This work reports on the development of an analyte sampling strategy on a plasmonic substrate to amplify the detection capability of a dual analytical system, paper spray ionization–mass spectrometry (PSI-MS) and surface-enhanced Raman spectroscopy (SERS). While simply applying only an analyte solution to the plasmonic paper results in a limited degree of SERS enhancement, the introduction of plasmonic gold nanoparticles (AuNPs) greatly improves the SERS signals without sacrificing PSI-MS sensitivity. It is initially revealed that the concentration of AuNPs and the type of analytes highly influence the SERS signals and their variations due to the "coffee ring effect" flow mechanism induced during sampling and the degree of the interfacial interactions (e.g., van der Waals, electrostatic, covalent) between the plasmonic substrate and analyte. Subsequent PSI treatment at high voltage conditions further impacts the overall SERS responses, where the signal sensitivity and homogeneity significantly increase throughout the entire substrate, suggesting the ready migration of adsorbed analytes regardless of their interfacial attractive forces. The PSI-induced notable SERS enhancements are presumably associated with creating unique conditions for local aggregation of the AuNPs to induce effective plasmonic couplings and hot spots (i.e., electromagnetic effect) and for repositioning analytes in close proximity to a plasmonic surface to increase polarizability (i.e., chemical effect). The optimized sampling and PSI conditions are also applicable to multi-analyte analysis by SERS and MS, with greatly enhanced detection capability and signal uniformity. [ABSTRACT FROM AUTHOR]

While extensive research has focused on understanding the degradation mechanisms of Poly(vinyl acetate) (PVAC) paint under different environmental conditions, limited attention has been paid to the long-term stability of PVAC-based white glues, especially when used in artworks. This study investigates the accelerated degradation, under simulated photoaging, and isothermal treatment of a commercial PVAC-based white glue considered representative of this class of materials used in contemporary artworks to predict its durability and assess its behavior in art objects. Through accelerated aging experiments and comparison with natural aging observed in artworks, the study reveals the formation of chromophores and the release of plasticizers as key processes; in particular, the progressive darkening was considered an early indicator of degradation processes, before structural changes could be detected by FTIR or NMR spectroscopies. The plasticizer loss induces an increase in glass transition temperature, from 7 °C to temperatures higher than room temperature, affecting the adhesive's cohesive strength and contributing to the detachment of materials in artworks. Mycotoxin contamination on food and feed can have deleterious effect on human and animal health. Agricultural crops may contain one or more mycotoxin compounds; therefore, a good multiplex detection method is desirable to ensure food safety. In this study, we developed a rapid method using label-free surface-enhanced Raman spectroscopy (SERS) to simultaneously detect three common types of mycotoxins found on corn, namely aflatoxin B1 (AFB1), zearalenone (ZEN), and ochratoxin A (OTA). The intrinsic chemical fingerprint from each mycotoxin was characterized by their unique Raman spectra, enabling clear discrimination between them. The limit of detection (LOD) of AFB1, ZEN, and OTA on corn were 10 ppb (32 nM), 20 ppb (64 nM), and 100 ppb (248 nM), respectively. Multivariate statistical analysis was used to predict concentrations of AFB1, ZEN, and OTA up to 1.5 ppm (4.8 µM) based on the SERS spectra of known concentrations, resulting in a correlation coefficient of 0.74, 0.89, and 0.72, respectively. Fenton technology is one of advanced oxidation process (AOP) methods to treat wastewater through chemical oxidation. Due to the limitations of classical iron-based catalysts, it is still challenging to find suitable catalysts for Fenton-like reactions. Here, MoS2/Au heterojunctions were successfully synthesized by reduction of chloroauric acid in the solution of layered MoS2 prepared by hydrothermal method. As a model molecule, methylene blue (MB) was used as the species to be degraded to evaluate the performance of the catalyst. It was determined by UV–visible spectra that the optimal catalyst can be obtained when MoS2 (mg): HAuCl4 (wt. % mL) is 2:2. The Fenton-like reaction process was monitored by introducing highly sensitive surface enhanced Raman spectroscopy (SERS). The results show that MB can be degraded by 83% in the first 10 min of the reaction, indicating that MoS2/Au has good catalytic performance. In addition, as a fingerprint spectrum, SERS was used to preliminarily analyze the molecular structure changes during the degradation process. The result showed that C-N-C bond was easier to break than the C-S-C bond. NH2 group and the fused ring were destroyed at the comparable speed at the first 30 min. In terms of application applicability, it was showed that MB degradation had exceeded 95% at all the three pH values of 1.4, 5.0, and 11.1 after the reaction was carried out for 20 min. The test and analysis of the light environment showed that the catalytic efficiency was significantly improved in the natural light of the laboratory compared to dark conditions. The possible mechanism based on ·OH and ·O2− from ESR data was proposed. In addition, it was demonstrated to be a first-order reaction from the perspective of kinetics. This study made a positive contribution to broaden of the applicable conditions and scope of Fenton-like reaction catalysts. It is expected to be used as a non-iron catalyst in practical industrial applications. From the perspective of detection method, we expect to develop SERS as a powerful tool for the in situ monitoring of Fenton-like reactions, and to further deepen our understanding of the mechanism. Surface-enhanced Raman spectroscopy (SERS) is an emerging technology that combines Raman spectroscopy and nanotechnology with great potential. This technology can accurately characterize molecular adsorption behavior and molecular structure. Moreover, it can provide rapid and sensitive detection of molecules and trace substances. In practical application, SERS has the advantages of portability, no need for sample pretreatment, rapid analysis, high sensitivity, and 'fingerprint' recognition. Thus, it has great potential in food safety detection. Alcoholic beverages have a long history of production in the world. Currently, a variety of popular products have been developed. With the continuous development of the alcoholic beverage industry, simple, on-site, and sensitive detection methods are necessary. In this paper, the basic principle, development history, and research progress of SERS are summarized. In view of the chemical composition, the beneficial and toxic components of alcoholic beverages and the practical application of SERS in alcoholic beverage analysis are reviewed. The feasibility and future development of SERS are also summarized and prospected. Edoxaban is a direct oral anticoagulant (DOAC) that has been recently indicated for the treatment of pulmonary embolism (PE) in SARS-CoV-2 infections. Due to its pharmacokinetic variability and a narrow therapeutic index, the safe administration of the drug requires its therapeutic drug monitoring (TDM) in patients receiving the treatment. In this work, we present a label-free method for the TDM of edoxaban by surface enhanced Raman spectroscopy (SERS). The new method utilises the thiol chemistry of the drug to chemisorb its molecules onto a highly sensitive SERS substrate. This leads to the formation of efficient hotspots and a strong signal enhancement of the drug Raman bands, thus negating the need for a Raman reporter for its SERS quantification. The standard samples were run with a concentration range of 1.4 × 10−4 M to 10−12 M using a mobile phase comprising of methanol/acetonitrile (85:15 v/v) at 291 nm followed by the good linearity of R2 = 0.997. The lowest limit of quantification (LOQ) by the SERS method was experimentally determined to be 10−12 M, whereas LOQ for HPLC-UV was 4.5 × 10−7 M, respectively. The new method was used directly and in a simple HPLC-SERS assembly to detect the drug in aqueous solutions and in spiked human blood plasma down to 1 pM. Persistent organic pollutants (POPs) have attracted significant attention because of their bioaccumulation, persistence, and toxicity. As anthropogenic products, POPs mainly contain polychlorinated biphenyls (PCBs), organochlorine pesticides (OPs), and polycyclic aromatic hydrocarbons (PAHs), and they pose a great threat to human health and the environment. To deal with these toxic contaminants, many different kinds of strategies for sensitively detecting POPs have been developed, such as high performance liquid chromatography (HPLC), surface enhanced Raman spectroscopy (SERS), and fluorescence. This paper mainly summarized the achievements of spectroscopy technologies, which generally consist of SERS, surface plasmon resonance (SPR), and fluorescence, in the detection of low-concentration POPs in different matrices. In addition, a retrospective summary is made on several critical considerations, such as sensitivity, specificity and reproducibility of these spectroscopy technologies in practical applications. Featured Application: In this research we propose a novel method for detecting Alzheimer's disease. This method involves the use of surface enhanced Raman spectroscopy in combination with multivariate statistical analysis. Based on the results of the proof-of-concept study, we have successfully demonstrated the potential of the method to identify Alzheimer's disease through analysis of blood serum. With further work, this method could be developed into a novel clinical assay for the effective and accurate diagnosis of Alzheimer's disease. Alzheimer's disease (AD) is the most common form of dementia worldwide and is characterized by progressive cognitive decline. Along with being incurable and lethal, AD is difficult to diagnose with high levels of accuracy. Blood serum from Alzheimer's disease (AD) patients was analyzed by surface-enhanced Raman spectroscopy (SERS) coupled with multivariate statistical analysis. The obtained spectra were compared with spectra from healthy controls (HC) to develop a simple test for AD detection. Serum spectra from AD patients were further compared to spectra from patients with other neurodegenerative dementias (OD). Colloidal silver nanoparticles (AgNPs) were used as the SERS-active substrates. Classification experiments involving serum SERS spectra using artificial neural networks (ANNs) achieved a diagnostic sensitivity around 96% for differentiating AD samples from HC samples in a binary model and 98% for differentiating AD, HC, and OD samples in a tertiary model. The results from this proof-of-concept study demonstrate the great potential of SERS blood serum analysis to be developed further into a novel clinical assay for the effective and accurate diagnosis of AD. Featured Application: The prepared MIHs doped with positively charged Au NPs show excellent surface-enhanced Raman scattering activity, could be used as efficient SERS substrates for the detection of synthetic colorants. A polyacrylamide-based molecularly imprinted hydrogel (MIH) doped with positively charged gold nanoparticles (Au NPs) has been synthesized via a free radical polymerization of acrylamide (AM) aqueous solution containing positively charged Au NPs as a Raman active substrate, New Red colorant as a template molecule, N,N'-methylenebis(acrylamide) as a crosslinking agent, and potassium persulfate as an initiator. The Au NPs-doped MIHs were subsequently explored as a Raman active substrate for the sensitive and selective detection of New Red colorant via surface-enhanced Raman spectroscopy (SERS). The logarithmic intensity of the characteristic peak of New Red at 1572 cm−1 was proportional to the logarithmic concentration of New Red with a detection linear range of 1.64 × 10−6 to 1.64 × 10−4 M and a limit of detection (LOD) of 1.64 × 10−7 M. The recoveries ranged from 86.3% to 100.6% with a relative standard deviation (RSD) in the range of 2.3% to 7.7%. The RSD and recovery rates for the detection of New Red spiked in a sports drink sample were 1.8% to 7.7% and 91.0% to 97.1%, respectively. These results showed that SERS combined with MIHs as Raman active substrates could provide a sensitive, selective, and effective approach for the detection of the New Red colorant in beverage matrix. Surface-enhanced Raman spectroscopy (SERS) is a promising analytical tool due to its label-free detection ability and superior sensitivity, which enable the detection of single molecules. Since its sensitivity is highly dependent on localized surface plasmon resonance, various methods have been applied for electric field-enhanced metal nanostructures. Despite the intensive research on practical applications of SERS, fabricating a sensitive and reproducible SERS sensor using a simple and low-cost process remains a challenge. Here, we report a simple strategy to produce a large-scale gold nanoparticle array based on laser interference lithography and the electrophoretic deposition of gold nanoparticles, generated through a pulsed laser ablation in liquid process. The fabricated gold nanoparticle array produced a sensitive, reproducible SERS signal, which allowed Rhodamine 6G to be detected at a concentration as low as 10–8 M, with an enhancement factor of 1.25 × 105. Hollow noble metal nanoparticles have excellent performance not only in surface catalysis but also in optics. In this work, the hollow Au-Ag alloy nanorices are fabricated by the galvanic replacement reaction. The dark-field spectrum points out that there is a big difference in the optical properties between the pure Ag nanorices and the hollow alloy nanorices that exhibit highly tunable localized surface plasmon resonances (LSPR) and that possess larger radiative damping, which is also indicated by the finite element method. Furthermore, the surface enhanced Raman scattering (SERS) and oxidation test indicate that hollow Au-Ag alloy nanorices show good anti-oxidation and have broad application prospects in surface-plasmon-related fields. Mercury ion (Hg2+) is recognized as one of the most toxic metal ions for the environment and for human health. Techniques utilized in the detection of Hg2+ are an important factor. Herein, a simple thymine was successfully employed as the surface enhanced Raman spectroscopy sensor for Hg2+ ion detection. The limit of detection (LOD) of the developed sensor is better than 0.1 nM (0.02 ppb). This sensor can also selectively distinguish Hg2+ ions over 7 types of alkali, heavy metal and transition-metal ions. Moreover, the LOD of the sensor can even achieve 1 ppb in practical application in the nature system, which is half the maximum allowable level (10 nM, 2 ppb) stipulated in the US Environmental Protection Agency standard. Further investigation of the thymine-Hg2+-thymine coordination mechanism provides a possible means of detecting other metal ions by replacing the metal ion-specific ligands. Surface Enhanced Raman Spectroscopy presents a rapid, non-destructive method to identify chemical and biological samples with up to single molecule sensitivity. Since its discovery in 1974, the technique has become an intense field of interdisciplinary research, typically generating >2000 publications per year since 2011. The technique relies on the localised surface plasmon resonance phenomenon, where incident light can couple with plasmons at the interface that result in the generation of an intense electric field. This field can propagate from the surface from the metal-dielectric interface, so molecules within proximity will experience more intense Raman scattering. Localised surface plasmon resonance wavelength is determined by a number of factors, such as size, geometry and material. Due to the requirements of the surface optical response, Ag and Au are typical metals used for surface enhanced Raman applications. These metals then need to have nano features that improve the localised surface plasmon resonance, several variants of these substrates exist; surfaces can range from nanoparticles in a suspension, electrochemically roughened electrodes to metal nanostructures on a substrate. The latter will be the focus of this review, particularly reviewing substrates made by oblique angle deposition. Oblique angle deposition is the technique of growing thin films so that the material flux is not normal to the surface. Films grown in this fashion will possess nanostructures, due to the atomic self-shadowing effect, that are dependent mainly on the deposition angle. Recent developments, applications and highlights of surface enhanced Raman scattering substrates made by oblique angle deposition will be reviewed. [ABSTRACT FROM AUTHOR]

A microfluidic bioreactor with an easy to fabricate nano-plasmonic surface is demonstrated for studies of biofilms and their precursor materials via Surface Enhanced Raman Spectroscopy (SERS). The system uses a novel design to induce sheath flow confinement of a sodium citrate biofilm precursor stream against the SERS imaging surface to measure spatial variations in the concentration profile. The unoptimised SERS enhancement was approximately 2.5 x 104, thereby improving data acquisition time, reducing laser power requirements and enabling a citrate detection limit of 0.1 mM, which was well below the concentrations used in biofilm nutrient solutions. The flow confinement was observed by both optical microscopy and SERS imaging with good complementarity. We demonstrate the new bioreactor by growing flow-templated biofilms on the microchannel wall. Oral cancer is among the most common malignancies worldwide, therefore early detection and treatment is imperative. The 5-year survival rate has remained at a dismal 50% for the past several decades. The main reason for the poor survival rate is the fact that most of the oral cancers, despite the general accessibility of the oral cavity, are not diagnosed until the advanced stage. Early detection of the oral tumors and its precursor lesions may be the most effective means to improve clinical outcome and cure most patients. One of the emerging technologies is the use of non-invasive in vivo tissue imaging to capture the molecular changes at high-resolution to improve the detection capability of early stage disease. This review will discuss the use of optical probes and highlight the role of optical imaging such as autofluorescence, fluorescence diagnosis (FD), laser confocal endomicroscopy (LCE), surface enhanced Raman spectroscopy (SERS), optical coherence tomography (OCT) and confocal reflectance micr scopy (CRM) in early oral cancer detection. FD is a promising method to differentiate cancerous lesions from benign, thus helping in the determination of adequate resolution of surgical resection margin. LCE offers in vivo cellular imaging of tissue structures from surface to subsurface layers and has demonstrated the potential to be used as a minimally invasive optical biopsy technique for early diagnosis of oral cancer lesions. SERS was able to differentiate between normal and oral cancer patients based on the spectra acquired from saliva of patients. OCT has been used to visualize the detailed histological features of the oral lesions with an imaging depth down to 2-3 mm. CRM is an optical tool to noninvasively image tissue with near histological resolution. Surface-Enhanced Raman Spectroscopy (SERS)-based biomolecule detection has been a challenge due to large variations in signal intensity, spectral profile, and nonlinearity. Recent advances in machine learning offer great opportunities to address these issues. However, well-documented procedures for model development and evaluation, as well as benchmark datasets, are lacking. Towards this end, we provide the SERS spectral benchmark dataset of Rhodamine 6G (R6G) for a molecule detection task and evaluate the classification performance of several machine learning models. We also perform a comparative study to find the best combination between the preprocessing methods and the machine learning models. Our best model, coined as the SERSNet, robustly identifies R6G molecule with excellent independent test performance. Substrates based on the metal-polymer nanocomposites 2,3-dichloro-p-xylylene-silver (PCPX-Ag) that realize the effect of surface-enhanced Raman scattering (SERS) were developed. To obtain nanocomposites, the vapor-phase polymerization method was used (VDP), which makes it possible to control the nanocomposite microstructure. In the process of self-assembly during VDP, nanocomposite films with inclusions of silver particles were formed on the polycore substrates. Silver content varied from 2.5 to 16% vol. The possibility of using such substrates for the detection of low-molecular substances, for example 5,5′-dithiobis- (2-nitrobenzoic acid) (DTNB) analyte, by the SERS method with an enhancement factor of up to 104, was demonstrated. The dependence of the SERS spectra enhancement on the microstructure of the nanocomposite and the silver content was determined. The optical and morphological properties of nanocomposites were also investigated and their dependence on the silver content was shown. It has been demonstrated that the nanocomposite is SERS selective since when working with complex solutions in the presence of high molecular weight substances, signal enhancement was only observed for low molecular weight substances. Due to the harmful effects of chemical fertilizers and pesticides, the need for an eco-friendly solution to improve soil fertility has become a necessity, thus microbial biofertilizer research is on the rise. Plant endophytic bacteria inhabiting internal tissues represent a novel niche for research into new biofertilizer strains. However, the number of species and strains that need to be differentiated and identified to facilitate faster screening in future plant-bacteria interaction studies, is enormous. Surface enhanced Raman spectroscopy (SERS) may provide a platform for bacterial discrimination and identification, which, compared with the traditional methods, is relatively rapid, uncomplicated and ensures high specificity. In this study, we attempted to differentiate 18 bacterial isolates from two oaks via morphological, physiological, biochemical tests and SERS spectra analysis. Previous 16S rRNA gene fragment sequencing showed that three isolates belong to Paenibacillus, 3—to Pantoea and 12—to Pseudomonas genera. Additional tests were not able to further sort these bacteria into strain-specific groups. However, the obtained label-free SERS bacterial spectra along with the high-accuracy principal component (PCA) and discriminant function analyses (DFA) demonstrated the possibility to differentiate these bacteria into variant strains. Furthermore, we collected information about the biochemical characteristics of selected isolates. The results of this study suggest a promising application of SERS in combination with PCA/DFA as a rapid, non-expensive and sensitive method for the detection and identification of plant-associated bacteria. Layers formed from single-stranded DNA on nanostructured plasmonic metals can be applied as "working elements" in surface–enhanced Raman scattering (SERS) sensors used to sensitively and accurately identify specific DNA fragments in various biological samples (for example, in samples of blood). Therefore, the proper formation of the desired DNA layers on SERS substrates is of great practical importance, and many research groups are working to improve the process in forming such structures. In this work, we propose two modifications of a standard method used for depositing DNA with an attached linking thiol moiety on certain SERS-active structures; the modifications yield DNA layers that generate a stronger SERS signal. We propose: (i) freezing the sample when forming DNA layers on the nanoparticles, and (ii) when forming DNA layers on SERS-active macroscopic silver substrates, using ω-substituted alkanethiols with very short alkane chains (such as cysteamine or mercaptopropionic acid) to backfill the empty spaces on the metal surface unoccupied by DNA. When 6-mercapto-1-hexanol is used to fill the unoccupied places on a silver surface (as in experiments on standard gold substrates), a quick detachment of chemisorbed DNA from the silver surface is observed. Whereas, using ω-substituted alkanethiols with a shorter alkane chain makes it possible to easily form mixed DNA/backfilling thiol monolayers. Probably, the significantly lower desorption rate of the thiolated DNA induced by alkanethiols with shorter chains is due to the lower stabilization energy in monolayers formed from such compounds. In this study, we developed highly sensitive substrates for Surface-Enhanced-Raman-Scattering (SERS) spectroscopy, consisting of silicon nanowires (SiNWs) decorated by silver nanostructures using single-step Metal Assisted Chemical Etching (MACE). One-step MACE was performed on p-type Si substrates by immersion in AgNO3/HF aqueous solutions resulting in the formation of SiNWs decorated by either silver aggregates or dendrites. Specifically, dendrites were formed during SiNWs' growth in the etchant solution, whereas aggregates were grown after the removal of the dendrites from the SiNWs in HNO3 aqueous solution and subsequent re-immersion of the specimens in a AgNO3/HF aqueous solution by adjusting the growth time to achieve the desired density of silver nanostructures. The dendrites had much larger height than the aggregates. R6G was used as analyte to test the SERS activity of the substrates prepared by the two fabrication processes. The silver aggregates showed a considerably lower limit of detection (LOD) for SERS down to a R6G concentration of 10−13 M, and much better uniformity in terms of detection in comparison with the silver dendritic structures. Extracellular vesicles (EVs) are membrane-enclosed structures ranging in size from about 60 to 800 nm that are released by the cells into the extracellular space; they have attracted interest as easily available biomarkers for cancer diagnostics. In this study, EVs from plasma of control and prostate cancer patients were fractionated by differential centrifugation at 5000× g, 12,000× g and 120,000× g. The remaining supernatants were purified by ultrafiltration to produce EV-depleted free-circulating (fc) fractions. Spontaneous Raman and surface-enhanced Raman spectroscopy (SERS) at 785 nm excitation using silver nanoparticles (AgNPs) were employed as label-free techniques to collect fingerprint spectra and identify the fractions that best discriminate between control and cancer patients. SERS spectra from 10 µL droplets showed an enhanced Raman signature of EV-enriched fractions that were much more intense for cancer patients than controls. The Raman spectra of dehydrated pellets of EV-enriched fractions without AgNPs were dominated by spectral contributions of proteins and showed variations in S-S stretch, tryptophan and protein secondary structure bands between control and cancer fractions. We conclude that the AgNPs-mediated SERS effect strongly enhances Raman bands in EV-enriched fractions, and the fractions, EV12 and EV120 provide the best separation of cancer and control patients by Raman and SERS spectra. Surface Enhanced on Infrared Absorption (SEIRAS) and Raman Spectroscopy (SERS) are nondestructive analytic techniques used to detect low concentrations and recognize the fingerprints of molecules. The recognition of the absorption from samples by conventional infrared spectroscopy (IR) via Attenuated Total Reflection (ATR) is difficult for molecules with a low signal strength. However, developed structures with SERS and SEIRAS effect present problems such as high cost, low stability, and low compatibility. Research into new media to obtain greater amplification is largely based on the creation of nanoscale structures with symmetrical arrangements and reproducible distances, resulting in aggregates of nanoparticles that help generate hot spots which are active for amplification. The sol-gel and electrospinning method for the obtention of ceramics provides an alternative means by which to produce said substrates. Fibers of nanometric scale provide an increase of surface area which allows more contact to occur with analytes. Consequently, in this study, a silica-titania-silver nanostructured support that amplifies signal intensity for Raman and infrared spectroscopy was developed. The silica-titania support was developed by sol-gel and electrospinning techniques, and the as spun fibers were treated at 800 °C. Then, the ceramic fibrous membrane was placed on conductive indium tin oxide plastic to be doped with silver using an electroplating technique, varying the silver nitrate concentration (5, 10 and 20 mM), as well as electrodeposition times (1, 2, 5 and 10 min), with a constant voltage (1 V). Twelve different supports were obtained that showed amplification. The enhancement of infrared signals from pyridine and crystal violet molecules adsorbed on silica-titania-silver (SiO2–TiO2–Ag) nanofibers was studied in situ by Attenuated Total Reflection-Fourier Transformed Infrared Spectroscopy (ATR-FTIR). The highest amplification was obtained by the support doped at 10 min in a 10 mM concentration, with an amplification factor of 2.74 in the band localized at 3301 cm−1. A sensitive and recyclable plasmonic nickel foam sensor has been developed for surface-enhanced Raman spectroscopy (SERS). A simple electrochemical method was used to deposit flower-shaped gold nanostructures onto nickel foam substrate. The high packing of the gold nanoflowers onto the nickel foam led to a high enhancement factor (EF) of 1.6 × 1011. The new SERS sensor was utilized for the direct determination of the broad-spectrum β-lactam carbapenem antibiotic meropenem in human blood plasma down to one pM. The sensor was also used in High Performance Liquid Chromatography (HPLC)-SERS assembly to provide fingerprint identification of meropenem in human blood plasma. Moreover, the SERS measurements were reproducible in aqueous solution and human blood plasma (RSD = 5.5%) and (RSD = 2.86%), respectively at 200 µg/mL (n = 3), and successfully recycled using a simple method, and hence, used for the repeated determination of the drug by SERS. A sensitive and accurate identification of specific DNA fragments (usually containing a mutation) can influence clinical decisions. Standard methods routinely used for this type of detection are PCR (Polymerase Chain Reaction, and its modifications), and, less commonly, NGS (Next Generation Sequencing). However, these methods are quite complicated, requiring time-consuming, multi-stage sample preparation, and specially trained staff. Usually, it takes weeks for patients to obtain their results. Therefore, different DNA sensors are being intensively developed by many groups. One technique often used to obtain an analytical signal from DNA sensors is Raman spectroscopy. Its modification, surface-enhanced Raman spectroscopy (SERS), is especially useful for practical analytical applications due to its extra low limit of detection. SERS takes advantage of the strong increase in the efficiency of Raman signal generation caused by a local electric field enhancement near plasmonic (typically gold and silver) nanostructures. A gold-coated array of flow-through inverse pyramids applicable as substrate for entrapment and immobilization of micro-objects and for surface enhanced Raman spectroscopic measurements was fabricated using bulk micromachining techniques from silicon. Surface morphology, optical reflectance, immobilization properties, and surface enhanced Raman amplification of the array were modelled and characterized. It was found that the special perforated periodic 3D structure can be used for parallel particle and cell trapping and highly sensitive molecular analysis of the immobilized objects. Sildenafil (SD) and its related compounds are the most common adulterants found in herbal preparations used as sexual enhancer or man's virility products. However, the abuse of SD threatens human health such as through headache, back pain, rhinitis, etc. Therefore, it is important to accurately detect the presence of SD in alcoholic beverages. In this study, the Opto Trace Raman 202 (OTR 202) was used as a surface-enhanced Raman spectroscopy (SERS) active colloids to detect SD. The results demonstrated that the limit of detection (LOD) of SD was found to be as low as 0.1 mg/L. Moreover, 1235, 1401, 1530, and 1584 cm−1 could be qualitatively determined as SD characteristic peaks. In a practical application, SD in cocktail could be easily detected using SERS based on OTR 202. Also, there was a good linear correlation between the intensity of Raman peaks at 1235, 1401, 1530, and 1584 cm−1 and the logarithm of SD concentration in cocktail was in the range of 0.1–10 mg/L (0.9822 < R2 < 0.9860). The relative standard deviation (RSD) was less than 12.7% and the recovery ranged from 93.0%–105.8%. Moreover, the original 500–1700 cm−1 SERS spectra were pretreated and the partial least squares (PLS) was applied to establish the prediction model between SERS spectra and SD content in cocktail and the highest determination coefficient (Rp2) reached 0.9856. In summary, the SD in cocktail could be rapidly and quantitatively determined by SERS, which was beneficial to provide a rapid and accurate scheme for the detection of SD in alcoholic beverages. During the last 30 years, the World Health Organization (WHO) reported a gradual increase in the number of patients with cardiovascular disease (CVD), not only in developed but also in developing countries. In particular, acute myocardial infarction (AMI) is one of the severe CVDs because of the high death rate, damage to the body, and various complications. During these harmful effects, rapid diagnosis of AMI is key for saving patients with CVD in an emergency. The prompt diagnosis and proper treatment of patients with AMI are important to increase the survival rate of these patients. To treat patients with AMI quickly, detection of a CVD biomarker at an ultra-low concentration is essential. Cardiac troponins (cTNs), cardiac myoglobin (cMB), and creatine kinase MB are typical biomarkers for AMI detection. An increase in the levels of those biomarkers in blood implies damage to cardiomyocytes and thus is related to AMI progression. In particular, cTNs are regarded as a gold standard biomarker for AMI diagnosis. The conventional TN detection system for detection of AMI requires long measurement time and is labor-intensive and tedious. Therefore, the demand for sensitive and selective TN detection techniques is increasing at present. To meet this demand, several approaches and methods have been applied to develop a TN detection system based on a nanostructure. In the present review, the authors reviewed recent advances in TN biosensors with a focus on four detection systems: (1) An electrochemical (EC) TN nanobiosensor, (2) field effect transistor (FET)-based TN nanobiosensor, (3) surface plasmon resonance (SPR)-based TN nanobiosensor and (4) surface enhanced Raman spectroscopy (SERS)-based TN nanobiosensor. Optofluidics, a research discipline combining optics with microfluidics, currently aspires to revolutionize the analysis of biological and chemical samples, e.g., for medicine, pharmacology, or molecular biology. In order to detect low concentrations of analytes in water, we have developed an optofluidic device containing a nanostructured substrate for surface enhanced Raman spectroscopy (SERS). The geometry of the gold surface allows localized plasmon oscillations to give rise to the SERS effect, in which the Raman spectral lines are intensified by the interaction of the plasmonic field with the electrons in the molecular bonds. The SERS substrate was enclosed in a microfluidic system, which allowed transport and precise mixing of the analyzed fluids, while preventing contamination or abrasion of the highly sensitive substrate. To illustrate its practical use, we employed the device for quantitative detection of persistent environmental pollutant 1,2,3-trichloropropane in water in submillimolar concentrations. The developed sensor allows fast and simple quantification of halogenated compounds and it will contribute towards the environmental monitoring and enzymology experiments with engineered haloalkane dehalogenase enzymes. Deltamethrin is widely used in pest prevention and control such as red spiders, aphids, and grubs in strawberry. It is important to accurately monitor whether the deltamethrin residue in strawberry exceeds the standard. In this paper, density functional theory (DFT) was used to theoretically analyze the molecular structure of deltamethrin, gold nanoparticles (AuNPs) and silver nanoparticles (AgNPs) were used to enhance the surface enhanced Raman spectroscopy (SERS) detection signal. As a result, the theoretical Raman peaks of deltamethrin calculated by DFT were basically similar to the measured results, and the enhancing effects based on AuNPs was better than that of AgNPs. Moreover, 554, 736, 776, 964, 1000, 1166, 1206, 1593, 1613, and 1735 cm−1 could be determined as deltamethrin characteristic peaks, among which only three Raman peaks (736, 1000, and 1166 cm−1) could be used as the deltamethrin characteristic peaks in strawberry when the detection limit reached 0.1 mg/L. In addition, the 500–1800 cm−1 SERS of deltamethrin were analyzed by the partial least squares (PLS) and backward interval partial least squares (BIPLS). The prediction accuracy of deltamethrin in strawberry ( R p 2 = 0.93, RMSEp = 4.66 mg/L, RPD = 3.59) was the highest when the original spectra were pretreated by multiplicative scatter correction (MSC) and then modeled by BIPLS. In conclusion, the deltamethrin in strawberry could be qualitatively analyzed and quantitatively determined by SERS based on AuNPs enhancement, which provides a new detection scheme for deltamethrin residue determination in strawberry. We present computational studies of quasi three-dimensional nanowell (NW) and nanopost (NP) plasmonic crystals for applications in surface enhanced Raman spectroscopy (SERS). The NW and NP plasmonic crystals are metal coated arrays of cylindrical voids or posts, respectively, in a dielectric substrate characterized by a well/post diameter (D), relief depth ( R D ), periodicity (P), and metal thickness ( M T ). Each plasmonic crystal is modeled using the three-dimensional finite-difference time-domain (FDTD) method with periodic boundary conditions in the x- and y-directions applied to a computational unit cell to simulate the effect of a periodic array. Relative SERS responses are calculated from time-averaged electric field intensity enhancements at λ exc and λ scat or at λ mid via G SERS 4 = g 2 ( λ exc ) × g 2 ( λ scat ) or G mid 4 = g 4 ( λ mid ) , respectively, where g 2 = | E | 2 / | E 0 | 2 . Comparisons of G SERS 4 and G mid 4 are made to previously reported experimental SERS measurements for NW and NP geometries. Optimized NW and NP configurations based on variations of D, P, R D , and M T using G SERS 4 are presented, with 6× and 2× predicted increases in SERS, respectively. A novel plasmonic crystal based on square NP geometries are considered with an additional 3× increase over the optimized cylindrical NP geometry. NW geometries with imbedded spherical gold nanoparticles are considered, with 10× to 10 3 × increases in SERS responses over the NW geometry alone. Two-dimensional (2D) materials, such as graphene and hexagonal boron nitride, are new kinds of materials that can serve as substrates for surface enhanced Raman spectroscopy (SERS). When combined with traditional metallic plasmonic structures, the hybrid 2D materials/metal SERS platform brings extra benefits, including higher SERS enhancement factors, oxidation protection of the metal surface, and protection of molecules from photo-induced damages. This review paper gives an overview of recent progress in the 2D materials-coated plasmonic structure in SERS application, focusing on the fabrication of the hybrid 2D materials/metal SERS platform and its applications for Raman enhancement.
