition has been extensively studied with a focus on large-scale gradients (i.e. regional drivers), however special focus on local drivers in shaping spatial community composition is still lacking. Here, the spatial impact of anthropogenic stressors on local and regional scales on coral reefs north of Jakarta was investigated. Results indicate that the direct impact of Jakarta is mainly restricted to inshore reefs, separating reefs in Jakarta Bay from reefs along the Thousand Islands further north. A spatial patchwork of differentially degraded reefs is present along the islands as a result of localized anthropogenic effects rather than regional gradients. Pollution is the main anthropogenic stressor, with over 80% of variation in benthic community composition driven by sedimentation rate, NO2, PO4 and Chlorophyll a. Thus, the spatial structure of reefs is directly related to intense anthropogenic pressure from local as well as regional sources.
