Research on soil biochar fertilization has mainly been conducted on cereal crops, and information on its potential for radish production remains inconsistent. Therefore, a pot experiment was conducted to examine the short-term effects of rice husk biochar on radish growth and nitrogen use efficiency (NUE). An investigation was conducted with two application rates of biochar alone, (10 t ha−1 (B10) and 25 t ha−1 (B25), and biochar + chicken manure application with and without NPK fertilizer. The results indicated that the application of biochar 25 t ha−1 + chicken manure (B25:CHM) and the combination of biochar 25 t ha−1 + chicken manure + NPK fertilizer (B25:CHM:NPK) significantly increased root yield by improving NUE, fertilizer recovery efficiency (REN), agronomic efficiency (AE), nitrogen harvest index (NHI), and retaining soil NH4+-N. Although biochar application alone did not significantly influence radish growth on a short-term basis, B10 and B25 increased root yields by 10% and 20%, respectively, compared with the control. Notably, the role of biochar application when combined with organic and inorganic fertilizers was to retain fertilizer N and promote N uptake efficiency by radishes, as higher rates of biochar resulted in higher NUE. Our results suggest that B25:CHM is a suitable combination for organic farming.

Radish is an important root vegetable that is widely grown in Asia. The root-knot nematode (RKN) Meloidogyne incognita seriously affects the growth and development of radish root and causes poor appearance quality. However, the molecular mechanism of radish response to RKNs remains poorly understood. In this study, a total of 220 lncRNAs, 1144 mRNAs, 20 miRNAs and 153 proteins were differential expressed between the RKN-infected and WT samples. Correlation analysis of all DEPs compared with all DGEs showed that 8 mRNAs-DEPs showed a changed abundance. The results showed that 18 DEmiRNAs have 167 target DEGs in 220 miRNA-target modules and 29 DElncRNAs were predicted as putative targets of 16 DEmiRNAs in 37 miRNA-target modules. In all, 6 DGEs in the ABA pathway and 2 DGEs in the JA pathway were identified under RKN infection, respectively. The four regulatory networks of lncRNA-miRNA-mRNA were constructed in response to RKN infection. qRT-PCR analysis found that the expression pattern of 6 DElncRNAs, 6 DEmRNAs, 6 DEmiRNAs and 6 DEPs were consistent with sequencing results. These results provide a theoretical basis for studying the molecular mechanism of radish in response to M. incognita and breeding resistant varieties to this nematode.

In the cosmetics industry, the extract from Raphanus sativus L. is fermented using specific starter cultures. These cosmetic ingredients act as preservatives and skin conditioners. Kombucha is traditionally made by fermenting sweetened tea using symbiotic cultures of bacteria and yeast and is used in cosmetic products. The aim of this study was to evaluate the cosmetic properties of radish leaf and root extract fermented with the SCOBY. Both unfermented water extracts and extracts after 7, 14, and 21 days of fermentation were evaluated. The analysis of secondary plant metabolites by UPLC-MS showed higher values for ferments than for extracts. A similar relationship was noted when examining the antioxidant properties using DPPH and ABTS radicals and the protective effect against H2O2-induced oxidative stress in fibroblasts and keratinocytes using the fluorogenic dye H2DCFDA. The results also showed no cytotoxicity to skin cells using Alamar Blue and Neutral Red tests. The ability of the samples to inhibit IL-1β and COX-2 activity in LPS-treated fibroblasts was also demonstrated using ELISA assays. The influence of extracts and ferments on bacterial strains involved in inflammatory processes of skin diseases was also assessed. Additionally, application tests were carried out, which showed a positive effect of extracts and ferments on TEWL and skin hydration using a TEWAmeter and corneometer probe. The results obtained depended on the concentration used and the fermentation time.

Pithiness is one of the physiological diseases of radishes, which is accompanied by the accumulation of reactive oxygen species (ROS) during the sponging of parenchyma tissue in the fleshy roots. A respiratory burst oxidase homolog (Rboh, also known as NADPH oxidase) is a key enzyme that catalyzes the production of ROS in plants. To understand the role of Rboh genes in radish pithiness, herein, 10 RsRboh gene families were identified in the genome of Raphanus sativus using Blastp and Hmmer searching methods and were subjected to basic functional analyses such as phylogenetic tree construction, chromosomal localization, conserved structural domain analysis, and promoter element prediction. The expression profiles of RsRbohs in five stages (Pithiness grade = 0, 1, 2, 3, 4, respectively) of radish pithiness were analyzed. The results showed that 10 RsRbohs expressed different levels during the development of radish pithiness. Except for RsRbohB and RsRbohE, the expression of other members increased and reached the peak at the P2 (Pithiness grade = 2) stage, among which RsRbohD1 showed the highest transcripts. Then, the expression of 40 genes related to RsRbohD1 and pithiness were analyzed. These results can provide a theoretical basis for improving pithiness tolerance in radishes.

Radish (Raphanus sativus L.) is a globally significant vegetable and relies on cytoplasmic male sterile (CMS) lines for hybrid seed production. The NWB CMS type is favored over Ogura CMS for its ease in maintainer screening. Despite its varied mitochondrial configurations and unvalidated sterile gene, we re-sequenced the mitochondrial genome of NWB CMS Tibet A and verified the function of the sterility gene via genetic transformation of Arabidopsis thaliana. The mitochondrial genomes of Tibet A could be assembled into circular DNA molecules, with a mitochondrial genome size of 239,184 bp. Our analysis indicated that the specific orf463a was the CMS-associated gene in Tibet A, sharing sequence consistency with the CMS gene in DCGMS and NWB CMS YB-A. Collinearity analysis showed that the mitochondrial genomes of NWB CMS Tibet A, DCGMS, and NWB CMS YB-A share the same mitotype, with structural variations due to recombination via a 9731 bp long repeat sequence and a 508 bp short repeat sequence. Driven by the Ap3 promoter, transgenic Arabidopsis with orf463a exhibited male sterility, confirming the gene’s potential role in CMS. In this study, we assembled a new isomeric form of NWB CMS mitochondrial genome and proved the function of the candidate sterile gene.

This study examined the absorption and concentration of anthracene (AN) in the garden radish. Experiments were conducted to grow radishes from the sowing stage in soil contaminated with AN and to grow radishes in soil contaminated with AN following maturity (27 days after sowing). Regarding growth in the AN-containing soil from sowing onward, the AN concentrations during the growing period in both the soil and radishes decreased exponentially over time, albeit with a time lag. Regarding growth in the AN-containing soil after maturity, the AN concentrations in the roots and leaves had a higher density than those in the stems. In these experiments, positive relationships between the AN levels in the soil and radishes were observed. It was inferred that the greater the degree of soil contamination, the greater the effect was on the amount of AN assimilated by radishes. The concentration ratio (radish-to-soil; Cr/Cs) decreased exponentially with the number of days following AN application. It was interpreted that AN was significantly absorbed in the early stages of radish growth and some of the AN returned to the soil as the growth period progressed. According to the results regarding the change over time in the Cr/Cs content in radish parts, it is suggested that soil AN was well absorbed through the roots to circulate within the organism, and it is preferentially distributed and accumulated in the roots and leaves, which have high organic matter content.

The quality of radish roots depends largely on its cultivar, production environment, and postharvest management along the supply chain. Quality monitoring of fresh products is of utmost importance during the postharvest period. The purpose of this study is to nondestructively estimate the weight of a radish using random forests based on color and shape information obtained from images, as well as volumetric information obtained by analyzing a point cloud obtained by combining multiple forms of shape information. The explanatory variables were color and shape information obtained through an image analysis of still images of radishes captured in a constructed photographic environment. The volume information was calculated from the bounding box and convex hull applied to the point cloud by combining the shape information obtained from the image analysis. We then applied random forests to relate the radish weight to the explanatory variables. The experimental results showed that the models using color, shape, or volume information all exhibited good performance with a Pearson’s correlation coefficient (COR) ≥ 0.80, suggesting the potential of nondestructive monitoring of radish weight based on color, shape, and volume information. Specifically, the model using volume information showed very high performance, with a COR of 0.95 or higher.

Site-specific measurements of the crop yield during harvesting are essential for successfully implementing precision management techniques. This study aimed to estimate the mass of radish tubers using the impact principle under simulated vibration and sloped-field harvesting conditions with a laboratory test bench. These conditions included the conveyor speed (CS), impact plate layout (IP), falling height onto the impact plate (FH), the plate angle relative to the horizontal (PH), the field slope, and the vibration of the harvesting machine. Two layouts of impact-type sensors were fabricated and tested, one with a single load cell (SL) and the other with two load cells (DL). An adjustable slope platform and a vibration table equipped with vibration blades were utilized to simulate the slope and vibration effects, respectively. Calibrations were conducted to verify the accuracy of the sensor outputs, processed with the finite impulse response and moving average filters. Radish mass was estimated using an asymmetrically trimmed mean method. The relative percentage error (RE), standard error (SE), coefficient of determination (R²), and analysis of variance (ANOVA) were used to assess the impact plate performance. The results indicated that the SE for both impact plates was less than 4 g in the absence of vibration and slope conditions. The R2 for the single and double impact plates ranged from 0.58 to 0.89 and 0.69 to 0.81, respectively. The FH had no significant impact, while the PH significantly affected the mass measurements for both impact plates. On the other hand, the CS significantly affected the plate performance, except for the double-load-cell impact plate. Both vibration and slope affected the mass measurements, with RE values of 9.89% and 13.92%, respectively. The RE for filtered radish signals was reduced from 9.13% to 5.42%. The tests demonstrated the feasibility of utilizing the impact principle to assess the mass of radishes, opening up possibilities for the development of yield-monitoring systems for crops harvested in a similar manner.

Ogura male sterile cytoplasm is widely used for radish breeding. In this study, high-resolution melting (HRM) markers associated with Rft and Rfo, major restorer-of-fertility genes in Ogura cytoplasmic male sterility (CMS) in radish, were developed. Genetic mapping was carried out using F2 populations derived from crosses between male-sterile Ogura CMS lines and male-fertile lines. Identification of the Rft and Rfo loci was achieved through SNP-based genotyping and linkage grouping. HRM markers were subsequently developed based on flanking sequences of SNPs linked to these loci. For the Rft gene, a set of 117 SNPs was selected within a candidate region on chromosome 5, and 14 HRM markers were successfully developed. Genotyping of F2 showed high correlation between three markers and the phenotype. Regarding the Rfo gene, a set of 27 HRM markers was designed based on flanking sequences of SNPs located on chromosomes 9 and 0. Genotyping in the Rfo segregating population identified a single marker, RSRF27, that accurately distinguished the male sterility phenotype. Validation of the developed markers was performed in populations containing both Rft and Rfo genes, confirming their utility for genotyping and demonstrating that these two genes independently contribute to male sterility recovery. Overall, this study provides HRM markers that can be used for genotyping Rft and Rfo and contributes to a deeper understanding of male sterility restoration mechanisms in Ogura CMS.

The aim of the study was to determine the effect of ozone gas fumigation on the mechanical, chemical, and microbiological parameters of radish tubers. Radish plants were grown in the ground in accordance with the principles of good agricultural practice and condition suitable for the soil and climatic conditions of south-eastern Poland. At the end of the growing season, 24 h before harvest, radish plants were exposed to a variable factor, i.e., fumigation with ozone gas at various doses (1 ppm for 1 and 5 min; 5 ppm for 1 and 5 min) in order to modify selected metabolic pathways of bioactive compounds. Then, 24 h after ozonation, radish tubers were harvested and placed in a climatic chamber with controlled conditions, i.e., 2 °C and 90% humidity. Laboratory analyses were performed during storage on days 1, 5, and 10. The ozonation used did not significantly improve the mechanical properties and water content of radish tubers. There was a beneficial effect of selected gaseous ozone doses (1 ppm for 1 and 5 min; 5 ppm for 1 min on the 10th day of storage) on the biosynthesis of selected bioactive compounds, i.e., ascorbic acid content, total polyphenol content, and antioxidant potential during storage. The most beneficial effects of the use of gaseous ozone were observed in the storage process in reducing the microbiological load of radish tubers. Among the ozonation doses used, the dose of 5 ppm for 5 min had the most beneficial effect on reducing the microbiological load. It reduced the number of yeasts and molds by 14.2% and aerobic mesophilic bacteria by 20.9% compared to the control sample on the last day of storage. Additionally, between the 5th and 10th day of storage, a significant effect of each ozone dose applied on reducing the occurrence of yeasts, molds, and mesophilic aerobic bacteria during tuber storage was noted.

Microgreens have become an important specialty crop valued by their varying texture, vibrant colors, and nutrient-dense features. As the number of species and cultivars rapidly increases for microgreen production, fertigation requirements in relation to shoot production and nutrient compositions remain unclear. This study aimed to investigate the shoot yield, visual quality, and mineral nutrient concentrations of six microgreens in the Brassicaceae family including the ‘Waltham’ broccoli, ‘Red Acre’ cabbage, Daikon radish, ‘Red Russian’ kale, pea, and Rambo radish in two experiments in December 2020 and January 2021. Each microgreen was fertigated with 120 mL of fertilizer solution daily for five consecutive days with a rate of 0, 70, 140, 210, or 280 mg·L−1 N from a general-purpose fertilizer. Broccoli, Daikon radish, and kale similarly produced the highest fresh shoot weights of 916.5 to 984 g·m−2 in December 2020, while pea produced the highest fresh shoot weight of 2471 g·m−2 in January 2021 among cultivars. The fertigation rates of 140, 210, and 280 mg·L−1 N resulted in similar fresh and dry shoot weights of selected microgreens, suggesting 140 mg·L−1 N should be sufficient for microgreen fertilization. Mineral nutrients in microgreens varied among cultivars: pea microgreens had the highest nitrogen (N) concentrations of 70.6 to 75.2 mg·g−1 in December 2020 and 72.1 to 75.4 mg·g−1 in January 2021; and cabbage microgreens were rich in calcium (Ca) in both experiments. The kale, pea, and Rambo radish microgreens contained the highest concentrations of iron (Fe) and manganese (Mn) in December 2020. The fertigation rate affected macronutrient concentrations but did not affect micronutrient concentrations including Fe, Mn, or zinc (Zn).

During the Advanced Plant Habitat experiment 2, radish plants were grown in two successive grow-outs on the International Space Station (ISS) for 27 days each. On days 10, 18, and 24, leaf punch (LP) samples were collected and frozen. At harvest, bulb tissue was sampled with oligo-dT functionalized Solid Phase Gene Extraction (SPGE) probes. The space samples were compared with samples from ground controls (GC) grown at the Kennedy Space Center (KSC) under the same conditions as on the ISS, with notably elevated CO2 (about 2500 ppm), and from lab plants grown under atmospheric CO2 but with light and temperature conditions similar to the KSC control. Genes corresponding to peroxidase (RPP), glucosinolate biosynthesis (GIS), protein binding (CBP), myrosinase (RMA), napin (RSN), and ubiquitin (UBQ) were measured by qPCR. LP from day 24 and bulb samples collected at harvest were compared with RNA-seq data from material that was harvested, frozen, and analyzed after return to Earth. The results showed stable transcription in LP samples in GC but decreasing values in ISS samples during both grow-outs, possibly indicative of stress. SPGE results were similar between GC and ISS samples. However, the RNA-seq analyses showed different transcription profiles than SPGE or LP results, possibly related to localized sampling. RNA-seq of leaf samples showed greater variety than LP data, possibly because of different sampling times. RSN and RPP showed the lowest transcription regardless of method. Temporal analyses showed relatively small changes during plant development in space and in ground controls. This is the first study that compares developmental changes in space-grown plants with ground controls based on a comparison between RNA-seq and qPCR analyses.

To understand the coloring mechanism in black radish, the integrated metabolome and transcriptome analyses of root skin from a black recombinant inbred line (RIL 1901) and a white RIL (RIL 1911) were carried out. A total of 172 flavonoids were detected, and the analysis results revealed that there were 12 flavonoid metabolites in radish root skin, including flavonols, flavones, and anthocyanins. The relative concentrations of most flavonoids in RIL 1901 were higher than those in RIL 1911. Meanwhile, the radish root skin also contained 16 types of anthocyanins, 12 of which were cyanidin and its derivatives, and the concentration of cyanidin 3-o-glucoside was very high at different development stages of black radish. Therefore, the accumulation of cyanidin and its derivatives resulted in the black root skin of radish. In addition, a module positively related to anthocyanin accumulation and candidate genes that regulate anthocyanin synthesis was identified by the weighted gene co-expression network analysis (WGCNA). Among them, structural genes (RsCHS, RsCHI, RsDFR, and RsUGT75C1) and transcription factors (TFs) (RsTT8, RsWRKY44L, RsMYB114, and RsMYB308L) may be crucial for the anthocyanin synthesis in the root skin of black radish. The anthocyanin biosynthesis pathway in the root skin of black radish was constructed based on the expression of genes related to flavonoid and anthocyanin biosynthesis pathways (Ko00941 and Ko00942) and the relative expressions of metabolites. In conclusion, this study not only casts new light on the synthesis and accumulation of anthocyanins in the root skin of black radish but also provides a molecular basis for accelerating the cultivation of new black radish varieties.

It is becoming increasingly challenging to maintain crop yields and quality as the global climate changes. The aim of this study was to determine whether and how the profile of health-promoting and taste-related compounds of radishes changes within a growing season. A total of 16 radish (Raphanus sativus L.) genotypes that are commercially available on the Czech market were assessed by means of chemical analysis. Radishes were cultivated in three independent growing cycles under controlled conditions, and the effects of the genotype and growing cycle, as well as their interactions, on the chemical traits were evaluated. Most of the variability in chemical composition was associated with the growing cycle, which accounted for 51.53% of total variance, followed by the genotype (26% of total variance). The interaction between the growing cycle and genotype explained 22.47% of total variance. The growing cycle had the strongest effect on amino acid profiles. More specifically, the amino acids that are known to contribute to overall taste (glycine, along with glutamic and aspartic acids) showed the highest degree of variation, while the amino acids related to glucosinolate biosynthesis (methionine, isoleucine, tryptophan, and phenylalanine) showed relatively low variability. On the other hand, indole glucosinolates were found to differ the most between genotypes.

This study investigated the degradation characteristics and conducted a risk assessment of four pesticides (Diniconazole, Dinotefuran, Metconazole, and Tebuconazole) in the leaves and roots of radish. Radish was cultivated in two greenhouse fields, and samples were collected at 0, 1, 2, 3, 5, 7, and 10 days after pesticide application. Sample analysis was performed using LC-MS/MS, and the recovery rates ranged from 70.1% to 118.6%. The biological half-life of Diniconazole was found to be 6.2 days (leaf and root), Dinotefuran was 5.3 days (leaf) and 4.6 days (root), Metconazole was 9.3 days (leaf) and 3.2 days (root), and Tebuconazole was 8.0 days (leaf) and 5.1 days (root). After comparing the maximum residue limits (MRL) of each pesticide in Korea with the residues during the pre-harvest interval (PHI), Diniconazole showed a Hazard quotient (HQ) exceeding 1, indicating potential risks for true consumers. Furthermore, Tebuconazole showed an HQ of 0.3 or higher, indicating a significant level of risk.

SWEET (Sugars Will Eventually be Exported Transporter) genes play essential roles in various biological processes, including phloem loading, sugar efflux, plant development and stress response. In this study, a total of 33 RsSWEET gene members were identified in the radish genome. They could be divided into four subfamilies and are distributed on eight radish chromosomes. Cis-acting regulatory element analysis indicated that these RsSWEET genes were potentially involved in the radish growth and development and stress response process, including circadian control and light response and responses to numerous stresses, including low-temperature and drought stress. Transcriptome data analysis revealed that a number of RsSWEET genes exhibited specific expression patterns in different tissues and developmental stages of radish. Moreover, several RsSWEET genes (e.g., RsSWEET2a, RsSWEET3a, RsSWEET16b and RsSWEET17) showed differential expression profiles under various abiotic stresses, including cold, heat, salt, Cd and Pb stress. Remarkably, the RsSWEET17 was specifically expressed in the cambium of radish. RsSWEET17 was heterologously expressed in yeast strain EBY.VW4000, which suggested that it has the ability to transport sugar. Notably, RsSWEET17-overexpressing Arabidopsis plants exhibited excessive root length, greater fresh weight and higher soluble sugar content (SSC) accumulation compared with wild-type (WT) plants, indicating that RsSWEET17 might positively regulate radish taproot development by strategically manipulating sugar accumulation. Collectively, these results clarify the molecular mechanisms underlying RsSWEET-mediated sugar accumulation and root growth and development in radish.

White radish is a nutritious and delectable vegetable that is enjoyed globally. Conventional techniques for monitoring radish growth are arduous and time-consuming, encouraging the development of novel methods for quicker measurements and greater sampling density. This research introduces a mathematical model working on high-resolution images to measure radish’s biophysical properties automatically. A color calibration was performed on the dataset using a color checker panel to minimize the impact of varying light conditions on the RGB images. Subsequently, a Mask-RCNN model was trained to effectively segment different components of the radishes. The observations of the segmented results included leaf length, leaf width, root width, root length, leaf length to width, root length to width, root shoulder color, and root peel color. The automated real-life measurements of these observations were then conducted and compared with actual results. The validation results, based on a set of white radish samples, demonstrated the models’ effectiveness in utilizing images for quantifying phenotypic traits. The average accuracy of the automated method was confirmed to be 96.2% when compared to the manual method.

Interspecific hybridization and accompanying backcross between crops and relatives have been recognized as a powerful method to broaden genetic diversity and transfer desirable adaptive traits. Crosses between radish (Raphanus sativus, RR, 2n = 18) and Brassica oleracea (CC, 2n = 18), which formed allotetraploid Raphanobrassica (RRCC, 2n = 36), initiated the construction of resynthetic allopolyploids. However, these progenies from the backcrosses between Raphanobrassica and the two diploid parents have not been well deciphered. Herein, thousands of backcrosses using both Raphanobrassica and the two diploid parents as pollen donors were employed. Several hybrids with expected (2n = 27) and unexpected chromosome numbers (2n = 26 and 2n = 36) were obtained. Fluorescence in situ hybridization (FISH) analysis with R-genome-specific sequences as probes demonstrated that the genome structures of the two expected hybrids were RRC and CCR, and the genome structures of the three unexpected hybrids were RRRC, CCCR, and RRC’ (harbouring an incomplete C genome). The unexpected hybrids with extra R or C genomes showed similar phenotypic characteristics to their expected hybrids. FISH analysis with C-genome-specific sequences as probes demonstrated that the unexpected allotetraploid hybrids exhibited significantly more intergenomic chromosome pairings than the expected hybrids. The expected and unexpected hybrids provide not only novel germplasm resources for the breeding of radish and B. oleracea but also very important genetic material for genome dosage analysis.

The aim of this work was to enhance the use of a food-grade antioxidant extract obtained from lemon processing byproducts (peel, pulp and seeds) to extend the shelf life of minimally processed radishes. The extract (LPE) was previously characterized in terms of total phenolic (6.75 ± 0.34 mg GAE g−1 d.w.) and flavonoid content (2.04 ± 0.09 mg CE g−1 d.w.) and antioxidant activity, and eriocitrin and hesperidin were identified as the most prevalent phenolic compounds by a UHPLC system. The effects of different dipping aqueous solutions (UCR, DRa, DRb) and alginate-based edible coating formulations (CRc, CRd) with and without the antioxidant extract were studied on the quality parameters of minimally processed radishes, characterized regarding their microbiological and physicochemical characteristics for up to 14 days at 3 °C. The coating formulated with LPE delayed the radish respiration process, as well as resulting in less color variation (ΔE < 3) and reduced mesophilic aerobic count values (4.49 ± 1.43 log CFU g−1), proving the effectiveness of LPE as a value-added ingredient in developing post-harvest strategies to prolong the shelf life of minimally processed vegetables. Indeed, coated samples without the extract showed a clear development of rotting, which led to the end of their shelf life on their 7th day of storage.

Radish is an economically important root vegetable worldwide. In this study, the 217 cultivated radish accessions were collected and genotyped. To detect the genotypes of these accessions, a total of 24 structure variation (SV) markers distributed on nine chromosomes were employed to analyze genetic diversity and construct a core germplasm collection of radish. The results of polymorphism information content (PIC) indicated a good polymorphism of these SV markers. Population structure analysis and principal component analysis (PCA) results showed that the 217 radish accessions fell into three main populations (P1, P2, and P3). Genetic diversity analysis showed that these populations were highly associated with geographical distribution. The values of the fixation index (FST) indicated a high genetic diversity between P2 and P3, and a moderate genetic diversity between P1 and P2, and P1 and P3. Furthermore, the 43 core germplasm were exploited for creating cytoplasmic male sterility (CMS) lines and cultivating new radish varieties. The high genetic diversity of 217 radish germplasms will not only provide valuable resources for future genetic mapping and functional genomic research, but also facilitate core germplasm utilization and the molecular breeding of radish.

Pickled radish (Raphanus sativus) is a traditional Asian ingredient, but the traditional method takes decades to make this product. To optimize such a process, this study compared the saponin content of pickled radishes with different thermal processing and traditional processes (production time of 7 days, 10 years, and 20 years) and evaluated the effects of different thermal processes on the formation of radish saponin through kinetics study and mass spectrometry. The results showed that increasing the pickling time enhanced the formation of saponin in commercial pickled radishes (25 °C, 7 days, 6.50 ± 1.46 mg g−1; 3650 days, 23.11 ± 1.22 mg g−1), but these increases were lower than those induced by thermal processing (70 °C 30 days 24.24 ± 1.01 mg g−1). However, it was found that the pickling time of more than 10 years and the processing temperature of more than 80 °C reduce the saponin content. Liquid chromatography–mass spectrometry (LC-MS) analysis showed that the major saponin in untreated radish was Tupistroside G, whereas treated samples contained Asparagoside A and Timosaponin A1. Moreover, this study elucidated the chemical structure of saponins in TPR. The findings indicated that thermal treatment could induce functional saponin conversion in plants, and such a mechanism can also be used to improve the health efficacy of plant-based crops.

Alcohol and drug overdoses cause liver diseases such as cirrhosis, hepatitis, and liver cancer globally. In particular, an overdose of acetaminophen (APAP), which is generally used as an analgesic and antipyretic agent, is a major cause of acute hepatitis, and cases of APAP-induced liver damage are steadily increasing. Potential antioxidants may inhibit the generation of free radicals and prevent drug-induced liver damage. Among plant-derived natural materials, radishes (RJ) and turnips (RG) have anti-inflammatory, anticancer, and antioxidant properties due to the presence of functional ingredients, such as glucosinolate and isothiocyanate. Although various functions have been reported, in vivo studies on the antioxidant activity of radishes are insufficient. Therefore, we aim to evaluate the hepatoprotective effects of RG and RJ in APAP-induced liver-damaged mice. RG and RJ extracts markedly improved the histological status, such as inflammation and infiltration, of mice liver tissue, significantly decreased the levels of alanine transaminase, aspartate aminotransferase, and malondialdehyde, and significantly increased the levels of glutathione, superoxide dismutase and catalase in the APAP-induced liver-damaged mice. In addition, RG and RJ extracts significantly increased the expression of Nrf-2 and HO-1, which are antioxidative-related factors, and regulated the BAX and BCL-2, thereby showing anti-apoptosis activity. These results indicated that RG and RJ extracts protected mice against acute liver injury, attributed to a reduction in both oxidative stress and apoptosis. These findings have clinical implications for the use of RG and RJ extracts as potential natural candidates for developing hepatoprotective agents.

In most higher plants, sucrose is the significant form of carbohydrate for long-distance transportation. Sucrose transporters/sucrose carriers (SUTs/SUCs) are involved in the loading and unloading of sucrose in phloem and play an important role in the growth and development of plants. In this study, 12 RsSUC genes were first identified from the radish genome, and their phylogenetic relationships, gene structure, and conserved motifs were further analyzed. RT-qPCR results indicated that RsSUC genes exhibited various expression patterns in different tissues and development stages of the radish. Overexpression of RsSUC1b in Arabidopsis significantly improved the uptake efficiency of exogenous sucrose, and promoted leaves and lateral root growth. In addition, the transgenic plants flowered significantly earlier than wild-type (WT) plants, and the soluble sugar contents (SSCs) including sucrose, glucose, and fructose in the mature leaves and pods were increased. It could be inferred that RsSUC1b is a plasma membrane sucrose transporter and plays a vital role in sucrose transportation and sugar accumulation during plant growth and development. These findings provided novel insights into the biological function of RsSUC genes and facilitate dissecting the molecular mechanism underlying sugar transport during radish development.

Pithiness is one of the physiological diseases of radishes, which is accompanied by the accumulation of reactive oxygen species (ROS) during the sponging of parenchyma tissue in the fleshy roots. A respiratory burst oxidase homolog (Rboh, also known as NADPH oxidase) is a key enzyme that catalyzes the production of ROS in plants. To understand the role of Rboh genes in radish pithiness, herein, 10 RsRboh gene families were identified in the genome of Raphanus sativus using Blastp and Hmmer searching methods and were subjected to basic functional analyses such as phylogenetic tree construction, chromosomal localization, conserved structural domain analysis, and promoter element prediction. The expression profiles of RsRbohs in five stages (Pithiness grade = 0, 1, 2, 3, 4, respectively) of radish pithiness were analyzed. The results showed that 10 RsRbohs expressed different levels during the development of radish pithiness. Except for RsRbohB and RsRbohE , the expression of other members increased and reached the peak at the P2 (Pithiness grade = 2) stage, among which RsRbohD1 showed the highest transcripts. Then, the expression of 40 genes related to RsRbohD1 and pithiness were analyzed. These results can provide a theoretical basis for improving pithiness tolerance in radishes.

Root shape is an important characteristic that affects the commodity of radish (Raphanus sativus L.), which can be measured using the ratio of root length (RL) to root diameter (RD). Although it is known that root shape is controlled by quantitative trait loci (QTLs), reliable QTLs for radish root shape are still lacking. In the present study, we used three F2 populations (1902, 1908 and 1909) derived from the crossing of five radish cultivars with highly divergent root shapes to perform QTL-seq. A total of 1282 individuals of the three F2 populations were measured to determine the root length and maximum diameter. High-depth resequencing of six extreme pools and five parents was performed, and QTL-seq was used to detect the QTLs controlling the radish root shape. We identified seven QTLs for root shape distributing on five radish chromosomes (R1, R2, R4, R5 and R7), among which rs7.1 and rs7.2 had an overlap of 1.02 Mb (13.79–14.81 Mb). In addition, two QTLs, rs4.1 and rs4.2, were adjacent to each other on chromosome R4. In conclusion, this study provides an important foundation for the fine mapping and functional analysis of the QTLs controlling the root shape and breeding for root shape in radish.

Radish is a typical self-incompatible crop. The rapid and accurate identification of S haplotypes can circumvent the blindness of the hybrid combination process, which is critical in radish heterosis utilization and the breeding of new varieties. In this study, based on the gene sequence which encodes the S-locus receptor kinase (SRK) of radish, and the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis, the S haplotypes were identified among 79 cultivated radish genotypes. The PCR results indicated that 79 radish genotypes could be divided into 48 Class I, 13 Class II, and 17 Class I/II S haplotypes. Sequence alignment confirmed that the Class I materials contained 19 S haplotypes, of which three haplotypes (‘NAU-S53’, ‘NAU-S54’ and ‘NAU-S55’) were identified for the first time in radish. After digestion using the Hinf I restriction endonuclease, the SRK domain of DNA fragments of different genotypes showed high polymorphism. Homozygous materials S haplotypes could be quickly distinguished by the differences in the digested bands. Molecular identification of the S haplotype was highly consistent with the field pollination and pollen tube germination results. These results would provide an important approach for the rapid identification of radish S haplotypes and the efficient utilization of self-incompatibility in heterosis breeding.

TCP transcription factors are a unique transcription family in higher plants, and play important roles in plant development, responses to environmental stresses and phytohormones. Radish is an important crop and widely cultivated worldwide. However, genome-wide identification and expression analysis of TCP family in radish has not been reported yet. In this study, 32 RsTCP genes were identified from the whole genome. Phylogenetic analysis showed that the RsTCP family can be divided into two major groups and three subgroups, namely Class I (PCF), Class II (CIN), and Class II (CYC/TB1). Chromosome mapping showed that 32 genes were distributed on all nine chromosomes of radish. Transcriptome data indicated that the transcription of RsTCP genes differed between tissues and developmental stages. Multiple phytohormone-related cis-acting elements in the promoter region of RsTCPs and several post-translational modification sites in RsTCP protein sequences were identified. The real-time quantitative PCR (RT-qPCR) analysis indicated that several RsTCP genes changed significantly in response to the treatments, including low temperature, drought, salt, gibberellin (GA3), and abscisic acid. Among them, RsTCP16 showed significantly higher levels in leaves than in other radish tissues, and the transcription of RsTCP16 was significantly upregulated under the five treatments, especially during low temperature, salt shock, and GA3 response, suggesting that RsTCP16 could be involved in the development of radish leaves and the response to stress. In summary, these results provided a basis for further exploring the molecular mechanism of RsTCP genes in radish.

CCT genes play vital roles in flowering, plant growth, development, and response to abiotic stresses. Although they have been reported in many plants, the characterization and expression pattern of CCT genes is still limited in R. sativus. In this study, a total of 58 CCT genes were identified in R. sativus. Phylogenetic tree, gene structure, and conserved domains revealed that all CCT genes were classified into three groups: COL, CMF, and PRR. Genome-wide identification and evolutionary analysis showed that segmental duplication expanded the CCT gene families considerably, with the LF subgenome retaining more CCT genes. We observed strong purifying selection pressure for CCT genes. RsCCT genes showed tissue specificity, and some genes (such as RsCCT22, RsCCT36, RsCCT42 and RsCCT51) were highly expressed in flowers. Promoter cis-elements and RNA-seq data analysis showed that RsCCT genes could play roles in controlling flowering through the photoperiodic pathway and vernalization pathway. The expression profiles of RsCCT genes under Cd, Cr, Pb, and heat and salt stresses revealed that many RsCCT genes could respond to one or more abiotic stresses. Our findings could provide essential information for further studies on the function of RsCCT genes.

Leaf chlorophyll content is used as a major indicator of plant stress and growth, and hyperspectral remote sensing is frequently used to monitor the chlorophyll content. Hyperspectral reflectance has been used to evaluate vegetation properties such as pigment content, plant structure and physiological features using portable spectroradiometers. However, the prices of these devices have not yet decreased to consumer-affordable levels, which prevents widespread use. In this study, a system based on a cost-effective fingertip-sized spectrometer (Colorcompass-LF, a total price for the proposed solution was approximately 1600 USD) was evaluated for its ability to estimate the chlorophyll contents of radish and wasabi leaves and was compared with the Analytical Spectral Devices FieldSpec4. The chlorophyll contents per leaf area (cm2) of radish were generally higher than those of wasabi and ranged from 42.20 to 94.39 μg/cm2 and 11.39 to 40.40 μg/cm2 for radish and wasabi, respectively. The chlorophyll content was estimated using regression models based on a one-dimensional convolutional neural network (1D-CNN) that was generated after the original reflectance from the spectrometer measurements was de-noised. The results from an independent validation dataset confirmed the good performance of the Colorcompass-LF after spectral correction using a second-degree polynomial, and very similar estimation accuracies were obtained for the measurements from the FieldSpec4. The coefficients of determination of the regression models based on 1D-CNN were almost same (with R2 = 0.94) and the ratios of performance to deviation based on reflectance after spectral correction using a second-degree polynomial for the Colorcompass-LF and the FieldSpec4 were 4.31 and 4.33, respectively.

The high yield of seeds can reduce the cost of seed production for parental lines, as well as F1 cultivars in radish. The number of seeds per silique and silique length are two important traits among traits determining seed yield, but no study has been conducted on their quantitative trait loci (QTLs) in radish. A high-density linkage map was constructed, based on genotyping-by-sequencing (GBS) of the F2 population, derived from two parental lines, significantly differed by the two traits, which were grown in a controlled environment to minimize the environmental effects. Using the map with 848 SNPs, three significant QTLs were identified, two and one of which were associated with the number of seeds per silique and silique length, respectively. Ortholog analysis was conducted with Arabidopsis thaliana genes, related to the number of seeds per silique, and revealed five radish putative candidate genes. These putative candidate genes appear to be related to ovule, embryo sac, embryo, pollen and seed development, as well as a double fertilization process. The method to pollinate the F2 population, as well as preliminary QTLs and SNPs therein, can be helpful for future QTL studies to improve seed production in radish breeding programs.

This study investigated the impact of Methyl Jasmonate (MeJA) application on the nutritional content and yield of five different colored radish microgreens. Microgreens were produced without substrate and subjected to 0.5 mM and 1.0 mM MeJA treatments on the 7th day, three days before harvest. The parameters measured included yield, dry matter, minerals, amino acids, secondary metabolites such as chlorophylls (Chls), anthocyanins, flavonoids, phenolics, glucosinolates (GSLs), vitamin C, and antioxidant capacity. MeJA at 1.0 mM generally improved yield and dry weight across cultivars, and all microgreens exhibited rich mineral and amino acid composition, with the influence of cultivar being more significant than MeJA treatment. However, MeJA enhanced all cultivars' anthocyanins, GSLs, phenolics, flavonoids, and antioxidant activities. Generally, as the antioxidant capacity is the primary factor influencing the nutritional quality of microgreens, MeJA-treated microgreens, especially with selected superior cultivars such as 'Asia purple' and 'Koregon red', could offer a potential for cultivation of value-added, eco-friendly microgreens with substrate-free cultivation.

Kimchi made from small-type (Altari) radishes grown in late spring is more pungent than that made from autumn-grown Altari radishes, which poses a major challenge in the kimchi industry. The mechanism through which the pungency of Altari radish changes seasonally has not been intensively investigated. In this study, three small-type radish cultivars with different pungency levels were cultivated in spring and autumn to identify the factors affecting the seasonal-dependent pungency of small-type radishes. The contents of pungency-related metabolite glucoraphasatin and other polar metabolites were analyzed. Although a previous study reported that the glucoraphasatin concentration affects the pungency of radish, in the current study, the concentration of neither glucoraphasatin nor its hydrolysis product (raphasatin) could fully explain the change in the pungency associated with radish cultivars grown in the two seasons. The change in the pungency of radish by season may be explained by the ratio of raphasatin content to total sweetness of sugars. In addition, the polar metabolites that differ with season were analyzed to identify seasonal biomarkers and understand the seasonal changed physio-biochemistry.

Glucosinolates, lipid-soluble vitamins E and K contents, primary metabolites and plant hormones were analyzed from topped radish root and detached leaf during storage at 1 °C. The topped root was analyzed at 0, 5, 15, 30, and 90 days after storage while the detached leaf was analyzed at 0, 5, 15, 30, and 45 days in an airtight storage atmosphere environment. The results showed that aliphatic glucosinolates were gradually decreased in leaf but not in root. There was a highly significant correlation between tryptophan and 4-methoxyindoleglucobrassicin in both tissues (r = 0.922, n = 10). There was no significant difference in vitamins E and K in leaf and root during storage. Plant hormones partially explained the significantly changed metabolites by tissue and time, which were identified during cold storage. Phenylalanine, lysine, tryptophan, and myo-inositol were the most important biomarkers that explained the difference in leaf and root tissue during cold storage. The most different metabolism between leaf and root tissue was starch and sucrose metabolism. Therefore, different postharvest technology or regimes should be applied to these tissues.

Radish is a kind of moderately salt-sensitive vegetable. Salt stress seriously decreases the yield and quality of radish. The plasma membrane Na+/H+ antiporter protein Salt Overly Sensitive 1 (SOS1) plays a crucial role in protecting plant cells against salt stress, but the biological function of the RsSOS1 gene in radish remains to be elucidated. In this study, the RsSOS1 gene was isolated from radish genotype ‘NAU-TR17’, and contains an open reading frame of 3414 bp encoding 1137 amino acids. Phylogenetic analysis showed that RsSOS1 had a high homology with BnSOS1, and clustered together with Arabidopsis plasma membrane Na+/H+ antiporter (AtNHX7). The result of subcellular localization indicated that the RsSOS1 was localized in the plasma membrane. Furthermore, RsSOS1 was strongly induced in roots of radish under 150 mmol/L NaCl treatment, and its expression level in salt-tolerant genotypes was significantly higher than that in salt-sensitive ones. In addition, overexpression of RsSOS1 in Arabidopsis could significantly improve the salt tolerance of transgenic plants. Meanwhile, the transformation of RsSOS1△999 could rescue Na+ efflux function of AXT3 yeast. In summary, the plasma membrane Na+/H+ antiporter RsSOS1 plays a vital role in regulating salt-tolerance of radish by controlling Na+ homeostasis. These results provided useful information for further functional characterization of RsSOS1 and facilitate clarifying the molecular mechanism underlying salt stress response in radish.

In hydroponics, a continuous supply of mineral nutrients is essential for plant growth. However, constitutive nutrient-rich conditions also increase the nitrate content in the plants, which can be harmful to human health. Here, we investigated the effect of nutrient deficiency on the growth and component composition of hydroponic radish by changing the timing of nutrient removal from the hydroponic solution. Radish plants that were 14 days old were transferred to four different nutrient conditions for 14 days: nutrient deficiency for 14 days (WW), full nutrient for 14 days (NN), nutrient deficiency for the last 7 days (NW), and nutrient deficiency for the first 7 days (WN). After the treatments, the NW plants had similar taproot growth to NN plants. In contrast, the WN plants significantly reduced taproot growth. The WW plants reduced the shoot and taproot weight and their water contents. The nitrate content in the taproots was reduced in the NW and WW plants. The WW plants contained lower total phenol and higher ascorbic acid and sugar contents. These results suggest that the uptake of nutrient minerals at the young growth stage is important for the growth of radish taproot. Nutrient deficiency management can be one of the most effective tools for regulating radish growth and composition.

Iodine is an essential trace element for humans, and iodine deficiency is a significant health problem. In this study, an improved method for iodine biofortification based on seed germination was established. Solutions of KI (0.15, 0.30, 0.75 and 1.5 mg∙g−1 of seeds) were applied to germinating radish seeds of two cultivars Raphanus sativus L. var. sativus: Warta and Zlata. Compared with the control (seeds treated with water) the iodine content (in the radish sprouts produced by germinating seeds treated with KI were approximately 112.9–2730 times higher. The application KI rates did not adversely affect the biological quality of the radish sprouts. Regarding the biological quality of the iodine-enriched seedlings, we determined their length, dry mater, protein, soluble sugars, chlorophylls, total phenol, ascorbic acid, thiol group content and total antioxidant capacity. The effect of potassium iodide on the selected parameters of their biological quality varied depending on the KI doses and radish cultivars. The results showed that the most appropriate biofortification application rates were 0.15 and 0.30 mg KI per g seeds, because the enriched seedlings had excellent biological quality parameters.

The red or purple color of radish (Raphanus sativus L.) taproots is due to anthocyanins, which have nutritional and aesthetic value, as well as antioxidant properties. Moreover, the varied patterns and levels of anthocyanin accumulation in radish roots make them an interesting system for studying the transcriptional regulation of anthocyanin biosynthesis. The R2R3 MYB transcription factor RsMYB1 is a key positive regulator of anthocyanin biosynthesis in radish. Here, we isolated an allele of RsMYB1, named RsMYB1Short, in radish cultivars with white taproots. The RsMYB1Short allele carried a 4 bp insertion in the first exon causing a frame-shift mutation of RsMYB1, generating a truncated protein with only a partial R2 domain at the N-terminus. Unlike RsMYB1Full, RsMYB1Short was localized to the nucleus and the cytoplasm and failed to interact with their cognate partner RsTT8. Transient expression of genomic or cDNA sequences for RsMYB1Short in radish cotyledons failed to induce anthocyanin accumulation, but that for RsMYB1Full activated it. Additionally, RsMYB1Short showed the lost ability to induce pigment accumulation and to enhance the transcript level of anthocyanin biosynthetic genes, while RsMYB1Full promoted both processes when co-expressed with RsTT8 in tobacco leaves. As the result of the transient assay, co-expressing RsTT8 and RsMYB1Full, but not RsMYB1Short, also enhanced the promoter activity of RsCHS and RsDFR. We designed a molecular marker for RsMYB1 genotyping, and revealed that the RsMYB1Short allele is common in white radish cultivars, underscoring the importance of variation at the RsMYB1 locus in anthocyanin biosynthesis in the radish taproot. Together, these results indicate that the nonsense mutation of RsMYB1 generated the truncated protein, RsMYB1Short, that had the loss of ability to regulate anthocyanin biosynthesis. Our findings highlight that the frame shift mutation of RsMYB1 plays a key role in anthocyanin biosynthesis in the radish taproot.

The European radish is one of the most unresponsive crops in the Brassicaceae family to embryogenesis in in vitro microspore culture. The aim of this work was to study the process of embryogenesis of European radish and its biological features. In this study, the embryogenesis of European radish is described in detail with illustrative data for the first time. For the first time for the entire family Brassicaceae, the following were found: microspores with intact exines with ordered-like divisions; microspores completely free of exines; and a new scheme of suspensors attachment to the apical parts of embryoids. The morphology of double and triple twin embryoids was described, and new patterns of their attachment to each other were discovered. Uneven maturation of European radish embryoids at all stages of embryogenesis was noted. The period of embryoid maturation to the globular stage of development corresponded, in terms of time, to the culture of B. napus, and into the cotyledonary stage of development, maturation was faster and amounted to 17–23 days. The rate of embryoid development with and without suspensors was the same.

Radish flower color, bolting time, and flowering time are important traits for attracting certain pollinators and affect fleshy root quality. In this study, an analysis of the anthocyanidins in radish flowers by high-performance liquid chromatography revealed that differences in the cyanidin content are likely to be associated with the variability in radish flower colors (i.e., purple and white petals). A quantitative trait loci (QTL) analysis identified nine QTLs on three Raphanus sativus linkage groups. Three QTLs—qRFC1, qRBT1, and qRFT1—which were consistently detected and explained a high proportion of the observed variation (10.30% to 34.57%), were considered as the major QTLs responsible for flower color, bolting time, and flowering time, respectively. A total of 16 and 11 candidate genes within the major QTL regions for flower color and bolting/flowering times, respectively, were preliminarily annotated. Six genes (Rs018140, Rs018950, Rs019220, Rs020080, Rs020590, and Rs021450) related to flower color were differentially expressed in the parental lines. On the basis of nucleotide and amino acid sequence diversity between the parental lines, Rs314940, Rs315000, Rs315310, and Rs315960 were identified as candidate genes mediating the radish bolting and flowering times. This study revealed the genetic complexity of the radish flower color, bolting time, and flowering time traits. The identified candidate genes in the QTL regions may be useful for radish breeding programs and also for functional characterization in radish.

The effects of elicitors on broccoli (Brassica oleracea L. var. Italica) and radish (Raphanus sativus L.) sprouts were evaluated. Seeds and then sprouts were soaked daily for 30 min over 6 days in water (control) or a mixture of FeEDTA and sodium silicate or sodium silicate alone. The contents of the flavonoids and phenolic acids (free, esters, and glycosides) were determined using HPLC-ESI-MS/MS. Phenolic compounds were released from the esters after acid hydrolysis and from the glycosides using alkaline hydrolysis. Quercetin, kaempferol, (‒)-epicatechin, naringenin, apigenin, and luteolin derivatives were found in broccoli and radish sprouts, while derivatives of iso-rhamnetin, orientin, and vitexin were not present at measurable levels. The flavonoid contents, especially derivatives of quercetin, were considerably higher in the broccoli sprouts than in the radish sprouts. The quantitatively major phenolic acid content in the sprouts of both species was found to be p-hydroxybenzoic acid. Its content in the radish sprouts was several times higher than in the broccoli sprouts. The total flavonoid content of broccoli sprouts was 507–734 µg/g DW, while that of the radish sprouts ranged from 155 µg/g DW to 211 µg/g DW. In contrast, total phenolic acids were higher in radish sprouts, ranging from 11,548 to 13,789 µg/g DW, while in broccoli sprouts, they ranged from 2652 to 4527 µg/g DW, respectively. These differences resulted radish sprouts having higher antioxidant activity compared to broccoli sprouts. The applied elicitors increased the content of the total phenolic acids and the antioxidant activity of radish and broccoli sprouts, while they decreased the level of the total flavonoids in broccoli sprouts.

Brassica cover crops have been widely used for improving soil health and as a feed resource for grazing cows, but their use in backgrounding diets is unknown. The objective of this study was to determine the impact of feeding a brassica cover crop mixture during backgrounding on beef palatability. Thirty steers were assigned to one of two dietary treatments during backgrounding with (1) ad libitum access to freshly cut brassica cover crop forage (CC) containing radish, turnip, rapeseed, and rye grass, or (2) common Midwestern dry lot backgrounding diet (CON). The steers were transitioned to a common finishing diet after backgrounding. Striploins were collected after harvest, and were analyzed for evaluation of the Warner–Bratzler shear force (WBSF), collagen content, autolysis of calpain-1, proteolysis of desmin, and troponin-T; in addition, the tenderness, juiciness, and flavor evaluated by a trained sensory panel. A treatment x day interaction was observed for WBSF (p = 0.02). Steaks from the CON diet were less tender than CC steaks on days 3 and 7, but did not differ on days 14 and 21. Feeding a brassica mixture cover crop during the backgrounding phase of production did not impact the collagen content, autolysis of calpain-1, or proteolysis of desmin and troponin-T. Thus, additional investigation into the mechanisms responsible for the differences observed in instrumental tenderness is warranted.

Two pot experiments were conducted to evaluate the response of radish crops against different plant growth regulators, biostimulants, and leaf extracts at Yale University, USA. The first experiment examined the marginal effect of vitamin B12 when added to the Berlyn Laboratory’s proprietary biostimulant formula (GPB Core). Increasing concentrations of vitamin B12 were added, as investigated in groups SL (0 mg/L), SB1 (0.5 mg/L), SB2 (1.0 mg/L), and SB3 (1.5 mg/L). The addition of vitamin B12 conferred no significant incremental benefit over the GPB Core. However, the GPB Core formula (SL) increased fresh shoot biomass by 172.9%, dry shoot biomass by 136.4%, fresh root biomass by 64.7%, and dry root biomass by 29.1% over plant treated with inorganic fertilizer alone (p < 0.01). The second experiment examined the combined marginal effect of vitamin B12 and coenzyme Q10 (CoQ10) when added to the GPB Core. The three experimental groups included the GPB Core plus inorganic fertilizer (S+); GPB Core, vitamin B12, CoQ10, and inorganic fertilizer (SBQ+); and GPB Core, vitamin B12, CoQ10, and no inorganic fertilizer (SBQ0). SBQ0 outperformed the inorganic fertilizer control in fresh shoot, dry shoot, fresh root, and dry root biomass by 190.3%, 127.1%, 128.5%, and 41.3%, respectively (p < 0.01), indicating that inorganic fertilizer can be replaced by biostimulants while simultaneously increasing yield. Additionally, the differences between SBQ+ and SBQ0 in the biomass metrics were statistically insignificant, indicating that in the presence of biostimulants, inorganic fertilizers confer a slight marginal benefit. There was no evidence, however, that the addition of CoQ10 and vitamin B12 conferred benefits over S+. Overall, the application of biostimulants statistically significantly improves radish biomass. Both experiments indicate that under low stress conditions, biostimulants can replace inorganic fertilizer while simultaneously increasing yield.

In the North Central USA, intersowing cover crops into standing maize (Zea mays L.) is required to establish plants large enough to afford the benefits of a cover since there is limited favorable weather for cover crop growth after maize harvest. The objective of this study was to quantify the impacts of the planting method and time of planting of three cover crop species when grown with or without maize competition on their establishment. Experiments were conducted in three environments during 2018 and 2019. Experiments consisted of a factorial combination of timing of cover crop planting (V7 and R4 growth stage of maize), cover crop species (camelina (Camelina sativa (L.) Crantz), rye (Secale cereale L.), or radish (Raphanus sativus L.), method of sowing (drilled or broadcast), and maize removal. Initial cover crop populations were similar regardless of maize removal or stage of maize when sown, but intersown cover crops produced only 3% of the fall biomass, compared with treatments with maize-removed when sown at the V7 stage of maize and 14% when sown at the R4 stage. Limited light intensity (less than 20%) under the maize canopy was the main factor affecting interseeded cover crop development. Radish was more sensitive to shading than the other cover crops. Camelina and rye sown at the R4 stage of corn produced similar spring biomass as earlier-sown cover crops. Intersown cover crops had no negative effect on maize grain yield.

Rhizofiltration experiments were conducted using uranium-contaminated groundwater and lettuce (Lactuca sativa), Chinese cabbage (Brassica campestris L.), radish (Raphanus sativus L.), and buttercup (Oenanthe javanica), which are commonly grown and consumed in South Korea. The results of the rhizofiltration experiments with artificial solutions with different initial uranium concentrations (18, 32, 84, 116, 173, and 263 μg/L) show that the uranium accumulation and bioconcentration factor (BCF) of plant roots increase with increasing uranium concentration in the groundwater. Among the four plants, the uranium concentration in the roots of Raphanus sativus L. is 1215.8 μg/g dry weight, with a maximum BCF value of 2692.7. The BCF value of the artificial solutions with various pH values (pH 3, 5, 7, and 9) is the highest under acidic conditions (pH 3) for all four plants. The uranium BCF values based on different hydroponic conditions range from 170.5 to 11580.3 and the results are comparable with those of other studies using similar methods; the highest BCF value was determined for Brassica campestris L. at pH 3. The BCF values of Raphanus sativus L. after the rhizofiltration experiments with genuine groundwater contaminated with uranium are the highest among the four species; that is, 1684.7 and 1700.1 in Oesam-dong and Bugokdong groundwater samples with uranium concentrations of 83 and 173 μg/L, respectively. The results of the scanning electron microscope/electron dispersive X-ray spectroscope analyses show that uranium in contaminated groundwater is adsorbed as a solid phase on the root surface. These results demonstrate that Raphanus sativus L. has a high tolerance to high concentrations of uranium and low pH conditions and a remarkable potential for uranium accumulation.

Solidago canadensis L. is an expansive perennial that forms persistent, species-poor plant communities. It often spreads in fallow areas, displacing native floristic ingredients. Its expansiveness is largely due to morphological features, but it can also be the effect of allelopathic interaction. The aim of the experiment was to investigate the effect of aqueous extracts (decoction, infusion, and maceration) from dry S. canadensis leaves on germination and early growth stages of Raphanus sativus L. var. radicula Pers., in three cultivars: ‘Rowa’, ‘Półdługa’, and ‘Krakowianka’. In comparison to the control, the percentage of germinated radish seeds of ‘Rowa’ cultivar was statistically lower on the infusion and macerate. Regardless of the cultivar, the smallest changes in germination were found in seeds watered with decoction, and the largest with macerate. Seedlings length was most inhibited on macerate substrates, and least with infusion. Regardless of the form of the extract, each of them negatively affected the initial growth of radish seedlings. A fresh mass of ‘Rowa’ seedlings was inhibited by all Canadian goldenrod extracts. In relation to the control, the ‘Krakowianka’ cultivar was the least sensitive to S. canadensis extracts. The total chlorophyll content was the lowest in the seedlings of the ‘Rowa’ and ‘Półdługa’ cultivars germinated on macerate, compared to the control and two others extracts. The percentage of electrolyte leakage depended on the type of extract used and the radish cultivar. The study showed that depending on the cultivar, the aqueous extracts from S. canadensis leaves decreasing of germination and early growth of R. sativus.

The overturning stability and vibration of upland crop machinery under development are important issues for analysis because farms for upland crops are usually uneven, which may cause work-related fatalities, and vibration affects user comfort and reduces the durability of components. In this study, the overturning stability and vibration of a tractor-mounted radish collector were investigated to ensure safety during radish collection. To analyze lateral stability, the center of gravity (CG) of the tractor-mounted radish collector system was calculated mathematically. Then, a simulation was performed to determine the lateral overturning angles at different folding positions of the radish conveyor belt and load conditions, and the results were validated through tests. Vibration sensors were used to measure the vibration levels and the power spectrum density (PSD) was obtained to check the cyclic apparatuses of the major frequencies. The load conditions, different conveyor speeds, and locations were considered as factors affecting the vibration levels. Considering the physical parameters of the tractor–collector system, the analytical overturning angle was 30.5°. The average overturning angle difference between the simulation and validation was 5°, and the difference between loaded and unloaded conditions was 2°. For 0, 45, and 90° folding positions of the conveyor belt, overturning angles increased and varied from 0.5 to 1°. The vibration level was greater under the unloaded conditions and increased with an increase in the conveyor speed. Vibrations under the loaded condition (0.37~0.48 ms−2) satisfied the ISO (International Organization for Standardization) standard (except the first conveyor belt). According to the PSD analysis, high magnitude peaks (>25 dB) appeared frequently in all directions, which indicates a high possibility of damage to the first conveyor belt. This study provides useful information for improving the safety and durability of agricultural machinery for uneven and sloped field conditions.

The CPA (cation proton antiporter) family plays an essential role during plant stress tolerance by regulating ionic and pH homeostasis of the cell. Radish fleshy roots are susceptible to abiotic stress during growth and development, especially salt stress. To date, CPA family genes have not yet been identified in radish and the biological functions remain unclear. In this study, 60 CPA candidate genes in radish were identified on the whole genome level, which were divided into three subfamilies including the Na+/H+ exchanger (NHX), K+ efflux antiporter (KEA), and cation/H+ exchanger (CHX) families. In total, 58 of the 60 RsCPA genes were localized to the nine chromosomes. RNA-seq. data showed that 60 RsCPA genes had various expression levels in the leaves, roots, cortex, cambium, and xylem at different development stages, as well as under different abiotic stresses. RT–qPCR analysis indicated that all nine RsNHXs genes showed up regulated trends after 250 mM NaCl exposure at 3, 6, 12, and 24h. The RsCPA31 (RsNHX1) gene, which might be the most important members of the RsNHX subfamily, exhibited obvious increased expression levels during 24h salt stress treatment. Heterologous over-and inhibited-expression of RsNHX1 in Arabidopsis showed that RsNHX1 had a positive function in salt tolerance. Furthermore, a turnip yellow mosaic virus (TYMV)-induced gene silence (VIGS) system was firstly used to functionally characterize the candidate gene in radish, which showed that plant with the silence of endogenous RsNHX1 was more susceptible to the salt stress. According to our results we provide insights into the complexity of the RsCPA gene family and a valuable resource to explore the potential functions of RsCPA genes in radish.

Allelopathic potential of 10 teff varieties was assessed in laboratory experimentation (conducted in NIBIO, Norway), and determined with an agar-based bioassay using ryegrass and radish as model weeds. Field experiments were conducted in Tigray, Ethiopia during 2015 and 2016 to identify the most important agronomic traits of teff contributing to its weed competitive ability. A split plot design with three blocks was used considering hand weeding as the main plot and varieties as the subplot. Randomized complete block design (RCBD) with four blocks was used in the laboratory experiment. The highest potential allelopathic activity (PAA) and specific potential allelopathic activity (SPAA) were recorded from a local landrace with an average PAA value of 11.77% and SPAA value of 1.21%/mg respectively, when ryegrass was used as the model weed. ‘Boset’ had the highest average PAA value of 16.25% and an SPAA value of 1.53%/mg, when using radish as the model weed. The lowest PAA and SPAA values were recorded from ‘DZ-Cr-387′ when using ryegrass and radish as model weeds. Days to emergence, height, tiller no./plant, biomass yield, and PAA of the crop significantly contributed to the variance of the weed biomass, cover, and density. Hence, they were the most important agronomic traits enhancing the competitive ability of teff.
