Vertical federated learning is designed to protect user privacy by building local models over disparate datasets and transferring intermediate parameters without directly revealing the underlying data. However, the intermediate parameters uploaded by participants may memorize information about the training data. With the recent legislation on the"right to be forgotten", it is crucial for vertical federated learning systems to have the ability to forget or remove previous training information of any client. For the first time, this work fills in this research gap by proposing a vertical federated unlearning method on logistic regression model. The proposed method is achieved by imposing constraints on intermediate parameters during the training process and then subtracting target client updates from the global model. The proposed method boasts the advantages that it does not need any new clients for training and requires only one extra round of updates to recover the performance of the previous model. Moreover, data-poisoning attacks are introduced to evaluate the effectiveness of the unlearning process. The effectiveness of the method is demonstrated through experiments conducted on four benchmark datasets. Compared to the conventional unlearning by retraining from scratch, the proposed unlearning method has a negligible decrease in accuracy but can improve training efficiency by over 400%. Therapeutic oligonucleotides, such as antisense oligonucleotide (ASO) and small interfering RNA (siRNA), are a new class of therapeutics rapidly growing in drug discovery and development. A sensitive and reliable method to quantify oligonucleotides in biological samples is critical to study their pharmacokinetic and pharmacodynamic properties. Hybridization LC-MS/MS was recently established as a highly sensitive and specific methodology for the quantification of single-stranded oligonucleotides, e.g., ASOs, in various biological matrices. However, there is no report of this methodology for the bioanalysis of double-stranded oligonucleotides (e.g., siRNA). In this work, we investigated hybridization LC-MS/MS methodology for the quantification of double-stranded oligonucleotides in biological samples using an siRNA compound, siRNA-01, as the test compound. The commonly used DNA capture probe and a new peptide nucleic acid (PNA) probe were compared for the hybridization extraction of siRNA-01 under different conditions. The PNA probe achieved better extraction recovery than the DNA probe, especially for high concentration samples, which may be due to its stronger hybridization affinity. The optimized hybridization method using the PNA probe was successfully qualified for the quantitation of siRNA-01 in monkey plasma, cerebrospinal fluid (CSF), and tissue homogenates over the range of 2.00–1000 ng/mL. This work is the first report of the hybridization LC-MS/MS methodology for the quantification of double-stranded oligonucleotides. The developed methodology will be applied to pharmacokinetic and toxicokinetic studies of siRNA-01. Transparent electrodes (TEs) are important components in organic optoelectronic devices. ITO is the mostly applied TE material, which is costly and inferior in mechanical performance, and could not satisfy the versatile need for the next generation of transparent optoelectronic devices. Recently, many new TE materials emerged to try to overcome the deficiency of ITO, including graphene, ultrathin metal, and oxide-metal-oxide structure. By finely control of the fabrication techniques, the main properties of conductivity, transmittance, and mechanical stability, have been studied in the literatures, and their applicability in the potential optoelectronic devices has been reported. Herein, in this work, we summarized the recent progress of the TE materials applied in optoelectronic devices by focusing on the fabrication, properties, such as Graphene, ultra-thin metal film, and metal oxide and performance. The advantages and insufficiencies of these materials as TEs have been summarized and the future development aspects have been pointed out to guide the design and fabrication TE materials in the next generation of transparent optoelectronic devices. Electrochromic devices are the preferred devices for smart windows because they work independently of uncontrollable environmental factors and rely more on the user's personal feelings to adjust actively. However, in practical applications, the ambient temperature still has an impact on device performance, such as durability, reversibility and switching performance, etc. These technical issues have significantly slowed down the commercialization of electrochromic devices (ECDs). It is necessary to investigate the main reasons for the influence of temperature on the device and make reasonable optimization to enhance the effectiveness of the device and extend its lifetime. In recent years, with the joint efforts of various outstanding research teams, the performance of electrochromic devices has been rapidly improved, with a longer lifetime, richer colors, and better color contrast. This review highlights the important research on temperature–dependent electrochromic properties in recent years. Also, the reported structures, mechanisms, characteristics, and methods for improving electrochromic properties are discussed in detail. In addition, the challenges and corresponding strategies in this field are presented in this paper. This paper will inspire more researchers to enrich the temperature–dependent properties of ECDs and their related fields with innovative means and methods to overcome the technical obstacles faced. With the increase in population aging, the tendency of osteochondral injury will be accelerated, and repairing materials are increasingly needed for the optimization of the regenerative processes in bone and cartilage recovery. The local environment of the injury sites and the deficiency of Mg2+ retards the repairing period via inhibiting the progenitor osteogenesis and chondrogenesis cells' recruitment, proliferation, and differentiation, which results in the sluggish progress in the osteochondral repairing materials design. In this article, we elucidate the Mg2+-concentration specified effect on the cell proliferation, osteochondral gene expression, and differentiation of modeling chondrocytes (extracted from New Zealand white rabbit) and osteoblasts (MC3T3-E1). The concentration of Mg2+ in the culture medium affects the proliferation, chondrogenesis, and osteogenesis: (i) Appropriate concentrations of Mg2+ promote the proliferation of chondrocytes (1.25–10.0 mM) and MC3T3-E1 cells (2.5–30.0 mM); (ii) the optimal concentration of Mg2+ that promotes the gene expression of noncalcified cartilage is 15 mM, calcified cartilage 10 mM, and subchondral bone 5 mM, respectively; (iii) overdosed Mg2+ leads to the inhibition of cell activity for either chondrocytes (>20 mM) or osteoblasts (>30 mM). The biomimetic elucidation for orchestrating the allocation of gradient concentration of Mg2+ in accordance of the physiological condition is crucial for designing the accurate microenvironment in osteochondral injury defects for optimization of bone and cartilage repairing materials in the future. For mechanical equipment, bearings have a high incidence area of faults. A problem for bearings is that their fault characteristics include a vibrating screen exciter which is weak and thus easily covered in strong background noise, hence making the noise difficult to remove. In this paper, a noise reduction method based on singular value decomposition, improved by singular value's unilateral ascent method (SSVD), and a fault feature enhancement method, i.e., variational mode decomposition, improved by revised whale algorithm optimization (RWOA-VMD), are proposed. These two methods are used in vibration signal processing with early faults of bearings having a vibrating screen and they have achieved significant application results. This paper also aims to construct a multi-modal feature matrix composed of energy entropy, singular value entropy, and power spectrum entropy, and then the early fault diagnosis of bearings of a vibrating screen exciter bearing is realized by using the proposed support vector machine, improved by the aquila optimizer algorithm (AO-SVM). Metakaolin was used as a raw material for the preparation of geopolymers, where two types of alkali activators (Na2SiO3 + NaOH and Na2SiO3 + NaOH) were used to prepare metakaolin geopolymers at room temperature. The mechanical properties and microstructures of the metakaolin geopolymers were analyzed. A three-factor, four-level orthogonal test was designed to investigate the mechanical properties of the metakaolin geopolymer with different ratios. The compressive and flexural strength of different specimens were tested for 7 and 28 days. Both the Na-based and K-based geopolymers exhibited excellent mechanical properties, but the K-based geopolymer had better mechanical properties. The optimal compressive strength and flexural strength of the K-based geopolymer were 73.93 MPa and 9.37 MPa, respectively. The 28-day optimal compressive strength of the Na-based polymer was 65.79 MPa, and the flexural strength was 8.71 MPa. SEM, XRD, and FTIR analyses showed that the mechanical properties of the geopolymers could be greatly improved by using a higher alkaline solution concentration, proper Na2SiO3/MOH mass ratio, and proper mass ratio of alkali exciter to metakaolin. Amorphous silicoaluminate was more favorable for the dissolution of silicon–alumina raw materials, promoted the formation of an amorphous silicoaluminate gel, and caused the internal structure of the geopolymer to be more compact. Technological advances in unmanned aerial vehicles (UAVs) have made it possible to employ drone deliveries for packaged products, but currently no standards of practice are available to qualify packaged products traveling through this distribution channel. This research proposes a methodology to collect field data from a UAV to develop simulation techniques for use with package testing equipment. This project utilized data recorders to measure the vibration, shock, and atmospheric field data on two models of the DJI drones. The root mean square G value (Grms), the power spectral density (PSD), maximum G-values and shock response spectrum, and atmospheric data were reported in this study. The study found that the general shape of the PSD profile of the drones differed from the PSD air profiles of the aircraft. The overall recorded Grms levels of the drones were also significantly higher than those of the published air profile of the aircraft. Moreover, the study found that the drone's in-flight vibration intensities in the horizontal level were consistently higher than those in the vertical direction. The major sources of vibration and shock in both drones' models originated from the two-propeller rotations. Shocks recorded during the flight reached 14 G, took place in the horizontal plane with the drone accelerating as opposed to the vertical plane where the drone is landing. [ABSTRACT FROM AUTHOR]

Patulin, a potent mycotoxin, contaminates fruits and derived products worldwide, and is a serious health concern. Several yeast strains have shown the ability to effectively degrade patulin. However, the mechanisms of its biodegradation still remain unclear at this time. In the present study, biodegradation and involved mechanisms of patulin by an antagonistic yeast Candida guilliermondii were investigated. The results indicated that C. guilliermondii was capable of not only multiplying to a high population in medium containing patulin, but also effectively reducing patulin content in culture medium. Degradation of patulin by C. guilliermondii was dependent on the yeast cell viability, and mainly occurred inside cells. E-ascladiol was the main degradation product of patulin. An iTRAQ-based proteomic analysis revealed that the responses of C. guilliermondii to patulin were complex. A total of 30 differential proteins involved in 10 biological processes were identified, and more than two-thirds of the differential proteins were down-accumulated. Notably, a short-chain dehydrogenase (gi|190348612) was markedly induced by patulin at both the protein and mRNA levels. Carbon dot (CD)-based multi-mode sensing has drawn much attention owing to its wider application range and higher availability compared with single-mode sensing. Herein, a simple and green methodology to construct a CD-based dual-mode fluorescent sensor from the waste biomass of flowers of wintersweet (FW-CDs) for parallel and semi-quantitative visual detection of Cr(VI) and Fe3+ was firstly reported. The FW-CD fluorescent probe had a high sensitivity to Cr(VI) and Fe3+ with wide ranges of linearity from 0.1 to 60 µM and 0.05 to 100 µM along with low detection limits (LOD) of 0.07 µM and 0.15 µM, respectively. Accordingly, the FW-CD-based dual-mode sensor had an excellent parallel sensing capacity toward Cr(VI) and Fe3+ with high selectivity and strong anti-interference capability by co-using dual-functional integration and dual-masking strategies. The developed parallel sensing platform was successfully applied to Cr(VI) and Fe3+ quantitative detection in real samples with high precision and good recovery. More importantly, a novel FW-CD-based fluorescent hydrogel sensor was fabricated and first applied in the parallel and semi-quantitative visual detection of Cr(VI) and ferrous ions in industrial effluent and iron supplements, further demonstrating the significant advantage of parallel and visual sensing strategies. RIDR-PI-103 is a novel reactive oxygen species (ROS)-induced drug release prodrug with a self-cyclizing moiety linked to a pan-PI3K inhibitor (PI-103). Under high ROS, PI-103 is released in a controlled manner to inhibit PI3K. The efficacy and bioavailability of RIDR-PI-103 in breast cancer remains unexplored. Cell viability of RIDR-PI-103 was assessed on breast cancer cells (MDA-MB-231, MDA-MB-361 and MDA-MB-453), non-tumorigenic MCF10A and fibroblasts. Matrigel colony formation, cell proliferation and migration assays examined the migratory properties of breast cancers upon treatment with RIDR-PI-103 and doxorubicin. Western blots determined the effect of doxorubicin ± RIDR-PI-103 on AKT activation and DNA damage response. Pharmacokinetic (PK) studies using C57BL/6J mice determined systemic exposure (plasma concentrations and overall area under the curve) and T1/2 of RIDR-PI-103. MDA-MB-453, MDA-MB-231 and MDA-MB-361 cells were sensitive to RIDR-PI-103 vs. MCF10A and normal fibroblast. Combination of doxorubicin and RIDR-PI-103 suppressed cancer cell growth and proliferation. Doxorubicin with RIDR-PI-103 inhibited p-AktS473, upregulated p-CHK1/2 and p-P53. PK studies showed that ~200 ng/mL (0.43 µM) RIDR-PI-103 is achievable in mice plasma with an initial dose of 20 mg/kg and a 10 h T1/2. Rare-earth labeling in biological apatite could provide critical information for the pathologic transition (osteoclastic) and physiologic regeneration (osteogenesis) of bone and teeth because of their characteristic site-sensitive fluorescence in different coordinative conditions of various tissues in many biological processes. However, the rare-earth labeling method for biological apatites, i.e., carbonated-hydroxyapatite, has been rarely found in the literature. In this paper, we report a Pourbaix-diagram guided mineralizing strategy to controllable carbonation and doping of rare-earth ions in the hydroxyapatite (HA) lattice. The carbonation process of hydroxyapatite was achieved by controllable mineralization in hydrothermal condition with K2CO3 as the carbonate source, which results into the pure B-type carbonated hydroxyapatite (CHA) with tunable carbonate substitution degree. All of the as-synthesized materials crystalized into P63/m (No. 176) space group with the lattice parameter of a decreases and c increases with the increasing of carbonate content in the reactants. Structural refinement results revealed that the substitution of planar CO32− is superimposed on one of the faces of PO43− tetrahedral sub-units with a rotation angle of 30° in reference to c-axis. All of the hydrothermally synthesized CHA nanocrystals show hexagonal rod-like morphology with the length of 70–110 nm and diameter of 21–35 nm, and the decreasing length/diameter ratio from 3.61 to 2.96 from low to high carbonated level of the samples. Five rare-earth cations, of Pr3+, Sm3+, Eu3+, Tb3+, and Ho3+, were used as possible probe ions that can be doped into either HA or CHA lattice. The site-preference of Tb3+ doping is the same in the crystallographic site of HA and CHA according to characteristic emission peaks of 5D4–7Fj (j = 3–6) transitions in their photoluminescent spectroscopy. Our work provides a controllable carbonation method for rare-earth labeling hydroxyapatite nanomaterials with potential biologically active implant powders for bone repair and tissue regeneration. Uniaxial compressive strength (UCS) and peak strain (PS) are essential indices for studying the mechanical properties of coal and rock masses, and they are closely related to mechanical parameters such as the elastic modulus (E), Poisson's ratio (υ), cohesion (C) and internal friction angle (Φ) of coal and rock masses. This study took the No. 2-1 coal seam of Zhaogu No. 2 Mine, in Henan Province, China, as the research object. An RMT-150B servo testing machine was used to test all mechanical parameters, including the E, υ, C and Φ of coal and rock masses. Based on the principle of orthogonal testing, Three Dimensions Fast Lagrangian Analysis of Continua (FLAC3D) was used to select E, υ, C, Φ, tensile strength (Rm) and dilation angle (Ψ) as initial participation factors. Using these six parameters and a five-level combination scheme (L25 (56)), the influence of coal mechanical parameters on UCS and PS was investigated, using the software SPSS for stepwise regression analysis, and a uniaxial pressure-resistant regression prediction equation was established. The research showed that, under uniaxial compression conditions, the main parameters controlling UCS of coal masses are C and Φ; conversely, the main parameters controlling PS are E and C. UCS and PS exhibit significant linear relationships with these main controlling parameters. Here, a stepwise regression prediction equation was established through reliability verification analysis using the main controlling parameters. This prediction method produces very small errors and a good degree of fit, thus allowing the rapid prediction of UCS. The precision of the stepwise regression model depends on the number of test samples, which can be increased in the later stages of a design project to further improve the precision of the projection model. B-Rapidly Accelerated Fibrosarcoma (BRAF) mutations are found in about 50% of melanoma patients. Treatment with Food and Drug Administration (FDA)-approved BRAF and MAP/ERK kinase (MEK) inhibitors has improved progression free and overall survival of patients with BRAF mutant melanoma. However, all responders develop resistance typically within 1 year of treatment with these inhibitors. Evidence indicates that reactive oxygen species (ROS) levels are elevated after BRAF pathway inhibition treatment. We aim to decipher the role of mitochondrial antioxidant proteins relative to ROS levels and BRAF pathway inhibitor resistance. We observed BRAF mutant melanoma cells treated with the combination of a MEK inhibitor (trametinib) and a BRAF inhibitor (dabrafenib), exhibited elevated ROS levels, both in in vitro and in vivo melanoma models. We next generated trametinib- and dabrafenib-resistant (TDR) cells and found increased ROS levels after acquisition of resistance. An immunofluorescence experiment showed an increase of DNA damage in TDR cell lines. Furthermore, we observed that TDR cells increased superoxide dismutase 2 (SOD2), an antioxidant, at both mRNA and protein levels, with the upregulation of the transcription factor Nuclear Factor (NF)-κB. Knockdown of SOD2 significantly reduced the growth of BRAF pathway inhibitor-resistant cells. In addition, the results indicate that TDR cells can be re-sensitized to BRAF pathway inhibitors by the ROS scavenger, N-Acetyl Cysteine (NAC). Overall, these data indicate that BRAF pathway inhibitor-resistant cells can compensate for elevated ROS via increased expression of the antioxidant SOD2. [ABSTRACT FROM AUTHOR]

Melanoma is the most lethal form of skin cancer. Melanoma is usually curable with surgery if detected early, however, treatment options for patients with metastatic melanoma are limited and the five-year survival rate for metastatic melanoma had been 15–20% before the advent of immunotherapy. Treatment with immune checkpoint inhibitors has increased long-term survival outcomes in patients with advanced melanoma to as high as 50% although individual response can vary greatly. A mutation within the MAPK pathway leads to uncontrollable growth and ultimately develops into cancer. The most common driver mutation that leads to this characteristic overactivation in the MAPK pathway is the B-RAF mutation. Current combinations of BRAF and MEK inhibitors that have demonstrated improved patient outcomes include dabrafenib with trametinib, vemurafenib with cobimetinib or encorafenib with binimetinib. Treatment with BRAF and MEK inhibitors has met challenges as patient responses began to drop due to the development of resistance to these inhibitors which paved the way for development of immunotherapies and other small molecule inhibitor approaches to address this. Resistance to these inhibitors continues to push the need to expand our understanding of novel mechanisms of resistance associated with treatment therapies. This review focuses on the current landscape of how resistance occurs with the chronic use of BRAF and MEK inhibitors in BRAF-mutant melanoma and progress made in the fields of immunotherapies and other small molecules when used alone or in combination with BRAF and MEK inhibitors to delay or circumvent the onset of resistance for patients with stage III/IV BRAF mutant melanoma. Shortly after the cessation of seafloor spreading, intraplate magmatism affected large areas in the South China Sea (SCS) region. The origin and geodynamic setting of the post-spreading volcanism is still in debate, for many previous studies have focused on petrogenesis and mantle source of the late Cenozoic basalts from the SCS region. In this study, we obtained in situ major element compositions (by using Electron microprobe analysis—EMPA) and trace element compositions (by using laser ablation inductively coupled plasma mass spectrometry— LA-ICP-MS) for minerals (clinopyroxenes (Cpx), plagioclases (Pl), and olivines (Ol)) hosted by late Cenozoic basaltic rocks from Thailand. The results showed that the olivines had forsterite contents between 60.12% and 84.74%. Clinopyroxene were diopside and augite, and they were enriched in light rare earth elements (LREEs) (LaN/YbN = 1.93–4.27) and depleted in large-ion lithophile elements (LILEs). Mineral compositions (mainly based on clinopyroxene) confirmed that these late Cenozoic basaltic rocks were of an intraplate affinity and were similar to contemporaneous basaltic fields in the SCS region (Southern Vietnam, Northern Hainan, and SCS seamounts). Plagioclases were predominantly labradorite, with a few andesine and bytownite, and they were enriched in LREEs and Ba, Sr, and Pb, and most of them exhibited strong positive Eu anomalies. The source lithology of Thailand basaltic rocks could be garnet pyroxenite. The mantle potential temperature beneath Thailand is in the range of 1448–1467 °C, which can be comparable to those beneath Southern Vietnam and Northern Hainan, indicating the Thailand basaltic rocks could be produced by the Hainan mantle plume. In addition, the crystallization temperature of clinopyroxenes (1145–1214 °C) and plagioclase (1067–1133 °C) and their composition characteristics indicate that the magmatic processes have a conspicuous characteristic of fast rate of magma upwelling. Thus, we proposed that the deep geodynamic setting of Thailand late Cenozoic basaltic rocks is similar to those of the whole SCS region, and Hainan mantle plume plays a significant role in the petrogenesis of these basaltic rocks. When asbestos containing materials (ACM) must be removed from the building before demolition, additional greenhouse gas (GHG) emissions are generated. However, precedent studies have not considered the removal of ACM from the building. The present study aimed to develop a model for estimating GHG emissions created by the ACM removal processes, specifically the removal of asbestos cement slates (ACS). The second objective was to use the new model to predict the total GHG emission produced by ACM removal in the entire country of Korea. First, an input-equipment inventory was established for each step of the ACS removal process. Second, an energy consumption database for each equipment type was established. Third, the total GHG emission contributed by each step of the process was calculated. The GHG emissions generated from the 1,142,688 ACS-containing buildings in Korea was estimated to total 23,778 tonCO₂eq to 132,141 tonCO₂eq. This study was meaningful in that the emissions generated by ACS removal have not been studied before. Furthermore, the study deals with additional problems that can be triggered by the presence of asbestos in building materials.
