Introduction: This study aimed to evaluate the relationship between pathological and clinical risk classifications in newly diagnosed prostate cancer patients, and 68Ga-PSMA PET/CT data and serum Prostate Specific Antigen (PSA) values. Method: A total of 203 patients who were diagnosed with prostate cancer between 2019 and 2023, who had not yet received treatment and who underwent 68Ga-PSMA PET/CT for staging purposes were included in this study. Results: There was a substantial correlation between D'Amico risk classification, Gleason score, ISUP classification, and the presence or absence of metastasis (p < 0.0001). The median SUVmax value of the prostate gland and the D'Amico risk classification were statistically significantly correlated. (p < 0.0001). There was a statistically significant correlation between the ISUP classification and the PSA value and prostate gland SUVmax value (p < 0.0001). There was a significant correlation between the median SUVmax values of the prostate gland at the time of diagnosis and the patients with and without metastases (p < 0.0001). According to the data obtained from ROC analysis, patients with prostate gland SUVmax values of 8.75 and above were found to have a high probability of metastasis with a sensitivity of 78.9% and a specificity of 59.05%. Conclusion: Our study showed that 68Ga-PSMA PET/CT is a highly effective method for staging newly diagnosed high-risk prostate cancer. The probability of metastasis was found to be dramatically increased in Gleason 8 and above. According to D'Amico risk classification, metastasis was detected in at least half of high-risk patients. Since the sensitivity of metastasis was 78.9% in patients with prostate gland SUVmax value above 8.75, we think that these patients should be carefully reported in terms of metastasis. The increasing global reliance on renewable energy sources, particularly solar energy, underscores the critical importance of accurate solar irradiance forecasting. As solar capacity continues to grow, precise predictions of solar irradiance become essential for optimizing the performance and reliability of photovoltaic (PV) systems. This study introduces a novel hybrid forecasting model that integrates Nonlinear Autoregressive with Exogenous Inputs (NARX) with Long Short-Term Memory (LSTM) networks. The purpose is to enhance the precision of predicting daily solar irradiance in fluctuating meteorological scenarios, particularly in southwestern France. The hybrid model employs the NARX model's capacity to handle complex non-linear relationships and the LSTM's aptitude to manage long-term dependencies in time-series data. The performance metrics of the hybrid NARX-LSTM model were thoroughly assessed, revealing a mean absolute error (MAE) of 9.58 W/m2, a root mean square error (RMSE) of 16.30 W/m2, and a Coefficient of Determination (R2) of 0.997. Consequently, the proposed hybrid model outperforms the benchmark model in all metrics, showing a significant improvement in prediction accuracy and better alignment with the observed data. These results highlight the model's effectiveness in enhancing forecasting accuracy under unpredictable conditions, improving solar energy integration into power systems, and ensuring more reliable energy predictions. Objectives: There are several endovascular treatment options to treat aortic arch and thoracic aortic pathologies with custom-made or surgeon-modified aortic stent grafts. This study seeks to assess endovascular treatment methods for aortic arch and thoracic aortic pathologies with no acceptable proximal landing zone for standard thoracic endovascular aortic repair (TEVAR), comparing different treatment methods and evaluating technical success, intraoperative parameters and short-term outcomes. Methods: All patients undergoing elective or emergency endovascular treatment of aortic arch and thoracic aortic pathologies, with no acceptable landing zone for standard TEVAR, between 1 January 2010 and 31 March 2024, at the University Hospital Düsseldorf, Germany were included. An acceptable landing zone was defined as a minimum of 2 cm for sufficient sealing. All patients were not suitable for open surgery. Patients were categorized by an endovascular treatment method for a comprehensive comparison of pre-, intra- and postoperative variables. IBM SPSS29 was used for data analysis. Results: The patient cohort comprised 21 patients, predominantly males (81%), with an average age of 70.9 ± 9 years with no acceptable proximal landing zone for standard TEVAR procedure. The most treated aortic pathologies were penetrating aortic ulcers and chronic post-dissection aneurysms. Patients were sub-grouped according to the applied procedure as follows: five patients with chimney thoracic endovascular aortic repair (chTEVAR), seven patients with in situ fenestrated thoracic endovascular aortic repair (isfTEVAR), six patients with custom-made fenestrated thoracic endovascular aortic repair (cmfTEVAR) and three patients with custom-made branched thoracic endovascular aortic repair (cmbTEVAR). Emergency procedures involved two patients. There were significant differences in the total procedure and fluoroscopy time, as well as in contrast agent usage among the treatment groups. cmfTEVAR had the shortest total procedure time, while chTEVAR exhibited the highest contrast agent usage. The overall mortality rate among all procedures was 9.5% (two patients) and 4.7% for elective procedures, respectively. Deaths were associated with either retrograde type A dissection or stent graft infection. Both patients were treated with chTEVAR. There was one minor and one major stroke; these patients were treated with isfTEVAR. No endoleak occurred during any procedure. The reintervention rate for chTEVAR was 20% and 0% for all other procedures during the in-hospital stay. The patients who were treated with cmfTEVAR had no complications, the shortest operating and fluoroscopy time, and less contrast agent was needed in comparison with other treatment methods. Conclusions: Complex endovascular procedures of the aortic arch with custom-made or surgeon-modified aortic stent grafts offer a safe solution, with acceptable complication rates for patients who are not suitable for open aortic arch repair. Regardless of their nationality, all children are unfortunately at risk since nations across the world are not doing enough to reduce the impacts of climate change. However, teenagers in developing countries face more severe challenges as a consequence of climate change. This research portrays teenagers living in a rural area in terms of their awareness, beliefs, attitudes, and actions relevant to climate change. Beliefs regarding climate change are reflected in the context of skepticism about the existence, causes, and impacts of climate change as well as belief in climate change mitigation. We also magnify ecocentrism and anthropocentrism, as a socio-psychological factor, and gender, a demographic variable, when exploring their power as the antecedents of climate change-related actions. The data were gathered from 650 students attending a middle school selected from a rural area located in the inner west region of Türkiye. The results indicated that less than half of the teenagers believed in the notion that something could be done to tackle climate change. According to discriminant analysis, female teenagers tend to believe in climate change more and have stronger ecocentric attitudes compared to males. Furthermore, female teenagers seemed to engage in climate change-related actions more than males. This study provided further evidence on the role of beliefs and attitudes in relation to this issue. To be more specific, teenagers believing that anthropogenic factors lead to climate change and that we, as humans, should therefore do our bit to reduce climate change, also tend to take the necessary actions to combat climate change. Furthermore, these individuals, valuing the natural environment for its own sake, engage in actions for climate change mitigation. By examining the awareness, beliefs, attitudes, and actions of teenagers in rural areas towards climate change, this research underscores the critical role of the quality of formal education in equipping teenagers to effectively engage with climate change issues. Objectives: The impact of the tourniquet on cardiac efficiency remains unknown. This study aimed to assess the impact of the tourniquet on cardiac cycle efficiency (CCE) and to interpret how general anesthesia (GA) or combined spinal epidural anesthesia (CSEA) affects this during surgery using cardiac energy parameters. Methods: This prospective observational study included 43 patients undergoing elective unilateral total knee arthroplasty (TKA) with a tourniquet divided into GA (n = 22) and CSEA (n = 21) groups. Cardiac energy parameters were measured before anesthesia (T1), pre-tourniquet inflation (T2), during inflation (T3–T8), and post-deflation (T9). The estimated power of the study was 0.99 based on the differences and standard deviations in CCE at T2–T3 for all patients (effect size: 0.88, alpha error: 0.05). Results: CCE decreased significantly more at T3 in the GA group than in the CSEA group, whereas dP/dtmax and Ea increased more (p < 0.05, p < 0.001, and p < 0.01, respectively). At T9, CCE increased significantly in the GA group, whereas dP/dtmax and Ea decreased (p < 0.05, p < 0.001, and p < 0.001, respectively). Nanoporous gold (np-Au) has found its use in applications ranging from catalysis to biosensing, where pore morphology plays a critical role in performance. While the morphology evolution of bulk np-Au has been widely studied, knowledge about its thin-film form is limited. This work hypothesizes that the mechanical compliance of the thin film substrate can play a critical role in the morphology evolution. Via experimental and finite-element-analysis approaches, we investigate the morphological variation in np-Au thin films deposited on compliant silicone (PDMS) substrates of a range of thicknesses anchored on rigid glass supports and compare those to the morphology of np-Au deposited on glass. More macroscopic (10 s to 100 s of microns) cracks and discrete islands form in the np-Au films on PDMS compared to on glass. Conversely, uniformly distributed microscopic (100 s of nanometers) cracks form in greater numbers in the np-Au films on glass than those on PDMS, with the cracks located within the discrete islands. The np-Au films on glass also show larger ligament and pore sizes, possibly due to higher residual stresses compared to the np-Au/PDMS films. The effective elastic modulus of the substrate layers decreases with increasing PDMS thickness, resulting in secondary np-Au morphology effects, including a reduction in macroscopic crack-to-crack distance, an increase in microscopic crack coverage, and a widening of the microscopic cracks. However, changes in the ligament/pore widths with PDMS thickness are negligible, allowing for independent optimization for cracking. In this study, we designed a high-performance, compact E-shaped microstrip antenna optimized for intelligent transportation systems, operating at 5.8 GHz. Utilizing simulation tools such as CST Studio Suite 2022 Learning Edition, Ansys HFSS 2022 R1, and MATLAB 2022b PCB Antenna Designer, we ensured consistent physical parameters. Fabricated with a 1.6 mm thick FR-4 substrate and a 50 Ω microstrip line-feeding technique, the antenna measures 35 × 50 × 1.6 mm³, smaller than already existing designs. At 5.75 GHz, it exhibits a return loss of −23.68 dB and a VSWR of 1.140 dB, ensuring stable performance within the desired frequency band. Our findings recommend its integration into vehicle-to-infrastructure wireless communication systems. Comparison across simulation environments and laboratory measurements highlights the close alignment of results with those from Ansys HFSS 2022 R1, affirming its reliability. [ABSTRACT FROM AUTHOR]

Background: Classifications of AAFD/PCFD have evolved with an increased understanding of the pathology involved. A review of classification systems helps identify deficiencies and respective contributions to the evolution in understanding the classification of AAFD/PCFD. Methods: Using multiple electronic database searches (Medline, PubMed) and Google search, original papers classifying AAFD/PCFD were identified. Nine original papers were identified that met the inclusion criteria. Results: Johnson's original classification and multiple variants provided a significant leap in understanding and communicating the pathology but remained tibialis posterior tendon-focused. Drawbacks of these classifications include the implication of causality, linearity of progression through stages, an oversimplification of stage 2 deformity, and a failure to understand that multiple tendons react, not just tibialis posterior. Later classifications, such as the PCFD classification, are deformity-centric. Early ligament laxity/instability in normal attitude feet and all stages of cavus feet can present with pain and instability with minor/no deformity. These may not be captured in deformity-based classifications. The authors developed the 'Triple Classification' (TC) understanding that primary pathology is a progressive ligament failure/laxity that presents as tendon reactivity, deformity, and painful impingement, variably manifested depending on starting foot morphology. In this classification, starting foot morphology is typed, ligament laxities are staged, and deformity is zoned. Conclusions: This review has used identified deficiencies within classification systems for AAFD/PCFD to delink ligament laxity, deformity, and foot type and develop the 'Triple classification'. Advantages of the TC may include representing foot types with no deformity, defining complex secondary instabilities, delinking foot types, tendon reactivity/ligament instability, and deformity to represent these independently in a new classification system. Compared to natural images in artificial datasets, it is more challenging to improve the spatial resolution of remote sensing optical image data using super-resolution techniques. Historical aerial images are primarily grayscale due to single-band acquisition, which further limits their recoverability. To avoid data limitations, it is advised to employ a data collection consisting of images with homogeneously distributed intensity values of land use/cover objects at various resolution values. Thus, two different datasets were created. In line with the proposed approach, images of bare land, farmland, residential areas, and forested regions were extracted from orthophotos of different years with different spatial resolutions. In addition, images with intensity values in a more limited range for the same categories were obtained from a single year's orthophoto to highlight the contribution of the suggested approach. Training of two different datasets was performed independently using a deep learning-based super-resolution model, and the same test images were enhanced individually with the weights of both models. The results were assessed using a variety of quality metrics in addition to visual interpretation. The findings indicate that the suggested dataset structure and content can enable the recovery of more details and effectively remove the smoothing effect. The power system is one of the most susceptible systems to failures, which are most frequently caused by transmission line faults. Transmission line failures account for 85% of all power system malfunctions. However, over the last decade, numerous fault detection methods have been developed to ensure the reliability and stability of power systems. A hybrid detection method based on the idea of redundancy property is presented in this paper. Because the continuous wavelet transform itself does not extract fault features for small defects effectively, the stationary wavelet transform approach is employed to assist in their detection. As a result of its ability to decompose the signal into high- and low-frequency components, undecimated reconstruction by using the algebraic summation operation (ASO) is used. This approach creates redundancy, which is useful for the feature extraction of small defects and makes faulty parts more evident. The numerical value of the redundancy ratio's contribution to the original signal is approximately equal to 36%. Following this method for redundant signal reconstruction, a continuous wavelet transform is used to extract the fault characteristic significantly easier in the time-scale (frequency) domain. Finally, the suggested technique has been demonstrated to be an efficient fault detection and identification tool for use in power systems. In fact, using this advanced signal processing technique will help with early fault detection, which is mainly about predictive maintenance. Ornithine transcarbamylase deficiency (OTCD) is the most common urea cycle disorder with high unmet needs, as current dietary and medical treatments may not be sufficient to prevent hyperammonemic episodes, which can cause death or neurological sequelae. To date, liver transplantation is the only curative choice but is not widely available due to donor shortage, the need for life-long immunosuppression and technical challenges. A field of research that has shown a great deal of promise recently is gene therapy, and OTCD has been an essential candidate for different gene therapy modalities, including AAV gene addition, mRNA therapy and genome editing. This review will first summarise the main steps towards clinical translation, highlighting the benefits and challenges of each gene therapy approach, then focus on current clinical trials and finally outline future directions for the development of gene therapy for OTCD. Ornithine transcarbamylase deficiency (OTCD) is the most common inherited disorder of the urea cycle and, in general, is transmitted as an X-linked recessive trait. Defects in the OTC gene cause an impairment in ureagenesis, resulting in hyperammonemia, which is a direct cause of brain damage and death. Patients with late-onset OTCD can develop symptoms from infancy to later childhood, adolescence or adulthood. Clinical manifestations of adults with OTCD vary in acuity. Clinical symptoms can be aggravated by metabolic stressors or the presence of a catabolic state, or due to increased demands upon the urea. A prompt diagnosis and relevant biochemical and genetic investigations allow the rapid introduction of the right treatment and prevent long-term complications and mortality. The gut–brain axis embodies the bi-directional communication between the gastrointestinal tract and the central nervous system (CNS), where vagal afferent neurons (VANs) serve as sensors for a variety of gut-derived signals. The gut is colonized by a large and diverse population of microorganisms that communicate via small (effector) molecules, which also act on the VAN terminals situated in the gut viscera and consequently influence many CNS processes. However, the convoluted in vivo environment makes it difficult to study the causative impact of the effector molecules on VAN activation or desensitization. Here, we report on a VAN culture and its proof-of-principle demonstration as a cell-based sensor to monitor the influence of gastrointestinal effector molecules on neuronal behavior. We initially compared the effect of surface coatings (poly-L-lysine vs. Matrigel) and culture media composition (serum vs. growth factor supplement) on neurite growth as a surrogate of VAN regeneration following tissue harvesting, where the Matrigel coating, but not the media composition, played a significant role in the increased neurite growth. We then used both live-cell calcium imaging and extracellular electrophysiological recordings to show that the VANs responded to classical effector molecules of endogenous and exogenous origin (cholecystokinin serotonin and capsaicin) in a complex fashion. We expect this study to enable platforms for screening various effector molecules and their influence on VAN activity, assessed by their information-rich electrophysiological fingerprints. Numerous deep learning techniques have been explored in pursuit of achieving precise road segmentation; nonetheless, this task continues to present a significant challenge. Exposing shadows and the obstruction of objects are the most important difficulties associated with road segmentation using optical image data alone. By incorporating additional data sources, such as LiDAR data, the accuracy of road segmentation can be improved in areas where optical images are insufficient to segment roads properly. The missing information in spectral data due to the object blockage and shadow effect can be compensated by the integration of 2D and 3D information. This study proposes a feature-wise fusion strategy of optical images and point clouds to enhance the road segmentation performance of a deep learning model. For this purpose, high-resolution satellite images and airborne LiDAR point cloud collected over Florida, USA, were used. Eigenvalue-based and geometric 3D property-based features were calculated based on the LiDAR data. These optical images and LiDAR-based features were used together to train, end-to-end, a deep residual U-Net architecture. In this strategy, the high-level features generated from optical images were concatenated with the LiDAR-based features before the final convolution layer. The consistency of the proposed strategy was evaluated using ResNet backbones with a different number of layers. According to the obtained results, the proposed fusion strategy improved the prediction capacity of the U-Net models with different ResNet backbones. Regardless of the backbone, all models showed enhancement in prediction statistics by 1% to 5%. The combination of optical images and LiDAR point cloud in the deep learning model has increased the prediction performance and provided the integrity of road geometry in woodland and shadowed areas. Simple Summary: Lateralized overgrowth may be isolated or accompany syndromes. Recently, international BWS consensus proposed that the patients with isolated lateralized overgrowth and epigenetic change related to this syndrome should be evaluated within the Beckwith–Wiedemann spectrum. The risk of cancer is high in patients with lateralized overgrowth, some patients also may be admitted presenting with a tumor. The aim of this study was to classify patients with lateralized overgrowth into isolated, atypical, and classic phenotypes according to consensus scoring, to investigate epigenetic alterations on chromosome 11p15.5, and to raise awareness for early detection and prevention of cancer. The Beckwith–Wiedemann spectrum (BWSp) ranges from isolated lateralized overgrowth (ILO) to classic phenotypes. In this broad clinical spectrum, an epigenetic alteration on chromosome 11p15.5 can be detected. The risk for embryonal tumors is high, especially in patients with lateralized overgrowth (LO). The aim of this study is to investigate epigenetic alterations in 11p15.5 and tumor risk in 87 children with LO. The methylation level of 11p15.5 was examined in the blood of all patients and in skin samples or buccal swabs from 40 patients with negative blood tests; 63.2% of patients were compatible with the ILO phenotype, 18.4% were atypical, and 18.4% were classic. The molecular diagnosis rate was 81.2% for the atypical and classic phenotypes, and 10.9% for the ILO phenotype. In patients with epigenetic alterations, LO was statistically significantly more severe than in test negatives. Tumors developed in six (6.9%) of the total 87 patients with LO; four belonged to the atypical or classical phenotype (12.5%) and two to ILO (3.5%). Three of the four patients with atypical/classical phenotypes had pUPD11, one had IC1-GOM alteration, and two ILO patients were negative. Civilizational populism is readily observable in Turkish politics. This study analyzes how the ruling political parties in Turkey (Islamist JDP and ultra-nationalist NAP) reframe extant civilizational ideas into a populist format to reproduce civilizational populism for ideological and pragmatic reasons. The research implements a two-level analysis of civilizational populism. On the first level, the article reveals the intellectual origins of the ideas on civilizations. On the second level, it analyses how these ideas are used by political actors in constructing populist civilizational discourse in daily politics. This methodological approach to civilizational populism shows that politicians use old ideas that are well-known by their supporters while framing them as their populist narrative. Thus, civilizational populism occurs in Turkey as a continuity. This study finds out that (i) Civilizational populism is a salient phenomenon proving how Islam and nationalism are socially and politically coded as friendly categories in Turkey; (ii) Political actors are the agents of populism but not the inventors of the many ideas they instrumentalize; (iii) Civilizational populism in Turkey is constructed on an anti-Western narrative which is essentialist and culturalist. Consequently, the article reveals that the culturist and essentialist critique levelled by other civilizations at Western political actors can be levelled similarly at the Muslim political elite. Neuroinflammation plays a central role in many neurological disorders, ranging from traumatic brain injuries to neurodegeneration. Electrophysiological activity is an essential measure of neuronal function, which is influenced by neuroinflammation. In order to study neuroinflammation and its electrophysiological fingerprints, there is a need for in vitro models that accurately capture the in vivo phenomena. In this study, we employed a new tri-culture of primary rat neurons, astrocytes, and microglia in combination with extracellular electrophysiological recording techniques using multiple electrode arrays (MEAs) to determine the effect of microglia on neural function and the response to neuroinflammatory stimuli. Specifically, we established the tri-culture and its corresponding neuron-astrocyte co-culture (lacking microglia) counterpart on custom MEAs and monitored their electrophysiological activity for 21 days to assess culture maturation and network formation. As a complementary assessment, we quantified synaptic puncta and averaged spike waveforms to determine the difference in excitatory to inhibitory neuron ratio (E/I ratio) of the neurons. The results demonstrate that the microglia in the tri-culture do not disrupt neural network formation and stability and may be a better representation of the in vivo rat cortex due to its more similar E/I ratio as compared to more traditional isolated neuron and neuron-astrocyte co-cultures. In addition, only the tri-culture displayed a significant decrease in both the number of active channels and spike frequency following pro-inflammatory lipopolysaccharide exposure, highlighting the critical role of microglia in capturing electrophysiological manifestations of a representative neuroinflammatory insult. While, for both patients in Turkey and in the UK, dual therapy with a combination of sodium benzoate and sodium/glycerol phenylbutyrate was favoured, almost 30% of the patients were only on sodium benzoate in France (Figure 6). 4. Building segmentation is crucial for applications extending from map production to urban planning. Nowadays, it is still a challenge due to CNNs' inability to model global context and Transformers' high memory need. In this study, 10 CNN and Transformer models were generated, and comparisons were realized. Alongside our proposed Residual-Inception U-Net (RIU-Net), U-Net, Residual U-Net, and Attention Residual U-Net, four CNN architectures (Inception, Inception-ResNet, Xception, and MobileNet) were implemented as encoders to U-Net-based models. Lastly, two Transformer-based approaches (Trans U-Net and Swin U-Net) were also used. Massachusetts Buildings Dataset and Inria Aerial Image Labeling Dataset were used for training and evaluation. On Inria dataset, RIU-Net achieved the highest IoU score, F1 score, and test accuracy, with 0.6736, 0.7868, and 92.23%, respectively. On Massachusetts Small dataset, Attention Residual U-Net achieved the highest IoU and F1 scores, with 0.6218 and 0.7606, and Trans U-Net reached the highest test accuracy, with 94.26%. On Massachusetts Large dataset, Residual U-Net accomplished the highest IoU and F1 scores, with 0.6165 and 0.7565, and Attention Residual U-Net attained the highest test accuracy, with 93.81%. The activation of toll-like receptors (TLRs) in the central nervous system (CNS) can lead to neuroinflammation and contribute to many neurological disorders, including autoimmune diseases. Cell culture models are powerful tools for studying specific molecular and cellular mechanisms that contribute to these disease states and identifying potential therapeutics. However, most cell culture models have limitations in capturing biologically relevant phenomena, due in part to the non-inclusion of necessary cell types. Neurons, astrocytes, and microglia (critical cell types that play a role in neuroinflammation) all express at least a subset of TLRs. However, the response of each of these cell types to various TLR activation, along with their relative contribution to neuroinflammatory processes, is far from clear. In this study, we demonstrate the screening capabilities of a primary cortical cell tri-culture of neuron, astrocyte, and microglia from neonatal rats. Specifically, we compare the neuroinflammatory response of tri-cultures to that of primary neuron-astrocyte co-cultures to a suite of known TLR agonists. We demonstrate that microglia are required for observation of neurotoxic neuroinflammatory responses, such as increased cell death and apoptosis, in response to TLR2, 3, 4, and 7/8 activation. Additionally, we show that following TLR3 agonist treatment, microglia and astrocytes play opposing roles in the neuroinflammatory response, and that the observed response is dictated by the degree of TLR3 activation. Overall, we demonstrate that microglia play a significant role in the neuroinflammatory response to TLR activation in vitro and, hence, the tri-culture has the potential to serve as a screening platform that better replicates the in vivo responses. Simple Summary: ALK translocation is present in 4–5% of patients with non-small-cell lung cancer. Patients treated with alectinib invariably develop subclinical hemolysis, which is presumably caused by acanthocytic deformation and splenic trapping of blood erythrocytes. Morphologic red blood cell changes, decreased EMA testing, and reactive reticulocytosis develop in literally all patients early after the initiation of alectinib, but not other ALK inhibitors. These alterations are not associated with the efficacy of alectinib or the molecular features of ALK+ disease and resolve within a few months after a switch to other ALK–TKI but complicate any hematologic workup. Furthermore, the risk of long-term complications, such as cholelithiasis due to increased serum bilirubin in the majority of cases, remains unclear at present. Alectinib is a standard initial treatment for patients with advanced anaplastic lymphoma kinase (ALK) rearranged non-small-cell lung cancer (NSCLC). The current study analyzed a prospective cohort of 24 consecutive alectinib-treated patients and controls in order to comprehensively characterize longitudinal erythrocyte changes under treatment with ALK inhibitors. Upon starting alectinib, all examined patients developed reticulocytosis and abnormal erythrocyte morphology with anisocytosis and a predominance of acanthocytes (64% of red blood cells on average, range 36–100%) in the peripheral blood smear within approximately 2 weeks. Changes were accompanied by a gradual reduction in Eosin-5-maleimide (EMA) binding, which became pathologic (<80% of cells) within 1–2 months in all cases, mimicking an abortive form of hereditary spherocytosis. The latter could be ruled out in 3/3 of analyzed cases by normal sequencing results for the ANK1, EPB42, SLC4A1, SPTA1, or SBTB genes. The direct Coombs test was also negative in 11/11 tested cases. Besides, anemia, increased LDH, and increased bilirubin were noted in a fraction of patients only, ranging between 42 and 68%. Furthermore, haptoglobin decreases were infrequent, occurring in approximately 1/3 of cases only, and mild, with an average value of 0.93 g/L within the normal range of 0.3–2 g/dL, suggesting that hemolysis occurred predominantly in the extravascular compartment, likely due to splenic trapping of the deformed erythrocytes. These changes showed no association with progression-free survival under alectinib or molecular features, i.e., ALK fusion variant or TP53 status of the disease, and resolved upon a switch to an alternative ALK inhibitor. Thus, alectinib induces mild, reversible erythrocyte changes in practically all treated patients, whose most sensitive signs are aberrant red cell morphology in the peripheral smear, a pathologic EMA test, and reactive reticulocytosis. Frank hemolytic anemia is rare, but mild subclinical hemolysis is very frequent and poses differential-diagnostic problems. Alectinib can be continued under the regular control of hemolysis parameters, but the risk of long-term complications, such as cholelithiasis due to increased serum bilirubin in most patients, remains unclear at present. Nanoporous gold (np-Au) electrode coatings have shown improved neural electrophysiological recording fidelity in vitro, in part due to reduced surface coverage by astrocytes. This reduction in astrocytic spreading has been attributed to the influence of electrode nanostructure on focal adhesion (FA) formation. This study describes the development and use of a microfluidic flow cell for imposing controllable hydrodynamic shear on astrocytes cultured on gold surfaces of different morphologies, in order to study the influence of nanostructure on astrocyte adhesion strength as a function of np-Au electrode morphology. Astrocyte detachment (a surrogate for adhesion strength) monotonically increased as feature size was reduced from planar surfaces to np-Au, demonstrating that adhesion strength is dependent on nanostructure. The Equality-Generalized Travelling Salesman Problem (E-GTSP), which is an extension of the Travelling Salesman Problem (TSP), is stated as follows: given groups of points within a city, like banks, supermarkets, etc., find a minimum cost Hamiltonian cycle that visits each group exactly once. It can model many real-life combinatorial optimization scenarios more efficiently than TSP. This study presents five spatially driven search-algorithms for possible transformation of E-GTSP to TSP by considering the spatial spread of points in a given urban city. Presented algorithms are tested over 15 different cities, classified by their street-network's fractal-dimension. Obtained results denote that the R-Search algorithm, which selects the points from each group based on their radial separation with respect to the start-end point, is the best search criterion for any E-GTSP to TSP conversion modelled for a city street network. The construction industry is known as a hazardous industry because of its complexity and strategic nature. Therefore, it is important to know the main causes of occupational accidents to prevent fatal occupational accidents in construction industry. At building construction sites, workers performing tasks are continuously exposed to risks, not only emerging from their own mistakes but also from the mistakes of their co-workers. A great deal of studies investigating risks and preventing occupational hazards for the construction industry has been carried out in the literature. The quantitative conventional methods mostly use either probabilistic techniques or statistics, or both, but they have limitations dealing with the ambiguity and fuzziness in information. In this study, to overcome these limitations, an applicable and improved approach, which helps construction managers to propose preventive measures for accidents on construction sites, is proposed to simplify the risk assessment. It is shown that the Fuzzy Decision Making Trial and Evaluation Laboratory (DEMATEL) method can evaluate causal factors of occupational hazards by a cause-effect diagram and improve certain safety measures on construction sites. Remarkable progress has been made to date in the discovery of material binding peptides and their utilization in nanotechnology, which has brought new challenges and opportunities. Nowadays phage display is a versatile tool, important for the selection of ligands for proteins and peptides. This combinatorial approach has also been adapted over the past decade to select material-specific peptides. Screening and selection of such phage displayed material binding peptides has attracted great interest, in particular because of their use in nanotechnology. Phage display selected peptides are either synthesized independently or expressed on phage coat protein. Selected phage particles are subsequently utilized in the synthesis of nanoparticles, in the assembly of nanostructures on inorganic surfaces, and oriented protein immobilization as fusion partners of proteins. In this paper, we present an overview on the research conducted on this area. In this review we not only focus on the selection process, but also on molecular binding characterization and utilization of peptides as molecular linkers, molecular assemblers and material synthesizers. Nanoporous gold (np-Au) has intriguing material properties that offer potential benefits for many applications due to its high specific surface area, well-characterized thiol-gold surface chemistry, high electrical conductivity, and reduced stiffness. The research on np-Au has taken place on various fronts, including advanced microfabrication and characterization techniques to probe unusual nanoscale properties and applications spanning from fuel cells to electrochemical sensors. Here, we provide a review of the recent advances in np-Au research, with special emphasis on microfabrication and characterization techniques. Simple Summary: The estimated survival time for glioblastoma patients is extremely low; only about 5% of patients survive five years post diagnosis. The standard of care for glioblastoma patients involves surgery, radiation therapy, and chemotherapy with temozolomide. However, due to the extremely invasive capability of glioblastoma cells, tumors develop very diffusely, integrating into the healthy brain tissue. Indeed, the separation of healthy brain tissue and the tumor boundaries, by standard surgical microscopy, is very challenging. Therefore, the maximum safe removal of the tumor mass is difficult, leaving some tumor cells behind. Therefore, understanding the molecular mechanisms of tumor cell infiltration and developing anti-invasive approaches are of the utmost priority. Here, we provide a review of the characteristics and molecular mechanisms of glioblastoma invasion, and include a perspective of clinical applications. Glioblastoma is the most common and malignant primary brain tumor, defined by its highly aggressive nature. Despite the advances in diagnostic and surgical techniques, and the development of novel therapies in the last decade, the prognosis for glioblastoma is still extremely poor. One major factor for the failure of existing therapeutic approaches is the highly invasive nature of glioblastomas. The extreme infiltrating capacity of tumor cells into the brain parenchyma makes complete surgical removal difficult; glioblastomas almost inevitably recur in a more therapy-resistant state, sometimes at distant sites in the brain. Therefore, there are major efforts to understand the molecular mechanisms underpinning glioblastoma invasion; however, there is no approved therapy directed against the invasive phenotype as of now. Here, we review the major molecular mechanisms of glioblastoma cell invasion, including the routes followed by glioblastoma cells, the interaction of tumor cells within the brain environment and the extracellular matrix components, and the roles of tumor cell adhesion and extracellular matrix remodeling. COVID-19 has brought serious challenges to education and many other sectors. Within the educational context, the main difficulties experienced have been reported as challenges related to not only mechanical impediments—such as technology or infrastructure—but also to methodological and personal features, such as lack of motivation or online learning/teaching experiences. The abrupt transition from face-to-face to online education has created the need for some specific abilities, such as digital literacy on the side of the learners at all educational levels. In this context, this mixed-method study aims to determine the digital literacy levels of learners belonging to different school levels and whether age, gender and school degree were significant variables. This study also investigates the technology-related challenges students experienced during COVID-19 pandemic. The sample consisted of 510 participants representing different school levels, age groups, and genders; in addition, a smaller representative group (n = 30) revealed their own evaluations related to their levels of digital literacy and the technology-related challenges they had about online learning. The findings suggest that there is a statistically significant relationship between students' digital literacy and their gender and school degree, while age was not found to be a statistically significant variable. The qualitative self-reported data suggested that learners have sufficient levels of digital literacy, and that the major technology-related challenges were reported to be lack of the necessary technologies and difficulties in adapting to a new approach to learning. 3D scene classification has become an important research field in photogrammetry, remote sensing, computer vision and robotics with the widespread usage of 3D point clouds. Point cloud classification, called semantic labeling, semantic segmentation, or semantic classification of point clouds is a challenging topic. Machine learning, on the other hand, is a powerful mathematical tool used to classify 3D point clouds whose content can be significantly complex. In this study, the classification performance of different machine learning algorithms in multiple scales was evaluated. The feature spaces of the points in the point cloud were created using the geometric features generated based on the eigenvalues of the covariance matrix. Eight supervised classification algorithms were tested in four different areas from three datasets (the Dublin City dataset, Vaihingen dataset and Oakland3D dataset). The algorithms were evaluated in terms of overall accuracy, precision, recall, F1 score and process time. The best overall results were obtained for four test areas with different algorithms. Dublin City Area 1 was obtained with Random Forest as 93.12%, Dublin City Area 2 was obtained with a Multilayer Perceptron algorithm as 92.78%, Vaihingen was obtained as 79.71% with Support Vector Machines and Oakland3D with Linear Discriminant Analysis as 97.30%. 3D imaging of Earth's surface layers (such as canopy, sub-surface, or ice) requires not just the penetration of radar signal into the medium, but also the ability to discriminate multiple scatterers within a slant-range and azimuth resolution cell. The latter requires having multiple radar channels distributed in across-track direction. Here, we describe the theory of multi-static radar tomography with emphasis on resolution, SNR, sidelobes, and nearest ambiguity location vs. platform distribution, observation geometry, and different multi-static modes. Signal-based 1D and 2D simulations are developed and results for various observation geometries, target distributions, acquisition modes, and radar parameters are shown and compared with the theory. Pros and cons of multi-static modes are compared and discussed. Results for various platform formations are shown, revealing that unequal spacing is useful to suppress ambiguities at the cost of increased multiplicative noise. In particular, we demonstrate that the multiple-input multiple-output (MIMO) mode, in combination with nonlinear spacing, outperforms the other modes in terms of ambiguity, sidelobe levels, and noise suppression. Sustained release and replenishment of the drug depot are essential for the long-term functionality of implantable drug-delivery devices. This study demonstrates the use nanoporous gold (np-Au) thin films for in-plane transport of fluorescein (a small-molecule drug surrogate) over large (mm-scale) distances from a distal reservoir to the site of delivery, thereby establishing a constant flux of molecular release. In the absence of halides, the fluorescein transport is negligible due to a strong non-specific interaction of fluorescein with the pore walls. However, in the presence of physiologically relevant concentration of ions, halides preferentially adsorb onto the gold surface, minimizing the fluorescein–gold interactions and thus enabling in-plane fluorescein transport. In addition, the nanoporous film serves as an intrinsic size-exclusion matrix and allows for sustained release in biofouling conditions (dilute serum). The molecular release is reproducibly controlled by gating it in response to the presence of halides at the reservoir (source) and the release site (sink) without external triggers (e.g., electrical and mechanical). Niemann Pick disease type C (NPC) is a neurovisceral disorder due to mutations in NPC1 or NPC2. This review focuses on poorly characterized clinical and molecular features of early infantile form of NPC (EIF) and identified 89 cases caused by NPC1 (NPC1) and 16 by NPC2 (NPC2) mutations. Extra-neuronal features were common; visceromegaly reported in 80/89 NPC1 and in 15/16 NPC2, prolonged jaundice in 30/89 NPC1 and 7/16 NPC2. Early lung involvement was present in 12/16 NPC2 cases. Median age of neurological onset was 12 (0–24) and 7.5 (0–24) months in NPC1 and NPC2 groups, respectively. Developmental delay and hypotonia were the commonest first detected neurological symptoms reported in 39/89 and 18/89 NPC1, and in 8/16 and 10/16 NPC2, respectively. Additional neurological symptoms included vertical supranuclear gaze palsy, dysarthria, cataplexy, dysphagia, seizures, dystonia, and spasticity. This study analyses the spatio-temporal pattern of parolees using electronic monitoring, where the developed spatial framework supports the Environmental Criminology concepts such as crime patterns or crime attractive locations. A grid-based solution for spatio-temporal analyses is introduced to ensure the anonymity of the parolees. In order to test these developed concepts, the Istanbul Metropolitan Area was selected as the pilot study area. Following the developed concepts of the Crime Pattern Theory, a spatial framework was designed. A novel grid-based weighted algorithm for the most attractive areas was generated via spatial point-of-interest data and a conducted survey among police officers. The designed framework and the spatio-temporal analyses were carried out for 77 parolees using geostatistical methods. The major findings of the study are (a) 24-hour trajectories of each parolee was monitored; (b) the most attractive grids within the city were defined; and (c) for each parolee, the entrance time to the grid and the time spent within that grid were reported and analyzed. This study is a preliminary study for spatio-temporal detection of parolees' trajectories, where location-based analyses serve well. High mortality rates of glioblastoma (GBM) patients are partly attributed to the invasive behavior of tumor cells that exhibit extensive infiltration into adjacent brain tissue, leading to rapid, inevitable, and therapy-resistant recurrence. In this study, we analyzed transcriptome of motile (dispersive) and non-motile (core) GBM cells using an in vitro spheroid dispersal model and identified SERPINE1 as a modulator of GBM cell dispersal. Genetic or pharmacological inhibition of SERPINE1 reduced spheroid dispersal and cell adhesion by regulating cell-substrate adhesion. We examined TGFβ as a potential upstream regulator of SERPINE1 expression. We also assessed the significance of SERPINE1 in GBM growth and invasion using TCGA glioma datasets and a patient-derived orthotopic GBM model. SERPINE1 expression was associated with poor prognosis and mesenchymal GBM in patients. SERPINE1 knock-down in primary GBM cells suppressed tumor growth and invasiveness in the brain. In higher organisms, epithelia separate compartments in order to guarantee their proper function. Such structures are able to seal but also to allow substances to pass. Within the paracellular pathway, a supramolecular structure, the tight junction transport is largely controlled by the temporospatial regulation of its major protein family called claudins. Besides the fact that the expression of claudins has been identified in different forms of human diseases like cancer, clearly defined mutations in the corresponding claudin genes have been shown to cause distinct human disorders. Such disorders comprise the skin and its adjacent structures, liver, kidney, the inner ear, and the eye. From the phenotype analysis, it has also become clear that different claudins can cause a complex phenotype when expressed in different organs. To gain deeper insights into the physiology and pathophysiology of claudin-associated disorders, several mouse models have been generated. In order to model human disorders in detail, they have been designed either as full knockouts, knock-downs or knock-ins by a variety of techniques. Here, we review human disorders caused by CLDN mutations and their corresponding mouse models that have been generated thus far and assess their usefulness as a model for the corresponding human disorder. Adsorption on a functionalized surface can be an effective way of purifying polyphenols from complex plant extracts. Polymeric resins that rely on hydrophobic interactions suffer from low selectivity, weak affinity towards polyphenols, and lack tunability therefore making the purification of polyphenols less efficient. In this study, a purification process for the recovery of polyphenols from grape pomace extract was successfully developed using hydrogen bonding affinity ligands grafted on silica particles and PEG-assisted elution solvents. Bare silica (SiO2) and polyethylene glycol (mPEG)-grafted silica microparticles with molecular weights of 2000 and 5000 were tested to determine their polyphenol binding and release characteristics. Functionalizing the surface of bare silica with mPEG ligands increased the adsorption capacity by 7.1- and 11.4-fold for mPEG-2000 and mPEG-5000 compared to bare silica particles, respectively. This was likely due to the introduction of more polyphenol binding sites with mPEG functionalization. Altering the molecular weight (MW) of mPEG grafted on silica surfaces provided tunability in the adsorption capacity. A complete recovery of polyphenols (~99.9%) from mPEG-grafted silica particles was achieved by utilizing PEG–ethanol or PEG–water cosolvent systems. Recovered polyphenols showed up to ~12-fold antioxidant activity compared to grape pomace extract. This study demonstrates that mPEG-grafted silica particles and elution of polyphenols with PEG cosolvents can potentially be used for large-scale purification of polyphenols from complex plant extracts and simplify the use of polyphenols, as PEG facilitates remarkable solvation and is an ideal medium for the final formulation of polyphenols. The collection of solid waste is a challenging issue, especially in highly urbanized areas. In developing countries, landfilling is currently the preferred method for disposing of solid waste, but each landfill has a limited lifecycle. Therefore, changes in the amount of stored waste should be monitored for the sustainable management of such areas. In this study, volumetric changes in a landfill were examined using a low-cost unmanned aerial vehicle (UAV). Aerial photographs obtained from five different flights, covering approximately two years, were used in the volume calculations. Values representing the amount of remaining space between the solid waste and a reference plane were determined using digital elevation models, which were produced based on the structure from motion (SfM) approach. The obtained results and potential of UAVs in the photogrammetric survey of a landfill were further evaluated and interpreted by considering other possible techniques, ongoing progress, and the information existing in an environmental impact assessment report. As a result of the study, it was proved that SfM carried out using a low-cost UAV has a high potential for use in the reconstruction of a landfill. Outcomes were obtained over a short period, without the need for direct contact with the solid waste, making the UAV preferable for use in planning and decision-making studies. Molecular diagnostics have significantly advanced the early detection of diseases, where the electrochemical sensing of biomarkers (e.g., DNA, RNA, proteins) using multiple electrode arrays (MEAs) has shown considerable promise. Nanostructuring the electrode surface results in higher surface coverage of capture probes and more favorable orientation, as well as transport phenomena unique to nanoscale, ultimately leading to enhanced sensor performance. The central goal of this study is to investigate the influence of electrode nanostructure on electrically-guided immobilization of DNA probes for nucleic acid detection in a multiplexed format. To that end, we used nanoporous gold (np-Au) electrodes that reduced the limit of detection (LOD) for DNA targets by two orders of magnitude compared to their planar counterparts, where the LOD was further improved by an additional order of magnitude after reducing the electrode diameter. The reduced electrode diameter also made it possible to create a np-Au MEA encapsulated in a microfluidic channel. The electro-grafting reduced the necessary incubation time to immobilize DNA probes into the porous electrodes down to 10 min (25-fold reduction compared to passive immobilization) and allowed for grafting a different DNA probe sequence onto each electrode in the array.
