The distribution of container mosquitoes in relation to distances from forests was studied in temperate Japan. Mosquito larvae were collected between May and September in 4 years from tree holes, bamboo stumps, riverine rock pools, and artificial containers; sampling ranged spatially from the mountain forest across the deforested plain developed as agriculture and urban areas to the seacoast. Although tree holes, bamboo stumps, and artificial containers existed throughout the deforested plain area, 10 container species of 6 genera were found virtually only within 5 km from the nearest forest edge. Worldwide invasive Aedes albopictus (Skuse) and Aedes japonicus (Theobald) of Asian origin showed unique occurrence patterns different from other container species and from each other. Ae. japonicus was dominant in artificial containers in and near the forest but minor in forest natural containers and only occurred within 5 km from the forest. Ae. albopictus was minor in the forest irrespective of container types but not bound to the forest and dominant in natural and artificial containers throughout rural and urban areas. The 5-km range was designated as the circum-forest zone for container mosquitoes (except Ae. albopictus) in Japan, and an expanded concept, circum-boundary zone, is proposed. The widths of these zones primarily depend on the dispersal traits of mosquitoes. Whether the relation of Ae. albopictus and Ae. japonicus to forests we observed are common in the native and invasive ranges is discussed. Recent studies on invasive Aedes koreicus (Edwards 1917) have been conducted to elucidate the source population introduced to Europe. However, current information about the native range of Ae. koreicus is not consistent. The purpose of this study is to resolve confusion in the native distribution of Ae. koreicus by reviewing available literature from the first description of the species in its native range in 1917 to the first invasion in Europe in 2008. Aedes koreicus have been recorded in China, Japan, Korea, and eastern Russia. The 2 existing records of Ae. koreicus from Hokkaido, Japan, however, is likely due to the misidentification of 2 different morphologically similar species, Ae. koreicoides (Sasa, Kano & Hayashi 1950) and Aedes japonicus (Theobald 1901). Objectives: The purpose of this study is to review the short- and long-term outcomes of high-risk neonates with Ebstein anomaly treated with a newly developed rapid 2-stage Starnes procedure, which is aimed at reducing the size of the enlarged right side of the heart., Methods: Fifty-two foetuses with Ebstein anomaly were analysed in this study and divided into 2 groups. The control group comprised 25 foetuses, referred to us before 2008, and the study group was composed of 27 foetuses, referred to us after 2009. The right atrial area index was defined as high risk when it was >1.5. We applied our management approach to 6 high-risk neonates in the study group. This approach consisted of reducing the size of the right side of the heart through a 2-stage process: (i) right atrial plication without the use of a bypass and (ii) a Starnes procedure. Cox proportional hazards models were used to evaluate the effects of our management approach on the survival rates of the neonates., Results: The mean follow-up period was 7.5 ± 3.3 years. All 6 high-risk neonates in the study group survived. The overall hazard ratio was 0.12 (95% confidence interval of 0.03-0.43) in the study group as compared with the control group (P = 0.0007). A Fontan operation was completed in all but 1 case, with the remaining case awaiting a Fontan operation., Conclusions: We suggest that a rapid 2-stage Starnes approach can be effective in the treatment of high-risk neonates with Ebstein anomaly., (© The Author(s) 2020. The Asian mosquitoes, Aedes albopictus (Skuse) and Aedes japonicus japonicus (Theobald), have invaded North America, Europe, and other regions since the late 20th century. In invasive ranges, Ae. albopictus has well been recognized as urban, domestic species, whereas views about the macrohabitats of Ae. j. japonicus are inconsistent. Previous reports in Japan suggest the disappearance of Ae. j. japonicus from metropolises. However, container-mosquito larvae have not been inspected simultaneously for various macro and microhabitats in metropolises in Japan. The current study in Fukuoka City, a metropolis in southwest Japan, confirmed the absence of Ae. j. japonicus irrespective of macrohabitats (temples with graveyards, shrines, public graveyards, cultivated bamboo groves, and urban forests, all within the area densely inhabited by humans) and microhabitats (container types). In contrast, Ae. albopictus was dominant throughout the macro and microhabitats except forest tree holes rich with competitive species. Past records indicate the disappearance of Ae. j. japonicus from metropolitan Fukuoka within the last 70 yr. Based on careful examination of available evidence, we concluded that 1) both species benefit from human-made environments with artificial containers free from competitors, 2) Ae. j. japonicus disappeared due to hot, dry summer conditions facilitated by urban heat-island effects and a decrease in favored mammal hosts, and 3) Ae. albopictus has proliferated with higher tolerance to hot, dry climate and a wider blood-feeding host-range including humans. Gigantomastia is characterised by excessive breast growth and can occur as a rare, drug-induced adverse event. D-penicillamine is the most frequent cause of drug-induced gigantomastia. Only one case of gigantomastia due to bucillamine, an analogue of D-penicillamine, has been reported so far. We herein report a case of bucillamine-induced gigantomastia presenting with acute enlargement of the bilateral breasts and accessory breast tissue in the axillae 7 months after the start of bucillamine therapy. Awareness about this rare adverse event is important since bucillamine is still widely used in Japan and Korea.

Rugosin G, an ellagitannin trimer, was isolated from the water-soluble fraction of red rose petals, and its inhibitory activity against recombinant human histidine decarboxylase was investigated. Rugosin G showed potent inhibition compared to ellagitannin monomers and a dimer with macrocyclic structure (oenothein B), suggesting the potent inhibition of rugosin G was attributed to its linear oligomeric conformation. Aedes albopictus (Skuse) has expanded its distribution worldwide during the past decades. Despite attempts to explain and predict its geographic occurrence, analyses of the distribution of Ae. albopictus in the context of broad climatic regions (biomes) has not been performed. We analyzed climate conditions at its distribution sites in the range before the worldwide invasions (from the easternmost Hawaii through westernmost Madagascar) by using thermal and aridity-humidity indices descriptive of major biomes. A significant advantage of this approach is that it uses simple indices clearly related to the population dynamics of Ae. albopictus. Although Ae. albopictus has been regarded as a forest species preferring humid climate, in areas with significant human habitation, the distribution sites extended from the perhumid, rain forest zone to the semiarid, steppe zone. This pattern was common from the tropics through the temperate zone. Across the distribution range, there was no seasonal discordance between temperature and precipitation; at sites where winter prevents Ae. albopictus reproduction (monthly means<10°C), precipitation was concentrated in warm months (>10°C) under the Asian summer monsoon. Absence of the species in northern and eastern coastal Australia and eastern coastal Africa was not attributable solely to climate conditions. However, Asia west of the summer monsoon range was climatically unsuitable because of low precipitation throughout the year or in warm months favorable to reproduction (concentration of precipitation in winter). Objectives: This study examined the effects of palate depth, modifications of the arm shape, and anchor screw placement in the mid-palatal area on rapid maxillary expansion (RME) using finite element (FE) analysis., Materials and Methods: Three-dimensional FE models were constructed that included the maxilla (cortical and cancellous bone), maxillary sinus, maxillary first molar and first premolar, periodontal membrane, and an RME appliance with arms, bands, and anchor screws. The expansion screws were activated 0.2mm transversely., Results: The deepest palate model had the smallest lateral displacement of the tooth and expansion of the mid-palatal suture and the greatest strain of the arm among the models with different palate heights. The model with a larger diameter arm had the smallest arm strain among the models with various arm shapes. The model with an anchor screw had the greatest lateral displacement of the tooth and expansion of the mid-palatal suture among all models., Conclusions: For a deeper palate, the arm strain increased and the effect of RME decreased. Modified arm shapes such as a larger diameter arm, arms connected by a diagonal wire, a straight arm, and a shorter arm efficiently expanded the maxillary dental arch. Anchor screws increased the effect of RME, generated more and closer bodily movement of the tooth, and parallel expansion of the mid-palatal suture. The model with an anchor screw without arms decreased the displacement of the teeth compared to the models with arms, so the arms are necessary for effective RME., (© The Author 2014. Resorcinomycin (1) is composed of a nonproteinogenic amino acid, (S)-2-(3,5-dihydroxy-4-isopropylphenyl)-2-guanidinoacetic acid (2), and glycine. A biosynthetic gene cluster was identified in a genome database of Streptoverticillium roseoverticillatum by searching for orthologs of the genes responsible for biosynthesis of pheganomycin (3), which possesses a (2)-derivative at its N-terminus. The cluster contained a gene encoding an ATP-grasp-ligase (res5), which was suggested to catalyze the peptide bond formation between 2 and glycine. A res5-deletion mutant lost 1 productivity but accumulated 2 in the culture broth. However, recombinant RES5 did not show catalytic activity to form 1 with 2 and glycine as substrates. Moreover, heterologous expression of the cluster resulted in accumulation of only 2 and no production of 1 was observed. These results suggested that a peptide with glycine at its N-terminus may be used as a nucleophile and then maturated by a peptidase encoded by a gene outside of the cluster.

On a molecular level, cells sense changes in oxygen availability through the PHDs, which regulate the protein stability of the α-subunit of the transcription factor HIF. Especially, PHD3 has been additionally associated with apoptotic cell death. We hypothesized that PHD3 plays a role in cell-fate decisions in macrophages. Therefore, myeloid-specific PHD3(-/-) mice were created and analyzed. PHD3(-/-) BMDM showed no altered HIF-1α or HIF-2α stabilization or increased HIF target gene expression in normoxia or hypoxia. Macrophage M1 and M2 polarization was unchanged likewise. Compared with macrophages from WT littermates, PHD3(-/-) BMDM exhibited a significant reduction in TUNEL-positive cells after serum withdrawal or treatment with stauro and SNAP. Under the same conditions, PHD3(-/-) BMDM also showed less Annexin V staining, which is representative for membrane disruption, and indicated a reduced early apoptosis. In an unbiased transcriptome screen, we found that Angptl2 expression was reduced in PHD3(-/-) BMDM under stress conditions. Addition of rAngptl2 rescued the antiapoptotic phenotype, demonstrating that it is involved in the PHD3-mediated response toward apoptotic stimuli in macrophages., (© 2014 Society for Leukocyte Biology.)

Background: Orthodontic miniscrews placed in growing subjects often loosen during orthodontic treatment. The ability to place miniscrews, regardless of age, would be clinically beneficial., Objectives: To assess the effects of low-intensity pulsed ultrasound (LIPUS) on the stability of orthodontic miniscrews in growing rats., Materials/methods: The mobility of miniscrews after placement was recorded and the miniscrew-bone interface was examined histomorphometrically using tibiae of seven male Sprague-Dawley rats (aged 6 weeks). Field-emission scanning electron microscopic images were used to evaluate the bone-miniscrew interface, and a mobility test device was used to assess the stiffness of miniscrew placement. Fourteen custom-made miniscrews with 1.4mm diameters and 4.0mm lengths were placed in the right and left tibiae. LIPUS was used to stimulate right tibiae at the sites of miniscrew placement, and left tibiae were left untreated as controls., Results: Significantly lower mobility was observed in the LIPUS-treated group compared with the control group (P < 0.05). Histomorphometric evaluation indicated that bone-miniscrew adhesion was significantly better in the LIPUS-treated group than in the control group (P < 0.05)., Limitations: This in vivo study used tibiae rather than jaw bones because the jaw bones of 6-week-old rats were too small to allow miniscrew placement., Conclusions: LIPUS was able to increase the bone-miniscrew contact and reduce the mobility of miniscrews in growing subjects., Implications: LIPUS may accelerate the bone healing process after miniscrew placement in growing subjects and can reduce the latent period., (© The Author 2013. The impact of climate change on the distribution of Aedes albopictus (Skuse) was analyzed in northern Japan, where chronological distribution records are incomplete. We analyzed local climate data using linear regression of the thermal suitability index (TSI) for the mosquito and mean annual temperature as functions of time. In northern Japan, thermal conditions since the early 20th century have become increasingly suitable for Ae. albopictus, more as a result of decreasing coldness in the overwintering season than increasing warmth in the reproductive season. Based on recent discovery records of Ae. albopictus in the northern border range, we determined thermal criteria for estimating when its persistent establishment became thermally possible. Retrospective analyses indicated that those criteria were reached in most coastal lowlands of northern Honshu before the accelerated temperature increase after the mid-1980s and the first records of this species after 1990; at some sites, temperature criteria were reached during or before the early 20th century. Expansion of the thermally suitable range after 1990 was supported only for inland areas and the northernmost Pacific coast. The estimated expansion rate was approximately 26 km per decade. Our analyses also demonstrated the importance of local climate heterogeneity (apart from north-south or altitudinal temperature gradients) in determining the expansion pattern.

A putative indole diterpene biosynthetic gene cluster composed of eight genes was identified in a genome database of Phomopsis amygdali, and from it, biosynthetic genes of fusicoccin A were cloned and characterized. The six genes showed significant similarities to pax genes, which are essential to paxilline biosynthesis in Penicillium paxilli. Recombinants of the three putative prenyltransferase genes in the cluster were overexpressed in Escherichia coli and characterized by means of in vitro experiments. AmyG is perhaps a GGDP synthase. AmyC and AmyD were confirmed to be prenyltransferases catalyzing the transfer of GGDP to IGP and a regular di-prenylation at positions 20 and 21 of paxilline, respectively. AmyD is the first know example of an enzyme with this function. The Km values for AmyD were calculated to be 7.6 ± 0.5 μM for paxilline and 17.9 ± 1.7 μM for DMAPP at a kcat of 0.12 ± 0.003/s.

The purpose of this study was to analyse distal movements of molars in a force system using a trans-palatal arch (TPA), fixed to the maxillary first molar, and mini-implants placed at the palatal midline, considering the diagnostic standard for placement site in association with variation in upper molar locations, using finite element (FE) analysis. Three-dimensional FE models, divided by the differing direction of traction force, mesiodistal locations of the left and right molars, and the lateral location of the mini-implant were constructed. (1) When a traction force was fixed from the height of alveolar crest to the mini-implant placed at the middle of palate, the molars underwent bodily movement. (2) When the location of the mini-implant was moved to the left of the midline, the amount of distal movement of the left molar increased. When the mesiodistal locations of the left and right molars differed, the amount of distal movement of the molar located mesially was larger than that of the contralateral molar, even when the mini-implant was located on the midline.

Worldwide invasion of Aedes albopictus (Skuse) has become a serious concern in vector control. In temperate regions, an important goal has been to predict northern range limits after new introductions or global climate change. We propose a simple thermal suitability index (TSI) that can be calculated from monthly mean temperatures and may predict the northern distributional limits of Ae. albopictus. Analyses of known distributions revealed that TSI yields a common criterion (TSI = 1) below which establishment is thermally prohibited throughout the northern hemisphere. However, temperature conditions around northern limits differ among regions, depending on thermal continentality (the degree of seasonal temperature variation). Thermal continentality is high on the Asian Continent, intermediate in North America and Japan, and low (i.e., oceanic) in Europe. On the Asian Continent, there may be maximal winter coldness above which establishment is prohibited irrespective of summer reproduction. In Europe, establishment is possible under summer warmth lower than in other regions but there may be minimal summer warmth below which establishment is difficult despite high winter survival during mild winters. Among the thermal indices that have been commonly used as establishment criteria, the annual mean temperature of 10 degrees C proved to be a common criterion irrespective of thermal continentality, whereas coldest month means and warmth during the reproductive season (often expressed as accumulated degree-days above 11 degrees C) are strongly influenced by thermal continentality. Although annual mean temperatures can be a practical criterion, TSI was more informative in evaluating thermal suitability for this species.

This study used an experimental dental arch model to examine the orthodontic forces generated by a quadhelix appliance in terms of parallel expansion, fan expansion, or a combination of the two. Strain gauges were attached to experimental brass rods that represented the teeth arranged in the shape of an average dental arch to detect forces in the buccal, lingual, mesial, and distal directions. Orthodontic forces generated by different types of activation were compared by Scheffe's multiple test. The largest orthodontic force generated during parallel expansion was observed at the first molar in the buccal direction. When fan expansion was applied, significant orthodontic force was observed at the canine in the mesial and labial directions, whereas force in the mesial and lingual directions was noted at the first molar. When a combination of 3 mm parallel and 5 mm fan expansion was used, the forces generated at the canine and first and second premolar, and first molar were nearly equivalent. Depending on the type of malocclusion, the most appropriate expansion technique may be parallel or fan expansion or a combination of the two. When expanding the entire dental arch simultaneously, a combination of 3 mm parallel and 5 mm fan expansion may be the most suitable.

One notable complication of mini-implants that are used to provide anchorage in orthodontic treatment is loosening. The aim of this study was to evaluate the relationship between mini-implant mobility during the healing phase and the prognosis for implant stability. Twenty male Wistar rats (aged 20 weeks) were used. Drills with diameters of 0.8, 0.9, 1.0, and 1.1 mm were used to make pilot holes in the rat tibiae. The inserted mini-implants (diameter 1.4 mm; spearhead 1.2 mm; halfway between maximum and minimum 1.3 mm; length 4.0 mm) were subjected to an experimental traction of force for 3 weeks. Bone-to-implant contact (BIC) was observed histologically. Another 20 male rats (aged 20 weeks) underwent an identical procedure, and the stability of the mini-implants was measured using the Periotest before and after traction. The data were statistically analysed using Scheffé's test. The BIC ratios of the 0.9 and 1.0 mm groups were significantly greater than those of the other groups. The Periotest values measured 3 weeks after implant insertion were significantly lower (P < 0.05) than those measured at insertion, except in the 1.1 mm group. To obtain mini-implant stability, the hole diameter should be between 69 and 77 per cent of the diameter of the mini-implant. A significant decrease in the mobility of the mini-implants 3 weeks post-insertion implies a good prognosis for the subsequent mini-implant stability.

The aim of this study was to investigate the relationship between masseter muscle size and craniofacial morphology, focusing on the maxilla. Twenty-four patients (11 males and 13 females; mean age 27.6 ± 5.6 years) underwent cephalometric analyses. Ultrasonography was used to measure the cross-sectional area (CSA) of the masseter muscle and bite force was measured using pressure sensitive film. The results showed that CSA-relaxed was positively correlated with upper anterior face height (UAFH)/total anterior face height (TAFH) and negatively with lower anterior face height (LAFH)/TAFH and LAFH (P < 0.05). CSA-clenched was correlated positively with SN-palatal, FH-palatal, UAFH/TAFH, and lower posterior face height (LPFH)/total posterior face height (TPFH) and negatively with LAFH/TAFH, LAFH, upper posterior face height (UPFH)/TPFH, and UPFH (P < 0.05). Bite force was positively correlated with LPFH/TPFH and negatively with UPFH/TPFH (P < 0.05). As the masseter became larger, the anterior maxillary region tended to shift downwards relative to the cranial base, whereas the posterior region tended to shift upwards. The decrease in LAFH/TAFH and increase in LPFH/TPFH as the size of the masseter muscle increases may be influenced not only by the inclination of the mandibular plane but also by the clockwise rotation of the maxilla.

Variation in cold hardiness was studied in the laboratory for the nondiapausing eggs of nine Aedes (Stegomyia) species from eastern Asia and Pacific islands, ranging from the tropics to the cool-temperate zone. Species included were Aedes aegypti (L.), Aedes albopictus (Skuse), Aedes flavopictus Yamada, Aedes galloisi Yamada, Aedes hensilli Farner, Aedes palauensis Bohart, Aedes paullusi Stone and Farner, Aedes riversi Bohart and Ingram, and Aedes scutellaris (Walker). For species with distributions covering two or three climatic zones, at least one population was included from each zone, except for Ae. aegypti (subtropical populations were unavailable). Some eggs of four species confined to the tropical zone survived short exposure to freezing temperature (> or = -5 degrees C) when humidity was high. There were inter- and intraspecies variations in cold hardiness of nondiapausing eggs. Cold hardiness in each climatic zone differed in that populations of species with more northerly distributions were more cold-hardy than species with southerly distributions. In Ae. albopictus and Ae. flavopictus that cover three climatic zones, populations from cooler regions were more cold-hardy than populations from warmer regions. A possibility of southern species without diapausing eggs to colonize temperate eastern Asia was discussed.

We used two mitochondrial loci (nicotinamide adenine dinucleotide dehydrogenase subunit 4 and cytochrome oxidase II) and a nuclear locus (28S-D2 spacer) for a total of 1337 bp to evaluate the relationships among the four subspecies of Aedes (Finlaya) japonicus Theobald. Ae. j. japonicus was recently introduced into the United States and has been expanding rapidly. We also included in our analysis a morphologically very closely related species, Aedes (Finlaya) koreicus Edwards, as well as three more distantly related species: Aedes (Finlaya) togoi Theobald, Aedes (Finlaya) hatorii Yamada, and Aedes (Aedimorphus) vexans Meigen. We found that the four subspecies in the Ae. japonicus complex are genetically quite distinct but seem to form a monophyletic group that surprisingly also includes Ae. koreicus, suggesting the need for a taxonomic reconsideration of the group. We also found that the two southern subspecies are more closely related to each other than to any of the remaining subspecies or to Ae. koreicus and may indicate an ancient north-south split of the lineage. Considering the overlap between Ae. j. japonicus and Ae. koreicus, but the stronger association between Ae. koreicus and humans, we are surprised it also has not expanded from its original range. As a proactive reaction to this possibility, we designed and tested a DNA-based rapid assay to differentiate Ae. koreicus from some of the species with which it may be confused in the United States. These Aedes are putative vectors of several important viral encephalitides.

The expression of C/EBP homologous protein (CHOP), which is an endoplasmic reticulum (ER) stress-induced transcription factor, induces apoptosis. Our previous study demonstrated that lipopolysaccharide (LPS)-induced CHOP expression does not induce apoptosis, but activates a pro-IL-1beta activation process. However, the mechanism by which CHOP activates different pathways, depending on the difference in the inducing stimuli, remains to be clarified. The present study shows that LPS rapidly activates the ER function-protective pathway, but not the PERK pathway in macrophages. PERK plays a major role in CHOP induction, and other ER stress sensors-mediated pathways play minor roles. The induction of CHOP by LPS was delayed and weak, in comparison with CHOP induction by ER stress-inducer thapsigargin. In addition, LPS-pre-treatment or overexpression of ER chaperone, IgH chain binding protein (BiP), prevented ER stress-mediated apoptosis. LPS plus IFN-gamma-treated macrophages produce a larger amount of nitric oxide (NO) in comparison with LPS-treated cells. Treatment with the NO donor, SNAP (S-nitro-N-acetyl-dl-penicillamine), induces CHOP at an earlier period than LPS treatment. The depletion of NO retards CHOP induction and prevents apoptosis in LPS plus IFN-gamma-treated cells. We concluded that apoptosis is prevented in LPS-treated macrophages, because the ER function-protective mechanisms are induced before CHOP expression, and induction level of CHOP is low.

It is thought that the stress concentration at the root apex caused by orthodontic force induces root resorption. The purpose of this study was to investigate stress distribution at the root in cases of deviated root shapes using finite element models (FEMs). To clarify this, five three-dimensional FEMs divided by deviated root shape (normal, short, blunt, bent root apex, pipette shape) were constructed and, experimental orthodontic forces, applied in a vertical (intrusive) and horizontal (lingual) direction to the tooth axis. In the short-root model, significant stress was concentrated at the middle of the root. The blunt-shaped root model showed no significant stress concentration at the root. In the models with a bent or pipette-shaped root, significant stress was concentrated at the root apex. During orthodontic force application, stress concentration was observed in the root of the models with short, bent, and pipette-shaped roots, indicating that attention must be paid to root shape during orthodontic treatment.

Using lateral cephalograms and a jaw movement-recording system, the relationship between the masticatory movement path and dentofacial morphology was investigated in 17 subjects (9 males and 8 females, mean age 23.5 years) without a history of orthodontic treatment. The masticatory movement path was measured at the right and left lower first molar while the subjects chewed gum. The angle between the Frankfort horizontal plane and the masticatory axis (FH-masticatory angle), defined as the axis passing the opening and closing turning point on the sagittal masticatory path, was also measured. The correlation between the angular measurements derived from the lateral cephalogram and the FH-masticatory angle was then investigated. A positive correlation was observed in the FH-masticatory, occlusal plane (P < 0.05), and mandibular plane (P < 0.01) angles. Furthermore, it was found that the angle between the masticatory axis and the occlusal plane (69.1 +/- 4.2 degrees) remained constant even as the masticatory axis showed a tendency to incline forward as the mandibular plane angle became steeper; the rates of change of the FH-masticatory and the occlusal plane angles were approximately 1:1. This finding suggests that the masticatory movement path is closely associated with the occlusal plane.

A large number of complete microorganism genomes has been sequenced and submitted to the public database and then incorporated into our complete genome database, Genome Information Broker (GIB, http://gib.genes.nig.ac.jp/). However, when comparative genomics is carried out, researchers must be aware that there are protein-coding genes not confirmed by homology or motif search and that reliable protein-coding genes are missing. Therefore, we developed a protocol (Gene Trek in Prokaryote Space, GTPS) for finding possible protein-coding genes in bacterial genomes. GTPS assigns a degree of reliability to predicted protein-coding genes. We first systematically applied the protocol to the complete genomes of all 123 bacterial species and strains that were publicly available as of July 2003, and then to those of 183 species and strains available as of September 2004. We found a number of incorrect genes and several new ones in the genome data in question. We also found a way to estimate the total number of orthologous genes in the bacterial world.

The aim of this study was to investigate the initial stability of tapered orthodontic miniscrews (T-type screws) after placement, the necessity of a healing period, and the propriety of immediate loading. Twenty male Wistar rats with a mean age of 20 weeks were divided into two groups. In the immediate-loading groups, straight orthodontic miniscrews (S-type screws) and T-type screws (five rats each) underwent experimental traction force for 2 weeks (W) immediately after placement. In the healing groups (S- and T-type, five rats each), force was applied for 2 W after a 6-W healing period. The right tibia in each rat was identified as the test limb, while the left tibia in each rat was used as the control group, and underwent no experimental force during the experimental period. The screw-to-bone contact was observed histologically and the bone-screw contact ratio was calculated. Scheffe's test was performed to compare the bone-screw contact ratio in each group using statistical software package (SPSS 8.0 for Windows). In the control group, the bone-screw contact ratio improved from 34.8 +/- 16.0 to 74.8 +/- 12.0 per cent with S-type screws in proportion to the experimental period (2 to 8 W, respectively). With the T-type screws in the test group, there was no significant difference between the immediate-loading and healing groups. In the immediate-loading group, the bone-screw contact ratio with T-type screws was significantly greater (82.3 +/- 15.0 per cent) than with the S-type screws (33.3 +/- 11.8 per cent; P < 0.05), suggesting that T-type screws can be used for orthodontic anchorage immediately after placement.

Two mosquito species of the subgenus Stegomyia (genus Aedes) (Diptera: Culicidae) on the islands of Palau and Yap (Aedes dybasi Bohart and Aedes maehleri Bohart) are adapted to aquatic habitats occupied by Nepenthes pitcher plants. To reveal the origin of these pitcher plant mosquitoes, we attempted a molecular phylogenetic analysis with 11 Stegomyia species by using sequence data from mitochondrial cytochrome oxidase subunit I and 16SrRNA genes as well as the nuclear 28SrRNA gene. Ae. dybasi, a pitcher plant specialist, was sister to Aedes palauensis Bohart within the scutellaris group from the same islands. Ae. maehleri, an opportunistic pitcher plant mosquito, was in a distinct lineage related to the scutellaris group. The adaptation to pitcher plants could have occurred independently in these two species, and recent differentiation of the pitcher plant mosquito Ae. dybasi from the nonpitcher plant mosquito Ae. palauensis was suggested by a relatively small sequence divergence between these species. We also discuss the implications of this analysis for the phylogeny of some other Stegomyia species.

CHOP is a C/EBP family transcription factor involved in endoplasmic reticulum (ER) stress-mediated apoptosis. To determine if the ER stress pathway is involved in the pathogenesis of LPS-treated mouse lung injury, mice were given lipopolysaccharide (LPS) intraperitoneally. The mRNAs for activating transcription factor (ATF) 4 and X-box binding protein (XBP) 1, transcriptional activators of the CHOP gene, and that for CHOP were induced by or after the LPS treatment. Apoptosis induced by LPS treatment was suppressed in the lungs of Chop-knockout mice. Overexpression of CHOP induced apoptosis in a lung cancer-derived cell line. These results suggest that the ER stress pathway, involving CHOP, is activated and plays a role in the pathogenesis of septic shock lung.

In recent years, mandibular expansion has been increasingly performed in conjunction with orthodontic treatment. Lateral tipping of the molars associated with mandibular expansion should, however, be considered, because excessive expansion may result in excessive buccal tooth inclination, which may disturb the occlusal relationship. This study was conducted to quantitatively clarify molar movement during mandibular expansion using the Schwarz appliance to determine the permissible limit of mandibular expansion as a clinical index for inclination movement. Inclinations in the masticatory surface of the first molar and intermolar width were measured before expansion (T1), after expansion (T2), and before edgewise treatment (T3). Lower plaster models from 29 subjects treated with expansion plates were used and compared with models from 11 control subjects with normal occlusion. The average treatment change (T1-T2) in intermolar width was 5.42 mm (standard deviation 1.98), and the average angle of buccal tooth inclination was 10.16 degrees (standard deviation 3.83). No significant correlation was found between age prior to treatment and the treatment period when they were compared with the intermolar width increments and inclination angles. There was a significant positive correlation between retention duration and the amount of expansion. The regression coefficient of the angle of buccal tooth inclination during expansion to the increment of the intermolar width was approximately 0.2. This means that 1 mm of expansion is accompanied by 5 degrees of molar lateral tipping. This coefficient is clinically useful for estimating the permissible limit for mandibular expansion.

We found that a synthetic peptide fragment of MEPE containing the RGD and SGDG sequence stimulated new bone formation in vitro and in vivo., Introduction: Matrix extracellular phosphoglycoprotein (MEPE) was recently identified as a candidate for the phosphaturic hormone phosphatonin, which has been implicated in disturbed phosphate metabolism, rickets, and osteomalacia associated with X-linked hypophosphatemic rickets (XLH) and oncogenic hypophosphatemic osteomalacia (OHO). MEPE expression was predominantly found in osteoblasts, and mice deficient in a homolog of MEPE showed increased bone density, suggesting that MEPE produced in osteoblasts negatively regulates bone formation. In this study, we examined the effects of a synthetic 23mer peptide fragment of MEPE (AC-100, region 242-264) containing the RGD (integrin-binding) and SGDG (glycosaminoglycan-attachment) motif on bone formation in vitro and in vivo., Materials and Methods: The osteogenic activity of AC-100 was examined in organ cultures of neonatal mouse calvariae and in vivo by injecting AC-100 onto the calvariae of mice., Results: Histomorphometric examination showed that AC-100 stimulated new bone formation with increased numbers of osteoblasts in neonatal mouse calvariae in organ culture. In contrast, synthetic MEPE fragment peptides without either the RGD or SGDG motif failed to increase new bone formation. Repeated daily subcutaneous injections of AC-100 onto the calvariae in mice increased bone thickness and stimulated new bone formation as determined by the calcein double-labeling technique. However, peptides in which the RGD or SGDG sequence was scrambled did not stimulate new bone formation in vivo. AC-100 increased cell proliferation and alkaline phosphatase activity and activated focal adhesion kinase (FAK) and extracellular signal-regulated protein kinase (ERK) in human primary osteoblasts., Conclusion: Our results show that a synthetic peptide corresponding with the sequence of human MEPE fragment stimulates new bone formation with increased number of osteoblasts. The results also suggest that the RGD and SGDG motifs are critical to the osteogenic activity of AC-100, presumably through activating integrin signaling pathways in osteoblasts. The anabolic effects of AC-100 may be beneficial for bone diseases associated with decreased bone formation.

The characteristics of mandibular lateral displacement include lateral inclination of the occlusal plane and the differences between the right and left masticatory muscles. The aims of this investigation were to compare the mandibular stress distribution and displacement of the cervical spine using three-dimensional finite element models (3D FEM) to simulate masticatory movements and to clarify the association between morphological and functional characteristics and head posture. A symmetrical standard model was produced (model-A). Model-B had higher masticatory muscle strength on the left side, model-C had symmetrical masticatory muscle strength but the occlusal plane was inclined upwards towards the right and model-D had the occlusal plane inclined upwards towards the right with higher masticatory muscle strength on the left side. Model-A showed a completely symmetrical stress distribution pattern, while in model-B there was an uneven distribution in the mandible with higher stress on the left side. In addition, the stress distribution in the cervical spine was asymmetrical, showing displacement to the right. Model-C showed a similar mandibular tendency to model-B but the opposite tendency in the cervical spine. In model-D, the mandibular stress distribution was markedly asymmetrical, but almost symmetrical in the cervical spine with markedly decreased lateral displacement. These results suggest that lateral inclination of the occlusal plane and imbalance between the right and left masticatory muscles antagonistically act on displacement of the cervical spine, i.e. the morphological and functional characteristics in patients with mandibular lateral displacement may play a compensatory role in posture control.

The biomechanical effects on cervical vertebral columns (C1-C7) during mastication were calculated using a three-dimensional (3D) finite element method. To verify the biomechanical influences of vertical occlusal alteration to the cervical column, three finite element models (FEM) showing a normal (model A), a steep (model B), and a flat occlusal plane (model C) were constructed. The occlusal stress distribution showed various patterns for the three models; the stress extended to the anterior area as the occlusal plane became steeper. The plots of the stresses on the mid sagittal section of the cervical columns showed different patterns for the three models; the stress converged at the odontoid process in models A and B, whereas the stresses at C7 in model B tended to decrease compared with model A. Concentrated stress was observed at C5 in model C, supporting the hypothesis that vertical occlusal alteration could influence stress distribution in the cervical columns.

Background: Laminin-1, a multifunctional glycoprotein of the basement membrane, is thought to be important in embryogenesis, embryonic implantation, and placentation. We recently showed that serum IgG anti-laminin-1 autoantibodies (auto-Abs) are associated with recurrent first-trimester miscarriages. The present study assessed the clinical significance of anti-laminin-1 Abs with infertility, accompanied with or without endometriosis., Methods: Sixty-eight infertile patients who underwent laparoscopy or laparotomy and 39 healthy non-pregnant women were tested for IgG anti-laminin-1 Abs. The association between the Abs and endometriosis was analysed. The presence of laminin-1 mRNA was detected in endometriotic lesions., Results: Twenty infertile patients were positive for anti-laminin-1 Abs. The Ab levels in those patients were significantly higher than those in healthy non-pregnant women (P = 0.0005). The presence of the Abs was significantly associated with endometriosis in those patients (P = 0.0096). The Abs recognized a particular domain, i.e., the laminin-alpha1 chain G domain. mRNA encoding laminin-alpha1, -beta1, and -gamma1 chains was expressed in 90% of endometriotic lesions., Conclusions: IgG anti-laminin-1 Abs were significantly associated with endometriosis in infertile patients. The Abs might be clinically important in the development of autoimmune-mediated reproductive failures and the assessment of the Abs may provide a novel non-invasive diagnosis of endometriosis.

Maxillae and mandibles of males of 44 species of 12 mosquito genera and females of three autogenous genera and two partially autogenous species were examined under light microscopy and scanning electron microscopy. The maxillae and mandibles of male mosquitoes are delicate, tape-like structures with lengths characterizing genera or higher level classification units. Five patterns are recognized: (A) long maxillae and mandibles with mandibles longer than maxillae in Anopheles; (B) long maxillae and mandibles with maxillae longer than mandibles in Toxorhynchites; (C) short or intermediate lengths of maxillae with short mandibles in Aedes, Armigeres, Culex, Ochlerotatus, Orthopodomyia, and Uranotaenia; (D) short or intermediate length of maxillae with no mandibles in Mimomyia and Tripteroides; and (E) no maxillae and mandibles in Malaya and Topomyia. Maxillary and mandibular lengths of male mosquitoes show a positive correlation. Length of maxillae and mandibles of autogenous females are reduced to the same level as conspecific males. In contrast, females of partially autogenous species have complete maxillae and mandibles as in females of anautogenous species.

Since 1988 malaria epidemics have occurred in multiple sites in western Kenya highlands. Climatic variability has been associated with some of the recent epidemics. We examined influences of climatic factors on the distribution and abundance of three malaria vector species, Anopheles gambiae, Anopheles arabiensis, and Anopheles funestus in western Kenya and in the Great Rift Valley. Mosquito samples were collected from the lowland and highland areas with various climatic conditions. The three vector species were abundant in the lower part of western Kenya. An. arabiensis was not found in the areas above 1,400 m elevation in western Kenya Although An. gambiae and An. funestus were found in the sites above 1,700 m in western Kenya, their densities were < 1 per house. In the Great Rift Valley, An. gambiae was not recorded. An. funestus was more widely distributed than the other two species. A stepwise multiple regression analysis found that moisture index was the most important variable in shaping species composition of the An. gambiae complex. Relative abundance of An. gambiae was positively associated with moisture index, suggesting that An. gambiae is more adapted to moist climate. Seasonal differences in species composition were significant in western Kenya, and the proportion of An. funestus was higher in the dry season than the rainy season. Influence of temperature on vector density was significant for all three species. These results imply that climate changes alter the distribution and abundance of malaria vectors in future.

The biomechanical influences of head posture on the cervical column and craniofacial complex during masticatory simulation were quantified using three-dimensional (3D) finite element analysis (FEA). Three types of finite element model (FEM) were designed to examine relationships between the position of the head and malocclusion. Model A was constructed to have a standardized cervical column curve, model B a forward inclined posture, and model C a backward inclined posture. The results of the spinal displacements revealed that model B moved in a forward direction and model C in a backward direction during masticatory simulation. The stress distributions on the cervical column (C1-C7) for models A, B, and C showed differences; stress converged at the atlas in model A, high-level stresses were observed at the spinous processes of C6 and C7 in model C, and the stress converged at the anterior edge in the vertebral body of C4 of model B. Stress distribution on the occlusal plane and maxillofacial structure did not show absolute differences among the three models. Alteration of head posture was directly related to stress distribution on the cervical column, but may not always directly influence the occlusal state.

The purpose of this study was to estimate the increase in arch perimeter associated with mandibular lateral expansion. The mandibular expansion was simulated using a three-dimensional (3D) finite element method (FEM) and a computer graphics technique (3D simulation). The centre of rotation of molars during movement accompanied by lateral expansion was calculated using 3D FEM. The geometry of the model was determined using the mandibular bone of an East Indian skeletal specimen and 1 mm computer tomogram (CT) slices. The 3D set-up simulation was then conducted using 3D computer graphics instead of performing a manual set-up. Rotational movement was induced in the buccal segment, from the first premolar to second molar, in the 3D set-up model around the location of the centre of rotation (4.5 mm below the root apex of the first molar) derived from the FEM. According to 3D simulation, the model showed an opening space of 1.43 mm between the canine and first premolar, and thus a change in arch perimeter of 2.86 mm. The tip of the mesio-lingual cusp of the first molar moved 3.88 mm laterally, resulting in a change in inter-molar width of 7.76 mm. These values mean that a 1 mm increase in arch width resulted in an increase in arch perimeter of 0.37 mm. This result would be of value clinically for prediction of the effects of mandibular expansion.
