Plesiomonas shigelloides, a Gram-negative bacillus, is the only member of the Enterobacteriaceae family able to produce polar and lateral flagella and cause gastrointestinal and extraintestinal illnesses in humans. The flagellar transcriptional hierarchy of P. shigelloides is currently unknown. In this study, we identified FlaK, FlaM, FliA, and FliAL as the four regulators responsible for polar and lateral flagellar regulation in P. shigelloides. To determine the flagellar transcription hierarchy of P. shigelloides, the transcriptomes of the WT and ΔflaK, ΔflaM, ΔfliA, and ΔfliAL were carried out for comparison in this study. Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) and luminescence screening assays were used to validate the RNA-seq results, and the Electrophoretic Mobility Shift Assay (EMSA) results revealed that FlaK can directly bind to the promoters of fliK, fliE, flhA, and cheY, while the FlaM protein can bind directly to the promoters of flgO, flgT, and flgA. Meanwhile, we also observed type VI secretion system (T6SS) and type II secretion system 2 (T2SS-2) genes downregulated in the transcriptome profiles, and the killing assay revealed lower killing abilities for ΔflaK, ΔflaM, ΔfliA, and ΔfliAL compared to the WT, indicating that there was a cross-talk between the flagellar hierarchy system and bacterial secretion system. Invasion assays also showed that ΔflaK, ΔflaM, ΔfliA, and ΔfliAL were less effective in infecting Caco-2 cells than the WT. Additionally, we also found that the loss of flagellar regulators causes the differential expression of some of the physiological metabolic genes of P. shigelloides. Overall, this study aims to reveal the transcriptional hierarchy that controls flagellar gene expression in P. shigelloides, as well as the cross-talk between motility, virulence, and physiological and metabolic activity, laying the groundwork for future research into P. shigelloides’ coordinated survival in the natural environment and the mechanisms that infect the host.

China has been facing a contradiction between its water supply and demand for a long time, and it is urgent that it builds a “water-saving society”. Previous research on water pricing mainly focused on the policy level, only considering the supply and demand sides and ignoring the participation of local governments. Due to the inconsistent approval mechanisms, different times of cost formation, and lack of a scientific pricing relationship between multiple water sources and multiple users, with the current water pricing in China, it is difficult to solve the complex problem of water resource allocation. This study aimed to promote the qualitative supply of water and the reuse of reclaimed water, achieve a high-quality and low-cost water supply, and explore the role of water-pricing mechanisms in the regulation of multi-source water supply configurations. The mechanism of regulation is constructed for the complex, multi-source, multi-user water resource system, and a three-party game model is established among local governments, water supply enterprises, and water users. A reasonable pricing system with a rational price relationship is obtained, and Pinghu City in Zhejiang Province is taken as the research area. The prices of the external water supply to residential users, non-resident users, and special users are 4.48 CNY/m3, 6.28 CNY/m3, and 7.12 CNY/m3. The external supply prices for resident users, non-resident users, and special users are 3.81 CNY/m3, 5.25 CNY/m3, and 6.05 CNY/m3. The external supply price for non-resident users is 2.62 CNY/m3 for reclaimed water. The results of the study showed that when the government’s contribution did not exceed the amount that it would have been without the inclusion of recycled water and the annual payment of water users did not exceed their ability to pay, the income of water suppliers increased by 69%. The three balanced objectives of rational water resource configuration by the local government, reasonable profit for the water supply enterprise, and reasonable payment by water users have been achieved. This study provides theoretical and methodological support for the implementation of water pricing for multi-source and multi-user water supplies in water-scarce areas.

The charging behavior of electric vehicle users is highly stochastic, which makes the short-term prediction of charging load at electric vehicle charging stations difficult. In this paper, a data-driven hybrid model optimized by the improved dung beetle optimization algorithm (IDBO) is proposed to address the problem of the low accuracy of short-term prediction. Firstly, the charging station data are preprocessed to obtain clear and organized load data, and the input feature matrix is constructed using factors such as temperature, date type, and holidays. Secondly, the optimal CNN-BiLSTM model is constructed using convolutional neural network (CNN) and Bi-directional Long Short-Term Memory (BiLSTM), which realizes the feature extraction of the input matrix and better captures the hidden patterns and regularities in it. Then, methods such as Bernoulli mapping are used to improve the DBO algorithm and its hyperparameters; for example, hidden neurons of the hybrid model are tuned to further improve the model prediction accuracy. Finally, a simulation experiment platform is established based on MATLAB R2023a to validate the example calculations on the historical data of EV charging stations in the public dataset of ANN-DATA, and comparative analyses are carried out. The results show that compared with the traditional models such as CNN, BiLSTM and PSO-CNN-BiLSTM, the coefficient of determination of the model exceeds 0.8921 and the root mean square error is maintained at about 4.413 on both the training and test sets, which proves its effectiveness and stability.

In this study, we develop an Atmospheric Motion Vector (AMV)-based method for retrieving wind vectors using 183.31 GHz water-vapor absorption channels. The method involves tracking water-vapor features from image triplets and subsequently deriving wind fields from motion vectors. The height of the derived wind for each channel is determined by calculating the weighing function peak using monthly averaged ERA5 reanalysis data. By utilizing Microwave Humidity Sounder-II (MWHS-II) brightness temperatures from the five channels centered around 183.31 GHz, wind vectors are retrieved within the Arctic region for the entire year of 2022. The retrieval quality is evaluated through comparative analysis with ERA5 reanalysis data and the Visible Infrared Imaging Radiometer Suite (VIIRS) wind product. The resultant vector root mean square errors (RMSEs) are approximately 4.5 m/s for the three lower-height channels and 5.5 m/s for the two upper-height channels. These findings demonstrate a wind retrieval performance comparable to the existing methods, highlighting its potential for augmenting wind availability at lower height levels.

Background: As one of the important components of immunotherapies, mRNA vaccines have displayed promising clinical outcomes in solid tumors. Nonetheless, their efficacy remains unclear in pancreatic adenocarcinoma (PAAD). Given the interaction of pyroptosis with anticancer immunity, our study aims to identify pyroptosis-related antigens for mRNA vaccine development and discern eligible candidates for vaccination. Methods: Utilizing gene expression data from TCGA and ICGC, we integrated RNA-seq data and compared genetic alterations through cBioPortal. Differential gene expressions were integrated using GEPIA. Relationships between immune cell abundance and tumor antigens were analyzed and visualized via TIMER. WGCNA facilitated the clustering of pyroptosis-related genes, identification of hub genes, and pathway enrichment analyses. Pyroptosis landscape was depicted through graph learning-based dimensional reduction. Results: Four overexpressed and mutant pyroptosis-related genes associated with poor prognosis were identified as potential antigens for mRNA vaccines in PAAD, including ANO6, PAK2, CHMP2B, and RAB5A. These genes displayed positive associations with antigen-presenting cells. PAAD patients were stratified into three pyroptosis subtypes. Notably, the PS3 subtype, characterized by a lower mutation count and TMB, exhibited “cold” immunological traits and superior survival compared to other subtypes. The pyroptosis landscape exhibited considerable heterogeneity among individuals. Furthermore, the turquoise module emerged as an independent prognostic indicator and patients with high expressions of hub genes might not be suitable candidates for mRNA vaccination. Conclusions: In PAAD, ANO6, PAK2, CHMP2B, and RAB5A are prospective pyroptosis-related antigens for mRNA vaccine development, which holds potential benefits for patients classified as PS3 and those with diminished hub gene expressions, providing insights into personalized mRNA vaccine strategies.

At present, the new energy industry represented by photovoltaics has become the main force to realize the optimization of China’s energy structure and the goal of “double carbon”; with the absence of land resources, the waterborne bifacial photovoltaic has ushered in a new opportunity. Therefore, in order to address the problem that the maximum power point tracking (MPPT) of photovoltaics (PV) could not take into account, the dynamic economic performance in the control process, an economic model predictive control (EMPC), is proposed in this work to realize the MPPT of the waterborne bifacial PV power generation system. Firstly, the model of the bifacial PV module is constructed by combining the ray-tracing irradiance model and considering the effect of water surface albedo on the irradiance absorbed by the module. Secondly, the EMPC controller is designed based on the state-space model of the system to maximize the power generation as the economic performance index, and to solve the optimal input variables time by time to achieve a rolling optimization with the operational requirements of the system itself as the constraints. Thirdly, the MATLAB/Simulink (R2022a) simulation experimental results verify that the EMPC strategy could be utilized to achieve MPPT of the waterborne bifacial PV power generation system, according to the changes of environment. Finally, it is also demonstrated that the bifacial PV power generation system that employed the EMPC strategy outperformed the traditional MPPT algorithm, with respect to both output power tracking velocity and accuracy, and the power generation could be improved by about 6% to 14.5%, which significantly enhances the system’s dynamic process economics.

Conventional maps of soil parent material (SPM) types obtained by field survey and manual mapping or predictions from other map data have limited accuracy. Digital soil mapping of SPM types necessitates accurate acquisition of SPM distribution information, which is still a challenge in hilly areas. This study developed a high-accuracy method for SPM identification in hilly areas at the county scale. Based on geographic information system technology, seven feature variables were extracted from the geological map, geomorphic map, digital elevation model, and remote sensing image data of Shanggao County, Jiangxi Province, China. Different feature combination schemes were designed to develop SPM identification models based on random forest (RF), support vector machine (SVM), and maximum likelihood classification (MLC) algorithms. The best SPM identification results were obtained from the RF algorithm using the combination of geological type, geomorphic type, elevation, and slope. Confusion matrices were constructed based on a field survey of 586 validation samples, and the results were evaluated in terms of overall accuracy, precision, recall, F1 score, and Kappa coefficient. The overall accuracy and Kappa coefficient of the results from the optimal RF model were 83.11% and 0.79, respectively, which were 26.11% and 0.31 higher than those of the conventional map, respectively. Its precision and recall for various SPM types were greater than 75%. A comprehensive comparison of the accuracy, uncertainty, and plotting performance of the SPM recognition results reveals that the RF algorithm outperforms the SVM algorithm and the MLC algorithm. Geological type was the largest contributor to SPM identification, followed by geomorphic type, elevation, and slope. The importance of different feature variables varied for distinct SPM types. The accuracy of SPM identification was not improved by selecting more feature variables, such as land use type, normalised difference vegetation index, and topographic wetness index. This study demonstrates the feasibility of high-accuracy county-level SPM mapping in hilly areas based on the RF algorithm using geological type, geomorphic type, elevation, and slope as feature variables. As hilly areas have typical topographic features and SPM types, the proposed method of SPM mapping can be useful for application in other similar areas. There are a few limitations in this study with regard to data quality and resolution, feature variable selection, classification algorithm generalisation, and study area representativeness, which may affect the outcomes and need to be solved.

The road transportation of explosives is highly concerning due to its substantial impact on social safety. For the safety management of explosive transportation, e.g., transport route planning and emergency rescue, explosion consequence evaluation is of paramount importance. The consequence evaluation of explosion accidents is affected by many factors, especially spatial features, such as the location of transport vehicles, the distribution of buildings, and the presence of individuals around the road, etc. However, there is still a lack of quantification methods for building damage evaluation, human casualty evaluation that considers real-time population density, and efficient interactive damage evaluation methods. In this paper, we formalize three typical scenarios of damage evaluation for explosive road transportation accidents, i.e., explosion point-based, road segment-based, and route-based damage evaluation. For each scenario, we propose a Height-aware Hierarchical Building Damage (HHBD) model and a Shelter-aware Human Casualty (SHC) model for building damage evaluation and human casualty evaluation, respectively. We also develop a GIS-based interactive visualization platform that integrates multi-source geospatial data and that enables efficient geospatial computation. In addition, a case study of liquefied natural gas (LNG) transportation in Wuhan is demonstrated in order to verify the effectiveness and efficiency of the proposed system. The research results can support the decision-making process of explosive transportation safety warnings and emergency rescue.

The O-antigen is one of the outermost surface components of Gram-negative bacteria. Its large structural variation provides the molecular basis for bacterial serological diversity. Here, we established the structure of the O-antigen from an Escherichia coli strain, SD2019180, which appeared to be completely different from the known E. coli serogroups. The O-antigen tetrasaccharide biological repeating unit was identified as → 2)-[β-d-GlcpA-(1 → 4)]-[α-d-Galp-(1 → 3)]-α-l-Fucp-(1 → 3)-α-d-GlcpNAc-(1 →. Furthermore, we analyzed the O-antigen gene cluster of SD2019180 and confirmed its role in O-antigen synthesis by using deletion and complementation experiments. Our findings indicate that SD2019180 is a novel serogroup of Escherichia coli.

Prolonged exposure to high concentrations of suspended particulate matter (SPM), especially aerodynamic fine particulate matter that is ≤2.5 μm in diameter (PM2.5), can cause serious harm to human health and life via the induction of respiratory diseases and lung cancer. Therefore, accurate prediction of PM2.5 concentrations is important for human health management and governmental environmental management decisions. However, the time-series processing of PM2.5 concentration based only on a single region and a special time period is less explanatory, and thus, the spatial-temporal applicability of the model is more restricted. To address this problem, this paper constructs a PM2.5 concentration prediction optimization model based on Convolutional Neural Networks-Long Short-Term Memory (CNN-LSTM). Hourly data of atmospheric pollutants, meteorological parameters, and Precipitable Water Vapor (PWV) of 10 cities in the Beijing-Tianjin-Hebei metropolitan area during the period of 1–30 September 2021/2022 were used as the training set, and the PM2.5 data of 1–7 October 2021/2022 were used for validation. The experimental results show that the CNN-LSTM model optimizes the average root mean square error (RMSE) by 25.52% and 14.30%, the average mean absolute error (MAE) by 26.23% and 15.01%, and the average mean absolute percentage error (MAPE) by 35.64% and 16.98%, as compared to the widely used Back Propagation Neural Network (BPNN) and Long Short-Term Memory (LSTM) models. In summary, the CNN-LSTM model is superior in terms of applicability and has the highest prediction accuracy in the Beijing-Tianjin-Hebei metropolitan area. The results of this study can provide a reference for the relevant departments in the Beijing-Tianjin-Hebei metropolitan area to predict PM2.5 concentration and its trend in specific time periods.

Ten Candida species strains were isolated from the first known fatal case of rhinofacial and rhino–orbital–cerebral candidiasis. Among them, five strains of Candida parapsilosis complex were isolated during the early stage of hospitalization, while five strains of Candida tropicalis were isolated in the later stages of the disease. Using whole-genome sequencing, we distinguished the five strains of C. parapsilosis complex as four Candida metapsilosis strains and one Candida parapsilosis strain. Antifungal susceptibility testing showed that the five strains of C. parapsilosis complex were susceptible to all antifungal drugs, while five C. tropicalis strains had high minimum inhibitory concentrations to azoles, whereas antifungal-drug resistance gene analysis revealed the causes of azole resistance in such strains. For the first time, we analyzed the microevolutionary characteristics of pathogenic fungi in human hosts and inferred the infection time and parallel evolution of C. tropicalis strains. Molecular clock analysis revealed that azole-resistant C. tropicalis infection occurred during the first round of therapy, followed by divergence via parallel evolution in vivo. The presence/absence variations indicated a potential decrease in the virulence of genomes in strains isolated following antifungal drug treatment, despite the absence of observed clinical improvement in the conditions of the patient. These results suggest that genomic analysis could serve as an auxiliary tool in guiding clinical diagnosis and treatment.

Under the influence of global climate change and urbanization processes, the number of available water resources (AWRs) in basins has become significantly more uncertain, which has restricted the sustainable development of basins. Therefore, it is important for us to understand the relationship between land use (LU) patterns and climate change on AWRs in a basin for sustainable development. To this end, the vector autoregressive (VAR) method was adopted to construct a quantitative model for AWRs in the basin in this study. Taking the Yiluo River Basin (YRB) as an example, the dynamic relationship between the five elements of agricultural land (AD), woodland (WD), grassland (GD), construction land (CD), and annual precipitation (PREP) and AWRs in the basin was studied. The results show the following: (1) The constructed VAR model was stable, indicating that the use of the proposed VAR model to characterize the degree of the effect of LU pattern and PREP on AWRs in the YRB was reasonable and effective. (2) AWRs in the YRB showed a downward trend, and their responses to the change in LU and PREP were delayed. The changes in the AWRs in the YRB tended to occur the year after changes to the LU pattern and PREP occurred. (3) In the long run, the degree of the contribution of each influencing factor to changes to AWRs was 23.76% (AD), 6.09% (PREP), 4.56% (CD), 4.40% (WD), and 4.34% (GD), which meant that the impact of the LU pattern was more than 90%. This study provides new ideas for similar research, water resource allocation, and LU planning in other river basins from a macroscopic perspective.

The trade-offs and synergies reveal the profit and loss relationship between ecosystem services, which is of great significance to the sustainable development of natural resources. The ecosystem services in Jiangxi Province, such as net primary productivity (NPP), soil conservation (SC) and water yield (WY) during 2000–2020, were estimated in this study. The correlation coefficient was adopted to analyze the trade-offs and synergies between the three ecosystem services by static space correlation and dynamic space correlation from such perspectives as Watershed, county and grid. Moreover, the influence of the three ecosystem services and the relations between them were explored from four aspects: landform, NDVI, accumulated temperature and precipitation. The results showed that the ecological environment quality in Jiangxi Province was improved and that the distribution of ecosystem services had significant regional characteristics. In the static analysis, ecosystem services at all scales were remarkably synergistic, and synergies weakened rapidly and even turned into trade-offs as the scale decreased. In the dynamic analysis, ecosystem services at all scales were mainly synergistic; the proportion of significant samples was much lower than that in the static analysis, the degree of trade-offs/synergies decreased with the decrease in scale, and the decrease was smaller than that in the static analysis. The major constraints for SC were landform and NDVI. The main constraint for WY was precipitation, and that for NPP was NDVI. Affected by various factors, NPP and SC were stably synergistic, NPP and WY were in a stable trade-off relationship, and the relationship between SC and WY was unstable. The trade-offs and synergies changed with factors and zoning.

As natural selenium (Se)-rich soil in China is generally characterized by a high geological background of cadmium (Cd), the safe utilization of such seleniferous soil remains a challenge. The accumulating evidence shows that the threshold value of the Se:Cd ratio is a determinant of regulating Cd accumulation in plants. However, the factors modulating the soil’s Se:Cd ratio in selenium-enriched regions are not well understood. Here, a comprehensive study aimed at quantitatively analyzing the effects of land-use types, parent-material types, and soil properties on the distribution and influencing factors of Se, Cd, and the Se:Cd ratios. According to land use and parent-material types, 77 soil samples were collected in Yuanzhou District, a typical naturally seleniferous area in the subtropical hilly area. The results suggested that, compared with quaternary red clays (qrc), the Se content of soils derived from river and lake sediments (rls) and weathered acidic crystalline rocks (wacr) decreased by 5.81%–19.75%, while the weathered quartzite (wq)-derived soils was increased significantly. The soil Cd content in an orchard was significantly reduced compared with that in a paddy field. A redundancy analysis (RDA) revealed that SOM, Total K, and Total P significantly affected the changes in Se and Cd contents. In addition, the land-use type had the most significant effect on the Se:Cd ratio, with a regression coefficient of −0.6999 analyzed by the binary logistic regression model (p < 0.05). Furthermore, pH and Total K were the critical soil properties in controlling the Se:Cd ratio. The study indicated that the Se:Cd ratio in natural selenium-rich soil was mainly regulated by land-use types. Therefore, it is a feasible measure to regulate the Se:Cd ratio by using agronomic practices, mainly regulating soil pH, for the safe utilization of selenium-rich soil with a high Cd background.

Robust soil organic matter (SOM) mapping is required by farms, but their generation requires a large number of samples to be chemically analyzed, which is cost prohibitive. Recently, research has shown that visible and near-infrared (vis-NIR) reflectance spectroscopy is a fast and accurate technique for estimating SOM in a cost-effective manner. However, few studies have focused on using vis-NIR spectroscopy as a covariate to improve the accuracy of spatial modeling. In this study, our objective was to compare the mapping accuracy from a spatial model using kriging methods with and without the covariate of vis-NIR spectroscopy. We split the 261 samples into a calibration set (104) for building the spectral predictive model, a test set for generating the vis-NIR augmented set from the prediction of the fitted spectral predictive model (131), and a validation set (26) for evaluating map accuracy. We used two datasets (235 samples) for Kriging: a laboratory-based dataset (Ld, observations from calibration and test datasets) and a laboratory-based dataset with vis-NIR augmented predictions (Au.p, observations from calibration and predictions from test dataset), a laboratory-based dataset with vis-NIR spectra as the covariance (Ld.co) and augmented dataset with predictions using vis-NIR with vis-NIR spectra for the covariance (Au.p.co). The first one to seven accumulated principal components of vis-NIR spectra were used as the covariates when we used the measurement of Ld.co and Au.p.co. The map accuracy was evaluated by the validation set for the four datasets using Kriging. The results indicated that adding vis-NIR spectra as covariates had great potential in improving the map accuracy using kriging, and much higher accuracies were observed for Ld.p.co (RMSE of 5.51 g kg−1) and Au.p.co (RMSE of 5.66 g kg−1) than without using vis-NIR spectra as covariates for Ld (RMSE of 7.12 g kg−1) and Au.p (RMSE of 7.69 g kg−1). With a similar model performance to Ld.p.co, Au.p.co can reduce the cost of laboratory analysis for 60% of soil samples, demonstrating its advantage in cost-efficiency for spatial modeling of soil information. Therefore, we conclude that vis-NIR spectra can be used as a cost-effective technique to obtain augmented data to improve fine-resolution spatial mapping of soil information.

The rapid quantitative assessment of soil organic carbon (SOC) is essential for understanding SOC dynamics and developing management strategies in forest ecosystems. Compared with traditional laboratory methods, visible and near-infrared spectroscopy is an efficient and inexpensive technique widely used to predict SOC content. Herein, we compared three different spiking strategies. That is, a large-scale global soil spectral library (global-SSL; 3122 samples) was used as the basis for predicting SOC content in a small-scale local soil spectral library (local-SSL; 89 samples) in Wugong Mountain, Jiangxi Province, China. Partial least squares regression models using global-SSL ‘spiking’ with local samples did not necessarily achieve more accurate predictions than models using local-SSL. Using the developed strategy, a calibration set can be established by selecting the top N spectral samples from global-SSL with high similarity to each local sample, together with the ‘spiking’ set from local-SSL. It is possible to individually improve the prediction results based on local samples (R2 = 0.90, RMSE = 7.19, RPD = 3.38) and still allow for quantitative prediction from fewer local calibration samples (R2 = 0.83, RMSE = 8.71, RPD = 2.68). The developed method is cost-effective and accurate for local-scale SOC assessment in target forest areas using a large soil spectral library.

In order to improve the mechanical properties, nutritional value and fresh-keeping ability of conventional sodium alginate edible composite membranes, a new type of edible composite film was prepared by adding water-blocking agent carnauba wax, plasticizer glycerin, antioxidant and nutritional enhancer sodium ascorbate on a basis of traditional sodium alginate composite film. In this study, the physical, mechanical and structural properties of different film components were investigated. The results showed the components did not simply combine, but produced interaction forces which improved the stability and mechanical properties of composite film. When the amount of calcium ascorbate was 0.4%, the water vapor transmittance of the composite film reached a minimum of 0.65 g·mm/(cm2·d·kPa), and the tensile strength and elongation at break reached the maximum, which were 398.64 MPa and 17.93%, respectively. Additionally, the sodium alginate-carnauba wax film exhibited better performance on the preservation of fresh-cut apples. Compared with other composite films, the color and hardness of fresh-cut apples coated with this composite film were better maintained, and the losses of titration acid content and soluble solid content were reduced. Moreover, the weight loss rate, increase in polyphenol oxidase activity and total colony count were inhibited. All results determined that the edible film has good application value in the field of fresh-cut fruit preservation, which provides a theoretical basis for further research on edible film.

Liposarcoma (LPS) is a rare and heterogeneous malignancy of adipocytic origin. Well-differentiated liposarcoma (WDLPS) and dedifferentiated liposarcoma (DDLPS) are two of the most common subtypes, showing similar genetic characterizations but distinct biological behaviors and clinical prognosis. Compared to WDLPS, DDLPS is more aggressive and has the potential of metastasis, as the malignant adipocytic tumor’s metabolic changes may have taken place during the tumorigenesis of LPSs. Therefore, to investigate the lipid alterations between the two subtypes, high-resolution liquid chromatography tandem mass spectrometry (LC-MS/MS) based untargeted lipidomic analysis was performed onto LPS tissues from 6 WDLPS and 7 DDLPS patients. The lipidomic analysis showed the upregulated phosphatidylcholines and phosphoethanolamines in DDLPS, and the upregulated triglycerides and diglycerides in WDLPS, which might be due to the uncompleted adipocytic dedifferentiation leading to such tumorigenesis. Such a finding was also confirmed by the similarity comparison of two LPS subtypes to the transcriptome of stromal vascular fraction at different differentiation stages. Transcriptomic analysis also demonstrated that metabolic pathways including the pentose phosphate pathway (PPP) were upregulated in WDLPS compared to DDLPS. Therefore, the cell line LPS853 was treated with the PPP inhibitor 6-aminonicotinamide ex vivo and the proliferation and invasion of LPS853 was significantly promoted by PPP inhibition, suggesting the potential role of PPP in the development and differentiation of LPS. In conclusion, this study described the altered lipid profiles of WDLPS and DDLPS for the first time, revealing the different differentiation stages of the two subtypes and providing a potential metabolic target for LPS treatment.

Objective: To evaluate the value of dynamic contrast-enhanced ultrasound (DCE-US) and quantitative parameters in early prediction of tumor response to chemoradiotherapy (CRT) in patients with locally advanced pancreatic ductal adenocarcinoma (LAPC). Patients and Methods: In this prospective study, patients with biopsy-proved and histopathologically proved LAPC who underwent regular CRT were recruited. DCE-US evaluations were performed before and four months after CRT. SonoVue-enhanced contrast-enhanced ultrasound (CEUS) was performed by an ultrasound system (ACUSON Sequoia; Siemens Medical Solutions, USA) equipped with a 5C1 MHz convex array transducer. Time–intensity curves were created by VueBox software (Bracco, Italy), and various DCE-US quantitative parameters were obtained. Taking Response Evaluation Criteria in Solid Tumors (RECIST) based on computed tomography (CT) or magnetic resonance imaging (MRI) as the gold standard, DCE-US parameters were compared between the treatment responder group (RG) and non-responder group (NRG). The correlation between the DCE-US parameters and the serum carbohydrate antigen 19-9 (CA 19-9) level was also analyzed. Results: Finally, 21 LAPC patients (mean age 59.3 ± 7.2 years) were included. In comparing the RG (n = 18) and NRG (n = 3), no significant change could be found among the mean size of the lesions (31.2 ± 8.1 mm vs. 27.2 ± 8.3 mm, p = 0.135). In comparing the TICs between the two groups, the LAPC lesions in the RG took a longer time to reach peak enhancement and to wash out. Among all the DCE-US parameters, RT (rise time), WiAUC (wash-in area under the curve), WoAUC (wash-out area under the curve) and WiWoAUC (wash-in and wash-out area under the curve) decreased significantly after CRT in the RG (p < 0.05). The RT ratio, WiAUC ratio, WoAUC ratio and WiWoAUC ratio were closely correlated with the change in serum CA 19-9 level in the RG (p < 0.05). Conclusion: DCE-US might be a potential imaging method for non-invasive follow-up for early response in LAPC patients treated by CRT.

Previous studies have demonstrated that the effects of monensin on methanogenesis and ruminal fermentation in ruminants were time-dependent. To elucidate the underlying mechanism, we investigated the ruminal bacterial community successions during the adaptation to monensin supplementation and subsequent withdrawal in goats. The experiment included a baseline period of 20 days followed by a treatment period of 55 days with 32 mg monensin/d and a washout period of 15 days. Monensin supplementation reduced the α diversity and changed the structure of ruminal microflora. The α diversity was gradually restored during adaption, but the structure was still reshaped. The temporal dynamics of 261 treatment- and/or time-associated ruminal bacteria displayed six patterns, with two as monensin-sensitive and four as monensin-resistant. The monensin sensitivity and resistance of microbes do not follow a clear dichotomy between Gram-positive and Gram-negative cell types. Moreover, the temporal dynamic patterns of different bacterial species within the same genus or family also displayed variation. Of note, the relative abundance of the total ruminal cellulolytic bacteria gradually increased following monensin treatment, and that of the total amylolytic bacteria were increased by monensin, independent of the duration. In conclusion, under the pressure of monensin, the ruminal ecosystem was reshaped through a series of succession processes, and the carbohydrate-degrading bacteria presented a higher level of adaptability.

Rumen microorganisms can be used in in vitro anaerobic fermentation to encourage the sustainable exploitation of agricultural wastes. However, the understanding of active microbiota under in vitro ruminal fermentation conditions is still insufficient. To investigate how rumen microbes actively participate in the fermentation process in vitro, we resolved the metaproteome generated from ruminal fermentation broth after seven days of in vitro incubation. Herein, the sample-specific database for metaproteomic analysis was constructed according to the metagenomic data of in vitro ruminal fermentation. Based on the sample-specific database, we found in the metaproteome that Bacteroidetes and Firmicutes_A were the most active in protein expression, and over 50% of these proteins were assigned to gene categories involved in energy conversion and basic structures. On the other hand, a variety of bacteria-derived extracellular proteins, which contained carbohydrate-active enzyme domains, were found in the extracellular proteome of fermentation broth. Additionally, the bacterial intracellular/surface moonlighting proteins (ISMPs) and proteins of outer membrane vesicles were detected in the extracellular proteome, and these ISMPs were involved in maintaining microbial population size through potential adherence to substrates. The metaproteomic characterizations of microbial intracellular/extracellular proteins provide new insights into the ability of the rumen microbiome to maintain in vitro ruminal fermentation.

Escherichia coli K1 is the most popular neonatal meningitis-causing Gram-negative bacterium. As a key virulence determinant, the K1 capsule enhances the survival of E. coli K1 in human brain microvascular endothelial cells (HBMECs) upon crossing the blood–brain barrier; however, the regulatory mechanisms of capsule synthesis during E. coli K1 invasion of HBMECs remain unclear. Here, we identified YbdO as a transcriptional regulator that promotes E. coli K1 invasion of HBMECs by directly activating K1 capsule gene expression to increase K1 capsule synthesis. We found that ybdO deletion significantly reduced HBMEC invasion by E. coli K1 and meningitis occurrence in mice. Additionally, electrophoretic mobility shift assay and chromatin immunoprecipitation–quantitative polymerase chain reaction analysis indicated that YbdO directly activates kpsMT and neuDBACES expression, which encode products involved in K1 capsule transport and synthesis by directly binding to the kpsM promoter. Furthermore, ybdO transcription was directly repressed by histone-like nucleoid structuring protein (H-NS), and we observed that acidic pH similar to that of early and late endosomes relieves this transcriptional repression. These findings demonstrated the regulatory mechanism of YbdO on K1 capsule synthesis, providing further insights into the evolution of E. coli K1 pathogenesis and host–pathogen interaction.

Reasonable delineation of the urban growth boundary (UGB) plays a vital role in guiding orderly urban space growth and ensuring urban environmental health. Existing methodologies for UGB delineation have failed to address the significance of ecological security. Therefore, this study presents a framework that couples ecological security pattern (ESP) establishment and construction land expansion (CLE) simulation to delineate the UGB. The proposed framework is applied to the Nanchang Metropolitan Area (NCMA) in southeastern China. First, we established the regional ESP of the NCMA in 2018 based on an improved minimum cumulative resistance model. The areas of low-, medium-, and high-level ESP were 1050.75, 736.42, and 720.59 km2, respectively. Second, we implemented a multi-scenario simulation of CLE in the NCMA in 2025 based on a cellular automata–Markov model. A natural development scenario was superior to urban growth and ecological protection scenarios for social, economic, and ecological development at the regional scale. Accordingly, we delineated the UGB of the NCMA in 2025 with a scale of 687.87 km2, based on dynamic adjustment using the results of ESP establishment and CLE simulation in the natural development scenario. The rationality and scientificity of the proposed framework were verified by comparing the scale and layout of the delineated UGB with the regional planning of Nanchang City. The framework incorporating dynamic adjustment with ESP establishment and multi-scenario CLE simulation provides a useful tool for the delineation of the UGB in similar urbanized cities. Its application is conducive to achieving a win–win outcome of regional ecological security and urban development.

The O-antigen is the outermost component of the lipopolysaccharide layer in Gram-negative bacteria, and the variation of O-antigen structure provides the basis for bacterial serological diversity. Here, we determined the O-antigen structure of an Escherichia coli strain, LL004, which is totally different from all of the E. coli serogroups. The tetrasaccharide repeating unit was determined as →4)-β-d-Galp-(1→3)-β-d-GlcpNAc6OAc(~70%)-(1→3)-β-d-GalpA-(1→3)-β-d-GalpNAc-(1→ with monosaccharide analysis and NMR spectra. We also characterized the O-antigen gene cluster of LL004, and sequence analysis showed that it correlated well with the O-antigen structure. Deletion and complementation testing further confirmed its role in O-antigen biosynthesis, and indicated that the O-antigen of LL004 is assembled via the Wzx/Wzy dependent pathway. Our findings, in combination, suggest that LL004 should represent a novel serogroup of E. coli.

Many future space missions to asteroids and comets will implement autonomous or near-autonomous navigation, in order to save costly observation time from Earth tracking stations, improve the security of spacecraft and perform real-time operations. Existing Earth-Spacecraft-Earth tracking modes rely on severely limited Earth tracking station resources, with back-and-forth delays of up to several hours. In this paper, we investigate the use of CONSERT ranging data acquired in direct visibility between the lander Philae and the Rosetta orbiter, in the frame of the ESA space mission to comet 67P/Churyumov-Gerasimenko, as a proxy of autonomous navigation and orbitography science capability.

Microwave Imager Combined Active and Passive (MICAP), which is a package of active and passive microwave instruments including L/C/K-band radiometers and L-band scatterometer, has been approved to be taken onbord the Chinese Ocean Salinity Mission. The L-band one-dimensional synthetic aperture radiometer (L-Rad) is the key part of MICAP to measure sea surface salinity (SSS). Since radio frequency interference (RFI) is reported as a serious threat to L-band radiometry, the RFI detection and mitigation techniques must be carefully designed before launch. However, these techniques need to be developed based on the knowledge of how RFI affects complex correlation, visibility function, and reconstructed brightness temperature. This paper presents a time-domain signal modeling method for the simulation of interferometric measurement under RFI’s presences, and a simulation system for L-Rad is established accordingly. Several RFI cases are simulated with different RFI types, parameters, and positions; and the RFI characteristics upon L-Rad’s measurement are discussed. The proposed simulation system will be further dedicated to the design of RFI processing strategy onboard MICAP.

Vascular endothelial growth factor (VEGF) is a well-known angiogenic factor that plays a critical role in various physiological and pathological processes. VEGF also contributes to the process of embryo implantation by enhancing embryo development, improving endometrial receptivity, and facilitating the interactions between the developing embryo and the endometrium. There is a correlation between the alteration of VEGF expression and reproductive failure, including recurrent implantation failure (RIF) and recurrent miscarriage (RM). In order to clarify the role of VEGF in embryo implantation, we reviewed recent literature concerning the expression and function of VEGF in the reproductive system around the time of embryo implantation and we provide a summary of the findings reported so far. We also explored the effects and the possible underlying mechanisms of action of VEGF in embryo implantation.

How to allocate the limited water consumption and pollutant carrying capacity of water function areas to sub-areas and water sectors is key for the practice of the most stringent water resource management decision-making. In response to the joint optimal allocation mechanism of water consumption and pollutant-carrying capacity, joint optimal allocation and the unified quantification problem of multiform benefits, the whole process of joint optimal distribution is refined into the links “water supply—water use and consumption—drainage—pollutant carrying” and the unified quantification method based on the emergy theory to achieve various economic, social, and ecological benefits in different links is proposed. Then the joint optimal allocation joint optimal allocation model of regional water consumption and pollutant carrying capacity is constructed, which realizes the reverse constraint relationship between water quantity and pollution carrying capacity in the water quality and quantity conversion processes. Using Zhengzhou city as an example, the models and methods were applied and researched. Countermeasures and suggestions for the rational allocation of available water resources and water pollution capacity were put forward accordingly.

Value accounting of water in the Yellow River Basin is a key issue in managing local water resources in an efficient, equitable, and sustainable way. In view of the dubious current theories of water resource value, the value transfer of water resources, based on energy flow, is discussed from the perspective of eco-economics. An emergy analysis method is introduced to quantify both the sediment transportation value and social value, and a quantitative system of eco-economic value indicators is constructed. The water resource values of 66 cities in the Basin were calculated, and the GIS atlas was used to describe their spatial distribution. Eight typical cities were selected for the key analysis. The results show that: (1) Among the sub-items, the social value of water per unit is the largest, reaching 30.67 Chinese Yuan/m³, and the difference between the maximum and minimum is only 0.04%, which reflects the social equity of water resources. (2) The eco-environmental value inside the river is generally higher than that of industry, and it is verified that industrial water should not intrude the eco-environmental water in the river. (3) The unit agricultural value of water is the lowest among the sub-items, and the construction of water-saving agriculture should be carried out.

Copy number variation (CNV) is a common type of structural variation in the human genome. Accurate detection of CNVs from tumor genomes can provide crucial information for the study of tumor genesis and cancer precision diagnosis. However, the contamination of normal genomes in tumor genomes and the crude profiles of the read depth make such a task difficult. In this paper, we propose an alternative approach, called CIRCNV, for the detection of CNVs from sequencing data. CIRCNV is an extension of our previously developed method CNV-LOF, which uses local outlier factors to predict CNVs. Comparatively, CIRCNV can be performed on individual tumor samples and has the following two new features: (1) it transfers the read depth profile from a line shape to a circular shape via a polar coordinate transformation, in order to improve the efficiency of the read depth (RD) profile for the detection of CNVs; and (2) it performs a second round of CNV declaration based on the truth circular RD profile, which is recovered by estimating tumor purity. We test and validate the performance of CIRCNV based on simulation and real sequencing data and perform comparisons with several peer methods. The results demonstrate that CIRCNV can obtain superior performance in terms of sensitivity and precision. We expect that our proposed method will be a supplement to existing methods and become a routine tool in the field of variation analysis of tumor genomes.

The novel compound JRS-15 was obtained through the chemical modification of xylocydine. JRS-15 exhibited much stronger cytotoxic and pro-apoptotic activity than its parent compound in various cancer cell lines, with IC50 values in HeLa, HepG2, SK-HEP-1, PC-3M and A549 cells ranging from 12.42 to 28.25 µM. In addition, it is more potent for killing cancer than non-cancerous cells. Mechanistic studies showed that JRS-15 treatment arrested cell cycle at the G1/S phase, which further triggered the translocation of Bax and Bak to the mitochondria, resulting in mitochondrial membrane potential (MMP) depolarization and the subsequent release of cytochrome c and the second mitochondria-derived activator of caspase (Smac). The sequential activation of caspase-9 and caspase-3/7 and the cleavage of poly (ADP-ribose) polymerase (PARP) were observed following these mitochondrial events. Caspase-8, an initiator caspase that is required to activate the membrane receptor-mediated extrinsic apoptosis pathway was not activated in JRS-15-treated cells. Further analysis showed that the levels of the anti-apoptotic proteins Bcl-xL and XIAP were significantly reduced upon JRS-15 treatment. Furthermore, the caspase-9 inhibitor z-LEHD-fmk, the pan-caspase inhibitor z-VAD-fmk, and Bcl-xL or XIAP overexpression all effectively prevented JRS-15-induced apoptosis. Taken together, these results indicate that JRS-15 induces cancer cell apoptosis by regulating multiple apoptosis-related proteins, and this compound may therefore be a good candidate reagent for anticancer therapy.

Ginsenoside Rh2 (G-Rh2) has been shown to induce apoptotic cell death in a variety of cancer cells. However, the details of the signal transduction cascade involved in G-Rh2-induced cell death is unclear. In this manuscript we elucidate the molecular mechanism of G-Rh2-induced apoptosis in human hepatoma SK-HEP-1 cells by demonstrating that G-Rh2 causes rapid and dramatic translocation of both Bak and Bax, which subsequently triggers mitochondrial cytochrome c release and consequent caspase activation. Interestingly, siRNA-based gene inactivation of caspase-8 effectively delays caspase-9 activation and apoptosis induced by G-Rh2, indicating that caspase-8 also plays an important role in the G-Rh2-induced apoptosis program. Taken together, our results indicate that G-Rh2 employs a multi pro-apoptotic pathway to execute cancer cell death, suggesting a potential role for G-Rh2 as a powerful chemotherapeutic agent.
