Equus caballus papillomavirus 2 (EcPV2) is known to cause genital neoplasms in horses. However, reports on EcPV2 in Japan and Asia are limited. Herein, we present the histopathological and virological findings of the first reported case of an EcPV2-associated penile mass in Japan. The patient was a 22-year-old stallion with a history of breeding in Japan and abroad. Histopathological examination contained RNA in situ hybridization targeting the E6/E7 region and an immunohistochemical approach, and whole-genome sequencing was conducted within the viral examination. Proliferating epidermal cells were observed, and EcPV2 E6/E7 mRNA was detected within the epidermis, which was interpreted as viral papilloma. The detected EcPV2 virus was genetically close to foreign strains and different from the strain previously reported from a Japanese mare. This suggests that various types of EcPV2 might already exist among horses in Japan. Although the mass reported herein was not malignant based on histopathological findings and the absence of recurrence, its presence on the penis would be an obstacle to breeding. Simple Summary: Racehorses race and train in either a clockwise or anticlockwise direction and may adapt how they move accordingly, potentially creating asymmetry of movement between their left and right sides. Wireless inertial sensors measured the vertical head and pelvic movement of 307 Thoroughbreds (156 from the anticlockwise exercising Singapore turf club, "STC", and 151 from the clockwise exercising Hong Kong Jockey Club, "HKJC") trotting in a straight line in-hand on a firm surface. Seven vertical movement symmetry variables comparing the vertical movement between the left- and right-side halves were extracted, and statistical analyses were conducted to compare the number of left and right asymmetrical horses between cohorts, for both the fore- and hindlimbs. Our results revealed that there were significantly more left forelimb asymmetrical horses in the HKJC cohort compared to the STC cohort. Additional analyses revealed significant differences between cohorts, with the HKJC horses on average showing higher levels of left fore- and hindlimb asymmetry and, for the STC horses, right fore- and hindlimb asymmetry. This suggests that horses may indeed adapt their movement to favor one side over the other based on the direction they typically train and race in. Background: Racehorses commonly train and race in one direction, which may result in gait asymmetries. This study quantified gait symmetry in two cohorts of Thoroughbreds differing in their predominant exercising direction; we hypothesized that there would be significant differences in the direction of asymmetry between cohorts. Methods: 307 Thoroughbreds (156 from Singapore Turf Club (STC)—anticlockwise; 151 from Hong Kong Jockey Club (HKJC)—clockwise) were assessed during a straight-line, in-hand trot on firm ground with inertial sensors on their head and pelvis quantifying differences between the minima, maxima, upward movement amplitudes (MinDiff, MaxDiff, UpDiff), and hip hike (HHD). The presence of asymmetry (≥5 mm) was assessed for each variable. Chi-Squared tests identified differences in the number of horses with left/right-sided movement asymmetry between cohorts and mixed model analyses evaluated differences in the movement symmetry values. Results: HKJC had significantly more left forelimb asymmetrical horses (Head: MinDiff p < 0.0001, MaxDiff p < 0.03, UpDiff p < 0.01) than STC. Pelvis MinDiff (p = 0.010) and UpDiff (p = 0.021), and head MinDiff (p = 0.006) and UpDiff (p = 0.017) values were significantly different between cohorts; HKJC mean values indicated left fore- and hindlimb asymmetry, and STC mean values indicated right fore- and hindlimb asymmetry. Conclusion: the asymmetry differences between cohorts suggest that horses may adapt their gait to their racing direction, with kinematics reflecting reduced 'outside' fore- and hindlimb loading. This study compared the incidence of fatalities, falls and injuries in horses participating in hurdle and steeplechase races with those participating in flat races at the same race meets, for all jumps races in the 2022 and 2023 Thoroughbred horse jumps racing seasons in Victoria, Australia. Overall, horse fatalities, falls and injuries occurred at higher rates in jumps races compared with flat races during the study period. The continuation of jumps racing is contentious due to the higher risk of fatalities, falls and injuries for horses, compared with flat racing. While measures have been introduced by the industry to improve the safety of riders and horses, the rates of fatalities, falls and injuries in horses participating in jumps races have not been collectively reported in Australia since the 2012 to 2014 race seasons. Although information on individual horse fatalities, falls and injuries is published by Racing Victoria in Stewards' Reports, the data are not aggregated, and so cannot readily be used to assess trends or evaluate the efficacy of safety measures introduced by the industry. The aim of this study was to determine the fatality, fall and injury rates for horses participating in hurdle and steeplechase races in Victoria in the 2022 and 2023 Thoroughbred horse jumps racing seasons compared with horses participating in flat races at the same race meets. Data on horse fatalities, falls and injuries were extracted from the published Racing Victoria race results and Stewards' Reports for the jumps races (n = 150) and corresponding flat races (n = 157) held at the 38 jumps race meets in Victoria in 2022 and 2023. Overall, horse fatalities, falls and injuries occurred at higher rates in jumps races compared with flat races during the study period. The rate of horse fatalities in jumps races was 3.3 per 1000 starts, with no fatalities in flat races. The rate of horse falls in hurdle races was 24 per 1000 starts and 41.6 per 1000 starts in steeplechase races, comparable with rates previously reported in the 2012 to 2014 seasons. There were no falls in flat races. Horse injuries occurred at a rate of 68.9 per 1000 starts in jumps races compared with 18.8 per 1000 starts in flat races. In hurdle and steeplechase races, veterinary clearance being required following horse injury was 5.4 times (OR 5.4, 95% CI 2.8–10.2) and 7.2 times (OR 7.2, 95% CI 3.3–15.6) more likely, respectively, compared with flat races. The risk of trauma was 4 times more likely in hurdle and steeplechase races (OR 4.8, 95% CI 1.7–13.3 and OR 4.1, 95% CI 1.2–13.4, respectively) and the risk of lameness was increased by 2.5 times in hurdles (OR 2.5, 95% CI 1.2–5.2) and 5.1 times in steeplechase races (OR 5.1, 95% CI 2.3–11.5), compared with flat races. Simple Summary: There is a lack of understanding as to whether the bacteria isolated from swabs of the surface of the lining of broodmares' uteri (endometrial swabs) are present due to infection or contamination, or whether they are, in fact, part of the normal flora. This study aimed to understand this further by investigating endometrial swabs taken from a population of Thoroughbred broodmares and submitted to a laboratory in the UK between 2014 and 2020; by describing bacterial prevalence (the estimated percentage of swabs from which bacteria are isolated); and by evaluating whether bacterial prevalence changed over time or between mares of different ages and with differing amounts of inflammation of the uterine lining. Over the 7-year period, 18,996 endometrial swabs were submitted from 6050 mares on 290 farms. The overall prevalence of bacteria was 35.5%. Beta-hemolytic Streptococcus (17.9%) and E. coli (10.3%) were the most prevalent types of bacteria. The prevalence of bacteria changed over time (between different years in which mares were sampled) and was also impacted by the mare age and by whether there was inflammation of the uterine lining at the time of sampling. Results from this study provide up-to-date prevalence estimates and suggest that the interpretation of Thoroughbred endometrial swab findings is complicated. Determining whether endometrial microbial isolates are pathogens, contaminants, or even part of the "normal" microbiome is extremely complex, particularly given the absence of "gold standard" tests for endometritis. Population-level benchmarking and temporal monitoring can provide novel insights and a wider context to improve understanding. This study aimed to (i) estimate the prevalence of endometrial isolates from swabs of Thoroughbred broodmares in Newmarket, UK between 2014 and 2020; and (ii) evaluate the effects of year, mare age, and cytology findings on isolate prevalence. Generalised linear mixed models with a logit link, both null models and models using year of sampling, mare age, or cytology findings as predictors, were fitted to estimate isolate prevalence. Over the 7-year period, data were available from 18,996 endometrial-swab samples from 6050 mares on 290 premises. The overall isolate prevalence was 35.5% (95% confidence interval (CI) 33.0–37.9), and this varied significantly between years. The most prevalent isolates were β-hemolytic Streptococcus (17.9; 95% CI: 17–19) and E. coli (10.3%; 95% CI: 9.0–11.6). Isolate prevalence increased with mare age except for E. coli isolates, and with increasing category of cytology findings except for α-hemolytic Streptococcus isolates. Simple Summary: The concept of altitude training became popular among human athletes following the 1968 Olympic Games, at which African runners were particularly successful. Culminating from these observations was the concept that during exercise training, local tissue hypoxia is an important adaptive stress for muscle that ultimately leads to superior physiological adaptations and enhanced endurance performance. The application of the concept of hypoxic training to the Thoroughbred horse is new, and now, with purpose-built hypoxic chambers, there has been a growing interest in its use in equine training programs. This paper provides a comprehensive discussion on the physiological impacts of hypoxic training, its benefits to endurance performance, and a rationale for utilizing it to improve performance in the equine athlete. All exercise-induced training adaptations are governed by genetics. Exercise prescriptions can be tailored to elicit the desired physiological adaptations. Although the application of hypoxic stimuli on its own is not ideal to promote favorable molecular responses, exercise training under hypoxic conditions provides an optimal environment for maximizing physiological adaptations to enhance endurance performance. The combination of exercise training and hypoxia increases the activity of the hypoxia-inducible factor (HIF) pathway compared to training under normoxic conditions. Hypoxia-inducible factor-1 alpha (HIF-1α) is known as a master regulator of the expression of genes since over 100 genes are responsive to HIF-1α. For instance, HIF-1-inducible genes include those critical to erythropoiesis, angiogenesis, glucose metabolism, mitochondrial biogenesis, and glucose transport, all of which are intergral in physiological adaptations for endurance performance. Further, hypoxic training could conceivably have a role in equine rehabilitation when high-impact training is contraindicated but a quality training stimulus is desired. Simple Summary: Vitamin D biology in equids is unique and poorly understood. Naturally managed (grazing) horses rely on dietary vitamin D2 (ergocalciferol) to provide adequate vitamin D, because endogenous ultraviolet radiation-mediated synthesis of vitamin D3 (cholecalciferol) is ineffective in this species. To test the hypothesis that the management of stabled, non-grazing racehorses is a risk factor for low vitamin D status, the vitamin D status of non-grazing Thoroughbred racehorses in Hong Kong (HK) and grazing Thoroughbred racehorses in the United Kingdom (UK) was compared. The HK horses had lower serum concentrations of 25-hydroxyvitamin D2 (25OHD2) and total 25-hydroxyvitamin D (total 25OHD: the index of vitamin D status), reflecting reduced dietary vitamin D2 intake. These data indicate that HK racehorses required dietary vitamin D3 supplementation to maintain adequate vitamin D status. The inverse relationship between the serum concentrations of 25OHD2 and 25OHD3, previously identified in humans, was observed for the first time in horses, indicating that further study is needed to determine the optimal form of dietary vitamin D supplementation for Thoroughbred racehorses. There is a paucity of data relating to the vitamin D status of racehorses. We hypothesised that the management of racehorses in Hong Kong (HK) predisposes to low vitamin D status unless they receive dietary supplementation. Serum concentrations of 25-hydroxyvitamin D2 (25OHD2), 25-hydroxyvitamin D3 (25OHD3) and total 25-hydroxyvitamin D (total 25OHD) for 79 non-grazing HK racehorses were compared with those for 22 racehorses training in the United Kingdom (UK) that grazed for ≥1 h/d, and for which published data exists. A nested group of 41 HK horses was sampled twice to determine the effect of the duration in HK on vitamin D status. The HK horses had significantly lower serum concentrations of total 25OHD and 25OHD2 than the UK horses; 25OHD2 was undetectable in 15/79 HK sera and serum concentrations of 25OHD2 declined with the duration in HK. The main determinants of vitamin D status were assessed using linear regression; the retained variables were the 25OHD3 concentration and the duration in HK. The inverse relationship between the serum concentrations of 25OHD2 and 25OHD3, previously identified in humans, was observed for the first time in horses. In conclusion, HK racehorses have low serum 25OHD2 and total 25OHD concentrations and rely on D3 supplementation to maintain adequate vitamin D status. In the genomes of diploid organisms, runs of homozygosity (ROH), consecutive segments of homozygosity, are extended. ROH can be applied to evaluate the inbreeding situation of individuals without pedigree data and to detect selective signatures via ROH islands. We sequenced and analyzed data derived from the whole-genome sequencing of 97 horses, investigated the distribution of genome-wide ROH patterns, and calculated ROH-based inbreeding coefficients for 16 representative horse varieties from around the world. Our findings indicated that both ancient and recent inbreeding occurrences had varying degrees of impact on various horse breeds. However, recent inbreeding events were uncommon, particularly among indigenous horse breeds. Consequently, the ROH-based genomic inbreeding coefficient could aid in monitoring the level of inbreeding. Using the Thoroughbred population as a case study, we discovered 24 ROH islands containing 72 candidate genes associated with artificial selection traits. We found that the candidate genes in Thoroughbreds were involved in neurotransmission (CHRNA6, PRKN, and GRM1), muscle development (ADAMTS15 and QKI), positive regulation of heart rate and heart contraction (HEY2 and TRDN), regulation of insulin secretion (CACNA1S, KCNMB2, and KCNMB3), and spermatogenesis (JAM3, PACRG, and SPATA6L). Simple Summary: Racehorse welfare has generated public attention for both on-track treatment and practices, as well as the fate of racehorses upon completion of their racing careers. As such, the successful transition from racing into sport horse and other disciplines is paramount to the welfare of retired racehorses whose racing career lasts, on average, just 4.5 years. Thoroughbreds are frequently used in sport horse disciplines, and the consistent supply of retiring racehorses provide an affordable and athletic breed option for equestrians. This study analyzes demand for thoroughbreds offered for sale through online, sporthorse auctions. The results show buyer preferences for coat color, age, sex, and those registered with USEF, USHJA, or USEA. These findings support the value of thoroughbreds to buyers interested in sport horse disciplines. The results from this study may help non-profit, off-the-track thoroughbred rehoming facilities, reduce the number of unwanted thoroughbreds, and improve the public image of welfare in the racing industry. Racehorse welfare is a prominent, public issue which affects nearly every aspect of the racing industry. Thoroughbred care after race career completion has garnered increasing attention from the equine industry, general public, and animal welfare groups alike. As the average racehorse's career lasts just 4.5 years, owner demand for thoroughbreds is essential for post-race careers and acceptable welfare standards. This study utilized data from and hedonic pricing models to analyze buyer demand for thoroughbreds sold in online auctions held from 2012 to 2020. The results indicate buyer preferences for age (p < 0.01), sex (p < 0.05), and organization registration (p < 0.05), with bid price premiums for age and registration status (USEF, USEA, USHJA, etc.) and price discounts for mares compared to geldings and horses listed for non-competition careers (trail, p < 0.01). The results of this study confirm and quantify the value potential buyers place on thoroughbreds offered for sale in sport disciplines. Simple Summary: By using the Thoroughbred genome variant database to identify orthologues to personality-related human genes, we identified 18 potential personality-related genes in horses. These candidate genes have a total of 55 variants that cause amino acid substitutions when compared to the EquCab3.0 reference genome that may impact the function of the proteins encoded by these genes. Moreover, 15 of the 18 genes have not previously been linked to personality in horses, suggesting that this exploratory approach of related genes using human evidence can be useful for equine behavioral genetics. Although using this bioinformatics approach is less useful for investigating genes affecting personality in horses than it is in humans due to a lack of supporting personality research, this study highlights the potential for the identification of candidate genes. If future studies with equine behavioral datasets validate these potential personality–gene associations, this bioinformatics strategy may become important in the field of equine genetics. Considering the personality traits of racehorses (e.g., flightiness, anxiety, and affability) is considered essential to improve training efficiency and decrease accident frequency, especially when retraining for a second career that may involve contact with inexperienced personnel after retiring from racing. Studies on human personality-related genes are frequently conducted; however, such studies are rare in horses because a consistent methodology for personality evaluation is lacking. Using the recently published whole genome variant database of 101 Thoroughbred horses, we compared horse genes orthologous to human genes related to the Big Five personality traits, and identified 18 personality-related candidate genes in horses. These genes include 55 variants that involve non-synonymous substitutions that highly impact the encoded protein. Moreover, we evaluated the allele frequencies and functional impact on the proteins in terms of the difference in molecular weights and hydrophobicity levels between reference and altered amino acids. We identified 15 newly discovered genes that may affect equine personality, but their associations with personality are still unclear. Although more studies are required to compare genetic and behavioral information to validate this approach, it may be useful under limited conditions for personality evaluation. Simple Summary: The association between poor performance and respiratory disease in Thoroughbred racehorses that do not have a structural abnormality of the respiratory tract is often based on anecdotal evidence. We examined peer reviewed publications to determine if there was scientific evidence to link conditions such as inflammation of the airways, asthma, tracheal mucous, and exercise-induced pulmonary haemorrhage (EIPH) with decreased athletic performance. This is a complex field and studies have yielded conflicting results as to the impact of such conditions on performance. For example, some investigators found a significant association between poor racing performance and low-grade EIPH and mild to moderate asthma. In contrast, others have suggested that they may represent a normal response to training and the stabling environment. The key outcome of the review was that there is a dearth of studies unequivocally linking non-structural airway disease with poor performance, and there is a lack of an internationally harmonised approach to the assessment of racehorse performance. Improved investigations providing better quality evidence would facilitate comparison across studies, increase our understanding of the conditions associated with poor performance, safeguard horse welfare, and assist trainers to achieve their goals. The association between poor performance and respiratory disease in Thoroughbred racehorses that do not have a structural abnormality of the respiratory tract, is often based on anecdotal evidence. The objective of this scoping review was to examine the scientific evidence for such associations. Publications were selected based on a search of three databases (PubMed, Scopus, and CAB Direct), in English and without date restriction, followed by a screening process to exclude non-relevant papers, duplicates, and reviews. This process identified 996 publications of which 20 were analysed using the Quality in Prognosis Studies (QUIPS) tool. The results indicated that the evidence supporting the relationship between proposed diagnostic indicators and poor performance is variable. There is a need for better quality evidence. In particular, there are conflicting reports relating to the impact of equine asthma and EIPH on athletic performance. Furthermore, a lack of standardisation in the measurement of racehorse performance makes it difficult to compare findings from different studies. The industry would benefit from high-level guidance concerning the design of controlled performance studies in Thoroughbred racehorses to collect comprehensive data and facilitate targeted interventions. Simple Summary: The objective of this study is to detect copy number variations (CNVs) in four horse populations (Jeju horses, Thoroughbreds, Jeju riding horses, and Hanla horses) in South Korea. We found a total of 843 CNV regions (CNVRs) (164.3 Mb), which coincided with 7.2% of the reference horse genome. Overall, copy number losses were found more than gains and mixed CNVRs. A comparison of the CNVRs among the populations showed that a substantial number of CNVRs overlapped each other, while some CNVRs were found specifically in each population. We retrieved parts of CNVRs that overlapped with genes; these overlapping areas are potentially associated with traits of interest in horses. The Thoroughbred and crossbred populations had shared CNVRs overlapping with QTLs (Quantitative trait loci) that were associated with withers height and racing performance. Using gene ontology (GO) analysis, a total of 1884 functional genes were identified within the 577 CNVRs. GO analysis further showed that several of the genes are involved in the olfactory pathway and the nervous system. In this study, genome-wide CNVs were identified using a total of 469 horses from four horse populations (Jeju horses, Thoroughbreds, Jeju riding horses, and Hanla horses). We detected a total of 843 CNVRs throughout all autosomes: 281, 30, 301, and 310 CNVRs for Jeju horses, Thoroughbreds, Jeju riding horses, and Hanla horses, respectively. Of the total CNVRs, copy number losses were found to be the most abundant (48.99%), while gains and mixed CNVRs accounted for 41.04% and 9.96% of the total CNVRs, respectively. The length of the CNVRs ranged from 0.39 kb to 2.8 Mb, while approximately 7.2% of the reference horse genome assembly was covered by the total CNVRs. By comparing the CNVRs among the populations, we found a significant portion of the CNVRs (30.13%) overlapped; the highest number of shared CNVRs was between Hanla horses and Jeju riding horses. When compared with the horse CNVRs of previous studies, 26.8% of CNVRs were found to be uniquely detected in this study. The CNVRs were not randomly distributed throughout the genome; in particular, the Equus caballus autosome (ECA) 7 comprised the largest proportion of its genome (16.3%), while ECA 24 comprised the smallest (0.7%). Furthermore, functional analysis was applied to CNVRs that overlapped with genes (genic-CNVRs); these overlapping areas may be potentially associated with the olfactory pathway and nervous system. Simple Summary: Shiitake mushrooms (Lentinula edodes) are a valuable source of many important nutrients, vitamins and minerals. However, there are currently no standard protocols for testing the efficacy of shiitake mushrooms in equine nutritional supplementation. In total, 20 Thoroughbreds were divided into either the supplemented group or the control group. The supplemented group was given 30 g of L. edodes daily for four months. One blood sample was collected four times from each horse at four-week intervals. Our study showed differences between individual days of blood sampling in the supplement and control groups. Blood hematology showed that supplementation had an effect on monocytes. In a biochemical analysis, the shiitake mushroom affected the level of alkaline phosphatase, glucose, aspartate aminotransferase, lactic acid and cholesterol levels measured in the individual days of blood sampling. Our study is the first to demonstrate the influence of shiitake mushroom supplementation on blood parameters in young racehorses undergoing regular training. The greatest effects of supplementation were found for monocytes and alkaline phosphatase. Further studies are needed to show the influence of supplementation with shiitake mushrooms in larger groups of horses over a longer period. The aim of this study was to evaluate the effect of shiitake mushroom (Lentinula edodes) supplementation on the hematology and biochemical blood parameters of young Thoroughbred racehorses. The study was conducted with 20 horses divided into two groups: the supplemented and the control group. The supplemented group was given 30 g of L. edodes daily for four months. One blood sample was collected four times from each horse at four-week intervals. The hematology analysis in the supplemented group showed a higher level of monocytes at day 56 when compared to the control group (p = 0.000986). Biochemical analysis showed that alkaline phosphatase is most sensitive to shiitake mushroom supplementation, with statistically significant lower levels in supplemented group compared to the control group on all individual days of blood sampling. It was also found that supplementation had an effect on the decrease of glucose levels on days 28 (p = 0.009109) and 56 y (p = 0.025749), on reduction aspartate aminotransferase level on day 56 (p = 0.017258) and a decrease of lactic acid on day 28 of sampling (p = 0.037636). Cholesterol levels decreased consistently in all individual days of blood sampling. Simple Summary: Mitigating the risk of catastrophic injuries in the horse racing industry remains a challenge. Non-invasive methods such as CT imaging in combination with machine learning could be used to screen horses at risk of injury, but there remain questions on the feasibility of such an approach. In this work, we investigated whether machine learning models could be developed from in vitro harvested μCT images of intact proximal sesamoid bones to predict whether the bone was from a horse that suffered a catastrophic injury or from a control group. The average accuracy in differentiating whether a sesamoid bone came from a case or control horse using our approach was 0.754. Our work suggests it may be possible to develop similar models using CT images of horses in the clinical setting. Proximal sesamoid bone (PSB) fractures are the most common musculoskeletal injury in race-horses. X-ray CT imaging can detect expressed radiological features in horses that experienced catastrophic fractures. Our objective was to assess whether expressed radiomic features in the PSBs of 50 horses can be used to develop machine learning models for predicting PSB fractures. The μCTs of intact contralateral PSBs from 50 horses, 30 of which suffered catastrophic fractures, and 20 controls were studied. From the 129 intact μCT images of PSBs, 102 radiomic features were computed using a variety of voxel resampling dimensions. Decision Trees and Wrapper methods were used to identify the 20 top expressed features, and six machine learning algorithms were developed to model the risk of fracture. The accuracy of all machine learning models ranged from 0.643 to 0.903 with an average of 0.754. On average, Support Vector Machine, Random Forest (RUS Boost), and Log-regression models had higher performance than K-means Nearest Neighbor, Neural Network, and Random Forest (Bagged Trees) models. Model accuracy peaked at 0.5 mm and decreased substantially when the resampling resolution was greater than or equal to 1 mm. We find that, for this in vitro dataset, it is possible to differentiate between unfractured PSBs from case and control horses using μCT images. The creation of genetically modified horses is prohibited in horse racing as it falls under the banner of gene doping. In this study, we developed a test to detect gene editing based on amplicon sequencing using next-generation sequencing (NGS). We designed 1012 amplicons to target 52 genes (481 exons) and 147 single-nucleotide variants (SNVs). NGS analyses showed that 97.7% of the targeted exons were sequenced to sufficient coverage (depth > 50) for calling variants. The targets of artificial editing were defined as homozygous alternative (HomoALT) and compound heterozygous alternative (ALT1/ALT2) insertion/deletion (INDEL) mutations in this study. Four models of gene editing (three homoALT with 1-bp insertions, one REF/ALT with 77-bp deletion) were constructed by editing the myostatin gene in horse fibroblasts using CRISPR/Cas9. The edited cells and 101 samples from thoroughbred horses were screened using the developed test, which was capable of identifying the three homoALT cells containing 1-bp insertions. Furthermore, 147 SNVs were investigated for their utility in confirming biological parentage. Of these, 120 SNVs were amenable to consistent and accurate genotyping. Surrogate (nonbiological) dams were excluded by 9.8 SNVs on average, indicating that the 120 SNV could be used to detect foals that have been produced by somatic cloning or embryo transfer, two practices that are prohibited in thoroughbred racing and breeding. Simple Summary: Multiple pregnancies (MPs), or twins/triplets, are commonly diagnosed in pregnant mares. Whilst some studies have identified factors associated with an increased risk of MPs, they have only looked at each factor individually and not accounted for the effects of factors when they occur together. This study used statistical modelling to identify risk factors for MPs, looking at 27 factors as well as the contribution of the mare, stallion (both proxies for genetics), farm, and veterinarian. We found that multiple ovulations and the use of a drug that mimics prostaglandin F2α to induce oestrus, both increased the risk of a mare having an MP. Mares that had a foal that same year, had a uterine cyst, or who did not get pregnant on the first cycle they were bred on were at a decreased risk of having an MP. Factors that impact the early embryonic environment are more important influences of multiple pregnancies when compared to the genetics of the mare. The increased incidence of MP but not MOs over the previous decades may well reflect improved management of the endometrium as opposed to selection of mares with increased risk for MPs. A limitation of the study is the reliance on clinical records being completed accurately. Multiple pregnancies (MPs) are commonly diagnosed during breeding management of mares. Whilst some studies have reported on factors associated with the risk of MPs, few have utilised multivariable data analysis to control for confounding variables. A prospective cohort study of Thoroughbred broodmares was conducted with information collected on 27 factors. Mixed-effects logistic regression was used to determine risk factors for MPs. Mare, stallion, stud, and veterinarian were evaluated as random effects. The prevalence of MPs in 1754 mares and 2245 pregnancies was 16.06% (95% confidence interval [CI] = 14.54, 17.58). Multiple ovulations (OR = 15.57, 95% CI = 11.88, 20.53) and treatment with cloprostenol (OR = 1.35, 95% CI = 1.015, 1.80) were associated with increased odds of MPs following multivariable analysis. Mares that foaled at the start of the breeding season (OR = 0.66, 95% CI = 0.47, 0.94), conceived at the second or more oestrus cycles (OR = 0.60, 95% CI= 0.43, 0.84), or identified with a uterine cyst (OR = 0.63, 95% CI = 0.40, 0.97) were at reduced odds of conceiving MPs. Mare, stallion, stud, and veterinarian were not associated with MPs. These findings provide possible explanations as to why the prevalence of MPs but not MOs have increased over the last decade. Simple Summary: Shiitake mushrooms (Lentinula edodes) have remarkable health properties that have been used in Far Eastern medicine for centuries. There is limited evidence that L. edodes has a direct effect on cell metabolism, with overall beneficial effects on animal health. As shiitake mushroom products appear to have a broad spectrum of effects, from changes in the immune response to changes in performance, a study was conducted to evaluate the effects of shiitake mushroom supplementation on the blood morphological and biochemical parameters in horses. This pilot study showed that there were single statistical differences between sessions between the supplemented and the control groups. The blood morphology showed that shiitake mushroom supplementation had an effect on white blood cells, hemoglobin, and hematocrit levels. In the biochemical analysis, shiitake mushrooms affected the levels of alkaline phosphatase, calcium, gamma-glutamyl transferase, bilirubin, and glucose and the albumin/globulin ratio. The observed differences between the supplemented and the control group sessions suggest that shiitake mushrooms may be a beneficial nutritional supplement for horses. Shiitake mushrooms have been highly regarded as possessing enormous nutritive and medicinal values. No clinical studies have yet investigated the effect of shitake supplementation on the health of horses. The aim of this study was to evaluate the effect of shiitake mushroom supplementation on the morphological and biochemical blood properties in horses. A total of 17 adult horses were divided into two groups: supplemented and control. The supplemented group was fed 60 g of shiitake mushrooms per day for 5 months. Blood samples were collected in five sessions. Blood morphological analysis showed higher levels of lymphocytes in session 3 and monocytes in session 4 in the supplemented group. In addition, basophils, hemoglobin, and hematocrit were elevated compared to the control group. Biochemical analysis showed that the shiitake mushrooms affected a large number of parameters. In particular, alkaline phosphatase was found to be the most sensitive to shitake mushroom supplementation, for which the statistical differences were significant for sessions 2, 4, and 5. Furthermore, calcium was found to be affected by supplementation only in session 4, and gamma-glutamyl transferase in session 2. In addition, the bilirubin and glucose levels were lower in the supplemented group, and the albumin/globulin ratio was higher compared to the control group. (1) Background: The intestinal microbiota plays an essential role in maintaining the host’s health. Dysbiosis of the equine hindgut microbiota can alter the fermentation patterns and cause metabolic disorders. (2) Methods: This study compared the fecal microbiota composition of horses with intestinal disease and their healthy counterparts living in Korea using 16S rRNA sequencing from fecal samples. A total of 52 fecal samples were collected and divided into three groups: horses with large intestinal disease (n = 20), horses with small intestinal disease (n = 8), and healthy horses (n = 24). (3) Results: Horses with intestinal diseases had fewer species and a less diverse bacterial population than healthy horses. Lactic acid bacteria, Lachnospiraceae, and Lactobacillaceae were overgrown in horses with large intestinal colic. The Firmicutes to Bacteroidetes ratio (F/B), which is a relevant marker of gut dysbiosis, was 1.94, 2.37, and 1.74 for horses with large intestinal colic, small intestinal colic, and healthy horses, respectively. (4) Conclusions: The overgrowth of two lactic acid bacteria families, Lachnospiraceae and Lactobacillaceae, led to a decrease in hindgut pH that interfered with normal fermentation, which might cause large intestinal colic. The overgrowth of Streptococcus also led to a decrease in pH in the hindgut, which suppressed the proliferation of the methanogen and reduced methanogenesis in horses with small intestinal colic. The objective was to describe the incidence of failure to finish a race in flat-racing Thoroughbreds in New Zealand as these are summary indicators of falls, injuries and poor performance. Retrospective data on six complete flat racing seasons (n = 188,615 race starts) of all Thoroughbred flat race starts from 1 August 2005 to 31 July 2011 were obtained. The incidence of failure to finish events and binomial exact 95% confidence intervals were calculated per 1000 horse starts. The association between horse-, rider- and race-level variables with the outcomes failure to finish, pulled-up/fell and lost rider were examined with a mixed effects Poisson regression model. A total of 544 horses failed to finish in 188,615 race starts with an overall incidence of 2.88 per 1000 horse starts (95% CI 2.64–3.12). The incidence of failure to finish horses across each race year showed little variability. In the univariable analysis race distance, larger field size, season, and ratings bands showed association with failing to finish a race. The overall failure to finish outcome was associated with season, race distance and ratings bands (horse experience and success ranking criteria). In the multivariable analysis, race distance and ratings bands were associated with horses that pulled-up/fell; season, apprentice allowances and ratings bands were associated with the outcome lost rider. The failure to finish rate was lower than international figures for race day catastrophic injury. Racing and environmental variables were associated with failure to finish a race highlighting the multifactorial nature of race-day events. Further investigation of risk factors for failure to finish is required to better understand the reasons for a low failure to finish rate in Thoroughbred flat races in New Zealand. Thoroughbred jump racing sits in the spotlight of contemporary welfare and ethical debates about horse racing. In Australia, jump racing comprises hurdle and steeplechase races and has ceased in all but two states, Victoria and South Australia. This paper documents the size, geography, composition, and dynamics of Australian jump racing for the 2012, 2013, and 2014 seasons with a focus on debate about risks to horses. We found that the majority of Australian jump racing is regional, based in Victoria, and involves a small group of experienced trainers and jockeys. Australian jump horses are on average 6.4 years of age. The jump career of the majority of horses involves participating in three or less hurdle races and over one season. Almost one quarter of Australian jump horses race only once. There were ten horse fatalities in races over the study period, with an overall fatality rate of 5.1 fatalities per 1000 horses starting in a jump race (0.51%). There was significant disparity between the fatality rate for hurdles, 0.75 fatalities per 1000 starts (0.075%) and steeplechases, 14 fatalities per 1000 starts (1.4%). Safety initiatives introduced by regulators in 2010 appear to have significantly decreased risks to horses in hurdles but have had little or no effect in steeplechases. Simple Summary: Early detection of racehorses at risk of stress fracture is key to reducing the number of horses with catastrophic fractures while racing. Bone changes are often visible in the limbs of Thoroughbred racehorses in work, particularly in the fetlock region. However, it is currently unknown whether some of these changes indicate an impending fracture or are a healthy adaptation to high-speed exercise. This study looks at imaging and gross changes in a specific area (parasagittal grooves (PSGs) of the cannon bone) and the utility of X-ray, computed tomography (CT) and magnetic resonance imaging (MRI) to detect the changes. All fetlock joints were assessed from twenty horses that died during racing or training, including horses with and without fetlock fracture. Overall, X-ray was poor for detecting PSG changes. Some PSG changes on CT and MRI were common in Thoroughbred racehorses and possibly represent normal bone adaptation when seen in clinical cases. However, certain CT and MRI findings were more prevalent in horses with a fracture, possibly indicating microdamage accumulation and increased risk of fracture. Bilateral advanced imaging is recommended in clinical cases of suspected fetlock pathology. (1) Background: Parasagittal groove (PSG) changes are often present on advanced imaging of racing Thoroughbred fetlocks and have been suggested to indicate increased fracture risk. Currently, there is limited evidence differentiating the imaging appearance of prodromal changes in horses at risk of fracture from horses with normal adaptive modelling in response to galloping. This study aims to investigate imaging and gross PSG findings in racing Thoroughbreds and the comparative utility of different imaging modalities to detect PSG changes. (2) Methods: Cadaver limbs were collected from twenty deceased racing/training Thoroughbreds. All fetlocks of each horse were examined with radiography, low-field magnetic resonance imaging (MRI), computed tomography (CT), contrast arthrography and gross pathology. (3) Results: Horses with fetlock fracture were more likely to have lateromedial PSG sclerosis asymmetry and/or lateral PSG lysis. PSG lysis was not readily detected using MRI. PSG subchondral bone defects were difficult to differentiate from cartilage defects on MRI and were not associated with fractures. The clinical relevance of PSG STIR hyperintensity remains unclear. Overall, radiography was poor for detecting PSG changes. (4) Conclusions: Some PSG changes in Thoroughbred racehorses are common; however, certain findings are more prevalent in horses with fractures, possibly indicating microdamage accumulation. Simple Summary: The training and racing schedules of Thoroughbred racehorses vary within a racing jurisdiction. Changes in regulation at a local or national level can alter the frequency of racing, age profile and the number of and duration of spells (rest periods) for horses in active race training. In order to consider any intervention to reduce injury risk at an industry level, it is important to understand all the parameters pertinent to the racing jurisdiction; both those constrained by biology and by the economics of the racing system. The number of horses and races in New Zealand is decreasing, with a concurrent increase in both the age of horses and the proportion of fillies racing. However, the pattern of race starts remained the same, highlighting the individual biological constraints of the horses in the system. If the industry is considered as a bio-economic system, then the racing system will converge towards efficiency and optimisation of resources. Within this framework, the described trend is likely to continue if the number of horses entering the system continues to decrease. Therefore, these data present a basis for the modelling of changes in racing regulations and injury intervention strategies within this conceptual framework to account for the economic and biological constraints. This study aimed to examine thirteen seasons of flat racing starts (n = 388,964) in the context of an ecological system and identify metrics that describe the inherent characteristics and constraints of the New Zealand Thoroughbred racing industry. During the thirteen years examined, there was a 2–3% per year reduction in the number of races, starts and number of horses. There was a significant shift in the racing population with a greater number of fillies (aged 2–4 years) having a race start, and subsequent longer racing careers due to the inclusion of one more racing preparation post 2008 (p < 0.05). Additionally, there was an increasingly ageing population of racehorses. These changes resulted in more race starts in a career, but possibly because of biological constraints, there was no change in the number of race starts per season, starts per preparation, or days spelling between preparations (p < 0.05). There was no change in the proportion of horses having just one race start (14% of new entrants), indicating that the screening for suitability for a racing career remained consistent. These data identify key industry parameters which provide a basis for future modelling of intervention strategies to improve economic performance and reduce horse injury. Consideration of the racing industry as a bio-economic or ecological model provides framework to test how the industry may respond to intervention strategies and signal where changes in system dynamics may alter existing risk factors for injury. Simple Summary: In modern times, horse breeds, mostly in horse racing, are the Thoroughbred varieties obtained by breeding three Godolphin Arabians with British mares in England. Especially in Jeju Island, Korea, Jeju horses have been introduced from Mongolia since the 13th century. They have contributed a lot to the agricultural community, but their population has been rapidly decreasing due to rapid agricultural industrialization. Therefore, we sympathize with Jeju horse-specific genetic variation and compare and analyze evolutionary correlations by utilizing Whole Genome Sequencing analysis to evaluate the genetic diversity of Jeju horses and preserve genetic information. We explored Jeju horse-specific genetic differences through a comparative analysis of large-capacity genomic data between the public database and a Thoroughbred variety. In order to adapt to the barren external environment, it is predicted that Jeju horses have experienced strong positive selection in the direction of accumulating many genetic variations, enough to cause functional differences in the eqCD1a6 gene to have an efficient immune function. In addition, we further validate the Jeju horse-specific single nucleotide polymorphisms in the aqCD1a6 gene by employing the digital PCR method, a diagnostic technique for genetic variations. The Jeju horse is a native Korean species that has been breeding on Jeju Island since the 13th century. Their shape has a distinct appearance from the representative species, Thoroughbred. Here, we performed a comparison of the Jeju horse and Thoroughbred horse for the identification of genome-wide structure variation by using the next-generation sequencing (NGS) technique. We generated an average of 95.59 Gb of the DNA sequence, resulting in an average of 33.74 X sequence coverage from five Jeju horses. In addition, reads obtained from WGRS data almost covered the horse reference genome (mapped reads 98.4%). Based on our results, we identified 1,244,064 single nucleotide polymorphisms (SNPs), 113,498 genomic insertions, and 114,751 deletions through bioinformatics analysis. Interestingly, the results of the WGRS comparison indicated that the eqCD1a6 gene contains signatures of positive natural selection in Jeju horses. The eqCD1a6 gene is known to be involved in immunity. The eqCD1a6 gene of Jeju horses commonly contained 296 variants (275 SNPs and 21 INDELs) that were compared with its counterpart of two Thoroughbred horses. In addition, we used LOAA, digital PCR, to confirm the possibility of developing a molecular marker for species identification using variant sites. As a result, it was possible to confirm the result of the molecular marker with high accuracy. Nevertheless, eqCD1a6 was shown to be functionally intact. The development of training and management strategies to reduce injuries has been challenging due to conflicting findings about the risk factors for injury, and limited understanding of the role of different training methodologies. We describe training methodologies for 535 two-year-old horses providing 1258 training preparations and 7512 weeks of exercise. We investigated the variation in training methodologies between trainers from small, medium and large stable sizes. Significant differences were observed between trainers, with horses from larger stables accumulating a greater high-speed exercise volume, attaining training milestones more frequently and taking less time to reach their training milestones. We also highlight similarities and differences in training methods used in Queensland, Australia, and those previously reported from other geographic locations. A better understanding of training methods is an essential step towards reducing the impact of injuries. This prospective study of two-year-old racehorses was conducted in Queensland, Australia and involved weekly personal structured interviews with 26 trainers over 56 weeks. Detailed daily exercise data for 535 horses providing 1258 training preparations and 7512 weeks at risk were collected. Trainers were categorised into three groups by the mean number of two-year-old horses that they had in work each week over the study duration: (1) Small stables with five or less, (2) Medium stables with 6 to 15 and (3) Large stables with greater than 15 horses in training. Differences between trainers with small, medium and large stable sizes were evaluated using linear regression, Kruskal–Wallis equality-of-populations rank test if linear models were mis-specified or Chi-squared tests for categorical variables. Significant differences were observed between trainers, with horses from larger stables accumulating a greater high-speed exercise volume (p < 0.001), attaining training milestones more frequently (p = 0.01) and taking less time to reach their training milestones (p = 0.001). This study provides detailed data to which training practices from other locations can be compared. Presenting actual training data rather than trainers' estimation of a typical program provides a more accurate assessment of training practices. Understanding how training practices vary between regions improves comparability of studies investigating risk factors and is an important step towards reducing the impact of musculoskeletal injuries. Clenbuterol, the β2-adrenoceptor agonist, is gaining growing popularity because of its effects on weight loss (i.e., chemical liposuction). It is also popular in bodybuilding and professional sports, due to its effects that are similar to anabolic steroids. However, it is prohibited by anti-doping control. On the other hand, it is suggested that clenbuterol can inhibit the inflammatory process. The cells from 14 untrained and 14 well-trained race horses were collected after acute exercise and cultured with clenbuterol. The expressions of CD4, CD8, FoxP3, CD14, MHCII, and CD5 in PBMC, and reactive oxygen species (ROS) production, as well as cell proliferation, were evaluated by flow cytometry. In addition, IL-1β, IL-4, IL-6, IL-10, IL-17, INF-γ and TNF-α concentrations were evaluated by ELISA. β2-adrenoceptor stimulation leads to enhanced anti-inflammatory properties in well-trained horses, as do low doses in untrained animals. In contrast, higher clenbuterol doses create a pro-inflammatory environment in inexperienced horses. In conclusion, β2-adrenoceptor stimulation leads to a biphasic response. Simple Summary: The microbiome of the digestive system of both animals and humans is affected by many factors including diet, environment, age, physical condition, and genetic factors, as well as the "host effect". Physical effort, as a factor influencing the physiological state of an animal, can affect the microbial composition of the digestive system. The research so far is largely based on human and laboratory animals, but not enough research has been done on the effect of physical effort on horse digestive system. The study conducted suggests that effort has a significant impact on the studied bacterial phyla level and that there is also variation in the proportions of the studied phyla between individual horses, as well as "reacting" to changes in different ways, which also suggests the occurrence of a phenomenon called "host influence". Exercise significantly affects the body of both animals and humans, including the composition of the digestive microbiome. This study aimed to determine the changes in the composition of the most numerous bacterial phyla (Firmicutes and Bacteroidetes, as well as the level of the Lactobacillaceae family) in the digestive system of horses under the influence of physical effort. The study included a group of 17 Thoroughbred racehorses at the age of 3 years, fed the same forage, from whom feces samples were collected individually before and 48 h after physical effort. The obtained samples were subjected to DNA isolation and RT-PCR analysis. The results showed a significant increase in the level of both phyla after exercise compared to the state before physical effort; there were no such differences in the level of facultative aerobes, i.e., the Lactobacillaceae family (although a decreasing tendency was found after exercise). Simple Summary: Musculoskeletal injuries (MSI) continue to affect Thoroughbred racehorses internationally, despite over thirty years of research into this problem. Studies of risk factors for musculoskeletal injuries report inconsistent findings. Consequently, developing training strategies to mitigate the risk of MSI is difficult. We identified factors associated with particularly high odds of injury in this population of racehorses. Two-year-old horses from primiparous mares (first foals born) are at increased odds of MSI, particularly dorsal metacarpal disease ("shinsoreness"). Two-year-old horses that have had a total preparation length of between 10 and 14 weeks also have increased odds of injury. Horses of all ages that travelled a total distance of 2.4–3.8 km (12–19 furlongs) at a gallop (faster than 15 m/s; 13 s/furlong; 900 m/min; 55 km/h) in the last four weeks and horses three years and older that travelled 3.0–4.8 km (15–24 furlongs) at three-quarter pace and above (faster than 13 m/s; 15 s/furlong; 800 m/min; 48 km/h) also have increased odds of injury. We recommend that these horses should be monitored closely for impending signs of injury. We also observed a non-linear relationship between high-speed exercise and musculoskeletal injuries. This highlights the importance of high-speed exercise to enable tissue adaptation to training. Finally, in some situations, increasing the number of days worked at a slow pace may be more effective for preventing MSI, if horses are perceived at a higher risk, than resting the horse altogether. Early identification of horses at increased risk of injury and appropriate intervention could substantially reduce the impact of musculoskeletal injuries in Thoroughbred racehorses. By combining racing and training data a larger exposure to risk factors and a larger number of musculoskeletal injuries are captured and the true effect of risk factors may be more accurately represented. Furthermore, modifications to reduce the impact of MSI are more readily implemented at the training level. Our study aimed to: (1) determine the risk factors for musculoskeletal injuries and whether these are different for two-year-old and older horses and (2) determine whether risk factors vary with type of injury. This was performed by repeating analyses by age category and injury type. Data from 202 cases and 202 matched controls were collected through weekly interviews with trainers and analysed using conditional logistic regression. Increasing dam parity significantly reduced the odds of injury in horses of all age groups because of the effect in two-year-old horses (odds ratio (OR) 0.08; 95% confidence interval (CI) 0.02, 0.36; p < 0.001). Increasing total preparation length is associated with higher odds of injury in horses of all ages (OR 5.56; 95% CI 1.59, 19.46; p = 0.01), but particularly in two-year-old horses (OR 8.05; 95% CI 1.92, 33.76; p = 0.004). Increasing number of days exercised at a slow pace decreased the odds of injury in horses of all ages (OR 0.09; 95% CI 0.03, 0.28; p < 0.001). The distance travelled at three-quarter pace and above (faster than 13 m/s; 15 s/furlong; 800 m/min; 48 km/h) and the total distance travelled at a gallop (faster than 15 m/s; 13 s/furlong; 900 m/min; 55 km/h) in the past four weeks significantly affected the odds of injury. There was a non-linear association between high-speed exercise and injury whereby the odds of injury initially increased and subsequently decreased as accumulated high-speed exercise distance increased. None of the racing career and performance indices affected the odds of injury. We identified horses in this population that have particularly high odds of injury. Two-year-old horses from primiparous mares are at increased odds of injury, particularly dorsal metacarpal disease. Two-year-old horses that have had a total preparation length of between 10 and 14 weeks also have increased odds of injury. Horses of all ages that travelled a total distance of 2.4–3.8 km (12–19 furlongs) at a gallop in the last four weeks and horses three years and older that travelled 3.0–4.8 km (15–24 furlongs) at three-quarter pace and above also have increased odds of injury. We recommend that these horses should be monitored closely for impending signs of injury. Increasing the number of days worked at a slow pace may be more effective for preventing injury, if horses are perceived at a higher risk, than resting the horse altogether. Early identification of horses at increased risk and appropriate intervention could substantially reduce the impact of musculoskeletal injuries in Thoroughbred racehorses. [ABSTRACT FROM AUTHOR]

North American Thoroughbred racing is conducted on three types of surfaces—dirt, turf, and synthetic. The tracks are oval, and races are run counterclockwise. The loading on right and left limbs is expected to differ as a function of turn radius, banking, surface, and gait asymmetry. Hind limbs and forelimbs also have different functions related to propulsion and turning, respectively. This study uses the Equine Injury Database for race starts from 1 January 2009 through 31 December 2014, to compare injury rates across participating North American racetracks. The data are limited to catastrophic injuries in which horses died or were euthanized due to a fracture within 72 h of the start of the race. Overall injury rates were lower on turf and synthetic surfaces and the pattern of limb injuries in left vs. right and fore vs. hind limbs were different. Regardless of surface, forelimbs were more likely to fracture. Dirt surfaces showed higher rates of forelimb injuries compared to other surfaces, hind limbs were more likely to experience a fatal fracture on turf than on dirt. Simple Summary: Whilst the fate of horses after racing has received widespread attention, there is little research into this field. A recent independent inquiry in Queensland, Australia, highlighted that the true outcomes for horses after retirement from racing are largely unknown. This study investigated the retirement of racehorses over a 13-month period. It was found that 0.4% of horses in training per week were retired, and the season and training track did not affect this proportion. The decision for retirement was involuntary, whereby musculoskeletal injuries, respiratory or cardiac conditions and behavioural problems prevented the horse from racing in 56/110 horses (51%). Musculoskeletal injuries were the most common reason for retirement (40/110 horses, 36%). Medium-term follow-up (mostly 14 months, range 8–21) revealed that most horses (108/110; 98%) were repurposed after retirement, almost half as performance horses (50/110; 46%). Horses that were voluntarily retired (retired due to racing form or an impending injury), had 2.28 times the odds of being repurposed as performance horses than those retired involuntarily (p = 0.03). There was no association between voluntary or involuntary retirement and whether horses were used for breeding or pleasure. There is a need for traceability and accountability for these horses to ensure that their welfare is maintained in their new careers. There is international public concern regarding retirement of racehorses, including the reason for retirement and the outcome for horses after racing. However, there are currently no prospective studies investigating these factors. There was a low incidence of retirements, namely 0.4% of horses in training per week. The season and training track did not affect the incidence of retirement. Musculoskeletal injuries were the most common reason for retirement (40/110 horses, 36%). Involuntary retirements accounted for 56/100 (51%) of retirements, whereby musculoskeletal injuries, respiratory or cardiac conditions and behavioural problems prevented the horse from racing The odds of voluntary retirement, whereby the horse was retired due to racing form or impending injury, increased with each additional race start (OR 1.05; p = 0.01) and start/year of racing (OR 1.21; p = 0.03) but decreased with increasing percentage of first, second and third places (OR 0.94; p < 0.001). Medium-term follow-up (median 14 months, IQR 11, 18, range 8–21) revealed that most horses (108/110; 98%) were repurposed after retirement, almost half as performance horses (50/110; 46%). Horses that voluntarily retired had 2.28 times the odds of being repurposed as performance horses than those retired involuntarily (p = 0.03). Whether retirement was voluntary or involuntary did not influence whether horses were used for breeding or pleasure. The primary limitation of this study is that our results reflect retirement in racehorses in South East Queensland, Australia, and may not be globally applicable. Furthermore, we were unable to monitor the long-term outcome and welfare of horses in their new careers. Simple Summary: The genetic mechanisms occurring in organisms are shaped by selection pressure. Features that ought to be useful under given conditions leave their marks on the genome in the form of mutations, thereby creating different alleles. In this study, five different horse breeds were examined to find the connection between an individual's lifestyle and the presence of the peroxisomal acyl-coenzyme A oxidase 1 (ACOX1) gene, which is necessary for some metabolic pathways. Results indicated that different ACOX1 gene alleles play various roles in primitive breeds and domesticated horses. This led to the conclusion that the DNA profile can be rated on the basis of adaptation to living conditions, opening the gate for further investigation. The ACOX1 gene encodes peroxisomal acyl-coenzyme A oxidase 1, the first enzyme in the fatty acid β-oxidation pathway, which could be significant for organisms exposed to long periods of starvation and harsh living conditions. We hypothesized that variations within ACOX1, revealed by RNA Sequencing (RNA-Seq), might be based on adaptation to living conditions and had resulted from selection pressure. There were five different horse breeds used in this study, representing various utility types: Arabian, Thoroughbred, Polish Konik, draft horses, and Hucul. The single-nucleotide polymorphism (SNP) located in the ACOX1 (rs782885985) was used as a marker and was identified using the PCR restriction fragment length polymorphism method (PCR-RFLP). Results indicated extremely different genotype and allele distributions of the ACOX1 gene across breeds. A predominance of the G allele was exhibited in horses that had adapted to difficult environmental conditions, namely, Polish Konik and Huculs, which are considered to be primitive breeds. Simple Summary: Despite extensive international research, musculoskeletal injuries remain a key problem for the Thoroughbred racing industry, with multiple welfare and ethical consequences. High-speed exercise (HSE) history is an important risk factor for injury. However, studies report conflicting findings concerning the effect of HSE on the risk of injury. Although most injuries and fatalities occur during training rather than racing, many research studies only evaluate injuries reported on race day, which likely underrepresents the injuries acquired by these horses. This study aimed to determine the effect of combined training and racing HSE on injuries in racehorses. We performed a systematic search of the relevant literature and evaluated the effect of seven measures of HSE on injury through meta-analyses. The total career HSE distance significantly affected the odds of musculoskeletal injuries (MSI). With every 5-furlong (1 km) increase in career HSE distance, the odds of MSI increased by 2%. The average HSE distance per day also affected the odds of MSI. With every additional furlong (200 m) of average HSE per day, the odds of MSI increased by 73%. However, the strength of the available evidence is restricted by methodological limitations, which will need to be addressed in the future, to explore the level of risk and mechanisms for injury in more detail. Despite over three decades of active research, musculoskeletal injuries (MSI) remain a global problem for the Thoroughbred (TB) racing industry. High-speed exercise history (HSEH) has been identified as an important risk factor for MSI. However, the nature of this relationship remains unclear, with an apparent protective effect of HSE against injury, before it becomes potentially harmful. Many MSI cases and fatalities occur during training rather than during racing, resulting in an underestimation of injury from studies focused on race day. The objective of this study was to examine the current evidence of the effect of combined training and racing HSEH on MSI in TB flat racehorses, through a systematic review and meta-analysis. A systematic search of the relevant literature was performed using PubMed®, Scopus®, Web of Science®, and Embase® online databases and the gray literature using sites containing ".edu" or ".edu.au". Studies included in the review had explored seven different measures of HSE, including total career HSE distance, cumulative HSE distance in the 30 and 60 days before MSI, average HSE distance per day, per event and per 30 days, and the total number of HSE events. The total cumulative career HSE distance significantly affected the odds of MSI, with every 5-furlong increase, the odds of MSI increased by 2% (OR = 1.02; 95% CI 1.01, 1.03; p = 0.004). The average HSE distance per day also affected the odds of MSI, with every additional furlong increasing the odds of MSI by 73% (OR = 1.73; 95% CI 1.29, 2.31; p < 0.001). Other measures of HSE were not found to be consistently associated with risk of MSI, but these results should be interpreted with caution. Significant methodological limitations were identified and influence the comparability of studies. Standardizing the measures of HSE in studies of MSI, and describing training conditions in more detail, would support a more thorough investigation of the relationship between HSE and MSI. (1) Background: The intestinal microbiota plays an essential role in maintaining the host's health. Dysbiosis of the equine hindgut microbiota can alter the fermentation patterns and cause metabolic disorders. (2) Methods: This study compared the fecal microbiota composition of horses with intestinal disease and their healthy counterparts living in Korea using 16S rRNA sequencing from fecal samples. A total of 52 fecal samples were collected and divided into three groups: horses with large intestinal disease ( n = 20), horses with small intestinal disease ( n = 8), and healthy horses ( n = 24). (3) Results: Horses with intestinal diseases had fewer species and a less diverse bacterial population than healthy horses. Lactic acid bacteria, Lachnospiraceae, and Lactobacillaceae were overgrown in horses with large intestinal colic. The Firmicutes to Bacteroidetes ratio (F/B), which is a relevant marker of gut dysbiosis, was 1.94, 2.37, and 1.74 for horses with large intestinal colic, small intestinal colic, and healthy horses, respectively. (4) Conclusions: The overgrowth of two lactic acid bacteria families, Lachnospiraceae and Lactobacillaceae, led to a decrease in hindgut pH that interfered with normal fermentation, which might cause large intestinal colic. The overgrowth of Streptococcus also led to a decrease in pH in the hindgut, which suppressed the proliferation of the methanogen and reduced methanogenesis in horses with small intestinal colic.

Simple Summary: The Thoroughbred horse breed provides a unique model for studying the theoretical bases and practical methods of selection in horse breeding. To assess the diversity in the modern population of Russian Thoroughbred horses, we used classic parameters, such as Wright's inbreeding coefficient, the heterozygosity level, and population inbreeding level, over the period 1990–2018. The average inbreeding coefficient of the Thoroughbred horses was 0.83% and slowly increased over the past three decades. The individual observed heterozygosity levels of the tested horses varied within 23.1–100%, and the population's parameters of observed heterozygosity tended to decrease. The relationship between the level of inbreeding and the rate of homozygosity was not significant (r = 0.022; p > 0.05). The Thoroughbred (TB) horse has hugely impacted the development of horse breeding around the world. This breed has unique genetic qualities due to having had a closed studbook for approximately 300 years. In Russia, TBs have been bred since the second half of the 18th century. Here, we analyzed the genetic diversity and the inbreeding level in TB horses (n = 9680) for the period from 1990 to 2018 using polymorphisms of 17 microsatellite loci. We found that the genetic structure of the TB breed in Russia is represented by 100 alleles of panel STR (short tandem repeat) loci and has been stable for the past three decades. The conducted monitoring revealed a slight increase in the Wright's inbreeding coefficient in all age and sex groups of TB horses (stallions, broodmares, and foals) from 0.68% to 0.90%, which was followed by a decrease in the degree of heterozygosity, Ho, from 68.5% to 67.6%. The Spearman's rank correlation coefficient between the level of inbreeding and the degree of homozygosity was estimated (r = 0.022; p > 0.05). The obtained data on the DNA genotypes of horses of different breeds provide a unique base for the evaluation of genetic variability and the control of genetic variability of horses in selection programs. Simple Summary: Technology is becoming increasingly popular across scientific and general population groups. Pedometers, activity and health trackers, are widely used and commercially available. However, these technologies do not appear to be used in field-based equestrian health and performance. Irish industry stakeholders were interviewed regarding their perceived value of technology in equestrian sport. The interviews resulted in four primary themes around horse health, training and management. The movement assessment of the horse is undertaken subjectively through the eye and is incorporated into a holistic management structure that is based on years of experience. There is no such thing as the perfect horse and each one must be treated as an individual. The stakeholders are aware of technologies for movement analysis but demonstrate a healthy scepticism towards new, unproven technologies. Finally, the economic impact of applying technology across the herd is a major barrier to technology use. The results of this study suggest technology design needs to take place in consultation with industry stakeholders to improve field-based use. Wearable sensing technologies are increasingly used in human and equine gait research to improve ecological validity of research findings. It is unclear how these tools have penetrated the equine industry or what perspectives industry stakeholders' hold in relation to these relatively new devices. Semi-structured interviews were conducted with Irish equine industry stakeholders to understand their perception of objective tools for biomechanical analysis in the field. The study participants came from professional/elite backgrounds in both the sport horse (n = 6) and thoroughbred (n = 6) sectors. The interview data were analysed using thematic analysis, resulting in four analytical themes. The first theme conveys the importance of tacit knowledge and experience in the holistic analysis of a horse. Theme two highlights that the perfect horse does not exist therefore, equine athlete management is complex and requires a multi-layered problem-solving approach. Theme three describes an awareness among stakeholders of technologies, however they are sceptical of their value. The final theme identified that one of the key barriers to technology adoption is the economic value of the horse and the cost of implementing technology herd-wide. Our findings highlight the need for a user-centred design in this domain, which requires greater consultation and learning between technology developers and equine stakeholders to develop fit-for-purpose analysis and monitoring tools. Simple Summary: The acceptability of jumps racing has been debated in the media due to the high risk of horse fatality. Investigation of jumps racing articles in New Zealand for the 2016/2017 and 2017/2018 seasons demonstrated that this debate was vocal but localised and short-lived. Using rhetorical analysis, two different argumentative positions were identified, linked by a common acceptance of the inherent risks posed by jumps racing. Proponents of jumps racing advocated acceptance and minimisation of these inherent risks. This position was supported by emphasising the 'wealth of expertise and care within the racing industry' and was contrasted to the 'naïve opinions' characterising those who opposed jumps racing. Opponents of jumps racing personified the horse as a rights-bearing individual, resulting in the inherent risks becoming an unacceptable danger perpetrated by an exploitative racing industry for economic and personal gains. These different lines of reasoning provided a basis for arguments for and against the continuation of jumps racing in New Zealand. Jumps racing involves a higher risk of accident and fatality than flat racing. The wide accessibility of media, combined with alternate views regarding the place of animals in society, raises the question of the acceptability of the continuation of jumps racing. Racing data and media articles from Newztext and Google news search were collected for the 2016/2017 and 2017/2018 jumps racing seasons, during which the fatality rate was 5.8 per 1000 starters. Jumps racing articles comprised 3.4% of all race reporting, and the volume of discussion about jumps racing was minimal (2.9% of jumps race articles related to the continuation of jumps racing), short-lived and related to horse fatalities. Articles were categorised and analysed using rhetorical analysis to determine the main arguments. The inherent risk posed by jumps racing to the horse formed a basis for two argumentative positions. Proponents of jumps racing argued that risks were reasonable, with risk minimisation measures best determined by expertise and care from within the racing industry, labelling opponents as naïve extremists. Opponents of jumps racing used anthropomorphism of the horse to argue that any risk was unacceptable and jumps racing should be banned. Horses were attributed with rights, and from this perspective, the racing industry exploited horses for entertainment. These two different arguments were used to shape claims for and against the continuation of jumps racing, allowing both to be built upon a shared acceptance of inherent risk. [ABSTRACT FROM AUTHOR]

Simple Summary: Musculoskeletal injury rates for Thoroughbred racehorses in training and racing differ between racing jurisdictions. The aetiology of these injuries is multifactorial, but one potentially important and modifiable risk factor is the track surface on which horses train. However, the extent to which different track surfaces are used by trainers has not been clearly established. Similarly, the extent of use of alternatives to ridden exercise between different jurisdictions is unknown. Trainers in Victoria, Australia, use a combination of turf, sand, synthetic and dirt training track surfaces. Sand or synthetic surfaces were most commonly used for slow workouts and turf or synthetic tracks for fast workouts. A high proportion of trainers raced horses on surfaces that were not regularly used for training, and 89% of trainers used alternative exercise methods in addition to overground ridden workouts. Determining types of surfaces and alternatives to ridden exercise used during training, and to what extent they are used, is the first step in understanding their association with the risk of injury. The future aim is mitigating injury risk by recommending safer track surfaces. Little is known about the types of surfaces used during training of Thoroughbred racehorses or methods of exercise used in addition to ridden track-work. Our aims were to (1) describe the types of surfaces used in the training of Thoroughbred racehorses and to (2) identify alternative approaches used to exercise horses in addition to, or in place of, ridden overground track-work. Information regarding surface and alternative exercise methods was collected as part of an in-person survey of training practices of 66 registered Thoroughbred trainers in Victoria, Australia. Sand and synthetic surfaces were used by 97% and 36% of trainers respectively for slow-workouts, with galloping on turf training tracks used in training regimens by 82% and synthetic by 58% of trainers. Of those trainers utilising turf tracks, only 34% of gallop training was completed on turf despite turf being the predominant racing surface. Almost 90% of trainers used alternatives to ridden exercise. There is substantial variation in training surface used and alternative types of exercise undertaken by Victorian trainers. Future research should focus on how such practices relate to injury risk, particularly as it relates to the importance of musculoskeletal adaptation to specific race-day surfaces.
