Among forbidden substances included in the European Union legislation, endogenous steroids constitute a challenge in the framework of veterinary Official Monitoring Plans. They can be naturally present in body fluids at variable levels depending on the species, sex and age of the animals. Considering the significant advances achieved in breeding conditions and in the selection of producing traits in meat cattle, the aim of this study was to verify by analytical method, in veal calves housed under controlled conditions, if the level of natural steroids hormones assumed as physiological are still actual. The second aim of the study was to verify if the normal histological pattern of growth promoters in target organs is influenced accordingly. Bovine male sex organs are currently analysed in the frame of the Italian histological plan to monitor illicit treatments trend, highlighting microscopic, induced alterations. The levels of 17β-estradiol and progesterone residues resulted under the Limit of Quantitation of the approved official methods and the level of testosterone resulted below the level stated in the Italian Ministerial Decree in force. Male target organs appeared within the limits of the standard histological features.

Ocular ultrasonography is seldom performed in cattle. Here, we compared three ultrasound probes (linear, convex, transrectal gynecological) for the measurement of eight eye biometry parameters on vertical and horizontal scans. The sample population was 20 healthy calves (n = 10 Holstein, n = 10 Piedmontese breed). Intragroup (same probe for vertical vs. horizontal scanning) and intergroup (different probes measuring the same biometric parameter) comparisons were performed using Student’s t-test and the Mann–Whitney U test. Statistical significance was set at p ≤ 0.05. Intragroup comparison revealed few significant differences. Ultrasound examination with the convex transducer detected more differences than either of the two other probes on vertical (linear six out of eight; transrectal gynecological four out of eight) and horizontal (linear six out of eight; transrectal gynecological six out of eight) scans. Similar results were obtained for both breeds. More non-valuable parameters on the horizontal (77 out of 320, 24%) and the vertical (85 out of 320, 26%) (p ≤ 0.001) scans were obtained with the convex transducer. Both linear transducers were found comparable for ocular ultrasonography in field conditions. However, given its widespread application in the field, the transrectal gynecological transducer may offer veterinarians the added advantage of familiarity and ease-of-use without any additional costs.

The up-to-date literature suggests that the compost-bedded pack barn housing system is capable of remarkably improving productive and reproductive performance, as well as health status and welfare, in dairy cattle. However, there is currently limited knowledge available on the endocrine and biochemical changes in animals housed in such alternative systems. Therefore, this study aimed to measure blood cortisol (COR) and beta-endorphins (BE) in 22 two-year-old primiparae Fleckvieh cows, who were randomly allotted to the following two different housing systems: CB (n = 11) and FB (n = 11). Blood samples were collected at the beginning of the experiment (T0) and every two months thereafter (T1, T2, and T3). The COR and BE were measured through an immunoenzymatic kit. With the only exception being T0, no differences were observed over time between the two groups, neither for COR nor for BE. However, the blood cortisol levels of the CB cows decreased over time, while a T1 peak was identified in the FB group. On the contrary, both the housing systems displayed numerically higher BE at T3 than at the other experimental times. Therefore, the overall data suggest that the compost-bedded pack barn did not significantly affect the studied parameters. Accordingly, cow welfare should be assessed using a wider panel of animal-based indicators.

Intravenous iodinated contrast (IVIC) medium is routinely administered to dogs. Scattered information exists regarding the serum biochemical or urinary profiles associated with the administration of IVIC in dogs. The aim of the study was to describe, compare, and discuss from the perspective of previous studies the alterations in serum biochemical and urinary parameters before (T0) and within one week (T1) of the IVIC administration during routine computed tomography (CT) scan evaluation of 22 dogs. Mature dogs presenting for CT scan evaluation for preoperative oncology staging/surgical planning were included. T1 evaluation was performed within one week of IVIC administration. Statistically significant differences in serum total protein, albumin, chloride, calcium, and phosphorus concentrations, urine protein to creatinine ratio, and urine specific gravity were found between T1 and T0. At T1, the serum creatinine concentration was within reference ranges in all dogs but one. An increase in the urine protein to creatinine ratio was observed in four samples, one of which was non-proteinuric at T0. Changes in biochemistry and urine parameters between T0 and T1 were not considered clinically significant.

The abuse or misuse of antimicrobials in animal production is thought to be a potential factor in the development of antimicrobial resistance in veterinary and human medicine. With this study, we wanted to quantify antimicrobial usage in beef fattening operations in northwestern Italy and to identify factors potentially influencing antimicrobial usage. The sample was composed of 26 beef fattening operations that import heifers and bulls from France. Data were extracted from the 2014 and 2015 treatment registers kept by the farmers. The mean (±SD) number of animal daily doses per animal (nADDa) per year for each farm was 3 (±2.1) during the study period (2014–2015). Group antimicrobial treatments (57.5% of all treatments) were often administered orally (70.5%) and consisted overwhelmingly of doxycycline (97%). Individual treatments (42.5% of all treatments) were administered parenterally (98.1%) and the most often used active substances were florfenicol (19.9%), marbofloxacin (19.5%), and tylosin (12.4%). There was a negative correlation between the nADDa for total and group treatments and average batch weight at arrival and between the amount of straw added per animal per day and the nADDa (p ≤ 0.05). Our data show that antimicrobials critical for human medicine were often used in beef fattening operations in northwestern Italy before the European guidelines for the prudent use of antimicrobials in veterinary medicine were issued. Additionally, the use of antimicrobials as a preventive group treatment was still widespread, mostly in lighter weight animals.

Horses kept for meat production are reared in intensive breeding farms. We employed a checklist adapted from the Animal Welfare Indicators (AWIN) assessment protocol. Our evaluation aims to assess whether welfare indicators are influenced by stocking densities (m2/horse) and feeding strategies applied. An analysis was carried out on the data obtained from 7 surveys conducted at a single horse farm designed for meat production. In each survey, the same 12 pens were assessed, but on each occasion, the horses in the pens had been changed as had the stocking densities. Briefly, 561 horses aged 16 ± 8 months (mean ± standard deviation) were evaluated. Two stocking density cut-off values (median and 75th percentile: 3.95 and 4.75 m2/horse, respectively) were applied to investigate the effect of stocking density on horse welfare. Data were analysed using Mann–Whitney U and Fisher’s exact tests (p < 0.05). When cut-off was set as the median percentile, lower stocking density was associated with improvements in body condition score (BCS), coat cleanliness and bedding quantity, less coughing, less resting in a standing position, and less feeding related to the greater space available at the feed bunk. When the 75th percentile cut-off was used, indicators that improved were coat cleanliness, bedding quantity and mane and tail condition, as well as less resting in standing position and less feeding related to the greater space available at the feed bunk. Accordingly, the use of two different stocking density cut-off values showed that the increase of space allowance affected specific welfare indicators. Further increment of space and/or changes in management regimes should be investigated to improve all the indicators. Moreover, results related to feeding indicated the need to intervene as starch intakes exceeded recommended safe levels, negatively affecting horse welfare.

Canine aggression is a major concern, affecting millions of people worldwide, and treatment can be challenging even for skilled veterinarians. Empiric use of fluoxetine is sometimes attempted, although few data regarding long-term effects in aggressive dogs are available. The aim of the study was to investigate clinical effectiveness of fluoxetine (1.5 mg/kg/die PO) combined with a behavior modification program for treatment of canine dominance-related aggression. Circulating levels of fluoxetine, norfluoxetine, and serotonin (5-HT) were also measured. Eight dogs with a diagnosis of dominance aggression (owner-directed) were enrolled. Before treatment (T0), and after one (T1), two (T2), four (T3), and six (T4) months of fluoxetine administration, clinical outcomes were graded using a five-point frequency scale (0–4), and blood samples were collected to measure fluoxetine/norfluoxetine (high-performance liquid chromatography) and 5-HT (ELISA) levels. Following treatment, a decrease in behavioral test scores was observed at T1–T4. Increasing concentrations of circulating fluoxetine and norfluoxetine were measured throughout the follow-up. Correlation between norfluoxetine levels and clinical scores was observed at T4. Starting from T1, a significant decrease in 5-HT levels was observed. Our data suggest that fluoxetine (1.5 mg/kg/day) when associated with behavior treatment is effective in controlling canine aggression over a six-month period, and that, in dogs norfluoxetine levels seem reliable in predicting clinical efficacy.

The use of reference genes is commonly accepted as the most reliable approach to normalize qRT-PCR and to reduce possible errors in the quantification of gene expression. The most suitable reference genes in sheep have been identified for a restricted range of tissues, but no specific data on whole blood are available. The aim of this study was to identify a set of reference genes for normalizing qRT-PCR from ovine whole blood. We designed 11 PCR assays for commonly employed reference genes belonging to various functional classes and then determined their expression stability in whole blood samples from control and disease-stressed sheep. SDHA and YWHAZ were considered the most suitable internal controls as they were stably expressed regardless of disease status according to both geNorm and NormFinder software; furthermore, geNorm indicated SDHA/HPRT, YWHAZ/GAPDH and SDHA/YWHAZ as the best reference gene combinations in control, disease-stressed and combined sheep groups, respectively. Our study provides a validated panel of optimal control genes which may be useful for the identification of genes differentially expressed by qRT-PCR in a readily accessible tissue, with potential for discovering new physiological and disease markers and as a tool to improve production traits (e.g., by identifying expression Quantitative Trait Loci). An additional outcome of the study is a set of intron-spanning primer sequences suitable for gene expression experiments employing SYBR Green chemistry on other ovine tissues and cells.
