Despite the huge potential of aquaculture in the Amazon, several producers have abandoned the activity due to a lack of assistance, technology, and innovation. Thus, the objective of this study was to identify factors that have contributed to the withdrawal of aquaculturists from the municipality of ‘Vigia de Nazaré’, state of Pará (Northern Region of Brazil). This case study took place in 2022 through a quanti-qualitative survey, applying structured questionnaires to former aquaculturists. A total of 30 fish farms were investigated, with 11 of them being abandoned. They are distributed across 10 rural communities that have developed fish farming, with ‘Vila de Itapuá’ (18.2%) being the most representative. When active, fish farming was practiced by men (100%) aged between 51 to 60 years (54.5%) with an incomplete primary education (100%), who carried out the activity for an average of 5 years (81.8%), quitting fish farming between the years 2019 and 2020 (72.7%). All former aquaculturists owned small properties, with excavated ponds in an extensive system and used family labor. The absence of technical assistance and the high cost of feed were pointed out as the main problems in the production chain. It is worth noting that the monoculture of Tambaqui (Colossoma macropomum) accounted for 63.6% of the species produced in rural communities. In conclusion, former aquaculturists express the desire to return to fish farming due to its socio-economic importance. For this, government actions supporting technical assistance and advanced studies in fish nutrition by educational, research, and extension institutions are necessary.

Background: The aim was to diagnose Candida in the oral cavity of subjects with type 2 diabetes mellitus (T2DM) using a genotyping technique and compare the results with those from conventional diagnosis by Papanicolaou (Pap) staining. Methods: Palatal mucosa smears were performed on 18 dental care patients diagnosed with T2DM and grade I, II, and III prosthetic stomatitis who met the inclusion criteria; 18 healthy control subjects were also included in the study. Hemoglobin A1c (HbA1c) levels were determined from total blood. Using exfoliative cytology, the Pap staining technique was used to diagnose candidiasis. Exfoliative cytology was also used for molecular diagnosis; DNA was obtained for Candida genotyping, and RNA was used for gene expression studies. Results: Clinical patterns indicated that all subjects were positive for Candida; however, Pap analysis revealed only three positive subjects, whereas end-point polymerase chain reaction (PCR) analysis revealed 15 subjects with some type of Candida. The most common Candida species found were Candida guilliermondii (38.8%), Candida krusei (33.3%), Candida tropicalis, and Candida lusitaniae (22.2%). Interestingly, the coexpression of different species of Candida was found in various patients. In all patients, HbA1c levels were increased. Gene expression analysis showed a significant decrease (p ≤ 0.05) in TLR2 expression in positive subjects, whereas TLR4 expression did not differ significantly among patients. Conclusions: The end-point PCR technique showed better sensitivity for the diagnosis of Candida when compared with the diagnosis by Pap staining. T2DM subjects showed an increased presence of C. guilliermondii that was correlated with decreased TLR2 expression.

This research developed smart integrated hybrid renewable systems for small energy communities and applied them to a real system to achieve energy self-sufficiency and promote sustainable decentralized energy generation. It compares stand-alone (SA) and grid-connected (GC) configurations using a developed optimized mathematical model and data-driven optimization, with economic analysis of various renewable combinations (PV, Wind, PHS, BESS, and Grid) to search for the optimal solution. Four cases were developed: two stand-alone (SA1: PV + Wind + PHS, SA2: PV + Wind + PHS + BESS) and two grid-connected (GC1: PV + PHS + Grid, GC2: Wind + PHS + Grid). GC2 shows the most economical with stable cash flow (−€123.2 annually), low CO2 costs (€367.2), and 91.7% of grid independence, requiring 125 kW of installed power. While GC options had lower initial investments (between €157k to €205k), the SA configurations provided lower levelized costs of energy (LCOE) ranging from €0.039 to €0.044/kWh. The integration of pumped hydropower storage enhances energy independence, supporting peak loads for up to two days with a storage capacity of 2.17 MWh.

Hydrogen production through electrolysis using renewable resources is highly promising for reducing greenhouse gas emissions. While significant efforts have focused on developing more efficient and cost-effective catalysts to lower hydrogen production costs, catalysts are not the primary expense in electrolyzer fabrication. In the case of Proton Exchange Membrane Water Electrolyzers (PEMWEs), other components—such as the proton membrane, gas diffusion layer (GDL), and bipolar plates—contribute more to overall costs. To explore this, a study was conducted on the performance of PEMWEs with various carbon paper GDLs, developed in the lab, on the cathodic side. This study examined how properties like electrical conductivity, porosity, and gas permeability affect performance. These findings emphasize the need to optimize components beyond catalysts to improve the cost-effectiveness of hydrogen production through electrolysis.

Air exchange in pressurized water pipelines is an essential but complex aspect of pipeline modeling and operation. Implementing effective air management strategies can yield numerous benefits, enhancing the system’s energy efficiency, reliability, and safety. This paper comprehensively evaluates an irregular profile pipeline filling procedure involving air-release through an air valve. The analysis includes real-time data tests and numerical simulations using Computational Fluid Dynamics (CFD). A Digital Twin model was proposed and applied to filling maneuvers in water installations. In particular, this research considers an often-overlooked aspect, such as filling a pipe with an irregular profile rather than a simple straight pipe. CFD simulations have proven to capture the main features of the transient event, which are suitable for tracking the air-water interface, the unsteady water flow, and the evolution of the trapped air pocket. Thus, they provide thorough and reliable information for real-time operational processes in the industry, focusing on the filling pressure and geometry of the air-valve hydraulic system. Additionally, this study provides details regarding the application of an efficient Digital Twin CFD approach, demonstrating its feasibility in optimizing the filling procedure in pipes with irregular profiles.

A thorough literature review was conducted on the effects of free surface oscillation in open channels, highlighting the risks of the occurrence of positive and negative surge waves that can lead to overtopping. Experimental analyses were developed to focus on the instability of the flow due to constrictions, gate blockages, and the start-up and shutdown of hydropower plants. A forebay at the downstream end of a tunnel or canal provides the right conditions for the penstock inlet and regulates the temporary demand of the turbines. In tests with a flow of 60 to 100 m3/h, the effects of a gradually and rapidly varying flow in the free surface profile were analyzed. The specific energy and total momentum are used in the mathematical characterization of the boundaries along the free surface water profile. A sudden turbine stoppage or a sudden gate or valve closure can lead to hydraulic drilling and overtopping of the infrastructure wall. At the same time, a PID controller, if programmed appropriately, can reduce flooding by 20–40%. Flooding is limited to 0.8 m from an initial amplitude of 2 m, with a dissipation wave time of between 25 and 5 s, depending on the flow conditions and the parameters of the PID characteristics.

Background/Objectives: 8-hydroxy-2′-deoxyguanosine (8-OHdG) is a form of oxidative DNA damage caused by oxidative stress (OS), which is considered a major factor in male infertility. The cellular defense system against 8-OHdG involves base excision repair (BER) with the enzyme 8-Oxoguanine DNA glycosylase 1 (OGG1). However, studies on the single-nucleotide polymorphism (SNP) OGG1 Ser326Cys have demonstrated that the Cys326Cys genotype could be the cause of an increment in oxidative DNA damage. In this study, the OGG1 Ser326Cys polymorphism and its effect on DNA oxidation were evaluated in 118 infertile men. Methods: Polymorphic screening was performed using TaqMan allelic discrimination assays, and oxidative DNA damage was evaluated through the quantification of 8-OHdG and total antioxidant capacity (TAC); in addition, electrical bioimpedance spectroscopy (EBiS) measurements were used as a reference for different electrical properties associated with 8-OHdG concentrations. Results: The detected Cys (G) allele frequency (0.4) was higher compared to the allele frequency reported in the “Allele Frequency Aggregator” (ALFA) and “Haplotype Map” (HapMap) projects for American populations (0.21–0.29), suggesting that the Cys (G) allele carrier could be a factor associated with American infertile populations. The values of 8-OHdG were twofold higher in carriers of the Cys326Cys (GG) genotype than the other genotypes and, in concordance, the TAC levels were threefold lower in Cys326Cys (GG) genotype carriers compared to the other genotypes. Moreover, the EBiS magnitude exhibited potential for the detection of different oxidative damage in DNA samples between genotypes. Conclusions: The Cys326Cys (GG) genotype is associated with oxidative DNA damage that could contribute to male infertility.

Integrated crop–livestock systems (ICLS) have sustainably intensified modern agricultural practices worldwide. This research assessed how production systems and crop types impact the chemical properties of an Oxisol in the Brazilian Cerrado, the grain yield of corn intercropped with palisade grass (Urochloa) in the off-season in an ICLS, and the grain yield (GY) of soybean in succession. Intercropped and monocropped systems were assessed in a three-year field experiment: corn + Urochloa ruziziensis–soybean; corn + U. brizantha cv. Piatã–soybean; corn + U. brizantha cv. Paiaguás–soybean (ICL–Paiaguás); corn–soybean under a no-tillage system (NTS); corn–soybean under a conventional tillage system (CTS); Piatã grass–continuous grazing (Perennial Piatã); and Paiaguás grass–continuous grazing (Perennial Paiaguás). The residual impact of phosphate fertilization was more pronounced in the ICLS treatments. In the soil layer from 0.0 to 0.2 m depth, ICLS–Paiaguás and Perennial Piatã had the most positive effects on soil chemical quality. In the last year, grain yield was highest in corn monoculture under the NTS and soybean in succession under the ICLS. ICL–Paiaguás improved soil chemical properties for soybean in succession. These results confirm that an intermittent pasture system for legume crops in sequence is an alternative that can maintain or improve soil chemical composition, and that CTS should be avoided in tropical sandy soils.

Oysters are sessile, filter-feeding bivalve molluscs widely distributed in estuarine and coastal habitats worldwide. They constitute a valuable resource for fisheries and extensive aquaculture and provide essential ecological services. Yet, their genetic diversity and distribution remain understudied. The variability in shell morphology complicates species classification, which is influenced by environmental and genetic factors. Although molecular phylogenetics research has refined oyster taxonomy and identified approximately 100 extant species, numerous taxonomic uncertainties persist. In the present study, we aimed to document the occurrence of small flat oysters of the genus Ostrea along the Mediterranean coastal areas of Liguria and Sardinia (Italy). Specifically, 16S rRNA sequence data were used to identify Ostrea species. Our findings offer novel insights into the O. stentina species complex and O. neostentina, a new species in the Mediterranean coastal areas of Italy. The study data further our understanding of Ostrea species diversity, distribution, and evolutionary patterns.

A new methodology, called HY4RES models, includes hybrid energy solutions (HESs) based on the availability of renewable sources, for 24 h of water allocation, using WaterGEMS 10.0 and PVGIS 5.2 as auxiliary calculations. The optimization design was achieved using Solver, with GRG nonlinear/evolutionary programming, and Python, with the non-dominated sorting genetic algorithm (NSGA-II). The study involves the implementation of complex multi-objective and multi-variable algorithms with different renewable sources, such as PV solar energy, pumped hydropower storage (PHS) energy, wind energy, grid connection energy, or battery energy, and also sensitivity analyses and comparisons of optimization models. Higher water allocations relied heavily on grid energy, especially at night when solar power was unavailable. For a case study of irrigation water needs of 800 and 1000 m3/ha, the grid is not needed, but for 3000 and 6000 m3/ha, grid energy rises significantly, reaching 5 and 14 GWh annually, respectively. When wind energy is also integrated, at night, it allows for reducing grid energy use by 60% for 3000 m3/ha of water allocation, yielding a positive lifetime cashflow (EUR 284,781). If the grid is replaced by batteries, it results in a lack of a robust backup and struggles to meet high water and energy needs. Economically, PV + wind + PHS + grid energy is the most attractive solution, reducing the dependence on auxiliary sources and benefiting from sales to the grid.

The aim of this study was to evaluate the effect of genetic groups and diets with different sources of nonprotein nitrogen (NPN) on the carcass and meat characteristics of beef heifers. The meat from 40 heifers (20 ½ Angus ½ Nellore (A × N) and 20 ½ Charolais ½ Nellore (L × N)), finished in feedlots, was used. The heifers were fed diets containing different sources of NPN—(1) a diet with livestock urea and protected urea (LPU) and (2) a diet with extruded urea (EU)—in a completely randomized design with a 2 × 2 factorial arrangement. Carcass, composition and meat quality evaluations were carried out. There were no significant interactions between diet and genetic group for most of the variables evaluated (p > 0.05). The A × N heifers had higher hot carcass weights (305.73 vs. 279.80 kg), loin eye areas (80.87 vs. 75.45 cm2), subcutaneous fat thicknesses (8.69 vs. 6.35 mm) and lower shear forces (6.98 vs. 7.7 kg) compared to the C × N heifers (p < 0.05). The meat from the A × N heifers had higher proportions of saturated fatty acids (49.41 vs. 47.95%), with no effects on the proportions of monounsaturated (47.57%) and polyunsaturated (4.01%) fatty acids. The A × N heifers had better carcass and meat characteristics, while the C × N heifers had meat and fat with better fatty acid profiles.

A new methodology for hybrid energy systems (HESs) was developed, namely the HY4RES model, tailored for the water sector, covering hybrid energy objective functions and grid or battery support using optimization algorithms in Solver, MATLAB, and Python, with evolutionary methods. HOMER is used for hybrid microgrids and allows for comparison with HY4RES, the newly developed model. Both models demonstrated flexibility in optimizing hybrid renewable solutions. This study analyzed an irrigation system for 3000 m3/ha (without renewables (Base case) and the Proposed system—with PV solar and pumped-hydropower storage to maximize cash flow over 25 years). Case 1—3000 m3/ha presented benefits due to PV supplying ~87% of energy, reducing grid dependency to ~13%. Pumped-hydropower storage (PHS) charges with excess solar energy, ensuring 24 h irrigation. Sensitivity analyses for Case 2—1000—and Case 3—6000 m3/ha—highlighted the advantages and limitations of water-energy management and system optimization. Case 2 was the most economical due to lower water-energy needs with noteworthy energy sales (~73.4%) and no need for the grid. Case 3 led to increased operating costs relying heavily on grid energy (61%), with PV providing only 39%. PHS significantly lowered operating costs and enhanced system flexibility by selling excess energy to the grid.

In recent years, various solutions have been developed to control power electronic converters using devices available on the market that are powerful and easy to use. These solutions, in most cases, offer high performance. However, these have high implementation costs because the required devices are expensive. For this reason, this document presents the implementation of two discrete-time controllers widely used in the literature for a boost converter implemented on a low-cost platform. The objective is to obtain a constant voltage at the converter’s output for photovoltaic system applications. The proportional-integral control is implemented as the first case, and the second case is a sliding mode control. In addition, a prior analysis is presented through simulation. Both control algorithms are implemented on the TMS320F28379D microcontroller from Texas Instruments through the same manufacturer’s integrated development software based on an optimized C/C++ language compiler. The results of the non-linear algorithm reveal better performance in reducing the time response, the overshoot of the transient state, and the steady-state error. Finally, the significant economic savings associated with the implementation costs of the controllers tested on a low-cost platform differentiate this work from other similar ones.

Water utilities are concerned about the issue of pipeline collapses, as service interruptions lead to water shortages. Pipeline collapses can occur during the maintenance phase when water columns compress entrapped air pockets, consequently increasing the pressure head. Analysing entrapped air pockets is complex due to the necessity of numerically solving a system of differential equations. Currently, water utilities need more tools to perform this analysis effectively. This research provides a numerical solution to the problem of entrapped air pockets in pipelines which can be utilised to predict filling operations. The study develops an analytical solution to examine the filling process. A practical application is shown, considering a 600 m long pipeline with an internal diameter of 400 mm. Compared with existing mathematical models, the results of the new analytical equations demonstrate their effectiveness as a new tool for computing the main hydraulic and thermodynamic variables involved in this issue.

Gymnema sylvestre (GS) and berberine (BBR) are natural products that have demonstrated therapeutic potential for the management of obesity and its comorbidities, as effective and safe alternatives to synthetic drugs. Although their anti-obesogenic and antidiabetic properties have been widely studied, comparative research on their impact on the gene expression of adipokines, such as resistin (Res), omentin (Ome), visfatin (Vis) and apelin (Ap), has not been reported. Methodology: We performed a comparative study in 50 adult Mexican patients with obesity treated with GS or BBR for 3 months. The baseline and final biochemical parameters, body composition, blood pressure, gene expression of Res, Ome, Vis, and Ap, and safety parameters were evaluated. Results: BBR significantly decreased (p < 0.05) body weight, blood pressure and Vis and Ap gene expression and increased Ome, while GS decreased fasting glucose and Res gene expression (p < 0.05). A comparative analysis of the final measurements revealed a lower gene expression of Ap and Vis (p < 0.05) in patients treated with BBR than in those treated with GS. The most frequent adverse effects in both groups were gastrointestinal symptoms, which attenuated during the first month of treatment. Conclusion: In patients with obesity, BBR has a better effect on body composition, blood pressure, and the gene expression of adipokines related to metabolic risk, while GS has a better effect on fasting glucose and adipokines related to insulin resistance, with minimal side effects.

Hydrogels consist of a network of highly porous polymeric chains with the potential for use as a wound dressing. Propolis is a natural product with several biological properties including anti-inflammatory, antibacterial and antioxidant activities. This study was aimed at synthesizing and characterizing a polyacrylamide/methylcellulose hydrogel containing propolis as an active ingredient, to serve as a wound dressing alternative, for the treatment of skin lesions. The hydrogels were prepared using free radical polymerization, and were characterized using scanning electron microscopy, infrared spectroscopy, thermogravimetry, differential scanning calorimetry, swelling capacity, mechanical and rheological properties, UV-Vis spectroscopy, antioxidant activity by the DPPH, ABTS and FRAP assays and biocompatibility determined in Vero cells and J774 macrophages by the MTT assay. Hydrogels showed a porous and foliaceous structure with a well-defined network, a good ability to absorb water and aqueous solutions simulating body fluids as well as desirable mechanical properties and pseudoplastic behavior. In hydrogels containing 1.0 and 2.5% propolis, the contents of total polyphenols were 24.74 ± 1.71 mg GAE/g and 32.10 ± 1.01 mg GAE/g and those of total flavonoids 8.01 ± 0.99 mg QE/g and 13.81 ± 0.71 mg QE/g, respectively, in addition to good antioxidant activity determined with all three methods used. Therefore, hydrogels containing propolis extract, may serve as a promising alternative wound dressing for the treatment of skin lesions, due to their anti-oxidant properties, low cost and availability.

In 2030, the world population will exceed 8.5 billion, increasing the challenges to satisfy basic needs for food, shelter, water, and/or energy. Irrigation plays a vital role in productive and sustainable agriculture. In the current context, it is determined not only by water availability but also by optimal management. Several authors have attempted to measure the performance of irrigation networks through various approaches in terms of technical indicators. To improve the sustainability in the pipe sizing of the pressurised irrigation networks, 25 different models were evaluated to discuss the advantages and disadvantages to consider in future methodologies to size water systems, which guarantee the network operation but contribute to improving the sustainability. They enable water managers to use them as tools to reduce a complex evaluation of the performance of a system, and focusing on better management of resources and sustainability indicators for agricultural ecosystems are clear and objective values.

Soy is Brazil’s main commodity, and due to its importance, farmers have adopted several technological innovations, such as light supplementation, to increase grain productivity and quality. However, the use of artificial light can interfere with weed dynamics and herbicide efficiency. The objective of this work was to evaluate the effect of herbicides on weed control and qualitative and quantitative attributes of soybeans grown under light supplementation. A field experiment was set up in the city of Monte Carmelo, MG, Brazil. The treatments were distributed in plots mounted under a central pivot in two environments: with light supplementation (LED panels) and without light supplementation. The treatments were distributed across four blocks, and plots were subdivided in space. The herbicides used were glyphosate, diclosulam and glyphosate, s-metolachlor and glyphosate, fomesafen + clethodim, flumioxazin + clethodim. The variables analyzed were the number of weeds, biomass, and control; chlorophyll levels; initial fluorescence; ratio of variable fluorescence to maximum fluorescence; and soybean electron transport rate—indicators of soybean yield. Light supplementation increases the density and biomass of weeds and impairs the effectiveness of herbicides. However, light supplementation with LED panels installed on the central irrigation pivot promotes increased productivity and improvements in the quantitative and qualitative parameters of soybeans. Due to these benefits, light supplementation can be considered advantageous for irrigated soybean production.

Previous studies have shown that some lamellarin-resembling annelated azaheterocyclic carbaldehydes and related imino adducts, sharing the 1-phenyl-5,6-dihydropyrrolo[2,1-a]isoquinoline (1-Ph-DHPIQ) scaffold, are cytotoxic in some tumor cells and may reverse multidrug resistance (MDR) mediated by P-glycoprotein (P-gp). Herein, several novel substituted 1-Ph-DHPIQ derivatives were synthesized which carry carboxylate groups (COOH, COOEt), nitrile (CN) and Mannich bases (namely, morpholinomethyl derivatives) in the C2 position, as replacements of the already reported aldehyde group. They were evaluated for antiproliferative activity in four tumor cell lines (RD, HCT116, HeLa, A549) and for the ability of selectively inhibiting P-gp-mediated MDR. Lipophilicity descriptors and molecular docking calculations helped us in rationalizing the structure–activity relationships in the P-gp inhibition potency of the investigated 1-Ph-DHPIQs. As a main outcome, a morpholinomethyl Mannich base (8c) was disclosed which proved to be cytotoxic to all the tested tumor cell lines in the low micromolar range (IC50 < 20 μM) and to inhibit in vitro the efflux pumps P-gp and MRP1 responsible for MDR, with IC50s of 0.45 and 12.1 μM, respectively.

The corrosion behavior of 1018 carbon steel in LiCl/H2O mixtures has been evaluated by using potentiodynamic polarization curves, electrochemical noise and electrochemical impedance spectroscopy techniques. Two different concentrations of LiCl were employed, namely 35 and 40 wt. %, and the testing temperatures included 25, 35 and 70 °C. It was found that the steel showed a passive zone; the corrosion current density value increased with an increase in the solution temperature and concentration. The pitting potential value decreased with an increase in the testing temperature and the solution concentration. The corrosion process was under charge transfer control. This mechanism was unaltered either by the solution temperature or concentration. The charge transfer resistance value decreased with an increase in both the solution temperature and concentration. A localized, pitting type of corrosion dominated the corrosion morphology at 25 and 35 °C, whereas at 70 °C, the main type of attack was a mixed type of corrosion.

Using pumps operating as turbines (PATs) offers the possibility of increasing the sustainability of water and energy systems by recovering the excess energy that would be otherwise lost in pressure-reducing valves or head loss chambers. Regarding on-grid applications, there have been many research works, and PATs have been implemented in several ways. However, more research still needs to be done on optimizing the efficiency and stability of PATs operating in off-grid systems. This work contributes to the development of stable direct current (DC) off-grid electric systems based on PATs using a self-excited induction generator (SEIG). In this context, a methodology is proposed, based on the hydraulic, mechanical, and electric subsystems, to define the PAT-SEIG operational area to maximize energy conversion and system efficiency. These limits depend highly on the capacitor value, rotational speed, and electric load. In addition, an analytical model is proposed to estimate the PAT-SEIG operation under specific conditions. With this, water managers can design and optimize an off-grid PAT-SEIG system and define the best hydraulic machines, electronic equipment, and control elements to maximize energy conversion within the target of operational limits. Two micro PAT-SEIG setups were implemented in the hydraulic laboratory of IST/CERIS under typical operating conditions to validate the proposed methodology. The system’s maximum efficiency and operational limits can be adapted using different capacitor values for the excitation of the SEIG. Considering the nominal efficiencies of the system’s components, the maximum p.u. efficiency obtained for each PAT-SEIG system was between 0.7 and 0.8 p.u.

Background: Diabetes Mellitus (DM) is an important chronic disease that occurs worldwide. Aims: This study aims to investigate how the use of the FreeStyle® Libre system in Unified Health System (SUS) patients impacts diabetes parameters in patients who receive education on proper insulin administration and the use of the continuous monitoring device, as well as how this affects patients without any concomitant multidisciplinary support in Sergipe, Brazil. Methods: We conducted a prospective randomized study in a diabetes clinic in Sergipe, Brazil, using the flash method FreeStyle® Libre (Abbott). The participants were divided into two groups: one receiving diabetes education on CGM (continuous glucose monitoring), while the other did not. Before the intervention, the patient’s treatment motivation and quality of life were assessed using a questionnaire, and baseline levels of glycated hemoglobin were measured using high-performance liquid chromatography (HPLC) and the point of care AlereTM Afinion with boronate fixation. We compared first- and second-phase data with respect to glycated hemoglobin, mean interstitial blood glucose, time on and above target for hypoglycemic and hyperglycemic events, and mean hypoglycemic duration. Results: In group A, which received the diabetes education intervention, there was a significant reduction in average HbA1c levels from 8.6% to 7.9% after 3 months (p = 0.001). However, there was no significant difference in average glycemic values. Time above target decreased significantly from 50.62% to 29.43% (p = 0.0001), while time below target decreased from 22.90% to 20.21% (p = 0.002). There was no significant change in the number of hypoglycemic events, but the duration of hypoglycemia decreased significantly from 130.35 min to 121.18 min after 3 months (p = 0.0001). In Group B, there was no significant difference in mean HbA1c levels before (7.07%) and after (7.28%) sensor installation. This group maintained lower HbA1c levels compared to the other group. Average blood glucose levels also remained similar before (148.37 mg/dL) and after (154.65 mg/dL) the intervention. Although the time above the target glucose level increased significantly from 35.94% to 48.17%, the time at target decreased from 50.40% to 37.97%. No significant changes were observed in the time below target, the number of hypoglycemic events, or the duration of hypoglycemia. Conclusions: Our findings indicate that utilizing continuous glucose monitoring technology can enhance glycemic control, particularly in motivated, educated, low-income patients dependent on the SUS. To achieve positive results with FreeStyle Libre, it is imperative to allocate resources for multidisciplinary support.

This article addresses challenges in the design of underground high-voltage transmission lines, focusing on thermal management and cable ampacity determination. It introduces an innovative proposal that adjusts the dimensions of the backfill to enhance ampacity, contrasting with the conventional approach of increasing the core cable’s cross-sectional area. The methodology employs a particle swarm optimization (PSO) technique with adaptive penalization and restart strategies, implemented in MATLAB for parameter autoadaptation. The article emphasizes more efficient solutions than traditional PSO, showcasing improved convergence and precise results (success probability of 66.1%). While traditional PSO is 81% faster, the proposed PSO stands out for its accuracy. The inclusion of thermal backfill results in an 18.45% increase in cable ampacity, considering variations in soil thermal resistivity, backfill properties, and ambient temperature. Additionally, a sensitivity analysis was conducted, revealing conservative values that support the proposal’s robustness. This approach emerges as a crucial tool for underground installation, contributing to continuous ampacity improvement and highlighting its impact on decision making in energy systems.

The world’s water infrastructures suffer from inefficiencies, such as high energy consumption and water losses due to inadequate management practices and feeble pressure regulation, leading to frequent water and energy losses. This strains vital water and energy resources, especially in the face of the worsening challenges of climate change and population growth. A novel method is presented that integrates micro-hydropower plants, with pumps as turbines (PATs), in the water network in the city of Funchal. Sensitivity analyses evaluated the microgrid’s response to variations in the cost of energy components, showing favorable outcomes with positive net present value (NPV). PV solar and micro-wind turbines installed exclusively at the selected PRV sites within the Funchal hydro grid generate a combined 153 and 55 MWh/year, respectively, supplementing the 406 MWh/year generated by PATs. It should be noted that PATs consistently have the lowest cost of electricity (LCOE), confirming their economic viability and efficiency across different scenarios, even after accounting for reductions in alternative energy sources and grid infrastructure costs.

Combat sports athletes seeking a competitive edge often engage in weight management practices to become larger than their opponents, which ultimately includes periods of gradual weight loss, rapid weight loss, and weight regain. This pattern of weight loss and regain is known as weight cycling and often includes periods of low energy availability, making combat sports athletes susceptible to metabolic dysfunction. This narrative review represents an effort to explore the metabolic perturbations associated with weight cycling and outline the short-, medium-, and long-term effects on metabolic flexibility, function, and health. The short-term effects of rapid weight loss, such as a reduced metabolic rate and alterations to insulin and leptin levels, may prelude the more pronounced metabolic disturbances that occur during weight regain, such as insulin resistance. Although definitive support is not currently available, this cycle of weight loss and regain and associated metabolic changes may contribute to metabolic syndrome or other metabolic dysfunctions over time.

Polyphenolic compounds, encompassing flavonoids (e.g., quercetin, rutin, and cyanidin) and non-flavonoids (e.g., gallic acid, resveratrol, and curcumin), show several health-related beneficial effects, which include antioxidant, anti-inflammatory, hepatoprotective, antiviral, and anticarcinogenic properties, as well as the prevention of coronary heart diseases. Polyphenols have also been investigated for their counteraction against the adverse effects of common anticancer chemotherapeutics. This review evaluates the outcomes of clinical studies (and related preclinical data) over the last ten years, with a focus on the use of polyphenols in chemotherapy as auxiliary agents acting against oxidative stress toxicity induced by antitumor drugs. While further clinical studies are needed to establish adequate doses and optimal delivery systems, the improvement in polyphenols’ metabolic stability and bioavailability, through the implementation of nanotechnologies that are currently being investigated, could improve therapeutic applications of their pharmaceutical or nutraceutical preparations in tumor chemotherapy.

Cognitive abilities tend to decline with aging, with variation between individuals, and many studies seek to identify genetic biomarkers that more accurately anticipate risks related to pathological aging. We investigated the influence of BDNF, NTRK2, and FNDC5 single nucleotide polymorphisms (SNPs) on the cognitive performance of young and older adults with contrasting educational backgrounds. We addressed three questions: (1) Is education associated with reduced age-related cognitive decline? (2) Does the presence of SNPs explain the variation in cognitive performance observed late in life? (3) Is education differentially associated with cognition based on the presence of BDNF, NTRK2, or FNDC5 polymorphisms? We measured the cognitive functions of young and older participants, with lower and higher education, using specific and sensitive tests of the Cambridge Automated Neuropsychological Test Assessment Battery. A three-way ANOVA revealed that SNPs were associated with differential performances in executive functions, episodic memory, sustained attention, mental and motor response speed, and visual recognition memory and that higher educational levels improved the affected cognitive functions. The results revealed that distinct SNPs affect cognition late in life differentially, suggesting their utility as potential biomarkers and emphasizing the importance of cognitive stimulation that advanced education early in life provides.

In this paper, we discuss a collaborative research project called Lacunae: Embodying the Untranslatable. The issue of untranslatability has been a much-discussed topic in translation studies, with recent debate linking it to performability. Although untranslatability has received some attention lately, the debate has been largely theoretical, confined to a textual conception of translation. In the study discussed in this article, we explored an applied approach to (un)translatability, working with/through the body in space, positing the body as the vehicle for deciphering the untranslatable. We draw on an embodied way of knowing as a phenomenological framework to construct knowledge as lived experience. The study aimed to investigate the lexical, intercultural, and aesthetic potential of performing untranslatability by exploring a series of untranslatable words through research-based theatre. The data generation process involved a retreat where nine researchers/artists/practitioners addressed the research question through practices like process drama, Butoh, physical theatre, improvisation, and visual arts on mixed media. In this paper, first, we introduce the theoretical framework and context of the study. Next, we illustrate the methodology, data analysis, and findings, with reference to one untranslatable word from the Chinese language, yuánfèn 缘分, loosely translated as ‘serendipity in relationships and life events’. We contemplate the practice in this workshop through a philosophical, pedagogical, and research-based lens. Finally, we contemplate future iterations of this project, reflecting on how performing yuánfèn could inform theatre-based research on migration and identity in education.

We show that Einstein’s conformal gravity can explain, simply, and on the geometric ground, galactic rotation curves, without the need to introduce any modification in both the gravitational as well as in the matter sector of the theory. The geometry of each galaxy is described by a metric obtained, making a singular rescaling of Schwarzschild’s spacetime. The new exact solution, asymptotically anti-de Sitter, manifests an unattainable singularity at infinity that cannot be reached in finite proper time; namely, the spacetime is geodetically complete. It deserves to be noticed that, in this paper, we have a different opinion from the usual one. Indeed, instead of making the metric singularity-free, we make it apparently but harmlessly even more singular than Schwarzschild’s. Finally, it is crucial to point out that Weyl’s conformal symmetry is spontaneously broken into the new singular vacuum rather than the asymptotically flat Schwarzschild’s one. The metric is unique according to the null energy condition, the zero acceleration for photons in the Newtonian regime, and the homogeneity of the Universe at large scales. Once the matter is conformally coupled to gravity, the orbital velocity for a probe star in the galaxy turns out to be asymptotically constant consistent with the observations and the Tully–Fisher relation. Therefore, we compare our model with a sample of 175 galaxies and show that our velocity profile very well interpolates the galactic rotation curves after a proper choice of the only free parameter in the metric. The mass-to-luminosity ratios of galaxies turn out to be close to 1, consistent with the absence of dark matter.

Cholecystokinin 8 (CCK8) is an entero-octapeptide that participates in crosstalk with components of intestinal immunity via the CCK receptor (CCKR), but its role in modulation of the IgA response is not fully known under physiological conditions. Male eight-week-old BALB/c mice each were intraperitoneally injected once during 7 days with CCK8, devazapide (CCKR1 antagonist), L365,260 (CCKR2 antagonist) or vehicle (sham group). In intestinal lavages, total and secretory IgA (SIgA) were determined by ELISA; in lamina propria, IgA+ B lymphocytes and IgA+ plasma cells were analyzed by flow cytometry; mRNA levels of polymeric immunoglobulin receptor (pIgR) in epithelial cells and α chain, interleukins (ILs) in lamina propria cells were assessed by qRTPCR. Regarding the sham conditions, IgA+ plasma-cell percentage and IL-2, IL-5, IL-10 and transforming growth factor-β (TGF-β) mRNA levels were either increased by CCK8 or decreased by both CCKR antagonists. For IgA/SIgA responses, IL-4/IL-6 mRNA levels were decreased by all drugs and pIgR mRNA was increased by CCK8 and reduced by L365,260. IgA+ B cell percentage and α chain mRNA levels were elicited by CCK8 and L365,260. Data suggested a presumable differential role of CCK/CCKR on the IgA-response; outcome of L365,260 on the elicitation of IgA+ B cells and α chain mRNA needs further examination.

Grumixama is an anthocyanin-rich berry commonly found in South America, yet not widely consumed by Brazilians due to the lack of studies covering its technological properties. Therefore, the stability of anthocyanins and the activity of oxidoreductase enzymes in grumixama juice during thermal treatment (60 °C to 100 °C) and storage (at 25 °C and 7 °C) were evaluated. As a result, the pasteurization of grumixama juice at 80 °C for 60 s completely inactivated peroxidase (POD) and polyphenol oxidase (PPO), eliminated microorganisms, and induced low degradation of the total anthocyanins (40%) and minimal degradation of the total phenolic compounds. Four anthocyanins were monitored in the pasteurized juice, namely delphinidin 3-glucoside, cyanidin 3-glucoside, and two unidentified anthocyanins. Despite the temperature range, the qualitative chromatographic profile of the anthocyanins did not change, while the contents were affected. The binominal temperature/time of 80 °C/60 s effectively inactivated PPO and POD enzymes, promoted moderate alterations in the contents of total anthocyanins and total phenolic compounds, and ensured the microbiological quality of grumixama juice. Grumixama juice can be used as a health food due to its high content of antioxidant phenolic compounds, especially anthocyanins.

Intracranial primary tumors (IPTs) are aggressive forms of malignancies that cause high mortality in both humans and domestic animals. Meningiomas are frequent adult IPTs in humans, dogs, and cats, and both benign and malignant forms cause a decrease in life quality and survival. Surgery is the primary therapeutic approach to treat meningiomas, but, in many cases, it is not resolutive. The chemotherapy and targeted therapy used to treat meningiomas also display low efficacy and many side effects. Therefore, it is essential to find novel pharmacological approaches to increase the spectrum of therapeutic options for meningiomas. This review analyzes the similarities between human and domestic animal (dogs and cats) meningiomas by evaluating the molecular and histological characteristics, diagnosis criteria, and treatment options and highlighting possible research areas to identify novel targets and pharmacological approaches, which are useful for the diagnosis and therapy of this neoplasia to be used in human and veterinary medicine.

Beverages and drinks play a significant role in maintaining the integral health of individuals. The aim of this study is to discover the pattern of beverage consumption in different groups of the Spanish population and to investigate its relationship with other nutritional variables and habits. To achieve the objectives, an observational, descriptive and cross-sectional study was conducted. For data collection, a questionnaire was designed and validated that explored different beverage and food consumption variables as well as socio-demographic and lifestyle variables. The instrument was disseminated, among the Spanish young adult population, through snowball sampling using social networks, collecting a sample of 17,541 valid surveys. Bivariate comparative analyses and correlation analyses were performed, and finally, the principal component analysis (PCA) method was used in order to study the relationships between variables related to drinking and health. The main results show significant differences in the pattern of beverage consumption between the socio-demographic variables of sex, age and educational level, as well as between different areas of Spain, while the PCA model shows the relationship between the consumption of sugar-sweetened beverages with the Healthy Nutrition Index of the population and sport practice. Based on the results of the study, the following conclusions were reached: the beverage consumption pattern of the Spanish population is affected by socio-demographic variables. Healthier drinking habits affect the nutrition and health of the population.

Our objectives were to evaluate the use of cottonseed cake in replacing corn silage in a diet without forage and to identify the model with higher precision and accuracy of adjustment of parameters of ruminal degradation kinetics. A diet containing corn silage and another with cottonseed cake as a fiber source were formulated. Gompertz, Dual-pool Logistic, Brody, and Ørskov models were evaluated for goodness of fit to gas production. There were significant differences in dry matter (DM), organic matter (OM), and neutral detergent fiber (NDF) in the in vitro digestibility for diets and fiber sources. The estimated values of the Gompertz (6.77), Brody (6.72), and Ørskov (6.73) models were similar to the observed mean of gas production in the corn silage diet (6.73 mL/100 mg DM). Similarly, the estimated values of the Brody (5.87) and Ørskov (5.89) models were similar to the observed mean of gas production in the cottonseed cake diet (5.87 mL/100 mg DM). The roughage-free diet containing cottonseed cake as a fiber source stimulated higher gas production. Brody and Ørskov models presented higher precision and accuracy in the fitting of kinetics of degradation independent of the fiber source in the diet.

The increasing number of food frauds, mainly targeting high quality products, is a rising concern among producers and authorities appointed to food controls. Therefore, the development or implementation of methods to reveal frauds is desired. The genetic traceability of traditional or high-quality dairy products (i.e., products of protected designation of origin, PDO) represents a challenging issue due to the technical problems that arise. The aim of the study was to set up a genetic tool for the origin traceability of dairy products. We investigated the use of Short Tandem Repeats (STRs) to assign milk and cheese to the corresponding producer. Two farms were included in the study, and the blood of the cows, bulk milk, and derived cheese were sampled monthly for one year. Twenty STRs were selected and Polymerase Chain Reactions for each locus were carried out. The results showed that bulk milk and derived cheese express an STR profile composed of a subset of STRs of the lactating animals. A bioinformatics tool was used for the exclusion analysis. The study allowed the identification of a panel of 20 markers useful for the traceability of milk and cheeses, and its effectiveness in the traceability of dairy products obtained from small producers was demonstrated.

Currently, the antioxidants (AOs) used in foods are mainly synthetic, often questioned on health grounds. So, the need for innocuous natural AOs has increased in last years. The peanut skin (PS) is an industrial by-product of low added value, but rich in bioactive phenolic compounds. In this study, the antioxidant capacity of a PS extract (PSE) was examined in a chia oil-based mayonnaise stored during six months (25 °C). The mayonnaise was made using chia oil (68% w/w) added with PSE (2 mg/g fat) and without any AO (control). For the storage test, 30 g were placed in 100 mL amber bottles and at 2, 4 and 6 months the oily phase was extracted (chloroform: methanol). Peroxide index (PI), acidity index (AI), K232, K270, p-Anisidine (pAnV) and TOTOX values were measured. Moreover, the presence of 2,4 heptadienal and 3,5-octadiene-2-one was analyzed by static headspace GC-MS. At the end of the assay, PI, AI, K232, K270, and pAnV for control and PSE mayonnaises were 74.7 and 13.4 meq O2/kg oil; 2.4 and 2.0 mg KOH/kg oil, 10 and 3.55, 1.34 and 0.64, 3.7 and 0.98, respectively. The TOTOX value of the control was approximately six times higher than PSE mayonnaise. 2,4 Heptadienal and 3,5-octadiene-2-one were not detected at initial time but in the control treatment at the end reached 3.75 and 2.15 µg/g, respectively. Differently, in PSE mayonnaise, 3,5-octadiene-2-one was undetected and 2,4 Heptadienal was 0.83 µg/g. In conclusion, PSE represents a potential natural AO to preserve the oxidative stability of chia oil-based mayonnaise.

Background and Objectives: C-arm-free MIS techniques can offer significantly reduced rates of postoperative complications such as inadequate decompression, blood loss, and instrumentation misplacement. Another advantageous long-term aspect is the notably diminished exposure to radiation, which is known to cause malignant changes. This study emphasizes that, in some cases of spinal conditions that require a procedural intervention, C-arm-free MIS techniques hold stronger indications than open surgeries guided by image intensifiers. Materials and Methods: This study includes a retrospective analysis and review of various cervical and thoracic spinal procedures, performed in our hospital, applying C-arm-free techniques. The course of this study explains the basic steps of the procedures and demonstrates postoperative and intraoperative results. For anterior cervical surgery, we performed OPLL resection, while for posterior cervical surgery, we performed posterior fossa decompression for Chiari malformation, minimally invasive cervical pedicle screw fixation (MICEPS), and modified Goel technique with C1 lateral mass screw for atlantoaxial subluxation. Regarding the thoracic spine, we performed anterior correction for Lenke type 5 scoliosis and transdiscal screw fixation for diffuse idiopathic skeletal hyperostosis fractures. Results: C-arm-free techniques are safe procedures that provide precise and high-quality postoperative results by offering sufficient spine alignment and adequate decompression depending on the case. Navigation can offer significant assistance in the absence of normal anatomical landmarks, yet the surgeon should always appraise the quality of the information received from the software. Conclusions: Navigated C-arm-free techniques are safe and precise procedures implemented in the treatment of surgically demanding conditions. They can significantly increase accuracy while decreasing operative time. They represent the advancement in the field of spine surgery and are hailed as the future of the same.

Small-scale hydropower plants (SHP) have been losing competitiveness compared to other renewable sources in the global electricity matrix due to technical, environmental, regulatory, and economic issues. In this context, and to contribute to the development and strengthening of SHP’s integration into the country’s electricity matrix, this study proposes the Balanced Scorecard (BSC) as a strategic evaluation tool for SHP projects from the prospecting phase to the pre-implementation phase, taking into account financial and non-financial performance indicators. This study aims to structure a strategic map of the SHP source composed of economic, socio-environmental, institutional, and technical perspectives, adapted to the four perspectives of the BSC, with the required performance indicators for the strategic evaluation of SHP. The development of a computer program allowed data visualization from eight case studies focusing on indicators or perspectives that jeopardize the project’s strategic viability. The results showed that (i) the adaptation of the BSC methodology for the strategic evaluation of SHP feasibility proved effective and (ii) the developed computer program demonstrated robustness and effectiveness in analyzing SHP, even in different stages of development and with different technical and environmental characteristics. Finally, the strategic evaluation of SHP feasibility is an effective tool in assisting decision makers, indicating the positive aspects and weaknesses of the project under evaluation and providing a better understanding of the asset and the areas of development that deserve greater attention to improve its performance.

The multitarget therapeutic strategy, as opposed to the more traditional ‘one disease-one target-one drug’, may hold promise in treating multifactorial neurodegenerative syndromes, such as Alzheimer’s disease (AD) and related dementias. Recently, combining a photopharmacology approach with the multitarget-directed ligand (MTDL) design strategy, we disclosed a novel donepezil-like compound, namely 2-(4-((diethylamino)methyl)benzylidene)-5-methoxy-2,3-dihydro-1H-inden-1-one (1a), which in the E isomeric form (and about tenfold less in the UV-B photo-induced isomer Z) showed the best activity as dual inhibitor of the AD-related targets acetylcholinesterase (AChE) and monoamine oxidase B (MAO-B). Herein, we investigated further photoisomerizable 2-benzylideneindan-1-one analogs 1b–h with the unconjugated tertiary amino moiety bearing alkyls of different bulkiness and lipophilicity. For each compound, the thermal stable E geometric isomer, along with the E/Z mixture as produced by UV-B light irradiation in the photostationary state (PSS, 75% Z), was investigated for the inhibition of human ChEs and MAOs. The pure E-isomer of the N-benzyl(ethyl)amino analog 1h achieved low nanomolar AChE and high nanomolar MAO-B inhibition potencies (IC50s 39 and 355 nM, respectively), whereas photoisomerization to the Z isomer (75% Z in the PSS mixture) resulted in a decrease (about 30%) of AChE inhibitory potency, and not in the MAO-B one. Molecular docking studies were performed to rationalize the different E/Z selectivity of 1h toward the two target enzymes.

Weed coexistence with an agricultural crop can negatively affect its growth, development, and yield. From this perspective, this study aimed to evaluate weed management strategies and their effect on the agronomic parameters of cowpea cultivation in direct (SPD) and conventional (SPC) planting systems. The experiment was set up in a completely randomized block design with a split-plot arrangement with four replications. The plots received a source of variation referring to the planting systems (direct and conventional planting), and the subplots corresponded to ten weed management strategies (manual hoeing 18 days after planting (DAP); at 36 DAP; at 54 DAP; at 18 and 36 DAP; at 18 and 54 DAP; at 18 and 72 DAP; at 36 and 54 DAP; at 36–72 DAP; at 18, 36, and 54 DAP; and a control with no hoeing). Density and dry mass evaluations of the cowpea plants were performed at harvest (72 DAP) by determining the number of pods per plant, pod length, number of grains per pod, 1000-grain mass, and yield. A total of 28 species distributed in 12 botanical families were identified in the two cultivation systems. The family Poaceae showed the highest frequency, with 25% of the species identified. At the end of the assay, treatment 20 had the highest positive influence and provided significant quantitative gains to the complex of traits related to cowpea production (SPD and hoeing at 18, 36, and 54 DAP). UPGMA cluster analysis and canonical discriminant analysis were performed and allowed a better classification of the evaluated treatments. It was observed that the first two canonical variables explained 90.8% of the total variance contained in the original variables. The use of SPD with weeding at 18, 36, and 54 days after planting provides greater weed control and significant quantitative gains for the complex of characteristics related to cowpea production. The results underscore the importance of choosing the correct cropping system and implementing effective weeding practices to optimize weed control and improve crop performance.

Intestinal bacteria establish a specific relationship with the host animal, which causes the acquisition of gut microbiota with a unique composition classified as the enterotype. As the name suggests, the Red River Hog is a wild member of the pig family living in Africa, in particular through the West and Central African rainforest. To date, very few studies have analysed the gut microbiota of Red River Hogs (RRHs) both housed under controlled conditions and in wild habitats. This study analysed the intestinal microbiota and the distribution of Bifidobacterium species in five Red River Hog (RRH) individuals (four adults and one juvenile), hosted in two different modern zoological gardens (Parco Natura Viva, Verona, and Bioparco, Rome) with the aim of disentangling the possible effects of captive different lifestyle and host genetics. Faecal samples were collected and studied both for bifidobacterial counts and isolation by means of culture-dependent method and for total microbiota analysis through the high-quality sequences of the V3–V4 region of bacterial 16S rRNA. Results showed a host-specific bifidobacterial species distribution. Indeed, B. boum and B. thermoacidophilum were found only in Verona RRHs, whereas B. porcinum species were isolated only in Rome RRHs. These bifidobacterial species are also typical of pigs. Bifidobacterial counts were about 106 CFU/g in faecal samples of all the individuals, with the only exception for the juvenile subject, showing 107 CFU/g. As in human beings, in RRHs a higher count of bifidobacteria was also found in the young subject compared with adults. Furthermore, the microbiota of RRHs showed qualitative differences. Indeed, Firmicutes was found to be the dominant phylum in Verona RRHs whereas Bacteroidetes was the most represented in Roma RRHs. At order level, Oscillospirales and Spirochaetales were the most represented in Verona RRHs compared with Rome RRHs, where Bacteroidales dominated over the other taxa. Finally, at the family level, RRHs from the two sites showed the presence of the same families, but with different levels of abundance. Our results highlight that the intestinal microbiota seems to reflect the lifestyle (i.e., the diet), whereas age and host genetics are the driving factors for the bifidobacterial population.

Introduction: Recent modifications in the epidemiology of oropharyngeal squamous cell carcinoma (OSCC) have led to the increase of Human papillomavirus (HPV) related metastatic head and neck cancer patients with high life expectancy even at advanced stage, low comorbidity and still restricted systemic therapy opportunities. In the recent era of ablative therapies’ development, oligometastatic HPV OSCC patients are indubitably good candidates for intensified treatment. However, data related to outcomes after optimised management of metastatic sites are dramatically missing. Materials and patients: In our cohort of 186 unselected consecutive OSCC patients treated with curative intent at our institution between 2009 and 2013, we analysed the incidence, treatment and outcomes of distant metastatic (DM) failure according to p16 status. Results: After a median follow-up of 4.2 years (95% CI: 3.8–4.4) from primary diagnosis of OSCC, 21/95 p16− patients (22.1%) vs. 8/91 (8.8%) p16+ patients presented DM failure with a median interval of 11 (range 0–46) and 28 months (range 0–71), respectively (p = 0.10). Overall survival (OS) after DM failure was significantly higher in p16+ patients with a two-year OS rate of 75% and 15% for p16+ and p16−, respectively (p = 0.002). In eight HPV-related metastatic patients, three underwent ablative lung metastasis treatment and are still complete responders four to five years later. Conclusion: This study highlights distinct outcomes of metastatic HPV-related OSCC patients emphasised by three long-term complete responders after lung ablative treatment. In patients with high life expectancy and limited tumour burden, the question of ablative treatment such as metastasectomy or stereotactic ablative radiotherapy (SBRT) should be addressed.

In the same way that specialized DNA polymerases (DNAPs) replicate cellular and viral genomes, only a handful of dedicated proteins from various natural origins as well as engineered versions are appropriate for competent exponential amplification of whole genomes and metagenomes (WGA). Different applications have led to the development of diverse protocols, based on various DNAPs. Isothermal WGA is currently widely used due to the high performance of Φ29 DNA polymerase, but PCR-based methods are also available and can provide competent amplification of certain samples. Replication fidelity and processivity must be considered when selecting a suitable enzyme for WGA. However, other properties, such as thermostability, capacity to couple replication, and double helix unwinding, or the ability to maintain DNA replication opposite to damaged bases, are also very relevant for some applications. In this review, we provide an overview of the different properties of DNAPs widely used in WGA and discuss their limitations and future research directions.

The interaction between variety, rootstock, and trellis height is important for grapevine management, mainly for producing new varieties of grapes for juice and wine in new wine-growing regions with high production potential. Then, this study aimed to evaluate the rootstocks and trellis height influence on photosynthesis, biochemical, and yield performance for grapevine hybrids. The experiment was carried out in a randomized block design using two factors, rootstocks (‘IAC 766’ and ‘106-8 Mgt’) and trellis height (until 1.6 and 2.0 m), evaluated for two grapevine hybrids (IAC 138-22 ‘Maximo’ and ‘BRS Violeta’). During grapevine flowering, it was evaluated photosynthesis and biochemical performance, for this, the gaseous exchanges were measured using the open system photosynthesis equipment with a CO2 analyzer and water vapor by infrared radiation, being net assimilation rate of CO2, stomatal conductance, transpiration rate, internal CO2 concentration, water use efficiency, carboxylation efficiency (Rubisco), and the flux density of photosynthetically active photons. At the stages of grapevine flowering and ripening berries were evaluated the antioxidant enzymes (peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT)), total soluble proteins, chlorophylls, and SPAD. The interaction between rootstock and trellis heigh influenced varieties’ photosynthesis, biochemical, and yield performance. In conclusion under subtropical conditions, better photosynthesis, biochemical, and yield performance were observed when both cultivars were grafted on the ‘IAC 766’ rootstock. The ‘IAC 138-22 Maximo’ was trained until 2.0 and grafted on the ‘IAC 766’ rootstock, increasing grape production and photosynthesis efficiency. In addition, this variety was more productive than ‘BRS Violeta’.

The adoption of diversified agricultural systems that employ integrated cultural practices appears to be the way to sustainably intensify tropical agriculture. Our objectives were to evaluate the dry matter (DM) accumulation of sorghum inoculated with Azospirillum brasilense, with or without a nitrogen fertilization split, intercropped with palisade grass (Urochloa brizantha cv. BRS Paiaguás), and how these practices influenced the nutrition and development of soybean in succession. The design was a randomized complete block in a 2 × 2 × 3 factorial, consisting of sorghum monoculture cropped or intercropped with palisade grass, sorghum either inoculated or not with A. brasilense, and nitrogen applied at 120 kg ha−1 N only at sowing, only at topdressing, or split—30% at sowing and 70% at topdressing at the beginning of the panicle initiation stage. The residual impacts of these treatments on the following soybean crop were also evaluated. Higher DM yield occurred in sorghum inoculated with A. brasilense, however, this result varied by year. The sorghum–palisade grass intercrop produced a higher amount of straw than sorghum monoculture. The nutrition of soybean was adequate regardless of treatments, but grain yield was higher when the sorghum residue was inoculated. The inoculation of A. brasilense in sorghum intercropped with palisade grass increased yield. The nutrition of soybean was adequate regardless of the treatments, while grain yield was higher on the inoculated sorghum residues. The inoculation of A. brasilense in sorghum intercropped with palisade grass increased DM yield. The intercropping increased the production of biomass for animal grazing and DM for soil coverage. The inoculation of sorghum by A. brasilense and its intercropping with palisade grass contributed to higher soybean yield in succession.

The selection of pumps as turbines (PATs) for their respective use in energy optimisation systems is a complicated task, because manufacturers do not provide the characteristic curves. For this reason, some research has been carried out to predict them with computational fluid dynamics (CFD) and mathematical models. The purpose of this study is to validate these two prediction methodologies of flow (Q) vs. head (H) curves through numerical modelling using the computational package OpenFOAM, together with a comparison with the experimental data obtained from a PAT for the case in which the nominal rotation speed of the machine varies. Depending on the configuration and working conditions of the PAT, the simulation performed with OpenFOAM was validated by calibrating it with the nominal curve of the pump and with another simulation performed with CFD workbench SOLIDWORKS FloEFD. Subsequently, the second methodology related to the analyses and mathematical models proposed to predict the Q vs. H curves were also validated with new models in OpenFOAM and the experimental data. The results show that these prediction methods are effective when a machine’s operating point is close to the BEP (best efficient point). The absolute error ranges obtained with these two prediction methodologies for rotation speeds of 880 rpm, 1020 rpm, 1200 rpm, and 1500 rpm are between 5 and 24%, 2 and 17%, 0 and 12%, and 1 and 24%, respectively.

About twenty molecules sharing 1H-chromeno[3,2-c]pyridine as the scaffold and differing in the degree of saturation of the pyridine ring, oxidation at C10, 1-phenylethynyl at C1 and 1H-indol-3-yl fragments at C10, as well as a few small substituents at C6 and C8, were synthesized starting from 1,2,3,4-tetrahydro-2-methylchromeno[3,2-c]pyridin-10-ones (1,2,3,4-THCP-10-ones, 1) or 2,3-dihydro-2-methyl-1H-chromeno[3,2-c]pyridines (2,3-DHPCs, 2). The newly synthesized compounds were tested as inhibitors of the human isoforms of monoamine oxidase (MAO A and B) and cholinesterase (AChE and BChE), and the following main SARs were inferred: (i) The 2,3-DHCP derivatives 2 inhibit MAO A (IC50 about 1 μM) preferentially; (ii) the 1,2,3,4-THCP-10-one 3a, bearing the phenylethynyl fragment at C1, returned as a potent MAO B inhibitor (IC50 0.51 μM) and moderate inhibitor of both ChEs (IC50s 7–8 μM); (iii) the 1H-indol-3-yl fragment at C10 slightly increases the MAO B inhibition potency, with the analog 6c achieving MAO B IC50 of 3.51 μM. The MAO B inhibitor 3a deserves further pharmacological studies as a remedy in the symptomatic treatment of Parkinson’s disease and neuroprotectant for Alzheimer’s disease. Besides the established neuroprotective effects of MAO inhibitors, the role of MAOs in tumor insurgence and progression has been recently reported. Herein, antiproliferative assays with breast (MCF-7), colon (HCT116) and cisplatin-resistant ovarian (SK-OV-3) tumor cells revealed that the 10-indolyl-bearing 2,3,4,10-THCP analog 6c exerts anti-tumor activity with IC50s in the range 4.83–11.3 μM.

The addition of Biofire® FilmArray® Blood Culture Identification panel 2 (BCID2) to the antimicrobial stewardship program (ASP) could improve outcomes in bloodstream infections (BSI) of patients with febrile neutropenia (FN). A pre- and post-quasi-experimental single-center study was conducted at a reference hospital in Peru. Three groups were considered: patients with BSI before ASP intervention (control group), patients with BSI after ASP intervention (group 1), and patients with BSI after ASP intervention plus BCID2 PCR Panel implementation (group 2). Overall, 93 patients were identified (32 control, 30 group 1, 31 group 2). The median time to effective therapy was significantly shorter in group 2 compared to group 1 and control group, respectively (3.75 vs. 10 h, p = 0.004; 3.75 vs. 19 h, p < 0.001). No significant differences in terms of relapse of bacteremia, in-hospital mortality (all cause), and 30-day-all-cause hospital readmission between the three study periods were found. The appropriateness of empirical antimicrobial use, adding or change, and the following de-escalation or discontinuation was significant when the two intervention periods were compared with the control group (p < 0.001). In addition to the lack of local studies documenting the microbiological profile of FN episodes, adding syndromic panels-based testing could allow for the consolidation of ASP strategies.

The tropical glaciers of the Cordillera Blanca have played host to some of the most significant mass movements ever recorded in the world and Peru; many proglacial lakes formed in this mountain range have natural dikes made of moraine material, which, if they collapse, would present a risk for the cities located downstream of a proglacial lake, where the proglacial lake Palcacocha has a remarkable background regarding floods. The Sentinel-2 MSI (Multi-Spectral Instrument, Level-2A) has a specific band for snow probability mapping that indicates glaciers and snow cover; this is effective for recognizing proglacial lakes by calculating the NDWIice. It is also helpful for lithology with SWIR for granite moraine deposits and slate moraines in the proglacial environment Palcacocha; these deposits surround the proglacial lake, with NDWIice determining the perimeter where sediment interacts with the rocks and meltwater. In addition, there are high radon concentrations made by ice avalanche impacts on the proglacial lake. Unstable glacier blocks cause ice avalanches into this proglacial lake, and the radon responds to flow variations from these high-impact avalanches. We used the device RadonEye PLus2, which allows real-time detection of radon flux changes in the proglacial environment. Our results indicated that ice avalanches making a high impact in the proglacial lake cause turbulent flow and generate radon concentration marks with a rising magnitude, while the absence of ice avalanches in the lake will cause the values to go down. The relationships of radon concentrations in the atmosphere for a tropical proglacial environment are radon and temperature (R2 = 0.364), radon and humidity (R2 = 0.469). In a passive proglacial environment with prolonged rainfall, radon concentrations tend to decrease, with an inversely proportional relationship between humidity and radon in the tropical proglacial environment. Proglacial lakes in the tropical zone often have large volumes of freshwater with high slopes from tropical glaciers, and climate change effects are an imminent danger for nearby cities.

Acute lymphoblastic leukemia (ALL) is the most common childhood cancer in the world. Single nucleotide variants (SNVs) in miRNA and genes encoding proteins of the miRNA synthesis complex (SC) may affect the processing of drugs used in the treatment of ALL, resulting in treatment-related toxicities (TRTs). We investigated the role of 25 SNVs in microRNA genes and genes encoding proteins of the miRNA SC, in 77 patients treated for ALL-B from the Brazilian Amazon. The 25 SNVs were investigated using the TaqMan® OpenArray™ Genotyping System. SNVs rs2292832 (MIR149), rs2043556 (MIR605), and rs10505168 (MIR2053) were associated with an increased risk of developing Neurological Toxicity, while rs2505901 (MIR938) was associated with protection from this toxicity. MIR2053 (rs10505168) and MIR323B (rs56103835) were associated with protection from gastrointestinal toxicity, while DROSHA (rs639174) increased the risk of development. The rs2043556 (MIR605) variant was related to protection from infectious toxicity. SNVs rs12904 (MIR200C), rs3746444 (MIR499A), and rs10739971 (MIRLET7A1) were associated with a lower risk for severe hematologic toxicity during ALL treatment. These findings reveal the potential for the use of these genetic variants to understand the development of toxicities related to the treatment of ALL in patients from the Brazilian Amazon region.
