Rosa roxburghii, a calciphilic species native to the mountainous regions of Southwest China, is renowned for its high vitamin C and bioactive components, making it valuable for culinary and medicinal uses. This species exhibits remarkable tolerance to the high-calcium conditions typical of karst terrains. However, the underlying mechanisms of this calcium resilience remain unclear. The Ca2+/cation antiporter (CaCA) superfamily plays a vital role in the transport of Ca2+ and other cations and is crucial for plant tolerance to metal stress. However, the roles and evolutionary significance of the CaCA superfamily members in R. roxburghii remain poorly understood. This study identified 22 CaCA superfamily genes in R. roxburghii, categorized into four subfamilies. The gene structures of these RrCaCAs show considerable conservation across related species. Selection pressure analysis revealed that all RrCaCAs are subject to purifying selection. The promoter regions of these genes contain numerous hormone-responsive and stress-related elements. qRT-PCR analyses demonstrated that H+/cation exchanger (CAX) RrCAX1a and RrCAX3a were highly responsive to Ca2+ stress, cation/Ca2+ exchanger (CCX) RrCCX4 to Mg2+ stress, and RrCCX11a to Na+ stress. Subcellular localization indicated that RrCAX1a is localized to the plant cell membrane, and its stable transformation in tobacco confirmed its ability to confer enhanced resistance to heavy Ca2+ stresses, highlighting its crucial role in the high-calcium tolerance mechanisms of R. roxburghii. This research establishes a foundation for further molecular-level functional analyses of the adaptation mechanisms of R. roxburghii to high-calcium environments.

The echo traces in the ionograms contain key information about the ionosphere. Therefore, the accurate extraction of these traces is crucial for the subsequent work. This paper transforms the original signal processing problem into a semantic segmentation task, combines it with the currently popular deep learning techniques, and proposes a multiscale Transformer network to achieve pixel-level trace extraction. To train the proposed model, we built a dataset by discretizing the original echo data, labeling, and other preprocessing work. A series of advanced semantic segmentation networks are utilized for comparative experiments. The analysis of the results indicates that the proposed network excels in performance, achieving the highest scores on key semantic segmentation evaluation metrics, including mIoU, Kappa, Dice, and AUC-ROC. In addition, this paper also designs a series of ablation experiments to observe the changes in network performance and to evaluate the rationality of the network design. The experimental results demonstrate the effectiveness of the network in the trace extraction task, which plays a positive role in the subsequent electron density reversal work.

In this paper, a matching method for altimeter and transponder signals in Sub-optimal Maximum Likelihood Estimate (SMLE) tracking mode is proposed. In the in-orbit calibration of the altimeter in SMLE tracking mode using the reconstructive transponder, it is necessary to separate the forwarding signal from the ground echo signal. At the same time, the fluctuations in the received signal of the altimeter, which are caused by the forwarding signal of the transponder, can be eliminated. The transponder generates a bias when measuring the arrival time of the transmitting signal from the altimeter and embeds this bias in both the transponder-recorded data and the altimeter-recorded data. Therefore, the two sets of data have one-to-one correspondence, and they are superimposed using the sliding sum method. Moreover, the distance between the altimeter and the transponder is a parabolic geometric relationship, and the outliers are eliminated by the fitting error minimization decision, and the transponder signal is separated from the ground echo. The final altimeter transmitting–receiving signal path is obtained. Furthermore, the principles underlying this method can be used for any transponder that can adjust the response signal delay during calibration.

Vaccinium duclouxii, a wild blueberry species native to the mountainous regions of southwestern China, is notable for its exceptionally high anthocyanin content, surpassing that of many cultivated varieties and offering significant research potential. Glutathione S-transferases (GSTs) are versatile enzymes crucial for anthocyanin transport in plants. Yet, the GST gene family had not been previously identified in V. duclouxii. This study utilized a genome-wide approach to identify and characterize the GST gene family in V. duclouxii, revealing 88 GST genes grouped into seven distinct subfamilies. This number is significantly higher than that found in closely related species, with these genes distributed across 12 chromosomes and exhibiting gene clustering. A total of 46 members are classified as tandem duplicates. The gene structure of VdGST is relatively conserved among related species, showing closer phylogenetic relations to V. bracteatum and evidence of purifying selection. Transcriptomic analysis and qRT-PCR indicated that VdGSTU22 and VdGSTU38 were highly expressed in flowers, VdGSTU29 in leaves, and VdGSTF11 showed significant expression in ripe and fully mature fruits, paralleling trends seen with anthocyanin accumulation. Subcellular localization identified VdGSTF11 primarily in the plasma membrane, suggesting a potential role in anthocyanin accumulation in V. duclouxii fruits. This study provides a foundational basis for further molecular-level functional analysis of the transport and accumulation of anthocyanins in V. duclouxii, enhancing our understanding of the molecular mechanisms underlying anthocyanin metabolism in this valuable species.

This study evaluates the effects of a combined rice-crayfish farming model and compares this model with traditional paddy fields. The focus is on soil aggregate characteristics, organic matter content, and also the distribution of soil aggregates. This research was conducted in Qianjiang, Hubei Province. The surface soil samples were collected from two types of arable land: paddy fields (WR) and rice-crayfish fields (CR). We performed an analysis of soil aggregate distribution and organic matter content. Results reveal that the majority of soil aggregates exceed 2 mm in size (≥74.94%). The integrated rice-crayfish farming model significantly enhances the presence of large soil aggregates. And these parameters such as the average weight diameter (MWD), average geometric diameter (GWD), and agglomerate stability (PAD) also increase. Moreover, it mitigates agglomerate fragmentation (WASR). However, the net increase in total soil organic matter due to the integrated farming model remains modest. Organic matter content within the agglomerates follows an initial increase followed by a decrease. The highest content occurs in the 0.25–0.5 mm grain size (D4). When examining the distribution of soil aggregates and organic matter, it becomes evident that organic matter primarily originates from grain sizes larger than 2 mm (≥71.92%). Notably, the rice-crayfish paddy field (CR) exhibits a substantially higher contribution compared to the traditional rice paddy field (WR). This study demonstrates several positive outcomes of the integrated rice-crayfish farming model compared to traditional paddy farming. It promotes the development of larger soil aggregates, enhances the structural integrity of soil aggregates, and improves their mechanical and hydrological stability. Additionally, it marginally increases the organic matter content within each component of soil aggregates. Furthermore, integrated modelling increases the impact of larger soil aggregates on soil organic matter. This improves the quality of the soil and as a result, crop yields are increased. The health of the soil is also improved and this contributes positively to food security.

Gastrodia elata (G. elata) Blume is widely used as a health product with significant economic, medicinal, and ecological values. Due to variations in the geographical origin, soil pH, and content of organic matter, the levels of physiologically active ingredient contents in G. elata from different origins may vary. Therefore, rapid methods for predicting the geographical origin and the contents of these ingredients are important for the market. This paper proposes a visible–near-infrared (Vis-NIR) spectroscopy technology combined with machine learning. A variety of machine learning models were benchmarked against a one-dimensional convolutional neural network (1D-CNN) in terms of accuracy. In the origin identification models, the 1D-CNN demonstrated excellent performance, with the F1 score being 1.0000, correctly identifying the 11 origins. In the quantitative models, the 1D-CNN outperformed the other three algorithms. For the prediction set of eight physiologically active ingredients, namely, GA, HA, PE, PB, PC, PA, GA + HA, and total, the RMSEP values were 0.2881, 0.0871, 0.3387, 0.2485, 0.0761, 0.7027, 0.3664, and 1.2965, respectively. The Rp2 values were 0.9278, 0.9321, 0.9433, 0.9094, 0.9454, 0.9282, 0.9173, and 0.9323, respectively. This study demonstrated that the 1D-CNN showed highly accurate non-linear descriptive capability. The proposed combinations of Vis-NIR spectroscopy with 1D-CNN models have significant potential in the quality evaluation of G. elata.

External interference in ionospheric sounding seriously degrades the quality of echo signals and data; thus, it should be eliminated. This paper presents a method for suppressing interference using chaotic coding with a set of Bernoulli map sequences; compared with other commonly used coding methods such as Barker code, complementary code, and Barker-like codes, through simulation, the ambiguity function (AF) of Bernoulli map codes has better performance in terms of peak sidelobe level (PSL), integral sidelobe ratio (ISL), noise suppression (NS), and signal-to-noise ratio (SNR). Experimental tests were performed using a vertical ionosonde in Yinchuan, Ningxia Hui Autonomous Region, China, and the ionosonde was operated by alternating 40-bit Barker-like coding and 40-bit Bernoulli map coding each day to compare the effectiveness of interference suppression. The results showed that using Bernoulli map coding could remove interference and improve SNR significantly, thereby improving the data quality of the resulting ionograms.

Titanium-based implants are often utilized in oral implantology and craniofacial reconstructions. However, the biological inertness of machined titanium commonly results in unsatisfactory osseointegration. To improve the osseointegration properties, we modified the titanium implants with nanotubular/superhydrophilic surfaces through anodic oxidation and thermal hydrogenation and evaluated the effects of the machined surfaces (M), nanotubular surfaces (Nano), and hydrogenated nanotubes (H-Nano) on osteogenesis and osseointegration in vitro and in vivo. After incubation of mouse bone marrow mesenchymal stem cells on the samples, we observed improved cell adhesion, alkaline phosphatase activity, osteogenesis-related gene expression, and extracellular matrix mineralization in the H-Nano group compared to the other groups. Subsequent in vivo studies indicated that H-Nano implants promoted rapid new bone regeneration and osseointegration at 4 weeks, which may be attributed to the active osteoblasts adhering to the nanotubular/superhydrophilic surfaces. Additionally, the Nano group displayed enhanced osteogenesis in vitro and in vivo at later stages, especially at 8 weeks. Therefore, we report that hydrogenated superhydrophilic nanotubes can significantly accelerate osteogenesis and osseointegration at an early stage, revealing the considerable potential of this implant modification for clinical applications.

High hydrostatic pressure (HHP, 600 MPa/15 min), pasteurization (72 °C/15 s) and pasteurization-HHP (72 °C/15 s + 600 MPa/15 min) processing of milk were comparatively evaluated by examining their effects on microorganisms and quality during 30 days of storage at 4 °C. The counts of total aerobic bacteria in HHP-treated milk were less than 2.22 lgCFU/mL during storage, while they exceeded 5.00 lgCFU/mL in other treated milk. Although HHP changed the color, it had more advantages in maintaining the nutrient (fat, calcium and β-lactoglobulin) properties of milk during storage. Moreover, the viscosity and particle size of HHP-treated milk were more similar to the untreated milk during storage. However, consumer habits towards heat-treated milk have led to poor acceptance of HHP-treated milk, resulting in a low sensory score. In sum, compared with pasteurization- and pasteurization-HHP-treated milk, HHP-treated milk showed longer shelf life and better nutritional quality, but lower sensory acceptance.

Dosage compensation is a mechanism first proposed by Susumu Ohno, whereby X inactivation balances X gene output between males (XY) and females (XX), while X upregulation balances X genes with autosomal gene output. These mechanisms have been actively studied in Drosophila and mice, but research regarding them lags behind in domestic species. It is unclear how the X chromosome is regulated in the sheep male germline. To address this, using single-cell RNA sequencing, we analyzed testes in three important developmental stages of sheep. We observed that the total RNA per cell from X and autosomes peaked in SSCs and spermatogonia and was then reduced in early spermatocytes. Furthermore, we counted the detected reads per gene in each cell type for X and autosomes. In cells experiencing dose compensation, close proximity to MSL (male-specific lethal), which is regulated the active X chromosome and was observed. Our results suggest that there is no dose compensation in the pre-meiotic germ cells of sheep testes and, in addition, MSL1 and MSL2 are expressed in early germ cells and involved in regulating mammalian X-chromosome inactivation and activation.

Pyrethrum (Tanacetum cinerariifolium) is one of the most important industrial crops for the extraction of pyrethrins, which are natural insecticidal compounds. Progress in pyrethrum molecular breeding with the objective of increasing pyrethrin content has been slow for lack of a suitable gene transfer system. Regeneration recalcitrance is a crucial barrier to establishing a genetic transformation system in pyrethrum. Therefore, in this study, an Agrobacterium-mediated transformation system in pyrethrum was developed using shoot apical meristems from germinated seedlings. Factors affecting transformation efficiency were optimized. Optimal conditions included explants at the “no true leaf” stage with a half apical meristem, an Agrobacterium tumefaciens cell density of OD600 = 0.5, two days of cocultivation, and the incorporation of 1.5 mg L−1 6-BA and 30 mg L−1 kanamycin into the selection medium. Under the optimized conditions, two expression cassettes (proTcCHS-GUS and proRbcS-TcCHS) were successfully transformed into pyrethrum. Polymerase chain reaction (PCR), Southern blotting, reverse-transcription quantitative PCR (RT-qPCR), and histochemical staining confirmed the identity of proTcCHS-GUS transgenic plants. PCR and RT-qPCR analyses confirmed the identity of proRbcS-TcCHS transgenic plants. The transformation efficiency was 0.83% (5 transgenic lines/600 infected explants). The relative concentration of pyrethrins in proRbcS-TcCHS transformants (OX T0-1: 1.50% or OX T0-2: 1.24%) was higher than that in nontransformed plants (WT: 0.76%). Thus, the genetic transformation system overcame the low regeneration efficiency and integrated a foreign gene into the pyrethrum genome. The new system is a suitable and effective tool for creating high-yielding cultivars of pyrethrum.

The short-tailed phenotype has long been considered one of the best traits for population genetic improvement in sheep breeding. In short-tailed sheep, not only is tail fat eliminated but also the pubic area is exposed due to the lack of a tail covering, giving them an advantage in reproduction. Recent studies have shown that two linked mutations in sheep TBXT at nucleotides 333 and 334 are associated with the short-tailed phenotype. In the population of short-tailed sheep, several heterozygous mutants of this gene are found. In our research, we used high-resolution melting (HRM) to identify homozygous and heterozygous genotypes in a flock of short-tailed sheep and compared the results with those of Sanger sequencing, which were identical. This demonstrates that our established HRM method, a rapid and inexpensive genotyping method, can be used to identify homozygous and heterozygous individuals in short-tailed sheep flocks.

Hydroxycamptothecin (SN38) is a natural plant extract isolated from Camptotheca acuminate. It has a broad spectrum of anticancer activity through inhibition of DNA topoisomerase I, which could affect DNA synthesis and lead to DNA damage. Thus, the action of SN38 against cancers could inevitably affect endogenous levels of ribonucleotide (RNs) and deoxyribonucleotide (dRNs) that play critical roles in many biological processes, especially in DNA synthesis and repair. However, the exact impact of SN38 on RNs and dRNs is yet to be fully elucidated. In this study, we evaluated the anticancer effect and associated mechanism of SN38 in human colorectal carcinoma HCT 116 cells. As a result, SN38 could decrease the cell viability and induce DNA damage in a concentration-dependent manner. Furthermore, cell cycle arrest and intracellular nucleotide metabolism were perturbed due to DNA damage response, of which ATP, UTP, dATP, and TTP may be the critical metabolites during the whole process. Combined with the expression of deoxyribonucleoside triphosphates synthesis enzymes, our results demonstrated that the alteration and imbalance of deoxyribonucleoside triphosphates caused by SN38 was mainly due to the de novo nucleotide synthesis at 24 h, and subsequently the salvage pathways at 48 h. The unique features of SN38 suggested that it might be recommended as an effective supplementary drug with an anticancer effect.
