Aim: Herbal products are increasingly used as therapeutic agents in the prevention and management of periodontal diseases to preclude the side effects of antibiotics. A recent addition to the list is the leaves of P guajava, as they possess antimicrobial, anti-inflammatory, and antioxidant properties. This study evaluates the effects of the adjunctive use of 3% P. guajava local drug delivery in the management of chronic periodontitis. Materials and Methods: This was a split-mouth study involving contralateral sites in 15 patients (30 sites), who were randomly treated with either 3% P. guajava gel local drug delivery along with scaling and root planing (SRP) or SRP alone. The primary objective was to evaluate pocket probing depth (PPD), clinical attachment level (CAL), and colony counts of A.actenomycetemcomitans (Aa) and P.gingivalis (Pg); the secondary objective was to evaluate plaque, gingival, and bleeding scores. All parameters were assessed at baseline, one month and three months. The data were analyzed by using SPSS 18.5. Results: There was a significant overall improvement in clinical parameters over the study period. There was a statistically significant reduction in site-specific indices, PPD (2.74 ± 0.283), and gain in CAL (2.8 ± 0.152) in the test sites at three months. Microbiological analysis showed a significant reduction in the colony counts of Aa (17.4 ± 0.026) and Pg (22.7 ± 1.225) in the test sites at one and three months. (P < 0.001). Conclusion: Improvements in clinical and microbiological parameters showed that adjunctive use of locally delivered 3% P. guajava gel is effective in the management of chronic periodontitis.

Aim To evaluate the antimicrobial potential of irreversible hydrocolloid impression material manipulated using chitosan impregnated solution at various time intervals. Setting and design Evaluative invivo study design. Materials and methods Maxillary impressions made for 20 dentulous volunteers using irreversible hydrocolloid impression material manipulated using distilled water as control and using 1% chitosan impregnated solution as test group using stock metal trays with one-week interval. Bacterial samples were collected using dry sterile cotton swab in the mid-palatal region at the time intervals of 0, 10, 20 minutes. Bacterial swabs were inoculated on nutrient agar media and incubated at 37° C for 24 hours. Bacterial colonies were counted with the aid of colony counter. Statistical analysis used The resultant data was subjected to statistical analysis using repeated measures ANOVA and independent t test. Results Adding water soluble chitosan to irreversible hydrocolloid impression material resulted in superior antimicrobial activity. With the passage of time there was a significant decrease in the microbial colony count upto 10min (p=0.016). However, the rate of decrease of microbial colony count was statistically insignificant between the samples collected at 10 and 20 min. Conclusion Incorporation of water soluble chitosan to irreversible hydrocolloid impression material showed significant antimicrobial activity in 10 minutes.

Introduction: For decades now, head and neck cancer (HNC) remains to be one of the deadliest; per se the treatment for the same has been evolving for the past years. Radiation therapy (RT) has been one among the various treatment modalities adopted to treat HNC. The fact that RT can affect the quality of life in these patients cannot be denied. In patients with HNC, these can bring about a wide array of lesions in oral cavity and its associated structures as these areas are invariably affected due to exposure to radiation as such. These include alteration in salivary parameters, changes in microbial flora, and occurrence of radiation caries. Aim: In our study we aimed at assessing the alteration in oral microbial flora inclusive of Streptococcus mutans and Lactobacillus colony count before and after RT. Materials and Methods: Saliva and plaque samples were collected from patients with HNC, who were prescribed to undergo intensity-modulated radiation therapy (IMRT) amounting to a mean radiation dosage of 60 Gy units. The aforementioned samples were collected at three different intervals, before, immediately after, and 6 months after RT. The samples were cultured in selective media in three different dilutions (1:10, 1:100, and 1:1000). Following with colony counting is carried out using a standardized colony counter. Conclusion: The results showed a significant alteration in microbial flora pre- and post-RT. Individual parameters do not show a single stand in causing these changes. Moreover, these changes seem to have earned a multifactorial contribution.

Introduction: Cross-infection or contamination are the major threats related to any medical profession. Microorganisms present in the dental clinic can cause cross-infection to the dentist, auxiliary staff and even towards the patients. Aims: The study was conducted to assess the level of atmospheric microbial contamination and composition of aerosols before, during and after dental treatment procedures in four clinical settings. Methods and Materials: The present study was conducted over a two-week period in a private dental college setting. An equal number of culture medium plates (blood agar) were placed 30 min prior to the initiation of work sessions in the selected area and 1 h after the working session began and after 2 h of cessation of the working period. After the collection of samples, the culture medium plates were incubated aerobically at 37°C in an incubator for 48 h. The number of colonies was expressed as colonies per media plate. After counting the colonies bacterial cell morphology was determined by a microscopic examination using a Reichert-Jung Series 150 light microscope. Statistical Analysis: Statistical analysis such as ANOVA test for mean values and post hock was done using statistical package for social sciences (SPSS). Results: It shows that colony count increased after the working session and which reduced by itself once the working session was concluded which was significant (P < 0.001). The highest increase in the mean colony count was found in the department of periodontology during the treatment sessions. In the blood agar plates, the S. epidermidis was found maximum 62%, micrococcus was 22%, diphtheroid was 10%, fungi 4% and the least S. aureus 2%. Conclusion: This study demonstrates that aerosols increase during and after work sessions and, therefore, increases the chance for infectious agent transmission in clinical settings.

Objectives: The purpose of this study is to assess the association of passive smoking (PS) with dental caries and salivary biomarkers among 5–10 years old children of Muradnagar, Ghaziabad. Methods: A case–control study was conducted among 160 children of age group 5–10 years who visited the outdoor patient department of a dental college. Regular smoking households were recognized and children who lived in smoking households were identified as PS subjects. Two categories of children were formed – PS (80 children) and control group (80 children). Parents completed a pretested questionnaire and clinical examination of children was done using dmft index and gingival index. This was followed by collection of stimulated saliva of children which was further subjected to determine salivary buffering capacity and pH. Inoculation on mitis salivarius-bacitracin agar for counting streptococcus colonies and Rogosa SL agar (Difco) for counting lactobacillus colonies was done. Cotinine level was then measured using enzyme-linked immunosorbent assay kit. Student's independent t-test, Mann–Whitney U test, and one-way analysis of variance test were used for analyzing data. Results: The mean streptococcus and lactobacillus colony count was higher in PS case subjects, that is, 348.9 ± 166.509 and 247.3 ± 15.86 in comparison to control group where the mean streptococcus and lactobacillus colony count was 63.03 ± 23.082 and 63.825 ± 12.638, respectively. The mean cotinine level among PS case subjects was 1.08 ± 0.265 which was higher than the control group, that is, 0.00 ± 0.00. The mean cotinine level was directly proportional to streptococcus colonies, lactobacillus colonies, dmft and gingival index (GI) scores, and smoking exposure. Conclusion: PS has deleterious impact on children which was reflected by their increased cotinine levels, streptococcus colonies, lactobacillus colonies, and poor dmft and GI scores in comparison to the control group.

Aims: Green tea is an antibacterial agent with no significant side effect. This feature makes green tea safe for children to use. The purpose of this study was to compare the effectiveness of green tea gel and mouth rinse on salivary level of Streptococcus mutans and Lactobacillus of teenagers aged 12–18 years. Subjects and Methods: In this randomized controlled clinical trial study, 30 children aged 12–18 years were included in the study according to the inclusion criteria and were randomly divided into two groups. Participants in mouth rinse group were asked to rinse their mouth with 0.5% green tea mouthwash twice a day for 2 weeks. In the gel group, participants were requested to brush their teeth with 0.5% green tea gel twice a day for 2 weeks. After 4 weeks of washout period, mouthwash group applied the gel and the gel group rinsed the mouthwash for 2 weeks and with the same instruction as mentioned. Colony count of S. mutans and Lactobacillus was determined before and after intervention and data were analyzed using t- test. Results: According to the independent t- test, there was no significant difference in the salivary levels of S. mutans before and after intervention regarding age and gender (P = 0.33). Results from paired t-test showed significant decrease in the mean count of S. mutans and Lactobacillus colonies in both groups before and after intervention (P < 0.001). Conclusions: Green tea gel and mouthwash contribute to a significant reduction of salivary levels of S. mutans and Lactobacillus colonies, with a greater effect of mouthwash than the green tea gel, which was not statistically significant.

To investigate and optimize microbial media that substitute peptone agar using brebra seed defatted flour.Defatted process, inoculums preparation, evaluation of bacterial growth, preparation of cooked and hydrolyzed media and growth turbidity of tested bacteria were determined.Two percent defatted flour was found to be suitable concentration for the growth of pathogenic bacteria: Escherichia coli (ATCC 25922) (E. coli), Pseudomonas aeruginosa (ATCC 27853), Salmonella (NCTC 8385) and Shigella flexneri (ATCC 12022) (S. flexneri), while 3% defatted flour was suitable for Staphylococcus aureus (ATCC 25923) (S. aureus). E. coli (93±1) and S. flexneri (524±1) colony count were significantly (P≤0.05) greater in defatted flour without supplement than in supplemented medium. E. coli [(3.72×10(9)±2) CFU/mL], S. aureus [(7.4×10(9)±2) CFU/mL], S. flexneri [(4.03×10(9)±2) CFU/mL] and Salmonella [(2.37×10(9)±1) CFU/mL] in non-hydrolyzed sample were statistically (P≤0.05) greater than hydrolyzed one and commercial peptone agar. Colony count of Salmonella [(4.55×10(9)±3) CFU/mL], S. flexneri [(5.40×10(9)±3) CFU/mL] and Lyesria moncytogenes (ATCC 19116) [(5.4×10(9)±3) CFU/mL] on raw defatted flour agar was significantly (P≤0.05) greater than cooked defatted flour and commercial peptone agar. Biomass of E. coli, S. aureus, Salmonella and Enterococcus faecalis in non-hydrolyzed defatted flour is highly increased over hydrolyzed defatted flour and commercial peptone broth.The defatted flour agar was found to be better microbial media or comparable with peptone agar. The substances in it can serve as sources of carbon, nitrogen, vitamins and minerals that are essential to support the growth of microorganisms without any supplements. Currently, all supplements of peptone agar are very expensive in the market.

Aims and Objectives: The aim of the study was to compare and to analyze the antimicrobial efficacy of 0.12% chlorhexidine and new formulated herbal mouthwash after using for 14 days. The objective was to signify whether the noval herbal combination could be a better alternative mouthwash to Chlorhexidine (CHX). Materials and Methods: This is a double-blinded, random controlled research study conducted in the Department of Oral Pathology and Microbiology. A total of 200 dental students were selected randomly, comprising of two groups, 100 in each, aged between 18 and 22 years with gingival index of score II. The first group was advised to oral rinse with 0.12% chlorhexidine mouthwash and the second group with new formulated herbal mouthwash for 14 days. Saliva samples were collected on the day 0 (baseline), followed by day 7 and 14 and microbial colony count was performed. The data obtained was statistically analyzed using SPSS version 16. Student's t-test was applied for comparison of the mean microbial count between the two groups. Repeated measures analysis of variance followed by Tukey's post hoc test was applied to assess the changes from day 0 to day 7 to day 14. The statistical significance level was set at P < 0.05. Results: Microbial colonies were reduced better in chlorhexidine group on the day 7 whereas, on day 14, greater reduction was observed in the herbal group in both gender groups with high statistical significance (P < 0.001). Conclusions: Herbal mouthwash formulation performed effectively well on long-term usage, could be used as an alternative mouthwash to overcome the disadvantages of chlorhexidine.

Background: Disinfection of dental impression is mandatory for preventing the cross infection between dental staff and dental laboratory. Ozone as a disinfectant can be used in various ways in dentistry. The aim of this study was to evaluate the efficacy of ozonated water for disinfection of irreversible hydrocolloid impression materials and preventing cross-contamination. Methods: Thirty-two circular specimens (1 cm diameter and 2 mm thickness) of irreversible hydrocolloid impression material were made. Thirty specimens were contaminated with Pseudomonas aeruginosa (ATCC 27853), Staphylococcus aureus (ATCC 51299), and Candida albicans (PTCC 5027). Except control samples (n = 2), the other specimens were immersed in ozonated water for 5 and 10 min (5 sample each group). Trypsin was used for separating microorganisms. Diluted microbial solution was analyzed microbiologically for colony-forming units (CFUs). Two-way repeated measure ANOVA and Scheffe post hoc test were used for data analysis (α = 0.05). Results: The number of CFUs after disinfection with ozonated water was decreased significantly (11.84%, 61.55%, and 20.27% after 5 min and 11.03%, 14.50%, and 16.99% after 10 min for P. aeruginosa, S. aureus, and C. albicans, respectively) (P < 0.001). Increasing the time of disinfection resulted in significant reduction in a number of S. aureus only (P < 0.001) although they did not vanish microorganisms completely. Conclusions: Immersion of irreversible hydrocolloid impression material in ozonated water for 10 min will not lead to complete disinfection but decreases the microorganisms to a level that can prevent infection transmission.

Context: Most of the adult population is colonized by Candida in their oral cavity. The process of colonization depends on several factors, including the interaction between Candida and salivary proteins. Therefore, salivary gland hypofunction may alter the oral microbiota and increase the risk for opportunistic infections, such as candidiasis. Hence, it is necessary to evaluate the relationship between salivary flow rates (SFRs) and Candida colony counts in the saliva of patients with xerostomia. Aims: This study aims to determine and evaluate the relationship between SFRs and Candida colony forming units (CFUs) in patients with xerostomia. Settings and Design: This study was a descriptive study. Subjects and Methods: The study participants were taken from the patients attending outpatient department in a private dental college. Fifty patients, who reported xerostomia in a questionnaire of the symptoms of xerostomia, were selected. Chewing stimulated whole saliva samples were collected from them and their SFRs were assessed. Saliva samples were inoculated in the Sabouraud dextrose agar culture media for 24–48 h, and Candida CFUs were counted. Statistical Analysis Used: Chi-squared test was used to analyze the data. Results: There was a significant inverse relationship between salivary flow and candida CFUs count when patients with high colony counts were analyzed (cutoff point of 400 or greater CFU/mL). Females had less SFR than males. Most of the patients who had hyposalivation were taking medication for the underlying systemic diseases. Candida albicans was the most frequent species. Conclusions: There was a significantly negative correlation between SFRs and Candida CFUs in the patients with xerostomia.

Background: The aim of this study was to assess the relationships of quantitative salivary levels of mutans streptococci (MS) in children, aged 3–6 years and 12–15 years, exhibiting variable patterns of caries activity, and to compare the association of MS in saliva using microbial and molecular (polymerase chain reaction [PCR]) assay. Materials and Methods: In this cross-sectional observational study, eighty children were included, forty children each in Group I (3–6 years) and Group II (12–15 years). Children were further divided into two subgroups (Group IC, INC and Group IIC, IINC) based on their dental caries status. Saliva samples were collected and plated onto Mitis Salivarius-Bacitracin agar plates. After detection of MS, DNA was isolated and purifi ed, and MS were evaluated using the PCR and AP-PCR. Results: Of the selected 80 children, 42 were male and 38 were female. In Group IC, the mean colony count was 2.27 ± 0.54 × 105 CFU/ml, and in Group INC, the mean colony was found to be 1.61 ± 0.54 × 105 CFU/ml. In Group II, where mean colony count of 3.31 ± 0.85 × 105 CFU/ml and 2.44 ± 0.54 × 105 CFU/ml was observed in Group IIC and Group IINC, respectively. Conclusion: The mean colony count increased with increasing age and was also more in children with dental caries. Based on the matrices generated by the PCR analysis using coeffi cient slipped-strand mispairing, wide range of genetic diversity was seen in cases of children with and without clinically detectable caries. Background: Today, most approaches to caries viewed that catastrophic change in normal plaque biofilm is responsible for the disease. The behavior and composition of the biofilm are a reflection of the oral environment; the caries is a reflection of adverse changes occurring in that environment. Thus, it is important to identify the pathogenicity of the plaque biofilm so as to predict the caries risk. The recently developed three-tone plaque disclosing agent was used to test its ability in identifying the pathogenicity of plaque. Aim: To assess the efficacy of three-tone plaque disclosing agent in identifying the plaque pathogenicity and correlate with the clinical caries status and microbiological findings. Materials and Methods: Sixty children of 6-13 years age group of both sexes were clinically examined for caries and plaque scores, and then disclosing agent was applied; the color stained plaque samples were collected and cultured for microbiological assessment, and the data were analyzed based on the caries status of the children. Results: There was a significant difference between the pathological plaque of caries active and caries free group (P < 0.05). The pathological plaque scores and the total colony counts, Streptococcus counts and mutans streptococci counts increased with the increase in caries. Conclusion: Three-tone plaque disclosing agent was effective in identifying pathological plaque and can be used as one of the chairside adjuvants in caries risk assessment.

Background: Previous investigations showed that a high-molecular-weight, nondialyzable material (NDM) from cranberries inhibits the adhesion of a number of bacterial species and prevents the coaggregation of many oral bacterial pairs. Aim: In the present study, the effect of mouthrinse containing high-molecular-weight component of cranberry was evaluated on colonization of Streptococcus mutans in children and compared it with a control mouthrinse without high-molecular-weight component on Streptococcus mutans counts. Materials and Methods: A high-molecular-weight NDM was isolated from cranberry juice concentrate after the dialysis of the cranberry concentrate; followed by lyophilization. A mouthwash was prepared especially for the study having NDM in the concentration of 3 mg/ml. Following 4 weeks of daily usage of cranberry-containing mouthwash by the children of an experimental group (n = 20), the Streptococcus mutans counts in plaque and saliva were compared with that in control group using placebo mouthwash (n = 20) with the help of Dentocult SM strips. Results: There was a highlysignificant reduction in Streptococcus mutans counts in saliva and plaque of children using mouthwash containing cranberry NDM (P < 0.05) compared to control. Conclusion: The data suggest that the high-molecular-weight cranberry extract in mouthwash has a significant potential in reducing the Streptococcus counts in the oral environment.

Background: Antimicrobial methods of controlling dental caries that include probiotic agents can play a valuable role in establishing caries control in children at moderate to high risk for developing dental caries. Several studies have demonstrated the beneficial effects of use of various Probiotic products including curd. The objective of this study was to compare the effect of short‑term consumption of probiotic curd containing Lactobacillus acidophilus and normal curd on salivary Streptococcus Mutans counts, as well as salivary pH. Materials and Methods: Forty, caries‑free, 10‑12 years old children were selected and randomly allocated to two groups. Test Group consisted of 20 children who consumed 200ml of probiotic curd daily for 30 days. Control Group consisted of 20 children who were given 200ml of regular curd for 30 days. Salivary pH and salivary Streptococcus Mutans counts were recorded at baseline and after 30 days and statistically compared using the Student’s t‑test. Results: Consumption of probiotic curd resulted in a statistically significant reduction in S. Mutans colony counts (P0.05) in both the groups. Conclusion: Short‑term consumption of probiotic curds can reduce oral S. Mutans counts. Antibacterial efficacy of formocresol, 2% gluteraldehyde and iodine-potassium iodide was assessed by obtaining cultures at consecutive appointments in multiple visit pulpectomies in primary molars. Formocresol and 2% gluteraldehyde were more effective as intracanal medicaments and caused significant reduction in the counts of aerobic and anaerobic microorganisms, thereby supporting the need for placing intracanal medicaments with antibacterial properties, in multiple visit pulpectomies in primary molars.

The present cross-sectional study was carried out in 372 school going children of Belgaum aged 13-15 years. The sample was selected by two stage random sampling method. The aim of the study was to estimate the salivary levels of Streptococcus mutans and Lactobacilli and to study the relationship between these microorganisms and dental caries experience. The DMFT was recorded according to the WHO criteria(1997).The microbial load was estimated from a single saliva sample obtained from the children. The mean DMFT recorded was 2.41. Streptococcus mutans was detected in 87.37% and Lactobacilli in only 36.71% of the sample. Statistically, highly significant relation was found between Streptococcus mutans and DMFT categories (chi2=125.36, P0.001), while Lactobacilli was not statistically related to DMFT categories (chi2=8.78, P0.05).
