Background: Accurate and rapid testing for COVID-19 is critical for effective disease management and control. The One-Step-COVID-2019-Test was developed as a rapid serological test to detect antibodies against SARS-CoV-2. Objective: To estimate the accuracy of the rapid serological test for COVID-19 using One-Step-COVID-2019. Methods: We conducted a population-based serological survey with a stratified sampling of 593 adults between October and December 2020, prior to mass vaccination and during a period of limited availability of rapid tests. Participants provided 7.5 mL of serum, which was tested using the One-Step-COVID-2019-Test for IgM-IgG antibodies without distinction, as well as an in-house ELISA for IgG against the spike protein. Statistical analysis accounted for sampling weights, with accuracy assessed through sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), Youden index, and kappa coefficient, using ELISA as the reference standard. McNemar’s test identified significant differences between the test results. Results: The ELISA-based prevalence of infection was 11.1%. The One-Step-COVID-2019-Test showed low sensitivity (27.0–30.8%) but high specificity (89.9–96.6%), with poor agreement (kappa: 0.290–0.337), particularly among asymptomatic individuals. Conclusions: The One-Step-COVID-2019 rapid test for COVID-19 demonstrated inadequate performance, characterized by low sensitivity and poor reliability, making it unsuitable for effective serological surveillance.

Making plant breeding programs less expensive, fast, practical, and accurate, especially for soybeans, promotes the selection of new soybean genotypes and contributes to the emergence of new varieties that are more efficient in absorbing and metabolizing nutrients. Using spectral information from soybean genotypes combined with nutritional information on secondary macronutrients can help genetic improvement programs select populations that are efficient in absorbing and metabolizing these nutrients. In addition, using machine learning algorithms to process this information makes the acquisition of superior genotypes more accurate. Therefore, the objective of the work was to verify the classification performance of soybean genotypes regarding secondary macronutrients by ML algorithms and different inputs. The experiment was conducted in the experimental area of the Federal University of Mato Grosso do Sul, municipality of Chapadão do Sul, Brazil. Soybean was sown in the 2019/20 crop season, with the planting of 103 F2 soybean populations. The experimental design used was randomized blocks, with two replications. At 60 days after crop emergence (DAE), spectral images were collected with a Sensifly eBee RTK fixed-wing remotely piloted aircraft (RPA), with autonomous takeoff control, flight plan, and landing. At the reproductive stage (R1), three leaves were collected per plant to determine the macronutrients calcium (Ca), magnesium (Mg), and sulfur (S) levels. The data obtained from the spectral information and the nutritional values of the genotypes in relation to Ca, Mg, and S were subjected to a Pearson correlation analysis; a PC analysis was carried out with a k-means algorithm to divide the genotypes into clusters. The clusters were taken as output variables, while the spectral data were used as input variables for the classification models in the machine learning analyses. The configurations tested in the models were spectral bands (SBs), vegetation indices (VIs), and a combination of both. The combination of machine learning algorithms with spectral data can provide important biological information about soybean plants. The classification of soybean genotypes according to calcium, magnesium, and sulfur content can maximize time, effort, and labor in field evaluations in genetic improvement programs. Therefore, the use of spectral bands as input data in random forest algorithms makes the process of classifying soybean genotypes in terms of secondary macronutrients efficient and important for researchers in the field.

The Addax nasomaculatus is an African antelope currently considered “critically endangered”. There is little scientific knowledge about the species, which could potentially aid in species conservation. The objective of this study was to determine how hair cortisol concentration and hair color vary according to sex, body site, and season across a whole year in zoo-housed addax. The addax population was housed at Parque Lecocq Zoo, Uruguay. Hair samples were taken from the shoulder and rump in autumn, winter, spring, and summer. Hair cortisol concentration in addax varied according to sex, with males exhibiting a higher hair cortisol concentration than females (1.03 ± 0.05 pg/mg vs. 0.91 ± 0.05 pg/mg, p = 0.016, respectively), with these differences being more significant in the spring (p < 0.0001). Hair cortisol concentration varied across seasons (p < 0.0001), with maximum values observed in summer. The color parameters (L*, a*, and b*) varied across seasons (p < 0.0001). The coat was blacker and yellower in winter and whiter in summer. Males had darker and greener hair color than females (p < 0.05). Sex and season-driven variation in hair cortisol concentration and hair color seem to be important elements to consider in relation to thermal comfort and animal welfare in addax, highlighting the warmer seasons as critical.

As microbes develop resistance to various drugs, the treatment of infections becomes increasingly challenging, leading to prolonged illness, heightened severity of infections, elevated mortality rates, and increased healthcare costs. Essential oils are lipophilic and volatile mixtures of compounds that have gained attention in research for novel antimicrobial agents. Therefore, the present study evaluated the essential oil of Syzygium cumini leaves (EOSC) in order to prospect its antifungal and trichomonacidal activities. The essential oil from the leaves was extracted by steam distillation and analyzed by GC-MS. Antifungal activity was evaluated using the serial microdilution method. Additionally, the potential of the EOSC as an enhancer of fluconazole (FCZ) action was tested at subinhibitory concentrations. To assess anti-Trichomonas vaginalis activity, concentrations ranging from 15.6 to 500 μg/mL of EOSC were tested. Finally, the SwissADME platform was employed to analyze the physicochemical and pharmacokinetic characteristics of the major component of EOSC. The GC-MS analysis identified 94.24% of the components of EOSC, with α-pinene (51.11%) and nerol (8.25%) as major constituents. EOSC exhibited low antifungal activity against the evaluated Candida strains. However, the combination of EOSC and FCZ reduced the IC50 against Candida krusei from 45.29 to 0.30 μg/mL. EOSC also demonstrated significant activity against T. vaginalis (IC50 = 88.2 μg/mL). In silico prediction with α-pinene showed low toxic action and important physicochemical aspects for drug production. The essential oil of Syzygium cumini emerges as a promising candidate for the discovery of molecules with potential antifungal and anti-Trichomonas vaginalis applications.

This study was conducted to survey the level of technification of quail sheds in Brazil. Data from 25 quail farms (5 in each Brazilian region) were collected by image analysis of videos available on the Internet. The analyzed variables were farm location, degree of technological adoption in quail sheds, housing conditions, structural conditions, wall conditions, and thermal comfort equipment. The data were subjected to descriptive analysis, and differences were assessed using the chi-squared test (p < 0.10). It was found that curtain walls were the most used system for air entry and renewal in quail sheds. Fan systems were present in only 12% of sheds, and evaporative cooling systems (or air conditioning) were observed in 4% of sheds, exclusively on large farms. Internal insulation was used in 20.83% of farms. In conclusion, Brazilian quail sheds have a low degree of technification; about 90% do not use implements such as ceiling, ventilation, and cooling systems. These conditions make it difficult to control environmental variables within quail sheds, impairing thermal comfort and, consequently, animal welfare and quail productivity.

This study aimed to identify behavioral and morphological traits of Nellore cattle that can influence calves’ survival and pre-weaning performance. It was performed in two units of a large-scale cow–calf operation farm located in the Central-West region of Brazil, with 975 cow–calf dyads, by scoring cows’ maternal protective behavior, body condition, horns, udder suspension (US), teat size (TS) and calves’ vigor. TS was used to determine teat symmetry (TSm). The production unit where calves were born, their birth date, sex, and birth (BW) and weaning weights were also recorded. Cow ages at calving and production units were used to categorize the cows into twelve classes (Cage). The Shapiro–Wilk test alongside tables of frequencies were used to determine if continuous and categorical variables followed a normal distribution, respectively. Two datasets were considered in statistical analyses: (i) 975 dyad records were used to identify potential factors influencing calf survivability and (ii) 936 records of weaned calves were used to assess pre-weaning performance. Generalized linear models were used to assess the effect of target traits on the calves’ weight adjusted for 210 days of age (W210, kg) and pre-weaning average daily gain (preADG, kg/day). Tukey’s test was used to compare W210 and preADG means. Pearson’s correlation coefficients were estimated between BW, W210 and preADG. Cage, US and the TSm significantly affected pre-weaning calf performance (p < 0.01). Older cows (independently of the production unit) and those with intermediate US and symmetrical teats weaned heavier calves. As expected, positive and significant correlation was observed between BW and preADG (r = 0.35), which, in turn, was highly correlated with W210 (r = 0.99). We concluded that none of the behavioral and morphological traits influenced calf survivability, but cow age, US and TSm impacted pre-weaning performance, with 8- to 11-year-old cows and those with intermediate US and symmetric teats leading to better calf performance at weaning.

The effects of Tagetes erecta flower extract (TFE) and increasing levels of vitamin E (VE) in the diet of Japanese quail breeders on progeny performance and oxidative status were studied. Methods: 480 Japanese quail breeders were distributed in a completely randomized design with five treatments and twelve replications of six females and two males each. A control diet (25 mg/kg VE) and four diets supplemented with TFE (3 g/kg) and VE (25, 100, 175, or 250 mg/kg) were used. Fresh yolk samples and the yolk sac and liver from embryos (11 and 15 days) and chicks (hatch and 3 days) were analysed. Data were subjected to ANOVA, a regression linear model, and contrast tests and the level of significance was set at p < 0.05. Results: TF and VE in the maternal diet improved the amount of alfa-tocopherol and total carotenoid content in the yolk. TFE + VE reduced lipid peroxidation and improved the oxidative status in the fresh yolk, in the embryo and chick yolk, and in the liver. Liver superoxide dismutase activity in hatched chicks increased linearly with the VE level and was not altered by TFE. Maternal diets did not influence progeny performance (1 to 28 days) or the relative expression of superoxide dismutase or glutathione peroxidase genes in the liver of chicks. Conclusions: TFE is an effective antioxidant in fresh eggs and supplementation of 3 g/kg TFE and high levels of VE in quail breeders improves the oxidative status of embryos and newly hatched chicks.

For bioactivity studies, it is necessary to use products with a high degree of purity, which may influence the cytotoxic effects. Supercritical technology presents itself as an alternative to obtain these products. Therefore, the objective of this work was to obtain the bioactive compounds of oil and pulp of açaí fat-free supercritical technology and evaluate the cytotoxicity of products in MRC-5 and VERO cells in vitro. The açaí pulp was subjected to extraction with supercritical CO2 to obtain the oil and pulp without fat, under conditions of 323.15 K at 35 MPa, 333.15 K at 42 MPa, and 343.15 K at 49 MPa. The largest yields (51.74%), carotenoids (277.09 µg/g), DPPH (2.55 μmol TE/g), ABTS (2.60 μmol TE/g), and FRAP (15.25 μm of SF/g) of oil and ABTS (644.23 μmol TE/g) of pulp without fat were found in the condition 343.15 K at 49 MPa. The highest levels of compounds phenolics (150.20 mg GAE/g), DPPH (414.99 μmol TE/g), and FRAP (746.2 μm SF/g) of the pulp without fat were found in the condition of 323.15 K to 35 MPa. The fat-free pulp presented high levels of anthocyanins without significant variation in cytotoxicity. The developed process was efficient in obtaining oil rich in carotenoids, and the supercritical technology elucidated an efficient way to obtain açaí fat-free pulp.

The increasing reliance on electric vehicle (EV) charging in buildings requires balancing the load from other building systems to support the new demand. This paper uses a study case in a Near-Zero Energy Building (NZEB) educational facility located in the Brazilian Amazon to verify how much the energy efficiency (EE) measures would improve the existing ratings of the building and supply the expansion of EV demand. A comprehensive building energy load and energy performance analysis were conducted in four steps, following the mandatory Brazilian requirements for EE in public buildings, using measured data, computer modeling, and thermoenergetic analyses using OpenStudio version 1.1.0 and EnergyPlus software version 9.4.0. First, the EE retrofit measures were proposed and evaluated, targeting the air conditioning and lighting systems. Subsequently, an equation was elaborated to indicate the maximum level of energy consumption that could be increased without compromising the building’s energy performance and NZEB classification. Finally, Open DSS software version 10.0.0.2 was used to simulate the increased availability of EV charging after the retrofit. With the proposed retrofit, the building improved the EE ratings by three levels, and the percentage of the NZEB rating increased by 33.28%. These measures also increased the EV charging load by 20%, maintaining the maximum EE level and the NZEB classification, although EV maximization reduced self-sufficiency by 9.78% compared to the retrofit-only scenario.

Background: Schistosomiasis is a parasitic infection that can cause pulmonary hypertension (PH). Th2 CD4 T cells are necessary for experimental Schistosoma-PH. However, if T cells migrate to the lung to initiate, the localized inflammation that drives vascular remodeling and PH is unknown. Methods: Mice were sensitized to Schistosoma mansoni eggs intraperitoneally and then challenged using tail vein injection. FTY720 was administered, which blocks lymphocyte egress from lymph nodes. T cells were quantified using flow cytometry, PH severity via heart catheterization, and cytokine concentration through ELISA. Results: FTY720 decreased T cells in the peripheral blood, and increased T cells in the mediastinal lymph nodes. However, FTY720 treatment resulted in no change in PH or type 2 inflammation severity in mice sensitized and challenged with S. mansoni eggs, and the number of memory and effector CD4 T cells in the lung parenchyma was also unchanged. Notably, intraperitoneal Schistosoma egg sensitization alone resulted in a significant increase in intravascular lymphocytes and T cells, including memory T cells, although there was no significant change in parenchymal cell density, IL-4 or IL-13 expression, or PH. Conclusion: Blocking T cell migration did not suppress PH following Schistosoma egg challenge. Memory CD4 T cells, located in the lung intravascular space following egg sensitization, appear sufficient to cause type 2 inflammation and PH.

Laser shock peening (LSP) is a formidable cold working surface treatment that provides high-energy precision to enhance the mechanical properties of materials. As the significance of LSP continues to grow, the collective findings underscore its potential as a transformative technology for fortifying materials against mechanical stress and improving their overall performance and longevity. The discourse encapsulates the evolving landscape of the LSP, emphasizing the pivotal role played by finite element analysis in advancing our understanding and application of this innovative surface treatment.

Brazil is the second largest beef producer and a leading exporter, contributing to some 3000 t CWE in global markets (27.7% of market share). The sector has experienced substantial development, but yields remain far below potential, and there are growing concerns regarding land use change and greenhouse gas emissions. The need for sustainable technologies, such as sound pasture management and integrated farming systems, is evident, but adoption may be low amongst farmers unable to keep up with technological advances. This article describes the historical developments of Brazilian beef farming towards sustainability and discusses possible socioenvironmental outcomes. We combined an extensive literature review, public data, and our own insights as senior researchers to achieve that. The trajectory shown here evidenced the technological intensification of Brazilian beef farming, with strong support of public policies for decarbonizing agriculture. Nonetheless, the pace of this transition may affect small to medium farmers with limited access to information, technologies, and credit. Our recommendations involve a broad program of technical assistance and training on sustainable technologies, including financial and digital literacy. A novel approach to financing farmers is suggested to support a sustainable and inclusive transition in beef farming in Brazil.

The present study evaluates the effect of including microencapsulated hot chili pepper (MHCP) in the diet of crossbred dairy cows on the volume and quality of milk and on ovarian morphofunctionality. Twenty-four crossbred females in their lactating period were used. The cows were divided into two experimental groups, a control (CT) and an MHCP -supplemented group (CP) given 1 g a day per animal of microencapsulated hot chili in concentrate for 42 days. Over seven weeks of daily milk production was measured, and sample milk was collected weekly for composition analysis. Animals were subject to an ovulation synchronization protocol on day 0 (D0), and an intravaginal progesterone (P4) implant, estradiol benzoate, and prostaglandin (PGF2α) were administered. On D8, the P4 implant was removed and PGF2α, equine chorionic gonadotropin, and estradiol cypionate were administered to the animals. The ovarian dynamics were evaluated in B mode and color Doppler. There were significant differences (p < 0.05) in the group X time interaction, the volume of milk produced, and the amount in kg/day of milk components. There was a higher percentage of vascularization in the preovulatory follicle in the CP group (p ≥ 0.10). The findings show that the inclusion of MHCP in the diet of dairy cows does influence their milk production and reproduction.

The growth of electric vehicles (EVs) and their integration into existing and future buildings bring new considerations for energy efficiency (EE) and balance when combined with renewable energy. However, for buildings with an energy efficiency label, such as Near Zero Energy Building (NZEB) or Positive Energy Building (PEB), the introduction of EVs may result in the declassification of the EE label due to the additional energy required for the charging infrastructure. This underscores the increasing relevance of demand-side management techniques to effectively manage and utilize energy consumption and generation in buildings. This paper evaluates the influence of electric vehicle (EV) charging on NZEB/PEB-labeled buildings of the Brazilian Building Labeling Program (PBE Edifica). Utilizing on-site surveys, computational modeling, and thermos-energetic analysis with software tools such as OpenStudio v. 1.1.0 and EnergyPlus v. 9.4.0, an energy classification was conducted in a building in the city of Belem, State of Para, Brazil. Subsequently, power flow simulations employing probabilistic models and Monte Carlo approaches were executed in the OpenDSS software v. 10.0.0.2 to examine the impact of EV integration, both with and without the implementation of demand-side management techniques. Analyses using the labeling methodology demonstrated that the building has EE level C and NZEB self-sufficiency classification. The assessment of the impact of EV integration on the building’s total energy consumption in the base (current) scenario was carried out in two scenarios, with (2) and without (1) supply management. Scenario 01 generated a 69.28% increase in energy consumption, reducing the EE level to D and resulting in the loss of the NZEB class. Scenario 02 resulted in a smaller increase in energy consumption of 40.50%, and guaranteed the return of the NZEB class lost in scenario 1, but it was not enough to return the EE level to class C. The results highlight the need for immediate and comprehensive energy management strategies, as the findings show that the two scenarios present a difference of 41.55% in energy consumption. Nonetheless, these strategies are not enough if other consumption restrictions or energy efficiency measures are not applied to other building systems.

Chikungunya (CHIKV) and Mayaro (MAYV) viruses are arthritogenic alphaviruses that promote an incapacitating and long-lasting inflammatory muscle–articular disease. Despite studies pointing out the importance of skeletal muscle (SkM) in viral pathogenesis, the long-term consequences on its physiology and the mechanism of persistence of symptoms are still poorly understood. Combining molecular, morphological, nuclear magnetic resonance imaging, and histological analysis, we conduct a temporal investigation of CHIKV and MAYV replication in a wild-type mice model, focusing on the impact on SkM composition, structure, and repair in the acute and late phases of infection. We found that viral replication and induced inflammation promote a rapid loss of muscle mass and reduction in fiber cross-sectional area by upregulation of muscle-specific E3 ubiquitin ligases MuRF1 and Atrogin-1 expression, both key regulators of SkM fibers atrophy. Despite a reduction in inflammation and clearance of infectious viral particles, SkM atrophy persists until 30 days post-infection. The genomic CHIKV and MAYV RNAs were still detected in SkM in the late phase, along with the upregulation of chemokines and anti-inflammatory cytokine expression. In agreement with the involvement of inflammatory mediators on induced atrophy, the neutralization of TNF and a reduction in oxidative stress using monomethyl fumarate, an agonist of Nrf2, decreases atrogen expression and atrophic fibers while increasing weight gain in treated mice. These data indicate that arthritogenic alphavirus infection could chronically impact body SkM composition and also harm repair machinery, contributing to a better understanding of mechanisms of arthritogenic alphavirus pathogenesis and with a description of potentially new targets of therapeutic intervention.

Staphylococcus aureus and Acinetobacter baumannii are opportunistic pathogens, and both are involved in different oral infections. This work aimed to analyze the phytochemical composition of Myrciaria cauliflora hydroethanolic extract and to evaluate its antimicrobial and antibiofilm action against Staphylococcus aureus (ATCC 6538) and Acinetobacter baumannii (ATCC 19606; multi-resistant clinical strains 58004, 50098, 566006, and H557). Myrciaria cauliflora hydroethanolic extract was prepared, and the content of soluble solids, flavonoids, and phenols was quantified. High-performance liquid chromatography (HPLC) was performed later. The minimum inhibitory concentration was determined using the broth microdilution method according to the Clinical and Laboratory Standards Institute, standard M7-A6, and subsequently, its minimum bactericidal concentration was determined. Then, the most effective concentrations were analyzed against biofilms. Statistical analysis was performed using the ANOVA method with Tukey’s test. The soluble solids content in the prepared hydroethanolic extract of M. cauliflora was 2.22%. Additionally, the total flavonoid content, measured using the quercetin standard curve, was 0.040 mg/mL. Furthermore, the total phenol content, determined using the gallic acid standard curve, was 0.729 mg/mL. HPLC analysis presented peaks of gallic acid (11.80 m), p-coumaric acid (12.09 m), cinnamic acid derivative (19.02 m), and ellagic acid (29.83 m). The extract demonstrated antimicrobial and antibiofilm action against all tested strains. However, the most effective antibacterial concentration against all the tested bacteria was 5.55 mg/mL. Therefore, these chemical components justify that M. cauliflora hydroethanolic extract is effective in reducing biofilm formation in S. aureus (standard strain) and A. baumannii (standard and clinical strains).

The Parecis Basin, one of Brazil’s most extensive intracratonic basins, holds significant potential for hydrocarbon exploration. Despite its vast size, Parecis has yet to be extensively explored, with only five wildcat wells drilled. So far, no commercial discoveries have been announced. Regional studies have suggested Paleozoic sedimentation, while recent analyses have revealed a Neoproterozoic infill. Its tectonic model is still a matter of debate, and to date, no detailed structural map for the whole basin has been published. The present work proposes a new detailed structural map of the Parecis Basin based on a four-step interpretation workflow integrating seismic and gravimetric data. The first step includes converting the public 2D seismic lines to the depth domain. The second step is estimating the residual Bouguer anomaly, where the computed residual anomalies should relate to the basin’s tectonic features. The third step comprises the 2D forward modeling of the gravimetric anomalies using the 2D seismic interpretation as a constraint. The final step compiled all the interpreted features into our new structural map. This map reveals the top of the basement, forming a complex framework of horsts and grabens. Normal faults define the main structural style in the basin. Further, we could recognize thick, high-density bodies embedded in the crystalline basement. These bodies consist of Orosian–Calimian (1.8–1.6 Ga) mafic and ultramafic rocks, which may be a potential source for hydrogen exploration in the basin. Subsequent geophysical and geochemical surveys will assess the hydrogen potential in the area.

Background: Leishmaniasis is a significant global public health issue that is caused by parasites from Leishmania genus. With limited treatment options and rising drug resistance, there is a pressing need for new therapeutic approaches. Molecular chaperones, particularly Hsp90, play a crucial role in parasite biology and are emerging as promising targets for drug development. Objective: This study evaluates the efficacy of 17-DMAG in treating BALB/c mice from cutaneous leishmaniasis through in vitro and in vivo approaches. Materials and Methods: We assessed 17-DMAG’s cytotoxic effect on bone marrow-derived macrophages (BMMΦ) and its effects against L. braziliensis promastigotes and intracellular amastigotes. Additionally, we tested the compound’s efficacy in BALB/c mice infected with L. braziliensis via intraperitoneal administration to evaluate the reduction in lesion size and the decrease in parasite load in the ears and lymph nodes of infected animals. Results: 17-DMAG showed selective toxicity [selective index = 432) towards Leishmania amastigotes, causing minimal damage to host cells. The treatment significantly reduced lesion sizes in mice and resulted in parasite clearance from ears and lymph nodes. It also diminished inflammatory responses and reduced the release of pro-inflammatory cytokines (IL-6, IFN-γ, TNF) and the regulatory cytokine IL-10, underscoring its dual leishmanicidal and anti-inflammatory properties. Conclusions: Our findings confirm the potential of 17-DMAG as a viable treatment for cutaneous leishmaniasis and support further research into its mechanisms and potential applications against other infectious diseases.

Background: Coronavirus disease 2019 (COVID-19) is a disease with a broad clinical spectrum, which may result in hospitalization in healthcare units, intensive care, and progression to death. This study aimed to describe and compare the clinical and epidemiological profile of COVID-19 during the three waves of the disease, in patients admitted to a public hospital in the city of Belém, Pará, in the Amazon region of Brazil. Methods: This descriptive, observational, and cross-sectional study was population-based on individuals who were hospitalized with a diagnosis of COVID-19, confirmed by real-time reverse transcription-polymerase chain reaction (RT-PCR), and who were interviewed and monitored at the public hospital, from February 2020 to April 2022. Results: The prevalence was male patients, older than 60 years. The most frequent symptoms were dyspnea, cough, and fever. Systemic arterial hypertension was the most prevalent comorbidity followed by diabetes mellitus. Less than 15% of patients were vaccinated. The nasal oxygen cannula was the most used oxygen therapy interface followed by the non-rebreathing reservoir mask. Invasive mechanical ventilation predominated and the median time of invasive mechanical ventilation ranged from 2 to 6 days among waves. As for the hospital outcome, transfers prevailed, followed by deaths and discharges. Conclusion: The presence of comorbidities, advanced age, and male sex were important factors in the severity and need for hospitalization of these patients, and the implementation of the vaccination policy was an essential factor in reducing the number of hospital admissions.

Avocado oil is rich in nutrients beneficial to human health, such as monounsaturated fatty acids, phenolic compounds, tocopherol, and carotenoids, with numerous possibilities for application in industry. This review explores, through a comparative approach, the effectiveness of the supercritical oil extraction process as an alternative to the conventional cold-pressing method, evaluating the differences in the extraction process steps through the effect of temperature and operating pressure on bioactive quality and oil yield. The results reveal that supercritical avocado oil has a yield like that of mechanical cold pressing and superior functional and bioactive quality, especially in relation to α-tocopherol and carotenoids. For better use and efficiency of the supercritical technology, the maturation stage, moisture content, fruit variety, and collection period stand out as essential factors to be observed during pre-treatment, as they directly impact oil yield and nutrient concentration. In addition, the use of supercritical technology enables the full use of the fruit, significantly reducing waste, and adds value to the agro-industrial residues of the process. It produces an edible oil free of impurities, microorganisms, and organic solvents. It is a green, environmentally friendly technology with long-term environmental and economic advantages and an interesting alternative in the avocado market.

The study explored the viability of using waste plastic oil (PO) as an alternative lubricant to petroleum-based lubricants in industrial settings. To enhance the lubrication performance of the PO, this study incorporated cost-efficient, oleic acid-modified, graphene nano platelets [GNP (f)] and hexagonal boron nitride [hBN (f)] nano solid lubricant additives into the PO in various concentrations, forming functionalized nano lubricants. The PO and its functionalized nano lubricant’s rheological, dispersion stability, thermal degradation, friction, and wear performance were investigated. Results manifest that incorporating GNP (f) and hBN (f) into the PO significantly enhanced the viscosity and dispersion stability. In addition, it was seen that GNP (f) and hBN (f) nano lubricants lowered the coefficient of friction (COF) by 53% and 63.63% respectively, compared to the PO. However, the GNP (f) and hBN (f) nano lubricants demonstrated a 3.16% decrease and a 50.08% increase in wear volume relative to the PO. Overall, the GNP (f) and hBN (f) nano lubricants displayed a synergistic friction behavior, while they exhibited an antagonistic behavior pertaining to the wear volume. The study elucidated the mechanisms underlying friction and wear performance of the nano lubricants.

Porous materials have recently attracted significant attention in the aerospace and biomedical fields for addressing issues related to friction and wear. Porous materials are beneficial in applications where continuous lubrication is not feasible or for components that operate under extreme conditions, such as high speeds, elevated temperatures, and heavy loads. The pores can serve as reservoirs for liquid lubricants, which are gradually released during the operation of the components. The tribological properties of these materials depend on their porosity, the lubricants used, and any additional additives incorporated into the porous materials. This review article provides insight into common fabrication techniques for porous materials and examines their tribological performance for all three classes of materials—polymers, metals, and ceramics. Additionally, it discusses design criteria for porous self-lubricating materials by highlighting the critical properties of both the substrate and lubricants.

Imbalanced dietary intake is associated with the development of inflammatory bowel diseases (IBDs) and is often observed during the active phases of Crohn’s disease (CD) and ulcerative colitis (UC). Cumulative data also suggest the potential for dietary manipulation in avoiding IBD relapse. However, there is a paucity of dietary data from patients in clinical remission to guide such an approach. Our study aimed to characterize the dietary pattern and adequacy of patients with IBD in clinical remission. Data on dietary intake (three alternate 24 h food records) were collected from 40 patients with IBD (20 CD and 20 UC) and 45 gender-matched healthy controls (HC). Statistical comparisons between patients and controls employed Student’s t-test, Mann–Whitney U, chi-squared, and Fisher’s exact tests. The adequacy of dietary intake of IBD patients was further studied by assessing the nutrient inadequacy prevalence, estimated using the Dietary Reference Intakes (DRI) framework and the Estimated Average Requirement (EAR) parameter. We observed significant dietary imbalances among patients with IBD compared to the HC group, marked by disparities in both macronutrient and micronutrient intakes. Inadequacies with frequencies >80% were observed for the ingestion of total fiber and 13 micronutrients in IBD patients. Our preliminary findings suggest that imbalanced dietary intake is also characteristic among individuals with IBD during clinical remission, corroborating the need for dietary interventions in this population.

This study examines the epidemiological and genomic characteristics, along with the transmission dynamics, of SARS-CoV-2 within prison units I and II in Campo Grande, Mato Grosso do Sul, Brazil. Conducted between May and October 2022, it reveals how the virus spreads in the confined settings of prisons, emphasizing the roles of overcrowded cells, frequent transfers, and limited healthcare access. The research involved 1927 participants (83.93% of the total prison population) and utilized nasopharyngeal swabs and RT-qPCR testing for detection. Contact tracing monitored exposure within cells. Out of 2108 samples, 66 positive cases were identified (3.13%), mostly asymptomatic (77.27%), with the majority aged 21–29 and varying vaccination statuses. Next-generation sequencing generated 28 whole genome sequences, identifying the Omicron variant (subtypes BA.2 and BA.5) with 99% average coverage. Additionally, the study seeks to determine the relationship between immunization levels and the incidence of SARS-CoV-2 cases within this enclosed population. The findings underscore the necessity of comprehensive control strategies in prisons, including rigorous screening, isolation protocols, vaccination, epidemiological monitoring, and genomic surveillance to mitigate disease transmission and protect both the incarcerated population and the broader community.

Four-pile caps made from concrete are essential elements for the force transfer from the superstructure to piles or pipes. Due to the difficulties in carrying out full-scale tests and all the instrumentation involved, the use of numerical models as a way to study the mechanical behavior of these elements presents itself as a good alternative. Such numerical studies usually provide useful information for the update and improvement of normative standards and codes. The concrete damaged plasticity (CDP) constitutive model, which combines damage and plasticity with smeared-crack propagation, stands out in the simulation of reinforced concrete. This model is composed of five parameters: dilatation angle (ψ), eccentricity (ϵ), ratio between biaxial and uniaxial compressive strength (σbo/σco), failure surface in the deviator plane normal to the hydrostatic axis (Kc), and viscosity (μ). For unidimensional elements, the values of the CDP parameters are well defined, but for volumetric elements, such as concrete pile caps, there is a gap in the literature regarding the definition of these values. This fact ends up limiting the use of the CDP on these structural elements due to the uncertainties involved. Therefore, the aim of this research was to calibrate two numerical models of concrete four-pile caps with different failure modes for the evaluation of the sensitivity of the CDP parameters, except for ϵ, which remained constant. As a result, the parameters σbo/σco and Kc did not significantly influence the calibration of the force × displacement curves of the simulated structures. Values of ψ and μ equal to 36° and 1 × 10−4, respectively, are recommended for “static” analysis, while for “quasi-static” analysis, ψ values ranging between 45° and 50° are suggested according to the failure mode. The results also showed to be sensitive to the constitutive relation of concrete tensile behavior in both modes of analysis. For geometric parameterization, the “static” analysis is recommended due to the lower coefficient of variation (3.29%) compared to the “quasi-static” analysis (19.18%). This conclusion is supported by the evaluation of the ultimate load of the numerical models from the geometrically parametric study compared to the results estimated by an analytical model.

Objectives: This investigation aimed to assess the optimal timing for lip repair in children with cleft lip and palate via 3D anthropometric analysis to evaluate their maxillofacial structures. Methods: The sample comprised 252 digitized dental models, divided into groups according to the following timing of lip repair: G1 (n = 50): 3 months; G2 (n = 50): 5 and 6 months; G3 (n = 26): 8 and 10 months. Models were evaluated at two-time points: T1: before lip repair; T2: at 5 years of age. Linear measurements, area, and Atack index were analyzed. Results: At T1, the intergroup analysis revealed that G1 had statistically significant lower means of I-C′, I-C, C-C′, and the sum of the segment areas compared to G2 (p = 0.0140, p = 0.0082, p = 0.0004, p < 0.0001, respectively). In addition, there was a statistically significant difference when comparing the cleft area between G2 and G3 (p = 0.0346). At T2, the intergroup analysis revealed that G1 presented a statistically significant mean I-C′ compared to G3 (p = 0.0461). In the I-CC’ length analysis, G1 and G3 showed higher means when compared to G2 (p = 0.0039). The I-T′ measurement was statistically higher in G1 than in G2 (p = 0.0251). In the intergroup growth rate analysis, G1 and G2 showed statistically significant differences in the I-C′ measurement compared to G3 (p = 0.0003). In the analysis of the Atack index, there was a statistically significant difference between G1 and the other sample sets (p < 0.0001). Conclusion: Children who underwent surgery later showed better results in terms of the growth and development of the dental arches.

This study aimed to evaluate the effect of inoculants of endophytic bacteria producing indoleacetic acid (IAA) on the physiological quality of seeds and the production of seedlings of Corymbia citriodora (Hook.) KD Hill & LAS Johnson. In the physiological quality test of the seeds, the treatments used were individual inoculation with Priestia megaterium, Exiguobacterium sibiricum, Pantoea vagans strain 45URP4-1, and Bacillus sp.; joint effect of the four strains (mix); inoculation only with the carrier (cassava starch and activated charcoal); carrier with 1.0 μg mL−1 of IAA; and non-inoculated control without IAA and without a carrier. In the production of seedlings in a greenhouse, the treatments were the same, except for the mix, which was replaced by P. vagans strain 7URP1-6 (Pvs7), as inoculation with the mix increased the number of abnormal seedlings. In the physiological quality test of seeds, seeds inoculated with the bacteria individually did not have the physiological quality impaired and the carrier created a microenvironment around the seeds, benefiting germination percentage, germination speed index, average germination time, and average germination speed. In the greenhouse, seedlings inoculated with Pvs7, P. megaterium and E. sibiricum were taller, with a larger stem diameter and dry mass of shoot, roots, and total. Seeds inoculated with E. sibiricum had higher averages for height, chlorophyll b content, and shoot and total dry mass, as well as a greater ability to colonize the rhizosphere and roots of C. citriodora, resulting in the production of higher-quality seedlings. Inoculation of seeds of C. citriodora with endophytic bacteria proved to be a promising alternative for plant development.

The Oropouche virus is an orthobunyavirus responsible for causing Oropouche fever, a disease that primarily affects thousands of people in South and Central America. Currently, no specific antiviral treatments or vaccines are available against this virus, highlighting the urgent need for safe, affordable, and effective therapies. Natural products serve as an important source of bioactive compounds, and there is growing interest in identifying natural bioactive molecules that could be used for treating viral diseases. Quercetin hydrate is a compound classified as a flavonoid, which has garnered scientific attention due to its potential health benefits and its presence in various plant-based foods. In this study, we aim to evaluate the in vitro antiviral activity of quercetin hydrate against the Oropouche virus (OROV). Furthermore, we intend to explore its mode of action through in silico approaches. The cytotoxicity and antiviral activity of the compound were assessed using Vero cells. In addition, in silico studies were also performed through molecular docking, molecular dynamics simulations, Molecular Mechanics Poisson–Boltzmann surface area (MM/PBSA), and quantum-mechanical analysis in order to evaluate the interaction with the Gc protein of OROV. The assay revealed that the compound was highly active against the virus, inhibiting OROV with an EC50 value of 53.5 ± 26.5 µM under post-infection treatment conditions. The present study demonstrates that the compound is a promising antiviral agent; however, the mechanisms of action proposed in this study need to be experimentally verified by future assays.

Background: The post-COVID-19 scenario has demonstrated the increasing importance of marketing for organizations, as retailers and entrepreneurs have had to adapt to new ways of selling their products and services. In this regard, this research aimed to identify challenges for developing the marketing plan of startups and validate them from the perspective of managers in the field, considering the market characteristics inherent to the post-COVID-19 era; Methods: To achieve this, a literature review and a survey were conducted among professionals in the field. The collected data were analyzed using the quantitative Lawshe method. Results: The results indicate that, for the development of the marketing plan of startups considering the post-COVID-19 reality, it is important to prioritize overcoming the challenges of “Consumer behavior pattern change”, “Differentiation from the competition”, “Digital expansion”, “Innovation capacity of companies”, “Creation of transformative marketing”, and “Reevaluation of marketing channels in the post-pandemic period”; Conclusions: Therefore, it can be concluded that these challenges reflect the main concerns and obstacles faced by startups in building effective marketing strategies and striving for a competitive position in the market. By recognizing and understanding these challenges, startups will be better prepared to face adversity and seize opportunities in this new market context.

Physical classification is the procedure adopted by the rice unloading, delivery, storage, and processing units for the commercial characterization of the quality of the grains. This step occurs mostly by the conventional method, which demands more time and specialized labor, and the results are subjective since the evaluation is visual. In order to make the operation faster, more accurate, and less dependent, non-destructive technologies and computational intelligence can be applied to characterize grain quality. Therefore, this study aimed to characterize and predict the quality of whole, processed rice grains, as well as classify any defects present. This was achieved by sampling from the upper and lower points of four silo dryers with capacities of up to 40,000 sacks. The grain samples had moisture contents of 16%, 17%, 18%, and 19% and were subjected to drying-aeration until reaching 12% moisture content (w.b.). Near-infrared spectroscopy technology and Machine Learning algorithm models (Artificial Neural Networks, decision tree algorithms Quinlan’s algorithm, Random Tree, REPTree, and Random Forest) were employed for this purpose. By analyzing Pearson’s correlation statistics, a strong negative correlation (R2 = 0.98) was found between moisture content and the yield of whole grains. Conversely, a strong positive correlation (R2 = 0.97) was observed between moisture content and classified physical defects across the various characterized physicochemical constituents. These findings indicate the effectiveness of near-infrared spectroscopy technology. The Random Tree model (RandT) successfully predicted the grain quality outcomes and is therefore recommended as the model of choice, obtained Pearson’s correlation coefficient (r = 0.96), mean absolute error (MAE = 0.017), and coefficient of determination (R2 = 0.92). The results obtained here reveal that the combination of near-infrared spectroscopy technology and Machine Learning algorithm models is an excellent non-destructive alternative to manual physical classification for characterizing the physicochemical quality of whole and defective rice grains.

The scientific community recognizes coffee grounds (Coffea arabica) as an important biological residue, which led to using the Eversa® Transform 2.0 lipase as an in silico enzymatic catalyst for coffee grounds’ free fatty acids (FFA). Molecular modeling studies, including molecular docking, were performed, which revealed the structures of the lipase and showed the primary interactions between the ligands and the amino acid residues in the active site of the enzyme. Of the ligands tested, 6,9-methyl octadienoate had the best free energy of −6.1 kcal/mol, while methyl octadecenoate and methyl eicosanoate had energies of −5.7 kcal/mol. Molecular dynamics confirmed the stability of the bonds with low Root Mean Square Deviation (RMSD) values. The MMGBSA study showed that methyl octadecenoate had the best free energy estimate, and CASTp identified key active sites for potential enzyme immobilization in experimental studies. Overall, this study provides efficient and promising results for future experimental investigations, showing a classification of oils present in coffee grounds and their binding affinity with Eversa.

Breast cancer is the most prevalent cancer among women worldwide. Therapeutic strategies to control tumors and metastasis are still challenging. Three-dimensional (3D) spheroid-type systems more accurately replicate the features of tumors in vivo, working as a better platform for performing therapeutic response analysis. This work aimed to characterize the epithelial–mesenchymal transition and doxorubicin (dox) response in a mammary tumor spheroid (MTS) model. We evaluated the doxorubicin treatment effect on MCF-7 spheroid diameter, cell viability, death, migration and proteins involved in the epithelial–mesenchymal transition (EMT) process. Spheroids were also produced from tumors formed from 4T1 and 67NR cell lines. MTSs mimicked avascular tumor characteristics, exhibited adherens junction proteins and independently produced their own extracellular matrix. Our spheroid model supports the 3D culturing of cells isolated from mice mammary tumors. Through the migration assay, we verified a reduction in E-cadherin expression and an increase in vimentin expression as the cells became more distant from spheroids. Dox promoted cytotoxicity in MTSs and inhibited cell migration and the EMT process. These results suggest, for the first time, that this model reproduces aspects of the EMT process and describes the potential of dox in inhibiting the metastatic process, which can be further explored.

Our study investigated the innate immune response to Toxoplasma gondii infection by assessing microglial phenotypic changes and sickness behavior as inflammatory response markers post-ocular tachyzoite instillation. Disease progression in Swiss albino mice was compared with the previously documented outcomes in BALB/c mice using an identical ocular route and parasite burden (2 × 105 tachyzoites), with saline as the control. Contrary to expectations, the Swiss albino mice displayed rapid, lethal disease progression, marked by pronounced sickness behaviors and mortality within 11–12 days post-infection, while the survivors exhibited no apparent signs of infection. Comparative analysis revealed the T. gondii-infected BALB/c mice exhibited reduced avoidance of feline odors, while the infected Swiss albino mice showed enhanced avoidance responses. There was an important increase in microglial cells in the dentate gyrus molecular layer of the infected Swiss albino mice compared to the BALB/c mice and their respective controls. Hierarchical cluster and discriminant analyses identified three microglial morphological clusters, differentially affected by T. gondii infection across strains. The BALB/c mice exhibited increased microglial branching and complexity, while the Swiss albino mice showed reduced shrunken microglial arbors, diminishing their morphological complexity. These findings highlight strain-specific differences in disease progression and inflammatory regulation, indicating lineage-specific mechanisms in inflammatory responses, tolerance, and resistance. Understanding these elements is critical in devising control measures for toxoplasmosis.

The role of nutrition in preventing non-communicable diseases has been widely studied in recent years, with indications that non-animal-based diets might improve body composition and therefore bring multiple health benefits. For all of these reasons, the main purpose was to compare body composition and metabolic status between vegetarian and omnivorous individuals and relate these values with cardiovascular risk. The present analysis included 176 participants (61 vegetarians and 115 omnivores). Body composition was assessed using a dual-energy X–ray absorptiometry, biochemical parameters obtained from capillary blood, and the 10-year cardiovascular risk (10RCVD) calculated by the QRISK3 score. No statistical differences were found between groups regarding body composition. Concerning metabolic markers, vegetarian individuals showed reduced values of total cholesterol, LDL cholesterol, and non–HDL cholesterol (p < 0.05). There were no differences in 10RCVD between groups. In both diets, moderate correlations between groups were found for cardiovascular risk and visceral adipose tissue. Our results suggest that the vegetarian regimen might be associated with better cardiometabolic biomarkers and better cardiovascular health, although controversial with the body composition trends observed. In conclusion, the results suggest that cardiovascular risk appears to be more influenced by body composition, mainly fat tissue, over dietary patterns itself.

The purpose of this study is to propose an integrative model for evaluating the effectiveness of performance management system (PMS). This model aims to systematize the dimensions and criteria used in the literature and provide clarity in terms of evaluation possibilities. A comprehensive review of the literature was conducted to identify the dimensions, criteria, and causal relationships used in evaluating PMS effectiveness. A sample of 57 articles was analyzed using content analysis. The study established dimensions and criteria that have been neglected in the literature. The review resulted in the proposal of an integrative model for evaluating PMS effectiveness, which incorporates individual and organizational dimensions and criteria identified in the literature. It sheds light on recurrently adopted dimensions, particularly those related to individual-level phenomena, and seeks to clarify current conceptual ambiguities. This study’s originality lies in its integrative approach, which diverges from the prevailing tendencies in the field. This study provides clarity regarding the conceptual confusion surrounding ambiguous concepts and generically applied measures that hinder the drawing of certain conclusions about the effectiveness of PMS.

Background/Objectives: Tibial diaphysis fractures are common injuries resulting from high-to-low-energy traumas in patients of all age groups, but few reports currently provide complementary parameters for the assessment of bone healing processes in the postoperative period. Serum alkaline phosphatase (ALP) and the scores from the Radiographic Union Scale for Tibial Fractures (RUST) can promote new horizons in this context. Therefore, the aim of this study was to assess the behavior of ALP and RUST through within-subject comparisons from immediately post-surgery to 49 days after tibial diaphysis fracture repair. Methods: This article included four case studies where patients underwent the same procedures. Adults of both sexes aged 18 to 60 years with tibial fractures requiring surgery were included. After surgical intervention (T1), the patients were followed for 49 days after surgery, returning for follow-up appointments on the 21st (T2) and 49th (T3) days. At the follow-up appointments, new X-ray images were obtained, and blood samples were collected for ALP measurement. Results: Serum ALP levels increased by T2 following tibial reamed intramedullary nailing surgery. While this increase persisted into T3 for two patients, a decline was observed during the same period for the other two patients. Both events are indicative of the bone consolidation process, and RUST scores at the T3 corroborate this perspective for all patients included in this study. Considering that delta ALP (T3-T1 value) was lower in patients who exhibited the highest RUST score, we suggest that a synchronized analysis between ALP and RUST allows medics to diagnose bone consolidation. Conclusions: Therefore, it can be concluded that the analysis of ALP alongside RUST may be complementary for evaluating bone consolidation following tibial reamed intramedullary nailing surgery, but future studies are needed to confirm this assertion.

Smart buildings play a key role in the complex ecosystem of cities and are often subject to barriers that limit their development. Although identifying these barriers is fundamental to creating an enabling environment for this segment’s expansion, few works aim to identify these challenges. This work has two main objectives: (1) to research the main barriers limiting the development of new smart building projects and (2) to prioritize these barriers from the perspective of professionals with experience in the field. We adopted an exploratory approach common in research that focuses on identifying and prioritizing variables related to a phenomenon, which is based on two main actions: obtaining information through a careful literature review and consulting professionals who work in the concerned field. The results showed that professionals assessed the 23 barriers identified through bibliographic research as important, with the most important being related to lack of qualified professionals, shortage of government policies, higher initial and construction costs, macroeconomic barriers and access to financing, high cost of intelligent systems and technologies, regulatory barriers, lack of knowledge about the current and potential benefits of smart buildings, and more complex design and construction.

Despite the central role of cats in the transmission and amplification of Sporothrix, studies regarding immune response in feline sporotrichosis are scarce. In cats with sporotrichosis, neutrophil-rich lesions are usually associated to good general condition and lower fungal burden. However, the role of neutrophils in anti-Sporothrix immunity has been little explored in cats. Thus, the aim of this study was to evaluate the neutrophil oxidative burst in the blood of cats with sporotrichosis. Cats with sporotrichosis included in the study were treated with itraconazole (ITZ) alone or combined with potassium iodide (KI). The neutrophil oxidative burst was evaluated through a flow-cytometry-based assay using dihydrorhodamine 123 (background) and stimulation with Zymosan and heat-killed Sporothrix yeasts. The cure rate was 50.0% in cats under treatment with ITZ monotherapy and 90.9% in cats treated with ITZ + KI (p = 0.014), endorsing the combination therapy as an excellent alternative for the treatment of feline sporotrichosis. Higher percentages of Sporothrix-stimulated neutrophils were associated with good general condition (p = 0.003). Higher percentages of Sporothrix- (p = 0.05) and Zymosan-activated (p = 0.014) neutrophils before and early in the treatment were related to clinical cure in ITZ-treated cats. The correlation between oxidative burst and successful use of KI could not be properly assessed given the low number of failures (n = 2) in this treatment group. Nasal mucosa involvement, typically linked to treatment failure, was related to lower percentages of activated neutrophils in the background at the treatment outcome (p = 0.02). Our results suggest a beneficial role of neutrophils in feline sporotrichosis and a positive correlation between neutrophil activation and the cure process in ITZ-treated cats.

This paper sets forth a thorough procedure to design surface-mounted permanent magnet synchronous generators. Since synchronous generators generate the majority of electrical energy, their relevance in society nowadays is substantial. As a consequence, the methodology to design these electrical machines also holds great importance. However, even though a considerable amount of works addresses the matter, it is difficult to find a complete and thoroughly explained design procedure. The proposed method is based on analytical equations to fully consider PM generator fundamentals with a few simplifications, which implies in a considerable number of design equations and parameters. Differently from most papers on the design of PM synchronous generators, a significant level of detail and explanation is presented, all design choices are discussed, and the suggested ranges for the design parameters are shown. This results in a straightforward procedure that allows non-experienced designers to easily replicate the results and effectively enhance the comprehension of permanent magnet synchronous machines, and provides a guideline for researchers from other fields who may need to understand and perform a synchronous generator design. To show the effectiveness of the proposed design procedure, a PM generator is designed, and the results are compared with a finite element simulation, showing good accuracy.

The rupture of an iron mining tailing dam in Brumadinho, Brazil, released around 10 million cubic meters of tailings, of which 1.6 Mm3 reached the Paraopeba River. In this work, a total of 30 samples from three bottom sediment cores were collected in the lower course of the Paraopeba River basin and analyzed for major, trace and rare earth elements by ICP-OES and ICP-MS. The sediments presented a range of compositions with different weathering histories, overall marked by depleted Ca2+, Na+ and K+ compared with the average UCC, PAAS and NASC and some advanced weathering trends. The samples presented a fractionation pattern characterized by a continuous depletion of light REEs from La to Sm and a regular decreased distribution of heavy REEs from Gd to Yb, and the Co/Th vs. La/Sc diagram indicates a predominant intermediate source. The upper samples presented the highest contents of REEs, probably due to the higher presence of iron and aluminum oxides and hydroxides, which can be related to more advanced weathering. The Al, Cu, Ni, V, Zn, Co, Mn, Ti, Fe and Si concentrations and the CF, EF and Igeo index values varied across the sediment core samples, demonstrating that there were long periods of geogenic or anthropogenic contributions.

In the New World, dogs are considered the main reservoir of visceral leishmaniasis (VL). Due to inefficacies in existing treatments and the lack of an efficient vaccine, dog culling is one of the main strategies used to control disease, making the development of new therapeutic interventions mandatory. We previously showed that Tanespimycin (17-AAG), a Hsp90 inhibitor, demonstrated potential for use in leishmaniasis treatment. The present study aimed to test the safety of 17-AAG in dogs by evaluating plasma pharmacokinetics, dose-proportionality, and the tolerability of 17-AAG in response to a dose-escalation protocol and multiple administrations at a single dose in healthy dogs. Two protocols were used: Study A: four dogs received variable intravenous (IV) doses (50, 100, 150, 200, or 250 mg/m2) of 17-AAG or a placebo (n = 4/dose level), using a cross-over design with a 7-day “wash-out” period; Study B: nine dogs received three IV doses of 150 mg/m2 of 17-AAG administered at 48 h intervals. 17-AAG concentrations were determined by a validated high-performance liquid chromatographic (HPLC) method: linearity (R2 = 0.9964), intra-day precision with a coefficient of variation (CV) ≤ 8%, inter-day precision (CV ≤ 20%), and detection and quantification limits of 12.5 and 25 ng/mL, respectively. In Study A, 17-AAG was generally well tolerated. However, increased levels of liver enzymes–alanine aminotransferase (ALT), aspartate aminotransferase (AST), and gamma-glutamyl transferase (GGT)–and bloody diarrhea were observed in all four dogs receiving the highest dosage of 250 mg/m2. After single doses of 17-AAG (50–250 mg/m2), maximum plasma concentrations (Cmax) ranged between 1405 ± 686 and 9439 ± 991 ng/mL, and the area under the curve (AUC) plotting plasma concentration against time ranged between 1483 ± 694 and 11,902 ± 1962 AUC 0–8 h μg/mL × h, respectively. Cmax and AUC parameters were dose-proportionate between the 50 and 200 mg/m2 doses. Regarding Study B, 17-AAG was found to be well tolerated at multiple doses of 150 mg/m2. Increased levels of liver enzymes–ALT (28.57 ± 4.29 to 173.33 ± 49.56 U/L), AST (27.85 ± 3.80 to 248.20 ± 85.80 U/L), and GGT (1.60 ± 0.06 to 12.70 ± 0.50 U/L)–and bloody diarrhea were observed in only 3/9 of these dogs. After the administration of multiple doses, Cmax and AUC 0–48 h were 5254 ± 2784 μg/mL and 6850 ± 469 μg/mL × h in plasma and 736 ± 294 μg/mL and 7382 ± 1357 μg/mL × h in tissue transudate, respectively. In conclusion, our results demonstrate the potential of 17-AAG in the treatment of CVL, using a regimen of three doses at 150 mg/m2, since it presents the maintenance of high concentrations in subcutaneous interstitial fluid, low toxicity, and reversible hepatotoxicity.

Studies on the interaction sites of peptide toxins and ion channels typically involve site-directed mutations in toxins. However, natural mutant toxins exist among them, offering insights into how the evolutionary process has conserved crucial sequences for activities and molecular target selection. In this study, we present a comparative investigation using electrophysiological approaches and computational analysis between two alpha toxins from evolutionarily close scorpion species of the genus Tityus, namely, Tst3 and Ts3 from T. stigmurus and T. serrulatus, respectively. These toxins exhibit three natural substitutions near the C-terminal region, which is directly involved in the interaction between alpha toxins and Nav channels. Additionally, we characterized the activity of the Tst3 toxin on Nav1.1-Nav1.7 channels. The three natural changes between the toxins did not alter sensitivity to Nav1.4, maintaining similar intensities regarding their ability to alter opening probabilities, delay fast inactivation, and induce persistent currents. Computational analysis demonstrated a preference for the down conformation of VSD4 and a shift in the conformational equilibrium towards this state. This illustrates that the sequence of these toxins retained the necessary information, even with alterations in the interaction site region. Through electrophysiological and computational analyses, screening of the Tst3 toxin on sodium isoform revealed its classification as a classic α-NaTx with a broad spectrum of activity. It effectively delays fast inactivation across all tested isoforms. Structural analysis of molecular energetics at the interface of the VSD4-Tst3 complex further confirmed this effect.

Hypertrophic cardiomyopathy (HCM) is a heart condition characterized by cellular and metabolic dysfunction, with mitochondrial dysfunction playing a crucial role. Although the direct relationship between genetic mutations and mitochondrial dysfunction remains unclear, targeting mitochondrial dysfunction presents promising opportunities for treatment, as there are currently no effective treatments available for HCM. This review adhered to the Preferred Reporting Items for Systematic Reviews and Meta-Analysis Extension for Scoping Reviews guidelines. Searches were conducted in databases such as PubMed, Embase, and Scopus up to September 2023 using “MESH terms”. Bibliographic references from pertinent articles were also included. Hypertrophic cardiomyopathy (HCM) is influenced by ionic homeostasis, cardiac tissue remodeling, metabolic balance, genetic mutations, reactive oxygen species regulation, and mitochondrial dysfunction. The latter is a common factor regardless of the cause and is linked to intracellular calcium handling, energetic and oxidative stress, and HCM-induced hypertrophy. Hypertrophic cardiomyopathy treatments focus on symptom management and complication prevention. Targeted therapeutic approaches, such as improving mitochondrial bioenergetics, are being explored. This includes coenzyme Q and elamipretide therapies and metabolic strategies like therapeutic ketosis. Understanding the biomolecular, genetic, and mitochondrial mechanisms underlying HCM is crucial for developing new therapeutic modalities.

In this work, a self-lubricating composite was manufactured using a novel hybrid 3D printing/in situ spraying process that involved the printing of an acrylonitrile butadiene styrene (ABS) matrix using fused deposition modeling (FDM), along with the in situ spraying of alumina (Al2O3) and hexagonal boron nitride (hBN) reinforcements during 3D printing. The results revealed that the addition of the reinforcement induced an extensive formation of micropores throughout the ABS structure. Under tensile-loading conditions, the mechanical strength and cohesive interlayer bonding of the composites were diminished due to the presence of these micropores. However, under tribological conditions, the presence of the Al2O3 and hBN reinforcement improved the frictional resistance of ABS in extreme loading conditions. This improvement in frictional resistance was attributed to the ability of the Al2O3 reinforcement to support the external tribo-load and the shearing-like ability of hBN reinforcement during sliding. Collectively, this work provides novel insights into the possibility of designing tribologically robust ABS components through the addition of in situ-sprayed ceramic and solid-lubricant reinforcements.

Drought is the most complex natural hazard that can occur over large spatial scales and during long time periods. It affects more people than any other natural hazard, particularly in areas with a dry climate, such as the semiarid region of the Brazilian Northeast (NEB), which is the world’s most populated dry area. In this work, we analyzed trends and the spatial distribution of drought characteristics (frequency, affected area, and intensity) based on the Standardized Precipitation Index (SPI) on annual (SPI-12) and seasonal (SPI-3) scales. The study used monthly precipitation data recorded between 1962 and 2012 at 133 meteorological stations in Pernambuco State, Brazil, which is located in the eastern part of the NEB and has more than 80% of its territory characterized by a semiarid climate. The regions of Sertão, Agreste, and Zona da Mata of Pernambuco were considered for comparison. The Mann–Kendall and Sen’s slope tests were used to detect the trend and determine its magnitude, respectively. The results indicated that annual droughts in the state of Pernambuco became more frequent from the 1990s onwards, with summer having the greatest spatial coverage, followed by winter, autumn, and spring. Sertão presented a greater number of stations with a significant positive trend in drought frequency. Regarding the drought-affected area, global events occurred in a greater number of years on an annual scale and during the summer. Trend analysis pointed to an increase in areas with drought events on both scales. As for the drought intensity, the entire state of Pernambuco experienced drought events with high intensity during the autumn. The relationship between drought characteristics indicated an increase in the affected area as the result of an increase in drought intensity.

Background: School meals represent a significant supply of nutrients for children in Brazil, especially those in conditions of social vulnerability. Objectives: This study aimed to assess the levels of arsenic (As), cadmium (Cd), and lead (Pb) in meals served in public elementary schools in four municipalities in the state of Bahia, Brazil, and assess the risk posed to children’s health. Methods: Ninety-six samples were collected from 16 schools, freeze-dried, and subjected to microwave-assisted digestion. The As, Cd, and Pb levels were determined by graphite furnace atomic absorption spectrometry. The risk assessment was based on calculating each element’s hazard quotient (HQ). Results: None of the samples reached or exceeded the tolerable levels for the elements analyzed. Pb was the metal that obtained the most significant result, reaching maximum levels of 39–157 µg·kg−1. Conclusions: No element exceeded the PTWI proposed by JECFA; thus, the toxic metal content in school meals poses a negligible risk to children’s health.

Ischemic heart disease represents a significant global burden of morbidity and mortality. While revascularization strategies are well defined in acute settings, there are uncertainties regarding chronic coronary artery disease treatment. Recent trials have raised doubts about the necessity of revascularization for “stable”, chronic coronary syndromes or disease, leading to a shift towards a more conservative approach. However, the issue remains far from settled. In this narrative review, we offer a summary of the most pertinent evidence regarding revascularization for chronic coronary disease, while reflecting on less-often-discussed details of major clinical trials. The cumulative evidence available indicates that there can be a prognostic benefit from revascularization in chronic coronary syndrome patients, provided there is significant ischemia, as demonstrated by either imaging or coronary physiology. Trials that have effectively met this criterion consistently demonstrate a reduction in rates of spontaneous myocardial infarction, which holds both prognostic and clinical significance. The prognostic benefit of revascularization in patients with heart failure with reduced ejection fraction remains especially problematic, with a single contemporary trial favouring surgical revascularization. The very recent publication of a trial focused on revascularizing non-flow-limiting “vulnerable” plaques adds further complexity to the field. The ongoing debates surrounding revascularization in chronic coronary syndromes emphasize the importance of personalized strategies. Revascularization, added to the foundational pillar of medical therapy, should be considered, taking into account symptoms, patient preferences, coronary anatomy and physiology, ischemia tests and intra-coronary imaging.

Drug-resistant bacteria constitute a big barrier against current pharmacotherapy. Efforts are urgent to discover antibacterial drugs with novel chemical and biological features. Our work aimed at the synthesis, evaluation of antibacterial effects, and toxicity of licochalcone C (LCC), a naturally occurring chalcone. The synthetic route included six steps, affording a 10% overall yield. LCC showed effects against Gram-positive bacteria (MIC = 6.2–50.0 µg/mL), Mycobacterium species (MIC = 36.2–125 µg/mL), and Helicobacter pylori (MIC = 25 µg/mL). LCC inhibited the biofilm formation of MSSA and MRSA, demonstrating MBIC50 values of 6.25 μg/mL for both strains. The investigations by fluorescence microscopy, using PI and SYTO9 as fluorophores, indicated that LCC was able to disrupt the S. aureus membrane, similarly to nisin. Systemic toxicity assays using Galleria mellonella larvae showed that LCC was not lethal at 100 µg/mL after 80 h treatment. These data suggest new uses for LCC as a compound with potential applications in antibacterial drug discovery and medical device coating.

Understanding climate change and land use impacts is crucial for mitigating environmental degradation. This study assesses the environmental vulnerability of the Doce River Basin for 2050, considering future climate change and land use and land cover (LULC) scenarios. Factors including slope, elevation, relief dissection, precipitation, temperature, pedology, geology, urban distance, road distance, and LULC were evaluated using multicriteria analysis. Regional climate models Eta-HadGEM2-ES and Eta-MIROC5 under RCP 4.5 and RCP 8.5 emission scenarios were employed. The Land Change Modeler tool simulated 2050 LULC changes and hypothetical reforestation of legal reserve (RL) areas. Combining two climate and two LULC scenarios resulted in four future vulnerability scenarios. Projections indicate an over 300 mm reduction in average annual precipitation and an up to 2 °C temperature increase from 2020 to 2050. Scenario 4 (RCP 8.5 and LULC for 2050 with reforested RLs) showed the greatest basin area in the lowest vulnerability classes, while scenario 3 (RCP 4.5 and LULC for 2050) exhibited more high-vulnerability areas. Despite the projected relative improvement in environmental vulnerability by 2050 due to reduced rainfall, the complexity of associated relationships must be considered. These results contribute to mitigating environmental damage and adapting to future climatic conditions in the Doce River Basin.

To evaluate differences in the morphology of the frontal sinus in adolescents and adults with different craniofacial patterns, searches up to April 2024 were conducted in six databases and other information sources to identify observational studies. Study selection, data extraction, and quality assessment using the NOS scale were performed independently by two reviewers. Random effects meta-analyses were conducted to estimate the difference in frontal sinus measurements between different craniofacial skeletal patterns (α = 0.05). The certainty of the evidence was evaluated according to GRADE. Fourteen studies were included in the review. All studies had methodological limitations that affected their quality. The syntheses showed that skeletal Class II subjects presented a significantly smaller width of the frontal sinus than skeletal Class I subjects (MD = 0.56; 95% CI: 0.38, 0.74; p < 0.0001; I2 = 3%). Skeletal Class III subjects showed a frontal sinus width (MD = −0.91; 95% CI: −1.35, −0.47; p < 0.0001; I2 = 36%) and area (MD = −28.13; 95% CI: −49.03, −7.23; p = 0.0084; I2 = 66%) significantly larger than those of the skeletal Class I subjects. The available evidence suggests a positive relationship between mandibular and frontal sinus size. There is limited evidence to make reliable estimates of the association of other craniofacial patterns and frontal sinus characteristics. These reported results are not conclusive and should be evaluated carefully due to the very low certainty of the evidence. The current evidence is scarce and consists of studies with methodological limitations; the results of the studies are often inconsistent, and the pooled estimates are imprecise. New high-quality research is still necessary.
