To improve maneuverability, the focus of photoelectric theodolites is on reducing the weight of the primary mirror and enhancing its optical performance. This study uses MOAT and Sobol methods to identify key parameters that affect design. Using the high-sensitivity part as the optimization domain, six optimization results were obtained based on the multi-objective SIMP topology optimization method and synthesized into a compromise optimization structure. The performance of the mirror before and after optimization was compared on the opto-mechanical-thermal level. Modal analysis shows the optimized structure has a first natural frequency of 716.84 Hz, indicating excellent stiffness and avoiding low-frequency resonance, with a 30.37% weight reduction. Optical performance is also improved, with a 6 μm reduction in the spot diagram radius and an 8.95 nm decrease in RMS. Simulations under real-world conditions show that the lightweight mirror performs better in resisting gravity deformation and maintaining imaging quality. At maximum thermal deformation, the spot diagram radius of the optimized mirror is 1521.819 μm, with only a 0.145% difference in imaging quality compared to the original. In conclusion, the optimized structure shows comprehensive advantages. Constructing the optical system components and the real physical environment of the site provides a valuable reference for the optimization and analysis of the mirror.

Gallium nitride (GaN) exhibits distinctive physical and chemical properties that render it indispensable in a multitude of electronic and optoelectronic devices. Given that GaN is a typical hard and brittle material that is difficult to machine, femtosecond laser technology provides an effective and convenient tool for processing such materials. However, GaN undergoes complex physical and chemical changes during high-power ablation, which poses a challenge to high-precision processing with controllable geometry. In this study, the quantitative relationship between the parameters of a single-pulse femtosecond laser and GaN ablation morphology was investigated using isotherm distribution. A multiphysics model using COMSOL Multiphysics ® was developed to generate the isothermal distributions. Experiments were conducted on the femtosecond laser ablation of GaN at various single-pulse energies, and the resulting ablation morphologies were compared with the predictions from the multiphysics model. The comparison demonstrated that the calculated isotherm distribution accurately predicted not only the ablation diameter and depth but also the crater shape across a broad range of laser fluences. The predicted errors of the ablation diameters and depths were within 4.71% and 10.9%, respectively. The root mean square error (RMSE) and coefficient of determination (R 2 ) were employed to evaluate the prediction errors associated with the crater shapes, which fell within the range of 0.018-0.032 μm and 0.77-0.91, respectively. This study can provide an important reference for utilizing femtosecond lasers in the precise ablation of GaN to achieve desired geometries.

Recent decades have seen growing attention on viruses in the environment and their potential impacts as a result of global epidemics. Due to the diversity of viral species along with the complexity of environmental and host factors, virus extraction and detection methods have become key for the study of virus ecology. This review systematically summarises the methods for extracting and detecting pathogens from different environmental samples (e.g., soil, water, faeces, air) and biological samples (e.g., plants, animals) in existing studies, comparing their similarities and differences, applicability, as well as the advantages and disadvantages of each method. Additionally, this review discusses future directions for research in this field. The aim is to provide a theoretical foundation and technical reference for virus ecology research, facilitating further exploration and applications in this field.

The medicinal plant Stephania yunnanensis is rich in aporphine alkaloids, a type of benzylisoquinoline alkaloid (BIA), with aporphine being the representative and most abundant compound, but our understanding of the biosynthesis of BIAs in this plant has been relatively limited. Previous research reported the genome of S. yunnanensis and preliminarily identified the norcoclaurine synthase (NCS), which is involved in the early stages of the BIA biosynthetic pathways. However, the key genes promoting the formation of the aporphine skeleton have not yet been reported. In this study, based on the differences in the content of crebanine and several other BIAs in different tissues, we conducted transcriptome sequencing of roots, stems, and leaves. We then identified candidate genes through functional annotation and sequence alignment and further analyzed them in combination with the genome. Based on this analysis, we identified three CYP80 enzymes (SyCYP80Q5-1, SyCYP80Q5-3, and SyCYP80G6), which exhibited different activities toward ( S )- and ( R )-configured substrates in S. yunnanensis and demonstrated strict stereoselectivity enroute to aporphine. This study provides metabolomic and transcriptomic information on the biosynthesis of BIAs in S. yunnanensis , offers valuable insights into the elucidation of BIA biosynthesis, and lays the foundation for the complete analysis of pathways for more aporphine alkaloids.

Post-stroke spasticity (PSS), characterized by a velocity-dependent increase in muscle tone and exaggerated reflexes, affects a significant portion of stroke patients and presents a substantial obstacle to post-stroke rehabilitation. Effective management and treatment for PSS remains a significant clinical challenge in the interdisciplinary aspect depending on the understanding of its etiologies and pathophysiology. We systematically review the relevant literature and provide the main pathogenic hypotheses: alterations in the balance of excitatory and inhibitory inputs to the descending pathway or the spinal circuit, which are secondary to cortical and subcortical ischemic or hemorrhagic injury, lead to disinhibition of the stretch reflex and increased muscle tone. Prolongation of motoneuron responses to synaptic excitation by persistent inward currents and secondary changes in muscle contribute to hypertonia. The guidelines for PSS treatment advocate for a variety of therapeutic approaches, yet they are hindered by constraints such as dose-dependent adverse effects, high cost, and limited therapeutic efficacy. Taken together, we highlight key processes of PSS pathophysiology and summarize many interventions, including neuroprotective agents, gene therapy, targeted therapy, physiotherapy, NexTGen therapy and complementary and alternative medicine. We aim to confer additional clinical benefits to patients and lay the foundation for the development of new potential therapies against PSS.

More than 70% of cancer patients receive radiotherapy during their treatment, with consequent various side effects on normal cells due to high ionizing radiation doses despite tumor shrinkage. To date, many radioprotectors and radiosensitizers have been investigated in preclinical studies, but their use has been hampered by the high toxicity to normal cells or poor tumor radiosensitization effects. Genistein is a naturally occurring isoflavone found in soy products. It selectively sensitizes tumor cells to radiation while protecting normal cells from radiation-induced damage, thus improving the efficacy of radiotherapy and consequent therapeutic outcomes while reducing adverse effects. Genistein protects normal cells by its potent antioxidant effect that reduces oxidative stress and mitigates radiation-induced apoptosis and inflammation. Conversely, genistein increases the radiosensitivity of tumor cells through specific mechanisms such as the inhibition of DNA repair, the arrest of the cell cycle in the G 2 /M phase, the generation of reactive oxygen species (ROS), and the modulation of apoptosis. These effects increase the cytotoxicity of radiation. Preclinical studies demonstrated genistein efficacy in various cancer models, such as breast, prostate, and lung cancer. Despite limited clinical studies, the existing evidence supports the potential of genistein in improving the therapeutic effect of radiotherapy. Future research should focus on dosage optimization and administration, the exploration of combination therapies, and long-term clinical trials to establish genistein benefits in clinical settings. Hence, the unique ability of genistein to improve the radiosensitivity of tumor cells while protecting normal cells could be a promising strategy to improve the efficacy and safety of radiotherapy.

RAD52 plays crucial roles in several aspects of mammalian cells, including DNA double-strand breaks repair, viral infection, cancer development, and antibody class switching. To comprehensively elucidate the role of RAD52 in maintaining genome stability and uncover additional functions of RAD52 in mammals, we performed the transcriptomics and proteomics analysis of the liver of RAD52 knockout mice. Transcriptomics analysis reveals overexpression of mitochondrial genes in the liver of RAD52 knockout (RAD52KO) mice. Proteomics analysis of RAD52KO mice shows that damage recognition proteins Cul4b and Rad23a in the process of nucleotide excision repair pathway are overexpressed. Furthermore, gene ontology and KEGG enrichment analysis (accessed on 20 November 2024) from integrated omics shows that differentially expressed genes are significantly enriched in pathways related to mitochondrial oxidative phosphorylation and nucleotide metabolism in the liver of RAD52KO mice. In addition, mRNA and protein levels of Bhmt1b are elevated in the liver of RAD52KO mice. Taken together, this study provides valuable insights into the function and mechanism of RAD52.

This study presents a facile one-pot synthesis method to fabricate BiFeO 3 -Bi 25 FeO 40 -Bi 2 O 3 heterojunction photocatalysts with controllable compositions and pure phases. Three different binary heterojunctions (BiFeO 3 /Bi 25 FeO 40 , BiFeO 3 /Bi 2 O 3 , and Bi 25 FeO 40 /Bi 2 O 3 ) and a ternary BiFeO 3 /Bi 25 FeO 40 /Bi 2 O 3 heterojunction were formed, all exhibiting significantly enhanced photocatalytic performance for the degradation of methylene blue (MB) and phenol under visible light irradiation, outperforming the individual compositions. Notably, the BiFeO 3 /Bi 25 FeO 40 heterojunction achieved the highest degradation efficiency (93.68% and 83.3% for MB and phenol, respectively) as well as excellent stability. Impressively, the phenol degradation efficiency of BiFeO 3 /Bi 25 FeO 40 was even over twice that of BiFeO 3 and Bi 25 FeO 40 , and four times higher than that of Bi 2 O 3 . The enhanced photocatalytic activity of the BiFeO 3 /Bi 25 FeO 40 heterojunction is primarily attributed to the advantageous S-scheme band alignments that facilitate efficient charge separation and enhance redox capabilities. While other heterojunctions also exhibited improved MB and phenol degradation efficiency, each unique combination of materials led to distinct electronic structures and diverse reaction mechanisms. The simplicity and scalability of the synthesis method, combined with the remarkable photocatalytic performance of these BiFeO 3 -Bi 25 FeO 40 -Bi 2 O 3 heterojunction materials, position them as highly promising candidates for applications in environmental remediation and solar energy conversion.

IoT (Internet of Things) networks are vulnerable to network viruses and botnets, while facing serious network security issues. The prediction of payload states in IoT networks can detect network attacks and achieve early warning and rapid response to prevent potential threats. Due to the instability and packet loss of communications between victim network nodes, the constructed protocol state machines of existing state prediction schemes are inaccurate. In this paper, we propose a network payload predictor called IoTGuard, which can predict the payload states in IoT networks based on real-time IoT network traffic. The steps of IoTGuard are briefly as follows: Firstly, the application-layer payloads between different nodes are extracted through a module of network payload separation. Secondly, the classification of payload state within network flows is obtained via a payload extraction module. Finally, the predictor of payload state in a network is trained on a payload set, and these payloads have state labels. Experimental results on the Mozi botnet dataset show that IoTGuard can predict the state of payloads in IoT networks more accurately while ensuring execution efficiency. IoTGuard achieves an accuracy of 86% in network payload prediction, which is 8% higher than the state-of-the-art method NetZob, and the training time is reduced by 52.8%.

This study aimed to predict and fit the nonlinear dynamic grip force of the human upper limb using surface electromyographic (sEMG) signals. The research employed a time-series-based neural network, NARX, to establish a mapping relationship between the electromyographic signals of the forearm muscle groups and dynamic grip force. Three-channel electromyographic signal acquisition equipment and a grip force sensor were used to record muscle signals and grip force data of the subjects under specific dynamic force conditions. After preprocessing the data, including outlier removal, wavelet denoising, and baseline drift correction, the NARX model was used for fitting analysis. The model compares two different training strategies: regularized stochastic gradient descent (BRSGD) and conjugate gradient (CG). The results show that the CG greatly shortened the training time, and performance did not decline. NARX demonstrated good accuracy and stability in dynamic grip force prediction, with the model with 10 layers and 20 time delays performing the best. The results demonstrate that the proposed method has potential practical significance for force control applications in smart prosthetics and virtual reality.

Hemp fibers, recognized for their breathability, specific strength, and ultraviolet resistance, are widely utilized in textile manufacturing and composite materials. Bio-degumming is a promising alternative technology to traditional chemical degumming that can be used to produce hemp fibers due to its eco-friendly nature. However, its lower efficiency has hindered its widespread adoption. The unclear and complex structure of the gums leads to a poor understanding on the enzyme types required for bio-degumming, thereby restricting improvements in its efficiency. In this study, the morphological characteristics, polysaccharide composition, and branched structure of hemp stem, roving fibers, and refined fibers were investigated using scanning electron microscopy and laser scanning confocal microscopy in combination with immunofluorescence techniques, with a view to identify the enzymes necessary for the efficient bio-degumming of hemp. The results revealed that the gums were primarily located in the middle lamella, phloem parenchyma, and certain xylem tissues. These tissues showed chunk-like, fence-like, and plate-like shapes, respectively, and tightly wrapped around the fiber bundles. In these tissues, pectin comprised low-esterified homogalacturonan, along with rhamnogalacturonan carrying galactan and arabinan branches. Xylan exhibited acetyl, arabinose, and glucuronic acid branches, while mannan displayed acetyl and galactose branches. Partial xylan and mannan were masked by pectin, and the branching structures impeded their enzymatic removal. As a consequence, the necessary enzymes and their synergistic effects for effective hemp roving degumming were elucidated. Pectin degradation was facilitated by pectate lyase and rhamnogalacturonan-degrading enzymes. Xylan and mannan were effectively removed by endo-xylanase and endo-mannanase, a process necessitating the synergistic action of branched-chain-degrading enzymes, including the esterase, α-L-arabinofuranosidase, α-galactosidase, and α-glucuronidase. This study provided practical strategies to enhance the efficiency of hemp bio-degumming.

Owing to its extensive use and intrinsic toxicity, NH 3 detection is very crucial. Moisture can cause significant interference in the performance of sensors, and detecting NH 3 in high humidity is still a challenge. In this work, a humidity-activated NH 3 sensor was prepared by urocanic acid (URA) modifying poly (ethylene glycol) diacrylate (PEGDA) via a thiol-ene click cross-linking reaction. The optimized sensor achieved a response of 70% to 50 ppm NH 3 at 80% RH, with a response time of 105.6 s and a recovery time of 346.8 s. The sensor was improved for response and recovery speed. In addition, the prepared sensor showed excellent selectivity to NH 3 in high-humidity environments, making it suitable for use in some areas with high humidity all the year round or in high-humidity areas such as the detection of respiratory gas. A detailed investigation of the humidity-activated NH 3 -sensing mechanism was conducted using complex impedance plot (CIP) measurements.

There is increasing interest in the potential therapeutic role of 5-HT (serotonin) in the treatment of neurodegenerative diseases, which are characterized by the progressive degeneration and death of nerve cells. 5-HT is a vital neurotransmitter that plays a central role in regulating mood, cognition, and various physiological processes in the body. Disruptions in the 5-HT system have been linked to several neurological and psychiatric disorders, making it an attractive target for therapeutic intervention. Although the exact causes of neurodegenerative diseases such as Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS) are not fully understood, researchers believe that regulating the 5-HT system could help alleviate symptoms and potentially slow the progression of these diseases. Here, we delve into the potential of harnessing 5-HT as a therapeutic target for the treatment of neurodegenerative diseases. It is important to note that the current clinical drugs targeting 5-HT are still limited in the treatment of these complex diseases. Therefore, further research and clinical trials are needed to evaluate the feasibility and effectiveness of its clinical application.

In the context of escalating energy demands and environmental sustainability, the paradigm of global energy systems is undergoing a transformative shift to innovative and reliable energy-harvesting techniques ranging from solar cells to triboelectric nanogenerators (TENGs) to hybrid energy systems, where a fever in the study of perovskite materials has been set off due to the excellent optoelectronic properties and defect tolerance features. This review begins with the basic properties of perovskite materials and the fundamentals of TENGs, including their working principles and general developing strategy, then delves into the key role of perovskite materials in promoting TENG-based hybrid technologies in terms of energy conversion. While spotlighting the coupling of triboelectric-optoelectronic effects in harnessing energy from a variety of sources, thereby transcending the limitations inherent to single-source energy systems, this review pays special attention to the strategic incorporation of perovskite materials into TENGs and TENG-based energy converting systems, which heralds a new frontier in enhancing efficiency, stability, and adaptability. At the end, this review highlights the remaining challenges such as stability, efficiency, and functionality for applications in TENG-based energy-harvesting systems, aiming to provide a comprehensive overview of the current landscape and the prospective trajectory of the role of perovskite materials in TENG-based energy-harvesting technologies within the renewable energy sector.

In this paper, uniaxial compression tests and numerical simulation were conducted on specimens of five sizes, and the influence of specimen size on the failure characteristics and mechanics of specimens was studied. The results show that when the bottom size of the specimen is the same, with the increase in the height-width ratio of the specimen size (from 1 to 3), the peak stress of the specimen gradually decreases, but when the decrease is greatly reduced, the concentration of contact force chains in the model increases. The failure mode of the specimen changes from tensile failure to shear failure, and the distribution of cracks changes from multiple vertical cracks uniformly to a concentrated main oblique crack. The failure characteristics change from the overall failure to the serious failure of the near stressed end of the specimen, while the far stressed end is not failure or slight failure. When the height-width ratio of the specimen is the same, with the increase in the overall size, the peak stress decreases, and the dense vertical cracks change into a small amount of concentrated oblique cracks, and the integrity of the specimen and model is better. There is a good effect using PFC2D software to simulate the crack evolution and failure characteristics of plastic concrete.

The oil bean is a high-quality, economically valuable variety of kidney bean ( Phaseolus vulgaris L.) that is widely cultivated in Northeast China. However, the prevalence of anthracnose, caused by a combination of factors, including continuous cropping over many years, has led to significant declines in both yield and quality. The mitogen-activated protein kinase (MAPK) cascade is a highly conserved plant cell signaling pathway that plays a pivotal role in plant growth and development, as well as responses to biotic stress. However, its role in the response of P. vulgaris to anthracnose infection has not previously been reported. We identified and characterized thirteen MAPK genes ( PvMAPK01-PvMAPK13 ) in the P. vulgaris genome. These genes were found on eight of the eleven chromosomes of P. vulgaris , and phylogenetic analyses classified them into four previously established subgroups (A-D). Analysis of the cis -acting elements in their promoter regions revealed the presence of multiple elements associated with light, hormone regulation, stress responses, and growth and development. An analysis of intraspecific collinearity revealed that whole-genome and/or segmental duplication, rather than tandem duplication, has been the primary driver of PvMAPK family expansion in P. vulgaris . Transcriptome data revealed that the PvMAPKs differed in their tissue-specific expression patterns, with PvMAPK05 showing particularly high expression in stems and stem tips and PvMAPK07 and PvMAPK11 showing relatively low expression across all tissues. In general, expression of the PvMAPKs was higher in stems, stem tips, and pods than in other tissues and organs, suggesting that they may be particularly important for regulating stem and pod development. Analysis of the expression of PvMAPKs in field-grown plants infected or uninfected with anthracnose revealed that the relative expression levels of PvMAPK05 , PvMAPK07 , PvMAPK09 , and PvMAPK11 exhibited particularly significant changes in response to anthracnose infection across different varieties, suggesting their potential involvement in the anthracnose response of Phaseolus vulgaris. This study reports the fundamental characteristics of the thirteen MAPK genes in P. vulgaris , documents their expression patterns in diverse tissues, and offers preliminary insights into their responses to anthracnose infection, establishing a foundation for subsequent functional validation.

Background and Aim: The prevalence and adverse outcomes of metabolic dysfunction associated with steatotic liver disease (MAFLD) are increasing. The changes in the gut microbiota and metabolites associated with metabolic dysfunction-associated steatohepatitis (MASH) are regarded as an essential part of the progression of MAFLD. This study aimed to identify the gut microbiota and metabolites involved in the development of MAFLD in patients., Method: This study enrolled 90 patients (healthy controls, HC: n = 30; MASH: n = 30; MASH-related cirrhosis, MC: n = 30), and their fecal samples were collected for 16S rRNA sequencing and non-targeted LC-MS/MS metabolomics analysis. Data preprocessing and statistical analyses were performed using QIIME2 software, Pynast, QIIME2 package, Progenesis QI, and R program., Results: The abundance of Prevotellaceae at the family level and Prevotella at the genus level was lower in the MASH and NC samples than in the HC samples. Both Prevotellaceae and Prevotella showed the strongest correlation with MASH progression via random forest analysis. Untargeted metabolomics was used to quantitatively screen for discrepant metabolites in the stool samples from the three groups. Linolenic acid (LA)-related metabolite levels were significantly lower in MASH and NC samples. Associations between Prevotella- or LA-related metabolites and liver function were discovered. A high abundance of Prevotella was associated with LA-related metabolites and MASH., Conclusion: This study identified that gut microbiota and metabolites are associated with MASH-related metabolic dysfunction. LA and Prevotella are depleted during MASH progression, and additional supplementation with Prevotella may be a potential strategy for the future treatment of MAFLD.

Platanus officinalis fibers (PFs) taking advantage of high-availability, eco-friendly and low-cost characteristics have attracted significant focus in the field of biomaterial application. Polyethyleneimine grafted with polydopamine on magnetic Platanus officinalis fibers (PEI-PDA@M-PFs) were prepared through a two-step process of mussel inspiration and the Michael addition reaction, which can work as an effective multifunctional biomass adsorbent for anionic dye with outstanding separation capacity and efficiency. The as-prepared PEI-PDA@M-PFs possess desirable hydrophilicity, magnetism and positive charge, along with abundant amino functional groups on the surface, facilitating efficient adsorption and the removal of Eriochrome Black T (EBT) dyes from water. In addition to the formation mechanism, the adsorption properties, including adsorption isotherms, kinetics, and the reusability of the absorbent, were studied intensively. The as-prepared PEI-PDA@M-PFs achieved a theoretical maximum adsorption capacity of 166.11 mg/g under optimal conditions (pH 7.0), with 10 mg of the adsorbent introduced into the EBT solution. The pseudo-second-order kinetic and Langmuir models were well matched with experimental data. Moreover, thermodynamic data ΔH > 0 revealed homogeneous chemical adsorption with a heat-absorption reaction. The adsorbent remained at high stability and recyclability even after five cycles of EBT adsorption processes. These above findings provide new insights into the adsorption processes and the development of biologic material for sustainable applications.

In this work, we demonstrated the epitaxial growth of a gallium nitride (GaN) buffer structure on 200 mm SOI (silicon-on-insulator) substrates. This epitaxial layer is grown using a reversed stepped superlattice buffer (RSSL), which is composed of two superlattice (SL) layers with different Al component ratios stacked in reverse order. The upper layer, with a higher Al component ratio, introduces tensile stress instead of accumulative compressive stress and reduces the in situ curvature of the wafer, thereby achieving a well-controlled wafer bow ≤ ±50 µm for a 3.3 µm thick buffer. Thanks to the compliant SOI substrate, good crystal quality of the grown GaN layers was obtained, and a breakdown voltage of 750 V for a 3.3 µm thick GaN buffer was achieved. The breakdown field strength of the epitaxial GaN buffer layer on the SOI substrate is estimated to be ~2.27 MV/cm, which is higher than the breakdown field strength of the GaN-on-Si epitaxial buffer layer. This RSSL buffer also demonstrated a low buffer dispersion of less than 10%, which is good enough for the further processing of device and circuit fabrication. A D-mode GaN HEMT was fabricated on this RSSL buffer, which showed a good on/off ratio of ~10 9 and a breakdown voltage of 450 V.

Background/Objectives : Chronic Limb-Threatening Ischemia (CLTI) is a chronic limb ischemic disease caused by vascular lesions, characterized by pain, ulcers, and gangrene, which can be life-threatening in severe cases. The objective of this study is to explore whether Berbamine (BBM) can protect against and repair ischemic muscle tissue in the lower limbs; Methods : Using a mouse hindlimb ischemia (HLI) model, 36 C57BL6 mice were divided into sham, HLI, and HLI+BBM treatment groups. Results : Our findings indicate that BBM can restore motor function and muscle tissue pathology in mice, potentially by inhibiting the nuclear translocation of nuclear factor kappa-B (NF-κB), thereby alleviating tissue inflammation caused by chronic ischemia, reducing muscle cell apoptosis, inhibiting M1 macrophage polarization, and promoting angiogenesis. Conclusions : Our research suggests that BBM has the potential to protect against ischemic damage in lower limb muscle tissue, providing a new approach to the treatment of CLTI.

Intramuscular fat (IMF) content plays a crucial role in determining pork quality. Recent studies have highlighted transcriptional mechanisms controlling adipogenesis in porcine IMF. However, the changes in chromatin accessibility during adipogenic differentiation are still not well understood. In this study, we performed the assay for transposase-accessible chromatin with high-throughput sequencing (ATAC-seq) and transcriptome sequencing (RNA-Seq) analyses on porcine intramuscular preadipocytes to explore their adipogenic differentiation into mature adipocytes. We identified a total of 56,374 differentially accessible chromatin peaks and 4226 differentially expressed genes at day 0 and day 4 during adipogenic differentiation. A combined analysis of the ATAC-seq and RNA-seq data revealed that 1750 genes exhibited both differential chromatin accessibility and differential RNA expression during this process, including selenium-binding protein 1 ( SELENBP1 ), PLIN1 , ADIPOQ , and FASN . Furthermore, we found that vitamin D receptor (VDR) could bind to the promoter region of the SELENBP1 gene, activate SELENBP1 transcription, and ultimately promote lipid accumulation during adipogenic differentiation. This study provides a detailed overview of chromatin accessibility and gene expression changes during the adipogenic differentiation of porcine intramuscular preadipocytes. Moreover, we propose a novel regulatory mechanism involving the VDR-SELENBP1 signaling axis in adipogenic differentiation.

Cannabigerol (CBG), a non-psychoactive cannabinoid found in cannabis, has emerged as a promising therapeutic agent with a diverse range of potential applications. Unlike its well-known counterpart tetrahydrocannabinol (THC), CBG does not induce intoxication, making it an attractive option in the clinic. Recent research has shed light on CBG's intriguing molecular mechanisms, highlighting its potential to modulate multiple physiological processes. This review delves into the current understanding of CBG's molecular interactions and explores its therapeutic power to alleviate various conditions, including cancer, metabolic, pain, and inflammatory disorders, amongst others. We discuss how CBG interacts with the endocannabinoid system and other key signaling pathways, such as CB1, CB2, TPR channels, and α2-adrenoceptor, potentially influencing inflammation, pain, neurodegeneration, and other ailments. Additionally, we highlight the ongoing research efforts aimed at elucidating the full spectrum of CBG's therapeutic potential and its safety profile in clinical settings. Through this comprehensive analysis, we aim to provide a deeper understanding of CBG's role in promoting human health and pave the way for future research endeavors.

This study aimed to improve the mechanical properties and microstructure of recycled aggregate concrete (RAC) by incorporating carbon fibers (CFs) and nano-SiO 2 (NS) to promote the optimal utilization of RAC. The mechanical properties of the RAC were enhanced by both single and hybrid additions of CFs and NS, and the hybrid addition had a better strengthening effect. From the experimental results, it was found that the addition of CFs could increase the 28 d compressive strength and splitting strength of the RAC by 9.05% and 22.36%, respectively. The hybrid CFs and NS were more conducive to improving the mechanical properties of the RAC, and the enhancement effect increased first and then decreased with an increase in the NS content. The optimal content of NS was 0.8 wt%, which increased the 28 d compressive strength and splitting strength of the RAC by 20.51% and 14.53%, respectively. The microstructure results indicated that the addition of CFs had little effect on the optimized pore structure of the RAC, but the crack inhibition action of the CFs could improve the mechanical properties of the RAC. The addition of NS reduced the content of CH and facilitated the formation of more (C-S-H) gel. The hydrated calcium silicate (C-S-H) gel significantly decreased the porosity and transformed harmful capillary pores and harmful pores into harmless capillary pores and gel pores, thus improving the mechanical properties of the RAC. Therefore, the use of hybrid CFs and NS was more conducive to enhancing the performance of RAC for building materials.

Background/Objectives : Cutaneous infections of fungal and bacterial origins are common. An accurate diagnosis-especially concerning pathogens that are difficult to isolate on culture-can be achieved using molecular methods (PCR) with a short turnaround time. Methods : We reviewed records of skin specimens (superficial scrapings) submitted by dermatologists across the United States with a clinically suspected dermatitis. As per physician's order, specimens were tested for infections either fungal (N = 4262) or bacterial (N = 1707) in origin. All unique specimens (one per patient) were subjected to real-time PCR assays where cases suspected of a fungal etiology were tested for dermatophytes, Malassezia and Candida , and cases suspected of a bacterial etiology were tested for Streptococcus pyogenes , Staphylococcus aureus , and the mecA gene potentially conferring β-lactam resistance. Results : Fungal agents were detected in 32.8% (SD: 4.5) of the submitted specimens, with most attributed to dermatophytes (19.3% (SD: 4.9)), followed by Malassezia (8.7% (SD: 2.8)) and Candida (2.9% (SD: 1.0)). Dermatophyte detection was more common in the elderly (≥65 years) compared to young adults (18-44 years) (OR: 1.8 (95% CI: 1.5, 2.2)), whereas Malassezia was more commonly detected in younger age groups (12.1-13.6%) than the elderly (5.6%). Candida was more frequently observed in females while dermatophytes and Malassezia were more frequently observed in males. Approximately one quarter of the submitted skin specimens tested positive for S. aureus (23.6% (SD: 3.4)), of which 34.4% (SD: 9.8) exhibited concurrent detection of the mecA gene. An S. aureus detection was more frequently observed in males (OR: 1.5 (95% CI: 1.2, 1.9)) and in children (OR: 1.7 (95% CI: 1.2, 2.5)). Streptococcus pyogenes was rarely detected. Among specimens positive for dermatophytes, 12.0% (20/166) showed co-detection of S. aureus and mecA , which is in contrast to 6.8% (70/1023) detected in samples without a fungal co-detection and 6.2% (8/130) in samples positive for Malassezia . Conclusions : PCR testing, when available, can be valuable as a part of routine care for diagnosing patients with clinically suspected skin infections. Further studies are warranted to survey the prevalence of resistant S. aureus isolates in dermatology outpatients, in particular with regard to the association with dermatophyte infections.

Terahertz (THz) communication is a crucial technique in sixth generation (6G) mobile networks, which allow for ultra-wide bandwidths to enable ultra-high data rate wireless communication. However, the current subcarrier spacing and the size of fast Fourier transform (FFT) of the orthogonal frequency division multiplexing (OFDM) in 5G NR are insufficient regarding the bandwidth requirements of terahertz scenarios. In this paper, a novel waveform is proposed to address the ultra-wideband issue, namely the generalized filter bank orthogonal frequency division multiplexing (GFB-OFDM) waveform. The main advantages are summarized as follows: (1) The K -point IFFT is implemented by two levels of IFFTs in smaller sizes, i.e, performing M -point IFFT in N times and performing N -point IFFT in M times, where K=N×M. (2) The proposed waveform can accommodate flexible subcarrier spacings and different numbers of the subbands to provide various services in a single GFB-OFDM symbol. (3) Different bandwidths can be supported using a fixed filter since the filtering is performed on each subband. In contrast, the cyclic prefix orthogonal frequency division multiplexing (CP-OFDM) in 4G/5G requires various filters. (4) The existing detection for CP-OFDM can be directly employed as the detector of the proposed waveform. Lastly, the comprehensive simulation results demonstrate that GFB-OFDM outperforms CP-OFDM in terms of the out-of-band leakage, complexity and error performance.

Objective: This study assessed the pharmacokinetic (PK) interactions between clarithromycin (a P-glycoprotein [P-gp] inhibitor) and four direct oral anticoagulants (DOACs) (P-gp substrates) using physiologically based PK (PBPK) models to elucidate the influence of P-gp in the interaction between them. Methods: PBPK models for clarithromycin, DABE-dabigatran (DAB), rivaroxaban, apixaban, and edoxaban were constructed using GastroPlus™ (version 9.9), based on physicochemical data and PK parameters from the literature. The models were optimized and validated in healthy subjects. We evaluated the predictive performance of the established model and further assessed the impact of P-gp on the PK of the four DOACs. Successfully validated models were then used to evaluate potential drug-drug interactions (DDIs) between clarithromycin and the DOACs. Results: The established PBPK models accurately described the PK of clarithromycin, DABE-DAB, rivaroxaban, apixaban, and edoxaban. The predicted PK parameters (C max , T max , AUC 0-t ) were within 0.5-2 times the observed values. A sensitivity analysis of P-gp parameters indicated that an increase in P-gp expression was reduced by in vivo exposure to DOACs. The models demonstrated good predictive ability for DDIs between clarithromycin and the anticoagulants, and the ratio of the predicted values to the observed values of C max and the area under the curve (AUC) in the DDI state was within the range of 0.5-2. Conclusions: Comprehensive PBPK models for clarithromycin, DABE-DAB, rivaroxaban, apixaban, and edoxaban were developed, which can effectively predict DDIs mediated by P-gp's function. These models provide theoretical support for clinical dose adjustments and serve as a foundation for future PBPK model development for DOACs under specific pathological conditions.

To achieve effective consolidation of fine particles in moraine and enhance the freeze-thaw resistance of the consolidated body, this study developed a novel grouting material using sodium silicate, lipid-based curing agents, and acidic catalysts. The gelation time and rheological properties of this material were tested. The freeze-thaw resistance was studied through changes in uniaxial compressive strength (UCS) after freeze-thaw cycles, while the consolidation mechanism was analyzed using X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and scanning electron microscopy (SEM). The experimental results indicate that the material's gelation time can be controlled between 30s and 1600s, with an initial viscosity ranging from 5.9 to 9.8 mPa·s. Predictive models for these two indicators were established, and variance analysis revealed the influence ranking for gelation time: phosphoric acid dosage had the greatest effect, followed by EGDA content, with the Baume degree of sodium silicate having the least effect. The initial viscosity positively correlated with the Baume degree of sodium silicate and exhibited exponential growth over time. EGDA addition enhanced UCS by over 450%, reaching 1.2 MPa. During freeze-thaw cycles, strength degradation of the consolidated body was reduced by 10% to 30%. Microstructural tests showed that EGDA promotes silica gel formation and creates a network structure with unreacted sodium silicate, forming a dense consolidated body with moraine fine particles, thereby enhancing freeze-thaw resistance. These findings provide design references and theoretical support for moraine grouting in cold regions.

Trichoderma is a widely studied ascomycete fungal genus, including more than 400 species. However, genetic information on Trichoderma is limited, with most species reporting only DNA barcodes. Mitochondria possess their own distinct DNA that plays a pivotal role in molecular function and evolution. Here, we report 42 novel mitochondrial genomes (mitogenomes) combined with 18 published mitogenomes of Trichoderma . These circular mitogenomes exhibit sizes of 26,276-94,608 bp, typically comprising 15 core protein-coding genes (PCGs), 2 rRNAs, and 16-30 tRNAs; however, the number of endonucleases and hypothetical proteins encoded in the introns of PCGs increases with genome size enlargement. According to the result of phylogenetic analysis of the whole mitogenome, these strains diverged into six distinct evolutionary branches, supported by the phylogeny based on 2830 single-copy nuclear genes. Comparative analysis revealed that dynamic Trichoderma mitogenomes exhibited variations in genome size, gene number, GC content, tRNA copy, and intron across different branches. We identified three mutation hotspots near the regions encoding nad3 , cox2 , and nad5 that caused major changes in the mitogenomes. Evolutionary analysis revealed that atp9 , cob , nad4L , nad5 , and rps3 have been influenced by positive selection during evolution. This study provides a valuable resource for exploring the important roles of the genetic and evolutionary dynamics of Trichoderma mitogenome in the adaptive evolution of biocontrol fungi.

As the mainstream type of gas sensors, metal oxide semiconductor (MOS) gas sensors have garnered widespread attention due to their high sensitivity, fast response time, broad detection spectrum, long lifetime, low cost, and simple structure. However, the high power consumption due to the high operating temperature limits its application in some application scenarios such as mobile and wearable devices. At the same time, highly sensitive and low-power gas sensors are becoming more necessary and indispensable in response to the growth of the environmental problems and development of miniaturized sensing technologies. In this work, hierarchical indium oxide (In 2 O 3 ) sensing materials were designed and the pulse-driven microelectromechanical system (MEMS) gas sensors were also fabricated. The hierarchical In 2 O 3 assembled with the mass of nanosheets possess abundant accessible active sites. In addition, compared with the traditional direct current (DC) heating mode, the pulse-driven MEMS sensor appears to have the higher sensitivity for the detection of low-concentrations of nitrogen dioxide (NO 2 ). The limit of detection (LOD) is as low as 100 ppb. It is worth mentioning that the average power consumption of the sensor is as low as 0.075 mW which is one three-hundredth of that in the DC heating mode. The enhanced sensing performances are attributed to loose and porous structures and the reducing desorption of the target gas driven by pulse heating. The combination of morphology design and pulse-driven strategy makes the MEMS sensors highly attractive for portable equipment and wearable devices.

Heterogeneous Mg-Fe oxide/biochar (MgFeO@BC) nanocomposites were synthesized by a co-precipitation method and used as biochar-based catalysts to activate peroxymonosulfate (PMS) for sulfamethoxazole (SMX) removal. The optimal conditions for SMX degradation were examined as follows: pH 7.0, MgFeO@BC of 0.4 g/L, PMS concentration of 0.6 mM and SMX concentration of 10.0 mg/L at 25 ℃. In the MgFeO@BC/PMS system, the removal efficiency of SMX was 99.0% in water within 40 min under optimal conditions. In the MgFeO@BC/PMS system, the removal efficiencies of tetracycline (TC), cephalexin (CEX), ciprofloxacin (CIP), 4-chloro-3-methyl phenol (CMP) and SMX within 40 min are 95.3%, 98.4%, 98.2%, 97.5% and 99.0%, respectively. The radical quenching experiments and electron spin resonance (ESR) analysis suggested that both non-radical pathway and radical pathway advanced SMX degradation. SMX was oxidized by sulfate radicals (SO 4 •- ), hydroxyl radicals (•OH) and singlet oxygen ( 1 O 2 ), and SO 4 •- acted as the main active species. MgFeO@BC exhibits a higher current density, and therefore, a higher electron migration rate and redox capacity. Due to the large number of available binding sites on the surface of MgFeO@BC and the low amount of ion leaching during the catalytic reaction, the system has good anti-interference ability and stability. Finally, the intermediates of SMX were detected.

Studies using source localization results have shown that cortical involvement increased in treadmill walking with brain-computer interface (BCI) control. However, the reorganization of cortical functional connectivity in treadmill walking with BCI control is largely unknown. To investigate this, a public dataset, a mobile brain-body imaging dataset recorded during treadmill walking with a brain-computer interface, was used. The electroencephalography (EEG)-coupling strength of the between-region and within-region during the continuous self-determinant movements of lower limbs were analyzed. The time-frequency cross-mutual information (TFCMI) method was used to calculate the coupling strength. The results showed the frontal-occipital connection increased in the gamma and delta bands (the threshold of the edge was >0.05) during walking with BCI, which may be related to the effective communication when subjects adjust their gaits to control the avatar. In walking with BCI control, the results showed theta oscillation within the left-frontal, which may be related to error processing and decision making. We also found that between-region connectivity was suppressed in walking with and without BCI control compared with in standing states. These findings suggest that walking with BCI may accelerate the rehabilitation process for lower limb stroke.

Idiopathic pulmonary fibrosis (IPF) is a chronic, progressive lung disease with limited effective therapies. Interstitial macrophages (IMs), especially those derived from monocytes, play an unknown role in IPF pathogenesis. By using single-cell RNA sequencing (scRNA-seq), bleomycin (BLM)-induced pulmonary fibrosis mouse lungs were analyzed to characterize the cellular landscape and heterogeneity of macrophages in this model. scRNA-seq was used to identify distinct interstitial macrophage subpopulations in fibrotic lungs, with monocyte-derived macrophages exhibiting a pro-fibrotic gene expression profile enriched in wound healing, extracellular matrix (ECM) remodeling, and pro-fibrotic cytokine production functions. A pseudotime analysis revealed that IMs originated from monocytes and differentiated along a specific trajectory. A cell-cell communication analysis demonstrated strong interactions between monocyte-derived interstitial macrophages (Mo-IMs) and fibroblasts through the transforming growth factor beta (TGFβ), secreted phosphoprotein 1 (SPP1), and platelet-derived growth factor (PDGF) signaling pathways. Flow cytometry validated the presence and expansion of Mo-IMs subpopulations in BLM-treated mice. This study reveals the cellular heterogeneity and developmental trajectory of lung macrophages in early BLM-induced pulmonary fibrosis, highlighting the crucial role of Mo-IMs with a pro-fibrotic phenotype in IPF pathogenesis via interactions with fibroblasts. Targeting these specific macrophage subpopulations and associated signaling pathways may provide novel therapeutic strategies for IPF.

In this work, rice-husk-derived biochar (RBC) was synthesized by using simple one-step pyrolysis strategies and served as catalysts to activate peroxymonosulfate (PMS) for degrading sulfamethoxazole (SMX). When the annealing temperature (T) = 800 °C, RBC 800 exhibits the typical hardwood structure with several micropores and mesoporous. Furthermore, RBC 800 obtains more defect sites than RBC 600 , RBC 700 , and RBC 900 . In the RBC 800 /PMS system, the removal rate of the SMX reached 92.0% under optimal conditions. The kinetic reaction rate constant ( k obs ) of the RBC 800 /PMS system was 0.009 min -1 , which was about 1.50, 1.28, and 4.50 times that of the RBC 600 /PMS ( k obs = 0.006 min -1 ), RBC 700 /PMS ( k obs = 0.007 min -1 ), and RBC 900 /PMS ( k obs = 0.002 min -1 ) systems, respectively. In the RBC 800 /PMS system, sulfate radical (SO 4 •- ) is the main active species. Compared with other active sites, the hydroxyl group (C-OH) on the surface of RBC 800 interacts more strongly with PMS, which is more likely to promote the stretching of the O-O bond of the PMS, thus breaking into the activated state and significantly reducing the activation energy required for reaction. The degradation intermediates of SMX were speculated, and the toxicity analysis was conducted. Generally, this work reveals in depth the interaction between reactive sites of biochar-based catalysts and PMS at the molecular level.

The poor timeliness of routing has always been an urgent problem in practical operator networks, especially in situations with large-scale networks and multiple network domains. In this article, a pruning idea of routing integrated with Dijkstra's shortest path searching is utilized to accelerate the process of routing in large-scale multi-domain elastic optical networks (EONs). The layered-graph approach is adopted in the spectrum allocation stage. To this end, an efficient heuristic algorithm is proposed, called "Branch-and-Bound based Routing and Layered Graph based Spectrum Allocation algorithm (BBR-LGSA)", which is an integrated RSA algorithm. Notably, the significant reduction in algorithm time complexity is not only reflected in the pruning method used in the routing stage but also in the construction of auxiliary graphs during the spectrum allocation stage utilizing the Branch-and-Bound method. Simulation results show that the proposed BBR-LGSA significantly reduces the average running time by nearly 78% with higher spectrum utilization in large-scale multi-domain EONs, compared with benchmark algorithms. In addition, the impact of key parameters on performance comparisons of different algorithms is evaluated.

The flow of six kinds of fresh concrete under different flow rates and lubrication layer thickness (T LL ) values in the horizontal pipe was numerically simulated. The influence of the T LL on the pressure per unit length (P L ) was analyzed. It was determined that the formation of the lubrication layer (LL) significantly reduces the P L in concrete pumping. As the T LL increased, the P L decreased. However, the degree of reduction in the P L gradually decreased as the T LL increased. Relating the simulated P L with the experimental P L , the size of the T LL was obtained, which was between 1 and 3 mm. The minimum and maximum were 1.23 and 2.58 mm, respectively, and the average value was 1.97 mm. The strength (S24, S50), the size of the aggregate (A10, A20, A25), and the flow rate of pumping all affected the T LL . The type of fresh concrete and the flow rate of pumping significantly affected the P L , which impacted the T LL . However, the T LL also impacted the P L . Finally, this made the T LL change within a certain range. When P L > 14,000 Pa/m, 2 mm < T LL < 3 mm; on the other hand, 1 mm < T LL < 2 mm. Therefore, we can use CFD to simulate the flow of all types of concrete in the actual pumping pipeline with a T LL of 2 mm to obtain their pumping pressure and guide the actual construction.

Serine/arginine-rich (SR) proteins mostly function as splicing factors for pre-mRNA splicing in spliceosomes and play critical roles in plant development and adaptation to environments. However, detailed study about SR proteins in legume plants is still lacking. In this report, we performed a genome-wide investigation of SR protein genes in wild soybean ( Glycine soja ) and identified a total of 31 GsSR genes from the wild soybean genome. The analyses of chromosome location and synteny show that the GsSR s are unevenly distributed on 15 chromosomes and are mainly under the purifying selection. The GsSR proteins can be phylogenetically classified into six sub-families and are conserved in evolution. Prediction of protein phosphorylation sites indicates that GsSR proteins are highly phosphorylated proteins. The protein-protein interaction network implies that there exist numerous interactions between GsSR proteins. We experimentally confirmed their physical interactions with the representative SR proteins of spliceosome-associated components such as U1-70K or U2AF35 by yeast two-hybrid assays. In addition, we identified various stress-/hormone-responsive cis -acting elements in the promoter regions of these GsSR genes and verified their expression patterns by RT-qPCR analyses. The results show most GsSR genes are highly expressed in root and stem tissues and are responsive to salt and alkali stresses. Splicing analysis showed that the splicing patterns of GsSR s were in a tissue- and stress-dependent manner. Overall, these results will help us to further investigate the biological functions of leguminous plant SR proteins and shed new light on uncovering the regulatory mechanisms of plant SR proteins in growth, development, and stress responses.

In clinical settings, computed tomography (CT), magnetic resonance imaging (MRI), or positron emission tomography (PET) are commonly employed in brain imaging to assist clinicians in determining the type of stroke in patients. However, these modalities are associated with potential hazards or limitations. In contrast, microwave imaging emerges as a promising technique, offering advantages such as non-ionizing radiation, low cost, lightweight, and portability. The primary challenges faced by microwave tomography include the severe ill-posedness of the electromagnetic inverse scattering problem and the time-consuming nature and unsatisfactory resolution of iterative quantitative algorithms. This paper proposes a learning electric field enhancement imaging method (LEFEIM) to achieve quantitative brain imaging based on a microwave tomography system. LEFEIM comprises two cascaded networks. The first, based on a convolutional neural network, utilizes the electric field from the receiving antenna to predict the electric field distribution within the imaging domain. The second network employs the electric field distribution as input to learn the dielectric constant distribution, thereby realizing quantitative brain imaging. Compared to the Born Iterative Method (BIM), LEFEIM significantly improves imaging time, while enhancing imaging quality and goodness-of-fit to a certain extent. Simultaneously, LEFEIM exhibits anti-noise capabilities.

Identifying key genes involved in Cadmium (Cd) response pathways in plants and developing low-Cd-accumulating cultivars may be the most effective and eco-friendly strategy to tackle the problem of Cd pollution in crops. In our previous study, Stressseventy subfamily A 4 ( SSA4 ) was identified to be associated with Cd tolerance in yeast. Here, we investigated the mechanism of SSA4 in regulating Cd tolerance in yeast. ScSSA4 binds to POre Membrane 34 (POM34), a key component of nuclear pore complex (NPC), and translocates from the cytoplasm to the nucleus, where it regulates the expression of its downstream gene, Viable in a Hal3 Sit4 background 1 ( VHS1 ), resulting in reduced Cd accumulation in yeast cells. Additionally, we identified a Chinese cabbage SSA4 gene, BrSSA4c , which could enhance the Cd tolerance in Chinese cabbage. This study offers new insights into the regulatory mechanisms of Cd tolerance in yeast, a model organism, and paves the way for the genetic enhancement of Cd tolerance in Chinese cabbage.

The challenge of producing polymer vesicles remains difficult, despite numerous attempts to modulate the kinetics of polymer vesicle budding and achieve precise control over the membrane characteristics. An innovative approach that incorporates the use of copolymer-loaded single-emulsion droplets is proposed to address this challenge. This method enables the precise manipulation of micelles and polymer vesicles' composition, structures and dimensions. The emulsion contracts and forms microspheres when the copolymer concentrations exceed > 0.5 wt%, resulting in the formation of nano polymer vesicles. Conversely, the copolymer spontaneously forms micro polymer vesicles and micelles through vesicle budding at lower concentrations. The spontaneous production of vesicles and micelles can be induced by modifying the copolymer concentration in the emulsion. Our discoveries have a significant impact relative to the development of copolymer membranes and contribute to an enhanced comprehension of the mass manufacturing of polymer vesicles from single emulsions.

Drought stress is one of the main abiotic stresses that limit plant growth and affect fruit quality and yield. Plants primarily lose water through leaf transpiration, and wax effectively reduces the rate of water loss from the leaves. However, the relationship between water loss and the wax formation mechanism in goji ( Lycium barbarum ) leaves remains unclear. 'Ningqi I' goji and 'Huangguo' goji are two common varieties. In this study, 'Ningqi I' goji and 'Huangguo' goji were used as samples of leaf material to detect the differences in the water loss rate, chlorophyll leaching rate, wax phenotype, wax content, and components of the two materials. The differences in wax-synthesis-related pathways were analyzed using the transcriptome and metabolome methods, and the correlation among the wax components, wax synthesis genes, and transcription factors was analyzed. The results show that the leaf permeability of 'Ningqi I' goji was significantly lower than that of 'Huangguo' goji. The total wax content of the 'Ningqi I' goji leaves was 2.32 times that of the 'Huangguo' goji leaves, and the epidermal wax membrane was dense. The main components of the wax of 'Ningqi I' goji were alkanes, alcohols, esters, and fatty acids, the amounts of which were 191.65%, 153.01%, 6.09%, and 9.56% higher than those of 'Huangguo' goji, respectively. In the transcriptome analysis, twenty-two differentially expressed genes (DEGs) and six transcription factors (TFs) were screened for wax synthesis; during the metabolomics analysis, 11 differential metabolites were screened, which were dominated by lipids, some of which, like D-Glucaro-1, 4-Lactone, phosphatidic acid (PA), and phosphatidylcholine (PE), serve as prerequisites for wax synthesis, and were significantly positively correlated with wax components such as alkanes by the correlation analysis. A combined omics analysis showed that DEGs such as LbaWSD1 , LbaKCS1 , and LbaFAR2 , and transcription factors such as LbaMYB306 , LbaMYB60 , and LbaMYBS3 were strongly correlated with wax components such as alkanes and alcohols. The high expression of DEGs and transcription factors is an important reason for the high wax content in the leaf epidermis of 'Ningqi I' goji plants. Therefore, by regulating the expression of wax-synthesis-related genes, the accumulation of leaf epidermal wax can be promoted, and the epidermal permeability of goji leaves can be weakened, thereby reducing the water loss rate of goji leaves. The research results can lay a foundation for cultivating drought-tolerant goji varieties.

Axitinib (AXTN) is an oral tyrosine kinase inhibitor for the treatment of early to advanced renal cell carcinoma. In this work, solvates of AXTN were prepared in five solvents and subjected to desolvation treatment. The crystal form A of AXTN can form solvates in acetonitrile, DMF, acetic acid, acetic acid + water, and methanol. Different ratios of AXTN and acetic acid will form different products (solvate or directly crystallized into another crystal form (form IV)). The characterization results of thermal analyses confirmed the types of the five solvates. The obtained solvates were desolvated using methods of solid-phase desolvation (heating, exposure to solvent steam, microwave) and solvent-mediated phase transformation (SMPT). The desolvated solids were characterized by PXRD, TGA, DSC, FT-IR, and SEM, and it was ultimately inferred that a new crystal form (form Z) of AXTN could be formed after desolvation. In addition, the solvates obtained in this work experienced mutual transformation via SMPT, which depends on the type of solvents or mixed solvents. The phase transformations of different solid forms were summarized. This study is instructive for exploring solvates and polymorphs of AXTN and understanding phase transitions under different environments.

In this study, sodium alginate (SA) aerogel cross-linked with Ca 2+ was selected as the basic skeleton to construct a lightweight, flame retardant, and thermal insulating composite aerogel via modification with melamine and phytic acid. The resulting aerogel, SA-1.0 MP, achieved a thermal conductivity as low as 0.0379 W/(m·K). Compared to pristine SA aerogel, SA-1.0 MP demonstrated improved fire resistance, evidenced by a substantial increase in the limiting oxygen index (LOI) from 21.5% to 48.8% and a V-0 rating in the UL-94 test. Furthermore, a synergistic mechanism was proposed to explain its remarkable flame-retardant capability.

In this study, Fe, N co-doped biochar (Fe@N co-doped BC) was synthesized by the carbonization-pyrolysis method and used as a carbocatalyst to activate peroxymonosulfate (PMS) for sulfamethoxazole (SMX) removal. In the Fe@N co-doped BC/PMS system, the degradation efficiency of SMX (10.0 mg·L -1 ) was 90.2% within 40 min under optimal conditions. Radical quenching experiments and electron spin resonance (ESR) analysis suggested that sulfate radicals (SO 4 •- ), hydroxyl radicals ( • OH), and singlet oxygen ( 1 O 2 ) participated in the degradation process. After the reaction, the proportion of pyrrolic N decreased from 57.9% to 27.1%. Pyrrolic N served as an active site to break the inert carbon network structure and promote the generation of reactive oxygen species (ROS). In addition, pyrrolic N showed a stronger interaction with PMS and significantly reduced the activation energy required for the reaction (∆G = 23.54 kcal/mol). The utilization potentiality of Fe@N co-doped BC was systematically evaluated in terms of its reusability and selectivity to organics. Finally, the intermediates of SMX were also detected.

Objective: This study aims to investigate the association between the arteries and veins surrounding a pulmonary nodule and its malignancy., Methods: A dataset of 146 subjects from a LDCT lung cancer screening program was used in this study. AI algorithms were used to automatically segment and quantify nodules and their surrounding macro-vasculature. The macro-vasculature was differentiated into arteries and veins. Vessel branch count, volume, and tortuosity were quantified for arteries and veins at different distances from the nodule surface. Univariate and multivariate logistic regression (LR) analyses were performed, with a special emphasis on the nodules with diameters ranging from 8 to 20 mm. ROC-AUC was used to assess the performance based on the k-fold cross-validation method. Average feature importance was evaluated in several machine learning models., Results: The LR models using macro-vasculature features achieved an AUC of 0.78 (95% CI: 0.71-0.86) for all nodules and an AUC of 0.67 (95% CI: 0.54-0.80) for nodules between 8-20 mm. Models including macro-vasculature features, demographics, and CT-derived nodule features yielded an AUC of 0.91 (95% CI: 0.87-0.96) for all nodules and an AUC of 0.82 (95% CI: 0.71-0.92) for nodules between 8-20 mm. In terms of feature importance, arteries within 5.0 mm from the nodule surface were the highest-ranked among macro-vasculature features and retained their significance even with the inclusion of demographics and CT-derived nodule features., Conclusions: Arteries within 5.0 mm from the nodule surface emerged as a potential biomarker for effectively discriminating between malignant and benign nodules.

Non-thermal plasma (NTP) synergistic anticancer strategies are a current hotspot of interest at the intersection of plasma biomedicine. Melittin (MEL) has been shown to inhibit cancer in many malignant tumors; however, its clinical application is controversial. Therefore, the transmembrane process and mechanism of MEL activity in different cell systems were studied and the combination of MEL and NTP was proposed in this paper. The results showed that the electrostatic attraction between MEL and the lipid bilayer contributes to the stable orientation of MEL on the membrane surface. In addition, sialic acid overexpression affects the degree to which MEL binds the membrane system and the stability of the membrane structure. The use of NTP to reduce the dosage of MEL and its related nonspecific cytolysis activity has certain clinical application value. The results of this study provide theoretical support for improving the clinical applicability of MEL and contribute to the further development of plasma biomedicine.

The Mongolian sheep, emblematic of the Inner Mongolian grasslands, is renowned for its exceptional stress resistance and adaptability to harsh environments, drawing considerable attention. Recent research has unveiled the novel role of γ-aminobutyric acid (GABA) in combating oxidative stress. This investigation examined how GABA impacts renal-cortex and medulla cells from Mongolian sheep exposed to high-glucose stress conditions, utilizing gene expression analysis and non-targeted metabolomics. Elevated glucose levels significantly reduced the viability of Mongolian sheep renal cells and increased reactive oxygen species (ROS) levels. Conversely, the introduction of GABA notably enhanced cell viability, reduced ROS production, and stimulated the expression of antioxidant genes (e.g., Gpx , SOD , CAT ) in the renal cortex. In the renal medulla, CAT expression increased, while Gpx gene expression showed mixed responses. Metabolomics analysis indicated that high-glucose exposure altered various metabolites, whereas GABA alleviated the metabolic stress induced by high glucose through modulating glycolysis and the tricarboxylic acid cycle. In Mongolian sheep renal cells, GABA effectively mitigated oxidative damage triggered by high-glucose stress by upregulating antioxidant genes and regulating metabolic pathways, revealing insights into its potential mechanism for adapting to extreme environments. This finding offers a fresh perspective on understanding the stress resilience of Mongolian sheep and may provide valuable insights for research across diverse disciplines.

(1) Background: Target capture sequencing (TCS) is potentially a cost-effective way to detect single-nucleotide polymorphisms (SNPs) and an alternative to SNP array-based genotyping. (2) Methods: We evaluated the effectiveness and reliability of TCS in cattle breeding scenarios using 48 female and 8 male samples. DNA was extracted from blood samples, targeted for 71,746 SNPs with TWIST probes, and sequenced on an MGI platform. GATK and BCFtools were evaluated for the best genotyping calling tool. The genotypes were compared to existing genotypes from the Versa50K SNP array of the same animals by measuring accuracy as concordance (%) and R 2 . (3) Results: In this study, 71,553 SNPs and 166 indels were identified. The genotype comparison of 37,130 common SNPs between TCS and SNP arrays yielded high agreement, with a mean concordance of 98%, R 2 of 0.98 and Cohen's kappa of 0.97. The concordances of sex prediction, parent verification and validation of five genotype markers of interest important for Wagyu breeding were 100% between TCS and SNP array. The elements of the genomic relationship matrix (GRM) constructed from the SNP array and TCS data demonstrated a correlation coefficient approaching unity (r = 0.9998). (4) Conclusions: Compared to the SNP array, TCS is a comparable, cost-effective and flexible platform for genotyping SNPs, including non-model organisms and underrepresented commercial animal populations.

Background: Arterial thrombosis, a condition in which thrombi form in arteries, can lead to various acute cardiovascular diseases and impact the quality of life and survival of patients. Berberine (BBR), a quaternary ammonium alkaloid, has been shown to treat these diseases. However, further exploration is needed to understand underlying mechanisms of BBR., Methods and Results: Rats were administered BBR via intramuscular injection. Then, an FeCl 3 -coated filter paper was applied to a carotid artery to induce thrombosis. The size of the thrombus and the blood flow velocity were evaluated by carotid ultrasound. The shape of the thrombus was observed using staining and microscopy. The expression levels of mRNA and proteins were verified. Additionally, mass spectrometry and single-cell RNA sequencing analysis were conducted. The administration of BBR resulted in a significant reduction in the thrombus area and an extension of the thrombus-clogging time. Furthermore, BBR administration effectively reversed the decreasing tissue-plasminogen activator (t-PA) expression and alterations in fibrinolysis system of model group. Additionally, the expression of PKM2 was suppressed following BBR administration, and the overexpression of PKM2 inhibited t-PA expression., Conclusions: BBR ameliorates thrombosis by modulating expression of PKM2, subsequently impacting the expression of t-PA within fibrinolytic system. These preliminary findings suggest that BBR could be a potential preventive and therapeutic strategy for arterial thromboembolic diseases.

Maternal stress experienced during prenatal development is recognized as a significant risk factor for neurodevelopmental and neuropsychiatric disorders across the offspring's lifespan. The placental barrier serves a crucial function in safeguarding the fetus from detrimental exposures during gestation. However, previous investigations have not yet comprehensively elucidated the extensive connections between prenatal stress and the expression of placental proteins. In this study, we used iTRAQ-based quantitative proteomics to elucidate the placental adaptive mechanisms of pregnant rats in response to fear-induced stress. Our results showed that during pregnancy, exposure to fear-induced stress led to a pathological hypercoagulable state in the mother's body. Placental circulation was also disrupted, significantly reducing placental efficiency and blood oxygen saturation in newborn rats. Proteomic analyses showed that most of the DEPs were annotated to the PI3K-Akt and ECM-receptor interaction signaling pathway. In addition, the expressions of CDC37, HSP90β, AKT, p-AKT and p-mTOR were down-regulated significantly in the placenta. Our results demonstrated that prenatal fear-induced stress led to inhibition of the cellular signal transduction of placental PI3K/AKT/mTOR, which affected biological processes such as rRNA processing, translation, protein folding, protein stability, and oxygen transport in the placenta. These abnormalities in biological functions could potentially damage the barrier function of the placenta and thereby result in abnormal development in the offspring.

Sulforaphene (4-methylsufinyl-3-butenyl isothiocyanate, SFE), produced by myrosinase hydrolysis of glucoraphenin (4-methylsulfinyl-3-butenyl glucosinolate) found in radish seeds, is strongly associated with cancer prevention. In this study, we investigated the stability of SFE (purity above 98%) under various thiol-containing compounds at 25 °C, such as sodium hydrosulfide (NaHS), glutathione (GSH), and cysteine (Cys). We observed that the degradation of SFE was closely related to the presence and dissociation capacity of thiol-containing compounds in the solution, particularly the thiol group. We found that the degradation rate of SFE was influenced by incubation with NaHS, GSH, and Cys, with distinct degradation products detected for each of these thiol-containing compounds. Compared to GSH, sulfide and Cys played important roles in promoting the degradation of SFE. Furthermore, we found substantial quantities of hydrogen sulfide in conjunction with SFE during the hydrolysis process of seeds, and a heat treatment of the seeds resulted in increased production of SFE. However, the introduction of sulfide-oxidizing bacteria to the hydrolytic system did not exhibit any inhibitory effect on the degradation of SFE. These results provided a guideline for industries to improve the stability of SFE during preparation.
