In recent years, trafficking and abuse of hallucinogenic mushrooms have become a serious social problem. It is therefore imperative to identify hallucinogenic mushrooms of the genus Psilocybe for national drug control legislation. An internal transcribed spacer (ITS) is a DNA barcoding tool utilized for species identification. Many methods have been used to discriminate the ITS region, but they are often limited by having a low resolution. In this study, we sought to analyze the ITS and its fragments, ITS1 and ITS2, by using high-resolution melting (HRM) analysis, which is a rapid and sensitive method for evaluating sequence variation within PCR amplicons. The ITS HRM assay was tested for specificity, reproducibility, sensitivity, and the capacity to analyze mixture samples. It was shown that the melting temperatures of the ITS, ITS1, and ITS2 of Psilocybe cubensis were 83.72 &plusmn, 0.01, 80.98 &plusmn, 0.06, and 83.46 &plusmn, 0.08 &deg, C, and for other species, we also obtained species-specific results. Finally, we performed ITS sequencing to validate the presumptive taxonomic identity of our samples, and the sequencing output significantly supported our HRM data. Taken together, these results indicate that the HRM method can quickly distinguish the DNA barcoding of Psilocybe cubensis and other fungi, which can be utilized for drug trafficking cases and forensic science.

Mitochondrial DNA (mtDNA), coupled with maternal inheritance and relatively high mutation rates, provides a pivotal way for us to investigate the formation histories of populations. The Hui minority with Islamic faith is one of the most widely distributed ethnic groups in China. However, the exploration of Hui&rsquo, s genetic architecture from the complete mitochondrial genome perspective has not been detected yet. Therefore, in this study, we employed the complete mitochondrial genomes of 98 healthy and unrelated individuals from Northwest China, as well as 99 previously published populations containing 7274 individuals from all over the world as reference data, to comprehensively dissect the matrilineal landscape of Hui group. Our results demonstrated that Hui group exhibited closer genetic relationships with Chinese Han populations from different regions, which was largely attributable to the widespread of haplogroups D4, D5, M7, B4, and F1 in these populations. The demographic expansion of Hui group might occur during the Late Pleistocene. Finally, we also found that Hui group might have gene exchanges with Uygur, Tibetan, and Tajik groups in different degrees and retained minor genetic imprint of European-specific lineages, therefore, hinting the existence of multi-ethnic integration events in shaping the genetic landscape of Chinese Hui group.

Adsorbent material was prepared using distillers’ grains (DG), which is a waste product of distilleries. The DG was pre-treated with NaOH and esterification-modified with CS2, which is a commonly used anionic modifier. The structure and morphology of the adsorbent was characterized by FTIR, XRD, EDS, SEM, BET, and zeta potential. The related mechanism of adsorption of malachite green (MG) onto modified distiller’s grains (MDG) was studied by adsorption experiments and molecular simulation techniques. The experimental results showed that CS2 successfully modified the DG fiber, and simultaneously yielded the MDG with a uniform pore distribution. MDG had a considerable adsorption capacity of 367.39 mg/g and a maximum removal rate of 96.51%. After eight adsorption–desorption cycle experiments, the adsorption removal rate of MDG to MG dye remained at 82.6%. The adsorption process could be fitted well by a pseudo-second-order kinetic model (the correlation coefficient R2 > 0.998) and Freundlich isotherm adsorption equation (the correlation coefficient R2 > 0.972). Moreover, the adsorption of MG dye by MDG is a spontaneous, endothermic, and increased entropy process. The results of molecular simulation showed that the mechanism of MG molecules onto MDG was mainly chemical adsorption. The adsorption performance of MG onto MDG was better and more stable than DG. Molecular simulation also provided a theoretical guidance of MDG adsorption–desorption for the research on recycling of DG resources.

This study examined the correlation between non-metallic organic tanning agents (NOTAs) and wastewater containing double-triazine (bistriazine). Specifically, high-throughput sequencing was used to analyze how the physical and chemical properties of the bistriazine-containing wastewater affected the microbial community. In total, 9995 operating taxonomy units (OTUs) were detected at a similarity level of 97%. According to the taxonomy results and relative bacterial richness, the non-metallic organic tanning agent (NOTA) wastewater communities showed significant differences. Clostridium sensu stricto and Turicibacter bacteria were dominant in NOTA T2, T3, and the blank control (KB) wastewater, Trichococcus and Aeromonas were dominant in NOTA T4, and Pseudomonas was dominant in T2, T4, and the blank control (KB). Thus, the addition of bistriazine ring derivatives altered the composition of the bacterial community. Furthermore, Spearman’s correlation analysis revealed a substantial correlation in the composition of the communities and the physicochemical properties of the wastewater. Acidocella and norank c Cyanobacteria correlated positively with Total Phosphorus (TP) (p ≤ 0.001) and Mycobacterium showed a positively held correlation with pH (p ≤ 0.05). Conversely, norank_f__Elev-16S-1332 indicated a negative correlation with TP, Total Nitrogen (TN), and Total Organic Carbon (TOC) (p ≤ 0.05), whereas Zoogloea correlated negatively with NH3–N, TOC, and TN (p ≤ 0.05). Regarding water quality, a significant correlation between microbial communities and the physicochemical properties of bistriazine wastewater was also observed (p ≤ 0.05). We concluded that Trichococcus, Clostridium sensu stricto, Turicibacter, Pseudomonas, Intestinibacter, Acidocella, Bacillus, and Tolumonas showed a high tolerance for bistriazine wastewater, which could offer insight into its bioremediation.
