As triboelectric nanogenerator (TENG) technology continue to evolve, its application in harsh environments has increasingly captivated the interest of researchers. However, the current research on heat-resistant triboelectric materials remains predominantly focused on the development of tribo-negative materials, with scant attention given to their equally crucial tribo-positive counterparts. In this study, the tribo-positive polyimide (PI) material with enhanced tribo-positivity is developed by integrating amide groups with electron-donating effects into the molecular chain. Furthermore, the TENG devices based on this series of tribo-positive PI materials have demonstrated an open-circuit voltage (VOC) of 242 V, a short-circuit current (ISC) of 8.13 μA, and a transferred charge (QSC) of 117 nC. Notably, these devices also demonstrate the capability to efficiently generate electricity even under elevated temperature conditions. This work not only proposes a potential molecular design strategy for developing high-performance tribo-positive PI materials applicable in TENGs, but also markedly propels the advancement of robust energy-harvesting devices engineered for operation at elevated temperatures.

Cell death is of great significance in maintaining tissue homeostasis and bodily functions. With considerable research coming to the fore, it has been found that programmed cell death presents in multiple modalities in the body, which is not only limited to apoptosis, but also can be divided into autophagy, pyroptosis, ferroptosis, mitotic catastrophe, entosis, netosis, and other ways. Different forms of programmed cell death have disparate or analogous characteristics with each other, and their occurrence is accompanied by multiple signal transduction and the role of a myriad of regulatory factors. In recent years, scholars across the world have carried out considerable in-depth research on programmed cell death, and new forms of cell death are being discovered continually. Concomitantly, the mechanisms of intricate signaling pathways and regulators have been discovered. More critically, cancer cells tend to choose distinct ways to evade cell death, and different tumors adapt to different manners of death. Therefore, targeting the cell death network has been regarded as an effective tumor treatment strategy for a long time. The objective of our paper is to review the signaling pathways and gene regulation in several typical types of programmed cell death and their correlation with cancer.

Ophiocordyceps sinensis (Berk.) is a complex is formed by Hepialidae larvae and Hirsutella sinensis. Infestation by H. sinensis, interaction with host larvae, and fruiting body development are three crucial processes affecting the formation of O. sinensis. However, research on the molecular mechanism of O. sinensis formation has been hindered by the lack of effective genetic transformation protocols. Therefore, Agrobacterium tumefaciens-mediated transformation (ATMT) was adopted to genetically transform two H. sinensis strains and optimize the transformation conditions. The results revealed that the most suitable Agrobacterium strain for H. sinensis transformation was AGL1, and that the surfactant Triton X-100 could also induce ATMT, although less effectively than acetosyringone (AS). In addition, the endogenous promoters of H. sinensis genes had a stronger ability to drive the expression of the target gene than did the exogenous promoter. The optimal transformation conditions were as follows: AS and hygromycin B concentrations of 100 μM and 50 μg/mL, respectively; A. tumefaciens OD600 of 0.4; cocultivation at 18 °C for 24 h; and H. sinensis used within three passages. The results lay a foundation for the functional study of key regulatory genes involved in the formation of O. sinensis.

In this study, we investigated the surface-confined coupling reactions of 1,8-dibromobiphenylene (BPBr2) on Cu(111) to elucidate the details of the organometallic intermediates via Ullmann reactions. We used scanning tunneling microscopy (STM) to characterize the resulting organometallic intermediates. Moreover, submolecular resolution of the non-contact atomic force microscopy (nc-AFM) qPlus technique enables the bond-resolving within the organometallic dimer product. Our findings reveal the debromination of BPBr2 on Cu(111), leading to the formation of an organometallic dimer intermediate at room temperature. Through nc-AFM measurements, we confirm and visualize the formation of the C-Cu-C bond. These insights enhance our understanding of Ullmann reaction and hold potential implications for the design of novel two-dimensional electronic devices.

Nano-encapsulation and conjugation are the main strategies employed for drug delivery. Nanoparticles help improve encapsulation and targeting efficiency, thus optimizing therapeutic efficacy. Through nanoparticle technology, replacement of a defective gene or delivery of a new gene into a patient’s genome has become possible. Lipid nanoparticles (LNPs) loaded with genetic materials are designed to be delivered to specific target sites to enable gene therapy. The lipid shells protect the fragile genetic materials from degradation, then successfully release the payload inside of the cells, where it can integrate into the patient’s genome and subsequently express the protein of interest. This review focuses on the development of LNPs and nano-pharmaceutical techniques for improving the potency of gene therapies, reducing toxicities, targeting specific cells, and releasing genetic materials to achieve therapeutic effects. In addition, we discuss preparation techniques, encapsulation efficiency, and the effects of conjugation on the efficacy of LNPs in delivering nucleic acid materials.

This study evaluates the efficiency of China’s seven emission trading schemes (ETS) piloted in 2013. We evaluate seven pilots’ overall technical and scale efficiencies and temporal dynamics during 2014–2023. We use a bootstrap correction data envelopment analysis (bootstrap-DEA), which guarantees a more accurate efficiency estimation than the traditional DEA model. The results show that the average overall (pure technical) efficiency of the seven pilot markets increased from 0.612 (0.844) in 2014 to 0.898 (0.990) in 2023. Furthermore, we document that seven ETS pilots differ remarkably in efficiency and transaction price, whilst all have shortages. Specifically, the small-scale market transaction is the main constraint effect on the average scale efficiency of the ETS. This study provides concrete recommendations for policy makers to consummate institutional designs to improve ETS efficiency.

Liver fibrosis is an important step in the transformation of chronic liver disease into cirrhosis and liver cancer, and structural changes and functional disorders of liver sinusoidal endothelial cells (LSECs) are early events in the occurrence of liver fibrosis. Therefore, it is necessary to identify the key regulatory genes of endothelial dysfunction in the process of liver fibrosis to provide a reference for the diagnosis and treatment of liver fibrosis. In this study, we identified 230 common differentially expressed genes (Co-DEGs) by analyzing transcriptomic data of primary LSECs from three different liver fibrosis mouse models (carbon tetrachloride; choline-deficient, l-amino acid-defined diet; and nonalcoholic steatohepatitis). Enrichment analysis revealed that the Co-DEGs were mainly involved in regulating the inflammatory response, immune response, angiogenesis, formation and degradation of the extracellular matrix, and mediating chemokine-related pathways. A Venn diagram analysis was used to identify 17 key genes related to the progression of liver cirrhosis. Regression analysis using the Lasso–Cox method identified genes related to prognosis among these key genes: SOX4, LGALS3, SERPINE2, CD52, and LPXN. In mouse models of liver fibrosis (bile duct ligation and carbon tetrachloride), all five key genes were upregulated in fibrotic livers. This study identified key regulatory genes for endothelial dysfunction in liver fibrosis, namely SOX4, LGALS3, SERPINE2, CD52, and LPXN, which will provide new targets for the development of therapeutic strategies targeting endothelial dysfunction in LSECs and liver fibrosis.

The Sustainable Development Goals (SDGs) have set the agenda for 2030, calling for collective global efforts to deal with climate change while seeking a balance between economic development and environmental protection. Although many countries are exploring emission reduction paths, mainly from government and corporate perspectives, addressing climate change is also an individual responsibility and requires public participation in collective action. The millennial generation constitutes the current workforce and will be the leaders in climate action for the next 30 years. Therefore, our study focuses on the Chinese millennial generation, conducting in-depth semi-structured interviews with 50 participants in qualitative research to explore their low-carbon lifestyles, the barriers, and enablers in switching to a wider range of low-carbon lifestyles. There are three main results: (1) Based on our study samples, there is an indication that Chinese millennials have a positive attitude towards transitioning to a low-carbon lifestyle. Women demonstrate a stronger willingness to adopt low-carbon behaviors in their daily household activities compared to men. However, their involvement in governance in the context of transitioning to a low-carbon society is limited, with most women assuming execution roles in climate action rather than decision-making positions. (2) Millennial’s low-carbon life transition is accompanied by technological innovation and progress. However, this progress brings some new forms of resource waste, and reasonable policy-making is essential. (3) Personal economic interests and the satisfaction of their consumption needs will drive millennials to reduce carbon emissions in their daily lives, but it requires the guidance of reasonable policy-making and synergies among various stakeholders. This research will help policymakers better understand the current status and potential issues related to people’s low-carbon actions, enabling the formulation of more rational guiding policies. It can also help other stakeholders learn about millennials’ demands and take more effective collective action toward carbon reduction.

Chemical exchange saturation transfer (CEST) magnetic resonance imaging (MRI) is a novel MRI technology to image certain compounds at extremely low concentrations. Long acquisition time to measure signals at a set of offset frequencies of the Z-spectra and to repeat measurements to reduce noise pose significant challenges to its applications. This study explores correlations of CEST MR images along the spatial and Z-spectral dimensions to improve MR image quality and robustness of magnetization transfer ratio (MTR) asymmetry estimation via a joint k-ω reconstruction model. The model was formulated as an optimization problem with respect to MR images at all frequencies ω, while incorporating regularizations along the spatial and spectral dimensions. The solution was subject to a self-consistency condition that the Z-spectrum of each pixel follows a multi-peak data fitting model corresponding to different CEST pools. The optimization problem was solved using the alternating direction method of multipliers. The proposed joint reconstruction method was evaluated on a simulated CEST MRI phantom and semi-experimentally on choline and iopamidol phantoms with added Gaussian noise of various levels. Results demonstrated that the joint reconstruction method was more tolerable to noise and reduction in number of offset frequencies by improving signal-to-noise ratio (SNR) of the reconstructed images and reducing uncertainty in MTR asymmetry estimation. In the choline and iopamidol phantom cases with 10.5% noise in the measurement data, our method achieved an averaged SNR of 31.0 dB and 32.2 dB compared to the SNR of 24.7 dB and 24.4 dB in the conventional reconstruction approach. It reduced uncertainty of the MTR asymmetry estimation over all regions of interest by 54.4% and 43.7%, from 1.71 and 2.38 to 0.78 and 1.71, respectively.

Clostridium perfringens (C. perfringens) is an important veterinary pathogen and a noteworthy threat to human and animal health. Recently, there has been a significant rise in the number of moose fatalities caused by this rare, endemic species in China. Currently, there is an increasing trend in conducting whole-genome analysis of C. perfringens strains originating from pigs and chickens, whereas fewer studies have been undertaken on Elaphurus davidianus-originating strains at the whole-genome level. Our laboratory has identified and isolated five C. perfringens type A from affected Elaphurus davidianus. The current study identified the most potent strain of C. perfringens, which originated from Elaphurus davidianus, and sequenced its genome to reveal virulence genes and pathogenicity. Our findings show that strain CX1-4 exhibits the highest levels of phospholipase activity, hemolytic activity, and mouse toxicity compared to the other four isolated C. perfringens type A strains. The chromosome sequence length of the CX1-4 strain was found to be 3,355,389 bp by complete genome sequencing. The current study unveils the genomic characteristics of C. perfringens type A originating from Elaphurus davidianus. It provides a core foundation for further investigation regarding the prevention and treatment of such infectious diseases in Elaphurus davidianus.

AI is considered a key driver of industrial transformation and a strategic technology that will shape future development. With AI services continuing to permeate various sectors, concerns have emerged about the ethics of AI. This study investigates the effects of different types of AI services (mechanical, thinking, and affective AI services) on consumers’ attitudes through offline and online AI service experiments. We also construct a model to explore the mediating roles of identity threat and perceived control. The findings reveal that mechanical AI services negatively affect consumers’ attitudes while thinking and affective AI services have a positive effect. Additionally, we explore how consumers’ attitudes vary across different service scenarios and ethical judgments (utilitarianism and deontology). Our findings could offer practical guidance for enterprises providing AI services.

Objective: Functional magnetic resonance imaging (fMRI) has been applied to assess the microstructure of the kidney. However, it is not clear whether fMRI could be used in the field of kidney injury in patients with Antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV). Methods: This study included 20 patients with AAV. Diffusion kurtosis imaging (DKI) and blood oxygen level-dependent (BOLD) scanning of the kidneys were performed in AAV patients and healthy controls. The mean kurtosis (MK), mean diffusivity (MD), and fractional anisotropy (FA) parameters of DKI, the R2* parameter of BOLD, and clinical data were further analyzed. Results: In AAV patients, the cortex exhibited lower MD but higher R2* values compared to the healthy controls. Medullary MK values were elevated in AAV patients. Renal medullary MK values showed a positive correlation with serum creatinine levels and negative correlations with hemoglobin levels and estimated glomerular filtration rate. To assess renal injury in AAV patients, AUC values for MK, MD, FA, and R2* in the cortex were 0.66, 0.67, 0.57, and 0.55, respectively, and those in the medulla were 0.81, 0.77, 0.61, and 0.53, respectively. Conclusions: Significant differences in DKI and BOLD MRI parameters were observed between AAV patients with kidney injuries and the healthy controls. The medullary MK value in DKI may be a noninvasive marker for assessing the severity of kidney injury in AAV patients.

Considering the lack of studies on the transient vibro-acoustic properties of conical shell structures, a Jacobi–Ritz boundary element method for forced vibro-acoustic behaviors of structure is proposed based on the Newmark-β integral method and the Kirchhoff time domain boundary integral equation. Based on the idea of the differential element method and the first-order shear deformation theory (FSDT), the vibro-acoustic model of conical shells is established. The axial and circumferential displacement tolerance functions are expressed using Jacobi polynomials and the Fourier series. The time domain response of the forced vibration of conical shells is calculated based on the Rayleigh–Ritz method and Newmark-β integral method. On this basis, the time domain response of radiated noise is solved based on the Kirchhoff integral equation, and the acoustic radiation characteristics of conical shells from forced vibration are analyzed. Compared with the coupled FEM/BEM method, the numerical results demonstrate the high accuracy and great reliability of this method. Furthermore, the semi-vertex angle, load characteristics, and boundary conditions related to the vibro-acoustic response of conical shells are examined.

Popularity is of great developmental significance to early adolescents for its implications on behavioral and adjustment outcomes. Although extensive research on popularity has been conducted in Western cultures, little is known about Chinese adolescents’ indigenous understanding of popularity. The current study examined Chinese early adolescents’ understanding of popularity using focus group interviews to explore the characteristics of popularity and unpopularity, as well as popularity determinants for boys and girls. We conducted interviews on six focus groups comprising 37 Chinese fifth and sixth graders (18 boys, 19 girls). Transcriptions were coded and analyzed through developing coding schemes that included emerged codes and categories. The results showed both cross-cultural similarities and culturally specific findings in adolescents’ perceptions of popularity characteristics and determinants. Specifically, sociability, academic performance, and prosocial behaviors served as major characteristics of popularity, whereas aggressive and disruptive behaviors were regarded as typical attributes of unpopularity. More importantly, culturally unique aspects of popularity characteristics were reported by Chinese adolescents, such as a strong sense of collective honor. When responding what would make a peer popular, Chinese early adolescents reported various behavioral, social, and cultural factors. Findings of this study underscore the importance of examining perceptions of popularity in consideration of the cultural context.

Canada’s decentralized healthcare system may lead to regional disparities in survival among Canadians diagnosed with central nervous system (CNS) tumours. We identified 50,670 patients diagnosed with a first-ever primary CNS tumour between 2008 and 2017 with follow-up until 31 December 2017. We selected the four highest incidence histologies and used proportional hazard regression to estimate hazard ratios (HRs) for five regions (British Columbia, Prairie Provinces, Ontario, Atlantic Provinces and the Territories), adjusting for sex, tumour behaviour and patient age. Ontario had the best survival profile for all histologies investigated. The Atlantic Provinces had the highest HR for glioblastoma (HR = 1.26, 95% CI: 1.18–1.35) and malignant glioma not otherwise specified (NOS) (Overall: HR = 1.87, 95% CI:1.43–2.43; Pediatric population: HR = 2.86, 95% CI: 1.28–6.39). For meningioma, the Territories had the highest HR (HR = 2.44, 95% CI: 1.09–5.45) followed by the Prairie Provinces (HR = 1.52, 95% CI: 1.38–1.67). For malignant unclassified tumours, the highest HRs were in British Columbia (HR = 1.45, 95% CI: 1.22–1.71) and the Atlantic Provinces (HR = 1.40, 95% CI: 1.13–1.74). There are regional differences in the survival of CNS patients at the population level for all four specific histological types of CNS tumours investigated. Factors contributing to these observed regional survival differences are unknown and warrant further investigation.

The lifespan of core graphite under neutron irradiation in a commercial molten salt reactor (MSR) has an important influence on its economy. Flattening the fast neutron flux (≥0.05 MeV) distribution in the core is the main method to extend the graphite irradiation lifespan. In this paper, the effects of the key parameters of MSRs on fast neutron flux distribution, including volume fraction (VF) of fuel salt, pitch of hexagonal fuel assembly, core zoning, and layout of control rod assemblies, were studied. The fast neutron flux distribution in a regular hexagon fuel assembly was first analyzed by varying VF and pitch. It was demonstrated that changing VF is more effective in reducing the fast neutron flux in both global and local graphite blocks. Flattening the fast neutron flux distribution of a commercial MSR core was then carried out by zoning the core into two regions under different VFs. Considering both the fast neutron flux distribution and burnup depth, an optimized core was obtained. The fast neutron flux distribution of the optimized core was further flattened by the rational arrangement of control rod channels. The calculation results show that the final optimized core could reduce the maximum fast neutron flux of the graphite blocks by about 30% and result in a more negative temperature reactivity coefficient, while slightly decreasing the burnup and maintaining a fully acceptable core temperature distribution.

Previous studies have mainly focused on the impact of social media self-presentation on individuals’ happiness and mental health, whereas few have attended to its potential influence on consumer behavior. A mixed-method empirical study with an online survey (N = 408) and an experimental study (N = 160) investigated the effects of social media self-presentation on individuals’ conspicuous consumption in China. We found that consumers’ self-presentation behavior on social media promoted tendencies for social comparison, thereby enhancing their conspicuous consumption. However, compared with individuals using social media less often, self-presentation had a more significant impact on the social comparison tendencies of users who used social media more often. Our study explains why individuals’ self-presentation on social media increases their conspicuous consumption tendencies. The study results contribute to the existing knowledge of how consumers’ behavior on social media affects their product preferences.

To decrease privacy risks, consumers may choose to misrepresent themselves when they are asked to offer personal information. Using a game theoretic model, this study examines the impact of consumer misrepresentation on both a monopolistic firm and consumers. The results show that consumer misrepresentation may benefit the firm, but hurt consumers under certain conditions. In addition, we find that when the unit cost of personalized service is low, consumer misrepresentation may encourage the firm to provide a higher personalized service level. Moreover, when consumers misrepresent themselves and the firm only covers part of the market, a greater unit value of consumer private information will reduce the firm’s profit, while a greater unit cost of personalized service will increase the firm’s profit. The analysis reported here provides important insights regarding the application of consumer information in online personalized marketing and consumer privacy protection.

Software vulnerability detection aims to proactively reduce the risk to software security and reliability. Despite advancements in deep-learning-based detection, a semantic gap still remains between learned features and human-understandable vulnerability semantics. In this paper, we present an XAI-based framework to assess program code in a graph context as feature representations and their effect on code vulnerability classification into multiple Common Weakness Enumeration (CWE) types. Our XAI framework is deep-learning-model-agnostic and programming-language-neutral. We rank the feature importance of 40 syntactic constructs for each of the top 20 distributed CWE types from three datasets in Java and C++. By means of four metrics of information retrieval, we measure the similarity of human-understandable CWE types using each CWE type’s feature contribution ranking learned from XAI methods. We observe that the subtle semantic difference between CWE types occurs after the variation in neighboring features’ contribution rankings. Our study shows that the XAI explanation results have approximately 78% Top-1 to 89% Top-5 similarity hit rates and a mean average precision of 0.70 compared with the baseline of CWE similarity identified by the open community experts. Our framework allows for code vulnerability patterns to be learned and contributing factors to be assessed at the same stage.

Changes in intracellular nicotinamide adenine dinucleotide (NAD+) levels have been observed in various disease states. A decrease in NAD+ levels has been noted following spinal cord injury (SCI). Nicotinamide riboside (NR) serves as the precursor of NAD+. Previous research has demonstrated the anti-inflammatory and apoptosis-reducing effects of NR supplements. However, it remains unclear whether NR exerts a similar role in mice after SCI. The objective of this study was to investigate the impact of NR on these changes in a mouse model of SCI. Four groups were considered: (1) non-SCI without NR (Sham), (2) non-SCI with NR (Sham +NR), (3) SCI without NR (SCI), and (4) SCI with NR (SCI + NR). Female C57BL/6J mice aged 6–8 weeks were intraperitoneally administered with 500 mg/kg/day NR for a duration of one week. The supplementation of NR resulted in a significant elevation of NAD+ levels in the spinal cord tissue of mice after SCI. In comparison to the SCI group, NR supplementation exhibited regulatory effects on the chemotaxis/recruitment of leukocytes, leading to reduced levels of inflammatory factors such as IL-1β, TNF-α, and IL-22 in the injured area. Moreover, NR supplementation notably enhanced the survival of neurons and synapses within the injured area, ultimately resulting in improved motor functions after SCI. Therefore, our research findings demonstrated that NR supplementation had inhibitory effects on leukocyte chemotaxis, anti-inflammatory effects, and could significantly improve the immune micro-environment after SCI, thereby promoting neuronal survival and ultimately enhancing the recovery of motor functions after SCI. NR supplementation showed promise as a potential clinical treatment strategy for SCI.

Background: Machine learning (ML) and artificial intelligence (AI)-based classifiers can be used to diagnose diseases from medical imaging data. However, few of the classifiers proposed in the literature translate to clinical use because of robustness concerns. Materials and methods: This study investigates how to improve the robustness of AI/ML imaging classifiers by simultaneously applying perturbations of common effects (Gaussian noise, contrast, blur, rotation, and tilt) to different amounts of training and test images. Furthermore, a comparison with classifiers trained with adversarial noise is also presented. This procedure is illustrated using two publicly available datasets, the PneumoniaMNIST dataset and the Breast Ultrasound Images dataset (BUSI dataset). Results: Classifiers trained with small amounts of perturbed training images showed similar performance on unperturbed test images compared to the classifier trained with no perturbations. Additionally, classifiers trained with perturbed data performed significantly better on test data both perturbed by a single perturbation (p-values: noise = 0.0186; contrast = 0.0420; rotation, tilt, and blur = 0.000977) and multiple perturbations (p-values: PneumoniaMNIST = 0.000977; BUSI = 0.00684) than the classifier trained with unperturbed data. Conclusions: Classifiers trained with perturbed data were found to be more robust to perturbed test data than the unperturbed classifier without exhibiting a performance decrease on unperturbed test images, indicating benefits to training with data that include some perturbed images and no significant downsides.

The tandem solar cell presents a potential solution to surpass the Shockley–Queisser limit observed in single-junction solar cells. However, creating a tandem device that is both cost-effective and highly efficient poses a significant challenge. In this study, we present proof of concept for a four-terminal (4T) tandem solar cell utilizing a wide bandgap (1.6–1.8 eV) perovskite top cell and a narrow bandgap (1.2 eV) antimony selenide (Sb2Se3) bottom cell. Using a one-dimensional (1D) solar cell capacitance simulator (SCAPS), our calculations indicate the feasibility of this architecture, projecting a simulated device performance of 23% for the perovskite/Sb2Se3 4T tandem device. To validate this, we fabricated two wide bandgap semitransparent perovskite cells with bandgaps of 1.6 eV and 1.77 eV, respectively. These were then mechanically stacked with a narrow bandgap antimony selenide (1.2 eV) to create a tandem structure, resulting in experimental efficiencies exceeding 15%. The obtained results demonstrate promising device performance, showcasing the potential of combining perovskite top cells with the emerging, earth-abundant antimony selenide thin film solar technology to enhance overall device efficiency.

Farnesyl pyrophosphate synthase (FPPS) catalyzes the synthesis of C15 farnesyl diphosphate (FPP) from C5 dimethylallyl diphosphate (DMAPP) and two or three C5 isopentenyl diphosphates (IPPs). FPP is an important precursor for the synthesis of isoprenoids and is involved in multiple metabolic pathways. Here, farnesyl pyrophosphate synthase from Sporobolomyces pararoseus NGR (SpFPPS) was isolated and expressed by the prokaryotic expression system. The SpFPPS full-length genomic DNA and cDNA are 1566 bp and 1053 bp, respectively. This gene encodes a 350-amino acid protein with a predicted molecular mass of 40.33 kDa and a molecular weight of 58.03 kDa (40.33 kDa + 17.7 kDa), as detected by SDS-PAGE. The function of SpFPPS was identified by induction, purification, protein concentration and in vitro enzymatic activity experiments. Structural analysis showed that Y90 was essential for chain termination and changing the substrate scope. Site-directed mutation of Y90 to the smaller side-chain amino acids alanine (A) and lysine (K) showed in vitro that wt-SpFPPS catalyzed the condensation of the substrate DMAPP or geranyl diphosphate (GPP) with IPP at apparent saturation to synthesize FPP as the sole product and that the mutant protein SpFPPS-Y90A synthesized FPP and C20 geranylgeranyl diphosphate (GGPP), while SpFPPS-Y90K hydrolyzed the substrate GGPP. Our results showed that FPPS in S. pararoseus encodes the SpFPPS protein and that the amino acid substitution at Y90 changed the distribution of SpFPPS-catalyzed products. This provides a baseline for potentially regulating SpFPPS downstream products and improving the carotenoid biosynthesis pathway.

In recent years, with the rapid development of live-streaming commerce, the social dynamics and psychological impact of such online activities merit further discussion. In this study, we investigate the sensory experiences of viewers watching live streaming and examine how these online experiences influence consumer well-being. We developed a conceptual model to understand this mechanism based on the relationship between social presence, shopping enjoyment, familiarity, and consumer well-being. The results of 410 samples indicate that (1) social presence in live-streaming commerce has a significant positive effect on consumer well-being; (2) shopping enjoyment plays a mediating role in the process of social presence predicting consumer well-being; and (3) familiarity plays a moderating role in the second half of the indirect effect of social presence on well-being. This study examines the relationship between social presence and consumer well-being in the context of live-streaming marketing, expanding the research scenario of consumer well-being and clarifying the psychological mechanisms and boundary conditions of the effect of social presence on consumers well-being, which has important implications for online interactive marketing enterprises to enhance social presence and promote consumers long-term well-being.

Since reef-building corals rely on both heterotrophy and endosymbiotic dinoflagellate autotrophy to meet their metabolic needs, it is necessary to consider both food supply and light levels, respectively, when optimizing their cultivation ex situ. Herein nubbins of the model reef coral Pocillopora acuta cultured in recirculating aquaculture systems at photosynthetically active radiation levels of 370 or 670 μmol quanta m−2 s−1 were fed Artemia nauplii at concentrations of either 33 or 78 individuals mL−1 in a separate feeding tank for 6 hr in the dark thrice weekly. A subset of nubbins was experimentally wounded at the outset of the 84-day experiment to assess recovery, and 100% fully healed within 2–4 weeks. All cultured corals survived, and unwounded corals (1) grew at a specific growth rate approaching 0.5% day−1 and (2) demonstrated a mean total linear extension of 0.2% day−1 (~6–8 cm year−1); these are far higher than growth rates normally documented in situ. In the feeding tank, corals tolerated nitrate levels up to 25 mg L−1, but once concentrations reached 50 mg L−1 by day 84, tissue necrosis began to occur in nubbins of one tank. This highlights the importance of feeding in separate tanks during long-term culture of corals, and bio-filtration could reduce the possibility of organic matter accumulation in future coral culture studies.

Auriculotemporal neuralgia is a rare facial pain disorder with no therapeutic evidence for refractory cases. We described a male patient with right auriculotemporal neuralgia, refractory to anesthetic nerve blocks and botulinum toxin type A injections, who was successfully treated with pulsed radiofrequency without adverse events. Pulsed radiofrequency may be an effective and safe treatment for refractory auriculotemporal neuralgia.

An increased neutrophil-to-lymphocyte ratio (NLR) is a poor prognostic biomarker in various types of cancer, because it reflects the inhibition of lymphocytes in the circulation and tumors. In urologic cancers, upper tract urothelial carcinoma (UTUC) is known for its aggressive features and lack of T cell infiltration; however, the association between neutrophils and suppressed T lymphocytes in UTUC is largely unknown. In this study, we examined the relationship between UTUC-derived factors and tumor-associated neutrophils or T lymphocytes. The culture supernatant from UTUC tumor tissue modulated neutrophils to inhibit T cell proliferation. Among the dominant factors secreted by UTUC tumor tissue, apolipoprotein A1 (Apo-A1) exhibited a positive correlation with NLR. Moreover, tumor-infiltrating neutrophils were inversely correlated with tumor-infiltrating T cells. Elevated Apo-A1 levels in UTUC were also inversely associated with the population of tumor-infiltrating T cells. Our findings indicate that elevated Apo-A1 expression in UTUC correlates with tumor-associated neutrophils and T cells. This suggests a potential immunomodulatory effect on neutrophils and T cells within the tumor microenvironment, which may represent therapeutic targets for UTUC treatment.

The immune system plays a crucial role in defending the body against foreign invaders, and the balance of various polyunsaturated fatty acids, such as alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), can impact immune cell functions and overall immune responses. This study aimed to assess the effectiveness of mussel oil extracts in modulating inflammatory responses by analysing their effects on immune cell lines and cytokine expression. Four different mussel oil extracts were obtained using two extraction methods (organic solvent and supercritical CO2 extraction) from two tissue sources (fresh and commercial). These extracts were then tested at various concentrations on T lymphocyte (Jurkat) cells, monocytes, and macrophages (THP-1 and U-937). Cytokine levels were quantified using ELISA. The results showed that the solvent-extracted samples had a dose-dependent effect on tumour necrosis factor alpha (TNF-α) and interleukin-1 beta (IL-1β) cytokine secretion in THP-1 and U937 cells, with the extract from a commercial mussel powder being more efficient than the extract from fresh powder. However, supercritical CO2 samples showed elevated cytokine secretion levels despite their high omega-3 content. Furthermore, 100 ug/mL extract from fresh powder successfully reduced interleukin-2 (IL-2) secretion while maintaining cell viability after stimulation. The study demonstrated that solvent-extracted mussel oil can effectively regulate cytokine secretion, modulate immune cell activation, and alleviate inflammation. These findings offer valuable insights into using mussel oil extracts to treat inflammatory disorders and enhance immune responses.

In buried plastic water pipes, the predominantly fluid-borne wave is of particular interest, as it plays a key role in the propagation of leak noise. Consequently, it has been studied by several researchers to determine the speed of wave propagation and its attenuation with distance. These features are encapsulated in the wavenumber. By examining the factors that govern the behaviour of this wavenumber, this paper presents an in-depth examination of the physical mechanisms of leak noise propagation. To achieve this, an alternative physics-based model for the wavenumber is developed, using the concept of the wave dynamic stiffnesses of the individual components within the pipe system, i.e., the water in the pipe, the pipe wall, and the surrounding medium. This facilitates a clear interpretation of the wave behaviour in terms of the physical properties of the system, especially the interface between the pipe and the surrounding medium, which can have a profound influence on the leakage of acoustic energy from the pipe wall into the external medium. Three systems with different types of surrounding medium are studied, and the factors that govern leak noise propagation in each case are identified. Experimental results on two distinct test sites from different parts of the world are provided to validate the approach using leak noise as an excitation mechanism.

Based on the unique properties of optical Fano resonance and plasmonic-waveguide coupling systems, this paper explores a novel refractive index concentration sensor structure. The sensor structure is composed of a metal–insulator–metal (MIM) waveguide and two identically shaped and sized double-tooth ring couplers (DTR). The performance structure of the nanoscale refractive index sensor with DTR cavity was comprehensively assessed using the finite element method (FEM). Due to the impact of various geometric parameters on the sensing characteristics, including the rotation angles, the widths between the double-tooth rings, and the gaps between the cavity and the waveguide, we identified an optimal novel refractive index sensor structure that boasts the best performance indices. Finally, the DTR cavity sensor achieved a sensitivity of 4137 nm/RIU and Figure of merit (FOM) of 59.1. Given the high complexity and sensitivity of the overall structure, this nanoscale refractive index sensor can be applied to the detection of oil concentration in industrial oil–water mixtures, yielding highly precise results.

Autism spectrum disorder (ASD) is thought to result from susceptibility genotypes and environmental risk factors. The offspring of women who experience pregnancy infection have an increased risk for autism. Maternal immune activation (MIA) in pregnant animals produces offspring with autistic behaviors, making MIA a useful model for autism. However, how MIA causes autistic behaviors in offspring is not fully understood. Here, we show that NKCC1 is critical for mediating autistic behaviors in MIA offspring. We confirmed that MIA induced by poly(I:C) infection during pregnancy leads to autistic behaviors in offspring. We further demonstrated that MIA offspring showed significant microglia activation, excessive dendritic spines, and narrow postsynaptic density (PSD) in their prefrontal cortex (PFC). Then, we discovered that these abnormalities may be caused by overexpression of NKCC1 in MIA offspring’s PFCs. Finally, we ameliorated the autistic behaviors using PFC microinjection of NKCC1 inhibitor bumetanide (BTN) in MIA offspring. Our findings may shed new light on the pathological mechanisms for autism caused by pregnancy infection.

The diversity of leaf characteristics, particularly leaf color, underscores a pivotal area of inquiry within plant science. The synthesis and functionality of chlorophyll, crucial for photosynthesis, largely dictate leaf coloration, with varying concentrations imparting different shades of green. Complex gene interactions regulate the synthesis and degradation of chlorophyll, and disruptions in these pathways can result in abnormal chlorophyll production, thereby affecting leaf pigmentation. This study focuses on Bambusa multiplex f. silverstripe, a natural variant distinguished by a spectrum of leaf colors, such as green, white, and green–white, attributed to genetic variations influencing gene expression. By examining the physiological and molecular mechanisms underlying chlorophyll anomalies and genetic factors in Silverstripe, this research sheds light on the intricate gene interactions and regulatory networks that contribute to leaf color diversity. The investigation includes the measurement of photosynthetic pigments and nutrient concentrations across different leaf color types, alongside transcriptomic analyses for identifying differentially expressed genes. The role of key genes in pathways such as ALA biosynthesis, chlorophyll synthesis, photosynthesis, and sugar metabolism is explored, offering critical insights for advancing research and plant breeding practices.

The coating effect of 1,2-bis(triethoxysilyl)ethane (BTES) on carbonyl iron particles (CIPs) was enhanced by etching with hydrochloric acid (HCl) of various concentrations, and magnetorheological fluids (MRFs) with significantly improved dispersion stability were obtained. The microstructures, coating effect, and magnetism of CIPs were examined using scanning electron microscopy (SEM), automatic surface and porosity analysis (BTE), Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), and a vibrating sample magnetometer (VSM), respectively. Furthermore, the rheological properties and dispersion stability of the MRFs were assessed by a rotating rheometer and a Turbiscan Tower. The results show that as the HCl concentration increased, nanopores appeared on CIPs and then disappeared, and the specific surface area of the particles increased and then decreased. When the concentration of HCl was 0.50 mol/L, the number of nanopores and the specific surface area of particles changed sharply. Not only that, the coated mass of BTES increased greatly and the saturation magnetization of particles decreased sharply. As the coated mass increased, without a magnetic field, the viscosity and shear stress of the MRFs increased, especially when the coated mass was more than 2.45 wt.%; while under a magnetic field, the viscosity and shear stress decreased, and the sedimentation rate of the MRFs decreased from 0.13 to 0.01 mm/h. By controlling the concentration of HCl for etching, the coating effect of CIPs was greatly enhanced, and thus an MRF with superior shear stress and excellent dispersion stability was obtained, which is significant in basic research and MRF-related applications.

Accurate crop disease classification is crucial for ensuring food security and enhancing agricultural productivity. However, the existing crop disease classification algorithms primarily focus on a single image modality and typically require a large number of samples. Our research counters these issues by using pre-trained Vision–Language Models (VLMs), which enhance the multimodal synergy for better crop disease classification than the traditional unimodal approaches. Firstly, we apply the multimodal model Qwen-VL to generate meticulous textual descriptions for representative disease images selected through clustering from the training set, which will serve as prompt text for generating classifier weights. Compared to solely using the language model for prompt text generation, this approach better captures and conveys fine-grained and image-specific information, thereby enhancing the prompt quality. Secondly, we integrate cross-attention and SE (Squeeze-and-Excitation) Attention into the training-free mode VLCD(Vision-Language model for Crop Disease classification) and the training-required mode VLCD-T (VLCD-Training), respectively, for prompt text processing, enhancing the classifier weights by emphasizing the key text features. The experimental outcomes conclusively prove our method’s heightened classification effectiveness in few-shot crop disease scenarios, tackling the data limitations and intricate disease recognition issues. It offers a pragmatic tool for agricultural pathology and reinforces the smart farming surveillance infrastructure.

Direct detection of miRNA is currently limited by the complex amplification and reverse transcription processes of existing methods, leading to low sensitivity and high operational demands. Herein, we developed a CRISPR/Cas13a-mediated photoelectrochemical (PEC) biosensing platform for direct and sensitive detection of miRNA-21. The direct and specific recognition of target miRNA-21 by crRNA-21 eliminates the need for pre-amplification and reverse transcription of miRNA-21, thereby preventing signal distortion and enhancing the sensitivity and precision of target detection. When crRNA-21 binds to miRNA-21, it activates the trans-cleavage activity of CRISPR/Cas13a, leading to the non-specific cleavage of biotin-modified DNA with uracil bases (biotin-rU-DNA). This cleavage prevents the biotin-rU-DNA from being immobilized on the electrode surface. As a result, streptavidin cannot attach to the electrode via specific biotin binding, reducing spatial resistance and causing a positively correlated increase in the photocurrent response. This Cas-PEC biosensor has good analytical capabilities, linear responses between 10 fM and 10 nM, a minimum detection limit of 9 fM, and an excellent recovery rate in the analysis of real human serum samples. This work presented an innovative solution for detecting other biomarkers in bioanalysis and clinical diagnostics.

Enteroviruses such as coxsackievirus B3 are identified as a common cause of viral myocarditis, but the potential mechanism of its replication and pathogenesis are largely unknown. The genomes of a variety of viruses contain N6-methyladenosine (m6A), which plays important roles in virus replication. Here, by using the online bioinformatics tools SRAMP and indirect immunofluorescence assay (IFA), we predict that the CVB3 genome contains m6A sites and found that CVB3 infection could alter the expression and cellular localization of m6A-related proteins. Moreover, we found that 3-deazaadenosine (3-DAA), an m6A modification inhibitor, significantly decreased CVB3 replication. We also observed that the m6A methyltransferases methyltransferase-like protein 3 (METTL3) and METTL14 play positive roles in CVB3 replication, whereas m6A demethylases fat mass and obesity-associated protein (FTO) or AlkB homolog 5 (ALKBH5) have opposite effects. Knockdown of the m6A binding proteins YTH domain family protein 1 (YTHDF1), YTHDF2 and YTHDF3 strikingly decreased CVB3 replication. Finally, the m6A site mutation in the CVB3 genome decreased the replication of CVB3 compared with that in the CVB3 wild-type (WT) strain. Taken together, our results demonstrated that CVB3 could exploit m6A modification to promote viral replication, which provides new insights into the mechanism of the interaction between CVB3 and the host.

Not-from-concentrate (NFC) juice effectively retains the original characteristics of the fruit. Despite the various health benefits of Goji berries, reports on the processing technology and quality changes of NFC goji juice are lacking. We optimised the processing technology of NFC goji juice. Employing fuzzy mathematics evaluation combined with response surface methodology based on single-factor experiments, the physicochemical, nutritional, and microbiological indicators of the juice were determined. Gas chromatography-ion mobility spectroscopy was used to analyse changes in volatile compounds before and after processing. The optimal processing parameters were: times for homogenisation, ultrasonication, and sterilisation of 2 min, 3 min, and 85 s, respectively, and sterilisation temperature of 102 °C. The resulting product had a sensory evaluation score of 85.5 and a rich, pleasant taste. The ratio of total soluble solids to titratable acidity and turbidity were 12.8 and 1420 NTU, respectively. Post-processing, the contents of β-carotene, polysaccharides, and betaine increased by 57.3%, 26.7%, and 31.5%, respectively. Microbiological indicators met Chinese national limits for food pollutants and microorganisms. The total relative content of volatile substances in NFC goji juice decreased by 19.86% after processing. This study provides a theoretical reference for the intensive processing and development of high-value goji berries.

Smoking is prevalent among people living with the human immunodeficiency virus (HIV), and it increases morbidity and mortality in this population. However, due to ethical constraints, there is limited information on the effects of smoking on susceptibility to HIV infection. To investigate whether smoking is associated with an increased susceptibility to HIV infection, we conducted a two-sample Mendelian randomization (MR) study using summary statistics from genome-wide association studies of individuals of European ancestry who have ever smoked (n = 99,996) and have HIV (n = 412,130). The random-effects inverse-variance weighted estimation method was used as the study’s primary approach, with the MR-Egger regression and the weighted-median method as complementary approaches. Using 100 single-nucleotide polymorphisms of genome-wide significance as instrumental variables for smoking, we observed a significant association between smoking and HIV infection (odds ratio 5.790, 95% confidence interval [1.785, 18.787], and p = 0.003). Comparable results were obtained using the weighted-median method. Our findings implied that smoking is probably associated with increased susceptibility to HIV infection. Given the exploratory nature of this study, further research is needed to confirm this relationship.

Deep learning technology can automatically learn features from large amounts of data, with powerful feature extraction and pattern recognition capabilities, thereby improving the accuracy and efficiency of object detection. [The objective of this study]: In order to improve the accuracy and speed of mask wearing deep learning detection models in the post pandemic era, the [Problem this study aimed to resolve] was based on the fact that no research work has been reported on standardized detection models for mask wearing with detecting nose targets specially. [The topic and method of this study]: A mask wearing normalization detection model (towards the wearing style exposing the nose to outside, which is the most obvious characteristic of non-normalized style) based on improved YOLOv5s (You Only Look Once v5s is an object detection network model) was proposed. [The improved method of the proposed model]: The improvement design work of the detection model mainly includes (1) the BottleneckCSP (abbreviation of Bottleneck Cross Stage Partial) module was improved to a BottleneckCSP-MASK (abbreviation of Bottleneck Cross Stage Partial-MASK) module, which was utilized to replace the BottleneckCSP module in the backbone architecture of the original YOLOv5s model, which reduced the weight parameters’ number of the YOLOv5s model while ensuring the feature extraction effect of the bonding fusion module. (2) An SE module was inserted into the proposed improved model, and the bonding fusion layer in the original YOLOv5s model was improved for better extraction of the features of mask and nose targets. [Results and validation]: The experimental results indicated that, towards different people and complex backgrounds, the proposed mask wearing normalization detection model can effectively detect whether people are wearing masks and whether they are wearing masks in a normalized manner. The overall detection accuracy was 99.3% and the average detection speed was 0.014 s/pic. Contrasted with original YOLOv5s, v5m, and v5l models, the detection results for two types of target objects on the test set indicated that the mAP of the improved model increased by 0.5%, 0.49%, and 0.52%, respectively, and the size of the proposed model compressed by 10% compared to original v5s model. The designed model can achieve precise identification for mask wearing behaviors of people, including not wearing a mask, normalized wearing, and wearing a mask non-normalized.

It is important to improve the separation ability of photogenerated electrons and the adsorption capacity of carbon dioxide (CO2) for efficient photoreduction of CO2. Here, we synthesized ZnCdS quantum dots (ZCS-QDs) and cerium dioxide nanosheets (CeO2) using the solvothermal method and calcination method. We combined CeO2 and ZCS-QDs to effectively enhance the charge separation efficiency, and the lifetime of photogenerated electrons was increased 4.5 times. The CO evolution rate of the optimized composite (ZCS-QDs/CeO2) was up to 495.8 μmol g−1 h−1, and it had 100% product selectivity. In addition, the stability remained high after five cycles. The CO2 adsorption capacity of the catalyst surface was observed by in situ FTIR. The test results showed that improving CO2 capture ability and promoting photogenic electron separation had positive effects on enhancing photoreduction of CO2. This study provides a reference for constructing a zero-dimensional–two-dimensional (0D–2D) heterojunction and explores potential CO2 reduction reaction mechanisms.

The shared energy storage device acts as an energy hub between multiple microgrids to better play the complementary characteristics of the microgrid power cycle. In this paper, the cooperative operation process of shared energy storage participating in multiple island microgrid systems is researched, and the two-stage research on multi-microgrid operation mode and shared energy storage optimization service cost is focused on. In the first stage, the output of each subject is determined with the goal of profit optimization and optimal energy storage capacity, and the modified grey wolf algorithm is used to solve the problem. In the second stage, the income distribution problem is transformed into a negotiation bargaining process. The island microgrid and the shared energy storage are the two sides of the game. Combined with the non-cooperative game theory, the alternating direction multiplier method is used to reduce the shared energy storage service cost. The simulation results show that shared energy storage can optimize the allocation of multi-party resources by flexibly adjusting the control mode, improving the efficiency of resource utilization while improving the consumption of renewable energy, meeting the power demand of all parties, and realizing the sharing of energy storage resources. Simulation results show that compared with the traditional PSO algorithm, the iterative times of the GWO algorithm proposed in this paper are reduced by 35.62%, and the calculation time is shortened by 34.34%. Compared with the common GWO algorithm, the number of iterations is reduced by 18.97%, and the calculation time is shortened by 22.31%.

Discarded unripe kiwifruits (DUKs) are regarded as the major agro-byproducts in the production of kiwifruits, which have abundantly valuable secondary metabolites. Nevertheless, owing to the limited knowledge about the differences in phytochemicals and bioactivity between DUKs and mature kiwifruits, the utilization of DUKs in the food industry remains scarce. Hence, to promote their food applications, the phenolic compounds and bioactivity of discarded unripe, mature, and overripe fruits from three red-fleshed kiwifruit cultivars were studied and compared. The results revealed that the levels of total phenolics, total flavonoids, and total procyanidins in kiwifruits varied significantly by maturity stage. In addition, our findings demonstrated that DUKs possessed much higher contents of valuable phenolic compounds (e.g., chlorogenic acid (CHA), neochlorogenic acid (NCHA), gallic acid (GA), protocatechuic acid (PA), procyanidin B1 (ProcB1), procyanidin B2 (ProcB2), procyanidin C1 (ProcC1), quercetin 3-O-glucoside (QueG), and quercetin 3-O-rhamnoside (QueR)) than mature and overripe kiwifruits. Furthermore, DUKs exerted much stronger in vitro antioxidant capacity, inhibitory effects on α-glucosidase, and anti-inflammatory activity than mature and overripe kiwifruits, which were mainly attributed to their higher contents of total polyphenols and individual phenolic components, such as GA, CHA, NCHA, PA, ProcB1, ProcB2, ProcC1, and QueR. Overall, these findings provide sufficient evidence for the development and utilization of DUKs in the food/functional food industry.

Maternal immune activation (MIA) during pregnancy has been increasingly recognized as a critical factor in the development of neurodevelopmental disorders, with potential sex-specific impacts that are not yet fully understood. In this study, we utilized a murine model to explore the behavioral and molecular consequences of MIA induced by lipopolysaccharide (LPS) administration on embryonic day 12.5. Our findings indicate that male offspring exposed to LPS exhibited significant increases in anxiety-like and depression-like behaviors, while female offspring did not show comparable changes. Molecular analyses revealed alterations in pro-inflammatory cytokine levels and synaptic gene expression in male offspring, suggesting that these molecular disruptions may underlie the observed behavioral differences. These results emphasize the importance of considering sex as a biological variable in studies of neurodevelopmental disorders and highlight the need for further molecular investigations to understand the mechanisms driving these sex-specific outcomes. Our study contributes to the growing evidence that prenatal immune challenges play a pivotal role in the etiology of neurodevelopmental disorders and underscores the potential for sex-specific preventative approaches of MIA.

Producing translucent eggs has been found to reduce the quality and safety of the eggs, as well as the demand from consumers. However, the intestinal function and the molecular mechanism for the production of translucent eggs remain uncertain. A total of 120 eggs from 276-day-old Jining Bairi were divided into two groups based on eggshell translucence: the translucent egg group (group T) and the normal group (group C). Group T exhibited thicker eggshells and a lower egg yolk color. Subsequently, we divided the chickens into translucent and normal groups based on their egg quality. We then assessed the plasma biochemical index, intestinal morphology and structure, enzyme activity, and antioxidant capacity of the hens producing translucent eggs compared to those producing normal eggs. The results showed that the ratio of duodenal villus length to crypt depth, succinate dehydrogenase (SDH) activity, chymotrypsin, total ATPase (T-ATPase), alkaline phosphatase (AKP), and glutathione peroxidase (GSH-Px) activities were decreased in the hens produced translucent eggs (p < 0.05), but malondialdehyde (MDA) content was increased (p < 0.05); jejunal lipase activity, Na+K+-ATPase activity, total antioxidant capacity (T-AOC), and GSH-Px activities were decreased (p < 0.05) in group T; ileal amylase and Ca2+Mg2+-ATPase activities were also decreased (p < 0.05) in group T. In addition, we identified a total of 471 differentially expressed genes (DEGs) in duodenal tissue, with 327 up-regulated genes and 144 down-regulated genes (|log2FC| ≥ 1 and p < 0.05). Enrichment analysis showed that the up-regulated genes, such as GSTT1, GSTO2, and GSTA3, were mostly enriched in metabolism of xenobiotics by cytochrome P450, drug metabolism-cytochrome P450, and oxidative phosphorylation pathways. The results of our study indicate that plasma lipid metabolism disorder, decreased intestinal antioxidant capacity, and altered intestinal metabolism capabilities may influence the formation of translucent eggs.

As one of the most important future clean energy sources, natural gas hydrate (NGH) is attracting widespread attention due to the vast reserves available and high energy density. How to extract this source in a safe, efficient, and environmentally friendly manner has become the key to the commercial utilization of its resources. This paper reviews the recent advances in the study of the fundamental reservoir properties of offshore NGH, summarizing the methods and technologies for testing the sedimentary properties of reservoirs, analyzing the characteristics in reservoir mechanics, electrics, thermodynamics, and fluid dynamics, and discusses the influence of reservoir fundamental properties on NGH exploitation. The aim is to provide guidance and reference for research on the exploitation of NGH in different target exploitation areas offshore.

5-Hydroxytryptamine (5-HT) is an inhibitory neurotransmitter widely distributed in mammalian tissues, exerting its effects through binding to various receptors. It plays a crucial role in the proliferation of granulosa cells (GCs) and the development of follicles in female animals, however, its effect on porcine follicle development is not clear. The aim of this study is to investigate the expression of 5-HT and its receptors in various parts of the pig ovary, as well as the effect of 5-HT on porcine follicular development by using ELISA, quantitative real-time PCR (qPCR) and EdU assays. Firstly, we examined the levels of 5-HT and its receptors in porcine ovaries, follicles, and GCs. The findings revealed that the expression of different 5-HT receptors varied among follicles of different sizes. To investigate the relationship between 5-HT and its receptors, we exposed the GCs to 5-HT and found a decrease in 5-HT receptor expression compared to the control group. Subsequently, the treatment of GCs with 0.5 μM, 5 μM, and 50 μM 5-HT showed an increase in the expression of cell cycle-related genes, and EdU results indicated cell proliferation after the 0.5 μM 5-HT treatment. Additionally, the expression of genes involved in E2 synthesis was examined after the treatment of granulosa cells with 0.5 μM 5-HT. The results showed that CYP19A1 and HSP17β1 expression was decreased. These results suggest that 5-HT might affect the development of porcine follicle by promoting the proliferation of GCs and inhibiting the synthesis of estrogen. This provides a new finding for exploring the effect of 5-HT on follicular development, and lays a foundation for further research on the mechanism of 5-HT in follicles.

A narrow genetic basis limits further the improvement of modern Gossypium hirsutum cultivar. The abundant genetic diversity of wild species provides available resources to solve this dilemma. In the present study, a chromosome segment substitution line (CSSL) population including 553 individuals was established using G. darwinii accession 5-7 as the donor parent and G. hirsutum cultivar CCRI35 as the recipient parent. After constructing a high-density genetic map with the BC1 population, the genotype and phenotype of the CSSL population were investigated. A total of 235 QTLs, including 104 QTLs for fiber-related traits and 132 QTLs for seed-related traits, were identified from four environments. Among these QTLs, twenty-seven QTLs were identified in two or more environments, and twenty-five QTL clusters consisted of 114 QTLs. Moreover, we identified three candidate genes for three stable QTLs, including GH_A01G1096 (ARF5) and GH_A10G0141 (PDF2) for lint percentage, and GH_D01G0047 (KCS4) for seed index or oil content. These results pave way for understanding the molecular regulatory mechanism of fiber and seed development and would provide valuable information for marker-assisted genetic improvement in cotton.

Turbidity, as a key indicator of water quality linked to underwater light attenuation, is crucial for evaluating water quality. Control in high-turbidity water environments plays a critical role in navigable rivers. For this purpose, our study proposed a framework for analyzing the spatio-temporal variation of turbidity and its driving factors in a navigable and turbid river using in situ measurement data, satellite data, socioeconomic data, a power index function model, and correlation analysis. The results show that the proposed model is feasible for quantitative turbidity monitoring of the Xitiaoxi River. Its upstream turbidity is lower than downstream, with seasonal averages for spring, summer, autumn, and winter of 93.9, 111.3, 113.5, and 120.9 NTU, respectively. Furthermore, the turbidity in the middle and lower reaches of the Xitiaoxi River continuously increased before 2005 and began to decline after 2005 due to the policy of mining moratorium. This trend is especially noticeable at monitoring points along the main stream of the Xitiaoxi River, such as downstream of the Xitiaoxi River (S1), Gangkou station (S2), middle reaches of the Xitiaoxi River (S4), Hengtangcun station (S6), upper stream of the Xitiaoxi River (S7), and Huxi River (S8). Mining and shipping have significantly contributed to the turbidity of the target river. This framework offers a practical approach for assessing the environmental impacts of both natural and anthropogenic factors, thereby providing valuable insights for river management practices.

Wheat dwarf virus (WDV, genus Mastrevirus, family Geminiviridae) is one of the causal agents of wheat viral disease, which severely impacts wheat production in most wheat-growing regions in the world. Currently, there is little information about natural resistance against WDV in common wheat germplasms. CRISPR/Cas9 technology is being utilized to manufacture transgenic plants resistant to different diseases. In the present study, we used the CRISPR/Cas9 system targeting overlapping regions of coat protein (CP) and movement protein (MP) (referred to as CP/MP) or large intergenic region (LIR) in the wheat variety ‘Fielder’ to develop resistance against WDV. WDV-inoculated T1 progenies expressing Cas9 and sgRNA for CP/MP and LIR showed complete resistance against WDV and no accumulation of viral DNA compared with control plants. Mutation analysis revealed that the CP/MP and LIR targeting sites have small indels in the corresponding Cas9-positive plants. Additionally, virus inhibition and indel mutations occurred in T2 homozygous lines. Together, our work gives efficient results of the engineering of CRISPR/Cas9-mediated WDV resistance in common wheat plants, and the specific sgRNAs identified in this study can be extended to utilize the CRISPR/Cas9 system to confer resistance to WDV in other cereal crops such as barley, oats, and rye.

Aiming to accurately identify apple targets and achieve segmentation and the extraction of branch and trunk areas of apple trees, providing visual guidance for a picking robot to actively adjust its posture to avoid branch trunks for obstacle avoidance fruit picking, the spindle-shaped fruit trees, which are widely planted in standard modern apple orchards, were focused on, and an algorithm for apple tree fruit detection and branch segmentation for picking robots was proposed based on an improved YOLOv8s model design. Firstly, image data of spindle-shaped fruit trees in modern apple orchards were collected, and annotations of object detection and pixel-level segmentation were conducted on the data. Training set data were then augmented to improve the generalization performance of the apple detection and branch segmentation algorithm. Secondly, the original YOLOv8s network architecture’s design was improved by embedding the SE module visual attention mechanism after the C2f module of the YOLOv8s Backbone network architecture. Finally, the dynamic snake convolution module was embedded into the Neck structure of the YOLOv8s network architecture to better extract feature information of different apple targets and tree branches. The experimental results showed that the proposed improved algorithm can effectively recognize apple targets in images and segment tree branches and trunks. For apple recognition, the precision was 99.6%, the recall was 96.8%, and the mAP value was 98.3%. The mAP value for branch and trunk segmentation was 81.6%. The proposed improved YOLOv8s algorithm design was compared with the original YOLOv8s, YOLOv8n, and YOLOv5s algorithms for the recognition of apple targets and segmentation of tree branches and trunks on test set images. The experimental results showed that compared with the other three algorithms, the proposed algorithm increased the mAP for apple recognition by 1.5%, 2.3%, and 6%, respectively. The mAP for tree branch and trunk segmentation was increased by 3.7%, 15.4%, and 24.4%, respectively. The proposed detection and segmentation algorithm for apple tree fruits, branches, and trunks is of great significance for ensuring the success rate of robot harvesting, which can provide technical support for the development of an intelligent apple harvesting robot.
