Gastroenterologists are exposed daily to musculoskeletal (MSK) stress during upper and lower gastrointestinal endoscopy, both during routine endoscopies and during long, demanding therapeutic procedures. There is evidence that endoscopy-related MSK injuries are becoming more common, particularly in the back, neck, shoulders, elbows, and hands. The aims of this study were to identify the most stressed muscle groups during endoscopy; to measure their activity using surface electromyographical (EMG) sensors; to detect areas of muscle overload; and to identify the number of microbreaks taken in specific muscles. Furthermore, we measured differences in the loading of these muscle groups with and without the use of special support systems such as a belt-like holder. Measurements were performed on 15 subjects (7 experienced endoscopists and 8 non-endoscopists). Due to this small sample size, inside each group, we achieved inconclusive results regarding statistically significant differences in different muscle groups. We increased the sample size by comparing all participants with and without the belt support system, disregarding their endoscopic background. There was a statistically significant difference (p < 0.05) in muscle tension and in levels of microbreaks in the muscles of the left forearm, biceps, and trapezius muscles. No statistically significant difference was observed in the muscle tension and level of microbreaks in the left deltoid muscle (p > 0.05). We hypothesize that the increased level of muscle loading and decreased level of microbreaks in the deltoid muscle are due to different muscle activity and different shoulder movements. Additionally, the deltoid muscle is not connected to the kinetic chain of body posture and stabilization. It is our belief that MSK injuries in gastrointestinal (GI) endoscopy can be prevented with the use of a belt-like support system.

A faulty voltage measurement can lead to the overcharging of a Li-Ion cell, resulting in gas formation and heating inside the cell, which can trigger thermal runaway. To mitigate this risk, cylindrical cells are equipped with a Current Interrupt Device (CID), which functions as a pressure relief valve, disconnecting the electrical circuit within the cell when internal pressure rises. However, this disconnection causes the cell to suddenly become highly resistant, posing a significant issue in series-connected cells. In such configurations, a portion or even the entire system voltage may drop across the disconnected cell, substantially increasing the likelihood of an electric arc. This arc could ignite any escaping flammable gases, leading to catastrophic failures. In a series of tests conducted on three different cell chemistries—NMC (Nickel Manganese Cobalt), NCA (Nickel Cobalt Aluminum), and LFP (Lithium Iron Phosphate)—it was found that the safe operation of the CID cannot be guaranteed for system voltages exceeding 120 V. Although comparative tests at double the nominal cell voltage did not exhibit the same behavior, these findings suggest that current safety standards, which recommend testing at double the nominal voltage, may not adequately address the risks involved. The tests further revealed that series connections of cells with CIDs are inherently dangerous, as, in the worst-case scenario, the entire system voltage can be concentrated across a single cell, leading to potential system failure.

The Microplastic Index (MPI) was presented in a previous paper as a method to assess the formation of microplastics during the application of impact and wear stresses, based on selected mechanical and physical properties of polymers. In this paper, the experimental validation of the MPI model is presented. A series of ten polymers was characterized to obtain the relevant parameters for the calculation of the MPI, i.e., the minimum particle size and volume of microplastics formed. The milling (addressing impact stress) and sanding experiments (addressing wear stress) resulted in particle sizes between 3 and 200 μm and 0.3 and 25 μm, respectively. These values were very well predicted by the MPI model, showing smaller particles for brittle polymers and larger ones for ductile polymers. In addition, the experimental-specific wear rates of impact and wear correlated well with the predicted ones, being 0.01–30 mm3/Nm for impact and 0.0002–0.012 mm3/Nm for wear. These results indicate that the MPI can be very well used to predict the tendency of a material to form microplastics. In the search for understanding and mitigating microplastic formation, the MPI can be used by both producers and end users to choose plastic grades that form fewer microplastics.

GI endoscopy forms an important part in the daily practice of a gastroenterologist. Musculoskeletal injuries related to GI endoscopy are on the rise. Observational studies and surveys depict the importance of a proper ergonomic environment when performing GI endoscopy. Our aim is to describe the pathophysiology, risk factors and possible preventive measures to reduce the risk of musculoskeletal injuries during gastrointestinal (GI) endoscopy. We review the most relevant studies that outline the gravity of the problem. A detailed analysis of proposed ergonomic recommendations is performed, outlining crucial steps in injury prevention. Proper ergonomic education is a key step in preventing occupational injury. Robotics and other mechanical solutions offer a variety of approaches to tackling the ergonomic challenge. Implementing proper ergonomic education and mechanical solutions lowers the possibility of occupational injury. The strategies and appliances presented are beneficial to all GI endoscopy practitioners.

Childhood critical illness can have long-term effects on families, but the extent and trajectory of recovery for parents are unknown. Using prospective longitudinal design, we describe the health outcomes of parents and their trajectory six months after paediatric intensive care unit (PICU) discharge. Parents reported health outcomes at PICU discharge (baseline), and 1-, 3-, and 6-months post-discharge. We used the Pediatric Quality-of-Life Family Impact Module, Patient Health Questionnaire-4, and post-traumatic stress disorder (PTSD) Checklist for DSM-5. The group-based trajectory model was used to identify recovery patterns. We included 128 parents of children aged 1 month to 18 years, admitted to the PICU for ≥48 h. Three post-discharge composite health trajectory groups were classified: 54 mild (42%), 68 moderate (53%), and 6 severe (4%). Parents in the mild and moderate groups returned to baseline health within the first 3 months, but those in the severe group exhibited worse outcomes at 6-months. The mean (SD) PICU stay durations for mild, moderate, and severe groups were 9 (16), 7 (10), and 38 (61) days; days of mechanical ventilation were 4 (5), 4 (7), and 18 (25) days; and readmission rates were 12 (22%), 23 (34%), and 4 (66%), respectively. Identifying these trajectories enables novel, targeted interventions for at-risk parents, underscoring the significance of integrated PICU follow-up care.

This study investigates the challenges and potential of conventional injection molding for producing thick-walled optical components. The research primarily focuses on optimizing process parameters and mold design to enhance product quality. The methods include software simulations and experimental validation using polycarbonate test samples (optical lenses). Significant parameters such as melt temperature, mold temperature, injection pressure, and packing pressure were varied to assess their impact on geometric accuracy and visual properties. The results show that lower melt temperatures and higher mold temperatures significantly reduce the occurrence of dimensional defects. Additionally, the design of the gate system was found to be crucial in minimizing defects and ensuring uniform material flow. Effective packing pressure was essential in reducing volumetric shrinkage and sink marks. Furthermore, we monitored the deviation between the predicted and actual defects relative to the thickness of the sample wall. After optimization, the occurrence of obvious defects was eliminated across all sample thicknesses (lenses), and the impact of the critical defect, the sink mark on the planar side of the lens, was minimized. These findings demonstrate the substantial potential of conventional injection molding to produce high-quality thick-walled parts when these parameters are precisely controlled. This study provides valuable insights for the efficient design and manufacturing of optical components, addressing the growing demand for high-performance thick-walled plastic products.

This study aimed to retrospectively assess the cost-effectiveness of various COVID-19 vaccination strategies in Ethiopia. It involved healthcare workers (HCWs) and community participants; and was conducted through interviews and serological tests. Local SARS-CoV-2 variants and seroprevalence rates, as well as national COVID-19 reports and vaccination status were also analyzed. A cost-effectiveness analysis was performed to determine the most economical vaccination strategies in settings with limited vaccine access and high SARS-CoV-2 seroprevalence. Before the arrival of the vaccines, 65% of HCWs had antibodies against SARS-CoV-2, indicating prior exposure to the virus. Individuals with prior infection exhibited a greater antibody response to COVID-19 vaccines and experienced fewer new infections compared to those without prior infection, regardless of vaccination status (5% vs. 24%, p < 0.001 for vaccinated; 3% vs. 48%, p < 0.001 for unvaccinated). The cost-effectiveness analysis indicated that a single-dose vaccination strategy is optimal in settings with high underlying seroprevalence and limited vaccine availability. This study underscores the need for pragmatic vaccination strategies tailored to local contexts, particularly in high-seroprevalence regions, to maximize vaccine impact and minimize the spread of COVID-19. Implementing a targeted approach based on local seroprevalence information could have helped Ethiopia achieve higher vaccination rates and prevent subsequent outbreaks.

Deficits in memory performance have been linked to a wide range of neurological and neuropsychiatric conditions. While many studies have assessed the memory impacts of individual conditions, this study considers a broader perspective by evaluating how memory recall is differentially associated with nine common neuropsychiatric conditions using data drawn from 55 international studies, aggregating 15,883 unique participants aged 15–90. The effects of dementia, mild cognitive impairment, Parkinson’s disease, traumatic brain injury, stroke, depression, attention-deficit/hyperactivity disorder (ADHD), schizophrenia, and bipolar disorder on immediate, short-, and long-delay verbal learning and memory (VLM) scores were estimated relative to matched healthy individuals. Random forest models identified age, years of education, and site as important VLM covariates. A Bayesian harmonization approach was used to isolate and remove site effects. Regression estimated the adjusted association of each clinical group with VLM scores. Memory deficits were strongly associated with dementia and schizophrenia (p < 0.001), while neither depression nor ADHD showed consistent associations with VLM scores (p > 0.05). Differences associated with clinical conditions were larger for longer delayed recall duration items. By comparing VLM across clinical conditions, this study provides a foundation for enhanced diagnostic precision and offers new insights into disease management of comorbid disorders.

Dilated cardiomyopathy (DCM) is characterized by reduced left ventricular ejection fraction (LVEF) and left or biventricular dilatation. We evaluated sex-specific associations of circulating proteins and metabolites with structural and functional heart parameters in DCM. Plasma samples (297 men, 71 women) were analyzed for proteins using Olink assays (targeted analysis) or LC-MS/MS (untargeted analysis), and for metabolites using LC MS/MS (Biocrates AbsoluteIDQ p180 Kit). Associations of proteins (n = 571) or metabolites (n = 163) with LVEF, measured left ventricular end diastolic diameter (LVEDDmeasured), and the dilation percentage of LVEDD from the norm (LVEDDacc. to HENRY) were examined in combined and sex-specific regression models. To disclose protein–metabolite relations, correlation analyses were performed. Associations between proteins, metabolites and LVEF were restricted to men, while associations with LVEDD were absent in both sexes. Significant metabolites were validated in a second independent DCM cohort (93 men). Integrative analyses demonstrated close relations between altered proteins and metabolites involved in lipid metabolism, inflammation, and endothelial dysfunction with declining LVEF, with kynurenine as the most prominent finding. In DCM, the loss of cardiac function was reflected by circulating proteins and metabolites with sex-specific differences. Our integrative approach demonstrated that concurrently assessing specific proteins and metabolites might help us to gain insights into the alterations associated with DCM.

Pancreatic ductal adenocarcinoma (PDAC) is the most lethal form of pancreatic cancer characterized by therapy resistance and early metastasis, resulting in a low survival rate. Histone deacetylase (HDAC) inhibitors showed potential for the treatment of hematological malignancies. In PDAC, the overexpression of HDAC 2 is associated with the epithelial–mesenchymal transition (EMT), principally accompanied by the downregulation of the epithelial marker E-cadherin and increased metastatic capacity. The effector cytokine transforming growth factor-β (TGF β) is known to be a major inducer of the EMT in PDAC, leading to high metastatic and invasive potential. In addition, the overexpression of HDAC 6 in PDAC is associated with reduced apoptosis. Here, we have demonstrated that a novel HDAC 2/6 inhibitor not only significantly increased E-cadherin expression in PANC-1 cells (5.5-fold) and in 3D PDAC co-culture spheroids (2.5-fold) but was also able to reverse the TGF-β-induced downregulation of E-cadherin expression. Moreover, our study indicates that the HDAC inhibitor mediated re-differentiation resulting in a significant inhibition of tumor cell invasion by approximately 60% compared to control. In particular, we have shown that the HDAC inhibitor induces both apoptosis (2-fold) and cell cycle arrest. In conclusion, the HDAC 2/6 inhibitor acts by suppressing invasion via upregulating E-cadherin mediated by HDAC 2 blockade and by inducing cell cycle arrest leading to apoptosis via HDAC 6 inhibition. These results suggest that the HDAC 2/6 inhibitor might represent a novel therapeutic strategy for the treatment of PDAC tumorigenesis and metastasis.

Rubber blends based on styrene–butadiene rubber, ethylene–propylene–diene monomer rubber and a combination of both rubbers were cured with different sulfur and peroxide curing systems. In sulfur curing systems, two type of accelerators, namely tetramethylthiuram disulfide, N-cyclohexyl-2-benzothiazole sulfenamide, and combinations of both accelerators were used. In peroxide curing systems, dicumyl peroxide, and a combination of dicumyl peroxide with zinc diacrylate or zinc dimethacrylate, respectively, were applied. The work was aimed at investigating the effect of curing systems composition as well as the type of rubber or rubber combinations on the curing process, cross-link density and physical–mechanical properties of vulcanizates. The dynamic mechanical properties of the selected vulcanizates were examined too. The results revealed a correlation between the cross-link density and physical–mechanical properties. Similarly, there was a certain correlation between the cross-linking degree and glass transition temperature. The tensile strength of vulcanizates based on rubber combinations was higher when compared to that based on pure rubbers, which points out the fact that in rubber combinations, not only are the features of both elastomers combined, but improvement in the tensile characteristics can also be achieved. When compared to vulcanizates cured with dicumyl peroxide, materials cured with a sulfur system exhibited higher tensile strength. With the application of co-agents in peroxide vulcanization, the tensile strength overcame the tensile behavior of sulfur-cured vulcanizates.

Currently, therapeutic and diagnostic applications of antibodies are primarily limited to cell surface-exposed and extracellular proteins. However, research has been conducted on cell-penetrating peptides (CPP), as well as cytosol-penetrating antibodies, to overcome these limitations. In this context, a heparin sulfate proteoglycan (HSPG)-binding antibody was serendipitously discovered, which eventually localizes to the cytosol of target cells. Functional characterization revealed that the tested antibody has beneficial cytosol-penetrating capabilities and can deliver cargo proteins (up to 70 kDa) to the cytosol. To achieve tumor-specific cell targeting and cargo delivery through conditional activation of the cell-penetrating antibody in the tumor microenvironment, a single-chain Fc fragment (scFv) and a VL domain were isolated as masking units. Several in vitro assays demonstrated that fusing the masking protein with a cleavable linker to the cell penetration antibody results in the inactivation of antibody cell binding and internalization. Removal of the mask via MMP-9 protease cleavage, a protease that is frequently overexpressed in the tumor microenvironment (TME), led to complete regeneration of binding and cytosol-penetrating capabilities. Masked and conditionally activated cytosol-penetrating antibodies have the potential to serve as a modular platform for delivering protein cargoes addressing intracellular targets in tumor cells.

Injection molding technology is widely utilized across various industries for its ability to fabricate complex-shaped components with exceptional dimensional accuracy. However, challenges related to injection quality often arise, necessitating innovative approaches for improvement. This study investigates the influence of surface roughness on the efficiency of conformal cooling channels produced using additive manufacturing technologies, specifically Direct Metal Laser Sintering (DMLS) and Atomic Diffusion Additive Manufacturing (ADAM). Through a combination of experimental measurements, including surface roughness analysis, scanning electron microscopy, and cooling system flow analysis, this study elucidates the impact of surface roughness on coolant flow dynamics and pressure distribution within the cooling channels. The results reveal significant differences in surface roughness between DMLS and ADAM technologies, with corresponding effects on coolant flow behavior. Following that fact, this study shows that when cooling channels’ surface roughness is lowered up to 90%, the reduction in coolant media pressure is lowered by 0.033 MPa. Regression models are developed to quantitatively describe the relationship between surface roughness and key parameters, such as coolant pressure, Reynolds number, and flow velocity. Practical implications for the optimization of injection molding cooling systems are discussed, highlighting the importance of informed decision making in technology selection and post-processing techniques. Overall, this research contributes to a deeper understanding of the role of surface roughness in injection molding processes and provides valuable insights for enhancing cooling system efficiency and product quality.

Alternative solvents are being tested as green solvents to replace the traditional organic solvents used in both academy and industry. Some of these are already available, such as ethyl lactate, cyrene, limonene, glycerol, and others. This alternative explores eco-friendly processes for extracting secondary metabolites from nature, thus increasing the number of unconventional extraction methods with lower environmental impact over conventional methods. In this context, the Peruvian Ambrosia arborescens was our model while exploring a microwave-assisted extraction (MAE) approach over maceration. The objective of this study was to perform a phytochemical study including UHPLC-ESI-MS/MS and the antioxidant activity of Ambrosia arborescens, using sustainable strategies by mixing both microwaves and ethyl lactate as a green solvent. The results showed that ethyl lactate/MAE (15.07%) achieved a higher extraction yield than methanol/maceration (12.6%). In the case of the isolation of psilostachyin, it was similar to ethyl lactate (0.44%) when compared to methanol (0.40%). Regarding UHPLC-ESI-MS/MS studies, the results were similar. Twenty-eight compounds were identified in the ethyl lactate/MAE and methanol/maceration extracts, except for the tentative identification of two additional amino acids (peaks 4 and 6) in the MeOH extract. In relation to the antioxidant assay, the activity of the ethyl lactate extract was a little higher than the methanol extract in terms of ORAC (715.38 ± 3.2) and DPPH (263.04 ± 2.8). This study on A. arborescens demonstrated that the unconventional techniques, such as MAE related to ethyl lactate, could replace maceration/MeOH for the extraction and isolation of metabolites from diverse sources. This finding showed the potential of unconventional methods with green solvents to provide eco-friendly methods based on green chemistry.

The corrosion behavior of a hybrid laminate consisting of laser-structured aluminum EN AW-6082 ∪ carbon fiber-reinforced polymer was investigated. Specimens were corroded in aqueous NaCl electrolyte (0.1 mol/L) over a period of up to 31 days and characterized continuously by means of scanning electron and light microscopy, supplemented by energy dispersive X-ray spectroscopy. Comparative linear sweep voltammetry was employed on the first and seventh day of the corrosion experiment. The influence of different laser morphologies and production process parameters on corrosion behavior was compared. The corrosion reaction mainly arises from the aluminum component and shows distinct differences in long-term corrosion morphology between pure EN AW-6082 and the hybrid laminate. Compared to short-term investigations, a strong influence of galvanic corrosion on the interface is assumed. No distinct influences of different laser structuring and process parameters on the corrosion behavior were detected. Weight measurements suggest a continuous loss of mass attributed to the detachment of corrosion products.

There is an urgent need for interventions in addressing the rapid and disproportionate impact of antimicrobial resistance (AMR) and climate change (CC) on low- and middle-income countries. Within this context, it is important to understand indigenous knowledge in rural communities, which are highly affected. This study examined knowledge, attitude, and practices (KAP) regarding AMR and CC in the Adadle district, Somali region, Ethiopia, utilizing mixed methods, including 362 surveys and 12 focus group discussions among rural communities. Findings showed that 39% and 63% of participants were familiar with AMR and CC, respectively. Of those surveyed, 57% attributed AMR to inappropriate antimicrobial use in animals and humans, while CC was often associated with Allah/God. Multivariable analysis indicated that males exhibited superior knowledge and a positive attitude towards AMR and CC. Additionally, individuals aged 26–35 and 36–45 years showed heightened awareness of AMR and CC, respectively. Moreover, participants who were government employees, pastoralists, and business owners showed better knowledge on CC compared to family caretaker. Religious education and households with more than six members were linked to lower AMR knowledge. This study underlines a greater awareness of CC than AMR and highlights gender-based disparities, recommending integrated educational AMR programs targeting different demographics through a One Health lens, actively involving females, and incorporating local beliefs and practices.

The sintering dynamics of nickel nanoparticles (Ni NPs) were investigated through a comprehensive approach that included in situ transmission electron microscopy annealing and molecular dynamics simulations. This study systematically examines the transformation behaviors of Ni NP agglomerates over a temperature spectrum from room temperature to 850 °C. Experimental observations, supported by molecular dynamics simulations, revealed the essential influence of rotational and translational motions of particles, especially at lower temperatures, on sintering outcomes. The effect of the orientation of particles on the sintering process was confirmed, with initial configurations markedly determining sintering efficiency and dynamics. Calculated activation energies from this investigation follow those reported in the literature, confirming surface diffusion as the predominant mechanism driving the sintering of Ni NPs.

In this study, we performed a detailed analysis of -sputtered-nylon 6,6 plasma polymer nanoparticles (NPs). Following a previous study using standard techniques such as X-ray photoelectron spectroscopy (XPS) and Fourier transform infrared (FTIR) spectroscopy, we employed unconventional approaches, specifically solid- and liquid-state high-resolution nuclear magnetic resonance (NMR) spectroscopy, supplemented by gel permeation chromatography (GPC). Scanning electron microscopy (SEM) was also used to examine changes in the size of the NPs after contact with solvents and after heating. Our investigations revealed suspected strong binding and networking of the NPs, and a soluble monomer/oligomer phase was identified and characterised. This fraction is removable using solvent or heat treatment without significantly affecting the size of the NPs. Additionally, we suggested the chemical structure of this soluble phase. Our findings support the proposed rubber-like character of plasma polymer NPs and explain their strong tendency to reflect from substrates upon high-speed impact.

In this paper, we investigate a validation process in order to assess the predictive capabilities of a single degree-of-freedom oscillator. Model validation is understood here as the process of determining the accuracy with which a model can predict observed physical events or important features of the physical system. Therefore, assessment of the model needs to be performed with respect to the conditions under which the model is used in actual simulations of the system and to specific quantities of interest used for decision-making. Model validation also supposes that the model be trained and tested against experimental data. In this work, virtual data are produced from a non-linear single degree-of-freedom oscillator, the so-called oracle model, which is supposed to provide an accurate representation of reality. The mathematical model to be validated is derived from the oracle model by simply neglecting the non-linear term. The model parameters are identified via Bayesian updating. This calibration process also includes a modeling error due to model misspecification and modeled as a normal probability density function with zero mean and standard deviation to be calibrated.

The modification of polymer materials’ useful properties can be applicable in many industrial areas due to the ability to make commodity and technical plastics (plastics that offer many benefits, such as processability, by injection molding) useful in more demanding applications. In the case of injection-molded parts, one of the most suitable methods for modification appears to be high-energy irradiation, which is currently used primarily for the modification of mechanical and thermal properties. However, well-chosen doses can effectively modify the properties of the surface layer as well. The purpose of this study is to provide a complex description of high-energy radiation’s (β radiation) influence on the useful properties of injection-molded parts made from common polymers. The results indicate that β radiation initiates the cross-linking process in material and leads to improved mechanical properties. Besides the cross-linking process, the material also experiences oxidation, which influences the properties of the surface layer. Based on the measured results, the main outputs of this study are appropriately designed regression models that determine the optimal dose of radiation.

Lithium–ion batteries are well established as traction batteries for electric vehicles. This has led to a growing market for second-life batteries that can be used in applications like home energy storage systems. Moreover, the recyclability and safe handling of aged or damaged cells and packs has become more important. While there are several indicators, like state of health (SOH), state of power (SOP), or state of safety (SOS), which describe the state of a battery before its defined end of life (EOL), there is no consistent classification methodology by which to describe the usability of a cell or pack after its EOL is reached. The proposed state of usability (SOU) provides a new indicator that accounts for the usability for second life, recyclability, and possible required safety handling of a lithium–ion battery after its first intended life cycle. This work presents a decision tree method, which in turn leads to five discrete usability levels enabling a fast and rough determination of the SOU for practical use. Further, a calculation methodology for reasonable continuous regions of the SOU is proposed. Both methods are based on a literature-based rating of all of the relevant defect and aging mechanisms displayed in a risk matrix. Finally, some experimental methods that can be used for SOU determination are proposed. The developed methodology and the hands-on approach using a decision tree are well-suited for real world application in recycling companies and battery test laboratories.

The carbon (C) fixed by crops, which is exported with harvest and retained as postharvest residues in a field, is important for calculating the C balance. The aim of this study was to determine the effect of water availability on the C content in whole wheat plants. In a three-year field trial, the weights of grain, straw, chaff, stubble, and roots of two cultivars of winter wheat (Triticum aestivum L.) and one cultivar of einkorn wheat (Triticum monococcum L.) and their carbon contents were determined in water stress, irrigation, and rain-fed control treatments. The water availability, year, and cultivar had a significant influence on the C content in aboveground plant parts, but the effect of water on grain C was weak. The C content decreased with irrigation and increased with drought, but the differences were small (at most, 3.39% in chaff). On average, the C contents of grain, straw, chaff, and roots reached 45.0, 45.7, 42.6, and 34.9%, respectively. The amount of C exported with grain and left on the field in the form of postharvest residues depended on the weight of the total biomass and the ratio of grain to straw and residue. Whole plant C yield reached 8.99, 7.46, and 9.65 t ha−1 in rain-fed control, stressed, and irrigated treatments, respectively, and 8.91, 9.45, and 7.47 t ha−1 in Artix, Butterfly, and Rumona, respectively. Irrigation significantly increased the C content in grain and straw (but not in chaff, stubble, and roots) in comparison with water shortage conditions. On average, a grain yield of 1 t ha−1 corresponded to an average export of 0.447–0.454 t C ha−1 in the grain of all cultivars and inputs of 0.721, 0.832, and 2.207 t C ha−1 of residue to the soil in the form of straw and postharvest residue in the two cultivars of common wheat and one of einkorn. The results of the study provided reliable data for the calculation of the C balance of wheat under conditions of different water availability.

Bioprinting provides a powerful tool for regenerative medicine, as it allows tissue construction with a patient’s specific geometry. However, tissue culture and maturation, commonly supported by dynamic bioreactors, are needed. We designed a workflow that creates an implant-specific bioreactor system, which is easily producible and customizable and supports cell cultivation and tissue maturation. First, a bioreactor was designed and different tissue geometries were simulated regarding shear stress and nutrient distribution to match cell culture requirements. These tissues were then directly bioprinted into the 3D-printed bioreactor. To prove the ability of cell maintenance, C2C12 cells in two bioinks were printed into the system and successfully cultured for two weeks. Next, human mesenchymal stem cells (hMSCs) were successfully differentiated toward an adipocyte lineage. As the last step of the presented strategy, we developed a prototype of an automated mobile docking station for the bioreactor. Overall, we present an open-source bioreactor system that is adaptable to a wound-specific geometry and allows cell culture and differentiation. This interdisciplinary roadmap is intended to close the gap between the lab and clinic and to integrate novel 3D-printing technologies for regenerative medicine.

Human papillomavirus (HPV) is a common sexually transmitted infection that can cause both benign and malignant lesions. HPV vaccines, preferably administered before the onset of sexual activity, have demonstrated remarkable efficacy in preventing HPV-related cancers. The impact of a healthcare provider’s recommendation on HPV vaccine acceptance is substantial. Therefore, medical students must undergo thorough training in this domain. This study compares fundamental understanding and viewpoints regarding HPV and anti-HPV vaccines among Polish students pursuing medical and non-medical sciences. This study was based on the authors’ questionnaire, and the results were statistically analyzed. The participants in this study were 1025 students (medical sciences students—520 respondents in total; and non-medical sciences students—505 respondents in total). According to the results, medical students’ knowledge about the consequences of HPV infection and vaccination against HPV was significantly greater. To date, numerous publications have investigated the understanding of particular social, gender, parental, etc., groups about vaccination, but the knowledge of students at different universities—medical and other—has not been compared. Social awareness is still insufficient, even in groups of medical students. There is much to be done to educate and encourage preventive behavior in those not receiving primary prevention in early childhood.

Introduction: Cone Beam Computed Tomography (CBCT) scans should be digitally oriented using a reference plane before being evaluated for patient growth and treatment outcome. Current orientation planes are usually dependent on the Porion points. Using more reliable landmarks than the Porion, we introduce the Acta plane and investigate its reliability as an alternative reference plane for cases in which the Porion point is inaccessible. Methods: To test its reliability, a dental student and an orthodontist located sixteen skeletal and dental landmarks in sixteen CBCT scans before and after reorientation. Also, four measurements were calculated based on these landmarks’ coordinates. The reorientation of these CBCT scans with the Acta plane was performed by another dental student. The Intraclass Correlation Coefficient (ICC) was calculated before and after orientation, as well as the intra and inter-examiner. The data were tested for significance through the Shapiro–Wilk test with a threshold set at 0.05. Through the ICC variation of the points’ coordinates, this article also evaluated the reliability of this plane with the t-paired and exact-sign tests. Results: The identification of landmarks was more reliable in the reoriented CBCT images than in the original images, with a significant increase of 0.012 in the median of the ICC. Linear and angular measures showed no significant differences between groups. Conclusion: This plane is a reliable alternative for the orientation of CBCT images. The design of the Acta plane might allow the use of a smaller field of view and, consequently, a reduction in the radiation dose.

The quality of wastewater greatly affects the aquatic environment. Currently, a significant amount of emerging pollutants are entering wastewater in the form of pharmaceutical contaminants, industrial chemicals, pesticides, toxins, etc. However, conventional wastewater treatment processes at WWTPs are not effective enough for these emerging pollutants. Therefore, emerging pollutants represent a significant source of wastewater pollution and associated pollution of surface water and even drinking water. The main sources of pharmaceutical contaminants are analgesics and anti-inflammatory drugs, and of these, the most common in wastewater are non-steroidal anti-inflammatory drugs (NSAIDs). The aim is to provide a comprehensive review of the available information on NSAIDs in the aquatic environment, i.e., their occurrence, effects on the environment, formation of main metabolites, and methods of NSAID removal, with a focus on current trends and possible directions for future research.

With the increasing prevalence of monitored micro-pollutants in the environment, new challenges arise in their determination. Currently, the issue of microplastics in the environment is a highly debated topic. This article focuses on the assessment of micro-particles in drinking water samples collected in the Czech Republic. The samples for analysis were collected at two locations, twice a day. Particle separation was achieved through vacuum filtration, and particle identification was performed using the µ-FTIR method. A total of 40 particles were analyzed, and their shape, color, and material composition were determined. The study results demonstrated the presence of microplastics and additives used in plastic production in the drinking water samples. The findings of this article contribute to understanding the issue of micro-particles in drinking water and raise questions for further research and measures to protect public health and the environment.

Boil-off losses occur when gaseous hydrogen has to be released from a cryogenic tank due to liquid hydrogen evaporating. These are a substantial drawback for all areas in which liquid hydrogen is discussed as a potential fuel to limit the climate impact. Especially boil-off losses during fuelling are one of the most significant source of losses along the liquid hydrogen pathway. To analyse and minimize such losses, simulations of the filling process are performed with the simulation tool “EcoSimPro”. The simulations are validated with an analytical solution. The results show that boil-off losses can be significantly reduced by pre-cooling the cryogenic tank with liquid nitrogen. This method is most effective for relatively small tanks that could be used e.g., in small air crafts or air taxis.

As an indispensable part of the engine manufacturer supply chain, the eco-efficiency of altitude test facility (ATF) operations must improve. Automation is a key enabler in this context since it not only increases precision and reproducibility but also allows for reducing the test time and energy consumption. A suitable controller and reliable validation are crucial to ensure the stability and appropriate response of the control system. Both aspects necessitate a thorough understanding of the control plant, expressed in a numerical model. These models have to be suitable for control system design and validation while covering asymmetric flow phenomena that occur in the pipe system and detailing the nonlinear system dynamics to a high degree of accuracy. One-dimensional network models, state-space models, highly resolving numerical models, and data-driven models are relevant applications for this task. We compare the results of state-of-the-art one-dimensional network models which mainly imply symmetric flow conditions with those of three-dimensional Reynolds-averaged-Navier–Stokes (RANS) simulations which cover asymmetric flow phenomena. The findings show that the assumptions of idealized, axis-symmetric flow in the one-dimensional flow elements do not hold true for the complex flows in an altitude test facility. In a second step, we have compared the results of one-dimensional simulations with in-service measurements taken during ATF test campaigns. Deviations were observed, which become explainable based on the insights gained from the comparison with the RANS simulation. The findings reveal that the one-dimensional simulation-based approach is insufficient to adequately reflect the plant and subsequently for validation due to the observed asymmetric flow phenomena. To overcome this limitation, the application of an empirical first-order transfer function using system identification methods is proposed. Its applicability is successfully demonstrated for the exhaust gas section of the ATF. Subsequently, essential criteria for the design of a suitable control concept for the outlet condition are derived.

The transferability of structure–property relationships for laser-pretreated metal adhesive joints to laser-pretreated metal–carbon-fiber-reinforced plastic (CFRP) bonds was investigated. Single-lap shear tests were performed on hybrid AW 6082-T6–CFRP specimens pretreated with the same pulsed laser surface parameter sets on the metal surface as previously tested, AW 6082-T6–E320 metal adhesive joints. The fracture surfaces were characterized to determine the type of failure and elucidate differences and commonalities in the link between surface structures and single-lap shear strengths. Digital image analyses of the hybrid specimens’ fractured surfaces were used to quantify remaining CFRP fragments on the metallic joint side. The results indicate that high surface enlargements and the presence of undercut structures lead to single-lap shear strengths exceeding 40 MPa and 35 MPa for unaged and aged hybrid specimens, respectively. Whereas for the metal–polymer joints, the trend from high strength to weakly bonded specimens is largely continuous with the degree of surface structuring, hybrid metal–CFRP joints exhibit a drastic drop in joint performance after aging if the laser-generated surface structures are less pronounced with low surface enlargements and crater depths. Surface features and hydrothermal aging determine whether the specimens fail cohesively or adhesively.

This paper deals with the optimization of reactive powder concrete mixtures with respect to the addition of silica fume and the type of polycarboxylate superplasticizer used. First, the properties of reactive powder concrete with eight different commercial polycarboxylate superplasticizers were tested in terms of workability, specific weight, and mechanical properties. It was found that different commercially available superplasticizers had significant effects on the slump flow, specific weight, and compressive and flexural strengths. The optimal superplasticizer (BASF ACE430) was selected for further experiments in order to evaluate the influences of silica fume and superplasticizer content on the same material properties. The results showed that the silica fume and superplasticizer content had considerable effects on the mini-cone slump flow value, specific weight, flexural and compressive strengths, and microstructure. There were clearly visible trends and local minima and maxima of the measured properties. The optimal reactive powder concrete mixture had a composition of 3.5–4.0% superplasticizer and 15–25% silica fume.

Although posaconazole tablets show relatively low variability in pharmacokinetics (PK), the proportion of patients achieving the PK/PD target at the approved uniform dose for both prophylaxis and therapy is not satisfactory. The aim of this study was to develop a posaconazole population PK model in lung-transplant recipients and to propose a covariate-based dosing optimization for both prophylaxis and therapy. In this prospective study, 80 posaconazole concentrations obtained from 32 lung-transplant patients during therapeutic drug monitoring were analyzed using nonlinear mixed-effects modelling, and a Monte Carlo simulation was used to describe the theoretical distribution of posaconazole PK profiles at various dosing regimens. A one-compartment model with both linear absorption and elimination best fit the concentration–time data. The population apparent volume of distribution was 386.4 L, while an apparent clearance of 8.8 L/h decreased by 0.009 L/h with each year of the patient’s age. Based on the covariate model, a dosing regimen of 200 mg/day for prophylaxis in patients ˃60 years, 300 mg/day for prophylaxis in patients ˂60 years and for therapy in patients ˃60 years, and 400 mg/day for therapy in patients ˂60 years has been proposed. At this dosing regimen, the PK/PD target for prophylaxis and therapy is reached in 95% and 90% of population, respectively, representing significantly improved outcomes in comparison with the uniform dose.

Robots are defined as programmable machines that can perform specified tasks. Medical robots are emerging solutions in the field of cardiology leveraging recent technological innovations of control systems, sensors, actuators, and imaging modalities. Robotic platforms are successfully applied for percutaneous coronary intervention, invasive cardiac electrophysiology procedures as well as surgical operations including minimally invasive aortic and mitral valve repair, coronary artery bypass procedures, and structural heart diseases. Furthermore, machines are used as staff-assisting tools to support nurses with repetitive clinical duties i.e., food delivery. High precision and resolution allow for excellent maneuverability, enabling the performance of medical procedures in challenging anatomies that are difficult or impossible using conventional approaches. Moreover, robot-assisted techniques protect operators from occupational hazards, reducing exposure to ionizing radiation, and limiting risk of orthopedic injuries. Novel automatic systems provide advantages for patients, ensuring device stability with optimized utilization of fluoroscopy. The acceptance of robotic technology among healthcare providers as well as patients paves the way for widespread clinical application in the field of cardiovascular medicine. However, incorporation of robotic systems is associated with some disadvantages including high costs of installation and expensive disposable instrumentations, the need for large operating room space, and the necessity of dedicated training for operators due to the challenging learning curve of robotic-assisted interventional systems.

Gripper finger design is a complex process that requires a lot of experience, time, and effort. For this reason, automating this design process is an important area of research that has the potential to improve the efficiency and effectiveness of robotic systems. The current approaches are aimed at the automated design of monolithic gripper fingers, which have to be manufactured additively or by machining. This paper describes a novel approach for the automated design of gripper fingers. The motivation for this work stems from the increasing demand for flexible, adaptable handling systems in various industries in response to the increasing individualization of products as well as the increasing volatility in the markets. Based on the CAD data of the handling objects, the most suitable configuration of gripper fingers can be determined from the existing modules of a construction kit for the respective handling object, which can significantly reduce the provisioning time for new gripper fingers. It can be shown that gripper fingers can be effectively configured for a variety of objects and two different grippers, increasing flexibility in industrial handling processes.

ORCHESTRA (“Connecting European Cohorts to Increase Common and Effective Response To SARS-CoV-2 Pandemic”) is an EU-funded project which aims to help rapidly advance the knowledge related to the prevention of the SARS-CoV-2 infection and the management of COVID-19 and its long-term sequelae. Here, we describe the early results of this project, focusing on the strengths of multiple, international, historical and prospective cohort studies and highlighting those results which are of potential relevance for vaccination strategies, such as the necessity of a vaccine booster dose after a primary vaccination course in hematologic cancer patients and in solid organ transplant recipients to elicit a higher antibody titer, and the protective effect of vaccination on severe COVID-19 clinical manifestation and on the emergence of post-COVID-19 conditions. Valuable data regarding epidemiological variations, risk factors of SARS-CoV-2 infection and its sequelae, and vaccination efficacy in different subpopulations can support further defining public health vaccination policies.

Multi-material additive manufacturing (AM) attempts to utilize the full benefits of complex part production with a comprehensive and complementary material spectrum. In this context, this research article presents new processing strategies in the field of polymer- and metal-based multi-material AM. The investigation highlights the current progress in powder-based multi-material AM based on three successfully utilized technological approaches: additive and formative manufacturing of hybrid metal parts with locally adapted and tailored properties, material-efficient AM of multi-material polymer parts through electrophotography, and the implementation of UV-curable thermosets within the laser-based powder bed fusion of plastics. Owing to the complex requirements for the mechanical testing of multi-material parts with an emphasis on the transition area, this research targets an experimental shear testing set-up as a universal method for both metal- and polymer-based processes. The method was selected based on the common need of all technologies for the sufficient characterization of the bonding behavior between the individual materials.

Synergies of optical, thermal and laser scanning remotely sensed data provide valuable information to study the structure and functioning of terrestrial ecosystems. One of the few fully operational airborne multi-sensor platforms for ecosystem research in Europe is the Flying Laboratory of Imaging Systems (FLIS), operated by the Global Change Research Institute of the Czech Academy of Sciences. The system consists of three commercial imaging spectroradiometers. One spectroradiometer covers the visible and near-infrared, and the other covers the shortwave infrared part of the electromagnetic spectrum. These two provide full spectral data between 380–2450 nm, mainly for the assessment of biochemical properties of vegetation, soil and water. The third spectroradiometer covers the thermal long-wave infrared part of the electromagnetic spectrum and allows for mapping of surface emissivity and temperature properties. The fourth instrument onboard is the full waveform laser scanning system, which provides data on landscape orography and 3D structure. Here, we describe the FLIS design, data acquisition plan and primary data pre-processing. The synchronous acquisition of multiple data sources provides a complex analytical and data framework for the assessment of vegetation ecosystems (such as plant species composition, plant functional traits, biomass and carbon stocks), as well as for studying the role of greenery or blue-green infrastructure on the thermal behaviour of urban systems. In addition, the FLIS airborne infrastructure supports calibration and validation activities for existing and upcoming satellite missions (e.g., FLEX, PRISMA).

13C discrimination (Δ13C) has been used in research as an indicator of water availability in crops; however, few data are available concerning fruit trees. The aim of this study was to examine the effect of irrigation on the Δ13C values of apple leaves. We assumed that Δ13C would increase with irrigation intensity. The Δ13C of apple trees (Malus domestica) cv. ‘Red Jonaprince’ was determined in the years 2019–2022. Leaf samples were collected in spring, in June, at the beginning of the irrigation campaign, and in autumn, in September, following the harvest. The irrigation doses were applied to replenish the water consumption, 0% (ET0), 50% (ET50), 75% (ET75), and 100% (ET100), of the calculated evapotranspiration (ET) levels. In November, the leaves collected from different positions on the shoots were sampled, assuming the Δ13C signature would reflect the changes occurring in the water supply during their growth. The irrigation rates had a significant effect on the Δ13C of the leaves when the data for the spring and summer months were pooled. On average, Δ13C increased from 20.77‰ and 20.73‰ for ET0 and ET50, respectively, to 20.80‰ and 20.95‰ for ET75 and ET100, respectively. When the data obtained for the spring and summer months were analysed separately, the effect of irrigation was weak (p < 0.043). The Δ13C value was always higher for treatment ET100 than treatment ET0, for individual experimental years and terms; however, the differences were minor and mostly insignificant. The leaf position had a strong significant effect on Δ13C; the values gradually decreased from the leaves growing from two-years-old branches (22.50‰) to the youngest leaves growing at the top of the current year’s shoots (21.07‰). This order was similar for all the experimental years. The results of the experiment suggest that 13C discrimination in apples is affected not only by water availability during growth, but also by the use of C absorbed in previous years.

In this work, an innovative manufacturing approach that includes a fully linked and integrated manufacturing system consisting of a laser-based directed energy deposition (DED-LB/M) module and a forming press is presented. The alternating additive manufacturing (AM) process is based on a combination of a DED-LB/M process using a laser power of 600 W and a feed rate of 400 mm/min and a subsequent forming process, in which the structure is upset with a hydraulic press using a constant forming force of 500 kN in order to smooth the surface and influence the accuracy of the components. For the generation of a fundamental process understanding, a cuboid, basic shape was chosen as geometry for the investigations. The aim is to improve part properties by applying the process steps to generate part properties, which are superior to solely additive manufactured material. It is shown that the geometry of additive manufactured structures can be adapted, and the top surface can be smoothed due to the forming operation. The mean roughness value Rz decreases up to 50% after the forming operation. The hardness can be increased by work hardening. Of special interest is that the higher hardness can be kept up even though a further DED-LB/M process step and forming operation are applied to the additively manufactured and formed structure again. Finally, an analysis of the new manufacturing approach regarding its potential is given.

The presence of microplastics in environmental compartments is generally recognized as a (potential) health risk. Many papers have been published on the abundance of microplastics at various locations around the globe, but only limited knowledge is available on possible mitigation routes. One of the mitigation routes is based on the choice of plastic materials used for products that may unintentionally end up in the environment. As a first approach, this paper presents a method to calculate the tendency of polymers to form microplastics, based on their mechanical and physical properties. A MicroPlastic Index (MPI) that correlates the microplastic formation to polymer properties is defined for both impact and wear of polymers via a theoretical particle size and the energy required to form these particles. A first comparison between calculated and experimental particle size is included. The MPI for impact and wear follow the same trend. Finally, these MPIs are correlated to the respective abundance of the microplastics in the environment, corrected for global production of the corresponding polymers: the higher the MPI, the more microplastics are found in the environment. Thus, the MPI can be used as a basis for choice or redesign of polymers to reduce microplastic formation.

Ultrashort pulse infrared lasers can simultaneously sample and homogenize biological tissue using desorption by impulsive vibrational excitation (DIVE). With growing attention on alterations in lipid metabolism in malignant disease, mass spectrometry (MS)-based lipidomic analysis has become an emerging topic in cancer research. In this pilot study, we investigated the feasibility of tissue sampling with a nanosecond infrared laser (NIRL) for the subsequent lipidomic analysis of oropharyngeal tissues, and its potential to discriminate oropharyngeal squamous cell carcinoma (OPSCC) from non-tumorous oropharyngeal tissue. Eleven fresh frozen oropharyngeal tissue samples were ablated. The produced aerosols were collected by a glass fiber filter, and the lipidomes were analyzed with mass spectrometry. Data was evaluated by principal component analysis and Welch’s t-tests. Lipid profiles comprised 13 lipid classes and up to 755 lipid species. We found significant inter- and intrapatient alterations in lipid profiles for tumor and non-tumor samples (p-value < 0.05, two-fold difference). Thus, NIRL tissue sampling with consecutive MS lipidomic analysis is a feasible and promising approach for the differentiation of OPSCC and non-tumorous oropharyngeal tissue and may provide new insights into lipid composition alterations in OPSCC.

Apart from increased fluid intake, patients with kidney stone disease (KSD) due to renal phosphate wasting require specific metaphylaxis. NaPi2a, NaPi2c, and NHERF1 regulate plasma phosphate concentration by reabsorbing phosphate in proximal kidney tubules and have been found altered in monogenic hypophosphatemia with a risk of KSD. In this study, we aimed at assessing the combined genetic alterations impacting NaPi2a, NaPi2c, and NHERF1. Therefore, we screened our hereditary KSD registry for cases of oligo- and digenicity, conducted reverse phenotyping, and undertook functional studies. As a result, we identified three patients from two families with digenic alterations in NaPi2a, NaPi2c, and NHERF1. In family 1, the index patient, who presented with severe renal calcifications and a bone mineralization disorder, carried digenic alterations affecting both NaPi transporter 2a and 2c. Functional analysis confirmed an additive genetic effect. In family 2, the index patient presented with kidney function decline, distinct musculature-related symptoms, and intracellular ATP depletion. Genetically, this individual was found to harbor variants in both NaPi2c and NHERF1 pointing towards genetic interaction. In summary, digenicity and gene dosage are likely to impact the severity of renal phosphate wasting and should be taken into account in terms of metaphylaxis through phosphate substitution.

Terrestrial laser scanners (TLSs) are a standard method for 3D point cloud acquisition due to their high data rates and resolutions. In certain applications, such as deformation analysis, modelling uncertainties in the 3D point cloud is crucial. This study models the systematic deviations in laser scan distance measurements as a function of various influencing factors using machine-learning methods. A reference point cloud is recorded using a laser tracker (Leica AT 960) and a handheld scanner (Leica LAS-XL) to investigate the uncertainties of the Z+F Imager 5016 in laboratory conditions. From 49 TLS scans, a wide range of data are obtained, covering various influencing factors. The processes of data preparation, feature engineering, validation, regression, prediction, and result analysis are presented. The results of traditional machine-learning methods (multiple linear and nonlinear regression) are compared with eXtreme gradient boosted trees (XGBoost). Thereby, it is demonstrated that it is possible to model the systemic deviations of the distance measurement with a coefficient of determination of 0.73, making it possible to calibrate the distance measurement to improve the laser scan measurement. An independent TLS scan is used to demonstrate the calibration results.

Freshwater fish production is significantly correlated with water temperature, which is expected to increase under climate change. This study evaluated changes in water temperature and their impact on productive ponds at fisheries in the Czech Republic. A model was developed to calculate surface water temperature based on the five-day mean of the air temperature and was then tested in several ponds in three major Czech fish production areas. The output of the surface water temperature model was compared with independently measured data (r = 0.79–0.96), and the verified model was then applied to predict climate change conditions. The results were evaluated with regard to the thresholds characterizing the water temperature requirements of fish species and indicated that the limitation of Czech fish farming results from (i) an increased number of continuous periods during which given fish species are threatened by high water temperatures and (ii) the extension of continuous periods with stressful water temperatures. For Czech fisheries, the model suggests a sharp increase in unprecedented temperature regimes, which will pose critical challenges to traditional forms of common carp farming within several decades. Although reducing the level of eutrophication and loading them with organic substances might alleviate expected threads, farming current fish species in deeper and colder ponds at higher elevations might be inevitable.

The present study investigated whether an unhealthy diet and other lifestyle behaviors may modify the genetic susceptibility to impulsivity. A total of 33,047 participants (mean age = 42.1 years, 59.8% females) from the Dutch Lifelines cohort were included. Each diet index and other lifestyle behaviors were tested for their interactions on the effect on the attention-deficit/hyperactivity disorder (ADHD) polygenic risk score (PRS) on impulsivity using a linear regression model with adjustment for covariates. The ADHD PRS was significantly associated with impulsivity (B = 0.03 (95% CI: 0.02, 0.04); p = 2.61 × 10−9). A poorer diet, a higher intake of energy, and a higher intake of fat were all associated with higher impulsivity, and a high intake of energy amplified the effect of ADHD PRS on impulsivity (e.g., for the interaction term of ADHD PRS and highest tertile on intake of energy, B = 0.038 (95% CI: 0.014, 0.062); p = 0.002. The other lifestyle factors, namely short and long sleep duration, current and past smoking, higher alcohol intake, and more time spent on moderate-to-vigorous physical activity were associated with higher impulsivity, but no interaction effect was observed. In conclusion, we found that a high intake of energy exacerbated the genetic susceptibility to impulsivity. Our study helps to improve our understanding of the role of diet and genetic factors on impulsivity.

In a three-year experiment (2019–2021), the roots of 7-year-old apple trees (Malus domestica cv. ‘Red Jonaprince’) grown under drip irrigation were studied. The aim of the study was to determine the effect of irrigation on root density at different depths and distances from the trunk. The working hypothesis assumed that irrigation significantly affects the total length of apple roots. The irrigation treatments corresponding to the calculated water evapotranspiration (ET100), 50% of the calculated ET (ET50), a control (ET0, no irrigation, under rainfed conditions), and a treatment using double-drip lines (2Drops) were monitored. Soil cores were collected in spring and autumn. The total length of the roots (TRLt) and the length of new vital roots (TRLv) to a depth of 80 cm were evaluated. The effects of treatments were mostly insignificant for the TRLt; only in the dry season in 2019 were the TRLt values of the irrigated treatments (ET50 and ET100) significantly higher, 18.67 km·m−2 and 17.45 km·m−2, in comparison to 11.16 km·m−2 for the ET0, at a 10 cm distance from the tree trunk. The irrigation treatments had a statistically significant effect on the TRLv values near the trunk in 2019 and 2020, while in autumn 2020 and 2021, irrigation significantly affected the TRLv at greater distances from the tree trunk. In summary, the irrigation treatments mostly had no significant effect on the total root length. However, an effect of irrigation on the root length of new vital roots was observed at certain sampling dates and distances from the trunk.

Vitamin D (VitD) and its receptor (VDR) have been intensively investigated in many cancers. As knowledge for head and neck cancer (HNC) is limited, we investigated the (pre)clinical and therapeutic relevance of the VDR/VitD-axis. We found that VDR was differentially expressed in HNC tumors, correlating to the patients’ clinical parameters. Poorly differentiated tumors showed high VDR and Ki67 expression, whereas the VDR and Ki67 levels decreased from moderate to well-differentiated tumors. The VitD serum levels were lowest in patients with poorly differentiated cancers (4.1 ± 0.5 ng/mL), increasing from moderate (7.3 ± 4.3 ng/mL) to well-differentiated (13.2 ± 3.4 ng/mL) tumors. Notably, females showed higher VitD insufficiency compared to males, correlating with poor differentiation of the tumor. To mechanistically uncover VDR/VitD’s pathophysiological relevance, we demonstrated that VitD induced VDR nuclear-translocation (VitD < 100 nM) in HNC cells. RNA sequencing and heat map analysis showed that various nuclear receptors were differentially expressed in cisplatin-resistant versus sensitive HNC cells including VDR and the VDR interaction partner retinoic acid receptor (RXR). However, RXR expression was not significantly correlated with the clinical parameters, and cotreatment with its ligand, retinoic acid, did not enhance the killing by cisplatin. Moreover, the Chou–Talalay algorithm uncovered that VitD/cisplatin combinations synergistically killed tumor cells (VitD < 100 nM) and also inhibited the PI3K/Akt/mTOR pathway. Importantly, these findings were confirmed in 3D-tumor-spheroid models mimicking the patients’ tumor microarchitecture. Here, VitD already affected the 3D-tumor-spheroid formation, which was not seen in the 2D-cultures. We conclude that novel VDR/VitD-targeted drug combinations and nuclear receptors should also be intensely explored for HNC. Gender-specific VDR/VitD-effects may be correlated to socioeconomic differences and need to be considered during VitD (supplementation)-therapies.

Drug-eluting stents (DES) demonstrated superior efficacy when compared to bare metal stents and plain-old balloon angioplasty and are nowadays used in almost all percutaneous revascularization procedures. The design of the stent platforms is constantly improving to maximize its efficacy and safety. Constant development of DES includes adoption of new materials used for scaffold production, new design types, improved overexpansion abilities, new polymers coating and, finally, improved antiproliferative agents. Especially nowadays, with the immense number of available DES platforms, it is crucial to understand how different aspects of stents impact the effect of their implantation, as subtle differences between various stent platforms could impact the most important issue—clinical outcomes. This review discusses the current status of coronary stents and the impact of stent material, strut design and coating techniques on cardiovascular outcomes.

Therapy for pancreatic ductal adenocarcinoma remains challenging, and the chances of a complete cure are very limited. As in other types of cancer, the expression and role of miRNAs in controlling the biological properties of this type of tumor have been extensively studied. A better insight into miRNA biology seems critical to refining diagnostics and improving their therapeutic potential. In this study, we focused on the expression of miR-21, -96, -196a, -210, and -217 in normal fibroblasts, cancer-associated fibroblasts prepared from a ductal adenocarcinoma of the pancreas, and pancreatic carcinoma cell lines. We compared these data with miRNAs in homogenates of paraffin-embedded sections from normal pancreatic tissues. In cancer-associated fibroblasts and cancer cell lines, miRNAs differed significantly from the normal tissue. In detail, miR-21 and -210 were significantly upregulated, while miR-217 was downregulated. Similar transcription profiles were earlier reported in cancer-associated fibroblasts exposed to hypoxia. However, the cells in our study were cultured under normoxic conditions. We also noted a relation to IL-6 production. In conclusion, cultured cancer-associated fibroblasts and carcinoma cells reflect miR-21 and -210 expression similarly to the cancer tissue samples harvested from the patients.

Over the past 10 years, ground-based radar interferometry has become a frequently used technology for determining dynamic deflections of bridge structures induced by vehicle passages. When measuring with only one radar device, the so-called Interpretation Error (EI) considerably rises. When using two radars, it is possible to simultaneously determine, for example, vertical and longitudinal displacements and to eliminate the Interpretation Error. The aim of the article is to establish a suitable strategy for determining dynamic and quasi-static response of bridge structures based on the accuracy analysis of measurement by two radars. The necessary theory for displacements determination by means of two radar devices is presented. This is followed by an analysis of errors when measuring with only one radar. For the first time in the literature, mathematical formulas are derived here for determining the accuracy of the resulting displacements by simultaneous measurement with two radars. The practical examples of bridge structures displacements determination by measuring with two radar devices in the field are presented. The key contribution of the paper is the possibility to estimate and plan in advance the achievable accuracy of the resulting displacements for the given radar configurations in relation to the bridge structure.
