Bovine milk proteins account for 10% of the global protein supply, which justifies the importance of thoroughly understanding their digestive processes. Extensive research on digestion is being conducted both in vivo and in vitro . However, interpretations and comparisons across different studies require a thorough understanding of the methodologies used. Both the rate and extent of milk protein digestion can be affected by several intrinsic and extrinsic factors with potential implications for overall digestibility and physiological responses. Among intrinsic factors, the impact of genetic variants in native milk proteins has emerged as a growing research area. To these, further complexity is added by the processing conditions frequently applied to milk prior to consumption. The main aim of this work is to provide an overview of the current knowledge on the impact of variations in milk protein profiles (particularly whey: casein ratio and protein polymorphisms), the treatments applied during processing (pasteurisation, homogenisation) and consumption (temperature changes) on protein digestion. To support the interpretation of the current literature, this manuscript also presents a historical perspective into research in this field and summarizes the protocols that are most frequently used, presently, on in vitro digestion studies.

The objective of this study was to investigate the effect of pH (6.1, 6.3, 6.5, and 6.7) on heat-induced changes in concentrated skim milk as related to fouling in heat exchangers. Skim milk (30%, w / w , total solids) was recirculated in a laboratory-scale fouling rig at an initial target temperature of 85 °C for 90 min to simulate thermal processing and preheating of evaporated liquid concentrate feeds in dairy processing. This study investigated key changes in relevant physicochemical properties, such as viscosity, particle size, and sedimentation, as major contributors to fouling at lower pHs (6.1 and 6.3). Additionally, protein aggregation and calcium phosphate precipitation were identified as significant contributors to fouling deposits. Possible strategies to mitigate fouling were determined, including optimizing pH and adjusting heat treatment parameters to minimize protein denaturation and mineral deposition. The findings indicate that carefully controlling pH and processing parameters can greatly enhance the efficiency of milk concentration by evaporation and tailor finished product quality. Moreover, this study showed that monitoring of CIP solutions for protein content and turbidity provides valuable information on the intensity of fouling and the efficiency of cleaning.

The purpose of this study was to examine the impact of a variety of supplemental feeds on the composition and quality of milk in a pasture-based dairy system. Four pasture-supplemented feeding systems were compared: Group 1 supplementation with 16% crude protein parlour concentrate (CONC); Group 2 supplementation with palm kernel expeller plus parlour concentrate (PKE); Group 3 supplemented with soya hulls plus parlour concentrate (SOYA); Group 4 was supplemented with molassed beet pulp plus parlour concentrate (BEET). Supplemental feeding system was demonstrated to have a significant effect on the size of native casein micelles and the gelation properties of milks. While CONC feeding produced significantly higher casein micelle size, gel strength (Young's Modulus) was significantly negatively correlated with casein micelle size. Supplemental feeding system had a significant effect on a number of fatty acids (FA) and indices derived therefrom, including total saturated and unsaturated fatty acids, de novo produced FA, omega 3, and omega 6 FA. The volatile profile of milks was also affected by supplemental feed choice, whereby multivariate analysis demonstrated that the CONC diet was distinctly different to that of the PALM, SOYA, and BEET milks. Multivariate analysis demonstrated that it is possible to distinguish milks from different pasture-supplemented feeding systems by their FA profile.
