Rosalia funebris (RFUNE; Cerambycidae), the banded alder borer, is a longhorn beetle whose larvae feed on the wood of various economically and ecologically significant trees in western North America. Adults are short-lived and not known to consume plant material substantially. We sequenced, assembled, and annotated the RFUNE genome using HiFi and RNASeq data. We documented genome architecture and gene content, focusing on genes putatively involved in plant feeding (phytophagy). Comparisons were made to the well-studied genome of the Asian longhorned beetle (AGLAB; Anoplophora glabripennis) and other Cerambycidae. The 814 Mb RFUNE genome assembly was distributed across 42 contigs, with an N50 of 30.18 Mb. Repetitive sequences comprised 60.27% of the genome, and 99.0% of expected single-copy orthologous genes were fully assembled. We identified 12,657 genes, fewer than in the four other species studied, and 46.4% fewer than for Aromia moschata (same subfamily as RFUNE). Of the 7,258 orthogroups shared between RFUNE and AGLAB, 1,461 had more copies in AGLAB and 1,023 had more copies in RFUNE. We identified 240 genes in RFUNE that putatively arose via horizontal transfer events. The RFUNE genome encoded substantially fewer putative plant cell wall degrading enzymes than AGLAB, which may relate to the longer-lived plant-feeding adults of the latter species. The RFUNE genome provides new insights into cerambycid genome architecture and gene content and provides a new vantage point from which to study the evolution and genomic basis of phytophagy in beetles. Bt technologies have played a major role in the control of bollworm, Helicoverpa zea (Boddie) (Lepidoptera: Noctuidae), in cotton.Variation in expression levels among varieties and plant parts, along with selection pressure on bollworm populations, has led to the development of resistance to some Bt proteins. Trials were conducted to evaluate how cotton varieties expressing different Bt proteins affect bollworm larval behavior and their damage in flowering cotton. Differences in larval recovery were observed among cotton varieties at 3 d with 3-gene Bt cotton having the lowest recovery and non-Bt cotton having the greatest recovery. Loss of bloom tags and abscission of small bolls at the site of infestation affected bollworm larval recovery among varieties. Day after infestation was the main factor that affected bollworm movement across all varieties. Number of total damaged fruiting forms by an individual bollworm larva was different among all varieties. Overall, flower bud (square) and fruit (boll) damage by an individual larva was lower on 3-gene cotton than 2-gene cotton and non-Bt cotton. An individual larva damaged fewer squares on 2-gene cotton than non-Bt cotton, but boll damage from bollworm was similar among 2-gene cotton and non-Bt cotton. The level of square and boll damage in 2-gene cotton has increased compared to previous research further supporting the occurrence of bollworm resistance to Cry proteins. The 3-gene cotton containing the Vip3A gene experienced low levels of damage and survival. The ability to visualize cell and tissue morphology at a high magnification using scanning electron microscopy (SEM) has revolutionized plant sciences research. In plant–insect interactions studies, SEM-based imaging has been of immense assistance to understand plant surface morphology including trichomes [plant hairs; physical defense structures against herbivores], spines, waxes, and insect morphological characteristics such as mouth parts, antennae, and legs, that they interact with. While SEM provides finer details of samples, and the imaging process is simpler now with advanced image acquisition and processing, sample preparation methodology has lagged. The need to undergo elaborate sample preparation with cryogenic freezing, multiple alcohol washes, and sputter coating makes SEM imaging expensive, time consuming, and warrants skilled professionals, making it inaccessible to majority of scientists. Here, using a desktop version of SEM (SNE- 4500 Plus Tabletop), we show that the "plug and play" method can efficiently produce SEM images with sufficient details for most morphological studies in plant–insect interactions. We used leaf trichomes of Solanum genus as our primary model, and oviposition by tobacco hornworm (Manduca sexta ; Lepidoptera: Sphingidae) and fall armyworm (Spodoptera frugiperda ; Lepidoptera: Noctuidae), and leaf surface wax imaging as additional examples to show the effectiveness of this instrument and present a detailed methodology to produce the best results with this instrument. While traditional sample preparation can still produce better resolved images with less distortion, we show that even at a higher magnification, the desktop SEM can deliver quality images. Overall, this study provides detailed methodology with a simpler "no sample preparation" technique for scanning fresh biological samples without the use of any additional chemicals and machinery. Swede midge (Contarinia nasturtii Kieffer) is an invasive, economic pest of canola (Brassica napus L.) that threatens production throughout Canada. Swede midge has up to four overlapping generations, placing canola at risk of multiple infestations in the field. The relationship between single and multiple swede midge infestations at different canola stages, and the resulting impacts on yield, are unknown. Laboratory experiments were conducted to evaluate the impact of single and multiple infestations of swede midge on four plant stages of canola: three-leaf, seven-leaf, primary bud, and secondary bud. Previously, the seven-leaf and primary bud stages were considered vulnerable to swede midge, but we determined that the secondary bud stage is also vulnerable. Evidence of compensation by canola in response to herbivory by swede midge was discovered. Compensation occurred mainly through increased production of tertiary racemes and pods and was maximized with exposure of 4.5–6.5 female midges per plant. Although compensation may increase potential yield, it also results in uneven crop maturation resulting in delayed harvest and yield loss. Consequently, to prevent delays in maturation and harvest, insecticides should be applied before these densities are reached. [ABSTRACT FROM AUTHOR]

Detection of host plant DNA from sap-feeding insects can be challenging due to potential low concentration of ingested plant DNA. Although a few previous studies have demonstrated the possibility of detecting various fragments of plant DNA from some sap-feeders, there are no protocols available for potato leafhopper, Empoasca fabae (Harris) (Hemiptera: Cicadellidae), a significant agricultural pest. In this study we focused on optimizing a DNA-based method for host plant identification of E. fabae and investigating the longevity of the ingested plant DNA as one of the potential applications of the protocol. We largely utilized and modified our previously developed PCR-based method for detecting host plant DNA from grasshopper and the spotted lanternfly gut contents. We have demonstrated that the trn L (UAA) gene can be successfully utilized for detecting ingested host plant DNA from E. fabae and determining plant DNA longevity. The developed protocol is a relatively quick and low-cost method for detecting plant DNA from E. fabae. Chemical ecology is the highly interdisciplinary study of biochemicals that mediate the behavior of organisms and the regulation of physiological changes that alter intraspecific and/or interspecific interactions. Significant advances are often achieved through the collaboration of chemists and biologists working to understand organismal survival strategies with an eye on the development of targeted technologies for controlling agricultural, forestry, medical, and veterinary pests in a sustainable world. Plants play a pivotal role in interactions involving herbivores and their natural enemies. Variation in plant primary and secondary metabolites not only affects herbivores but, directly and indirectly, also their natural enemies. Here, we used a commercial NPK fertilizer to test the impact of three fertilizer, namely 50, 100, and 200 ppm nitrogen, and one control (i.e. water) treatments, on the weight of the nymphs of the whitefly Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae). Subsequently, the whitefly parasitoid Eretmocerus mundus Mercet (Hymenoptera: Aphelinidae) was reared on the different groups of whitefly nymphs and upon parasitoid emergence, the number of oocytes was determined as a measure of reproductive capacity. Trials were done on tomato and tobacco plants. The level of nitrogen concentration in tobacco leaves was directly correlated with the fertilizer applications, thus confirming the effect of our fertilizer treatments. Both in tomato and tobacco plants, healthy as well as parasitized whitefly nymphs, were heaviest in the 200 ppm nitrogen treatment. The highest number of oocytes per female parasitoid was recorded in the 200 ppm nitrogen treatment in tomato (31% more oocytes as compared with the control) and in the 100 and 200 ppm nitrogen treatments in tobacco (200% more oocytes). We suggest that the increase in oocytes was the result of the enhanced size (food quantity) and/or nutritional quality of the whitefly host. Temperature has a strong influence on the development, survival, and fecundity of herbivorous arthropods, and it plays a key role in regulating the growth and development of their host plants. In addition, temperature affects the production of plant secondary chemicals as well as structural characteristics used for defense against herbivores. Thus, temperature has potentially important implications for host plant resistance. Because temperature directly impacts arthropod pests, both positively and negatively, distinguishing direct effects from indirect effects mediated through host plants poses a challenge for researchers and practitioners. A more comprehensive understanding of how temperature affects plant resistance specifically, and arthropod pests in general, would lead to better predictions of pest populations, and more effective use of plant resistance as a management tactic. Therefore, the goals of this paper are to 1) review and update knowledge about temperature effects on plant resistance, 2) evaluate alternative experimental approaches for separating direct from plant-mediated indirect effects of temperature on pests, including benefits and limitations of each approach, and 3) offer recommendations for future research. The insect order Hymenoptera originated during the Permian nearly 300 Mya. Ancestrally herbivorous hymenopteran lineages today make up the paraphyletic suborder 'Symphyta', which encompasses c. 8200 species with very diverse host-plant associations. We use phylogeny-based statistical analyses to explore the drivers of diversity dynamics within the 'Symphyta', with a particular focus on the hypothesis that diversification of herbivorous insects has been driven by the explosive radiation of angiosperms during and after the Cretaceous. Our ancestral-state estimates reveal that the first symphytans fed on gymnosperms, and that shifts onto angiosperms and pteridophytes – and back – have occurred at different time intervals in different groups. Trait-dependent analyses indicate that average net diversification rates do not differ between symphytan lineages feeding on angiosperms, gymnosperms or pteridophytes, but trait-independent models show that the highest diversification rates are found in a few angiosperm-feeding lineages that may have been favoured by the radiations of their host taxa during the Cenozoic. Intriguingly, lineages-through-time plots show signs of an early Cretaceous mass extinction, with a recovery starting first in angiosperm-associated clades. Hence, the oft-invoked assumption of herbivore diversification driven by the rise of flowering plants may overlook a Cretaceous global turnover in insect herbivore communities during the rapid displacement of gymnosperm- and pteridophyte-dominated floras by angiosperms. Widely distributed polyphagous insects provide opportunities to understand the influence of local plant communities on geographical variation in patterns of host plant use. Larval performance in the widespread butterfly Acraea terpsicore appears to vary geographically. This prompted us to investigate in detail the host plant-associated divergences in behaviour and life-history traits between two Indian populations of this species. Given that the two populations differ in abundances of the host plants, we tested whether local availability of host plants has influenced the evolution of larval performance and female oviposition preference. We also investigated whether females prefer to oviposit on host plants that are optimal for larval development. We found that host plants influence larval performance in a similar manner for both populations. Female preference is not correlated with larval performance; however, the preference varied between the populations. In one population, the females preferred a locally abundant host plant for oviposition although the plant was suboptimal for larval development. The negative correlation between preference and performance correlation suggests that the high cost of searching for the optimal host plant has influenced female preference. [ABSTRACT FROM AUTHOR]

Microbes are known to influence insect-plant interactions; however, it is unclear if host-plant diet influences the regulation of nutritional insect symbioses. The pea aphid, Acyrthosiphon pisum, requires its nutritional endosymbiont, Buchnera, for the production of essential amino acids. We hypothesize that key aphid genes that regulate the nutritional symbioses respond to host-plant diet when aphids feed on a specialized (alfalfa) compared to a universal host-plant diet (fava), which vary in amino acid profiles. Using RNA-Seq and whole genome bisulfite sequencing, we measured gene expression and DNA methylation profiles for such genes when aphids fed on either their specialized or universal host-plant diets. Our results reveal that when aphids feed on their specialized host-plant they significantly up-regulate and/or hypo-methylate key aphid genes in bacteriocytes related to the amino acid metabolism, including glutamine synthetase in the GOGAT cycle that recycles ammonia into glutamine and the glutamine transporter ApGLNT1. Moreover, regardless of what host-plant aphids feed on we observed significant up-regulation and differential methylation of key genes involved in the amino acid metabolism and the glycine/serine metabolism, a metabolic program observed in proliferating cancer cells potentially to combat oxidative stress. Based on our results, we suggest that this regulatory response of key symbiosis genes in bacteriocytes allows aphids to feed on a suboptimal host-plant that they specialize on. At least half of metazoan species are herbivorous insects. Why are they so diverse? Most herbivorous insects feed on few plant species, and adaptive host specialization is often invoked to explain their diversification. Nevertheless, it is possible that the narrow host ranges of many herbivorous insects are nonadaptive. Here, we test predictions of this hypothesis with comparative phylogenetic analyses of scale insects, a group for which there appear to be few host-use trade-offs that would select against polyphagy, and for which passive wind-dispersal should make host specificity costly. We infer a strong positive relationship between host range and diversification rate, and a marked asymmetry in cladogenetic changes in diet breadth. These results are consonant with a system of pervasive nonadaptive host specialization in which small, drift- and extinction-prone populations are frequently isolated from persistent and polyphagous source populations. They also contrast with the negative relationship between diet breadth and taxonomic diversification that has been estimated in butterflies, a disparity that likely stems from differences in the average costs and benefits of host specificity and generalism in scale insects versus butterflies. Volatile phytochemicals play a role in orientation by phytophagous insects. We studied antennal and behavioral responses of the Asian citrus psyllid, Diaphorina citri, vector of the citrus greening disease pathogen. Little or no response to citrus leaf volatiles was detected by electroantennography. Glass cartridges prepared with β-ocimene or citral produced no response initially but became stimulatory after several days. Both compounds degraded completely in air to a number of smaller molecules. Two peaks elicited large antennal responses and were identified as acetic and formic acids. Probing by D. citri of a wax substrate containing odorants was significantly increased by a blend of formic and acetic acids compared with either compound separately or blends containing β-ocimene and/or citral. Response surface modeling based on a 4-component mixture design and a 2-component mixture-amount design predicted an optimal probing response on wax substrate containing a blend of formic and acetic acids. Our study suggests that formic and acetic acids play a role in host selection by D. citri and perhaps by phytophagous insects in general even when parent compounds from which they are derived are not active. These results have implications for the investigation of arthropod olfaction and may lead to elaboration of attract-and-kill formulations to reduce nontarget effects of chemical control in agriculture. Ecological specialization is widely recognized as a major determinant of the emergence and maintenance of biodiversity. We studied two critical facets of specialization - local adaptation and habitat choice - in the host races of the leaf beetle Lochmaea capreae on willow and birch. Our results revealed that there is asymmetric disruptive selection for host use traits, and host races achieved different adaptive sets of life history traits through association with their host plant. Beetles from each host race exhibited food and oviposition preference for their own host plant. Reciprocal transplant displayed significant variation in host acceptance and performance: all families from the willow race rejected the alternative host plant before initiation of feeding and all died on this host plant. By contrast, all families from the birch race accepted willow for feeding, but they consumed less and performed less well. Intriguingly, families that performed well on birch also performed well on willow, suggesting positive genetic correlation rather than genetic trade-offs. Our results suggest that the major proximal determinant of host specialization in the willow race is the behavioural acceptance of a plant rather than the toxicity of the food resource. However, in the birch race a combination of behavioural host acceptance and performance may play a role in specialization. In an environment with changing availability and quality of host plants, phytophagous insects are under selection pressure to find quality hosts. They need to maximize their fitness by locating suitable plants and avoiding unsuitable ones. Thus, they have evolved a finely tuned sensory system, for detection of host cues, and a nervous system, capable of integrating inputs from sensory neurons with a high level of spatio-temporal resolution. Insect responses to cues are not fixed but depend on the context in which they are perceived, the physiological state of the insect, and prior learning experiences. However, there are examples of insects making 'mistakes' and being attracted to poor quality hosts. While insects have evolved ways of finding hosts, plants have been under selection pressure to do precisely the opposite and evade detection or defend themselves when attacked. Once on the plant, insect-associated molecules may trigger or suppress defence depending on whether the plant or the insect is ahead in evolutionary terms. Plant volatile emission is influenced by defence responses induced by insect feeding or oviposition which can attract natural enemies but repel herbivores. Conversely, plant reproductive fitness is increased by attraction of pollinators. Interactions can be altered by other organisms associated with the plant such as other insects, plant pathogens, or mycorrhizal fungi. Plant phenotype is plastic and can be changed by epigenetic factors in adaptation to periods of biotic stress. Linking adaptive divergence to hybrid unfitness is necessary to understand the ecological factors contributing to reproductive isolation and speciation. To date, this link has been demonstrated in few model systems, most of which encompass ecotypes that occupy relatively early stages in the speciation process. Here we extend these studies by assessing how host-plant adaptation conditions hybrid fitness in the pea aphid, Acyrthosiphon pisum. We made crosses between and within five pea aphid biotypes adapted to different host plants and representing various stages of divergence within the complex. Performance of F1 hybrids and nonhybrids was assessed on a 'universal' host that is favorable to all pea aphid biotypes in laboratory conditions. Although hybrids performed equally well as nonhybrids on the universal host, their performance was much lower than nonhybrids on the natural hosts of their parental populations. Hence, hybrids, rather than being intrinsically deficient, are maladapted to their parents' hosts. Interestingly, the impact of this maladaptation was stronger in certain hybrids from crosses involving the most divergent biotype, suggesting that host-dependent postzygotic isolation has continued to evolve late in divergence. Understanding the genetic bases of biological diversification is a long-standing goal in evolutionary biology. Here, we investigate whether replicated cases of adaptive divergence involve the same genomic regions in the pea aphid, Acyrthosiphon pisum, a large complex of genetically differentiated biotypes, each specialized on different species of legumes. A previous study identified genomic regions putatively involved in host-plant adaptation and/or reproductive isolation by performing a hierarchical genome scan in three biotypes. This led to the identification of 11 FST outliers among 390 polymorphic microsatellite markers. In this study, the outlier status of these 11 loci was assessed in eight biotypes specialized on other host plants. Four of the 11 previously identified outliers showed greater genetic differentiation among these additional biotypes than expected under the null hypothesis of neutral evolution ( α < 0.01). To protect against loss of photo-assimilate-rich phloem sap, plants have evolved several mechanisms to plug phloem sieve tubes in response to damage. In many Fabaceae, each sieve element contains a discrete proteinaceous body called a forisome, which, in response to damage, rapidly transforms from a condensed configuration that does not impede the flow of sap to a dispersed configuration that plugs the sieve element. Aphids and other specialized phloem sap feeders can ingest phloem sap from a single sieve element for hours or days, and to do this, they must be able to suppress or reverse phloem plugging. A recent study provided in vitro evidence that aphid saliva can reverse forisome plugs. The present study tested this hypothesis in vivo by inducing forisome plugs which triggered aphids to switch behaviour from phloem sap ingestion to salivation into the sieve element. After salivating into the sieve element for various periods of time, the aphids were instantaneously cryofixed (freeze fixed) in situ on their leaf. The state of the forisome was then determined in the penetrated sieve element and in nearby non-penetrated sieve elements which served as controls for sieve elements not subjected to direct aphid salivation. Forisomes were almost always in close contact with the stylet tips and thus came into direct contact with the saliva. Nonetheless, forisome plugs in the penetrated sieve element did not revert back to a non-plugging state any faster than those in neighbouring sieve elements that were not subjected to direct aphid salivation. Background and Aims Trichomes are epidermal outgrowths generally associated with protection against herbivores and/or desiccation that are widely distributed from ferns to angiosperms. Patterns of topological variation and morphological evolution of trichomes are still scarce in the literature, preventing valid comparisons across taxa. This study integrates detailed morphoanatomical data and the evolutionary history of the tribe Bignonieae (Bignoniaceae) in order to gain a better understanding of current diversity and evolution of trichome types. Methods Two sampling schemes were used to characterize trichome types: (1) macromorphological characterization of all 105 species currently included in Bignonieae; and (2) micromorphological characterization of 16 selected species. Individual trichome morphotypes were coded as binary in each vegetative plant part, and trichome density and size were coded as multistate. Ancestral character state reconstructions were conducted using maximum likelihood (ML) assumptions. Key Results Two main functional trichome categories were found: non-glandular and glandular. In glandular trichomes, three morphotypes were recognized: peltate (Pg), stipitate (Sg) and patelliform/cupular (P/Cg) trichomes. Non-glandular trichomes were uniseriate, uni- or multicellular and simple or branched. Pg and P/Cg trichomes were multicellular and non-vascularized with three clearly distinct cell layers. Sg trichomes were multicellular, uniseriate and long-stalked. ML ancestral character state reconstructions suggested that the most recent common ancestor (MRCA) of Bignonieae probably had non-glandular, Pg and P/Cg trichomes, with each trichome type presenting alternative histories of appearance on the different plant parts. For example, the MRCA of Bignonieae probably had non-glandular trichomes on the stems, prophylls, petiole, petiolule and leaflet veins while P/Cg trichomes were restricted to leaflet blades. Sg trichomes were not present in the MRCA of Bignonieae independently of the position of these trichomes. These trichomes had at least eight independent origins in tribe. Conclusions The patterns of trichome evolution indicate that most morphotypes are probably homologous in Bignonieae and could be treated under the same name based on its morphological similarity and common evolutionary history, in spite of the plethora of names that have been previously designated in the literature. The trichome descriptions presented here will facilitate comparisons across taxa, allowing inferences on the relationsthips between trichome variants and future studies about their functional properties. Many studies have reported the effect of nitrogen (N) fertilization on plant constitutive defense responses; however, little is known about their effects on plant induced defense patterns and its consequence for insect herbivores. In our experiments, the effects of N availability on growth, nutritional quality (N content, protein/carbohydrate [P:C] ratio, modified gross energy [MGE] ), and constitutive phenolics of corn, Zea mays L. were quantified. Moreover, the indirect effects of N fertilization on the beet armyworm, Spodoptera exigua Hiibner through larval performance and feeding preference were examined. N fertilization increased plant growth, and depressed defense traits by increasing N content and the P:C ratio, as well as decreasing the constitutive concentration of phenolics. Subsequently, beet arnwworm showed higher performance and preferentially fed on high-N corn because of its low defense traits. After beet armyworm feeding, high-N corn significantly deterred larval feeding, and had negative effects on the performance of beet armyworm through decreasing P:C ratio and increasing induced pheno]ies. On the contrary, there were no significant changes in P:C ratio and phenolics in low-N corn after feeding damage. Larval performance and preference were also not afffected by induced compounds in low-N corn, which suggested that the expression of induced defense was dependent on N availability. The result indicates that N availability can exert a variety of bottom-up effect on plant defense patterns to influence insect population dynamics, and thereby may represent a source of variation in plant-insect interactions. Endophytic fungi, which live within host plant tissues without causing any visible symptom of disease, are important mediators of plant-herbivore interactions. These endophytes enhance resistance of host plant against insect herbivores mainly by productions of various alkaloid based defensive compounds in the plant tissue or through alterations of plant nutritional quality. Two endophytic fungi, i.e., Nigrospora sp. and Cladosporium sp., were isolated from Tinospora cordifolia (Thunb.) Miers, a traditional indian medicinal plant. Cauliflower (Brassica oleracea L.) plants were inoculated with these two endophytic fungi. The effect of endophyte infected and uninfected cauliflower plants were measured on the survival and development of Spodoptera litura (Fab.), a polyphagous pest. Endophyte infected cauliflower plants showed resistance to S. litura in the form of significant increase in larval and pupal mortality in both the fungi. Inhibitory effects of endophytic fungi also were observed on adult emergence, longevity, reproductive potential, as well as hatchability of eggs. Thus, it is concluded that antibiosis to S. litura could be imparted by artificial inoculation of endophytes and this could be used to develop alternative ecologically safe control strategies. For animals that lay eggs, a longstanding question is, why do females choose particular opposition sites? For insects that lay eggs on plants there are three hypotheses: maximizing suitable habitat for juveniles, maximizing female lifespan, and maximizing egg survival. We investigated the function of the oviposition-site choice behavior of a gall midge, the Hessian fly, Mayetiola destructor (Say). In spite of living less than a day and having hundreds of eggs, the ovipositing female is choosy about the placement of eggs. Choosiness makes sense. The tiny gall-making neonate larva has limited movement and strict requirements for colonization. We examined whether offspring benefit from the Hessian fly female's preference for the plant's youngest leaf. To do this we restricted the female's access to the first, second, or third leaf of a seedling (wheat Triticum aestivum L.) plant. Being placed on older leaves did not impact egg survival or larval survival during migration to attack sites at the base of the plant, but did have negative impacts on egg-to-adult survival (reduced by 48%) and reproductive potential (reduced by 30-45%). These negative impacts appear to come from larvae having to search harder to find the limited number of reactive plant cells that can be reprogrammed to form the gall nutritive tissue. We propose that the ability of larvae to find these reactive cells in spite of being placed on an older leaf is important because it creates leeway for female behavior to evolve in the face of other selection pressures, e.g., attack by egg parasitoids. Establishment of the saltcedar leaf beetle (Diorhabda spp.) has been unpredictable when caged or released in the field for saltcedar (Tamarix spp.) biocontrol. It has been observed that one caged tree might be voraciously fed upon by beetles while an adjacent tree in the cage is left untouched. We hypothesized that differences in the nutrient content of individual trees may explain this behavior. We evaluated survival, development rate, and egg production of beetles fed in the laboratory on saltcedar foliage from trees that had been grown under a range of fertilizer treatments. Tissue samples from the experimental trees and from the field were analyzed for percent nitrogen, phosphorus, and potassium. There was essentially no survival of beetle larvae fed foliage from saltcedar trees at nitrogen levels below 2.0%. At levels above 2.0% N, beetle larvae had corresponding increased survival rates and shorter development times. Multiple regression analyses indicated that nitrogen and phosphorus are important for larval survival and faster development rates. Higher levels of potassium were important for increased egg cluster production. The plant tissue analysis showed that the percentage of nitrogen in the experimental trees reflected the range of trees in the field and also that there is high variability within trees in the field. The emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), is an exotic woodborer first detected in 2002 in Michigan and Ontario and is threatening the ash resource in North America. We examined the effects of light exposure and girdling on green ash (Fraxinus pennsyluanica Marsh) volatile production, and effects of light exposure, girdling, and leaf age on emerald ash borer adult feeding preferences and phototaxis. Green ash seedlings grown under higher light exposure had lower amounts of three individual volatile compounds, (Z)-3-hexenol, (E)-β- ocimene, and (Z,E)-α-farnesene, as well as the total amount of six detected volatile compounds. Girdling did not affect the levels of these volatiles. Emerald ash borer females preferred mature leaves, leaves from girdled trees, and leaves grown in the sun over young leaves, leaves from nongirdled trees, and leaves grown in the shade, respectively. These emerald ash borer preferences were most likely because of physical, nutritional, or biochemical changes in leaves in response to the different treatments. Emerald ash borer females and males showed positive phototaxis in laboratory arenas, a response consistent with emerald ash borer preference for host trees growing in sunlight. Insect herbivores typically oviposit on the most suitable hosts, but choices can be modulated by detection of potential competition among conspecifics, especially when eggs are deposited cryptically. Larvae of the wheat stem sawfly, Cephus cinctus Norton, developing within an already infested stem, experience elevated risk when only one will survive because of cannibalism. To increase our understanding of host selection when the choices made by females can lead to severe intraspecific competition, females were presented with either uninfested wheat plants or with plants previously exposed to other females in laboratory choice tests. The oviposition behavior of this insect was described by recording the behavioral sequences that lead to and follow the insertion of the ovipositor in both previously infested and uninfested stems. No significant differences were found in frequencies of specific behaviors or behavioral transitions associated with oviposition. In choice tests, there was no difference in the numbers of eggs laid in infested and uninfested plants. Taller plants received more eggs, irrespective of infestation. Females neither preferred nor avoided previously infested hosts. Other characteristics of the host, such as stem height, may be more important in determining suitability for oviposition. These findings support the use of management tactics relying on the manipulation of oviposition behavior, such as trap cropping. Given that there is no evidence for response to previously infested hosts, the infested plants in a trap crop would remain as suitable as they were when uninfested, which could also lead to an increase in mortality caused by intraspecific competition. The effect of taxa [common Bermuda grass, Cynodon dactylon (L.); centipedegrass, Eremochloa ophiuroides Munro Hack; St. Augustinegrass, Stenotaphrum secundatuni [Walt.] Kuntze; and zoysiagrass, Zoysia spp.], density, height, and weed density on abundance of natural enemies, and their potential prey were evaluated in residential turf. Total predatory Heteroptera were most abundant in St. Augustinegrass and zoysiagrass and included Anthocoridae, Lasiochilidae, Geocoridae, and Miridae. Anthocoridae and Lasiochilidae were most common in St. Augustinegrass, and their abundance correlated positively with species of Blissidae and Delphacidae. Chinch bugs were present in all turf taxa, but were 23-47 times more abundant in St. Augustinegrass. Anthocorids/lasiochilids were more numerous on taller grasses, as were Blissidae, Delphacidae, Cicadellidae, and Cercopidae. Geocoridae and Miridae were most common in zoysiagrass arid were collected in higher numbers with increasing weed density. However, no predatory Heteroptera were affected by grass density. Other beneficial insects such as staphylinids and parasitic Hymerioptera were captured most often in St. Augustinegrass and zoysiagrass. These differences in abundance could be in response to primary or alternate prey, or reflect the influence of turf microenvironmental characteristics. In this study, Simpson's diversity index for predatory Heteroptera showed the greatest diversity and evenness in centipedegrass, whereas the herbivores and detritivores were most diverse in St. Augustinegrass lawns. These results demonstrate the complex role of plant taxa in structuring arthropod communities in turf. An increased understanding of how turf species and cultivars help shape pest and beneficial arthropod communities will enhance predictive abilities and further pest management objectives. A 2-yr study was conducted in a citrus orchard (Citrus sinensis L. Osbeck cultivar Valencia) to determine the influence of plant water stress on the population dynamics of glassy-winged sharpshooter, Hornalodisca vitripennis (Germar). Experimental treatments included irrigation at 100% of the crop evapotranspiration rate (ETc) and continuous deficit-irrigation regimens at 80 and 60% ETc. Microclimate and plant conditions monitored included temperature and humidity in the tree canopy, leaf surface temperature, water potential, and fruit quality and yield. Glassy-winged sharp-shooter population densities and activity were monitored weekly by a combination of visual inspections, beat net sampling, and trapping. Glassy-winged sharpshooter populations were negatively affected by severe plant water stress; however, population densities were not linearly related to decreasing water availability in plants. Citrus trees irrigated at 60% ETc had significantly warmer leaves, lower xylem water potential, and consequently hosted fewer glassy-winged sharpshooter eggs, nymphs, and adults than trees irrigated at 80% ETc. Citrus trees irrigated at 100% ETc hosted similar numbers of glassy-winged sharpshooter stages as trees irrigated at 60% ETc and a lower number of glassy-winged sharpshooter nymphs than the 80% ET, treatment, specifically during the nymphal density peak in mid-April to early July. Irrigation treatments did not affect populations of monitored natural enemies. Although the adult glassy-winged sharpshooter population was reduced, on average, by 50% in trees under severe water stress, the total number of fruit and number of fruit across several fruit grade categories were significantly lower in the 60% ETc than in the 80 and 100% ETc irrigation treatments. Holymenia clavigera Herbst and Anisosceis foliacea marginella Dallas (Hemiptera: Coreidae: Anisoscelini) use the same host plants (Passifloraceae), especially Passilfora suberosa L., and they have a very similar egg and nymphal morphology, This study compares the mouthparts of these coreids both intra- and interspecifically through scanning electron microscopy. Also, it determines the feeding sites for the fifth instars and for adults when offered some of their preferred structures (green fruit and mature leaves) of P. suberosa. Histological sections were performed on feeding sites, on plant pieces having the penetrated stylet in situ. Mouthpart morphologies were extremely similar between H. clavigera and A. foliacea marginella throughout ontogeny. The stylets reached the xylem, in almost all situations, followed by a low use of phloem. Nymphs and adults of both species consumed several fruit parts, including the seeds. When feeding upon the latter, endosperm and embryo were used for feeding. Feeding frequencies indicated that seeds are the main resource used by these coreids and that xylem feeding is probably related to water acquisition. Two models for assessing the performance of herbivores on a variety of host plants are developed by combining knowledge of population genetics and population ecology, especially that of the fixation probability of mutant genes. The absolute host performance model precisely assesses host performance for one herbivore population based on parameters of fecundity, larval survivorship, and selection pressure. The relative host performance model compares host performance for one population among different host plant species and for several populations on the same host species. Two herbivore populations, Bemisia tabaci (Gennadius) and Tetranychus truncates Ehara, were used to validate the absolute and relative host performance models. Results indicated that the assessment systems of host performance were reasonable and reliable. Speciation of plant-feeding insects is typically associated with host-plant shifts, with subsequent divergent selection and adaptation to the ecological conditions associated with the new plant. However, a few insect groups have apparently undergone speciation while remaining on the same host-plant species, and such radiations may provide novel insights into the causes of adaptive radiation. We used mitochondrial and nuclear DNA to infer a phylogeny for 14 species of gall-inducing Asphondylia flies (Diptera: Cecidomyiidae) found on Larrea tridentata (creosote bush), which have been considered to be monophyletic based on morphological evidence. Our phylogenetic analyses provide strong support for extensive within-host plant speciation in this group, and it demonstrates that diversification has involved numerous shifts between different plant organs (leaves, buds, flowers, and stems) of the same host-plant species. Within-plant speciation of Asphondylia is thus apparently facilitated by the opportunity to partition the plant ecologically. One clade exhibits temporal isolation among species, which may have facilitated divergence via allochronic shifts. Using a novel method based on Bayesian reconstruction, we show that the rate of change in an ecomorphological trait, ovipositor length, was significantly higher along branches with inferred shifts between host-plant organs than along branches without such shifts. This finding suggests that Larrea gall midges exhibit close morphological adaptation to specific host-plant parts, which may mediate ecological transitions via disruptive selection. Butterflies in the family Pieridae (Lepidoptera: Papilionoidea) feed as larvae on plants belonging primarily to three distantly related angiosperm orders: Fabales (legumes and allied plants), Brassicales (crucifers and related plants containing mustard oil glucosides), and Santalales (‘mistletoes’). However, some utilize plants from 13 other families in a further eight orders. We investigated the evolutionary history of host plant use of the Pieridae in the context of a recent phylogenetic hypothesis of the family, using simple character optimization. Although there is a close association between host plant and butterfly higher classification, we find no evidence for cospeciation but a pattern of repeated colonization and specialization. The ancestral host of the family appears to be Fabaceae or Fabales, with multiple independent shifts to other orders, including three to Santalales. The shift to Brassicales, which contain secondary compounds (glucosinolates), promoted diversification and adaptive radiation within the subfamily Pierinae. Subsequent shifts from crucifers to mistletoes (aerial-stem hemiparasites) facilitated further diversification, and more recent shifts from mistletoes to mistletoe host trees led to exploitation of novel host plants outside the conventional three orders. Possible mechanisms underlying these host shifts are briefly discussed. In the Pierinae, a striking association between host plant, larval and adult behaviour, adult phenotype, and mimicry calls for further research into possible relationships between host specialization, plant chemistry and butterfly palatability. Adaptive radiations consist of two intertwined processes, diversification of species and diversification of their ecological niches, but it is unclear whether there is a causal link between the processes. In phytophagous insects, ecological diversification mainly involves shifts in host-plant associations and in larval feeding habits (internal or external) on different plant parts, and several observations indicate that speciation is facilitated by host shifts. Data on host use in individual species suggest that internal feeders are less likely to colonize new hosts than external-feeding taxa and, consequently, increases in collective host ranges and species numbers should be slowed down in endophagous lineages. We tested these related hypotheses by using phylogenetic information to reconstruct the evolutionary history of larval resource use in the sawfly subfamily Nematinae. a group of 1000 plus species with a broad range of niches: the subfamily's combined host range includes over 20 plant families, and larvae may feed externally on leaves or needles, or internally, for example, in buds, fruits, leaves, or galls, The results show that: (I) Most internally feeding groups have evolved independently from external-feeding ancestors, but several distinct internal habits have appeared convergently multiple times; (2) Shifts among host taxa are clearly more common than changes in larval habits; (3) The majority of host switches have occurred among phylogenetically close plant groups, but many shifts are manifest among distantly related, ecologically proximate hosts; (4) Although external feeding characteristic of the common ancestor of Nematinae is associated with relatively high rates of host-shifting, internal feeders are very conservative in their host use; (5) In contrast, the effect of endophagy on speciation probabilities is more variable: net speciation rates are lowered in most internal-feeding groups, but a striking exception is found in species that induce galls on Salicaceae. The loose connection between collective host ranges and species diversity provides empirical support for theoretical models suggesting that speciation rates are a function of a complex interplay between "intrinsic" niche width and resource heterogeneity. Survival of nymphs and adult males and females of potato leafhopper, Empoasca fabae (Harris), on six potato (Solanum spp.) genotypes with varied levels of resistance was evaluated and related to type, density, and droplet/head size of foliar glandular trichomes. Survival was greater on Solarium tuberosum cultivars ('Allegany' and 'Elba') compared with accessions of the wild S. berthaultii (P1473331 and PT 473334). The barrier provided by glandular trichomes may affect survival of E. fabae by restricting nymph and adult feeding. PI 473331 was the most pubescent host, bearing type A and type B trichomes and the most unsuitable host for E. fabae. Allegany possesses leaflets with low trichome densities and small gland diameter and was the most suitable host for adult E. fabae survival. Females generally had higher survival rates than males. Nymphal and adult leathoppers differed greatly in their survival rates, particularly on S. tuberosum × S. berthaultii hybrids (Q174-2 and NY123). Interestingly, nymphs had longer (190 d) mean lethal time (LT50) than adults (31 d) when held on Q174-2, and a lower LT50 (19 d) when held on NY123 compared with adults (104 d). Starvation, dehydration, or both were likely associated with mortality of nymphs confined on PI 473331 and PI 473334. Nymphs on these accessions died at a comparable rate to those confined on a starvation treatment and more rapidly than those supplied with water only, sucrose solution or other potato genotypes. Darkened trichome exudates were observed on insects held on PI 473331 and PI 473334, consistent with previous reports that glandular trichomes of S. berthaultii contain phenolic oxidation chemistry. Field studies at sites with two contrasting soil types investigated effects from the presence of yellow foxtail [Setaria pumila (Poir.) Roem. and Sehult.], established in bands parallel to corn rows, on western corn rootworm (Diabrotica virgifera virgifera LeConte) survival, corn root injury, lodging, biomass production, and yield. Results suggested that the presence of foxtail as an alternate host influenced the degree and progression of corn rootworm damage and adult emergence in a given locality. Rootworm adults emerged later from foxtail band areas and had smaller head capsule size than did individuals from areas without foxtail, consistent with earlier findings that foxtail in the diet of western corn rootworm was a poor nutritional substitute for corn. Lodging was reduced in the presence of yellow foxtail in some cases, but corn stover biomass and yield also were lower. Influences, if any, of soil type on rootworm survival were unclear because of differences in planting date between the two sites. Foxtail may function as a buffer to reduce rootworm damage to corn and serves as an alternate host that should be considered in the development of resistance management strategies for transgenic corn modified for rootworm resistance. Vibrational communication is widespread in insect social and ecological interactions. Of the insect species that communicate using sound, water surface ripples, or substrate vibrations, we estimate that 92% use substrate vibrations alone or with other forms of mechanical signaling. Vibrational signals differ dramatically from airborne insect sounds, often having low frequencies, pure tones, and combinations of contrasting acoustic elements. Plants are the most widely used substrate for transmitting vibrational signals. Plant species can vary in their signal transmission properties, and thus host plant me may influence signal divergence. Vibrational communication occurs in a complex environment containing noise from wind and rain, the signals of multiple individuals and species, and vibration-sensitive predators and parasitoids. We anticipate that many new examples and functions of vibrational communication will be discovered, and that study of this modality will continue to provide important insights into insect social behavior, ecology, and evolution. Herbivorous insects and other small consumers are often specialized both in use of particular host taxa and in use of particular host tissues. Such consumers also often seem to show consistent differences in the rates of evolution of these two dimensions of host use, implying common processes, but this has been little studied. Here we quantify these rates of change in host use evolution in a major radiation of herbivorous insects, the Chrysomeloidea, whose diversity has been attributed to their use of flowering plants. We find a significant difference in the rates of evolutionary change in these two dimensions of host use, with host taxon associations most labile. There are apparently similar differences in rates of host use evolution in other parasite groups, suggesting the generality of this pattern. Molecular genetic analyses of populations showing phenotypic variation can provide important insights into the nature and limits of species. This is especially true when interpopulation variation is assessed in a phylogenetic context. The current study applies this approach to evaluate the evolutionary status of the leaf beetle Calligrapha multipunctata variety suturella, a taxon described from a single locality in New Hampshire, but not further discussed in the literature. Here, we report on the collection of material that greatly extends the geographic range of this taxon and on the analysis of cytochrome oxidase I mitochondrial DNA sequences from these specimens and related Calligrapha species. We find that sequences from geographically overlapping populations of C. m. suturella and C. m. bigsbyana form two distinct, phylogenetically divergent clades that exhibit appreciable differentiation (4.5%) at the nucleotide level. This mtDNA differentiation was also evident in each of three localities where both types were collected together on the same host plant. Our results indicate an absence of gene flow between C. m. suturella and C. m. bigsbyana and argue for the recognition of C. m. suturella as a distinct biological species. Accordingly, we elevate its status to C. suturella and provide a detailed diagnosis and description for this new species. Based on an absence of male specimens, a phenotypic similarity to C. m. bigsbyana, and the existence of other parthenogenetic Calligrapha, we speculate that C. suturella may be a parthenogenetic species of hybrid origin. Island archipelagos and insect-plant associations have both independently provided many useful systems for evolutionary study. The arytainine psyllid (Sternorrhyncha: Hemiptera) radiation on broom (Fabaceae: Genisteae) in the Canary Island archipelago provides a discrete system for examining the speciation of highly host-specific phytophagous insects in an island context. Phylogenetic reconstructions based on three datasets (adult and nymph morphological characters, and two mitochondrial DNA regions: part of the small subunit rRNA, and part of cytochrome oxidase I, cytochrome oxidase II and the intervening tRNA leucine) are generally consistent. The combined molecular tree provides a well-supported estimate of psyllid relationships and shows that there have been several colonizations of the Macaronesian islands but that only one has resulted in a significant radiation. Psyllid diversification has apparently been constrained by the presence of suitable host groups within the genistoid legumes, and the diversity, distribution, and abundance of those groups. The phylogeny, by indicating pairs of sister species, allows putative mechanisms of speciation to be assessed. The most common conditions associated with psyllid speciation are geographical allopatry with a host switch to closely related hosts (six examples), or geographical allopatry on the same host (four examples). Where allopatric speciation involves a host switch, these have all been to related hosts. There is some evidence that switches between unrelated host plants may be more likely in sympatry. Only one sister pair (Arytainilla cytisi and A. telonicola) and the putative host races of Arytinnis modica are sympatric but on unrelated hosts, which may be a necessary condition for sympatric speciation in these insects. Ambrosia beetles can be important pests of nursery production. The beetles are difficult to control with insecticides, requiring that pesticides be closely timed before tree attack, applied repeatedly, or have long residual activity. The goal of this project was to improve management decisions for ambrosia beetle control in nurseries. This study used ethanol-baited traps, field observations of tree attacks, and emergence cages over beetle galleries to determine the following: (1) composition of ambrosia beetle fauna in middle Tennessee, (2) species responsible for attacks on chestnut (Castanea mollissima Blume), a susceptible tree species, (3) timing of tree attacks and progeny emergence, and (4) the relationship between tree attacks, progeny emergence, and beetle collections in ethanol-baited traps. Ambrosia beetles were surveyed using ethanol-baited Lindgren traps at the Tennessee State University Nursery Crop Research Station in McMinnville, TN, and at two commercial nurseries near Dibrell and Tarlton, TN, during 1998 and 1999. At the Nursery Station, species composition of ambrosia beetles attacking chestnut trees was determined in 1999. Xyleborinus saxeseni Ratzeburg, Xylosandrus crassiusculus Motschulsky, and Monarthrum fasciatum Say were the dominant ambrosia beetle species collected in traps. Xyleborinus saxeseni was the dominant species at all three locations when both 1998 and 1999 collections were totaled. Other commonly trapped species included Monarthrum mali Fitch, Xyleborus atratus Eichhoff, and Xyleborus pelliculosus Eichhoff. Tree attacks began on 2 April before trees broke dormancy. The majority of chestnut attacks occurred in April and May. Progeny emerged from 48% of the caged galleries, including 35.9, 10.3, 3.3, and 1.1% X. germanus, X. crassiusculus, Hypothenemus spp., and X. saxeseni, respectively. Beetles exhibited several unusual behaviors during this study, including emergence of female X. germanus from trees the following spring, emergence of live male X. germanus and X. crassiusculus, a staggered chronology of progeny emergence, and presence of multiple beetle species emerging from the same gallery. Xylosandrus crassiusculus and X. germanus were the dominant species attacking chestnut, but total trap collections of X. germanus were small (=1.7%). Several findings from this study have significance to the nursery industry. The timing of peak trap collections during April (particularly collections of X. crassiusculus and X. saxeseni) coincided with peak tree attacks. The factors responsible for chestnut susceptibility to attack were not measured in this study, but since the majority of trees were attacked before dormancy break, tree phenological state probably is an important determinant of tree vulnerability. The collection of some species like X. germanus in trap collections may be a more important indicator of tree attack than abundance in the trap. Progeny emergence from chestnut trees during June and July did not coincide with increased trap collections or renewed attacks on chestnut. Therefore, traps may not always indicate ambrosia beetle abundance. We investigated the interaction between the honeydew-producing whitefly Aleurothrixus aepim and tending ants on shrubs of Croton floribundus (Euphorbiaceae) in a semideciduous forest in southeast Brazil. Whitefly eggs underside leaves were tagged during early March 1998 and randomly divided into two experimental groups: control (ants present, n = 75) and treatment (ants excluded, n = 52). During the 75-d experiment, honeydew-collecting ants visited 79% of the control groups of A. aepim. Adults emerged in significantly greater numbers from control than from treatment groups, the latter being heavily attacked by fungi due to accumulation of honeydew. Complete contamination by fungi was three times more frequent at ant-excluded (39%) than at ant-tended (13%) groups. Control groups with low levels of ant-tending produced significantly fewer adults than those more frequently tended by ants. Encarsia parasitoid wasps were more frequently seen on ant-excluded than on control A. aepim groups. Predatory arthropods, however, were equally frequent in either experimental group. This is the first experimental study to demonstrate ant-derived protection in honeydew-producing whiteflies. Given that aleyrodid honeydew can cause considerable damage to the host plant, our results suggest that the honeydew-gathering activity by tending ants is an important factor mediating such multitrophic interaction.
