This paper reports the fossil zokors (Myospalacinae) collected from the lower Pliocene (~4.4 Ma) of Zanda Basin, southwestern Tibet, which is the first record in the hinterland of Tibetan Plateau within the Himalayan Range. Materials include 29 isolated molars belonging to Prosiphneus eriksoni (Schlosser, 1924) by having characters including large size, highly fused roots, upper molars of orthomegodont type, m1 anterior cap small and centrally located, and first pair of m1 reentrants on opposing sides, high crowns, and high value of dentine tract parameters. Based on the cladistics analysis, all seven species of Prosiphneus and P. eriksoni of Zanda form a monophyletic clade. P. eriksoni from Zanda, on the other hand, is nearly the terminal taxon of this clade. The appearance of P. eriksoni in Zanda represents a significant dispersal in the early Pliocene from its center of origin in north China and Mongolian Plateau, possibly via the Hol Xil-Qiangtang hinterland in northern Tibet. The fast evolving zokors are highly adapted to open terrains at a time when regional climates had become increasingly drier in the desert zones north of Tibetan Plateau during the late Miocene to Pliocene. The occurrence of this zokor in Tibet thus suggests a rather open steppe environment. Based on fossils of large mammals, we have formulated an “out of Tibet” hypothesis that suggests earlier and more primitive large mammals from the Pliocene of Tibet giving rise to the Ice Age megafauna. However, fossil records for large mammals are still too poor to evaluate whether they have evolved from lineages endemic to the Tibetan Plateau or were immigrants from outside. The superior record of small mammals is in a better position to address this question. With relatively dense age intervals and numerous localities in much of northern Asia, fossil zokors provide the first example of an “into Tibet” scenario–earlier and more primitive taxa originated from outside of the Tibetan Plateau and the later the lineage became extinct in southwestern Tibet. We have used bias-corrected data from CMIP6 models to drive a regional climate model and project climate on the Tibetan Plateau (TP) in the 21st century. Changes in two background fields, namely, 2-meter air temperature and total precipitation, were analyzed. The results show that the WRF simulations capture the terrain effect that cannot be represented in low-resolution models. The simulation of temperature is better in summer than in winter, while the simulated precipitation is the opposite. By the end of the 21st century, the entire TP region experiences significant warming, with an average warming of 3°C and 7°C in the SSP245 and SSP585 scenarios, respectively. The western region shows a greater warming amplitude, with a maximum of more than 10°C in the SSP585 scenario. Most regions of the TP had significant increases in precipitation by the end of the 21st century, with precipitation increasing by 90 mm and 200 mm in the two scenarios, respectively. However, in the low-altitude areas of southeastern TP, total winter precipitation is significantly reduced in the SSP585 scenario. The strengthening of the East Asian summer monsoon and westerly disturbances collectively leads to a significant increase in precipitation within the TP region. The Tibetan Plateau (TP), once considered a pristine environment, is now facing increased heavy metal pollution due to human activities, causing unprecedented ecological risks to soil organisms. However, little is known about the sensitivity and tolerance of different soil organisms to heavy metal toxicity in the high-altitude areas of the TP under the background of human activity intensity and future risk control priorities. In this study, we conducted an ecological risk assessment and threshold calculation for 10 heavy metals in soil for typical soil organisms, including Cd, Co, Cr, Cu, Ni, Pb, Zn, Mn, Sb, and Sn, using the species sensitivity distribution (SSD) method in the zone between Ranwu town and Renlongba glacier on the TP. The results revealed that most sampling sites had average levels of Cd, Sb and Ni exceeding their regional soil environmental background values and were the major contaminants. Impressively, the hitherto undeveloped Renlungba Glacier showed relatively high contamination levels of Sb and Ni. From the view of sensitivity differences, the toxicity risks of Cd, Cr, Cu, Ni and Pb were higher for terrestrial plants than for soil invertebrates based on the fitted heavy metal SSD curve trends. In terms of the ecological risk level, the average potentially affected fraction values of Zn and Ni reached 18.5% and 17.0%, respectively, with significant ecological risk at a few sampling sites. In terms of ecological risk thresholds, the Cd concentration at the 5% hazard concentration (HC5) control level was 0.05 mg/kg, which was the heavy metal with the highest risk in this study. Comparing the HC5 values of each heavy metal with the limit values in the current Chinese soil environmental quality standards, the existing administrative policies as a whole lack a powerful prevention of the potential ecological risk posed by heavy metals to soil organisms. This research was designed to explore the variation characteristics of soil C:N:P stoichiometry and enzyme activity in the Qilian Mountains different grassland types. Thus, 7 grassland types (Upland meadow: UM, Alpine meadow: AM, Temperate steppe: ST, Alpine steppe: AS, Temperate Desert Steppe: TDS, Temperate Desert: TD, Alpine desert: AD) of Qilian Natural Reserve were selected to analyze the variation characteristics of soil enzyme activities and stoichiometry of different grassland types and its relationship with environmental factors. The study indicated that the C/N, C/P, and N/P of different grasslands ranged from 5.08 to 17.35, 2.50 to 72.29, and 0.53 to 4.02.The ranking of different types grassland for the C/N was TS ≥ AM ≥ UM ≥ AS ≥ TDS > AD > TD, and the changing pattern of C/P and N/P is similar to that of C/N. The ranking of different types grassland for the urease enzyme activity was UM ≈AS > AD ≈TDS ≈TS ≈AM > TD, and TS ≈AM ≈UM ≈AS ≈AD > TDS > TD for alkaline phosphatase enzyme activity, and AS ≈AM ≈TS ≈TDS≥UM ≥TD ≈AD for catalase enzyme activity. Based on N/P ratio and RDA analysis, nitrogen was the main factor limiting the grassland productivity, and pH, TN, SOC, Richness index and Simpson diversity index were the main environmental factors affecting the soil C:N:P stoichiometry and enzyme activities. Cluster analysis showed that 7 grassland types were clustered into three categories. In conclusion, the stoichiometric characteristics and soil enzyme activities of different grasslands vary with grassland types. Nitrogen was the main factor limiting the grasslands productivity, and pH, TN, SOC, Richness index and Simpson diversity index were the main environmental factors affecting the soil C:N:P stoichiometry and enzyme activities, and the grassland Qilian Mountain can be managed in the ecological district according to the clustering results. The immense and towering Tibetan Plateau acts as a heating source and, thus, deeply shapes the climate of the Eurasian continent and even the whole world. However, due to the scarcity of meteorological observation stations and very limited climatic data, little is quantitatively known about the heating effect of the plateau and its implications. This paper firstly collects climate data (2001–2007) from 109 observation stations and MODIS-based estimated monthly mean temperature data in the plateau and the neighboring Sichuan Basin, and conducts correlation and simple linear regression to reveal the altitudinal pattern of temperature. Then, according to the linear relationships of temperature and altitude for each month, it compares air temperature differences on the same elevation between the main plateau and surrounding mountains and the Sichuan Basin so as to quantify the heating effect and discuss its implication on timberline of the plateau. The results show that: 1) the heating effect of the plateau is significant. The temperature of the main plateau area was higher than that of free air on the same elevation above the neighboring areas; on the elevation of 4500 m (the main plateau), temperature is 1–6°C higher in the main Plateau than over the Sichuan Basin for different months and 5.9–10.7°C higher than in the Qilian Mountains in the northeastern corner of the plateau. 2) Even at altitudes of 5000–6000 m in the main Plateau, there are 4 months with a mean temperature above 0°C. The mean temperature of the warmest month (July) can reach 10°C at about 4600–4700 m. Steep canyons surrounded by high mountains resulting from large-scale landslides characterize the study area located in the southeastern part of the Tibetan Plateau. A total of 1766 large landslides were identified based on integrated remote sensing interpretations utilizing multisource satellite images and topographic data that were dominated by 3 major regional categories, namely, rockslides, rock falls, and flow-like landslides. The geographical detector method was applied to quantitatively unveil the spatial association between the landslides and 12 environmental factors through computation of the q values based on spatially stratified heterogeneity. Meanwhile, a certainty factor (CF) model was used for comparison. The results indicate that the q values of the 12 influencing factors vary obviously, and the dominant factors are also different for the 3 types of landslides, with annual mean precipitation (AMP) being the dominant factor for rockslide distribution, elevation being the dominant factor for rock fall distribution and lithology being the dominant factor for flow-like distribution. Integrating the results of the factor detector and ecological detector, the AMP, annual mean temperature (AMT), elevation, river density, fault distance and lithology have a stronger influence on the spatial distribution of landslides than other factors. Furthermore, the factor interactions can significantly enhance their interpretability of landslides, and the top 3 dominant interactions were revealed. Based on statistics of landslide discrepancies with respect to diverse stratification of each factor, the high-risk zones were identified for 3 types of landslides, and the results were contrasted with the CF model. In conclusion, our method provides an objective framework for landslide prevention and mitigation through quantitative, spatial and statistical analyses in regions with complex terrain. As the two limiting nutrients for plants in most terrestrial ecosystems, nitrogen (N) and phosphorus (P) are essential for the development of succession forests. Vegetation N:P stoichiometry is a useful tool for detecting nutrient limitation. In the present work, chronosequence analysis was employed to research N and P accumulation dynamics and their stoichiometry during forest primary succession in a glacier retreat area on the Tibetan Plateau. Our results showed that: (1) total ecosystem N and P pools increased from 97 kg hm−2 to 7186 kg hm−2 and 25 kg hm−2 to 487 kg hm−2, respectively, with increasing glacier retreat year; (2) the proportion of the organic soil N pool to total ecosystem N sharply increased with increasing glacier retreat year, but the proportion of the organic soil and the vegetation P pools to the total ecosystem P was equivalent after 125 y of recession; (3) the N:P ratio for tree leaves ranged from 10.1 to 14.3, whereas the N:P ratio for total vegetation decreased form 13.3 to 8.4 and remained constant after 35 y of recession, and the N:P ratio for organic soil increased from 0.2 to 23.1 with increasing glacier retreat. These results suggested that organic soil N increased with increasing years of glacier retreat, which may be the main sink for atmospheric N, whereas increased P accumulation in vegetation after 125 y of recession suggested that much of the soil P was transformed into the biomass P pool. As the N:P ratio for vegetation maintained a low level for 35–125 y of recession, we suggested that N might be the main limiting element for plant growth in the development of this ecosystem. Batrachuperus yenyuanensis, commonly known as Yenyuan Stream Salamander, is a hynobiid species inhabiting high-altitude (2440–4025 m above sea level) mountain stream and pond environments along the eastern fringe of the Qinghai-Tibetan Plateau in western Sichuan Province, China. Although the species has been known for almost 70 years since its initial discovery in 1950, a thorough osteological description has never been provided. Our study provides a detailed account of the bony anatomy of this species, based on micro computed tomography scanning of multiple specimens collected from the type locality Shuangertang at Bailinshan, Yanyuan County, and several other localities in Sichuan Province. Our revised species diagnosis utilizes both bony and soft anatomical features. Comparative study of the specimens from the type locality in Yanyuan area with those from the nearby Xichang and Mianning areas confirms that they all pertain to Batrachuperus yenyuanensis, thereby removing doubt on the occurrence of the species in the latter areas. With this confirmation, the distribution of the species is extended from the type locality northwards some 180 km to the Mianning area, on both the west and east sides of the Yalong River, which is a major tributary of the upper Yangtze River. This distribution pattern indicates that the biogeographic origin and historical evolution of the species are closely associated with the orogeny of the Hengduan Mountains and formation of the Yalong River. Given the basalmost position of Batrachuperus yenyuanensis in relation to other congeneric species based on molecular studies, however, early expansion of the species distribution by dispersal is expected following the origin of the genus in early–middle Miocene in western Sichuan Province. Dissolved organic carbon (DOC) released from glaciers has an important role in the biogeochemistry of glacial ecosystems. This study focuses on DOC from glaciers of the southeastern Tibetan Plateau, where glaciers are experiencing rapid shrinkage. We found that concentrations of DOC in snowpits (0.16±0.054 μg g−1), aged snow (0.16±0.048 μg g−1), and bare ice (0.18±0.082 μg g−1) were similar across the southeastern Tibetan Plateau, but were slightly lower than those in other glaciers on the Tibetan Plateau. Vertical variations of DOC, particulate organic carbon, black carbon, and total nitrate in snowpit showed no systematic variations in the studied glaciers, with high values of DOC occurring in the ice or dusty layers. We estimated the export of DOC and particulate organic carbon from glaciers to be 1.96±0.66 Gg yr–1 and 5.88±2.15 Gg yr–1 in this region, respectively, indicating that organic carbon released from glacier meltwater may be affecting downstream ecosystems. Potential sources of the air masses arriving at the southeastern Tibetan glaciers include South Asia, Central Asia, Middle East, and northwest China. Emissions from biomass burning of South Asia played an important role in the deposition of DOC to the glacier, which can be evidenced by backward trajectories and fire spot distributions from MODIS and CALIPSO images. Our findings suggest that anthropogenic aerosols contribute abundant DOC to glaciers on the southeastern Tibetan Plateau. The Late Cenozoic East Asian winter monsoon (EAWM) enhancement has been attributed to several factors, such as uplift of the Tibetan Plateau, retreat of the Paratethys Sea, and global cooling related to polar ice volume increment. However, the fundamental forcing factors remain enigmatic due to the absence of long and continuous climate records and sensitive indicators. Here we reanalyzed the published grain-size record of Sikouzi fine sediments in the western Chinese Loess Plateau through end-member (EM) modeling. The results indicate that EM 2 with grain-size peaks between 10–100 μm decreased in content from 20.1 to 17 Ma and stepwise increased from 17 to 0.07 Ma during the following six stages (17–15 Ma, 15–12 Ma, 12–8 Ma, 8–6 Ma, 6–4 Ma and 4–0 Ma). Such varying trends can be successively correlated in seven stages with the integrated benthic δ18O record, implying that global warming weakened the EAWM from 20.1 to 17 Ma and global cooling has stepwise strengthened the EAWM since 17 Ma. Therefore, we conclude that global temperature change played a major role on the evolution of EAWM during the Neogene period. By contrast, Late Cenozoic palaeogeographic reorganization caused by uplift of the Tibetan Plateau and retreat of the Paratethys Sea contributed less to the evolutionary evolution of EAWM. Spectral analysis of the EM 2 data first provided direct evidence of orbitally influenced deposition in the study area and thus the EAWM variations during the Neogene period. The 100-kyr period became weak since ~10 Ma, possibly due to the decrease in sensitivity of a more stable, continental-scale ice sheet in Antarctica to local insolation forcing, deserving further investigation. Phylogeographical studies have suggested that several plant species on the Tibetan Plateau (TP) underwent recolonization during the Quaternary and may have had distinct range dynamics in response to the last glacial. To further test this hypothesis and locate the possible historical dispersal routes, we selected 20 plant species from different parts of the TP and modeled their geographical distributions over four time periods using species distribution models (SDMs). Furthermore, we applied the least-cost path method together with SDMs and shared haplotypes to estimate their historical dispersal corridors. We identified three general scenarios of species distribution change during the late Quaternary: the ‘contraction-expansion’ scenario for species in the northeastern TP, the ‘expansion-contraction’ scenario for species in the southeast and the ‘stable’ scenario for widespread species. During the Quaternary, we identified that these species were likely to recolonize along the low-elevation valleys, huge mountain ranges and flat plateau platform (e.g. the Yarlung Zangbo Valley and the Himalaya). We inferred that Quaternary cyclic glaciations along with the various topographic and climatic conditions of the TP could have resulted in the diverse patterns of range shift and dispersal of Tibetan plant species. A new species, Ptilagrostis contracta, endemic to Qinghai-Tibet Plateau is described and illustrated. It is distinguished from other species in Ptilagrostis by having contracted panicles, 1-geniculate awns with hairy columns and scabrous bristles and evenly pubescent lemmas. Evidence from lemma epidermal pattern, cytology and molecular phylogenetic analyses based on the nuclear ITS sequence data confirm its systematic position in Ptilagrostis. [ABSTRACT FROM AUTHOR]

Pastoralists on the Tibetan alpine rangeland suffered great economic loss in cold season, due to serious live-weight loss of domestic livestock under traditional grazing management. This study aimed to evaluate the effect of dietary types (crude protein levels) on feed intakes, growth performance and economic returns of local Tibetan sheep and yaks during cold season. Twenty-four yearling Tibetan sheep (25.29±3.95 kg LW) and twenty two-year-old yaks (100.62±4.55 kg LW) with familiar body conditions were randomly assigned to four groups, fed oats hay (OH), oats silage (OS), total mixed ration (TMR) and traditionally grazed on the local cool-season pasture (TG), respectively, over a 135-day experiment. Daily dry matter intake was determined; all animals were weighed at the beginning and every 15 days of the 135-day experiment. Then, the total live-weight gain, average daily live-weight gain, gain rate, feed efficiency and net economic benefit were calculated. Results indicated that feed and nutrient intakes (DMI, DMI/kg LW, DMI/kg LW0.75 and CPI) of TMR, OH and OS were higher than TG (P < 0.05). Grazing animals suffered serious live-weight loss, while TMR, OS and OH significantly (P < 0.05) improved total live-weight gain and gain rate in both Tibetan sheep and yaks during the entire experiment. TMR worked better in animal performance and feed efficiency, obtained the highest breeding profit in both Tibetan sheep and yaks among four treatments (P < 0.05). When expressed on net economic benefit, TMR shared the highest net economic benefit in Tibetan sheep, OH shared the highest net economic benefit in yaks, but, no significant difference of net economic benefit in yaks fed TMR and OH diets was determined (P > 0.05). Results indicated that TMR was a reasonable diet in promoting feed intakes, animal performance, feed efficiency and economic returns in domestic livestock, which should be considered by local herdsmen to increase their breeding profit during cold season. This work analyzed carbon dioxide exchange and its controlling factors over an alpine grassland on the eastern Qinghai-Tibetan Plateau. The main results show that air temperature and photosynthetically active radiation are two dominant factors controlling daily gross primary production. Soil temperature and soil water content are the main factors controlling ecosystem respiration. Canopy photosynthetic activity is also responsible for the variation of daily ecosystem respiration other than environmental factors. No clear correlation between net ecosystem exchange and environmental factors was observed at daily scale. Temperature sensitive coefficient was observed to increase with larger soil water content. High values of temperature sensitive coefficient occurred during the periods when soil water content was high and grass was active. Annual integrated net ecosystem exchange, gross primary production and ecosystem respiration were -191, 1145 and 954 g C m-2 for 2010, and -250, 975 and 725 g C m-2 for 2011, respectively. Thus, this alpine grassland was a moderate carbon sink in both of the two years. Compared to alpine grasslands on the Qinghai-Tibetan Plateau, this alpine grassland demonstrated a much greater potential for carbon sequestration than others. Annual precipitation is a dominant factor controlling the variation of annual net ecosystem exchange over this grassland. The difference in gross primary production between the two years was not caused by the variation in annual precipitation. Instead, air temperature and the length of growing season had an important impact on annual gross primary production. Hypoxia and low temperature at high altitudes are the main environmental pressures for alpine animals, inducing phenotypic plasticity at several levels. To investigate the effect of these variables on the organ mass of Phrynocephalus vlangalii, 138 individuals belonging to four populations living along an altitudinal gradient in the Qinghai-Tibet Plateau (China) were dissected to remove heart, lungs, stomach, and intestinal tract. Organ dry mass, individuals’ sex, and body mass, as well as mean annual temperature and average air pressure (calculated from a 30-year-data series obtained from the National Climatic Data Center) were subjected to two-way analyses of covariance and generalized linear mixed models (GLMMs). Except for the heart, organ mass varied significantly among populations, although only lung and stomach mass increased significantly with increasing altitude. Males’ heart and lung mass was higher than that of females, which might be due to their different behavior and reproductive efforts. GLMM analyses indicated that air pressure had a positive effect on heart, lung and intestinal tract mass, whereas temperature had a negative effect on these three organs. In order to explain the effect of hypoxia and low temperature on P. vlangalii’s organ mass, further rigorous study on respiration, energy budget and food intake was encouraged. Estimating the potential of species to cope with rapid environmental climatic modifications is of vital importance for determining their future viability and conservation. The variation between existing populations along a climatic gradient may predict how a species will respond to future climate change. Stipa purpurea is a dominant grass species in the alpine steppe and meadow of the Qinghai-Tibetan Plateau (QTP). Ecological niche modelling was applied to S. purpurea, and its distribution was found to be most strongly correlated with the annual precipitation and the mean temperature of the warmest quarter. We established a north-to-south transect over 2000 km long on the QTP reflecting the gradients of temperature and precipitation, and then we estimated the morphological by sampling fruited tussocks and genetic divergence by using 11 microsatellite markers between 20 populations along the transect. Reproductive traits (the number of seeds and reproductive shoots), the reproductive-vegetative growth ratio and the length of roots in the S. purpurea populations varied significantly with climate variables. S. purpurea has high genetic diversity (He = 0.585), a large effective population size (Ne >1,000), and a considerable level of gene flow between populations. The S. purpurea populations have a mosaic genetic structure: some distant populations (over 1000 km apart) clustered genetically, whereas closer populations (< 100 km apart) had diverged significantly, suggesting local adaptation. Asymmetrical long-distance inter-population gene flow occurs along the sampling transect and might be mediated by seed dispersal via migratory herbivores, such as the chiru (Pantholops hodgsonii). Although only relatively few freshwater invertebrate families are reported from the Tibetan Plateau, the degree of endemism may be high. Many endemic lineages occur within permafrost areas, raising questions about the existence of isolated intra-plateau glacial refugia. Moreover, if such refugia existed, it might be instructive to learn whether they were associated with lakes or with more dynamic ecosystems such as ponds, wetlands, or springs. To study these hypotheses, we used pulmonate snails of the plateau-wide distributed genus Radix as model group and the Lake Donggi Cona drainage system, located in the north-eastern part of the plateau, as model site. First, we performed plateau-wide phylogenetic analyses using mtDNA data to assess the overall relationships of Radix populations inhabiting the Lake Donggi Cona system for revealing refugial lineages. We then conducted regional phylogeographical analyses applying a combination of mtDNA and nuclear AFLP markers to infer the local structure and demographic history of the most abundant endemic Radix clade for identifying location and type of (sub-)refugia within the drainage system. Our phylogenetic analysis showed a high diversity of Radix lineages in the Lake Donggi Cona system. Subsequent phylogeographical analyses of the most abundant endemic clade indicated a habitat-related clustering of genotypes and several Late Pleistocene spatial/demographic expansion events. The most parsimonious explanation for these patterns would be a scenario of an intra-plateau glacial refugium in the Lake Donggi Cona drainage system, which might have consisted of isolated sub-refugia. Though the underlying processes remain unknown, an initial separation of lake and watershed populations could have been triggered by lake-level fluctuations before and during the Last Glacial Maximum. This study inferred the first intra-plateau refugium for freshwater animals on the Tibetan Plateau. The alpine grassland ecosystem can sequester a large quantity of carbon, yet its significance remains controversial owing to large uncertainties in the relative contributions of climate factors and grazing intensity. In this study we surveyed 115 sites to measure ecosystem carbon storage (both biomass and soil) in alpine grassland over the Qinghai Plateau during the peak growing season in 2011 and 2012. Our results revealed three key findings. (1) Total biomass carbon density ranged from 0.04 for alpine steppe to 2.80 kg C m-2 for alpine meadow. Median soil organic carbon (SOC) density was estimated to be 16.43 kg C m-2 in alpine grassland. Total ecosystem carbon density varied across sites and grassland types, from 1.95 to 28.56 kg C m-2. (2) Based on the median estimate, the total carbon storage of alpine grassland on the Qinghai Plateau was 5.14 Pg, of which 94% (4.85 Pg) was soil organic carbon. (3) Overall, we found that ecosystem carbon density was affected by both climate and grazing, but to different extents. Temperature and precipitation interaction significantly affected AGB carbon density in winter pasture, BGB carbon density in alpine meadow, and SOC density in alpine steppe. On the other hand, grazing intensity affected AGB carbon density in summer pasture, SOC density in alpine meadow and ecosystem carbon density in alpine grassland. Our results indicate that grazing intensity was the primary contributing factor controlling carbon storage at the sites tested and should be the primary consideration when accurately estimating the carbon storage in alpine grassland. Knowledge about the spatiotemporal tree growth variability and its associations with climate provides key insights into forest dynamics under future scenarios of climate change. We synthesized 17 tree-ring width chronologies from four tree species at the high-elevation sites in the southeast Tibetan Plateau (SETP) to study the regional tree growth variability and climate-growth relationships. Despite of diverse habitats and different physiological characteristics of these species, these tree-ring chronologies shared a significant common variance in SETP. An unprecedented increase in the shared variance is found along the latter half of the 20th century, coinciding with the enhancement of the frequency of extreme rings among chronologies. It is found that minimum winter temperature tends to be the dominant climate for trees in this region. The site-specific responses in cold (1965–1980) and warm (1990–2005) intervals by means of Fuzzy Cmeans (FCM) clustering reveal that the remarkable enhancement of growth synchrony among trees mainly occur in warm conditions. This is different from previous findings indicating that increased consistence among temperature sensitive tree rings in cold periods. This may be related to the reduced temperature sensitivity of regional tree growth as winter minimum temperature is lower than a certain threshold, which is in agreement with the “principle of ecological amplitude”. In addition, it is worth noting that precipitation in June have started to restrain the tree growth since the beginning of the 1980s, which is possibly an important contributor for synchronized growth among trees in SETP. Water is a limiting factor for plant growth and vegetation dynamics in alpine sandy land of the Tibetan Plateau, especially with the increasing frequency of extreme precipitation events and drought caused by climate change. Therefore, a relatively stable water source from either deeper soil profiles or ground water is necessary for plant growth. Understanding the water use strategy of dominant species in the alpine sandy land ecosystem is important for vegetative rehabilitation and ecological restoration. The stable isotope methodology of δD, δ18O, and δ13C was used to determine main water source and long-term water use efficiency of Salix psammophila and S. cheilophila, two dominant shrubs on interdune of alpine sandy land in northeastern Tibetan Plateau. The root systems of two Salix shrubs were investigated to determine their distribution pattern. The results showed that S. psammophila and S. cheilophila absorbed soil water at different soil depths or ground water in different seasons, depending on water availability and water use strategy. Salix psammophila used ground water during the growing season and relied on shallow soil water recharged by rain in summer. Salix cheilophila used ground water in spring and summer, but relied on shallow soil water recharged by rain in spring and deep soil water recharged by ground water in fall. The two shrubs had dimorphic root systems, which is coincident with their water use strategy. Higher biomass of fine roots in S. psammophila and longer fine roots in S. cheilophila facilitated to absorb water in deeper soil layers. The long-term water use efficiency of two Salix shrubs increased during the dry season in spring. The long-term water use efficiency was higher in S. psammophila than in S. cheilophila, as the former species is better adapted to semiarid climate of alpine sandy land. [ABSTRACT FROM AUTHOR]

In the alpine steppe zone on the Central Tibetan Plateau, a large amount of area has been degraded due to natural and artificial factors. N & P fertilization is widely accepted to recover degraded pastures in other regions all over the world. However, it is not clear how alpine steppe communities respond to N & P fertilization, and what is the optimal application rate, in the perspective of forage production. To attempt to explore these questions, in July 2013, two fencing sites were designed in Baingoin County with 12 treatments of different levels of nitrogen (N0: 0; N1: 7.5 g m-2 yr-1; N2: 15 g m-2 yr-1) & phosphate (P0: 0; P1: 7.5 gP2O5 m-2 yr-1; P2: 15 gP2O5 m-2 yr-1; P3: 30 gP2O5 m-2 yr-1). The results indicated N&P addition was capable to ameliorate the quality of the two sites in the Tibetan Plateau steppe. Increasing N application level resulted in significant increment in Gramineae and total biomass in the two sites. P addition significantly improved the quantity of Compositae, total biomass and the biomasss of other species in site II, while it only significantly improved the total biomass in site I. Gramineae was much more sensitive to N-induced changes than P-induced changes, and this indicated N addition was better to ameliorate the quality of plateau steppe than P-induced changes. No strong evidence was found for critical threshold within 15 g N m-2 yr-1, and there was decreasing tendency when P addition rate was above 15 g m-2 yr-1. N&P has the potential to accelerate soil acidification, which improved the content of available K, likely as a result of nonsignificant correlation between biomass and soil moisture. Glaciochemistry can provide important information about climatic change and environmental conditions, as well as for testing regional and global atmospheric trace transport models. In this study, δ18O and selected chemical constituents records in snowpits collected from eight glaciers in the Tibetan Plateau and adjacent areas have been investigated. Drawing on the integrated data, our study summarized the seasonal and spatial characteristics of snow chemistry, and their potential sources. Distinct seasonal patterns of δ18O values in snowpits indicated more negative in the south TP controlled by Indian monsoon, and less negative in the north TP and Tien Shan. Overall increasing concentrations of microparticles and crustal ions from south to north indicated a strength of dust deposition on glaciers from semi-arid and arid regions. Principal component analysis and air mass trajectories suggested that chemical constituents were mainly attributable to crustal sources as demonstrated by the high concentrations of ions occurring during the non-monsoon seasons. Nevertheless, other sources, such as anthropogenic pollution, played an important role on chemical variations of glaciers near the human activity centers. Biomass partitioning has been explored across various biomes. However, the strategies of allocation in plants still remain contentious. This study investigated allocation patterns of above- and belowground biomass at the community level, using biomass survey from the Tibetan Plateau. We explored above- and belowground biomass by conducting three consecutive sampling campaigns across shrub biomes on the northeast Tibetan Plateau during 2011–2013. We then documented the above-ground biomass (AGB), below-ground biomass (BGB) and root: shoot ratio (R/S) and the relationships between R/S and environment factors using data from 201 plots surveyed from 67 sites. We further examined relationships between above-ground and below-ground biomass across various shrub types. Our results indicated that the median values of AGB, BGB, and R/S in Tibetan shrub were 1102.55, 874.91 g m-2, and 0.85, respectively. R/S showed significant trend with mean annual precipitation (MAP), while decreased with mean annual temperature (MAT). Reduced major axis analysis indicated that the slope of the log-log relationship between above- and belowground biomass revealed a significant difference from 1.0 over space, supporting the optimal hypothesis. Interestingly, the slopes of the allometric relationship between log AGB and log BGB differed significantly between alpine and desert shrub. Our findings supported the optimal theory of above- and belowground biomass partitioning in Tibetan shrub, while the isometric hypothesis for alpine shrub at the community level. The yak is primarily found throughout the Tibetan high plateau and the surrounding mountainous area of south central Asia; among its others attributes, its milk is very important for the local population. A key concern in the field of yak research is the better understanding of which genes control the production and composition of milk. The most accurate and sensitive method for gene expression analysis is quantitative reverse transcription polymerase chain reaction (RT-qPCR). It is essential for reliable RT-qPCR to be able to the normalize the data using internal control genes (ICGs). However, it is critical to assess the reliability of the normalization by testing multiple ICGs. Our objective was to uncover a reliable normalization for RT-qPCR data obtained from yak mammary tissue during the lactation cycle. We assessed the reliability of 10 ICGs (ACTB, EIF6, GAPDH, LRP10, MRPL39, MRPS15, MTG1, RPS8, RPS23, and UXT) using geNorm. The analysis revealed that all of the tested ICGs can be considered to be reliable, but the use of the 6 most stable ICGs should be applied to yield a reliable normalization factor (NF). We compared the results of 3 target genes (CSN1S1, ESR1, and MYC) normalized using 6, 3, or 1 of the best ICGs. We did not observe overall differences between the 3 normalization strategies with the exception of 1 time point in MYC. The use of only a single ICG is not recommended; thus, we concluded that the calculation of the NF using the 3 best ICGs, MRPS15, RPS23, and UXT, is a reliable normalization strategy for RT-qPCR data obtained from yak mammary tissue during pregnancy and lactation. A dilution effect of the ICGs due to a large increase in the mRNA of abundantly expressed genes in bovine and porcine mammary tissue during the lactation cycle was previously observed. To test for the presence of a dilution effect in our study, we evaluated the pattern of non-normalized RT-qPCR data of ICGs from pregnancy to lactation and compared them with the total RNA concentration, milk yield, and non-normalized RT-qPCR data of 3 target genes. With a few exceptions, the non- normalized RT-qPCR data for the tested ICGs was significantly increased by lactation and had a positive correlation with total RNA and the non-normalized RT-qPCR data of CSN1S1. These data clearly indicated the presence of a “concentration effect” of single mRNA that remains unexplained but needs to be accounted for during the normalization of RT-qPCR data. Based on our findings, we recommend that the NF of the MRPS15, RPS23, and UXT genes should be used in the normalization of RT-qPCR data obtained from mammary tissue of lactating yaks during pregnancy and lactation. Understanding the processes that influence the structure of biotic communities is one of the major ecological topics, and both stochastic and deterministic processes are expected to be at work simultaneously in most communities. Here, we investigated the vertical distribution patterns of bacterial communities in a 10-m-long soil core taken within permafrost of the Qinghai-Tibet Plateau. To get a better understanding of the forces that govern these patterns, we examined the diversity and structure of bacterial communities, and the change in community composition along the vertical distance (spatial turnover) from both taxonomic and phylogenetic perspectives. Measures of taxonomic and phylogenetic beta diversity revealed that bacterial community composition changed continuously along the soil core, and showed a vertical distance-decay relationship. Multiple stepwise regression analysis suggested that bacterial alpha diversity and phylogenetic structure were strongly correlated with soil conductivity and pH but weakly correlated with depth. There was evidence that deterministic and stochastic processes collectively drived bacterial vertically-structured pattern. Bacterial communities in five soil horizons (two originated from the active layer and three from permafrost) of the permafrost core were phylogenetically random, indicator of stochastic processes. However, we found a stronger effect of deterministic processes related to soil pH, conductivity, and organic carbon content that were structuring the bacterial communities. We therefore conclude that the vertical distribution of bacterial communities was governed primarily by deterministic ecological selection, although stochastic processes were also at work. Furthermore, the strong impact of environmental conditions (for example, soil physicochemical parameters and seasonal freeze-thaw cycles) on these communities underlines the sensitivity of permafrost microorganisms to climate change and potentially subsequent permafrost thaw. Hypoxia has been reported to cause the significant enhancement of hemoglobin (Hb) and hematocrit (Hct), which stabilizes at relatively high levels after an individual ascends to a high altitude. However, the quality of the suspended red blood cells (SRBCs) obtained from individuals at high altitudes such as Tibetan plateau migrants after storage has not been studied. In this study, we compared the storage quality of SRBCs prepared from Tibetan plateau and Deyang lowland populations by adding a normal volume of mannitol-adenine-phosphate (MAP), which is a common additive solution used in blood storage in Asian countries. The storage cell characteristics were examined on days1, 7, 14 and 35.We found higher Hct and Hb levels and viscosity in the high altitude samples. The metabolic rates, including those for electrolytes and lactate, were higher in plateau SRBCs than in lowland SRBCs; these findings were consistent with the higher osmotic fragility and hemolysis of plateau SRBCs throughout the entire storage period. In addition, the reduction rates of 2,3-diphosphoglycerate (2,3-DPG) and oxygen tension to attain 50% oxygen saturation of Hb (P50) in plateau SRBCs were higher than those in lowland SRBCs, and the oxygen delivering capacity in plateau SRBCs was weaker than that in lowland SRBCs. We concluded that the storage quality of plateau SRBCs was inferior to that of lowland SRBCs when using the same concentration of MAP. Tibetan pigs live between 2500 and 4300 m above sea level on the Tibetan Plateau, and are better adapted to hypoxia than lowland pigs. MicroRNAs (miRNAs) are involved in a wide variety of cellular processes; however, their regulatory role in hypoxia adaptation remains unclear. In this study, miRNA-seq was used to identify differentially expressed miRNAs (DE miRNAs) in the cardiac muscle of Tibetan and Yorkshire pigs, which were both raised in high elevation environments. We obtained 108 M clean reads and 372 unique miRNAs, which included 210 known porcine miRNAs, 136 conserved in other mammals, and 26 novel pre-miRNAs. In addition, 20 DE miRNAs, including 10 up-regulated and 10 down-regulated miRNAs, were also found after comparison between Tibetan and Yorkshire pigs. We predicted miRNA targets based on differential expression and abundance in the two populations. Furthermore, the results of a Kyoto Encyclopedia of Genes and Genomes pathway analysis suggested that DE miRNAs in Tibetan and Yorkshire pigs are involved in hypoxia-related signaling pathways such as the mitogen-activated protein kinase, which is the mechanistic target of rapamycin, and the vascular endothelial growth factor, as well as cancer-related signaling pathways. Five DE miRNAs were randomly selected to validate the results of miRNA-seq using real-time polymerase chain reaction, and the results corresponded to those from the miRNA-seq, confirming that deep-sequencing methods are feasible and efficient. In our study, we identified various previously unknown hypoxia-related miRNAs in pigs, and the data obtained suggest that hypoxia-related miRNA expression patterns are significantly altered in the Tibetan pig compared to other species. Pasture degradation caused by overgrazing and inappropriate fertiliser management is a major production and environmental threat in Qinghai-Tibet Plateau. Previous research has focused on the effects of mixed nitrogen (N) and phosphorus (P) fertiliser and reduced grazing pressure on the plant community of the grassland; however, the role of P and how it interacts with various defoliation (the process of the complete or partial removal of the above-ground parts of plants by grazing or cutting) intensities on the plant and soil of the grassland ecosystem have not been quantified. A field experiment was conducted to quantify how P application in combination of defoliation pressure could impact the dynamic change of the plant and soil in a native alpine grassland ecosystem of the Qinghai-Tibet Plateau, China, from May 2012 to September 2014. A split-plot design with 4 replicates and repeated measures was used to determine the growth and composition of plant community and soil physical and chemical properties under various levels of P fertiliser and defoliation intensity. The results showed that applying 20 kg P/ha increased the herbage yield of Melissitus ruthenica by 68% and total pasture yield by 25%. Close defoliation favoured the growth and plant frequency of the shorter species, whereas lax defoliation favoured that of the taller plant species. Medium P rate and cutting to 3 cm above ground gave an overall best outcome in pasture yield, quality and frequency and soil moisture and nutrient concentration. Application of P fertiliser with a moderate defoliation pressure to promote legume growth and N fixation has the potential to achieve multiple benefits in increasing pasture and livestock production and improving environmental sustainability in the alpine pasture of Qinghai-Tibet Plateau, a fragile and P-deficient ecosystem zone in China and its western neighbouring countries. Disturbances, both human-induced and natural, may re-shape ecosystems by influencing their composition, structure, and functional processes. Plateau zokor (Eospalax baileyi) is a typical subterranean rodent endemic to Qinghai-Tibetan Plateau (QTP), which are considered ecosystem engineers influencing the alpine ecosystem function. It is also regarded as a pest aggravating the degradation of overgrazed grassland and subject to regular control in QTP since 1950s. Climate change has been predicted in this region but little research exists exploring its impact on such subterranean rodent populations. Using plateau zokor as a model, through maximum entropy niche-based modeling (Maxent) and sustainable habitat models, we investigate zokor habitat dynamics driven by the future climate scenarios. Our models project that zokor suitable habitat will increase by 6.25% in 2050 in QTP. The predication indicated more threats in terms of grassland degradation as zokor suitable habitat will increase in 2050. Distribution of zokors will shift much more in their southern range with lower elevation compare to northern range with higher elevation. The estimated distance of shift ranges from 1 km to 94 km from current distribution. Grassland management should take into account such predictions in order to design mitigation measures to prevent further grassland degradation in QTP under climate change scenarios. Background: Infections of Echinococcus granulosus sensu stricto (s.s), E. multilocularis and E. shiquicus are commonly found co-endemic on the Qinghai-Tibet plateau, China, and an efficient tool is needed to facilitate the detection of infected hosts and for species identification. Methodology/Principal Findings: A single-tube multiplex PCR assay was established to differentiate the Echinococcus species responsible for infections in intermediate and definitive hosts. Primers specific for E. granulosus, E. multilocularis and E. shiquicus were designed based on sequences of the mitochondrial NADH dehydrogenase subunit 1 (nad1), NADH dehydrogenase subunit 5 (nad5) and cytochrome c oxidase subunit 1 (cox1) genes, respectively. This multiplex PCR accurately detected Echinococcus DNA without generating nonspecific reaction products. PCR products were of the expected sizes of 219 (nad1), 584 (nad5) and 471 (cox1) bp. Furthermore, the multiplex PCR enabled diagnosis of multiple infections using DNA of protoscoleces and copro-DNA extracted from fecal samples of canine hosts. Specificity of the multiplex PCR was 100% when evaluated using DNA isolated from other cestodes. Sensitivity thresholds were determined for DNA from protoscoleces and from worm eggs, and were calculated as 20 pg of DNA for E. granulosus and E. shiquicus, 10 pg of DNA for E. multilocularis, 2 eggs for E. granulosus, and 1 egg for E. multilocularis. Positive results with copro-DNA could be obtained at day 17 and day 26 after experimental infection of dogs with larval E. multilocularis and E. granulosus, respectively. Conclusions/Significance: The multiplex PCR developed in this study is an efficient tool for discriminating E. granulosus, E. multilocularis and E. shiquicus from each other and from other taeniid cestodes. It can be used for the detection of canids infected with E. granulosus s.s. and E. multilocularis using feces collected from these definitive hosts. It can also be used for the identification of the Echinococcus metacestode larva in intermediate hosts, a stage that often cannot be identified to species on visual inspection. Biomass allocation is an essential concept for understanding above- vs. below-ground functions and for predicting the dynamics of community structure and ecosystem service under ongoing climate change. There is rare available knowledge of grazing effects on biomass allocation in multiple zonal alpine grassland types along climatic gradients across the Northern Tibetan Plateau. We collected the peak above- and below-ground biomass (AGB and BGB) values at 106 pairs of well-matched grazed vs. fenced sites during summers of 2010–2013, of which 33 pairs were subject to meadow, 52 to steppe and 21 to desert-steppe. The aboveground net primary productivity (ANPP) was represented by the peak AGB while the belowground net primary productivity (BNPP) was estimated from ANPP, the ratio of living vs. dead BGB, and the root turnover rate. Two-ways analyses of variance (ANOVA) and paired samples comparisons with t-test were applied to examine the effects of pasture managements (PMS, i.e., grazed vs. fenced) and zonal grassland types on both ANPP and BNPP. Allometric and isometric allocation hypotheses were also tested between logarithmically transformed ANPP and BNPP using standardized major axis (SMA) analyses across grazed, fenced and overall sites. In our study, a high community-dependency was observed to support the allometric biomass allocation hypothesis, in association with decreased ANPP and a decreasing-to-increasing BNPP proportions with increasing aridity across the Northern Tibetan Plateau. Grazing vs. fencing seemed to have a trivial effect on ANPP compared to the overwhelming influence of different zonal grassland types. Vegetation links above- and below-ground ecological functions through integrated meta-population adaptive strategies to the increasing severity of habitat conditions. Therefore, more detailed studies on functional diversity are essentially to achieve conservation and sustainability goals under ongoing climatic warming and intensifying human influences. The authenticity of controversial species is a significant challenge for systematic biologists. Moschidae is a small family of musk deer in the Artiodactyla, composing only one genus, Moschus. Historically, the number of species in the Moschidae family has been debated. Presently, most musk deer species were restricted in the Tibetan Plateau and surrounding/adjacent areas, which implied that the evolution of Moschus might have been punctuated by the uplift of the Tibetan Plateau. In this study, we aimed to determine the evolutionary history and delimit the species in Moschus by exploring the complete mitochondrial genome (mtDNA) and other mitochondrial gene. Our study demonstrated that six species, M. leucogaster, M. fuscus, M. moschiferus, M. berezovskii, M. chrysogaster and M. anhuiensis, were authentic species in the genus Moschus. Phylogenetic analysis and molecular dating showed that the ancestor of the present Moschidae originates from Tibetan Plateau which suggested that the evolution of Moschus was prompted by the most intense orogenic movement of the Tibetan Plateau during the Pliocene age, and alternating glacial-interglacial geological eras. Qinghai-Tibet Plateau grasslands are unique geographical regions and store substantial soil organic matter (SOM) in the soil surface, which make them very sensitive to global climate change. Here, we focused on three main grassland types (alpine meadow, steppe, and desert) and conducted a soil incubation experiment at five different temperatures (5, 10, 15, 20, and 25°C) to investigate SOM decomposition rates (R), temperature sensitivity (Q10), and activation energy (Ea). The results showed that grassland type and incubation temperature had significant impact on R (P < 0.001), and the values of R were exponential correlated with incubation temperature in three alpine grasslands. At the same temperature, R was in the following order: alpine meadow > alpinesteppe > alpine desert. The Q10 values differed significantly among different grasslands, and the overall trends were as follows: alpine meadow (1.56 ± 0.09) < alpine steppe (1.88 ± 0.23) < alpine desert (2.39 ± 0.32). Moreover, the Ea values differed significantly across different grassland types (P < 0.001) and increased with increasing incubation time. The exponential negative correlations between Ea and R at 20°C across all grassland types (all Ps < 0.001) indicated that the substrate-quality temperature hypothesis is applicable to the alpine grasslands. Little information is available on the seasonal response of soil microbial biomass to climate warming even though it is very sensitive to climate change. A two-year field experiment was conducted in the subalpine and alpine forests of the eastern Tibetan Plateau, China. The intact soil cores from 3,600 m site were incubated in three elevations (3,000 m, 3,300 m and 3,600 m) to simulate climate warming. Soil microbial biomass carbon (MBC) and nitrogen (MBN) were measured at different periods (early growing season [EG], late growing season [LG], onset of soil freezing period [OF], deep soil frozen period [DF] and soil thawing period [ET]) from May 2010 to August 2012. Average air temperature and soil temperature increased with the decrease of elevation during the experimental period. MBC and MBN showed a sharp decrease during the OF and ET in both organic layer and mineral layer at the three sites. Additionally, a relatively high MBC was observed during the DF. MBC and MBN in the soil organic layer decreased with the decrease of elevation but the opposite was true in the mineral soil layer. Warming had stronger effects on soil microbial biomass in the organic layer than in the mineral soil layer. The results indicated that future warming would alter soil microbial biomass and biogeochemical cycling in the forest ecosystems on the eastern Tibetan Plateau. [ABSTRACT FROM AUTHOR]

To analyze CO2 fluxes under conditions of climate change in an alpine meadow on the central Qinghai–Tibetan Plateau, we simulated the effect of warming using open top chambers (OTCs) from 2012 to 2014. The OTCs increased soil temperature by 1.62°C (P < 0.05), but decreased soil moisture (1.38%, P < 0.05) during the experiments. The response of ecosystem CO2 fluxes to warming was variable, and dependent on the year. Under conditions of warming, mean gross ecosystem productivity (GEP) during the growing season increased significantly in 2012 and 2014 (P < 0.05); however, ecosystem respiration (ER) increased substantially only in 2012 (P < 0.05). The net ecosystem CO2 exchange (NEE) increased marginally in 2012 (P = 0.056), did not change in 2013(P > 0.05), and increased significantly in 2014 (P = 0.034) under conditions of warming. The GEP was more sensitive to climate variations than was the ER, resulting in a large increase in net carbon uptake under warming in the alpine meadow. Under warming, the 3-year averages of GEP, ER, and NEE increased by 19.6%, 15.1%, and 21.1%, respectively. The seasonal dynamic patterns of GEP and NEE, but not ER, were significantly impacted by warming. Aboveground biomass, particularly the graminoid biomass increased significantly under conditions of warming. Soil moisture, soil temperature, and aboveground biomass were the main factors that affected the variation of the ecosystem CO2 fluxes. The effect of warming on inter- and intra-annual patterns of ecosystem CO2 fluxes and the mechanism of different sensitivities in GEP and ER to warming, require further researched. The Qinghai–Tibetan Plateau is a very large land unit and an important terrestrial ecosystem within the Eurasian continent. Because of the harsh climate associated with the high altitude, alpine meadows on the plateau are susceptible to degradation from overgrazing. For this region, and for other alpine meadow pastures internationally, there is a need to define the sustainable stocking rate, to develop sound policy pertaining to future land use. Here we report biomass and liveweight gain per animal and per ha for pastures grazed by yaks at high, medium, or low stocking rates over 4 growing seasons from 2010 to 2013. Measures of herbage nutritive value are reported. The influence of inter-year variation in precipitation on standing herbage biomass was also evaluated. Higher precipitation increased standing herbage biomass and herbage nutritive value, indicating that vegetation suffered summer water deficit even in this environment. The sustainable stocking rate in this environment was determined to be approximately 1 yak ha-1 (grown from 80 kg to 120 kg liveweight in 90 d). At this stocking rate, yak weight gain per ha was 88% of that achieved at higher stocking rates typically used by farmers, but with little or no evidence of land degradation. [ABSTRACT FROM AUTHOR]

As CO2 concentrations continue to rise and drive global climate change, much effort has been put into estimating soil carbon (C) stocks and dynamics over time. However, the inconsistent methods employed by researchers hamper the comparability of such works, creating a pressing need to standardize the methods for soil organic C (SOC) quantification by the various methods. Here, we collected 712 soil samples from 36 sites of alpine grasslands on the Tibetan Plateau covering different soil depths and vegetation and soil types. We used an elemental analyzer for soil total C (STC) and an inorganic carbon analyzer for soil inorganic C (SIC), and then defined the difference between STC and SIC as SOCCNS. In addition, we employed the modified Walkley-Black (MWB) method, hereafter SOCMWB. Our results showed that there was a strong correlation between SOCCNS and SOCMWB across the data set, given the application of a correction factor of 1.103. Soil depth and soil type significantly influenced on the recovery, defined as the ratio of SOCMWB to SOCCNS, and the recovery was closely associated with soil carbonate content and pH value as well. The differences of recovery between alpine meadow and steppe were largely driven by soil pH. In addition, statistically, a relatively strong correlation between SOCCNS and STC was also found, suggesting that it is feasible to estimate SOCCNS stocks through the STC data across the Tibetan grasslands. Therefore, our results suggest that in order to accurately estimate the absolute SOC stocks and its change in the Tibetan alpine grasslands, adequate correction of the modified WB measurements is essential with correct consideration of the effects of soil types, vegetation, soil pH and soil depth. There is increasing evidence of environmental change impacts on ecosystem processes and services, yet poor understanding of the relative contributions of land use and climate change to ecosystem services variations. Based on detailed meteorological, hydrological records and satellite data over the Yellow River Source Area (YRSA) in Tibetan Plateau from 1980s to 2008, together with a water-yield module of Integrated Valuation of Ecosystem Services and Tradeoffs (InVEST) model and also a Residual Trends (RESTREND) method, we assessed the water supply variations in YRSA during the past three decades and disentangled the relative contributions of land use and climate change. Results show that water supply significantly decreased from 1980 to 2005 and then increased from 2005 to 2008. The quantity slightly decreased from 283.01mm in 1980 to 276.95mm in 1995, 270.12mm in 2000 and 267.97mm in 2005, and it then rebounded slightly to 275.26mm in 2008. The water supply variation ranged from 283.01mm to 267.97mm. Climate change contributed dominantly to water supply decrease from 1980 to 1995, which accounts for approximately 64% of the decrease. During 1995 to 2000, land use contributed more and about 58% to the water supply decrease as the intense human activities. From 2000 to 2005, climate change became a positive contribution to the water supply as the increased precipitation, but the land use still contributed negatively. From 2005 to 2008, both climate and land use have positive impacts, but land use contributed about 61% to the water supply increase. The implementation of the Three Rivers Source Area Ecological Protection Project has greatly improved the vegetation coverage conditions and the water retention ability during this period. We recommend that the implementation of ecological projects, grazing policies and artificial improvement of degraded grassland would help to conserve the water retention ability and increase water supply. Estimate of net ecosystem carbon exchange (NEE) between the atmosphere and terrestrial ecosystems, the balance of gross primary productivity (GPP) and ecosystem respiration (Reco) has significant importance for studying the regional and global carbon cycles. Using models driven by satellite data and climatic data is a promising approach to estimate NEE at regional scales. For this purpose, we proposed a semi-empirical model to estimate NEE in this study. In our model, the component GPP was estimated with a light response curve of a rectangular hyperbola. The component Reco was estimated with an exponential function of soil temperature. To test the feasibility of applying our model at regional scales, the temporal variations in the model parameters derived from NEE observations in an alpine grassland ecosystem on Tibetan Plateau were investigated. The results indicated that all the inverted parameters exhibit apparent seasonality, which is in accordance with air temperature and canopy phenology. In addition, all the parameters have significant correlations with the remote sensed vegetation indexes or environment temperature. With parameters estimated with these correlations, the model illustrated fair accuracy both in the validation years and at another alpine grassland ecosystem on Tibetan Plateau. Our results also indicated that the model prediction was less accurate in drought years, implying that soil moisture is an important factor affecting the model performance. Evidence from molecular genetics demonstrates that Pinus densata is a natural homoploid hybrid originating from the parent species Pinus tabuliformis and Pinus yunnanensis, and ecological selection may have played a role in the speciation of P. densata. However, data on differentiation in adaptive traits in the species complex are scarce. In this study, we performed a common garden test on 16 seedling traits to examine the differences between P. densata and its parental species in a high altitude environment. We found that among the 16 analyzed traits, 15 were significantly different among the species. Pinus tabuliformis had much earlier bud set and a relatively higher bud set ratio but poorer seedling growth, and P. yunnanensis had opposite responses for the same traits. P. densata had the greatest fitness with higher viability and growth rates than the parents. The relatively high genetic contribution of seedling traits among populations suggested that within each species the evolutionary background is complex. The correlations between the seedling traits of a population within a species and the environmental factors indicated different impacts of the environment on species evolution. The winter temperature is among the most important climate factors that affected the fitness of the three pine species. DNA barcoding, the identification of species using one or a few short standardized DNA sequences, is an important complement to traditional taxonomy. However, there are particular challenges for barcoding plants, especially for species with complex evolutionary histories. We herein evaluated the utility of five candidate sequences — rbcL, matK, trnH-psbA, trnL-F and the internal transcribed spacer (ITS) — for barcoding Rhodiola species, a group of high-altitude plants frequently used as adaptogens, hemostatics and tonics in traditional Tibetan medicine. Rhodiola was suggested to have diversified rapidly recently. The genus is thus a good model for testing DNA barcoding strategies for recently diversified medicinal plants. This study analyzed 189 accessions, representing 47 of the 55 recognized Rhodiola species in the Flora of China treatment. Based on intraspecific and interspecific divergence and degree of monophyly statistics, ITS was the best single-locus barcode, resolving 66% of the Rhodiola species. The core combination rbcL+matK resolved only 40.4% of them. Unsurprisingly, the combined use of all five loci provided the highest discrimination power, resolving 80.9% of the species. However, this is weaker than the discrimination power generally reported in barcoding studies of other plant taxa. The observed complications may be due to the recent diversification, incomplete lineage sorting and reticulate evolution of the genus. The β radioactivity of snow-pit samples collected in the spring of 2011 on four Tibetan Plateau glaciers demonstrate a remarkable peak in each snow pit profile, with peaks about ten to tens of times higher than background levels. The timing of these peaks suggests that the high radioactivity resulted from the Fukushima nuclear accident that occurred on March 11, 2011 in eastern Japan. Fallout monitoring studies demonstrate that this radioactive material was transported by the westerlies across the middle latitudes of the Northern Hemisphere. The depth of the peak β radioactivity in each snow pit compared with observational precipitation records, suggests that the radioactive fallout reached the Tibetan Plateau and was deposited on glacier surfaces in late March 2011, or approximately 20 days after the nuclear accident. Sophora moorcroftiana, a Leguminosae shrub species that is restricted to the arid and semi-arid regions of the Qinghai-Tibet Plateau, is an ecologically important foundation species and exhibits substantial drought tolerance in the Plateau. There are no functional genomics resources in public databases for understanding the molecular mechanism underlying the drought tolerance of S. moorcroftiana. Therefore, we performed a large-scale transcriptome sequencing of this species under drought stress using the Illumina sequencing technology. A total of 62,348,602 clean reads were obtained. The assembly of the clean reads resulted in 146,943 transcripts, including 66,026 unigenes. In the assembled sequences, 1534 transcription factors were identified and classified into 23 different common families, and 9040 SSR loci, from di- to hexa-nucleotides, whose repeat number is greater than five, were presented. In addition, we performed a gene expression profiling analysis upon dehydration treatment. The results indicated significant differences in the gene expression profiles among the control, mild stress and severe stress. In total, 4687, 5648 and 5735 genes were identified from the comparison of mild versus control, severe versus control and severe versus mild stress, respectively. Based on the differentially expressed genes, a Gene Ontology annotation analysis indicated many dehydration-relevant categories, including ‘response to water ‘stimulus’ and ‘response to water deprivation’. Meanwhile, the Kyoto Encyclopedia of Genes and Genomes pathway analysis uncovered some important pathways, such as ‘metabolic pathways’ and ‘plant hormone signal transduction’. In addition, the expression patterns of 25 putative genes that are involved in drought tolerance resulting from quantitative real-time PCR were consistent with their transcript abundance changes as identified by RNA-seq. The globally sequenced genes covered a considerable proportion of the S. moorcroftiana transcriptome, and the expression results may be useful to further extend the knowledge on the drought tolerance of this plant species that survives under Plateau conditions. The evolution and current distribution of the Sino-Tibetan flora have been greatly affected by historical geological events, such as the uplift of the Qinghai-Tibetan Plateau (QTP), and Quaternary climatic oscillations. Rhodiola kirilowii, a perennial herb with its distribution ranging from the southeastern QTP and the Hengduan Mountains (HM) to adjacent northern China and central Asia, provides an excellent model to examine and disentangle the effect of both geological orogeny and climatic oscillation on the evolutionary history of species with such distribution patterns. We here conducted a phylogeographic study using sequences of two chloroplast fragments (trnL-F and trnS-G) and internal transcribed spacers in 29 populations of R. kirilowii. A total of 25 plastid haplotypes and 12 ITS ribotypes were found. Molecular clock estimation revealed deep divergence between the central Asian populations and other populations from the HM and northern China; this split occurred ca. 2.84 million year ago. The majority of populations from the mountains of northern China were dominated by a single haplotype or ribotype, while populations of the HM harbored both high genetic diversity and high haplotype diversity. This distribution pattern indicates that HM was either a diversification center or a refugium for R. kirilowii during the Quaternary climatic oscillations. The present distribution of this species on mountains in northern China may have resulted from a rapid glacial population expansion from the HM. This expansion was confirmed by the mismatch distribution analysis and negative Tajima's D and Fu's FS values, and was dated to ca. 168 thousand years ago. High genetic diversity and population differentiation in both plastid and ITS sequences were revealed; these imply restricted gene flow between populations. A distinct isolation-by-distance pattern was suggested by the Mantel test. Our results show that in old lineages, populations may harbour divergent genetic forms that are sufficient to maintain or even increase overall genetic diversity despite fragmentation and low within-population variation. Most glaciers in the Himalayas and the Tibetan Plateau are retreating, and glacier melt has been emphasized as the dominant driver for recent lake expansions on the Tibetan Plateau. By investigating detailed changes in lake extents and levels across the Tibetan Plateau from Landsat/ICESat data, we found a pattern of dramatic lake changes from 1970 to 2010 (especially after 2000) with a southwest-northeast transition from shrinking, to stable, to rapidly expanding. This pattern is in distinct contrast to the spatial characteristics of glacier retreat, suggesting limited influence of glacier melt on lake dynamics. The plateau-wide pattern of lake change is related to precipitation variation and consistent with the pattern of permafrost degradation induced by rising temperature. More than 79% of lakes we observed on the central-northern plateau (with continuous permafrost) are rapidly expanding, even without glacial contributions, while lakes fed by retreating glaciers in southern regions (with isolated permafrost) are relatively stable or shrinking. Our study shows the limited role of glacier melt and highlights the potentially important contribution of permafrost degradation in predicting future water availability in this region, where understanding these processes is of critical importance to drinking water, agriculture, and hydropower supply of densely populated areas in South and East Asia. Leaf nitrogen (N) and phosphorus (P) have been used widely in the ecological stoichiometry to understand nutrient limitation in plant. However,few studies have focused on the relationship between root nutrients and environmental factors. The main objective of this study was to clarify the pattern of root and leaf N and P concentrations and the relationships between plant nitrogen (N) and phosphorus (P) concentrations with climatic factors under low temperature conditions in the northern Tibetan Plateau of China. We conducted a systematic census of N and P concentrations, and the N∶P ratio in leaf and root for 139 plant samples, from 14 species and 7 families in a dry Stipa purpurea alpine steppe on the northern Tibetan Plateau of China. The results showed that the mean root N and P concentrations and the N∶P ratios across all species were 13.05 mg g−1, 0.60 mg g−1 and 23.40, respectively. The mean leaf N and P concentrations and the N∶P ratio were 23.20 mg g−1, 1.38 mg g−1, and 17.87, respectively. Compared to global plant nutrients concentrations, plants distributing in high altitude area have higher N concentrations and N∶P, but lower P concentrations, which could be used to explain normally-observed low growth rate of plant in the cold region. Plant N concentrations were unrelated to the mean annual temperature (MAT). The root and leaf P concentrations were negatively correlated with the MAT, but the N∶P ratios were positively correlated with the MAT. It is highly possible this region is not N limited, it is P limited, thus the temperature-biogeochemical hypothesis (TBH) can not be used to explain the relationship between plant N concentrations and MAT in alpine steppe. Plant senescence is a critical life history process accompanied by chlorophyll degradation and has large implications for nutrient resorption and carbohydrate storage. Although photoperiod governs much of seasonal leaf senescence in many plant species, temperature has also been shown to modulate this process. Therefore, we hypothesized that climate warming would significantly impact the length of the plant growing season and ultimate productivity. To test this assumption, we measured the effects of simulated autumn climate warming paradigms on four native herbaceous species that represent distinct life forms of alpine meadow plants on the Tibetan Plateau. Conditions were simulated in open-top chambers (OTCs) and the effects on the degradation of chlorophyll, nitrogen (N) concentration in leaves and culms, total non-structural carbohydrate (TNC) in roots, growth and phenology were assessed during one year following treatment. The results showed that climate warming in autumn changed the senescence process only for perennials by slowing chlorophyll degradation at the beginning of senescence and accelerating it in the following phases. Warming also increased root TNC storage as a result of higher N concentrations retained in leaves; however, this effect was species dependent and did not alter the growing and flowering phenology in the following seasons. Our results indicated that autumn warming increases carbohydrate accumulation, not only by enhancing activities of photosynthetic enzymes (a mechanism proposed in previous studies), but also by affecting chlorophyll degradation and preferential allocation of resources to different plant compartments. The different responses to warming can be explained by inherently different growth and phenology patterns observed among the studied species. The results implied that warming leads to changes in the competitive balance among life forms, an effect that can subsequently shift vegetation distribution and species composition in communities. Understanding the impact of grazing intensity on grassland production and soil fertility is of fundamental importance for grassland conservation and management. We thus compared three types of alpine steppe management by studying vegetation traits and soil properties in response to three levels of grazing pressure: permanent grazing (M1), seasonal grazing (M2), and grazing exclusion (M3) in the alpine steppe in Xainza County, Tibetan Plateau. The results showed that community biomass allocation did not support the isometric hypothesis under different grassland management types. Plants in M1 had less aboveground biomass but more belowground biomass in the top soil layer than those in M2 and M3, which was largely due to that root/shoot ratios of dominant plants in M1 were far greater than those in M2 and M3. The interramet distance and the tiller size of the dominant clonal plants were greater in M3 than in M1 and M2, while the resprouting from rhizome buds did not differ significantly among the three greezing regimes. Both soil bulk density and soil available nitrogen in M3 were greater than in M1 at the 15–30 cm soil depth (P = 0.05). Soil organic carbon and soil total nitrogen were greater in M3 than in M1 and M2 (P = 0.05). We conclude that the isometric hypothesis is not supported in this study and fencing is a helpful grassland management in terms of plant growth and soil nutrient retention in alpine steppe. In grassland ecosystems, N and P fertilization often increase plant productivity, but there is no consensus if fertilization affects soil C fractions. We tested effects of N, P and N+P fertilization at 5, 10, 15 g m−2 yr−1 (N5, N10, N15, P5, P10, P15, N5P5, N10P10, and N15P15) compared to unfertilized control on soil C, soil microbial biomass and functional diversity at the 0–20 cm and 20–40 cm depth in an alpine meadow after 5 years of continuous fertilization. Fertilization increased total aboveground biomass of community and grass but decreased legume and forb biomass compared to no fertilization. All fertilization treatments decreased the C:N ratios of legumes and roots compared to control, however fertilization at rates of 5 and 15 g m−2 yr−1 decreased the C:N ratios of the grasses. Compared to the control, soil microbial biomass C increased in N5, N10, P5, and P10 in 0–20 cm, and increased in N10 and P5 while decreased in other treatments in 20–40 cm. Most of the fertilization treatments decreased the respiratory quotient (qCO2) in 0–20 cm but increased qCO2 in 20–40 cm. Fertilization increased soil microbial functional diversity (except N15) but decreased cumulative C mineralization (except in N15 in 0–20 cm and N5 in 20–40 cm). Soil organic C (SOC) decreased in P5 and P15 in 0–20 cm and for most of the fertilization treatments (except N15P15) in 20–40 cm. Overall, these results suggested that soils will not be a C sink (except N15P15). Nitrogen and phosphorus fertilization may lower the SOC pool by altering the plant biomass composition, especially the C:N ratios of different plant functional groups, and modifying C substrate utilization patterns of soil microbial communities. Background: Although the effect of water level on germination in soil seed banks has been documented in many ecosystems, the mechanism is not fully understood, and to date no empirical studies on this subject exist. Further, no work has been done on the effect of water level on seed banks of drying and saline-alkaline wetlands in alpine areas on the Tibetan Plateau. Methodology: We examined the effects of water level (0 cm, 5 cm and 10 cm) on seed germination and seedling establishment from soil seed banks at 0–5 cm and 5–10 cm depths in typical, drying, and saline-alkaline wetlands. We also explore the potential role of soil seed bank in restoration of drying and saline-alkaline wetlands. Principal Findings: Species richness decreased with increase in water level, but there almost no change in seed density. A huge difference exists in species composition of the seed bank among different water levels in all three wetlands, especially between 0 cm and 5 cm and 0 cm and 10 cm. Similarity of species composition between seed bank and plant community was higher in 0 cm water level in drying wetland than in the other two wetlands. The similarity was much higher in 0 cm water level than in 5 cm and 10 cm water levels in all three wetlands. Species composition of the alpine wetland plant community changed significantly after drying and salinization, however, species composition of the seed bank was unchanged regardless of the environment change. Conclusions/Significance: Water level greatly affects seed bank recruitment and plant community establishment. Further, different water levels in restored habitats are likely to determine its species composition of the plant community. The seed bank is important in restoration of degraded wetlands.
