Water erosion significantly affects soil properties, yet comparative studies between different soil types are limited. This study examines the impact of erosion on the physical, chemical, and biochemical properties of Chernozems and Cambisols in the South Moravian and Vysočina regions of the Czech Republic. We combined field sampling, laboratory analysis, and geospatial modeling to identify erosion, transition, and deposition zones on slopes. Soil samples were collected from these zones in both soil types and analyzed for particle size distribution, organic carbon content, humus quality, stability of aggregates, pH, nutrient levels, enzyme activities, and glomalin content. Chernozems showed more pronounced differences between the erosion and deposition zones, particularly in organic matter content, nutrient levels, and enzyme activities. Cambisols exhibited different patterns, with less pronounced differences between the zones, except for humic substances and cellulase activity. Chernozems had higher carbonate content and pH in erosion areas, while Cambisols showed an opposite trend. These differences relate to both substrate and slope shape: concave–convex for Chernozems and convex to convex–concave for Cambisols. The findings highlight the importance of considering soil type and topography in erosion studies and have significant implications for tailored soil management and erosion control strategies in different agricultural landscapes.

This article deals with the rationalization of manufacturing processes within the product life cycle with emphasis on the pre-production phase of production. A new methodology for evaluating the applicability of modern visualization tools in manufacturing processes is presented. This methodology includes a modified Z-score for categorizing manufacturing processes and has been validated by the successful implementation of 10 real projects. Ultimately, the methodology provides a practical decision-making aid for manufacturing companies in deploying such Computer Aided Instruction tools. For the pre-production phase of products and their development, the possibilities of using modern visualization tools to support CAD instructions and assembly instructions are being explored. These modern visualization tools are video tutorials, augmented reality tutorials and virtual reality tutorials. This paper explores the use of these tools for rationalization of pre-production processes. A methodology was designed to select the most appropriate tool for rationalizing process execution in preparation for production. The functionality of the methodology was verified by applying the methodology in industrial practice and subsequent implementation of the recommended solutions. The methodology was validated by testing key combinations that can arise based on the methodology directly in the operations of manufacturing companies. A total of 10 implementations in real production processes were tailored to this study and carried out over 2 years and the functionality of the methodology was confirmed (that consisted also of a new software development). It was found that there is a dependency between the visualization tools chosen to create the instructions in the context of organizational production preparation and the nature of the production processes.

Catalase–peroxidases (KatGs) are unique bifunctional oxidoreductases that contain heme in their active centers allowing both the peroxidatic and catalatic reaction modes. These originally bacterial enzymes are broadly distributed among various fungi allowing them to cope with reactive oxygen species present in the environment or inside the cells. We used various biophysical, biochemical, and bioinformatics methods to investigate differences between catalase–peroxidases originating in thermophilic and mesophilic fungi from different habitats. Our results indicate that the architecture of the active center with a specific post-translational modification is highly similar in mesophilic and thermophilic KatG and also the peroxidatic acitivity with ABTS, guaiacol, and L-DOPA. However, only the thermophilic variant CthedisKatG reveals increased manganese peroxidase activity at elevated temperatures. The catalatic activity releasing molecular oxygen is comparable between CthedisKatG and mesophilic MagKatG1 over a broad temperature range. Two constructed point mutations in the active center were performed selectively blocking the formation of described post-translational modification in the active center. They exhibited a total loss of catalatic activity and changes in the peroxidatic activity. Our results indicate the capacity of bifunctional heme enzymes in the variable reactivity for potential biotech applications.

The agricultural landscape of the Czech Republic is facing climate change, and drought is among the most severe stress factors. Thousands of small ponds and naturally wet areas have been drained and transformed into agricultural parcels. Their restoration could increase the landscape’s resilience to climate change. Therefore, we describe the possibility of using hyperspectral aerial surveying for the identification of waterlogged areas in the agricultural landscape based on the example of one of the warmest and driest regions of the Czech Republic—the South Moravian region, an area where water retention in the landscape is highly relevant. Within our study, a total of 33 spectral indices related to the waterlogging of soil selected from previous studies were evaluated. The maximum entropy model (MAXENT) was used in the analysis of these indices. The analysis, which was carried out in several locations during different periods of the year (spring and autumn), shows the varying applicability of individual groups of indices. Regardless of the season, chlorophyll-based indices (MCARI—31.8, CARI—26.3, TCARI2—24.3 average percentage contribution) made the most significant contribution to the creation of probability maps of the occurrence of waterlogged areas. However, more accurate results could be achieved in the spring period by using the NVI index (40.5 average percentage contribution). The results show that remote sensing could be used for the identification of waterlogged sites, especially for initial identification, which should then be confirmed by field survey. Furthermore, the research points out the role of the LAI and chlorophyll content. According to the NVI, low LAI contributes the most to the probability of occurrence in the spring season, while chlorophyll-based indices prove to be the best, contributing high values, which is rather contradictory but could be resolved only by subsequent field research.

Since the year 2012, nationwide monitoring of erosion on agricultural land has been carried out in the Czech Republic with more than 2,300 cases of erosion events recorded so far. The evaluation of the relationships between the actual manifestation of erosion and the characteristics of the 5600 erosion enclosed areas (EEAs), on which surface runoff develops after erosion-forming rainfall, are presented in this contribution. Most frequently grown crops during an erosion event include maize (49.8% of cases), rapeseed (13.7%), potatoes (8.1%) and sugar beets (2.1%). The distributions of affected erosion enclosed areas (EEAs) in terms of shares of soils with low susceptibility and medium susceptibility to compaction, drainage systems and contributing areas of critical points are bimodal; the low percentage (90%). The percentage of hydrologic soil group B in EEA has the high share dominant over the low one. Unsurprisingly, erosion events have been recorded predominantly in EEAs with low forest cover and on arable land (share >90%). The distribution of EEAs across altitudes corresponds with the relief of the country. Obtained results will be used to validate the implementation of erosion-monitoring systems and for the adaptation of soil erosion prevention and mitigation measures in the sustainable land use policy.

An archaeological excavation in Prostějov (Czech Republic) revealed a workshop of a local potter with colourless, pink, and blue powders presumably used to produce faience/surface decoration. A comprehensive analytical study, which combined elemental and molecular analysis techniques, was performed to shed light on the chemical composition of these unique findings. Scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM EDX), inductively coupled-plasma mass spectrometry (ICP MS), flow injection analysis (FIA) with electrospray ionisation mass spectrometry (ESI MS), laser desorption ionisation mass spectrometry (LDI MS), and Raman spectroscopy were applied to reveal the elemental composition of the powders and identify the colouring agents in the pink and blue powders. The colouring agents in the pink powder were probably iron and the agent in the blue powder is Prussian blue. On top of that, it was also possible to determine the organic additives in these powders through pyrolysis gas chromatography with mass spectrometric detection (Py GC/MS), atmospheric solids analysis probe ion mobility mass spectrometry (ASAP IM MS), and LDI MS. The organic constituents were identified as plant resin, beeswax, and fats. These results point to the preparation of faience/pigment mixtures as oil paint.

Energy constitutes an essential share of costs across any economy. The percentage of electricity in the overall energy consumption is steadily increasing. This increase, however, is not reflected correctly in the consumption basket relevant for measuring inflation and, therefore, for monetary policy formulation. We argue that the energy mix reflected in inflation should be revised in favour of electricity. We present an analysis of inflationary pressures across Europe and decompose the impact of energy categories on headline inflation. Building on the inflation expectations framework, this study examines the characteristics and magnitude of the current energy price dynamics and quantifies its share in the countries’ effective inflation. Our research also confirms a compelling insight into the country’s energy structure and inflationary pressures when a larger share of renewable electricity sources proves to be associated with lower inflation. Finally, we argue that the energy price shock cannot be viewed as a one-shot event as in the case of oil price shocks in the past. We draw recommendations for monetary policy formulation. The implication of renewable sources on inflation should be of interest to policymakers, especially in times of high, almost galloping inflation rates in some European countries, unstable fossil energy sources supply due to geopolitical instability, and climate crisis.

With the arrival of new technologies in modern smart factories, automated predictive maintenance is also related to production robotisation. Intelligent sensors make it possible to obtain an ever-increasing amount of data, which must be analysed efficiently and effectively to support increasingly complex systems’ decision-making and management. The paper aims to review the current literature concerning predictive maintenance and intelligent sensors in smart factories. We focused on contemporary trends to provide an overview of future research challenges and classification. The paper used burst analysis, systematic review methodology, co-occurrence analysis of keywords, and cluster analysis. The results show the increasing number of papers related to key researched concepts. The importance of predictive maintenance is growing over time in relation to Industry 4.0 technologies. We proposed Smart and Intelligent Predictive Maintenance (SIPM) based on the full-text analysis of relevant papers. The paper’s main contribution is the summary and overview of current trends in intelligent sensors used for predictive maintenance in smart factories.

Soil is negatively affected by many degradation factors, of which soil erosion is the most serious, affecting soil quality, crop production, and environmental components. Soil quality is an issue dealt with in the New European Green Deal. In order to meet the set goals, it will be necessary to address soil degradation and water erosion in the agricultural landscape, and increase the area of green infrastructure within the landscape (e.g., fragments of woodland, windbreaks, and grassland). In this context, climate change is also expected to affect the frequency and intensity of torrential rainfall, leading to increased runoff, reduced infiltration, and greater soil loss. Therefore, in this study, we have elaborated the issue of agricultural landscape and erosion, looking at erosion control measures necessary in dealing with existing erosion processes in an intensively farmed area with chernozem soils, and compared these with scenarios assumed for 2050. In these future scenarios, the commonly applied agrotechnical measures will not suffice to keep soil loss at a tolerable level. In the future, it will be necessary to discuss a further reduction in the size of land blocks, with the inclusion of green infrastructure in the landscape. In addition to solving problems of erosion, this would increase diversity in the area and enable sustainable agricultural management.

In this study, we focus on a detailed bioinformatics analysis of hyBpox genes, mainly within the genomes of Sclerotiniaceae (Ascomycota, Leotiomycetes), which is a specifically evolved fungal family of necrotrophic host generalists and saprophytic or biotrophic host specialists. Members of the genus Sclerotium produce only sclerotia and no fruiting bodies or spores. Thus, their physiological role for peroxidases remains open. A representative species, S. cepivorum, is a dangerous plant pathogen causing white rot in Allium species, particularly in onions, leeks, and garlic. On a worldwide basis, the white rot caused by this soil-borne fungus is apparently the most serious threat to Allium-crop production. We have also found very similar peroxidase sequences in the related fungus S. sclerotiorum, although with minor yet important modifications in the architecture of its active centre. The presence of ScephyBpox1-specific mRNA was confirmed by transcriptomic analysis. The presence of Hybrid B peroxidase at the protein level as the sole extracellular peroxidase of this fungus was confirmed in the secretome of S. cepivorum through detailed proteomic analyses. This prompted us to systematically search for all available genes coding for Hybrid B heme peroxidases in the whole fungal family of Sclerotiniaceae. We present here a reconstruction of their molecular phylogeny and analyse the unique aspects of their conserved-sequence features and structural folds in corresponding ancestral sequences.

In this article, we discuss the influence of soil erosion on crop yield in the erosion-prone chernozem region of South Moravia. Erosional and depositional areas show significant differences in soil properties, which are also reflected in total crop yield. Plots of winter wheat, grown during the years 2016–2019 were used for analysis. The Enhanced Vegetation Index (EVI), referred to in literature as one of the best correlates of yield, was used to provide indirect information on yield. Although erosional areas are visible on orthophoto images on chernozem soils, the necessary orthophoto images are not always available. Thus, we have proposed a method for the identification of such erosion-affected areas based on the use of Sentinel 2 satellite images and NDVI or NBR2 indices. The relationship between yield and erosion was expressed through Pearson’s correlation on a sample of pixels randomly selected on the studied plots. The results showed a statistically significant linear reduction in yield depending on the level of degradation. All plots were further reclassified, according to level of degradation, as high, medium, or low state of degradation, where the average EVI values were subsequently calculated. Yield on non-degraded soil is 16 ± 1% higher on average.

The antifungal and insecticidal effect of the essential oil from Ocimum sanctum L. was evaluated using a model set of harmful organisms hazardous for health and the economy. Toxigenic and plant pathogenic filamentous fungi, including causal agents of human infections, were chosen as exemplary fungal groups—Fusarium verticillioides, Penicillium expansum and Aspergillus flavus. Spodoptera littoralis (African cotton leafworm), Culex quinquefasciatus (Southern house mosquito), the lymphatic filariasis vector and potential Zika virus vector, and the common housefly, Musca domestica were chosen as model insects. Major and minor active substances were detected and quantified using GC/MS analysis. Environmental safety was verified using the non-target useful organism Eisenia fetida. Significant antifungal and insecticidal activity, as well as environmental safety, were confirmed. The essential oil showed the highest efficacy against A. flavus according to MIC50/90, and against S. littoralis larvae according to LD50/90. The monoterpenoid alcohol linalool, t-methyl cinnamate, and estragole as phenylpropanoids were detected as effective major components (85.4%). The essential oil from Ocimum sanctum L. was evaluated as universal and significantly efficient, providing a high potential for use in environmentally safe botanical pesticides.

The Late Neolithic palafitte site, Ustie na Drim, in the northern part of Lake Ohrid (North Macedonia), excavated in 1962, offered ceramic fragments of large, flat, elongated pans. These artifacts could be dated by relative chronology to roughly around 5200–5000 BC. According to their shape and technological traits, the ceramic pans were probably used for baking. The attached materials on the surface of studied pan fragments were sampled for consequent chemical and microscopical analyses (i.e., analyses of starch, phytoliths, and microscopic animal remains). An immunological method revealed the presence of pork proteins in samples. The presence of organic residues of animal origin was, moreover, confirmed by the detection of cholesterol using gas chromatography coupled to mass spectrometry. Analysis of detected microscopic botanical objects revealed starch grains of several plants (i.e., oak, cattail, and grasses). An interesting find was the hair of a beetle larva, which could be interpreted contextually as the khapra beetle, a pest of grain and flour. Based on our data, we suppose that the ceramic pans from Ustie na Drim were used for the preparation of meals containing meat from common livestock in combination with cereals and wild plants.

Geographical and botanical origin of honeys can be characterized on the basis of physico-chemical composition, sensory properties and on the basis of melissopalynological analysis. No comprehensive description of the characteristics of Czech honey has been published so far. This study provides insights that are important for correct classification. The study analysed 317 samples of authentic honey from randomly selected localities. Due to the diversity of the landscape, the typical honey of the region is blend honey with a predominance of blossom honey. According to the pollen profile and electric conductivity, the honeys were sorted into the following: Brassica honey (BH), Floral honey (FH), Fruit tree honey (PH), Honeydew (HD), Lime tree honey (LH), Robinia pseudoacacia honey (RH), and Trifolium honey (TH). Physico-chemical properties, including higher carbohydrates, were determined for the honeys and their pollen profiles were examined. The physico-chemical properties and pollen profile are partially in compliance with the description of European monofloral honeys, except for RH and TH. Although they had the highest proportion of acacia pollen, amounting to >10% of all the Czech honeys, these RH honeys differ from the European standard, so they cannot be considered acacia honey. Further, PH honeys and FH polyfloral honeys were described. Most honeys contained a significant proportion of rapeseed pollen, which is one of the common agricultural crops grown in the Czech Republic. All the analysed honeys met the parameters defined by the legislation. Due to direct on-site sampling, honeys were characterized by a low 5-(hydroxymethyl)furfural (HMF) content (3.0 mg/kg) and high diastase activity (24.4 DN). Honeydew honeys had the highest proportion of higher carbohydrates, primarily of Melezitose (4.8 g/100 g) and Trehalose (1.3 g/100 g). The presence of higher carbohydrates was also confirmed in LH for Maltose (4.6 g/100 g) and Turanose (2.4 g/100 g).

The aim of this work was to find an effective protocol for in vitro propagation and to perform the in vitro polyploidization of diploid Thymus vulgaris (2n = 30) using two experimental methods based on the use of oryzalin, an antimitotic agent. The ploidy level of the obtained shoots was checked by flow cytometric analysis. The most efficient conditions for inducing polyploidy were oryzalin concentrations of 0.346 and 1.73 mg L−1 present in the medium for two weeks. The vital polyploid shoots were multiplied for further evaluation, rooting and final transfer to nonsterile glasshouse and field conditions. The chemical compositions of the essential oils (EOs)—which were obtained from dried field grown plants by steam distillation—were analyzed by gas chromatography/mass spectrometry (GC/MS). The identified substances contributed approximately 95% to the total peak area. Statistical analysis revealed that the tetraploid subclone and the diploid reference plant do not differ in total terpene content, but they do differ in the relative proportions of all the individual terpenes with the exception of α-pinene and UN5, indicating that both clones produce EOs of different quality. The obtained results showed the possibility of developing more efficient botanical insecticides based on EOs obtained from the tetraploid plants.

Cannabis sativa L. is an herbaceous plant belonging to the family of Cannabaceae. It is classified into three different chemotypes based on the different cannabinoids profile. In particular, fiber-type cannabis (hemp) is rich in cannabidiol (CBD) content. In the present work, a rapid nano liquid chromatographic method (nano-LC) was proposed for the determination of the main cannabinoids in Cannabis sativa L. (hemp) inflorescences belonging to different varieties. The nano-LC experiments were carried out in a 100 µm internal diameter capillary column packed with a C18 stationary phase for 15 cm with a mobile phase composed of ACN/H2O/formic acid, 80/19/1% (v/v/v). The reverse-phase nano-LC method allowed the complete separation of four standard cannabinoids in less than 12 min under isocratic elution mode. The nano-LC method coupled to ultraviolet (UV) detection was validated and applied to the quantification of the target analytes in cannabis extracts. The nano-LC system was also coupled to an electrospray ionization–mass spectrometry (ESI-MS) detector to confirm the identity of the cannabinoids present in hemp samples. For the extraction of the cannabinoids, three different approaches, including dynamic maceration (DM), ultrasound-assisted extraction (UAE), and an extraction procedure adapted from the French Pharmacopeia’s protocol on medicinal plants, were carried out, and the results achieved were compared.

Degradation of anthocyanins involves scission of the flavonoid skeleton yielding 2,4,6-trihydroxybenzaldehyde (phloroglucinaldehyde, PGA) and a phenolic acid. However, the process is not finished with the formation of PGA, as the consequent condensation of two PGA molecules providing colored hydroxylated anthraquinones was observed for the first time. This process was studied using a combination of preparative column chromatography, nuclear magnetic resonance, liquid chromatography/high resolution tandem mass spectrometry (LC/HRMS2), and quantum calculations using density functional theory. 1,3,5,7-tetrahydroxyanthraquinone (anthrachrysone) and its isomers were found to rise during heating (95 °C) in a buffered PGA model solution (phosphate buffer, pH 7). These compounds were detected in heated red wine after an increase of its pH value. The concentration of the identified anthrachrysone in the red wine reached 0.01 mg·L−1. Presence of those compounds could therefore indicate involvement of certain steps in the processing of plant materials rich in anthocyanins (e.g., utilization of a higher temperature and/or reduction of acidity) or long-term transformation of anthocyanins (potentially, for instance, in archaeological findings such as wine or fruit residues). Additionally, measurement of wine–soil suspensions proved an increase of their pH to the values suitable for anthocyanin cleavage (neutral to slightly alkaline; reached using soil from archaeologically well-known Bull Rock Cave). Although not found in artificially prepared samples (imitations) or authentic materials so far, according to our results the above mentioned conditions are suitable for the formation of tetrahydroxylated anthraquinone derivatives and their monitoring would be beneficial.

Antimicrobial materials are widely used for inhibition of microorganisms in the environment. It has been established that bacterial growth can be restrained by silver nanoparticles. Combining these with other antimicrobial agents, such as ZnO, may increase the antimicrobial activity and the use of carrier substrate makes the material easier to handle. In the paper, we present an antimicrobial nanocomposite based on silver nanoparticles nucleated in general silicate nanostructure ZnO·mSiO2. First, we prepared the silicate fine net nanostructure ZnO·mSiO2 with zinc content up to 30 wt% by precipitation of sodium water glass in zinc acetate solution. Silver nanoparticles were then formed within the material by photoreduction of AgNO3 on photoactive ZnO. This resulted into an Ag-ZnO·mSiO2 composite with silica gel-like morphology and the specific surface area of 250 m2/g. The composite, alongside with pure AgNO3 and clear ZnO·mSiO2, were successfully tested for antimicrobial activity on both gram-positive and gram-negative bacterial strains and yeast Candida albicans. With respect to the silver content, the minimal inhibition concentration of Ag-ZnO·mSiO2 was worse than AgNO3 only for gram-negative strains. Moreover, we found a positive synergistic antimicrobial effect between Ag and Zn agents. These properties create an efficient and easily applicable antimicrobial material in the form of powder.

In this study, a soil from two ceramic vessels belonging to Corded Ware culture, 2707⁻2571 B.C., found in a cremation grave discovered in Central Moravia, Czech Republic, was analyzed using matrix-assisted laser desorption/ionization⁻mass spectrometry (MALDI⁻MS) combined with advanced statistical treatment (principal component analysis, PCA, and orthogonal projection to latent structures discriminant analysis, OPLS-DA) and by enzyme-linked immunosorbent assay (ELISA). MALDI⁻MS revealed the presence of triacylglycerols in both vessels. This analytical technique was used for the analysis of the soil content from archaeological ceramic vessels for the first time. Targeted ELISA experiments consequently proved the presence of milk proteins in both ceramic vessels. These results represent the first direct evidence of the use of milk or dairy products in the Eneolithic period in Moravian Corded Ware Culture and help to better understand the diet habits and living conditions of Eneolithic populations in Central Europe.

Ultra-wide-field of view (UWFOV) imaging systems are affected by various aberrations, most of which are highly angle-dependent. A description of UWFOV imaging systems, such as microscopy optics, security camera systems and other special space-variant imaging systems, is a difficult task that can be achieved by estimating the Point Spread Function (PSF) of the system. This paper proposes a novel method for modeling the space-variant PSF of an imaging system using the Zernike polynomials wavefront description. The PSF estimation algorithm is based on obtaining field-dependent expansion coefficients of the Zernike polynomials by fitting real image data of the analyzed imaging system using an iterative approach in an initial estimate of the fitting parameters to ensure convergence robustness. The method is promising as an alternative to the standard approach based on Shack–Hartmann interferometry, since the estimate of the aberration coefficients is processed directly in the image plane. This approach is tested on simulated and laboratory-acquired image data that generally show good agreement. The resulting data are compared with the results of other modeling methods. The proposed PSF estimation method provides around 5% accuracy of the optical system model.
