Musculoskeletal disorders are prevalent among agricultural workers, increasing the risk of work-related diseases due to manual labor, repetitive tasks, and prolonged postures. This study evaluates the association between physical behaviors during work and non-work, measured objectively, and musculoskeletal pain in the neck, upper back, and lower back in viticulture workers. A cross-sectional quantitative study was conducted with 75 viticulturists of both sexes aged 18 years or older. An accelerometer measured physical behaviors (lying down, sitting, standing, moving, walking, and sleeping) during work and non-work periods. Pain intensity was quantified using a 0–10 scale and categorized as "Low" and "High" pain intensity. Binary logistic regression tested the association between pain and time spent on physical behaviors. Results indicated a high prevalence of pain: 46.7% cervical, 52% upper back, and 60% lower back. Standing was the most common behavior during work, while lying and sitting were predominant during non-work. An increased sleeping time was associated with a decreased probability of experiencing high-intensity neck pain. Increased time spent lying down during non-work hours was associated with an increased probability of experiencing high-intensity upper back pain. No physical behavior was associated with high-intensity lower back pain. In conclusion, sedentary behaviors worsen upper back pain, and sleep reduces neck pain in viticulture workers. Background: Onychomycosis numbers among the most common fungal infections in humans affecting finger- or toenails. Histology remains a frequently applied screening technique to diagnose onychomycosis. Screening slides for fungal elements can be time-consuming for pathologists, and sensitivity in cases with low amounts of fungi remains a concern. Convolutional neural networks (CNNs) have revolutionized image classification in recent years. The goal of our project was to evaluate if a U-NET-based segmentation approach as a subcategory of CNNs can be applied to detect fungal elements on digitized histologic sections of human nail specimens and to compare it with the performance of 11 board-certified dermatopathologists. Methods: In total, 664 corresponding H&E- and PAS-stained histologic whole-slide images (WSIs) of human nail plates from four different laboratories were digitized. Histologic structures were manually annotated. A U-NET image segmentation model was trained for binary segmentation on the dataset generated by annotated slides. Results: The U-NET algorithm detected 90.5% of WSIs with fungi, demonstrating a comparable sensitivity with that of the 11 board-certified dermatopathologists (sensitivity of 89.2%). Conclusions: Our results demonstrate that machine-learning-based algorithms applied to real-world clinical cases can produce comparable sensitivities to human pathologists. Our established U-NET may be used as a supportive diagnostic tool to preselect possible slides with fungal elements. The genetic diversity of pear local varieties prospected in mountainous areas from Northeastern Spain (Pyrenees and Iberian Cordillera) is not well known so far. In this study, an overall set of 252 accessions (178 prospected in mountainous areas from Aragon and a diverse set of 74 reference cultivars) was analyzed using 14 SSRs in order to estimate its genetic diversity and to identify the genetic structure and relationships among the pear germplasm studied. A total of 251 distinct alleles were successfully amplified with an average of 17.9 alleles per locus and with a wide genetic diversity (mean expected heterozygosity of 0.82). In total, 228 unique genotypes were identified and 210 genotypes were represented by a single accession indicating a situation of extreme vulnerability of these pear genetic resources held in the CITA collection. An amount of 32.9% of accessions were considered triploids displaying three alleles at least into two loci. Genetic analyses performed by a model-based Bayesian procedure, principal coordinate analysis and analysis of molecular variance supported the presence of a genetic stratification with the existence of four sub-groups among the accessions, with a highly significant differentiation (FST = 0.132; p < 0.001). These results shed light on the characterization and genetic relatedness between these local accessions and currently cultivated pear cultivars and highlight the importance to safeguarding this diversity that might be essential for new breeding programs. Japanese plum (Prunus salicina Lindl.) is widely distributed in temperate zones across the world. Since its introduction to USA in the late 19th century, this species has been hybridized with up to 15 different diploid Prunus species. This high level of introgression has resulted in a wide range of traits and agronomic behaviors among currently grown cultivars. In this work, 161 Japanese plum-type accessions were genotyped using a set of eight Simple Sequence Repeats (SSR) markers to assess the current genetic diversity and population structure. A total of 104 alleles were detected, with an average of 13 alleles per locus. The overall Polymorphic Informative Content (PIC) value of SSR markers was 0.75, which indicates that these SSR markers are highly polymorphic. The Unweighted Pair Group Method with Arithmetic (UPGMA) dendrogram and the seven groups inferred by Discriminant Analysis of Principal Components (DAPC) revealed a strong correlation of the population structure to the parentage background of the accessions, supported by a moderate but highly significant genetic differentiation. In the last years, an important renewal of plant material from different breeding programs is taking place in apricot in order to improve resistance to biotic stresses, extension of the harvest season, fruit quality, and productivity. However, the graft compatibility of many of these cultivars with most popular Prunus rootstocks is unknown, and this is an essential agronomical trait for their better performance and longevity. Hence, the introduction of new cultivars requires knowledge of the extent and nature of incompatibility reactions before releasing these cultivars on the market. In this study, the determination of graft compatibility was carried out in 13 new apricot cultivars grafted on four Prunus rootstocks: 'Marianna2624' (P. cerasifera × P. musoniana), 'Miragreen' (P. cerasifera × P. davidiana), 'Mirared' (P. cerasifera × Nemared), and 'Montclar' (P. persica L. seedlings) at early stages of development. By combining cytomorphological, anatomical, and phenylalanine ammonia-lyase (PAL) gene expression analysis at the graft interface, as well as different vegetative parameters, the results highlighted 'Miragreen' and 'Mirared' as promising rootstocks for apricot, showing the highest degree of compatibility with more than 90% of the apricot cultivars. These results provide useful information for breeders and growers by selecting the most suitable scion-rootstock for efficient orchard design by planting compatible graft combinations. In viticulture, grafting is used to propagate Phylloxera-susceptible European grapevines, thereby using resistant American rootstocks. Although scion–rootstock reciprocal signaling is essential for the formation of a proper vascular union and for coordinated growth, our knowledge of graft partner interactions is very limited. In order to elucidate the scale and the content of scion–rootstock metabolic interactions, we profiled the metabolome of eleven graft combination in leaves, stems, and phloem exudate from both above and below the graft union 5–6 months after grafting. We compared the metabolome of scions vs. rootstocks of homografts vs. heterografts and investigated the reciprocal effect of the rootstock on the scion metabolome. This approach revealed that (1) grafting has a minor impact on the metabolome of grafted grapevines when tissues and genotypes were compared, (2) heterografting affects rootstocks more than scions, (3) the presence of a heterologous grafting partner increases defense-related compounds in both scion and rootstocks in shorter and longer distances from the graft, and (4) leaves were revealed as the best tissue to search for grafting-related metabolic markers. These results will provide a valuable metabolomics resource for scion–rootstock interaction studies and will facilitate future efforts on the identification of metabolic markers for important agronomic traits in grafted grapevines. Type 2 diabetes (T2D) heterogeneity is a major determinant of complications risk and treatment response. Using cluster analysis, we aimed to stratify glycemia within metabolic multidimensionality and extract pathophysiological insights out of metabolic profiling. We performed a cluster analysis to stratify 974 subjects (PREVADIAB2 cohort) with normoglycemia, prediabetes, or non-treated diabetes. The algorithm was informed by age, anthropometry, and metabolic milieu (glucose, insulin, C-peptide, and free fatty acid (FFA) levels during the oral glucose tolerance test OGTT). For cluster profiling, we additionally used indexes of metabolism mechanisms (e.g., tissue-specific insulin resistance, insulin clearance, and insulin secretion), non-alcoholic fatty liver disease (NAFLD), and glomerular filtration rate (GFR). We found prominent heterogeneity within two optimal clusters, mainly representing normometabolism (Cluster-I) or insulin resistance and NAFLD (Cluster-II), at higher granularity. This was illustrated by sub-clusters showing similar NAFLD prevalence but differentiated by glycemia, FFA, and GFR (Cluster-II). Sub-clusters with similar glycemia and FFA showed dissimilar insulin clearance and secretion (Cluster-I). This work reveals that T2D heterogeneity can be captured by a thorough metabolic milieu and mechanisms profiling—metabolic footprint. It is expected that deeper phenotyping and increased pathophysiology knowledge will allow to identify subject's multidimensional profile, predict their progression, and treat them towards precision medicine. Grafting is the most used propagation method in viticulture and is the unique control strategy against Phylloxera. Nevertheless, its practice remains limited mainly due to inconsistent graft success and difficulties in predicting graft compatibility responses of proposed scion–rootstock combinations, slowing down the selection of elite rootstocks. Aiming to identify optimal phenotypic parameters related to graft (in)compatibility, we used four clones of two grapevine cultivars that show different compatibility behavior when grafted onto the same rootstock. Several physiological parameters, internal anatomy of the graft union, chlorophyll fluorescence, and pigment contents of homo- and heterografts were monitored in a nursery-grafting context. The measurements highlighted enhanced performance of the heterografts due to rooting difficulties of Vitis vinifera homografts. This suggests that in viticulture, homografts should only be used as compatibility controls regarding qualitative attributes. By observing the internal anatomy of the union, we found that grapevines might require longer times for graft healing than anticipated. While Affinity Coefficients were not informative to assess incompatibility, leaf chlorophyll concentration analysis proved to be a more sensitive indicator of stress than the analysis of chlorophyll fluorescence.
