Invasive fungal diseases (IFDs) comprise a growing healthcare burden, especially given the expanding population of immunocompromised hosts. Early diagnosis of IFDs is required to optimise therapy with antifungals, especially in the setting of rising rates of antifungal resistance. Molecular techniques including nucleic acid amplification tests and whole genome sequencing have potential to offer utility in overcoming limitations with traditional phenotypic testing. However, standardisation of methodology and interpretations of these assays is an ongoing undertaking. The utility of targeted Aspergillus detection has been well-defined, with progress in investigations into the role of targeted assays for Candida, Pneumocystis, Cryptococcus, the Mucorales and endemic mycoses. Likewise, whilst broad-range polymerase chain reaction assays have been in use for some time, pathology stewardship and optimising diagnostic yield is a continuing exercise. As costs decrease, there is also now increased access and experience with whole genome sequencing, including metagenomic sequencing, which offers unparalleled resolution especially in the investigations of potential outbreaks. However, their role in routine diagnostic use remains uncommon and standardisation of techniques and workflow are required for wider implementation.

The reuse of waste materials has recently become appealing due to pollution and cost reduction factors. Using waste materials can reduce environmental pollution and product costs, thus promoting sustainability. Approximately 95% of calcium carbonate-containing waste eggshells end up in landfills, unused. These eggshells, a form of bio-waste, can be repurposed as catalytic electrode material for various applications, including supercapacitors, after being converted into CaO. Similarly, used waste battery electrode materials pose environmental hazards if not properly recycled. Various types of batteries, particularly lithium-ion batteries, are extensively used worldwide. The recycling of used lithium-ion batteries has become less important considering its low economic benefits. This necessitates finding alternative methods to recover and reuse the graphite rods of spent batteries. Therefore, this study reports the conversion of waste eggshell into calcium oxide by high-temperature calcination and extraction of nanographite from spent batteries for application in energy storage fields. Both CaO and CaO/graphite were characterized for their structural, morphological, and chemical compositions using XRD, SEM, TEM, and XPS techniques. The prepared CaO/graphite nanocomposite material was evaluated for its efficiency in electrochemical supercapacitor applications. CaO and its composite with graphite powder obtained from used lithium-ion batteries demonstrated improved performance compared to CaO alone for energy storage applications. Using these waste materials for electrochemical energy storage and conversion devices results in cheaper, greener, and sustainable processes. This approach not only aids in energy storage but also promotes sustainability through waste management by reducing landfills.

This study analyzes the dynamics of surface melting in Antarctica, which are crucial for understanding glacier and ice sheet behavior and monitoring polar climate change. Specifically, we focus on the Nivlisen ice shelf in East Antarctica, examining melt ponds, supra glacial lakes (SGLs), seasonal surface melt extent, and surface ice flow velocity. Spatial and temporal analysis is based on Landsat and Sentinel-1 data from the austral summers of 2000 to 2023. Between 2000 and 2014, melt ponds and SGLs on the ice shelf covered roughly 1 km2. However, from 2015 to 2023, surface melting increased consistently, leading to more extensive melt ponds and SGLs. Significant SGL depths were observed in 2016, 2017, 2019, and 2020, with 2008, 2016, and 2020 showing the highest volumes and progressive SGL area growth. We also examined the relationship between seasonal surface melt extent and ice flow velocity. Validation efforts involved ground truth data from a melt pond in central Dronning Maud Land (cDML) during the 2022–2023 austral summer, along with model-based results. The observed increase in melt pond depth and volume may significantly impact ice shelf stability, potentially accelerating ice flow and ice shelf destabilization. Continuous monitoring is essential for accurately assessing climate change’s ongoing impact on Antarctic ice shelves.

This paper presents an experimental study utilizing the vision-aided inertial navigation system (VaINS) on a fixed-wing unmanned aerial vehicle (UAV). VaINS operates as a data-logging box, collecting sensor data, processing them, and transmitting the navigation solution to an operator terminal in real time. The flight data contain buildings, vegetation, a river, a lake, and mountains. The camera’s pitch angle was tested at both a grazing angle of 22° and a nadir angle of 90°. The aim of this experiment was twofold: to explore the application of VaINS on a fixed-wing UAV and to assess the performance of the image-aided navigation system. The image-aided inertial navigation system (INS) solution was compared with a reference solution during GNSS-denied time intervals. The camera’s pitch angle at 90° exhibited a slight advantage over the grazing angle at 22°. Moreover, in the linear flight segments, the image-aided navigation system demonstrated significantly better performance compared to the free inertial solution during the GNSS-denied periods.

This study aimed to optimize machining parameters to obtain better surface roughness and remnant depth ratio values under dry turning of NiTi-shape memory alloy (SMA). During the turning experiments, various machining parameters were used, including three different cutting speeds vc (105, 144, and 200 m/min), three different feed rates f (0.05, 0.1, and 0.15 mm/rev), and three different depths of cut ap (0.1, 0.15, and 0.2 mm). The effects of machining parameters in turning experiments were investigated on the response surface methodology (RSM) with Box–Behnken design (BBD) using the Design Expert 11; how the cutting parameters affect the surface quality is discussed in detail. In this context, the cutting parameters were successfully optimized using a genetic algorithm (GA). The optimized processing parameters are vc = 126 m/min, f = 0.11 mm/rev, ap = 0.14 mm, resulting in surface roughness and remnant depth ratio values of 0.489 μm and 64.13%, respectively.

Glioblastoma (GBM) mesenchymal (MES) transition can be regulated by long non-coding RNAs (lncRNAs) via modulation of various factors (Epithelial-to-Mesenchymal (EMT) markers, biological signalling, and the extracellular matrix (ECM)). However, understanding of these mechanisms in terms of lncRNAs is largely sparse. This review systematically analysed the mechanisms by which lncRNAs influence MES transition in GBM from a systematic search of the literature (using PRISMA) performed in five databases (PubMed, MEDLINE, EMBASE, Scopus, and Web of Science). We identified a total of 62 lncRNAs affiliated with GBM MES transition, of which 52 were upregulated and 10 were downregulated in GBM cells, where 55 lncRNAs were identified to regulate classical EMT markers in GBM (E-cadherin, N-cadherin, and vimentin) and 25 lncRNAs were reported to regulate EMT transcription factors (ZEB1, Snai1, Slug, Twist, and Notch); a total of 16 lncRNAs were found to regulate the associated signalling pathways (Wnt/β-catenin, PI3k/Akt/mTOR, TGFβ, and NF-κB) and 14 lncRNAs were reported to regulate ECM components (MMP2/9, fibronectin, CD44, and integrin-β1). A total of 25 lncRNAs were found dysregulated in clinical samples (TCGA vs. GTEx), of which 17 were upregulated and 8 were downregulated. Gene set enrichment analysis predicted the functions of HOXAS3, H19, HOTTIP, MEG3, DGCR5, and XIST at the transcriptional and translational levels based on their interacting target proteins. Our analysis observed that the MES transition is regulated by complex interplays between the signalling pathways and EMT factors. Nevertheless, further empirical studies are required to elucidate the complexity in this process between these EMT factors and the signalling involved in the GBM MES transition.

Mucormycosis is an uncommon, yet deadly invasive fungal infection caused by the Mucorales moulds. These pathogens are a WHO-assigned high-priority pathogen group, as mucormycosis incidence is increasing, and there is unacceptably high mortality with current antifungal therapies. Current diagnostic methods have inadequate sensitivity and specificity and may have issues with accessibility or turnaround time. Patients with diabetes mellitus and immune compromise are predisposed to infection with these environmental fungi, but COVID-19 has established itself as a new risk factor. Mucorales also cause healthcare-associated outbreaks, and clusters associated with natural disasters have also been identified. Robust epidemiological surveillance into burden of disease, at-risk populations, and emerging pathogens is required. Emerging serological and molecular techniques may offer a faster route to diagnosis, while newly developed antifungal agents show promise in preliminary studies. Equitable access to these emerging diagnostic techniques and antifungal therapies will be key in identifying and treating mucormycosis, as delayed initiation of therapy is associated with higher mortality.

Dengue infection, caused by the dengue virus, is a global threat which requires immediate attention and appropriate disease management. The current diagnosis of dengue infection is largely based on viral isolation, RT-PCR and serology-based detection, which are time-consuming and expensive, and require trained personnel. For early diagnosis of dengue, the direct detection of a dengue antigenic target is efficacious, and one such target is NS1. NS1-based detection is primarily antibody-centric and is beset by drawbacks pertaining to antibodies such as the high cost of synthesis and large batch-to-batch variation. Aptamers are potential surrogates of antibodies and are much cheaper, without exhibiting batch-to-batch variation. Given these advantages, we sought to isolate RNA aptamers against the NS1 protein of dengue virus serotype 2. A total of 11 cycles of SELEX were carried out, resulting in two potent aptamers, DENV-3 and DENV-6, with dissociation constant values estimated at 37.57 ± 10.34 nM and 41.40 ± 9.29 nM, respectively. These aptamers can be further miniaturized to TDENV-3 and TDENV-6a with an increased LOD upon their usage in direct ELASA. Moreover, these truncated aptamers are highly specific against the dengue NS1 while showing no cross-reactivity against the NS1 of the Zika virus, the E2 protein of the Chikungunya virus or the LipL32 protein of Leptospira, with target selectivity retained even in human serum. The usage of TDENV-3 as the capturing probe and TDENV-6a as the detection probe underpinned the development of an aptamer-based sandwich ELASA for the detection of dengue NS1. The sensitivity of the sandwich ELASA was further improved with the stabilization of the truncated aptamers and the repeated incubation strategy, which enabled a LOD of 2 nM when used with the target NS1 spiked in human serum diluted at 1:2000.

Stroke is one of the main causes of mortality and disability, and it is due to be included in monetary implications on wellbeing frameworks around the world. Ischemic stroke is caused by interference in cerebral blood flow, leading to a deficit in the supply of oxygen to the affected region. It accounts for nearly 80–85% of all cases of stroke. Oxidative stress has a significant impact on the pathophysiologic cascade in brain damage leading to stroke. In the acute phase, oxidative stress mediates severe toxicity, and it initiates and contributes to late-stage apoptosis and inflammation. Oxidative stress conditions occur when the antioxidant defense in the body is unable to counteract the production and aggregation of reactive oxygen species (ROS). The previous literature has shown that phytochemicals and other natural products not only scavenge oxygen free radicals but also improve the expressions of cellular antioxidant enzymes and molecules. Consequently, these products protect against ROS-mediated cellular injury. This review aims to give an overview of the most relevant data reported in the literature on polyphenolic compounds, namely, gallic acid, resveratrol, quercetin, kaempferol, mangiferin, epigallocatechin, and pinocembrin, in terms of their antioxidant effects and potential protective activity against ischemic stroke.

Guava is an important revenue generating crop for small, medium, and commercial guava cultivators all over the world. Nematode infestation is one of the factors that cause declines in fruit production. Researches have proven that the application of plant growth regulators induces the synthesis of defense-related proteins in Guava. IBA is one such plant growth regulator, and its effects on guava plants has not yet been elucidated. Thus, this research is focused on the optimization of IBA concentrations, which results in the induction and production of maximum defense-related proteins to defend against root knot nematode. The present study includes the application of IBA on M. enterolobii-infested experimental guava plants at different concentrations ranging from 100 ppm to 2000 ppm. The synthesis of defense-related proteins is identified with 1000 ppm of IBA. At this concentration, IBA influences the morphological, physiological, and biochemical characteristics and enhances the induction of defense-related proteins in M. enterolobii-infested experimental guava plants. Thus, 1000 ppm of IBA prevents nematode infestation in Lucknow-49 guava plants.

To study their physicochemical and antimicrobial properties, zinc oxide nanoparticles were synthesized using a simple chemical route and 4-dimethylaminobenzaldehyde (4DB) as an organic additive. ZnO nanoparticles were characterized with XRD analysis, which confirmed the presence of a hexagonal wurtzite structure with different crystalline sizes. The SEM morphology of the synthesized nanoparticles confirmed the presence of nanorods in both modifications of ZnO nanoparticles. EDS analysis proved the chemical composition of the synthesized samples via different chemical approaches. In addition, the optical absorption results indicated that the use of 4DB increased the band gap energy of the synthesized nanoparticles. The synthesized Zn8O8 and Zn8O8:4DB clusters were subjected to HOMO–LUMO analysis, and their ionization energy (I), electron affinity (A), global hardness (η), chemical potential (σ), global electrophilicity index (ω), dipole moment (μ), polarizability (αtot), first-order hyperpolarizability (βtot), and other thermodynamic properties were determined. Furthermore, the antimicrobial properties of the ZnO nanoparticles were studied against G+ (S. aureus and B. subtilis) and G− (K. pneumoniae and E. coli) bacteria in a nutrient agar according to guidelines of the Clinical and Laboratory Standards Institute (CLSI).

Chemical warfare agents (CWAs) have been threatening human civilization and its existence because of their rapid response, toxic, and irreversible nature. The hybrid nanostructured composites were synthesized by the hydrothermal process to detect the dimethyl methyl phosphonate (DMMP), a simulant of G-series nerve agents, especially sarin. Cellulose (CE), manganese oxide cellulose (MnO2@CE), and MnO2@CE/polypyrrole (PPy) exhibited a frequency shift of 0.4, 4.8, and 8.9 Hz, respectively, for a DMMP concentration of 25 ppm in the quartz crystal microbalance (QCM). In surface acoustic wave (SAW) sensor, they exhibited 187 Hz, 276 Hz, and 78 Hz, respectively. A comparison between CE, MnO2@CE, and MnO2@CE/PPy demonstrated that MnO2@CE/PPy possesses excellent linearity with a coefficient of determination (COD or R2) of 0.992 and 0.9547 in the QCM and SAW sensor. The hybrid composite materials showed a reversible adsorption and desorption phenomenon in the reproducibility test. The response and recovery times indicated that MnO2@CE/PPy showed the shortest response (~23 s) and recovery times (~42 s) in the case of the QCM sensor. Hence, the pristine CE and its nanostructured composites were compared to analyze the sensing performance based on sensitivity, selectivity, linearity, reproducibility, and response and recovery times to detect the simulant of nerve agents.

Chemical warfare agents (CWAs) have inflicted monumental damage to human lives from World War I to modern warfare in the form of armed conflict, terrorist attacks, and civil wars. Is it possible to detect the CWAs early and prevent the loss of human lives? To answer this research question, we synthesized hybrid composite materials to sense CWAs using hydrothermal and thermal reduction processes. The synthesized hybrid composite materials were evaluated with quartz crystal microbalance (QCM) and surface acoustic wave (SAW) sensors as detectors. The main findings from this study are: (1) For a low dimethyl methyl phosphonate (DMMP) concentration of 25 ppm, manganese dioxide nitrogen-doped graphene oxide (NGO@MnO2) and NGO@MnO2/Polypyrrole (PPy) showed the sensitivities of 7 and 51 Hz for the QCM sensor and 146 and 98 Hz for the SAW sensor. (2) NGO@MnO2 and NGO@MnO2/PPy showed sensitivities of more than 50-fold in the QCM sensor and 100-fold in the SAW sensor between DMMP and potential interferences. (3) NGO@MnO2 and NGO@MnO2/PPy showed coefficients of determination (R2) of 0.992 and 0.975 for the QCM sensor and 0.979 and 0.989 for the SAW sensor. (4) NGO@MnO2 and NGO@MnO2/PPy showed repeatability of 7.00 ± 0.55 and 47.29 ± 2.69 Hz in the QCM sensor and 656.37 ± 73.96 and 665.83 ± 77.50 Hz in the SAW sensor. Based on these unique findings, we propose NGO@MnO2 and NGO@MnO2/PPy as potential candidate materials that could be used to detect CWAs.

Today’s world requires high-performance energy storage devices such as hybrid supercapacitors (HSc), which play an important role in the modern electronic market because supercapacitors (Sc) show better electrical properties for electronics devices. In the last few years, the scientific community has focused on the coupling of Sc and battery-type materials to improve energy and power density. Recently, various hybrid electrode materials have been reported in the literature; out of these, coordination polymers such as metal-organic frameworks (MOFs) are highly porous, stable, and widely explored for various applications. The poor conductivity of classical MOFs restricts their applications. The composite of MOFs with highly porous graphene (G), graphene oxide (GO), or reduced graphene oxide (rGO) nanomaterials is a promising strategy in the field of electrochemical applications. In this review, we have discussed the strategy, device structure, and function of the MOFs/G, MOFs/GO, and MOFs/rGO nanocomposites on Sc. The structural, morphological, and electrochemical performance of coordination polymers composites towards Sc application has been discussed. The reported results indicate the considerable improvement in the structural, surface morphological, and electrochemical performance of the Sc due to their positive synergistic effect. Finally, we focused on the recent development in preparation methods optimization, and the opportunities for MOFs/G based nanomaterials as electrode materials for energy storage applications have been discussed in detail.

Arunachali yak, the only registered yak breed of India, is crucial for the economic sustainability of pastoralist Monpa community. This study intended to determine the genomic diversity and to identify signatures of selection in the breed. Previously available double digest restriction-site associated DNA (ddRAD) sequencing data of Arunachali yak animals was processed and 99,919 SNPs were considered for further analysis. The genomic diversity profiled based on nucleotide diversity, π (π = 0.041 in 200 bp windows), effective population size, Ne (Ne = 83) and Runs of homozygosity (ROH) (predominance of shorter length ROHs) was found to be optimum. Subsequently, 207 regions were identified to be under selective sweeps through de-correlated composite of multiple signals (DCMS) statistic which combined three individual test statistics viz. π, Tajima’s D and |iHS| in non-overlapping 100 kb windows. Mapping of these regions revealed 611 protein-coding genes including KIT, KITLG, CDH12, FGG, FGA, FGB, PDGFRA, PEAR1, STXBP3, olfactory receptor genes (OR5K3, OR5H6 and OR1E1) and taste receptor genes (TAS2R1, TAS2R3 and TAS2R4). Functional annotation highlighted that biological processes like platelet aggregation and sensory perception were the most overrepresented and the associated regions could be considered as breed-specific signatures of selection in Arunachali yak. These findings point towards evolutionary role of natural selection in environmental adaptation of Arunachali yak population and provide useful insights for pursuing genome-wide association studies in future.

The adult human body contains about 4 g of iron. About 1–2 mg of iron is absorbed every day, and in healthy individuals, the same amount is excreted. We describe a patient who presents with severe iron deficiency anemia with hemoglobin levels below 6 g/dL and ferritin levels below 30 ng/mL. Although red blood cell concentrates and intravenous iron have been substituted every month for years, body iron stores remain depleted. Diagnostics have included several esophago-gastro-duodenoscopies, colonoscopies, MRI of the liver, repetitive bone marrow biopsies, psychological analysis, application of radioactive iron to determine intact erythropoiesis, and measurement of iron excretion in urine and feces. Typically, gastrointestinal bleeding is a major cause of iron loss. Surprisingly, intestinal iron excretion in stool in the patient was repetitively increased, without gastrointestinal bleeding. Furthermore, whole exome sequencing was performed in the patient and additional family members to identify potential causative genetic variants that may cause intestinal iron loss. Under different inheritance models, several rare mutations were identified, two of which (in CISD1 and KRI1) are likely to be functionally relevant. Intestinal iron loss in the current form has not yet been described and is, with high probability, the cause of the severe iron deficiency anemia in this patient.

In this research work, the machinability of turning Hastelloy X with a PVD Ti-Al-N coated insert tool in dry, wet, and cryogenic machining environments is investigated. The machinability indices namely cutting force (CF), surface roughness (SR), and cutting temperature (CT) are studied for the different set of input process parameters such as cutting speed, feed rate, and machining environment, through the experiments conducted as per L27 orthogonal array. Minitab 17 is used to create quadratic Multiple Linear Regression Models (MLRM) based on the association between turning parameters and machineability indices. The Moth-Flame Optimization (MFO) algorithm is proposed in this work to identify the optimal set of turning parameters through the MLRM models, in view of minimizing the machinability indices. Three case studies by considering individual machinability indices, a combination of dual indices, and a combination of all three indices, are performed. The suggested MFO algorithm’s effectiveness is evaluated in comparison to the findings of Genetic, Grass-Hooper, Grey-Wolf, and Particle Swarm Optimization algorithms. From the results, it is identified that the MFO algorithm outperformed the others. In addition, a confirmation experiment is conducted to verify the results of the MFO algorithm’s optimal combination of turning parameters.

Lower urinary tract obstruction (LUTO) is, in most cases, caused by anatomical blockage of the bladder outlet. The most common form are posterior urethral valves (PUVs), a male-limited phenotype. Here, we surveyed the genome of 155 LUTO patients to identify disease-causing CNVs. Raw intensity data were collected for CNVs detected in LUTO patients and 4.392 healthy controls using CNVPartition, QuantiSNP and PennCNV. Overlapping CNVs between patients and controls were discarded. Additional filtering implicated CNV frequency in the database of genomic variants, gene content and final visual inspection detecting 37 ultra-rare CNVs. After, prioritization qPCR analysis confirmed 3 microduplications, all detected in PUV patients. One microduplication (5q23.2) occurred de novo in the two remaining microduplications found on chromosome 1p36.21 and 10q23.31. Parental DNA was not available for segregation analysis. All three duplications comprised 11 coding genes: four human specific lncRNA and one microRNA. Three coding genes (FBLIM1, SLC16A12, SNCAIP) and the microRNA MIR107 have previously been shown to be expressed in the developing urinary tract of mouse embryos. We propose that duplications, rare or de novo, contribute to PUV formation, a male-limited phenotype.

In this work, wire electrical discharge machining (WEDM) of aluminum (LM25) reinforced with fly ash and boron carbide (B4C) hybrid composites was performed to investigate the influence of reinforcement wt% and machining parameters on the performance characteristics. The hybrid composite specimens were fabricated through the stir casting process by varying the wt% of reinforcements from 3 to 9. In the machinability studies, the WEDM process control parameters such as gap voltage, pulse-on time, pulse-off time, and wire feed were varied to analyze their effects on machining performance including volume removal rate and surface roughness. The WEDM experiments were planned and conducted through the L27 orthogonal array approach of the Taguchi methodology, and the corresponding volume removal rate and surface roughness were measured. In addition, the multi-parametric ANOVA was performed to examine the statistical significance of the process control parameters on the volume removal rate and surface roughness. Furthermore, the spatial distribution of the parameter values for both the responses were statistically analyzed to confirm the selection of the range of the process control parameters. Finally, the quadratic multiple linear regression models (MLRMs) were formulated based on the correlation between the process control parameters and output responses. The Grass–Hooper Optimization (GHO) algorithm was proposed in this work to identify the optimal process control parameters through the MLRMs, in light of simultaneously maximizing the volume removal rate and minimizing the surface roughness. The effectiveness of the proposed GHO algorithm was tested against the results of the particle swarm optimization and moth-flame optimization algorithms. From the results, it was identified that the GHO algorithm outperformed the others in terms of maximizing volume removal rate and minimizing the surface roughness values. Furthermore, the confirmation experiment was also carried out to validate the optimal combination of process control parameters obtained through the GHO algorithm.

Nanostructured materials synthesized by the hydrothermal and thermal reduction process were tested to detect the dimethyl methylphosphonate (DMMP) as a simulant for chemical warfare agents. Manganese oxide nitrogen-doped graphene oxide with polypyrrole (MnO2@NGO/PPy) exhibited the sensitivity of 51 Hz for 25 ppm of DMMP and showed the selectivity of 1.26 Hz/ppm. Nitrogen-doped multi-walled carbon nanotube (N-MWCNT) demonstrated good linearity with a correlation coefficient of 0.997. A comparison between a surface acoustic wave and quartz crystal microbalance sensor exhibited more than 100-times higher sensitivity of SAW sensor than QCM sensor.

Pulmonary vein stenosis (PVS) is a rare, but high mortality and resource intensive disease caused by mechanical obstruction or intraluminal myofibroproliferation, which can be post-surgical or idiopathic. There are increasing options for management including medications, cardiac catheterization procedures, and surgery. We queried the International Quality Improvement Collaborative for Congenital Heart Disease (IQIC) database for cases of PVS and described the cohort including additional congenital lesions and surgeries as well as infectious and mortality outcomes. IQIC is a quality improvement project in low-middle-income countries with the goal of reducing mortality after congenital heart surgery. Three cases were described in detail with relevant images. We identified 57 cases of PVS surgery, with similar mortality to higher income countries. PVS should be recognized as a global disease. More research and collaboration are needed to understand the disease, treatments, and outcomes, and to devise treatment approaches for low resource environments.

Endometrial cancer is the most common malignancy of the female genital tract and a major cause of morbidity and mortality in women. Early detection is key to ensuring good outcomes but a lack of minimally invasive screening tools is a significant barrier. Most endometrial cancers are obesity-driven and develop in the context of severe metabolomic dysfunction. Blood-derived metabolites may therefore provide clinically relevant biomarkers for endometrial cancer detection. In this study, we analysed plasma samples of women with body mass index (BMI) ≥ 30 kg/m2 and endometrioid endometrial cancer (cases, n = 67) or histologically normal endometrium (controls, n = 69), using a mass spectrometry-based metabolomics approach. Eighty percent of the samples were randomly selected to serve as a training set and the remaining 20% were used to qualify test performance. Robust predictive models (AUC > 0.9) for endometrial cancer detection based on artificial intelligence algorithms were developed and validated. Phospholipids were of significance as biomarkers of endometrial cancer, with sphingolipids (sphingomyelins) discriminatory in post-menopausal women. An algorithm combining the top ten performing metabolites showed 92.6% prediction accuracy (AUC of 0.95) for endometrial cancer detection. These results suggest that a simple blood test could enable the early detection of endometrial cancer and provide the basis for a minimally invasive screening tool for women with a BMI ≥ 30 kg/m2.

This study presents a series of experiments to test the integration of syngas fermentation into a single-cell microbial electrosynthesis (MES) process. Minimal gas–liquid mass transfer is the primary bottleneck in such gas-fermentation processes. Therefore, we hypothesized that MES integration could trigger the thermodynamic barrier, resulting in higher gas–liquid mass transfer and product-formation rates. The study was performed in three different phases as batch experiments. The first phase dealt with mixed-culture fermentation at 1 bar H2 headspace pressure. During the second phase, surface electrodes were integrated into the fermentation medium, and investigations were performed in open-circuit mode. In the third phase, the electrodes were poised with a voltage, and the second phase was extended in closed-circuit mode. Phase 2 demonstrated three times the gas consumption (1021 mmol) and 63% more production of acetic acid (60 mmol/L) than Phase 1. However, Phase 3 failed; at –0.8 V, acetic acid was oxidized to yield hydrogen gas in the headspace.

Cocaine abuse is known to alter mitochondrial biogenesis and induce epigenetic modification linked with neuronal dysfunction. Cocaine-induced epigenetic modification of DNA methylation and the mitochondrial genome may affect mitochondrial DNA (mtDNA) and nuclear DNA (nDNA), as epigenetic DNA methylation is key to maintaining genomic integrity in the central nervous system (CNS). However, the impact of cocaine-mediated epigenetic changes in astrocytes has not yet been elucidated. In this study, we explored the neuroprotective effect of piracetam against cocaine-induced epigenetic changes in DNA methylation in astrocytes. To study our hypothesis, we exposed human astrocytes to cocaine alone or in combination with the nootropic drug piracetam. We examined the expression of the DNA methyltransferases (DNMTs) DNMT-1, DNMT-3A, and DNMT-3B; global DNA methylation levels of 5-methycytosine (5-mC); and induction of ten–eleven translocation (TET) enzymes in astrocytes. In addition, we analyzed mtDNA methylation by targeted next-generation bisulfite sequencing. Our data provide evidence that cocaine impairs DNMT activity and thereby has impacts on mtDNA, which might contribute to the neurodegeneration observed in cocaine users. These effects might be at least partially prevented by piracetam, allowing neuronal function to be maintained.

Vasicinone is a quinazoline alkaloid isolated from the Adhatoda vasica plant. In this study, we explored the neuroprotective effect and underlying molecular mechanism of vasicinone against paraquat-induced cellular apoptosis in SH-SY5Y cells. Vasicinone reduced the paraquat-induced loss of cell viability, rescued terminal deoxynucleotide transferase-mediated dUTP nick end-labeling (TUNEL)-positive apoptotic nuclei, and suppressed generation of reactive oxygen species (ROS) in a dose-dependent manner. Western blotting analysis revealed that vasicinone increased the phosphorylation of IGF1R/PI3K/AKT cell survival signaling molecules and downregulated the paraquat-induced, mitogen-activated protein kinase (MAPK)/c-Jun N-terminal kinase (JNK)-mediated apoptotic pathways compared to that observed in cells not treated with vasicinone. This protection depended critically on the activation of IGF1R, and the silencing of IGF1R by siRNA completely abrogated the protective effect of vasicinone in SH-SY5Y cells. Our findings indicated that vasicinone is a potential candidate for the treatment of Parkinson’s disease and possibly other oxidative stress-related neurodegenerative disorders.

Gas sensing properties of diverse phthalocyanines functionalized TiO2/ZnO heterojunction thin films were investigated respect to a number of volatile organic compounds (VOCs) in both dark and light conditions. These studies showed that influence of heterojunction along with functionalization alter the optical properties and gas sensing of sensors. Results show that each sensor exhibits a different pattern of relative sensitivity, and this feature can be used to discriminate among a wide range of VOCs.

Ischemic heart disease (IHD) constitutes the leading global cause of mortality and morbidity. Although significant progress has been achieved in the diagnosis, treatment, and prognosis of IHD, more robust diagnostic biomarkers and therapeutic interventions are still needed to circumvent the increasing incidence of IHD. MicroRNAs (miRNAs) are critical regulators of cardiovascular function and are involved in various facets of cardiovascular biology. While the knowledge of the role of miRNAs in IHD as diagnostic biomarkers has improved, research emphasis on how miRNAs can be effectively used for diagnosis and prognosis of IHD is crucial. This review provides an overview of the biology, therapeutic and diagnostic potential, as well as the caveats of using miRNAs in IHD based on existing research.

We estimated the effectiveness of two doses of the ChAdOx1 nCoV-19 (Covishield) vaccine against any COVID-19 infection among individuals ≥45 years in Chennai, Tamil Nadu, India. A community-based cohort study was conducted from May to September 2021 in a selected geographic area in Chennai. The estimated sample size was 10,232. We enrolled 69,435 individuals, of which 21,793 were above 45 years. Two-dose coverage of Covishield in the 18+ and 45+ age group was 18% and 31%, respectively. Genomic analysis of 74 out of the 90 aliquots collected from the 303 COVID-19-positive individuals in the 45+ age group showed delta variants and their sub-lineages. The vaccine’s effectiveness against COVID-19 disease in the ≥45 age group was 61.3% (95% CI: 43.6–73.4) at least 2 weeks after receiving the second dose of Covishield. We demonstrated the effectiveness of two doses of the ChAdOx1 vaccine against the delta variant in the general population of Chennai. We recommend similar future studies considering emerging variants and newer vaccines. Two-dose vaccine coverage could be ensured to protect against COVID-19 infection.

Over the past decades, the rising antibiotic resistance bacteria (ARB) are continuing to emerge as a global threat due to potential public health risk. Rapidly evolving antibiotic resistance and its persistence in the environment, have underpinned the need for more studies to identify the possible sources and limit the spread. In this context, not commonly studied and a neglected genetic material called extracellular DNA (eDNA) is gaining increased attention as it can be one of the significant drivers for transmission of extracellular ARGS (eARGs) via horizontal gene transfer (HGT) to competent environmental bacteria and diverse sources of antibiotic-resistance genes (ARGs) in the environment. Consequently, this review highlights the studies that address the environmental occurrence of eDNA and encoding eARGs and its impact on the environmental resistome. In this review, we also brief the recent dedicated technological advancements that are accelerating extraction of eDNA and the efficiency of treatment technologies in reducing eDNA that focuses on environmental antibiotic resistance and potential ecological health risk.

A binder-free process for the electrode preparation for supercapacitor application was suggested by drop casting graphene nanoplatelets on a carbon fiber (GnP@CF) followed by electrodeposition of Ni nanoparticles (NPs). The microstructure of the electrode showed that Ni was homogeneously distributed over the surface of the GnP@CF. XRD analysis confirmed the cubic structure of metallic Ni NPs. The Ni-GnP@CF electrode showed excellent pseudocapacitive behavior in alkaline solution by exhibiting a specific capacitance of 480 F/g at 1.0 A/g, while it was 375 F/g for Ni@CF. The low value of series resistance of Ni-GnP@CF (1 Ω) was attributed to the high capacitance. The enhanced capacitance of the electrode could be correlated to the highly nanoporous structure of the composite material, synergetic effect of the electrical double layer charge-storage properties of graphene, and the pseudocapacitive nature of Ni NPs.

There is an ongoing worldwide coronavirus disease 2019 (Covid-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). At present, confirmatory diagnosis is by reverse transcription polymerase chain reaction (RT-PCR), typically taking several hours and requiring a molecular laboratory to perform. There is an urgent need for rapid, simplified, and cost-effective detection methods. We have developed and analytically validated a protocol for direct rapid extraction-free PCR (DIRECT-PCR) detection of SARS-CoV-2 without the need for nucleic acid purification. As few as six RNA copies per reaction of viral nucleocapsid (N) gene from respiratory samples such as sputum and nasal exudate can be detected directly using our one-step inhibitor-resistant assay. The performance of this assay was validated on a commercially available portable PCR thermocycler. Viral lysis, reverse transcription, amplification, and detection are achieved in a single-tube homogeneous reaction within 36 min. This minimizes hands-on time, reduces turnaround-time for sample-to-result, and obviates the need for RNA purification reagents. It could enable wider use of Covid-19 testing for diagnosis, screening, and research in countries and regions where laboratory capabilities are limiting.

Mitochondrial dysfunction and disturbed mitochondrial dynamics were found to be common phenomena in the pathogenesis of Parkinson’s disease (PD). Vasicinone is a quinazoline alkaloid from Adhatoda vasica. Here, we investigated the autophagy/mitophagy-enhancing effect of vasicinone and explored its neuroprotective mechanism in paraquat-mimic PD modal in SH-SY5Y cells. Vasicinone rescued the paraquat-induced loss of cell viability and mitochondrial membrane potential. Subsequently, the accumulation of mitochondrial reactive oxygen species (ROS) was balanced by an increase in the expression of antioxidant enzymes. Furthermore, vasicinone restored paraquat-impaired autophagy and mitophagy regulators DJ-1, PINK-1 and Parkin in SH-SY5Y cells. The vasicinone mediated autophagy pathways were abrogated by treatment with the autophagy inhibitor 3-MA, which lead to increases α-synuclein accumulation and decreased the expression of p-ULK and ATG proteins and the autophagy marker LC3-II compared to that observed without 3-MA treatment. These results demonstrated that vasicinone exerted neuroprotective effects by upregulating autophagy and PINK-1/Parkin mediated mitophagy in SH-SY5Y cells.

Endometrial cancer is the sixth most common cancer in women, with a rising incidence worldwide. Current approaches for the diagnosis and screening of endometrial cancer are invasive, expensive or of moderate diagnostic accuracy, limiting their clinical utility. There is a need for cost-effective and minimally invasive approaches to facilitate the early detection and timely management of endometrial cancer. We analysed blood plasma samples in a cross-sectional diagnostic accuracy study of women with endometrial cancer (n = 342), its precursor lesion atypical hyperplasia (n = 68) and healthy controls (n = 242, total n = 652) using attenuated total reflection-Fourier transform infrared (ATR-FTIR) spectroscopy and machine learning algorithms. We show that blood-based infrared spectroscopy has the potential to detect endometrial cancer with 87% sensitivity and 78% specificity. Its accuracy is highest for Type I endometrial cancer, the most common subtype, and for atypical hyperplasia, with sensitivities of 91% and 100%, and specificities of 81% and 88%, respectively. Our large-cohort study shows that a simple blood test could enable the early detection of endometrial cancer of all stages in symptomatic women and provide the basis of a screening tool in high-risk groups. Such a test has the potential not only to differentially diagnose endometrial cancer but also to detect its precursor lesion atypical hyperplasia—the early recognition of which may allow fertility sparing management and cancer prevention.

Water from anaerobic sludge dewatering (reject water that is recycled to the inlet main process treatment) from the Knarrdalstrand municipal wastewater treatment plant in Porsgrunn, Norway, contains 2.4 g/L of total chemical oxygen demand (TCOD) and 550 mg/L NH4-N (annual average). The high concentration of ammonium causes disturbances in the mainstream physical and chemical processes, while only a small fraction of the organics is biodegradable. A pilot-scale hybrid vertical anaerobic biofilm (HyVAB) reactor combining anaerobic and aerobic treatment was tested for reject water treatment to reduce process disturbances. The pilot HyVAB was prepared for the study with continuous aeration of the aerobic part of the reactor for 200 days, while two intermittent aeration schemes were applied during the three-month test period. Ammonium removal efficiency increased from 8% during the continuous aeration period to 50% at the end of the test when a short (7 min) aeration cycle was applied. COD removal was close to 20%, which was mainly obtained in the anaerobic stage and not significantly influenced by the aerations schemes. Simultaneous partial nitrification and denitrification were established in the biofilm that alternated between aerobic and anoxic conditions. The observed high ammonium removal is explained by two alternative shortcut processes via nitrite. The lack of biodegradable organics in the aerated stage suggests that most of the nitrogen removal was via the anammox pathway (autotrophic denitrification). The HyVAB, combining an anaerobic sludge bed and an intermittently aerated biofilm, appears to be an efficient process to treat high ammonium containing reject water.

Worldwide, depression and bipolar disorder affect a large and growing number of people. However, current pharmacotherapy options remain limited. Despite adequate treatment, many patients continue to have subsyndromal symptoms, which predict relapse in bipolar illness and often result in functional impairments. Aspirin, a common nonsteroidal anti-inflammatory drug (NSAID), has purported beneficial effects on mood symptoms, showing protective effects against depression in early cohort studies. This systematic review thus aimed to investigate the role of aspirin in mood disorders. Using the keywords (aspirin or acetylsalicy* or asa) and (mood or depress* or bipolar or mania or suicid*), a comprehensive search of PubMed, EMBASE, Medline, PsycINFO, Clinical Trials Register of the Cochrane Collaboration Depression, Anxiety and Neurosis Group (CCDANTR), Clinicaltrials.gov and Google Scholar databases found 13,952 papers published in English between 1 January 1988 and 1 May 2019. A total of six clinical studies were reviewed. There were two randomized, placebo-controlled, double-blind trials and populations drawn from two main cohort studies (i.e., the Geelong Osteoporosis Study and the Osteoarthritis Initiative study). Using a random-effects model, the pooled hazard ratio of the three cohort studies was 0.624 (95% confidence interval: 0.0503 to 1.198, p = 0.033), supporting a reduced risk of depression with aspirin exposure. Overall, the dropout rates were low, and aspirin appears to be well-tolerated with minimal risk of affective switch. In terms of methodological quality, most studies had a generally low risk of bias. Low-dose aspirin (80 to 100 mg/day) is safe, well-tolerated and potentially efficacious for improving depressive symptoms in both unipolar and bipolar depression. Due to its ability to modulate neuroinflammation and central nervous system processes, aspirin may also have valuable neuroprotective and pro-cognitive effects that deserve further exploration. Further randomized, controlled trials involving the adjunctive use of aspirin should be encouraged to confirm its therapeutic benefits.

Notably, carbapenem-resistant Enterobacteriaceae (CRE) infections at global level attribute a vast, dangerous clinical threat. In most cases, there are enormous difficulties for CRE infection except a few last resort toxic drugs such as tigecycline and colistin (polymyxin E). Due to this, CRE has now been categorized as one among the three most dangerous multidrug resistance (MDR) pathogens by the US Centres for Disease Control and Prevention (CDC). Considering this, the study of the frequency of CRE infections and the characterization of CRE is an important area of research in clinical settings. However, MDR bacteria are not only present in hospitals but are spreading more and more into the environment, thereby increasing the risk of infection with resistant bacteria outside the hospital. In this context, developing countries are a global concern where environmental regulations are often insufficient. It seems likely that overcrowding, poor sanitation, socioeconomic status, and limited infrastructures contribute to the rapid spread of MDR bacteria, becoming their reservoirs in the environment. Thus, in this review, we present the occurrence of CRE and their resistance determinants in different environmental compartments in India.

Low-temperature stress severely affects the growth, development, and geographical distribution of various crop plants, resulting in significant economic loss to producers. In a quest to identify cold-regulated genes, we constructed a cDNA suppression subtractive library from a high altitude adapted ecotype of Lepidium. We cloned a cold-induced gene LlaCIPK from the subtracted cDNA library which gave homology to Arabidopsis CIPK15 gene. The predicted 3D structure of LlaCIPK protein also showed homology with Arabidopsis CIPK protein. Quantitative real-time PCR analysis in Lepidium seedlings exposed to 6 h of cold stress shows a 3-fold increase in the expression of LlaCIPK transcript. The expression of LlaCIPK was also differentially regulated by ethylene, CaCl2, ABA, and SA treatments. Ethylene and CaCl2 treatments up regulated LlaCIPK expression, whereas ABA and SA treatments down regulated the LlaCIPK expression. Transgenic plants overexpressing LlaCIPK gene under constitutive promoter show an increased level of proline and cell membrane stability. Taken together, our results suggest that the LlaCIPK contributes to the cold-response pathway in Lepidium plants.

Darrieus-type Vertical Axis Wind Turbines (VAWT) are promising for small scale decentralized power generation because of their unique advantages such as simple design, insensitive to wind direction, reliability, and ease of maintenance. Despite these positive aspects, poor self-starting capability and low efficiency in weak and unsteady winds deteriorate further development. Adaptive Hybrid Darrieus Turbine (AHDT) was proposed by the author in the past study as a potential solution to enhance low wind speed characteristics. The objective of the current research is to optimize the parameters of AHDT. AHDT integrates a dynamically varying Savonius rotor with a Darrieus rotor. A fully detailed 2D numerical study employing Reynold-Averaged Navier Stokes (RANS) is carried out to investigate the impact of the Darrieus rotor diameter (DR) on the Savonius rotor (DT) with regard to hybrid turbine performance. The power coefficient of the Darrieus rotor is evaluated when the Savonius rotor is in the closed condition (cylinder) of various diameters. The influence of Reynolds number (Re) on the torque coefficient is examined. Power loss of 58.3% and 25% is reported for DR/DT ratio of 1.5 and 2 respectively for AHDT with solidity 0.5 at 9 m/s. The flow interaction between the Savonius rotor in closed configuration reveals the formation of von Karman vortices that interact with Darrieus blades resulting in flow detachment. An optimum diametrical ratio (DR/DT) of 3 is found to yield the maximum power coefficient of the Darrieus rotor.

Aerodynamic performance of a floating offshore wind turbine (FOWT) is significantly influenced by platform surging motions. Accurate prediction of the unsteady aerodynamic loads is imperative for determining the fatigue life, ultimate loads on key components such as FOWT rotor blades, gearbox and power converter. The current study examines the predictions of numerical codes by comparing with unsteady experimental results of a scaled floating wind turbine rotor. The influence of platform surge amplitude together with the tip speed ratio on the unsteady aerodynamic loading has been simulated through unsteady CFD. It is shown that the unsteady aerodynamic loads of FOWT are highly sensitive to the changes in frequency and amplitude of the platform motion. Also, the surging motion significantly influences the windmill operating state due to strong flow interaction between the rotating blades and generated blade-tip vortices. Almost in all frequencies and amplitudes, CFD, LR-BEM and LR-uBEM predictions of mean thrust shows a good correlation with experimental results.
