Research indicates that, owing to the enhanced grain-filling rate of tetraploid rice, its yield has notably improved compared to previous levels. Studies conducted on diploid rice have revealed that optimal planting density and fertilization rates play crucial roles in regulating rice yield. In this study, we investigated the effects of different nitrogen application and planting density treatments on the growth, development, yield, and nitrogen utilization in tetraploid (represented by T7, an indica–japonica conventional allotetraploid rice) and diploid rice (Fengliangyou-4, represented by FLY4, a two-line super hybrid rice used as a reference variety for the approval of super rice with a good grain yield performance). The results indicated that the highest grain-filling rate of T7 could reach 77.8% under field experimental conditions due to advancements in tetraploid rice breeding. This is a significant improvement compared with the rate seen in previous research. Under the same conditions, T7 exhibited a significantly lower grain yield than FLY4, which could be attributed to its lower grain-filling rate, spikelets per panicle, panicle number m−2, and harvest index score. Nitrogen application and planting density displayed little effect on the grain yield of both genotypes. A higher planting density significantly enhanced the leaf area index and biomass accumulation, but decreased the harvest index score. Compared with T7, FLY4 exhibited a significantly higher nitrogen use efficiency (NUEg), which was mainly due to the higher nitrogen content in the straw. Increasing nitrogen application significantly decreased NUEg due to its minimal effect on grain yield combined with its significant enhancement of nitrogen uptake. Our results suggest that the yield and grain-filling rate of T7 have been improved compared with those of previously tested polyploid rice, but are still lower than those of FLY4, and the yield of tetraploid rice can be further improved by enhancing the grain-filling rate, panicle number m−2, and spikelets per panicle via genotype improvement.

Photosynthetic induction and stomatal kinetics are acknowledged as pivotal factors in regulating both plant growth and water use efficiency under fluctuating light conditions. However, the considerable variability in methodologies and light regimes used to assess the dynamics of photosynthesis (A) and stomatal conductance (gs) during light induction across studies poses challenges for comparison across species. Moreover, the influence of stomatal morphology on both steady-state and non-steady-state gs remains poorly understood. In this study, we show the strong impact of IRGA Chamber Illumination and Whole Plant Illumination on the photosynthetic induction of two rice species. Our findings reveal that these illuminations significantly enhance photosynthetic induction by modulating both stomatal and biochemical processes. Moreover, we observed that a higher density of smaller stomata plays a critical role in enhancing the stomatal opening and photosynthetic induction to fluctuating light conditions, although it exerts minimal influence on steady-state gs and A under constant light conditions. Therefore, future studies aiming to estimate photosynthetic induction and stomatal kinetics should consider the light environments at both the leaf and whole plant levels.

Manipulating the photoprotective mechanism has been demonstrated to be an effective way to enhance the photosynthetic productivity of crop plants. NPQ(T) is a chlorophyll fluorescence parameter for rapid estimation and imaging of non-photochemical quenching (NPQ) of excitons in the photoprotective mechanism. However, the variation and genetic basis of NPQ(T) are rarely reported in the Oryza sativa L. In this study, we collected 173 rice cultivars and investigated the NPQ(T) value. We found that the NPQ(T) has a wide variation, although it had not been under-selected in the different subspecies. A genome-wide association study (GWAS) utilizing 1,566,981 high-quality SNPs identified three significant associated signals on chromosomes 02, 05, and 07. Furthermore, one likely candidate gene Os02g0184100, underlying the associated signal on chromosome 02, was uncovered by identifying the expression pattern in flag leaves and testing the correlation between functional polymorphisms and phenotypic variation. The significant SNPs and candidate genes identified in this study provide us a comprehensive understanding of the genetic architecture of NPQ(T) and could be used for genetic improvement of rice photoprotection.

Stomata control carbon and water exchange between the leaves and the ambient. However, the plasticity responses of stomatal traits to growth conditions are still unclear, especially for monocot leaves. The current study investigated the leaf anatomical traits, stomatal morphological traits on both adaxial and abaxial leaf surfaces, and photosynthetic traits of Oryza leaves developed in two different growth conditions. Substantial variation exists across the Oryza species in leaf anatomy, stomatal traits, photosynthetic rate, and stomatal conductance. The abaxial stomatal density was higher than the adaxial stomatal density in all the species, and the stomatal ratios ranged from 0.35 to 0.46 across species in two growth environments. However, no difference in the stomatal ratio was observed between plants in the growth chamber and outdoors for a given species. Photosynthetic capacity, stomatal conductance, leaf width, major vein thickness, minor vein thickness, inter-vein distance, and stomatal pore width values for leaves grown outdoors were higher than those for plants grown in the growth chamber. Our results indicate that a broad set of leaf anatomical, stomatal, and photosynthetic traits of Oryza tend to shift together during plasticity to diverse growing conditions, but the previously projected sensitive trait, stomatal ratio, does not shape growth conditions.

Conventional transplanted rice (TPR) has been increasingly replaced by direct-seeded rice (DSR) because of its low water and labour requirements. Whether and how DSR can be as productive as TPR has received widespread attention. Here, a comprehensive meta-analysis was performed to quantify the effects of direct seeding on rice yield and identify the management and environmental factors that contribute to the yield gap between DSR and TPR. The results showed that, overall, the yield of DSR was 12% lower than that of TPR. However, the yield loss of DSR relative to TPR was highly variable depending on management practices, soil type, and climate conditions, ranging from −2% to −42%. Weed and water management and climatic stress had the largest impact on yield performance, resulting in over 15% yield variation. With respect to soil properties, the yield gap can be significantly reduced by planting in areas with high organic carbon content, such as clayed and acidic soils. Furthermore, the DSR yield penalty was only 4% in a high-yielding condition compared to 14% in a low-yielding condition. All these factors indicate that optimizing management practices is necessary to improve DSR yield performance and narrow the yield gap between DSR and TPR. In conclusion, DSR could produce comparable yields to TPR but is more prone to yield losses due to inappropriate management practices, unsuitable soil properties, and climatic stresses.
