Endothelial–mesenchymal transition (EnMT) is the transversion of endothelial cells to mesenchymal cells under certain physiological or pathological conditions. When EnMT occurs in the corneal endothelium, corneal endothelial cells (CECs) lose their normal function and thus cannot maintain corneal clarity. Studies have shown that the mechanism of EnMT in CECs involves the transforming growth factor-β (TGF-β) signaling pathway, and one of the important inhibitors of the TGF-β/Smad2/3 pathway is sirtuin-1 (SIRT1). In this study, we used a rat model of corneal endothelium injury and TGF-β1-treated human CECs to induce EnMT, aiming to explore whether SIRT1 activation inhibits corneal EnMT in vivo and in vitro. SIRT1 was activated and suppressed using resveratrol (RSV) and EX527, respectively. The endothelial markers and mesenchymal markers were measured by immunofluorescence and Western blot assays. Co-immunoprecipitation was used to detect the interaction between SIRT1 and Smad2/3. The results showed that after mechanical injury, the group treated with RSV-activated SIRT1 regained corneal transparency and recovered from edema faster than the control group. Moreover, RSV-activated SIRT1 downregulated the expression levels of alpha smooth muscle actin (α-SMA), vimentin, and Snail and upregulated the expression levels of E-cadherin and Na+/K+-ATPase both in vivo and in vitro, but these effects were reversed when SIRT1 was inhibited by EX527. SIRT1 also upregulated the expression levels of TGF-β receptor 1 and phosphorylated Smad2/3. The interaction between SIRT1 and Smad2/3 in vitro was confirmed by co-immunoprecipitation. Overall, our results indicate that SIRT1 activation inhibits corneal EnMT via the TGF-β/Smad2/3 pathway, which may be a potential therapeutic target for corneal endothelium dysfunction.

The diverse content of and ease of listening to podcasts have made podcasts popular, particularly during the COVID-19 pandemic. Advertisers have begun to recognize their marketing potential and are now hiring podcasters to recommend their products. This study sought to determine the factors influencing podcast commitment, parasocial interaction (PSI), and the intention to purchase podcaster-endorsed items. It was conducted in Taiwan with a sample size of 578 participants and an online questionnaire. Structural equation modeling and mediation analysis were applied to the collected data from the perspective of uses and gratifications theory. We found that podcast commitment is positively related to edutainment, storytelling transportation, and social engagement. Social engagement is positively related to PSI, while storytelling transportation has a negative relationship with PSI. Additionally, there is a strong positive correlation between podcast commitment and PSI and both factors positively influence the intention to purchase podcaster-endorsed items. PSI also significantly moderates the positive relationship between podcast commitment and the intent to buy podcaster-endorsed items.

In this study, we present the HOAc-catalyzed selective cleavage of the C=C double bond of enaminones, enabling the formation of a new C–N bond and a new C=N bond for the one-pot synthesis of 2-substituted 3,4-dihydroquinazolines directly from ynones and 2-(aminomethyl)anilines. This method operates in ethanol under transition-metal-free and oxidant-free conditions, offering a sustainable and efficient approach for the synthesis of 3,4-dihydroquinazolines with broad functional group tolerance.

Background: SCTV01E is a tetravalent recombinant COVID-19 vaccine authorized for emergency use in China for adults 18 years and older but not for those under 18. Objective: This Phase 2 trial assessed the safety and immunogenicity of SCTV01E in healthy children and adolescents aged 3 to 17 years, to establish immunobridging with that observed in adults from the efficacy pivotal trial (NCT05308576). Methods: Participants were randomly assigned to receive either 30 µg of SCTV01E or a placebo. Primary endpoints were safety and immunogenicity focused on the geometric mean titer (GMT) and seroresponse rate (SRR) of neutralizing antibodies (nAb) against Omicron BA.5. Results: In total, 268 participants (214 SCTV01E vs. 54 placebo) were included in the safety analysis, with 241 participants (191 vs. 50) in the immunogenicity analysis. Overall, 127 (59.3%) participants receiving SCTV01E and 9 (16.7%) receiving a placebo reported adverse events (AEs), most of which were Grade 1 or 2. No serious adverse events (SAEs) or adverse events of special interest (AESIs) were reported. In the immunogenicity bridging analysis, data from 95 youths were compared with data from 188 adults; the geometric mean ratio (GMR) of the titers was 8.78 (95% CI: 6.05–12.74, p < 0.001), with the lower bound of the 95% CI exceeding 0.67. The difference in the SRR was 6.34% (95% CI: 0.93–11.22%) (p = 0.029), and the lower bound of the 95%CI was >−5%, indicating superiority. Conclusions: SCTV01E was found to be safe and well tolerated in children and adolescents, generating a robust immune response against Omicron BA.5. This supports its potential use in younger populations.

Maize leaf area offers valuable insights into physiological processes, playing a critical role in breeding and guiding agricultural practices. The Azure Kinect DK possesses the real-time capability to capture and analyze the spatial structural features of crops. However, its further application in maize leaf area measurement is constrained by RGB–depth misalignment and limited sensitivity to detailed organ-level features. This study proposed a novel approach to address and optimize the limitations of the Azure Kinect DK through the multimodal coupling of RGB-D data for enhanced organ-level crop phenotyping. To correct RGB–depth misalignment, a unified recalibration method was developed to ensure accurate alignment between RGB and depth data. Furthermore, a semantic information-guided depth inpainting method was proposed, designed to repair void and flying pixels commonly observed in Azure Kinect DK outputs. The semantic information was extracted using a joint YOLOv11-SAM2 model, which utilizes supervised object recognition prompts and advanced visual large models to achieve precise RGB image semantic parsing with minimal manual input. An efficient pixel filter-based depth inpainting algorithm was then designed to inpaint void and flying pixels and restore consistent, high-confidence depth values within semantic regions. A validation of this approach through leaf area measurements in practical maize field applications—challenged by a limited workspace, constrained viewpoints, and environmental variability—demonstrated near-laboratory precision, achieving an MAPE of 6.549%, RMSE of 4.114 cm2, MAE of 2.980 cm2, and R2 of 0.976 across 60 maize leaf samples. By focusing processing efforts on the image level rather than directly on 3D point clouds, this approach markedly enhanced both efficiency and accuracy with the sufficient utilization of the Azure Kinect DK, making it a promising solution for high-throughput 3D crop phenotyping.

Glaesserella parasuis is the etiological agent of Glässer’s disease, which causes high morbidity and mortality in pigs worldwide. Macrolide resistance poses an urgent threat to their treatment, as macrolides are widely used for preventing and treating G. parasuis infections. Here, we determined the susceptibilities to five macrolides and characterized the genetic markers of macrolide resistance. The antimicrobial susceptibility of 117 G. parasuis isolates to erythromycin, tulathromycin, gamithromycin, tylosin, and tilmicosin was evaluated using broth microdilution method. Erythromycin-resistant isolates were sequenced using whole-genome sequencing. Further analysis of these sequences revealed the genetic basis of macrolide resistance in G. parasuis. Our results show that most G. parasuis isolates remained susceptible to the macrolide drugs. For commonly used agents (e.g., tylosin and tilmicosin), elevated minimum inhibitory concentrations (MICs) were observed, whereas for the newer macrolides (e.g., tulathromycin and gamithromycin), the MICs remained almost unchanged. The macrolide resistance gene erm(T) and the A2059G mutation in 23S rRNA were detected in the current study. To the best of our knowledge, integrative and conjugative element (ICE)-borne erm(T) in G. parasuis is reported for the first time in this study. Taken together, these results provide insights into the susceptibility of G. parasuis to macrolides. The presence of erm(T) on ICEs may facilitate its transfer, reducing the effectiveness of macrolide treatment.

Honey robbing, which typically occurs during times of food scarcity, is a perilous foraging strategy for bee colonies and presents a formidable challenge in the realm of beekeeping. This article provides a comprehensive and multifaceted exploration of honey robbing, including the morphology, behavioral traits, timing, and scope of this phenomenon. This exploration elucidates the specific manifestations of honey robbing, offering readers a deeper understanding of its various facets. Next, this article investigates the root causes of honey robbing by examining both abiotic and biotic factors. The resulting harms are outlined, and corresponding preventive and control measures are suggested. Finally, the article succinctly summarizes the current obstacles in research related to honey robbing and outlines promising avenues for future exploration. The objective of this study was to elucidate the occurrence mechanism of honey robbing, ultimately aiming to contribute to the sustainable growth of the beekeeping industry.

A 40-year-old woman who had obstetric history of one vaginal delivery and two surgical abortions to terminate early pregnancy received regular prenatal care without any systemic maternal diseases. During the detailed second trimester ultrasound, a homogenous adhesion-induced pseudocystic lesion of 8.6 × 7.4 cm was found between the inlet of the endocervix and the uterine cavity in the lower segment of the uterus. There was a clear septum with an inlet of about 2.6 cm near the right lower segment of the uterus. Transvaginal sonography showed a cervical length of 3.29 cm without dilatation. No gross fetal anomalies were found. Sometimes, the fetal head or limbs moved into this cystic space. At 36 3/7 weeks of gestation, a cesarean section was arranged for fetal breech presentation and pre-labor rupture of the membrane. After the delivery of the baby and its placenta, there was no obvious septum in the uterine cavity but only a very short fibrous tissue from the posterior wall of uterus, which could be destroyed when the baby was delivered. No adverse outcomes for the mother or the neonate were observed.

Poly (ADP-ribose) polymerase inhibitors (PARPis) show cytotoxicity in homologous recombination deficiency (HRD) seen in BRCA-mutant ovarian cancer (OvCa). Despite initial responses, resistance often develops. The reintroduction of different PARPis, such as niraparib or rucaparib, has shown some clinical activity in BRCA mutation-associated OvCa patients with prior olaparib treatment, yet the underlying mechanisms remain unclear. To investigate the differential sensitivity to different PARPis, we established an olaparib-resistant BRCA1-mutant OvCa cell line (UWB-OlaJR) by exposing UWB1.289 cells to gradually increasing concentrations of olaparib. UWB-OlaJR exhibited restored HR capability without BRCA1 reversion mutation or increased drug efflux. We examined cell viability, DNA damage, and DNA replication fork dynamics in UWB-OlaJR treated with various PARPis. UWB-OlaJR exhibits varying sensitivity to PARPis, showing cross-resistance to veliparib and talazoparib, and sensitivity with increased cytotoxicity to niraparib and rucaparib. Indeed, DNA fiber assay reveals that niraparib and rucaparib cause higher replication stress than the others. Moreover, S1 nuclease fiber assay shows that niraparib and rucaparib induce greater DNA single-strand gaps than other PARPis, leading to increased DNA damage and cell death. Our study provides novel insights into differential PARPi sensitivity in olaparib-resistant BRCA-mutant OvCa, which requires further investigation of inter-agent differences in large prospective studies.

Autophagy is a complex cellular process that can either promote or inhibit cancer progression and development, depending on the context and molecular regulation involved. This study investigates how LSD1 regulates autophagy in ovarian cancer by interacting with the autophagy protein LC3B. Utilizing the bioinformatic analysis of TCGA, CPTAC, and GEO datasets, as well as immunohistochemistry in ovarian cancer patients, we explored the expression association between LSD1 and LC3B. Molecular mechanisms were further analyzed using Western blotting, immunoprecipitation, and GST pull-down assays. Our findings reveal that LSD1 binds to LC3B via its SWIRM domain, and high levels of LSD1 are closely associated with aggressive ovarian cancer and poor patient outcomes. Mechanistically, LSD1 demethylates LC3B, leading to decreased LC3B stability. The observed inverse correlation between LSD1 expression and LC3B protein levels in clinical samples underscores the need for further investigation to elucidate how reduced LC3B protein levels induced by LSD1 demethylation may contribute to ovarian cancer.

Background: Influenza vaccine uptake among healthcare workers is crucial for preventing influenza infections, yet its effectiveness needs further investigation. Objectives: This prospective observational study aimed to assess the protective effect of influenza vaccination among healthcare workers in Shenzhen. Methods: We enrolled 100 participants, with 50 receiving the 2023–2024 quadrivalent influenza vaccine (QIV) and 50 serving as unvaccinated controls. Epidemiological data were collected when the participants presented influenza-like illness. Serum samples were collected at three time points (pre-vaccination and 28 and 180 days after vaccination). Hemagglutination inhibition (HI) assay was performed against the strains included in the 2023–2024 QIV (H1N1, H3N2, BV and BY strains) to assess antibody protection levels. Demographics comparisons revealed no significant differences between the vaccinated and control groups (p > 0.05), ensuring group comparability. Results: The incidence of influenza-like illness was significantly lower in the vaccinated (18%) compared to the control group (36%; p = 0.046; OR = 0.39; 95% CI: 0.15 to 0.98). The vaccinated group also exhibited a higher rate of consecutive two-year vaccinations (48% vs. 24% in the control group, p < 0.05). Additionally, the vaccinated healthcare workers were more inclined to recommend vaccination to their families (80% vs. 48%, p < 0.05). HI titers against H1N1 (p < 0.01), H3N2 (p < 0.01), BV (p < 0.001) and BY (p < 0.01) significantly increased in the vaccinated group at 28 days post-vaccination. Moreover, a marked and sustained increase in HI titers against the H3N2 strain (p < 0.001) was observed at 180 days post-vaccination, highlighting the vaccine’s enduring impact on the immune response. The fold change in the HI titers, indicative of the magnitude of the immune response, was significantly higher for H1N1 (p < 0.01), H3N2 (p < 0.001), BV (p < 0.01) and BY (p < 0.05) among the vaccinated individuals compared to the control group, underscoring the vaccine’s efficacy in eliciting a robust and sustained antibody response. Conclusion: Influenza vaccination significantly reduces the incidence of influenza-like illness among healthcare workers and promotes a sustained immune response. The study supports the importance of annual vaccination for this group to enhance personal and public health.

This study investigated the effect of substances on nonspecific immune responses of head kidney leukocytes, the antimicrobial activity against Vibrio sp., as well as the time-kill of Vibrio sp. by combining the substances with lactic acid bacteria (LAB) and Pediococcus sp. The substances are B. pilosa hot water extract, B. pilosa powder extract, B. pilosa methanol extract, B. pilosa ethanol extract, B. alba hot water extract, B. alba powder extract, B. alba methanol extract, B. alba ethanol extract, and bioactive compounds, namely cytopiloyne, flavonoid, phenol, ethyl caffeate, luteolin, chlorogenic acid, butein, and linoleic acid. The results showed that some of them were nontoxic to the head kidney leukocytes, which can increase the phagocytic rate, phagocytic index, and respiratory burst. These substances were able to inhibit the growth of Vibrio sp.; they can even completely kill the pathogenic bacteria. The largest of the inhibition zone formed from the EC group at a concentration range of 5–50 µg/mL against V. parahaemolyticus, V. alginolyticus, and V. harveyi with a value of 19.7 ± 0.56, 19.3 ± 1.53, and 20.6 ± 1.53 mm. Furthermore, the time-kill studies showed that the LAB and P. acidilactici can completely kill the Vibrio sp. at 6 h incubation time, mainly in the group of combination with EC.

Background/Objectives: Frozen shoulder is a common shoulder disorder that often places limitations on the range of motion of the shoulder. The disease may induce neck pain due to overuse of the neck muscle in an attempt to compensate for lack of shoulder movement. In clinical practice, swelling and inflammation of the scalene and levator scapulae may be detected via sonography in patients with frozen shoulder. The aim of this study was, therefore, to determine whether the involvement of the scalene complex or levator scapulae could compensate for the limited motion of the shoulder in patients with frozen shoulder. Methods: We retrospectively reviewed the medical records of 362 patients with unilateral frozen shoulder. These patients were divided into four groups depending on the involvement of the scalene complex or levator scapulae muscle. The range of motion of the shoulder—encompassing flexion, abduction, and external rotation—was measured with a goniometer. We also performed an ultrasound scan on each shoulder. The involvement of the scalene complex and levator scapulae muscle was also assessed via musculoskeletal ultrasound. Results: The range of motion of the shoulder in terms of flexion, abduction, external rotation, and total range of motion differed significantly between the four groups (p < 0.05). Patients in whom the scalene complex or levator scapulae muscle was involved demonstrated a significantly wider range of motion in different shoulder directions than patients without the involvement of those muscles (p < 0.05). Conclusions: A greater range of motion in the shoulder can be obtained through the activation of the scalene complex or levator scapulae muscle, which act to compensate for the lack of shoulder movement in patients with frozen shoulder. These two muscles showed thickening and hypoechoic changes upon sonography.

As one of the most important future clean energy sources, natural gas hydrate (NGH) is attracting widespread attention due to the vast reserves available and high energy density. How to extract this source in a safe, efficient, and environmentally friendly manner has become the key to the commercial utilization of its resources. This paper reviews the recent advances in the study of the fundamental reservoir properties of offshore NGH, summarizing the methods and technologies for testing the sedimentary properties of reservoirs, analyzing the characteristics in reservoir mechanics, electrics, thermodynamics, and fluid dynamics, and discusses the influence of reservoir fundamental properties on NGH exploitation. The aim is to provide guidance and reference for research on the exploitation of NGH in different target exploitation areas offshore.

Concurrent chemoradiotherapy is an effective treatment option for patients with low-grade colorectal cancer (CRC) in the local disease stage. At present, the principle of the Taiwan Medical Center is to treat CRC patients with combination radiotherapy and chemotherapy (high-dose 5-FU) for a period of about five weeks prior to surgery. Radical resection of the tumor is performed at least six to eight weeks after concurrent chemoradiotherapy (CCRT). However, this approach fails to produce the desired therapeutic effect in approximately 20% to 30% of patients, and such patients are unnecessarily exposed to the risks of radiation and drug toxicity posed by this therapy. Therefore, it is crucial to explore new biomarkers to predict the prognosis of CRC. SUMO-activating enzyme subunit 1 (SAE1) plays an important role in SUMOylation, a post-translational modification involved in cellular functions, such as cell proliferation, cell cycle, and apoptosis. In our study, to explore the clinical–pathological role of SAE1 protein in CRC, we evaluated the clinical data and paraffin sections from CRC patients. The expression of SAE1 was evaluated using immunohistochemical analysis, and clinical parameters were analyzed using chi-square and Kaplan–Meier survival tests. The results of in vitro proliferation and radiosensitive assays were compared between control groups and SAE1 siRNA groups. Western blotting was also used to detect the expressions of the SAE1, PARP, cyclin D1, p-NF-κB, and NF-κB proteins. Flow cytometry and colony formation assays were used to detect the effect of SAE-1 on radiosensitivity. In vivo, we detected the growth curve in a mouse xenograft model. The results showed that SAE-1 was revealed to be an independent prognostic biomarker of CRC. SAE1 knockdown inhibited CRC proliferation in vitro and in vivo, and led to the cleavage of PARP, downregulation of cyclin D1 protein expression, and downregulation of p-NF-κB/NF-κB. Additionally, SAE1 knockdown promoted radiosensitivity in CRC cells. Therefore, it was inferred that SAE1 may be used as a potential therapeutic target in CRC treatment.

Adding cement to soft soils may lead to brittle behavior and the occurrence of sudden damage. Methods to further improve the tensile and flexural properties of cemented clay are noteworthy topics. This paper mainly focuses on the effect of cement and moisture content on the strength and flexural properties of cemented clay reinforced by PVA fiber. The selected clayey soil was a kaolin with cement content of 5%, 10%, and 15% and moisture content of 50%, 56%, 63%, and 70%. The results show that the incorporation of 0.6% fiber can effectively improve the deformability of cemented clay in unconfined compression tests (UCS). The strengthening effect of fiber, as seen in the peak strength and post-peak strength of UCS, was significantly related to cement content. As the water content increased, the compressive strength of the fiber-reinforced cemented clay decreased, but its load-bearing capacity enhanced. When the cement content was 15%, the splitting tensile strength of fiber-reinforced cemented specimens increased by 11% compared to cemented soil, but the deformability of the specimens became poor. In the cement-content interval from 5% to 10%, the bending toughness was significantly improved. Sufficient cement addition ensures the enhancement of PVA fibers on strength and flexural properties of cement-stabilized clayey soil.

This meta-synthesis crosses international borders by analyzing successful principalship across nine countries. With careful, detailed, systematic, and analytic synthesis, it critically explores research-informed understandings of successful principal leadership over the last 20 years. The paper is an interim report of reviews of the 85 articles and 23 chapters produced by the International Successful School Principal Project (ISSPP), the largest, longest-running international research project in this field. In that project, success is defined as the principal’s ability to grow and sustain students’ academic, personal, and social developments and achievements over time. The results add to existing knowledge of how successful school principals achieve success by examining high-quality, published case study evidence from Australia, Cyprus, Israel, New Zealand, Norway, Sweden, Spain, the UK, and the US. Results suggest common sets of core values, qualities, and practices among school principals, regardless of national contexts, conditions, or cultures. Because the research design is multi-perspective and multi-layered, the case studies also show significant, context-informed differences in timing, pace, and extent of application of improvement strategies. Thus, the findings of this synthesis paint a thorough and cohesive picture of who successful principals are and what they do over time at the global level to achieve success, as they navigate values-led school success in different contexts and improvement phases within complex internal and external ecological chrono-, macro-, exo-, meso-, and microsystems.

Extracellular HSP90α (eHSP90α) is a promoter of tumor development and malignant progression. Patients with malignancies, including pancreatic ductal adenocarcinoma (PDAC), have generally shown 5~10-fold increases in serum/plasma eHSP90α levels. In this study, we developed a humanized antibody HH01 to target eHSP90α and evaluated its anticancer efficacy. HH01, with novel complementarity-determining regions, exhibits high binding affinity toward HSP90α. It recognizes HSP90α epitope sites 235AEEKEDKEEE244 and 251ESEDKPEIED260, with critical amino acid residues E237, E239, D240, K241, E253, and K255. HH01 effectively suppressed eHSP90α-induced invasive and spheroid-forming activities of colorectal cancer and PDAC cell lines by blocking eHSP90α’s ligation with the cell-surface receptor CD91. In mouse models, HH01 potently inhibited the tumor growth of PDAC cell grafts/xenografts promoted by endothelial-mesenchymal transition-derived cancer-associated fibroblasts while also reducing serum eHSP90α levels, reflecting its anticancer efficacy. HH01 also modulated tumor immunity by reducing M2 macrophages and reinvigorating immune T-cells. Additionally, HH01 showed low aggregation propensity, high water solubility, and a half-life time of >18 days in mouse blood. It was not cytotoxic to retinal pigmented epithelial cells and showed no obvious toxicity in mouse organs. Our data suggest that targeting eHSP90α with HH01 antibody can be a promising novel strategy for PDAC therapy.

The encapsulation of sodium sulfobutylether-β-cyclodextrin (SBE-β-CD) is influenced not only by the degree of substitution (DS) but also by the presence of strong-bonded water (SBW). Guests compete with SBW for positions within the cavity of SBE-β-CD. However, the correlation between DS and SBW was not clear. This study revealed a positive correlation between DS and SBW utilizing Karl Fischer titration. The mechanism may be attributed to molecular polarizability. To explore the impact of SBW inside SBE-β-CD with different DS on encapsulation, density functional theory was employed. Throughout the release process, an increase in enthalpy is unfavorable, while an increase in entropy favors spontaneous reaction occurrence. For SBE-β-CD (DS = 2, 3), enthalpy increase is the primary factor, leading to the retention of SBW within the cavities and consequently hindering guest entry. In contrast, for SBE-β-CD (DS = 4, 7), the situation differs. For SBE10-β-CD, the influence of SBW is minimal. This study aims to elucidate the relationship between DS and SBW, as well as the effect of SBW inside SBE-β-CD with different DS on encapsulation. It is crucial for a comprehensive understanding of the factors affecting the encapsulation of SBE-β-CD, thereby promoting quality control and functional development of SBE-β-CD.

Medium-nickel cobalt-free cathode materials have attracted much attention in recent years for their low cost and high energy density. However, the structural stability of nickel-based cathode materials becomes compromised when accompanied by the increasing of voltage, leading to poor cycling performance and, thus, hindering their widespread industrial application. In this work, we investigated the optimal charge cut-off voltage for the nickel-based cathode material LiNi0.6Mn0.4O2 (NM64). Within the voltage range of 3.0 to 4.5 V, the electrode energy density reached 784.08 Wh/kg, with an initial Coulombic efficiency of 84.49%. The reversible specific capacity at 0.1 C reached 197.84 mAh/g, and it still maintained a high reversible specific capacity of nearly 150 mAh/g, with a capacity retention rate of 86% after 150 cycles at 1 C. Furthermore, NM64 exhibited an intact morphological structure without noticeable cracking after 150 cycles, indicating excellent structural stability. This study emphasizes the relationship between the stability of NM64 cathodes and different operating voltage ranges, thereby promoting the development of high-voltage layered nickel-based cathode materials.

The PI3K signaling pathway plays an essential role in cancer cell proliferation and survival. PI3K pathway inhibitors are now FDA-approved as a single agent treatment or in combination for solid tumors such as renal cell carcinoma or breast cancer. However, despite the high prevalence of PI3K pathway alterations in gynecological cancers and promising preclinical activity in endometrial and ovarian cancer models, PI3K pathway inhibitors showed limited clinical activity in gynecological cancers. In this review, we provide an overview on resistance mechanisms against PI3K pathway inhibitors that limit their use in gynecological malignancies, including genetic alterations that reactivate the PI3K pathway such as PIK3CA mutations and PTEN loss, compensatory signaling pathway activation, and feedback loops causing the reactivation of the PI3K signaling pathway. We also discuss the successes and limitations of recent clinical trials aiming to address such resistance mechanisms through combination therapies.

Aims: This study aims to compare the outcomes of immediate (followed by closed-incision negative-pressure therapy use) versus delayed ORIF in patients with Schatzker type IV–VI TPFs. Patients and Methods: A prospective study of patients undergoing ORIF between January 2018 and December 2019 was performed. The inclusion criteria were patients (>18 years) with a closed fracture sent to the emergency room (ER) within 24 h of injury. All the patients underwent preoperative image evaluation. Two senior orthopedic trauma surgeons evaluated the soft tissue condition in the ER by 5P’s of the compartment syndrome, judging the timing of the operation of definitive ORIF. Group 1 (n = 16) received delayed ORIF. Group 2 (n = 16) received immediate ORIF and ciNPT use. Patient follow-up occurred after 2 and 6 weeks and 3, 6, and 12 months after surgery. The assessments included the time to definitive fixation, the length of hospital stay, the time to bone union, surgical site complications, and reoperation within 12 months. A universal goniometer was used to measure the postoperative 3 m, 6 m, and 12 m ROM. Results: The patient demographics were similar between the groups (p > 0.05). Group 2 displayed significantly a shorter time to definitive fixation (5.94 ± 2.02 vs. 0.61 ± 0.28, p < 0.0001) and hospital stay (14.90 ± 8/78 vs. 10.30 ± 6.78, p = 0.0016). No significant difference was observed in the time to bone union, surgical site complication incidence, and reoperation rates (p > 0.05). Flexion and flexion–extension knee ROM were demonstrated to be significantly improved in Group 2, 3, 6, and 12 months postoperatively (p < 0.0001). Conclusions: In this study, early ORIF and ciNPT use resulted in a shorter hospital length of stay, a reduced time to early active motion of the knee, and improved knee ROM. These results suggest that early ORIF with ciNPT for Schatzker type IV–VI TPFs is safe and effective in some patients. However, further research to confirm these findings across larger and more diverse populations is needed.

The silkworm cocoon (SC), both as a traditional Chinese medicine and as the raw material for biocompatible carriers, has been extensively used in the medical and biomedical fields. This review elaborates on the multiple functions of SC, with an in-depth analysis of its chemical composition, biological activities, as well as its applications in modern medicine. The primary chemical components of SC include silk fibroin (SF), silk sericin (SS), and other flavonoid-like bioactive compounds demonstrating various biological effects. These include hypoglycemic, cardioprotective, hypolipidemic, anti-inflammatory, antioxidant, and antimicrobial actions, which highlight its potential therapeutic benefits. Furthermore, the review explores the applications of silk-derived materials in drug delivery systems, tissue engineering, regenerative medicine, and in vitro diagnostics. It also highlights the progression of SC from laboratory research to clinical trials, emphasizing the safety and efficacy of SC-based materials across multiple medical domains. Moreover, we discuss the market products developed from silk proteins, illustrating the transition from traditional uses to contemporary medical applications. This review provides support in understanding the current research status of SC and the further development and application of its derived products.

CRISPR-Cas systems are adaptive immune mechanisms present in most prokaryotes that play an important role in the adaptation of bacteria and archaea to new environments. Shewanella algae is a marine zoonotic pathogen with worldwide distribution, which accounts for the majority of clinical cases of Shewanella infections. However, the characterization of Shewanella algae CRISPR-Cas systems has not been well investigated yet. Through whole genome sequence analysis, we characterized the CRISPR-Cas systems in S. algae. Our results indicate that CRISPR-Cas systems are prevalent in S. algae, with the majority of strains containing the Type I-F system. This study provides new insights into the diversity and function of CRISPR-Cas systems in S. algae and highlights their potential role in the adaptation and survival of these marine pathogens.

With the large-scale development of distributed energy on the demand side, the trend of “supply exceeding demand” has gradually become prominent, and regional peer-to-peer (P2P) energy trading has become an important measure to improve the local consumption of distributed energy. However, most existing studies usually assume that prosumers behave entirely rationally with the goal of maximum benefit, which has been proved to deviate from the observed actual behaviors. Aiming at the optimal energy of prosumers participating in the P2P market, a prospect theory-based two-stage stochastic optimization model considering the bounded rationality was proposed to accurately simulate the decision-making behavior. Then, a benefit maximization model for the energy trading service provider (ETSP) was constructed considering the power flow constraint to ensure the safe operation of the system. Finally, an improved R-ADMM algorithm considering timeout was proposed to solve the above model and improve the convergence speed. The effectiveness of the proposed model and algorithm was verified via simulation.

Making judgments of learning (JOLs) can reactively alter memory itself, a phenomenon termed the reactivity effect. The current study recorded electroencephalography (EEG) signals during the encoding phase of a word list learning task to explore the neurocognitive features associated with JOL reactivity. The behavioral results show that making JOLs reactively enhances recognition performance. The EEG results reveal that, compared with not making JOLs, making JOLs increases P200 and LPC amplitudes and decreases alpha and beta power. Additionally, the signals of event-related potentials (ERPs) and event-related desynchronizations (ERDs) partially mediate the reactivity effect. These findings support the enhanced learning engagement theory and the elaborative processing explanation to account for the JOL reactivity effect.

The process of soil genesis unfolds as pioneering microbial communities colonize mineral substrates, enriching them with biomolecules released from bedrock. The resultant intricate surface units emerge from a complex interplay among microbiota and plant communities. Under these conditions, host rocks undergo initial weathering through microbial activity, rendering them far from pristine and challenging the quest for biomarkers in ancient sedimentary rocks. In addressing this challenge, a comprehensive analysis utilizing Gas Chromatography Mass Spectrometry (GC-MS) and Time-of-Flight Secondary Ion Mass Spectrometry (ToF-SIMS) was conducted on a 520-Ma-old Cambrian rock. This investigation revealed a diverse molecular assemblage with comprising alkanols, sterols, fatty acids, glycerolipids, wax esters, and nitrogen-bearing compounds. Notably, elevated levels of bacterial C16, C18 and C14 fatty acids, iso and anteiso methyl-branched fatty acids, as well as fungal sterols, long-chained fatty acids, and alcohols, consistently align with a consortium of bacteria and fungi accessing complex organic matter within a soil-type ecosystem. The prominence of bacterial and fungal lipids alongside maturity indicators denotes derivation from heterotrophic activity rather than ancient preservation or marine sources. Moreover, the identification of long-chain (>C22) n-alkanols, even-carbon-numbered long chain (>C20) fatty acids, and campesterol, as well as stigmastanol, provides confirmation of plant residue inputs. Furthermore, findings highlight the ability of contemporary soil microbiota to inhabit rocky substrates actively, requiring strict contamination controls when evaluating ancient molecular biosignatures or extraterrestrial materials collected.

Plants must adapt to the complex effects of several stressors brought on by global warming, which may result in interaction and superposition effects between diverse stressors. Few reports are available on how drought stress affects Xanthomonas albilineans (Xa) infection in sugarcane (Saccharum spp. hybrids). Drought and leaf scald resistance were identified on 16 sugarcane cultivars using Xa inoculation and soil drought treatments, respectively. Subsequently, four cultivars contrasting to drought and leaf scald resistance were used to explore the mechanisms of drought affecting Xa–sugarcane interaction. Drought stress significantly increased the occurrence of leaf scald and Xa populations in susceptible cultivars but had no obvious effect on resistant cultivars. The ROS bursting and scavenging system was significantly activated in sugarcane in the process of Xa infection, particularly in the resistant cultivars. Compared with Xa infection alone, defense response via the ROS generating and scavenging system was obviously weakened in sugarcane (especially in susceptible cultivars) under Xa infection plus drought stress. Collectively, ROS might play a crucial role involving sugarcane defense against combined effects of Xa infection and drought stress.

The escalating problem of copper (Cu) and cadmium (Cd) pollution in aquatic environments poses a significant threat to the ovarian tissue and reproductive capacity of fish, hindering the development of the aquaculture industry. However, the combined effects of Cu and Cd on fish gonadal development remain unclear. In this study, the fish species Nile tilapia was stressed with rearing water containing 300 μg/L Cu2+ and 100 μg/L Cd2+ for 30 days, followed by an intraperitoneal injection of luteinizing hormone-releasing hormone (LHRH-α) and human chorionic gonadotropin (HCG) at various concentrations. We investigated the ovarian transcriptome profiles before and after injection. Prior to injection, combined treatment with Cu and Cd resulted in reproductive dysfunction and metal ion imbalance in tilapia. Transcriptomic profiling revealed differential gene annotation concentrated in the MAPK signaling pathway and regulation of GTPase activity. Post-injection, all concentrations of LHRH-α and HCG groups showed an upregulated gonadosomatic index (G.S.I) and higher levels of vitellogenin (VTG), gonadotropin-releasing hormone (GnRH), gonadotropin (GTH), and estrogen (E2) in serum compared to the negative control group. Transcriptomic analysis revealed alterations in various ovarian signaling pathways, preliminarily revealing the in vivo molecular mechanisms and differences in LHRH-α and HCG. The findings from this study could help us better understand how to counteract the effects of combined Cu and Cd exposure on tilapia ovarian development, which has significant implications for the Nile tilapia aquaculture industry.

The development of severe multidrug-resistant bacterial infections has recently intensified because of the COVID-19 pandemic. According to the guidelines issued by the World Health Organization (WHO), routine antibiotic administration is not recommended for patients with supposed or confirmed mild SARS-CoV-2 infection or pneumonia, unless bacterial infection is clinically suspected. However, recent studies have pointed out that the proportion of non-essential antibiotic use in patients infected with SARS-CoV-2 remains high. Therefore, the silent pandemic of antibiotic resistance remains a pressing issue regardless of the present threats presented by the COVID-19 pandemic. To prevent or delay entry into the postulated post-antibiotic era, the long-term advocacy for the rational use of antibiotics, the optimization of infection control procedures, and the development of new antibacterial agents and vaccines should be underscored as vital practices of the antibacterial toolbox. Recently, the development of vaccines and monoclonal antibodies has gradually received attention following the advancement of biotechnology as well as enhanced drug discovery and development in cancer research. Although decent progress has been made in laboratory-based research and promising results have been obtained following clinical trials of some of these products, challenges still exist in their widespread clinical applications. This article describes the current advantages of antibacterial monoclonal antibodies, the development of associated clinical trials, and some perceived future perspectives and challenges. Further, we anticipate the development of more therapeutic agents to combat drug-resistant bacterial infections as well as to increase the resilience of current or novel agents/strategies.

Effective and economical processes for the advanced treatment of coking wastewater were urgently needed to reduce the persistent organic pollutants of external drainage. In the present work, we investigated the degradation of organic pollutants in coking wastewater through IHC/FO (imping stream hydrodynamic cavitation (IHC) coupled with the Fenton oxidation (FO) process) and IHC alone for their feasibility in the advanced treatment of coking wastewater. To select the optimum parameters, attention was paid to the effects of main operation conditions including inlet fluid pressure, medium temperature, initial pH, reaction time, and initial Fe(II) and initial H2O2 concentrations. The results showed that the effects of conditions that need energy to be maintained (such as initial pH and inlet pressure) on the organic pollutant removal efficiency through IHC/FO were less pronounced than those through IHC alone. Moreover, the application of IHC/FO could remove more organic pollutants from coking wastewater than IHC even at an energy-efficient condition. For example, the highest COD removal efficiency of 12.5% was achieved in the IHC treatment at 0.4 MPa, pH 3, and 60 min for the reaction time. In the case of IHC/FO, the maximum COD removal of 33.2% was obtained at pH 7, 0.1 MPa, 12 mmol/L H2O2, and 3 mmol/L Fe2+ after reacting for 15 min. The ultraviolet and visible spectrophotometry (UV-Vis) absorption spectra and gas chromatography and mass spectrometry (GC–MS) analysis further revealed that the kinds and amounts of pollutants (especially those that had benzenes) remaining in water treated through IHC/FO were much fewer and smaller than in water treated through IHC alone. The better performances of IHC/FO than IHC alone were likely related to the more hydroxyl radicals produced through IHC/FO. Taken together, our findings indicate that IHC/FO has great application potential in the advanced treatment of coking wastewater.

B0 field inhomogeneity is a long-lasting issue for Cardiac MRI (CMR) in high-field (3T and above) scanners. The inhomogeneous B0 fields can lead to corrupted image quality, prolonged scan time, and false diagnosis. B0 shimming is the most straightforward way to improve the B0 homogeneity. However, today’s standard cardiac shimming protocol requires manual selection of a shim volume, which often falsely includes regions with large B0 deviation (e.g., liver, fat, and chest wall). The flawed shim field compromises the reliability of high-field CMR protocols, which significantly reduces the scan efficiency and hinders its wider clinical adoption. This study aims to develop a dual-channel deep learning model that can reliably contour the cardiac region for B0 shim without human interaction and under variable imaging protocols. By utilizing both the magnitude and phase information, the model achieved a high segmentation accuracy in the B0 field maps compared to the conventional single-channel methods (Dice score: 2D-mag = 0.866, 3D-mag = 0.907, and 3D-mag-phase = 0.938, all p < 0.05). Furthermore, it shows better generalizability against the common variations in MRI imaging parameters and enables significantly improved B0 shim compared to the standard method (SD(B0Shim): Proposed = 15 ± 11% vs. Standard = 6 ± 12%, p < 0.05). The proposed autonomous model can boost the reliability of cardiac shimming at 3T and serve as the foundation for more reliable and efficient high-field CMR imaging in clinical routines.

A bilinear map whose domain and target sets are identical is called a self-bilinear map. Original self-bilinear maps are defined over cyclic groups. Since the map itself reveals information about the underlying cyclic group, the Decisional Diffie–Hellman Problem (DDH) and the computational Diffie–Hellman (CDH) problem may be solved easily in some specific groups. This brings a lot of limitations to constructing secure self-bilinear schemes. As a compromise, a self-bilinear map with auxiliary information was proposed in CRYPTO’2014. In this paper, we construct this weak variant of a self-bilinear map from generic sets and indistinguishable obfuscation. These sets should own several properties. A new notion, One Way Encoding System (OWES), is proposed to summarize these properties. The new Encoding Division Problem (EDP) is defined to complete the security proof. The OWES can be built by making use of one level of graded encoding systems (GES). To construct a concrete self-bilinear map scheme, Garg, Gentry, and Halvei(GGH13) GES is adopted in our work. Even though the security of GGH13 was recently broken by Hu et al., their algorithm does not threaten our applications. At the end of this paper, some further considerations for the EDP for concrete construction are given to improve the confidence that EDP is indeed hard.

Chlorophyll content reflects plants’ photosynthetic capacity, growth stage, and nitrogen status and is, therefore, of significant importance in precision agriculture. This study aims to develop a spectral and color vegetation indices-based model to estimate the chlorophyll content in aquaponically grown lettuce. A completely open-source automated machine learning (AutoML) framework (EvalML) was employed to develop the prediction models. The performance of AutoML along with four other standard machine learning models (back-propagation neural network (BPNN), partial least squares regression (PLSR), random forest (RF), and support vector machine (SVM) was compared. The most sensitive spectral (SVIs) and color vegetation indices (CVIs) for chlorophyll content were extracted and evaluated as reliable estimators of chlorophyll content. Using an ASD FieldSpec 4 Hi-Res spectroradiometer and a portable red, green, and blue (RGB) camera, 3600 hyperspectral reflectance measurements and 800 RGB images were acquired from lettuce grown across a gradient of nutrient levels. Ground measurements of leaf chlorophyll were acquired using an SPAD-502 m calibrated via laboratory chemical analyses. The results revealed a strong relationship between chlorophyll content and SPAD-502 readings, with an R2 of 0.95 and a correlation coefficient (r) of 0.975. The developed AutoML models outperformed all traditional models, yielding the highest values of the coefficient of determination in prediction (Rp2) for all vegetation indices (VIs). The combination of SVIs and CVIs achieved the best prediction accuracy with the highest Rp2 values ranging from 0.89 to 0.98, respectively. This study demonstrated the feasibility of spectral and color vegetation indices as estimators of chlorophyll content. Furthermore, the developed AutoML models can be integrated into embedded devices to control nutrient cycles in aquaponics systems.

Grouper aquaculture is a rapidly growing and dynamic industry within the Taiwan aquaculture sector that serves as an important source of income generation. Farming fishes at a better location (FFABL) is a promising production strategy. This study evaluated the production and economic performance of grouper farming in Taiwan. This investigation employed two steps: (1) a cost–benefit analysis to determine the main component affecting profitability; and (2) multivariate statistics to clarify the relationship between biological and economic factors. The findings suggest that the best choice of species is hybrid giant grouper that generates and highlights a positive income, viable benefit–cost ratio and profit rate. The study found the feed management of hybrid giant groupers requires more consideration due to its low feed conversion ratio. The producers need to replace trash fish in commercial feed during the production cycle to ensure better feed management. Finally, this paper argues that feed is the main constraint for grouper farming, which is sensitive to any change in feed productivity, and this constraint must be solved to sustain the grouper industry in Taiwan.

Titanium dioxide is a compound that is used in the food, cosmetic, and paint industries; however, it is still toxic to humans and the environment. This study determined the toxicities of titanium dioxide nanoparticles (TiO2 NPs) in a Caenorhabditis elegans (C. elegans) model. The effects of commercially available (C-TiO2) and synthetically (S-TiO2) prepared TiO2 NP solutions on lethality, lifespan, growth, reproduction, locomotion, and gene expression were studied in C. elegans. Exposure to TiO2 NPs (0.0, 0.01, 0.1, 1.0, and 10 mg/L) did not result in any change to the survival rate or body length of the nematodes, regardless of the concentration. However, there was a decrease in the reproduction (brood size) and locomotion (body bending and head thrashing) of the nematodes as the TiO2 NP concentration increased. The longevity of the nematodes was shortened following TiO2 NP exposure. The gene expression of sod-1, sod-3, ctl-1, ctl-2, cyp35A2, mlt-1, and mlt-2 in the nematodes showed that there was an overexpression of all genes when the worms were exposed to 1 mg/L C-TiO2 or 10 mg/L S-TiO2. It was therefore concluded that compared with S-TiO2, C-TiO2 possibly causes more toxicity or genotoxicity in the C. elegans model.

The applications of image classification are useful, especially in the prediction of object categories at a high accuracy rate. The technique helps industrial designers classify the existing products by product appearance and assess the extent to which newly designed products belong to their brand or product family. By doing this, designers classify products by typicality. Yet, the application of image classification of products and brand styles based on typicality is still limited. Thus, we presented a case study in which the extent to which a chair was recognized as a typical Apple product was predicted. In the technique, Waikato Environment for Knowledge Analysis (WEKA) was used. The results show that the industrial designer can use product typicality for the image classification of products to manage the style of a brand and its products.

To evaluate the potential of a bacterial strain as a fungal disease control agent and plant growth promoter, its inhibitory effects on phytopathogens such as Bipolaris sorokiniana, Botrytis cinerea, Colletotrichum capsici, Fusarium graminearum, F. oxysporum, Neocosmospora rubicola, Rhizoctonia solani, and Verticillium dahliae were investigated. The results showed that the inhibitory rates in dual-culture and sterile filtrate assays against these eight phytopathogens ranged from 57% to 83% and from 36% to 92%. The strain was identified as Bacillus velezensis based on morphological and physiological characterization as well as phylogenetic analyses of 16S rRNA and the gyrase subunit A protein (gyrA) regions. The results demonstrated that B. velezensis was able to produce fungal cell-wall-degrading enzymes, namely, protease, cellulase, and β-1,3-glucanase, and the growth-promotion substances indole-3-acetic acid (IAA) and siderophore. Furthermore, B. velezensis BV01 had significant control effects on wheat root rot and pepper Fusarium wilt in a greenhouse. Potted growth-promotion experiments displayed that BV01 significantly increased the height, stem diameter, and aboveground fresh and dry weights of wheat and pepper. The results imply that B. velezensis BV01, a broad-spectrum biocontrol bacterium, is worth further investigation regarding its practical applications in agriculture.

Si is a highly promising anode material due to its superior theoretical capacity of up to 3579 mAh/g. However, it is worth noting that Si anodes experience significant volume expansion (>300%) during charging and discharging. Due to the weak adhesion between the anode coating and the smooth Cu foil current collector, the volume-expanded Si anode easily peels off, thus damaging anode cycling performance. In the present study, a femtosecond laser with a wavelength of 515 nm is used to texture Cu foils with a hierarchical microstructure and nanostructure. The peeling and cracking phenomenon in the Si anode are successfully reduced, demonstrating that volume expansion is effectively mitigated, which is attributed to the high specific surface area of the nanostructure and the protection of the deep-ablated microgrooves. Moreover, the hierarchical structure reduces interfacial resistance to promote electron transfer. The Si anode achieves improved cycling stability and rate capability, and the influence of structural features on the aforementioned performance is studied. The Si anode on the 20 μm-thick Cu current collector with a groove density of 75% and a depth of 15 μm exhibits a capacity of 1182 mAh/g after 300 cycles at 1 C and shows a high-rate capacity of 684 mAh/g at 3 C.

Elderly users are unable to adapt to the technological dividend brought by the internet of everything as soon as possible due to the deterioration in cognitive and perceptual functions, especially in the state of development of modern intelligent homes whose users’ practical operational capabilities cannot be matched. This situation creates a digital divide in which older users also need helpmate intelligent home systems. Current research on innovative home systems often lacks a focus on the elderly and a matching mapping between smart homes and operational capabilities. Therefore, this study proposes a theoretical approach and model based on the matching between smart home functions and older users’ abilities. This modelling path provides specific guidance for actual smart home design practices. This study outlines the methodology, theoretical derivation, and construction of the user capability gradient for matching functions and requirements of elderly users. Based on a theoretical model, experimental data, and threshold law in practical application, the age-appropriate intelligent home control system is developed independently, and the integrated development of hardware and software cloud synergy is realised for the user pain points of elderly users to make up for the gap at this research level.

(1) Background: Klotho is a multifunctional protein, but its biophysiological roles during fish early development are unclear. (2) Methods: We carried out anti-sense Klotho morpholino injection and whole-mount in situ hybridization experiments in this study. (3) Results: Our results showed that in the Klotho MO1-injected group, embryos displayed longitudinal shapes and swelling yolks compared to those of the un-injected groups. Their length/width ratio by 9 hpf for the uninfected group is 1 ± 0.1; but for 0.1 mM-, 0.2 mM-, and 0.5 mM-MO-injected are 1.1 ± 0.1, 1.2 ± 0.1, and 1.3 ± 0.1, respectively, suggesting that the klotho knockdown-induced length/width ratio changes are in a dose-dependent manner. Whole-mount in situ hybridization showed that several cell migration-related gene expressions, including goosecoid, floating head, bone morphogenetic protein 4, and even-skipped-liked 1 were affected in the Klotho MO-injected embryos. (4) Conclusions: We suggest that knockdown of klotho leads to cell movement impairment during zebrafish gastrulation.

Wolfberry (Lycium barbarum L.) is a small Solanaceae shrub with medicinal and edible homology, and widely used as ethnobotanical medicine and nutraceutical food. The wolfberry fruits mainly have red, purple, and yellow phenotypes. Wolfberries are rich in flavonoids, which are natural water-soluble pigments that endow a variety of colors in plants There are very few investigations on mechanism of flavonoids biosynthesis and fruit coloring reported about wolfberry. The widely targeted metabolome and transcriptome analysis were performed to obtain metabolite and gene expression profiles of red, yellow, and purple wolfberries and to explain the underlying molecular mechanism of the color differences in wolfberry. As result, metabolomics analysis revealed that the bluish anthocyanins Malvidin and petunidin trended to accumulate in purple wolf-berry, while red and yellow wolfberries trended to accumulate more yellowish flavonoids. And transcriptome analysis showed that flavonoid synthesis-related genes, such as CHS, F3H, ANS and DFR, and several MYB and bHLH genes were differentially expressed among wolfberries in different colors: most of them were more highly expressed in purple wolfberries than in red and yellow ones. In conclusion, the different flavonoids’ accumulation patterns may result in the different fruit colors of wolfberry, and the MYB or bHLH transcription factors could regulate the expression of flavonoids biosynthesis related genes to change the composition of flavonoids or anthocyanins in wolfberry fruits and result in varied fruit colors. These findings provide new insights into the underlying molecular mechanism of the fruit color differences in wolfberry and provide new ideas for molecular breeding of wolfberry.

This paper focuses on investigating the bifurcation of limit cycles and centers within a specific class of three-dimensional cubic systems possessing Z3-equivariant symmetry. By calculating the singular point values of the systems, we obtain a necessary condition for a singular point to be a center. Subsequently, the Darboux integral method is employed to demonstrate that this condition is also sufficient. Additionally, we demonstrate that the system can bifurcate 15 small amplitude limit cycles with a distribution pattern of 5−5−5 originating from the singular points after proper perturbation. This finding represents a novel contribution to the understanding of the number of limit cycles present in three-dimensional cubic systems with Z3-equivariant symmetry.

The present study introduces an optimized overlay target design to minimize the overlay error caused by asymmetric sidewall structures in semiconductor manufacturing. To achieve this goal, the overlay error formula was derived by separating the asymmetric bottom grating structure into symmetric and asymmetric parts. Based on this formula, it was found that the overlay target design with the linewidth of the bottom grating closed to the grating period could effectively reduce the overlay error caused by the sidewall asymmetry structure. Simulation results demonstrate that the proposed design can effectively control the measurement error of different wavelengths within ±0.3 nm, even under varying sidewall angles and film thicknesses. Overall, the proposed overlay target design can significantly improve the overlay accuracy in semiconductor manufacturing processes.

We numerically and experimentally investigated the behavior of high-frequency underwater ultrasounds reflected by gradient acoustic metasurfaces. Metasurfaces were fabricated with a periodic array of gradient slits along the surface of a steel specimen. The finite element method was adopted for the acoustics–structure interaction problem to design the metasurfaces and simulate the reflected fields of the incident ultrasound. Our metasurfaces yielded anomalous reflection, specular reflection, apparent negative reflection, and radiation of surface-bounded modes for ultrasonic waves impinging on the metasurfaces at different incident angles. The occurrence of these reflection behaviors could be explained by the generalized Snell’s law for a gradient metasurface with periodic supercells. We showed that at some incident angles, strong anomalous reflection could be generated, which could lead to strong retroreflection at specific incident angles. Furthermore, we characterized the time evolution of the reflections using pulsed ultrasound. The simulated transient process revealed the formation of propagating reflected ultrasound fields. The experimentally measured reflected ultrasound signals verified the distinct reflection behaviors of the metasurfaces; strong anomalous reflection steering the ultrasound pulse and causing retroreflection was observed. This study paves the way for designing underwater acoustic metasurfaces for ultrasound imaging and caustic engineering applications using pulsed ultrasound in the high-frequency regime.

Leaf scald caused by Xanthomonas albilineans (Xa) is a major bacterial disease in sugarcane that represents a threat to the global sugar industry. Little is known about the population structure and genetic evolution of this pathogen. In this study, 39 Xa strains were collected from 6 provinces in China. Of these strains, 15 and 24 were isolated from Saccharum spp. hybrid and S. officinarum plants, respectively. Based on multilocus sequence analysis (MLSA), with five housekeeping genes, these strains were clustered into two distinct phylogenetic groups (I and II). Group I included 26 strains from 2 host plants, Saccharum spp. hybrid and S. officinarum collected from 6 provinces, while Group II consisted of 13 strains from S. officinarum plants in the Zhejiang province. Among the 39 Xa strains, nucleotide sequence identities from 5 housekeeping genes were: ABC (99.6–100%), gyrB (99.3–100%), rpoD (98.4–100%), atpD (97.0–100%), and glnA (97.6–100%). These strains were clustered into six groups (A–F), based on the rep-PCR fingerprinting, using primers for ERIC2, BOX A1R, and (GTG)5. UPGMA and PCoA analyses revealed that group A had the most strains (24), followed by group C with 11 strains, while there was 1 strain each in groups B and D–F. Neutral tests showed that the Xa population in S. officinarum had a trend toward population expansion. Selection pressure analysis showed purification selection on five concatenated housekeeping genes from all tested strains. Significant genetic differentiation and infrequent gene flow were found between two Xa populations hosted in Saccharum spp. hybrids and S. officinarum. Altogether, these results provide evidence of obvious genetic divergence and population structures among Xa strains from China.

The emergence of carbapenem-resistant Klebsiella pneumoniae poses a significant threat to public health. In this study, we aimed to investigate the distribution and genetic diversity of plasmids carrying beta-lactamase resistance determinants in a collection of carbapenem-resistant K. pneumoniae blood isolates. Blood isolates of carbapenem-resistant K. pneumoniae bacteremia were collected and identified. Whole-genome sequencing, assembly and analysis were performed for the prediction of antimicrobial resistance determinants. Plasmidome analysis was also performed. Our plasmidome analysis revealed two major plasmid groups, IncFII/IncR and IncC, as key players in the dissemination of carbapenem resistance among carbapenem-resistant K. pneumoniae. Notably, plasmids within the same group exhibited conservation of encapsulated genes, suggesting that these plasmid groups may serve as conservative carriers of carbapenem-resistant determinants. Additionally, we investigated the evolution and expansion of IS26 integrons in carbapenem-resistant K. pneumoniae isolates using long-read sequencing. Our findings revealed the evolution and expansion of IS26 structure, which may have contributed to the development of carbapenem resistance in these strains. Our findings indicate that IncC group plasmids are associated with the endemic occurrence of carbapenem-resistant K. pneumoniae, highlighting the need for targeted interventions to control its spread. Although our study focuses on the endemic presence of carbapenem-resistant K. pneumoniae, it is important to note that carbapenem-resistant K. pneumoniae is indeed a global problem, with cases reported in multiple regions worldwide. Further research is necessary to better understand the factors driving the worldwide dissemination of carbapenem-resistant K. pneumoniae and to develop effective strategies for its prevention and control.

The aim of this study was to propose a finite element method based numerical approach for evaluating various hallux valgus treatment strategies. We developed three-dimensional hallux valgus deformity models, with different metatarsal osteotomy methods and Kirschner wire fixation strategies, under two types of standing postures. Ten Kirschner wire fixations were analyzed and compared. The fixation stability, bone stress, implant stress, and contact pressure on the osteotomy surface were calculated as the biomechanical indexes. The results showed that the biomechanical indexes of the osteotomy and Kirschner wire fixations for hallux valgus deformity could be effectively analyzed and fairly evaluated. The distal metatarsal osteotomy method provided better biomechanical indexes compared to the proximal metatarsal osteotomy method. This study proposed a finite element method based numerical approach for evaluating various osteotomy and Kirschner wire fixations for hallux valgus deformity before surgery.

We demonstrate a narrow linewidth 980 nm laser by self-injection locking of an electrically pumped distributed-feedback (DFB) laser diode to a high quality (Q) factor (>105) lithium niobate (LN) microring resonator. The lithium niobate microring resonator is fabricated by photolithography-assisted chemo-mechanical etching (PLACE) technique, and the Q factor of lithium niobate microring is measured as high as 6.91 × 105. The linewidth of the multimode 980 nm laser diode, which is ~2 nm measured from its output end, is narrowed down to 35 pm with a single-mode characteristic after coupling with the high-Q LN microring resonator. The output power of the narrow-linewidth microlaser is about 4.27 mW, and the wavelength tuning range reaches 2.57 nm. This work explores a hybrid integrated narrow linewidth 980 nm laser that has potential applications in high-efficient pump laser, optical tweezers, quantum information, as well as chip-based precision spectroscopy and metrology.

Global travel and climate change have drastically increased the number of countries with endemic or epidemic dengue. The largest dengue outbreak in Taiwan, with 43,419 cases and 228 deaths, occurred in 2015. Practical and cost-effective tools for early prediction of clinical outcomes in dengue patients, especially the elderly, are limited. This study identified the clinical profile and prognostic indicators of critical outcomes in dengue patients on the basis of clinical parameters and comorbidities. A retrospective cross-sectional study was conducted in a tertiary hospital from 1 July 2015 to 30 November 2015. Patients diagnosed with dengue were enrolled, and the initial clinical presentations, diagnostic laboratory data, details of the underlying comorbidities, and initial management recommendations based on 2009 World Health Organization (WHO) guidelines were used to evaluate prognostic indicators of critical outcomes in dengue patients. Dengue patients from another regional hospital were used to evaluate accuracy. A group B (4 points) classification, temperature < 38.5 °C (1 point), lower diastolic blood pressure (1 point), prolonged activated partial thromboplastin time (aPTT) (2 points), and elevated liver enzymes (1 point) were included in the scoring system. The area under the receiver operating characteristic curve of the clinical model was 0.933 (95% confidence interval [CI]: 0.905–0.960). The tool had good predictive value and clinical applicability for identifying patients with critical outcomes.
