Increasing urbanization and consequent habitat fragmentation and loss call for studies of the influence of urbanization on native fauna. South American Coati (Procyonidae: Nasua nasua) live in groups and are common in urban areas of South America. Here, we aimed to understand how coatis occupy and select space, and whether they change their circadian activity in an urban environment. We estimated home range sizes, verified any overlap, and evaluated the effects of sex and weight on home range size. We also described coati activity patterns associated with habitat selection. Animals established their home ranges only within the study area, with few exploratory exits to surrounding areas. Furthermore, our home range estimates of 0.66 km2 are lower than previously found in natural habitats. We did not observe an effect of sex or weight on home range size. Coatis predominantly selected forested areas and explored houses only early in the morning. We suggest that this demonstrates the need for forested areas for survival of the species. Regarding activity patterns, coatis were active only during the day, as described for the species in its natural environment. Human barriers—in addition to human refuse that serves as food—could reduce the home range sizes. Our data demonstrate differences in home range size, but not circadian activity or habitat selection when compared to coatis in their natural environment. [ABSTRACT FROM AUTHOR]

The article presents a study which identified clusters of anti-Ku-positive patients according to their spectrum of antinuclear antibodies (ANA) specificities and analyzed their clinical and analytical features. Topics discussed include features identified from the patients' clinical charts, autoantibody clusters identified in the analysis, and limitations of the study.

Motivation: Our society has become data-rich to the extent that research in many areas has become impossible without computational approaches. Educational programmes seem to be lagging behind this development. At the same time, there is a growing need not only for strong data science skills, but foremost for the ability to both translate between tools and methods on the one hand, and application and problems on the other. Results: Here we present our experiences with shaping and running a masters' programme in bioinformatics and systems biology in Amsterdam. From this, we have developed a comprehensive philosophy on how translation in training may be achieved in a dynamic and multidisciplinary research area, which is described here. We furthermore describe two requirements that enable translation, which we have found to be crucial: sufficient depth and focus on multidisciplinary topic areas, coupled with a balanced breadth from adjacent disciplines. Finally, we present concrete suggestions on how this may be implemented in practice, which may be relevant for the effectiveness of life science and data science curricula in general, and of particular interest to those who are in the process of setting up such curricula. Experimental evolution of microbes has gained lots of interest in recent years, mainly due to the ease of strain characterisation through next-generation sequencing. While evolutionary and systems biologists use experimental evolution to address fundamental questions in their respective fields, studies with lactic acid bacteria are often more directed by applied questions. Insight into population and genome dynamics are valuable for experimental design and data interpretation, and it is becoming increasingly apparent how different constraints limit and govern the outcome of microbial adaptation to a selective environment. Examples for such constraints are the finite membrane and cellular space which can lead to trade-offs between cellular strategies. A powerful perspective is that of resource allocation, which allows cells to maximise fitness. This impacts on metabolic strategies that have different protein/resource demands. This review focuses on parameters and forces that shape cellular optimisation processes and that are determining for the outcome of laboratory evolution experiments. Phenotypic changes of experimentally evolved lactic acid bacteria will be discussed in the light of the selection conditions and the prevailing constraints. [ABSTRACT FROM AUTHOR]

An effective long-term cell therapy for skeletal muscle regeneration requires donor contribution to both muscle fibers and the muscle stem cell pool. Although satellite cells have these abilities, their therapeutic potential so far has been limited due to their scarcity in adult muscle. Myogenic progenitors obtained from Pax3-engineered mouse embryonic stem (ES) cells have the ability to generate myofibers and to improve the contractility of transplanted muscles in vivo, however, whether these cells contribute to the muscle stem cell pool and are able to self-renew in vivo are still unknown. Here, we addressed this question by investigating the ability of Pax3, which plays a critical role in embryonic muscle formation, and Pax7, which is important for maintenance of the muscle satellite cell pool, to promote the derivation of self-renewing functional myogenic progenitors from ES cells. We show that Pax7, like Pax3, can drive the expansion of an ES-derived myogenic progenitor with significant muscle regenerative potential. We further demonstrate that a fraction of transplanted cells remains mononuclear, and displays key features of skeletal muscle stem cells, including satellite cell localization, response to reinjury, and contribution to muscle regeneration in secondary transplantation assays. The ability to engraft, self-renew, and respond to injury provide foundation for the future therapeutic application of ES-derived myogenic progenitors in muscle disorders.
