Elevational migration can be defined as roundtrip seasonal movement that involves upward and downward shifts in elevation. These shifts incur physiological challenges that are proportional to the degree of elevational change. Larger shifts in elevation correspond to larger shifts in partial pressure of oxygen, air density, temperature, and ultraviolet (UV) exposure. Although most avian examples of elevational migration involve subtle shifts that would have minimal impacts on physiology, shifts of any magnitude have previously been considered under the broad umbrella of "elevational migration". Here, we consider extreme seasonal elevational movements (≥2,000 m), sufficient to shift the elevational dimension of the eco-climatic niche. Migratory bird populations typically maintain inter-seasonal stability in the temperature, precipitation, and elevational aspects of their climatic niches, a tendency that likely reflects genetic physiological specialization on environmental conditions such as atmospheric pressure. A shift of ≥2,000 m involves a ≥20% change in air density and oxygen partial pressure, sufficient to incur functionally impactful declines in arterial blood-oxygen saturation and require compensatory shifts in respiratory physiology. We refer to this phenomenon as elevational niche-shift migration (ENSM). In this review, we analyzed >4 million occurrence records to identify 105 populations, representing 92 bird species, that undergo complete or partial ENSM. We identified key ecological and evolutionary questions regarding the causes and consequences of ENSM. Our synthesis reveals that ENSM has evolved independently in at least 29 avian families spanning 10 orders. Nonetheless, ENSM is rare relative to other forms of seasonal migration, consistent with the general tendency of seasonal niche conservatism by migratory species and evolutionarily conserved elevational range limits. For many migratory species and populations, within-species patterns of migratory connectivity are not sufficiently understood to determine ENSM status. ENSM is distinguished by its scale within the broader phenomenon of elevational migration. Critical examination of ENSM illustrates fundamental constraints on the ecology and evolution of migration systems, topographical influences on geographic patterns of migratory connectivity, and the remarkable metabolic flexibility of certain bird species that allows them to occupy disparate elevations across different seasons. Precise phosphorus (P) application requires a mechanistic understanding of mineral effects on crop biology and physiology. Photosynthate assimilation, metabolism, and transport require phosphorylation, and we postulated that P is critical for the bloom and fruit-set of almond trees that rely on stored carbohydrate reserves. Hence, we studied the growth, physiology and carbohydrate dynamics in 2-year-old almond trees irrigated with P concentrations between 1 mg l−1 and 20 mg l−1. Almond trees attained maximal photosynthesis, transpiration, and growth by 6 mg P l−1 irrigation. Nevertheless, almond trees continued to extract P in 10 mg P l−1 and 15 mg P l−1 irrigations, which corresponded to larger yields. We attributed the augmented productivity to increased fruit-set (59% between 6 mg P l−1 and 15 mg P l−1), caused by more frequent (29%) honeybee visits. High P improved pollinator visitation by enabling almond trees to utilize more of their starch reserves for nectar secretion (which increased by ~140% between 6 mg P l−1 and 15 mg P l−1). This work elucidates the benefits of P fertilization to plant–pollinator mutualism, critical to almond productivity, and reveals novel indices for optimal P application in almond orchards. The motivation for this review comes from the emerging complexity of the autonomic innervation of the carotid body (CB) and its putative role in regulating chemoreceptor sensitivity. With the carotid bodies as a potential therapeutic target for numerous cardiorespiratory and metabolic diseases, an understanding of the neural control of its circulation is most relevant. Since nerve fibres track blood vessels and receive autonomic innervation, we initiate our review by describing the origins of arterial feed to the CB and its unique vascular architecture and blood flow. Arterial feed(s) vary amongst species and, unequivocally, the arterial blood supply is relatively high to this organ. The vasculature appears to form separate circuits inside the CB with one having arterial venous anastomoses. Both sympathetic and parasympathetic nerves are present with postganglionic neurons located within the CB or close to it in the form of paraganglia. Their role in arterial vascular resistance control is described as is how CB blood flow relates to carotid sinus afferent activity. We discuss non-vascular targets of autonomic nerves, their possible role in controlling glomus cell activity, and how certain transmitters may relate to function. We propose that the autonomic nerves sub-serving the CB provide a rapid mechanism to tune the gain of peripheral chemoreflex sensitivity based on alterations in blood flow and oxygen delivery, and might provide future therapeutic targets. The term 'vacuum phenomenon' (VP), is characterized by gas-like density areas due to a rapid increase in the joint space volume ('acute VP') or represent a chronic gas collection. It can occur within a collapsed vertebral body, the spinal canal, joints but mainly the intervertebral disc. Studies support that VP is originated by a dynamic process involving the balance between tissues' liquid and gaseous components, influenced by the duration and the depth of mechanical and metabolic alterations, by the nature of neighboring tissues and the variability in both pressure and permeability of disc or vertebral or joint structures. Prevalence of VP in the general population is about 2%, reaching 20% in the elderly with disc degeneration. Although it's often a random finding in asymptomatic patients, VP is an eventually painful expression of disc degeneration, or disc or vertebral fracture, or bone lesions. In sporadic cases, intradiscal gas can be expelled (all-in-one or gradually), resulting in a gaseous cyst, causing pain and neurological symptoms. Considering that spontaneous resolution and recurrence after surgery are both possible, most of the authors recommend conservative treatment in patients with intradiscal and intravertebral VP; occasionally percutaneous CT(computed tomography) -guided aspiration or vertebral stabilization. The production of bulk chemicals mostly depends on exhausting petroleum sources and leads to emission of greenhouse gases. Within the last decades the urgent need for alternative sources has increased and the development of bio-based processes received new attention. To avoid the competition between the use of sugars as food or fuel, other feedstocks with high availability and low cost are needed, which brought acetogenic bacteria into focus. This group of anaerobic organisms uses mixtures of CO2, CO and H2 for the production of mostly acetate and ethanol. Also methanol, a cheap and abundant bulk chemical produced from methane, is a suitable substrate for acetogenic bacteria. In methylotrophic acetogens the methyl group is transferred to the Wood-Ljungdahl pathway, a pathway to reduce CO2 to acetate via a series of C1-intermediates bound to tetrahydrofolic acid. Here we describe the biochemistry and bioenergetics of methanol conversion in the biotechnologically interesting group of anaerobic, acetogenic bacteria. In wheat, temperature affects the timing and intensity of stem elongation. Genetic variation for this process is therefore important for adaptation. This study investigates the genetic response to temperature fluctuations during stem elongation and its relationship to phenology and height. Canopy height of 315 wheat genotypes (GABI wheat panel) was scanned twice weekly in the field phenotyping platform (FIP) of ETH Zurich using a LIDAR. Temperature response was modelled using linear regressions between stem elongation and mean temperature in each measurement interval. This led to a temperature-responsive (slope) and a temperature-irresponsive (intercept) component. The temperature response was highly heritable (H 2=0.81) and positively related to a later start and end of stem elongation as well as final height. Genome-wide association mapping revealed three temperature-responsive and four temperature-irresponsive quantitative trait loci (QTLs). Furthermore, putative candidate genes for temperature-responsive QTLs were frequently related to the flowering pathway in Arabidopsis thaliana , whereas temperature-irresponsive QTLs corresponded to growth and reduced height genes. In combination with Rht and Ppd alleles, these loci, together with the loci for the timing of stem elongation, accounted for 71% of the variability in height. Extracellular vesicles (EVs), such as exosomes and microvesicles, are small membrane vesicles secreted by almost all cell types and are abundant in blood, body fluids, such as urine, spinal fluid, tears and saliva, and cell culture media. From an evolutionary perspective, they are biologically significant as a means for expelling unwanted cellular contents. Recently, EVs have received considerable attention as messengers of intercellular communication networks, allowing the exchange of proteins and lipids between the cells producing them and target cells that trigger various cellular responses. EVs also carry mRNAs and microRNAs inside them, transferring genetic information among cells. In addition, the expression pattern of these molecules is related to the cellular state and the progression of diseases, and the search for biomarkers within the EV is underway in many research fields. However, the physiological and pathophysiological roles of EVs remain largely elusive. Therefore, in this special issue, we have compiled reviews of the latest research findings on EV research. Nutritional and contaminant stressors influence organismal physiology, trophic interactions, community structure, and ecosystem-level processes; however, the interactions between toxicity and elemental imbalance in food resources have been examined in only a few ecotoxicity studies. Integrating well-developed ecological theories that cross all levels of biological organization can enhance our understanding of ecotoxicology. In the present article, we underline the opportunity to couple concepts and approaches used in the theory of ecological stoichiometry (ES) to ask ecotoxicological questions and introduce stoichiometric ecotoxicology, a subfield in ecology that examines how contaminant stress, nutrient supply, and elemental constraints interact throughout all levels of biological organization. This conceptual framework unifying ecotoxicology with ES offers potential for both empirical and theoretical studies to deepen our mechanistic understanding of the adverse outcomes of chemicals across ecological scales and improve the predictive powers of ecotoxicology. Alternative reproductive tactics occur when conspecifics of the same sex use different behavioral processes to maximize reproductive success. Resource availability can have significant impacts on the proximate factors impacting the success and persistence of alternative tactics, such as an individual's physiology, immunity, and body condition. Male Cape ground squirrels (Xerus inauris) display two alternative tactics, with some mature males dispersing from their natal groups and joining bands of nonrelated males (band males), whereas others do not disperse and remain philopatric in their natal groups past sexual maturation (natal males). Despite differences in metabolism, home ranges, and behavior, there is no difference in reproductive success between the two tactics, allowing us to test if rainfall (as a measure of primary productivity) influences body condition and physiological indices in tactics when fitness is equal. Over a period of 6 years, rainfall was positively related to body condition (mass–spine length residuals) in band males (N = 76), but not in natal males (N = 75). When we followed 18 males prior to and after dispersal, we also found a positive relationship between body condition and age following dispersal, but not prior to dispersal, and natal males maintained a better body condition in low rainfall years. During 2 years of low rainfall, band males (N = 39) had higher total ectoparasite abundance, and white blood cell differentials, implying they are less able to tolerate parasitic infection than natal males (N = 31). We found no difference in bacterial growth inhibition between tactics, suggesting that band and natal males both are equally resistant to parasites. The poorer body condition of band males during lower rainfall suggests they are less tolerant to parasites in these years. Dispersal during years of low rainfall thus may increase the costs to body condition and parasite infection, but the reproductive benefit of overlapping with more females may contribute to maintaining equal fitness between tactics. Synchronized movements are often key elements in activities where social bonding and emotional connection are a shared goal, such as religious gatherings, sporting events, parties and weddings. Previous studies have shown that synchronous movements enhance prosocial attitudes and affiliative behaviors. Similarly, observers attribute more social closeness to people moving synchronously together than people moving asynchronously. The mechanisms by which synchrony modulates these attributions are not well understood. In the present study, we ask whether viewing synchronous activities influences physiological arousal as measured by skin conductance and whether group size impacts this effect. Undergraduates viewed a series of short videos depicting people moving either (1) in or out of synchrony with each other and (2) in a large or small group. Participants' skin conductance was measured. Change in skin conductance levels and response counts were attenuated while watching synchronous movement, but only in the large-group condition. Post-hoc analyses suggest that viewer enjoyment/interest in the large-group synchronous videos mediated this association for phasic skin conductance responses, but no evidence of mediation was found for tonic skin conductance levels. Results extend previous research on affiliative effects of first-person interpersonal synchrony and demonstrate that watching others moving synchronously has an attenuating effect on observers' physiological state. When the developing embryo enters the uterus, before the placenta develops, the embryo receives nourishment from endometrially derived histotroph produced by the uterine glands that remain functional throughout gestation. Throughout pregnancy, this histotroph remains necessary for proper embryonic development. Maternal nutrition may affect histotroph composition resulting in altered embryo development. The objectives were to determine the effects of dietary intake on histotroph amino acid (AA) concentrations. Eighteen multiparous nonpregnant cows (initial BW = 597 kg) were housed at the North Dakota State University Animal Nutrition and Physiology Center (n = 6 cows/pen). Cows were individually fed a TMR once daily via a Calan feeding system and had ad libitum access to fresh water. Cows were randomly assigned to 1 of 2 treatments (n = 9/treatment): dietary intake designed to maintain body weight (CON) or dietary intake designed to lose body weight at a moderate rate (-0.7 kg/d; NEG), for 63 days. Slaughter group 1 contained 4 CON and 5 NEG cows, and group 2 contained 5 CON and 4 NEG cows. Cows were estrus-synchronized with the Co-Synch + CIDR protocol to ensure a common day of the estrous cycle at the time of slaughter (3 d after completion of the Co-Synch + CIDR protocol). When total uterine histotroph AA was calculated (luteal + non-luteal horns) and analyzed by treatment with group in the model, the differences were: CON > NEG for 1) Trp as % of essential AA, and 2) Ser as % of total AA (P = 0.04 and 0.05, respectively). Tendencies for total tract histotroph were observed for Phe and Trp as a % of total AA (P = 0.07 and 0.06, respectively) for CON > NEG. Luteal histotroph Ser as a % of total AA was greater (P = 0.05) for CON vs. NEG. When evaluating non-luteal histotroph, CON had greater (P < 0.05) Cys as a % of total AA compared with NEG cows. Nonluteal histotroph Arg concentration tended (P = 0.10) to be greater for NEG vs. CON. These results demonstrate that feed intake affects cow AA supply in the uterine histotroph, and these AA (Trp, Ser, Phe, Cys and Arg) may function in critical nutrient pathways, reproductive tract vascularization, and antioxidant production, which are necessary for optimal fetal development. Maternal gestational nutrition is known to impact gut physiology and the gut microbiome of offspring early in life; however, it remains unknown if these changes persist later in life. We hypothesized that the impacts of maternal feed restriction in late gestating beef cows on early colonization of the calf gut would impact the rumen microbiome post-weaning. Our objective was to compare the post-weaning rumen microbiome of calves born with feed restricted dams and those born to dams with adequate nutrition. Starting roughly 60 d before expected calving, dams were stratified to either receive 100% of NRC requirements (CON) or 70% intake of NRC requirements (FR) through 28 d post-parturition. Pairs were then moved on to native grass until weaning. Rumen fluid was collected from the calves at the time of weaning (PRE) and after a 70 d backgrounding feed efficiency study (POST). Microbial DNA was isolated and the V4 region of the 16S rRNA was sequenced on the MiSeq platform. Data were analyzed within each time point using QIIME2. Maternal treatment had no impact on alpha diversity (Faith's phylogeny, Shannon's diversity, and Evenness) in the PRE group, but CON calves had greater (q = 0.05) evenness than FR calves in the POST samples. There were differences in composition between CON and FR in the PRE samples (unweighted unifrac q = 0.04; bray-curtis q = 0.03). No differences (q = 0.14) in composition were found in the POST samples. These data indicate that at the time of weaning (PRE) differences in composition existed in CON vs. FR calves, specifically in the rare taxa, but the microbial composition converged by the end of the feeding trial. Interestingly, previous findings in our lab reported differences in alpha and beta diversity 28 d after birth in these calves. Together, these data indicate the potential for long-term impacts of maternal nutrition on the gut microbiome. This coupled with the previously published work where feed efficiency tended to differ between CON and FR, suggests that early perturbations during gestation may have had lasting impacts on gut physiology and subsequent performance in the presence of rumen microbial shifts. The rumen microbial consortium has long been viewed as a "black box", where we recognize what goes in, and most of what comes out, but the ruminal catabolic and anabolic activity remains shrouded in uncertainty. Microbiologists from the 1940s through the early 2000s identified microbes responsible for ruminal feedstuff degradation, highlighted animal-important fermentation endproducts (such as volatile fatty acids, VFA), and examined how microbes interacted with each other (e.g., interspecies hydrogen transfer), and with the host animal. However, our knowledge was dependent on our ability to grow anaerobic microbes in pure or mixed cultures, but it became apparent that our limiting factor was that we could culture <10% of the ruminal microbes. In the past 15 years, there has been a proliferation of new techniques to identify what is occurring in the rumen of live ruminants such as, culturable and unculturable microorganism populations, microbial biochemical pathways and important metabolic intermediates, potential interactions between microbes, and biologically relevant endproducts that impact host animal physiology, health, and production efficiency. As Next Generation Sequencing (NGS) has become increasingly affordable, there has been a surge in research examining the "microbiome" with a resultant influx of massive datasets. However, without the ability to connect microbial populations to biochemical degradative pathways, endproducts, microbial interactions or animal production parameters, this information is of limited impact in ruminant nutrition. Ruminant nutrition is on a precipice where we can finally fully integrate microbiology with metabolomics and animal performance and nutrition. Advancements in ruminal microbiology will allow us to fully grasp the functional nature of the microbial ecosystem and where it fits within ruminant nutrition, which will improve animal productivity and ruminant-derived protein food production sustainability. The diverse rumen microbiome converts lowquality feedstuffs into usable energy and provides approximately 70% of the energy required by its ruminant host through fermentation byproducts. Due to microbial impacts on host health and productivity, numerous studies have associated rumen microbiome variation with important sustainability phenotypes, such as feed efficiency and methane emissions. Although diet, genetics, and environment contribute to variation in feed efficiency, selection for feed-efficient beef cattle using genetic improvement technologies has helped to understand and improve the persistence and longevity of such phenotypes within the herd. While diet has been considered to be the primary driver for the composition and structure of the rumen microbiome, recent studies have indicated the microbial communities present in the rumen are under low-to-moderate host genetic control. The mutualistic, commensal, and parasitic microorganisms that reside in the rumen and lower gastrointestinal tract of cattle and other ruminants exert enormous influence over animal physiology and performance. The ability to interrogate these systems at great depth has permitted a greater understanding of the microbiological and molecular mechanisms involved in ruminant nutrition and health. The work in our group in the field of beef cattle gut microbiology, as it relates to feed efficiency, permits the exploration of these critical microbial community networks. This knowledge will aid researchers seeking to address the grand challenge of maintaining host-efficient gut microbiomes throughout cattle production operations. Improving feed efficiency is essential for the sustainability of the US beef production system. However, achieving this goal is challenging due to various factors such as diet types, genetics, environments, and industry segmentation. To address these difficulties, it is critical to have a fundamental understanding of what contributes to efficient nutrient utilization and feed resources, which can guide future genetic selection and production practices. Despite recent progress, the regulation of feed intake in growing and finishing cattle remains incompletely understood, hindering advances in feed efficiency. Recent research has aimed to advance the hepatic oxidation theory to growing and finishing cattle fed high-concentrate diets and explored the control of appetite by gut peptide hormones. Other factors contributing to variation in feed efficiency include nutrient metabolism and hormonal control of metabolism. Previous research is limited by a lack of in-depth analysis of metabolism due to the inability to time sample collection relative to feed consumption and account for correlations among metabolites. The role of fat metabolism in the rumen has been associated with epithelial inflammation, while inflammation of the gut has been identified as a contributing factor to inefficiencies in nutrient utilization. However, there is still a need to further understand the role of fatty acid metabolism in the induction of inflammation. An issue that limits the further understanding of efficiency is that a wide variety of feedstuffs are utilized by the industry and small changes in the nutrient composition of a diet can have large impacts on efficiency. Therefore, it is important to consider the utilization efficiency of specific nutrients rather than general feed efficiency. Nitrogen utilization efficiency is becoming increasingly important due to the significant economic and environmental ramifications of inefficiencies in nitrogen utilization. There is growing evidence that precision feeding of protein to match requirements can reduce nitrogen excretion without compromising growth and efficiency. In conclusion, a better understanding of the underlying biology contributing to nutrient utilization is crucial for improving feed utilization efficiency in the US beef production system, which will enhance the sustainability of the US beef production system. First described in 2012, the glymphatic system is responsible for maintaining homeostasis within the central nervous system, including nutrient delivery, waste clearance, and consistency of the ionic microenvironment. It is comprised of glial cells and barrier systems that modulate neurofluid production, circulation, and exchange. Experimental interrogation of neurofluid dynamics is restricted to ex vivo and in vitro studies in animals and humans, therefore diagnostic imaging plays an important role in minimally invasive evaluation. This review article will synthesize current knowledge and theories regarding neurofluid circulation and implications for neuroimaging. First, we will discuss the anatomy of the neurogliovascular unit, including paravascular and perivascular pathways of fluid exchange. In addition, we will summarize the structure and function of barrier systems including the blood–brain, blood–cerebrospinal fluid, and brain–cerebrospinal fluid barriers. Next, we will mention physiologic factors that yield normal variations in neurofluid circulation, and how various disease pathologies can disrupt glymphatic drainage pathways. Lastly, we will cover the spectrum of diagnostic imaging and interventional techniques with relevance to glymphatic structure, flow, and function. Historically, animals growing well were believed to be in a positive welfare state. Evidence shows that pigs will seek out enrichment using play and complex puzzle-solving, which is beyond the physical needs required for positive growth. Physiological and psychological indicators of welfare can now be used to address positive affect in swine. Further, early research may show that those animals living with enrichment may be less likely to fall ill and are better prepared to handle sudden life changes (such as transport). However, measuring animal pleasure successfully is difficult. Understanding ways to measure animal welfare using behavior and physiology can provide insight into validating positive affective states beyond historically used measurements such as average daily gain, reproductive performance, or cortisol. This experiment compared incidence of frothy bloat, as well as ruminal, physiological, and performance responses of beef heifers receiving a bloat-provoking diet and supplemented with Yucca schidigera extract. Sixteen ruminally cannulated Angus-influenced heifers were ranked by body weight (BW) and assigned to 4 groups of 4 heifers each. Groups were enrolled in a replicated 4 × 4 Latin square design containing 4 periods of 28 d, and a 21-d washout interval between periods. Groups were assigned to receive no Y. schidigera extract (CON), or Y. schidigera extract at (as-fed basis) 1 g/heifer daily (YS1), 2 g/heifer daily (YS2), or 4 g/heifer daily (YS4). During each period, heifers (n = 16/treatment) were housed in individual pens, and fed a sorghum (Sorghum bicolor L.)-based bloat-provocative diet at 2% of their BW. Diet and treatments were individually fed to heifers, twice daily in equal proportions (0700 and 1600 hours). Heifers were assessed for bloat score (0 to 5 scale, increasing according to bloat severity) 3 hr after the morning feeding. Blood samples were collected on days 0, 7, 14, 21, and 28 prior to (0 hr) and at 3, 6, and 9 hr relative to the morning feeding. Rumen fluid samples were collected at the same time points on days 0 and 28. Orthogonal contrasts were tested to determine whether inclusion of Y. schidigera extract (0, 1, 2, or 4 g/heifer daily) yielded linear or quadratic effects, and explore an overall effect of Y. schidigera extract supplementation (CON vs. YS1 + YS2 + YS4). Rumen fluid viscosity was impacted quadratically by Y. schidigera extract inclusion (P = 0.02), being greatest in YS1, followed by YS2, and equivalent between CON and YS4 heifers. Heifers receiving Y. schidigera extract had greater (P ≤ 0.05) rumen propionate, iso-valerate, and valerate concentrations, as well as less (P < 0.01) acetate : propionate ratio compared with CON heifers. Inclusion of Y. schidigera extract linearly increased (P ≤ 0.04) average daily gain and feed efficiency. No other treatment effects were noted (P ≥ 0.19) including bloat score (1.07 ± 0.03 across treatments), ruminal protozoa count, plasma concentrations of cortisol, haptoglobin, urea N, total protein, and rumen concentration of total volatile fatty acids. Precise genetic manipulation of specific cell types or tissues to pinpoint gene function requirement is a critical step in studies aimed at unraveling the intricacies of organismal physiology. Drosophila researchers heavily rely on the UAS/Gal4/Gal80 system for tissue-specific manipulations; however, it is often unclear whether the reported Gal4 expression patterns are indeed specific to the tissue of interest such that experimental results are not confounded by secondary sites of Gal4 expression. Here, we surveyed the expression patterns of commonly used Gal4 drivers in adult Drosophila female tissues under optimal conditions and found that multiple drivers have unreported secondary sites of expression beyond their published cell type/tissue expression pattern. These results underscore the importance of thoroughly characterizing Gal4 tools as part of a rigorous experimental design that avoids potential misinterpretation of results as we strive for understanding how the function of a specific gene/pathway in one tissue contributes to whole-body physiology. While the general effect of CO2 enrichment on photosynthesis, stomatal conductance, N content, and yield has been documented, there is still some uncertainty as to whether there are interactive effects between CO2 enrichment and other factors, such as temperature, geographical location, water availability, and cultivar. In addition, the metabolic coordination between leaves and grains, which is crucial for crop responsiveness to elevated CO2, has never been examined closely. Here, we address these two aspects by multi-level analyses of data from several free-air CO2 enrichment experiments conducted in five different countries. There was little effect of elevated CO2 on yield (except in the USA), likely due to photosynthetic capacity acclimation, as reflected by protein profiles. In addition, there was a significant decrease in leaf amino acids (threonine) and macroelements (e.g. K) at elevated CO2, while other elements, such as Mg or S, increased. Despite the non-significant effect of CO2 enrichment on yield, grains appeared to be significantly depleted in N (as expected), but also in threonine, the S-containing amino acid methionine, and Mg. Migraine is the most common neurological disorder worldwide and it has been shown to have complex polygenic origins with a heritability of estimated 40-70%. Both common and rare genetic variants are believed to underlie the pathophysiology of the prevalent types of migraine, migraine with typical aura and migraine without aura. However, only common variants have been identified so far. Here we identify for the first time a gene module with rare mutations through a systems genetics approach integrating RNA sequencing data from brain and vascular tissues likely to be involved in migraine pathology in combination with whole genome sequencing of 117 migraine families. We found a gene module in the visual cortex, based on single nuclei RNA sequencing data, that had increased rare mutations in the migraine families and replicated this in a second independent cohort of 1930 patients. This module was mainly expressed by interneurons, pyramidal CA1, and pyramidal SS cells, and pathway analysis showed association with hormonal signalling (thyrotropin-releasing hormone receptor and oxytocin receptor signalling pathways), Alzheimer's disease pathway, serotonin receptor pathway and general heterotrimeric G-protein signalling pathways. Our results demonstrate that rare functional gene variants are strongly implicated in the pathophysiology of migraine. Furthermore, we anticipate that the results can be used to explain the critical mechanisms behind migraine and potentially improving the treatment regime for migraine patients. Embryo implantation is a hallmark of the female reproductive biology of eutherian (placental) mammals and does not exist in a sustainable form in any other vertebrate group. Implantation is the initial process that leads to a sustained fetal-maternal unit engendering a complex functional relationship between the mother and the embryo/fetus. The nature of this relationship is often portrayed as one of conflict between an aggressive embryo and a passive or defensive maternal organism. Recent progress in elucidating the evolutionary origin of eutherian pregnancy leads to a different picture. The emerging scenario suggests that the very initial stages in the evolution of embryo implantation required evolutionary changes to the maternal physiology which modified an ancestral generic mucosal inflammation in response to the presence of the embryo into an active embedding process. This "female-first" evolutionary scenario also explains the role of endometrial receptivity in human pregnancy. On the marsupial side, where in most animals the fetal–maternal interaction is short and does not lead to a long term sustainable placentation, the relationship is mutual. In these mammals, uterine inflammation is followed by parturition in short order. The inflammatory signaling pathways, however, are cooperative, i.e. they are performed by both the fetus and the mother and therefore we call this relationship "cooperative inflammation." Myths, misconceptions, and taboos about sexual anatomy and physiology are common and can affect sexual health and maintain harmful practices and beliefs. To construct a female and a preliminary male 3-dimensional (3D) pelvic model on the basis of in vivo imaging, which could be studied in sex education and clinical practice. We retrospectively studied the images of 200 female pelvic magnetic resonance examinations and reviewed the literature to choose the optimum magnetic resonance imaging (MRI) protocol for the study of the clitoris and surrounding organs. We also conducted a cross-sectional study of 30 women who were undergoing a pelvic MRI. 15 women had undergone female genital mutilation/cutting involving the clitoris and 15 had not. The best-quality MRI images of 3 uncut and 1 cut clitoris, together with the principal surrounding pelvic organs, were selected to generate 3D reconstructions using dedicated software. The same software was used to reconstruct the anatomy of the penis and the principal surrounding pelvic organs, based on contrast-enhanced computer tomography images. Images of both models were exported in.stl format and cleaned to obtain single manifold objects in free, open source software. Each organ model was sliced and 3D printed. A preliminary feedback was collected from 13 potential users working in urology, gynaecology, sexual medicine, physiotherapy, and education. The main outcomes of this study are a kit of 3D pelvic models, 2-dimensional figures of female and male sexual anatomy, and files for 3D printing. We present a kit containing 3D models and 2-dimensional figures of female and male sexual anatomy, based on in vivo imaging and, feedbacks and suggestions received from potential users. Our kit can be used in anatomy and sex education among and by health professionals, teachers, sex educators, students, and the general population. The strengths are that the models were based on in vivo imaging, can be dismantled/reassembled, and show analogous anatomic structures of the clitoris and the penis. The female models represent diversity, including women with female genital mutilation/cutting. The limitations are that the male model is preliminary and can be improved if based on an MRI; that imaging-based anatomic representations can differ from anatomic dissections; and that the models represent the sexual organs at rest or during an unknown state of arousal only. Anti-N-methyl-d-aspartate receptor (NMDAR) encephalitis is an immune-mediated disease characterized by a complex neuropsychiatric syndrome in association with an antibody-mediated decrease of NMDAR. About 85% of patients respond to immunotherapy (and removal of an associated tumour if it applies), but it often takes several months or more than 1 year for patients to recover. There are no complementary treatments, beyond immunotherapy, to accelerate this recovery. Previous studies showed that SGE-301, a synthetic analogue of 24(S)-hydroxycholesterol, which is a potent and selective positive allosteric modulator of NMDAR, reverted the memory deficit caused by phencyclidine (a non-competitive antagonist of NMDAR), and prevented the NMDAR dysfunction caused by patients' NMDAR antibodies in cultured neurons. An advantage of SGE-301 is that it is optimized for systemic delivery such that plasma and brain exposures are sufficient to modulate NMDAR activity. Here, we used SGE-301 to confirm that in cultured neurons it prevented the antibody-mediated reduction of receptors, and then we applied it to a previously reported mouse model of passive cerebroventricular transfer of patient's CSF antibodies. Four groups were established: mice receiving continuous (14-day) infusion of patients' or controls' CSF, treated with daily subcutaneous administration of SGE-301 or vehicle (no drug). The effects on memory were examined with the novel object location test at different time points, and the effects on synaptic levels of NMDAR (assessed with confocal microscopy) and plasticity (long-term potentiation) were examined in the hippocampus on Day 18, which in this model corresponds to the last day of maximal clinical and synaptic alterations. As expected, mice infused with patient's CSF antibodies, but not those infused with controls' CSF, and treated with vehicle developed severe memory deficit without locomotor alteration, accompanied by a decrease of NMDAR clusters and impairment of long-term potentiation. All antibody-mediated pathogenic effects (memory, synaptic NMDAR, long-term potentiation) were prevented in the animals treated with SGE-301, despite this compound not antagonizing antibody binding. Additional investigations on the potential mechanisms related to these SGE-301 effects showed that (i) in cultured neurons SGE-301 prolonged the decay time of NMDAR-dependent spontaneous excitatory postsynaptic currents suggesting a prolonged open time of the channel; and (ii) it significantly decreased, without fully preventing, the internalization of antibody-bound receptors suggesting that additional, yet unclear mechanisms, contribute in keeping unchanged the surface NMDAR density. Overall, these findings suggest that SGE-301, or similar NMDAR modulators, could potentially serve as complementary treatment for anti-NMDAR encephalitis and deserve future investigations. Background: Haem oxygenases (HO) catabolise haem, which is the prosthetic group of numerous haemoproteins. Thus, multiple primary cellular pathways and functions rely on haem availability. HO exists in two isoforms, both expressed in the placenta, namely HO-1 and HO-2, the first being inducible. Haem oxygenases, particularly HO-1, have garnered specific interest in the field of physiological and pathological placental function. These enzymes mediate haem degradation by cleaving the alpha methene bridge to produce biliverdin, which is subsequently converted to bilirubin, carbon monoxide and iron. Folates are one of the essential micronutrients for all living organisms. Due to inadequate dietary intake, folate deficiency remains prevalent in humans. Genetically diverse germplasms can potentially be used as parents in breeding programs and also for understanding the folate regulatory network. Therefore, we investigated the natural genetic diversity of folates and their physiological regulation in pak choi (Brassica rapa subsp. Chinensis) germplasm. The total folate concentration ranged from 52.7 μg 100 gFW–1 to 166.9 μg 100 gFW–1, with 3.2-fold variation. The main folate vitamer was represented by 5-CH3-H4folate, with 4.5-fold variation. The activities of GTP cyclohydrolase I and aminodeoxy chorismate synthase, the first step of folate synthesis, were high in high folate accessions and low in low folate accessions. Analysis of the transcription levels of 11 genes associated with folate metabolism demonstrated that the difference in folate concentrations may be primarily controlled at the post-transcriptional level. A general correlation between total folate and their precursors was observed. Folate diversity and chlorophyll content were tightly regulated through the methyl cycle. The diverse genetic variation in pak choi germplasm indicated the great genetic potential to integrate breeding programs for folate biofortification and unravel the physiological basis of folate homeostasis in planta. Over the last 100 years, fishes native to the Southwestern United States have faced a myriad of biotic and abiotic pressures which has resulted in most being federally listed as endangered or threatened. Most notably, water diversions and the introduction of non-native fishes have been the primary culprits in causing the downfall of native fish populations. We describe how recent studies of morphology, physiology, and behavior yield insights into the failed (occasionally successful) management of this vanishing biota. We describe how understanding locomotor morphologies, physiologies, and behaviors unique to Southwestern native fishes can be used to create habitats that favor native fishes. Additionally, through realizing differences in morphologies and behaviors between native and non-native fishes, we describe how understanding predator–prey interactions might render greater survivorship of native fishes when stocked into the wild from repatriation programs. Understanding fundamental form–function relationships is imperative for managers to make educated decisions on how to best recover species of concern in the Southwestern United States and worldwide. Discovering widespread microbial processes that create variation in soil carbon (C) cycling within ecosystems may improve soil C modeling. Toward this end, we screened 206 soil communities decomposing plant litter in a common garden microcosm environment and examined features linked to divergent patterns of C flow. C flow was measured as carbon dioxide (CO2) and dissolved organic carbon (DOC) from 44-days of litter decomposition. Two large groups of microbial communities representing 'high' and 'low' DOC phenotypes from original soil and 44-day microcosm samples were down-selected for fungal and bacterial profiling. Metatranscriptomes were also sequenced from a smaller subset of communities in each group. The two groups exhibited differences in average rate of CO2 production, demonstrating that the divergent patterns of C flow arose from innate functional constraints on C metabolism, not a time-dependent artefact. To infer functional constraints, we identified features – traits at the organism, pathway or gene level – linked to the high and low DOC phenotypes using RNA-Seq approaches and machine learning approaches. Substrate use differed across the high and low DOC phenotypes. Additional features suggested that divergent patterns of C flow may be driven in part by differences in organism interactions that affect DOC abundance directly or indirectly by controlling community structure. Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive cyst formation, leading to growth in kidney volume and renal function decline. Although therapies have emerged, there is still an important unmet need for slowing the rate of disease progression in ADPKD. High intracellular levels of adenosine 3′,5′-cyclic monophosphate (cAMP) are involved in cell proliferation and fluid secretion, resulting in cyst formation. Somatostatin (SST), a hormone that is involved in many cell processes, has the ability to inhibit intracellular cAMP production. However, SST itself has limited therapeutic potential since it is rapidly eliminated in vivo. Therefore analogues have been synthesized, which have a longer half-life and may be promising agents in the treatment of ADPKD. Insulin/insulin-like growth factor signaling (IIS) is a conserved mechanism to regulate animal physiology in response to nutrition. IIS activity controls gene expression, but only a subset of transcriptional regulators (TRs) targeted by the IIS pathway is currently known. Here we report the results of an unbiased screen for Drosophila TRs phosphorylated in an IIS-dependent manner. To conduct the screen, we built a library of 857 V5/Strep-tagged TRs under the control of Copper-inducible metallothionein promoter (pMt). The insulin-induced phosphorylation changes were detected by using Phos-tag SDS-PAGE and Western blotting. Eight proteins were found to display increased phosphorylation after acute insulin treatment. In each case, the insulin-induced phosphorylation was abrogated by mTORC1 inhibitor rapamycin. The hits included two components of the NURF complex (NURF38 and NURF55), bHLHZip transcription factor Max, as well as the Drosophila ortholog of human proliferation-associated 2G4 (dPA2G4). Subsequent experiments revealed that the expression of the dPA2G4 gene was promoted by the mTOR pathway, likely through transcription factor Myc. Furthermore, NURF38 was found to be necessary for growth in larvae, consistent with the role of IIS/mTOR pathway in growth control. [ABSTRACT FROM AUTHOR]

Anthropogenic change has well-documented impacts on stress physiology and behavior across diverse taxonomic groups. Within individual organisms, physiological and behavioral traits often covary at proximate and ultimate timescales. In the context of global change, this means that impacts on physiology can have downstream impacts on behavior, and vice versa. Because all organisms interact with members of their own species and other species within their communities, the effects of humans on one organism can impose indirect effects on one or more other organisms, resulting in cascading effects across interaction networks. Human-induced changes in the stress physiology of one species and the downstream impacts on behavior can therefore interact with the physiological and behavioral responses of other organisms to alter emergent ecological phenomena. Here, we highlight three scenarios in which the stress physiology and behavior of individuals on different sides of an ecological relationship are interactively impacted by anthropogenic change. We discuss host–parasite/pathogen dynamics, predator–prey relationships, and beneficial partnerships (mutualisms and cooperation) in this framework, considering cases in which the effect of stressors on each type of network may be attenuated or enhanced by interactive changes in behavior and physiology. These examples shed light on the ways that stressors imposed at the level of one individual can impact ecological relationships to trigger downstream consequences for behavioral and ecological dynamics. Ultimately, changes in stress physiology on one or both sides of an ecological interaction can mediate higher-level population and community changes due in part to their cascading impacts on behavior. This framework may prove useful for anticipating and potentially mitigating previously underappreciated ecological responses to anthropogenic perturbations in a rapidly changing world. Although poor nutrition is cited as one of the crucial factors in global pollinator decline, the requirements and role of several important nutrients (especially micronutrients) in honey bees are not well understood. Micronutrients, viz. phytosterols, play a physiologically vital role in insects as precursors of important molting hormones and building blocks of cellular membranes. There is a gap in comprehensive understanding of the impacts of dietary sterols on honey bee physiology. In the present study, we investigated the role of 24-methylenecholesterol—a key phytosterol—in honey bee nutritional physiology. Artificial diets with varying concentrations of 24-methylenecholesterol (0%, 0.1%. 0.25%, 0.5%, 0.75%, and 1% dry diet weight) were formulated and fed to honey bees in a laboratory cage experiment. Survival, diet consumption, head protein content, and abdominal lipid contents were significantly higher in dietary sterol-supplemented bees. Our findings provide additional insights regarding the role of this important sterol in honey bee nutritional physiology. The insights gleaned from this study could also advance the understanding of sterol metabolism and regulation in other bee species that are dependent on pollen for sterols, and assist in formulation of a more complete artificial diet for honey bees (Apis mellifera Linnaeus, 1758) (Hymenoptera: Apidae). The thermophilic hydrogenotrophic methanogen Methanothermobacter sp. CaT2 aggregates by itself. CaT2 is known to have a surface sugar layer and extracellular proteins that may be related to its aggregation. Aggregation-enhanced mutants, CHA001 and CHA002, were isolated after repeated cultivation for more than two years. When treated with proteinase K, CHA001 and CaT2 similarly exhibited a very low degree of aggregation and CHA002 exhibited less aggregation but still retained aggregation, suggesting protein-based aggregation via extracellular proteins in both CHA001 and CHA002, presumably via a putative membrane-bound and extracellularly protruding protein, MTCT_1020, identified previously. Genomic analysis revealed that CHA001 and CHA002 shared a missense mutation of MTCT_1348 and had distinct mutations. These results suggested that the MTCT_1348 mutation provides subsidiary support to the adhesive function of extracellular proteins and that there is an additional mutation(s) in CHA002 for the non-proteinous aggregation capability. Acquired immune function shows recognizable changes over time with organismal aging. These changes include T-cell dysfunction, which may underlie diminished resistance to infection and possibly various chronic age-associated diseases in the elderly. T-cell dysfunction may occur at distinct stages, from naive cells to the end stages of differentiation during immune responses. The thymus, which generates naive T cells, shows unusually early involution resulting in progressive reduction of T-cell output after adolescence, but peripheral T-cell numbers are maintained through antigen-independent homeostatic proliferation of naive T cells driven by the major histocompatibility complex associated with self-peptides and homeostatic cytokines, retaining the diverse repertoire. However, extensive homeostatic proliferation may lead to the emergence of dysfunctional CD4+ T cells with features resembling senescent cells, termed senescence-associated T (SA-T) cells, which increase and accumulate with age. In situations such as chronic viral infection, T-cell dysfunction may also develop via persistent antigen stimulation, termed exhaustion, preventing possible immunopathology due to excessive immune responses. Exhausted T cells are developed through the effects of checkpoint receptors such as PD-1 and may be reversed with the receptor blockade. Of note, although defective in their regular T-cell antigen-receptor-mediated proliferation, SA-T cells secrete abundant pro-inflammatory factors such as osteopontin, reminiscent of an SA-secretory phenotype. A series of experiments in mouse models indicated that SA-T cells are involved in systemic autoimmunity as well as chronic tissue inflammation following tissue stresses. Domesticated buffaloes have been integral to rice-paddy agro-ecosystems for millennia, yet relatively little is known about the buffalo genomics. Here, we sequenced and assembled reference genomes for both swamp and river buffaloes and we re-sequenced 230 individuals (132 swamp buffaloes and 98 river buffaloes) sampled from across Asia and Europe. Beyond the many actionable insights that our study revealed about the domestication, basic physiology and breeding of buffalo, we made the striking discovery that the divergent domestication traits between swamp and river buffaloes can be explained with recent selections of genes on social behavior, digestion metabolism, strengths and milk production. In tall conifers, leaf structure can vary dramatically with height due to decreasing water potential (Ψ) and increasing light availability. This variation in leaf structure can have physiological consequences such as increased respiratory costs, reduced internal carbon dioxide conductance rates and ultimately reduced maximum photosynthetic rates (A max). In Picea sitchensis (Bong.) Carrière, the leaf structure varies along the vertical gradient in ways that suggest compensatory changes to enhance photosynthesis, and this variation seems to be driven largely by light availability rather than by Ψ. These trends in leaf structure coupled with remarkably fast growth rates and dependence on moist environments inspire two important questions about P. sitchensis : (i) does foliar water uptake minimize the adverse effects of decreasing Ψ with height on leaf structure, and (ii) do trends in leaf structure increase photosynthetic rates despite increasing height? To answer these questions, we measured foliar water uptake capacity, predawn (Ψpd) and midday water potential and gas-exchange rates as they varied between 25- and 89-m heights in 300-year-old P. sitchensis trees in northwestern California. Our major findings for P. sitchensis include the following: (i) foliar water uptake capacity was quite high relative to published values for other woody species; (ii) foliar water uptake capacity increased between the crown base and treetop; (iii) wet season Ψpd was higher than predicted by the gravitational potential gradient, indicating foliar water uptake; and (iv) the maximum photosynthetic rate increased with height, presumably due to shifts in leaf structure between the crown base and treetop, mitigating height-related decreases in A max. These findings suggest that together, the use of fog, dew and rain deposits on leaves and shifts in the leaf structure to conserve and possibly enhance photosynthetic capacity likely contribute to the rapid growth rates measured in this species. Ocean acidification (OA) and its subsequent changes in seawater carbonate chemistry are threatening the survival of calcifying organisms. Due to their use of calcium carbonate to build their shells, marine molluscs are particularly vulnerable. This study investigated the effect of CO2-induced OA on adult European abalone (Haliotis tuberculata) using a multi-parameter approach. Biological (survival, growth), physiological (pHT of haemolymph, phagocytosis, metabolism, gene expression), and structural responses (shell strength, nano-indentation measurements, Scanning electron microscopy imaging of microstructure) were evaluated throughout a 5-month exposure to ambient (8.0) and low (7.7) pH conditions. During the first 2 months, the haemolymph pH was reduced, indicating that abalone do not compensate for the pH decrease of their internal fluid. Overall metabolism and immune status were not affected, suggesting that abalone maintain their vital functions when facing OA. However, after 4 months of exposure, adverse effects on shell growth, calcification, microstructure, and resistance were highlighted, whereas the haemolymph pH was compensated. Significant reduction in shell mechanical properties was revealed at pH 7.7, suggesting that OA altered the biomineral architecture leading to a more fragile shell. It is concluded that under lower pH, abalone metabolism is maintained at a cost to growth and shell integrity. This may impact both abalone ecology and aquaculture. [ABSTRACT FROM AUTHOR]

As 2020 is "The Year of the Rat" in the Chinese astrological calendar, it seems an appropriate time to consider whether we should bring back the laboratory rat to front-and-center in research on the basic biology of mammalian aging. Beginning in the 1970s, aging research with rats became common, peaking in 1992 but then declined dramatically by 2018 as the mouse became preeminent. The purpose of this review is to highlight some of the historical contributions as well as current advantages of the rat as a mammalian model of human aging, because we suspect at least a generation of researchers is no longer aware of this history or these advantages. Herein, we compare and contrast the mouse and rat in the context of several biological domains relevant to their use as appropriate models of aging: phylogeny/domestication, longevity interventions, pathology/physiology, and behavior/cognition. It is not the goal of this review to give a complete characterization of the differences between mice and rats, but to provide important examples of why using rats as well as mice is important to advance our understanding of the biology of aging. Extrachromosomal circular DNA (eccDNA) elements of chromosomal origin are known to be common in a number of eukaryotic species. However, it remains to be addressed whether genomic features such as genome size, the load of repetitive elements within a genome, and/or animal physiology affect the number of eccDNAs. Here, we investigate the distribution and numbers of eccDNAs in a condensed and less repeat-rich genome compared with the human genome, using Columba livia domestica (domestic rock pigeon) as a model organism. By sequencing eccDNA in blood and breast muscle from three pigeon breeds at various ages and with different flight behavior, we characterize 30,000 unique eccDNAs. We identify genomic regions that are likely hotspots for DNA circularization in breast muscle, including genes involved in muscle development. We find that although eccDNA counts do not correlate with the biological age in pigeons, the number of unique eccDNAs in a nonflying breed (king pigeons) is significantly higher (9-fold) than homing pigeons. Furthermore, a comparison between eccDNA from skeletal muscle in pigeons and humans reveals ∼9-10 times more unique eccDNAs per human nucleus. The fraction of eccDNA sequences, derived from repetitive elements, exist in proportions to genome content, that is, human 72.4% (expected 52.5%) and pigeon 8.7% (expected 5.5%). Overall, our results support that eccDNAs are common in pigeons, that the amount of unique eccDNA types per nucleus can differ between species as well as subspecies, and suggest that eccDNAs from repeats are found in proportions relative to the content of repetitive elements in a genome. In many animals, catabolic and anabolic periods are temporally separated. Migratory birds alternate energy expenditure during flight with energy accumulation during stopover. The size of the energy stores at stopover affects the decision to resume migration and thus the temporal organization of migration. We now provide data suggesting that it is not only the size of the energy stores per se that may influence migration scheduling, but also the physiological consequences of flying. In two subspecies of the northern wheatear Oenanthe oenanthe , a long-distance migrant, estimated energy stores at a stopover during autumn migration were positively related with both constitutive innate and acquired immune function, and negatively related with oxidative damage to lipids. In other words, migrants' physiological condition was associated with their energetic condition. Although time spent at stopover before sampling may have contributed to this relationship, our results suggest that migrants have to trade-off the depletion of energy stores during flight with incurring physiological costs. This will affect migrants' decisions when to start and when to terminate a migratory flight. The physiological costs associated with the depletion of energy stores may also help explaining why migrants often arrive at and depart from stopover sites with larger energy stores than expected. We propose that studies on the role of energy stores as drivers of the temporal organization of (avian) migration need to consider physiological condition, such as immunological and oxidative states. The relationship of genotypes to phenotypes can be modified by environmental inputs. Such crucial environmental inputs include metabolic cues derived from microbes living together with animals. Thus, the analysis of genetic effects on animals' physiology can be confounded by variations in the metabolic profile of microbes. Caenorhabditis elegans exposed to distinct bacterial strains and species exhibit phenotypes different at cellular, developmental, and behavioral levels. Here we reported metabolomic profiles of three Escherichia coli strains, B strain OP50, K-12 strain MG1655, and B-K-12 hybrid strain HB101, as well as different mitochondrial and fat storage phenotypes of C. elegans exposed to MG1655 and HB101 vs. OP50. We found that these metabolic phenotypes of C. elegans are not correlated with overall metabolic patterning of bacterial strains, but their specific metabolites. In particular, the fat storage phenotype is traced to the betaine level in different bacterial strains. HT115 is another K-12 E. coli strain that is commonly utilized to elicit an RNA interference response, and we showed that C. elegans exposed to OP50 and HT115 exhibit differences in mitochondrial morphology and fat storage levels. We thus generated an RNA interference competent OP50 (iOP50) strain that can robustly and consistently knockdown endogenous C. elegans genes in different tissues. Together, these studies suggest the importance of specific bacterial metabolites in regulating the host's physiology and provide a tool to prevent confounding effects when analyzing genotype-phenotype interactions under different bacterial backgrounds. Background: Military aircrews' health status is critical to their mission readiness, as they perform physically and cognitively demanding tasks in nontraditional work environments. Research Objectives: Our objective is to develop a broad operational risk assessment framework and demonstrate its applicability to health risks to aircrews because of airborne chemical exposure, considering stressors such as heat and exertion.Methods: Extrapolation of generic exposure standards to military aviation-specific conditions can include computation of risk-relevant internal dosimetry estimates by incorporating changes in breathing patterns and blood flow distribution because of aspects of the in-flight environment. We provide an example of the effects of exertion on peak blood concentrations of 1,2,4-trimethylbenzene computed using a physiologically based pharmacokinetic model.Results: Existing published collections on the effects of flight-related stressors on breathing patterns and blood flow address only a limited number of stressors. Although data exist that can be used to develop operational exposure limits specific to military aircrew activities, efforts to integrate this information in specific chemical assessments have been limited.Conclusions: Efforts to develop operational exposure limits would benefit from guidance on how to make use of existing assessments and expanded databases of the impact of environmental stressors on adult human physiology. Introduction: This study develops and demonstrates an analysis approach to understand the statistics of cumulative pressure exposure of the brain to repetitive blasts events.Materials and Methods: A finite element model of blast loading on the head was used for brain model biomechanical responses. The cumulative pressure exposure fraction (CPEF), ranging from 0.0 to 1.0, was used to characterize the extent and repetition of high pressures. Monte Carlo simulations were performed to generate repetitive blast cumulative exposures.Results: The blast orientation effect is as influential as the blast overpressure magnitudes. A 75° (from the side) blast orientation can produce CPEF values exceeding traumatic brain injury pressure thresholds >0.95 while, for the same blast overpressure, a 0° (front) blast orientation results in a CPEF <0.25. Monte Carlo results for different sequences reflecting notional operational and training environments show that both mean values and standard deviations of CPEF reach the statistically equilibrium state at a finite value of n exposures for each sequence.Conclusions: Statistical convergence of the brain pressure response metrics versus number of blasts for different exposures characterizes the transitions from "low" to "high" number of blasts and quantitatively highlights the differences between operational and training exposures. Introduction: Military and civil aviation have documented physiological episodes among aircrews. Therefore, continued efforts are being made to improve the internal environment. Studies have shown that exposures to many organic compounds present in emissions are known to cause a variety of physiological symptoms. We hypothesize that these compounds may reversibly inhibit acetylcholinesterase, which may disrupt synaptic signaling. As a result, neural proteins leak through the damaged blood-brain barrier into the blood and in some, elicit an autoimmune response.Materials and Methods: Neural-specific autoantibodies of immunoglobulin-G (IgG) class were estimated by the Western blotting technique in the sera of 26 aircrew members and compared with the sera of 19 normal healthy nonaircrew members, used as controls.Results: We found significantly elevated levels of circulating IgG-class autoantibodies to neurofilament triplet proteins, tubulin, microtubule-associated tau proteins (Tau), microtubule-associated protein-2, myelin basic protein, and glial fibrillary acidic protein, but not S100 calcium-binding protein B compared to healthy controls.Conclusion: Repetitive physiological episodes may initiate cellular injury, leading to neuronal degeneration in selected individuals. Diagnosis and intervention should occur at early postinjury periods. Use of blood-based biomarkers to assess subclinical brain injury would help in both diagnosis and treatment. Objective: This work describes the isolation of anti‐Leishmania amazonensis metabolites from Saururus cernuus (Saururaceae). Additionally, ultrastructural changes in promastigotes were evidenced by electron microscopy. Methods: The MeOH extract from the leaves of S. cernuus was subjected to bioactivity‐guided fractionation. Anti‐L. amazonensis activity of purified compounds was performed in vitro against promastigote and amastigote forms. Key findings: Bioactivity‐guided fractionation of the MeOH extract from the leaves of S. cernuus afforded two related tetrahydrofuran dineolignans: threo,threo‐manassantin A (1) and threo,erythro‐manassantin A (2). Compounds 1 and 2 displayed activity against promastigotes (EC50 of 35.4 ± 7.7 and 17.6 ± 4.2 μm, respectively) and amastigotes (EC50 of 20.4 ± 1.9 and 16.0 ± 1.1 μm, respectively), superior to that determined for the positive control miltefosine (EC50 of 28.7 ± 3.5 μm). Reduced cytotoxicity for host cells was observed for both compounds. Additionally, ultrastructural changes in promastigotes leading to an alteration of structural morphology were observed, as evidenced by electron microscopy. Furthermore, these compounds altered the morphology and physiology of the plasmatic membrane of L. amazonensis. Conclusions: The obtained results indicated that dineolignans 1 and 2 could be considered as a scaffold for the design of novel and selective drug candidates for the treatment of leishmaniasis. Frontotemporal dementia (FTD), a neurodegenerative disease broadly characterized by socioemotional impairments, includes three clinical subtypes: behavioral variant FTD (bvFTD), semantic variant primary progressive aphasia (svPPA) and non-fluent variant primary progressive aphasia (nfvPPA). Emerging evidence has shown emotional reactivity impairments in bvFTD and svPPA, whereas emotional reactivity in nfvPPA is far less studied. In 105 patients with FTD (49 bvFTD, 31 svPPA and 25 nfvPPA) and 27 healthy controls, we examined three aspects of emotional reactivity (physiology, facial behavior and subjective experience) in response to a sad film. In a subset of the sample, we also examined the neural correlates of diminished aspects of reactivity using voxel-based morphometry. Results indicated that all three subtypes of FTD showed diminished physiological responding in respiration rate and diastolic blood pressure; patients with bvFTD and svPPA also showed diminished subjective experience, and no subtypes showed diminished facial behavior. Moreover, there were differences among the clinical subtypes in brain regions where smaller volumes were associated with diminished sadness reactivity. These results show that emotion impairments extend to sadness reactivity in FTD and underscore the importance of considering different aspects of sadness reactivity in multiple clinical subtypes for characterizing emotional deficits and associated neurodegeneration in FTD.
