Fifty-four Angus-cross steers (297 kg +/- 12) were stratified by body weight (BW) to pens (six steers per pen) to determine the effects of supplemental Zn on posttransit growth performance and blood and muscle metabolites. Dietary treatments started 25 d before trucking: control (CON; analyzed 54 mg Zn/kg DM), industry (IND; CON + 70 mg supplemental Zn/kg DM), and supranutritional Zn (SUPZN; CON + 120 mg supplemental Zn/kg DM). Supplemental Zn was bis-glycinate bound Zn (Plexomin Zn; Phytobiotics North America, Cary, NC). On day 0, steers were loaded onto a commercial trailer and transported in 18 h (1,822 km). Individual BW was recorded on days -26, -25, -1, and 0 (pre-transit), 1 (posttransit), 6, 27, and 28. Blood was collected on days -1, 1, 6, and 27. Longissimus thoracis biopsies were collected on days -1, 1, and 28. Daily individual feed disappearance was recorded via GrowSafe bunks. Data were analyzed using Proc Mixed of SAS with fixed effect of diet and steer as the experimental unit (growth performance, blood: n = 18 steers per treatment; muscle: n = 12 steers per treatment). Individual initial BW was used as a covariate in BW analysis. Contrast statements to test linear, quadratic, and Zn effects were used to analyze performance and blood parameters. Repeated measures analysis was used for posttransit DMI recovery and weekly posttransit DMI and Zn intake with the repeated effect of time. MetaboAnalyst 5.0 was utilized for statistical analysis of day 1 (off truck) muscle metabolites. Plasma Zn linearly increased due to Zn on days 1, 6, and 27 (P = 0.01), and off-truck (day 1) serum lactate increased over day -1 by 20%, 0%, and 20% in CON, IND, and SUPZN, respectively (Quadratic: P = 0.01). α-Tocopherol (αT) in its natural form [2'R, 4'R, 8'R αT (RRR-αT)] is more bioactive than synthetic α-tocopherol (all rac-αT). All rac-αT is widely used in infant formulas, but its accretion in formula-fed infant brain is unknown.We sought to compare αT and stereoisomer status in infant rhesus macaques (Macaca mulatta) fed infant formula (RRR-αT or all rac-αT) with a reference group fed a mixed diet of breast milk and maternal diet.From 1 d after birth until 6 mo of age, infants (n = 23) were either nursery reared and exclusively fed 1 of 2 formulas by staff personnel or were community housed with their mothers and consumed a mixed reference diet of breast milk (69 mL/d at 6 mo) transitioning to monkey diet at ∼2 mo (MF; n = 8). Formulas contained either 21 μmol RRR-αT/L (NAT-F; n = 8) or 30 μmol all rac-αT/L (SYN-F; n = 7). Total αT and αT stereoisomers were analyzed in breast milk at 2, 4, and 6 mo and in monkey plasma and liver and 6 brain regions at 6 mo of age. α-Tocopherol transfer protein (α-TTP), lipoprotein αT, and urinary α-carboxyethyl-hydroxychroman (α-CEHC) were measured. One-way ANOVA with Tukey's post-hoc test was used for analysis.At study termination, plasma, liver, lipoprotein, and brain total αT did not differ between groups. However, the NAT-F-fed group had higher RRR-αT than the SYN-F-fed group (P 0.01) and the MF group (P 0.0001) in plasma (1.7- and 2.7-fold) and brain (1.5- and 2.5-fold). Synthetic αT 2R stereoisomers (SYNTH-2R) were generally 3- and 7-fold lower in brain regions of the NAT-F group compared with those of the SYN-F and MF groups (P 0.05). SYNTH-2R stereoisomers were 2-fold higher in MF than SYN-F (P 0.0001). The plasma percentage of SYNTH-2R was negatively correlated with the brain percentage of RRR-αT (r = -0.99, P 0.0001). Brain αT profiles were not explained by α-TTP mRNA or protein expression. Urine α-CEHC was 3 times higher in the NAT-F than in the MF group (P 0.01).Consumption of infant formulas with natural (NAT-F) compared with synthetic (SYN-F) αT differentially impacted brain αT stereoisomer profiles in infant rhesus macaques. Future studies should assess the functional implications of αT stereoisomer profiles on brain health.

Oral administration of indigestible markers and subsequent urine collection is a useful method to determine in vivo gastrointestinal tract (GIT) permeability in cattle for research purposes. However, urine sampling techniques often rely on total waste collection, which reduces the ability to perform more frequent sampling and obtain accurate volumes and sterile samples. An alternative is urethral catheterization, though the feasibility of this technique has not been thoroughly tested in preweaned Holstein heifer calves. The study objective was to develop a urethral catheter placement procedure in preweaned Holstein heifer calves for continuous and accurate urine collection to evaluate GIT permeability using an indigestible marker. Fifteen Holstein heifer calves had catheters placed at approximately 1 wk (8.0 ± 1.5 d) and 6 wk (40.0 ± 1.5 d) of age. During the procedure, calves were individually housed and restrained. The vulva was sterilized and then a sterile, lubricated speculum was inserted into the vagina. A sterile 0.09 cm diameter guidewire was guided into a lubricated, sterile 10 French Foley catheter. The catheter was inserted at approximately 5 through 7 cm into the urethral opening, guided into the bladder, and the catheter balloon was filled with 10 mL of water. The guidewire was removed, and urine flow confirmed correct placement before a 4-L urinary drainage bag was attached to the catheter. After catheterization (24 h), 1 L of chromium (Cr)-ethylenediaminetetraacetic acid was orally dosed to the calves. Calf health observations were made six times over a 48-h period, and any occurrence of vaginal discharge, tissue discharge in catheter, bleeding, inflammation, or abnormal urine was considered a localized reaction. The proportion of localized reactions for each age group was determined using Microsoft Excel, and the total Cr output was analyzed using PROC GLIMMIX. Localized reactions occurred for 20.0% of the 1-wk-old calves and 13.3% of the 6-wk-old calves. In the first 4 h, urine was collected every 15 min, and there were no overall Cr output differences (P = 0.38; 10.28 ± 3.21 mg Cr) when comparing 1- and 6-wk-old calves. However, 1-wk-old calves tended (P = 0.08) to have greater overall Cr output at 480 min (19.2%) and 1,440 min (41.9%) when compared with 6-wk-old calves. In summary, urinary catheterization is a viable urinary collection method for the determination of in vivo GIT permeability in preweaned Holstein heifer calves.Neonatal calves are highly susceptible to enteric disease during their first few weeks of life, and enteric disease is the leading cause of preweaning morbidity and mortality. A consequence of enteric disease is greater gastrointestinal tract (GIT) permeability in neonatal calves, which is also influenced by reduced intestinal maturity and environmental factors. Therefore, an accurate and precise method of evaluating GIT permeability in neonatal calves is necessary to develop appropriate treatment and mitigation strategies. The oral administration of indigestible markers and measurement of their presence in urine is an accepted method to determine the total GIT permeability in mature heifers. However, current urine collection methods in preweaned heifer calves may not be reliable. Therefore, the study objective was to develop a urinary catheterization method to collect urine accurately and precisely for the in vivo determination of GIT permeability in 1- and 6-wk-old Holstein heifer calves. It was determined that the urinary catheterization procedure and collection system developed in this study were viable and could be applied when evaluating GIT permeability in preweaned Holstein heifer calves using orally dosed indigestible markers.

Oral microbiota play a prominent role in canine periodontal disease and wet foods are often blamed for poor oral health, but canine oral microbial communities have been poorly studied. We aimed to determine differences in oral health measures, breath odor, and oral microbiota populations of dogs fed wet or dry food. Twelve adult dogs fed either a commercial dry (extruded) or commercial wet (canned) food for 6 wk were studied. Breath samples were measured for sulfur compounds, teeth were scored for plaque, calculus, and gingivitis by a blinded veterinary dentist, salivary pH was measured, and supragingival (SUP) and subgingival (SUB) plaque samples were collected for microbiota analysis. Plaque DNA was extracted and Illumina sequencing was conducted. Phylogenetic data were analyzed using the CosmosID bioinformatics platform and SAS 9.4, with P0.05 being significant and P0.10 being trends. Plaque coverage tended to be higher (P0.10) in dogs fed wet vs. dry food, but other oral health scores were not different. Dogs fed dry food had higher (P0.05) salivary pH and lower (P0.05) breath sulfur concentrations than those consuming wet food. Bacterial alpha diversity was higher in SUP than SUB samples, and a clear separation in beta diversity was observed between sample sites on principal coordinates analysis (PCoA) plots. In SUP samples, dogs fed wet food had a higher alpha diversity than dogs fed dry food, with PCoA plots showing a separation between wet and dry food. Relative abundances of Firmicutes, Synergistetes, and 10 bacterial genera were different (P0.05) in SUB samples of dogs fed wet vs. dry food. Relative abundances of Fusobacteria and over 20 bacterial genera were different (P0.05) in SUP samples of dogs fed wet vs. dry food. In general, oral health-associated bacterial taxa (Pasteurella, Capnocytophaga, Corynebacterium) were higher, while bacteria associated with poor oral health (Fretibacterium fastidiosum, Filifactor alocis, Treponema medium, Tannerella forsythia, Porphyromonas canoris, Porphyromonas gingivalis) were lower in dogs fed dry food. Such shifts in the oral microbiota may impact periodontal disease risk, but longer dietary intervention studies are required to confirm their role in the disease process. Our results suggest that dogs fed dry extruded foods have lower breath odor and tooth plaque buildup and an oral microbiota population more closely associated with oral health than dogs fed wet canned foods.Canned wet foods are often blamed for poor oral health in dogs, but comparison between wet and dry foods is not commonly done. Mixed parity sows (n = 3,451; PIC, Hendersonville, TN; parities 2 through 9) and their litters were used to evaluate the effects of essential fatty acid (EFA) intake on sow reproductive performance, piglet growth and survivability, and colostrum and milk composition. Our hypothesis, like observed in earlier research, was that increasing linoleic acid (LA) and α-linolenic acid (ALA) would improve sow and litter performance. At approximately day 112 of gestation, sows were randomly assigned within parity groups to 1 of 4 corn-soybean meal-wheat-based lactation diets that contained 0.5 (Control) or 3% choice white grease (CWG), 3% soybean oil (SO), or a combination of 3% soybean oil and 2% choice white grease (Combination). Thus, sows were provided diets with low LA and ALA in diets with CWG or high LA and ALA in diets that included soybean oil. Sows received their assigned EFA treatments until weaning and were then fed a common gestation and lactation diet in the subsequent reproductive cycle. Average daily feed intake during the lactation period increased (P0.05) for sows fed the Combination and CWG diets compared with sows fed the Control or SO diet. However, daily LA and ALA intakes of sows fed the Combination and SO diets were still greater (P0.05) than those of sows fed 0.5 or 3% CWG. Overall, sows consuming high EFA from the Combination or SO diets produced litters with heavier (P0.05) piglet weaning weights and greater (P0.05) litter ADG when compared with litters from sows fed diets with CWG that provided low EFA. Despite advantages in growth performance, there was no impact of sow EFA intake on piglet survivability (P0.10). Additionally, lactation diet EFA composition did not influence sow colostrum or milk dry matter, crude protein, or crude fat content (P0.10). However, LA and ALA content in colostrum and milk increased (P0.05) in response to elevated dietary EFA from SO. There was no evidence for differences (P0.10) in subsequent sow reproductive or litter performance due to previous lactation EFA intake. In conclusion, increased LA and ALA intake provided by soybean oil during lactation increased overall litter growth and pig weaning weights, reduced sow ADFI, but did not affect piglet survivability or subsequent performance of sows.Supplemental fat sources are an effective and widely accepted strategy to increase energy density of sow lactation diets that can also provide essential fatty acids such as linoleic acid (LA) and α-linolenic acid (ALA). Currently, the effects of supplemental LA and ALA provided shortly before farrowing on colostrum and milk composition are not fully understood. Additionally, the influence of elevated LA and ALA provided in sow lactation diets on litter growth and survivability responses has not been extensively evaluated. Therefore, this trial was conducted to evaluate the effects of fat sources providing low and high LA and ALA intake on sow performance, litter growth and survivability, colostrum and milk composition, and subsequent reproductive performance. Overall, sows consuming diets with high LA and ALA provided by soybean oil produced litters with heavier piglet weaning weights and greater litter average daily gain when compared with sows consuming diets with low LA and ALA content. Increasing LA and ALA by added soybean oil also increased their content in colostrum and milk. However, there was no influence of sow LA and ALA intake on litter survivability or subsequent reproductive performance of sows.

As the hemp industry continues to develop in the United States, there is an increasing interest in feeding byproducts of industrial hemp production to livestock. A completely randomized design experiment using crossbred finishing heifers (initial body weight [BW] ± SE = 494 ± 10 kg) was conducted to determine the effects of feeding hempseed cake in a corn-based finishing diet (10% forage) formulated to meet or exceed ruminally degradable and metabolizable protein requirements on growth performance, carcass characteristics, feeding behavior, and plasma parameters. Dietary treatments were the inclusion of 20% (dry matter [DM] basis) of dried corn distillers grains plus solubles (DDGS, n = 16) or hempseed cake (HEMP, n = 15). Cattle were housed in two pens, had ad libitum access to feed and water, and individual intakes and feeding behavior were monitored using the Insentec feeding system. Cattle were fed treatment diets for 111 d, and every 14 d BW were measured and blood samples were collected. Blood plasma was analyzed for glucose, urea nitrogen, and individual amino acids, and results were analyzed using repeated-measures analysis in SAS. Final BW, average daily gain, gain:feed, and hot carcass weight decreased (P ≤ 0.05) by 2.3%, 7.7%, 7.7%, and 2.6%, respectively, in heifers fed the HEMP diet than in heifers fed the DDGS diet. Net energy for maintenance and gain (Mcal/kg of feed, DM basis), estimated based on heifer intake and performance, were greater (P = 0.02) for the DDGS diet than for the HEMP diet. All other performance and carcass characteristics were not different (P ≥ 0.20) between treatments. Heifers fed the HEMP diet had greater (P 0.05) plasma urea nitrogen concentration in samples from each collection day compared with heifers fed the DDGS diet, although there was a treatment-by-day interaction (P 0.01) because of variability in the magnitude of treatment differences over time. Plasma glucose concentration was not influenced (P = 0.17) by dietary treatment. Plasma concentrations of total amino acids, nonessential amino acids, and essential amino acids were not different between treatments (P ≥ 0.09), although there were several interactions between treatment and day (P ≤ 0.04) for individual amino acids. These data suggest that hempseed cake has a lower net energy for maintenance and gain relative to DDGS when adequate metabolizable protein is supplied, while still providing adequate nutrition to support the acceptable performance of finishing cattle.This experiment evaluated the effects of dietary inclusion of hempseed cake in comparison with dried corn distillers grains plus solubles (DDGS) on growth performance, feeding behavior, carcass characteristics, and blood parameters in finishing heifers. Angus-crossbred heifers were assigned randomly to one of two treatments (20% hempseed cake [HEMP] or 20% DDGS; dry matter basis) and were fed for 111 d until slaughter. Heifers receiving the DDGS treatment had greater final body weights, average daily gain, gain efficiency, dietary concentration of net energy for maintenance and gain, and carcass weight than heifers fed the HEMP treatment. All other carcass characteristics, as well as feeding behavior, were not influenced by treatment. Plasma urea nitrogen was greater in heifers fed the HEMP diet compared with the DDGS diet, while glucose was not influenced by treatment. Several plasma amino acid concentrations were influenced by treatment. Although the inclusion of hempseed cake decreased growth performance, it could be a viable alternative feed source for cattle.

This study was aimed to investigate the effects of dietary arctiin (ARC) supplementation (100, 200, and 400 mg/kg) on the growth performance and immune response of broilers after a Salmonella pullorum (S. pullorum) challenge, and we conducted in vitro antibacterial test to explore the bacteriostatic mechanism of ARC. The in vivo trial was randomly assigned to six groups: noninfected control (NC) group and positive control (PC) group received a basal diet; TET group, received a basal diet supplemented with 100 mg/kg chlortetracycline; ARC100, ARC200, and ARC400 groups received a basal diet containing 100, 200, and 400 mg/kg ARC, respectively. From days 14 to 16, all birds (except the NC group) were infected with 1 mL (1 × 108 CFU per mL) fresh S. pullorum culture by oral gavage per day. In vivo results showed that dietary supplementation of 200 mg/kg ARC significantly increased average daily gain (P0.05) and decreased feed-to-gain ratio of broilers vs. the PC group during days 15 to 28 after being challenged with S. pullorum (P0.05). The jejunal crypt depth (CD) was decreased by supplementing 100 or 200 mg/kg ARC in diets compared with PC birds at day 19 (P0.05). The jejunal villi height (VH) was increased by supplementing 100, 200, or 400 mg/kg ARC in diets compared with PC birds at day 28 (P0.05). Besides, dietary supplementation of 200 mg/kg ARC increased the jejunal VH to CD ratio than the PC group both at days 19 and 28 (P0.05). Notably, the broilers had lower serum lipopolysaccharide and diamine oxidase levels in the ARC100 and ARC200 groups at day 28 than those in the PC group (P0.05). Furthermore, in comparison to PC birds, the birds in ARC groups (100, 200, and 400 mg/kg) had higher serum contents of IgM and IL-10, and the birds in the ARC200 group had higher serum contents of IgA at day 19 (P0.05). At day 28, the birds in ARC groups (100, 200, and 400 mg/kg) had lower serum contents of IL-8, and the birds in the ARC200 group had lower serum contents of IFN-γ compared with PC birds (P0.05). The in vitro experiment showed that ARC significantly inhibited the biofilm formation and adhesion of S. pullorum (P0.05). Metabonomics analysis revealed that ARC can restrain the formation of the biofilm by affecting a variety of metabolic pathways of S. pullorum. Therefore, dietary supplementation of 200 mg/kg ARC might be a potential way to substitute antibiotics to control S. pullorum infection in broilers.Pullorosis caused by Salmonella pullorum (S. pullorum) is a severe contagious disease and could cause great economic loss to the poultry industry. Antibiotics are usually used to control pullorosis, while prolonged use of antibiotics has led to the emergence of multidrug-resistant bacterial strains. Therefore, it is necessary to find safer and more effective alternatives to substitute antibiotics. In this study, we established a model of S. pullorum infection in broilers and conducted in vitro antibacterial test to explore the preventive effect and mechanisms of dietary arctiin (ARC) supplementation on S. pullorum infection in broilers. The results showed that ARC could not only improve the immune function of infected broilers by regulating the immune system but also directly inhibit the invasion of S. pullorum to broilers by inhibiting the formation and adhesion rate of S. pullorum biofilm. Dietary supplementation of 200 mg/kg ARC might be a potential way to substitute antibiotics to control S. pullorum infection in broilers.

Pyridox (am) ine 5'-phosphate oxidase (PNPO) is a rate-limiting enzyme in converting dietary vitamin B6 (VB6) to pyridoxal 5'-phosphate (PLP), the biologically active form of VB6 and involved in the synthesis of neurotransmitters including γ-aminobutyric acid (GABA), dopamine, and serotonin. In humans, PNPO mutations have been increasingly identified in neonatal epileptic encephalopathy and more recently also in early-onset epilepsy. Till now, little is known about the neurobiological mechanisms underlying PNPO-deficiency-induced seizures due to the lack of animal models. Previously, we identified a c.95 CA missense mutation in sugarlethal (sgll)-the Drosophila homolog of human PNPO (hPNPO)-and found mutant (sgll95) flies exhibiting a lethal phenotype on a diet devoid of VB6. Here, we report the establishment of both sgll95 and ubiquitous sgll knockdown (KD) flies as valid animal models of PNPO-deficiency-induced epilepsy. Both sgll95 and sgll KD flies exhibit spontaneous seizures before they die. Electrophysiological recordings reveal that seizures caused by PNPO deficiency have characteristics similar to that in flies treated with the GABA antagonist picrotoxin. Both seizures and lethality are associated with low PLP levels and can be rescued by ubiquitous expression of wild-type sgll or hPNPO, suggesting the functional conservation of the PNPO enzyme between humans and flies. Results from cell type-specific sgll KD further demonstrate that PNPO in the brain is necessary for seizure prevention and survival. Our establishment of the first animal model of PNPO deficiency will lead to better understanding of VB6 biology, the PNPO gene and its mutations discovered in patients, and can be a cost-effective system to test therapeutic strategies.

Galactomannan and its degradation products have been gaining attention based on their possible means for improving the natural defense of the host through modulation of the bacterial population in the gut. Herein, incomplete degradation products of galactomannan (IDPG) was supplemented into the diet of aged laying hens to investigate the efficacy of IDPG on the gut microbiome. Four treatments with six replicates of twelve 68-wk-old laying hens (Hy-Line variety brown) each were fed a basal diet supplemented with 0%, 0.01%, 0.025%, and 0.05% IDPG for 8 wk. Results showed that the propionate concentration significantly increased in laying hens fed a diet supplemented with 0.025% or 0.05% IDPG relative to the control diet (P0.05). Moreover, the results of 16S rRNA gene sequencing revealed that there was a notable elevation of microbiome species diversity due to the addition of IDPG, with a noted enrichment to phyla Bacteroidetes at the expense of Firmicutes and Proteobacteria. Metabolic prediction of the cecal microbiome suggested significant improvements to carbohydrate and lipid metabolism and a significant depletion for energy metabolism and infectious diseases. More importantly, a strong positive correlation between levels of genera Bacteroides, Rikenellaceae_RC9_gut_group, and Prevotellaceae_UCG-001 with high production of propionate was found using multivariate analysis. Our study demonstrated that IDPG acted by mainly enriching the phyla Bacteroidetes in the cecum, increasing species diversity, and cecal propionate concentrations. It seems that IDPG can be used as feed additives in laying hen farming due to its capacity to positively modulate the cecal microbiome and aid improve overall health.The health and nutritional status of poultry are largely interconnected with the gut microbiome, which directly or indirectly affects gut morphology, nutrition, and immune responses. Dietary fiber is resistant to digestion in the small intestines of monogastric animals but is completely or partially fermented in the distal gut, thus it is understood that they could stimulate gut health. Incomplete degradation products of galactomannan (IDPG) is an important member of the dietary fiber family of molecules, however, there exists scant research on their beneficial effects on human or animal health. Our study demonstrated that IDPG acted by mainly enriching the phyla Bacteroidetes in the cecum, which are common bacteria in the gut that are involved in the fermentation of carbohydrates. Thereafter, the enriched phyla Bacteroidetes produced propionate and reduced the abundance of phyla Firmicutes and Proteobacteria by competitive inhibition. IDPG has also increased species diversity and enhanced the stability of intestinal flora, thereby exhibiting excellent prebiotic activity.

There is a growing interest in using black soldier fly larvae (BSFL) due to its supposed sustainability and nutritional qualities. Because little research has been conducted to evaluate the use of BSFL in cats, our objective was to determine the palatability and apparent total tract macronutrient digestibility (ATTD) of BSFL-containing canned diets and the fecal characteristics of healthy adult cats consuming them. First, three palatability tests were conducted to compare the following diets: 1) diet with poultry byproduct meal (PBPM) and chicken serving as the primary protein sources (control) vs. diet with BSFL meal replacing PBPM (BSFL meal); 2) control vs. diet with whole BSFL replacing some PBPM and poultry fat (BSFL whole); and 3) control vs. diet with BSFL oil replacing poultry fat (BSFL oil). All diets were formulated to meet Association of American Feed Control Officials nutrient profiles for adult cats and were produced using a still retort. A paired t-test was conducted to analyze data from each palatability test, with a higher (P0.05) consumption ratio being observed for BSFL meal (1.93:1), BSFL whole (2.03:1), and BSFL oil (1.57:1). Second, 32 adult cats (20 females and 12 males; BW: 4.19 ± 0.55 kg; age: 3.3 ± 0.38 yr) were used in a completely randomized design study composed of a 21-d baseline period and a 70-d experimental period. Cats consumed the control diet during the baseline and were then allotted to one of four experimental diets (n = 8 per group): 1) control, 2) BSFL meal, 3) whole BSFL, and 4) BSFL oil. Fecal samples were collected after baseline and experimental periods for ATTD and fecal characteristic analysis. Fecal output was higher (P0.05) and fecal dry matter percentage was lower (P0.05) in cats fed BSFL meal than those fed BSFL oil. Organic matter, crude protein (CP), and energy ATTD were lower (P0.05) in cats fed BSFL meal than those fed BSFL oil or control. CP and energy ATTD were lower (P0.05) in cats fed BSFL whole than those fed BSFL oil. A few serum metabolites were affected by diet (P0.05) but remained within reference ranges. Hematology was not affected by diet (P0.05). Overall, our results suggest that BSFL-containing diets are palatable and do not negatively affect fecal characteristics or serum chemistry but may have slightly lower nutrient digestibilities in adult cats.The pet food industry is interested in black soldier fly larvae (BSFL) because of its supposed sustainability and nutritional qualities. BSFL has not been well tested in cats, so our objective was to determine the palatability and apparent total tract macronutrient digestibility (ATTD) of BSFL-containing canned diets and how they affected the fecal characteristics of adult cats. Palatability tests were conducted to compare diets containing BSFL meal, whole BSFL, or BSFL oil against a poultry-based control. Higher consumption ratios were observed for all BSFL-containing diets. A second study determined the digestibility of BSFL-containing diets and how they affected fecal characteristics. Cats were allotted to a control diet or diets containing BSFL meal, whole BSFL, or BSFL oil. Fecal output was higher and fecal dry matter percentage was lower in cats fed BSFL meal than those fed BSFL oil. Organic matter, crude protein (CP), and energy ATTD were lower in cats fed BSFL meal than those fed BSFL oil or control. CP and energy ATTD were lower in cats fed whole BSFL than those fed BSFL oil. Overall, BSFL-containing diets were shown to be palatable and did not negatively affect fecal characteristics, but reduced nutrient digestibilities in adult cats.

The objective of this study was to investigate the effects of whole corn high-grain diet feeding on growth performance, ruminal bacterial community, and epithelial morphology and gene expression related to VFA absorption and metabolism in fattening lambs. Fourteen male (castrated) lambs were randomly assigned to either a group fed a ground corn high-grain diet (50.4% grain; HGC, n = 7) or a group fed a whole corn high-grain diet (50.4% grain; HWC, n = 7). After 7 wk of feeding, HWC group increased the average daily gain (ADG) (P = 0.036) and decreased the feed: gain value (P = 0.010) significantly. HWC group had a greater crude protein apparent digestibility (P = 0.028) in the third week and dry matter and neutral detergent fiber apparent digestibility (P0.05) in the seventh week. Pyrosequencing of the 16S ribosomal RNA gene revealed that HWC feeding increased the relative abundance of genera Anaerovibrio, Schwartzia and Unclassified Veillonellaceae in the rumen content and Howardella, Schwartzia and Unclassified Veillonellaceae in the rumen epithelia (P0.05), while decreased the proportion of Lachnospira and Unclassified Synergistaceae in the rumen content and Anaerovorax, Papillibacter, Ruminococcus, Fibrobacter, Unclassified Lachnospiraceae, Unclassified Bacteroidales and Unclassified Prevotellaceae in the rumen epithelia (P0.05). HWC group increased the rumen papilla length (P = 0.001) and surface area (P = 0.002). Furthermore, HWC diet feeding up-regulated the relative mRNA expression of putative anion transporter isoform 1 (PAT1) (P = 0.032) in the rumen epithelia. In summary, compared with ground corn high-grain diet feeding, whole corn high-grain diet feeding improved animal performance, changed ruminal bacterial composition and diversity, and increased VFA absorption of epithelial papilla in fattening lambs. These findings provided theoretical guidance for the actual application of whole corn high-grain diet in ruminants.Long-term feeding of high-grain diets with too fine crushed grains to sheep will cause a series of health issues. Therefore, our study was to investigate the effects of whole corn (unprocessed corn) high-grain diet feeding on growth performance and rumen development in fattening lambs. Fourteen male (castrated) lambs were randomly assigned to either a group fed a ground corn high-grain diet (50.4% grain; HGC, n = 7) or a group fed a whole corn high-grain diet (50.4% grain; HWC, n = 7). After 7 wk of feeding, the whole corn group significantly increased the body weight. The sheep in the 2 groups ate the same amount, but the HWC group had a greater body weight. Whole corn group changed rumen microbial community, among which starch-degrading bacteria were easier to survive in the rumen. Whole corn group promoted the rumen epithelial papilla growth. All in all, compared to ground corn, feeding whole corn high-grain diet to the fattening lambs directly had a better effect. Whole corn did not require processing, which saved costs and was of great significance to actual sheep production.

The goal of this project was to determine if standardized ileal digestible (SID) lysine provided at 40% above estimated requirements, with the concomitant increase in protein intake, from days 90 to 110 of gestation would stimulate mammary development in gilts. From day 90 of gestation, Yorkshire × Landrace gilts were fed 2.65 kg of either a conventional diet (CTL, control, n = 19) providing 18.6 g/d of SID Lys or a diet providing 26.0 g/d of SID Lys via additional soybean meal (HILYS, n = 19). Both diets were isoenergetic. Jugular blood samples obtained on days 90 and 110 of gestation were used to measure concentrations of insulin-like growth factor-1 (IGF-1), metabolites, and amino acids (AA). Gilts were necropsied on day 110 ± 1 of gestation to obtain mammary glands for compositional analyses, immunohistochemistry, and analysis of mRNA abundance for AA transporters and markers of cell proliferation and differentiation. The HILYS gilts gained more body weight (P0.01) during the experimental period compared with CTL gilts, and had greater fetal weights (1.29 vs. 1.21 ± 0.03 kg, P0.05). There was no difference in circulating IGF-1, glucose, or albumin (P0.10) between HILYS and CTL gilts on day 110 of gestation, whereas concentrations of urea and free fatty acids were greater (P0.01), and those of Trp and Ala were lower (P0.05), in HILYS than CTL gilts. The provision of lysine at 40% above estimated requirements increased total mammary parenchymal mass by 44%, as well as total parenchymal fat, protein, DNA, and RNA (P0.01). The mRNA abundance of ACACA was greater (P0.05) in HILYS than CTL gilts, while only the AA transporter SLC6A14 tended (P0.10) to be greater. Results demonstrate that providing dietary Lys above current National Research Council recommendations in late gestation increases mammary development in gilts. Results also indicate that Lys may have been limiting for protein retention. These data suggest that the use of a two-phase feeding strategy during gestation, whereby dietary Lys is increased from day 90, could benefit potential sow milk yield in the subsequent lactation.Results indicate that the current National Research Council recommendations for dietary lysine during late pregnancy in pigs, the period when most mammary gland development takes place, are underestimated. From days 90 to 110 of gestation, gilts were fed 2.65 kg of either a conventional diet providing 18.6 g/d of standardized ileal digestible (SID) lysine, or a diet providing 26.0 g/d of SID lysine via the inclusion of additional soybean meal. Findings suggest that a greater mammary uptake of lysine in supplemented sows supported enhanced accretion of mammary parenchyma. Such information is most pertinent in the actual context where milk yield of hyperprolific sows must be maximized to sustain optimal growth of all their piglets. Furthermore, these data indicate that the use of a two-phase feeding strategy during gestation, whereby dietary lysine is increased from day 90, could benefit potential sow milk yield in the subsequent lactation.

The objective was to determine effects of old and new rubber matting in a slatted, indoor cattle feeding facility on cattle growth performance, locomotion, and carcass characteristics. In experiment 1, fall-born Angus × Simmental steers (N = 207; body weight = 222 ± 38 kg) were blocked by weight and assigned to 32 pens. Pens were randomly assigned to 1 of 4 treatments: no matting/concrete (CONC1), 12-yr-old Animat Pebble matting (OLD1), new Animat Maxgrip matting (MG), and new Animat Pebble matting (PEB1). Steers were fed a common diet for 209 d with a minimum stocking density of 3.40 m2 per animal. Final body weight (BW) and average daily gain (ADG) were affected (P = 0.02 and P0.01, respectively) by treatment with steers on PEB1 finishing heaviest with the greatest growth, MG and CONC1 intermediate, and OLD1 finishing at the lightest final BW with the least growth. Flooring treatment did not affect overall dry matter intake (DMI; P = 0.16) or gain to feed ratio (G:F; P = 0.94). Flooring treatment did not affect (P ≥ 0.19) any carcass traits. Locomotion scores (LS) were affected (P0.01) by flooring treatment with CONC1 having the worst mobility while OLD1, MG, and PEB1 were similar (P ≥ 0.24). Locomotion score had a day effect (P0.01) where cattle gait and mobility worsened as days on feed increased. In experiment 2, fall-born Angus × Simmental steers (N = 189; BW = 352 ± 43 kg) were blocked by weight and assigned to 21 pens. Pens were randomly assigned to 1 of 3 treatments: no matting/concrete (CONC2), 15-yr-old Animat Pebble matting (OLD2), and new Animat Pebble matting (PEB2). Steers were fed a common diet for 152 d with a stocking density of 2.65 m2 per steer. After 152 d on feed, flooring treatment did not affect (P ≥ 0.30) BW, ADG, or DMI nor did treatment affect (P ≥ 0.17) carcass traits. However, steers housed on OLD2 or PEB2 had improved locomotion scores (P = 0.02) compared with steers housed on CONC2. Locomotion score had a day effect (P0.01) as cattle gait and mobility worsened with greater number of days on feed, regardless of treatment. Overall, results suggest that new rubber matting increased ADG and HCW during a 209-d trial when cattle were stocked at 3.4 m2 in small pens and that rubber matting regardless of age improved cattle locomotion scores in slatted indoor feeding facilities.Many feedlots in the Midwest have constructed slatted indoor cattle feeding facilities to improve winter pen conditions and decrease land requirements. Finishing cattle in indoor feeding facilities can increase cattle growth and carcass value, but greater prevalence of cattle lameness is a concern. Rubber matting is commonly used in these facilities, but it is a consumable product with a variable lifespan. This study evaluated the effects of old and new rubber matting in a slatted, indoor cattle feeding facility on cattle growth performance, locomotion, and carcass characteristics in two experiments. In experiment 1, steers in pens with new Animat Pebble matting had increased growth compared with steers in pens with no matting and 12-yr-old Animal Pebble matting. Furthermore, steers in pens with no matting had the poorest locomotion scores. In experiment 2, flooring treatments did not affect cattle growth or carcass traits, but steers in pens with old or new rubber matting had more desirable locomotion scores than cattle in pens with no matting. Therefore, new rubber matting can improve cattle growth in small pens with slatted floors when cattle were stocked at 3.4 m2 per steer (experiment 1) and that rubber matting regardless of age improved cattle locomotion.

This 2-yr study investigated the timing of dried distillers grains (DDG) supplementation during the third trimester of gestation of Bos indicus-influenced beef cows and its impact on their offspring performance. On day 0 of each year (84 d before calving), Brangus cows (n = 84/yr; cow age = 8 ± 3 yr) were stratified by initial body weight (BW; 482 ± 75 kg) and body condition score (BCS; 5.3 ± 0.8) and assigned randomly to one of six bahiagrass (Paspalum notatum) pastures (experimental units; 14 cows/pasture). Treatments were assigned randomly to pasture (2 pastures/treatment/yr) and consisted of no prepartum supplementation (CON), 2 kg/d of DDG from day 0 to 42 (LATE42), or 1 kg/d of DDG from day 0 to 84 (LATE84). Following calving (day 84), cow-calf pairs remained in their respective pastures, and cows were offered sugarcane molasses + urea (1.82 kg of dry matter/cow/d) from day 85 until the end of the breeding season (day 224). On day 347, steer calves (n = 38/yr; 11 to 15 steers/treatment/yr) were weaned and transported to the feedlot (1,193 km). Steers were penned according to cow prepartum pasture and managed similarly until the time of harvest. BCS at calving was greater (P0.01) for LATE42 and LATE84 vs. CON cows but did not differ (P = 0.16) between LATE42 and LATE84 cows. Calving date, calving percentage, and birth BW of the first offspring did not differ (P ≥ 0.22) among treatments. However, LATE42 cows calved their second offspring 8 d earlier (P = 0.04) compared with CON and LATE84 cows. At weaning (first offspring), LATE84 calves were the heaviest (P ≤ 0.05), CON calves were the lightest, and LATE42 calves had intermediate BW (P ≤ 0.05). Steer plasma concentrations of cortisol and haptoglobin and serum bovine viral diarrhea virus type-1 titers did not differ (P ≥ 0.21) between treatments. Steer serum parainfluenza-3 titers were greater (P = 0.03) for LATE42 vs. CON steers, tended to be greater (P = 0.10) for LATE84 compared with CON steers, and did not differ (P = 0.38) between LATE42 and LATE84 steers. Steer feedlot BW, average daily gain, dry matter intake, and hot carcass weight did not differ (P ≥ 0.36) between treatments. Marbling and the percentage of steers grading choice were greater (P ≤ 0.04) for LATE42 vs. CON steers, whereas LATE84 steers were intermediate. In summary, different timing of DDG supplementation during the third trimester of gestation could be explored to optimize cow BCS and offspring preweaning growth and carcass quality.This 2-yr study evaluated the effect of the timing of dried distillers grains (DDG) supplementation during the last trimester of gestation in Bos indicus-influenced beef cows and the subsequent impact on their offspring. Brangus cows were allocated to one of the three prepartum treatments consisting of no prepartum supplementation, 2 kg/d of DDG for the first half of the last trimester of gestation, or 1 kg/d of DDG for the entire length of the last trimester of gestation. Prepartum supplementation, regardless of supplementation length, improved cow body condition scores at the time of calving. Calf birth weights were not affected by prepartum maternal treatment. Calves born to cows that received prepartum supplementation had greater weaning weight compared with no prepartum supplementation. However, weaning weights were improved to the greatest extent when calves were born to cows that received supplementation for the entire length of late gestation. Steer antibody response to parainfluenza-3 was improved with prepartum maternal supplementation, regardless of supplementation length. Furthermore, concentrating the total amount of supplement offered to the first half of the last trimester of gestation improved marbling and increased the percentage of steers grading choice compared with no prepartum supplementation or supplementation during the entire last trimester.

Heat stress (HS) triggers oxidative stress, systemic inflammation, and disrupts growth efficiency of livestock. β-adrenergic agonists supplemented to ruminant livestock improve growth performance, increase skeletal muscle mass, and decrease carcass fat. The objective of this study was to understand the independent and interacting effects of HS and zilpaterol hydrochloride (ZH) supplementation on the transcriptome of subcutaneous white adipose tissue and the longissimus dorsi muscle in steers. Twenty-four Red Angus-based steers were assigned to thermoneutral (TN; Temperature Humidity Index [THI] = 68) or HS (THI = 73-85) conditions and were not supplemented or supplemented with ZH (8.33 mg/kg/d) for 21 d in a 2 × 2 factorial. Steers in the TN condition were pair-fed to the average daily feed intake of HS steers. RNA was isolated from adipose tissue and skeletal muscle samples collected via biopsy on 3, 10, and 21 d and sequenced using 3' Tag-Seq to an achieved average depth of 3.6 million reads/sample. Transcripts, mapped to ARS-UCD1.2, were quantified. Differential expression (DE) analyses were performed in DESeq2 with a significance threshold for false discovery rate of 0.05. In adipose, 4 loci (MISP3, APOL6, SLC25A4, and S100A12) were DE due to ZH on day 3, and 2 (RRAD, ALB) were DE due to the interaction of HS and ZH on day 10 (Padj0.05). In muscle, 40 loci (including TENM4 and OAZ1) were DE due to ZH on day 10, and 6 loci (HIF1A, LOC101903734, PDZD9, HNRNPU, MTUS1, and TMCO6) were DE due to environment on day 21 (Padj0.05). To explore biological pathways altered by environment, supplement, and their interaction, loci with DE (Praw0.05) were evaluated in Ingenuity Pathway Analysis. In adipose, 509 pathways were predicted to be altered (P0.01): 202 due to HS, 126 due to ZH, and 181 due to the interaction; these included inflammatory pathways predicted to be upregulated due to HS but downregulated due to the interaction of HS and ZH. In muscle, 113 pathways were predicted to be altered (P0.01): 23 due to HS, 66 due to ZH, and 24 due to the interaction of HS and ZH. Loci and pathway data in muscle suggest HS induced oxidative stress and that the stress response was moderated by ZH. Metabolic pathways were predicted to be altered due to HS, ZH, and their interaction in both tissues. These data provide evidence that HS and ZH interact to alter expression of genes in metabolic and immune function pathways and that ZH moderates some adverse effects of HS.Heat stress (HS) negatively impacts livestock health and carcass quality. Supplementation of livestock with β-adrenergic agonists (β-AA) increases muscle mass and decreases fat deposition. The purpose of this study was to understand how HS and zilpaterol hydrochloride (ZH), a β-AA, alter gene expression in muscle and in adipose of cattle. Twenty-four steers were assigned to thermoneutral (TN) or HS conditions and were not supplemented (NS) or supplemented with ZH for 21 d. RNA was isolated from muscle and adipose collected on days 3, 10, and 21 to identify changes in gene expression. Several individual loci were differentially expressed (DE) due to HS or ZH in both tissues while the interaction of HS and ZH altered expression in adipose. A less stringent definition of DE used to explore biological pathways predicted that both treatments alter metabolism. Pathway analyses also supported that HS increased inflammation in adipose, but that these inflammatory pathways were downregulated by ZH. HS also was predicted to induce oxidative stress in muscle although ZH moderated this response. This study provides information on how HS and β-AA act independently and interact to alter physiology, lending insight useful for the development of management and mitigation strategies for stress.

Soybean accounts for more than half of the global production of oilseed and more than a quarter of the protein used globally for human food and animal feed. Soybean domestication involved parallel increases in seed size and oil content, and a concomitant decrease in protein content. However, science has not yet discovered whether these effects were due to selective pressure on a single gene or multiple genes. Here, re-sequencing data from >800 genotypes revealed a strong selection during soybean domestication on GmSWEET10a. The selection of GmSWEET10a conferred simultaneous increases in soybean-seed size and oil content as well as a reduction in the protein content. The result was validated using both near-isogenic lines carrying substitution of haplotype chromosomal segments and transgenic soybeans. Moreover, GmSWEET10b was found to be functionally redundant with its homologue GmSWEET10a and to be undergoing selection in current breeding, leading the the elite allele GmSWEET10b, a potential target for present-day soybean breeding. Both GmSWEET10a and GmSWEET10b were shown to transport sucrose and hexose, contributing to sugar allocation from seed coat to embryo, which consequently determines oil and protein contents and seed size in soybean. We conclude that past selection of optimal GmSWEET10a alleles drove the initial domestication of multiple soybean-seed traits and that targeted selection of the elite allele GmSWEET10b may further improve the yield and seed quality of modern soybean cultivars.

Prebiotics and dietary fibers are nondigestible ingredients that may confer benefits to the host by selectively stimulating beneficial intestinal bacteria and microbial-derived metabolites that support gut and host health. This experiment evaluated the effects of a blend of prebiotics and dietary fibers on apparent total tract digestibility (ATTD) and fecal metabolites related to gastrointestinal health in adult dogs. Four diets containing either 5% cellulose (control; CT), 5% dietary fiber and prebiotic blend (FP), 0.02% saccharin and eugenol (SE), or 5% fiber blend plus 0.02% saccharin and eugenol (FSE) were formulated to meet or exceed the AAFCO (2017) nutritional requirements for adult dogs. Eight adult female beagles (mean age 4.2 ± 1.1 yr; mean BW = 10.8 ± 1.4 kg; mean BCS = 5.8 ± 0.6) were randomly assigned to 1 of the 4 dietary treatments using a replicated 4 × 4 Latin square design. Each experimental period consisted of 14 d (10 d of diet adaptation and 4 d of total and fresh fecal and total urine collection). All animals remained healthy throughout the study, with serum metabolites being within reference ranges for adult dogs. All diets were well accepted by the dogs, resulting in similar (P > 0.05) daily food intakes among treatments. Likewise, fecal output and scores did not differ (P > 0.05) among dietary treatments, with the latter being within the ideal range (2.5–2.9) in a 5-point scale. All diets were highly digestible and had similar (P > 0.05) ATTD of dry matter (81.6%–84.4%), organic matter (86.4%–87.3%), and crude protein (86.6%–87.3%). However, total dietary fiber (TDF) digestibility was greater for dogs fed the FSE diet (P < 0.05) in contrast with dogs fed the CT and SE diets, whereas dogs fed FP diets had intermediate TDF digestibility, but not different from all other treatments. Fecal acetate and propionate concentrations were greater (P < 0.05) for dogs fed FP and FSE diets. Fecal concentrations of isobutyrate and isovalerate were greater for dogs fed CT (P < 0.05) compared with dogs fed the other three treatments. No shifts in fecal microbial richness and diversity were observed among dietary treatments. Overall, the data suggest that dietary supplementation of fiber and prebiotic blend was well tolerated by dogs, did not cause detrimental effects on fecal quality or nutrient digestibility, and resulted in beneficial shifts in fecal metabolites that may support gut health.

Heat stress (HS) decreases lactation output in sows due to an attempt to reduce metabolic heat production. However, this negatively affects litter growth performance. Therefore, the study objective was to determine whether electronically controlled cooling pads (ECP) would improve indirect measures of lactation output (e.g., total heat production [THP]) and litter growth performance in HS exposed sows. Over two repetitions, 12 multiparous (2.69 ± 0.85) lactating sows (265.4 ± 26.1 kg body weight [BW]) and litters were assigned to either an ECP (n = 3/repetition) or a non-functional ECP (NECP; n = 3/repetition) and placed into farrowing crates within indirect calorimeters from days 3.7 ± 0.5 to 18.7 ± 0.5 of lactation. Litters were standardized across all sows (11.4 ± 0.7 piglets/litter), and sows were provided ad libitum feed and water. All sows were exposed to cyclical HS (28.27 ± 0.26 °C nighttime to 33.09 ± 0.19°C daytime). On days 4, 8, 14, and 18 of lactation, indirect calorimetry was performed on each individual sow and litter to determine THP and THP/kg BW0.75. Body temperature (TB) was measured hourly using vaginal implants, and respiration rate (RR; breaths per minute [bpm]) was measured daily at 0700, 1100, 1300, 1500, and 1900 h. Sow feed intake (FI) was assessed daily. Litter weights were obtained at birth, on days 4, 8, 14, and 18 of lactation, and at weaning. Data were analyzed using PROC GLIMMIX with sow and/or litter as the experimental unit. An overall decrease (P0.01; 25 bpm) in RR and maximum daily TB (P = 0.02; 0.40 °C) was observed in ECP vs. NECP sows. An increase in THP (P0.01; 20.4%) and THP/kg BW0.75 (P0.01; 23.1%) was observed for ECP when compared with NECP sows and litters. Litter average daily gain and weaning weight were increased (P0.05; 25.0 and 19.2%, respectively) for ECP vs. NECP litters. No FI differences were observed (P = 0.40) when comparing ECP (5.66 ± 0.31 kg/d) and NECP (5.28 ± 0.31 kg/d) sows. In summary, ECPs improve litter growth, thermoregulatory measures, and bioenergetic parameters associated with greater milk production in lactating sows exposed to cyclical HS.Lactating sows are heat stress sensitive due to greater feed intake and metabolic heat production to support milk production. Therefore, heat-stressed lactating sows reduce their feed intake and undergo physiological changes that prioritize survival over productivity which reduces milk production. Reduced milk production has negative downstream effects on the litter and can decrease litter growth performance and health status. The negative effects of heat stress on lactating sows may be ameliorated by using electronically controlled cooling pads. Electronically controlled cooling pads improve the ability of lactating sows to maintain a normal body temperature under heat stress conditions. However, it is unknown whether these pads can improve lactating sow feed intake, milk production, and subsequently litter performance under heat stress conditions. It was determined that electronically controlled cooling pads were effective in increasing piglet body weight gain and weaning weight and increasing indirect measures of sow milk production independent of greater sow feed intake. In addition, the electronically controlled cooling pads allowed sows to maintain a normal body temperature under heat stress conditions. These data suggest that electronically controlled cooling pads can be an effective technology to improve lactating sow and litter performance under heat stress conditions.

To determine the effect of CaO-treated Pensacola bahiagrass (Paspalum notatum) hay on intake, ruminal fermentation parameters, and apparent total-tract digestibility of nutrients, nine ruminally cannulated Angus-crossbred steers were used in a triplicated 3 × 3 Latin square design. Steers had ad libitum access to either 1) untreated dry hay (DH; n = 8); 2) hay at 50% DM treated with 8.9% CaCO3 (dry matter [DM] basis; CC; n = 9); or 3) hay at 50% DM treated with 5% CaO (DM basis; CO; n = 8). Water was added to reach 50% DM in the CC and CO diets. Ruminal fluid and blood samples were collected every 3 h for 24 h. Ruminal fluid was analyzed for pH, volatile fatty acids (VFA), and ammonia-nitrogen (NH3-N). Blood was analyzed for plasma urea nitrogen (PUN). Hay and fecal samples were collected for 4 d, four times daily for hay and twice daily for feces, to determine apparent total-tract digestibility of nutrients. The hay provided to steers during the digestibility period was analyzed for in vitro organic matter digestibility (IVOMD) for 48 h. Data were analyzed as repeated measures for blood and ruminal fermentation parameters. Total DM intake was not affected (P ≥ 0.674) by treatment. A treatment effect (P0.001) was observed for average ruminal pH, where steers consuming CO had the greatest pH (P0.001). Ruminal concentration of NH3-N tended (P = 0.059) to be reduced in steers consuming CO. There was a treatment × time interaction (P = 0.023) on concentrations of PUN, where at 3 h DH and CO were lesser than CC (P ≤ 0.050) and at 21 h DH was lesser than CC (P = 0.020). Total VFA, acetate, propionate, butyrate, branched-chain VFA, and valerate concentrations were affected by treatment (P ≤ 0.035), where a reduction (P ≤ 0.034) occurred in steers consuming CO. No treatment differences were observed for total-tract digestibility of DM (P = 0.186), organic matter (P = 0.169), or crude protein (P = 0.152); however, steers consuming DH had greater neutral detergent fiber (P = 0.038) than CC and tended to be greater than CO (P = 0.082). The CO hay had greater (P = 0.005) IVOMD compared with DH and tended (P = 0.100) to be greater than CC. Bahiagrass hay treated with CaO may reduce ruminal fermentation, as indicated by decreased total VFA concentration without altering DM intake. The addition of CaO did not improve the digestibility of bahiagrass hay in vivo; however, in vitro results are contradictory and warrant further elucidation.With the ever-growing desire to increase efficiency in beef cattle production, researchers have developed strategies such as treating poor-quality forages with chemicals to increase the digestibility of fiber fractions, consequently increasing their energy value for cattle feeding. Calcium oxide has been proposed as a replacement to more caustic chemicals used in the past (e.g., NaOH) and data indicate that it can promote similar and effective outcomes. The current study evaluated the effects of bahiagrass hay treated with calcium oxide on ruminal fermentation parameters, apparent total-tract digestibility of nutrients, and intake by beef steers consuming hay ad libitum as the sole ingredient in their diet. Additionally, in vitro organic matter digestibility was evaluated on the hay provided to steers to assess treatment effectiveness. Results indicated that steers consuming bahiagrass hay treated with calcium oxide had 1) increased pH and reduced volatile fatty acids concentrations in the rumen; 2) reduced or tendency for reduction on total-tract digestibility of fiber fractions; and 3) no effect on intake, all when compared with steers consuming untreated hay. In contrast, in vitro results indicated that organic matter digestibility was increased when the forage was treated with calcium oxide.

This study was aimed to explore the effect of using cassava as an amylopectin source in low protein (LP) diets on growth performance, nitrogen efficiency, and postprandial changes in plasma glucose and related hormones concentrations of growing pigs. Three animal experiments were included in the present study. Treatments included corn-soybean meal LP (Corn LP), corn-cassava-soybean meal LP (Corn + Cassava LP), and cassava-soybean meal LP (Cassava LP). The in vitro digestion proved that Corn + Cassava LP and Cassava LP diets induced more rapid starch digestion and glucose release, compared with Corn LP diet. The results of animal experiments are as follows: Cassava LP diet caused the most rapid changes in plasma glucose and relevant hormones concentrations after a meal. It decreased the concentrations of fasting plasma insulin, glucagon, and leptin concentrations compared with other treatments (P < 0.05). These modulations above led to a strong desire to eat and increased feed intake and then weight gain in growing pigs fed Cassava LP diet. Besides, feeding Cassava LP diet caused diarrhea, increased noxious gas release from feces, and increased concentrations of fecal isobutyrate and isovalerate (P < 0.05). Compared with Corn LP group, Corn + Cassava LP group showed significantly decreased urinary nitrogen (P < 0.05) and improved post-absorptive amino acid utilization efficiency. In conclusion, the use of cassava as an amylopectin source in LP diets could modulate glucose absorption and related gut secreted hormones secretion, subsequently strengthened the desire to eat, improved growth performance, and enhanced nitrogen efficiency in growing pigs.

This study used 18 calves (295 ± 29 kg) and 18 yearlings (521 ± 29 kg) fed whole, cracked, or steam-flaked corn (SFC) to evaluate nutrient digestion and energy balance across different types of processed corn and sizes of cattle. Cattle were fed a diet comprised of 75% corn (dry matter [DM]-basis) from whole, cracked, or SFC to 2.5-times maintenance energy requirements. Subsequently, cattle were placed in individual stanchions, and urine and feces were collected together with measures of gas production via indirect calorimetry. Data were analyzed using the MIXED procedure of SAS. There was no interaction between corn processing and cattle size (P ≥ 0.40). Time spent ruminating (min/d) and rumination rate (min/kg DM intake [DMI]) were not affected by corn processing or cattle size. The eating rate (min/kg DMI) was faster (P < 0.01) for yearlings compared with calves. Total tract starch digestion was greatest (P = 0.01) for cattle fed SFC (97.5%), intermediate in cattle fed cracked (92.4%), and least in cattle fed whole corn (89.5%). Dietary digestible energy and metabolizable energy (Mcal/kg DMI) were greater (P ≤ 0.05) for cattle fed SFC compared with cracked or whole. A greater proportion of digestible energy was lost to heat production (P = 0.01) in cattle fed whole corn compared with cracked and tended to be greater (P = 0.08) in cattle fed SFC than cracked. Conversion of digestible energy to metabolizable energy in this study was more closely related to a dynamic model used to estimate metabolizable energy of feeds to dairy cows than to a linear model used to predict metabolizable energy of feeds to beef cattle. If library estimates of net energy for maintenance are correct, then retained energy (Mcal/d) should have been similar between each type of processed corn; however, retained energy was greater (P < 0.01) for cattle fed cracked compared with whole corn and tended to be greater (P = 0.06) compared with SFC. Yet, observed amounts of net energy based on measures of retained energy were not different (P ≥ 0.60) between cracked and SFC. Nitrogen balance was not affected (P ≥ 0.30) by corn processing or cattle size, although cattle fed cracked had numerically greater (P ≤ 035) N retention. These data indicate that physical processing of corn provides greater net energy to cattle in comparison to whole corn.

A study investigating the use of a nonviable Lactobacillus acidophilus (NVL: Culbac; TransAgra, Storm Lake, IA) and a mixed prebiotic (MP) blend (beet pulp, fructooligosaccharide (FOS), mannanoligosaccharide (MOS), inulin, and kelp) was done to evaluate changes in fecal microbiota, fermentative end products, and gut immune health in healthy female and male adult Beagle dogs (n = 24; 5.74 ± 2.18 yr; 9.30 ± 1.32 kg). The study protocol was first approved by the facility’s Institutional Animal Care and Use Committee (Summit Ridge Farms; Susquehanna, PA) and followed throughout. Each of four test diets (control, NVL, MP, and MP + NVL [formulated to crude protein 25%, crude fat 14%, crude fiber 10% as-fed]) was fed once daily to maintain body weight for 21 d in a randomized-crossover design (four treatment periods and four washout periods). Fecal samples were collected on days 0 and 21 only for immunoglobulin A (IgA) and microbiota evaluation (16S rRNA V4 region and qPCR for Escherichia coli and Bifidobacterium), and fecal fermentative end-products and fecal pH were assessed only on day 21. Over the test periods, apparent total tract nutrient digestibility and stool quality were assessed. Data were analyzed by ANOVA (SAS v9.4, Cary, NC) or Kruskal–Wallis for between-diet effects, and paired t-test or Wilcoxon for time effects. Statistical significance was set at P ≤ 0.05. Apparent total tract nutrient digestibility revealed feeding MP-containing diets resulted in lower (P < 0.05) crude protein and fat digestibility vs. control and NVL diets. When dogs were fed MP, they had lower (P < 0.05) fecal pH compared with control and NVL diets, whereas fecal pH was lower in (P < 0.05) MP + NVL- vs. NVL-fed dogs. Fecal E. coli was (P < 0.05) lower at day 21 vs. day 0 when dogs were fed MP. Fecal Fusobacterium spp. was lower (P < 0.05) in both MP diets vs. control. Fecal Lactobacillus spp. increased (P < 0.05) from baseline with MP. Both diets with MP elicited greater (P < 0.05) fecal acetate and propionate concentration vs. control diet. At day 21, fecal IgA was greater (P < 0.05) in MP and MP + NVL compared with NVL diet. Only when dogs were fed MP did they have increased (P < 0.05) fecal IgA from day 21 vs. day 0. The MP + NVL diet decreased (P < 0.05) fecal isovalerate, isobutyrate, phenol, and indole vs. control. Overall, the MP elicited the most changes on microbiota, fermentative end-products, and IgA. Further investigation into NVL’s gut health benefits is warranted.

Genetic selection of pigs over recent decades has sought to reduce carcass fat content to meet consumer demands for lean meat in many countries (e.g., Australia). Due to the impacts of genetic changes, it is unknown whether the carcass fat measures are still responsive to energy intake. Thus, the present experiment aimed to quantify the relationship between tissue composition and dietary energy intake in finisher pigs selected for low carcass backfat. Intact male and female pigs (n = 56 for each sex; Primegro Genetics, Corowa, NSW, Australia) were fed seven different amounts of an amino acid adequate wheat-based diet containing 14.3 MJ digestible energy (DE)/kg to provide the following daily DE intakes- 25.8, 29.0, 32.6, 35.3, 38.5, 41.5, and 44.2 (ad libitum) MJ DE/d for males, and 25.8, 28.9, 32.0, 35.6, 38.3, 40.9, and 44.5 (ad libitum) MJ DE/d for females between 60 and 108 kg live weight. Body composition of anesthetized pigs was measured using the dual energy X-ray absorptiometry (DXA) method when individual pigs reached 108 kg, and protein, fat, and ash deposition rates were calculated. Pigs were slaughtered on the second day post-DXA scan for carcass backfat measurement. The results showed that the carcass backfat thickness (standardized at 83.7 kg carcass) increased by 0.125 mm for every MJ increase in daily DE intake in male pigs (P = 0.004; R2 = 0.130), but carcass backfat of female pigs (standardized at 85.1 kg carcass) was not responsive to daily DE intake. Whole-body fat composition and fat deposition rate increased linearly (both P < 0.01) in male pigs but quadratically (both P < 0.01) in female pigs in response to DE intake. Every MJ increase of daily DE intake increased the rate of daily protein deposition by 3.8 g in intact male pigs (P < 0.001; R2 = 0.781) and by 2.5 g in female pigs (P < 0.001; R2 = 0.643). In conclusion, the selection for low backfat thickness over the last two decades has altered the response of fat deposition and backfat thickness to energy intake, particularly in female pigs. Despite this change, the linear relationship between DE intake and protein deposition rate was maintained in these modern genetics.

Intensive pastoral systems have moved away from diverse and varied diets towards overly simple monotonous diets. Feed choice through time is an obsolete way of providing forage to animals, as intensive management schemes generally allocate a single herbage or a dyad mixed sward. Monotonous feeding regimes impose nutritional repetition, which may impair animal performance and welfare. The objective of this experiment was to determine the impact of a diverse diet [DIV; free choice from perennial ryegrass (Lolium perenne L.), plantain (Plantago lanceolata L.), alfalfa (Medicago sativa L.), and chicory (Cichorium intybus L.) at all times], a varied diet [VAR; choice from ryegrass and plantain in the AM (0700–1600 h), and chicory and alfalfa in PM (1600–0700 h)], and a single forage diet of alfalfa [SFA; alfalfa at all times], on DMI, performance, and welfare of lambs. Six-month-old Coopworth ram lambs (n = 21) were offered their respective fresh-forage treatment (n = 7) diet indoors for 20 d. The DIV lambs consumed 1.64 ± 0.03 kg DM/d (mean ± SEm), which was 6% more (P < 0.05; 1.54 ± 0.03 kg DM/d) than the SFA and were not different (P > 0.05; 1.59 ± 0.03 kg DM/d) to the VAR lambs. Average daily gain (ADG) of DIV (296 g/d) and VAR (378 g/d) was 30% and 67% greater (P < 0.05) than that in the SFA lambs (227 g/d), respectively. The VAR lambs had 28% greater (P < 0.05) ADG than the DIV lambs. Differences among treatments were detected (P < 0.05) for the proportion of the day spent conducting the following behaviors: eating, ruminating, idling, lying, and standing. In addition, the number of bouts of stereotypic behaviors recorded from the SFA lambs (13.2 ± 2.2) was 150% greater (P < 0.05) than the DIV (5.1 ± 1.0) and VAR (5.5 ± 1.0) lambs. Our results suggest that the varied diet offered can improve animal performance and welfare compared to a monotonous SFA diet. Feeding management to provide a varied diet can improve performance relative to giving lambs free choice from taxonomically diverse forage options. Moreover, performance is affected by more than the primary chemical composition of the diet consumed, but how the diet is presented through time and the herbage species and quantities of each that are consumed to reach that chemical composition.

A total of 360 weanling barrows (Line 200 ×400, DNA, Columbus NE; initially 5.6 ± 0.03 kg) were used in a 42-d study to evaluate yeast-based pre- and probiotics (Phileo by Lesaffre, Milwaukee, WI) in diets with or without pharmacological levels of Zn on growth performance and antimicrobial resistance (AMR) patterns of fecal Escherichia coli. Pens were assigned to one of four dietary treatments with five pigs per pen and 18 pens per treatment. Dietary treatments were arranged in a 2 × 2 factorial with main effects of yeast-based pre- and probiotics (none vs. 0.10% ActiSaf Sc 47 HR+, 0.05% SafMannan, and 0.05% NucleoSaf from days 0 to 7, then concentrations were lowered by 50% from days 7 to 21) and pharmacological levels of Zn (110 vs. 3,000 mg/kg from days 0 to 7, and 2,000 mg/kg from days 7 to 21 with added Zn provided by ZnO). All pigs were fed a common diet from days 21 to 42 post-weaning. There were no yeast ×Zn interactions or effects from yeast additives observed on any response criteria. From days 0 to 21 and 0 to 42, pigs fed pharmacological levels of Zn had increased (P < 0.001) ADG and ADFI. Fecal samples were collected on days 4, 21, and 42 from the same three pigs per pen for fecal dry matter (DM) and AMR patterns of E. coli. On day 4, pigs fed pharmacological levels of Zn had greater fecal DM (P = 0.043); however, no differences were observed on day 21 or 42. Escherichia coli was isolated from fecal samples and the microbroth dilution method was used to determine the minimal inhibitory concentrations (MIC) of E. coli isolates to 14 different antimicrobials. Isolates were categorized as either susceptible, intermediate, or resistant based on Clinical and Laboratory Standards Institute (CLSI) guidelines. The addition of pharmacological levels of Zn had a tendency (P = 0.051) to increase the MIC values of ciprofloxacin; however, these MIC values were still well under the CLSI classified resistant breakpoint for ciprofloxacin. There was no evidence for differences (P > 0.10) for yeast additives or Zn for AMR of fecal E. coli isolates to any of the remaining antibiotics. In conclusion, pharmacological levels of Zn improved ADG, ADFI, and all isolates were classified as susceptible to ciprofloxacin although the MIC of fecal E. coli tended to be increased. Thus, the short-term use of pharmacological levels of Zn did not increase antimicrobial resistance. There was no response observed from live yeast and yeast extracts for any of the growth, fecal DM, or AMR of fecal E. coli criteria.

A recent association between the inclusion of pulses in canine diets and taurine deficiency has become a prevalent issue in the pet food industry. Although dogs do not currently have a nutritional requirement for taurine, taurine deficiencies that do occur can result in serious health issues, such as dilated cardiomyopathy. The objective of this study was to determine the circulating concentrations of plasma and whole blood taurine, indispensable and dispensable amino acid concentrations in the plasma, and taurine and creatinine concentrations in the urine of adult dogs fed a green lentil diet. Twelve adult, intact, female beagles were randomly assigned to a diet containing 45% green lentils (GLD) or a poultry byproduct meal diet (CON) for 90 d. Fresh urine samples were collected every 30 d and analyzed for taurine and creatinine concentrations. A blood sample was also collected every 30 d and analyzed for amino acids including taurine. Animal procedures were approved by the University of Illinois Institutional Animal Care and Use Committee. All diets were formulated to meet or exceed the nutrient requirements for adult dogs at maintenance. The concentrations of taurine in the plasma and whole blood showed no differences (P > 0.05) between dietary treatments or across time points. Similarly, no differences (P > 0.05) in plasma methionine concentrations were observed between treatments or across time points. A treatment effect (P < 0.05) showed dogs fed GLD had higher total primary fecal bile acid excretion compared with dogs fed CON. The differential abundance of fecal microbial communities showed Firmicutes as the predominant phyla in dogs fed both GLD and CON, with Bacteroidaceae, Erysipelotrichaceae, and Lactobacillaceae as predominant families in dogs fed GLD. The α-diversity of dogs fed GLD (P < 0.05) was lower than in dogs fed CON. These data suggest that the inclusion of 45% green lentil in extruded diets does not lower whole blood and plasma taurine concentrations during a 90-d period and is appropriate for use in a complete and balanced formulation for dogs.

An experiment was conducted to test two hypotheses: 1) reducing dietary Ca and P reduces gastric pH and diarrhea in weanling pigs; 2) negative effects of low Ca and P on pig growth performance may be overcome if phytase is added to the diets. A total of 320 weanling pigs (6.35 ± 0.87 kg) were allotted to eight corn-soybean meal-based diets in a randomized complete block design with five pigs per pen. Two phase 1 (days 1 to 14) control diets containing 100 or 50% of total Ca and digestible P relative to the requirement, and six diets in which 500, 2,000, or 16,000 units of phytase/kg feed (FTU) were added to each control diet were formulated. Phytase was assumed to release 0.16% total Ca and 0.11% digestible P. Common diets were fed in phases 2 (days 15 to 27) and 3 (days 28 to 42). Growth performance data were recorded within each phase. Data for fecal scores and gastrointestinal pH were recorded for phase 1. Colon content (day 14), the right femur (days 14 and 42), and blood samples (days −1, 14, 27, and 42) were collected from one pig per pen. In phase 1, reducing Ca and P did not reduce gastric pH or fecal score, but pigs fed the 50% diets had reduced (P < 0.05) average daily gain (ADG) and average daily feed intake (ADFI) compared with pigs fed the 100% diets. In both 50% and 100% diets, phytase above 500 FTU increased (P < 0.05) gain:feed ratio (G:F) and tended (P < 0.10) to reduce gastric pH of pigs. From days 1 to 42, pigs fed the 50% diets tended (P < 0.10) to have reduced ADG and ADFI compared with pigs fed the 100% diets, but among the 100% diets, pigs tended (P < 0.10) to have a linear increase in G:F as phytase level increased. Pigs fed the 50% diets had reduced (P < 0.05) concentrations of inositol phosphate esters (IP) in the colon and reduced bone ash (days 14 and 42) compared with pigs fed the 100% diets. Phytase did not affect bone ash or most blood metabolites. Concentrations of IP in the colon decreased, whereas plasma inositol increased (d 14; P < 0.05) in pigs fed diets with phytase (≥ 500 FTU). In pigs fed the 100% diets, IP in the colon linearly decreased (P < 0.05), but plasma inositol linearly increased (P < 0.05) with increasing levels of phytase. In conclusion, reducing Ca and P in diets for weanling pigs did not influence gastric pH or fecal score, but compromised growth performance and bone ash. However, regardless of dietary Ca and P, high doses of phytase increased phytate degradation and inositol absorption, which consequently increased G:F of pigs.

The negative impacts of stress on gastrointestinal tract (GIT) barrier function can result in compromised animal growth and health. Aspirin is known to cause mucosal injury leading to increased gut permeability and tight junction damage and can be used as a model to study leaky gut in cattle. The objective of this study was to determine the long-term impact of aspirin-induced chronic leaky gut on cattle growth and carcass attributes. Two treatments were evaluated in two studies: control (no aspirin) or 0.25% of the diet dry matter (DM) aspirin fed daily. Diets consisted of 50% corn, 24% dried distillers grains, 20% corn silage, and 6% supplement on a DM basis. In experiment 1, sixteen Angus × Simmental heifers, allotted by body weight (BW) and breed composition, were fed diets for 154 d. On day 155, heifers were dosed with 1 liter of a 180-mM Cr–ethylenediaminetetraacetic acid solution using an esophageal tube and had urine collected every 1.5 to 3 h for 48 h for analysis of Cr as a measure of gut leakiness. In experiment 2, ninety-six Simmental × Angus steers (355.0 ± 14.8 kg) were allotted by BW and breed composition and fed treatment diets for 159 d. Weight was recorded monthly and serum was collected on day 159 and analyzed for lipopolysaccharide-binding protein (LBP), interleukin-6 (IL-6), serum amyloid A (SAA), haptoglobin, and aspartate aminotransferase (AST). Data were analyzed using the MIXED procedure of SAS. Heifers fed 0.25% aspirin in experiment 1 excreted more Cr into urine compared with heifers not fed aspirin (overall treatment effect, P = 0.01). In experiment 2, aspirin tended to increase serum LBP (P = 0.06) but had no effect on concentrations of IL-6, haptoglobin, SAA, or AST (P ≥ 0.25). Aspirin tended to decrease average daily gain (P = 0.10), decreased hot carcass weight and rib-eye area (P ≤ 0.05), and increased fat thickness, marbling score, and yield grade (P ≤ 0.02). Aspirin tended to increase kidney, pelvic, and heart fat percentage (P = 0.10) and had no effect on liver abscesses (P ≥ 0.80). This study indicates that leaky gut induced by long-term administration of aspirin has negative impacts on feedlot performance and carcass composition. The negative impact of aspirin-induced leaky gut on animal performance suggests that chronic leaky gut caused by other factors (subacute acidosis, stress) may be a significant problem for the feedlot industry.

The objective was to determine the nutritional and functional values of lysed Corynebacterium glutamicum cell mass (CGCM) as a protein supplement and a source of cell wall fragments supporting the growth and intestinal health of nursery pigs. Thirty-two pigs (21 d of age) were allotted to four treatments (n = 8) based on the randomized block design with sex and initial body weight (BW) as blocks. The main effect was the dietary supplementation of lysed CGCM (0, 0.7, 1.4, and 2.1%) replacing blood plasma and fed in two phases (10 and 11 d, respectively). Feed intake and BW were measured at the end of each phase. Pigs were euthanized on day 21 to collect jejunal tissue and mucosa to evaluate intestinal health. Ileal digesta were collected to measure the apparent ileal digestibility of nutrients in diets. Data were analyzed using Proc Mixed and Reg of SAS. Increasing daily intake of CGCM increased (linear; P < 0.05) ADG of pigs. Increasing CGCM supplementation affected (quadratic; P < 0.05) the relative abundance of Lactobacillaceae (minimum: 26.4% at 1.2% CGCM), Helicobacteraceae (maximum: 29.3% at 1.2% CGCM), and Campylobacteraceae (maximum: 9.0% at 1.0% CGCM). Increasing CGCM supplementation affected (quadratic; P < 0.05) the concentrations of immunoglobulin G (maximum: 4.94 µg/mg of protein at 1.0% CGCM) and protein carbonyl (PC; maximum: 6.12 nmol/mg of protein at 1.1% CGCM), whereas linearly decreased (P < 0.05) malondialdehyde (MDA) in the proximal jejunal mucosa. Increasing CGCM supplemention affected (quadratic; P < 0.05) intestinal enterocyte proliferation rate (maximum: 13.3% at 1.0% CGCM), whereas it did not affect intestinal morphology and the nutrient digestibility. In conclusion, supplementing 1.0% to 1.2%, reducing blood plasma supplementation by 0.7% to 0.9%, respectively, increased potential pathogenic microbiota associated in the jejunal mucosa resulting in increased immune response, enterocyte proliferation, and PC concentration. However, supplementing diets with 2.1% CGCM, replacing 1.5% blood plasma, improved growth performance, and reduced MDA without affecting nutrient digestibility, intestinal morphology, and microbiota in the jejunal mucosa. In this study, based on the polynomial contrast, supplementing 1.0% to 1.2% CGCM suppressed the benefits from blood plasma, whereas supplementing 2.1% CGCM showed functional benefits of CGCM with similar effects from blood plasma supplementation.

Our aim was to determine whether the method used to estimate truly digestible neutral detergent fiber (tdNDF) affects calculated concentrations of total digestible nutrients (TDN(1x)) and net energy of lactation (NE(L3x)) of canola meal (CM). Samples were collected from 12 CM processing plants in Canada over 4 yr (2011 to 2014, n = 47) and analyzed for dry matter (DM), crude protein (CP), ether extract (EE), ash, neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin (ADL), and neutral detergent insoluble CP (NDICP). Ruminal in situ incubation of CM samples was performed at 0, 24, 48, 96, and 288 h to determine NDF fractions (A, B, and C), effective ruminal NDF digestibility (ERNDFD), and indigestible NDF (iNDF) of CM. Three tdNDF-estimation methods were evaluated: 1) National Research Council (NRC) = 0.75 × (NDF − NDICP − ADL) × {1− [ADL/ (NDF − NDICP)](0.667)}; 2) iNDF = 0.75 × (NDF − NDICP − NDF remaining after 288 h in situ); and 3) ERNDFD estimated from in situ NDF digestion kinetics. Resulting tdNDF values were used for calculation of TDN(1x) and NE(L3x) according to NRC (2001) equations. Data were analyzed with MIXED procedure of SAS 9.4 to determine the effect of processing plant on chemical composition, NDF degradation kinetics and NE(L3x) of CM. Effect of tdNDF estimation method on calculated TDN(1x) and NE(L3x) of CM was also evaluated. Model for analysis of processing plant included the fixed effect of plant and the random effect of year (plant) as replication, while analysis of tdNDF methods included the fixed effect of tdNDF estimation method and the random effects of processing plant and of year(plant) as replication. There was an effect of processing plant on DM (P = 0.03), CP (P < 0.01), EE (P < 0.01), and NDF (P < 0.01) of CM. Processing plant also had an effect on NDF fractions A (P < 0.01) and B (P = 0.02) but did not affect fraction C and ERNDFD. The tdNDF estimation method had an effect on tdNDF (P < 0.01), TDN(1x) (P < 0.01), and NE(L3x) (P < 0.01) of CM, yielding average NE(L3x) values of 1.72, 1.87, and 2.07 Mcal/kg for NRC, iNDF, and ERNDFD, respectively. Our results indicate that calculated energy concentration of CM according to NRC (2001) equations varies depending on the method used for estimation of tdNDF. Further research will be needed to determine the most accurate estimation method.

This study was conducted to evaluate the effects of feeding fermented wheat bran (FWB) and yeast culture (YC) on growth performance, immune levels, and intestinal microflora in growing-finishing pigs. In total, 96 crossbred pigs were randomly distributed into four treatments with four replicates pens and six pigs per pen. This study was performed using a 2 × 2 factor design: 1) CON (basal diet), 2) FWB (basal diet + 5% FWB), 3) YC (basal diet + 2% YC), and 4) FWB + YC (basal diet + 5% FWB + 2% YC). Dietary FWB supplementation significantly increased the average daily gain and significantly decreased the feed gain ratio of growing-finishing pigs (P < 0.05). Supplementation of FWB and YC improved the immune capacity and reduced the inflammation level of growing-finishing pigs (P < 0.05). In addition, pigs fed FWB, YC, and FWB + YC diets showed better intestinal development and morphology compared with those CON pigs. The relative abundance of Streptococcus in the FWB group was significantly lower than that in the CON group (P < 0.05), and the relative abundance of probiotics (unclassified_f_Lachnospiraceae, Turicibacter) increased significantly (P < 0.05). Furthermore, the relative abundance of probiotics (Lactobacillus, norank_f_Muribaculaceae) in the YC group was significantly increased compared with the CON group (P < 0.05). The results of this study observed positive effects of FWB and YC on growing-finishing pigs, which provides insights into the application of biological feed in swine industry.

This study was conducted to investigate the effects of combined supplementation of sodium humate (HNa) and glutamine (Gln) on growth performance, diarrhea incidence, serum parameters, intestinal microbiome, and metabolites of weaned calves. In Exp. 1, 40 calves were randomly assigned to four treatments: 1) NC (negative control, basal diet), 2) 1% H+1% G (basal diet extra orally gavaged with 1 g of HNa and 1 g of Gln daily), 3) 3% H+1% G (basal diet extra orally gavaged with 3 g of HNa and 1 g of Gln daily), and 4) 5% H+1% G (basal diet extra orally gavaged with 5 g of HNa and 1 g of Gln daily). The HNa and Gln were together mixed with 100 mL of milk replacer (51 to 58 d of age) or water (59 to 72 d of age) and orally administrated to each calf from a bottle before morning feeding. In a 21-d trial, calves on the 5% HNa+1% Gln group had higher (P < 0.05) average daily gain (ADG) and lower (P < 0.05) diarrhea incidence than those in the control group. In Exp. 2, 20 calves were randomly assigned to two treatments fed with a basal diet and a basal diet supplemented with 100 mL of 5% HNa+1% Gln. In a 21-d trial, calves supplemented with HNa and Gln had higher (P < 0.05) ADG, IgG concentration and glutathione peroxidase (GSH-Px), and total antioxidant capacity (T-AOC) activities in the serum, but lower (P < 0.05) diarrhea incidence, as well as serum diamine oxidase (DAO), D-isomer of lactic acid (D-lac), tumor necrosis factor-α (TNF-α), and malondialdehyde (MDA) concentrations compared with control group. Results of intestinal microbiota indicated that supplementation with HNa and Gln significantly increased (P < 0.05) the abundance of intestinal beneficial microbiota. Moreover, supplementation with HNa and Gln altered 18 metabolites and enriched 6 Kyoto Encyclopedia of Genes and Genomes pathways in weaned calves. In conclusion, combined supplementation with HNa and Gln could decrease diarrhea incidence of weaned calves via altering intestinal microbial ecology and metabolism profile.

The objective of this study was to investigate the effects of water quality on water intake (WI), forage intake, diet digestibility, and blood constituents in beef cows and growing beef heifers. This was a replicated 5 × 5 Latin square with five drinking water treatments within each square: 1) fresh water (Control); 2) brackish water (100 BRW treatment) with approximately 6,000 mg/kg total dissolved solids (TDS); 3) same TDS level as 100 BRW achieved by addition of NaCl to fresh water (100 SLW); 4) 50% brackish water and 50% fresh water to achieve approximately 3,000 mg/kg TDS (50 BRW); and 5) same TDS level as 50 BRW achieved by addition of NaCl to fresh water (50 SLW). Each of the five 21-d periods consisted of 14 d of adaptation and 5 d of data collection. Animals were housed individually and fed mixed alfalfa (Medicago sativa) grass hay cubes. Feed and WI were recorded daily. Data were analyzed with animal as the experimental unit. Age, treatment, and age × treatment were fixed effects, and animal ID within age was the random variable for intake, digestibility, and blood parameter data. Water and feed intake were greater than expected, regardless of age or water treatment. No treatment × age interactions were identified for WI (P = 0.71), WI expressed as g/kg body weight (BW; P = 0.70), or dry matter intake (DMI; P = 0.21). However, there was an age × treatment tendency for DMI when scaled to BW (P = 0.09) in cows consuming 100 BRW compared with fresh water. No differences were found for the other three treatments. Heifers provided 50 SLW water consumed less (P < 0.05) feed (g/kg BW) compared with heifers provided fresh water and 100 BRW. No differences (P > 0.05) in water, DMI, feed intake, or diet digestibility were found due to water quality treatment. In conclusion, under these conditions, neither absolute WI, absolute DMI, nor diet digestibility was influenced by the natural brackish or saline water used in this experiment. These results suggest that further research is necessary to determine thresholds for TDS or salinity concentration resulting in reduced water and/or feed intake and diet digestibility.

This study examined the effect of a water acidifier containing free and buffered short-chain fatty acids (SCFA-WA) on growth performance and microbiota of weaned piglets. In total, 192 male piglets, approximately 4 wk of age, were allocated to 24 pens (12 per treatment) with 8 piglets per pen. The piglets received either regular drinking water (negative control) or drinking water with the acidifier supplied at 2 L/1,000 L. Body weight and feed intake were measured weekly on pen level. During the first 2 wk, daily visual assessment and scoring of the feces was conducted. Fecal samples of three piglets per pen were collected on days 14 and 42 for high-throughput sequencing analysis of the microbiota. Piglets offered SCFA-WA had significantly improved feed efficiency in the third week (P = 0.025) and over the whole study period (days 0 to 42, P = 0.042) compared with piglets in the negative control group, with a strong tendency observed during the first feeding phase (days 0 to 21, P = 0.055). Furthermore, the water acidifier group had a higher water intake than piglets provided with control water during the second feeding phase (days 21 to 42, P = 0.028) and over the whole study period (days 0 to 42, P = 0.043). There was no significant difference in body weight, average daily gain, or average daily feed intake (days 0 to 21, 21 to 42, 0 to 42). Furthermore, there was no overall significant difference in fecal scoring between the treatments. In terms of the fecal microbiota response, piglets offered the water acidifier showed a significantly higher relative abundance (RA) of genus Clostridium sensu stricto 1 and a lower RA of genus Streptococcus compared to the control. Furthermore, the redundancy analysis showed a positive association between improved feed efficiency and daily weight gain and RA of Butyricicoccus and Faecalibacterium. In conclusion, consumption of the water acidifier containing free and buffered SCFA modulated the microbiota and improved feed efficiency in piglets.

The objectives of the present study were to estimate genetic parameters for several feeding behavior traits in growing cattle, as well as the genetic associations among and between feeding behavior and both performance and feed efficiency traits. An additional objective was to investigate the use of feeding behavior traits as predictors of genetic merit for feed intake. Feed intake and live-weight data on 6,088 growing cattle were used of which 4,672 had ultrasound data and 1,548 had feeding behavior data. Feeding behavior traits were defined based on individual feed events or meal events (where individual feed events were grouped into meals). Univariate and bivariate animal linear mixed models were used to estimate (co)variance components. Heritability estimates (± SE) for the feeding behavior traits ranged from 0.19 ± 0.08 for meals per day to 0.61 ± 0.10 for feeding time per day. The coefficient of genetic variation per trait varied from 5% for meals per day to 22% for the duration of each feed event. Genetically heavier cattle, those with a higher daily energy intake (MEI), or those that grew faster had a faster feeding rate, as well as a greater energy intake per feed event and per meal. Better daily feed efficiency (i.e., lower residual energy intake) was genetically associated with both a shorter feeding time per day and shorter meal time per day. In a validation population of 321 steers and heifers, the ability of estimated breeding values (EBV) for MEI to predict (adjusted) phenotypic MEI was demonstrated; EBVs for MEI were estimated using multi-trait models with different sets of predictor traits such as liveweight and/or feeding behaviors. The correlation (± SE) between phenotypic MEI and EBV for MEI marginally improved (P < 0.001) from 0.64 ± 0.03 to 0.68 ± 0.03 when feeding behavior phenotypes from the validation population were included in a genetic evaluation that already included phenotypic mid-test metabolic live-weight from the validation population. This is one of the largest studies demonstrating that significant exploitable genetic variation exists in the feeding behavior of young crossbred growing cattle; such feeding behavior traits are also genetically correlated with several performance and feed efficiency metrics. Nonetheless, there was only a marginal benefit to the inclusion of time-related feeding behavior phenotypes in a genetic evaluation for MEI to improve the precision of the EBVs for this trait.

Two experiments were performed to evaluate the effects of bismuth subsalicylate (BSS) and calcium-ammonium nitrate (CAN) on in vitro ruminal fermentation, growth, apparent total tract digestibility of nutrients, liver mineral concentration, and carcass quality of beef cattle. In Exp. 1, four ruminally cannulated steers (520 ± 30 kg body weight [BW]) were used as donors to perform a batch culture and an in vitro organic matter digestibility (IVOMD) procedure. Treatments were arranged in a 2 × 2 factorial with factors being BSS (0 or 0.33% of substrate dry matter [DM]) and CAN (0 or 2.22% of substrate DM). In Exp. 2, 200 Angus-crossbred steers (385 ± 27 kg BW) were blocked by BW and allocated to 50 pens (4 steers/pen) in a randomized complete block design with a 2 × 2 + 1 factorial arrangement of treatments. Factors included BSS (0 or 0.33% of the diet DM) and nonprotein nitrogen (NPN) source (urea or encapsulated CAN [eCAN] included at 0.68% or 2.0% of the diet, respectively) with 0.28% ruminally available S (RAS). A low S diet was included as a positive control containing urea (0.68% of DM) and 0.14% RAS. For Exp. 1, data were analyzed using the MIXED procedure of SAS with the fixed effects of BSS, CAN, BSS × CAN, and the random effect of donor. For Exp. 2, the MIXED procedure of SAS was used for continuous variables and the GLIMMIX procedure for categorical data. For Exp. 1, no differences (P > 0.230) were observed for IVOMD. There was a tendency (P = 0.055) for an interaction regarding H2S production. Acetate:propionate increased (P = 0.003) with the addition of CAN. In Exp. 2, there was a NPN source effect (P = 0.032) where steers consuming urea had greater carcass-adjusted final shrunk BW than those consuming eCAN. Intake of DM (P < 0.001) and carcass-adjusted average daily gain (P = 0.024) were reduced by eCAN; however, it did not affect (P = 0.650) carcass-adjusted feed efficiency. Steers consuming urea had greater (P = 0.032) hot carcass weight, and a BSS × NPN interaction (P = 0.019) was observed on calculated yield grade. Apparent absorption of S decreased (P < 0.001) with the addition of BSS. Final liver Cu concentration was reduced (P = 0.042) by 58% in cattle fed BSS, indicating that BSS may decrease Cu absorption and storage in the liver. The results observed in this experiment indicate that BSS does not have negative effects on feedlot steer performance, whereas CAN may hinder performance of steers fed finishing diets.

In a previous study, we examined the synergistic effects of the dietary supranutritional supplementation with vitamin E, vitamin C, and Se on the in vivo antioxidative status of broilers under conditions of dietary oxidative stress induced by feeding a diet high in n-3 PUFA. In this study, we examined the effect of their inclusion on the quality characteristics and oxidative stability of raw or cooked meat, both fresh or after a long-term frozen storage. Four hundred 21-day-old Ross 308 male broilers were allocated to 5 experimental groups fed 5% linseed oil-enriched finisher diets (days 21 to 40): Cont (recommended levels of vitamin E, C, and selenium), +E (200 IU vitamin E/kg feed), +C (250 mg vitamin C/kg feed), +Se (0.2 mg selenium/kg feed), or +ECSe (concentrations as in the sole supplementation, combined). Animal performance and carcass characteristics were monitored at the age of 40 D. Breast meat samples of 12 chickens per group were analysed fresh, fresh after frozen storage, cooked fresh, and cooked after frozen storage (2 × 2 factorial design) for parameters of meat quality (water-holding capacity—WHC, pH, and color) and oxidative stability (concentrations of vitamin E, malondialdehyde—MDA, antioxidant capacity of the water-soluble compounds—ACW, and fatty acid composition). Vitamin E alone (+E) and combined with Se and vitamin C (+ECSe) increased the α-tocopherol concentration in breast muscle, and showed similar protective effects against lipid peroxidation measured as MDA regardless of the frozen storage or cooking. The sole supplementation of vitamin C or selenium showed no effects on the meat quality parameters. In conclusion, the dietary supranutritional inclusion of vitamin E inhibited the lipid peroxidation in fresh, frozen stored, cooked fresh, and frozen stored meat in broilers fed with diets rich in n-3 PUFAs. Even though no clear synergistic effects of the supranutritional supplementation of vitamin C and Se with vitamin E were detected, their dietary inclusion did not negatively affect broilers carcass and meat quality parameters.

This study aimed to evaluate the impact of grading levels of deoxynivalenol (DON) in the diet of weaned pigs, as well as the effects of a supplementation with antioxidants (AOX), hydrated sodium calcium aluminosilicates (HSCAS), and their combination on the growth, AOX status, and immune and vaccine responses against the porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2). At weaning, 336 piglets were allocated to six dietary treatments according to a randomized complete block design. Treatments were as follows: basal diet (CTRL); basal diet containing DON at 1.2 mg/kg (DON1.2); basal diet containing DON at 2.4 mg/kg (DON2.4); DON2.4 diet + a mix of AOX which included vitamins A and E at 20,000 IU and 200 IU/kg feed respectively, selenized yeast at 0.3 mg/kg, and a grape seed extracts at 100 mg/kg feed (DON2.4 + AOX); DON2.4 diet + the mix of AOX and the modified HSCAS mentioned above (DON2.4 + AOX + HSCAS); DON2.4 + AOX + HSCAS. Pigs were vaccinated against PRRSV and PCV2 at 7 d; on 0, 14, and 35 d, growth performance was recorded, and blood samples were collected in order to evaluate the oxidative status, inflammatory blood markers, lymphocyte blastogenic response, and vaccine antibody response. Increasing intake of DON resulted in a quadratic effect at 35 d in the lymphocyte proliferative response to concanavalin A and PCV2 as well as in the anti-PRRSV antibody response, whereas the catalase activity decreased in DON2.4 pigs compared with the CTRL and DON1.2 groups (P ≤ 0.05). Compared with the DON2.4 diet, the AOX supplementation slightly reduced gain to feed ratio (P = 0.026) and increased the ferric reducing ability of plasma as well as α-tocopherol concentration (P < 0.05), whereas the association of AOX + HSCAS increased the anti-PRRSV IgG (P < 0.05). Furthermore, the HSCAS supplement reduced haptoglobin levels in serum at 14 d compared with the DON2.4 group; however, its concentration decreased in all the experimental treatments from 14 to 35 d and particularly in the DON2.4 + AOX pigs, whereas a different trend was evidenced in the DON2.4 + HSCAS group, where over the same period haptoglobin concentration increased (P < 0.05). Overall, our results show that the addition of AOX and HSCAS in the diet may alleviate the negative effects due to DON contamination on the AOX status and immune response of vaccinated weanling pigs.

Adequate drinking water is essential to maintain acceptable production levels in beef cattle operations. In the context of global climate change, the water scarcity forecasted for the future is a growing concern and it would determine an increase in the use of poorer quality water by the agricultural sector in many parts of the world. However, consumption of high-salt water by cattle has consequences often overlooked. A meta-analysis was carried out to assess the impact of utilizing high-salt water on dry matter (DMI) and water intake (WI), and performance in beef cattle. The dataset was collected from 25 studies, which were conducted between 1960 and 2020. Within the dataset, the water quality was divided into three categories according to the ratio of sulfates (SO4) or sodium chloride (NaCl) to total dissolved solids (TDS): 1) TDS = all studies included (average SO4:TDS = 0.4); 2) NaCl = considered studies in which water salinity was dominated by NaCl (average SO4:TDS = 0.1); and 3) SO4 = considered studies in which water salinity was dominated by SO4 (average SO4:TDS = 0.8). Results showed that DMI and WI were negatively affected by high-salt water consumption, although the magnitude of the effect is dependent on the type of salt dissolved in the water. There was a quadratic effect (P < 0.01) for the WI vs. TDS, WI vs. NaCl, DMI vs. TDS, and DMI vs. NaCl, and a linear effect (P < 0.01) for WI vs. SO4 and WI vs. SO4. Average daily gain (ADG) and feed efficiency (FE) were quadratically (P < 0.01) affected by high-salt water, respectively. This study revealed significant negative effects of high-salt water drinking on beef cattle WI, DMI, and performance. However, the negative effects are exacerbated when cattle drink high-sulfate water when compared with high-chloride water. To the best of our knowledge, this is the first approach to evaluate animal response to high-salt water consumption and could be included in the development of future beef cattle models to account for the impact of water quality on intake and performance. In addition, this meta-analysis highlights the need for research on management strategies to mitigate the negative effects of high-salt water in cattle.

Six ruminally cannulated steers (average BW = 791 + 71 kg) were used in a replicated 3 × 3 Latin square experiment to determine the effects of roughage type on rumination, fiber mat characteristics, and rumen fermentation variables. Three roughages were included at 7% (DM basis) in a steam flaked corn-based diet; cotton burrs (CB), wheat silage (WS), or corn stalks (CS). Steers were fitted with a sensory collar to record rumination behaviors in 2-h intervals at the beginning of the experiment. Each 30-d period consisted of a 7-d of recovery, 14-d of diet adaptation, 7-d of rumination data collection (daily and bi-hourly average rumination), 1-d of rumen fluid collection, and 1-d of rumen evacuations. In situ degradation of individual roughages was determined for 4-d after period 3 evacuations. During rumen evacuations, ruminal contents were removed; the rumen fiber mat (RF) was separated from the liquid portion with a 2 mm sieve, weighed, and a subsample was dried. Data were analyzed using the MIXED procedure of SAS with steer as the experimental unit and roughage (CB, WS, and CS) as the main effect. Dry matter intake (DMI) was not different for CB and WS (P = 0.25) and greatest for steers consuming CS diet (P < 0.01). Roughage type did not influence the weight of the RF dry matter (%; DM; P = 0.92), RF weight (P = 0.69), or RF:DMI ratio (P = 0.29). Daily rumination (min/d) did not differ among roughages (P = 0.40), but min of rumination/kg of DMI was greatest for CS (18.0 min), min/kg of NDF was greatest for WS (89.8 min; P = 0.02), and min/kg of peNDF was greatest for CS (132.4 min; P < 0.01). Wheat silage had the greatest percentage of soluble DM and CB-R and CS-R (P < 0.01) had the greatest ruminal degraded DM fraction. Rumen fiber mat did not differ for roughages, although rumination min/kg of DMI and peNDF was greatest for steers consuming CS and WS. In situ degradation determined that CB-R and CS-R had the greatest percentage of ruminal degraded DM. Based on the objective of the experiment, roughage type did not influence daily rumination or fiber mat characteristics.
