Pyrrolizidine alkaloids (PAs) are toxic compounds that occur naturally in certain plants, however, there are many secondary pathways causing PA contamination of other plants, including medicinal herbs and plant-based food products, which pose a risk of human intoxication. It is proven that chronic exposure to PAs causes serious adverse health consequences resulting from their cytotoxicity and genotoxicity. This review briefly presents PA occurrence, structures, chemistry, and toxicity, as well as a set of analytical methods. Recently developed sensitive electrochemical and chromatographic methods for the determination of PAs in honey, teas, herbs, and spices were summarized. The main strategies for improving the analytical efficiency of PA determination are related to the use of mass spectrometric (MS) detection; therefore, this review focuses on advances in MS-based methods. Raising awareness of the potential health risks associated with the presence of PAs in food and herbal medicines requires ongoing research in this area, including the development of sensitive methods for PA determination and rigorous legal regulations of PA intake from herbal products. The maximum levels of PAs in certain products are regulated by the European Commission; however, the precise knowledge about which products contain trace but significant amounts of these alkaloids is still insufficient.

Rifampicin is an essential medicine for treating and preventing tuberculosis (TB). TB is a life-threatening infectious disease and its prevention and treatment are public health imperatives. In the time of a global crisis of nitrosamine contamination of medicinal products, patient safety and a reduction in the number of drug recalls at the same time are crucial. In this work, the LC-MS/MS method was developed for the determination of the 1-methyl-4-nitrosospiperazine (MNP), a genotoxic nitrosamine impurity in various products containing rifampicin at a 5.0 ppm limit level according to Food and Drug Administration (FDA). Extraction with neutralization was necessary due to the matrix and solvent effect associated with the complexity of the rifampicin product. The developed method was validated in accordance with regulatory guidelines. Specificity, accuracy, precision, limit of detection, and limit of quantification parameters were evaluated. The recovery of the MNP was 100.38 ± 3.24% and the intermediate precision was 2.52%. The contamination of MNP in Rifampicin originates in the manufacturing process of the drug. Furthermore, the results of the forced degradation experiments show that the formation of MNP is possible by two mechanisms: through degradation of rifampicin and the oxidation of 1-amino-4-methyl-piperazine. This article points out that it is necessary to monitor and describe degradation products and the mechanism of degradation of potentially affected active pharmaceutical ingredient (API) with respect to the formation of nitrosamines during stress testing, as it was done in the following work for rifampicin in multicomponent products.

Over the last decade, an increase in the use of locally available, recycled, and waste materials as growing media components have occurred in various regions of the world in extensive green roof technology. For eco-concept reasons, such a strategy appears to be appropriate, but can be problematic due to difficulties in obtaining proper parameters of growing substrate. The growing media should be properly engineered in order to enable the proper functioning of green roofs and provide suitable environment for ideal root growth. The aim of the study was to assess the utility of locally occurring waste materials for growing media composition and estimate plant- and time-dependent changes in the physico-chemical parameters of waste-based substrates in a simulated extensive green roof system during a two-year Sedum acre L. cultivation. Five different substrate compositions were prepared using silica waste, crushed brick, Ca- and Zn-aggregates, melaphyre, tuff, sand, muck soil, urban compost, spent mushroom, and coconut fibres. Optimal water capacity, particle-size distribution, pH and salts concentration were found in all substrates. A higher concentration of macronutrients (N, P, K, Mg) and trace elements (B, Cu, Fe, Mn, Zn, Cd, Ni, Pb, and Cr) was found in waste-based substrates than in the commercial medium. In comparison to the parameters determined before establish the experiment, bulk density of tested growing media decreased, except for the substrates where the source of organic matter was the rapidly mineralising spent mushroom. The organic matter content in substrates after the two-year vegetation increased in relation to the ready-made substrate, with the exception of the composition with spent mushroom. After two years of the experiment, all available macronutrients and trace elements (with the exception of mineral N, K, SO4-S, and B) concentration were higher than in 2014, while pH, salt concentration was lower. In general, plants grown in waste substrates had lower dry matter content and higher biomass. A significantly higher biomass of S. acre L. was found in the first year of the experiment. In the second year of the research, the plants grown in the commercial medium, the substrate with silica waste, and the substrate with spent mushroom produced higher biomass than in the first year. No symptoms of abnormal growth were observed, despite the higher trace element concentrations in plants collected from waste-based substrate. Waste-based growing media can be considered as a valuable root environment for S. acre L. in an extensive green roof system.

The common ice plant (Mesembryanthemum crystallinum L.) is a widely studied model due to its tolerance to numerous biotic and abiotic stresses. In this study, carried out in model pots, the plants were treated with variant doses of Cd(II) and Cr(VI) and proved resistant to extreme levels of these heavy metals. Initial toxicity symptoms were observed upon final concentrations of 818 mg Cd kg−1 soil d.w., and 1699 mg Cr kg−1 applied as potassium chromate. Biometric analyses revealed that none of the Cr(VI) doses affected dry weight of the plant organs thus maintaining the shoot-to-root ratio. The Cd and Cr hypertolerance strategies were divergent and resulted in different accumulation patterns. For the case of Cd(II), an excluder-like mechanism was developed to prevent the plant from toxicity. For chromate, high accumulation potential together with Cr(VI) root-to-shoot translocation at sublethal concentrations was revealed (up to 6152 mg Cr kg−1 shoot at 4248 mg Cr kg−1 soil). It is concluded that M. crystallinum reveals considerable phytoremediation capabilities due to unique growth potential in contaminated substrates and is suitable for bioreclamation of degraded soils. The plant is especially applicable for efficient phytoextraction of chromate-contamination, whereas for Cd-affected areas it may have a phytostabilizing effect.

Polymer-drug conjugates are currently being more widely investigated for the treatment of hypertension. In view of the above, in the first stage of our work, we used nontoxic β-cyclodextrin (β-CD) as effective, simple, inexpensive, and safe for the human body initiator for the synthesis of biocompatible and biodegradable functionalized polymers suitable for the medical and pharmaceutical applications. The obtained polymeric products were synthesized through a ring-opening polymerization (ROP) of ε-caprolactone (CL), d,l-, and l,l-lactide (LA and LLA). The chemical structures of synthesized materials were elucidated based on 1H NMR and solid-state carbon-13 cross-polarization/magic angle spinning nuclear magnetic resonance (13C CP/MAS NMR) analysis, while the incorporation of β-CD molecule into the polymer chain was confirmed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Furthermore, molecular modeling has been applied to investigate the intrachain rigidities and chain architectures for several representative structures. The obtained and thoroughly characterized branched matrices were then used to generate the first β-cyclodextrin/biodegradable polymer/β-blocker conjugate through the successful conjugation of pindolol. The conjugates were fabricated by carbodiimide-mediated coupling reaction. The branched biodegradable materials released the drug in vitro in a sustained manner and without “burst release” and thus have the ability to treat different heart diseases.

In many areas of image processing, we deal with focused images. Indeed, the most important object is focused and the background is smooth. Finding edges in such images is difficult, since state-of-the-art edge detection methods assume that edges should be sharp. In this way, smooth edges are not detected. Therefore, these methods can detect the main object edges that skip the background. However, we are often also interested in detecting the background as well. Therefore, in this paper, we propose an edge detection method that can efficiently detect the edges of both a focused object and a smooth background alike. The proposed method is based on the local use of the k-Means algorithm from Machine Learning (ML). The local use is introduced by the proposed enhanced image filtering. The k-Means algorithm is applied within a sliding window in such a way that, as a result of filtering, we obtain a given square image area instead of just a simple pixel like in classical filtering. The results of the proposed edge detection method were compared with the best represented methods of different approaches of edge detection like pointwise, geometrical, and ML-based ones.

The results of research on the efficiency and technological reliability of domestic wastewater purification in two household wastewater treatment plants (WWTPs) with activated sludge are presented in this paper. The studied facilities were located in the territory of the Roztocze National Park (Poland). The mean wastewater flow rate in the WWTPs was 1.0 and 1.6 m3/day. In 2017–2019, 20 series of analyses were done, and 40 wastewater samples were taken. On the basis of the received results, the efficiency of basic pollutant removal was determined. The technological reliability of the tested facilities was specified using the Weibull method. The average removal efficiencies for the biochemical oxygen demand in 5 days (BOD5) and chemical oxygen demand (COD) were 66–83% and 62–65%, respectively. Much lower effects were obtained for total suspended solids (TSS) and amounted to 17–48%, while the efficiency of total phosphorus (TP) and total nitrogen (TN) removal did not exceed 34%. The analyzed systems were characterized by the reliability of TSS, BOD5, and COD removal at the level of 76–96%. However, the reliability of TN and TP elimination was less than 5%. Thus, in the case of biogenic compounds, the analyzed systems did not guarantee that the quality of treated wastewater would meet the requirements of the Polish law during any period of operation. This disqualifies the discussed technological solution in terms of its wide application in protected areas and near lakes, where the requirements for nitrogen and phosphorus removal are high.

In this paper, the research methodology and the results of the analysis carried out using the acoustic emission (AE) and thermal imaging for a selected oil power transformer are presented. The basis for the research, by means of the AE method, was the author’s patented research method. The AE descriptor maps on the side walls of the tested transformer along with the location of areas with increased AE activity and an analysis of the properties of AE signals recorded at the measurement points located in these areas have been performed. The results showed no partial discharges that could threaten further operation of the tested transformer as well as three areas where increased magnetoacoustic emission occurred. Thermal imaging studies were carried out in the 7.5 μm < λ < 13 μm band. Three areas were located on the calculated thermograms: the entire upper surface of the transformer tank and two areas on the side walls of the tested transformer in which increased IR radiation occurred. The results of the analysis of the research results for the two methods correspond with each other, having a common part, and complement each other giving a broader description of studied phenomena.
