Panicum maximum is planted extensively in tropical and subtropical areas, due to its high-quality forage and high biomass yield. This study aims to assess the varied metabolic dynamics of P. maximum subject to different pollution-related wastewater levels, thus providing information for sustainable agriculture and soil restoration. We analyzed the primary and secondary metabolites in P. maximum subject to two different types of polluted wastewater (WW), compared to a control group. The alterations observed in the metabolite profiles were affected by several factors, including nutrient imbalances and oxidative stress induced by heavy metal accumulation. Initially, the increased nutrient availability stemming from wastewater treatment promoted plant growth; however, this positive effect was later diminished by the adverse impacts of heavy metals, which generated oxidative stress, resulting in metabolic disturbances and a decrease in the plant biomass. Importantly, the substantial increase in antioxidant enzymes, related to primary (e.g., sugars) and secondary metabolites (e.g., phenolics and flavonoids), underscores plants' adaptive strategies to cope with stress. The increased biosynthesis of flavonoids and phenolic compounds is a protective mechanism against oxidative stress, which also improves the antimicrobial activity, following the activation of key biosynthetic pathways involved in their synthesis. These complex interactions among diverse metabolites suggest that plants exposed to polluted wastewater use various biochemical strategies to increase both their survival and defenses against pathogens. Collectively, these findings emphasize the significance of understanding how wastewater management practices can affect plant health, metabolic responses, and the broader implications for food safety and ecosystem stability. Drought stress is an important challenge to global food security and agricultural output, and dramatic and rapid climate change has made the problem worse, causing unexpected impacts on the growth, development, and yield of different plants. Understanding the biochemical, ecological, and physiological reactions to these pressures is essential for improved management. Carbon materials' impacts on plants subjected to different stresses are still poorly studied. Thus, this study was carried out investigate the feasibility of applying carbon nanotubes (CNTs) (0, 20, and 40 mg/L) as a foliar treatment for mitigating the effect of water stress (100%, 75%, and 50% irrigation water, IW) on peanut plants growing in sandy soil through assessments of growth and productivity and some physiological and biochemical measurements. Exposure of peanuts to decreased irrigation water led to significant decreases in growth, yield, photosynthetic pigments, indole acetic acid (IAA), and some nutritional components in peanut seeds, but increased levels of osmolytes such as total soluble carbohydrates (TSS) and proline, in addition to free amino acids and phenolics. However, foliar spraying with CNTs could ameliorate the impacts of decreased irrigation water on growth and production via enhancing the studied physiological parameters, such as photosynthetic pigments, IAA, osmolytes, and phenolics. Furthermore, the application of carbon nanotubes improved the nutrient contents, as expressed by the oil yield, protein yield, total carbohydrates, antioxidant activities (DPPH), B-carotene, lycopene, and flavonoids in peanut seeds, either under normal or water stress conditions. The higher level of CNTs (40 mg/L) was more effective than the lower one (20 mg/L) at increasing the above-mentioned parameters. Textile dyes are one of the major water pollutants released into water in various ways, posing serious hazards for both aquatic organisms and human beings. Bioremediation is a significantly promising technique for dye decolorization. In the present study, the fungal strain Lasiodiplodia sp. was isolated from the fruiting bodies of Schizophyllum for the first time. The isolated fungal strain was examined for laccase enzyme production under solid-state fermentation conditions with wheat bran (WB) using ABTS and 2,6-Dimethoxyphenol (DMP) as substrates, then the fermented wheat bran (FWB) was evaluated as a biosorbent for Congo red dye adsorption from aqueous solutions in comparison with unfermented wheat bran. A Box–Behnken design was used to optimize the dye removal by FWB and to analyze the interaction effects between three factors: fermentation duration, pH, and dye concentration. Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and scanning electron microscopy (SEM) were applied to study the changes in the physical and chemical characteristics of wheat bran before and after fermentation. An additional experiment was conducted to investigate the ability of the Lasiodiplodia sp. YZH1 to remove Congo red in the dye-containing liquid culture. The results showed that laccase was produced throughout the cultivation, reaching peak activities of ∼6.2 and 22.3 U/mL for ABTS and DMP, respectively, on the fourth day of cultivation. FWB removed 89.8% of the dye (100 mg L−1) from the aqueous solution after 12 h of contact, whereas WB removed only 77.5%. Based on the Box–Behnken design results, FWB achieved 93.08% dye removal percentage under the conditions of 6 days of fermentation, pH 8.5, and 150 mg L−1 of the dye concentration after 24 h. The fungal strain removed 95.3% of 150 mg L−1 of the dye concentration after 8 days of inoculation in the dye-containing liquid culture. Exotic plants usually exhibit problems for native species where they coexist. This study evaluated the effect of naturalized alien Cenchrus echinatus L. on native plants in urban vegetation. A field trial was conducted to assess the effect of this species on the cover and diversity of the native vegetation. The allelopathic potential of such species was examined. Sites comprising C. echinatus had a lower cover than some native species. Lower floristic diversity was observed at higher densities of this plant. The soil under this plant attained lower N, P, and K contents. This soil had no effect on the germination and growth of native species. It also comprised germinable seeds of some species which were absent from the standing vegetation. Exotic C. echinatus may exert negative effects on the native vegetation of the urban plant communities. A dense cover of this species may inhibit the germination of native species, leading to a reduction in their cover. Reduction in cover and diversity of native species may not be attributed to allelopathy. These results suggest that naturalized C. echinatus may be more competitive than the native ones, particularly at higher densities. Maintaining the plant architecture of Psidium guajava L. (guava tree) is essential for enhancing capture and distribution in the plant, directly affecting the fruit quality. The lifespan of the harvest period can be extended by proper pruning. Both timeliness and proper pruning play crucial roles in achieving high-quality fruit production and in maintaining a consistent fruit size while stimulating ascorbic acid levels, sugar content, total soluble solids (TSS), and titratable acidity. From this perspective, this study aimed to characterize the influence of different intensities of fruit pruning and thinning on guava trees grown under a seasonal tropical climate in two growing seasons in Currais, Piauí, Brazil. The experiment was set up in a randomized block design with a 3 × 3 × 2 factorial arrangement corresponding to short, medium, and long pruning intensities and 0%, 10%, and 20% thinning intensities during the two growth seasons, respectively. An analysis was performed to discriminate the treatment groups according to the physicochemical variables of the guava tree cv. Paluma and canonical discriminant analysis. There was significant variation in the SS, titratable acidity, ascorbic acid, and pH contents. Cluster analysis of all treatments allowed division into five different groups for the two pruning times. Canonical discriminant analysis showed that the first two canonical variables explained 91% of the total variance. The fruits of the second harvest exhibited a lower level of acidity, higher levels of soluble solids, and higher levels of ascorbic acid contents. In addition, these fruits also obtained better nutrient contents. Short pruning with up to 20% thinning, medium pruning with up to 10%, and long pruning without thinning favored better levels of macronutrients and micronutrients and, consequently, better fruit quality. Medium or long pruning with up to 20% thinning resulted in higher average fruit weights and nutrient contents (especially of Fe and Cu), lower acidity, and higher ascorbic acid contents. Sugarcane harvesting requires improvements, particularly in cutting tools. Continuous cutting saws have been introduced as a solution to this issue. This study evaluates the performance of two basal sugarcane cutting systems in different fields: a traditional impact cut system (ICS) with knives and a continuous cut system (CCS) with saw blades. Tests were conducted during two crop cycles in three areas, using a 3 × 2 factorial design with two cutting devices and four replications per treatment. Cut quality indices and ratoon damage were analyzed using descriptive statistics. Raw material losses were subjected to the Shapiro–Wilk normality test, ANOVA, and Tukey's test at 5% probability. Significant differences in cutting quality were found across different areas. The total crop productivity influenced sugarcane cut quality, with the CCS showing (0.8 Mg ha−1) visible losses in higher productivity areas, which is a 74% increase compared to the ICS. In lower productivity areas, the CCS demonstrated better loss performance (0.8 Mg ha−1). Additionally, the stumps damage rate for the CCS was lower than that for the ICS (0.15 and 0.28, respectively), indicating that saws can preserve cane fields and enhance longevity. Delayed sowing of wheat is a common problem in Punjab that exacerbates serious yield loss. However, individual or combined applications of seed rate and slow-release nitrogen fertilizer significantly impacted their efficacy in improving crop growth and productivity. In this regard, the present study explored the potential of slow-release nitrogen fertilizers (control (CK), simple urea (SU), neem-coated urea (NCU), and sulfur-coated urea (SCU)) to improve the growth, yield, and physiological and biochemical attributes of delayed sown wheat with three seed rate [(100 kg ha−1 (S1), 125 kg ha−1 (S2), and 150 kg ha−1 (S3)]. The study was carried out at the Postgraduate Agricultural Research Station of the University of Agriculture Faisalabad in 2018–19 and 2019–20. The study findings revealed that the sulfur-coated urea performed better under S3 seed rate. Combining SCU with S3 significantly increased leaf area index by 0.99 cm2 and plant height by 8.24% compared to simple urea, maximum spikelets, and spike length by 3.9 and 3.8 cm, respectively. The SCU with S3 treatment also increased the biological yield by 43% and 41% and the grain yield by 46% in 2018–2019 and 2019–2020, respectively, compared to CK. Similarly, higher N contents in straw and grain were recorded in the interaction of SCU with S3 treatment. Likewise, the SCU with S3 treatment enhanced the physiological attributes, i.e., chlorophyll contents and transpiration rate, by 18% and 25%, respectively, and decreased internal CO2 by 25.5%, compared to CK. In conclusion, applying sulfur-coated urea with a seed rate of 150 kg ha−1 could be considered a potential strategy for improving the growth and productivity of delayed sown wheat. Plants are sessile and mostly exposed to various environmental stresses which hamper plant growth, development, and significantly decline its production. Drought stress is considered to be one of the most significant limiting factors for crop plants, notably in arid and semi-arid parts the world. Therefore, the present study aimed to evaluate the potential impact of different concentrations (10, 100, and 200 µg/mL) of kinetin capped zinc oxide nanoparticles (Kn-ZnONPs) on Vigna radiata (L.) R. Wilczek under varying levels (5%, 10%, 15%) of PEG-induced drought stress. ZnONPs were synthesized by a co-precipitation method using Zinc acetate as a precursor at pH-12, incinerated to 500 °C, and kinetin was used as a surface functionalizing agent. The resulting Kn-ZnONPs were characterized by various contemporary analytical techniques, including SEM, SEM-EDS, XRD, DLS, and Zeta potential and IR spectroscopy. Crystalline Kn-ZnONPs, with a zeta potential of 27.8 mV and a size of 67.78 nm, of hexagonal wurtzite structure and vibrational stretches associated with N-H, C-O, C-N, etc., were confirmed. PEG-induced drought stress significantly reduced the growth of V. radiata by declining the chlorophyll and carotenoid contents. Moreover, a significant decrease in the levels of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), soluble sugar contents, proline, protein contents, phenol, and tannin were observed compared to the control. However, the exogenous application of Kn-ZnONPs ameliorated all photosynthetic parameters by up-regulating the antioxidant defense system through the promotion of SOD, POD, CAT, and lipid peroxidation levels. The biochemical parameters, such as proteins, soluble sugars, and proline, were observed to be maximum in plants treated with 200 µg/mL Kn-ZnONPs under 5% drought stress. The application of Kn-ZnONPs also enhanced the total phenol contents, flavonoid, and tannin contents. In conclusion, the findings of this study demonstrate that the exogenous application of Kn-ZnONPs provides beneficial effects to V. radiata by attenuating the damaging effects of drought stress through the up-regulation of the antioxidant defense system and osmolytes. Highlights: Jatropha curcas L. has great potential to be used as a model plant in several studies involving native forest species. The immersion in the 2,4-D solution accelerated the emission of primary roots in hardwood cuttings. Studies on vegetative propagation of native species can use Jatropha curcas L. species as a model for obtaining important information in a short time and reducing labor costs. The immersion of cuttings of native species in solutions with low concentrations of 2,4-D can favor the rooting process and vegetative propagation. Even though it is a forest native plant, there are already several studies evaluating the small genome of Jatropha curcas L., which belongs to the Euphorbiaceae family, and may be an excellent representative model for the other plants from the same family. Jatropha curcas L. plant has fast growth, precocity, and great adaptability, facilitating silvicultural studies, allowing important information to be obtained quickly, and reducing labor costs. This information justifies the use of the species as a model plant in studies involving the reproduction of native plants. This study aimed to evaluate the possibility of using Jatropha curcas L. as a model plant for studies involving native forest plants and establish possible recommendations for the vegetative propagation of the species using hardwood cuttings. The information collected can be helpful to other native forest plant species, similar to Jatropha curcas L. To this end, the effects of hardwood cutting length (10, 20, and 30 cm) and the part of the hardwood cuttings (basal, middle, and apex) were evaluated. Moreover, the influence of immersing the hardwood cuttings in solutions containing micronutrients (boron or zinc) or plant regulators (2,4-D, GA3) and a biostimulant composed of kinetin (0.09 g L−1), gibberellic acid (0.05 g L−1), and 4-indole-3-butyric acid (0.05 g L−1). The experiments were carried out in duplicates. In one duplicate, sand was used as the substrate, and rooting evaluations were made 77 days after planting. In another duplicate, a substrate composed of 50% soil, 40% poultry litter, and 10% sand was used, and the evaluations of the saplings were performed 120 days after planting. The GA3 solutions inhibited the roots' and sprouts' emissions, while immersion in 2,4-D solution increased the number of primary roots at 77 days after planting. This study aims to provide possible utilization of sunflower head byproduct (SFH) as a feedstuff by implementing chemical pretreatments (4% sodium hydroxide (SFHNaOH) or 4% urea (SFHurea) and supplementation with either exogenous fibrolytic enzymes (EFE) or functional feed additive (FFA). The experimental EFE was a complex (1:1, v/v) of two enzyme products with high activity of β-1,3-1,4-glucanase and endo-1,4-β-D-xylanase and applied at 0 (SFHout), 1, 2, 5, and 10 µL/ gdry matter, while FFA was a fermentation byproduct rich in cellulase and xylanase activities, applied at 0 (SFHout), 0.5, 1, 2, and 4 mg/g DM. SFHurea had the highest (p < 0.05) crude protein (CP) content compared to other SFH substrates. Linear enhancements (p < 0.05) in kinetics of gas production (GP), metabolizable energy (ME), organic matter digestibility (OMD) and total short-chain fatty acids (SCFAs) concentrations were observed for all SFH substrates supplemented with EFE. The SFHout had the highest (p < 0.05) potential GP, maximum rate (Rmax) of GP, ME, OMD and SCFAs. Supplementation of EFE was more pronounced than FFA in affecting the kinetic parameters of in vitro GP for all SFH substrates. Type 2 diabetes mellitus (T2DM) is a notable health care load that imposes a serious impact on the quality of life of patients. The small amount of reported data and multiple spectra of pathophysiological mechanisms of T2DM make it a challenging task and serious economic burden in health care management. Abrus precatorius L. is a slender, perennial, deciduous, and woody twining plant used in various regions of Asia to treat a variety of ailments, including diabetes mellitus. Various in vitro studies revealed the therapeutic significance of A. precatorius against diabetes. However, the exact molecular mechanism remains unclarified. In the present study, a network pharmacology technique was employed to uncover the active ingredients, their potential targets, and signaling pathways in A. precatorius for the treatment of T2DM. In the framework of this study, we explored the active ingredient–target–pathway network and figured out that abrectorin, abrusin, abrisapogenol J, sophoradiol, cholanoic acid, precatorine, and cycloartenol decisively contributed to the development of T2DM by affecting AKT1, MAPK3, TNFalpha, and MAPK1 genes. Later, molecular docking was employed to validate the successful activity of the active compounds against potential targets. Lastly, we conclude that four highly active constituents, namely, abrusin, abrisapogenol J, precatorine, and cycloartenol, help in improving the body's sensitivity to insulin and regulate the expression of AKT1, MAPK3, TNFalpha, and MAPK1, which may act as potential therapeutic targets of T2DM. Integrated network pharmacology and docking analysis revealed that A. precatorius exerted a promising preventive effect on T2DM by acting on diabetes-associated signaling pathways. Pelargonium graveolens, rose-scented geranium, is commonly used in the perfume industry. P. graveolens is enriched with essential oils, phenolics, flavonoids, which account for its tremendous biological activities. Laser light treatment and arbuscular mycorrhizal fungi (AMF) inoculation can further enhance the phytochemical content in a significant manner. In this study, we aimed to explore the synergistic impact of these two factors on P. graveolens. For this, we used four groups of surface-sterilized seeds: (1) control group1 (non-irradiated; non-colonized group); (2) control group2 (mycorrhizal colonized group); (3) helium-neon (He-Ne) laser-irradiated group; (4) mycorrhizal colonization coupled with He-Ne laser-irradiation group. Treated seeds were growing in artificial soil inculcated with Rhizophagus irregularis MUCL 41833, in a climate-controlled chamber. After 6 weeks, P. graveolens plants were checked for their phytochemical content and antibacterial potential. Laser light application improved the mycorrhizal colonization in P. graveolens plants which subsequently increased biomass accumulation, minerals uptake, and biological value of P. graveolens. The increase in the biological value was evident by the increase in the essential oils production. The concomitant application of laser light and mycorrhizal colonization also boosted the antimicrobial activity of P. graveolens. These results suggest that AMF co-treatment with laser light could be used as a promising approach to enhance the metabolic content and yield of P. graveolens for industrial and pharmaceutical use. Germination models are quite helpful in predicting emergence times, dormancy periods, and their applications in crop management. This study investigated the germination behaviors of Eruca sativa Mill. in response to fluctuations in temperatures (Ts) and water potentials (ψs). Germination percentage (GP) increased 95% with rising temperature within the range of 20–30 °C, and decreased 25% at 5 °C. Moreover, each ψ and T resulted in a decrease in GP as ψ decreased. Further, we noted that the θT1 value was substantially high at 30 °C and in (0 MPa), whereas the θT2 value was maximum at 10 °C (−0.02 MPa) and it decreased with decreasing Ψ. The maximum hydrothermal time constant (θHTT) and hydrotime (θH) values were obtained at 10 and 30 °C, respectively. In addition, a linear increase in the GR(g) pattern was observed at Tb and a decrease below the To. The calculated cardinal Ts was 5 °C for the base T, and 30 °C for both the optimum and ceiling T. The germination characteristics were higher at 30 °C having (0 MPa). Therefore, using cardinal temperatures, germination results, and the hydrothermal time model (HTT) could reveal the independent and interactive impacts of both T and the Ψ on the response of seed germination subjected to diverse environmental conditions. The study aimed to analyze the agronomic performance of 11 safflower genotypes using adaptability and stability methods, while identifying safflower genotypes with stable behavior and a high grain yield in different environments of the Brazilian Cerrado. Ten lines and a cultivar of safflower were evaluated in four environments in the Brazilian conditions. Our results revealed the genotypes P30, P35, P9, P11, and P31 to be superior for grain yield and P43, P7, P11, and P31 to be superior for oil content. The lowest Wricke index, an indication of genotype stability, was observed for P9 (0.41%), which is considered the most stable genotype, followed by P35 (1.29%) and P31 (1.98%). For the predictability of the behavior of genotypes in the environments, P7 (80.85%), P35 (86.10%), P31 (85.90%), and P9 (97.42%) were considered predictable genotypes. The genotypes P11 (1045.6 kg ha−1 and 19.7%) and P21 (952.7 kg ha−1 and 20.6%) are recommended for cultivation in this region, considering both their grain yield and oil content. Safflower is viable to use out of season in the Brazilian Cerrado. The crop can generate profits for farmers and be used for oil production in periods of uncertain corn production. Jasmonates (JAs) are lipid-derived compounds that function in plants as key signaling compounds during stressful conditions. This study aimed to examine the effects of exogenous fo-liar-JA application (100 μmol L−1) on the morpho-physiological response of two soybean varieties (parachinar-local and swat-84) grown under different NaCl regimes (0, 40, 80, and 120 mM). Results show that exogenous JA application alone and in combination with salt stress altered the growth and metabolism of both soybeans. For instance, they accumulated significant amounts of Na+ and Cl–, while their K+, Mg2+, Fe2+, Mn2+, B3+, and P3+ contents were low. Further, photosynthetic pigments Chl a and Chl b increased at low concentrations of salt and exogenous JA. Car decreased under both salt and exogenous JA as compared with untreated control. In addition, sugar, phenol, and protein content increased under both salt and exogenous JA application. In contrast, the exogenous JA application alleviated the negative impact of salt stress on the growth and metabolism of both soybeans. Further, the high concentrations of soluble protein and phenol in the leaves of both soybeans may contribute to their ability to adapt to salinity. However, molecular studies are necessary to understand the ameliorative role of exogenous JA in the growth and metabolism of salt-treated young seedlings in both soybean varieties. Marrubium vulgare is a valuable source of natural bioactive molecules with high preventive and therapeutic effectiveness. Therefore, this study aimed to study the chemical polymorphism of natural populations of M. vulgare in Tunisia by quantitative chemical markers and the estimation of divergence between populations. Phytochemical analyses of the eight natural populations of Tunisian Marrubium vulgare prospected in different bioclimatic stages, revealed 42 compounds of essential oils representing 96.08% to 100% of the total oil. Hydrocarbon sesquiterpenes were the main fraction of all the populations studied and β-bisabolene was the major compound (from 30.11% to 71.35% of the total oil). The phytochemical investigation of the M. vulgare plant indicated the presence of essential oil with significant percentages of phenolic compounds. A significant quantitative and qualitative variation in the essential oils is detected for both major and minor compounds. The principal components analysis (PCA) performed in the single and combined traits provides a good distinction among populations, not according to their geographical and/or bioclimatic origins. Moreover, the phytochemical analysis of the leaves showed that the Tunisian populations, i.e., the populations of Kasserine, Kef, and Beja, were very rich in phenolic compounds (from 20.8 to 44.65 mg GAE/g DW). Flavonoids compounds were also the main class of total polyphenols present in all the tested populations (from 8.91 to 37.48 mg RE/g DW). The quantitative genetic diversity estimated by the population's structure, based on PCA analysis, was an adaptation to the changes in the environmental conditions. Overall, our study indicated that natural populations of M. vulgare had different chemotypes of essential oils and they were rich in phenolic compounds, particularly flavonoids, which opens a new prospect for industrial use and differential exploitation of this species. Cadmium (Cd) toxicity strongly influences plants growth and seed germination in crop plants. This pot trial had aimed evaluate the benefits of two different kinds of phosphorus (P)-fertilizer in the phytoremediation of Cd by Solanum nigrum L. The current pot experiment was conducted to evaluate the role of P-fertilizers in phytoremediation of Cd by Solanum nigrum L. Single superphosphate (SSP) contain 7 to 9% P and Di-ammonium Phosphate (DAP) contain 46% P had been applied in single and combine form in soil with different ratios (0:0, 100:0, 0:100, 50:50%) accompanied by diverse Cd levels (0, 25, 50 mg kg−1). Three weeks seeding were transferred into pots, and plants had been harvested afterward seventy days of growth in the pots. Significantly inhibited plant growth was observed in shoots and roots of Cd contaminated plants. Cadmium stress had stimulated oxidative stress in subjected plants. However, supplementation of P-fertilizers in an optimum manner significantly increased plant biomass along with enhancing antioxidants enzymatic activities and inhibiting oxidative stress. Maximum plant-growth had been noted in SSP + DAP supplemented plants in contrast to single SSP, DAP supplemented plants. Higher Cd concentrations observed in SSP + DAP supplemented plants over single treatment. It has been concluded that combination of SSP + DAP might be a better option to improve growth as well as uptake capacity of Solanum nigrum L. under Cd stress. Barley (Hordeum vulgare L.) is a salt-tolerant crop with considerable economic value in salinity-affected arid and semiarid areas. In the laboratory experiment, the halothermal time (HaloTT) model was used to examine barley seed germination (SG) at six constant cardinal temperatures (Ts) of 15, 20, 25, 30, 35, and 40 °C under five different water potentials (ψs) of 0, −0.5, −1.5, −1.0, and −2.0 MPa. Results showed that at optimum moisture (0 MPa), the highest germination percentage (GP) was recorded at 20 °C and the lowest at 40 °C. Moreover, GP increased with the accelerated aging period (AAP) and significantly (p ≤ 0.05) decreased with high T. In addition, with a decrease of ψ from 0 to −0.5, −1, 1.5, and −2.0 MPa, GP decreased by 93.33, 76.67, 46.67, and 33.33%, respectively, in comparison with 0 MPa. The maximum halftime constant (θHalo) and coefficient of determination (R2) values were recorded at 20 °C and 30 °C, respectively. The optimum temperature (To) for barley is 20 °C, base Ψ of 50th percentile (Ψb (50)) is −0.23 Mpa, and standard deviation of Ψb (σΨb) is 0.21 MPa. The cardinal Ts for germination is 15 °C (Tb), 20 °C (To), and 40 °C (Tc). The GP, germination rate index (GRI), germination index (GI), coefficient of the velocity of germination (CVG), germination energy (GE), seed vigor index I and II (SVI-I & II), Timson germination index (GI), and root shoot ratio (RSR) were recorded maximum at 0 MPa at 20 °C and minimum at −2.0 MPa at 40 °C. Mean germination time (MGT) and time to 50% germination (T 50%) were maximum at −2 MPa at 40 °C, and minimum at 20 °C, respectively. In conclusion, the HaloTT model accurately predicted the germination time course of barley in response to T, Ψ, or NaCl. Low nitrogen-use efficiency (NUE) is a serious issue for cotton production and environmental sustainability in arid climates. A pot study was conducted to evaluate the effect of K nutrition on NUE and performance of low- and high-K-efficiency cotton cultivars under two moisture regimes. Treatments included two soil moisture levels—i.e., normal irrigation, 100% available water content (AWC); reduced irrigation, 50% AWC—three levels of nitrogen (N)—i.e., 0, 375, and 750 mg N pot−1—and two K levels, i.e., 0 and 208 mg K pot−1. Results reveal that 208 mg K pot−1 application with nitrogen significantly enhanced the N-use efficiency, growth, and yield attributes of both cotton cultivars compared with sole N fertilization. Similarly, the combined application of NK @ 375 N + 208 K mg pot−1 caused up to 83% increase in NUE under AWC50% and AWC100%, as compared with NK control (0 N + 0 K). Compared with the control, imposed low-moisture stress caused a decrease of 13.9% in stomatal conductance (gs), 2.5% in transpiration rate (E), and 6.5% in net photosynthetic rate (PN), respectively. The physiological water use efficiency (PN/E) decreased by 13.2% under AWC50%. Applied NK @ 375 N:208 K, mg pot−1 caused 27.39 and 27.56% improvement in the PN/E in HKE and LKE cultivars under AWC50%, respectively. The HKE cultivar, i.e., CIM-554, maintained the highest gs and PN than FH-901, that was low-K-efficiency cultivar. The study suggests that varietal selection and adequate K fertilization have the prospects to improve NUE and save considerable quantities of fertilizer and irrigation water in cotton production under arid environments. The use of natural products as therapeutic agents is rapidly growing recently. In the current study, we investigated the protective effects of green tea supplementation on lead-induced toxicity in mice. Forty albino mice were divided into four groups as follows: A: control group; B: green tea receiving group; C: lead-intoxicated group; and D: lead-intoxicated group supplemented with green tea. At the end of the experiment, the animals were tested for neurobehavioral and biochemical alterations. Green tea was analyzed through Gas Chromatography–Mass Spectrometry (GC/MS) analysis. We found that supplementation with green tea ameliorated the lead-associated increase in body weight and blood glucose. Green tea supplementation also changed the blood picture that was affected due to lead toxicity and ameliorated lead-induced dyslipidemia. The group of mice that were supplemented with green tea has shown positive alterations in locomotory, anxiety, memory, and learning behaviors. The GC/MS analysis revealed many active ingredients among which the two most abundant were caffeine and 1,2-benzenedicarboxylic acid, mono(2-ethylhexyl) ester. We concluded that green tea supplementation has several positive effects on the lead-induced neurotoxicity in mice and that these effects may be attributed to its main two active ingredients. The present study was designed to evaluate polarity-dependent extraction efficiency and pharmacological profiling of Polygonum glabrum Willd. Crude extracts of leaves, roots, stems, and seeds, prepared from solvents of varying polarities, were subjected to phytochemical, antioxidant, antibacterial, antifungal, antidiabetic, and cytotoxicity assays. Maximum extraction yield (20.0% w/w) was observed in the case of an acetone:methanol (AC:M) root extract. Distilled water:methanol (W:M) leaves extract showed maximum phenolic contents. Maximum flavonoid content and free radical scavenging potential were found in methanolic (M) seed extract. HPLC-DAD quantification displayed the manifestation of substantial quantities of quercetin, rutin, gallic acid, quercetin, catechin, and kaempferol in various extracts. The highest ascorbic acid equivalent total antioxidant capacity and reducing power potential was found in distilled water roots and W:M leaf extracts, respectively. Chloroform (C) seeds extract produced a maximum zone of inhibition against Salmonella typhimurium. Promising protein kinase inhibition and antifungal activity against Mucor sp. were demonstrated by C leaf extract. AC:M leaves extract exhibited significant cytotoxic capability against brine shrimp larvae and α-amylase inhibition. Present results suggest that the nature of pharmacological responses depends upon the polarity of extraction solvents and parts of the plant used. The current study was intended to explore the phytochemical profiling and therapeutic activities of Putranjiva roxburghii Wall. Crude extracts of different plant parts were subjected to the determination of antioxidant, antimicrobial, antidiabetic, cytotoxic, and protein kinase inhibitory potential by using solvents of varying polarity ranges. Maximum phenolic content was notified in distilled water extracts of the stem (DW-S) and leaf (DW-L) while the highest flavonoid content was obtained in ethyl acetate leaf (EA-L) extract. HPLC-DAD analysis confirmed the presence of various polyphenols, quantified in the range of 0.02 ± 0.36 to 2.05 ± 0.18 μg/mg extract. Maximum DPPH scavenging activity was expressed by methanolic extract of the stem (MeOH-S). The highest antioxidant capacity and reducing power was shown by MeOH-S and leaf methanolic extract (MeOH-L), respectively. Proficient antibacterial activity was shown by EA-L extract against Bacillus subtilis and Escherichia coli. Remarkable α-amylase and α-glucosidase inhibition potential was expressed by ethyl acetate fruit (EA-F) and n-Hexane leaf (nH-L) extracts, respectively. In case of brine shrimp lethality assay, 41.67% of the extracts (LC50 < 50 µg/mL) were considered as extremely cytotoxic. The test extracts also showed mild antifungal and protein kinase inhibition activities. Edible sprouts are rich in flavonoids and other polyphenols, as well as proteins, minerals, and vitamins. Increasing sprout consumption necessitates improving their quality, palatability, and bioactivity. The purpose of this study was to test how KNO3 priming affects the sprouting process species on three Medicago species (Medicago indicus, Medicago interexta, and Medicago polymorpha) and their nutritional values. Targeted species of Medicago were primed with KNO3, and the levels of different primary and secondary metabolites were determined. KNO3 induced biomass accumulation in the sprouts of the three species, accompanied by an increased content of total mineral nutrients, pigments, vitamins, and essential amino acids. Besides, our results showed that KNO3 enhanced the activity of nitrate reductase (NR), glutamate dehydrogenase (GDH), and glutamine synthetase (GS) enzymes, which are involved in the nitrogen metabolism and GOGAT cycle, which, in turn, increase the nitrogen and protein production. KNO3 treatment improved the bioactive compound activities of Medicago sprouts by increasing total phenolic and flavonoid contents and enhancing the antioxidant and antidiabetic activities. Furthermore, species-specific responses toward KNO3 priming were noticeable, where Medicago interexta showed the highest antioxidant and antidiabetic activities, followed by Medicago polymorpha. Withaferin A (WA) is a pivotal withanolide that has conquered a conspicuous place in research, owning to its multidimensional biological properties. It is an abundant constituent in Withania somnifera Dunal. (Ashwagandha, WS) that is one of the prehistoric pivotal remedies in Ayurveda. This article reviews the literature about the pharmacological profile of WA with special emphasis on its anticancer aspect. We reviewed research publications concerning WA through four databases and provided a descriptive analysis of literature without statistical or qualitative analysis. WA has been found as an effective remedy with multifaceted mechanisms and a broad spectrum of pharmacological profiles. It has anticancer, anti-inflammatory, antiherpetic, antifibrotic, antiplatelet, profibrinolytic, immunosuppressive, antipigmentation, antileishmanial, and healing potentials. Evidence for wide pharmacological actions of WA has been established by both in vivo and in vitro studies. Further, the scientific literature accentuates the role of WA harboring a variable therapeutic spectrum for integrative cancer chemoprevention and cure. Solubility of phytoconstituents depends on the polarity of the extraction medium used, which might result in the different pharmacological responses of extracts. In line with this, ethnomedicinally important food plant (i.e., Caralluma tuberculata extracts) have been made in fourteen distinct solvent systems that were then analyzed phytochemically via total phenolic amount estimation, total flavonoid amount estimation, and HPLC detection and quantification of the selected polyphenols. Test extracts were then subjected to a battery of in vitro assays i.e., antioxidants (DDPH scavenging, antioxidant capacity, and reducing power estimation), antimicrobial (antibacterial, antifungal, and antileishmanial), cytotoxic (brine shrimps, THP-1 human leukemia cell lines and normal lymphocytes), and protein kinase inhibition assays. Maximum phenolic and flavonoid contents were computed in distilled water–acetone and acetone extracts (i.e., 16 ± 1 μg/mg extract and 8 ± 0.4/mg extract, respectively). HPLC-DAD quantified rutin (0.58 µg/mg extract) and gallic acid (0.4 µg/mg extract) in methanol–ethyl acetate and methanol extracts, respectively. Water–acetone extract exhibited the highest DPPH scavenging of 36 ± 1%. Total reducing potential of 76.0 ± 1 μg/mg extract was shown by ethanol chloroform while maximum total antioxidant capacity was depicted by the acetone extract (92.21 ± 0.70 μg/mg extract). Maximal antifungal effect against Mucor sp., antileishmanial, brine shrimp cytotoxicity, THP-1 cell line cytotoxicity, and protein kinase inhibitory activities were shown by ethyl acetate-methanol (MIC: 50 µg/disc), n-hexane (IC50: 120.8 ± 3.7 µg/mL), ethyl acetate (LD50: 29.94 ± 1.6 µg/mL), distilled water–acetone (IC50: 118 ± 3.4 µg/mL) and methanol–chloroform (ZOI: 19 ± 1 mm) extracts, respectively. Our findings show the dependency of phytochemicals and bioactivities on the polarity of the extraction solvent and our preliminary screening suggests the C. tuberculata extract formulations to be tested and used in different ailments, however, detailed studies remain necessary for corroboration with our results. A worldwide increase in the incidence of fungal infections, emergence of new fungal strains, and antifungal resistance to commercially available antibiotics indicate the need to investigate new treatment options for fungal diseases. Therefore, the interest in exploring the antifungal activity of medicinal plants has now been increased to discover phyto-therapeutics in replacement to conventional antifungal drugs. The study was conducted to explore and identify the mechanism of action of antifungal agents of edible plants, including Cinnamomum zeylanicum, Cinnamomum tamala, Amomum subulatum, Trigonella foenumgraecum, Mentha piperita, Coriandrum sativum, Lactuca sativa, and Brassica oleraceae var. italica. The antifungal potential was assessed via the disc diffusion method and, subsequently, the extracts were assessed for phytochemicals and total antioxidant activity. Potent polyphenols were detected using high-performance liquid chromatography (HPLC) and antifungal mechanism of action was evaluated in silico. Cinnamomum zeylanicum exhibited antifungal activity against all the tested strains while all plant extracts showed antifungal activity against Fusarium solani. Rutin, kaempferol, and quercetin were identified as common polyphenols. In silico studies showed that rutin displayed the greatest affinity with binding pocket of fungal 14-alpha demethylase and nucleoside diphosphokinase with the binding affinity (Kd, −9.4 and −8.9, respectively), as compared to terbinafine. Results indicated that Cinnamomum zeylanicum and Cinnamomum tamala exert their antifungal effect possibly due to kaempferol and rutin, respectively, or possibly by inhibition of nucleoside diphosphokinase (NDK) and 14-alpha demethylase (CYP51), while Amomum subulatum and Trigonella foenum graecum might exhibit antifungal potential due to quercetin. Compared to seeds and mature tissues, sprouts are well known for their higher nutritive and biological values. Fruits of Pimpinella anisum (anise) are extensively consumed as food additives; however, the sprouting-induced changes in their nutritious metabolites are hardly studied. Herein, we investigated the bioactive metabolites, phytochemicals, and antioxidant properties of fruits, sprouts (9-day-old), and mature tissue (5-week-old) of anise under laser irradiation treatment (He-Ne laser, 632 nm). Laser treatment increased biomass accumulation of both anise sprouts and mature plants. Bioactive primary (e.g., proteins and sugars) and secondary metabolites (e.g., phenolic compounds), as well as mineral levels, were significantly enhanced by sprouting and/or laser light treatment. Meanwhile, laser light has improved the levels of essential oils and their related precursors (e.g., phenylalanine), as well as enzyme activities [e.g., O–methyltransferase and 3-Deoxy-D-arabino-heptulosonate-7-phosphate synthase (DAHPS)] in mature tissues. Moreover, laser light induced higher levels of antioxidant and anti-lipidemic activities in sprouts as compared to fruits and mature tissues. Exploration of leads with therapeutic potential in inflammatory disorders is worth pursuing. In line with this, the isolated natural compound daturaolone from Datura innoxia Mill. was evaluated for its anti-inflammatory potential using in silico, in vitro and in vivo models. Daturaolone follows Lipinski's drug-likeliness rule with a score of 0.33. Absorption, distribution, metabolism, excretion and toxicity prediction show strong plasma protein binding; gastrointestinal absorption (Caco-2 cells permeability = 34.6 nm/s); no blood–brain barrier penetration; CYP1A2, CYP2C19 and CYP3A4 metabolism; a major metabolic reaction, being aliphatic hydroxylation; no hERG inhibition; and non-carcinogenicity. Predicted molecular targets were mainly inflammatory mediators. Molecular docking depicted H-bonding interaction with nuclear factor kappa beta subunit (NF-κB), cyclooxygenase-2, 5-lipoxygenase, phospholipase A2, serotonin transporter, dopamine receptor D1 and 5-hydroxy tryptamine. Its cytotoxicity (IC50) value in normal lymphocytes was >20 µg/mL as compared to cancer cells (Huh7.5; 17.32 ± 1.43 µg/mL). Daturaolone significantly inhibited NF-κB and nitric oxide production with IC50 values of 1.2 ± 0.8 and 4.51 ± 0.92 µg/mL, respectively. It significantly reduced inflammatory paw edema (81.73 ± 3.16%), heat-induced pain (89.47 ± 9.01% antinociception) and stress-induced depression (68 ± 9.22 s immobility time in tail suspension test). Caraway plants have been known as a rich source of phytochemicals, such as flavonoids, monoterpenoid glucosides and alkaloids. In this regard, the application of elevated CO2 (eCO2) as a bio-enhancer for increasing plant growth and phytochemical content has been the focus of many studies; however, the interaction between eCO2 and plants at different developmental stages has not been extensively explored. Thus, the present study aimed at investigating the changes in growth, photosynthesis and phytochemicals of caraway plants at two developmental stages (sprouts and mature tissues) under control and increased CO2 conditions (ambient CO2 (a CO2, 400 ± 27 μmol CO2 mol−1 air) and eCO2, 620 ± 42 μmol CO2 mol−1 air ppm). Moreover, we evaluated the impact of eCO2-induced changes in plant metabolites on the antioxidant and antibacterial activities of caraway sprouts and mature plants. CO2 enrichment increased photosynthesis and biomass accumulation of both caraway stages. Regarding their phytochemical contents, caraway plants interacted differently with eCO2, depending on their developmental stages. High levels of CO2 enhanced the production of total nutrients, i.e., carbohydrates, proteins, fats and crude fibers, as well as organic and amino acids, in an equal pattern in both caraway sprouts and mature plants. Interestingly, the eCO2-induced effect on minerals, vitamins and phenolics was more pronounced in caraway sprouts than the mature tissues. Furthermore, the antioxidant and antibacterial activities of caraway plants were enhanced under eCO2 treatment, particularly at the mature stage. Overall, eCO2 provoked changes in the phytochemical contents of caraway plants, particularly at the sprouting stage and, hence, improved their nutritive and health-promoting properties. Molybdenum ions (Mo) can improve plants' nutritional value primarily by enhancing nitrogenous metabolism. In this study, the comparative effects of seed priming using Mo were evaluated among sproutings of Canavalia species/cultivars, including Canavalia ensiformis var. gladiata (CA1), Canavalia ensiformis var. truncata Ricker (CA2), and Canavalia gladiata var. alba Hisauc (CA3). Mo impacts on growth, metabolism (e.g., nitrogen and phenolic metabolism, pigment and total nutrient profiles), and biological activities were assayed. Principal component analysis (PCA) was used to correlate Mo-mediated impacts. The results showed that Mo induced photosynthetic pigments that resulted in an improvement in growth and increased biomass. The N content was increased 0.3-fold in CA3 and 0.2-fold in CA1 and CA2. Enhanced nitrogen metabolism by Mo provided the precursors for amino acids, protein, and lipid biosynthesis. At the secondary metabolic level, phenolic metabolism-related precursors and enzyme activities were also differentially increased in Canavalia species/cultivars. The observed increase in metabolism resulted in the enhancement of the antioxidant (2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) free radical scavenging, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), ferric reducing antioxidant power (FRAP)) and antidiabetic potential (Glycemic index (GI) and inhibition activity of α-amylase, and α-glucosidase) of species. The antioxidant activity increased 20% in CA3, 14% in CA1, and 8% in CA2. Furthermore, PCA showed significant variations not only between Mo-treated and untreated samples but also among Canavalia species. Overall, this study indicated that the sprouts of Canavalia species have tremendous potential for commercial usage due to their high nutritive value, which can be enhanced further with Mo treatment to accomplish the demand for nutritious feed. Herbal and traditional medicines can play a pivotal role in combating cancer and neglected tropical diseases. Ajuga bracteosa, family Lamiaceae, is an important medicinal plant. The genetic transformation of A. bracteosa with rol genes of Agrobacterium rhizogenes further enhances its metabolic content. This study aimed at undertaking the molecular, phytochemical, and in vitro biological analysis of A. bracteosa extracts. We transformed the A. bracteosa plant with rol genes and raised the regenerants from the hairy roots. Transgenic integration and expression of rolB were confirmed by conventional polymerase chain reaction (PCR) and qPCR analysis. The methanol: chloroform crude extracts of wild-type plants and transgenic regenerants were screened for in vitro antibacterial, antihemolytic, cytotoxic, anticancer, and leishmanial activity. Among all plants, transgenic line 3 (ABRL3) showed the highest expression of the rolB gene. Fourier transform infra-red (FTIR) analysis confirmed the enhanced number of functional groups of active compounds in all transgenic lines. Moreover, ABRL3 exhibited the highest antibacterial activity, minimum hemolytic activity (CC50 = 7293.05 ± 7 μg/mL) and maximum antileishmanial activity (IC50 of 56.16 ± 2 μg/mL). ABRL1 demonstrated the most prominent brine shrimp cytotoxicity (LD5039.6 ± 4 μg/mL). ABRL3 was most effective against various human cancer cell lines with an IC50 of 57.1 ± 2.2 μg/mL, 46.2 ± 1.1 μg/mL, 72.4 ± 1.3 μg/mL, 73.3 ± 2.1 μg/mL, 98.7 ± 1.6 μg/mL, and 97.1 ± 2.5 μg/mL against HepG2, LM3, A549, HT29, MCF-7, and MDA-MB-231, respectively. Ajuga bracteosa Wall. ex Benth. is an endangered medicinal herb traditionally used against different ailments. The present study aimed to create new insight into the fundamental mechanisms of genetic transformation and the biological activities of this plant. We transformed the A. bracteosa plant with rol genes of Agrobacterium rhizogenes and raised the regenerants from the hairy roots. These transgenic regenerants were screened for in vitro antioxidant activities, a range of in vivo assays, elemental analysis, polyphenol content, and different phytochemicals found through HPLC. Among 18 polyphenolic standards, kaempferol was most abundant in all transgenic lines. Furthermore, transgenic line 3 (ABRL3) showed maximum phenolics and flavonoids content among all tested plant extracts. ABRL3 also demonstrated the highest total antioxidant capacity (8.16 ± 1 μg AAE/mg), total reducing power, (6.60 ± 1.17 μg AAE/mg), DPPH activity (IC50 = 59.5 ± 0.8 μg/mL), hydroxyl ion scavenging (IC50 = 122.5 ± 0.90 μg/mL), and iron-chelating power (IC50 = 154.8 ± 2 μg/mL). Moreover, transformed plant extracts produced significant analgesic, anti-inflammatory, anticoagulant, and antidepressant activities in BALB/c mice models. Elevated atmospheric CO2 (eCO2) can significantly enhance plant metabolism and improve their nutritional quality. Although several studies addressed the effect of eCO2 on plants, to our knowledge, there are no studies that have applied eCO2 to improve growth, chemical composition, and biological properties of ajwain (Trachyspermum ammi) during sprouting. Herein, eCO2 (620 µmol mol−1) was employed to enhance photosynthetic reactions. Improved photosynthesis induced primary and bioactive secondary metabolism, which led to improved biological activities of T. ammi sprouts in comparison with control sprouts and seeds. eCO2-treated sprouts showed significantly (p < 0.05) higher levels of most measured nutritional primary and secondary metabolites like soluble sugars, essential amino acids, organic acids, essential oils, phenolics, and flavonoids than control sprouts, which had significantly higher values than control seeds. eCO2 significantly improved the antimicrobial properties of T. ammi sprouts against 11 out of 13 microbial species than control sprouts, which had more potent antimicrobial activities than seeds. The significant increase in the antioxidant potential of treated sprouts was indicated by improved levels of ferric reducing antioxidant power (FRAP), DPPH, and oxygen radical absorbance capacity (ORAC). In addition, the anticancer activity against four different human tumor cell lines was significantly (p < 0.05) boosted by both sprouting and eCO2 exposure. Thus, the present study confirms the synergistic effect of sprouting with eCO2 exposure as promising approaches to produce ajwain sprouts with rich nutraceuticals, effective phytochemicals, and enhanced biological activities. Compared to seeds and mature plants, sprouts are well characterized based on their nutritive values and biological properties. Moreover, laser light application is known to be a promising approach to improving plant growth, photosynthesis, and nutraceutical values. However, no studies have investigated the phytochemicals and biological activity of lemongrass (Cymbopogon proximus (Hochst. ex A.Rich.) Chiov.) sprouts or the further improvement of their quality by applying laser light treatment. We carried out a preliminary experiment for the optimization of laser treatment conditions, finding that a helium neon (He–Ne) laser at 632 nm and 5 mW for 5 min provided the most favorable conditions. We then investigated fresh weight, photosynthetic reactions, and primary and secondary metabolites, including sugars, amino acids, organic acids, essential oils, and phenolic compounds. Moreover, we studied the effect of laser light-induced changes in chemical compositions on the antioxidant, anti-diabetic, and anti-cholesterol activities of Cymbopogon proximus sprouts grown from laser-treated seeds. Laser light treatment increased the photosynthesis and respiration and hence the fresh weight of Cymbopogon proximus sprouts. Overall, sprouting increased most bioactive primary and secondary metabolites as compared to seeds. Increased photosynthesis by laser light improved carbon allocation and raised non-structural carbohydrates, which in turn led to improved synthesis of amino acids, organic acids, and essential oils, as well as phenolic and flavonoid compounds. As a result, laser light significantly improved the antioxidant capacity in terms of increasing the levels of ferric reducing antioxidant power (FRAP) (from 9.5 to 21 µmole trolox/g fresh weight (FW)), oxygen radical absorbance (ORAC) (from 400 to 1100 µmole trolox/100 g FW), and DPPH (from 5% to 25% of inhibation) and enhanced the hypocholesterolemic and antidiabetic activity through increasing the percentage of cholesterol micellar solubility (CMS) inhibition (from 42% to 62%) and glycemic index (from 33 to 17 µmole/g) over sprouts and seeds. In conclusion, the synergism of seed laser treatment and sprouting induced the health-promoting bioactive compounds in Cymbopogon proximus as compared to seeds, which can be applied at a large scale to improve the biochemical, physiological, and nutraceutical values of medicinal and crop sprouts. Increased problems associated with side effects and bacterial resistance of chemical drugs has prompted the research focus on herbal medicines in the past few decades. In the present investigation, the antimicrobial activity of the various parts of Avicennia marina (AM), a mangrove plant, has been evaluated. The plants were collected from the Jazan area of the Kingdom of Saudi Arabia. Primary extracts of roots, stem, leaves, fruits, and seeds were made in ethanol and fractioned in ethanol, ethyl acetate, petroleum ether, chloroform, and water. Minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of the extracts were determined against Bacillussubtilis, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. It has been observed that the chloroform extract of roots of the AM exhibited inhibitory effects against both S. aureus (MIC = 1.5 ± 0.03 mg/mL) and E. coli (MIC = 1.7 ± 0.01 mg/mL). The ethanolic extract of the AM roots has shown antibacterial activity against Pseudomonas aeruginosa (MIC = 10.8 ± 0.78 mg/mL), Bacillussubtilis (MIC = 6.1 ± 0.27 mg/mL), Staphylococcus aureus (MIC = 2.3 ± 0.08 mg/mL), and Escherichia coli (MIC = 6.3 ± 0.28 mg/mL). The leaf extract of the AM in ethyl acetate showed antibacterial activity against S. aureus and E. coli. Antifungal activity of these extracts was also investigated against Aspergillus fumigatus and Candida albicans. Ethanolic extract of roots and seeds of the AM has shown antifungal activity against Aspergillus fumigatus when applied individually. Ethanolic extract of the AM fruits has shown an inhibitory effect on the growth of Aspergillus fumigatus and Candida albicans. It is suggested that the plant extracts of AM have tremendous antimicrobial activity against a group of microbes, and this effect depends on both the plant part and the solvent used for extraction. Sulfur (S) is an essential element for all forms of life. It is involved in numerous essential processes because S is considered as the primary source of one of the essential amino acids, methionine, which plays an important role in biological events. For the control and regulation of sulfate in a metabolic network through fluxomics, a non-invasive tool is highly desirable that opens the door to monitor the level of the sulfate in real time and space in living cells without fractionation of the cells or tissue. Here, we engineered a FRET (fluorescence resonance energy transfer) based sensor for sulfate, which is genetically-encoded and named as FLIP-SP (Fluorescent indicator protein for sulfate). The FLIP-SP can measure the level of the sulfate in live cells. This sensor was constructed by the fusion of fluorescent proteins at the N- and C-terminus of sulfate binding protein (sbp). The FLIP-SP is highly specific to sulfate, and showed pH stability. Real-time monitoring of the level of sulfate in prokaryotic and eukaryotic cells showed sensor bio-compatibility with living cells. (+)-Catechin is an important antioxidant of green tea (Camelia sinensis (L.) O. Kuntze). Catechin is known for its positive role in anticancerous activity, extracellular matrix degradation, cell death regulation, diabetes, and other related disorders. As a result of enormous interest in and great demand for catechin, its biosynthesis using metabolic engineering has become the subject of concentrated research with the aim of enhancing (+)-catechin production. Metabolic flux is an essential concept in the practice of metabolic engineering as it helps in the identification of the regulatory element of a biosynthetic pathway. In the present study, an attempt was made to analyze the metabolic flux of the (+)-catechin biosynthesis pathway in order to decipher the regulatory element of this pathway. Firstly, a genetically encoded fluorescence resonance energy transfer (FRET)-based nanosensor (FLIP-Cat, fluorescence indicator protein for (+)-catechin) was developed for real-time monitoring of (+)-catechin flux. In vitro characterization of the purified protein of the nanosensor showed that the nanosensor was pH stable and (+)-catechin specific. Its calculated Kd was 139 µM. The nanosensor also performed real-time monitoring of (+)-catechin in bacterial cells. In the second step of this study, an entire (+)-catechin biosynthesis pathway was constructed and expressed in E. coli in two sets of plasmid constructs: pET26b-PT7-rbs-PAL-PT7-rbs-4CL-PT7-rbs-CHS-PT7-rbs-CHI and pET26b-T7-rbs-F3H-PT7-rbs- DFR-PT7-rbs-LCR. The E. coli harboring the FLIP-Cat was transformed with these plasmid constructs. The metabolic flux analysis of (+)-catechin was carried out using the FLIP-Cat. The FLIP-Cat successfully monitored the flux of catechin after adding tyrosine, 4-coumaric acid, 4-coumaroyl CoA, naringenin chalcone, naringenin, dihydroquercetin, and leucocyanidin, individually, with the bacterial cells expressing the nanosensor as well as the genes of the (+)-catechin biosynthesis pathway. Dihydroflavonol reductase (DFR) was identified as the main regulatory element of the (+)-catechin biosynthesis pathway. Information about this regulatory element of the (+)-catechin biosynthesis pathway can be used for manipulating the (+)-catechin biosynthesis pathway using a metabolic engineering approach to enhance production of (+)-catechin. An efficient protocol of plant regeneration through indirect organogenesis in Viola serpens was developed in the present study. Culture of leaf explants on MS (Murashige and Skoog) medium supplemented with 2.0 mg/L 6-benzyladenine and 0.13 mg/L 2,4-dichloro phenoxy acetic acid. Adventitious shoot formation was observed when calli were transferred on to MS medium containing 0.5 mg/L α-naphthalene acetic acid and 2.25 mg/L kinetin, which showed the maximum 86% shoot regeneration frequency. The highest root frequency (80.92%) with the 5.6 roots per explant and 1.87 cm root length was observed on MS medium supplemented with 2 mg/L indole-3-butyric acid. The plantlets were transferred to the mixture of sand, coffee husk and soil in the ratio of 1:2:1 in a pot, and placed under 80% shade net for one month. It was then transferred to 30% shade net for another one month, prior to transplantation in the field. These plantlets successfully acclimatized under field conditions. A Sequence Characterized Amplified Region (SCAR) marker was also developed using a 1135 bp amplicon that was obtained from RAPD (Random Amplification of Polymorphic DNA) analysis of six accessions of V. serpens. Testing of several market samples of V. serpens using the SCAR marker revealed successful identification of the genuine samples of V. serpens. This study, therefore, provides a proficient in vitro propagation protocol of V. serpens using leaf explants and a SCAR marker for the authentic identification of V. serpens. Water scarcity has greatly increased the need for research into alternative irrigation methods. Irrigation with treated wastewater (TW) is considered an important alternative in terms of reducing our dependence upon groundwater and freshwater. In this study, we examined the effects of irrigation with TW on the nutrient contents of two alfalfa cultivars and compared them with the nutrient contents of plants irrigated with well water (WW). The two cultivars (Alhassawy—a local cultivar and CUV101) were cultivated from 2013 to 2015 and sampled twice. For both cultivars and in both sampling periods, irrigation with TW significantly affected the macronutrient and micronutrient contents of alfalfa tissues. Plants irrigated with TW had higher K and Ca contents, but lower N, P, and Mg contents. No significant variation in S content was noted between plants irrigated with WW and TW. Furthermore, cultivar and cutting time had significant effects on nutrient contents, and these variables interacted with the effect of the water type used for irrigation. However, before TW can be regularly used in irrigation processes, further long-term studies are needed that consider the variations in water treatment efficiency and differences between cultivated sites in addition to the crop being irrigated with TW. In the present work, essential oils (EOs) extracted from different parts of sour orange Citrus aurantium (green leaves/twigs, small branches, wooden branches, and branch bark) were studied through gas chromatography coupled with mass spectrometry (GC/MS). Furthermore, the EOs in the amounts of 5, 10, 15, 20, and 25 µL were studied for their antibacterial activity against three pathogenic bacteria, Agrobacterium tumefaciens, Dickeya solani, and Erwinia amylovora. The main EO compounds in the leaves/twigs were 4-terpineol (22.59%), D-limonene (16.67%), 4-carvomenthenol (12.84%), and linalool (7.82%). In small green branches, they were D-limonene (71.57%), dodecane (4.80%), oleic acid (2.72%), and trans-palmitoleic acid (2.62%), while in branch bark were D-limonene (54.61%), γ-terpinene (6.68%), dodecane (5.73%), and dimethyl anthranilate (3.13%), and in branch wood were D-limonene (38.13%), dimethyl anthranilate (8.13%), (-)-β-fenchol (6.83%), and dodecane (5.31%). At 25 µL, the EO from branches showed the highest activity against A. tumefaciens (IZ value of 17.66 mm), and leaves/twigs EO against D. solani and E. amylovora had an IZ value of 17.33 mm. It could be concluded for the first time that the wood and branch bark of C. aurantium are a source of phytochemicals, with D-limonene being the predominant compound in the EO, with potential antibacterial activities. The compounds identified in all the studied parts might be appropriate for many applications, such as antimicrobial agents, cosmetics, and pharmaceuticals. [ABSTRACT FROM AUTHOR]
