Acorn size is of great importance, both ecologically and economically. However, the long lifespan is an obstacle to improvement in breeding. Keumsura1ho is a recently bred cultivar of oak (Quercus acutissima Carruth) selected after eight years of selection due to its larger acorns and higher weight compared to the control. In the present study, we investigated the transcriptional mechanisms underlying the outstanding morphological characteristics of Keumsura1ho. For this purpose, one- and two-year-old acorns from the control and Keumsura1ho groups were used for comparative transcriptome analyses of their seeds. Among morphological characteristics, the Keumsura1ho seeds were 18.3% longer than those of the control. Seed width and depth were 33.9% greater than those of the control. In the transcriptome analysis, genes related to seed size were further enriched, with biennial Keumsura1ho showing higher enrichment in comparison to control genes. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that the upregulated genes of the two-year-old Keumsura1ho seeds showed higher enrichment in phenylpropanoid metabolism compared to the control, with OMT1 and PRXs exhibiting high expression levels. This pathway has a significant impact on seed coat lignification and size in Keumsura1ho. The results of this study will benefit the development of breeding techniques by identifying marker genes for seed size.

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is crucial in plant metabolism and responses to various abiotic stresses. In the glycolysis pathway, glyceraldehyde-3-phosphate (G3P) is oxidized to 1,3-bisphosphate glycerate (1,3-BPG) through the catalytic action of GAPDH. However, the GAPDH gene family in Quercus rubra has been minimally researched. In this study, we identified 13 GAPDH-encoding genes in Q. rubra through a bioinformatics analysis of genomic data. Evolutionary studies suggest that these QrGAPDH genes are closely related to those in Glycine max and Triticum aestivum. We conducted a comprehensive whole-genome study, which included predictions of subcellular localization, gene structure analysis, protein motif identification, chromosomal placement, and analysis of cis-acting regions. We also examined the expression of GAPDH proteins and genes in various tissues of Q. rubra and under drought stress. The results indicated diverse expression patterns across different tissues and differential expression under drought conditions. Notably, the expression of Qurub.02G290300.1, Qurub.10G209800.1, and Qrub.M241600.1 significantly increased in the leaf, stem, and root tissues under drought stress. This study provides a systematic analysis of QrGAPDH genes, suggesting their pivotal roles in the drought stress response of trees.

Drought stress affects plant productivity by altering plant responses at the morphological, physiological, and molecular levels. In this study, we identified physiological and genetic responses in Populus alba × Populus glandulosa hybrid clones 72-30 and 72-31 after 12 days of exposure to drought treatment. After 12 days of drought treatment, glucose, fructose, and sucrose levels were significantly increased in clone 72-30 under drought stress. The Fv/Fo and Fv/Fm values in both clones also decreased under drought stress. The changes in proline, malondialdehyde, and H2O2 levels were significant and more pronounced in clone 72-30 than in clone 72-31. The activities of antioxidant-related enzymes, such as catalase and ascorbate peroxidase, were significantly higher in the 72-31 clone. To identify drought-related genes, we conducted a transcriptomic analysis in P. alba × P. glandulosa leaves exposed to drought stress. We found 883 up-regulated and 305 down-regulated genes in the 72-30 clone and 279 and 303 in the 72-31 clone, respectively. These differentially expressed genes were mainly in synthetic pathways related to proline, abscisic acid, and antioxidants. Overall, clone 72-31 showed better drought tolerance than clone 72-30 under drought stress, and genetic changes also showed different patterns.

Plants produce two phosphorylated enzymes: pyrophosphate-dependent fructose-6-phosphate phosphotransferase (PFP) and ATP-dependent phosphofructokinase (PFK). However, the characterization of phosphofructokinase (PFK) family members and their functions remains unexplored in Eastern Cottonwood (Populus deltoides). This study comprehensively analyzed 17 phosphofructokinase genes in the P. deltoides genome, including their gene structure, conserved domains, evolutionary relationships, and expression patterns. Genome-wide analysis of the poplar genome identified 11 genes belonging to PdPFK and 6 genes belonging to PdPFP. Phylogenetic, gene structure, and motif analyses revealed two main categories of PFK genes: PFK with eleven genes and PFP with six genes. Quantitative PCR (qPCR) analysis demonstrated that all 17 phosphofructokinase genes were induced in various tissues and developmental stages of P. deltoides. PFPs were strongly expressed throughout P. deltoides’ growth and development, notably in the roots, according to the transcript analysis. This genome-wide analysis provides insights into PFK and PFP genes and establishes a foundation for future investigations into the role of genes in P. deltoids.

Physiological response and transcriptome changes were observed to investigate the effects on the growth, metabolism and genetic changes of Pinus densiflora grown for a long time in an environment with an elevated atmospheric CO2 concentration. Pine trees were grown at ambient (400 ppm) and elevated (560 ppm and 720 ppm) CO2 concentrations for 10 years in open-top chambers. The content of nonstructural carbohydrates was significantly increased in elevated CO2. It was notable that the contents of chlorophylls significantly decreased at an elevated CO2. The activities of antioxidants were significantly increased at an elevated CO2 concentration of 720 ppm. We analyzed the differences in the transcriptomes of Pinus densiflora at ambient and elevated CO2 concentrations and elucidated the functions of the differentially expressed genes (DEGs). RNA-Seq analysis identified 2415 and 4462 DEGs between an ambient and elevated CO2 concentrations of 560 ppm and 720 ppm, respectively. Genes related to glycolysis/gluconeogenesis and starch/sucrose metabolism were unchanged or decreased at an elevated CO2 concentration of 560 ppm and tended to increase at an elevated CO2 concentration of 720 ppm. It was confirmed that the expression levels of genes related to photosynthesis and antioxidants were increased at an elevated CO2 concentration of 720 ppm.

It is difficult to accurately simulate the cornering performance of a vehicle using a simplified vehicle model; therefore, a proper evaluation process should be conducted from an experimental perspective. In addition, there are several possible combinations of subparts in suspension modules, such as springs, dampers, stabilizers, or other parts, and the characteristic identification of each part may require significant costs and time. In this study, the cornering performance of a vehicle system was experimentally evaluated using several combinations of subparts in the suspension module as well as candidate tire cases. All characteristic parameters used in the two indices were previously identified as separate steps, and vehicle tests were performed according to all the required test scenarios for cornering performance. The evaluation of the cornering performance of a vehicle was proposed using the suspension characteristic cornering performance (SCCP) index and suspension and tire characteristic cornering performance based-on-tests (STCT) index, respectively, using measured data from a vehicle test. The STCT index is revised from the SCCP index by considering the tires’ effects, and the optimal selection of the suspension and tires can be obtained from the proposed indices. The feasibility of the proposed STCT index was verified by evaluating the cornering performance from all the prepared test results.

Global climate change is heavily affected by an increase in CO2. As one of several efforts to cope with this, research on poplar, a representative, fast growing, and model organism in plants, is actively underway. The effects of elevated atmospheric CO2 on the metabolism, growth, and transcriptome of poplar were investigated to predict productivity in an environment where CO2 concentrations are increasing. Poplar trees were grown at ambient (400 ppm) or elevated CO2 concentrations (1.4× ambient, 560 ppm, and 1.8× ambient, 720 ppm) for 16 weeks in open-top chambers (OTCs). We analyzed the differences in the transcriptomes of Populusalba × Populus glandulosa clone “Clivus” and Populus euramericana clone “I-476” using high-throughput sequencing techniques and elucidated the functions of the differentially expressed genes (DEGs) using various functional annotation methods. About 272,355 contigs and 207,063 unigenes were obtained from transcriptome assembly with the Trinity assembly package. Common DEGs were identified which were consistently regulated in both the elevated CO2 concentrations. In Clivus 29, common DEGs were found, and most of these correspond to cell wall proteins, especially hydroxyproline-rich glycoproteins (HRGP), or related to fatty acid metabolism. Concomitantly, in I-476, 25 were identified, and they were related to heat shock protein (HSP) chaperone family, photosynthesis, nitrogen metabolism, and carbon metabolism. In addition, carbohydrate contents, including starch and total soluble sugar, were significantly increased in response to elevated CO2. These data should be useful for future gene discovery, molecular studies, and tree improvement strategies for the upcoming increased-CO2 environments.

Cinnamyl alcohol dehydrogenase (CAD) is involved in the final step of the phenylpropanod pathway, catalyzing the NADPH-dependent reduction of hydroxy-cinnamaldehydes into the corresponding alcohols. The rice genome contains twelve CAD and CAD-like genes, collectively called OsCADs. To elucidate the biochemical function of the OsCADs, OsCAD1, 2, 6, and 7, which are highly expressed in rice, were cloned from rice tissues. The cloned OsCADs were heterologously expressed in Escherichia coli as His-tag fusion proteins. The activity assay of the recombinant OsCADs showed that OsCAD2, 6, and 7 have CAD activity toward hydroxycinnamaldehydes, but OsCAD1 has no detectable catalytic activity. The kinetic parameters of the enzyme reactions demonstrated that OsCAD2 has the highest catalytic activity among the examined enzymes. This result agrees well with the finding that the Zn binding and NADPH binding motifs and the residues constituting the substrate binding pocket in bona fide plant CADs were fully conserved in OsCAD2. Although they have large variations in the residue for the substrate binding pocket, OsCAD6 and 7 catalyzed the reduction of hydroxycinnamaldehydes with a similar efficiency. Alignment of amino acid sequences showed that OsCAD1 lacks the GxxxxP motif for NADPH binding and has mismatches in residues important in the reduction process, which could be responsible for the loss of catalytic activity. OsCAD2 belongs to CAD Class I with bona fide CADs from other plant species and is constitutively expressed throughout the developmental stages of rice, with preferential expression in actively lignifying tissues such as the root, stem, and panicle, suggesting that it is mainly involved in developmental lignification in rice. The expression of OsCAD2 was also induced by biotic and abiotic stresses such as Xanthomonas oryzae pv. oryzae (Xoo) infection and UV-irradiation, suggesting that it plays a role in the defense response of rice, in addition to a bona fide role in developmental lignification. OsCAD6 and 7 belong in CAD Class II. Their expression is relatively lower than that of OsCAD2 and is confined to certain tissues, such as the leaf sheath, stem, and panicle. The expression of OsCAD6 was stimulated by Xoo infection and UV-irradiation. Thus OsCAD6 appears to be an inducible OsCAD that is likely involved in the defense response of rice against biotic and abiotic stresses.
