The Ozone Mapping and Profiler Suites (OMPS) Nadir Mapper (NM) is a grating spectrometer within the OMPS nadir instruments onboard the SNPP, NOAA-20, and NOAA-21 satellites. It is designed to measure Earth radiance and solar irradiance spectra in wavelengths from 300 nm to 380 nm for operational retrievals of the nadir total column ozone. This study presents calibration and validation analysis results for the NOAA-21 OMPS NM SDR data to meet the JPSS scientific requirements. The NOAA-21 OMPS SDR calibration derives updates of several previous OMPS algorithms, including the dark current correction algorithm, one-time wavelength registration from ground to on-orbit, daily intra-orbit wavelength shift correction, and stray light correction. Additionally, this study derives an empirical scale factor to remove 2.2% of systematic biases in solar flux data, which were caused by pre-launch solar calibration errors of the OMPS nadir instruments. The validation of the NOAA-21 OMPS SDR data is conducted using various methods. For example, the 32-day average method and radiative transfer model are employed to estimate inter-sensor radiometric calibration differences from either the SNPP or NOAA-20 data. The quality of the NOAA-21 OMPS NM SDR data is largely consistent with that of the SNPP and NOAA-20 OMPS data, with differences generally within ±2%. This meets the scientific requirements, except for some deviations mainly in the dichroic range between 300 nm and 303 nm. The deep convective cloud target approach is used to monitor the stability of NOAA-21 OMPS reflectance above 330 nm, showing a variation of 0.5% over the observed period. Data from the NOAA-21 VIIRS M1 band are used to estimate OMPS NM data geolocation errors, revealing that along-track errors can reach up to 3 km, while cross-track errors are generally within ±1 km.

Post-disaster housing, constructed on a massive scale, often overlooks the indoor thermal environment, despite being a crucial design factor for residential satisfaction. This study examined the indoor thermal environment in post-Gorkha earthquake-reconstructed prototype and traditional vernacular houses in the Dolakha district of Nepal. It employed a questionnaire survey and measurement of indoor and outdoor temperature in both house types across two study locations: Panipokhari and Jillu, during the coldest winter month. Despite the indoor temperature in both house types falling below the ASHRAE comfort standard, the study found that prototype houses’ nighttime indoor temperatures were 2.1 °C lower in Panipokhari and 1 °C lower in Jillu compared to vernacular houses. This difference is attributed to the use of local building materials with low U-values, substantial thermal mass in vernacular houses, and a low window-to-wall ratio. Occupants expressed dissatisfaction with the thermal environment in prototype houses compared to vernacular ones. By incorporating climate-responsive features seen in vernacular houses, heating energy could have been reduced by approximately 21% in Panipokhari and 10% in Jillu, easing the economic burden on vulnerable households. These findings hold significance for policy-makers, implementers, designers, and other stakeholders involved in post-disaster resettlement housing programs, offering insights for enhancing long-term satisfaction and sustainability in such programs.

Osteoarthritis (OA) is the progressive destruction of articular cartilage with severe symptoms, including pain and stiffness. We investigated the anti-osteoarthritic effects of Prunella vulgaris (PV) and Gentiana lutea (GL) extract in primary cultured chondrocytes RAW 264.7 cells in vitro and destabilization of the medial meniscus (DMM)-induced OA mice in vivo. Primary chondrocytes were induced with IL-1β, and RAW 264.7 cells were treated with LPS and co-incubated with either individual extracts of PV and GL or different ratios of PV and GL mixture. For the OA animal model, the medial meniscus (DMM) was destabilized in 9-week-old male C57BL/6 mice. Treatment of individual PV and GL and combination of PV and GL extracts inhibited the mRNA expression level of COX2 in chondrocytes and RAW 264.7 cells. The optimized inhibitory effect was attained with a PV and GL combination at an 8:2 ratio (PG) without cytotoxic effects. PG extracts prevented the expression of catabolic factors (COX2, Mmp3, Mmp9, and Mmp13) and inflammatory mediator levels (PGE2 and collagenase). In addition, PG decreased subchondral sclerosis and increased BMD in the subchondral region of DMM-induced OA mice with protection of articular cartilage destruction by inhibiting inflammatory processes. This study suggests that PG may be an alternative medicinal herb for treatment of OA.

In this study, we used RVUS data from RadCalNet as a benchmark to verify the radiometric accuracy and stability of operational and reprocessed SNPP/VIIRS data and the accuracy of NOAA-20/VIIRS data, as well as to assess the efficiency of the SNPP/VIIRS reprocessing algorithm. In addition, to remove the uncertainty of the RVUS site itself, we used Landsat 8/OLI as another benchmark with which to validate the accuracy and stability of VIIRS data through the RUVS site. The radiometric biases of the operational and reprocessed SNPP VIIRS bands were within ±4% and ±2%, respectively, as compared with the RUVS site and OLI, except for the M10 and M11 bands. In particular, the biases of the M5 and M7 bands were reduced by ~2% in this study. NOAA-20 VIIRS, on the other hand, was consistently lower than SNPP by ~2 to ~4% for all the bands. For the equivalent bands, the drift differences between operational and reprocessed SNPP/VIIRS and OLI were no larger than 0.24%/year and 0.1%/year, respectively. The reprocessing algorithm of SNPP VIIRS efficiently improved the radiometric accuracy and stability of the SNPP/VIIRS dataset to meet its specifications.

Energy is a fundamental need of modern society and a basis for economic and social development, and one of the major Sustainable Development Goals (SDG), particularly SDG7. However, the UN’s SDG Report 2021 betrays millions of people living without electricity and one-third of the world’s population deprived of using modern energy cooking services (MECS) through access to electricity. Achieving the SDG7 requires standard approaches and tools that effectively address the geographical, infrastructural, and socioeconomic characteristics of a (rural) municipality of Nepal. Furthermore, Nepal’s Constitution 2015 incorporated a federal system under the purview of a municipality as the local government that has been given the mandate to ensure electricity access and clean energy. To address this, a methodology is developed for local government planning in Nepal in order to identify the optimal mix of electrification options by conducting a detailed geospatial analysis of renewable energy (RE) technologies by exploring accessibility and availability ranging from grid extensions to mini-grid and off-grid solutions, based on (a) life cycle cost and (b) levelized cost of energy. During energy assessment, geospatial and socio-economic data are coupled with household and community level data collected from a mobile survey app, and are exploited to garner energy status-quo and enable local governments to assess the existing situation of energy access/availability and planning. In summary, this paper presents a geo-enabled municipal energy planning method and a comprehensive toolkit to facilitate sustainable energy access to local people.

Antibodies to influenza D virus (IDV) have been detected in horses, but no evidence of disease in the field has been reported. To determine whether IDV is infectious, immunogenic, and pathogenic in horses, four 2-year-old horses seronegative for both influenza A (H3N8) and D viruses were intranasally inoculated with 6.25 × 107 TCID50/animal of D/bovine/California/0363/2019 (D/CA2019) virus, using a portable equine nebulizer system. Horses were observed daily for clinical signs including rectal temperature, nasal discharge, coughing, lung sounds, tachycardia, and tachypnea. No horses exhibited clinical signs of disease. Nasopharyngeal swabs collected from 1–8 days post-infection demonstrated virus shedding by qRT-PCR. The horses showed evidence of seroconversion as early as 13 days post-infection (dpi) and the geometric mean of the antibody titers (GMT) of all four horses ranged from 16.82–160 as demonstrated by the microneutralization assay. Further, deep RNA sequencing of the virus isolated in embryonated chicken eggs revealed no adaptive mutations indicating that IDV can replicate in horses, suggesting the possibility of interspecies transmission of IDV with bovine reservoir into equids in nature.

Despite some significant advancements, breast cancer has become the most prevalent cancer in the world. One of the main reasons for failure in treatment and metastasis has been attributed to the presence of cancer initiating cells—cancer stem cells. Consequently, research is now being focussed on targeting cancer cells along with their stem cell population. Non-oncology drugs are gaining increasing attention for their potent anticancer activities. Metformin, a drug commonly used to treat type 2 diabetes, is the best example in this regard. It exerts its therapeutic action by activating 5′ adenosine monophosphate-activated protein kinase (AMPK). Activated AMPK subsequently phosphorylates and targets several cellular pathways involved in cell growth and proliferation and the maintenance of stem-like properties of cancer stem cells. Therefore, AMPK is emerging as a target of choice for developing effective anticancer drugs. Vanadium compounds are well-known PTP inhibitors and AMPK activators. They find extensive applications in treatment of diabetes and obesity via PTP1B inhibition and AMPK-mediated inhibition of adipogenesis. However, their role in targeting cancer stem cells has not been explored yet. This review is an attempt to establish the applications of insulin mimetic vanadium compounds for the treatment of breast cancer by AMPK activation and PTP1B inhibition pathways.

Tumour cells maintain a local hypoxic and acidic microenvironment which plays a crucial role in cancer progression and drug resistance. Urease is a metallohydrolases that catalyses the hydrolysis of urea into ammonia and carbon dioxide, causing an abrupt increase of pH. This enzymatic activity can be employed to target the acidic tumour microenvironment. In this study, we present the anticancer activities of urease mimetic cobalt (III) complexes on A549 cells. The cells were treated with different doses of cobalt (III) complexes to observe the cytotoxicity. The change in cellular morphology was observed using an inverted microscope. The cell death induced by these complexes was analysed through ATP proliferation, LDH release and caspase 3/7 activity. The effect of extracellular alkalinization by the cobalt (III) complexes on the efficacy of the weakly basic drug, doxorubicin (dox) was also evaluated. This combination therapy of dox with cobalt (III) complexes resulted in enhanced apoptosis in A549 cells, as evidenced by elevated caspase 3/7 activity in treated groups. The study confirms the urease mimicking anticancer activity of cobalt (III) complexes by neutralizing the tumour microenvironment. This study will motivate the applications of transition metal-based enzyme mimics in targeting the tumour microenvironment for effective anticancer treatments.

Activation of the neuropeptide S receptor (NPSR) system has been shown to produce anxiolytic-like actions, arousal, and enhance memory consolidation, whereas blockade of the NPSR has been shown to reduce relapse to substances of abuse and duration of anesthetics. We report here the discovery of a novel core scaffold (+) N-benzyl-3-(2-methylpropyl)-1-oxo-3-phenyl-1H,3H,4H,5H,6H,7H-furo[3,4-c]pyridine-5-carboxamide with potent NPSR antagonist activity in vitro. Pharmacokinetic parameters demonstrate that 14b reaches pharmacologically relevant levels in plasma and the brain following intraperitoneal (i.p.) administration, but is cleared rapidly from plasma. Compound 14b was able to block NPS (0.3 nmol)-stimulated locomotor activity in C57/Bl6 mice at 3 mg/kg (i.p.), indicating potent in vivo activity for the structural class. This suggests that 14b can serve as a useful tool for continued mapping of the pharmacological functions of the NPS receptor system.

Equine rotavirus group A (ERVA) is one of the most common causes of foal diarrhea. Starting in February 2021, there was an increase in the frequency of severe watery to hemorrhagic diarrhea cases in neonatal foals in Central Kentucky. Diagnostic investigation of fecal samples failed to detect evidence of diarrhea-causing pathogens including ERVA. Based on Illumina-based metagenomic sequencing, we identified a novel equine rotavirus group B (ERVB) in fecal specimens from the affected foals in the absence of any other known enteric pathogens. Interestingly, the protein sequence of all 11 segments had greater than 96% identity with group B rotaviruses previously found in ruminants. Furthermore, phylogenetic analysis demonstrated clustering of the ERVB with group B rotaviruses of caprine and bovine strains from the USA. Subsequent analysis of 33 foal diarrheic samples by RT-qPCR identified 23 rotavirus B-positive cases (69.69%). These observations suggest that the ERVB originated from ruminants and was associated with outbreaks of neonatal foal diarrhea in the 2021 foaling season in Kentucky. Emergence of the ruminant-like group B rotavirus in foals clearly warrants further investigation due to the significant impact of the disease in neonatal foals and its economic impact on the equine industry.

Due to complex radiometric calibration, the imagery collected by the Day/Night Band (DNB) of the Visible Infrared Imaging Radiometer Suite (VIIRS) onboard the Suomi National Polar Partnership (Suomi-NPP) and the NOAA-20 follow-on satellite is subject to artifacts such as striping, which eventually affect Earth remote sensing applications. Through comprehensive analysis using the NOAA-20 VIIRS DNB prelaunch-test and on-orbit data, it is revealed that the striping results from flaws in the calibration process. In particular, a discrepancy between the low-gain stage (LGS) Earth view (EV) gain and the onboard calibrator solar diffuser view gain makes the operational LGS gain coefficients of a few aggregation modes and detectors biased. Detector nonlinearity at low radiance level also induces errors to the mid-gain stage (MGS) and high-gain stage (HGS) gain through the biased gain ratios. These systematic errors are corrected by scaling the operational LGS gains using the factors derived from the NOAA-20 VIIRS DNB prelaunch test data and by adopting linear regression for evaluating the gain ratios. Striping in the NOAA-20 VIIRS DNB imagery is visibly reduced after the upgraded gain calibration process was implemented in the operational calibration.

In this study, we investigated the occurrence of Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) RNA in primary influent (n = 42), secondary effluent (n = 24) and tertiary treated effluent (n = 34) collected from six wastewater treatment plants (WWTPs A–F) in Virginia (WWTP A), Florida (WWTPs B, C, and D), and Georgia (WWTPs E and F) in the United States during April–July 2020. Of the 100 wastewater samples analyzed, eight (19%) untreated wastewater samples collected from the primary influents contained SARS-CoV-2 RNA as measured by reverse transcriptase quantitative polymerase chain reaction (RT-qPCR) assays. SARS-CoV-2 RNA were detected in influent wastewater samples collected from WWTP A (Virginia), WWTPs E and F (Georgia) and WWTP D (Florida). Secondary and tertiary effluent samples were not positive for SARS-CoV-2 RNA indicating the treatment processes in these WWTPs potentially removed SARS-CoV-2 RNA during the secondary and tertiary treatment processes. However, further studies are needed to understand the log removal values (LRVs) and transmission risks of SARS-CoV-2 RNA through analyzing wastewater samples from a wider range of WWTPs.

Suomi NPP has been successfully operating since its launch on 28 October 2011. As one of the major payloads, along with microwave and infrared sounders (Advanced Technology Microwave Sounder (ATMS), Cross-track Infrared Sounder (CrIS)), and ozone mapping/profiling (OMPS) instruments, the Visible Infrared Imaging Radiometer Suite (VIIRS) has performed for well beyond its mission design life. Its data have been used for a variety of applications for nearly 30 environmental data products, including global imagery twice daily with 375 and 750 m resolutions, clouds, aerosol, cryosphere, ocean color and sea-surface temperature, a number of land products (vegetation, land-cover, fire and others), and geophysical and social economic studies with nightlights. During the early days of VIIRS operational calibration and data production, there were inconsistencies in both algorithms and calibration inputs, for several reasons. While these inconsistencies have less impact on nowcasting and near real-time applications, they introduce challenges for time series analysis due to calibration artifacts. To address this issue, we developed a comprehensive algorithm, and recalibrated and reprocessed the Suomi NPP VIIRS radiometric data that have been produced since the launch. In the recalibration, we resolved inconsistencies in the processing algorithms, terrain correction, straylight correction, and anomalies in the thermal bands. To improve the stability of the reflective solar bands, we developed a Kalman filtering model to incorporate onboard solar, lunar, desert site, inter-satellite calibration, and a deep convective cloud calibration methodology. We further developed and implemented the Solar Diffuser Surface Roughness Rayleigh Scattering model to account for the sensor responsivity degradation in the near infrared bands. The recalibrated dataset was validated using vicarious sites and alternative methods, and compared with independent processing from other organizations. The recalibrated radiometric dataset (namely, the level 1b or sensor data records) also incorporates a bias correction for the reflective solar bands, which not only addresses known calibration biases, but also allows alternative calibrations to be applied if so desired. The recalibrated data have been proven to be of high quality, with much improved stability (better than 0.3%) and accuracy (by up to 2%). The recalibrated radiance data are now available from 2012 to 2020 for users and will eventually be archived on the NOAA CLASS database.

The launch of the National Oceanic and Atmospheric Administration (NOAA)/ National Aeronautics and Space Administration (NASA) Suomi National Polar-orbiting Partnership (S-NPP) and its follow-on NOAA Joint Polar Satellite Systems (JPSS) satellites marks the beginning of a new era of operational satellite observations of the Earth and atmosphere for environmental applications with high spatial resolution and sampling rate. The S-NPP and JPSS are equipped with five instruments, each with advanced design in Earth sampling, including the Advanced Technology Microwave Sounder (ATMS), the Cross-track Infrared Sounder (CrIS), the Ozone Mapping and Profiler Suite (OMPS), the Visible Infrared Imaging Radiometer Suite (VIIRS), and the Clouds and the Earth’s Radiant Energy System (CERES). Among them, the ATMS is the new generation of microwave sounder measuring temperature profiles from the surface to the upper stratosphere and moisture profiles from the surface to the upper troposphere, while CrIS is the first of a series of advanced operational hyperspectral sounders providing more accurate atmospheric and moisture sounding observations with higher vertical resolution for weather and climate applications. The OMPS instrument measures solar backscattered ultraviolet to provide information on the concentrations of ozone in the Earth’s atmosphere, and VIIRS provides global observations of a variety of essential environmental variables over the land, atmosphere, cryosphere, and ocean with visible and infrared imagery. The CERES instrument measures the solar energy reflected by the Earth, the longwave radiative emission from the Earth, and the role of cloud processes in the Earth’s energy balance. Presently, observations from several instruments on S-NPP and JPSS-1 (re-named NOAA-20 after launch) provide near real-time monitoring of the environmental changes and improve weather forecasting by assimilation into numerical weather prediction models. Envisioning the need for consistencies in satellite retrievals, improving climate reanalyses, development of climate data records, and improving numerical weather forecasting, the NOAA/Center for Satellite Applications and Research (STAR) has been reprocessing the S-NPP observations for ATMS, CrIS, OMPS, and VIIRS through their life cycle. This article provides a summary of the instrument observing principles, data characteristics, reprocessing approaches, calibration algorithms, and validation results of the reprocessed sensor data records. The reprocessing generated consistent Level-1 sensor data records using unified and consistent calibration algorithms for each instrument that removed artificial jumps in data owing to operational changes, instrument anomalies, contaminations by anomaly views of the environment or spacecraft, and other causes. The reprocessed sensor data records were compared with and validated against other observations for a consistency check whenever such data were available. The reprocessed data will be archived in the NOAA data center with the same format as the operational data and technical support for data requests. Such a reprocessing is expected to improve the efficiency of the use of the S-NPP and JPSS satellite data and the accuracy of the observed essential environmental variables through either consistent satellite retrievals or use of the reprocessed data in numerical data assimilations.

Few studies have investigated the gas-sensing properties of graphene oxide/titanium dioxide (GO/TiO2) composite combined with photocatalytic effect. Room temperature gas-sensing properties of the GO/TiO2 composite were investigated towards various reducing gases. The composite sensor showed an enhanced gas response and a faster recovery time than a pure GO sensor due to the synergistic effect of the hybridization, such as creation of a hetero-junction at the interface and modulation of charge carrier density. However, the issue of long-term stability at room temperature still remains unsolved even after construction of a composite structure. To address this issue, the surface and hetero-junction of the GO/TiO2 composite were engineered via a UV process. A photocatalytic effect of TiO2 induced the reduction of the GO phase in the composite solution. The comparison of gas-sensing properties before and after the UV process clearly showed the transition from n-type to p-type gas-sensing behavior toward reducing gases. This transition revealed that the dominant sensing material is GO, and TiO2 enhanced the gas reaction by providing more reactive sites. With a UV-treated composite sensor, the function of identifying target gas was maintained over a one-month period, showing strong resistance to humidity.

The Visible Infrared Imaging Radiometer Suite (VIIRS) is a key instrument onboard the Suomi NPP (S-NPP) and the NOAA-20 satellites that provides state-of-the-art Earth observations for ocean, land, aerosol, and cloud applications. VIIRS Reflective Solar Band (RSB) Sensor Data Records (SDR, or Level 1b products) are calibrated and produced independently by The National Oceanic and Atmospheric Administration (NOAA) and the National Aeronautics and Space Administration (NASA) VIIRS science teams. Multiple versions of S-NPP and NOAA-20 VIIRS SDRs are available to date. This study evaluates the long-term calibration stability, biases, and inter-channel consistency of S-NPP and NOAA-20 VIIRS SDRs generated by NOAA and NASA over Deep Convective Clouds (DCC) to support downstream applications, especially climate data record studies. Five VIIRS RSB SDRs were analyzed in this study: (1) NOAA version 2 S-NPP VIIRS reprocessed SDRs (NOAA-NPP-V2, 2012–2020), (2) NASA Collection 1 S-NPP VIIRS SDRs (NASA-NPP-C1, 2012–2021), (3) NASA Collection 2 S-NPP VIIRS SDRs (NASA-NPP-C2, 2012–2021), (4) NOAA constant F-factor calibrated NOAA-20 VIIRS SDRs (NOAA-N20-ConstF, 2018–2021), and (5) NASA Collection 2 NOAA-20 VIIRS SDRs (NASA-N20-C2, 2018–2021). The DCC time series analysis results indicate that the calibrations of the three S-NPP VIIRS RSB SDRs are generally stable, with trends within ±0.1%/year for all RSBs, except for M3–M4 (all three S-NPP SDRs) and I3 (NASA-NPP-C1 only). The calibration of NASA-NPP-C2 SDRs is more uniform at individual detector levels. NOAA-NPP-V2 and NASA-NPP-C1 SDRs exhibit non-negligible time-dependent detector level degradation in M1–M4 (up to 1.5% in 2020–2021), causing striping in the SDR imagery. The biases between NOAA and NASA S-NPP VIIRS RSB SDRs are from 0.1% to 2.4%. The calibrations of the two NOAA-20 VIIRS RSB SDRs are also generally stable, with trends within ±0.16%/year. Small downward trends were observed in the visible and near-infrared (VIS/NIR) bands, and small upward trends were observed in the shortwave infrared (SWIR) bands for both NOAA and NASA NOAA-20 SDRs. The biases between NOAA and NASA NOAA-20 VIIRS RSB SDRs are nearly constant over time and within ±0.2% for VIS/NIR bands and ±0.7% for SWIR bands. There exists large inter-satellite biases between S-NPP and NOAA-20 VIIRS SDRs, especially in the VIS/NIR bands (up to 4.5% for NOAA SDRs and up to 7% for NASA SDRs). In addition, the DCC reflectance of S-NPP VIIRS RSB spectral bands is more consistent with that of the SCanning Imaging Absorption SpectroMeter for Atmospheric CHartographY (SCIAMACHY) than that of NOAA-20. Bands M4 and M9 seem out of family in all five S-NPP and NOAA-20 RSB SDRs evaluated.

Influenza viruses infect the epithelial cells of the swine respiratory tract. Cell lines derived from the respiratory tract of pigs could serve as an excellent in vitro model for studying the pathogenesis of influenza viruses. In this study, we examined the replication of influenza viruses in the MK1-OSU cell line, which was clonally derived from pig airway epithelium. MK1-OSU cells expressed both cytokeratin and vimentin proteins and displayed several sugar moieties on the cell membrane. These cells also expressed both Sial2-3Gal and Sial2-6Gal receptors and were susceptible to swine influenza A, but not to human B and C viruses. Interestingly, these cells were also permissive to infection by influenza D virus that utilized 9-O-acetylated glycans. To study the differences in the expression of pattern recognition receptors (PRRs) upon influenza virus infection in the respiratory and digestive tract, we compared the protein expression of various PRRs in MK1-OSU cells with that in the SD-PJEC cell line, a clonally derived cell line from the porcine jejunal epithelium. Toll-like receptor 7 (TLR-7) and melanoma differentiation-associated protein 5 (MDA5) receptors showed decreased expression in influenza A infected MK1-OSU cells, while only TLR-7 expression decreased in SD-PJEC cells. Further research is warranted to study the mechanism behind the virus-mediated suppression of these proteins. Overall, this study shows that the porcine respiratory epithelial cell line, MK1-OSU, could serve as an in-vitro model for studying the pathogenesis and innate immune responses to porcine influenza viruses.
