This paper presents a passive Envelope Detector (ED) to be used for reception of OOK-modulated signals, such as in Wake-Up Receivers employed within Wireless Sensor Networks, widely used in the IoT. The main goal is implementing a temperature compensation mechanism in order to keep the passive ED input resistance roughly constant over temperature, making it a constant load for the preceding matching network and ultimately keeping the overall receiving chain sensitivity constant over temperature. The proposed ED was designed using STMicroelectronics 90 nm CMOS technology to receive 1 kbps OOK-modulated packets with a 433 MHz carrier frequency and a 0.6 V supply. The use of a block featuring a Proportional-to-Absolute Temperature (PTAT) current yields a 5 dB reduction in sensitivity temperature variation across the −40 °C to 120 °C range. Moreover, two different implementations were compared, one targeting minimal mismatch and the other one targeting minimal area. The minimal area version appears to be better in terms of estimated overall chain sensitivity at all temperatures despite a higher sensitivity spread.

Infections caused by KPC-producing K. pneumoniae continue to pose a significant clinical challenge due to their emerging resistance to new antimicrobials. We investigated the association between two drugs whose roles have been repurposed against multidrug-resistant bacteria: fosfomycin and temocillin. Temocillin exhibits unusual stability against KPC enzymes, while fosfomycin acts as a potent “synergizer”. We conducted in vitro antimicrobial activity studies on 100 clinical isolates of KPC-producing K. pneumoniae using a combination of fosfomycin and temocillin. The results demonstrated synergistic activity in 91% of the isolates. Subsequently, we assessed the effect on Galleria mellonella larvae using five genetically different KPC-Kp isolates. The addition of fosfomycin to temocillin increased larvae survival from 73 to 97% (+Δ 32%; isolate 1), from 93 to 100% (+Δ 7%; isolate 2), from 63 to 86% (+Δ 36%; isolate 3), from 63 to 90% (+Δ 42%; isolate 4), and from 93 to 97% (+Δ 4%; isolate 10). Among the temocillin-resistant KPC-producing K. pneumoniae isolates (24 isolates), the addition of fosfomycin reduced temocillin MIC values below the resistance breakpoint in all isolates except one. Temocillin combined with fosfomycin emerges as a promising combination against KPC-producing K. pneumoniae, warranting further clinical evaluation.

The need to use renewable sources and matrices with energy potential is widely recognized. The development of innovative technologies aimed at the improvement of energy conversion processes and reducing environmental impacts is currently receiving increasing attention from the scientific community and policymakers. The presence of sugars in airborne particle materials is attributed to biomass combustion. For this reason, these compounds are considered markers of biomass burning. The purpose of this work was to evaluate the emissions produced by agroforestry biomass burning (citrus pruning) by simultaneously sampling both stack emissions and atmospheric particulates in the area around a biomass boiler to understand the real contribution of biomass burning to atmospheric pollution. The combustion tests were carried out by comparing the processes with and without particulate abatement system to see how biomass combustion’s contribution to particulate emission can be controlled and reduced. During the tests, the focus was on particulate matter (PM) speciation in terms of sugar marker identification and determination. This study aims to increase knowledge to better understand the contribution of biomass plants to air pollution and differentiate it from the contributions of other sources, such as vehicular traffic or domestic heating.

The injudicious usage of antibiotics during infections caused by Gram-negative bacteria leads to the emergence of β-lactamases. Among them, the NDM-1 enzyme poses a serious threat to human health. Developing new antibiotics or inhibiting β-lactamases might become essential to reduce and prevent bacterial infections. Nanobodies (Nbs), the smallest antigen-binding single-domain fragments derived from Camelidae heavy-chain-only antibodies, targeting enzymes, are innovative alternatives to develop effective inhibitors. The biopanning of an immune VHH library after phage display has helped to retrieve recombinant antibody fragments with high inhibitory activity against recombinant-NDM-1 enzyme. Nb02NDM-1, Nb12NDM-1, and Nb17NDM-1 behaved as uncompetitive inhibitors against NDM-1 with Ki values in the nM range. Remarkably, IC50 values of 25.0 nM and 8.5 nM were noted for Nb02NDM-1 and Nb17NDM-1, respectively. The promising inhibition of NDM-1 by Nbs highlights their potential application in combating particular Gram-negative infections.

Maritime safety and the protection of the marine environment were the primary objectives of two European projects that the National Research Council of Italy had participated in, with numerical applications in two areas located in the northern part of Sardinia, Italy. Specifically, two operational Numerical Prediction Systems (NPS) for pollution risk management were developed; the first was applied to the area of the Bonifacio Strait and the Gulf of Asinara and the second to the port of Olbia. These systems are composed of many oceans and particle tracking numerical models. They are forced with meteorological and ocean data provided by the European Centre for Medium-Range Weather Forecasts and Copernicus Marine Service and their outputs have been compared with in situ measurements for preliminary calibration. A web graphical interface was ad hoc designed, specifically responding to projects’ needs, providing online access to a 3-day oceanographic forecast and advanced diagnostic variables like Oil Stranding Time, Risk Score and Water Age. These products, along with the interactive web platform, prove invaluable for marine spatial planning, prevention and emergency management at sea, for the use of competent governmental and local bodies.

In the present study, we analyzed the genome of two S. enterica strains TS1 and TS2 from stool and blood cultures, respectively, and one strain of C. freundii TS3, isolated from a single hospitalized patient with acute myeloid leukemia. The S. enterica Goldcoast ST358 (O:8 (C2-C3) serogroup), sequenced by the MiSeq Illumina system, showed the presence of β-lactamase genes (blaVIM-1, blaSHV-12 and blaOXA-10), aadA1, ant(2″)-Ia, aac(6′)-Iaa, aac(6′)-Ib3, aac(6′)-Ib-cr, qnrVC6, parC(T57S), and several incompatibility plasmids. A wide variety of insertion sequences (ISs) and transposon elements were identified. In C. freundii TS3, these were the blaVIM-1, blaCMY-150, and blaSHV-12, aadA1, aac(6′)-Ib3, aac(6′)-Ib-cr, mph(A), sul1, dfrA14, ARR-2, qnrVC6, and qnrB38. IncA plasmid isolated from E.coli/K12 transconjugant and C. freundii exhibited a sequence identity >99.9%. The transfer of IncA plasmid was evaluated by conjugation experiments.

The dramatic increase in infections caused by critically multidrug-resistant bacteria is a global health concern. In this study, we characterized the antimicrobial resistance genes (ARGs) of K. pneumoniae, P. mirabilis, E. cloacae and A. baumannii isolated from both surgical wound and rectal swab of a single Italian patient. Bacterial identification was performed by MALDI-TOF MS and the antimicrobial susceptibility was carried out by Vitek 2 system. The characterization of ARGs was performed using next-generation sequencing (NGS) methodology (MiSeq Illumina apparatus). K. pneumoniae, P. mirabilis and E. cloacae were resistant to most β-lactams and β-lactam/β-lactamases inhibitor combinations. A. baumannii strain was susceptible only to colistin. The presence of plasmids (IncN, IncR, IncFIB, ColRNAI and Col (MGD2)) was detected in all Enterobacterales but not in A. baumannii strain. The IncN plasmid and blaNDM-1 gene were found in K. pneumoniae, P. mirabilis and E. cloacae, suggesting a possible transfer of this gene among the three clinical species. Conjugation experiments were performed using K. pneumoniae (1 isolate), P. mirabilis (2 isolates) and E. cloacae (2 isolates) as donors and E. coli J53 as a recipient. The blaNDM-1 gene was identified by PCR analysis in all transconjugants obtained. The presence of four different bacterial species harboring resistance genes to different classes of antibiotics in a single patient substantially reduced the therapeutic options.

Infections caused by carbapenem-resistant Acinetobacter baumannii (CRAB) have limited therapeutic options. Sulbactam-durlobactam is a combination of two βlactamase inhibitors with activity against CRAB under phase 3 clinical investigation. We performed a systematic review on in vitro studies reporting A. baumannii resistances against sulbactam/durlobactam. We considered “resistant” species to be those with MIC ≥ 8 mg/L. Ten studies were included in the review (9754 tested isolates). Overall, 2.3% of A. baumannii were resistant to sulbactam/durlobactam, and this percentage rose to 3.4% among CRAB subgroups and to 3.7% among colistin-resistant strains. Resistance was 100% among metallo β-lactamase-producing strains. Overall, in 12.5% of cases, sulbactam/durlobactam resistance was associated with the production of NDM-1, in 31.7% of cases with the substitutions in the PBP3 determinants, and in the remaining cases the resistance mechanism was unknown. In conclusion, A. baumannii resistance towards sulbactam/durlobactam is limited, except for MBL-producing strains.

Introduction. The Biolabo Solea 100 is a fully automated coagulation analyser using an optical system to detect coagulation designed to meet the needs of small- and medium-sized laboratories. This study aimed to evaluate the analytical performance in terms of bias, precision, and interference of the Biolabo Solea 100 coagulometer under routine laboratory conditions. In addition, a comparison was made with Stago STA-R MAX. Materials and Methods. Imprecision and bias were evaluated for activated partial thromboplastin time (APTT), fibrinogen (FIB), and prothrombin time (PT) at the medical decision levels. The results of 200, 181, and 206 plasma samples for APTT, FIB, and PT, respectively, were compared with those obtained by Stago STA-R MAX. In addition, the interference level of bilirubin, haemoglobin, triglycerides, and fractionated heparin was evaluated. Results. Repeatability, intermediate imprecision, bias, and total error are overall below the defined limits of acceptability. Of interest is the high degree of agreement between Solea 100 and STA-R MAX with respect to PT (s), which fits perfectly with the theoretical line of identity (y = 0 + 1.00x). No interferences were found within the limits stated by the manufacturer, with some exceptions for APTT with heparin and APTT and PT for higher bilirubin concentrations. Conclusions. In conclusion, the performance of the Solea 100 optical analyser is satisfactory and adequate for the determination of routine coagulation tests. Moreover, they are perfectly comparable to mechanical systems, such as STA-R MAX and other upper-level analysers, even considering the low interference levels under routine conditions.

Agricultural activities lead to the production of large quantities of pruning and residual biomass, the correct disposal of which is a topic of great interest. Unfortunately, although it is an environmentally unacceptable practice, disposal often takes place with uncontrolled combustion in field (open burning). Disposal using biomass boilers produces energy from controlled combustion and the use of abatement systems reduces emissions that are inevitably generated in the combustion processes of solid fuel (CO, CO2, NOx, SO2, PM, etc.). Among the pollutants produced in combustion processes, Persistent Organic Pollutants (POPs) are one of the most relevant groups of chemicals that must be monitored for their effect on human health and the environment. This paper reports the data obtained for the emissive characterization of POPs of three different wood biomass prunings (Olive, Citrus and Grapevine) focusing on VOCs, PAHs, PCBs, PCDD/Fs. Emissions samplings were performed at the stack during combustion in a 30 kWth boiler equipped with a multicyclone filter bag for PM abatement in an isokinetic mode in accordance with the CEN and ISO standards methods. Through this work, a complete emission profile of VOCs, PAHs, PCBs, and PCDD/Fs of three different biomass types is provided for the first time, showing that the use of biomass boilers produces quantities of pollutants below the regulatory limits and still lower than those produced by open burning phenomena. Citrus has shown, regarding the other two essences, elevated concentrations in terms of Σ PAH = 174.247 ng/m3, while Olive gave higher results in terms of PCBs and PCDD/Fs (respectively, 39.769 and 3390.63 TEQ pg/Nm3). This is probably due to the high concentration of iron in this biomass (711.44 mg/kg), which during combustion is a catalyst (together with copper) of PCDD/Fs.

Background: Alcohol consumption during pregnancy, even at low doses, may damage the fetus. Pregnant women tend to underreport their alcohol consumption generating the need for sensitive and specific biomarkers, among which PEth has emerged due to its high specificity and possibility to be measured in both maternal and neonatal blood. The aim of this study is to systematically review the latest 20 years of literature for depicting the state of the art, the limitations, and the prospects of PEth for estimating alcohol consumption during pregnancy. Materials and methods: A systematic search, adhering to PRISMA guidelines, of the latest 20 years of literature through “MeSH” and “free-text” protocols in the databases PubMed, SCOPUS, and Web of Science, with time limits 1 January 2002–1 March 2022, was performed. The inclusion criteria were as follows: PEth used for detecting alcohol consumption during pregnancy, quantified in blood through liquid chromatography coupled to mass spectrometry, and full texts in the English language. Opinion papers, editorials, and narrative reviews were excluded. Results: Sixteen (16) papers were included in the present review (0.81% of total retrieved records). All the included records were original articles, of which there were seven prospective cohort/longitudinal studies, six cross-sectional studies, two observational-descriptive studies, and one retrospective study. All studies assayed PEth in at least one biological matrix; seven (7) studies quantified PEth in maternal blood, seven studies in newborn blood, and only two studies in both maternal and neonatal blood. In several included papers, PEth proved more sensitive than self-reports for identifying pregnant women with an active alcohol intake with the diagnostic efficiency improving with the increase of the maternal alcohol intake. Conclusions: Further studies, performed on wider and well-stratified populations, are needed to drive any definitive conclusion. PEth is a promising marker for monitoring alcohol use in pregnancy; however, at the present time, its use is still limited mainly by the absence of a globally agreed interpretative cut-off, the paucity of data regarding its specificity/sensitivity, and the lack of standardization on the diagnostic efficiency of the different isoforms.

The whole-genome sequencing (WGS) of eighteen S. marcescens clinical strains isolated from 18 newborns hospitalized in the Neonatal Intensive Care Unit (NICU) at Pescara Public Hospital, Italy, was compared with that of S. marcescens isolated from cradles surfaces in the same ward. The identical antibiotic resistance genes (ARGs) and virulence factors were found in both clinical and environmental S. marcescens strains. The aac(6′)-Ic, tetA(41), blaSRT-3, adeFGH, rsmA, and PBP3 (D350N) genes were identified in all strains. The SRT-3 enzyme, which exhibited 10 amino acid substitutions with respect to SST-1, the constitutive AmpC β-lactamase in S. marcescens, was partially purified and tested against some β-lactams. It showed a good activity against cefazolin. Both clinical and environmental S. marcescens strains exhibited susceptibility to all antibiotics tested, with the exception of amoxicillin/clavulanate.

This study aimed to improve the overall fatigue properties of WAAM-produced SDSS by changing the interpass temperatures. Micro-computed tomography was used to quantitatively characterise the internal defects, such as porosity, in large-volume WAAM-fabricated SDSS materials. An increase in the interpass temperature led to a reduction in the ferrite phase balance by up to 20%. The fatigue anisotropy was still evident, but the fatigue limit in the weakest (transverse) direction was increased to 250 MPa or by approximately 40%. Meanwhile, the increased interpass temperature had no significant effect on fatigue resistance in the longitudinal direction. This study suggests that the interpass temperature can be critical for both achieving isotropic mechanical properties and increasing fatigue life of structural components fabricated with the WAAM method.

In the present study, the in vitro activity of the sulbactam–durlobactam (SUL–DUR) combination was evaluated against 141 carbapenem-resistant A. baumannii (CRAb) clinical strains collected from six Italian laboratories. Over half (54.6%) of these isolates were resistant to colistin. The SUL–DUR combination was active against these CRAb isolates with MIC50 and MIC90 values of 0.5 mg/L and 4 mg/L, respectively. Only eleven isolates were resistant to SUL–DUR with MIC values ranging from 8 to 128 mg/L. The SUL–DUR resistant A. baumannii exhibited several antimicrobial resistance genes (ARGs) such as blaOXA-20, blaOXA-58, blaOXA-66, blaADC-25, aac(6′)-Ib3 and aac(6′)-Ib-cr and mutations in gyrA (S81L) and parC (V104I, D105E). However, in these isolates, mutations Q488K and Y528H were found in PBP3. Different determinants were also identified in these CRAb isolates, including adeABC, adeFGH, adeIJK, abeS, abaQ and abaR, which encode multidrug efflux pumps associated with resistance to multiple antibacterial agents. This is the first report on the antimicrobial activity of SUL–DUR against carbapenem-resistant A. baumannii isolates selected from multiple regions in Italy.

A total of 43 A. baumannii strains, isolated from 43 patients affected by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and by bacterial sepsis, were analyzed by antimicrobial susceptibility testing. All strains were resistant to almost three different classes of antibiotics, including carbapenems and colistin. The whole-genome sequencing (WGS) of eight selected A. baumannii isolates showed the presence of different insertion sequences (ISs), such as ISAba13, ISAba26, IS26, ISVsa3, ISEc29, IS6100 and IS17, giving to A. baumannii a high ability to capture and mobilize antibiotic resistance genes. Resistance to carbapenems is mainly mediated by the presence of OXA-23, OXA-66 and OXA-82 oxacillinases belonging to OXA-51-like enzymes. The presence of AmpC cephalosporinase, ADC-25, was identified in all A. baumannii. The pathogenicity of A. baumannii was exacerbated by the presence of several virulence factors. The multi-locus sequence typing (MLST) analysis showed that all strains belong to sequence type 2 (ST) international clone.

The study investigated the inhibitory activity of protocetraric and salazinic acids against SARS-CoV-2 3CLpro. The kinetic parameters were determined by microtiter plate-reading fluorimeter using a fluorogenic substrate. The cytotoxic activity was tested on murine Sertoli TM4 cells. In silico analysis was performed to ascertain the nature of the binding with the 3CLpro. The compounds are slow-binding inactivators of 3CLpro with a Ki of 3.95 μM and 3.77 μM for protocetraric and salazinic acid, respectively, and inhibitory efficiency kinact/Ki at about 3 × 10−5 s−1µM−1. The mechanism of inhibition shows that both compounds act as competitive inhibitors with the formation of a stable covalent adduct. The viability assay on epithelial cells revealed that none of them shows cytotoxicity up to 80 μM, which is well below the Ki values. By molecular modelling, we predicted that the catalytic Cys145 makes a nucleophilic attack on the carbonyl carbon of the cyclic ester common to both inhibitors, forming a stably acyl-enzyme complex. The computational and kinetic analyses confirm the formation of a stable acyl-enzyme complex with 3CLpro. The results obtained enrich the knowledge of the already numerous biological activities exhibited by lichen secondary metabolites, paving the way for developing promising scaffolds for the design of cysteine enzyme inhibitors.

Human salmonellosis incidence is increasing in the European Union (EU). Salmonellaenterica subsp. enterica serovar Enteriditis, Salmonellaenterica subsp. enterica serovar Typhimurium (including its monophasic variant) and Salmonellaenterica subsp. enterica serovar Infantis represent targets in control programs due to their frequent association with human cases. This study aimed to detect the most prevalent serotypes circulating in Abruzzo and Molise Regions between 2015 and 2020 in the framework of the Italian National Control Program for Salmonellosis in Poultry (PNCS)]. A total of 332 flocks of Abruzzo and Molise Regions were sampled by veterinary services in the period considered, and 2791 samples were taken. Samples were represented by faeces and dust from different categories of poultry flocks: laying hens (n = 284), broilers (n = 998), breeding chickens (n = 1353) and breeding or fattening turkeys (n = 156). Breeding and fattening turkeys had the highest rate of samples positive for Salmonella spp. (52.6%; C.I. 44.8%–60.3%). Faeces recovered through boot socks represented the greatest number of positive samples (18.2%). Salmonellaenterica subsp. enterica serovar Infantis was the prevalent serotype in breeding and fattening turkeys (32.7%; C.I. 25.8%–40.4%) and in broiler flocks (16.5%; C.I. 14.4%–19.0%). Salmonellaenterica subsp. enterica serovar Typhimurium was detected at low levels in laying hens (0.7%; C.I. 0.2%–2.5%) followed by breeding and fattening turkeys (0.6%; C.I. 0.2%–2.5%). Salmonellaenterica subsp. enterica serovar Enteriditis was also detected at low levels in laying hens (2.5%; C.I. 1.2%–5.0%). These findings highlight the role of broilers and breeding/fattening turkeys as reservoirs of Salmonella spp. and, as a consequence, in the diffusion of dangerous serotypes as Salmonellaenterica subsp. enterica serovar Infantis. This information could help veterinary services to analyze local trends and to take decisions not only based on indications from national control programs, but also based on real situations at farms in their own competence areas.

Long-term care facilities (LTCFs) are important reservoirs of antimicrobial-resistant (AMR) bacteria which colonize patients transferred from the hospital, or they may emerge in the facility as a result of mutation or gene transfer. In the present study, we characterized, from a molecular point of view, 43 E. coli strains collected from residents of LTCFs in Northern Italy. The most common lineage found was ST131, followed by sporadic presence of ST12, ST69, ST48, ST95, ST410 and ST1193. All strains were incubators of several virulence factors, with iss, sat, iha and senB being found in 84%, 72%, 63% and 51% of E. coli, respectively. Thirty of the ST131 analyzed were of the O25b:H4 serotype and H30 subclone. The ST131 isolates were found to be mainly associated with IncF plasmids, CTX-M-1, CTX-M-3, CTX-M-15, CTX-M-27 and gyrA/parC/parE mutations. Metallo-β-lactamases were not found in ST131, whereas KPC-3 carbapenemase was found only in two ST131 and one ST1193. In conclusion, we confirmed the spread of extended-spectrum β-lactamase genes in E. coli ST131 isolated from colonized residents living inside LTCFs. The ST131 represents an incubator of fluoroquinolones, aminoglycosides and other antibiotic resistance genes in addition to different virulence factors.

The results of air pollution assessment during a 2017 cruise of the research ship “Minerva Uno” in the Tyrrhenian Sea are reported. Volatile Organic Compounds (VOCs), Oxygenated Volatile Organic Compounds (OVOCs), and pollutants such as nitrogen oxides, ozone, and sulphur dioxide were monitored throughout the cruise. The shallow waters at ten sites of the investigated area were also analyzed. Organic compounds such as n-alkanes showed a bimodal distribution with a maximum at C5–C6 and C10–C11 at sites the most affected by anthropic impact, whereas remote sites showed a unimodal distribution with maximum at C10–C11. The most abundant atmospheric OVOC was acetone (3.66 μg/m3), accounting for 38%; formaldehyde (1.23 μg/m3) and acetaldehyde (0.99 μg/m3) made up about 22–29% of the total. The influence of some natural sources as volcanoes, in the southern part of the Tyrrhenian Sea near the Aeolian arc was studied. This source did not induce any noticeable effect on the total amount of hydrocarbons nor on the levels of trace gases such as CFCs, whereas the trends of sulphur dioxide seemed to confirm a possible contribution. The impact of underwater emissions was observed near the Panarea and Vulcano islands, where lower pHs, high levels of Fe and Mn, and diagnostic of vent activity, were measured.

In building construction, it is very important to reduce energy consumption and provide thermal comfort. In this regard, defects in insulating panels can compromise the capability of these panels of reducing the heat flow by conduction with the surroundings. In recent years, both experimental techniques and numerical methods have been used for investigating the effect of defects on the thermal behavior of building panels. The main novelty of this work regards the application of both numerical and experimental approaches based on infrared thermography techniques for studying the effects of defects such as debonding on the insulation properties of cork panels. In particular, the effects of defects were investigated by using the Long Pulse Thermography technique and then by analyzing the thermal behavior of the panel during the cooling phase. Results show the capability of the proposed approaches in describing the effects of defects in cork panels such as detachments and the benefit effect of a shield coating in improving the insulation properties of the panel.

The Western Mediterranean basin (WMED) is characterized by the presence of energetic and dynamic mesoscale cyclonic and anticyclonic eddies. They mainly originate along the Algerian and the Northern currents and have a large influence on the basin circulation. Eddies can last for months, with longer lifetimes associated with the anticyclones, which can move far from their areas of origin. As they partially isolate and transfer water masses, they also have an impact on water properties (physical, chemical and biological), pollutant’s dispersion and transport of eggs, larvae and planktonic organisms. In this study, a connectivity analysis method is applied to the anticyclonic eddies (AEs) identified by an automated hybrid detection and tracking algorithm south of 42°N in the WMED. The same methodology is also applied to the trajectories of Lagrangian surface drifters available in the study area. The purpose is to highlight the connections between different areas of the basin linked to eddy activities in addition to the connectivity due to the mean surface circulation. Drifter data analysis showed that all the WMED sub-basins are strongly interconnected, with the mean surface circulation allowing a shortcut connection among many areas of the basin. The connectivity analysis of the AEs tracks shows that although AEs are ubiquitous in the WMED, their connectivity is limited to well-defined regions, depending on their origin location. Three main regions: the south-western, the south-eastern and the northern parts of the basin are characterized by AEs recirculation, with sporadic export of eddies to the other WMED zones.

Natural fibres present ozone-friendly solutions in the field of construction. The attenuation of the sound and heat losses is an important feature in such type of materials above all, when used in non-woven fabrics and fibre-reinforced composites. Hemp fibres show robust insulation performance; this research work should be considered beneficial to the development of a non-destructive thermographic methodology, which can address the thermal barrier (typically applied on multi-layer panel) effects. The intent is to assess the integrity of the sub-surface reinforcing glass fibres; such integrity state will help confer the rigidity and the resistance to mechanical stresses. The testing proposed in this study can be further developed in a laboratory right after the manufacturing process of similar type of components. The testing needs preliminary numerical simulations to help guide the selection of the appropriate pre- and post-processing algorithms combined with or without segmentation operators. A set of numerical and experimental tests were performed through controlled thermal stimulation while recording the thermal responses. The study also highlights the advantages, disadvantages, and future development of the presented technique and methodologies.

K. pneumoniae (KPN) is one of the widest spread bacteria in which combined resistance to several antimicrobial groups is frequent. The most common β-lactamases found in K. pneumoniae are class A carbapenemases, both chromosomal-encoded (i.e., NMCA, IMI-1) and plasmid-encoded (i.e., GES-enzymes, IMI-2), VIM, IMP, NDM, OXA-48, and extended-spectrum β-lactamases (ESBLs) such as CTX-M enzymes. In the present study, a total of 68 carbapenem-resistant KPN were collected from twelve long-term care facilities (LTCFs) in the Northern Italian region. The whole-genome sequencing (WGS) of each KPN strain was determined using a MiSeq Illumina sequencing platform and analysed by a bacterial analysis pipeline (BAP) tool. The WGS analysis showed the prevalence of ST307, ST512, and ST37 as major lineages diffused among the twelve LTCFs. The other lineages found were: ST11, ST16, ST35, ST253, ST273, ST321, ST416, ST1519, ST2623, and ST3227. The blaKPC-2, blaKPC-3, blaKPC-9, blaSHV-11, blaSHV-28, blaCTX-M-15, blaOXA-1, blaOXA-9, blaOXA-23, qnrS1, qnrB19, qnrB66, aac(6′)-Ib-cr, and fosA were the resistance genes widespread in most LTCFs. In this study, we demonstrated the spreading of thirteen KPN lineages among the LTCFs. Additionally, KPC carbapenemases are the most widespread β-lactamase.

Four NDM-1 mutants (L218T, L221T, L269H and L221T/Y229W) were generated in order to investigate the role of leucines positioned in L10 loop. A detailed kinetic analysis stated that these amino acid substitutions modified the hydrolytic profile of NDM-1 against some β-lactams. Significant reduction of kcat values of L218T and L221T for carbapenems, cefazolin, cefoxitin and cefepime was observed. The stability of the NDM-1 and its mutants was explored by thermofluor assay in real-time PCR. The determination of TmB and TmD demonstrated that NDM-1 and L218T were the most stable enzymes. Molecular dynamic studies were performed to justify the differences observed in the kinetic behavior of the mutants. In particular, L218T fluctuated more than NDM-1 in L10, whereas L221T would seem to cause a drift between residues 75 and 125. L221T/Y229W double mutant exhibited a decrease in the flexibility with respect to L221T, explaining enzyme activity improvement towards some β-lactams. Distances between Zn1-Zn2 and Zn1-OH- or Zn2-OH- remained unaffected in all systems analysed. Significant changes were found between Zn1/Zn2 and first sphere coordination residues.

Fibre metal laminates are widely implemented in the aerospace industry owing to the merits of fatigue resistance and plastic properties. An effective defect assessment technique needs to be investigated for this type of composite materials. In order to achieve accurate impact-induced damage evaluation, a multi-excitation infrared fusion method is introduced in this study. Optical excitation thermography with high performance on revealing surface and subsurface defects is combined with vibro-thermography to improve the capability of detection on defects. Quantitative analysis is carried out on the temperature curve to assess the impact-induced deformation. A new image fusion framework including feature extraction, feature selection and fusion steps is proposed to fully utilize the information from two excitation modalities. Six fibre metal laminates which contain aluminium-basalt fibre reinforced plastic and aluminium-glass fibre reinforced plastic are investigated. Features from different perspectives are compared and selected via intensity contrast on deformation area for fusion imaging. Both types of defects (i.e., surface and sub-surface) and the internal deformation situation of these six samples are characterized clearly and intuitively.

The dramatic intensification of antimicrobial resistance occurrence in pathogenic bacteria concerns the global community. The revitalisation of inactive antibiotics is, at present, the only way to go through this health system crisis and the use of antimicrobial adjuvants is turning out the most promising approach. Due to their low toxicity, eco-friendly characteristics and antimicrobial activity, amphoteric surfactants are good candidates. This study investigated the adjuvant potentialities of commercial acyclic and newly cyclic N-oxide surfactants combined with therapeutically available antibiotics against MDR methicillin-resistant Staphylococcus aureus (MRSA). The safety profile of the new cyclic compounds, compared to commercial surfactants, was preliminarily assessed, evaluating the cytotoxicity on human peripheral mononuclear blood cells and the haemolysis in human red blood cells. The compounds show an efficacious antimicrobial activity strongly related to the length of the carbon atom chain. In drug–drug interaction assays, all surfactants act synergistically, restoring sensitivity to oxacillin in MRSA, with dodecyl acyclic and cyclic derivatives being the most effective. After evaluating the cytotoxicity and considering the antimicrobial action, the most promising compound is the L-prolinol amine-oxide C12NOX. These findings suggest that the combination of antibiotics with amphoteric surfactants is a valuable therapeutic option for topical infections sustained by multidrug-resistant S. aureus.

The ability to form biofilms is a recognized trait of Stenotrophomonas maltophilia, but the extent of its clinical relevance is still unclear. The present multicenter prospective study (ANSELM) aims at investigating the association between biofilm formation and clinical outcomes of S. maltophilia infections. One hundred and nine isolates were collected from various geographical origins and stratified according to their clinical relevance. Biofilm formation was evaluated by the microtiter plate assay and correlated with microbiological and clinical data from the associated strains. Antibiotic susceptibility of the planktonic cells was tested by the disk diffusion technique, while antibiotic activity against mature biofilms was spectrophotometrically assessed. Most strains (91.7%) were able to form biofilm, although bloodborne strains produced biofilm amounts significantly higher than strains causing hospital- rather than community-acquired infections, and those recognized as “definite” pathogens. Biofilm formation efficiency was positively correlated with mechanical ventilation (p = 0.032), whereas a negative relationship was found with antibiotic resistance (r2 = 0.107; p < 0.001), specifically in the case of the pathogenic strains. Mature S. maltophilia biofilms were markedly more resistant (up to 128 times) to cotrimoxazole and levofloxacin compared with their planktonic counterparts, especially in the case of bloodborne strains. Our findings indicate that biofilm formation by S. maltophilia is obviously a contributing factor in the pathogenesis of infections, especially in deep ones, thus warranting additional studies with larger cohort of patients and isolates.

Oil spill models are used to simulate the evolution of an oil slick that occurs after an accidental ship collision, malfunctioning of oil extraction platforms, or illegal discharges intentionally released by ships into the marine environment. We present an integrated operational oil spill prediction system that improves capacities in preventing and mitigating maritime risks from oil spills. The objective is to provide forecast information about the transport and the fate of a hypothetical oil spill under Nearly-Real Time hydrodynamic conditions in the western and central Mediterranean Sea. This complex forecast system is developed in the framework of the project SOS-Piattaforme & Impatti Off-Shore to the needs of Italian Coast Guard and other institutions, such as the Ministry of the Environment. This service has been operational since July 2020. The innovative aspect of this work is a graphical user interface (the GUI), which allows to select properties, time, and location of a potential oil spill and show the evolution of oil slick concentration and oil fate parameters. This platform represents the first component of a future Decision Support System aimed to identify the risk assessment of oil spills in order to better manage emergencies and minimize economic damages.

Nowadays, infrared thermography, as a widely used non-destructive testing method, is increasingly studied for impact evaluation of composite structures. Sparse pattern extraction is attracting increasing attention as an advanced post-processing method. In this paper, an enhanced sparse pattern extraction framework is presented for thermographic sequence processing and defect detection. This framework adapts cropping operator and typical component extraction as a preprocessing step to reduce the dimensions of raw data and applies sparse pattern extraction algorithms to enhance the contrast on the defect area. Different cases are studied involving several defects in four basalt-carbon hybrid fiber-reinforced polymer composite laminates. Finally, comparative analysis with intensity distribution is carried out to verify the effectiveness of contrast enhancement using this framework.

New Delhi Metallo-β-lactamase-1 (NDM-1) is the most prevalent type of metallo-β-lactamase, able to hydrolyze almost all antibiotics of the β-lactam group, leading to multidrug-resistant bacteria. To date, there are no clinically relevant inhibitors to fight NDM-1. The use of dromedary polyclonal antibody inhibitors against NDM-1 represents a promising new class of molecules with inhibitory activity. In the current study, immunoreactivities of dromedary Immunoglobulin G (IgG) isotypes containing heavy-chain and conventional antibodies were tested after successful immunization of dromedary using increasing amounts of the recombinant NDM-1 enzyme. Inhibition kinetic assays, performed using a spectrophotometric method with nitrocefin as a reporter substrate, demonstrated that IgG1, IgG2, and IgG3 were able to inhibit not only the hydrolytic activity of NDM-1 but also Verona integron-encoded metallo-β-lactamase (VIM-1) (subclass B1) and L1 metallo-β-lactamase (L1) (subclass B3) with inhibitory concentration (IC50) values ranging from 100 to 0.04 μM. Investigations on the ability of IgG subclasses to reduce the growth of recombinant Escherichia coli BL21(DE3)/codon plus cells containing the recombinant plasmid expressing NDM-1, L1, or VIM-1 showed that the addition of IgGs (4 and 8 mg/L) to the cell culture was unable to restore the susceptibility of carbapenems. Interestingly, IgGs were able to interact with NDM-1, L1, and VIM-1 when tested on the periplasm extract of each cultured strain. The inhibitory concentration was in the micromolar range for all β-lactams tested. A visualization of the 3D structural basis using the three enzyme Protein Data Bank (PDB) files supports preliminarily the recorded inhibition of the three MBLs.

The disciplines of biosciences included in the curricula of a nursing degree represent a daunting but crucial type of knowledge that a well-prepared nurse should acquire. Given the importance and the objective difficulties of these courses, nursing students experience anxiety, especially for the anatomy course. This anxiety and the related rate of exam failures lead professors to analyze their teaching approach, by diversifying the lecturing methods. The aim of our study was to test the use of a virtual dissection table (DT) during the anatomy lectures of a nursing course, evaluating the anxiety level before the exam and evaluating the exam score. The feedback of the evaluated student population was positive overall. The integration of the DT in anatomy lectures improved the learning performance and mostly enhanced the self-confidence of the first year nursing students.

The relationship between wood and its degree of humidity is one of the most important aspects of its use in construction and restoration. The wood presents a behavior similar to a sponge, therefore, moisture is related to its expansion and contraction. The nondestructive evaluation (NDE) of the amount of moisture in wood materials allows to define, e.g., the restoration procedures of buildings or artworks. In this work, an integrated study of two non-contact techniques is presented. Infrared thermography (IRT) was able to retrieve thermal parameters of the wood related to the amount of water added to the samples, while the interference pattern generated by speckles was used to quantify the expansion and contraction of wood that can be related to the amount of water. In twenty-seven wooded samples, a known quantity of water was added in a controlled manner. By applying advanced image processing to thermograms and specklegrams, it was possible to determine fundamental values controlling both the absorption of water and the main thermophysical parameters that link the samples. On the one hand, results here shown should be considered preliminary because the experimental values obtained by IRT need to be optimized for low water contents introduced into the samples. On the other hand, speckle interferometry by applying an innovative procedure provided robust results for both high and low water contents.
