Pig producers in Europe adopt different production methods for male pig fattening. More than half of the animals are surgically castrated. The different interpretations of animal welfare in different countries lead to market differentiation and economically different production conditions, which do not restrict trade, but economically lead to drastic competitive changes for local producers. While the Netherlands has already implemented surgical castration for the export market, using CO2 narcosis (NL), Denmark and Germany are each introducing their strategies with local anesthesia (DK) and isoflurane anesthesia (DE), respectively. Using typical pig farms from the agri benchmark Pig Network, the additional costs and economic impacts of animal welfare regulations are calculated. In Germany, isoflurane anesthesia increases costs by EUR 28.54 to EUR 49.86 per sow, or EUR 1.93 to EUR 3.81 per male piglet. This corresponds to a cost increase of around 5% per piglet. In Denmark, the costs of local anesthesia with procaine increase more moderately by EUR 3.55 to EUR 5.05 per sow, or around EUR 0.30 per male piglet. The cost increase here is less than 1% per piglet. The additional costs are leading to a loss of competitiveness for Denmark and Germany compared to the Netherlands. However, Germany is also losing profit significantly compared to Denmark. This study highlights that animal welfare regulations can negatively impact the competitiveness of the pig sector. It emphasizes the need for countries to implement such regulations carefully, ensuring that they do not lead to the loss of production or international competitiveness. Surgical castration can effectively avoid boar taint and improve pork quality by removing the synthesis of androstenone in the testis, thereby reducing its deposition in adipose tissue. The expression of genes involved in testis-derived hormone metabolism was altered following surgical castration, but the upstream regulatory factors and underlying mechanism remain unclear. In this study, we systematically profiled chromatin accessibility and transcriptional dynamics in liver tissue of castrated and intact full-sibling Yorkshire pigs. First, we identified 897 differentially expressed genes and 6864 differential accessible regions (DARs) using RNA- and ATAC-seq. By integrating the RNA- and ATAC-seq results, 227 genes were identified, and a significant positive correlation was revealed between differential gene expression and the ATAC-seq signal. We constructed a transcription factor regulatory network after motif analysis of DARs and identified a candidate transcription factor (TF) SP1 that targeted the HSD3B1 gene, which was responsible for the metabolism of androstenone. Subsequently, we annotated DARs by incorporating H3K27ac ChIP-seq data, marking 2234 typical enhancers and 245 super enhancers involved in the regulation of all testis-derived hormones. Among these, four typical enhancers associated with HSD3B1 were identified. Furthermore, an in-depth investigation was conducted on the androstenone-related enhancers, and an androstenone-related mutation was identified in a newfound candidatetypical enhancer (andEN) with dual-luciferase assays. These findings provide further insights into how enhancers function as links between phenotypic and non-coding area variations. The discovery of upstream TF and enhancers of HSD3B1 contributes to understanding the regulatory networks of androstenone metabolism and provides an important foundation for improving pork quality. Simple Summary: The use of boars for breeding purposes is common on small farms in rural regions of Italy. These animals are butchered at the end of their reproductive life for the production of charcuterie. In this regard, orchiectomy is required to remove boar taint caused by testosterone metabolites. The purpose of this study is to validate the efficacy of a safe, rapid and inexpensive castration procedure. To accomplish this objective, 91 boars were orchiectomised using the suture ligation technique. Following castration, characteristics of the severed ductus deference were evaluated in order to understand how much force would be required to tie knots without breaking the deferens and achieving proper haemostasis. After identifying a correlation between tensile strength and the age and weight of boars, a sutureless technique was successfully applied to 20 animals. The heterogeneity of Italian manufacturing processes results in the production of a large variety of pork products. In Sardinia, boars are raised and butchered to produce charcuterie. These animals are castrated before slaughter as androstenone would otherwise taint the meat, rendering it unfit for human consumption. However, to date, the literature concerning surgical orchiectomy in adult boars is limited. The goal of this study is to assess whether a sutureless swine orchiectomy procedure is feasible. Additionally, this study aims to determine the appropriate traction force needed to tie knots in the deferens duct of pigs of different weights and ages. Two groups were created: the first (n = 91) underwent orchiectomy by suture ligation; the second (n = 20) was castrated using the sutureless technique. Deferens ducts of animals in the first group (n = 182) were collected following castration, and their tensile strength was measured. Pearson's linear correlation was used to determine the relationship between the maximum traction force and weight and age groups. A correlation of 0.99 and 0.96 was shown between traction force and age and traction force and weight, respectively. In accordance with these results, sutureless castration was performed on 20 animals, calibrating the pulling force needed according to the age and weight of the boars. Simple Summary: This study aimed to analyze if the well-known positive effects of immunocastration on the behavior and welfare of pigs are robust to varying environments. One hundred forty-four male pigs were studied with regard to their sex category (EM: entire males, IC: immunocastrates, BA: barrows) and housing environment (ENR: enriched, STD: standard, MIX: repeated social mixing). The second (=effective) vaccination was administered to the immunocastrates 5 or 6 weeks before slaughter according to the standard protocol. Regardless of housing conditions, beneficial effects of immunocastration on a reduction in undesired agonistic and sexual behavior, including penis biting and penile injuries, were observed. Enriched housing showed a beneficial effect on play, whereas the social mixing environment reduced the number of social nosing events. The novel result is that the beneficial effects of immunocastration on behavioral and welfare aspects apply independent from the housing environments assessed in this study. The aim of this study was to investigate whether the well-known positive effects of immunocastration on the behavior and welfare of pigs persist under varying environments. The vaccination of immunocastrates included two injections at the age of 12 and 22 weeks. Regardless of the housing conditions, frequencies of sexual and fighting behavior expressed by immunocastrates shifted from boar-like to barrow-like behavior after the second immunocastration vaccination (Mixed model analysis, p < 0.05). Penis biting decreased in IC after the second vaccination (Wilcoxon signed-rank tests, p = 0.036) and penile injuries were lower in IC animals compared to EM (Mixed model analysis, p < 0.001). Housing-dependent effects on behavior could also be observed in the animals at a relatively young age. Enriched housing showed a beneficial effect on play behavior (Chi-square test, p < 0.001) and the social mixing environment reduced the number of social nosing events (Mixed model analysis, p < 0.05). Differences in adipose tissue deposition and properties between pig male sex categories, i.e., entire males (EM), immunocastrates (IC) and surgical castrates (SC) are relatively well-characterized, whereas the underlying molecular mechanisms are still not fully understood. To gain knowledge about the genetic regulation of the differences in adipose tissue deposition, two different approaches were used: RNA-sequencing and candidate gene expression by quantitative PCR. A total of 83 differentially expressed genes were identified between EM and IC, 15 between IC and SC and 48 between EM and SC by RNA-sequencing of the subcutaneous adipose tissue. Comparing EM with IC or SC, upregulated genes related to extracellular matrix dynamics and adipogenesis, and downregulated genes involved in the control of lipid and carbohydrate metabolism were detected. Differential gene expression generally indicated high similarity between IC and SC as opposed to EM, except for several heat shock protein genes that were upregulated in EM and IC compared with SC. The candidate gene expression approach showed that genes involved in lipogenesis were downregulated in EM compared with IC pigs, further confirming RNA-sequencing results. [ABSTRACT FROM AUTHOR]

Simple Summary: Male piglets designated for pork production have been surgically castrated for centuries. The fattening of intact boars is more environmentally friendly due to anabolic effects, but entails a higher risk of aggressive and sexual behavior, and some boar carcasses may exhibit boar taint, which reduces pork quality. Immunocastration as a further alternative to surgical castration is considered as a reliable and animal welfare-friendly method, but currently still has a very small market share. A crucial factor for the period following surgical castration is that pork production, either with boars or immunocastrates, is competitive and produces valuable carcasses with regard to lean meat content and the respective amounts of primal pork cuts. In this study, we therefore evaluated AutoFOM III data from a commercial abattoir. The results show that carcasses from immunocastrates were of similar quality to those of barrows, with carcass characteristics that lay between the values of gilts and barrows. Boar carcasses are leaner in comparison to barrows, but produce the same amount of valuable pork per carcass. Boars and immunocastrates are therefore suitable for the processing industry and both techniques are able to compete with pork production with barrows in terms of carcass characteristics and primal pork cuts. The surgical castration of male piglets as a routine procedure in modern pig production is facing increasing societal criticism. Pork production with boars and immunocastrates are available alternatives, but both have low market shares as it is so far uncertain how the carcass characteristics and primal pork cuts of boars and immunocastrates will be estimated in comparison to barrows and gilts. This article therefore evaluates the impact of sex group (gilts, boars, immunocastrates and barrows) on carcass characteristics and primal pork cuts using AutoFOM III data from a commercial abattoir. In our study, weekly slaughter data from a total of n = 36,994 pigs between 2018 and 2019 were analyzed. The results show that gilts had the highest amount of pork per carcass of all sex groups, whereas non-significant differences between boars, immunocastrates and barrows could be observed. Boars had the highest lean meat content, followed by gilts, immunocastrates and finally barrows with the lowest lean meat content. These results suggest that both immunocastration and pork production with boars are sustainable techniques that can replace pork production with barrows without affecting carcass quality. Simple Summary: Surgical castration of male piglets is societally criticized as it is painful and violates the integrity of the animals. Pork production with boars and immunocastrates are possible alternatives. Even if immunocastration is an animal-welfare-friendly alternative, its market share is low and the reliability of this technique is discussed controversially within the pork chain. Currently, the number and the reason for non-responders to vaccination are not clear. Various factors may contribute to impaired immune response including adverse and stressful housing conditions. This study, therefore, examines the influence of different housing conditions on the immune response after two Improvac® vaccinations. To determine vaccination success, testosterone concentrations, GnRH-binding, and boar taint compounds were evaluated. Furthermore, the growth performance of male pigs was compared. The results show that immunocastration is reliable under different housing systems and prevents boar taint. Moreover, the growth performance of immunocastrates is high and even superior to that of boars and barrows after the 2nd vaccination. Accordingly, immunocastration is not only animal-welfare-friendly but also economically attractive and suitable for different housing systems. Immunocastration is a sustainable alternative to piglet castration but faces limited market acceptance. The phenomenon of non-responders has not to date been examined in detail, but adverse and stressful housing conditions (e.g., mixing of groups) might impair the success of vaccinations. Therefore, we evaluated the influence of housing conditions on the immune response after two Improvac® vaccinations at an age of 12 and 22 weeks, respectively. Boars, immunocastrates and barrows (n = 48 each) were assigned to three different housing conditions (n = 36 enriched, n = 36 standard n = 72 repeated social mixing). Immune response was quantified by measuring GnRH-binding and its consequences for testosterone concentrations, development of the genital tract and boar taint. Growth performance was evaluated via average daily gain (ADG). GnRH-binding and testosterone levels revealed that immunocastration reliably suppressed testicular functions after the 2nd vaccination. Housing conditions did not modify testicular function but influenced ADG as animals under mixing grew slower than those under enriched conditions. Gonadal status had only a slight impact on ADG except in immunocastrates, which showed a temporarily higher ADG after the 2nd vaccination. From 2021 onwards, surgical castration of male piglets without pain relief will be banned in Germany. In Europe, stakeholders have committed themselves to end piglet castration from 2018 onwards. Alternatives to surgical castration are pork production with boars or immunocastrates. The competitiveness of these production systems is required to increase their market acceptance. The aim of this study was to test the profitability of pork production with boars and immunocastrates under different carcass pricing systems and penalty systems linked to boar taint. The calculations were based on the performance parameters of 36 animals (n = 12 immunocastrates, n = 12 boars, n = 12 barrows) from an experimental study. In order to analyze the economic effects of both alternatives under different regional German production systems, the performance data were set in relation to the data of agri benchmark. Both boars and immunocastrates performed economically worse than barrows in all the scenarios tested. If immunocastrates are sold according to the boar pricing system, the profitability of this technique is even lower, but still more profitable than boar fattening. Immunocastration, a technique to replace surgical castration of piglets, consists of two consecutive vaccinations to induce antibodies which transiently suppress testicular functions and avoid boar taint. It is a method to ensure both a high product quality and a high level of animal welfare. The impact of immunocastration on the three pillars of sustainability has been studied extensively. While all aspects of sustainability have been studied separately, however, a contemporary global overview of different aspects is missing. In immunocastrates, performance results are better than in barrows, but worse than in boars. The environmental impact of pork production with immunocastrates is lower than with barrows, but higher than with boars. The level of aggression is considerably lower in immunocastrates compared to boars. Societal concerns are mainly related to food safety, and are not supported by scientific evidence. After second vaccination, immunocastrates switch from a boar- to a barrow-like status. Therefore, the timing of second vaccination is a fine-tuning tool to balance advantages of boars with environmental and economic benefits against increased risk of welfare problems and boar taint. Nevertheless, both synergic and conflicting relationships between the pillars of sustainability must be communicated along the value chain to produce tailored pork products. [ABSTRACT FROM AUTHOR]
