Despite improvements in treatment options for advanced colorectal cancer (CRC), survival outcomes are still best for patients with non-metastasised disease. Diagnostic tools to identify blood-based biomarkers and assist in CRC subtype classification could afford a means to track CRC progression and treatment response. Cancer cell-derived small extracellular vesicles (EVs) circulating in blood carry an elevated cargo of lipids and proteins that could be used as a signature of tumour suppressor/promoting events or stages leading up to and including metastasis. Here, we used pre-characterised biobanked plasma samples from surgical units, typically with a low volume (~100 µL), to generate and discover signatures of CRC-derived EVs. We employed nanostructured porous silicon (pSi) surface assisted-laser desorption/ionisation (SALDI) coupled with high-resolution mass spectrometry (HR-MS), to allow sensitive detection of low abundant analytes in plasma EVs. When applied to CRC samples, SALDI-HR-MS enabled the detection of the peptide mass fingerprint of cancer suppressor proteins, including serine/threonine phosphatases and activating-transcription factor 3. SALDI-HR-MS also allowed the detection of a spectrum of glycerophospholipids and sphingolipid signatures in metastatic CRC. We observed that lithium chloride enhanced detection sensitivity to elucidate the structure of low abundant lipids in plasma EVs. pSi SALDI can be used as an effective system for label-free and high throughput analysis of low-volume patient samples, allowing rapid and sensitive analysis for CRC classification.

Recurrent pregnancy loss is a distressing condition affecting 1–2% of couples. Traditionally investigations have focused on the female, however more recently researchers have started to explore the potential contribution of the male partner. Seminal reactive oxygen species have a physiological function in male reproduction but in excess are suspected to generate structural and functional damage to the sperm. Evidence is mounting to support an association between elevated seminal reaction oxygen species and recurrent pregnancy loss. Studies suggest that the rates of sperm DNA damage are higher in the male partners of women affected by recurrent pregnancy loss compared with unaffected men. However, the available pool of data is conflicting, and interpretation is limited by the recent change in nomenclature and the heterogeneity of study methodologies. Furthermore, investigation into the effects of oxidative stress on the epigenome show promise. The value of antioxidant therapy in the management of recurrent pregnancy loss currently remains unclear.

Innovations in nanogenerator technology foster pervading self-power devices for human use, environmental surveillance, energy transfiguration, intelligent energy storage systems, and wireless networks. Energy harvesting from ubiquitous ambient mechanical, thermal, and solar energies by nanogenerators is the hotspot of the modern electronics research era. Ferroelectric materials, which show spontaneous polarization, are reversible when exposed to the external electric field, and are responsive to external stimuli of strain, heat, and light are promising for modeling nanogenerators. This review demonstrates ferroelectric material-based nanogenerators, practicing the discrete and coupled pyroelectric, piezoelectric, triboelectric, and ferroelectric photovoltaic effects. Their working mechanisms and way of optimizing their performances, exercising the conjunction of effects in a standalone device, and multi-effects coupled nanogenerators are greatly versatile and reliable and encourage resolution in the energy crisis. Additionally, the expectancy of productive lines of future ensuing and propitious application domains are listed.

The main focus of the current study was to sustain the releasing behavior of theophylline by fabricated polymeric microgels. The free radical polymerization technique was used for the development of aspartic acid-co-poly(2-acrylamido-2-methylpropanesulfonic acid) microgels while using various combinations of aspartic acid, 2-acrylamido-2-methylpropanesulfonic acid, and N′,N′-methylene bisacrylamide as a polymer, monomer, and cross-linker, respectively. Ammonium peroxodisulfate and sodium hydrogen sulfite were used as initiators. Characterizations such as DSC, TGA, SEM, FTIR, and PXRD were performed for the fabricated microgels to assess their thermal stability with unreacted polymer and monomer, their surface morphology, the formation of a new polymeric system of microgels by evaluating the cross-linking of functional groups of the microgels’ contents, and to analyze the reduction in crystallinity of the theophylline by fabricated microgels. Various studies such as dynamic swelling, drug loading, sol–gel analysis, in vitro drug release studies, and kinetic modeling were carried out for the developed microgels. Both dynamic swelling and percent drug release were found higher at pH 7.4 as compared to pH 1.2 due to the deprotonation of functional groups of aspartic acid and AMPS. Similarly, sol–gel analysis was performed and an increase in gel fraction was observed with the increasing concentration of microgel contents, while sol fraction was decreased. Conclusively, the prepared carrier system has the potential to sustain the release of the theophylline for an extended period of time.

Zinc oxide nanoparticles (ZnONPs) are the top candidate in the field of biological applications because of their high surface area and excellent catalytic activities. In the present study, the cyanobacteria-mediated biosynthesis of zinc oxide NPs using Nostoc sp. extract as a stabilizing, chelating, and reducing agent is reported. ZnONPs were biologically synthesized using an eco-friendly and simple technique with a minimal reaction time and calcination temperature. Various methods, including X-ray diffraction (XRD), ultraviolet spectroscopy (UV), Fourier transform infrared (FTIR), scanning electron microscopy (SEM), and energy-dispersive X-ray spectroscopy (EDX) were used to characterize the biosynthesized zinc oxide NPs. XRD analysis depicted the crystalline form of zinc oxide NPs, and the Scherrer equation determined a mean crystalline size of ~28.21 nm. The SEM results reveal the spherical shape of the biosynthesized nanoparticles. Various functional groups were involved in the capping and stabilization of the zinc oxide NPs, which were confirmed by FTIR analysis. The zinc oxide NPs showed strong UV-vis absorption at 340 nm. Multiple in vitro biological applications showed significant therapeutic potential for zinc oxide NPs. Potential antimicrobial assays were reported for zinc oxide NPs via the disc-diffusion method and food poisoning method, respectively. All other activities mentioned below are described with the concentration and IC50 values. Biocompatibility with human erythrocytes and macrophages (IC50: 433 µg/mL, IC50 > 323 µg/mL) and cytotoxic properties using brine shrimps (IC50: 11.15 µg/mL) and Leishmania tropics (Amastigotes IC50: 43.14 µg mL−1 and Promastigotes IC50: 14.02 µg mL−1) were determined. Enzyme inhibition assays (protein kinase and alpha amylase) were performed and showed strong potential. Free radical scavenging tests showed strong antioxidant capacities. These results indicate that zinc oxide NPs synthesized by Nostoc sp. have strong biological applications and are promising candidates for clinical development.

Soil with low fertility is a big problem for achieving citrus productivity. In this regard, the management of macro and micronutrients is essential. Macro and micronutrient deficiency decreased the yield and the quality of citrus fruit. It is the need of the hour to classify the soil fertility status under changing climatic scenarios. The current soil fertility survey was conducted to examine the macro and micronutrient status in the citrus production area. In soil, three depths (0–15, 15–30, and 30–45 cm) were taken for sampling. For leaves, 4–6-months-old non-bearing twigs were sampled from 20 trees per orchard at breast height. Cadmium (Cd) and lead (Pb) are global environmental pollutants. In this study, Nostoc sp. MK-11 was used as an environmentally safe, economical, and efficient biosorbent for the removal of Cd and Pb ions from synthetic aqueous solutions. Nostoc sp. MK-11 was identified on a morphological and molecular basis using light microscopic, 16S rRNA sequences and phylogenetic analysis. Batch experiments were performed to determine the most significant factors for the removal of Cd and Pb ions from the synthetic aqueous solutions using dry Nostoc sp. MK1 biomass. The results indicated that the maximum biosorption of Pb and Cd ions was found under the conditions of 1 g of dry Nostoc sp. MK-11 biomass, 100 mg/L of initial metal concentrations, and 60 min contact time at pH 4 and 5 for Pb and Cd, respectively. Dry Nostoc sp. MK-11 biomass samples before and after biosorption were characterized using FTIR and SEM. A kinetic study showed that a pseudo second order kinetic model was well fitted rather than the pseudo first order. Three isotherm models Freundlich, Langmuir, and Temkin were used to explain the biosorption isotherms of metal ions by Nostoc sp. MK-11 dry biomass. Langmuir isotherm, which explains the existence of monolayer adsorption, fitted well to the biosorption process. Considering the Langmuir isotherm model, the maximum biosorption capacity (qmax) of Nostoc sp. MK-11 dry biomass was calculated as 75.757 and 83.963 mg g−1 for Cd and Pb, respectively, which showed agreement with the obtained experimental values. Desorption investigations were carried out to evaluate the reusability of the biomass and the recovery of the metal ions. It was found that the desorption of Cd and Pb was above 90%. The dry biomass of Nostoc sp. MK-11 was proven to be efficient and cost-effective for removing Cd and especially Pb metal ions from the aqueous solutions, and the process is eco-friendly, feasible, and reliable.

BackgroundPost-viral syndromes (PVS), including Long COVID, are symptoms sustained from weeks to years following an acute viral infection. Non-pharmacological treatments for these symptoms are poorly understood. This review summarises evidence for the effectiveness of non-pharmacological treatments for symptoms of PVS. It also summarises the symptoms and health impacts of PVS in individuals recruited to studies evaluating treatments.Methods and findingsWe conducted a systematic review to evaluate the effectiveness of non-pharmacological interventions for PVS, as compared to either standard care, alternative non-pharmacological therapy, or placebo. The outcomes of interest were changes in symptoms, exercise capacity, quality of life (including mental health and wellbeing), and work capability. We searched five databases (Embase, MEDLINE, PsycINFO, CINAHL, MedRxiv) for randomised controlled trials (RCTs) published between 1st January 2001 to 29th October 2021. We anticipated that there would be few RCTs specifically pertaining to Long COVID, so we also included observational studies only if they assessed interventions in individuals where the viral pathogen was SARS-COV-2. Relevant outcome data were extracted, study quality appraised using the Cochrane Risk of Bias tool, and the findings were synthesised narratively. Quantitative synthesis was not planned due to substantial heterogeneity between the studies. Overall, five studies of five different interventions (Pilates, music therapy, telerehabilitation, resistance exercise, neuromodulation) met the inclusion criteria. Aside from music-based intervention, all other selected interventions demonstrated some support in the management of PVS in some patients.ConclusionsIn this study, we observed a lack of robust evidence evaluating non-pharmacological treatments for PVS, including Long COVID.
