African swine fever virus (ASFV) poses a significant threat to the global pig industry, necessitating accurate and efficient diagnostic methods for its infection. Previous studies have often focused on a limited number of epitopes from a few proteins for detecting antibodies against ASFV. Therefore, the current study aimed to use multiple B-cell epitopes in developing an indirect Enzyme-Linked Immunosorbent Assay (ELISA) for enhanced detection of ASFV antibodies. For the expression of recombinant protein, k3 derived from 27 multiple peptides of 11 ASFV proteins, such as p72, pA104R, pB602L, p12, p14.5, p49, pE248R, p30, p54, pp62, and pp220, was used. To confirm the expression of the recombinant protein, we used the Western blotting analysis. The purified recombinant K3 protein served as the antigen in our study, and we employed the indirect ELISA technique to detect anti-ASFV antibodies. The present finding showed that there was no cross-reactivity with antibodies targeting Foot-and-mouth disease virus (FMDV), Porcine circovirus type 2 (PCV2), Pseudorabies virus (PRV), Porcine reproductive and respiratory syndrome virus (PRRSV), and Classical swine fever virus (CSFV). Moreover, the current finding was sensitive enough to find anti-ASFV in serum samples that had been diluted up to 32 times. The test (k3-iELISA) showed diagnostic specificity and sensitivity of 98.41% and 97.40%, respectively. Moreover, during the present investigation, we compared the Ingenasa kit and the k3-iELISA to test clinical pig serum, and the results revealed that there was 99.00% agreement between the two tests, showing good detection capability of the k3-iELISA method. Hence, the current finding showed that the ELISA kit we developed can be used for the rapid detection of ASFV antibodies and used as an alternative during serological investigation of ASF in endemic areas. Volatile accumulation during tomato ripening greatly affects the fruit flavor. In this study, four accessions from each of the three tomato subgroups (BIG, S. lycopersicum, CER, S. lycopersicumvar. Cerasiforme, and PIM, S. pimpinellifolium) were subjected to a sensory evaluation. The CER subgroup had the highest fruit-flavor score. Using a Headspace solid-phase microextraction/gas chromatography-mass spectrometer (HP-SPME/GC-MS), a volatile database containing 94 volatiles was created. Pentanal accumulated in green fruits and 1-pentanol in red fruits. 1-Octen-3-ol was discovered to underlie the bitterness of green tomatoes, and it was most abundant in PIM green fruits. Phenylethyl alcohol affected the acidity and sweetness of red tomatoes, and it was most abundant in CER red fruits. Branched-chain volatiles were most abundant in PIM and BIG red fruits, while apocarotenoids were most abundant in CER red fruits. These findings suggest that domestication and improvement have influenced volatile content, and apocarotenoids and branched-chain volatiles synergistically mediated aromatic flavors in red fruits. The diversity of anthocyanins is largely due to the action of glycosyltransferases, which add sugar moieties to anthocyanidins. Although a number of glycosyltransferases have been identified to glycosylate anthocyanidin in plants, the enzyme that catalyzes malvidin galactosylation remains unclear. In this study, we identified three rice varieties with different leaf color patterns, different anthocyanin accumulation patterns, and different expression patterns of anthocyanin biosynthesis genes (ABGs) to explore uridine diphosphate (UDP)-glycosyltransferases (UGTs) responsible for biosynthesis of galactosylated malvidin. Based on correlation analysis of transcriptome data, nine candidate UGT genes coexpressed with 12 ABGs were identified (r values range from 0.27 to 1.00). Further analysis showed that the expression levels of one candidate gene, OsUGT88C3, were highly correlated with the contents of malvidin 3-O-galactoside, and recombinant OsUGT88C3 catalyzed production of malvidin 3-O-galactoside using UDP-galactose and malvidin as substrates. OsUGT88C3 was closely related to UGTs with flavone and flavonol glycosylation activities in phylogeny. Its plant secondary product glycosyltransferase (PSPG) motif ended with glutamine. Haplotype analysis suggeste d that the malvidin galactosylation function of OsUGT88C3 was conserved among most of the rice germplasms.  OsUGT88C3 was highly expressed in the leaf, pistil, and embryo, and its protein was located in the endoplasmic reticulum and nucleus. Our findings indicate that OsUGT88C3 is responsible for the biosynthesis of malvidin 3-O-galactoside in rice and provide insight into the biosynthesis of anthocyanin in plants. Leaf area index (LAI) stands as a pivotal parameter for the quantitative assessment of vegetation growth dynamics, and the rapid acquisition of the effective leaf area index (LAIe) in different scales is crucial for forest ecological monitoring. In this study, forest structure parameters were derived from fusion point cloud data obtained through Airborne Laser Scanning and Terrestrial Laser Scanning in three coniferous forests. The influence of point diameter on the extraction of different forest structure parameters was examined, and an in-depth analysis of the correlations between these parameters and measured LAIe was undertaken. The LAIe inversion model was constructed, and its performance for different forest types was studied. The results show that the precision of the extracted forest structure parameters was highest when the point diameter was set to 0.1 cm. Among the 10 forest structure parameters, internal canopy structures such as canopy openness (CO), gap fraction (GF) and canopy closure (CC) were significantly correlated with measured LAIe (p < 0.01), and the correlations between different forest types were significantly different. In addition, the multiparameter LAIe inversion model was able to distinguish forest type and thus better stimulate measured LAIe; also, it appeared closer to the 1:1 relationship line than the voxel model. This study made up for the inefficiency of LAIe measurement with optical instruments and the inaccuracy of passive remote sensing measurement and proved the possibility of LAIe extraction at a large scale via LiDAR in the future. This paper introduces a novel distributionally robust mean-variance portfolio estimator based on the projection robust Wasserstein (PRW) distance. This approach addresses the issue of increasing conservatism of portfolio allocation strategies due to high-dimensional data. Our simulation results show the robustness of the PRW-based estimator in the presence of noisy data and its ability to achieve a higher Sharpe ratio than regular Wasserstein distances when dealing with a large number of assets. Our empirical study also demonstrates that the proposed portfolio estimator outperforms classic "plug-in" methods using various covariance estimators in terms of risk when evaluated out of sample. The rapid urbanization process and high-intensity construction mode have greatly changed the underlying surface structure and spatial distribution of the natural land surface, further amplified the possibility of urban floods, and made urban security face more serious threats. Urban forest could help to mitigate urban floods through water holding and interception by its unique structures, especially the litter layer. This paper compared the ability of different forest tree species on urban floods mitigation, through analyzing their litter accumulation, litter water holding characteristics, and water interception features of different decomposed layers. The results concluded that Quercus mongolica Fisch. ex Ledeb. (QM) forest, Betula platyphylla Sukaczev (BP) forest, Larix gmelinii (Rupr.) Kuzen. (LG) forest, and Picea koraiensis Nakai (PK) forest were the best choices for improving urban floods resistance in a high-urbanization winter city, for they had larger litter mass and higher maximum water holding and interception capacity. Rice (Oryza sativa L.) is one of the most globally important crops, nutritionally and economically. Therefore, analyzing the genetic basis of its nutritional quality is a paramount prerequisite for cultivating new varieties with increased nutritional health. To systematically compare the nutritional quality differences between landraces and cultivated rice, and to mine key genes that determine the specific nutritional traits of landraces, a seed metabolome database of 985 nutritional metabolites covering amino acids, flavonoids, anthocyanins, and vitamins by a widely targeted metabolomic approach with 114 rice varieties (35 landraces and 79 cultivars) was established. To further reveal the molecular mechanism of the metabolic differences in landrace and cultivated rice seeds, four cultivars and six landrace seeds were selected for transcriptome and metabolome analysis during germination, respectively. The integrated analysis compared the metabolic profiles and transcriptomes of different types of rice, identifying 358 differentially accumulated metabolites (DAMs) and 1982 differentially expressed genes (DEGs), establishing a metabolite–gene correlation network. A PCA revealed anthocyanins, flavonoids, and lipids as the central differential nutritional metabolites between landraces and cultivated rice. The metabolite–gene correlation network was used to screen out 20 candidate genes postulated to be involved in the structural modification of anthocyanins. Five glycosyltransferases were verified to catalyze the glycosylation of anthocyanins by in vitro enzyme activity experiments. At the same time, the different mechanisms of the anthocyanin synthesis pathway and structural diversity in landrace and cultivated rice were systematically analyzed, providing new insights for the improvement and utilization of the nutritional quality of rice landrace varieties. Pseudouridine (Ψ), the isomer of uridine (U), is the most abundant type of RNA modification, which is crucial for gene regulation in various cellular processes. Pseudouridine synthases (PUSs) are the key enzymes for the U-to-Ψ conversion. However, little is known about the genome-wide features and biological function of plant PUSs. In this study, we identified 20 AtPUSs and 22 ZmPUSs from Arabidopsis and maize (Zea mays), respectively. Our phylogenetic analysis indicated that both AtPUSs and ZmPUSs could be clustered into six known subfamilies: RluA, RsuA, TruA, TruB, PUS10, and TruD. RluA subfamily is the largest subfamily in both Arabidopsis and maize. It's noteworthy that except the canonical XXHRLD-type RluAs, another three conserved RluA variants, including XXNRLD-, XXHQID-, and XXHRLG-type were also identified in those key nodes of vascular plants. Subcellular localization analysis of representative AtPUSs and ZmPUSs in each subfamily revealed that PUS proteins were localized in different organelles including nucleus, cytoplasm and chloroplasts. Transcriptional expression analysis indicated that AtPUSs and ZmPUSs were differentially expressed in various tissues and diversely responsive to abiotic stresses, especially suggesting their potential roles in response to heat and salt stresses. As an important improvement to naive Bayes, Tree-Augmented Naive Bayes (TAN) exhibits excellent classification performance and efficiency since it allows that every attribute depends on at most one other attribute in addition to the class variable. However, its performance might be lowered as some attributes might be redundant. In this paper, we propose an attribute Selective Tree-Augmented Naive Bayes (STAN) algorithm which builds a sequence of approximate models each involving only the top certain attributes and searches the model to minimize the cross validation risk. Five different approaches to ranking the attributes have been explored. As the models can be evaluated simultaneously in one pass learning through the data, it is efficient and can avoid local optima in the model space. The aberrant activation of a signal transducer and activator of transcription 3 (STAT3) restrains type I interferon (IFN) α/β-induced antiviral responses and is associated with the development of cancer. Designing specific STAT3 inhibitors will thus provide new options for use as IFN therapy. Herein, we identified a novel small molecule, dimethyl 2-(4-(2-(methyl(phenyl(p-tolyl)methyl)amino)ethoxy)benzyl)malonate (CIB-6), which can inhibit the IFN-α-induced interferon stimulated response element (ISRE) luciferase reporter (IC50 value = 6.4 μM) and potentiate the antiproliferative effect of IFN-α in human hepatocellular carcinoma (HCC) cells. CIB-6 was found to bind to the STAT3 Src homology 2 (SH2) domain, thereby selectively inhibiting STAT3 phosphorylation without affecting Janus kinases and STAT1/2. CIB-6 also inhibited the migration and invasion of HCC cells by inhibiting the epithelial–mesenchymal transition (EMT) process. Mechanistically, CIB-6 reduced the expression of β-catenin (an EMT key protein) via upregulating β-transducin repeat-containing protein (β-TrCP) and curbed nuclear factor kappa-B (NF-κB) activation through restricting the phosphorylation of the inhibitor of NF-κB (IκB) kinase (IKK) via STAT3 inhibition. Treatment with CIB-6 significantly retarded tumor growth in nude mice with SK-HEP-1 xenografts. In addition, clinical sample analysis revealed that lower β-TrCP and higher β-catenin expression could affect the median survival time of HCC patients. Our findings suggest that CIB-6 could be a new therapeutic strategy for HCC therapy through STAT3-mediated β-TrCP/β-catenin/NF-κB axis. [ABSTRACT FROM AUTHOR]

Net Primary Productivity (NPP) is one of the significant indicators to measure environmental changes; thus, the relevant study of NPP in Northeast China, Asia, is essential to climate changes and ecological sustainable development. Based on the Global Production Efficiency (GLO-PEM) model, this study firstly estimated the NPP in Northeast China, from 2001 to 2019, and then analyzed its spatio-temporal evolution, future changing trend and phenology regularity. Over the years, the NPP of different forests type in Northeast China showed a gradual increasing trend. Compared with other different time stages, the high-value NPP (700–1300 gC·m−2·a−1) in Changbai Mountain, from 2017 to 2019, is more widely distributed. For instance, the NPP has an increasing rate of 6.92% compared to the stage of 2011–2015. Additionally, there was a significant advance at the start of the vegetation growth season (SOS), and a lag at the end of the vegetation growth season (EOS), from 2001 to 2019. Thus, the whole growth period of forests in Northeast China became prolonged with the change of phenology. Moreover, analysis on the sustainability of NPP in the future indicates that the reverse direction feature of NPP change will be slightly stronger than the co-directional feature, meaning that about 30.68% of the study area will switch from improvement to degradation. Unidirectional pultruded glass/carbon hybrid fiber-reinforced polymer (HFRP) bars with a diameter of 19 mm have recently been developed for various structural applications. In this study, the creep behavior of HFRP bars caused by bending was experimentally evaluated under different conditions. Our creep study included freeze–thaw preconditioned and unconditioned HFRP bars. The rate of strain and deflection were monitored continuously for a duration of 5000 h. The bars were further tested for creep under the combined effects of mechanical loading and induced thermal cycles, while continuously monitoring the strain rate. Stress levels of 50% to 70% were selected for our creep study. The creep behavior of the bars was analyzed utilizing Findley's power-law model. On the basis of the linear approximation of Findley's power law, modulus reductions of approximately 21%, 19%, and 10.75% were calculated for combined freeze–thaw/creep-loaded, freeze–thaw pretreated, and unconditioned HFRP bars, respectively, over a service period of 50 y. The time-dependent deflection of HFRP bars was analyzed by coupling Findley's power-law model with Euler Bernoulli's beam theory. The creep deflection intensified by 26.6% and 11.1% for preconditioned and untreated bars, respectively, after a service period of 50 y. Urban lakes play a vital role in the sustainable development of urbanized areas. In this freshwater ecosystem, massive microbial communities can drive the recycling of nutrients and regulate the water quality. However, water bacterial and fungal communities in the urban lakes are not well understood. In the present work, scanning electron microscopy (SEM) was combined with community level physiological profiles (CLPPs) and Illumina Miseq sequence techniques to determine the diversity and composition of the water bacterial and fungal community in three urban lakes, namely Xingqing lake (LX), Geming lake (LG) and Lianhu lake (LL), located in Xi'an City (Shaanxi Province, China). The results showed that these three lakes were eutrophic water bodies. The highest total nitrogen (TN) was observed in LL, with a value of 12.1 mg/L, which is 2 times higher than that of LG. The permanganate index (COD Mn ) concentrations were 21.6 mg/L, 35.4 mg/L and 28.8 mg/L in LG, LL and LX, respectively ( p < 0.01). Based on the CLPPs test, the results demonstrated that water bacterial communities in the LL and LX urban lakes had higher carbon source utilization ability. A total of 62,742 and 55,346 high quality reads were grouped into 894 and 305 operational taxonomic units (OTUs) for bacterial and fungal communities, respectively. Water bacterial and fungal community was distributed across 14 and 6 phyla. The most common phyla were Proteobacteriaand Cyanobacteria. Cryptomycota was particularly dominant in LL, while Chytridiomycota and Entomophthormycota were the most abundant fungal phyla, accounting for 95% of the population in the LL and 56% in the LG. Heat map and redundancy analysis (RDA) highlighted the dramatic differences of water bacterial communities among three urban lakes. Meanwhile, the profiles of fungal communities were significantly correlated with the water quality parameters (e.g., COD Mn and total nitrogen, TN). Several microbes ( Legionella sp. and Streptococcus sp.) related to human diseases, such as infectious diseases, were also found.
