Cancer research in Rwanda is estimated to be less than 1% of the total African cancer research output with limited research on colorectal cancer (CRC). Rwandan patients with CRC are young, with more females being affected than males, and most patients present with advanced disease. Considering the paucity of oncological genetic studies in this population, we investigated the mutational status of CRC tissues, focusing on the Adenomatous polyposis coli (APC), Kirsten rat sarcoma (KRAS), and Homeobox B13 (HOXB13) genes. Our aim was to determine whether there were any differences between Rwandan patients and other populations. To do so, we performed Sanger sequencing of the DNA extracted from formalin-fixed paraffin-embedded adenocarcinoma samples from 54 patients (mean age: 60 years). Most tumors were located in the rectum (83.3%), and 92.6% of the tumors were low-grade. Most patients (70.4%) reported never smoking, and 61.1% of patients had consumed alcohol. We identified 27 variants of APC, including 3 novel mutations (c.4310_4319delAAACACCTCC, c.4463_4470delinsA, and c.4506_4507delT). All three novel mutations are classified as deleterious by MutationTaster2021. We found four synonymous variants (c.330C>A, c.366C>T, c.513T>C, and c.735G>A) of HOXB13. For KRAS, we found six variants (Asp173, Gly13Asp, Gly12Ala, Gly12Asp, Gly12Val, and Gln61His), the last four of which are pathogenic. In conclusion, here we contribute new genetic variation data and provide clinicopathological information pertinent to CRC in Rwanda.

Dietary exogenous proteases (ENZ) can be used in poultry production to improve the growth of chickens fed low-protein (LP) diets. We hypothesized that ENZ supplemented in an LP diet would improve growth performance and physiological response in broilers for 8−35 days. To investigate this, we used a 2 × 2 factorial design with crude protein (CP, normal diet (NP) and LP) and ENZ. The LP diet contained low in 1% CP and ca. 8−12% amino acids compared to the NP diet and both NP and LP diets were added without or with (1 g/kg of diet) ENZ. We randomly allocated 720 1-week-old Ross 308 male chicks to 48 pens and experimental diets. At 21 days, dietary ENZ, but not CP, increased (p = 0.007) live body weight. Body weight gain from 8−21 days was affected (p = 0.006) by dietary ENZ, but was not affected (p = 0.210) by CP. The feed conversion ratio was affected by both CP and ENZ during the starter period (p < 0.05), by ENZ (p = 0.034) during the finisher period, and by CP (p < 0.001) during the whole period. However, the interaction between CP and ENZ did not significantly affect growth performance (p > 0.05). Dietary ENZ increased (p = 0.013) the relative weight of liver at 21 days. CP and ENZ affected (p = 0.043) total short-chain fatty acids at 21 days. However, this effect was not seen (p = 0.888) at 35 days. Dietary CP increased (p < 0.05) the serum concentrations of both uric acid and creatinine in broilers. We concluded that dietary ENZ is more beneficial to younger broilers, independent of CP levels, and that its effect was restricted to body weight and the feed conversion ratio.

Low HDL-cholesterol (HDL-C), using as cut-offs 1.03 mmol/L in men and 1.29 mmol/L in women, was observed in more than 25% apparently healthy adults (n = 541) in a cross-sectional study on nutrition transition and cardiometabolic risk factors (CMRF) in Benin, West Africa. Both overweight/obesity (35.3%) and underweight (11.3%) were present, displaying the double burden of malnutrition. We examined in more depth the association of low HDL-C with nutrition and with other CMRF. Metabolic syndrome components were assessed, plus the ratio of total cholesterol (TC)/HDL-C and serum homocysteine. Insulin resistance was based on Homeostasis Model Assessment. We also measured BMI and body composition by bio-impedance. Dietary quality was appraised with two non-consecutive 24 h recalls. Low HDL-C was associated with much higher TC/HDL-C and more abdominal obesity in men and women and with more insulin resistance in women. The rate of low HDL-C was highest (41.9%) among the overweight/obese subjects (BMI ≥ 25), but it also reached 31.1% among the underweight (BMI < 18.5), compared with 17.3% among normal-weight subjects (p < 0.001). Lower dietary micronutrient adequacy, in particular, in vitamins A, B3, B12, zinc and calcium, was associated with low HDL-C when controlling for several confounders. This suggests that at-risk lipoprotein cholesterol may be associated with either underweight or overweight/obesity and with poor micronutrient intake.
