Background: Multiparametric MRI markers of liver health corrected T1 (cT1) and proton density fat fraction (PDFF) have shown utility in the management of various chronic liver diseases. We assessed the normal population reference range of both cT1 and PDFF in healthy child and adult volunteers without any known liver disease. Methods: A retrospective multi-centre pooled analysis of 102 child and young adult (9.1 years (6–18)) volunteers from three centres: Children’s Memorial Health Institute (N = 21), University Hospital Southampton (N = 28) and Hospital Infantil de Mexico (N = 53). Sex and ethnic differences were investigated for both cT1 and PDFF. Age effects were investigated with comparison to a pooled adult cohort from the UK Biobank (N = 500) and CoverScan (N = 71), covering an age range of 21 to 81 years. Results: cT1 values were normally distributed with a median of 748 ms (IQR: 725–768 ms; 2.5–97.5 percentiles: 683–820 ms). PDFF values followed a normal distribution with a median of 1.7% (IQR: 1.3–1.9%; 2.5–97.5 percentiles: 1–4.4%). There were no significant age and sex differences in cT1 and PDFF between children and young adults. No differences in cT1 and PDFF were found between ethnicities. Age comparisons showed statistically significant, but clinically negligible, cT1 (748 ms vs. 732 ms) and PDFF (2.4% vs. 1.9%) differences between paediatric and adult groups, respectively. Conclusions: Median healthy cT1 and PDFF reference ranges in children and young adults fall within the reported limits for normal of 800 ms and 5%, respectively.

The digital representation of seafloor, a challenge in UNESCO’s Ocean Decade initiative, is essential for sustainable development support and marine environment protection, aligning with the United Nations’ 2030 program goals. Accuracy in seafloor representation can be achieved through remote sensing measurements, including acoustic and laser sources. Ground truth information integration facilitates comprehensive seafloor assessment. The current seafloor mapping paradigm benefits from the object-based image analysis (OBIA) approach, managing high-resolution remote sensing measurements effectively. A critical OBIA step is the segmentation process, with various algorithms available. Recent artificial intelligence advancements have led to AI-powered segmentation algorithms development, like the Segment Anything Model (SAM) by META AI. This paper presents the SAM approach’s first evaluation for seafloor mapping. The benchmark remote sensing dataset refers to Puck Lagoon, Poland and includes measurements from various sources, primarily multibeam echosounders, bathymetric lidar, airborne photogrammetry, and satellite imagery. The SAM algorithm’s performance was evaluated on an affordable workstation equipped with an NVIDIA GPU, enabling CUDA architecture utilization. The growing popularity and demand for AI-based services predict their widespread application in future underwater remote sensing studies, regardless of the measurement technology used (acoustic, laser, or imagery). Applying SAM in Puck Lagoon seafloor mapping may benefit other seafloor mapping studies intending to employ AI technology.

The concept of homogeneity in the real estate market is a well-known analysis aspect, yet it remains a significant challenge in practical implementation. This study aims to fill this research gap by introducing the HELIOS concept (Homogeneity Estate Linguistic Intelligence Omniscient Support), presenting a new approach to real estate market analyses. In a world increasingly mindful of environmental, social, and economic concerns, HELIOS is a novel concept grounded in linguistic intelligence and machine learning to reshape how we perceive and analyze real estate data. By exploring the synergies between human expertise and technological capabilities, HELIOS aims not only to enhance the efficiency of real estate analyses but also to contribute to the broader goal of sustainable and responsible data practices in the dynamic landscape of property markets. Additionally, the article formulates a set of assumptions and suggestions to improve the effectiveness and efficiency of homogeneity analysis in mass valuation, emphasizing the synergy between human knowledge and the potential of machine technology.

This paper proposes a mathematical model for the shading profiles of a PV module with thin, long linear elements. The model includes the brightness distribution over the entire shading region (umbra, penumbra, and antumbra). A corresponding calculation code in the form of m-files has been prepared for the MATLAB environment. The input data for the calculations are the coordinates of the Sun’s position in the sky, the dimensions and spatial orientation of the shading element, and the spatial orientation of the shaded PV module. The correctness of the model was verified by a measurement experiment carried out under actual outdoor weather conditions. Statistical analysis of the comparison between the measurement data from the experiment and the model showed its high accuracy. As part of this research work, it was also checked how shading with thin linear elements affects the current–voltage characteristics of the module. It turned out that even a small linear shading could reduce the power output of the module by more than 6%, with the distribution of this shading across the individual cells of the module being extremely important.

Fluconazole (2-(2,4-difluorophenyl)-1,3-bis(1H-1,2,4-triazol-1-yl)propan-2-ol), which was patented in 1981 and introduced for commercial use in 1988, is a widely utilized antifungal drug whose mechanism of action involves inhibition of the activity of 14-α lanosterol demethylase. Its safety and effectiveness have established it as one of the most frequently employed antifungal agents. Resistance to azole antifungal drugs is becoming more common. It may be related to a mutation of the gene encoding the enzyme. To address this issue, molecules with modifications in three main regions of fluconazole, namely the hydroxyl group, the aromatic ring, and the 1,2,4-triazole rings, have been synthesized in an attempt to create more potent antifungal drugs. These modifications aim at enhancing the effectiveness against microorganisms and improving pharmacokinetic parameters and safety profiles of the synthesized compounds. The present review explores the synthesis of fluconazole derivatives, accompanied by insights into the results of biological studies evaluating the therapeutic effects of these compounds.

This article presents the results of an experiment to measure the mass of contaminants naturally deposited on the surface of photovoltaic modules. Six types of PV modules included in the installation located on the roof of the C3 building (AGH campus, Krakow, Poland) were tested. More than 120 contamination samples were collected during the experiment, which lasted from 23 March to 15 June 2022. Detailed analysis showed a clear relationship between the tilt angle of the photovoltaic modules and the amount of contamination accumulating on them. The impact of the backsheet color and the way the module was mounted (vertical or horizontal) on the amount of dirt deposited on a given module was also recorded. Because the experiment lasted for many weeks, it was possible to investigate the intensity of the contamination build-up over the following weeks (in the absence of module cleaning) and the effect of precipitation on the module self-cleaning. During one measurement, a layer of contamination with a normalized mass of 181 mg/m2 was found to reduce PV module peak power by almost 4%.

This work aims to engineer a new stable injectable Mn-based methacrylated gellan gum (Mn/GG-MA) hydrogel for real-time monitored cell delivery into the central nervous system. To enable the hydrogel visualization under Magnetic Resonance Imaging (MRI), GG-MA solutions were supplemented with paramagnetic Mn2+ ions before its ionic crosslink with artificial cerebrospinal fluid (aCSF). The resulting formulations were stable, detectable by T1-weighted MRI scans and also injectable. Cell-laden hydrogels were prepared using the Mn/GG-MA formulations, extruded into aCSF for crosslink, and after 7 days of culture, the encapsulated human adipose-derived stem cells remained viable, as assessed by Live/Dead assay. In vivo tests, using double mutant MBPshi/shi/rag2 immunocompromised mice, showed that the injection of Mn/GG-MA solutions resulted in a continuous and traceable hydrogel, visible on MRI scans. Summing up, the developed formulations are suitable for both non-invasive cell delivery techniques and image-guided neurointerventions, paving the way for new therapeutic procedures.

The treatment of infectious diseases is a challenging issue faced by the medical community. The emergence of drug-resistant strains of bacteria and fungi is a major concern. Researchers and medical professionals are working to develop new and innovative treatments for infectious diseases. Schiff bases are one a promising class of compounds. In this work, new derivatives were obtained of the 4-amino-5-(3-fluorophenyl)-2,4-dihydro-3H-1,2,4-triazole-3-thione reaction, with corresponding benzaldehydes with various substituents at position 4. The antibacterial and antifungal activities of all synthesized compounds were tested. Several new substances have shown moderate antifungal activity against Candida spp. The highest activity directed against C. albicans was shown by compound RO4, with a 4-methoxyphenyl moiety and an MIC value of 62.5 µg/mL. In order to check the toxicity of the synthesized compounds, their effect on cell lines was examined. Additionally, we tried to elucidate the mechanism of the antibacterial and antifungal activity of the tested compounds using molecular docking to topoisomerase IV, D-Alanyl-D-Alanine Ligase, and dihydrofolate reductase.

Soil fungi are crucial microorganisms in the functioning of ecosystems. They shape the soil properties, facilitate nutrient circulation, and assist with plant growth. However, their biogeography and distribution studies are limited compared to other groups of organisms. This review aims to provide an overview of the main factors shaping the spatial distribution of soil fungi (with a special focus on mycorrhizal fungi). The review also tries to identify the field frontier where further studies are needed. The main drivers of soil fungal distribution were classified and reviewed into three groups: soil properties, plant interactions, and dispersal vectors. It was apparent that ectomycorrhizal and arbuscular fungi are relatively overrepresented in the body of research, while the other mycorrhiza types and endophytes were grossly omitted. Notwithstanding, soil pH and the share of ectomycorrhizal plants in the plant coverage were repeatedly reported as strong predictors of mycorrhizal fungal distribution. Dispersal potential and vector preferences show more variation among fungi, especially when considering long-distance dispersal. Additionally, special attention was given to the applications of the island biogeography theory to soil fungal assemblages. This theory proves to be a very efficient framework for analyzing and understanding not only the soil fungal communities of real islands but even more effective islands, i.e., isolated habitats, such as patches of trees discontinuous from more enormous forests.

Purpose: Mannitol is a hyperosmolar agent for reducing intracranial pressure and inducing osmotic blood–brain barrier opening (OBBBO). There is a great clinical need for a non-invasive method to optimize the safety of mannitol dosing. The aim of this study was to develop a label-free Chemical Exchange Saturation Transfer (CEST)-based MRI approach for detecting intracranial accumulation of mannitol following OBBBO. Methods: In vitro MRI was conducted to measure the CEST properties of D-mannitol of different concentrations and pH. In vivo MRI and MRS measurements were conducted on Sprague-Dawley rats using a Biospec 11.7T horizontal MRI scanner. Rats were catheterized at the internal carotid artery (ICA) and randomly grouped to receive either 1 mL or 3 mL D-mannitol. CEST MR images were acquired before and at 20 min after the infusion. Results: In vitro MRI showed that mannitol has a strong, broad CEST contrast at around 0.8 ppm with a mM CEST MRI detectability. In vivo studies showed that CEST MRI could effectively detect mannitol in the brain. The low dose mannitol treatment led to OBBBO but no significant mannitol accumulation, whereas the high dose regimen resulted in both OBBBO and mannitol accumulation. The CEST MRI findings were consistent with 1H-MRS and Gd-enhanced MRI assessments. Conclusion: We demonstrated that CEST MRI can be used for non-invasive, label-free detection of mannitol accumulation in the brain following BBBO treatment. This method may be useful as a rapid imaging tool to optimize the dosing of mannitol-based OBBBO and improve its safety and efficacy.

Based on the results of previous work, we designed and synthesized 1,3,4-thiadiazole derivatives. The cytotoxic activity of the obtained compounds was then determined in biological studies using MCF-7 and MDA-MB-231 breast cancer cells and a normal cell line (fibroblasts). The results showed that all compounds displayed weak anticancer activity towards two breast cancer lines: an estrogen-dependent cell line (MCF-7) and an estrogen-independent cell line (MDA-MB-231). The compound most active towards MCF-7 breast cancer cells was SCT-4, which decreased DNA biosynthesis to 70% ± 3 at 100 µM. The mechanism of the anticancer action of 1,3,4-thiadiazole was also investigated. We choose a set of the most investigated proteins, which are attractive anticancer targets. In silico studies demonstrated a possible multitarget mode of action for the synthesized compounds but the most likely mechanism of action for the new compounds is connected with the activity of caspase 8.

Respiratory infections with newly emerging zoonotic viruses such as SARS-CoV-2, the etiological agent of COVID-19, often lead to the perturbation of the human innate and adaptive immune responses causing severe disease with high mortality. The responsible mechanisms are commonly virus-specific and often include either over-activated or delayed local interferon responses, which facilitate efficient viral replication in the primary target organ, systemic viral spread, and rapid onset of organ-specific and harmful inflammatory responses. Despite the distinct replication strategies, human infections with SARS-CoV-2 and highly pathogenic avian influenza viruses demonstrate remarkable similarities and differences regarding the mechanisms of immune induction, disease dynamics, as well as the long-term sequelae, which will be discussed in this review. In addition, we will highlight some important lessons about the effectiveness of antiviral and immunomodulatory therapeutic strategies that this pandemic has taught us.

The fact that personality traits play an important role when it comes to predicting people’s entrepreneurial behavior is currently indisputable. However, so far, the majority of subject literature has focused on employee characteristics in developed countries. To address this gap, research was conducted, including 188 entrepreneurs of small and medium enterprises (SME) and 21 highest-level employees in Poland—one of the countries with the most hostile and turbulent environment for entrepreneurial operations in the world. The five personality traits (Big Five) were evaluated using the 60-item scale. There were three objectives of this study: to identify the differences between entrepreneurs and the most effective, highest-level employees, in the context of personality trait intensities, to determine the level of specific trait(s) intensities (OCEAN) of an effective entrepreneur. Finally, to determine whether it is possible to construct a model based on BIG5 to estimate the probability of success as an entrepreneur. The research results imply there are significant differences between successful entrepreneurs and highly effective employees and their personality trait intensities, described in the BIG5 model. Moreover, it is possible to distinguish trait intensity ranges, determining the success as an entrepreneur. Finally, we constructed the empirically based model, which allows us to estimate the chances of an individual succeeding as an entrepreneur with only a 5% error rate. The main factor and common denominator of entrepreneurial effectiveness is the trait of extraversion. The findings of our study are particularly important for the renewable energy sector in Poland, as the last part of green energy implementing blockchain (e.g., the installation and maintenance of wind turbines and solar panels) is being undertaken, in vast majority, by independent contractors and SME owners (entrepreneurs).

Throughout the 16th and 17th centuries, many builders, artists, and architects living on the shores of Italian lakes decided to settle in Poland. Upon arrival, they pursued brilliant careers in various areas of life. Over time, they became Polonized. This was also the case for Giovanni Battista Trevano, who was active in Krakow in the first half of the 17th century and whose lifetime achievement was to become the royal architect of the Vasa kings. This article presents Trevano’s artistic oeuvre and provides insight into his social, economic, and intellectual status in the new community, including the architect’s offspring, who pursued successful careers in army, church, and state offices throughout the 17th and 18th centuries. These new findings are based on manuscripts that have recently been discovered by the author of the article in both Polish and Swiss archives. They allow for expanding the knowledge of the Trevano family’s genealogy and biography, and correcting some unjustified views in the discourse. On the basis of research on new archival sources, one can conclude that Giovanni Battista Trevano was a prominent architect, who is credited with introducing in Poland the early Baroque style, which soon became dominant in northern European art.

Amyotrophic lateral sclerosis (ALS) is a progressive, fatal disease with no effective therapy. The neurodegenerative character of ALS was an appealing target for stem cell-based regenerative approaches. Different types of stem cells have been transplanted in both preclinical and clinical settings, but no convincing outcomes have been noted. Human glial restricted precursors (hGRPs) transplanted intraventricularly to neonatal, immunodeficient mice rescued lifespan of dysmyelinated mice. Intraspinal injection of hGRPs also provided benefits in the mouse model of ALS. Therefore, we have recently developed an immunodeficient model of ALS (double mutant SOD1/rag2), and, in this study, we tested the strategy previously used in dysmyelinated mice of intraventricular transplantation of hGRPs to immunodeficient mice. To maximize potential therapeutic benefits, the cells were implanted into neonates. We used magnetic resonance imaging to investigate the progression of neurodegeneration and therapeutic responses. A cohort of animals was devoted to survival assessment. Postmortem analysis included immunohistochemistry, Nissl staining, and Western blots. Cell transplantation was not associated with improved animal survival, slowing neurodegeneration, or accumulation of misfolded superoxide dismutase 1. Postmortem analysis did not reveal any surviving hGRPs. Grafting into neonatal immunodeficient recipients did not prevent ALS-induced cell loss, which might explain the lack of positive therapeutic effects. The results of this study are in line with the modest effects of clinical neurotransplantations. Therefore, we urge stem cell and ALS communities to develop and implement cell tracking methods to better understand cell fates in the clinic.

Wilson disease (WD) is a liver disorder characterized by improper copper metabolism. Although non-invasive tools are currently used to support diagnosis and management, this is still an area of unmet need, as patients present with a wide range of symptoms. Our aim was to investigate the potential utility of multiparametric magnetic resonance imaging (mpMRI) and quantitative magnetic resonance cholangiopancreatography (MRCP+) to support patient management. MRI examinations of 7 children and young adults aged 8–16 years (six at diagnosis) were performed alongside a standard of care clinical and histological examination. Images were quantitatively analyzed to derive metrics of liver (corrected T1 (cT1; fibro-inflammation), MR liver fat (proton density fat fraction; PDFF)), and biliary health (MRCP+). MRI–PDFF provided a more dynamic characterization of fat compared with that provided by ultrasound. Those with elevated histological scores of fibrosis, inflammation, and steatosis had elevated mpMRI values. MRCP+ managed to identify dilatations in the biliary tree which were not observed during the standard of care examination. mpMRI and MRCP+ metrics show early promise as markers to assess both liver and biliary health in Wilson disease. Investigations to understand and explore the utility of these markers are warranted and should be performed.

We present a unique case of a young woman with acute myeloid leukemia (AML) with complex karyotype. The presence of the t(4;11)(q23;p15) is extremely rare in myeloid leukemias, while t(4;8)(q32;q13) has not yet been described in any leukemia reference. Another interesting issue is the familial aggregation of myeloid malignancies and worse course of the disease in each subsequent generation, as well as an earlier onset of the disease. Our report emphasizes the need for thorough pedigree examination upon myeloid malignancy diagnosis as there are relatives for whom counseling, gene testing, and surveillance may be highly advisable.

Over the years, the idea of corporate social responsibility has attracted the interest both of practitioners and researchers. While a discourse has occurred in the context of the meaning of Corporate Social Responsibility (CSR), there is no extensive analysis of the latest contributions. To address this gap, three objectives have been formulated for the purpose of this paper: to analyse development trends in CSR definitions, to assess the applicability of scientific efforts in the above-mentioned area and to demonstrate future research opportunities and threats based on the state-of-the-art in CSR. An organized literature review and bibliometric methods have been proposed based on 55 articles published in the “Corporate Social Responsibility and Environmental Management Journal”, the top ranked periodical with the highest Impact Factor, from its beginnings to January 2021. This research provides a clear overview of the recent CSR trends that are present in the literature, by pinpointing those works that refer to significant characteristics and indicate future trajectories of CSR in a managerial practical context, particularly important for the effectiveness of future CSR-oriented organizational activities. This paper summarises the existing CSR-related knowledge and constitutes the first attempt to investigate the process of the creation and development of the corporate social responsibility concept from its origins to its contemporary multidimensionality.

Background: Dysfunction of glia contributes to the deterioration of the central nervous system in a wide array of neurological disorders, thus global replacement of glia is very attractive. Human glial-restricted precursors (hGRPs) transplanted intraventricularly into neonatal mice extensively migrated and rescued the lifespan in half of the studied mice, whereas mouse GRPs (mGRPs) presented no therapeutic benefit. We studied in the same experimental setting canine GRPs (cGRP) to determine whether their therapeutic potential falls between hGRPs and mGRPs. Additional motivation for the selection of cGRPs was a potential for use in veterinary medicine. Methods: cGRPs were extracted from the brain of dog fetuses. The cells were transplanted into the anterior or posterior aspect of the lateral ventricle (LV) of neonatal, immunodeficient, dysmyelinated mice (Mbpshi, Rag2 KO; shiv/rag2). Outcome measures included early cell biodistribution, animal survival and myelination assessed with MRI, immunohistochemistry and electron microscopy. Results: Grafting of cGRP into posterior LV significantly extended animal survival, whereas no benefit was observed after anterior LV transplantation. In contrast, myelination of the corpus callosum was more prominent in anteriorly transplanted animals. Conclusions: The extended survival of animals after transplantation of cGRPs could be explained by the vicinity of the transplant near the brain stem.

Acoustic seafloor measurements with multibeam echosounders (MBESs) are currently often used for submarine habitat mapping, but the MBESs are usually not acoustically calibrated for backscattering strength (BBS) and cannot be used to infer absolute seafloor angular dependence. We present a study outlining the calibration and showing absolute backscattering strength values measured at a frequency of 150 kHz at around 10–20 m water depth. After recording bathymetry, the co-registered backscattering strength was corrected for true incidence and footprint reverberation area on a rough and tilted seafloor. Finally, absolute backscattering strength angular response curves (ARCs) for several seafloor types were constructed after applying sonar backscattering strength calibration and specific water column absorption for 150 kHz correction. Thus, we inferred specific 150 kHz angular backscattering responses that can discriminate among very fine sand, sandy gravel, and gravelly sand, as well as between bare boulders and boulders partially overgrown by red algae, which was validated by video ground-truthing. In addition, we provide backscatter mosaics using our algorithm (BBS-Coder) to correct the angle varying gain (AVG). The results of the work are compared and discussed with the published results of BBS measurements in the 100–400 kHz frequency range. The presented results are valuable in extending the very sparse angular response curves gathered so far and could contribute to a better understanding of the dependence of backscattering on the type of bottom habitat and improve their acoustic classification.

Neurological disorders (NDs) comprise a heterogeneous group of conditions that affect the function of the nervous system. Often incurable, NDs have profound and detrimental consequences on the affected individuals’ lives. NDs have complex etiologies but commonly feature altered gene expression and dysfunctions of the essential chromatin-modifying factors. Hence, compounds that target DNA and histone modification pathways, the so-called epidrugs, constitute promising tools to treat NDs. Yet, targeting the entire epigenome might reveal insufficient to modify a chosen gene expression or even unnecessary and detrimental to the patients’ health. New technologies hold a promise to expand the clinical toolkit in the fight against NDs. (Epi)genome engineering using designer nucleases, including CRISPR-Cas9 and TALENs, can potentially help restore the correct gene expression patterns by targeting a defined gene or pathway, both genetically and epigenetically, with minimal off-target activity. Here, we review the implication of epigenetic machinery in NDs. We outline syndromes caused by mutations in chromatin-modifying enzymes and discuss the functional consequences of mutations in regulatory DNA in NDs. We review the approaches that allow modifying the (epi)genome, including tools based on TALENs and CRISPR-Cas9 technologies, and we highlight how these new strategies could potentially change clinical practices in the treatment of NDs.

Protein crystallization can serve as a purification step in biotechnological processes but is often limited by the non-crystallizability of proteins. Enabling or improving crystallization is mostly achieved by high-throughput screening of crystallization conditions and, more recently, by rational crystal contact engineering. Two selected rational crystal contact mutations, Q126K and T102E, were transferred from the alcohol dehydrogenases of Lactobacillus brevis (LbADH) to Lactobacillus kefir (LkADH). Proteins were expressed in E. coli and batch protein crystallization was performed in stirred crystallizers. Highly similar crystal packing of LkADH wild type compared to LbADH, which is necessary for the transfer of crystal contact engineering strategies, was achieved by aligning purification tag and crystallization conditions, as shown by X-ray diffraction. After comparing the crystal sizes after crystallization of LkADH mutants with the wild type, the mean protein crystal size of LkADH mutants was reduced by 40–70% in length with a concomitant increase in the total amount of crystals (higher number of nucleation events). Applying this measure to the LkADH variants studied results in an order of crystallizability T102E > Q126K > LkADH wild type, which corresponds to the results with LbADH mutants and shows, for the first time, the successful transfer of crystal contact engineering strategies.

Cell therapy is a promising tool for treating central nervous system (CNS) disorders; though, the translational efforts are plagued by ineffective delivery methods. Due to the large contact surface with CNS and relatively easy access, the intrathecal route of administration is attractive in extensive or global diseases such as stroke or amyotrophic lateral sclerosis (ALS). However, the precision and efficacy of this approach are still a challenge. Hydrogels were introduced to minimize cell sedimentation and improve cell viability. At the same time, contrast agents were integrated to allow image-guided injection. Here, we report using manganese ions (Mn2+) as a dual agent for cross-linking alginate-based hydrogels and magnetic resonance imaging (MRI). We performed in vitro studies to test the Mn2+ alginate hydrogel formulations for biocompatibility, injectability, MRI signal retention time, and effect on cell viability. The selected formulation was injected intrathecally into pigs under MRI control. The biocompatibility test showed a lack of immune response, and cells suspended in the hydrogel showed greater viability than monolayer culture. Moreover, Mn2+-labeled hydrogel produced a strong T1 MRI signal, which enabled MRI-guided procedure. We confirmed the utility of Mn2+ alginate hydrogel as a carrier for cells in large animals and a contrast agent at the same time.

The aim of the study was to evaluate the influence of administration of aglepristone in mid-proestrus on progesterone concentration, LH release, and occurrence of ovulation in the bitch. Experimental bitches (n = 7) were treated on days 4 and 5 of proestrus with aglepristone at the dose of 10 mg/kg body weight s.c. (i.e., the two treatments were 24 h apart). Control animals (n = 7) received s.c. injections of saline. For progesterone determination, blood was collected daily until the first day of cytological diestrus. For LH determination, blood was collected daily and in the periovulatory phase every 8 h. The progesterone concentration showed a similar pattern in both groups. The LH peak value in bitches treated with aglepristone was significantly lower (p < 0.05) than that in control bitches (4.83 ± 1.20 vs. 13.66 ± 1.21 ng/mL). The area under the curve (AUC) for LH was significantly (p < 0.05) lower in treated than in control animals (6.85 ± 1.21 ng/mL/d vs. 12.25 ± 1.35 ng/mL/d). The ovulation occurred in all animals in both groups. The study showed that administration of aglepristone in the mid-proestrus significantly reduced the preovulatory LH surge, but it had no effect on progesterone concentration and the occurrence of ovulation.

Rapidly ageing populations are beset by tissue wear and damage. Stem cell-based regenerative medicine is considered a solution. Years of research point to two important aspects: (1) the use of cellular imaging to achieve sufficient precision of therapeutic intervention, and the fact that (2) many therapeutic actions are executed through extracellular vesicles (EV), released by stem cells. Therefore, there is an urgent need to interrogate cellular labels in the context of EV release. We studied clinically applicable cellular labels: superparamagnetic iron oxide nanoparticles (SPION), and radionuclide detectable by two main imaging modalities: MRI and PET. We have demonstrated effective stem cell labeling using both labels. Then, we obtained EVs from cell cultures and tested for the presence of cellular labels. We did not find either magnetic or radioactive labels in EVs. Therefore, we report that stem cells do not lose labels in released EVs, which indicates the reliability of stem cell magnetic and radioactive labeling, and that there is no interference of labels with EV content. In conclusion, we observed that direct cellular labeling seems to be an attractive approach to monitoring stem cell delivery, and that, importantly, labels neither locate in EVs nor affect their basic properties.

Among the many different applications of geothermal energy, its use for agricultural production purposes is noteworthy. The use of geothermal water in greenhouse cultivation is the most modern form of production; it enables the implementation of the production cycle throughout the year while reducing energy costs. In this paper, based on basic heat transfer relations, the heat demand of modern greenhouse constructions was determined. The monthly and year-round heat demand was determined for facilities with diversified heights of the side walls. Depending on the type of unit of greenhouse area analysed (1 ha), the calculated annual heat demand ranged from 11.05 to 12.46 TJ. Based on data on the geothermal energy potential in Poland, investment costs and payback periods were determined for the analysed locations. The locations of the facilities were selected based on traditions of plant cultivation in the greenhouse facilities and the availability of geothermal energy. The analysis showed that, under the adopted conditions, without additional co-financing, and in one of the considered locations, the investment will result in positive financial effects after 15 years. In the case of co-financing, at the level of currently existing values, all locations—irrespective of the assumed period of exploitation—showed satisfactory financial effects.

Protein crystallization can function as an effective method for protein purification or formulation. Such an application requires a comprehensive understanding of the intermolecular protein–protein interactions that drive and stabilize protein crystal formation to ensure a reproducible process. Using alcohol dehydrogenase from Lactobacillus brevis (LbADH) as a model system, we probed in our combined experimental and computational study the effect of residue substitutions at the protein crystal contacts on the crystallizability and the contact stability. Increased or decreased contact stability was calculated using molecular dynamics (MD) free energy simulations and showed excellent qualitative correlation with experimentally determined increased or decreased crystallizability. The MD simulations allowed us to trace back the changes to their physical origins at the atomic level. Engineered charge–charge interactions as well as engineered hydrophobic effects could be characterized and were found to improve crystallizability. For example, the simulations revealed a redesigning of a water mediated electrostatic interaction (“wet contact”) into a water depleted hydrophobic effect (“dry contact”) and the optimization of a weak hydrogen bonding contact towards a strong one. These findings explained the experimentally found improved crystallizability. Our study emphasizes that it is difficult to derive simple rules for engineering crystallizability but that free energy simulations could be a very useful tool for understanding the contribution of crystal contacts for stability and furthermore could help guide protein engineering strategies to enhance crystallization for technical purposes.

Astroviruses are single stranded, positive-sense RNA viruses that have been historically associated with diseases of the gastrointestinal tract of vertebrates, including humans. However, there is now a multitude of evidence demonstrating the capacity of these viruses to cause extraintestinal diseases. The most striking causal relationship is neurological diseases in humans, cattle, pigs, and other mammals, caused by astrovirus infection. Astroviruses have also been associated with disseminated infections, localized disease of the liver or kidneys, and there is increasing evidence suggesting a potential tropism to the respiratory tract. This review will discuss the current understanding of the tissue tropisms for astroviruses and their emerging capacity to cause disease in multiple organ systems.

Examples from the Polish clastic and carbonate reservoirs from the Central Polish Anticlinorium, Carpathians and Carpathian Foredeep are presented to illustrate possibilities of using well logging to geothermal resources recognition and characterization. Firstly, there was presented a short description of selected well logs and methodology of determination of petrophysical parameters useful in geothermal investigations: porosity, permeability, fracturing, mineral composition, elasticity of orogeny and mineralization of formation water from well logs. Special attention was allotted to spectral gamma-ray and temperature logs to show their usefulness to radiogenic heat calculation and heat flux modelling. Electric imaging and advanced acoustic logs provided with continuous information on natural and induced fracturing of formation and improved lithology recognition. Wireline and production logging were discussed to present the wealth of methods that could be used. A separate matter was thermal conductivity provided from the laboratory experiments or calculated from the results of the comprehensive interpretation of well logs, i.e., volume or mass of minerals composing the rocks. It was proven that, in geothermal investigations and hydrocarbon prospection, the same petrophysical parameters are considered, and well-logging acquisition equipment and advanced methods of processing and interpretation, developed and improved for almost one hundred years, can be successfully used in the detection and characterization of the potential geothermal reservoirs. It was shown that the newest (current investment)—as well as the old type (archive)—logs provide useful information.

Scientific exploration of seabed substrata has significantly progressed in the last few years. Hydroacoustic methods of seafloor investigation, including multibeam echosounder measurements, allow us to map large areas of the seabed with unprecedented precision. Through time-series of hydroacoustic measurements, it was possible to determine areas with distinct characteristics in the inlets of the Lagoon of Venice, Italy. Their temporal variability was investigated. Monitoring the changes was particularly relevant, considering the presence at the channel inlets of mobile barriers of the Experimental Electromechanical Module (MoSE) project installed to protect the historical city of Venice from flooding. The detection of temporal and spatial changes was performed by comparing seafloor maps created using object-based image analysis and supervised classifiers. The analysis included extraction of 25 multibeam echosounder bathymetry and backscatter features. Their importance was estimated using an objective approach with two feature selection methods. Moreover, the study investigated how the accuracy of classification could be affected by the scale of object-based segmentation. The application of the classification method at the proper scale allowed us to observe habitat changes in the tidal inlet of the Venice Lagoon, showing that the sediment substrates located in the Chioggia inlet were subjected to very dynamic changes. In general, during the study period, the area was enriched in mixed and muddy sediments and was depleted in sandy deposits. This study presents a unique methodological approach to predictive seabed sediment composition mapping and change detection in a very shallow marine environment. A consistent, repeatable, logical site-specific workflow was designed, whose main assumptions could be applied to other seabed mapping case studies in both shallow and deep marine environments, all over the world.

The article presents the results of research on the efficiency of photovoltaic (PV) modules cooled with water. The aim of the experiment was to improve the working conditions of solar modules. A temperature decrease was obtained for the PV module by pouring cool tap water onto the upper surface of the modules, either in imitation of rain or as a water film. The power of the cooled and non-cooled devices were then compared. The temperature of the cooled modules dropped to almost 25 °C, whilst the temperature of the non-cooled module was 45 °C. The best results were achieved by cooling modules with a water film, since there were no water splashes, and the continuous cooling of the surface leads to a 20% increase in power. During the test, the non-cooled module attained a maximum power of 105.3 W/m2, compared to 125.5 W/m2 for its cooled counterpart. Cooling the module, therefore, resulted in a power increase of 20.2 W/m2. The results of the work may be of particular interest for small installations, especially because it cleans the modules while providing an increase in power.

We need data sets of images and subjective scores to develop robust no reference (or blind) visual quality metrics for consumer applications. These applications have many uncontrolled variables because the camera creates the original media and the impairment simultaneously. We do not fully understand how this impacts the integrity of our subjective data. We put forward two new data sets of images from consumer cameras. The first data set, CCRIQ2, uses a strict experiment design, more suitable for camera performance evaluation. The second data set, VIME1, uses a loose experiment design that resembles the behavior of consumer photographers. We gather subjective scores through a subjective experiment with 24 participants using the Absolute Category Rating method. We make these two new data sets available royalty-free on the Consumer Digital Video Library. We also present their integrity analysis (proposing one new approach) and explore the possibility of combining CCRIQ2 with its legacy counterpart. We conclude that the loose experiment design yields unreliable data, despite adhering to international recommendations. This suggests that the classical subjective study design may not be suitable for studies using consumer content. Finally, we show that Hoßfeld−Schatz−Egger α failed to detect important differences between the two data sets.

Real estate management, including real estate market analysis, is part of a so-called geosystem. In recent years, the popularity of creating various types of systems and automatic solutions in real estate management, including those related to property classification and valuation, has been growing in the world, mainly to reduce the impact of human subjectivity, to increase the scope of analyses and reduce research time. A very important fact that should be underlined is that properties are strongly related to geolocation (space) and strongly determine it. Authors proposed in the paper solutions that highlight implementation of geoscience and “geo-approach” combined with fuzzy logic methods that allow to decrease subjectivity in property analyses and diminish uncertainty in decision making process. The proposed methodology involves three main problematic components of decision support system in property investment analyses development with the use of geo-technologies such as: determination of the database model; elaboration geo-property-zones with geoprocessing activities; identification of homogeneous group of properties transactions. The influence of spatial decision factor determined in the study lead to objective and precise calculation of value differentiation from 22 to 43% depending on the property’s remoteness to the sea.

Modern Global Navigation Satellite Systems (GNSS) allow for positioning with accuracies ranging from tens of meters to single millimeters depending on user requirements and available equipment. A major disadvantage of these systems is their unavailability or limited availability when the sky is obstructed. One solution is to use additional range measurements from ground-based nodes located in the vicinity of the receiver. The highest accuracy of distance measurement can be achieved using ultra wide band (UWB) or ZigBee phase shift measurement. The position of the additional transmitter must be carefully selected in order to obtain the optimal improvement in the dilution of precision (DOP), which reflects the improvement in the geometry of solution. The presented case study depicts a method for selecting the optimal location of a ground-based ranging source. It is based on a search of a minimum DOP value as a transmitter location function. The parameters of objective function are the elevation and azimuth of the transceiver. The solution was based on a limited-memory Broyden⁻Fletcher⁻Goldfarb⁻Shanno with Box constraints (L-BFGS-B) method and a numerical optimization algorithm for parameter value estimation. The presented approach allows for the selection of the optimal location of a ground-based source of ranging signals in GNSS processing from a geometry of solution point of view. This can be useful at the design stage of an augmentation network of ground-based transceivers. This article presents a theoretical basis and a case study presenting the selection of the optimal location of a ground-based ranging source.

Recently, the rapid development of the seabed mapping industry has allowed researchers to collect hydroacoustic data in shallow, nearshore environments. Progress in marine habitat mapping has also helped to distinguish the seafloor areas of varied acoustic properties. As a result of these new developments, we have collected a multi-frequency, multibeam echosounder dataset from the valuable nearshore environment of the southern Baltic Sea using two frequencies: 150 kHz and 400 kHz. Despite its small size, the Rowy area is characterized by diverse habitat conditions and the presence of red algae, unique on the Polish coast of the Baltic Sea. This study focused on the utilization of multibeam bathymetry and multi-frequency backscatter data to create reliable maps of the seafloor. Our approach consisted of the extraction of 70 secondary features of bathymetric and backscatter data, including statistic and textural attributes of different scales. Based on ground-truth samples, we have identified six habitat classes and selected the most relevant features of the bathymetric and backscatter data. Additionally, five types of image processing pixel-based and object-based classifiers were tested. We also evaluated the performance of algorithms using an accuracy assessment based on the validation subset of the ground-truth samples. Our best results reached 93% overall accuracy and a kappa coefficient of 0.90, confirming that nearshore seabed habitats can be accurately distinguished based on multi-frequency, multibeam echosounder measurements. Our predictive habitat mapping of shallow euphotic zones creates a new scientific perspective and provides relevant data for the management of natural resources. Object-based approaches previously used in various environments and areas suggest that methodology presented in this study may be scalable.

We investigate the nonautonomous difference equation with real initial conditions and coefficients g i , i = 0 , 1 which are in general functions of n and/or the state variables x n , x n − 1 , … , and satisfy g 0 + g 1 = 1 . We also obtain some global results about the behavior of solutions of the nonautonomous non-homogeneous difference equation where g i , i = 0 , 1 , 2 are functions of n and/or the state variables x n , x n − 1 , … , with g 0 + g 1 = 1 . Our results are based on the explicit formulas for solutions. We illustrate our results by numerous examples.

Haemolysin BL is an important virulence factor regarding the diarrheal type of food poisoning caused by Bacillus cereus. However, the pathogenic importance of this three-component enterotoxin is difficult to access, as nearly all natural B. cereus culture supernatants additionally contain the highly cytotoxic Nhe, the second three-component toxin involved in the aetiology of B. cereus-induced food-borne diseases. To better address the toxic properties of the Hbl complex, a system for overexpression and purification of functional, cytotoxic, recombinant (r)Hbl components L2, L1 and B from E. coli was established and an nheABC deletion mutant was constructed from B. cereus reference strain F837/76. Furthermore, 35 hybridoma cell lines producing monoclonal antibodies (mAbs) against Hbl L2, L1 and B were generated. While mAbs 1H9 and 1D8 neutralized Hbl toxicity and thus, represent important tools for future investigations of the mode-of-action of Hbl on the target cell surface, mAb 1D7, in contrast, even enhanced Hbl toxicity by supporting the binding of Hbl B to the cell surface. By using the specific mAbs in Dot blots, indirect and hybrid sandwich enzyme immuno assays (EIAs), complex formation between Hbl L1 and B, as well as L1 and L2 in solution could be shown for the first time. Surface plasmon resonance experiments with the rHbl components confirmed these results with KD values of 4.7 × 10−7 M and 1.5 × 10−7 M, respectively. These findings together with the newly created tools lay the foundation for the detailed elucidation of the molecular mode-of-action of the highly complex three-component Hbl toxin.
