Nowadays, any research discipline is interested in tackling its problems with artificial intelligence and, therefore, is demanding knowledge and frameworks with the aim of developing and using intelligent methods. Within this scenario, neural networks stand out for the important results they have achieved. This paper introduces Nets4Learning, a web platform for designing, training and testing artificial/deep neural network models. The application deals with some of the most popular tasks in the data science field such as tabular classification, regression, image classification and object detection. Nets4Learning has been designed so that any researcher from any discipline can easily develop neural network models without special programming or digital skills. In fact, the user does not have to install anything as the application is publicly available and can be accessed from any device. The genus Pycnoporus includes fungi with great potential for the production of antibiotic substances. It is necessary to develop new models to assess their effectiveness against microorganisms with an economic impact, such as phytopathogenic fungi. The objective of this study is to evaluate three models of Pycnoporus sanguineus for the growth inhibition of the phytopathogens Botrytis cinerea and Fusarium oxysporum. Model 1 involves dual tests of the antagonistic activity of P. sanguineus vs. phytopathogens, Model 2 involves antifungal effectiveness tests of cinnabarin, and Model 3 involves antifungal effectiveness tests of P. sanguineus extract. Models 2 and 3 are contrasted with products containing benomyl and captan. The results show that Model 3 is the most effective in controlling B. cinerea, with an inhibition percentage of 74.34% (p < 0.05) and a decrease in the growth rate (3.85 mm/day; p < 0.05); the same is true for F. oxysporum, with an inhibition percentage of 47.14% (p < 0.05). In general, F. oxysporum exhibits greater resistance (p < 0.05). The results of this study indicate that P. sanguineus extracts may be used as control agents for fungal species in the same way as other Pycnoporus species. Although commercial products are very efficient at inhibiting phytopathogens, one must consider the disadvantages of their use. Retinitis pigmentosa (RP) is a heterogeneous inherited retinal disorder. Mutations in KIZ cause autosomal recessive (AR) RP. We aimed to characterize the genotype, expression pattern, and phenotype in a large cohort of KIZ cases. Sanger and whole exome sequencing were used to identify the KIZ variants. Medical records were reviewed and analyzed. Thirty-one patients with biallelic KIZ mutations were identified: 28 homozygous for c.226C>T (p.R76*), 2 compound heterozygous for p.R76* and c.3G>A (p.M1?), and one homozygous for c.247C>T (p.R83*). c.226C>T is a founder mutation among patients of Jewish descent. The clinical parameters were less severe in KIZ compared to DHDDS and FAM161A cases. RT-PCR analysis in fibroblast cells revealed the presence of four different transcripts in both WT and mutant samples with a lower percentage of the WT transcript in patients. Sequence analysis identified an exonic sequence enhancer (ESE) that includes the c.226 position which is affected by the mutation. KIZ mutations are an uncommon cause of IRD worldwide but are not rare among Ashkenazi Jews. Our data indicate that p.R76* affect an ESE which in turn results in the pronounced skipping of exon 3. Therefore, RNA-based therapies might show low efficacy since the mutant transcripts are spliced. [ABSTRACT FROM AUTHOR]

Current weather monitoring systems often remain out of reach for small-scale users and local communities due to their high costs and complexity. This paper addresses this significant issue by introducing a cost-effective, easy-to-use local weather station. Utilizing low-cost sensors, this weather station is a pivotal tool in making environmental monitoring more accessible and user-friendly, particularly for those with limited resources. It offers efficient in-site measurements of various environmental parameters, such as temperature, relative humidity, atmospheric pressure, carbon dioxide concentration, and particulate matter, including PM 1, PM 2.5, and PM 10. The findings demonstrate the station's capability to monitor these variables remotely and provide forecasts with a high degree of accuracy, displaying an error margin of just 0.67%. Furthermore, the station's use of the Autoregressive Integrated Moving Average (ARIMA) model enables short-term, reliable forecasts crucial for applications in agriculture, transportation, and air quality monitoring. Furthermore, the weather station's open-source nature significantly enhances environmental monitoring accessibility for smaller users and encourages broader public data sharing. With this approach, crucial in addressing climate change challenges, the station empowers communities to make informed decisions based on real-time data. In designing and developing this low-cost, efficient monitoring system, this work provides a valuable blueprint for future advancements in environmental technologies, emphasizing sustainability. The proposed automatic weather station not only offers an economical solution for environmental monitoring but also features a user-friendly interface for seamless data communication between the sensor platform and end users. This system ensures the transmission of data through various web-based platforms, catering to users with diverse technical backgrounds. Strawberry wilt is a disease caused by Fusarium solani, which it provokes the death of the plant. Farmers mainly use chemical methods for its control, which has a negative impact on the environment and human health. Given the growing demand for organic agricultural products, compatible alternatives must be sought for disease management that can reduce the doses of fungicides. A combination of pesticides and biological control agents could be an alternative for the management of F. solani. Consequently, investigations on fungicide compatibility and synergistic effects are recommended in relation to the biological control of strawberry wilt. In this study, potential antagonism was calculated according to the class of mycoparasitism and the percentage inhibition of radial growth in order to later design a compatibility model of the different species of Trichoderma with three protective fungicides at different concentrations. The potential antagonism showed that Trichoderma asperellum presented high compatibility with the fungicides Captan and Mancozeb added in concentrations of 450, 900, and 1350 mg L−1. The use of antagonistic strains together with the fungicide Chlorothalonil in its three concentrations showed a negative effect on the growth of Trichoderma species, which caused low and null compatibility against the MA-FC120 strain of F. solani in vitro. There are several by-products obtained in agricultural systems that can contribute to small-scale production systems within backyards, in this sense, the production of oyster mushrooms (Pleurotus ostreatus) has been integrated into the rural family economy in Mexico. After harvesting the fruiting bodies, the remainder is known as sold-out substrate, this by-product has been destined for other agricultural uses such as: medium for seedlings, vermiculture or fertilizer; however, there are studies where it has been used in the diet of bovine cattle. In this study, the effects of sold-out substrate (corn stubble) of P. ostreatus in the diet of broilers with different percentages of protein, on feed conversion ratio, carcass yield, and broiler meat quality were evaluated. A total of 120 broilers Ross 308 were randomly assigned in 12 pens with concrete floor and curly shaving with an area of 3 m2. During the initial period (1 to 28 days), all broilers were fed commercial feed® mixture. On day 29, the broilers were weighed and divided into four treatment groups and fed diets with different percentages of protein until the end of the experiment. Feed conversion ratio was significantly higher in treatment control (TC) compared to the other treatments; however, the performance parameters were not affected by the diet supplemented with sold-out substrate, likewise, the color and quality of the meat were in a normal range (48 < L* < 51) and with a good amount of crude protein. Sold-out substrate of P. ostreatus from corn stubble presented a high mineral content of Mg, Na, K, Fe, Cu, and Mn suitable to be considered in diets for feeding backyard broilers. Although the aetiology of inflammatory bowel diseases (IBDs) is still unknown, one of their main characteristics is that the immune system chronically affects the permeability of the intestinal lamina propria, in turn altering the composition of the microbiota. In this study, the TNBS rat model of colitis was used because it contains a complex inflammatory milieu of polymorphonuclear cells (PMN) and lymphocytes infiltrating the lamina propria. The aim of the present study was to investigate six dehydrogenases and their respective adaptations in the tissue microenvironment by quantifying enzymatic activities measured under substrate saturation conditions in epithelial cells and leukocytes from the lamina propria of rats exposed to TNBS. Our results show that in the TNBS group, an increased DAI score was observed due to the presence of haemorrhagic and necrotic areas in the colon. In addition, the activities of G6PDH and GADH enzymes were significantly decreased in the epithelium in contrast to the increased activity of these enzymes and increased lactate mediated by the LDH-A enzyme in leukocytes in the lamina propria of the colon. Over the past years, evidence has emerged illustrating how metabolism supports aspect of cellular function and how a metabolic reprogramming can drive cell differentiation and fate. Corn cultivation represents the largest type of agricultural production in Mexico, with great economic, social, and cultural importance. The health of corn producers could be compromised by the extensive and accumulated use of pesticides. The effects of pesticides in terms of their cytotoxic and genotoxic damage in two groups of peasant maize producers in Tlaxcala, Mexico, were considered here. The buccal micronucleus cytome assay was used as an indicator of cytotoxicity and genotoxicity, along with nuclear abnormalities present in farmers who had used pesticides in the last thirty years. In total, 21 commercial products used in corn production were identified, mainly herbicides belonging to the chlorophenoxy, triazine, and organophosphate compounds; in addition, it was observed that a small group of farmers use the active ingredient carbofuran, as well as insecticides and fungicides. The results show that farmers with higher pesticide use present higher rates of cytotoxic and genotoxic damage compared to the group of producers with higher incidence rates of agroecological practices and lower rates of pesticide use, as revealed by the micronucleus assay, as well as by nuclear abnormalities present in the epithelial cells of the buccal mucosa. The manzano pepper (Capsicum pubescens) is an exportation product that generates substantial earnings for local producers in Mexico. Herein we report on the most relevant metabolic changes that occur during the postharvest rot of manzano peppers caused by Fusarium temperatum. Simultaneously, we describe the effect of the Zanthoxylum limoncello leaf essential oil (ZlEO) and its major volatiles on the control of this devastating disease. According to our results, ZlEO, 2-undecanone (34%), 2-undecenal (32%), and 2-dodecenal (8%) exerted in vitro fungicide activity on F. temperatum (MIC, 104.6–218.3 mg L−1) and a strong in situ fungistatic effect in manzano peppers previously infected with F. temperatum. A differential fungistatic activity was observed for the natural agents assayed. However, the best results were confirmed with 2-dodecenal, which improved the shelf life of infected peppers up to 16 d post-inoculation. The protective effect of ZlEO and its major volatiles resulted in the conservation of fruit firmness, pH, protein, fat, fiber, ascorbic acid, and nutraceuticals of manzano peppers (carotenoids and capsaicinoids). Breast cancer (BC) is the most frequently diagnosed cancer and is the second-most common cause of death in women worldwide. Because of this, the search for new drugs and targeted therapy to treat BC is an urgent and global need. Histone deacetylase 6 (HDAC6) is a promising anti-BC drug target associated with its development and progression. In the present work, the design and synthesis of a new family of dihydropyrazole-carbohydrazide derivatives (DPCH) derivatives focused on HDAC6 inhibitory activity is presented. Computational chemistry approaches were employed to rationalize the design and evaluate their physicochemical and toxic-biological properties. The new family of nine DPCH was synthesized and characterized. Compounds exhibited optimal physicochemical and toxicobiological properties for potential application as drugs to be used in humans. The in silico studies showed that compounds with –Br, –Cl, and –OH substituents had good affinity with the catalytic domain 2 of HDAC6 like the reference compounds. Nine DPCH derivatives were assayed on MCF-7 and MDA-MB-231 BC cell lines, showing antiproliferative activity with IC50 at μM range. Compound 2b showed, in vitro, an IC50 value of 12 ± 3 µM on human HDAC6. The antioxidant activity of DPCH derivatives showed that all the compounds exhibit antioxidant activity similar to that of ascorbic acid. In conclusion, the DPCH derivatives are promising drugs with therapeutic potential for the epigenetic treatment of BC, with low cytotoxicity towards healthy cells and important antioxidant activity. Phaseolus vulgaris is the most legume cultivated in the world; in Mexico, it is considered the second most important crop after corn. The aim of this research was to determine the characteristics of Xanthomonas campestris strain "Xcf1-APJR" isolated from the leaves of bean crops, and determine the antimicrobial activity of cinnabarin on this strain. Bacterial cultures were obtained from leaves with necrotic leaf spot symptoms of bean plant variety "Flor de Mayo M38" in Puebla, Mexico. The antimicrobial activity of cinnabarin was tested at 7, 14 and 21 days on X. campestris pv. campestris. The Xcf1-APJR strain showed 100% identity with X. campestris pv. campestris as a causal agent of necrotic leaf spot. Treatment with a potato dextrose medium with a dehydrated sugar cane (PDA+C) showed a higher orange pigmentation than the other treatments after 7, 14 and 21 days of incubation and a higher concentration of cinnabarin (54.33 InU/g) with in vitro antimicrobial activity against X. campestris pv. campestris. Featured Application: The synthesis of the nutraceutical resveratrol by filamentous fungi represents a new and exciting area for bioprocess research. Considering that resveratrol was initially placed as a natural phytoalexin from plants, its biosynthesis by fungi may be regulated by several factors including a broad spectrum of small molecules and enzymes involved in chemical defense. This work reports on the effect of some elicitors that promote resveratrol biosynthesis in fungi and how they can be used to produce this stilbene under controlled conditions. The chemical factors that regulate the synthesis of resveratrol (RV) in filamentous fungi are still unknown. This work reports on the RV production by Arcopilus aureus MaC7A under controlled conditions and the effect of amino acid precursors (PHE and TYR), monoterpenes (limonone, camphor, citral, thymol, menthol), and mixtures of hydrolytic enzymes (Glucanex) as elicitors for boosting fungal RV. Batch cultures with variable concentrations of PHE and TYR (50–500 mg L−1) stimulated RV production from 127.9 ± 4.6 to 221.8 ± 5.2 mg L−1 in basic cultures developed in PDB (pH 7) added with 10 g L−1 peptone at 30 °C. Maximum levels of RV and biomass were maintained during days 6–8 under these conditions, whereas a dramatic RV decrease was observed from days 10–12 without any loss of biomass. Among the tested volatiles, citral (50 mg L−1) enhanced RV production until 187.8 ± 2.2 mg L−1 in basic cultures, but better results were obtained with Glucanex (100 mg L−1; 198.3 ± 7.6 mg L−1 RV). Optimized batch cultures containing TYR (200 mg L−1), citral (50 mg L−1), thymol (50 mg L−1), and Glucanex (100 mg L−1) produced up to 237.6 ± 4.7 mg L−1 of RV. Our results suggest that low concentrations of volatiles and mixtures of isoenzymes with β-1, 3 glucanase activity increase the biosynthesis of fungal RV produced by A. aureus MaC7A in batch cultures. Featured Application: This work was focused in the generation of six yeast lines able to produce resveratrol from p-coumaric acid. Industrial use of these strains may be contemplated. Resveratrol is a nutraceutical with relevant benefits to human health. This investigation reports on the generation and evaluation of six recombinant yeast lines that produce resveratrol from p-coumaric acid. The yeast lines contained a single p-coumaric acid-Co-A ligase from Plagiochasma appendiculatum combined with the stilbene synthases from Parthenocissus henryana, Polygonum cuspidatum, Morus alba var. atropurpurea, Rheum tataricum, Vitis vinifera and Arachis hypogaea. Codon optimized versions of these sequences were inserted in an expression vector flanked by the constitutive PGK and GPD promoters before expression in Saccharomyces cerevisiae. Batch fermentation (60 h) revealed that yeast lines had different capacities (p < 0.01) to produce resveratrol. Slightly acidic pH (6) and concentrations <100 mg L−1p-coumaric acid improved resveratrol yields. Among the six lines, those containing the stilbene synthases (STS) from P. cuspidatum and M. alba produced up to 39 mg L−1 using 70 mg L−1p-coumaric acid. On the other hand, lines expressing STS from V. vinifera, A. hypogaea and R. tataricum generated resveratrol faster than other lines but accumulated lower amounts at the end of the batch period (27–30 mg L−1). Laser ablation has several advantages over the chemical synthesis of nanoparticles due to its simplicity and because it is a faster and cleaner process. In this paper, we use femtosecond laser ablation to generate highly concentrated silver colloidal nanoparticle solutions. Those high concentrations usually lead to agglomeration of the nanoparticles, rendering the solution nearly useless. We employ two different organic stabilizers (hexadecyltrimethylammonium bromide, CTAB, and polyvinylpyrrolidone, PVP) to avoid this problem and study their effect on the nanoparticle size distribution, structural characteristics, and the solution concentration. Myeloperoxidase (MPO) is an enzyme present in human neutrophils, whose main role is to provide defenses against invading pathogens. However, highly reactive oxygen species (ROS), such as HOCl, are generated from MPO activity, leading to chronic diseases. Herein, we report the microwave-assisted synthesis of a new series of stable (E)-(2-hydroxy)-α-aminocinnamic acids, in good yields, which are structurally analogous to the natural products (Z)-2-hydroxycinnamic acids. The radical scavenging activity (RSA), MPO inhibitory activity and cytotoxicity of the reported compounds were evaluated. The hydroxy derivatives showed the most potent RSA, reducing the presence of DPPH and ABTS radicals by 77% at 0.32 mM and 100% at 0.04 mM, respectively. Their mechanism of action was modeled with BDEOH, IP and ΔEH-L theoretical calculations at the B3LYP/6 − 31 + G(d,p) level. Compounds showed in vitro inhibitory activity of MPO with IC50 values comparable to indomethacin and 5-ASA, but cytotoxicities below 15% at 100–200 µM. Docking calculations revealed that they reach the amino acid residues present in the distal cavity of the MPO active site, where both the amino and carboxylic acid groups of the α-aminopropenoic acid arm are structural requirements for anchoring. The use of synthetic dyes in the textile, leather, and paper industries is a source of groundwater pollution around the world. There are different methods for the treatment of wastewater that has been contaminated with dyes, among which adsorption with agro-industrial wastes is gaining relevance. In the present study, the adsorption capacity of cocoa bean shell powder was evaluated when it was used as a bioadsorbent for Congo red dye in an aqueous medium. A 24 central factorial design with central and axial points was proposed to determine the adsorption capacity. The factors that were studied were the adsorbent (0.06–0.15 g), Congo red (40–120 mg L−1), pH (3–11), and time (4–36 h). The bioadsorbent was characterized through scanning electron microscopy and Fourier-transform infrared spectroscopy. The effects of the factors on the adsorption capacity for Congo red using cocoa bean shell were nonlinear, and they were modeled with a second-order polynomial (p < 0.05) and with an R2 of 0.84. The bioadsorbent obtained a maximum adsorption of 89.96% in runs. The process of optimization by using the surface response allowed the maximization of the adsorption, and the validation showed that 95.79% adsorption of the dye was obtained. [ABSTRACT FROM AUTHOR]
