Hepatitis is the term used to describe inflammation in the liver. It is associated with a high rate of mortality, but the underlying disease mechanisms are not completely understood and treatment options are limited. We present a mathematical model of hepatitis that captures the complex interactions between hepatocytes (liver cells), hepatic stellate cells (cells in the liver that produce hepatitis-associated fibrosis) and the immune components that mediate inflammation. The model is in the form of a system of ordinary differential equations. We use numerical techniques and bifurcation analysis to characterize and elucidate the physiological mechanisms that dominate liver injury and its outcome to a healthy or unhealthy, chronic state. This study reveals the complex interactions between the multiple cell types and mediators involved in this complex disease and highlights potential problems in targeting inflammation in the liver therapeutically. Cancer therapeutics-related cardiac dysfunction (CTRCD) has emerged as a major cause of morbidity and mortality in cancer survivors. Effective clinical management of CTRCD is impeded by a lack of sensitive diagnostic and prognostic strategies. Circulating molecular markers could potentially address this need as they are often indicative of cardiac stress before cardiac damage can be detected clinically. A growing understanding of the underlying physiological mechanisms for CTRCD has inspired research efforts to identify novel pathophysiologically relevant biomarkers that may also guide development of cardio-protective therapeutic approaches. The purpose of this review is to evaluate current circulating biomarkers of cardiac stress and their potential role in diagnosis and management of CTRCD. Although environmental factors affecting adventitious root (AR) formation have been examined, how nutrient status affects ARs under waterlogging conditions remains unclear. In this study, plants' performance in responding to AR regulation based on nutrient supply was investigated in terms of plant morphology, physiology and AR traits. Results indicated that Cleistocalyx operculatus possesses higher waterlogging tolerance than Syzygium cumini according to the waterlogging tolerance coefficient, mainly because of the higher fresh weight, porosity and length of AR in C. operculatus. Nutrient supply treatment under a waterlogging condition significantly decreased the fresh weight, length, number, porosity, cortex area of AR and the ratio of cortex-to-stele area in both species relative to those in the waterlogging treatment, but significantly increased the activities and stele areas of AR, and leaf nutrient content. This result showed that nutrient supply caused variations in the morphological and anatomical structures of AR that were more beneficial to improve nutrient transportation than oxygen absorption under waterlogging conditions, supporting the nutrient-priority hypothesis. Moreover, nutrient supply under waterlogging conditions induced greater increase in stele area of ARs, fresh weight of the whole plant, total leaf area, leaf nitrogen level, total chlorophyll content, net photosynthesis rate and maximum photochemical quantum yield of PSII in S. cumini than in C. operculatus , suggesting that S. cumini can transport more nutrients and easily adapts to increase in nutrient supply under waterlogging conditions. Thus, S. cumini have better performance in extracting and utilizing nutrients in the water for plant growth. The findings showed that terrestrial arbor plants have physiological and microstructural mechanisms that respond to nutrient supply under waterlogging conditions and provide novel insights into the phytoremediation of eutrophic water bodies in wetland systems. Opsins are transmembrane proteins that have a covalent bond with a chromophore 11-cis-retinal in the intracellular region, opsins transiently bind to a G protein, which in turn is alternatively coupled to GTP or GDP. These characteristics allow opsins to function as photopigments and, after reacting with light, they launch the cascade of intracellular communication processes known as phototransduction. Commonly referred to as the starting point of the visual pathway, phototransduction as initiated by opsins, can also mediate physiological processes as varied as taxis, metabolic synchronization, and seasonal migration. This review analyzes such functions performed by opsins in crustaceans, a group with a huge diversity of species that, owing to their plasticity, have been able to invade habitats that other arthropods or terrestrial vertebrates have been unable to occupy. Our analysis emphasizes the way in which opsins take part in crustacean behavioral display under extreme habitat conditions as in the case of cave fauna), as well as in essential ontogenic events. The many functions of opsins have been scarcely investigated due to bias generated by their participation in visual processes. Light pollution at astronomical observatories is one of the main factors to be taken into account to preserve their scientific productivity and their useful lifetime. Using the Garstang–Cinzano model applied to the Visible Infrared Imaging Radiometer Suite (VIIRS) 2021 satellite radiance data, we have compared 28 sites, all hosting telescopes with apertures larger than 3 m, plus some additional selected sites. We computed and analysed five indicators of light pollution: radiance at zenith; averaged at 60° zenith distance; averaged over all the sky; averaged in the first 10° above the horizon; and horizontal irradiance. We found large variations of the values of the indicators, with a factor greater than 600 for the zenith artificial radiance between the least and most polluted major observatories. The results show that two-thirds of all large observatories have already surpassed the critical 10 per cent increase in radiance over the assumed natural levels. The results presented and the method described here can help to plan countermeasures in order to lower the impact of light pollution on observatories. These same methods can be also used to protect the night environment from the impact of artificial light (e.g. on biodiversity, on animal behaviour and physiology, on human health). Background and Aims Limiting maximum transpiration rate (TR) under high vapour pressure deficit (VPD) works as a water conservation strategy. While some breeding programmes have incorporated this trait into some crops to boost yields in water-limited environments, its underlying physiological mechanisms and genetic regulation remain unknown for faba bean (Vicia faba). Thus, we aimed to identify genetic variation in the TR response to VPD in a population of faba bean recombinant inbred lines (RILs) derived from two parental lines with contrasting water use (Mélodie/2 and ILB 938/2). Methods Plants were grown in well-watered soil in a climate-controlled glasshouse with diurnally fluctuating VPD and light conditions. Whole plant transpiration was measured in a gas exchange chamber that tightly regulated VPD around the shoot under constant light, while whole-plant hydraulic conductance and its components (root and stem hydraulic conductance) were calculated from dividing TR by water potential gradients measured with a pressure chamber. Key Results Although TR of Mélodie/2 increased linearly with VPD, ILB 938/2 limited its TR above 2.0 kPa. Nevertheless, Mélodie/2 had a higher leaf water potential than ILB 938/2 at both low (1.0 kPa) and high (3.2 kPa) VPD. Almost 90 % of the RILs limited their TR at high VPD with a break-point (BP) range of 1.5–3.0 kPa and about 10 % had a linear TR response to VPD. Thirteen genomic regions contributing to minimum and maximum transpiration, and whole-plant and root hydraulic conductance, were identified on chromosomes 1 and 3, while one locus associated with BP transpiration was identified on chromosome 5. Conclusions This study provides insight into the physiological and genetic control of transpiration in faba bean and opportunities for marker-assisted selection to improve its performance in water-limited environments. Sideroxydans sp. CL21 is a microaerobic, acid-tolerant Fe(II)-oxidizer, isolated from the Schlöppnerbrunnen fen. Since the genome size of Sideroxydans sp. CL21 is 21% larger than that of the neutrophilic Sideroxydans lithotrophicus ES-1, we hypothesized that strain CL21 contains additional metabolic traits to thrive in the fen. The common genomic content of both strains contains homologs of the putative Fe(II) oxidation genes, mtoAB and cyc2. A large part of the accessory genome in strain CL21 contains genes linked to utilization of alternative electron donors, including NiFe uptake hydrogenases, and genes encoding lactate uptake and utilization proteins, motility and biofilm formation, transposable elements, and pH homeostasis mechanisms. Next, we incubated the strain in different combinations of electron donors and characterized the fen microbial communities. Sideroxydans spp. comprised 3.33% and 3.94% of the total relative abundance in the peatland soil and peatland water, respectively. Incubation results indicate Sideroxydans sp. CL21 uses H2 and thiosulfate, while lactate only enhances growth when combined with Fe, H2, or thiosulfate. Rates of H2 utilization were highest in combination with other substrates. Thus, Sideroxydans sp. CL21 is a mixotroph, growing best by simultaneously using substrate combinations, which helps to thrive in dynamic and complex habitats. [ABSTRACT FROM AUTHOR]

Background The impact and management of subclinical hypoxia during hemodialysis is a significant medical challenge. As key determinants of O2 availability and delivery, proposed mechanisms contributing to hypoxia include ischemia, alkalemia and pulmonary leukocyte sequestration. However, no study has comprehensively investigated and compared these interrelated mechanisms throughout a typical hemodialysis treatment week. This study aimed to comprehensively assess the physiological mechanisms that contribute to hypoxia during hemodialysis. Methods In 76 patients, we measured arterial blood gases and pH at four time-points during hemodialysis (start, 15 min, 60 min, end) over the course of a standard treatment week. For the mid-week hemodialysis session, we additionally measured central hemodynamics (non-invasive cardiac output monitoring) and white blood cell count. Results Linear regression modelling identified changes in pH, but not central hemodynamics or white blood cell count, to be predictive of changes in PaO2 throughout hemodialysis (e.g. at 60 min, β standardized coefficient pH = 0.45, model R2 = 0.25, P  < .001). Alkalemia, hypokalemia, decreased calcium and increased hemoglobin–O2 affinity (leftward shift in the oxyhemoglobin dissociation curve) were evident at the end of hemodialysis. pH and hemoglobin–O2 affinity at the start of hemodialysis increased incrementally over the course of a standard treatment week. Conclusion These data highlight the important role of pH in regulating O2 availability and delivery during hemodialysis. Findings support routine pH monitoring and personalized dialysate bicarbonate prescription to mitigate the significant risk of alkalemia and subclinical hypoxia. [ABSTRACT FROM AUTHOR]

Plasma membrane (PM) proton-translocating adenosine triphosphatase (H+-ATPase) is a pivotal enzyme for plant growth and development that acts as a primary transporter and is activated by phosphorylation of the penultimate residue, threonine, at the C-terminus. Small Auxin-Up RNA family proteins maintain the phosphorylation level via inhibiting dephosphorylation of the residue by protein phosphatase 2C-D clade. Photosynthetically active radiation activates PM H+-ATPase via phosphorylation in mesophyll cells of Arabidopsis thaliana , and phosphorylation of PM H+-ATPase depends on photosynthesis and photosynthesis-related sugar supplementation, such as sucrose, fructose and glucose. However, the molecular mechanism and physiological role of photosynthesis-dependent PM H+-ATPase activation are still unknown. Analysis using sugar analogs, such as palatinose, turanose and 2-deoxy glucose, revealed that sucrose metabolites and products of glycolysis such as pyruvate induce phosphorylation of PM H+-ATPase. Transcriptome analysis showed that the novel isoform of the Small Auxin-Up RNA genes, SAUR30 , is upregulated in a light- and sucrose-dependent manner. Time-course analyses of sucrose supplementation showed that the phosphorylation level of PM H+-ATPase increased within 10 min, but the expression level of SAUR30 increased later than 10 min. The results suggest that two temporal regulations may participate in the regulation of PM H+-ATPase. Interestingly, a 15NO3− uptake assay in leaves showed that light increases 15NO3− uptake and that increment of 15NO3− uptake depends on PM H+-ATPase activity. The results opened the possibility of the physiological role of photosynthesis-dependent PM H+-ATPase activation in the uptake of NO3−. Temperature is a key variable in biological processes. However, a complete understanding of biological temperature adaptation is lacking, in part because of the unique constraints among different evolutionary lineages and physiological groups. Here we compared the genomes of cultivated psychrotolerant and thermotolerant methanogens, which are physiologically related and span growth temperatures from −2.5°C to 122°C. Despite being phylogenetically distributed amongst three phyla in the archaea, the genomic core of cultivated methanogens comprises about one-third of a given genome, while the genome fraction shared by any two organisms decreases with increasing phylogenetic distance between them. Increased methanogenic growth temperature is associated with reduced genome size, and thermotolerant organisms—which are distributed across the archaeal tree—have larger core genome fractions, suggesting that genome size is governed by temperature rather than phylogeny. Thermotolerant methanogens are enriched in metal and other transporters, and psychrotolerant methanogens are enriched in proteins related to structure and motility. Observed amino acid compositional differences between temperature groups include proteome charge, polarity and unfolding entropy. Our results suggest that in the methanogens, shared physiology maintains a large, conserved genomic core even across large phylogenetic distances and biology's temperature extremes. The use of biomarkers is undisputed in the diagnosis of primary myocardial infarction (MI), but their value for identifying MI is less well studied in the postoperative phase following coronary artery bypass grafting (CABG). To identify patients with periprocedural MI (PMI), several conflicting definitions of PMI have been proposed, relying either on cardiac troponin (cTn) or the MB isoenzyme of creatine kinase, with or without supporting evidence of ischaemia. However, CABG inherently induces the release of cardiac biomarkers, as reflected by significant cTn concentrations in patients with uncomplicated postoperative courses. Still, the underlying (patho)physiological release mechanisms of cTn are incompletely understood, complicating adequate interpretation of postoperative increases in cTn concentrations. Therefore, the aim of the current review is to present these potential underlying mechanisms of cTn release in general, and following CABG in particular (Graphical Abstract). Based on these mechanisms, dissimilarities in the release of cTnI and cTnT are discussed, with potentially important implications for clinical practice. Consequently, currently proposed cTn biomarker cut-offs by the prevailing definitions of PMI might warrant re-assessment, with differentiation in cut-offs for the separate available assays and surgical strategies. Sodium-glucose cotransporter 2 inhibitors (SGLT2i) are associated with a modest but significant increase in serum magnesium levels. This report describes improvement in serum magnesium and associated symptoms after initiating SGLT2i therapy in a patient with refractory hypomagnesemia. A 58-year-old woman presented with persistent hypomagnesemia refractory to oral magnesium supplements. She had history of type 2 diabetes mellitus, hypothyroidism, fibromyalgia, and degenerative disk disease. The cause of hypomagnesemia was attributed to excessive renal losses. Laboratory investigations revealed serum magnesium of 1.2 mg/dL with fractional excretion of magnesium of 8.9%. She was started on empagliflozin 10 mg daily. Within 4 weeks of therapy, her serum magnesium level corrected with symptomatic improvement, which was sustained a few weeks later. Subsequently, her oral magnesium supplements dose was reduced. SGLT2i has been shown to improve magnesium levels in patients with urinary magnesium wasting. Several mechanisms have been postulated, but the exact physiology remains unknown. SGLT2i have been efficacious for glycemic control, renal protection, decreasing the risk of atherosclerotic cardiovascular disease events, and cardiac mortality in patients with diabetes. In addition, renal and cardiac benefits are also demonstrated in patients without diabetes. This observation demonstrates that SGLT2i can improve the management of patients with otherwise intractable hypomagnesemia. [ABSTRACT FROM AUTHOR]

Recent advances in active materials, micro-fabrication techniques, ultra-miniaturized design approaches and hybrid device layouts form the foundations for emerging classes of unusual systems that can capture physiological signals from the human body in a physically imperceptible fashion at nearly any anatomical location, external on the skin or internal on vital organ systems. Of particular interest with these technologies are their thin geometries, flexible/stretchable physical properties and unique form factors, to enable conformal and gentle contacts with soft, curved and dynamic surfaces of living tissues. Commercial embodiments of skin-integrated devices with clinical-grade monitoring capabilities are just now becoming widely available not only in developed countries but also in the most resource-constrained areas of the globe. A specific example is in skin-like—sometimes referred to as 'epidermal'—wireless electronics for continuous monitoring of essential markers of physiological health status, with accuracy and reliability comparable to that of expensive, wired-based systems currently used with patients in intensive care units, but cost-effectively and applicable in any setting—in hospitals, health clinics, work environments or in the home. These technologies have the potential to revolutionize diagnostic and therapeutic approaches for the care of patients. Graduate education is an important aspect of the life of most academic scientists and a serious responsibility because it comes with the obligation to help students achieve their career and life goals. It can also be very fulfilling for the graduate mentor in terms of personal satisfaction and advancement of the research program. Learning to be a good major professor is an active process that depends on developing a formal framework of education and modifying that framework for each student based on past experiences and experimentation, advice from colleagues, and the individual personality of the student. Perhaps most important is for the graduate mentor to buy into the success and well-being of the student. Among the characteristics that a major professor could seek to instill in his or her students are critical and independent thinking, self-confidence, a thick skin, teamwork, laboratory skills and understanding, and the ability for hard work. Work to make science joyful by celebrating accomplishments, creating a fun environment in the lab, and stressing the societal value of science as compared to personal rewards or ambition. Apiculate yeasts belonging to the genus Hanseniaspora are predominant on grapes and other fruits. While some species, such as Hanseniaspora uvarum, are well known for their abundant presence in fruits, they are generally characterized by their detrimental effect on fermentation quality because the excessive production of acetic acid. However, the species Hanseniaspora vineae is adapted to fermentation and currently is considered as an enhancer of positive flavour and sensory complexity in foods. Since 2002, we have been isolating strains from this species and conducting winemaking processes with them. In parallel, we also characterized this species from genes to metabolites. In 2013, we sequenced the genomes of two H. vineae strains, being these the first apiculate yeast genomes determined. In the last 10 years, it has become possible to understand its biology, discovering very peculiar features compared to the conventional Saccharomyces yeasts, such as a natural and unique G2 cell cycle arrest or the elucidation of the mandelate pathway for benzenoids synthesis. All these characteristics contribute to phenotypes with proved interest from the biotechnological point of view for winemaking and the production of other foods. The house sparrow (Passer domesticus) is a valuable avian model for studying evolutionary genetics, development, neurobiology, physiology, behavior, and ecology, both in laboratory and field-based settings. The current annotation of the P. domesticus genome available at the Ensembl Rapid Release site is primarily focused on gene set building and lacks functional information. In this study, we present the first comprehensive functional reannotation of the P. domesticus genome using intestinal Illumina RNA sequencing (RNA-Seq) libraries. Our revised annotation provides an expanded view of the genome, encompassing 38592 transcripts compared to the current 23574 transcripts in Ensembl. We also predicted 14717 protein-coding genes, achieving 96.4% completeness for Passeriformes lineage BUSCOs. A substantial improvement in this reannotation is the accurate delineation of untranslated region (UTR) sequences. We identified 82.7% and 93.8% of the transcripts containing 5′- and 3′-UTRs, respectively. These UTR annotations are crucial for understanding post-transcriptional regulatory processes. Our findings underscore the advantages of incorporating additional specific RNA-Seq data into genome annotation, particularly when leveraging fast and efficient data processing capabilities. Objectives: Psychosocial working conditions of ready‐made garment (RMG) workers have been associated with poorer self‐reported health outcomes. However, no such research has been done with respect to physiological markers that are considered to reflect stress. We consequently aimed to investigate associations of psychosocial working conditions with such a marker, that is, hair cortisol, among RMG workers in Bangladesh. Methods: We conducted semi‐structured face‐to‐face interviews in labor colonies in the Mirpur area, Dhaka, Bangladesh, in February and March 2021 with individuals identifying as garment workers. The interview inquired after various workplace stressors and resources (i.e., workplace support, workplace bullying, vertical trust, beneficial leadership, work–family conflict, and financial issues including savings, debts, financial obligations, and financial support). In addition, hair samples of 2 cm length were collected from participants. Hair cortisol concentrations (HCC) were determined based on liquid chromatography–tandem mass spectrometry (LC‐MS/MS). Linear regression models were run to detect possible associations of workplace stressors and resources with HCC. Results: In total, data of 576 participants were included in the analysis (71.9% female, mean age = 25.9 years). Mean HCC was 4.4 pg/mg (standard deviation = 2.1 pg/mg). The sole variable significantly associated with increased HCC was "having to keep your job to support your children or spouse financially" (β = 0.28 [95% confidence interval 0.02–0.55]). Conclusions: The sole workplace stressor significantly associated with increased HCC was the necessity to keep one's job to support children or spouse financially. As online learning becomes increasingly popular in higher education, the quality of courses that utilize this modality is becoming a focus of inquiry. Quality Matters (QM) is a leading quality assurance organization that reviews online and hybrid (partially online, partially in-person) courses for standards of pedagogy and instructional design and certifies courses that sufficiently meet these standards. In this study, we examine student perceptions of course quality in a hybrid three-credit-hour animal science course that has been certified by QM. The class met twice a week for 1.25 h with one class period online and one in person. It consisted of 11 modules, each of which included learning content, learning activities, and assessments. Upon completion, 46 of 114 students completed a survey in which they rated the course on each of the 21 QM essential standards (Fifth edition). Descriptive analysis revealed that for 19 of the 21 specific review standards, 75% to 91% of students agreed or strongly agreed that the course reflected the best practice described in the standard. For the other two standards, over half of students (72%, 63%) agreed or strongly agreed that best practices were reflected in course design. Another way to examine the data is to collapse specific review standards into eight general review categories as specified by QM; the collapsed data revealed that 75% to 88% of students agreed or strongly agreed that the course design reflected the eight general course design standards. The percentage of students disagreeing that the course reflected each best practice was 11% or lower. Cronbach analysis to examine the internal consistency of the QM questionnaire (0.96), indicated instrument reliability and stability. A principal component analysis of the data conducted to further examine features and patterns of student responses revealed four primary factors that students rated highly (learning objectives, learner interaction and engagement, accessibility and usability, and clarity) that explained 78% of the data variance. This study demonstrates that the high quality of course design and delivery in a QM-certified course is clear to students. and provides justification for the investment in high-quality online and hybrid course design. In the future, we plan to compare student perceptions of course quality in a course that has not been QM-certified with one that has, as well as the impact of those revisions on student outcomes. This experiment compared ruminal, physiological, and productive responses of feedlot cattle receiving Yucca schidigera extract to replace or fed in conjunction with monensin + tylosin. Angus-influenced steers (n = 120) were ranked by body weight (BW ; 315 ± 3 kg) and allocated to 4 groups of 30 steers each. Groups were housed in 1 of 4 drylot pens (30 × 12 m) equipped with GrowSafe feeding systems (4 bunks/pen) during the experiment (day −14 to slaughter). On day 0, groups were randomly assigned to receive a diet containing (2 × 2 factorial): 1) no inclusion or inclusion of monensin + tylosin (360 mg and 90 mg/steer daily, respectively) and 2) no inclusion or inclusion of Y. schidigera extract (4 g/steer daily). Steers were slaughtered in 3 groups balanced by treatment combination (36 steers on day 114, 36 steers on day 142, and 48 steers on day 169). Blood was sampled on days 0, 28, 56, and 84, and the day before shipping to slaughter. On day 41, eight rumen-cannulated heifers (BW = 590 ± 15 kg) were housed with steers (1 pair/pen). Pairs rotated among groups every 21 d, resulting in a replicated 4 × 4 Latin square (n = 8/treatment combination) with 14-d washout intervals. Heifers were sampled for blood and rumen fluid at the beginning and end of each 21-d period. Monensin + tylosin inclusion decreased (P < 0.01) feed intake and improved (P = 0.02) feed efficiency of steers, but did not alter (P ≥ 0.17) steer BW gain or carcass merit traits. Inclusion of Y. schidigera extract did not impact (P ≥ 0.30) steer performance and carcass characteristics. Plasma glucose, insulin, insulin-like growth factor-I, and urea-N concentrations were not affected (P ≥ 0.16) by monensin + tylosin, nor by Y. schidigera extract inclusion in steers and heifers. Ruminal pH in heifers was increased (P = 0.04) by monensin + tylosin, and also by (P = 0.03) Y. schidigera extract inclusion. Rumen fluid viscosity was reduced (P = 0.04) by Y. schidigera extract, and rumen protozoa count was increased (P < 0.01) by monensin + tylosin inclusion. The proportion of propionate in the ruminal fluid was increased (P = 0.04) by monensin + tylosin, and tended (P = 0.07) to be increased by Y. schidigera extract inclusion. Hence, Y. schidigera extract yielded similar improvements in rumen fermentation compared with monensin + tylosin, but without increasing performance and carcass quality of finishing cattle. A study was conducted to evaluate the influence of in ovo feeding of clove and cinnamon on broilers. The experiment used 700 broiler (Ross 308) hatching eggs that were incubated at the recommended temperature of 37.8 °C. On day 17.5 of incubation, 100 eggs were randomly assigned to each of the following seven treatments: uninjected eggs (OE), eggs injected 0.5 mL distilled water (DH), 2 mg of clove, 4 mg of clove, 2 mg of cinnamon, 4 mg of cinnamon, and 3 mg of ascorbic acid (AC). During the posthatch period, the chicks were raised for 56 days. Data on physiological parameters, growth performance, and intestinal histomorphology were collected. Results revealed that the plasma triiodothyronine (T3) of AC and CV2 chicken was higher than the others. Additionally, the plasma malondialdehyde levels of the chickens of AC, CV2, and CM2 were improved significantly (P  < 0.05). The initial weights of CV2 birds were comparable with AC CV4, CM4, and CM2 birds but heavier than those of OE and DW. The bodyweight gain in the CV2 group was similar to AC, CV4, and CM2 groups but heavier than OE, DW, and CM4 birds. Feed intake of OE and DW groups was similar to AC, CV2, CV4, and CM2 but higher than CM4. The feed conversion ratio of OE and DW chickens was comparable but higher than the value obtained in chickens of other treatments. The intestinal morphology of the birds did not follow a particular trend. The study concluded that the in ovo injection of 2 mg of clove improved broiler birds' metabolic and antioxidant status at hatch. Context Hypoparathyroidism is the most frequent complication after total thyroidectomy (PT-hypoPTH). After 1 year, most patients recover parathyroid function; however, the implicated physiologic dynamics remain unknown. Vitamin D deficiency (VDD) is the main cause of secondary hyperparathyroidism. Whether this compensatory hyperparathyroidism could influence parathyroid function recovery (PFR) in the setting of PT-hypoPTH has not been studied. Objective This work aimed to evaluate the effect of preoperative VDD on PFR. Methods A retrospective study was conducted with a prospectively maintained database including patients undergoing a total thyroidectomy between May 2014 and June 2019. Preoperative vitamin D (25(OH)D) less than 20 mg/mL was defined as VDD. Intact PTH less than 14 pg/mL on postoperative day 1 was defined as PT-hypoPTH. Transient PT-hypoPTH displayed PFR within the first year (early recovery: < 30 days; protracted recovery: > 30 days) whereas definite PT-hypoPTH did not. Survival analysis evaluated the effect of preoperative VDD on PFR, and a binary logistic regression model identified associated factors. Results A total of 397 patients were identified. The observed rates of transient, protracted, and definite PT-hypoPTH were 32.9%, 15.1%, and 5.2%, respectively. Rates of VDD were higher in the early-recovery PT-hypoPTH group (55.2% vs 31.5%; P =.01). Preoperative VDD was associated with faster PFR (19 vs 35 days; P =.03) and behaved as a protective factor for protracted PT-hypoPTH (odds ratio 0.47; 95% CI, 0.25-0.881; P =.016) in the multivariable analysis. Conclusion Preoperative VDD could act as a preconditioning factor of the parathyroid glands prior to the surgical aggression exerted against them during surgery aiding PFR. Basic research studies and prospective clinical trials are needed to explain the underlying physiological mechanisms and to provide further evidence to improve clinical management. Contemporary evolutionary medicine has unified the idea of 'evolutionary mismatch', derived from the older idea of 'adaptive lag' in evolution, with ideas about the mismatch in development and physiology derived from the Developmental Origins of Health and Disease (DOHaD) paradigm. A number of publications in evolutionary medicine have tried to make this theoretical framework explicit. The integrative theory of mismatch captures how organisms track environments across space and time on multiple scales in order to maintain an adaptive match to the environment, and how failures of adaptive tracking lead to disease. In this review, we try to present this complex body of theory as clearly and simply as possible with the aim of facilitating its application in new domains. We introduce terminology, which is as far as possible consistent with earlier usage, to distinguish the different forms of mismatch. Mismatch in its modern form is a productive organizing concept that can help researchers articulate how physiology, development and evolution interact with one another and with environmental change to explain health outcomes. Catch-and-release (C&R) angling is a conservation-oriented practice intended to reduce the impact recreational angling has on fish populations. Even though most recreationally angled fish are released, little is known about how C&R angling impacts fish at the cellular or tissue level. As the first to explore the impacts of C&R angling on mRNA abundances, our study aimed to identify how the stress of angling influenced metabolism, acid–base regulation and cellular stress in the gills of lake trout (Salvelinus namaycush). Because gills are responsible for metabolic gas exchange, are crucial sites of acid–base homeostasis and respond to stressors quickly, we hypothesized that the relative mRNA abundance of genes related to these three physiological processes would be altered after angling. We took gill samples of live lake trout at 0, 2 or 48 h after fish were angled by rod and reel, and then used quantitative PCR (qPCR) to measure the relative abundance of nine candidate mRNA transcripts. Heat shock protein 70 (hsp70) mRNA levels significantly increased over 5-fold 2 h after angling, indicating a potential activation of a cytoprotective response. However, contrary to our hypothesis, we observed no change in the relative mRNA abundance of genes related to metabolism or acid–base regulation in response to C&R angling within a 48-h period. As C&R angling can negatively impact fish populations, further use of transcript-level studies will allow us to understand the impact C&R has on specific tissues and improve our knowledge of how C&R influences overall fish health. Cochlospermum fraseri ('Kapok', Bixaceae) is a deciduous tree widely distributed throughout semi-arid and monsoon tropical northern Australia, and an important species for ecological restoration in the region. We aimed to better understand the seed biology and ecology of C. fraseri to determine the mechanisms by which seed dormancy might be alleviated, and the conditions that support germination to inform the use of this species in restoration. Dormancy breaking treatments (wet heat, dry heat, scarification) commonly applied to species with physical seed dormancy (PY) were tested along with stratification at 5–35°C (nine treatments). Following dormancy alleviation, seeds were germinated at nine temperatures (5 to 40°C) and five water potentials (0 to −0.8 MPa) to understand environmental thresholds that regulate germination physiology in non-dormant seeds. A proportion of seeds (<0.3) lose dormancy naturally in response to warm (25 to 35°C) moist conditions, which dislodges the hypostase plug that prevents water uptake, whilst neither dry (≥100°C) nor wet (~100°C) heat were effective. Dormancy loss was also achieved by exposing seeds to concentrated (95–98% v/v) sulphuric acid for 3–7 hours, after which high proportions (>0.75) of germination were observed. Cochlospermum fraseri seeds possess PY, which is alleviated by seasonal temperatures that occur when soil moisture is high, allowing seeds to employ a risk-adverse strategy and maximize establishment success in episodic environments with stochastic rainfall events. The understanding of dormancy alleviation requirements gained here adds to our knowledge of PY worldwide and recruitment dynamics in the Australian monsoonal tropics and will aid land managers and restoration practitioners by informing both seeding sites and optimal time for in situ sowing as well as the potential capacity of this species to form a persistent soil seed bank. Anthropogenic-induced climate change is having profound impacts on aquatic ecosystems, and the resilience of fish populations will be determined by their response to these impacts. The northern Namibian coast is an ocean warming hotspot, with temperatures rising faster than the global average. The rapid warming in Namibia has had considerable impacts on marine fauna, such as the southern extension of the distribution of Argyrosomus coronus from southern Angola into northern Namibian waters, where it now overlaps and hybridizes with the closely related Namibian species, A. inodorus. Understanding how these species (and their hybrids) perform at current and future temperatures is vital to optimize adaptive management for Argyrosomus species. Intermittent flow-through respirometry was used to quantify standard and maximum metabolic rates for Argyrosomus individuals across a range of temperatures. The modelled aerobic scope (AS) of A. inodorus was notably higher at cooler temperatures (12, 15, 18 and 21°C) compared with that of A. coronus , whereas the AS was similar at 24°C. Although only five hybrids were detected and three modelled, their AS was in the upper bounds of the models at 15, 18 and 24°C. These findings suggest that the warming conditions in northern Namibia may increasingly favour A. coronus and promote the poleward movement of the leading edge of their southern distribution. In contrast, the poor aerobic performance of both species at cold temperatures (12°C) suggests that the cold water associated with the permanent Lüderitz Upwelling Cell in the south may constrain both species to central Namibia. ' QTL-hotspot ' is a genomic region on linkage group 04 (CaLG04) in chickpea (Cicer arietinum) that harbours major-effect quantitative trait loci (QTLs) for multiple drought-adaptive traits, and it therefore represents a promising target for improving drought adaptation. To investigate the mechanisms underpinning the positive effects of ' QTL-hotspot ' on seed yield under drought, we introgressed this region from the ICC 4958 genotype into five elite chickpea cultivars. The resulting introgression lines (ILs) and their parents were evaluated in multi-location field trials and semi-controlled conditions. The results showed that the ' QTL-hotspot ' region improved seed yield under rainfed conditions by increasing seed weight, reducing the time to flowering, regulating traits related to canopy growth and early vigour, and enhancing transpiration efficiency. Whole-genome sequencing data analysis of the ILs and parents revealed four genes underlying the ' QTL-hotspot ' region associated with drought adaptation. We validated diagnostic KASP markers closely linked to these genes using the ILs and their parents for future deployment in chickpea breeding programs. The CaTIFY4b-H2 haplotype of a potential candidate gene CaTIFY4b was identified as the superior haplotype for 100-seed weight. This paper reviews several aspects of immunoendocrinology and ecoimmunology in Brazilian species of anurans under investigation for more than a decade, including (1) patterns of annual covariation of circulating steroids, calling behavior and innate immunity, (2) endocrine and immune correlates of calling performance, (3) behavioral and physiological correlates of parasite load, (4) thermal sensitivity of immune function, and (5) endocrine and immunomodulation by experimental exposure to acute and chronic stressors, as well as to endocrine manipulations and simulated infections. Integrated results have shown an immunoprotective role of increased steroid plasma levels during reproductive activity in calling males. Moreover, a higher helminth parasite load is associated with changes in several behavioral and physiological traits under field conditions. We also found anuran innate immunity is generally characterized by eurythermy, with maximal performance observed in temperatures close to normal and fever thermal preferendum. Moreover, the aerobic scope of innate immune response is decreased at fever thermal preferendum. Experimental exposure to stressors results in increased corticosterone plasma levels and immune redistribution, with an impact on immune function depending on the duration of the stress exposure. Interestingly, the fate of immunomodulation by chronic stressors also depends in part on individual body condition. Acute treatment with corticosterone generally enhances immune function, while prolonged exposure results in immunosuppression. Still, the results of hormonal treatment are complex and depend on the dose, duration of treatment, and the immune variable considered. Finally, simulated infection results in complex modulation of the expression of cytokines, increased immune function, activation of the Hypothalamus-Pituitary-Interrenal axis, and decreased activity of the Hypothalamus-Pituitary-Gonadal axis, as well as reduced melatonin plasma levels, suggesting that anurans have a functional Immune-Pineal axis, homologous to that previously described for mammals. These integrated and complementary approaches have contributed to a better understanding of physiological mechanisms and processes, as well as ecological and evolutionary implications of anuran immunoendocrinology. Red oaks (Quercus sect. Lobatae) are a taxonomic group of hardwood trees, which occur in swamp forests, subtropical chaparral and savannahs from Columbia to Canada. They cover a wide range of ecological niches, and many species are thought to be able to cope with current trends in climate change. Genus Quercus encompasses ca. 500 species, of which ca. 80 make up sect. Lobatae. Species diversity is greatest within the southeastern USA and within the northern and eastern regions of Mexico. This review discusses the weak reproductive barriers between species of red oaks and the effects this has on speciation and niche range. Distribution and diversity have been shaped by drought adaptations common to the species of sect. Lobatae , which enable them to fill various xeric niches across the continent. Drought adaptive traits of this taxonomic group include deciduousness, deep tap roots, ring-porous xylem, regenerative stump sprouting, greater leaf thickness and smaller stomata. The complex interplay between these anatomical and morphological traits has given red oaks features of drought tolerance and avoidance. Here, we discuss physiological and genetic components of these adaptations to address how many species of sect. Lobatae reside within xeric sites and/or sustain normal metabolic function during drought. Although extensive drought adaptation appears to give sect. Lobatae a resilience to climate change, aging tree stands, oak life history traits and the current genetic structures place many red oak species at risk. Furthermore, oak decline, a complex interaction between abiotic and biotic agents, has severe effects on red oaks and is likely to accelerate species decline and fragmentation. Incorporating physiology into models of population dynamics will improve our understanding of how and why invasions succeed and cause ecological impacts, whereas others fail or remain innocuous. Targeting both organismal physiologists and invasion scientists, we detail how physiological processes affect every invasion stage, for both plants and animals, and how physiological data can be better used for studying the spatial dynamics and ecological effects of invasive species. We suggest six steps to quantify the physiological functions related to demography of nonnative species: justifying physiological traits of interest, determining ecologically appropriate time frames, identifying relevant abiotic variables, designing experimental treatments that capture covariation between abiotic variables, measuring physiological responses to these abiotic variables, and fitting statistical models to the data. We also provide brief guidance on approaches to modeling invasions. Finally, we emphasize the benefits of integrating research between communities of physiologists and invasion scientists. Plants activate a myriad of signaling cascades to tailor adaptive responses under environmental stresses, such as salinity. While the roles of exogenous karrikins (KARs) in salt stress mitigation are well comprehended, genetic evidence of KAR signaling during salinity responses in plants remains unresolved. Here, we explore the functions of the possible KAR receptor KARRIKIN-INSENSITIVE2 (KAI2) in Arabidopsis thaliana tolerance to salt stress by investigating comparative responses of wild-type (WT) and kai2 -mutant plants under a gradient of NaCl. Defects in KAI2 functions resulted in delayed and inhibited cotyledon opening in kai2 seeds compared with WT seeds, suggesting that KAI2 played an important role in enhancing seed germination under salinity. Salt-stressed kai2 plants displayed more phenotypic aberrations, biomass reduction, water loss and oxidative damage than WT plants. kai2 shoots accumulated significantly more Na+ and thus had a lower K+/Na+ ratio, than WT, indicating severe ion toxicity in salt-stressed kai2 plants. Accordingly, kai2 plants displayed a lower expression of genes associated with Na+ homeostasis, such as SALT OVERLY SENSITIVE (SOS) 1, SOS2, HIGH-AFFINITY POTASSIUM TRANSPORTER 1;1 (HKT1;1) and CATION–HYDROGEN EXCHANGER 1 (NHX1) than WT plants. WT plants maintained a better glutathione level, glutathione-related redox status and antioxidant enzyme activities relative to kai2 plants, implying KAI2's function in oxidative stress mitigation in response to salinity. kai2 shoots had lower expression levels of genes involved in the biosynthesis of strigolactones (SLs), salicylic acid and jasmonic acid and the signaling of abscisic acid and SLs than those of WT plants, indicating interactive functions of KAI2 signaling with other hormone signaling in modulating plant responses to salinity. BACKGROUND Emerging evidence has linked visit-to-visit, day-to-day and 24-h ABPM blood pressure variability (BPV) with cognitive impairment. Few studies have, however, considered beat-to-beat BPV. This study, therefore, evaluated the relationship between beat-to-beat BPV and cognitive function among community-dwellers aged 55 years and over. METHODS Data was obtained from the Malaysian Elders Longitudinal Research (MELoR) study, which employed random stratified sampling from three parliamentary constituencies within the Klang Valley. Beat-to-beat blood pressure (BP) was recorded using non-invasive BP monitoring (TaskforceTM, CNSystems). Low frequency (LF), high frequency (HF) and low-to-high frequency (LF:HF) ratio for BPV were derived using fast Fourier transformation. Cognition was evaluated using the Montreal Cognitive Assessment (MoCA) test, and categorized into normal aging, mild impairment and moderate-to-severe impairment. RESULTS Data from 1,140 individuals, mean age (SD) 68.48 (7.23) years, were included. Individuals with moderate-to-severe impairment had higher HF-BPV for systolic (SBP) and diastolic (DBP) blood pressure compared to individuals within the normal aging group [OR (95% CI) = 2.29 (1.62–3.24)] and [OR (95% CI) = 1.80 (1.32–2.45)], while HF-SBPV [OR (95% CI) = 1.41 (1.03–1.93)] but not HF-DBPV was significantly higher with mild impairment compared to normal aging after adjustments for potential confounders. Moderate-to-severe impairment was associated with significantly lower LF:HF-SBPV [OR (95% CI) = 0.29 (0.18–0.47)] and LF:HF-DBPV [OR (95% CI) = 0.49 (0.34–0.72)], while mild impairment was associated with significantly lower LF:HF-SBPV [OR (95% CI) = 0.52 (0.34–0.80)] but not LF:HF-DBPV [OR (95% CI) = 0.81 (0.57–1.17)], compared to normal aging with similar adjustments. CONCLUSION Higher HF-BPV, which indicates parasympathetic activation, and lower LF:HF-BPV, which addresses sympathovagal balance, were observed among individuals with moderate-to-severe cognitive impairment. Body size is arguably one of the most important traits influencing the physiology and ecology of animals. Shifts in animal body size have been observed in response to climate change, including in bumble bees (Bombus spp. [Hymenoptera: Apidae]). Bumble bee size shifts have occurred concurrently with the precipitous population declines of several species, which appear to be related, in part, to their size. Body size variation is central to the ecology of bumble bees, from their social organization to the pollination services they provide to plants. If bumble bee size is shifted or constrained, there may be consequences for the pollination services they provide and for our ability to predict their responses to global change. Yet, there are still many aspects of the breadth and role of bumble bee body size variation that require more study. To this end, we review the current evidence of the ecological drivers of size variation in bumble bees and the consequences of that variation on bumble bee fitness, foraging, and species interactions. In total we review: (1) the proximate determinants and physiological consequences of size variation in bumble bees; (2) the environmental drivers and ecological consequences of size variation; and (3) synthesize our understanding of size variation in predicting how bumble bees will respond to future changes in climate and land use. As global change intensifies, a better understanding of the factors influencing the size distributions of bumble bees, and the consequences of those distributions, will allow us to better predict future responses of these pollinators. In Drosophila spp. their often high number of annual generations, large population sizes and large amounts of standing genetic variation should predispose them to undergo contemporary adaptation to climatic warming. However, a number of laboratory experimental evolution studies in this group of organisms suggest strong limits on the rate and magnitude of contemporary thermal adaptation. Here, we explore this discrepancy by examining the potential for rapid evolutionary divergence between wild populations of Drosophila tripunctata Loew, 1862 from rural and urban sites. We performed a multi-generation common garden study and found evidence for the evolution of higher heat tolerance (critical thermal maximum) in flies from urban populations. We also detected evolutionary divergence in cold resistance (chill coma recovery time), with diminished cold resistance in flies from urban populations, although the effect was weaker than the shift in heat tolerance. Our study provides evidence of contemporary urban thermal adaptation, although the magnitude of phenotypic change lagged the magnitude of environmental temperature change across the urbanization gradient, suggesting potential limits on the evolution of urban thermal physiology. The reduction of genetic diversity due to genetic hitchhiking is widely used to find past selective sweeps from sequencing data, but very little is known about how spatial structure affects hitchhiking. We use mathematical modeling and simulations to find the unfolded site frequency spectrum left by hitchhiking in the genomic region of a sweep in a population occupying a 1D range. For such populations, sweeps spread as Fisher waves, rather than logistically. We find that this leaves a characteristic 3-part site frequency spectrum at loci very close to the swept locus. Very low frequencies are dominated by recent mutations that occurred after the sweep and are unaffected by hitchhiking. At moderately low frequencies, there is a transition zone primarily composed of alleles that briefly "surfed" on the wave of the sweep before falling out of the wavefront, leaving a spectrum close to that expected in well-mixed populations. However, for moderate-to-high frequencies, there is a distinctive scaling regime of the site frequency spectrum produced by alleles that drifted to fixation in the wavefront and then were carried throughout the population. For loci slightly farther away from the swept locus on the genome, recombination is much more effective at restoring diversity in 1D populations than it is in well-mixed ones. We find that these signatures of space can be strong even in apparently well-mixed populations with negligible spatial genetic differentiation, suggesting that spatial structure may frequently distort the signatures of hitchhiking in natural populations. Larger animals studied during ontogeny, across populations, or across species, usually have lower mass-specific metabolic rates than smaller animals (hypometric scaling). This pattern is usually observed regardless of physiological state (e.g. basal, resting, field, and maximally active). The scaling of metabolism is usually highly correlated with the scaling of many life-history traits, behaviors, physiological variables, and cellular/molecular properties, making determination of the causation of this pattern challenging. For across-species comparisons of resting and locomoting animals (but less so for across populations or during ontogeny), the mechanisms at the physiological and cellular level are becoming clear. Lower mass-specific metabolic rates of larger species at rest are due to (a) lower contents of expensive tissues (brains, liver, and kidneys), and (b) slower ion leak across membranes at least partially due to membrane composition, with lower ion pump ATPase activities. Lower mass-specific costs of larger species during locomotion are due to lower costs for lower-frequency muscle activity, with slower myosin and Ca++ ATPase activities, and likely more elastic energy storage. The evolutionary explanation(s) for hypometric scaling remain(s) highly controversial. One subset of evolutionary hypotheses relies on constraints on larger animals due to changes in geometry with size; for example, lower surface-to-volume ratios of exchange surfaces may constrain nutrient or heat exchange, or lower cross-sectional areas of muscles and tendons relative to body mass ratios would make larger animals more fragile without compensation. Another subset of hypotheses suggests that hypometric scaling arises from biotic interactions and correlated selection, with larger animals experiencing less selection for mass-specific growth or neurolocomotor performance. An additional third type of explanation comes from population genetics. Larger animals with their lower effective population sizes and subsequent less effective selection relative to drift may have more deleterious mutations, reducing maximal performance and metabolic rates. Resolving the evolutionary explanation for the hypometric scaling of metabolism and associated variables is a major challenge for organismal and evolutionary biology. To aid progress, we identify some variation in terminology use that has impeded cross-field conversations on scaling. We also suggest that promising directions for the field to move forward include (1) studies examining the linkages between ontogenetic, population-level, and cross-species allometries; (2) studies linking scaling to ecological or phylogenetic context; (3) studies that consider multiple, possibly interacting hypotheses; and (4) obtaining better field data for metabolic rates and the life history correlates of metabolic rate such as lifespan, growth rate, and reproduction. Females and males can exhibit striking differences in body size, relative trait size, physiology, and behavior. As a consequence, the sexes can have very different rates of whole-body energy use, or converge on similar rates through different physiological mechanisms. Yet many studies that measure the relationship between metabolic rate and body size only pay attention to a single sex (more often males), or do not distinguish between sexes. We present four reasons why explicit attention to energy-use between the sexes can yield insight into the physiological mechanisms that shape broader patterns of metabolic scaling in nature. First, the sexes often differ considerably in their relative investment in reproduction, which shapes much of life-history and rates of energy use. Second, males and females share a majority of their genome but may experience different selective pressures. Sex-specific energy profiles can reveal how the energetic needs of individuals are met despite the challenge of within-species genetic constraints. Third, sexual selection often pushes growth and behavior to physiological extremes. Exaggerated sexually selected traits are often most prominent in one sex, can comprise up to 50% of body mass, and thus provide opportunities to uncover energetic constraints of trait growth and maintenance. Finally, sex-differences in behavior such as mating-displays, long-distance dispersal, and courtship can lead to drastically different energy allocation among the sexes; the physiology to support this behavior can shape patterns of metabolic scaling. The mechanisms underlying metabolic scaling in females, males, and hermaphroditic animals can provide opportunities to develop testable predictions that enhance our understanding of energetic scaling patterns in nature. Sex‐based differences in physiological traits may be influenced by both evolutionary and environmental factors. Here we used male and female flies from >80 Drosophila species reared under common conditions to examine variance in a number of physiological traits including size, starvation, desiccation and thermal tolerance. Sex‐based differences for desiccation and starvation resistance were comparable in magnitude to those for size, with females tending to be relatively more resistant than males. In contrast thermal resistance showed low divergence between the sexes. Phylogenetic signal was detected for measures of divergence between the sexes, such that species from the Sophophora clade showed larger differences between the sexes than species from the Drosophila clade. We also found that sex‐based differences in desiccation resistance, body size and starvation resistance were weakly associated with climate (annual mean temperature/precipitation seasonality) but the direction and association with environment depended on phylogenetic position. The results suggest that divergence between the sexes can be linked to environmental factors, while an association with phylogeny suggests sex‐based differences persist over long evolutionary time‐frames. This article introduces the perspectivist interpretation of Nietzsche's On the Genealogy of Morality (GM), characterized by two core theses. According to the results thesis , the three treatises of GM introduce three types of critical results, respectively: psychological claims about the value of morality for the interests of various character types ; physiological claims about its value for the ' progress of the species '; and medical claims about its value for health. According to the distinction thesis , the critical results of GM are descriptive, while their reader-specific implications (and therapeutic effects) are normative. This interpretation illuminates the genealogical method of GM by answering four fundamental questions about (Q1) its historical method, (Q2) the history of morality that it reveals, (Q3) the critical results of GM, and (Q4) their intended therapeutic effects. Challenging the widespread assumption that the critical results of GM are evaluative claims, the perspectivist interpretation reveals the novelty of Nietzsche's " new demand" for a critique of values not only relative to his predecessors' approach to critique, but also to his own previous works. GM offers a critique that is grounded on factual claims, at once extramoral and external to (Christian) morality, and internal to its readers' evaluative perspectives, even those who remain beholden to (Christian) moral values. Bivalve molluscs have been the focus of behavioral and physiological studies for over a century, due in part to the relative ease with which their traits can be observed. The author reviews historical methods for monitoring behavior and physiology in bivalves, and how modern methods with electronic sensors can allow for a number of parameters to be measured in a variety of conditions using low-cost components and open-source tools. Open-source hardware and software tools can allow researchers to design and build custom monitoring systems to sample organismal processes and the environment, systems that can be tailored to the particular needs of a research program. The ability to leverage shared hardware and software can streamline the development process, providing greater flexibility to researchers looking to expand the number of traits they can measure, the frequency and duration of sampling, and the number of replicate devices they can afford to deploy. Aims In cardiomyocytes, acute disturbances to intracellular pH (pHi) are promptly corrected by a system of finely tuned sarcolemmal acid–base transporters. However, these fluxes become thermodynamically re-balanced in acidic environments, which inadvertently causes their set-point pHi to fall outside the physiological range. It is unclear whether an adaptive mechanism exists to correct this thermodynamic challenge, and return pHi to normal. Methods and results Following left ventricle cryo-damage, a diffuse pattern of low extracellular pH (pHe) was detected by acid-sensing pHLIP. Despite this, pHi measured in the beating heart (13C NMR) was normal. Myocytes had adapted to their acidic environment by reducing Cl−/ HCO 3 − exchange (CBE)-dependent acid-loading and increasing Na+/H+ exchange (NHE1)-dependent acid-extrusion, as measured by fluorescence (cSNARF1). The outcome of this adaptation on pHi is revealed as a cytoplasmic alkalinization when cells are superfused at physiological pHe. Conversely, mice given oral bicarbonate (to improve systemic buffering) had reduced myocardial NHE1 expression, consistent with a needs-dependent expression of pHi-regulatory transporters. The response to sustained acidity could be replicated in vitro using neonatal ventricular myocytes incubated at low pHe for 48 h. The adaptive increase in NHE1 and decrease in CBE activities was linked to Slc9a1 (NHE1) up-regulation and Slc4a2 (AE2) down-regulation. This response was triggered by intracellular H+ ions because it persisted in the absence of CO2/ HCO 3 − and became ablated when acidic incubation media had lower chloride, a solution manoeuvre that reduces the extent of pHi-decrease. Pharmacological inhibition of FAK-family non-receptor kinases, previously characterized as pH-sensors, ablated this pHi autoregulation. In support of a pHi-sensing role, FAK protein Pyk2 (auto)phosphorylation was reduced within minutes of exposure to acidity, ahead of adaptive changes to pHi control. Conclusions Cardiomyocytes fine-tune the expression of pHi-regulators so that pHi is at least 7.0. Hybrid larch (Larix decidua x L.kaempferi) is a fast growing species with high potential for reforestation outside the native range of European larch. Its superiority over its parents is well documented but still today little is known about the genetic background of heterosis and even less about the physiological mechanisms supporting it. In this paper, we hypothesize that a different allocation of biomass and/or growth efficiency could explain growth differences between larch species. To examine heterosis sensu stricto , we used a subset of progeny from an intra−/inter-specific diallel-mating design represented by connected pedigrees from the three species (Hybrid larch, European larch and Japanese larch). Six years after planting, we harvested the trees, excavated roots and measured dry weights of above- and belowground parts. The best allometric relationships were determined for predicting the biomass of each compartment. Heterosis for stem volume exceeded 108 per cent and 81 per cent for total tree biomass over mid-parent values. Hybrid superiority was visible for all biomass compartments and particularly for needles and branches, so that aboveground biomass heterosis reached 2.5 times that for the roots. In addition to a greater biomass, the hybrid benefited from a modified allocation of biomass with a slightly higher proportion of branch and needle biomasses and a reduced proportion of root biomass. Hybrid and European larches had a significantly greater growth efficiency than Japanese larch. Whereas the differential of productivity between European and Japanese larch did not increase with size, it greatly increased for hybrids with fine root mass increase. A heavier crown, a reduced proportion of root biomass and greater root efficiency in hybrid larch should enhance photosynthesis and reduce root respiration cost, providing more carbon-based resources for use in stem wood production. Host plant species will influence the population and physiological performance of insects. Frankliniella occidentalis is a well-known invasive pest commonly found on flowering plants. Herein, the population development of F. occidentalis was investigated on the flowers of different Rosa chinensis cultivars (Ruby, Love, Parade, Pink Peace, and Mohana), and the digestive enzyme activities in thrips were measured after feeding on these flowers. The developmental times of F. occidentalis from egg to adult were 10.07, 10.37, 11.64, 10.66, and 10.90 d on Ruby, Love, Parade, Pink Peace, and Mohana, respectively. Significant differences in fecundity were also observed, with the greatest fecundity levels of F. occidentalis on Ruby (82.96) and the lowest on Mohana (63.40). F. occidentalis showed the greatest R 0 on Ruby (43.57), followed by Love (36.46), Parade (33.00), Pink Peace (27.97), and Mohana (23.21). The r m showed a similar trend, with values of 0.156, 0.145, 0.141, 0.134, and 0.130, respectively. There were significant differences in digestive enzyme activities in F. occidentalis on different flowers, and different digestive enzymes showed different performance among these plants. The highest amylase and lipase activities in F. occidentalis were on Ruby, on which F. occidentalis had the fastest development rate and the highest R 0, whereas the highest trypsin activity was on Pink Peace. All three digestive enzymes in thrips showed the lowest activities on Mohana. The varied population development of F. occidentalis associated with R. chinensis cultivars may be related to their digestive enzyme performance, which plays important roles in nutrient metabolism and insect growth. Organoids are tridimensional structures cultured in the lab from stem cells. Organoids have been developed for many different organs, particularly those with Lgr5+ stem cells. The organs of the digestive system belong to this group of organs and to date, organoids have been established from all the digestive tract tissues and organs including liver, gall bladder and pancreas. Although most protocols to generate organoids have focused on human and mouse organs, organoids from domestic and production animals like dogs, cats, rabbits, pigs, chickens, turkeys, and horses are actively used in research. In this presentation, the principles of developing intestinal organoids will be covered, along with a summary of how they have been used to address intestinal function and disease in our lab and by others. In our lab, we use swine enteroids (intestinal organoids) to study the intestinal epithelial responses to nutrients and pathogens. We have explored differential responses of the intestinal epithelium to organic and inorganic sources of dietary zinc, and we are currently investigating the mechanism of pathogenesis of Lawsonia intracellularis, the causative agent of porcine intestinal adenopathy. We will also discuss methods for modeling intestinal physiology using enteroids, and the advantages and challenges of working with swine enteroids. [ABSTRACT FROM AUTHOR]

Sensorimotor experiences of going against the flow can affect the choices consumers make. Eight experiments show that consumers who experience the sensation of going against the flow pick alternatives that are normatively not preferred (experiments 1a and 1b). These effects are evident only when the sensations are dynamic and self-experienced (experiments 2a and 2b), subjective feelings are elicited (experiments 4a and 4b), and no other objective, external norm information is supplied (experiment 5). Experiences of going against the flow typically involve both movement and direction and are represented in memory schematically. Re- experiencing these sensations leads to the activation of this schematic representation and elicits a feeling-based behavioral disposition to do something different, or to go against one's initial inclination (experiment 3), leading participants to pick an option that is normatively not preferred.
