In order to synthesize chemical filters for the selective removal of volatile fluorides, commercial magnesium fluoride MgF2 with high specific surface area (HSA) was investigated. The amount of -OH groups substituting fluorine is not negligible, partly due to the high surface area, but also due to the synthesis route. These hydroxyl groups induce a Lewis basicity on the surface of metal fluorides. The amount of these Lewis basic sites has been tailored using fluorination with F2 gas. The sorption of VOF3, used as model gas, onto these fluorides was investigated. The versatility of surface chemistry as a function of a number of Lewis basic sites opens the way to filter selectivity mixture of volatile fluorides depending on their Lewis acidity. HSA MgF2 acts as a stable matrix towards the gas to be purified, and the selectivity may be achieved by a higher Lewis acidity of the gaseous impurity.

Canine atopic dermatitis (CAD) is an allergic, inflammatory, and pruritic skin disease associated with the production of IgE antibodies against environmental allergens and mainly house dust mite allergens. This complex dermatological pathology involves Interleukin 31 (IL-31) as a central itch mediator. One of the most effective CAD treatments is a caninized monoclonal antibody (mAb) called Lokivetmab. It is produced in CHO cells and targets specifically canine IL-31 (cIL-31) and blocks its cellular messaging. This treatment has undoubtedly contributed to a breakthrough in dermatitis-related pruritus. However, its production in mammalian cells requires time-consuming procedures, high production costs, and investment. Plants are considered an emerging protein production platform for recombinant biopharmaceuticals due to their cost-effectiveness and rapidity for production. Here, we use transient expression in Nicotiana benthamiana plants to produce recombinant canine Interleukin 31 (cIL-31) and an anti-IL-31 monoclonal antibody (M1). First, we describe the production and characterization of M1 and then its activity on an IL-31-induced pruritic model in dogs compared to its commercial homolog. Dogs treated with the plant-made M1 mAb have shown similar improvements to Lokivetmab-treated ones after different challenges using canine IL-31. Furthermore, M1 injections were not associated with any side effects. These results demonstrate the safety and efficacy of this plant-made Lokivetmab biosimilar to control dogs’ pruritus in a well-established model. Finally, this study shows that the plant-production platform can be utilized to produce rapidly functional mAbs and bring hope to the immunotherapy field of veterinary medicine.

We investigated the influence of a fuel change from pure hydrogen to a hydrogen–ammonia mixture at different percentages on the electrochemical behavior of 50 mm in diameter Solid Oxide Fuel Cells (SOFCs) with sputtered thin buffer layers of Gd-doped ceria, varying the working temperatures from 800 °C to 650 °C. The results show that the performances of the cells are not affected by the fuel change for high working temperatures (800 °C and 750 °C). As an example, a power density value of 802 mW∙cm−2 at 1 A∙cm−2 is found when directly feeding the cell with 8 NmL∙min−1cm−2 of ammonia and with an equivalent flowrate of 12 NmL∙min−1cm−2 of H2. These power density output values are higher than those obtained in industrial state-of-art (SoA) SOFCs with screen-printed buffer layers fed with equivalent hydrogen flowrates, thanks to the improved electrochemical performances obtained in the case of cells with sputtered thin buffer layers of Gd-doped ceria. At lower working temperatures (700 °C and 650 °C), slight changes in the electrochemical behavior of the cells are observed. Nevertheless, in this temperature range, we also obtain an output current density value of 0.54 A∙cm−2 in a pure ammonia flowrate of 12 NmL min−1cm−2 at 800 mV and 700 °C, equal to the value observed in SoA button cells with industrial screen-printed GDC barrier layer fueled with 16 NmL∙min−1cm−2 of H2. These results pave the way towards the use of innovative SOFC structures with sputtered thin buffer layers fueled by ammonia.

In recent years, the hydrothermal conversion of actinide (IV) oxalates into nanometric actinide dioxides (AnO2) has begun to be investigated as an alternative to the widely implemented thermal decomposition method. We present here a comparison between the hydrothermal and the conventional thermal decomposition of Pu(IV) oxalate in terms of particle size, morphology and residual carbon content. A parametric study was carried out in order to define the temperature and time applied in the hydrothermal conversion of tetravalent Pu-oxalate into PuO2 and to optimize the reaction conditions.

A promising strategy to neutralize HIV-1 is to target the gp41 spike subunit to block membrane fusion with the cell. We previously designed a series of single-chain proteins (named covNHR) that mimic the trimeric coiled-coil structure of the gp41 N-terminal heptad repeat (NHR) region and potently inhibit HIV-1 cell infection by avidly binding the complementary C-terminal heptad repeat (CHR) region. These proteins constitute excellent tools to understand the structural and thermodynamic features of this therapeutically important interaction. Gp41, as with many coiled-coil proteins, contains in core positions of the NHR trimer several highly conserved, buried polar residues, the role of which in gp41 structure and function is unclear. Here we produced three covNHR mutants by substituting each triad of polar residues for the canonical isoleucine. The mutants preserve their helical structure and show an extremely increased thermal stability. However, increased hydrophobicity enhances their self-association. Calorimetric analyses show a marked influence of mutations on the binding thermodynamics of CHR-derived peptides. The mutations do not affect however the in vitro HIV-1 inhibitory activity of the proteins. The results support a role of buried core polar residues in maintaining structural uniqueness and promoting an energetic coupling between conformational stability and NHR–CHR binding.
