To investigate the reinforcement–soil interfacial effects in fiber-reinforced soil, this study developed a novel horizontal pullout tester and conducted pullout tests on coarse polypropylene fibers in plain soil, cemented soil, and fine fiber-reinforced cemented soil. Three soil types were analyzed: low liquid limit clay, high liquid limit clay, and clay sand. The pullout tester proved to be both scientifically robust and efficient. Depending on the soil properties, coarse polypropylene fibers were pulled out intact or fractured. The pullout curves displayed distinct multi-peak patterns, with wavelengths closely linked to the fiber’s intrinsic characteristics. The pullout curve wavelength for plain soil matched the fiber’s intrinsic wavelength, while it was slightly greater in cemented soils. The peak pullout force increased with extended curing periods, higher cement content, more excellent compaction, and the addition of fine polypropylene fibers. Among these factors, compaction had the most significant impact on enhancing the soil–fiber interfacial effect. Friction, cohesion, and fiber interweaving created interlocking effects, inhibiting fiber sliding. Cement hydration processes further deformed the fiber, increasing its friction coefficient and sliding resistance. Hydration products also fill soil voids, improving soil compactness, enlarging the fiber–soil contact area, and enhancing frictional and occlusal forces at the interface.

Receptor-based biosensors often suffer from slow analyte diffusion, leading to extended assay times. Moreover, existing methods to enhance diffusion can be complex and costly. In response to this challenge, we presented a rapid and cost-effective technique for fabricating concave magnetic-responsive hydrogel discs (CMDs) by straightforward pipetting directly onto microscope glass slides. This approach enables immediate preparation and customization of hydrogel properties such as porosity, magnetic responsiveness, and embedded particles and is adaptable for use with microarray printers. The concave design increased the surface area by 43% compared to conventional hemispherical hydrogels, enhancing diffusion rates and accelerating reactions. By incorporating superparamagnetic particles, the hydrogels become magnetically responsive, allowing for stirring within reagent droplets using magnets to improve mixing. Our experimental results showed that CMDs dissolved approximately 2.5 times faster than hemispherical ones. Numerical simulations demonstrated up to a 46% improvement in diffusion speed within the hydrogel. Particles with lower diffusion coefficients, like human antibodies, benefited most from the concave design, resulting in faster biosensor responses. The increased surface area and ease of fabrication make our CMDs efficient and adaptable for various biological and biomedical applications, particularly in point-of-care diagnostics where rapid and accurate biomarker detection is critical.

Agroforestry is being promoted as a feasible land use management to improve understory economic benefits. However, there are few studies on species selection and the comprehensive evaluation of soil quality change in rhizoma bletillae (Bletilla striata) agroforestry systems. The soil quality index (SQI) and minimum dataset (MDS) methods can reflect the overall condition and were effective tools for understanding different cultivation systems. In this study, we evaluated the soil quality of four cultivation models (including three agroforestry systems: PeB, moso bamboo (Phyllostachys edulis)–rhizoma bletillae; PoB, plane trees (Platanus orientali)–rhizoma bletillae; CcB, pecan trees (Carya cathayensis)–rhizoma bletillae; and CK, rhizoma bletillae monoculture. The total dataset (TDS) consisted of 15 soil parameters containing physical, chemical, and biological characteristics. The results showed that soil organic carbon (SOC), total nitrogen (TN), and total phosphorus (TP) were finally selected and established as the MDS. Agroforestry could significantly influence soil quality. Compared with CK, the SQI in CcB significantly increased and decreased in PeB and PoB. Soil water content (SWC), nitrate nitrogen (NO3−-N), dissolved organic carbon (DOC), SOC, TN, and TP contents were higher in CcB than in the other cultivation models. Based on various soil indicators and SQI analysis, the CcB was the best in improving soil quality. These findings showed that the soil quality index based on the MDS can be used as an effective indicator for agroforestry systems selection. It provides theoretical guidance for the practice of bionic cultivation and the sustainable management of rhizoma bletillae.

Glycerol-3-phosphate acyltransferase (GPAT), as a rate-limiting enzyme engaged in lipid synthesis pathways, exerts an important role in plant growth and development as well as environmental adaptation throughout diverse growth stages. Alfalfa (Medicago sativa L.) is one of the most significant leguminous forages globally; however, its growth process is frequently exposed to environmental stress, giving rise to issues such as impeded growth and decreased yield. At present, the comprehension of the GPAT genes in alfalfa and their reactions to abiotic stresses is conspicuously deficient. This study identified 15 GPATs from the genome of “Zhongmu No. 1” alfalfa, which were phylogenetically categorized into three major groups (Groups I ~ III). Furthermore, Group III is further subdivided into three distinct subgroups. MsGPATs belonging to the same subfamily exhibited similar protein conserved motifs and gene structural characteristics, in which groups with simple conserved motifs had more complex gene structures. A multitude of regulatory cis-elements pertinent to hormones and responses to environmental stress were detected in their promoter regions. In addition, a spatial–temporal expression analysis showed that MsGPATs have significant tissue specificity. Furthermore, the transcriptomic analysis of ABA treatment and the qRT-PCR results under drought, salt, and cold stress demonstrated that the majority of MsGPATs respond to abiotic stress with pronounced timely characteristics. It was also ascertained that these GPAT genes might assume a crucial role in salt and drought stress. This research can further constitute a fundamental basis for the exploration of the alfalfa GPAT family, the screening of key GPATs, and the investigation of their functionalities.

Umami peptides have the ability to enhance food flavours and have potential health benefits. The objective of this study was to conduct a comprehensive investigation into the umami intensity, taste mechanism, and antioxidant activity of six umami peptides derived from wheat gluten hydrolysates (WGHs) and fermented WGHs. The e-tongue analysis demonstrated that the peptides exhibited a direct proportionality in terms of umami value and concentration, and were capable of enhancing the umami of commercially available condiments. The molecular dynamics simulations demonstrated that the peptides interacted with T1R1/T1R3 receptors via hydrogen bonds, hydrophobic interactions, ionic interactions, and water bridges, thereby producing umami. Furthermore, the DPPH, ABTS, hydroxyl radical-scavenging, and FRAP assays demonstrated that the six peptides exhibited antioxidant activity in vitro. Ultimately, the network pharmacology and molecular docking results indicated that AKT1, JUN, and CASP3 may serve as the core targets for the peptides in the treatment of oxidative diseases. In conclusion, this work offers novel insights into the use of bioactive umami peptides, emphasising their prospective applications in the food and health supplement industries.

Atractylodes macrocephala, a perennial herb widely used in traditional Chinese medicine, is highly prone to root rot, which significantly reduces its yield and quality. This study compared the physicochemical properties of soil from healthy and diseased A. macrocephala plants and analyzed the microbial diversity in the endophytic, rhizosphere, and root zone soils. The results showed that the diseased plants had higher levels of available potassium and electrical conductivity in the rhizosphere, both positively correlated with the severity of root rot, while soil pH was negatively correlated. The diversity and richness of endophytic bacterial and fungal communities were significantly reduced in diseased plants. Additionally, root rot led to major changes in the rhizosphere microbial community, with an increased abundance of Proteobacteria and Ascomycota, and a decrease in Firmicutes, Bacteroidetes, Actinobacteria, and Basidiomycota. Fusarium oxysporum, Fusarium solani, and Fusarium fujikuroi were identified as key pathogens associated with root rot. This study enhances our understanding of the microbial interactions in soils affected by root rot, offering a foundation for developing soil improvement and biological control strategies to mitigate this disease in A. macrocephala cultivation.

Alfin-like proteins (ALs) form a plant-specific transcription factor (TF) gene family involved in the regulation of plant growth and development, and abiotic stress response. In this study, 30 ALs were identified in Brassica napus ecotype ‘Zhongshuang 11’ genome (BnaALs), and unevenly distributed on 15 chromosomes. Structural characteristic analysis showed that all of the BnaALs contained two highly conserved domains: the N terminal DUF3594 domain and the C-terminal PHD-finger domain. The BnaALs were classified into four groups (Group I-IV), supported by conserved intron–exon and protein motif structures in each group. The allopolyploid event between B. oleracea and B. rapa ancestors and the small-scale duplication events in B. napus both contributed to the large BnaALs expansion. The promoter regions of BnaALs contained multiple abiotic stress cis-elements. The BnaALs in I-IV groups were mainly expressed in cotyledon, petal, root, silique, and seed tissues, and the duplicated gene pairs shared highly similar expression patterns. RNA-seq and RT-qPCR analysis showed that BnaALs were obviously induced by low nitrogen (LN) and low phosphorus (LP) treatments in roots. Overexpressing BnaAL02 and BnaAL28 in Arabidopsis demonstrated their functions in response to LN and LP stresses. BnaAL28 enhanced primary roots’ (PRs) length and lateral roots’ (LRs) number under LP and LN conditions, where BnaAL02 can inhibit LR numbers under the two conditions. They can promote root hair (RH) elongation under LP conditions; however, they suppressed RH elongation under LN conditions. Our result provides new insight into the functional dissection of this family in response to nutrient stresses in plants.

Fasting and refeeding systems can cause significant short-term fluctuations in nutrient and energy levels, triggering adaptive physiological responses in animals. This study examines the effects of fasting and refeeding on blood biochemical indicators and transcriptional profiles in the hypothalamus and subcutaneous adipose tissue of geese. Biochemical assays reveal that fasting significantly increases levels of free fatty acids and glucagon, while reducing concentrations of triglycerides, leptin, and insulin. Transcriptomic analyses identify a complex transcriptional response in both the hypothalamus and subcutaneous adipose tissue, affecting several metabolic pathways and key genes associated with feed intake and energy metabolism. In subcutaneous adipose tissue, fasting downregulates genes involved in fatty acid synthesis (LPL, SCD, and ACSL1) and upregulates PLIN2, a gene promoting lipid droplet degradation. Fasting affects a variety of metabolic pathways and critical genes in the hypothalamus, including Apelin, insulin, and mTOR signaling pathways. After fasting, the mRNA expression of NOG, GABRD, and IGFBP-1 genes in the hypothalamus are significantly upregulated, while proopiomelanocortin (POMC) gene expression is markedly downregulated. This study highlights the intricate biological responses to nutritional changes in geese, which adds to our understanding of energy balance and metabolic regulation in avian species.

Dendrobium sinense, an endemic medicinal herb in Hainan Island, is rich in bibenzyl compounds. However, few studies have explored the molecular mechanisms of bibenzyl biosynthesis. This study presents a comprehensive analysis of DsBBS1 and DsBBS2 function in D. sinense. A molecular docking simulation revealed high-resolution three-dimensional structural models with minor domain orientation differences. Expression analyses of DsBBS1 and DsBBS2 across various tissues indicated a consistent pattern, with the highest expression being found in the roots, implying that they play a pivotal role in bibenzyl biosynthesis. Protein expression studies identified optimal conditions for DsBBS2-HisTag expression and purification, resulting in a soluble protein with a molecular weight of approximately 45 kDa. Enzyme activity assays confirmed DsBBS2’s capacity to synthesize resveratrol, exhibiting higher Vmax and lower Km values than DsBBS1. Functional analyses in transgenic Arabidopsis demonstrated that both DsBBS1 and DsBBS2 could complement the Atchs mutant phenotype. The total flavonoid content in the DsBBS1 and DsBBS2 transgenic lines was restored to wild-type levels, while the total bibenzyl content increased. DsBBS1 and DsBBS2 are capable of catalyzing both bibenzyl and flavonoid biosynthesis in Arabidopsis. This study provides valuable insights into the molecular mechanisms underlying the biosynthesis of bibenzyl compounds in D. sinense.

Sacha inchi (Plukenetia volubilis) is a valuable oilseed crop with a high content of polyunsaturated fatty acids (PUFAs). However, there is a lack of in-depth understanding of the lipidomics in Sacha inchi seeds (SIDs). Saturated fatty acids occupied more than half of the proportion (59.31%) in early development, while PUFAs accounted for 78.92% at maturation. The main triacylglycerols were TAG(18:3/18:3/18:3), TAG(18:2/18:2/18:3), and TAG(16:0/18:2/18:2). The corresponding species (18:3/18:3, 18:2/18:2, and 16:0/18:2) were also the main ingredients in diacylglycerol and phosphatidic acid, indicating high PUFA composition in the sn-1 and sn-2 positions of TAG. Only LPC(18:3), LPC(18:2), and LPC(16:0) were identified in SIDs, implying that those PUFAs on the sn-2 positions of the PC(18:3/-), PC(18:2/-), and PC(16:0/-) categories were released into the acyl-CoA pool for the Kennedy pathway. Conversely, the PC(18:1/-) and PC(18:0/-) categories might be responsible for the generation of PC-derived DAG and TAG. The lipidomics data will contribute to understanding the TAG assembly in developing SIDs, especially for PUFAs.

B-box (BBX) proteins, a subfamily of zinc-finger transcription factors, are involved in various environmental signaling pathways. In this study, we conducted a comprehensive analysis of BBX family members in Brassica crops. The 482 BBX proteins were divided into five groups based on gene structure, conserved domains, and phylogenetic analysis. An analysis of nonsynonymous substitutions and (Ka)/synonymous substitutions (Ks) revealed that most BBX genes have undergone purifying selection during evolution. An analysis of transcriptome data from rapeseed (Brassica napus) organs suggested that BnaBBX3d might be involved in the development of floral tissue-specific RNA-seq expression. We identified numerous light-responsive elements in the promoter regions of BnaBBX genes, which were suggestive of participation in light signaling pathways. Transcriptomic analysis under shade treatment revealed 77 BnaBBX genes with significant changes in expression before and after shading treatment. Of these, BnaBBX22e showed distinct expression patterns in yellow- vs. black-seeded materials in response to shading. UPLC-HESI-MS/MS analysis revealed that shading influences the accumulation of 54 metabolites, with light response BnaBBX22f expression correlating with the accumulation of the flavonoid metabolites M46 and M51. Additionally, BnaBBX22e and BnaBBX22f interact with BnaA10.HY5. These results suggest that BnaBBXs might function in light-induced pigment accumulation. Overall, our findings elucidate the characteristics of BBX proteins in six Brassica species and reveal a possible connection between light and seed coat color, laying the foundation for further exploring the roles of BnaBBX genes in seed development.

Wheat stem rust, caused by Puccinia graminis f. sp. tritici (Pgt), is a devastating fungal disease that affects wheat globally. The planting of resistant cultivars is the most cost-effective strategy for controlling this disease. The Huanghuai region, as a major wheat-growing area, plays a crucial role in the spread and prevalence of wheat stem rust in China. In this study, 64 wheat accessions from this region were tested at the adult stage against two major Pgt races, 34MKGQM and 21C3CTHQM. DNA markers associated with the known resistance genes Sr31, Sr24, Sr25, Sr26, and Sr38 were measured to determine their presence in the tested accessions. In the 2023 field tests, 5 (7.8%) accessions were immune to 21C3CTHQM and 34MKGQM, while 35 (54.7%) and 39 (60.9%) were moderately resistant and resistant, respectively. The remaining 20 (30.7%) accessions were moderately susceptible and susceptible. In the 2024 tests, 12 (18.8%) and 14 (21.9%) entries were immune to both races; 29 (45.3%) and 30 (46.9%) were moderately resistant and resistant, respectively. Only two cultivars, Xinong 816 and Yimai 211, were immune in both years, and three entries showed some degrees of resistance in both years. Seven cultivars, including Zhongzhimai 23, Longxing 1, Yunong 937, Huaguan 301, Wanke 800, Shaanhe 285, and Yunong 612, showed increased susceptibility. DNA markers showed that 30 entries carried Sr31, while 6 entries carried Sr38. Genes Sr24, Sr25, and Sr26, which confer good resistance to the globally prevalent cultivars TKTTF and TTTRF, were absent from the set of tested entries. While this study surveyed the resistance levels of a cross-section of wheat from the southern part of the Huanghuai region and confirmed the presence of two known resistance genes, the basis of immunity or high levels of resistance in several lines remains obscure.

Reservoir impoundment imposes a significant triggering effect on bank landslides. Studying the early identification of landslides and their stability concerning reservoir water levels and rainfall is vital for guaranteeing the safety of residents and infrastructure in reservoir regions. This study proposed a method for establishing a dynamic inventory of active landslides at large hydropower stations using integrated remote sensing techniques, demonstrated at Lianghekou Reservoir. We employed interferometric stacking synthetic aperture radar (stacking-InSAR) technology, small baseline subset interferometric synthetic aperture radar (SBAS-InSAR) technology, and optical satellite images to identify and catalogue active landslides. Moreover, we conducted field investigations to examine the deformation characteristics of landslides. Finally, Pearson’s correlation analysis was employed to evaluate the response between deformation values, reservoir water levels, and rainfall. The results revealed 75 active landslides, including 12 long-term active landslides before impoundment and 63 new landslides after impoundment, which were primarily concentrated in the Waduo and Yazho–Zatou regions. The correlation coefficient between landslide deformation values and the reservoir level was high (0.93), while the correlation coefficient with rainfall was low (0.57). The results of this research offer a crucial foundation for preventing and mitigating landslides in reservoir areas.

Blood flow measurement of microvessels in human tissues is of vital importance for the diagnosis and treatment of many diseases. In this paper, the detection method of abnormal blood flow regions based on near-infrared correlation spectroscopy is studied. We used the NL-Bregman-TV imaging algorithm to realize Blood flow imaging. However, due to the limitation of the number and distribution of detectors, the pixels obtained from images are extremely low, which cannot meet the practical requirements of the visual and the abnormal blood flow range measurement. In this paper, the bicubic interpolation method is used to improve the resolution of low-pixel blood flow images. The parameter index of the normalized similarity was proposed to help judge the effect of the interpolation method on the resolution of this kind of image. Aiming at the extraction of abnormal regions, a threshold segmentation algorithm based on the histogram difference method and a morphological processing algorithm is proposed to extract the contour of abnormal blood flow. The method proposed in this paper can be used to accurately locate and extract the clear and smooth contour of abnormal blood flow.

The current deformation and stable state of slopes with historical shatter signs is a concern for engineering construction. Suspected landslide scarps were discovered at the rear edge of the Genie slope on the Tibetan Plateau during a field investigation. To qualitatively determine the current status of the surface deformation of this slope, this study used high-resolution optical remote sensing, airborne light detection and ranging (LiDAR), and interferometric synthetic aperture radar (InSAR) technologies for comprehensive analysis. The interpretation of high-resolution optical and airborne LiDAR data revealed that the rear edge of the slope exhibits three levels of scarps. However, no deformation was detected with differential InSAR (D-InSAR) analysis of ALOS-1 radar images from 2007 to 2008 or with Stacking-InSAR and small baseline subset InSAR (SBAS-InSAR) processing of Sentinel-1A radar images from 2017 to 2020. This study verified the credibility of the InSAR results using the standard deviation of the phase residuals, as well as in-borehole displacement monitoring data. A conceptual model of the slope was developed by combining field investigation, borehole coring, and horizontal exploratory tunnel data, and the results indicated that the slope is composed of steep anti-dip layered dolomite limestone and that the scarps at the trailing edges of the slope were caused by historical shallow toppling. Unlike previous remote sensing studies of deformed landslides, this paper argues that remote sensing results with reliable accuracy are also applicable to the study of undeformed slopes and can help make preliminary judgments about the stability of unexplored slopes. The study demonstrates that the long-term consistency of InSAR results in integrated remote sensing can serve as an indicator for assessing slope stability.

Hyperuricemia, i.e., increased plasma uric acid concentration, is a common problem in clinical practice, leading to gout or nephrolithiasis, and is associated with other disorders, such as metabolic syndrome, cardiovascular disease, and chronic renal disease. Xanthine oxidoreductase (XOR) is a critical rate-limiting enzyme involved in uric acid synthesis and a promising target for hyperuricemia therapy. However, XOR inhibitors currently face clinical problems such as a short half-life and side effects. Here, we found that specifically targeting liver Xor with GalNAc-siRNAs had a good therapeutic effect on hyperuricemia. First, siRNAs were designed to target various sites in the homologous region between Homo sapiens and Mus musculus Xor mRNA and were screened in primary mouse hepatocytes. Then, the siRNAs were modified to increase their stability in vivo and conjugated with GalNAc for liver-specific delivery. The effects of GalNAc-siRNAs were evaluated in three hyperuricemia mouse models, including potassium oxonate and hypoxanthine administration in WT and humanized XDH mice and Uox knockout mice. Febuxostat, a specific XOR inhibitor used for hyperuricemia treatment, was used as a positive control. Targeting liver Xor with GalNAc-siRNAs by subcutaneous administration reduced plasma uric acid levels, uric acid accumulation in the kidney, renal inflammation, and fibrosis, thereby alleviating kidney damage in hyperuricemia mouse models without hepatoxicity. The results demonstrated that targeting liver Xor with GalNAc-siRNAs was a promising strategy for hyperuricemia therapy.

In the traditional farming systems, the excessive application of chemical fertilizers to boost crop yields has resulted in a range of issues, such as soil quality degradation, soil structure deterioration, and pollution of the farmland ecological environment. Green manure, as a high-quality biological fertilizer source with rich nutrient content, is of great significance for enhancing the soil quality and establishing a healthy farmland ecosystem. However, there are few studies on the effects of different green manures on the soil nutrient levels, enzyme activities, and soil bacterial community composition in the rice–wheat rotation areas in southern China. Thus, we planted Chinese milk vetch (MV; Astragalus sinicus L.), light leaf vetch (LV; Vicia villosa var.), common vetch (CV; Vicia sativa L.), crimson clover (CC; Trifolium incarnatum L.), Italian ryegrass (RG; Lolium multiflorum L.), and winter fields without any crops as a control in the Taihu Lake area of Jiangsu. The soil samples collected after tilling and returning the green manure to the field during the bloom period were used to analyze the effects of the different green manures on the soil nutrient content, enzyme activity, and the structural composition of the bacterial community. This analysis was conducted using chemical methods and high-throughput sequencing technology. The results showed that the green manure returned to the field increased the soil pH, soil organic matter (SOM), alkali-hydrolyzed nitrogen (AN), available phosphorus (AP), available potassium (AK), sucrose (SC), urease (UE), and neutral phosphatase (NEP) contents compared to the control. They increased by 1.55% to 10.06%, 0.26% to 9.31%, 20.95% to 28.42%, 20.66% to 57.79%, 12.38% to 37.94%, 3.11% to 58.19%, 6.49% to 32.99%, and 50.0% to 80.36%, respectively. In addition, the green manure field increased the relative abundance of the genera Proteobacteria and Haliangium while decreasing the relative abundance of Gemmatimonadetes, Chloroflexi, SBR1031, and Anaeromyxobacter in the soil bacteria. Both the number of ASVs (amplicon sequence variants) and α-diversity of the soil bacterial communities were higher compared to the control, and the β-diversity varied significantly among the treatments. Alkali-hydrolyzed nitrogen and neutral phosphatase had the greatest influence on the soil bacterial community diversity, with alkali-hydrolyzed nitrogen being the primary soil factor affecting the soil bacterial community composition. Meanwhile, the results of the principal component analysis showed that the MV treatment had the most significant impact on soil improvement. Our study provides significant insights into the sustainable management of the soil quality in rice–wheat rotations. It identifies MV as the best choice among the green manure crops for improving the soil quality, offering innovative solutions for reducing chemical fertilizer dependence and promoting ecological sustainability.

Satellite products have shown great potential in estimating torrential rainfall due to their wide and consistent global coverage. This study assessed the monitoring capabilities of satellite products for the tropical cyclone remote precipitation (TRP) over the Yangtze River Delta region (YRDR) associated with severe typhoon Khanun (2017) and super-typhoon Mangkhut (2018). The satellite products include the CPC MORPHing technique (CMORPH) data, Tropical Rainfall Measuring Mission 3B42 Version 7 (TRMM 3B42), and Integrated Multi-satellite Retrievals for the Global Precipitation Measurement Mission (GPM IMERG). Eight precision evaluation indexes and statistical methods were used to analyze and evaluate the monitoring capabilities of CMORPH, TRMM 3B42, and GPM IMERG satellite precipitation products. The results indicated that the monitoring capability of TRMM satellite precipitation products was superior in capturing the spatial distribution, and GPM products captured the temporal distributions and different category precipitation observed from gauge stations. In contrast, the CMORPH products performed moderately during two heavy rainfall events, often underestimating or overestimating precipitation amounts and inaccurately detecting precipitation peaks. Overall, the three satellite precipitation products showed low POD, high FAR, low TS, and high FBIAS for heavy rainfall events, and the differences in monitoring torrential TRP may be related to satellite retrieval algorithms.

The beta T-cell receptor (TRB) expressed by beta T cells is essential for foreign antigen recognition. The TRB locus contains a TRBV family that encodes three complementarity determining regions (CDRs). CDR1 is associated with antigen recognition and interactions with MHC molecules. In contrast to domestic pigs, African suids lack a 284-bp segment spanning exons 1 and 2 of the TRBV27 gene that contains a sequence encoding CDR1. In this study, we used the African swine fever virus (ASFV) as an example to investigate the effect of deleting the TRBV27-encoded CDR1 on the resistance of domestic pigs to exotic pathogens. We first successfully generated TRBV27-edited fibroblasts with disruption of the CDR1 sequence using CRISPR/Cas9 technology and used them as donor cells to generate gene-edited pigs via somatic cell nuclear transfer. The TRBV-edited and wild-type pigs were selected for synchronous ASFV infection. White blood cells were significantly reduced in the genetically modified pigs before ASFV infection. The genetically modified and wild-type pigs were susceptible to ASFV and exhibited typical fevers (>40 °C). However, the TRBV27-edited pigs had a higher viral load than the wild-type pigs. Consistent with this, the gene-edited pigs showed more clinical signs than the wild-type pigs. In addition, both groups of pigs died within 10 days and showed similar severe lesions in organs and tissues. Future studies using lower virulence ASFV isolates are needed to determine the relationship between the TRBV27 gene and ASFV infection in pigs over a relatively long period.

The English grain aphid (EGA) (Sitobion avenae F.) is one of the most destructive species of aphids in wheat- (Triticum aestivum L.) planting areas worldwide. Large quantities of insecticides are usually used to control aphid damage. The identification of new EGA-resistant genes is necessary for sustainable wheat production. The objective of this study was to identify candidate genes for EGA resistance from stable quantitative trait loci (QTLs). We previously constructed a genetic map of unigenes (UG-Map) with 31,445 polymorphic sub-unigenes via the RNA sequencing of ‘TN18 × LM6’ recombinant inbred lines (TL-RILs). The relative aphid index (RAI) for the TL-RILs was investigated for two growing seasons, with three measured times (MTs) in each season. Using the UG-Map, 43 candidate genes were identified from 22 stable QTLs, with an average of 1.95 candidate genes per QTL. Among the 34 candidate genes annotated in the reference genome Chinese Spring (CS) RefSeq v1.1, the homologous genes of seven candidate genes, TraesCS1A02G-319900, TraesCS1B02G397300, TraesCS2D02G460800, TraesCS4A02G015600LC, TraesCS5B02G329200, TraesCS-6A02G000600 and TraesCS6A02G418600LC have been previously reported to play roles in aphid resistance. This suggests that these genes are strongly associated with EGA resistance in wheat. The candidate genes in this study should facilitate the cloning of EGA-resistant genes and genetic improvement in wheat breeding programs.

Clanis bilineata tsingtauica Mell (Lepidoptera: Sphingidae, CBT), as a traditional edible insect, is becoming popular in China due to its high nutritional value, but production needs to be improved to meet the expanding market. In the present study, CBT eggs were artificially inoculated on soybean leaves to evaluate the effects of leaf position, growth stage, sowing season, and soybean variety on CBT larval growth, respectively. The results showed that (1) the larval weight and survival rate were poorly correlated, so they could represent two different larval growth and development indicators for CBT. The 21-day-old larval weight was significantly different between the sowing seasons and between soybean growth stages, which was suitable as a key indicator for evaluating CBT larval rearing factors. (2) Compared with autumn-sown soybeans, the weight of 21-day-old larvae feeding on V6 stage (sixth trifoliolate) leaves of summer-sown soybeans was significantly higher, with an average increase of 44.7%. (3) Under autumn sowing conditions, the weight of 21-day-old larvae feeding on soybeans in the V6 stage was significantly higher than those fed on soybeans in the R3 stage (beginning pod), increasing by 33.9%. (4) Under summer sowing conditions, the weight of 21-day-old larvae feeding on the third-top leaf (the third leaf from the top of the soybeans’ main stem) was significantly higher than those feeding on the third-bottom leaf (the third leaf from the bottom of the soybeans’ main stem) at V6 stage by 35.7%. Similar results also appeared in autumn sowing; the average weights of 21-day-old larvae feeding on the third-top leaf increased significantly by 29.9% compared to those feeding on the third-bottom leaf. Moreover, the survival rate of larvae fed with the third-top leaf was significantly higher than that of those fed with the third-bottom leaf at the V6 stage in autumn sowing. Leaf position is the main factor affecting the survival rate of larvae. (5) Under summer sowing conditions, the weights of larvae fed with the third-top leaf of the susceptible-soybean varieties NN89-29 and NN1138-2 were significantly higher than that of those fed with the third-bottom leaf of these varieties. This difference was significantly reduced with autumn sowing. In conclusion, the CBT eggs inoculated on the third-top leaf of NN89-29 and NN1138-2 at the V6 stage in summer sowing could achieve maximum larval yield.

Tea is a healthy beverage made from the leaves of the tea plant [Camellia sinensis (L.) O. Kuntze]. The tea plant is a perennial evergreen plant that is widely distributed in tropical and subtropical regions. PR proteins (pathogenesis-related proteins, PRs) are defense-related proteins induced under pathogenesis-related conditions. Currently, there are a few studies on PRs in plants. Tea leaf color mutants have been of wide interest to scientists as special materials. A tea plant displaying variegated spontaneous albinism found in a local tea plantation in Huangshan City, Anhui Province, China, was used as the material sample in this study. An analysis of preexisting transcriptomic and proteomic data revealed the significant upregulation of three classes of PRs. In order to investigate the correlation between PRs and variegated tea plants, a series of studies were conducted on PR genes. The results revealed the identification of 17 CsPR1, 3 CsPR4 and 31 CsPR5 genes in tea plants using bioinformatics methods, and their structures and promoter sequences. The expression of three PR1, two PR4 and one PR5 genes was determined to be induced in stress treatment experiments involving mechanical damage, insect bites, low temperature treatment, and fungal infections. Additionally, the ribonuclease activity of CsPR4 was successfully verified. This is the first study to report the ribonuclease activity of CsPR4 in tea plants. The results can serve as a reference for future studies on PRs in tea plants, offering new insights into information on albinism in tea plants.

Huanglongbing (HLB) is one of the most devastating diseases of citrus worldwide. The phloem-restricted bacterium Candidatus Liberibacter asiaticus (CLas) is considered to be the main pathogen responsible for HLB. There is currently no effective practical strategy for the control of HLB. Our understanding of how pathogens cause HLB is limited because CLas has not been artificially cultured. In this study, 15 potential virulence factors were predicted from the proteome of CLas through DeepVF and PHI-base searches. One among them, FlgI, was found to inhibit yeast growth when expressed in Saccharomyces cerevisiae. The expression of the signal peptide of FlgI fused with PhoA in Escherichia coli resulted in the discovery that FlgI was a novel Sec-dependent secretory protein. We further found that the carboxyl-terminal HA-tagged FlgI was secreted via outer membrane vesicles in Sinorhizobium meliloti. Fluoresence localization of transient expression FlgI-GFP in Nicotiana benthamiana revealed that FlgI is mainly localized in the cytoplasm, cell periphery, and nuclear periphery of tobacco cells. In addition, our experimental results suggest that FlgI has a strong ability to induce callose deposition and cell necrosis in N. benthamiana. Finally, by screening a large library of compounds in a high-throughput format, we found that cyclosporin A restored the growth of FlgI-expressing yeast. These results confirm that FlgI is a novel Sec-dependent effector, enriching our understanding of CLas pathogenicity and helping to develop new and more effective strategies to manage HLB.

Wheat stem rust, caused by the fungus Puccinia graminis f. sp. tritici (Pgt), poses a substantial threat to global wheat production. Utilizing stem rust resistance (Sr) genes represents an economically viable, effective, and environmentally friendly approach to disease control. In this study, gene postulation, molecular testing, and pedigree analysis were used to identify the presence of Sr genes in 45 wheat cultivars. In addition, the resistance of these cultivars was evaluated against two predominant Pgt races, 34MRGQM and 21C3CTHTM, at the adult-plant stage during 2021–2022. The results identify seven Sr genes (Sr31, Sr38, Sr30, SrTmp, Sr22, Sr19, and Sr5) within 35 wheat cultivars. Among these, 23 cultivars contained Sr31, whereas Sr5 and SrTmp were present in four cultivars each. Han 5316, Shimai 15, Shiyou 20, and Kenong 1006 exhibited the presence of Sr19, Sr22, Sr30, and Sr38, respectively. Molecular studies confirmed the absence of Sr25 and Sr26 in any of the wheat cultivars. During field evaluation, 37 (82.2%) and 39 (86.7%) wheat cultivars demonstrated resistance to races 34MRGQM and 21C3CTHTM, respectively. Moreover, 33 wheat cultivars (73.3%) exhibited resistance to all the tested races. These study findings will significantly contribute to future research in wheat pre-breeding and abiotic stress tolerance.

Drought is a natural disaster that occurs globally and can damage the environment, disrupt agricultural production and cause large economic losses. The accurate prediction of drought can effectively reduce the impacts of droughts. Deep learning methods have shown promise in drought prediction, with convolutional neural networks (CNNs) being particularly effective in handling spatial information. In this study, we employed a deep learning approach to predict drought in the Fenhe River (FHR) basin, taking into account the meteorological conditions of surrounding regions. We used the daily SAPEI (Standardized Antecedent Precipitation Evapotranspiration Index) as the drought evaluation index. Our results demonstrate the effectiveness of the CNN model in predicting drought events 1~10 days in advance. We evaluated the predictions made by the model; the average Nash–Sutcliffe efficiency (NSE) between the predicted and true values for the next 10 days was 0.71. While the prediction accuracy slightly decreased with longer prediction lengths, the model remained stable and effective in predicting heavy drought events that are typically difficult to predict. Additionally, key meteorological variables for drought predictions were identified, and we found that training the CNN model with these key variables led to higher prediction accuracy than training it with all variables. This study approves an effective deep learning approach for daily drought prediction, particularly when considering the meteorological conditions of surrounding regions.

Metastasis is one of the leading causes of cancer-related deaths. A comprehensive comparison of the differences between primary and metastatic cancers within the same organ can aid in understanding the growth mechanisms of cancer cells at metastatic sites, thereby helping to develop more effective targeted treatment strategies. Primary liver cancer is one of the most common types of cancer, and the liver is also one of the main metastatic sites. In this paper, we utilize single-cell RNA-Seq data to compare primary liver cancer and colorectal liver metastases from multiple perspectives, including cell types and proportions, activity of various cell types, cell–cell communication, mRNA expression differences within the same types of cells, key factors associated with cell proliferation, etc. Our analysis results show the following: (i) Compared to primary tissue, metastatic tissue contains more cytotoxic T cells and exhausted T cells, and it retains some specific characteristics of the primary site. (ii) Cells of the same type exhibit functional differences between primary and metastatic cancers, with metastatic cancer cells showing lower metabolism levels and immune cells exhibiting stronger immune activity. (iii) Interactions between monocytes and hepato-associated cells are strong in primary cancer, while depleted T cells frequently communicate with hepatocytes in metastatic cancer. (iv) Proliferation-related genes in primary and metastatic cancers are mainly involved in cell energy supply and basic metabolism activity, respectively.

An experimental study is conducted to investigate the effects of different operating parameters on the performance of liquid–gas jet pumps. A square nozzle with an area ratio of 2.25 is designed for the liquid–gas jet pump, and an experimental setup for the liquid–gas jet pump system is constructed. By varying parameters such as inlet flow rate, temperature, and inlet pressure, the variations in the pumping capacity and pumping ratio of the system are studied. The performance of liquid–gas jet pumps with square nozzles and traditional circular nozzles under the same working conditions was compared through experimental data. Explore the performance advantages and disadvantages of liquid–gas jet pumps with different shaped nozzles under the same working conditions. The experimental results indicate that as the inlet flow rate of the liquid–gas jet pump increases, the pumping capacity of the system increases, leading to an increase in the pumping ratio. The operational efficiency slightly decreases with a rise in the working water flow rate. The pumping ratio of the system increases with an increase in the inlet pressure, reaching a peak value of around 4.0 when the inlet valve is fully open. Inlet pressure significantly affects the efficiency of the liquid–gas jet pump, with the highest efficiency point achieved at Pa (inlet air pressure) = 60 kPa, reaching an operational efficiency of 42.48%. When Pa exceeds 70 kPa, the operational efficiency rapidly declines. Comparing the performance of square and circular nozzle liquid–gas jet pumps under the same operating conditions, the performance of the square nozzle liquid–gas jet pump outperforms that of the circular nozzle counterpart. The pumping system’s performance decreases continuously with an increase in the working liquid temperature; however, the decline in pumping performance becomes gradual after exceeding 40 °C. As the water level rises, both the pumping capacity and pumping ratio of the system increase. After the liquid level reaches 40 cm, the changes in the pumping system’s performance become less pronounced.

Landslides are common natural disasters that cause serious damage to ecosystems and human societies. To effectively prevent and mitigate these disasters, an accurate assessment of landslide hazards is necessary. However, most traditional landslide hazard assessment methods rely on static assessment factors while ignoring the dynamic changes in landslides, which may lead to false-positive errors in the assessment results. This paper presents a novel landslide hazard assessment method for the Zagunao River basin, China. In this study, an updated landslide inventory was obtained for the Zagunao River basin using data from interferometric synthetic aperture radar (InSAR) and optical images. Based on this inventory, a landslide susceptibility map was developed using a random forest algorithm. Finally, an evaluation matrix was created by combining the results of deformation rates from both ascending and descending data to establish a hazard level that considers surface deformation. The method presented in this study can reflect recent landslide hazards in the region and produce dynamic assessments of regional landslide hazards. It provides a basis for the government to identify and manage high-risk areas.

The Upper Jinsha River (UJSR) has great water resource potential, but large-scale active landslides hinder water resource development and utilization. It is necessary to understand the spatial distribution and deformation trend of active landslides in the UJSR. In areas of high elevations, steep terrain or otherwise inaccessible to humans, extensive landslide studies remain challenging using traditional geological surveys and monitoring equipment. Stacking interferometry synthetic aperture radar (stacking-InSAR) technology, optical satellite images and unmanned aerial vehicle (UAV) photography are applied to landslide identification. Small baseline subset interferometry synthetic aperture radar (SBAS-InSAR) was used to obtain time-series deformation curves of samples to reveal the deformation types of active landslides. A total of 246 active landslides were identified within the study area, of which 207 were concentrated in three zones (zones I, II and III). Among the 31 landslides chosen as research samples, six were linear-type landslides, three were upward concave-type landslides, 10 were downward concave-type landslides, and 12 were step-type landslides based on the curve morphology. The results can aid in monitoring and early-warning systems for active landslides within the UJSR and provide insights for future studies on active landslides within the basin.

In order to estimate bayberry yield, a lightweight bayberry target detection count model, YOLOv7-CS, based on YOLOv7, was proposed to address the issues of slow detection and recognition speed, as well as low recognition rate, of high-density bayberry targets under complex backgrounds. In this study, 8990 bayberry images were used for experiments. The training set, validation set, and test set were randomly recreated in a ratio of 8:1:1. The new network was developed with SPD-Conv detection head modules to extract features at various scales, to better capture small and indistinct bayberry targets. To improve accuracy and achieve a lightweight design, a CNxP module that replaces the backbone’s ELAN structure is proposed. We propose a global attention mechanism (GAM) in the intermediate layers of the network, to enhance cross-dimensional interactions, and a new pyramid pooling module called SPPFCSPC, to extend the field of perception and improve boundary detection accuracy. Finally, we combine the Wise-IoU function to enhance the network’s ability to identify overlapping and occluded objects. Compared with the SSD, Faster-RCNN, DSSD, and YOLOv7X target detection algorithms, YOLOv7-CS increases mAP 0.5 by 35.52%, 56.74%, 12.36%, and 7.05%. Compared with basic YOLOv7, mAP 0.5 increased from 5.43% to 90.21%, while mAP 0.95 increased from 13.2% to 54.67%. This parameter is reduced by 17.3 m. Ablation experiments further show that the designed module improves the accuracy of bayberry detection, reduces parameter counts, and makes bayberry image detection more accurate and effective.

The sensitive and accurate detection of tumor cells is essential for successful cancer therapy and improving cancer survival rates. However, current tumor cell detection technologies have some limitations for clinical applications due to their complexity, low specificity, and high cost. Herein, we describe the design of a terahertz anti-resonance hollow core fiber (THz AR-HCF) biosensor that can be used for tumor cell detection. Through simulation and experimental comparisons, the low-loss property of the THz AR-HCF was verified, and the most suitable fiber out of multiple THz AR-HCFs was selected for biosensing applications. By measuring different cell numbers and different types of tumor cells, a good linear relationship between THz transmittance and the numbers of cells between 10 and 106 was found. Meanwhile, different types of tumor cells can be distinguished by comparing THz transmission spectra, indicating that the biosensor has high sensitivity and specificity for tumor cell detection. The biosensor only required a small amount of sample (as low as 100 μL), and it enables label-free and nondestructive quantitative detection. Our flow cytometry results showed that the cell viability was as high as 98.5 ± 0.26% after the whole assay process, and there was no statistically significant difference compared with the negative control. This study demonstrates that the proposed THz AR-HCF biosensor has great potential for the highly sensitive, label-free, and nondestructive detection of circulating tumor cells in clinical samples.

Electrochemical biosensors based on immobilized enzymes are among the most popular and commercially successful biosensors. The literature in this field suggests that modification of electrodes with nanomaterials is an excellent method for enzyme immobilization, which can greatly improve the stability and sensitivity of the sensor. However, the poor stability, weak reproducibility, and limited lifetime of the enzyme itself still limit the requirements for the development of enzyme electrochemical biosensors for food production process monitoring. Therefore, constructing sensing technologies based on enzyme electrochemical biosensors remains a great challenge. This article outlines the construction principles of four generations of enzyme electrochemical biosensors and discusses the applications of single-enzyme systems, multi-enzyme systems, and nano-enzyme systems developed based on these principles. The article further describes methods to improve enzyme immobilization by combining different types of nanomaterials such as metals and their oxides, graphene-related materials, metal–organic frameworks, carbon nanotubes, and conducting polymers. In addition, the article highlights the challenges and future trends of enzyme electrochemical biosensors, providing theoretical support and future perspectives for further research and development of high-performance enzyme chemical biosensors.

Bioactive substances exhibit various physiological activities—such as antimicrobial, antioxidant, and anticancer activities—and have great potential for application in food, pharmaceuticals, and nutraceuticals. However, the low solubility, chemical instability, and low bioavailability of bioactive substances limit their application in the food industry. Using nanotechnology to prepare protein nanoparticles to encapsulate and deliver active substances is a promising approach due to the abundance, biocompatibility, and biodegradability of proteins. Common protein-based nanocarriers include nano-emulsions, nano-gels, nanoparticles, and nano complexes. In this review, we give an overview of protein-based nanoparticle fabrication methods, highlighting their pros and cons. Additionally, we discuss the applications and current issues regarding the utilization of protein-based nanoparticles in the food industry. Finally, we provide perspectives on future development directions, with a focus on classifying bioactive substances and their functional properties.

The brassinazole-resistant (BZR) transcription factors (TFs) are key components of brassinosteroid (BR) signaling, which play an important role in regulating plant growth and development as well as responding to abiotic stress. However, a functional study of BZR transcription factors in lotuses has not been reported. A total 10 BZR1 genes (four NnBZR1 and six NlBZR1) were identified from the genomes of two lotus species (Nelumbo nucifera and Nelumbo lutea). The construction of the phylogenetic tree showed that the 10 BZR1 genes of the lotus were divided into four groups; the NnBZR1s and NlBZR1s were unevenly distributed on three and four chromosomes, respectively. Gene structure analysis showed that motif 1 and motif 9 are highly conserved in the lotus BZR1 protein, which might be related to the conserved domain BES_N of BZR1. The analysis of promoter cis-acting elements showed that the promoters of most of the BZR1 genes in the lotus contained elements related to light-responsive, ABA-responsive and abiotic stress-responsive factors, indicating that the BZR1 gene of the lotus played an important role in its response to abiotic stress. The responses of BZR1 genes to BR, ABA and four abiotic stresses (Cold, PEG6000, Cd and NaCl) were analyzed by qRT-PCR. The qRT-PCR results further verified that the lotus BZR1 genes play an important role in responding to hormone signals and resisting abiotic stress. This study laid the foundation for further research on the function of lotus BZR1 genes and provided a theoretical basis for future breeding and horticultural applications.

T cells are critical players in adaptive immunity, driving the tissue injury and organ damage of patients with autoimmune diseases. Consequently, investigations on T cell activation, differentiation, and function are valuable in uncovering the disease pathogenesis, thus exploring promising therapeutics for autoimmune diseases. In recent decades, accumulating studies have pinpointed immunometabolism as the fundamental determinant in controlling T cell fate. Specifically, mitochondria, as a hub of intracellular metabolism, connect glucose, lipid, and amino acid metabolic pathways. Herein, we summarize metabolic adaptations of mitochondrial oxidative phosphorylation and the relevant glucose, lipid, and amino acid metabolism during T cell activation, differentiation, and function. Further, we focused on current updates of the molecular bases for metabolic reprogramming in autoimmune T cells and advances in exploring metabolic-targeted therapeutics against autoimmune diseases. This might facilitate the in-depth understanding of autoimmune pathogeneses and the clinical management of autoimmune diseases.

In recent years, the research on ciphertext-policy attribute-based encryption (CP-ABE) scheme design based on learning with errors (LWE) has been a challenging problem, and many researchers have made a lot of attempts at it. At EUROCRYPT 2021, Datta, Komargodski, and Waters proposed the first provably secure direct CP-ABE construction that supports NC1 circuit access structures. Improving on their work, we propose a CP-ABE scheme. Technically, we use the new lattice two-stage sampling technique of Lai, Liu, and Wang (EUROCRYPT 2021) in the key generation phase instead of the extended trapdoor sampling technique. In this way, we obtain a CP-ABE scheme, which is at least as secure as the original scheme under the same conditions, and has a shorter ciphertext; we provide an innovative design idea for designing CP-ABE schemes only based on LWE, although this is only a partial theoretical work.

In the context of online consumption, consumers are often faced with a decision between buying now or later. This study examines intertemporal choice by conducting two mixed experimental designs on a total of 206 college students to investigate the impact of combining sales volume and limited quantity on online consumption decisions. Experiment 1 revealed that under unlimited conditions, the same option with high sales volume was relatively more attractive and participants were more inclined to choose it, leading to a herding effect. However, under limited conditions, there was no scarcity effect. Experiment 2 built upon the findings of Experiment 1 and added dynamic change in sales volume. It was found that even with dynamic changes in sales volume, high sales volumes could still lead to a herding effect. In the case of unlimited conditions, dynamic changes in sales offset the effect of sales volume on intertemporal choice. Sales volume and limited quantity are important factors that influence consumers’ purchasing decisions. Therefore, this study combines sales volume and limited quantity and adds dynamic changes in sales to explore individuals’ intertemporal choices in online consumption situations. The findings of the study have significant implications for both merchants and consumers.

In this paper, we investigate the problem of a dynamic event-triggered robust controller design for flexible robotic arm systems with continuous-time phase-type semi-Markov jump process. In particular, the change in moment of inertia is first considered in the flexible robotic arm system, which is necessary for ensuring the security and stability control of special robots employed under special circumstances, such as surgical robots and assisted-living robots which have strict lightweight requirements. To handle this problem, a semi-Markov chain is conducted to model this process. Furthermore, the dynamic event-triggered scheme is used to solve the problem of limited bandwidth in the network transmission environment, while considering the impact of DoS attacks. With regard to the challenging circumstances and negative elements previously mentioned, the adequate criteria for the existence of the resilient H∞ controller are obtained using the Lyapunov function approach, and the controller gains, Lyapunov parameters and event-triggered parameters are co-designed. Finally, the effectiveness of the designed controller is demonstrated via numerical simulation using the LMI toolbox in MATLAB.

With the advancement of robotics, the field of path planning is currently experiencing a period of prosperity. Researchers strive to address this nonlinear problem and have achieved remarkable results through the implementation of the Deep Reinforcement Learning (DRL) algorithm DQN (Deep Q-Network). However, persistent challenges remain, including the curse of dimensionality, difficulties of model convergence and sparsity in rewards. To tackle these problems, this paper proposes an enhanced DDQN (Double DQN) path planning approach, in which the information after dimensionality reduction is fed into a two-branch network that incorporates expert knowledge and an optimized reward function to guide the training process. The data generated during the training phase are initially discretized into corresponding low-dimensional spaces. An “expert experience” module is introduced to facilitate the model’s early-stage training acceleration in the Epsilon–Greedy algorithm. To tackle navigation and obstacle avoidance separately, a dual-branch network structure is presented. We further optimize the reward function enabling intelligent agents to receive prompt feedback from the environment after performing each action. Experiments conducted in both virtual and real-world environments have demonstrated that the enhanced algorithm can accelerate model convergence, improve training stability and generate a smooth, shorter and collision-free path.

The pH of saline–alkali soil is high because of carbonate salts, and the deleterious effects of saline–alkali soil on the growth of plants are greater than those of saline soil. Few studies have examined the saline–alkali tolerance of Betula platyphylla at the molecular level. To clarify the regulatory mechanism underlying saline–alkali tolerance in B. platyphylla, RNA sequencing analysis of B. platyphylla seedlings treated with NaHCO3 was conducted. Differences in gene expression in the roots of B. platyphylla seedlings under saline–alkali stress (induced via NaHCO3) for 3 h and 6 h were characterized, and a total of 595 and 607 alkali stress-responsive genes were identified, respectively. Most differentially expressed genes were involved in stress, signal transduction, secondary metabolic process, regulation of jasmonic acid, and the abiotic stimulus signaling pathway. The single nucleotide polymorphism loci in the differentially expressed genes were associated with the alkaline-salt tolerance in birch germplasm. In addition, birch plants overexpressing WRKY70 and NAC9 were obtained using the A. tumefaciens-mediated transient transformation method, and these two genes were found to play key roles in saline–alkali tolerance. Additional study revealed that WRKY70 and NAC9 can increase resistance to saline–alkali stress by enhancing reactive oxygen species scavenging and inhibiting cell death in birch plants. The results of this study enhance our understanding of the saline–alkali stress tolerance of B. platyphylla at the molecular level, and provide several key genes that could be used in the breeding of birch plants in the future.

Glutamic acid is an important amino acid that is used widely in the fields of food, medicine, and agriculture. One of the methods of glutamic acid production is direct microbial fermentation, so the genetic stability and glutamic-acid-producing capacity of the producing strain are the keys to improving glutamic acid concentration. Experiments were carried out using Corynebacterium glutamicum GL−6 as the parental strain, with two iterations of mutagenesis by atmospheric and room temperature plasma (ARTP) and screening with agar plates tolerant to high sugar and malonic acid, and the best strains with stable phenotypes were verified by fermentation in 20 L tanks. The results show that the optimal mutagenesis time of ARTP was 140 s, with lethality and positive mutation rates of 93.0% and 15.6%, respectively. The concentrations of the high-sugar and malonic acid agar plates were 240 g/L and 35 g/L, respectively. A mutant strain, P−45, with improved glutamic acid production capacity and genetic stability, was obtained through two rounds of iterative mutagenesis screening. The concentration of this strain in the Erlenmeyer flasks was 17.7 g/L, which was 18.8% higher than that of the parental strain, GL−6, and could be inherited stably for 10 generations. In the glutamic acid synthesis pathway, the upregulation of the gene encoding citrate synthase (cs), gene encoding isocitrate dehydrogenase (icdh), and gene encoding glutamate dehydrogenase (gdh), and the downregulation of the gene encoding oxoglutarate dehydrogenase complex (odhc) increased the carbon flows of the TCA cycle and its branch metabolic flow to glutamic acid synthesis. P−45 showed a glutamic acid concentration of 147.0 g/L under fed-batch fermentation conditions in 20 L tanks, which was 81.5% higher than the starting strain, GL−6. This study provides a new technical solution for improving microbial metabolites and genetic stability.

Chloramphenicol antibiotics (CAs) are broad-spectrum antibiotics which are widely used in the prevention and treatment of infectious diseases in livestock and poultry breeding. However, overused CAs can enter the watershed and eventually enter the sediment. Antibiotics in sediment can cause secondary pollution through disturbance and suspension. In this study, taking the Fenhe River Basin as the research area, the risk of CAs in sediment were assessed by collecting sediment samples. The results showed that CAs were detected in all sediment samples of the Fenhe River Basin. The mean concentration of CAs was 79.1 μg/kg, and the concentration of thiamphenicol (THI) was dominant, which was up to 58.3 μg/kg. Temporally, there are great differences in different seasons; the concentration of CAs was higher in winter than that in summer, up to 4.79–174 times. Spatially, the mean concentration of CAs in midstream was 83.5 μg/kg, which was higher than that in the upstream and downstream. The concentration of CAs in tributaries were generally higher than that in the main stream, and the mean concentration of tributaries was 1.1 times that of the main stream. CAs in S2 (Lanhe River) was the most prominent among all sample sites; the concentration of CAs was 190.8 μg/kg. The risk threshold of CAs in the sediment was calculated using the Equilibrium Partitioning approach (EqP), based on the distribution coefficient (Kp) and the predicted no-effect concentration (PNEC) in the water, and the values were 0.091–1.44 mg/kg. Based on the risk threshold, the ecological risk of the CAs in sediment was assessed using risk quotients (RQ). The results showed that the Chloramphenicol (CHL) was the most prominent in the Fenhe River Basin, and the proportion of medium-risk areas reached 21.7%, while all the other areas showed low risk. Secondly, the proportion of medium-risk areas was 17.4% for THI, and all the other areas showed low risk. The risk for Florfenicol (FF) was least among all CAs, and the proportion of low-risk areas was only 8.7%, while all the other areas were of insignificant risk.

Sorghum bicolor (L.) is one of the oldest crops cultivated by human beings which has been used in food and wine making. To understand the genetic diversity of sorghum breeding resources and further guide molecular-marker-assisted breeding, six yield-related traits were analyzed for 214 sorghum germplasm from all over the world, and 2,811,016 single-nucleotide polymorphisms (SNPs) markers were produced by resequencing these germplasms. After controlling Q and K, QTLs were found to be related to the traits using three algorisms. Interestingly, an important QTL was found which may affect multiple traits in this study. It was the most likely candidate gene for the gene SORBI_3008G116500, which was a homolog of Arabidopsis thaliana gene-VIP5 found by analyzing the annotation of the gene in the LD block. The haplotype analysis showed that the SORBI_3008G116500hap3 was the elite haplotype, and it only existed in Chinese germplasms. The traits were proven to be more associated with the SNPs of the SORBI_3008G116500 promoter through gene association studies. Overall, the QTLs and the genes identified in this study would benefit molecular-assisted yield breeding in sorghum.

Erythromycin (ERY) is one of the most common macrolides applied in veterinary medicine to treat diseases or as a feed additive for animal growth promotion. Long-term irrational use of ERY could lead to residues in animal-derived food and the emergence of drug-resistant strains, posing potential threats to human health. In this study, a highly sensitive, specific, robust, and rapid fluorescence polarization immunoassay (FPIA) for the determination of ERY in milk has been described. Herein, to achieve high sensitivity, five tracers of ERY with different fluorescein structures were synthesized and paired with three monoclonal antibodies (mAbs). Under the optimized conditions, the combination of mAb 5B2 and tracer ERM-FITC achieved the lowest IC50 value in the FPIA with 7.39 μg/L for ERM. The established FPIA was used to detect ERY in milk, revealing a limit of detection (LOD) of 14.08 μg/L with recoveries of 96.08–107.77% and coefficients of variations (CVs) of 3.41–10.97%. The total detection time of the developed FPIA was less than 5 min from the addition of samples to the result readout. All the above results showed that the proposed FPIA in this study was a rapid, accurate, and simple method for the screening of ERY in milk samples.

Lysosomes are essential organelles of eukaryotic cells and are responsible for various cellular functions, including endocytic degradation, extracellular secretion, and signal transduction. There are dozens of proteins localized to the lysosomal membrane that control the transport of ions and substances across the membrane and are integral to lysosomal function. Mutations or aberrant expression of these proteins trigger a variety of disorders, making them attractive targets for drug development for lysosomal disorder-related diseases. However, breakthroughs in R&D still await a deeper understanding of the underlying mechanisms and processes of how abnormalities in these membrane proteins induce related diseases. In this article, we summarize the current progress, challenges, and prospects for developing therapeutics targeting lysosomal membrane proteins for the treatment of lysosomal-associated diseases.

Flowering time is an important trait that determines the breeding process of ornamental plants. The flowering period of lotus (Nelumbo nucifera Gaertn.) is mainly concentrated in June–August. During this period, the weather is hot and there are few tourists, which made many lotus scenic spots difficult to operate. People have a strong demand for early flowering lotus cultivars. In this paper, 30 lotus cultivars with high ornamental value were selected as materials and their phenological periods were observed for two consecutive years in 2019 and 2020. A number of cultivars with early flowering potential and stable flowering periods, such as ‘Fenyanzi’, ‘Chengshanqiuyue’, ‘Xianghumingyue’ and ‘Wuzhilian’, were screened by K-Means clustering method. The relationship between accumulated temperature and flowering time of 19 lotus cultivars at different growth stages was analyzed. It was found that lotus cultivars with early flowering traits could adapt well to the changes of early environmental temperature and were not affected by low temperature. On the other hand, by analyzing the relationship between different traits and flowering time of three typical cultivars, such as rhizome weight, phenological period, etc., it shows that the nutrient content of the rhizome and the early morphology of plants will affect the flowering time. These results provide a reference for the formation of a systematic lotus early flowering cultivar breeding mechanism and the establishment of a perfect flowering regulation technology system, which can further improve the ornamental value of lotus and promote industrial development.

To address the issues of not accurately identifying ice types and thickness in current fiber-optic ice sensors, in this paper, we design a novel fiber-optic ice sensor based on the reflected light intensity modulation method and total reflection principle. The performance of the fiber-optic ice sensor was simulated by ray tracing. The low-temperature icing tests validated the performance of the fiber-optic ice sensor. It is shown that the ice sensor can detect different ice types and the thickness from 0.5 to 5 mm at temperatures of −5 °C, −20 °C, and −40 °C. The maximum measurement error is 0.283 mm. The proposed ice sensor provides promising applications in aircraft and wind turbine icing detection.

Summer mortality and morbidity are serious environment-related problems in cultured sea cucumbers (Apostichopus japonicus). Air exposure probably worsens the impact of high temperature on cultured sea cucumbers. In this present study, two laboratory experiments were designed to investigate the effects of artificial reefs on mortality, morbidity, crawling, feeding, and adhesion behaviors of small sea cucumbers (~1 g of wet body weight) after air exposure and disease outbreaks at 25 °C, respectively. Significantly lower mortality and morbidity occurred in the group with artificial reefs compared with those in the group without artificial reefs in the two experiments. This present study found that the stressed sea cucumbers cultured inside artificial reefs showed a significantly higher adhesion index, feeding behavior, and crawling frequency than those cultured without artificial reefs. In disease challenge assays, small sea cucumbers cultured inside the artificial reefs showed a significantly higher adhesion index and crawling frequency than those cultured without artificial reefs at 25 °C. Feeding, crawling, and adhesion behaviors of sea cucumbers cultured outside artificial reefs were not significantly different from those cultured without artificial reefs. The experimental results indicate that sea cucumbers with good fitness-related behaviors may be less affected by the disease and more likely to move into the crevices of artificial reefs. Fitness-related behaviors were poor in sea cucumbers cultured outside artificial reefs, so we considered them as affected individuals. Thus, artificial reefs provide a place to reduce the physical contact between unaffected and diseased/affected individuals, showing a potential to reduce disease transmission. Our present study establishes a cost-effective approach to increasing the survival of small sea cucumbers in seed production at high temperatures.

An Ms 6.8 earthquake struck Luding County, Ganzi Prefecture, Sichuan Province on 5 September 2022, with the epicenter about 10 km away from Hailuogou Glacier. How Hailuogou Glacier was affected by the earthquake was of major concern to society. Sentinel-1 SAR satellite imaging was used to monitor the glacier surface velocity during different periods before and after the Luding earthquake based on pixel offset tracking (POT) technology, which applies a feature-tracking algorithm to overcome the phase co-registration problems commonly encountered in large displacement monitoring. The results indicated that the velocity had a positive correlation with the average daily maximum temperature and the slope gradient on the small-slope surfaces. The correlation was not apparent on the steeper surfaces, which corresponded spatially with the identified ice avalanche region in the Planet images. It was deduced that this may be because of the occurrence of ice avalanches on surfaces steeper than 25°, or that the narrower front channel impeded the glacier’s movement. The Luding earthquake did not cause a significant increase in the velocity of Hailuogou Glacier within a large range, but it disturbed the front area of the ice cascade, where the maximum velocity reached 2.5 m/d. Although the possibility of directly-induced destruction by ice avalanches after the earthquake was low, and the buffering in the downstream glacier tongue further reduced the risk of ice avalanches, the risk of some secondary hazards such as debris flow increased. The proposed method in this study might be the most efficient in monitoring and evaluating the effects of strong earthquakes on glaciers because it would not be limited by undesirable weather or traffic blockage.

To improve the drying efficiency, drying uniformity and germination rate after hot air drying of Cyperus esculentus seeds, this paper proposed a drying method that combines tempering and hot air drying. The drying curve of Cyperus esculentus was obtained by experiment. The influence of the drying method on the drying quality of Cyperus esculentus seeds and the trend of drying quality with the tempering process parameters (the tempering temperature, the moisture content of Cyperus esculentus at the beginning of tempering and the tempering duration) were analyzed by single-factor experiments. The regression models were established by star point design response surface methodology, and the relationships between the tempering process parameters and seed production quality indexes were analyzed. The results showed that the optimal combination of tempering process parameters was a tempering temperature of 24 °C, a moisture content of Cyperus esculentus at the beginning of tempering of 23%, and a tempering duration of 3 h. Under this combination, the tempering process increased the drying uniformity, germination rate and drying efficiency of Cyperus esculentus seeds by 21.122%, 4.205% and 22.832%, respectively. The error between the verification test value and the software optimization parameter value was acceptable. The study showed that the optimized tempering drying process significantly improved the drying quality of Cyperus esculentus seed production, and the results could provide a theoretical basis for production practices.
