Laboratory mice are typically housed in "shoebox" cages with limited opportunities to engage in natural behaviour. Temporary access to environments with increased space and complexity (playpens) may improve mouse welfare. Previous work by our group has shown that mice are motivated to access and use these environments, but it is unknown how other aspects of welfare are impacted. Female C57BL/6J, BALB/cJ, and DBA/2J mice (n = 21; 7 mice per strain) were housed in mixed-strain trios and given temporary access to a large playpen with their cage mates three times per week. Control mice (n = 21; 7 mice per strain) remained in their home cages. Home cage behaviour (development of stereotypic behaviour over time, aggression following cage-changing) and anxiety tests were used to assess how playpen access impacted welfare. Contrary to our predictions, we found increased time spent performing stereotypies in playpen mice; this difference may be related to negative emotional states, increased motivation to escape the home cage, or active coping strategies. Playpen access resulted in strain-dependent improvements in aggression and some measures of anxiety. Aggression was lower for C57BL/6J mice in the playpen treatment following cage changing than it was for C57BL/6J control mice, while playpen mice, and particularly the C57BL/6J strain, spent more time in the center of the open field test and produced fewer fecal boli during anxiety testing, supporting other research showing that strain differences play an important role in behaviour and stress resiliency., (© 2024. 3. Exploring the effect of pain on response to reward loss in calves.

- Abstract
Negative emotional states are known to interact, potentially aggravating one another. In this study, we used a well validated paradigm (successive negative contrast, SNC) to determine if pain from a common procedure (disbudding) influences responses to a reward downshift. Holstein calves (n = 30) were trained to approach a 0.5 L milk reward. Latency to approach, number of vocalisations and pressure applied on the bottle were recorded during training. To assess how pain affected responses to reward downshift, calves were randomly assigned to one of three treatments before the downshift. Two groups were disbudded and provided the 'gold standard' of pain mitigation: intraoperative local anesthesia and analgesia. One of these disbudded groups was then provided supplemental analgesic before testing. The third group was sham disbudded. All calves were then subjected to the reward downshift by reducing the milk reward to just 0.1 L. Interactions were detected between test session and daily trials on pressure applied for the Disbudded group (estimate ± SEM: 0.08 ± 0.05), and on vocalisations for the Sham (0.3 ± 0.1) and Disbudding + Analgesia (0.4 ± 0.1) groups. Our results indicate that SNC is a promising paradigm for measuring negative affect in calves and suggests that pain potentially affects the response to a reward downshift., (© 2023. 4. Calves peak-end memory of pain.

In humans, the 'peak-end' rule states that recollection of an experience is most often influenced by the peak (the most intense moment) and end of the experience. We investigated whether calves followed the peak-end rule in their memory of a painful procedure: disbudding. As proxies for retrospective and 'real-time' reports of pain, we used conditioned place aversion, and reflex pain behaviours. In two separate trials, calves were subjected to two disbudding conditioning sessions (one horn per treatment), acting as their own control. In the first trial, calves (n = 22) were disbudded and remained in a pen for 4 h, and disbudded and left in another pen for 4 h with an additional 2 h following an analgesic treatment. In the second trial, calves (n = 22) were disbudded and left in pens for 6 h during both treatments, receiving the analgesic at either 2 h or 4 h after disbudding. Calves were then tested for place aversion. For both trials we did not observe a preference for the pens where calves received analgesic treatment towards the end of the session. We did not find an association between aversion and the sum, peak or end of pain behaviours. Results are not consistent with a peak-end effect in calves' memory of pain., (© 2023. The Author(s).)

An animal's social position within a group can influence its ability to perform important behaviours like eating and resting, but little is known about how social position affects the ability to express what are arguably less important but still rewarding behaviors, such as grooming. We set out to assess if dominance measured at the feeder is associated with increased use of a mechanical brush. Over a 2-year period, 161 dry cows were enrolled in a dynamically changing group of 20 individuals with access to a mechanical brush. We determined dominance using agonistic behaviors at the feeder and retrospectively analyzed brush use for the 12 most, and 12 least dominant individuals during the week before calving. Cows that were more dominant at the feeder used the brush more, especially during peak feeding times. Agonistic interactions at the brush did not differ between dominants and subordinates and were not related to brushing duration. These findings indicate that social position, calculated using competition for feed, affects mechanical brush access such that subordinates use the brush less than dominant cows independent of competition or time of day., (© 2021. 6. Using approach latency and anticipatory behaviour to assess whether voluntary playpen access is rewarding to laboratory mice.

Laboratory mice are typically housed in "shoebox" cages that limit the expression of natural behaviours. Temporary access to more complex environments (playpens) may improve their welfare. We aimed to assess if access to playpens is rewarding for conventionally-housed mice and to document mouse behaviour during playpen access. Female C57BL/6J, BALB/cJ, and DBA/2J mice were provided temporary access to a large enriched playpen three times per week; control mice remained in their home cages. We measured latency to enter playpens and anticipatory behaviour to determine if access was rewarding, and recorded mouse behaviour during playpen sessions. Over time, playpen mice entered the playpen more quickly; latency declined from 168 ± 22 to 13 ± 2 s over the 14-d trial. As expected, playpen mice showed an increase in anticipatory behaviour before playpen access (mean ± SE = 19.7 ± 2.6 behavioural transitions), while control mice showed no change in anticipatory behaviour relative to baseline values (2.4 ± 1.6 transitions). Mice in the playpen performed more ambulatory behaviours than control mice who remained in home cages (21.5 ± 0.7 vs 6.9 ± 1.1 observations of 25 total observations). We conclude that conventionally-housed mice find voluntary playpen access rewarding, and suggest this as a useful option for providing laboratory mice with access to more complex environments., (© 2021. 7. Effect of cow-calf contact on cow motivation to reunite with their calf.

Early cow-calf separation prevents much of cows' natural maternal behaviour. Early separation is thought to prevent the development of a cow-calf bond. To assess this bond, we measured motivation of dairy cows to reunite with their calf. To vary the degree of bonding, some cows were allowed continued contact with their calf and others were separated from their calf soon after birth, following standard practice on most farms. Among cows allowed continued contact, some were able to suckle their calf and others were prevented from suckling (by covering the cow's udder with an udder net). Cows were habituated to the weighted-gate apparatus before calving by daily training with the (un-weighted) gate. After calving, cow willingness to use the gate was assessed by determining if she would push open the gate to access to her own calf. Testing occurred once daily, with weight on the gate gradually increased. After passing through the gate, the dam's calf-directed behaviour was recorded. Suckled cows pushed a greater maximum weight (45.8 ± 7.8 kg) than separated cows (21.6 ± 6.7 kg) and non-suckled cows (24.3 ± 4.5 kg), with no differences between separated and non-suckled cows. Once reunited, latency to make nose contact and duration of licking did not differ between treatments. We conclude that motivation for calf contact is greater for cows that are suckled.

8. Use of a food neophobia test to characterize personality traits of dairy calves.

Food neophobia, i.e. the avoidance of novel foods, is common in ruminants and may provide a biologically relevant and practical way to test individual responses to novelty or challenge. We aimed to determine if behavioural responses in a food neophobia test (exposure to a novel total mixed ration) reflected boldness and exploratory personality traits derived from 3 traditional tests (open field, novel human and novel object) in dairy calves. We performed two Principal Component Analyses, one using behaviours from 3 traditional tests (3 factors: 'Bold', 'Exploratory' and 'Active'), and one using behaviours from the food neophobia test (3 factors: 'Eating', 'Inspecting', and 'Avoidance'). A regression analysis determined if individual factor scores from the food neophobia test predicted factor scores from the traditional tests. Contrary to our expectations, 'Avoidance' (latencies to approach and eat the novel food) did not predict boldness trait, and the factors 'Inspecting' (time spent inspecting food and empty buckets) and 'Eating' (time spent eating food and total intake) did not predict exploration trait, but they did predict active trait. These results suggest that the food neophobia test in our study resulted in context-specific behaviours, or that behavioural responses to a novel food present different underlying personality traits. The application of food neophobia to assess specific or generalized personality traits of dairy calves deserves further work.

9. Assessing the motivation to learn in cattle.

Cognitive challenges may provide a form of enrichment to improve the welfare of captive animals. Primates, dolphins, and goats will voluntarily participate in learning tasks suggesting that these are rewarding, but little work has been conducted on livestock species. We investigated the motivation of 10 pairs of Holstein heifers to experience learning opportunities using a yoked design. All heifers were trained to perform an operant response (nose touch) on a variable interval schedule. Learning heifers then performed this response to access a discrimination learning task in which colour and texture of feed-bin lids signified a preferred reward (grain) vs. a non-preferred reward (straw). Control heifers received the same feed without a choice of bins or association of feed with lid type. Learning heifers approached the target to begin sessions faster (p = 0.024) and tended to perform more operant responses (p = 0.08), indicating stronger motivation. Treatments did not differ in the frequency with which heifers participated in voluntary training sessions. We conclude that heifers are motivated to participate in learning tasks, but that aspects of the experience other than discrimination learning were also rewarding. Cognitive challenges and other opportunities to exert control over the environment may improve animal welfare.

Carbon dioxide (CO
2 ) is commonly used to kill laboratory rats. Rats find CO2 aversive and aversion varies between individuals, indicating that rats vary in CO2 sensitivity. Healthy humans experience feelings of anxiety at concentrations similar to those avoided by rats, and these feelings are diminished by the administration of benzodiazepines. Our aim was to assess the effects of the benzodiazepine midazolam on individual thresholds of rat aversion to CO2 . Six female Sprague Dawley rats were repeatedly exposed to CO2 gradual-fill in approach-avoidance testing. The first three exposures were to a control-treatment followed by three exposures to midazolam (0.375 mg/kg). Within each treatment aversion to CO2 was not affected by exposure number; however, tolerance increased from an average of 10.7% CO2 avoided during control sessions, to 15.5% CO2 avoided when treated with midazolam. These results indicate that rats experience anxiety when exposed to CO2 , and that variation in rat CO2 sensitivity is driven by individual differences in the onset of these feelings of anxiety. No rat tolerated CO2 concentrations required to induce loss of consciousness.- Published

11. Standard laboratory housing for mice restricts their ability to segregate space into clean and dirty areas.

Laboratory mice (Mus musculus) are typically housed in simple cages consisting of one open space. These standard cages may thwart mouse ability to segregate resting areas from areas where they eliminate, a behaviour that is prevalent across the animal kingdom. No scientific work has directly tested whether mice engage in such segregation behaviour, or whether the ability to do so may have welfare consequences. Here we show that mice, whether housed in standard cages or a complex housing system consisting of three interconnected standard cages, kept nesting and elimination sites highly segregated, with nest and urine co-occurring in the same location only 2% of the time. However, mice in the complex system established these clean and dirty sites in separate cages instead of separate locations within one cage, and carried bedding materials (cellulose pellets) from their nesting cages to their latrine cage. Moreover, mice in the complex system displayed more behaviours associated with positive welfare and were less disturbed by weekly husbandry procedures. We conclude that mice find waste products aversive, and that housing mice in a way that facilitates spatial segregation provides a simple way of allowing the expression of natural behaviours and improving welfare.

12. Calf aversion to hot-iron disbudding.

Dairy calves are routinely disbudded by cauterization with a hot iron. To mitigate the intra-operative and initial post-operative pain associated with this procedure some farmers provide calves general and local anesthetics, but it is unknown if the procedure remains aversive. We used a place-conditioning paradigm to assess aversion caused by hot-iron cautery with a local anesthetic compared to a sham procedure. A test area was divided into three equally sized pens: two 'treatment' pens with distinct visual cues were connected by a central 'neutral' pen. Each calf went through the disbudding procedure and a 6-h recovery period in one treatment pen and the control procedure in the other treatment pen. In three tests (48, 72 and 96 h after the second treatment), calves could freely roam among the pens until they chose to lie down, ending the session. Calves spent less time in either of the treatment pens compared to the central pen. When only comparing the two treatment pen, calves spent less time in the disbudding pen, especially during the first test. Calves were also less likely to lie down in the pen associated with the disbudding procedure. We conclude that even with the use of a local anesthetic, hot-iron disbudding is salient and aversive for calves, indicating the need to refine or avoid the procedure.

The assessment of individual traits requires that tests are reliable (i.e. consistency over time) and externally valid, meaning that they predict future responses in similar contexts (i.e. convergent validity) but do not predict responses to unrelated situations (i.e. discriminant validity). The aim of this study was to determine if dairy calf personality traits (Fearfulness, Sociability and Pessimism), derived from behaviours expressed in standardized tests, predict individuals' responses in related situations. The first experiment tested if the trait 'Sociability' was related to the expression of social behaviour in the home-pen, with calves assigned individual proximity scores (based on proximity to other calves) while they were in their home-pen at approximately 113 and 118 d of age. The second experiment aimed at exploring whether traits 'Fearfulness' and 'Pessimism' were related to the calves' emotional response to transportation. All calves were subjected to two 10-min transportation challenges done on two consecutive days. Emotional response was assessed using the maximum eye temperature (measured using infrared thermography) and the number of vocalizations emitted. Social proximity scores (Experiment 1), vocalizations emitted and maximum eye temperature after loading (Experiment 2) were consistent over time. In addition, the results showed good convergent validity with calves scoring higher in Sociability also having higher proximity scores in the home-pen, and animals scoring higher in Fearfulness and Pessimism showing a more intense emotional response to transportation. The results also showed good discriminant validity, as neither Fearfulness nor Pessimism were associated with the expression of social behaviours (Experiment 1) and Sociability was not associated with the animal's emotional response to transportation (Experiment 2). We conclude that the methodology used to measure personality traits shows good reliability and external validity.

We assessed aversion to injections using an avoidance-learning paradigm. Holstein calves (n = 24) were randomly assigned to one of four routes of administration for 0.5 ml of saline: intramuscular (IM), intranasal (IN), subcutaneous (SC) and a null control. Calves were first trained to approach a milk reward of 1 L. Once the latency to approach the reward was consistent, calves received their assigned treatment when approaching the bottle. For the first 3 treatment sessions calves received a 1 L milk reward. This reward was then reduced to 500 mL, and then to 250 mL, and finally to 0 mL, each for 3 sessions. Compared to control calves, calves receiving the intramuscular injections showed a longer latency to approach the milk reward, but only when the milk reward was 0.25 L (P = 0.05) and 0 L (P < 0.01). Calves receiving the intranasal injections showed longer latencies relative to the controls only for the 0 L reward (P = 0.01). Calves receiving the subcutaneous injections did not differ from controls for any of the milk rewards (P > 0.2). We conclude that IM injections are aversive and that SC and IN routes are a refinement to be considered when feasible.

Animals that experience situations likely to induce negative emotions show changes in judgment associated with pessimism. Few studies have focused on whether animals express stable differences in pessimism and whether these differences are related to personality traits. The first aim of this study was to explore if dairy calves are consistent over time in making judgments under ambiguous situations. Our second aim was to determine whether individual differences in judgment bias are related to conventional personality traits assessed using four standardized tests (Open field, Novel object, Human reactivity and Social motivation test). We subjected animals to two sessions of judgment bias and personality trait tests at 25 and 50 d of age. Individual differences in judgment bias were consistent over time with some animals persistently making more pessimistic choices compared to others. Two main dimensions of personality (Fearfulness and Sociability), obtained through principal component analysis, were also highly consistent over time. Pessimism was related to fearfulness, with more fearful calves making more pessimistic judgments. We conclude that dairy calves differ in the way they perceive and react to ambiguity and that this relates to individual differences in fearfulness.

Many dairy cows in the developed world are now housed exclusively indoors with fewer than 5% of the 10 million lactating cows in the United States having access to pasture during the grazing season. Indoor housing systems are designed to meet biological needs for food, water, hygiene, and shelter, but surveys of public and farmer opinion suggest that people think that pasture access is also important for the well-being of dairy cows. To determine if pasture access is important to the cows themselves, we investigated to what extent cows will work to access pasture (by pushing on a weighted gate), and compared it to the motivation to access fresh feed. Cows worked at least as hard to access pasture as they did to access the fresh feed and worked hardest for outdoor access in the evening hours. Echoing public views on what allows for a good life for cattle, these results show that cows are highly motivated for outdoor access.
