Head and neck squamous cell carcinoma (HNSCC) is the sixth leading cancer and the fifth cause of cancer-related deaths worldwide with a poor 5-year survival. SOX family genes play a role in the processes involved in cancer development such as epithelial–mesenchymal transition (EMT), the maintenance of cancer stem cells (CSCs) and the regulation of drug resistance. We analyzed the expression of SOX2-OT, SOX6, SOX8, SOX21, SOX30 and SRY genes in HNSCC patients using the Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) datasets, to assess their biological role and their potential utility as biomarkers. We demonstrated statistically significant differences in expression between normal and primary tumor tissues for SOX6, SOX8, SOX21 and SOX30 genes and pointed to SOX6 as the one that met the independent diagnostic markers criteria. SOX21 or SRY alone, or the panel of six SRY-related genes, could be used to estimate patient survival. SRY-related genes are positively correlated with immunological processes, as well as with keratinization and formation of the cornified envelope, and negatively correlated with DNA repair and response to stress. Moreover, except SRY, all analyzed genes were associated with a different tumor composition and immunological profiles. Based on validation results, the expression of SOX30 is higher in HPV(+) patients and is associated with patients’ survival. SRY-related transcription factors have vast importance in HNSCC biology. SOX30 seems to be a potential biomarker of HPV infection and could be used as a prognostic marker, but further research is required to fully understand the role of SOX family genes in HNSCC.

Magnesium alloys, because of their excellent strength-to-weight ratio, are increasingly used in many industries. When used in external elements, the key factor is to provide adequate anticorrosion protection. High-temperature, cured-powder coatings are widely used to protect most metals, but their use on magnesium alloys is difficult as a result of the instability of the magnesium substrate at elevated temperatures. Another problem is ensuring the proper adhesion of the organic coating to the magnesium substrate. This paper presents the procedure for the synthesis of a duplex coating on AZ91 magnesium alloy. The topcoat was a powder coating based on acrylic resin, the main ingredient of which was glycidyl methacrylate. Because of the presence of epoxy groups, the coating was cured using ultraviolet (UV) radiation (low-temperature technology). The conversion subcoating was produced by plasma electrolytic oxidation (PEO) in an alkaline silicate electrolyte. The synthesized coating system was tested, among others, for microscopic (SEM), adhesive (mesh of cuts), and anticorrosion (EIS). The duplex PEO/UV-curable powder coating showed very good adhesion to the metal and increased the anticorrosion properties of the magnesium substrate, compared to the powder coating produced directly on the magnesium alloy and on an alternative conversion coating (synthesized in the process of chemical zircon phosphating).

In this study, conversion coatings were produced on the AM50 magnesium alloy by a plasma electrolytic oxidation (PEO) process in alkaline-silicate electrolyte with the addition of potassium hexafluorophosphate, using a unipolar pulse power source. The coating microstructure and its composition were determined using scanning electron microscopy (SEM) and an X-ray photoelectron spectroscopy (XPS). The corrosion resistance of the conversion coatings was evaluated by means of potentiodynamic polarization tests (PDP) and electrochemical impedance spectroscopy (EIS) in a dilute Harrison solution (DHS). It has been found that the properties (microstructure, composition, and coating thickness) of the obtained layer and, therefore, their anticorrosive resistance strongly depend on the electrolyte composition. The best anticorrosive properties were observed in the layers obtained in the presence of 2.5 g/L KPF6. It was found that the conversion coating produced with the addition of hexafluorophosphate is characterized by a different morphology (sponge-like) and better anticorrosion properties, in comparison to the coating obtained with the addition of fluoride and orthophosphate salts commonly used in PEO synthesis. The sponge-like structure, which is similar to bone structure in combination with the presence of phosphates in the layer, can increase the biocompatibility and the possibility of self-healing of this coating. However, neither Mg(PF6)2, nor any other compounds containing PF6−, have been found in the layers produced.

The larvae of ermine moths from the Yponomeutidae family (Lepidoptera) feed on a range of species and varieties of fruit and ornamental trees. Some species of this family pose a serious threat to the environment, mainly because of the significant defoliation they cause but also due to the widespread use of insecticides used to control them. This study was designed to assess the sensitivity of Yponomeuta padella and Yponomeuta cagnagella larvae and pupae to a native strain of Steinernema feltiae ZAG15 nematodes under laboratory conditions and to test the biological activity of these nematodes against the larvae and pupae of these species in field studies. The following doses were used in the laboratory tests: 50 IJs/insect (Petri dish tests) and 100 IJs/insect (container tests). Petri dish and container tests were performed at 20 °C and 60% humidity. Mortality of two stages (larvae and pupae) was determined 3 days after treatment. In the field trials, the nematodes were applied at the following doses: 4000 IJs/web for the caterpillars of Y. padella and Y. cagnagella and 1000 IJs/web for the pupae of Y. padella and Y. cagnagella (this corresponded to approximately 200 IJs/insect). Nematodes were applied using a 1 L hand sprayer and a lance. The efficacy of the application was assessed after seven days. The results of our study showed that the larvae (81.7%) and pupae (88.3%) of Y. padella had a greater susceptibility to entomopathogenic nematodes (EPNs) than those of Y. cagnagella (50% and 33.3%, respectively). However, our promising laboratory results did not translate into results in field trials, where the application of EPNs proved to be ineffective.

In the last decade, fresh-cut plants have become a more popular flavoring additive in food. It is important to find an effective method for ensuring the safety and quality of plant materials used as food additives. Ozonated water is being considered by the EFSA for approval as a cidal agent for plant protection. The objective of this study was to evaluate the effectiveness of ozonated water in improving the microbial safety of fresh-cut parsley leaves, with a particular focus on mesophilic and psychrotrophic bacteria and fungi. The yeasts and bacteria were identified with the MALDI-TOF MS system. Color changes on the surface of the parsley samples were measured in the CIE L*a*b trichromatic color model. The chemical composition of the essential oil was evaluated via gas chromatography with mass spectrometry (GCMS). The microbial level of the fresh leaves varied and depended on the season. The highest microbial levels were found in the leaves picked in the summer and autumn, at 104 to 106 CFU/g for fungi and 106 to 108 CFU/g for bacteria. Among the isolates with the highest isolation frequency, bacteria belonging to Pseudomonas fluorescens, Staphylococcus warneri, and Bacillus megaterium dominated. The dominant yeasts and molds were Candida sp., Rhodotorula sp., Cladosporium sp., and Fusarium sp. The conditions for water ozonation (ozone dose and time) were established for both mono- and mixed cultures. Time of 3 min, ozone content of 0.5, O3 mg/L or 1 min, and 1.5 mg of O3 mg/L were sufficient for a 90% reduction in the number of living microorganisms. Yeasts and bacteria were sensitive to ozone treatment, excluding P. fluorescens rods. The tested mold strains were the most resistant. However, it was noted that organic matter might reduce the decontamination effect. The effectiveness of ozonation was negatively influenced by organic compound content above 1%. Spectrophotometric measurements of parsley leaves after ozonation, especially after 3 min treatment at 1.5 O3 mg/L, revealed morphological changes. The CIELAB color space (L*a*b*) changed in the direction of lightness and yellowness; however, ΔE showed no statistically significant differences in comparison with the untreated leaves. In preliminary studies, no differences were noted in GLC-MS chromatograms for essential oils of parsley leaves before and after ozonation. The aroma of parsley treated with ozonated water was more intensely herbal than the control sample, probably due to the higher content of α and β phellandrene. The results of this study show that decontamination of parsley leaves by ozonated water containing 1.5 O3 mg/L in a closed 5 min process can effectively ensure the microbiological quality of fresh-cut parsley leaves. It can be concluded that ozone treatments in aqueous form appear to provide promising qualitative and quantitative results for the decontamination of this fresh-cut plant material. However, more work is necessary to study chemical and volatilome changes. Especially the sensory analyses should be conducted before and after ozone treatment.

This article describes how crystalline or fibrous nanocellulose influences the mechanical properties of paper substrate. In this context, we used commercially available cellulose nanocrystals, mechanically prepared cellulose nanofibers dispersed in water or ethanol, and carboxy cellulose nanofibers. Selective reinforcement of the paper treated with the nanocellulose samples mentioned above was observed. The change in the fibre structure was assessed using scanning electron microscopy, roentgenography, and spectroscopy techniques. In addition, the effect of nanocellulose coating on physical properties was evaluated, specifically tensile index, elongation coefficient, Elmendorf tear resistance, Bendtsen surface roughness, Bendtsen air permeability, and bending strength. It can be concluded that the observed decrease in the strength properties of the paper after applying some NC compositions is due to the loss of potential disturbances in hydrogen bonds between the nanocellulose dispersed in ethanol and the paper substrate. On the other hand, significantly increased strength was observed in the case of paper reinforced with nanocellulose functionalized with carboxyl groups.

The BNT162b2 vaccine is reportedly effective in preventing severe disease in more than 90% of the general population, but its efficacy in transplant recipients remains controversial. We aimed to determine the immune response to the BNT162b2 vaccine in kidney (KTRs) and liver transplant recipients (LTRs). In this retrospective cohort study, we included randomly 65 KTRs and 65 LTRs, who received two 30 μg doses of BNT162b2 vaccine in 3-to6-week intervals. We analyzed the anti-SARS-CoV-2 spike protein IgG antibody (anti-S1 Ab) titer, biochemical liver and renal tests, immunosuppressive drug trough level, and clinical follow up 4–6 weeks after the first dose and 4–8 weeks after the second dose. The level of protective antibodies was 57.1% in KTRs and 88.9% in LTRs after the second dose. The anti-S1 Ab response was significantly associated with sex, age, and history of COVID-19. A tacrolimus dose at vaccination but not its trough level was significantly correlated with the increase in anti-S1 Ab titer after the second vaccine dose in LTRs. Rejection episodes did not occur after vaccination. Our results showed a higher than previously reported humoral response to the BNT162b2 vaccine in KTRs and LTRs, which was dependent upon age, type of transplanted organ, and immunosuppression.

This study aims to investigate if vaginal bacteriology obtained prior to treatment influences the 3′-deoxy-3 18F-fluorothymidine (FLT) [18F]FLT and 2-deoxy-2-[18F]fluoro-d-glucose (2-[18F]FDG) [18F]FDG parameters in positron emission tomography (PET/CT) in cervical cancer (CC) patients. Methods: Retrospective analysis was performed on 39 women with locally advanced histologically confirmed cervical cancer who underwent dual tracer PET/CT examinations. The [18F]FLT and [18F]FDG PET parameters in the primary tumor, including SUVmax, SUVmean, MTV, heterogeneity, before radiotherapy (RT) were analyzed, depending on the bacteriology. The p-values < 0.05 were considered statistically significant. Results: In the vaginal and/or cervical smears, there were 27 (79.4%) positive results. In seven (20.6%) cases, no opportunistic pathogen growth was observed (No Bacteria Group). In positive bacteriology, eleven (32%) Gram-negative bacilli (Bacteria group 2) and fifteen (44%) Gram-positive bacteria (Bacteria group 1) were detected. Five patients with unknown results were excluded from the analysis. Data analysis shows a statistically significant difference between the SUVmax, and SUVmin values for three independent groups for the [18F]FLT. Conclusions: The lowest values of SUVmax and SUVmin for [18F]FLT are registered in Gram-negative bacteria, higher are in Gram-positive, and the absence of bacteria causes the highest [18F]FLT values.

Endometriosis is a gynecological disease defined by the presence of endometrial tissue outside the uterus. To date, the effective treatment of this disease is still based on invasive surgery or laparoscopy. Chelidonium majus L. (Papaveraceae) belongs to medicinal, latex-bearing plants. Extracts from the plant are a rich source of pharmacologically active agents. Protoberberine compounds derived from C. majus possess anticancer and antiproliferative activities. In the present study of a rat model of endometriosis, we investigated the influence of the plant protoberberine-rich fraction (BBR) obtained from the medicinal plant C. majus on the development of endometriosis. To understand of BBR therapeutic potential for endometriosis, metabolomics has been applied to study. BBR was prepared from an ethanolic extract of dry plants C. majus. Rats (n = 16) with confirmed endometriosis were treated with BBR administered orally (1 g/kg) for 14 days. Blood serum samples were collected from all of the animals and metabolites were studied using the NMR method. The metabolomic pattern was compared before and after the protoberberine treatment. The performed analysis showed significant changes in the concentrations of metabolites that are involved in energy homeostasis, including glucose, glutamine, and lactate. Histopathological studies showed no recurrence of endometriosis loci after treatment with BBR. The results of the study found that BBR treatment prevents the recurrence of endometriosis in rats. Moreover, metabolomics profiling can be applied to better understand the mechanisms of action of these protoberberine secondary plant metabolites. Our findings provide new insights into the pharmaceutical activity of natural protoberberine plant compounds.

Although progress is observed in cancer treatment, this disease continues to be the second leading cause of death worldwide. The current understanding of cancer indicates that treating cancer should not be limited to killing cancer cells alone, but that the target is the complex tumor microenvironment (TME). The application of nanoparticle-based drug delivery systems (DDS) can not only target cancer cells and TME, but also simultaneously resolve the severe side effects of various cancer treatment approaches, leading to more effective, precise, and less invasive therapy. Nanoparticles based on proteins derived from silkworms’ cocoons (like silk fibroin and sericins) and silk proteins from spiders (spidroins) are intensively explored not only in the oncology field. This natural-derived material offer biocompatibility, biodegradability, and simplicity of preparation methods. The protein-based material can be tailored for size, stability, drug loading/release kinetics, and functionalized with targeting ligands. This review summarizes the current status of drug delivery systems’ development based on proteins derived from silk fibroin, sericins, and spidroins, which application is focused on systemic cancer treatment. The nanoparticles that deliver chemotherapeutics, nucleic acid-based therapeutics, natural-derived agents, therapeutic proteins or peptides, inorganic compounds, as well as photosensitive molecules, are introduced.

The article presents the results of estimates of thermodynamic parameters (oxygen balance, pressure, temperature, gaseous products produced in the reaction, specific energy, and the combustion heat) for 11 pyrotechnic mixtures and the final product—gas generator prototype (RSP), which can be used in mines for producing blocks. In addition, formal and legal considerations have been presented, as well as the results of gas generators in Polish mines. In order to compare the effects induced by using a detonating cord and a gas generator prototype, a test was carried out in granite quarries involving the measurement, at the same points, of vibration and the air blast. As a result of the tests, limitations and possibilities of using gas generators were indicated.
