Colon cancer is a common and lethal malignancy, ranking second in global cancer-related mortality, highlighting the urgent need for novel targeted therapies. The sea cucumber (Apostichopus japonicus) is a marine organism known for its medicinal properties. After conducting a bioinformatics analysis of the cDNA library of Apostichopus japonicus, we found and cloned a cDNA sequence encoding histidine-rich peptides, and the recombinant peptide was named rAj-HRP. Human histidine-rich peptides are known for their anti-cancer properties, raising questions as to whether rAj-HRP might exhibit similar effects. To investigate whether rAj-HRP can inhibit colon cancer, we used human colon cancer HCT116 cells as a model and studied the tumor suppressive activity in vitro and in vivo. The results showed that rAj-HRP inhibited HCT116 cell proliferation, migration, and adhesion to extracellular matrix (ECM) proteins in vitro. It also disrupted the cytoskeleton and induced apoptosis in these cells. In vivo, rAj-HRP significantly inhibited the growth of HCT116 tumors in BALB/c mice, reducing tumor volume and weight without affecting the body weight of the tumor-bearing mice. Western blot analysis showed that rAj-HRP inhibited HCT116 cell proliferation and induced apoptosis by upregulating BAX and promoting PARP zymogen degradation. Additionally, rAj-HRP inhibited HCT116 cell adhesion and migration by reducing MMP2 levels. Further research showed that rAj-HRP downregulated EGFR expression in HCT116 cells and inhibited key downstream molecules, including AKT, P-AKT, PLCγ, P38 MAPK, and c-Jun. In conclusion, rAj-HRP exhibits significant inhibitory effects on HCT116 cells in both in vitro and in vivo, primarily through the EGFR and apoptosis pathways. These findings suggest that rAj-HRP has the potential as a novel targeted therapy for colon cancer.

This study introduces a deep learning framework based on SMILES representations of chemical structures to predict drug–drug interactions (DDIs). The model extracts Morgan fingerprints and key molecular descriptors, transforming them into raw graphical features for input into a modified ResNet18 architecture. The deep residual network, enhanced with regularization techniques, efficiently addresses training issues such as gradient vanishing and exploding, resulting in superior predictive performance. Experimental results show that StructNet-DDI achieved an AUC of 99.7%, an accuracy of 94.4%, and an AUPR of 99.9%, demonstrating the model’s effectiveness and reliability. These findings highlight that StructNet-DDI can effectively extract crucial features from molecular structures, offering a simple yet robust tool for DDI prediction.

The dung beetle optimization (DBO) algorithm is acknowledged for its robust optimization capabilities and rapid convergence as an efficient swarm intelligence optimization technique. Nevertheless, DBO, similar to other swarm intelligence algorithms, often gets trapped in local optima during the later stages of optimization. To mitigate this challenge, we propose the Move-to-Escape dung beetle optimization (MEDBO) algorithm in this paper. MEDBO utilizes a good point set strategy for initializing the swarm’s initial population, ensuring a more uniform distribution and diminishing the risk of local optima entrapment. Moreover, it incorporates convergence factors and dynamically balances the number of offspring and foraging individuals, prioritizing global exploration initially and local exploration subsequently. This dynamic adjustment not only enhances the search speed but also prevents local optima stagnation. The algorithm’s performance was assessed using the CEC2017 benchmark suite, which confirmed MEDBO’s significant improvements. Additionally, we applied MEDBO to three engineering problems: pressure vessel design, three-bar truss design, and spring design. MEDBO exhibited an excellent performance in these applications, demonstrating its practicality and efficacy in real-world problem-solving contexts.

Ultrashort light pulses have strong research and application values, while nonlinear inverse Thomson scattering has been considered as a unique source of zepto-yoctosecond pulses. Here, the mechanism of nonlinear inverse Thomson scattering of a high-energy electron colliding with a tightly focused intense laser pulse is investigated through numerical simulation. The time-compression effect was proposed to explain the origin of ultrashort pulses and the nonlinear phenomenon of electron radiation in the time–space joint distribution. It is found that the time scale of electron radiation is orders of magnitude shorter than that of electron motion, and the increases in laser intensity and electron initial energy will result in stronger and shorter pulses. Yoctosecond pulses can be generated by a laser pulse with an intensity of 1.384×1020 W/cm2 and an electron with an initial energy of 51.1 MeV. These results provide theoretical and numerical basis for generating shorter light pulses.

Self-powered wearable pressure sensors based on flexible electronics have emerged as a new trend due to the increasing demand for intelligent and portable devices. Improvements in pressure-sensing performance, including in the output voltage, sensitivity and response time, can greatly expand their related applications; however, this remains challenging. The Dung beetle optimization (DBO) algorithm, devised by Jiankai Xue in 2022, is known for its strong optimization capabilities and fast convergence. However, it does have certain limitations, including insufficiently random population initialization, slow search speed, and inadequate global search capabilities. Drawing inspiration from the mathematical properties of the Sinh and Cosh functions, we proposed a new metaheuristic algorithm, Sinh–Cosh Dung Beetle Optimization (SCDBO). By leveraging the Sinh and Cosh functions to disrupt the initial distribution of DBO and balance the development of rollerball dung beetles, SCDBO enhances the search efficiency and global exploration capabilities of DBO through nonlinear enhancements. These improvements collectively enhance the performance of the dung beetle optimization algorithm, making it more adept at solving complex real-world problems. To evaluate the performance of the SCDBO algorithm, we compared it with seven typical algorithms using the CEC2017 test functions. Additionally, by successfully applying it to three engineering problems, robot arm design, pressure vessel problem, and unmanned aerial vehicle (UAV) path planning, we further demonstrate the superiority of the SCDBO algorithm.

Background: Cold stress usually occurs in winter and is one of the most significant environmental factors restricting the growth of the tea plant as well as its geographical distribution. Objective: It is necessary to identify the physiological and molecular mechanisms of plants under cold stress so that cold-tolerant crop varieties can be cultivated to limit production losses. At the same time, this would allow the crop planting area to be expanded, hence improving the economic benefits. Methods: In this study, the transcriptome data of Yunwu Tribute Tea under cold conditions were obtained using the Illumina HiSeq platform. By analyzing changes in transcriptome data associated with the antioxidant enzyme system, plant hormone signal transduction, proline and tyrosine metabolism pathways, and transcription factors, the molecular mechanisms involved in Yunwu Tribute Tea under cold stress were investigated. Results: In this study, Illumina HiSeq technology was applied to investigate the cold-tolerance mechanism. For this purpose, cDNA libraries were obtained from two groups of samples, namely the cold-treated group (DW) and the control group (CK). A total of 185,973 unigenes were produced from 511,987 assembled transcripts; among these, 16,020 differentially expressed genes (DEGs) (corrected p-value < 0.01, |log2(fold change)| >3), including 9606 up-regulated and 6414 down-regulated genes, were obtained. Moreover, the antioxidant enzyme system, plant hormone signal transduction, proline and tyrosine metabolism pathways, and transcription factors were analyzed; based on these results, a series of candidate genes related to cold stress were screened out and discussed. The physiological indexes related to the low-temperature response were tested, along with five DEGs which were validated by quantitative real-time PCR. Conclusions: Differential gene expression analysis has confirmed that substantial cold-responsive genes are related to the antioxidant enzyme system, plant hormone signal transduction, proline metabolism pathway, tyrosine metabolism pathway, and transcription factors.

Clustering protocols and simultaneous wireless information and power transfer (SWIPT) technology can solve the issue of imbalanced energy consumption among nodes in energy harvesting-cognitive radio sensor networks (EH-CRSNs). However, dynamic energy changes caused by EH/SWIPT and dynamic spectrum availability prevent existing clustering routing protocols from fully leveraging the advantages of EH and SWIPT. Therefore, a multi-hop uneven clustering routing protocol is proposed for EH-CRSNs utilizing SWIPT technology in this paper. Specifically, an EH-based energy state function is proposed to accurately track the dynamic energy variations in nodes. Utilizing this function, dynamic spectrum availability, neighbor count, and other information are integrated to design the criteria for selecting high-quality cluster heads (CHs) and relays, thereby facilitating effective data transfer to the sink. Intra-cluster and inter-cluster SWIPT mechanisms are incorporated to allow for the immediate energy replenishment for CHs or relays with insufficient energy while transmitting data, thereby preventing data transmission failures due to energy depletion. An energy status control mechanism is introduced to avoid the energy waste caused by excessive activation of the SWIPT mechanism. Simulation results indicate that the proposed protocol markedly improves the balance of energy consumption among nodes and enhances network surveillance capabilities when compared to existing clustering routing protocols.

Saturated aqueous salt solutions have diverse applications in food production, mineral processing, pharmaceuticals, and environmental monitoring. However, the random and disordered arrangement of ions in these solutions poses limitations across different fields. In this study, we employ magnetic fields to regulate the disordered arrangement by a comprehensive methodology combining contact angle measurement, Raman spectroscopy, X-ray diffraction, and molecular dynamics simulations on saturated KCl solutions. Our findings reveal that weak magnetic fields impede the formation of K-Cl contact pairs and disrupt hydrogen bond networks, particularly DDAA and free OH types. However, they facilitate the interaction between water molecules and ions, leading to an increase in the number of K-O and Cl-H contact pairs, along with an expansion in ion hydration radius. These changes affect macroscopic properties, including the interaction with solid substrates and potential solubility increases. Our experimental and simulation results mutually validate each other, contributing to a theoretical framework for studying magnetic field–material interactions.

Background: The PREDICT-HN study aimed to systematically assess the kinetics of imaging MR biomarkers during head and neck radiotherapy. Methods: Patients with intact squamous cell carcinoma of the head and neck were enrolled. Pre-, during, and post-treatment MRI were obtained. Serial GTV and ADC measurements were recorded. The correlation between each feature and the GTV was calculated using Spearman’s correlation coefficient. The linear mixed model was used to evaluate the change in GTV over time. Results: A total of 41 patients completed the study. The majority (76%) had oropharyngeal cancer. A total of 36 patients had intact primary tumours that can be assessed on MRI, and 31 patients had nodal disease with 46 nodes assessed. Median primary GTV (GTVp) size was 14.1cc. The rate of GTVp shrinkage was highest between pre-treatment and week 4. Patients with T3-T4 tumours had a 3.8-fold decrease in GTVp compared to T1-T2 tumours. The ADC values correlated with residual GTVp. The median nodal volume (GTVn) was 12.4cc. No clinical features were found to correlate with GTVn reduction. The overall change in ADC for GTVn from pre-treatment was significant for 35th–95th percentiles in weeks 1–4 (p < 0.001). Conclusion: A discrepancy in the trajectory of ADC between primary and nodal sites suggested that they exhibit different treatment responses and should be analysed separately in future studies.

Based on the resonance fluorescence scattering mechanism, a narrowband sodium (Na) lidar can measure temperature and wind in the mesosphere and lower thermosphere (MLT) region. By using a narrowband spectral filter, background light noise during the day can be suppressed, allowing for continuous observations. To obtain full-diurnal-cycle temperature and wind measurement results, a complex and precise retrieval process is required, along with necessary corrections to minimize measurement errors. This paper introduces the design of a data acquisition unit for three frequencies in three directions of the Na lidar system in the Chinese Meridian Project (Phase II) and investigates the calibration and retrieval methods for obtaining diurnal temperature and horizontal wind in the MLT region, using a Na Doppler lidar with Faraday anomalous dispersion optical filter (FADOF). Furthermore, these methods are applied to observations conducted by a Na lidar in Beijing, China. The wind and temperature results over full diurnal cycles obtained from the all-solid-state Na Doppler lidar are reported for the first time and compared with temperature measurements from satellite, as well as wind observations from a meteor radar. The comparison demonstrates a reasonable agreement between the results, indicating the rationality of the lidar-retrieved results and the feasibility and effectiveness of the data correction and retrieval method.

This study presents a novel degradation pathway of a human immunoglobulin G (IgG) molecule featuring a light chain N-terminal asparagine. We thoroughly characterize this pathway and investigate its charge profiles using cation exchange chromatography (CEX) and capillary isoelectric focusing (cIEF). Beyond the well-documented asparagine deamidation into isoaspartic acid, aspartic acid, and succinimide intermediate, a previously unreported clipping degradation pathway is uncovered. This newly identified clipped N-terminal IgG variant exhibits a delayed elution in CEX, categorized as a “basic variant”, while retaining the same main peak isoelectric point (pI) in cIEF. The influence of temperature and pH on N-terminal asparagine stability is assessed across various stressed conditions. A notable correlation between deamidation percentage and clipped products is established, suggesting a potential hydrolytic chemical reaction underlying the clipping process. Furthermore, the impact of N-terminal asparagine modifications on potency is evaluated through ELISA binding assays, revealing minimal effects on binding affinity. Sequence alignment reveals homology to a human IgG with the germline gene from Immunoglobulin Lambda Variable 6-57 (IGLV6-57), which has implications for amyloid light-chain (AL) amyloidosis. This discovery of the N-terminal clipping degradation pathway contributes to our understanding of immunoglobulin light chain misfolding and amyloid fibril deposition under physiological conditions.

Malto-oligosaccharides (MOSs) from starch conversion is advantageous for food and pharmaceutical applications. In this study, an efficient malto-oligosaccharide-forming α-amylase AmyCf was identified from myxobacter Cystobacter sp. strain CF23. AmyCf is composed of 417 amino acids with N-terminal 41 amino acids as the signal peptide, and conserved glycoside hydrolase family 13 (GH13) catalytic module and predicted C-terminal domain with β-sheet structure are also identified. Phylogenetic and functional analysis demonstrated that AmyCf is a novel member of GH13_6 subfamily. The special activity of AmyCf toward soluble starch and raw wheat starch is 9249 U/mg and 11 U/mg, respectively. AmyCf has broad substrate specificity toward different types of starches without requiring Ca2+. Under ideal circumstances of 60 °C and pH 7.0, AmyCf hydrolyzes gelatinized starch into maltose and maltotriose and maltotetraose as the main hydrolytic products with more than 80% purity, while maltose and maltotriose are mainly produced from the hydrolysis of raw wheat starch with more than 95% purity. The potential applicability of AmyCf in starch processing is highlighted by its capacity to convert gelatinized starch and raw starch granules into MOSs. This enzymatic conversion technique shows promise for the low-temperature enzymatic conversion of raw starch.

To achieve the non-destructive detection of peanut defects, a multi-target identification method based on the multispectral system and improved Faster RCNN is proposed in this paper. In terms of the system, the root-mean-square contrast method was employed to select the characteristic wavelengths for defects, such as mildew spots, mechanical damage, and the germ of peanuts. Then, a multispectral light source system based on a symmetric integrating sphere was designed with 2% nonuniformity illumination. In terms of Faster RCNN improvement, a texture-based attention and a feature enhancement module were designed to enhance the performance of its backbone. In the experiments, a peanut-deficient multispectral dataset with 1300 sets was collected to verify the detection performance. The results show that the evaluation metrics of all improved compared with the original network, especially in the VGG16 backbone network, where the mean average precision (mAP) reached 99.97%. In addition, the ablation experiments also verify the effectiveness of the proposed texture module and texture enhancement module in peanut defects detection. In conclusion, texture imaging enhancement and efficient extraction are effective methods to improve the network performance for multi-target peanut defect detection.

Flavin mononucleotides (FMNs) and flavin adenine nucleotide (FAD) play vital roles in the electron-transfer processes in diverse enzymatic reactions. Owing to the isoalloxazine chromophore, flavins are easily detectable by surface-enhanced Raman spectroscopy (SERS), a surface-sensitive technique. However, the details of the adsorption of flavins on SERS-active materials have never been investigated. In this study, a comprehensive SERS analysis of flavins containing lumichrome and lumiflavin on silver nanoparticles was conducted. With the aid of density-functional-theory calculations, our results suggested that the flavin molecules were adsorbed on the silver nanoparticles via the N3 site of the isoalloxazine moiety, which had a stronger adsorption ability than the adenine moiety in the FAD. The SERS spectra of the flavins at different pH values also supported this conclusion. This study demonstrated the feasibility of SERS for the structural characterization of flavins, paving the way for the functional exploration of flavin-labeled detection sensors and flavoprotein researches.

Electricity load prediction is an essential tool for power system planning, operation and management. The critical information it provides can be used by energy providers to maximise power system operation efficiency and minimise system operation costs. Long Short-Term Memory (LSTM) and Support Vector Machine (SVM) are two suitable methods that have been successfully used for analysing time series problems. In this paper, the two algorithms are explored further for load prediction; two load prediction algorithms are developed and verified by using the half-hourly load data from the University of Warwick campus energy centre with four different prediction time horizons. The novelty lies in comparing and analysing the prediction accuracy of two intelligent algorithms with multiple time scales and in exploring better scenarios for their prediction applications. High-resolution load forecasting over a long range of time is also conducted in this paper. The MAPE values for the LSTM are 2.501%, 3.577%, 25.073% and 69.947% for four prediction time horizons delineated. For the SVM, the MAPE values are 2.531%, 5.039%, 7.819% and 10.841%, respectively. It is found that both methods are suitable for shorter time horizon predictions. The results show that LSTM is more capable of ultra-short and short-term forecasting, while SVM has a higher prediction accuracy in medium-term and long-term forecasts. Further investigation is performed via blind tests and the test results are consistent.

Lodging is one of the major constraints in attaining high yield in crop production. Major factors associated with stalk lodging involve morphological traits and anatomical features along with the chemical composition of the stem. However, little relevant research has been carried out in sorghum, particularly on the anatomical aspects. In this study, with a high-throughput procedure newly developed by our research group, the nine parameters related to stem regions and vascular bundles were generated in 58 sorghum germplasm accessions grown in two successive seasons. Correlation analysis and principal component analysis were conducted to investigate the relationship between anatomical aspects and stalk mechanical traits (breaking force, stalk strength and lodging index). It was found that most vascular parameters were positively associated with breaking force and lodging index with the correlation coefficient r varying from −0.46 to 0.64, whereas stalk strength was only associated with rind area with the r = 0.38. The germplasm resources can be divided into two contrasting categories (classes I with 23 accessions and II with 30 accessions). Compared to class II, the class I was characterized by a larger number (+40.7%) and bigger vascular bundle (+30%), thicker stem (+19.6%) and thicker rind (+36.0%) but shorter internode (plant) (−91.0%). This study provides the methodology and information for the studies of the stem anatomical parameters in crops and facilitates the selective breeding of sorghum.

Based on the established LIDAR chain along the 120°E meridian in China, the gravity wave (GW) activity between 80 and 105 km and the seasonal behavior of temporal and spatial spectra at Beijing, Hefei and Hainan were studied with three years of continued observations. The averaged GW-induced atmospheric density perturbations are near 6%, which in summer are obviously larger than in winter. The semiannual maxima occur near the solstice and the minimum emerges around the equinox at different latitudes. Besides, as a disparity, the density perturbation of GW is still active considerably in winter at low latitude at Hainan. The spectra of vertical wave number Fa(m) and observed frequency Fa(ω) show power law shapes, of which the average is near −3 and −1.7, respectively, and both spectra with special values all exhibit similar seasonal behavior as the atmospheric density perturbations shows. This behavior is explained by multiple effects possibly originating from Tibet plateau (TP) and the main GW source in China was roughly calculated by the LIDAR observation method for the first time located at the TP boundary.

Sugarcane is cultivated mainly for its high sucrose content but it can also produce many metabolites with promising antioxidant potential. However, very few studies have been reported on the biosynthesis of metabolites in sugarcane to date. In this study, we have identified a wide range of amino acids and organic acids in the rind of six sugarcane varieties by the LC-MS/MS method. A total number of 72 amino acids and 55 organic acid compounds were characterized; among these, 100 were reported for the first time. Moreover, 13 amino acids and seven organic acids were abundantly distributed in all varieties tested and considered major amino acids and organic acids in sugarcane. The variety Taitang134 (F134) showed the highest content of total amino acids, whereas the varieties ROC16 and Yuetang93/159 (YT93/159) had maximum content of organic acids. The amino acids of the rind extract presented higher antioxidant capacity than the organic acids of the rind extract. In addition, the transcriptomic and metabolic integrated analysis highlighted some candidate genes associated with amino acid biosynthesis in sugarcane. We selected a transcription factor gene, MYB(t), and over-expressed it in Arabidopsis. The transgenic plants showed a higher accumulation of amino acids with higher antiradical activity compared with the wild-type Arabidopsis plants. Thus, we characterize a wide range of amino acids and organic acids and their antiradical activities in different sugarcane varieties and present candidate genes that can be potentially valuable for the genetic improvement of metabolites in sugarcane bagasse

In recent years, sea cucumber has become a favorite healthcare food due to its characteristic prevention of cardiovascular diseases, suppression of tumors, as well as enhancement of immunity. In order to screen the anti-tumoral proteins or peptides from sea cucumber (Apostichopus japonicus), its cDNA library was analyzed, and a disintegrin-like and metalloproteinase with thrombospondin type 1 motif, member 13 (ADAMTS13)-like was found. ADAMTS13-like contains 10 thrombospondin 1 (TSP1) domains. Based on analysis of bioinformatics, the third TSP1 domain of this protein, which is further named Aj-Tspin, contains an arginine-glycine-aspartate (RGD) motif. Since our previous studies showed that the recombinant RGD-containing peptide from lampreys showed anti-tumoral activity, the third TSP1 domain of ADAMTS13-like was chosen to evaluate it’s effect on tumor proliferation and metastasis, despite the fact it shares almost no homologue with disintegrins from other species. After artificial synthesis, its cDNA sequence, Aj-Tspin, which is composed of 56 amino acids, was subcloned into a pET23b vector and expressed as a recombinant Aj-Tspin (rAj-Tspin) in a soluble form with a molecular weight of 6.976 kDa. Through affinity chromatography, rAj-Tspin was purified as a single protein. Both anti-proliferation and immunofluorescence assays showed that rAj-Tspin suppressed the proliferation of Lewis lung carcinoma (LLC) cells through apoptosis. Adhesion assay also displayed that rAj-Tspin inhibited the adhesion of LLC cells to ECM proteins, including fibronectin, laminin, vitronectin and collagen. Lastly, rAj-Tspin also suppressed the migration and invasion of LLC cells across the filter in transwells. Thus, the above indicates that rAj-Tspin might act as a potential anti-tumoral drug in the future and could also provide information on the nutritional value of sea cucumber.

Bcl-2-associated athanogene (BAG), a group of proteins evolutionarily conserved and functioned as co-chaperones in plants and animals, is involved in various cell activities and diverse physiological processes. However, the biological functions of this gene family in rice are largely unknown. In this study, we identified a total of six BAG members in rice. These genes were classified into two groups, OsBAG1, -2, -3, and -4 are in group I with a conserved ubiquitin-like structure and OsBAG5 and -6 are in group Ⅱ with a calmodulin-binding domain, in addition to a common BAG domain. The BAG genes exhibited diverse expression patterns, with OsBAG4 showing the highest expression level, followed by OsBAG1 and OsBAG3, and OsBAG6 preferentially expressed in the panicle, endosperm, and calli. The co-expression analysis and the hierarchical cluster analysis indicated that the OsBAG1 and OsBAG3 were co-expressed with primary cell wall-biosynthesizing genes, OsBAG4 was co-expressed with phytohormone and transcriptional factors, and OsBAG6 was co-expressed with disease and shock-associated genes. β-glucuronidase (GUS) staining further indicated that OsBAG3 is mainly involved in primary young tissues under both primary and secondary growth. In addition, the expression of the BAG genes under brown planthopper (BPH) feeding, N, P, and K deficiency, heat, drought and plant hormones treatments was investigated. Our results clearly showed that OsBAGs are multifunctional molecules as inferred by their protein structures, subcellular localizations, and expression profiles. BAGs in group I are mainly involved in plant development, whereas BAGs in group II are reactive in gene regulations and stress responses. Our results provide a solid basis for the further elucidation of the biological functions of plant BAG genes.

Based on 139 nights of observational data of the Rayleigh lidar site in Beijing, China (40.5° N, 116.2° E), typical lower MIL cases and their temperature inversion evolution process were reported and compared with the SABER data from the same time. Meanwhile, the seasonal distribution of lower MIL cases over North China was also statistically analyzed. The average inversion temperature of the low MIL is 23.4 K, and the average layer thickness is 4.78 km with an average MIL bottom altitude of 68.2 km. Meanwhile, 65% of the MIL propagates vertically, most of which goes downward. These results show the temperature behavior properties of the lower MIL over North China, which may be helpful for us to further understand middle atmosphere chemical and dynamics processes.

Semi-dwarfism is a main agronomic trait in crop breeding. In this study, we performed genome-wide association study (GWAS) and identified a new quantitative trait nucleotide (QTN) for rice shoot length. The peak QTN (C/T) was located in the first coding region of a group III WRKY transcription factor OsWRKY21 (LOC_Os01g60640). Interestingly, further haplotype analysis showed that C/T difference only existed in the indica group but not in the japonica group, resulting in significant differences in plant height among the different indica rice varieties. OsWRKY21 was expressed in embryo, radicle, shoots, leaves, and stems. Most notably, overexpressing OsWRKY21 resulted in the semi-dwarf phenotype, early heading date and short internodes compared to the wild type, while the knockout mutant plants by CRISPR/Cas9 technology yielded the opposite. The overexpressing lines exhibited the decreased length of the cells near sclerenchyma epidermis, accompanied with the lower levels of indole-3-acetic acid (IAA) and gibberellin 3 (GA3), but increased levels of the abscisic acid (ABA) and salicylic acid (SA) in the internodes at heading stage. Moreover, the semi-dwarf phenotype could be fully rescued by exogenous GA3 application at seedling stage. The RNA-seq and qRT-PCR analysis confirmed the differential expression levels of genes in development and the stress responses in rice, including GA metabolism (GA20ox2, GA2ox6, and YABY1) and cell wall biosynthesis (CesA4, 7, and 9) and regulation (MYB103L). These data suggest the essential role of OsWRKY21 in regulation of internode elongation and plant height in rice.

Compressed air energy storage is a well-used technology for application in high voltage power systems, but researchers are also investing efforts to minimize the cost of this technology in medium and low voltage power systems. Integration of this energy storage requires a robust control of the power electronic converter to control the power injection due to the dynamic behavior of the system. The conventional linear control design requires a thorough knowledge of the system parameters, but the uncertain disturbances caused by the mechanical properties of the energy storage is neglected in the design and the system fails in presence of such instances. In this paper an adaptive control-based boost converter and sliding mode control-based three phase inverter for a grid integrated compressed air energy storage system of up to 1 kW has been presented that can mitigate any uncertain disturbances in the system without prior knowledge of the system parameters. The experimental results along with the simulation results are also presented to validate the efficiency of the system.

The efficiency of solar photovoltaic (PV) panels is greatly reduced by panel soiling and high temperatures. A mechanism for eliminating both of these sources of inefficiencies is presented by integrating solar PV generation with a compressed air system. High-pressure air can be stored and used to blow over the surface of PV panels, removing present dust and cooling the panels, increasing output power. A full-system mathematical model of the proposed system is presented, comprised of compressed air generation and storage, panel temperature, panel cleaning, and PV power generation. Simulation results indicate the benefit of employing compressed air for cleaning and cooling solar PV panels. For a fixed volume of compressed air, it is advantageous to blow air over the panels early in the day if the panel is soiled or when solar radiation is most abundant with the highest achievable flow rate if the panel is clean. These strategies have been shown to achieve the greatest energy captures for a single PV panel. When comparing the energy for air compression to the energy gain from cleaning a single PV over a two-week period, an energy ROI of 23.8 is determined. The system has the potential to eliminate the requirement for additional manual cleaning of solar PV panels.

The increasing integration of large-scale electricity generation from renewable energy sources in the grid requires support through cheap, reliable, and accessible bulk energy storage technologies, delivering large amounts of electricity both quickly and over extended periods. Compressed air energy storage (CAES) represents such a storage option, with three commercial facilities using salt caverns for storage operational in Germany, the US, and Canada, with CAES now being actively considered in many countries. Massively bedded halite deposits exist in the UK and already host, or are considered for, solution-mined underground gas storage (UGS) caverns. We have assessed those with proven UGS potential for CAES purposes, using a tool developed during the EPSRC-funded IMAGES project, equations for which were validated using operational data from the Huntorf CAES plant. From a calculated total theoretical ‘static’ (one-fill) storage capacity exceeding that of UK electricity demand of ≈300 TWh in 2018, filtering of results suggests a minimum of several tens of TWh exergy storage in salt caverns, which when co-located with renewable energy sources, or connected to the grid for off-peak electricity, offers significant storage contributions to support the UK electricity grid and decarbonisation efforts.

With the development of society, the energy crisis has become increasingly prominent, which greatly affects the sustainable development of the economy of various countries. Industrial energy consumption accounts for more than 70% of China’s total energy consumption, of which more than 50% is converted to industrial waste heat, and recyclable waste heat resources account for about 60% of the total waste heat resources, while China’s current utilization rate of industrial waste heat only reaches about 30%. The development of renewable energy and recovery of low-grade waste heat in industry is the key to solve the problem. As a type of volumetric expander with full flow expansion, the screw expander is extensively applied in the industrial waste heat recovery and geothermal energy generation industry because of its effective utilization of low enthalpy energy. Improving the performance of the screw expander as the core, the paper concludes and summarizes the research status of the leakage, rotor geometry, sealing and lubrication, processing and manufacturing, which can affect the performance of the screw expander. In addition, it also introduces the application status and potential utilization of screw expander.

In this paper, the structure and photoelectric characteristics of zincblende InxGa1−xN alloys are systematically calculated and analyzed based on the density functional theory, including the lattice constant, band structure, distribution of electronic states, dielectric function, and absorption coefficient. The calculation results show that with the increase in x, the lattice constants and the supercell volume increase, whereas the bandgap tends to decrease, and InxGa1−xN alloys are direct band gap semiconductor materials. In addition, the imaginary part of the dielectric function and the absorption coefficient are found to redshift with the increase in indium composition, expanding the absorption range of visible light. By analyzing the lattice constants, polarization characteristics, and photoelectric properties of the InxGa1−xN systems, it is observed that zincblende InxGa1−xN can be used as an alternative material to replace the channel layer of wurtzite InxGa1−xN heterojunction high electron mobility transistor (HEMT) devices to achieve the manufacture of HEMT devices with higher power and higher frequency. In addition, it also provides a theoretical reference for the practical application of InxGa1−xN systems in optoelectronic devices.

As an important part of a micro-compressed air energy storage system, the scroll expander directly affects the efficiency of the whole energy storage system. The effects of resistance on the efficiency of scroll expander caused by inlet structure and size are discussed with theory analysis and experimental methods in this paper. Micro-compressed air energy storage system has aroused widespread attention because of its pollution-free, high flexibility, in the community, remote areas power supply. Comprehensive experimental work with the selections of different size and structure of the air inlet of the scroll expander was performed with the cutting angle of air inlet chamber of the scroll expander. The results of the experiments are discussed on how exergy efficiency and inlet flow of the scroll expander were affected resulting from the cutting angles dissected. The results show that a maximum value exists for exergy efficiency of the scroll expander. Therefore, the exergy efficiency of the scroll expander can be effectively improved by enlarging the air inlet port dimension and modifying the size of air chamber.

This paper presents a critical review of the research conducted for modeling and controlling supercritical power plants. Thermal power plants are classified according to the boiler pressure to supercritical and subcritical. The modeling concepts and control strategies of supercritical generation units are far more complex than those of subcritical. On the other hand, supercritical generation technologies are more efficient and much cleaner than subcritical generation units. From a deep technical analysis of the literature, there is no review that is dedicated to models-based control of supercritical power plants and most previous reviews are found to be too general to modeling-based control of fossil fuelled energy sources. This review reports the advancements on modeling and control of supercritical and ultra-supercritical plants as cleaner generation technologies. The various published achievements for modeling supercritical and ultra-supercritical units have been reviewed. The control strategies that fulfill the practical load demand requirements while keeping optimum efficiencies are also reviewed. Finally, expected future directions are reported as recommendations to overcome future challenges. The paper can be used as a brief educational directory to the postgraduate students or future researchers in the field.

The low thermospheric sodium layer (LTSL) is the separate sodium atom layer above 105 km. Based on 11,607 h of lidar observations from Yanqing (40.5° N, 116.0° E) from 2010 to 2016, we found 38 LTSLs wherein the peak densities were more than five percent above those of the main sodium layers. This work presents the peak altitudes, peak local times and peak densities of the LTSLs as well as the long-term characteristics of the seasonal and inter-annual variations of LTSLs. We analyzed the correlation between the LTSL and sporadic E layer (Es). The seasonal variation trends of the occurrences of LTSL and Es are similar, and the results showed that 95% of the LTSLs were accompanied by Es. We also found that 69% of the LTSL cases exhibited apparent downward phase progressions, while the descending rates of the LTSLs are consistent with the phase speeds of the tide.

Based on application of the atomic filter technology in a signal detection system of lidar, the diurnal observation of sodium lidar were obtained using the system at the National Space Science Center of the Chinese Academy of Sciences at Beijing Yanqing station (40.5° N, 116° E) in April 2014. During the lidar observation period, among the 103 cases of continuous daytime observations, the longest time was 181 h. In the case of a continuous observation period of 5 days (13−18 October 2014), the signal-to-noise ratio reached to 19:1 at 12:00−13:00 Local Time of the daytime, when the spatial and time resolutions were respectively set to 96 m of 167 s. The improvements resulted in the highest detection level of any existing diurnal lidars in China. Some interesting phenomena such as the sporadic sodium layer have also been observed during the daytime. The daytime capability extended the observing time range of the earlier systems that were limited to only nighttime observations. This innovation provides a useful method for the studies of diurnal tides, photochemistry, gravity waves, and correlative modeling studies.

As the number of renewable energy sources connected to the grid has increased, the need to address the intermittency of these sources becomes essential. One solution to this problem is to install energy storage technologies on the grid to provide a buffer between supply and demand. One such energy storage technology is Compressed Air Energy Storage (CAES), which is suited to large-scale, long-term energy storage. Large scale CAES requires underground storage caverns, such as the salt caverns situated in the Cheshire Basin, UK. This study uses cavern data from the Cheshire Basin as a basis for performing an energy and exergy analysis of 10 simulated CAES systems to determine the exergy storage potential of the caverns in the Cheshire Basin and the associated work and power input and output. The analysis revealed that a full charge of all 10 caverns could store 25.32 GWh of exergy, which can be converted to 23.19 GWh of work, which requires 43.27 GWh of work to produce, giving a round trip efficiency of around 54%. This corresponds to an input power of 670.07 GW and an output power of 402.74 GW. The Cheshire Basin could support around 100 such CAES plants, giving a potential total exergy storage capacity of 2.53 TWh and a power output of 40 TW. This is a significant amount of storage which could be used to support the UK grid. The total exergy destroyed during a full charge, store, and discharge cycle for each cavern ranged from 299.02 MWh to 1600.00 MWh.

Thermal energy storage using the latent heat of phase change materials (PCMs) is a promising technique to solve the time mismatch between the availability and usage of flue gas heat in distributed generation systems (DGSs). A diesel-engine-powered DGS integrated with two-stage tube-type PCM modules for exhaust gas heat recovery was developed and studied. Energy and exergy analysis for the PCM storage unit was carried out to verify the effectiveness of the PCM modules for heat recovery and to highlight the merits of the cascaded configuration through a practical engineering case. Furthermore, the performance of the DGS was evaluated to study the contribution of PCM storage to improving system efficiency. The results showed that 56.4% energy and 48.3% exergy of the input flue gas were stored by the two-stage storage unit. Additional integration of the low-temperature PCM module to the high-temperature module improved the average storage efficiency from 33.6% to 62.3% for energy and 33.1% to 50.8% for exergy. By utilizing the stored energy for heating water, the thermal efficiency of the diesel engine was increased from the original 35.8% to 41.9%, while the exergy efficiency was improved from 29.5% to 29.7%.

Cystatins are a family of cysteine protease inhibitors which are associated with a variety of physiological and pathological processes in vivo. In the present study, the cDNA sequence of a cystatin F homologue called Lm-cystatin F was cloned from the buccal glands of Lampetra morii. Although Lm-cystatin F shares a lower homology with cystatin superfamily members, it is also composed of a signal peptide and three highly conserved motifs, including the G in the N-terminal, QXVXG, as well as the PW in the C-terminal of the sequence. After sequence optimization and recombination, the recombinant protein was expressed as a soluble protein in Escherichia coli with a molecular weight of 19.85 kDa. Through affinity chromatography and mass spectrometry analysis, the purified protein was identified as a recombinant Lm-cystatin F (rLm-cystatin F). Additionally, rLm-cystatin F could inhibit the activity of papain. Based on MTT assay, rLm-cystatin F inhibited the proliferation of human umbilical vein endothelial cells (HUVECs) dose dependently with an IC50 of 5 μM. In vitro studies show that rLm-cystatin F suppressed the adhesion, migration, invasion, and tube formation of HUVECs, suggesting that rLm-cystatin F possesses anti-angiogenic activity, which provides information on the feeding mechanisms of Lampetra morii and insights into the application of rLm-cystatin F as a potential drug in the future.

To ensure the stability and reliability of the power network operation, a number of Grid Codes have been used to specify the technical boundary requirements for different countries and areas. With the fast propagation of the usage of Electrical Energy Storage (EES), it is quite important to study how the EES technology with its development can help the Grid Code realization. The paper provides a comprehensive study of Great Britain (GB) Grid Code mainly on its voltage and frequency relevant specifications, with a comparison of other countries’ grid operation regulations. The different types of EES technologies with their technical characteristics in relation to meeting Grid Codes have been analysed. From the study, apart from direct grid-connection to provide grid services on meeting Grid Codes, EES devices with different technologies can be used as auxiliary units in fossil-fuelled power plants and renewable generation to support the whole systems’ operation. The paper also evaluates the potentials of different types of EES technologies for implementing the relevant applications based on the Grid Codes. Some recommendations are given at the end, for the EES technology development to help the Grid Code realization and to support the relevant applications.

In the previous study, rLj-RGD3, a recombinant toxin protein which contains three RGD motifs, was reported to not only inhibit the proliferation of an ovarian cancer cell line, HeyA8 cells, by inducing apoptosis, but also block their adhesion, migration and invasion processes. However, whether rLj-RGD3 could also suppress the tumor growth in HeyA8 xenografted mice has not been reported yet. In the present study, rLj-RGD3 was intraperitoneally injected in the nude mice bearing HeyA8 tumors. Compared with the control group (normal saline), rLj-RGD3 inhibited the tumor growth significantly in the HeyA8 xenografted mice in a dose-dependent manner without affecting their body weights. Based on the H&E, Hoechst 33258 and TUNEL staining assays, as well as western blot analysis, rLj-RGD3 reduced the weight and volume of the solid tumors, probably by disturbing the tissue structure, inducing apoptosis and suppressing the FAK/PI3K/AKT pathway. Most importantly, rLj-RGD3 was found to prolong the survival days of the ovarian tumor xenografted mice, which suggested rLj-RGD3 might act as an effective and safe drug to treat ovarian cancer patients.

With the increase of power generation from renewable energy sources and due to their intermittent nature, the power grid is facing the great challenge in maintaining the power network stability and reliability. To address the challenge, one of the options is to detach the power generation from consumption via energy storage. The intention of this paper is to give an overview of the current technology developments in compressed air energy storage (CAES) and the future direction of the technology development in this area. Compared with other energy storage technologies, CAES is proven to be a clean and sustainable type of energy storage with the unique features of high capacity and long-duration of the storage. Its scale and cost are similar to pumped hydroelectric storage (PHS), thus CAES has attracted much attention in recent years while further development for PHS is restricted by the availability of suitable geological locations. The paper presents the state-of-the-art of current CAES technology development, analyses the major technological barriers/weaknesses and proposes suggestions for future technology development. This paper should provide a useful reference for CAES technology research and development strategy.

The current load balance in the grid is managed mainly through peaking fossil-fuelled power plants that respond passively to the load changes. Intermittency, which comes from renewable energy sources, imposes additional requirements for even more flexible and faster responses from conventional power plants. A major challenge is to keep conventional generation running closest to the design condition with higher load factors and to avoid switching off periods if possible. Thermal energy storage (TES) integration into the power plant process cycle is considered as a possible solution for this issue. In this article, a technical feasibility study of TES integration into a 375-MW subcritical oil-fired conventional power plant is presented. Retrofitting is considered in order to avoid major changes in the power plant process cycle. The concept is tested based on the complete power plant model implemented in the ProTRAX software environment. Steam and water parameters are assessed for different TES integration scenarios as a function of the plant load level. The best candidate points for heat extraction in the TES charging and discharging processes are evaluated. The results demonstrate that the integration of TES with power plant cycle is feasible and provide a provisional guidance for the design of the TES system that will result in the minimal influence on the power plant cycle.
