A peat substrate is made from peat from drained peatlands, which is a limited resource. A realistic estimate is that 50% of the world's wetlands have been lost. Peat is used in horticulture, especially for the cultivation of vegetables in greenhouses. The consequences of peatland exploitation are an increase in the greenhouse effect and a decrease in carbon stocks. Wood fiber can be used as an alternative to peat. The chemical properties of growing media interact and change continuously due to the small volume of growing media, which is limited by the growing container. This study aims to gain new knowledge on the impact of nutrient changes in the microbial degradation of carbon compounds in wood fiber and mixtures with a peat substrate on the content and uptake of nutrients required by plants. The cucumber (Cucumis sativus L.) variety 'Dirigent H' developed in the Netherlands was cultivated in growing media of a peat substrate and wood fiber: (1) peat substrate (PS); (2) wood fiber (WF); (3) wood fiber and peat substrate 50/50 v/v (WF/PS 50/50); (4) wood fiber and peat substrate 25/75 v/v (WF/PS 25/75). The rates of fertilization were the following: (1) conventional fertilization (CF); (2) 13 g N per plant (N13); (3) 23 g N per plant (N23); (4) 30 g N per plant (N30). The experiment was carried out with three replications. As the amount of wood fiber increased, the humidity and pH of the growing media increased. The fertilization of the cucumbers with different quantities of nitrogen influenced the nutrient uptake. The plants grown in the 50/50 and 25/75 growing media had the best Cu uptake when fertilized with N23. When the plants grown in the wood fiber media and the 50/50 media were fertilized with N13, N23, and N30, the Mn content in the growing media at the end of the growing season was significantly lower than the Mn content in the media with conventional fertilization. Thus, nitrogen improved the uptake of Mn by the plants grown not only in the wood fiber, but also in the combinations with a peat substrate. Growing plants in wood fiber and fertilizing them with N13 can result in the optimum uptake of micronutrients. The number and biomass of cucumber fruits per plant were influenced by the amount of wood fiber in the growing media and the application of nitrogen fertilizer. The highest number of fruits and biomass of fruits per plant obtained were significantly higher when the cucumbers were grown in WF/PS 50/50 growing media with additional N13 fertilization. Information on the long-term effects of the addition of organics and fertilizers to wheat under the pearl millet–wheat cropping system with semi-arid conditions in north-western India is still lacking. The present research was conducted in an ongoing field experiment initiated during Rabi 1995 at the Research Farm of Chaudhary Charan Singh at Haryana Agricultural University, Hisar. After 25 years, the impacts of nutrient management practices on soil fertility and wheat productivity were evaluated. The experiment comprised a total of eight treatment combinations viz. half and full doses of recommended fertilizers (N and P), organic manures (FYM: farmyard manure, POM: poultry manure, and PRM: press mud) alone and in combination with NP fertilizers. The conjoint application of organic manure and chemical fertilizers resulted in a positive influx of nutrients via increasing total organic carbon (TOC), available N, P, K, and S, which ranged from 0.46 to 1.42%, 122.70 to 194.70, 15.66 to 74.92, 340.5 to 761.2, and 15.26 to 54.63 kg ha−1 in surface soil (0–15 cm), respectively. Carbon fractions and crop yield were significantly improved by adopting integrated nutrient management (INM). The TOC showed a positive and significant correlation with C fractions (r > 0.92) and with soil-available N, P, K, and S (r > 0.77) content. The data also revealed a strong relationship between TOC and soil-available (0–15 cm) nutrients i.e., available N (R2= 0.769), available P (R2 = 0.881), available K (R2 = 0.758), and available S (R2 = 0.914), respectively. Thus, practices that increased TOC were also beneficial in enhancing the availability of the nutrients in the soil. A positive and highly significant correlation was also found among wheat yield, nutrient (NPKS) content, and uptake. A polynomial relationship between grain yield and grain N (R2 = 0.962), P (R2 = 0.946), and K (R2 = 0.967) content, and between straw yield and straw N (R2 = 0.830), P (R2 = 0.541) and K (R2 = 0.976) content was obtained. Integrated use of PRM7.5 followed by FYM15 and POM5 coupled with NP fertilizers proved best, which could be beneficial for obtaining nutritious and highest wheat yield (grain: 6.01 t ha−1 and straw: 7.70 t ha−1) coupled with improved fertility within a sustained manner under the pearl millet–wheat sequence in prevailing semi-arid conditions of the North Indian state of Haryana. Lithium (Li) is the lightest metal in existence. Its effects on higher plants are still under discussion because both positive and toxic results have been reported in different species. In the last decade, the use of Li has increased considerably, and it is projected that Li waste will be an environmental problem in the near future, such that various organisms, including plants, may be altered by its presence. Interestingly, Li can trigger hormesis, with beneficial effects at low doses and inhibitory or harmful effects at high doses. Currently, numerous research groups are focusing their studies on agriculture to obtain crops fortified with Li, which represents a nutritional advantage in food if adequate concentrations are used. However, more studies are still needed in order to understand the biochemical mechanisms of the effects of Li on plants. This review describes the natural and anthropogenic sources of Li, as well as the concentrations of this element in different environments. Regarding the uses of Li in different areas, topics related to doses that cause toxicity and lethality in humans are addressed. Given its impact on crop production, mechanisms of Li uptake and transport in higher plants are reviewed, as well as the effects on plant metabolism and physiology. This study investigated the effect of light intensity on the growth and nutrient uptake of Chlorella sorokiniana cultivated in nitrogen-rich anaerobic digestion wastewater. Three light intensities (20, 68, and 162 µmol m⁻2 s⁻1) were applied over a 30-day period with a 16:8 h light–dark photoperiod. The goal was to understand how light affects biomass productivity, nutrient assimilation, and biochemical composition under varying nitrogen concentrations originating from biogas plant digestate, up to 5 g L⁻1. The results showed that higher light intensities significantly boosted biomass production, achieving a five-fold increase at 162 µmol m⁻2 s⁻1 compared to 20 µmol m⁻2 s⁻1. Nutrient uptake followed a similar pattern, with 94% of ammonium nitrogen removed in 7 days under high light, compared to 55% after 30 days under low light. Phosphorus content was also completely removed after 7 days under light intensities of 68 and 162 µmol m⁻2 s⁻1. Additionally, elevated light intensity led to increased lipid accumulation (from 29.7% to 34%) and reduced protein content (from 30.9% to 26.1%), with carbohydrate content not being affected by light intensity. These findings highlight light intensity as a critical factor for optimizing microalgae cultivation in nitrogen-rich biogas digestate, promoting both effective nutrient removal and biomass production for potential bioenergy applications. Beneficial microbes are crucial for improving crop adaptation and growth under various stresses. They enhance nutrient uptake, improve plant immune responses, and help plants tolerate stresses like drought, salinity, and heat. The yield potential of any crop is significantly influenced by its associated microbiomes and their potential to improve growth under different stressful environments. Therefore, it is crucial and exciting to understand the mechanisms of plant–microbe interactions. Maize (Zea mays L.) is one of the primary staple foods worldwide, in addition to wheat and rice. Maize is also an industrial crop globally, contributing 83% of its production for use in feed, starch, and biofuel industries. Maize requires significant nitrogen fertilization to achieve optimal growth and yield. Maize plants are highly susceptible to heat, salinity, and drought stresses and require innovative methods to mitigate the harmful effects of environmental stresses and reduce the use of chemical fertilizers. This review summarizes our current understanding of the beneficial interactions between maize plants and specific microbes. These beneficial microbes improve plant resilience to stress and increase productivity. For example, they regulate electron transport, downregulate catalase, and upregulate antioxidants. We also review the roles of plant growth-promoting rhizobacteria (PGPR) in enhancing stress tolerance in maize. Additionally, we explore the application of these microbes in maize production and identify major knowledge gaps that need to be addressed to utilize the potential of beneficial microbes fully. The coastal saline zone of West Bengal in India is the home to millions of the world's poorest and most vulnerable people. Due to a gradual increase in salt accumulation on soils of the coastal saline zone of West Bengal in India from winter to summer days, cultivation of the second crop in the winter season becomes possible in a limited area. To address these issues, field experiments was conducted in rainy and winter seasons of 2016–17 and 2017–18 at the farmer's field of the coastal saline zone (CSZ) of West Bengal, India. The experiment was carried out to study the system productivity, nutrient uptake, and profitability vis-à-vis salinity dynamics of the crops in rice-pulse-based cropping systems under different land elevations (medium upland and medium lowland). The experiment was conducted in a strip-split plot design having horizontal factors namely, Factor A: Six dates of sowing of rice at an interval of one week (2nd week of June to 3rd week of July), Factor B: Two land situations (medium upland and medium lowland) and Two Cropping Systems (Rice-Lathyrus and Rice-Lentil) as vertical factor, replicated four times. The results suggest that irrespective of land situation, early sown rice (15 June to 21 June) produces higher dry matter and grain yield compared to late sown crops. This early sowing of rice also facilitated the better performance of subsequent lathyrus and lentil, by avoiding the worst situation of the salinity build-up and drought stress later in the winter. Moreover, significantly higher productions were obtained from medium-lowland situations for both the cropping systems. Sowing date has also significantly influenced macro-nutrient uptake (NPK) by rice and pulse grains. Chickpea (Cicer arietinum L.) is one of the most important legumes currently grown. It is an important source of proteins and nutrients, such as calcium, potassium and iron. As a result, precise crop management is necessary for maximizing its production. The presented study deals with the effect of soil heterogeneity caused by variable contents of macro- and micronutrients on the uptake of nutrients by chickpea. The values measured (contents of macro- and micronutrients in plant samples) indicate that soil heterogeneity is an important factor for the contents of nutrients and soil reactions, which strongly affect the growth of chickpea. We investigated the soil heterogeneity in a chickpea field. Two zones (A and B) with different stand development were found in the model plot. Zone A showed a healthy (green) growth, while Zone B exhibited a yellow-coloured growth, indicating deficits in nutrient uptake. The contents of selected nutrients (P, K, Ca, Mg, Fe, Cu, Zn and Mn) in the soil and in the plant biomass (i.e., stems, leaves, pods and seeds) were analyzed. In the zone with the yellow-coloured biomass, the results showed significantly (p < 0.05) reduced contents of N, P, K, Mg, Fe, Mn, Cu and Zn in the leaves; higher values of soil reaction (pH); and higher contents of calcium and calcium carbonate in the soil. The uptake of nutrients by the plants and their translocation were affected by the above-mentioned soil parameters and by their mutual interactions. Therefore, it is possible to state that soil heterogeneity (caused by variable contents of nutrients in soil) should be taken into account in the precise crop management of chickpeas. Background: Climate change is leading to severe and long-term droughts in European forest ecosystems. can have profound effects on various physiological processes, including photosynthesis, gene expression patterns, and nutrient uptake at the developmental stage of young trees. Objectives: Our study aimed to test the hypothesis that the application of silica (SiO2) influences photosynthetic efficiency and gene expression in 1- to 2-year-old Fagus sylvatica (L.) seedlings. Additionally, we aimed to assess whether silicon application positively influences the structural properties of leaves and roots. To determine whether the plant physiological responses are genotype-specific, seedlings of four geographically different provenances were subjected to a one-year evaluation under greenhouse conditions. Methods: We used the Kruskal–Wallis test followed by Wilcoxon's test to evaluate the differences in silicon content and ANOVA followed by Tukey's test to evaluate the physiological responses of seedlings depending on treatment and provenance. Results: Our results showed a significantly higher Si content in the roots compared with the leaves, regardless of provenance and treatment. The most significant differences in photosynthetic performance were found in trees exposed to Si treatment, but the physiological responses were generally nuanced and provenance-dependent. Expression of hsp70 and hsp90 was also increased in leaf tissues of all provenances. These results provide practical insights that Si can improve the overall health and resilience of beech seedlings in nursery and forest ecosystems, with possible differences in the beneficial role of silicon application arising from the large differences in wild populations of forest tree species. The lateral root (LR) is an essential component of the plant root system, performing important functions for nutrient and water uptake in plants and playing a pivotal role in cereal crop productivity. Nitrate (NO3−) is an essential nutrient for plants. In this study, wheat plants were grown in 1/2 strength Hoagland's solution containing 5 mM NO3− (check; CK), 0.1 mM NO3− (low NO3−; LN), or 0.1 mM NO3− plus 60 mg/L 2,3,5-triiodobenzoic acid (TIBA) (LNT). The results showed that LN increased the LR number significantly at 48 h after treatment compared with CK, while not increasing the root biomass, and LNT significantly decreased the LR number and root biomass. The transcriptomic analysis showed that LN induced the expression of genes related to root IAA synthesis and transport and cell wall remodeling, and it was suppressed in the LNT conditions. A physiological assay revealed that the LN conditions increased the activity of IAA biosynthesis-related enzymes, the concentrations of tryptophan and IAA, and the activity of cell wall remodeling enzymes in the roots, whereas the content of polysaccharides in the LRP cell wall was significantly decreased compared with the control. Fourier-transform infrared spectroscopy and atomic microscopy revealed that the content of cell wall polysaccharides decreased and the cell wall elasticity of LR primordia (LRP) increased under the LN conditions. The effects of LN on IAA synthesis and polar transport, cell wall remodeling, and LR development were abolished when TIBA was applied. Our findings indicate that NO3− starvation may improve auxin homeostasis and the biological properties of the LRP cell wall and thus promote LR initiation, while TIBA addition dampens the effects of LN on auxin signaling, gene expression, physiological processes, and the root architecture. Methicillin-resistant Staphylococcus aureus (MRSA) is a highly threatening foodborne pathogen capable of causing severe organ and life-threatening diseases. Over the past years, various commercial antibiotics have been used to treat MRSA infections. However, these commercial antibiotics have not yielded efficient results and also cause other side effects; therefore, there is a need for the development of effective alternatives to replace these commercial antibiotics. Suberanilic acid, an amide alkaloid obtained from the endophytic fungus Pestalotiopsis trachycarpicola DCL44, has been identified as a significant antimicrobial agent. However, its antibiotic properties on multi-drug-resistant bacteria such as MRSA have not been fully explored. Therefore, to investigate the potential antimicrobial mechanism of suberanilic acid against MRSA, a quantitative proteomics approach using tandem mass tagging (TMT) was used. The results obtained in the study revealed that suberanilic acid targets multiple pathways in MRSA, including disruption of ribosome synthesis, inhibition of membrane translocation for nutrient uptake (ABC transporter system), and causing dysregulation of carbohydrate and amino acid energy metabolism. These results provide new insights into the mechanism of action of suberanilic acid against MRSA and offer technical support and a theoretical basis for the development of novel food antimicrobial agents derived from endophytic fungal origin. Hemp (Cannabis sativa L.), renowned for its applications in environmental, industrial, and medicinal fields, is critically evaluated in this comprehensive review focusing on the impacts of chemical and organic fertilizers on its cultivation. As hemp re-emerges as a crop of economic significance, the choice between chemical and organic fertilization methods plays a crucial role in determining not only yield but also the quality and sustainability of production. This article examines the botanical characteristics of hemp, optimal growth conditions, and the essential biochemical processes for its cultivation. A detailed comparative analysis is provided, revealing that chemical fertilizers, while increasing yield by up to 20% compared to organic options, may compromise the concentration of key phytochemicals such as cannabidiol by approximately 10%, highlighting a trade-off between yield and product quality. The review presents quantitative assessments of nitrogen (N), phosphorus (P), and potassium (K) from both fertilizer types, noting that K significantly influences the synthesis of terpenes and cannabinoids, making it the most impactful element in the context of medicinal and aromatic hemp varieties. Optimal rates and timing of application for these nutrients are discussed, with a focus on maximizing efficiency during the flowering stage, where nutrient uptake directly correlates with cannabinoid production. Furthermore, the challenges associated with the U.S. industrial hemp market are addressed, noting that reducing production costs and improving processing infrastructure is essential for sustaining industry growth, especially given the slow expansion in fiber and cannabidiol markets due to processing bottlenecks. The review concludes that while chemical fertilizers may offer immediate agronomic benefits, transitioning towards organic practices is essential for long-term environmental sustainability and market viability. Purslane (Portulaca oleracea L.) is a common weed that attracts research and agricultural attention because of its significant nutritional value, and it is used commercially. The present work rated the effect of different nitrogen (N) concentrations, i.e., N50: 50 mg L−1; N100: 100 mg L−1; N200: 200 mg L−1; and N300: 300 mg L−1, on the growth, physiology, antioxidant capacity, and nutrient accumulation in the different plant parts of hydroponically grown purslane. Seedlings were transplanted to a Nutrient Film Technique (NFT) system and exposed to different N levels. The plants grown in high N levels of 300 mg L−1 had decreased biomass production, leaf number, leaf stomatal conductance, and total flavonoid content because of the increased oxidative stress, as shown by the elevated lipid peroxidation levels. Several enzymatic (superoxide dismutase) and non-enzymatic (ascorbic acid, total phenolics) plant antioxidant activities were activated to counteract the oxidative factors. Plants grown in intermediate N levels in the NS (i.e., 100 mg L−1) had decreased oxidative stress as several enzymatic antioxidant activities, i.e., peroxidases and catalases, were increased. Additionally, the water use efficiency and nitrogen uptake, as well as leaf stomatal conductance and leaf chlorophyll fluorescence, were increased in plants grown at a N level of 200 mg L−1. The mineral accumulation in the leaves, stems, and roots fluctuated, with increased calcium, magnesium, and sodium content being observed in the plants grown at high N levels in the nutrient solution. The stems accumulated less N compared with the leaves, but the N content and accumulation rates in purslane leaves were not affected by the N levels in the nutrient solution. Therefore, to increase the yield, nutritional value, and water use efficiency of purslane that is grown hydroponically, a concentration of 200 mg L−1 N in the nutrient solution is recommended. Nutrients are absorbed by special transport proteins on the cell membrane; however, there is less information regarding transporters across the mycobacterial outer envelope, which comprises dense and intricate structures. In this study, we focus on the model organism Mycolicibacterium smegmatis, which has a cell envelope similar to that of Mycobacterium tuberculosis, as well as on the TiME protein secretion tube across the mycobacterial outer envelope. We present transcriptome results and analyze the protein compositions of a mycobacterial surface envelope, determining that more transporters and porins are induced to complement the deletion of the time gene in Mycolicibacterium smegmatis. The TiME protein is essential for nutrient utilization, as demonstrated in the uptake experiments and growth on various monosaccharides or with amino acids as the sole carbon source. Its deletion caused bacteria to be more sensitive to anti-TB drugs and to show a growth defect at an acid pH level, indicating that TiME promotes the survival of M. smegmatis in antibiotic-containing and acidic environments. These results suggest that TiME tubes facilitate bi-directional processes for both protein secretion and nutrient uptake across the mycobacterial outer envelope. [ABSTRACT FROM AUTHOR]

Macroalgal biomass blooms, including those causing the green and golden tides, have been rising along Chinese coasts, resulting in considerable social impacts and economic losses. To understand the links between the ongoing climate changes (ocean warming and acidification) and algal tide formation, the effects of temperature (20 and 24 °C), pCO2 concentration (Partial Pressure of Carbon Dioxide, 410 ppm and 1000 ppm) and their interaction on the growth of Ulva prolifera and Ulva lactuca (green tide forming species), as well as Sargassum horneri (golden tide forming species) were investigated. The results indicate that the concurrent rises in temperature and pCO2 level significantly boosted the growth and nutrient uptake rates of U. lactuca. For U. prolifera, the heightened growth and photosynthetic efficiency under higher CO2 conditions are likely due to the increased availability of inorganic carbon. In contrast, S. horneri exhibited negligible responsiveness to the individual and combined effects of the increased temperature and CO2 concentration. These outcomes indicate that the progressive climate changes, characterized by ocean warming and acidification, are likely to escalate the incidence of green tides caused by Ulva species, whereas they are not anticipated to precipitate golden tides. Nano-biochar is a source of blackish carbonaceous material, a prerequisite for sustainable crop productivity. By using a variety of feedstock materials, nanobiochar synthesis can be employed via pyrolysis. Therefore, a project was initiated to explore the morpho-physio-biochemical alteration at the vegetative stage of wheat crops after the foliar application of nanobiochar suspension (NBS). This investigation was conducted at the Botanical Research Area of the University of Lahore in a randomized complete block design (RCBD) arrangement, with four treatments (0, 1, 3, and 5% NBS) by maintaining three replications for each treatment using the wheat variety "Zincol". Nano biochar suspension in above mentioned concentrations were foliarly applied at the end of tillering/beginning of leaf sheath elongation of wheat seedlings to assess the morphological changes (root length, shoot length, number of leaves, fresh biomass/plant, dry biomass/plant), physio-biochemical alterations (total free amino acids, total sugars, chlorophyll content, protein, phenols, flavonoids), and nutrient uptake (Na, K, Ca, Mg, N, P contents. Our findings indicate that the foliar application of 3% NBS yielded the most favorable results across all measured attributes. Furthermore, Treatment-4 (5% NBS) specifically improved certain traits, including leaf area, total soluble proteins, and leaf calcium content. Microalgae offer distinct advantages as a nutritional source for aquaculture and as a means of wastewater bioremediation. Studying the phycosphere bacteria and understanding their complex interactions is essential to optimizing high-quality biomass growth. This study aimed to isolate, characterize, and identify bacteria from the phycosphere of marine microalgae and to determine their potential to enhance growth, metabolism, and bioremediation capabilities of Chaetoceros calcitrans in stress nutrient-poor media simulating aquaculture wastewater enriched with nitrate, nitrite, or phosphorus. Bacterial characterization included tests for auxin and siderophore production, biofilm formation, amylase activity, phosphate solubilization, mobility, and antagonism evaluation. When Alteromonas macleodii, Bacillus cereus, and Marinobacter sp. were selected and then enriched (107 CFU/mL) in co-culture with C. calcitrans, growth levels significantly increased in four of six Synthetic Aquaculture Wastewater (SAW) media. Pigment levels were higher in five of six SAW media, and lipid levels were higher in SAW rich in nitrite (SAWni50) and phosphorus (SAWpho50). In addition, C. calcitrans with or without the bacterial consortium demonstrated excellent phosphorus bioremediation, achieving 67.6% average removal in SAWpho50. Nitrate and nitrite assimilation rates were approximately 10% in SAWna and SAWni50. This study marks the inaugural identification of these bacteria as microalga growth-promoting bacteria (MGPB) for enhancing growth and lipid and pigment production in C. calcitrans, and it also documents a maximum of 69.13% phosphorus removal. The nitrogen (N) use efficiency (NUE) in the roots of seedlings is beneficial for increasing crop yield. Creating marker-assisted selection for wheat root traits can assist wheat breeders in choosing robust roots to maximize nutrient uptake. Exploring and identifying the effect of different N supply conditions on root system architecture (RSA) is of great significance for breeding N efficient wheat varieties. In this study, a total of 243 wheat varieties native to the Yellow and Huai Valley regions of China were utilized for genome-wide association studies (GWAS). Furthermore, a recombinant inbred line (RIL) population of 123 lines derived from the cross between Avocet and Chilero was utilized for linkage examination. A hydroponic seedling experiment using a 96-well tray was conducted in the lab with two treatments: normal N (NN) and low N (LN). Five RSA traits, including the relative number of root tips (RNRT), relative total root length (RTRL), relative total root surface area (RTRS), relative total root volume (RTRV), and relative average root diameter (RARD), were investigated. GWAS and linkage analysis were performed by integrating data from the wheat 660 k single nucleotide polymorphism (SNP) chip and diversity arrays technology (DArT) to identify genetic loci associated with RSA. The results showed that, based on the ratio of RSA-related traits under two N supply conditions, a total of 497 SNP markers, which are significantly associated with RSA-related traits, were detected at 148 genetic loci by GWAS. A total of 10 QTL loci related to RSA were discovered and identified by linkage mapping. Combining two gene localization methods, three colocalized intervals were found: AX-95160997/QRtrl.haust-3D, AX-109592379/QRnrt.haust-5A, and AX-110924288/QRtrl.haust-7D/QRtrs.haust-7D. According to the physical location of the colocalization of these two sites, between 39.61 and 43.74 Mb, 649.97 and 661.55 Mb, and 592.44 and 605.36 Mb are called qRtrl-3D, qRnrt-5A, and qRtrl-7D. This study has the potential to enhance the effectiveness of selecting root traits in wheat breeding programs, offering valuable insights into the genetic underpinnings of NUE in wheat. In cereal-based farming, there is significant influence of tillage and residue incorporation as sustainable practices, although their role in crop production is still unclear. Two field trials were executed during winter seasons to evaluate the best-performing crop residue and tillage system for the nutrition and yield of rice at the Soil Science Field Laboratory of Bangladesh Agricultural University. In the first trial, residues from soybean, black gram, and rice were incorporated into the soil with 75% recommended fertilizer doses (RFD). The grain yields were 5.97, 6.21, and 6.10 t ha−1, respectively in rice, soybean, and black gram residue-treated plots, which were increased by 77.15, 84.27, and 81.01%, respectively, over the control. In the second trial, the residues from black gram and rice were incorporated with 100% RFD under conventional tillage (CT) and minimum tillage (MT) for two years, where CT with black gram residue plus 100% fertilizer (CT-I2) exhibited the highest grain yield of 6.69 and 6.88 t ha−1, increasing by 7.61% and 8% over 100% RFD. Both CT and MT performed similarly, and their combination with legume residue strongly influenced crop performance and nutrient uptake. Drought stress is one of the major impediments to plant growth. Plant growth-promoting rhizobacteria (PGPR) can mitigate moisture stress in plants by increasing the ability of plant nutrient uptake and transport. In this study, we investigated the root phenotype, mineral nutrients (in leaves, roots, and soil), soil pH, water saturation deficit (WSD), free water content (FWC), and bound water content (BWC) of leaves of two alfalfa varieties, 'Galalxie Max' (drought-tolerant) and 'Saidi 7' (drought-sensitive), in the presence or absence of Bacillus amyloliquefaciens QST713 under drought stress conditions. The results showed that water stress negatively affected both cultivar root morphology (total root length, average diameter, total surface area, and volume) and the contents of K and Fe in leaves, roots, and soil. It also reduced the Mn and Zn contents in the soil while increasing the content of Na in the leaves and soil. Additionally, alfalfa plants under drought stress exhibited higher levels of soil pH, WSD, and BWC but lower contents of FWC and ratios of BWC/FWC in the leaves of both cultivars. However, QST713 application significantly enhanced the total root length, average root diameter, and the contents of K and Fe in alfalfa leaves, roots, and soil, as well as the BWC/FWC ratio in leaves under drought stress conditions. A significant reduction in the Na content was detected in QST713-treated alfalfa leaves and soil under drought stress. Furthermore, QST713 application noticeably decreased soil pH and WSD. The current findings showed that QST713 enhanced the water stress tolerance of alfalfa plants by ameliorating root morphology, reducing soil pH, and improving the BWC/FWC ratio, consequently promoting the accumulation of mineral nutrients (mainly K and Fe). Overall, Bacillus amyloliquefaciens QST713 can serve as a potential green fertilizer in sustainable agriculture to improve soil nutrients and enhance plant production under increasing drought conditions. Biochar is widely used to maintain crop yields and the sustainability of agroecosystems due to its characteristics. However, its effect on vegetable yield and quality in southern acidic soils has not been fully elucidated. The effects of two kinds of biochar application rates (3 t ha−1 and 12 t ha−1) on the nutrient availability, nutrient absorption, yield, and fruit quality of sandy acid oxide soil in South China were studied in a 2-year field experiment using two varieties of wax gourd (Tiezhu No. 2 and Dadao). The results showed that optimized fertilization (OPT, nitrogen, phosphorus, and potassium were reduced by 20.0%, 20.6%, and 21.1%, respectively) did not reduce the yield of the different varieties of wax gourd compared to FP (farmer practice). On the basis of the OPT treatment, the application of biochar increased the yield of Tiezhu No. 2 and Dadao wax gourd by 16.0–27.3% and 7.7–13.3%, respectively. The increase in yield was attributed to the improvement in soil organic carbon content which increased by 4.5–19.7%, nutrient effectiveness (NO3−-N and Olsen-P content enhanced by 23.7–27.0% and 15.3–23.4% in Tiezhu No. 2), and nutrient uptake (N, P, K, Ca, and Mg accumulation increased significantly by 21.1%, 46.1%, 36.8%, 25.7%, and 31.9%, respectively) by the plant after the biochar application. Different types of biochar also have some differences in these three aspects. Specifically, under the same dosage, rice biochar has a more significant effect on increasing the yield of winter melon, and has a better effect on the improvement of soil physical and chemical properties, while sawdust biochar has a more significant effect on nutrient absorption. In addition, the commercial quality, namely hardness and glossiness, and the nutritional quality, namely soluble sugar, soluble protein, VC content, and sugar–acid ratio of wax gourd fruits, were significantly improved after the biochar application. In summary, biochar application on acidic soils in South China could achieve a win–win situation in terms of increasing soil nutrient effectiveness to improve vegetable yield and quality while reducing chemical fertilizer. Phosphorus (P) and potassium (K) stress significantly affect the growth, physiological characteristics, and nutrient uptake of rice plants. This study investigated the photosynthetic nitrogen (N) metabolism, N uptake, and N utilization of plants under varied P and K supplies. Two local conventional high-yield rice varieties (Shennong 265 and Liaojing 294) were used. These varieties were subjected to the following hydroponic experimental treatments: HPHK (normal P and K concentrations), HPLK (normal P and 1/20 normal K concentration), LPHK (normal K and 1/20 normal P concentration), and LPLK (1/20 normal P and K concentrations). The results showed that the mesophyll cells had a relatively complete nuclear and chloroplast structures, and the antioxidant enzymes of the plants were significantly reduced under the HPHK treatment. Compared to the LP treatments (LPHK and LPLK), the HPHK treatment was found to have the following potential effects: effectively optimize plant configuration; promote leaf development (Pn, E, Ci, and Tr, chlorophyll, and leaf area index); significantly increase the N-metabolism-related enzyme activity of leaves and roots and the accumulation of N in the plant in the main growth stages; and significantly increase the rice yield and N-related efficiency. In conclusion, the HPHK treatment was found to be beneficial in improving the plant configuration, promoting photosynthetic N metabolism, and increasing grain yield and N-related utilization efficiency. The physical properties of nano-fertilizers (NFs) are important in determining their performance, efficacy, and environmental interactions. Nano-fertilizers, due to their small size and high surface area-to-volume ratio, enhance plant metabolic reactions, resulting in higher crop yields. The properties of nano-fertilizers depend on the synthesis methods used. The nanoparticle's nutrient use efficiency (NUE) varies among plant species. This review aims to analyze the relationship between the physical properties of NF and their influence on crop performance and nutrient uptake efficiency. The review focuses on the physical properties of NFs, specifically their size, shape, crystallinity, and agglomeration. This review found that smaller particle-sized nanoparticles exhibit higher nutrient use efficiency than larger particles. Nano-fertilizer-coated additives gradually release nutrients, reducing the need for frequent application and addressing limitations associated with chemical fertilizer utilization. The shapes of nano-fertilizers have varying effects on the overall performance of plants. The crystalline structure of nanoparticles promotes a slow release of nutrients. Amorphous nano-fertilizers improve the NUE and, ultimately, crop yield. Agglomeration results in nanoparticles losing their nanoscale size, accumulating on the outer surface, and becoming unavailable to plants. The importance of implementing recirculating drainage for greenhouse strawberries is often overlooked because of the low electrical conductivity (EC) of drainage and transpiration despite the large area of cultivation in Korea. In this study, we analyzed the growth of strawberry crops and their water and individual nutrient use efficiency when using closed-loop hydroponics in greenhouses. The study consisted of two parts: Experiment (Exp) 1 and Exp 2, each of which employed a different closed-loop hydroponic control method. In Exp 1, the system was controlled solely based on the EC of the drainage mixed with raw water. In Exp 2, the nutrient solution (NS) was corrected according to the ion concentrations in the drainage, with correction intervals of 2 weeks, 4 weeks, and no correction, while the control treatment was open-loop. In Exp 1, a prolonged increase in drainage EC resulted in an imbalance in NS ion composition and reduced fruit yield. In Exp 2, the NS composition was stabilized through periodic nutrient correction, and the fruit yield of the closed-loop treatments did not differ from the open-loop treatment. However, the closed-loop with 2- and 4-week correction in Exp 2 showed 94% and 88% higher nutrient use efficiency (NUE), respectively, than an open-loop system. Among the closed-loop treatments, the 2- and 4-week correction intervals had 36% and 32% higher NUE than the no-correction treatment. Furthermore, the 2-week correction interval showed a 3% improvement in NUE compared to the 4-week interval. These findings highlight the importance of considering a shorter ion correction interval for optimal NUE and normal plant growth in recirculating hydroponic systems of strawberry cultivation. The use of natural-origin biomaterials in bioengineering has led to innovative approaches in agroforestry. Bacterial cellulose (BC), sharing the same chemical formula as plant-origin cellulose (PC), exhibits significantly different biochemical properties, including a high degree of crystallinity and superior water retention capacity. Previous research showed that natural-origin glucose-based chitin enhanced plant growth in both herbaceous and non-herbaceous plants. In this study, we produced BC in the laboratory and investigated its effects on the substrate and on Solanum lycopersicum seedlings. Soil amended with BC increased root growth compared with untreated seedlings. Additionally, under limited irrigation conditions, BC increased global developmental parameters including fresh and dry weight, as well as total carbon and nitrogen content. Under non-irrigation conditions, BC contributed substantially to plant survival. RNA sequencing (Illumina®) on BC-treated seedlings revealed that BC, despite its bacterial origin, did not stress the plants, confirming its innocuous nature, and it lightly induced genes related to root development and cell division as well as inhibition of stress responses and defense. The presence of BC in the organic substrate increased soil availability of phosphorus (P), iron (Fe), and potassium (K), correlating with enhanced nutrient uptake in plants. Our results demonstrate the potential of BC for improving soil nutrient availability and plant tolerance to low irrigation, making it valuable for agricultural and forestry purposes in the context of global warming. The aim of this study was to compare Fe, Zn, Mn, Al and Li contents in table potato tubers, obtained from seven potato cultivars with different flesh colour, and one cultivar characterised by light-coloured flesh. The study material consisted of potato tubers produced in a one-factor field experiment carried out at the Agricultural Experimental Station in Zawady belonging to the University of Siedlce, Poland (52°03′ N, 22°33′ E) in 2021–2023. The following cultivars were tested: Eurostar (light yellow flesh), Rote Emmalie and Herbie 26 (red flesh), Provita, Salad Blue, Blaue Annelise, Vitelotte Noire and Bora Valley (purple and blue flesh). The study showed that the genetic characteristics of the cultivars determined the nutrient content and uptake with tuber yield. Of the tested minerals which are important for human nutrition (Fe, Zn, Mn), red and purple cultivars accumulated more Fe than the light yellow-fleshed Eurostar. Zn content was the highest in red-fleshed cultivars compared with light and purple-fleshed cultivars. Mn content was the highest in the tubers of the light yellow-fleshed Eurostar and lowest in red Rote Emmalie. Ammonium sulfate (AS) has been utilized in agriculture; however, there is a dearth of research on its application in maize cultivation subsequent to the implementation of nitrification inhibitors or coating treatments. This study aimed to analyze the impacts of various combinations of AS fertilizers on soil nutrients, plant nutrient uptake, yield, and fertilizer utilization efficiency in maize cultivation to establish an optimal and stabilized disposal method for AS. A completely randomized design was employed with five treatments (AU, the control using urea; AS, treatment using ammonium sulfate; ASN, treatment using ammonium sulfate with a nitrification inhibitor; ASG, treatment using oil-coated ammonium sulfate; and ASD, treatment using oil–humic acid-coated ammonium sulfate). The results show the following: (1) Compared with AU and AS, ASN, ASG, and ASD decreased the leaching rates of total nitrogen (TN), ammonium nitrogen (NH4+-N), and nitrate nitrogen (NO3−-N), and more residual N had accumulated in the soil. The first-order kinetic equation Nt = N0(1 − e−kt) could better fit the process of N accumulation and release, and the N-release rate constant was in the order of AU > CK > AS > ASG > ASN > ASD. (2) Compared with the AU and AS treatments, the plant dry weight, grain dry weight, spike width, spike length, and yields of maize increased by 8.85–11.08%, 12.98–14.15%, 2.95–3.52%, 5.50–5.65%, and 43.21–51.10%, respectively, under the ASG treatment. A path analysis revealed the main decision coefficient of the effective spike number on the maize yield. Furthermore, the accumulation levels of N, P, and K within above-ground plants significantly increased under the ASG treatment compared with those under the AU and AS treatments. N, P, and K partial factor productivity under the ASG treatment increased by 47.12%, 47.15%, and 73.40% on average, while grain N, P, and K balance increased by 50.45%, 47.10%, 55.61% on average, compared with the AU and AS treatments. Molecular hydrogen (H2) is crucial for agricultural microbial systems. However, the mechanisms underlying its influence on crop yields is yet to be fully elucidated. This study observed that H2-based irrigation significantly increased strawberry (Fragaria × ananassa Duch.) yield with/without nutrient fertilization. The reduction in soil available nitrogen (N), phosphorus (P), potassium (K), and organic matter was consistent with the increased expression levels of N/P/K-absorption-related genes in root tissues at the fruiting stage. Metagenomics profiling showed the alterations in rhizosphere microbial community composition achieved by H2, particularly under the conditions without fertilizers. These included the enrichment of plant-growth-promoting rhizobacteria, such as Burkholderia, Pseudomonas, and Cupriavidus genera. Rhizobacteria with the capability to oxidize H2 (group 2a [NiFe] hydrogenase) were also enriched. Consistently, genes related to soil carbon (C) fixation (i.e., rbcL, porD, frdAB, etc.), dissimilar nitrate reduction (i.e., napAB and nrfAH), and P solublization, mineralization, and transportation (i.e., ppx-gppA, appA, and ugpABCE) exhibited higher abundance. Contrary tendencies were observed in the soil C degradation and N denitrification genes. Together, these results clearly indicate that microbe-mediated soil C, N, and P cycles might be functionally altered by H2, thus increasing plant nutrient uptake capacity and horticultural crop yield. Surface runoff and subsurface flow patterns were monitored in hillside runoff plots in almond and olive orchards with soils covered with spontaneous plants over two hydrological years. The experimental runoff plots were located on the south flank of the Sierra Nevada (Lanjarón, SE Spain) at 580 m a.s.l. with an area of 40 m2 (10 m × 4 m). The surface and subsurface discharge were collected and measured at different soil depths (0, 5, 10, 25, and 50 cm), and the dissolved nutrient concentrations (NO3–N, NH4–N, PO4–P, and K) were determined. According to the findings, the subsurface flow pathways drained most of the rainfall water compared with surface runoff, which was affected by plant cover. The influence of rainfall intensity (I30) on surface runoff was more meaningful than that on subsurface flow. Throughout the monitoring period, the runoff coefficients at soil depths of 0, 5, 10, 25, and 50 cm averaged 0.04, 0.11, 0.14, 0.17, and 0.18, respectively. Subsurface flow was one of the dominant pathways for N and K loss, whereas P loss mainly occurred via surface runoff. Moreover, the concentrations in subsurface flow were higher than the recommended level for standard water quality for NO3–N, NH4–N, and PO4–P. Subsurface flow was the main route of dissolved nutrient delivery, making these nutrients available to the root systems of trees, where nutrient uptake is more likely to occur. Thus, by lessening surface runoff and encouraging surface vegetation coverage to facilitate the recycling of nutrients and buffer the rainfall's impact on the soil surface, nutrient loss control can be achieved. In modern agricultural production, cattle manure waste recovery is considered as a sustainable approach to agricultural waste management, reducing environmental pollution and chemical fertilizer use. This study aimed to investigate the effects of manure and digestate derived from a pilot-scale livestock waste-recycling system, in combination with a low copper concentration as a fungicide, on the physiological response of lettuce cv Rufus (Lactuca sativa L.) plants and the associated soil microbiome. A five-week microcosm experiment was conducted in a greenhouse under environmental conditions. Lettuce plant performance was assessed in terms of biomass, leaf area index, photosynthetic activity, chlorophyll measurements, lipid peroxidation, total phenolic content, and nutrient uptake. The results suggested that incorporating digestate into the potting soil mix significantly enhanced crop yields compared to the control and manure treatments. The soil microbial activity increased in the presence of fertilizers, improving the soil chemical and biological properties. The addition of copper negatively affected the growth and physiological performance of the lettuce plants under both the control and manure-treated conditions, except for those grown in the presence of digestate, where copper accumulation was reduced. These findings highlight the potential of growing horticultural crops using organic fertilization through livestock waste anaerobic digestate, establishing a waste-to-food recycling system. Colpodella species are free-living protists phylogenetically related to apicomplexans. Colpodella sp. have been detected in human and animal tissues, as well as in ticks and biting flies. The trophozoite and cyst stages of Colpodella species can be distinguished from stages of the prey Parabodo caudatus using Sam-Yellowe's trichrome staining. Colpodella species obtain nutrients by attaching to their prey, aspirating the prey's cytoplasmic contents into a posterior food vacuole and encysting. It is unclear whether both trophozoite and cyst stages are present in human and animal tissues. Molecular techniques have detected Colpodella species in human blood, cerebrospinal fluid, and in ticks and flies. However, no morphological information was reported to aid life-cycle stage identification of Colpodella species. This review discusses the increased reports of Colpodella species detection in animals and in arthropods and the need to identify stages present in human and animal tissues. We previously used Sam-Yellowe's trichrome staining to identify life-cycle stages of Colpodella sp. In this review, we examine the reports of Colpodella species detection in human and animal tissues to determine whether the identification of Colpodella species represents true infections or contaminations of samples collected during routine surveillance of piroplasm infections in animals and arthropods. This review also aims to provide insights regarding Colpodella, nutrient uptake, and the survival of Colpodella sp. within humans, animals, and arthropods, as well as whether the attachment of trophozoites to cells occurs in tissues leading to myzocytosis and endocytosis. Cadmium (Cd) is a naturally occurring toxic heavy metal that adversely affects plant germination, growth, and development. While the effects of Cd have been described on many crop species including rice, maize, wheat and barley, few studies are available on cadmium's effect on Tartary buckwheat which is a traditional grain in China. We examined nine genotypes and found that 30 µM of Cd reduced the root length in seedlings by between 4 and 44% and decreased the total biomass by 7 to 31%, compared with Cd-free controls. We identified a significant genotypic variation in sensitivity to Cd stress. Cd treatment decreased the total root length and the emergence and growth of lateral roots, and these changes were significantly greater in the Cd-sensitive genotypes than in tolerant genotypes. Cd resulted in greater wilting and discoloration in sensitive genotypes than in tolerant genotypes and caused more damage to the structure of root and leaf cells. Cd accumulated in the roots and shoots, but the concentrations in the sensitive genotypes were significantly greater than in the more tolerant genotypes. Cd treatment affected nutrient uptake, and the changes in the sensitive genotypes were greater than those in the tolerant genotypes, which could maintain their concentrations closer to the control levels. The induction of SOD, POD, and CAT activities in the roots and shoots was significantly greater in the tolerant genotypes than in the sensitive genotypes. We demonstrated that Cd stress reduced root and shoot growth, decreased plant biomass, disrupted nutrient uptake, altered cell structure, and managed Cd-induced oxidative stress differently in the sensitive and tolerant genotypes of Tartary buckwheat. More exact information on soil nutrient management is crucial due to environmental protection, nature conservation, decreasing sources for mining, general precaution, etc. Soil magnesium (Mg) analytical methods of potassium chloride (KCl), Mehlich 3 (M3), water (WA) and cobalt hexamine (CoHex) extractions are compared with an elemental analysis and X-ray fluorescence (XRF) analysis. The ratio of the available to the total Mg content was calculated and compared on the whole dataset. The results showed that the linear regressions between all the pairs of Mg content measurement methods were significant. The linear relationship between the KCl and CoHex methods has the highest determination coefficient (R2 = 0.96), followed by WA–M3 (R2 = 0.68), M3–CoHex (R2 = 0.66) and M3–KCl (R2 = 0.60). The M3 solution demonstrated a greater capacity for extracting Mg from the soil. The second part is the analysis of the influence of CaCO3, pH, soil texture and clay content on the measurable magnesium content of soils. It was established that the extraction methods, the soil and the classification method of the soil properties affect the evaluation. In order to solve the problem of water waste and environmental pollution in arid and semiarid areas, this study aimed to investigate the effects of different water and nitrogen treatments on the growth of apple leaves, photosynthetic physiology, nutrient uptake and nitrogen use efficiency. A two-factor experimental design was used in this experiment. The first factor was three levels of irrigation (40–50%, 50–65%, 65–80% RWC, relative water content) and the second factor was four levels of nitrogen application (0, 150, 300, 600 kg/ha), which were a completely randomized combination. The results showed that irrigation and nitrogen application had significant effects on the growth, photosynthetic rate, chlorophyl and mineral element contents of apple tree leaves, but excessive input of water and nitrogen would restrain the growth of apple tree leaves. The Ndff% of different organs for apple trees were evaluated by 15N isotope tracer technology, and the highest Ndff% value was found in leaves, ranging from 1.01–1.23‰. Next were roots with 0.29–0.43‰, and the lowest Ndff% was found in fruits with 0.03–0.08‰. The calculation results of 15N utilization in apple trees showed that nitrogen use efficiency (NUE) under the medium-water low nitrogen (W1N1) treatment had the highest value with 43.33%. In conclusion, considering the growth physiology and nitrogen utilization of apple trees, it is recommended that a water and fertilizer coupling combination scheme for apple trees in arid and semi-arid areas of the Loess Plateau should be that of a medium-water and medium-nitrogen mode (W1N2, 50–65% RWC, 300 kg/ha). The incorporation of S fertilizers into macronutrient fertilizers such as ammonium phosphate represents a straightforward and economically efficient approach to alleviating S deficiency, strengthening S supply, and improving crop yield. However, limited research has been conducted to assess the effect of monoammonium phosphate (MAP) and diammonium phosphate (DAP) with different S additions on agronomic outcomes. In this study, ammonium sulfate and elemental S with S set at 3%, 6%, 9%, and 12% (ensuring a 1:1 ratio of SO4−S to elemental S) were granulated with MAP and DAP, respectively. Maize was used as the test crop to evaluate its yield, nutrient uptake, and apparent sulfur recovery. The results showed that S-fortified MAP treatment increased crop yield and S uptake by an average of 9.3% and 10.6%, respectively. A significant difference in crop yield and S uptake was observed when the S addition in MAP exceeded 9% S. Nevertheless, no statistical difference was found among the DAP-based treatments in calcareous soil. There was a strong relationship between S applied in fertilizers and S uptake by crops for MAP-based treatments. However, the apparent sulfur recovery drastically dropped from 44.2% to 7.19% with the increased addition level of S for MAP-based fertilizers. The results of this study indicate that the addition of S to MAP could be a simple, low-cost, and effective approach with great potential to promote S fertilizer application, minimize soil S deficiency, and improve crop yield in calcareous soil. The long-term use of chemical fertilizers poses a serious threat to crop productivity and soil quality. Organic fertilizers are used to improve the soil fertility and crop productivity. The application of organic fertilizers improves soil health and plant growth by improving the soil organic matter (SOM), soil structure, aggregate stability, nutrient uptake, water-holding capacity, cation exchange capacity, nutrient use efficiency and microbial activities of soil. The intensity of abiotic stress is continuously increasing, which is a serious threat to crop productivity and global food security. However, organic fertilizers have been reported to improve tolerance against drought, salinity, heat and heavy metal (HM) stresses. The application of organic fertilizer improves the leaf water status, nutrient uptake, nutrient homeostasis, synthesis of chlorophyll, osmolytes, hormones, secondary metabolites, antioxidant activities and gene expression, resulting in improved tolerance against drought, salinity, heat, and heavy metals. In the present review, we have discussed the ability of organic fertilizers to improve soil fertility, crop yield, and the nutrient use efficiency. We have also presented the various mechanisms through which organic fertilizers improve tolerance against drought, salinity, heat, and heavy metals. Therefore, this review will put forth new directions for researchers working on the use of organic materials to improve soil fertility, crop productivity and tolerance against abiotic stresses. Grain filling is critical for determining yield and quality, raising the question of whether central coordinators exist to facilitate the uptake and storage of various substances from maternal to filial tissues. The duplicate NAC transcription factors ZmNAC128 and ZmNAC130 could potentially serve as central coordinators. By analyzing differentially expressed genes from zmnac128 zmnac130 mutants across different genetic backgrounds and growing years, we identified 243 highly and differentially expressed genes (hdEGs) as the core target genes. These 243 hdEGs were associated with storage metabolism and transporters. ZmNAC128 and ZmNAC130 play vital roles in storage metabolism, and this study revealed two additional starch metabolism-related genes, sugary enhancer1 and hexokinase1, as their direct targets. A key finding of this study was the inclusion of 17 transporter genes within the 243 hdEGs, with significant alterations in the levels of more than 10 elements/substances in mutant kernels. Among them, six out of the nine upregulated transporter genes were linked to the transport of heavy metals and metalloids (HMMs), which was consistent with the enrichment of cadmium, lead, and arsenic observed in mutant kernels. Interestingly, the levels of Mg and Zn, minerals important to biofortification efforts, were reduced in mutant kernels. In addition to their direct involvement in sugar transport, ZmNAC128 and ZmNAC130 also activate the expression of the endosperm-preferential nitrogen and phosphate transporters ZmNPF1.1 and ZmPHO1;2. Malaria parasites increase their host erythrocyte's permeability to obtain essential nutrients from plasma and facilitate intracellular growth. In the human Plasmodium falciparum pathogen, this increase is mediated by the plasmodial surface anion channel (PSAC) and has been linked to CLAG3, a protein integral to the host erythrocyte membrane and encoded by a member of the conserved clag multigene family. Whether paralogs encoded by other clag genes also insert at the host membrane is unknown; their contributions to PSAC formation and other roles served are also unexplored. Here, we generated transfectant lines carrying epitope-tagged versions of each CLAG. Each paralog is colocalized with CLAG3, with concordant trafficking via merozoite rhoptries to the host erythrocyte membrane of newly invaded erythrocytes. Each also exists within infected cells in at least two forms: an alkaline-extractable soluble form and a form integral to the host membrane. Like CLAG3, CLAG2 has a variant region cleaved by extracellular proteases, but CLAG8 and CLAG9 are protease resistant. Paralog knockout lines, generated through CRISPR/Cas9 transfection, exhibited uncompromised growth in PGIM, a modified medium with higher physiological nutrient levels; this finding is in marked contrast to a recently reported CLAG3 knockout parasite. CLAG2 and CLAG8 knockout lines exhibited compensatory increases in the transcription of the remaining clags and associated rhoph genes, yielding increased PSAC-mediated uptake for specific solutes. We also report on the distinct transport properties of these knockout lines. Selenium (Se) is an essential trace element for human physiological metabolism. The application of organic Se as a source to cultivate Se-rich plants for micronutrient supplementation has been receiving increasing attention. In our study, a bacterial strain named H1 was isolated from the soil in Heilongjiang Province, China, and under optimal culture conditions, the unit Se content could reach 3000 μg·g−1 and its 16S ribosomal DNA sequence seemed to be a new molecular record of an Enterobacter species. After the domestication of Se tolerance and Se-rich experiments, H1 can be used as a Se source for cultivation of Se-rich Auricularia auricula. The results showed that soluble protein, soluble sugar, free amino acid and vitamin C contents in Auricularia auricula were notably increased by 28.7%, 21.8%, 32.5% and 39.2% under the treatment of Se concentration of 0.24 mg·kg−1, respectively. Drought affects several plant physiological characteristics such as photosynthesis, carbon metabolism, and chlorophyll content, causing hormonal and nutritional imbalances and reducing nutrient uptake and transport, which inhibit growth and development. The use of bioinoculants based on plant growth-promoting microorganisms such as plant growth-promoting rhizobacteria (PGPR), yeasts, and arbuscular mycorrhizal fungi (AMF) has been proposed as an alternative to help plants tolerate drought. However, most studies have been based on the use of a single type of microorganism, while consortia studies have been scarcely performed. Therefore, the aim of this study was to evaluate different combinations of three PGPR, three AMF, and three yeasts with plant growth-promoting attributes to improve the biochemical, nutritional, and physiological behavior of strawberry plants growing under severe drought. The results showed that the growth and physiological attributes of the non-inoculated plants were significantly reduced by drought. In contrast, plants inoculated with the association of the fungus Claroideoglomus claroideum, the yeast Naganishia albida, and the rhizobacterium Burkholderia caledonica showed a stronger improvement in tolerance to drought. High biomass, relative water content, fruit number, photosynthetic rate, transpiration, stomatal conductance, quantum yield of photosystem II, N concentration, P concentration, K concentration, antioxidant activities, and chlorophyll contents were significantly improved in inoculated plants by up to 16.6%, 12.4%, 81.2%, 80%, 79.4%, 71.0%, 17.8%, 8.3%, 6.6%, 57.3%, 41%, and 22.5%, respectively, compared to stressed non-inoculated plants. Moreover, decreased malondialdehyde levels by up to 32% were registered. Our results demonstrate the feasibility of maximizing the effects of inoculation with beneficial rhizosphere microorganisms based on the prospect of more efficient combinations among different microbial groups, which is of interest to develop bioinoculants oriented to increase the growth of specific plant species in a global scenario of increasing drought stress. Human malignancies are one of the major health-related issues throughout the world and are anticipated to rise in the future. Despite huge investments made in anticancer drug development, limited success has been obtained and the average number of FDA approvals per year is declining. So, an increasing interest in drug repurposing exists. Metformin (MET) and aspirin (ASP) possess anticancer properties. This work aims to test the effect of these two drugs in combination on colorectal cancer (CRC) cells in vitro. The effects of MET and/or ASP on cell proliferation, viability, migratory ability, anchorage-independent growth ability (colony formation), and nutrient uptake were determined in two (HT-29 and Caco-2) human CRC cell lines. Individually, MET and ASP possessed antiproliferative, cytotoxic, and antimigratory effects and reduced colony formation in HT-29 cells (BRAF- and phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit α (PI3KCA)-mutant), although MET did not affect either 3H-deoxy-D-glucose or 14C-butyrate uptake and lactate production, and ASP caused only a small decrease in 14C-butyrate uptake. Moreover, in these cells, the combination of MET and ASP resulted in a tendency to an increase in the cytotoxic effect and in a potentiation of the inhibitory effect on colony formation, although no additive antiproliferative and antimigratory effects, and no effect on nutrient uptake and lactate production were observed. In contrast, MET and ASP, both individually and in combination, were almost devoid of effects on Caco-2 cells (BRAF- and PI3KCA-wild type). We suggest that inhibition of PI3K is the common mechanism involved in the anti-CRC effect of both MET, ASP and their combination and, therefore, that the combination of MET + ASP may especially benefit PI3KCA-mutant CRC cases, which currently have a poor prognostic. Nitrogen (N) is the most important nutrient in coffee, with a direct impact on productivity, quality, and sustainability. N uptake by the roots is dominated by ammonium (NH4+) and nitrates (NO3−), along with some organic forms at a lower proportion. From the perspective of mineral fertilizer, the most common N sources are urea, ammonium (AM), ammonium nitrates (AN), and nitrates; an appropriate understanding of the right balance between N forms in coffee nutrition would contribute to more sustainable coffee production through the better N management of this important crop. The aim of this research was to evaluate the influences of different NH4-N/NO3-N ratios in coffee from a physiological and agronomical perspective, and their interaction with soil water levels. Over a period of 5 years, three trials were conducted under controlled conditions in a greenhouse with different growing media (quartz sand) and organic soil, with and without water stress, while one trial was conducted under field conditions. N forms and water levels directly influence physiological responses in coffee, including photosynthesis (Ps), chlorophyll content, dry biomass accumulation (DW), nutrient uptake, and productivity. In all of the trials, the plants group in soils with N ratios of 50% NH4-N/50% NO3-N, and 25% NH4-N/75% NO3-N showed better responses to water stress, as well as a higher Ps, a higher chlorophyll content, a higher N and cation uptake, higher DW accumulation, and higher productivity. The soil pH was significantly influenced by the N forms: the higher the NO3−-N share, the lower the acidification level. The results allow us to conclude that the combination of 50% NH4-N/50% NO3-N and 25% NH4-N/75% NO3-N N forms in coffee improves the resistance capacity of the coffee to water stress, improves productivity, reduces the soil acidification level, and improves ion balance and nutrient uptake. Wheat (Triticum aestivum L.) is one of the most important cereal crops worldwide, and its production is challenged by global climate change and a shortage of resources. The root system plays a vital role in uptaking water and nutrients and sensing soil environmental signals, and it has great potential to improve the final yield and stress tolerance of wheat. In order to further explore the genes regulating root development, this study focused on qRS-6D, located on chromosome 6D and spanning from 462,701,391 to 465,068,943, which was significantly associated with the total root length, root volume, root surface, and root fresh weight in our previous GWAS analysis. Firstly, its genetic effects were validated using an F6 segregating population by comparing the root-related traits of homologous lines harboring the alternative haplotypes of this QTL. Then, the number of causal genes of this QTL was narrowed down to four with a transcriptome-wide association study. Additionally, qRS-6D has been demonstrated to have genetic effects on several yield- (kernel length, kernel width, and thousand-kernel weight) and plant structure-related traits (plant height, peduncle length, total tiller number, productive tiller number, flag leaf length, and flag leaf angle). Relatively, the frequency of the favorable haplotype increased with the wheat breeding practice. Legumes have a high demand for phosphorus (P) due to energetically costly biological nitrogen fixation, but they also have effective physiological and morphological strategies for P mobilization. To evaluate the inter- and intraspecific P efficiency of small-grain legumes supplied with different P recycling fertilisers, eight accessions each of alfalfa (Medicago sativa L.) and red clover (Trifolium pratense L.) were cultivated in two pot experiments under greenhouse conditions until the flowering stage. To substantiate the results, some accessions were used in both experiments. Five treatments (no P, triple-superphosphate (TSP), sewage sludge ash (SSA), biowaste compost (compost), and struvite) were considered P sources. In addition to plant P uptake, the soil P pools were analysed in detail. Red clover showed higher yields and nutrient uptakes compared to alfalfa, but intraspecific effects were marginal. The addition of P resulted only partly in an increase in yield, despite the low P content in the soil. While struvite application clearly enhanced the P uptake of the plants in both experiments, SSA application had no effect compared to the control. The same treatment effect occurs with the bio-available soil P contents, which were on average 72.6 mg kg−1 after struvite and 44.3 mg kg−1 after SSA addition. Struvite as a P source was especially effective when applied to red clover. Our study aligns with previous field results and underscores the high potential of P mobilization of small-grain legumes without pronounced inter- or intraspecific differences. A greenhouse experiment was realized to investigate the effects of plant-growth-promoting rhizobacteria (PGPR) and arbuscular mycorrhizal fungi (AMF) on soil quality and the growth, nutrient uptake, and physiological performance of Batavia lettuce (Lactuca sativa L. var. longifolia). For this purpose, six fertilization treatments were applied: (i) inorganic fertilization (I.F.—control, i.e., usual/conventional fertilization), (ii) I.F. + PGPR, (iii) I.F. + PGPR + AMF, (iv) manure (MAN), (v) MAN + PGPR, and (vi) MAN + PGPR + AMF. Soil fertility was influenced by the treatments, and soil respiration (CO2 flux) was significantly increased after applications of PGPR and AMF. Following MAN fertilization in particular, an approximately sixfold higher soil respiration value was recorded following the MAN + PGPR + AMF treatment compared to the control (I.F.). Root, leaf, and total biomass dry weights were significantly affected by the PGPR and AMF applications, mainly following the I.F. treatment. In contrast, K, Mg, and Fe uptake was significantly influenced by PGPR and AMF application following the MAN treatment. The SPAD value, performance index (PI), photosynthetic rate, and stomatal conductance were significantly higher in the I.F. + PGPR + AMF-treated plants compared to the control plants. Overall, these data prove the beneficial roles of PGPR and AMF in soil quality and fertility and the nutrient uptake and physiological performance of lettuce plants. Arbuscular mycorrhizal (AM) fungi can establish a mutualistic relationship with the roots of most terrestrial plants to increase plant nutrient uptake. The effects of potassium uptake and transport by AM symbiosis are much less reported compared to other nutrients. In this research, a heterologous yeast system was used to verify that the LbHAK has capacity for potassium uptake. The split-roots system implemented using seedlings of Lycium barbarum confirmed that R. irregularis locally induced LbHAK expression, which means that LbHAK is only expressed in mycorrhizal roots. Furthermore, the impacts of overexpression of LbHAK on the growth, nutrients and water uptake, and transport of mycorrhizal tobacco (inoculation with Rhizophagus irregularis) at 0.2 mM and 2 mM K conditions were assessed. The mycorrhizal tobacco growth and potassium accumulation were significantly enhanced through LbHAK overexpression in tobacco. In addition, overexpression of LbHAK substantially enhanced phosphorus content, while stimulating the expression of NtPT4, Rir-AQP1, and Rir-AQP2 in mycorrhizal tobacco. Moreover, LbHAK overexpression greatly promoted AM colonization. LbHAK has a potential role in facilitating potassium absorption through the mycorrhizal pathway, and overexpression of LbHAK in tobacco may promote the transport of potassium, phosphorus, and water from AM fungi to tobacco. These data imply the important roles played by the LbHAK in AM-fungi-induced potassium uptake in L. barbarum and in improving plant nutrients and AM colonization. [ABSTRACT FROM AUTHOR]

Soil salinity hampers durum wheat plant growth and development at various stages. The detrimental effects of salinity on plant cellular and physiological processes necessitate strategies to alleviate its negative impact and improve overall crop yield. This study investigates the efficacy of plant growth-promoting rhizobacteria (PGPR) bacteria inoculation in mitigating salinity stress on two durum wheat genotypes with contrasting degrees of salinity tolerance (Tamaroi, salt-sensitive and Line 5004, salt-tolerant) under greenhouse and field conditions. For this purpose, two halotolerant-PGPR strains, Pseudomonas jordanii strain G34 and Oceanobacillus jordanicus strain GSFE11, were utilized for the inoculation. For the greenhouse experiment, the two selected genotypes were subjected to salinity at the flag leaf stage with continuous irrigation with a Hoagland solution supplemented with 50 mM NaCl. Field experiments were conducted across two locations with contrasting salinity levels over two growing seasons. At the end of both experiments, various parameters including total weight, spike weight, grain weight, spike number, spikelet number, grains per spike and thousand kernel weight were measured. The halotolerant PGPRs, P. jordanii strain G34 and O. jordanicus strain GSFE11, proved effective in alleviating salinity-induced adverse effects and enhancing growth under greenhouse and field conditions. However, bacterial inoculation significantly improved growth in the salt-sensitive genotype and such effects were not observed in the tolerant genotype, emphasizing genotype-specific responses. Notably, inoculation with O. jordanicus increased Na+ and Ca2+ uptake in the salt-tolerant "Line 5004" without hindering growth, suggesting one of its potential mechanisms for salt tolerance. This research demonstrates the potential of halotolerant-PGPR inoculation in enhancing durum wheat production in saline environments, but also underscores the importance of understanding genotype-specific responses for tailored interventions. Using beneficial microorganisms along with sustainable strategies such as agronomic biofortification offers eco-friendly alternatives to combat climate change in ecosystems like dehesas. This study analyzes the combined effects of four wild Trichoderma spp. isolated from Extremadura, Spain (T. koningiopsis, two T. gamsii, and T. koningii, with negative and positive controls) and four Zn biofortification treatments (no Zn application; soil application of 5 mg of ZnSO4·7H2O per kg of soil, labeled soil Zn; two foliar applications of 5 mL 0.5% ZnSO4·7H2O, labeled foliar Zn; and soil + foliar combination, labeled SF) on Trifolium subterraneum performance. The combination of T. koningiopsis and T. gamsii with foliar Zn improved plant growth by up to 34.4%. Zinc accumulation was about 30% higher when T. gamsii and T. koningii were applied with SF, and their inoculation resulted in a 2.5-fold increase in ash. Trichoderma spp. affected nodulation differently; both T. gamsii inhibited nodulation by 24%, whereas neither T. koningiopsis nor T. koningii showed differences from the controls. These results highlight the potential of combining beneficial microorganisms with biofortification strategies to address nutrient deficiencies and improve agricultural sustainability. However, the complex interactions between both factors underscore the importance of strain selection and call for further research to optimize application strategies and elucidate underlying mechanisms. Water and soil salinization significantly reduce crop yields. Among the strategies developed to counteract salt stress, biostimulants can maintain crop productivity, reversing its impact. In this context, there is interest in finding new substances that could act as biostimulants. Recently, the biostimulatory potential of Lemna minor L. (duckweed) extracts has been shown. This work aimed to highlight whether an extract from duckweed (Lemna extract—LE) could protect maize grown in salinity, exploring the mechanisms induced to improve crop resistance. Plants were grown by applying two concentrations of NaCl (150 and 300 mM), and some physiological, morphological, and biochemical traits were studied in control and salt-stressed samples, treated or not with LE. Salinity decreased shoots, roots, pigment, and soluble protein. LE prompted ameliorative changes at the root level and increased photosynthetic pigment and soluble protein. Furthermore, concerning the oxidative impairment provoked by salt stress, LE enhanced the cellular redox state, contrasting H2O2 and MDA accumulation and positively affecting the activity of superoxide dismutase (SOD—EC 1.15.1.1) and catalase (CAT—EC 1.11.1.6). The assessment of some mineral nutrients showed that LE stimulated their acquisition, especially for the highest salt dosage, explaining some benefits found for the parameters investigated. The increasing cost of fossil-based energy sources has driven research in bio-based alternatives, such as perennial grasses for feedstock. The mid-Atlantic receives appreciable summer rainfall that may support a two-cut-per-year−1 harvest. At Virginia State University, a study on annual forage sorghum and two one-year stand perennials, miscanthus, and selected switchgrass ecotypes was carried out. The experimental design was a split-plot with harvest systems and feedstock grass species randomly assigned to the main and sub-plots, respectively. Only perennial grasses were assigned to the two-cut-per-year−1 system. The first cut occurred in early summer, and the second and single cut occurred after the frost-kill. Under the two-cut system, in 2022, the first-cut dry matter (DM) yield ranged from 8.9 Mg ha−1 in Blackwell to 14.7 Mg ha−1 in BoMaster. Additionally, except for BoMaster, the regrowth DM yields were within 10% of the first-cut DM yield. Under the one-cut system, the yield ranged from 10.8 Mg ha−1 in Blackwell to 23.2 Mg ha−1 in sorghum. Under the two-cut system, in 2023, miscanthus produced the greatest first-cut DM yield of 18.4 Mg ha−1, while other perennials averaged 10.1 Mg ha−1. Compared to the first cut, the hot and dry summer significantly reduced regrowth for all feedstock species, with the miscanthus DM yield dropping by 64%. While forage attributes differ among feedstock species, in general, both the first cut and regrowth showed greater crude protein and mineral elements, as well as lower ADF contents compared to a single cut following a killing freeze. Sorghum had better forage quality for the one-cut-per-year−1 feedstock material, and, along with the first cut and regrowth, it may have the potential for use as forage for maintenance energy in animal systems. For perennials, the two-cut-per-year−1 system removed the greatest quantities of nutrients during both years, with the first-cut harvest contributing about 65% of all removed N and K. Sorghum removed the greatest quantities of nutrients compared to the perennial under a one-cut-per-year−1 system. Therefore, while a two-cut-per-year−1 system can result in the greatest DM yields for dual-purpose use, its adoption calls for a critical analysis of economic benefits that considers feedstock bioenergy processing approaches, stand persistence, and fertilizer management strategies to address potential soil fertility depletion due to mineral element mining.
