Mediterranean Temporary Ponds (MTPs) represent a priority habitat according to Directive 92/43/EEC (Natura 2000 code: 3170*). These are very shallow water habitats only seasonally flooded, with a flora mainly composed of Mediterranean therophytic and geophytic species. Its extreme seasonality and small size make this habitat highly vulnerable and hard to manage. In recent Italian monitoring campaigns, the conservation status of MTP 3170* was considered inadequate. In Apulia, where the habitat is considered as "the most vulnerable type", 73 sites were censused, with a total coverage of about 10,000 m2. The present work refers to the monitoring for three years of a total of 16 habitat 3170* sites, with the aim of better describing faunal indicator species for this priority habitat. A total of 158 taxa of flora and 103 of fauna were identified from 54 floristic and 44 faunistic samplings in total, with a robust updating of the listed biodiversity. For the first time a group of faunal species is proposed as an indicator of the habitat MTP 3170*. The conservation status, assessed on the basis of structural and functional criteria, gave a satisfactory status for seven sites and an unsatisfactory one (variously rated as inadequate or bad) for nine. Fallopia japonica (FJ), an invasive plant species known for its rich bioactive compounds, has been used for centuries in traditional Chinese medicine. Despite its significant beekeeping potential, this aspect of FJ remains underexplored. This research aims to investigate the antimicrobial and antibiofilm properties of FJ plants and honey. Notably, this study is the first to identify individual phenolic compounds in both FJ plant tissues and FJ honey, highlighting resveratrol as a marker of FJ honey. The study tested inhibitory activity against seven bacterial strains: Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, Bacillus cereus, Salmonella enteritidis, and the yeast Candida albicans. Disk diffusion and microdilution methods were used to assess antimicrobial activity, while the crystal violet staining test evaluated antibiofilm activity. Results showed that FJ plant tissues and honey exhibited strong inhibition, particularly against Gram-negative bacterial strains. The most significant inhibition of biofilm formation, by both FJ plant tissues and honey, was observed against Staphylococcus aureus and Escherichia coli. A significant positive correlation was found between antimicrobial activity and individual polyphenols, especially resveratrol. The antibacterial and antibiofilm potential of FJ plant tissues and honey suggests promising applications in sustainable beekeeping. Medicinal and aromatic plants are a consistent component of the biodiversity heritage in numerous countries worldwide. Origanum dictamnus L. (Lamiaceae family), also known as Dittany, an endemic plant of the Greek island of Crete, has been widely used as traditional medicine since antiquity, all over Europe. The aim of the present review is to provide a thorough and detailed account of Dittany in antiquity, the plant's physical characteristics and ecology, and its cultivation methods, as well as its chemical components, biological properties, and pharmacological studies. The information is presented and analyzed in a critical manner. A total of 86 research studies were systematically reviewed based on the Preferred Reporting Items for Systematic Review and Meta-Analysis (PRISMA) guidelines. The findings indicate that Dittany is one of the most important medicinal and aromatic plants, with many uses not only in pharmacology but also in gastronomy. While a large body of literature exists regarding the application of essential oils, the number of publications concerning the plant's cultivation is rather small. Therefore, the main focus of this review is on the cultivation methods and the significance of cultivating and employing Dittany in Greece and the wider Mediterranean region in the future. Fallopia japonica (Japanese knotweed, Reynoutria japonica or Polygonum cuspidatum) is considered an extremely invasive plant worldwide and a bioindicator of heavy metals. Yet, its potential as a crop for honeybees is still underevaluated. This study employs atomic absorption spectrometry to quantitatively analyze the concentration of macro-elements, namely, calcium (Ca), potassium (K) and magnesium (Mg); micro-elements, such as copper (Cu), iron (Fe), manganese (Mn) and selenium (Se); and trace elements, i.e., cadmium (Cd), chromium (Cr), nickel (Ni) and lead (Pb) in different anatomic parts of Fallopia japonica (FJ) plants (roots, rhizomes, stems, leaves) and their traceability into honey. This research encompasses a thorough examination of samples collected from the northwestern and western part of Romania, providing insights into their elemental composition. The results showed that the level of trace elements decreases in terms of traceability in honey samples (Pb was not detected in any of the analyzed samples, while Cd had a minimum content 0.001 mg/kg), ensuring its quality and health safety for consumption. Geijera Schott is a plant genus of the Rutaceae Juss. (rue and citrus) family, comprising six species which are all native to Oceania. Of the plants belonging to this genus, the most significant species that has a customary use is Geijera parviflora, which was used by Indigenous Australians, primarily as a pain reliever. Herein, a comprehensive review of the literature published on the genus Geijera from 1930 to 2023 was conducted. This is the first review for this plant genus, and it highlights the chemical constituents reported to date, together with the range of pharmacological properties described from the various species and different parts of the plant. These properties include anti-inflammatory, anti-microbial, anti-parasitic, insect repellent, analgesic, neuroactive, and anti-cancer activities. Common sage is a versatile medicinal and aromatic plant that adapts well to unfavorable soil conditions. We determined the effect of Fluvisol and Chernozem soils on sage morphology, leaf yield, essential oil content, and composition under rainfed, temperate conditions for two consecutive years. Based on the plant height, diameter, shoot length, and yield, Chernozem soil was significantly superior to Fluvisol. However, S. officinalis is considered a drought-tolerant plant; our results confirmed the importance of the available water capacity of the different soil types since the yield on both soil types decreased significantly after an extreme drought event in 2022. The essential oil concentration (0.67–1.10 mL/100 g DM) was higher on the Fluvisol, but the total oil content was higher in the case of Chernozem (78.64 ± 20.50 mL/m2 DM). The ratio of essential oil components was also influenced by the soil types, but the highest amounts were analyzed for soils viridiflorol, α-humulen, and α-tujone. The essential oil yield showed a strong positive correlation with hydrocarbon sesquiterpenes (0.92) and oxygenated sesquiterpenes (0.95). Moreover, significant correlations were identified between the harvest time, seasonal precipitation patterns, soil and plant characteristics, and drug quality. On both soil types, the macronutrient content of the soils showed a strong positive correlation with the morphological parameters. The essential oil concentration was higher in the arid season of 2022 in the case of both soil types. The soil type and the seasonal effects can also change the ratio of essential oils. The threat to public health resulting from the emergence of antimicrobial resistance (AMR) is ever rising. One of the major bacterial pathogens at the forefront of this problem is methicillin-resistant Staphylococcus aureus, or MRSA, for which there is a great need to find alternative treatments. One of the most promising alternatives is endophytic fungi, which were shown to produce a vast array of bioactive compounds, including many novel antibacterial compounds. In this study, two endophytic Alternaria sp., EL 24 and EL 35, were identified from the leaves of Eremophila longifolia. Ethyl acetate (EtOAc) extracts of their culture filtrates were found to inhibit both methicillin-sensitive S. aureus ATCC 25923 and MRSA strains M173525 and M180920. The activity of each extract was shown to be greatly affected by the growth medium, with considerable reductions in minimum inhibitory concentrations (MICs) and minimum bactericidal concentrations (MBCs) observed when tested in tryptic soy broth with glucose (TSBG) compared with Mueller–Hinton broth (MHB). Both extracts displayed significant (p ≤ 0.05) antibiofilm activity against all three S. aureus strains, the greatest of which was that of EL 35, which reduced biofilm formation by M180920 by 72%, while that of EL 24 resulted in a 57% reduction against ATCC 25923. Both extracts also disrupted established biofilms, of which the most effective was EL 35, which reduced the M180920 biofilm by 64%, while EL 24 also performed best against M180920, reducing biofilm by 54%. Gas chromatography–mass spectrometry (GC-MS) analysis of the EL 24 EtOAc extract revealed five known compounds. This study highlights the promise of endophytic fungi from Australian plants as a potential source of substances effective against important bacterial pathogens. Further understanding of the responsible compounds and their mechanisms could lead to the development of treatments effective against MRSA, as well as novel biofilm-resistant biomedical materials, contributing towards reducing the burden of AMR. The current food system and food choices have resulted in the increased human use of natural resources such as water and soil, and have directly impacted the 'Global Syndemic'—climate change, obesity, and undernutrition. Revitalising Indigenous food systems and incorporating native plant-based foods into current food systems may have the potential to reduce diet-linked chronic diseases and environmental degradation, and are important steps toward Indigenous rights and self-determination. This study aims to identify and describe Aboriginal food practices and Australian native plant-based foods and their social, environmental, and economic impacts on sustainable food systems. A scoping review was conducted using the five-stage framework informed by Arksey and O'Malley. To describe the results, the framework for sustainable food systems from the Food and Agriculture Organization of the United Nations—FAO was used. Articles were included if they described the impacts of Aboriginal food practices on sustainable food systems, were confined to studies that were conducted in the Australian context, and included native Australian plant-based foods. A total of 57 studies were identified that met the inclusion criteria. The major social impacts incorporated the nutritional and health benefits of Australian native plant-based foods, such as antidiabetic properties, anticancer and antioxidant activities, and cultural identification, involving Aboriginal ecological knowledge and their connection to their country. Within the environmental impacts category, studies showed that Australian native plant-based foods have environmental stress tolerance and some ecosystem benefits. The main economic impacts discussed in the literature were the source of income for remote communities and the potential market for Australian native plant-based foods. This review demonstrates that Aboriginal food practices and Australian native plant-based foods can contribute to more sustainable food systems and diets and give more voice and visibility to Aboriginal knowledge and aspirations. Australia's endemic desert shrubs are commonly aromatic, with chemically diverse terpenes and phenylpropanoids in their headspace profiles. Species from the genus Eremophila (Scrophulariaceae ex. Myoporaceae) are the most common, with 215 recognised taxa and many more that have not yet been described, widely spread across the arid parts of the Australian continent. Over the years, our research team has collected multiple specimens as part of a survey to investigate the chemical diversity of the genus and create leads for further scientific enquiry. In the current study, the diversity of volatile compounds is studied using hydrodistilled essential oils and leaf solvent extracts from 30 taxa. Several rare terpenes and iridoids were detected in chemical profiles widely across the genus, and three previously undescribed sesquiterpenes were isolated and are assigned by 2D NMR—E-11(12)-dehydroisodendrolasin, Z-11-hydroxyisodendrolasin and 10-hydroxydihydro-α-humulene acetate. Multiple sampling from Eremophila longifolia, Eremophila arbuscular, Eremophila latrobei, Eremophila deserti, Eremophila sturtii, Eremophila oppositifolia and Eremophila alternifolia coneys that species in Eremophila are highly chemovariable. However, taxa are generally grouped according to the expression of (1) furanosesquiterpenes, (2) iridoids or oxides, (3) mixtures of 1 and 2, (4) phenylpropanoids, (5) non-furanoid terpenes, (6) mixtures of 4 and 5, and less commonly (7) mixtures of 1 and 5. Furthermore, GC–MS analysis of solvent-extracted leaves taken from cultivated specimens conveys that many heavier 'volatiles' with lower vapour pressure are not detected in hydrodistilled essential oils and have therefore been neglected in past chemical studies. Hence, our data reiterate that chemical studies of the genus Eremophila will continue to describe new metabolites and that taxon determination has limited predictive value for the chemical composition. The Australian rainforest is a rich source of medicinal plants that have evolved in the face of dramatic environmental challenges over a million years due to its prolonged geographical isolation from other continents. The rainforest consists of an inherent richness of plant secondary metabolites that are the most intense in the rainforest. The search for more potent and more bioavailable compounds from other plant sources is ongoing, and our short review will outline the pathways from the discovery of bioactive plants to the structural identification of active compounds, testing for potency, and then neuroprotection in a triculture system, and finally, the validation in an appropriate neuro-inflammatory mouse model, using some examples from our current research. We will focus on neuroinflammation as a potential treatment target for neurodegenerative diseases including multiple sclerosis (MS), Parkinson's (PD), and Alzheimer's disease (AD) for these plant-derived, anti-inflammatory molecules and highlight cytokine suppressive anti-inflammatory drugs (CSAIDs) as a better alternative to conventional nonsteroidal anti-inflammatory drugs (NSAIDs) to treat neuroinflammatory disorders. Natural constituents have been utilized to avoid humanity from various diseases, such as microbial infection and cancer, over several decades due to bioactive compounds. Myoporum serratum seeds extract (MSSE) was formulated via HPLC for flavonoid and phenolic analysis. Moreover, antimicrobial via well diffusion method, antioxidant via 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method, anticancer activities against HepG-2 cells (human hepatocellular cancer cell line), and MCF-7 cells (human breast cancer cell line), and molecular docking of the main detected flavonoid and phenolic compounds with the cancer cells were performed. The phenolic acids, including cinnamic acid (12.75 µg/mL), salicylic acid (7.14 µg/mL), and ferulic (0.97 µg/mL), while luteolin represents the main detected flavonoid with a concentration of 10.74 µg/mL, followed by apegenin 8.87 µg/mL were identified in MSSE. Staphylococcus aureus, Bacillus subtilis, Proteus vulgaris, and Candida albicans were inhibited by MSSE with 24.33, 26.33, 20.67, and 18.33 mm of inhibition zone, respectively. MSSE exhibited a low inhibition zone of 12.67 mm against Escherichia coli while showing no inhibitory activity against Aspergillus fumigatus. The values of MIC ranged from 26.58 to 136.33 µg/mL for all tested microorganisms. MBC/MIC index and cidal properties were attributed to MSSE for all tested microorganisms except E. coli. MSSE demonstrated anti-biofilm 81.25 and 50.45% of S. aureus and E. coli, respectively. IC50 of the antioxidant activity of MSSE was 120.11 µg/mL. HepG-2 and MCF-7 cell proliferation were inhibited with IC50 140.77 ± 3.86 µg/mL and 184.04 µg/mL, respectively. Species mapping methods play a central role in biogeographical questions, as they may generate a domino effect on further works based on species distribution. In light of the massive recent increase in the availability of online occurrence data, we highlight the strengths and limitations of the mapping methods most widely used to display the geographic distribution of plants, namely geographic range maps and occurrence record maps. We use the modern distribution of the genus Arbutus in western Eurasia, North Africa, and Macaronesia, for which no occurrence record map has been published yet, to discuss critical issues in data collection and representation. The occurrence record map of A. unedo, A. andrachne, A. canariensis, and A. pavarii shows how well this mapping method captures the details of peripheral and isolated stands as well as the variability of population density. A number of biogeographical issues are addressed by this approach, including the determination of the chorological centre of gravity in relation to historical dynamics, genetic patterns in relation to range porosity, and the autochthony status of marginal stands. These issues constitute the necessary foundation for additional palaeobotanical research and ecological modelling to investigate the past-to-future dynamics of Arbutus and other species of the Mediterranean–Atlantic area. Soybean is a worldwide crop that offers valuable proteins, fatty acids, and phytonutrients to humans but is always damaged by insect pests or pathogens. Plants have captured sophisticated defense mechanisms in resisting the attack of insects and pathogens. How to protect soybean in an environment- or human-friendly way or how to develop plant-based pest control is a hotpot. Herbivore-induced plant volatiles that are released by multiple plant species have been assessed in multi-systems against various insects, of which (E)-β-ocimene has been reported to show anti-insect function in a variety of plants, including soybean. However, the responsible gene in soybean is unknown, and its mechanism of synthesis and anti-insect properties lacks comprehensive assessment. In this study, (E)-β-ocimene was confirmed to be induced by Spodoptera litura treatment. A plastidic localized monoterpene synthase gene, designated as GmOCS, was identified to be responsible for the biosynthesis of (E)-β-ocimene through genome-wide gene family screening and in vitro and in vivo assays. Results from transgenic soybean and tobacco confirmed that (E)-β-ocimene catalyzed by GmOCS had pivotal roles in repelling a S. litura attack. This study advances the understanding of (E)-β-ocimene synthesis and its function in crops, as well as provides a good candidate for further anti-insect soybean improvement. [ABSTRACT FROM AUTHOR]

Myoporum sandwicense A. Gray (naio) is one of the characteristic trees of Hawaiian montane–subalpine mesic forests. In this study, lab-distilled oils of M. sandwicense leaves, wood, and twigs growing on the island of Hawaii, as well as industrially produced wood oils, were characterized by gas chromatography–mass spectrometry (GC-MS). The lab-distilled oils were screened for antimicrobial activity. M. sandwicense leaf essential oil was rich in β-caryophyllene (15.1%), α-humulene (12.8%), germacrene D (7.9%), bicyclogermacrene (12.5%), brigalow ketol (9.6%), and myoporone (16.8%), while the wood essential oils were dominated by α-bisabolol and trans-α-bisabolol oxide B. The sapwood oil was dominated by palmitic acid (35.5%), linoleic acid (19.7%), oleic acid (31.9%), and stearic acid (5.7%), whereas the oil from twigs was rich in tricosane (77.3%) and pentacosane (13.1%). M. sandwicense essential oils were screened for antimicrobial activity against a panel of potentially pathogenic bacteria and fungi. The leaf essential oil of M. sandwicense showed excellent antibacterial activity against S. pyogenes and antifungal activity against A. fumigatus. The wood essential oil showed notable activity against S. pyogenes, A. fumigatus, A. niger, and M. gypseum. The twig oil was remarkably active against mold species. Vietnam is extremely rich in biodiversity, with a remarkable range of habitats and more than 13,500 species of vascular plants recorded for the flora of Vietnam. This number represents about 3 to 5% of the world's diversity of vascular plants. Over the past 30 years, there were two important documents on the vascular plants of Vietnam published, An Illustrated Flora of Vietnam (IFV) and Checklist of Plant Species of Vietnam (CPSV). During the past half century, the advent of molecular phylogenetics has witnessed dramatic changes in the classifications of vascular plants, and some modern classification systems of vascular plants have been established, e.g., PPG I, GPG, and APG. However, the vascular plants of Vietnam have not yet been classified according to these modern classification systems. In this paper, we present the history of the classification of vascular plants in Vietnam, compare the circumscription of all families of vascular plants occurring within Vietnam in IFV, CPSV, and the modern classification systems when applicable, and summarize familial assignments of all controversial genera in the different classifications. Furthermore, we also arrange the 37 families of lycophytes and ferns occurring within Vietnam according to the latest classification system (PPG I) and the 8 families of gymnosperms according to the latest Christenhusz's system (GPG). The 246 families of angiosperms are arranged according to the fourth edition of the latest Angiosperm Phylogeny Group (APG IV). These results are the foundation stones and would be helpful for future research on the flora of Vietnam and the arrangement of plant collections in Vietnamese herbaria based on the updated classifications. Wound care is a global health issue with a financial burden of up to US $96.8 billion annually in the USA alone. Chronic non-healing wounds which show delayed and incomplete healing are especially problematic. Although there are more than 3000 dressing types in the wound management market, new developments in more efficient wound dressings will require innovative approaches such as embedding antibacterial additives into wound-dressing materials. The lack of novel antibacterial agents and the misuse of current antibiotics have caused an increase in antimicrobial resistance (AMR) which is estimated to cause 10 million deaths by 2050 worldwide. These ongoing challenges clearly indicate an urgent need for developing new antibacterial additives in wound dressings targeting microbial pathogens. Natural products and their derivatives have long been a significant source of pharmaceuticals against AMR. Scrutinising the data of newly approved drugs has identified plants as one of the biggest and most important sources in the development of novel antibacterial drugs. Some of the plant-based antibacterial additives, such as essential oils and plant extracts, have been previously used in wound dressings; however, there is another source of plant-derived antibacterial additives, i.e., those produced by symbiotic endophytic fungi, that show great potential in wound dressing applications. Endophytes represent a novel, natural, and sustainable source of bioactive compounds for therapeutic applications, including as efficient antibacterial additives for chronic wound dressings. This review examines and appraises recent developments in bioactive wound dressings that incorporate natural products as antibacterial agents as well as advances in endophyte research that show great potential in treating chronic wounds. Sages are medicinal and aromatic plants that constitute a large pool from which active compounds of great pharmaceutical potential can be derived, while at the same time, they also have ornamental value. The purpose of this study was to develop the micropropagation protocols of Salvia fruticosa, S. officinalis, S. ringens, S. tomentosa, and S. pomifera ssp. pomifera to facilitate their exploitation in the pharmaceutical and floriculture industries. In vitro cultures of S. ringens and S. pomifera ssp. pomifera was studied for the first time. Shoot tips and single node explants from in vitro seedlings were initially cultured on hormone free (Hf)-MS medium, followed by subcultures on MS medium supplemented with 6-benzyladenine (BA) for all species, as well as with zeatin (ZEA), kinetin (KIN), 6-(γ,γ-dimethylallyamino) purine (2iP), or meta-topolin (mT) for S. fruticosa and S. officinalis, at concentrations 0.0 to 3.2 mg L−1, in combination with 0.01 mg L−1 1-naphthaleneacetic acid (NAA). S. officinalis was the most efficient in shoot multiplication of all the studied species. The highest multiplication indices were found using 0.8 mg L−1 BA for S. fruticosa, 0.4 mg L−1 BA, or mT for S. officinalis, and lower than 0.8 mg L−1 BA for the other three species. Hyperhydricity was a problem at the multiplication stage, and was most pronounced in single node explants, increasing in proportion to cytokinin concentration. Microshoots rooted at high percentages (75–85%) on half-strength MS medium with 0.0 or 0.5 mg L−1 Indole-3-butyric acid (IBA), except for those of S. ringens, which rooted best at 1.0–2.0 mg L−1 IBA. Ex vitro acclimatization was highly successful (80–95%) on peat–perlite substrate (1:1 v/v). Thus, the present study resulted in efficient micropropagation protocols for five Mediterranean sage species native to Greece, which will facilitate breeding programs and the promotion of these species in the floriculture and pharmaceutical industries. The genus Eremophila (family Scrophulariaceae) consists of approximately 200 species that are widely distributed in the semi-arid and arid regions of Australia. Multiple Eremophila spp. are used as traditional medicines by the First Australians in the areas in which they grow. They are used for their antibacterial, antifungal, antiviral, antioxidant, anti-diabetic, anti-inflammatory, and cardiac properties. Many species of this genus are beneficial against several diseases and ailments. The antibacterial properties of the genus have been relatively well studied, with several important compounds identified and their mechanisms studied. In particular, Eremophila spp. are rich in terpenoids, and the antimicrobial bioactivities of many of these compounds have already been confirmed. The therapeutic properties of Eremophila spp. preparations and purified compounds have received substantially less attention, and much study is required to validate the traditional uses and to highlight species that warrant further investigation as drug leads. The aim of this study is to review and summarise the research into the medicinal properties, therapeutic mechanisms, and phytochemistry of Eremophila spp., with the aim of focussing future studies into the therapeutic potential of this important genus. The Asteraceae is a large family, rich in ornamental, economical, and medicinally valuable plants. The current study involves the analytical and pharmacological assessment of the methanolic extracts of three less investigated Asteraceae plants, namely Echinops ritro, Centaurea deflexa, and Tripleurospermum decipiens, obtained by three different extraction methodologies viz. maceration (MAC), ultrasound-assisted extraction (UAE), and homogenizer-assisted extraction (HAE). LC-MS-MS analysis of E. ritro, C. deflexa, and T. decipiens extracts led to the identification of ca. 29, 20, and 33 metabolites, respectively, belonging to flavonoids, phenolic acids, and fatty acids/amides. Although there were significant differences in the quantitative metabolite profiles in the extracts of E. ritro and T. decipiens based on the used extraction method, no significant variation was observed in the extracts of C. deflexa in the three implemented extraction techniques. The antioxidant activities of the nine extracts were assessed in vitro using six different assays viz. DPPH, ABTS, CUPRAC, FRAP, PDA, and metal chelation assay (MCA). The HAE/UAE extracts of E. ritro and the UAE/ MAC extracts of C. deflexa displayed the highest antioxidant activity in the DPPH assay, while the UAE extract of T. decipiens showed the strongest antioxidant activity in both the CUPRAC and MCA assays. The enzyme inhibitory activities of the nine extracts were studied in vitro on five different enzymes viz. tyrosinase, α-amylase, α-glucosidase, acetylcholinesterase (AChE), and butyrylcholinestrase (BChE), affecting various pathological diseases. Concerning C. deflexa, its MAC /UAE extracts showed the strongest inhibition on α-amylase, while its UAE/HAE extracts displayed strong inhibitory power on AChE. However, no significant difference was observed on their effects on tyrosinase or BChE. For T. decipiens, its UAE/HAE showed potent inhibition to α-glucosidase, MAC/ HAE significantly inhibited AChE and BChE, while UAE could strongly inhibit tyrosinase enzyme. For E. ritro, all extracts equally inhibited α-amylase and α-glucosidase, MAC/HAE strongly affected tyrosinase, HAE/MAC best inhibited BChE, while HAE inhibited AChE to a greater extent. Chemometric analysis using PCA plot was able to discriminate between the plant samples and between the implemented extraction modes. The in vitro enzyme inhibitory activities of the extracts were supported by in silico data, where metabolites, such as the lignan arctiin and the flavonoid vicenin-2, dominating the extract of C. deflexa, displayed strong binding to AChE. Similarly, chlorogenic and dicaffeoyl quinic acids, which are some of the major metabolites in the extracts of E. ritro and T. decipiens, bound with high affinity to α-glucosidase. The search for natural plant-based products as new pharmacological alternatives to treat various human pathologies has taken on great importance for researchers and research laboratories. In this context, research has intensified to extract and identify natural molecules endowed with biological effects. The objective of this study is to review the source and pharmacological properties of cirsimaritin. The identification and isolation of this flavonoid from various natural sources, including medicinal plants such as Artemisia judaica, Cirsium japonicum, Lithocarpus dealbatus, Microtea debilis, and Ocimum sanctum, has been carried out and verified using different spectral techniques. Biological effect investigations are carried out with a wide variety of experimental models in vitro and in vivo and laboratory techniques. The results of these research works showed the biological properties of cirsimaritin including anticancer, antimicrobial, antidiabetic, antiparasitic, antioxidant, and anti-inflammatory effects. The mechanisms involved in the multiple activities of this molecule are diverse and include sub-cellular, cellular, and molecular levels. Indeed, this bioactive induces anti-inflammatory and antiproliferative effects by inhibiting cell membrane receptors, interference with signaling pathways, and inhibiting transcriptional factors such as Nf-κB involved in cell promotion and proliferation. Calluna vulgaris, belonging to the Ericaceae family, is an invasive plant that has widely spread from Europe all across Asia, North America, Australia and New Zealand. Being able to survive in rigid soil and environmental conditions, it is nowadays considered to be of high nature-conservation value. Known for its nutritional and medicinal properties, C. vulgaris stands out for its varied physiochemical composition, spotlighting a wide range of biological activity. Among the most important bioactive compounds identified in C. vulgaris, the phenolic components found in different parts of this herbaceous plant are the main source of its diverse pro-health properties (antioxidant, anti-inflammatory, antimicrobial, chemoprotective, etc.). Nonetheless, this plant exhibits an excellent nectariferous potential for social insects such as honeybees; therefore, comparing the bioactive compounds observed in the plant and in the final product of the beehive, namely honey, will help us understand and find new insights into the health benefits provided by the consumption of C. vulgaris-related products. A flavone, chrysoeriol is synthetized in several plant species. It comes from several natural sources, especially medicinal plants. The identification and isolation of this compound has been carried out and verified by several research teams using different spectral methods. It seems that the concentration of this molecule is variable and fluctuating depending on the source, the part extracted, the region, and the methods of extraction and characterization. The aim of this paper is to highlight the in vitro and in vivo pharmacological properties of chrysoeriol and to provide insight into its pharmacokinetics. Anticancer, anti-inflammatory, antibacterial, antifungal, anti-osteoporosis, anti-insecticide, and neuroprotective actions have been shown in a number of studies on this chemical. Different mechanisms in theses pharmacological effects include subcellular, cellular, and molecular targets. In vivo pharmacokinetic analysis has proved the good stability of this molecule, showing its promising potential to prevent or treat diseases including cancer, diabetes, inflammation, osteoporosis, Parkinson's disease, and cardiovascular diseases. Acute bovine liver disease (ABLD) is a hepatic disease affecting cattle sporadically in southern Australia, characterised histologically by striking periportal hepatocellular necrosis. The cause of ABLD is unknown; however, the seasonality and acute presentation of outbreaks suggest mycotoxin involvement. We described the geographical and seasonal occurrence of ABLD reports from 2010 to 2020 in Victoria, Australia, and explored potential weather triggers preceding 26 outbreaks occurring across 23 properties using a case-crossover design. Outbreaks occurred most frequently in autumn/early winter and in herds located along the southern coastal plain of Victoria, and occasionally within the low-lying regions of the Great Dividing Range. Lactating adult dairy cattle represented the most reported cases. We observed a significant association between an increase in average daily dewpoint in the 15 days preceding an ABLD outbreak, suggesting that dew formation may be a key determinant for this disease. Plant secondary metabolites (PSMs) are vital for human health and constitute the skeletal framework of many pharmaceutical drugs. Indeed, more than 25% of the existing drugs belong to PSMs. One of the continuing challenges for drug discovery and pharmaceutical industries is gaining access to natural products, including medicinal plants. This bottleneck is heightened for endangered species prohibited for large sample collection, even if they show biological hits. While cultivating the pharmaceutically interesting plant species may be a solution, it is not always possible to grow the organism outside its natural habitat. Plants affected by abiotic stress present a potential alternative source for drug discovery. In order to overcome abiotic environmental stressors, plants may mount a defense response by producing a diversity of PSMs to avoid cells and tissue damage. Plants either synthesize new chemicals or increase the concentration (in most instances) of existing chemicals, including the prominent bioactive lead compounds morphine, camptothecin, catharanthine, epicatechin-3-gallate (EGCG), quercetin, resveratrol, and kaempferol. Most PSMs produced under various abiotic stress conditions are plant defense chemicals and are functionally anti-inflammatory and antioxidative. The major PSM groups are terpenoids, followed by alkaloids and phenolic compounds. We have searched the literature on plants affected by abiotic stress (primarily studied in the simulated growth conditions) and their PSMs (including pharmacological activities) from PubMed, Scopus, MEDLINE Ovid, Google Scholar, Databases, and journal websites. We used search keywords: “stress-affected plants,” “plant secondary metabolites, “abiotic stress,” “climatic influence,” “pharmacological activities,” “bioactive compounds,” “drug discovery,” and “medicinal plants” and retrieved published literature between 1973 to 2021. This review provides an overview of variation in bioactive phytochemical production in plants under various abiotic stress and their potential in the biodiscovery of therapeutic drugs. Investigation of the single plant source bee glue type originating from Southern Australia resulted in the isolation and structure elucidation of major serrulatane diterpenes, novel 7,8,18-trihydroxyserrulat-14-ene (1), along with its oxidized product, 5,18-epoxyserrulat-14-en-7,8-dione (3) and known (18RS)-5,18-epoxyserrulat-14-en-8,18-diol (2). Exploration into the botanical origin revealed Myoporum insulare R. Br, as the plant source of the bee glue materials. Myoporum bontioides is a traditional medicinal plant in Asia with various biological activities, including anti-inflammatory and anti-bacterial characteristics. To identify the bioactive constituents from M. bontioides, a newly-identified flavone, 3,4'-dimethoxy-3',5,7-trihydroxyflavone (compound 1), along with eight known compounds, were investigated in human MCF-7 breast cancer, SCC4 oral cancer, and THP-1 monocytic leukemia cells. Among these compounds, compound 1 exhibited the strongest antiproliferative activity with half-maximal inhibitory concentration (IC50) values ranging from 3.3 μM (MCF-7) to 8.6 μM (SCC4). Flow cytometric analysis indicated that compound 1 induced G2/M cell cycle arrest in MCF-7 cells. Mechanistic evidence suggests that the G2/M arrest could be attributable to compound 1's modulatory effects on the phosphorylation and expression of numerous key signaling effectors, including cell division cycle 2 (CDC2), CDC25C, and p53. Notably, compound 1 downregulated the expression of histone deacetylase 2 (HDAC2) and HDAC4, leading to increased histone H3 acetylation and p21 upregulation. The formyl peptide receptors (FPRs) are G protein-coupled receptors that transduce chemotactic signals in phagocytes and mediate host-defense as well as inflammatory responses including cell adhesion, directed migration, granule release and superoxide production. In recent years, the cellular distribution and biological functions of FPRs have expanded to include additional roles in homeostasis of organ functions and modulation of inflammation. In a prototype, FPRs recognize peptides containing N-formylated methionine such as those produced in bacteria and mitochondria, thereby serving as pattern recognition receptors. The repertoire of FPR ligands, however, has expanded rapidly to include not only N-formyl peptides from microbes but also non-formyl peptides of microbial and host origins, synthetic small molecules and an eicosanoid. How these chemically diverse ligands are recognized by the three human FPRs (FPR1, FPR2 and FPR3) and their murine equivalents is largely unclear. In the absence of crystal structures for the FPRs, site-directed mutagenesis, computer-aided ligand docking and structural simulation have led to the identification of amino acids within FPR1 and FPR2 that interact with several formyl peptides. Photosensitivity in animals is defined as a severe dermatitis that results from a heightened reactivity of skin cells and associated dermal tissues upon their exposure to sunlight, following ingestion or contact with UV reactive secondary plant products. Photosensitivity occurs in animal cells as a reaction that is mediated by a light absorbing molecule, specifically in this case a plant-produced metabolite that is heterocyclic or polyphenolic. In sensitive animals, this reaction is most severe in non-pigmented skin which has the least protection from UV or visible light exposure. Photosensitization in a biological system such as the epidermis is an oxidative or other chemical change in a molecule in response to light-induced excitation of endogenous or exogenously-delivered molecules within the tissue. Photo-oxidation can also occur in the plant itself, resulting in the generation of reactive oxygen species, free radical damage and eventual DNA degradation. Similar cellular changes occur in affected herbivores and are associated with an accumulation of photodynamic molecules in the affected dermal tissues or circulatory system of the herbivore. Recent advances in our ability to identify and detect secondary products at trace levels in the plant and surrounding environment, or in organisms that ingest plants, have provided additional evidence for the role of secondary metabolites in photosensitization of grazing herbivores. This review outlines the role of unique secondary products produced by higher plants in the animal photosensitization process, describes their chemistry and localization in the plant as well as impacts of the environment upon their production, discusses their direct and indirect effects on associated animal systems and presents several examples of well-characterized plant photosensitization in animal systems. In this comprehensive commentary, Australian essential oils and their components are listed and discussed in the context of their value to industry and aesthetics. The historic and cultural significance of endemic essential oils is explained. Several promising candidates are identified that have commercial potential and will enter the marketplace in the not-too-distant future. This text elaborates on the current progress in research, and explains the up-to-date view of 'bioactive,' with reference to insect repellence, antimicrobial activity, anti-inflammatory activity, and potential toxicity. The concept of chemotypes and chemophenetics is explained in detail to justify why chemically variable species in Australia require standardisation practices to ensure reproducibility of their derived natural products: standardisation practice includes cultivar development and authentication protocols. Thereafter, some of the more significant essential oils are defined and some background information provided. This review concludes with a comprehensive table of aromatic species that were studied by Joseph Brophy over the last 30 years, thereby providing the most comprehensive overview available, on the chemistry of Australian essential oil yielding species. Aiming to obtain Salvia hybrids with ornamental value and high drought resistance, for xeriscaping, crossbreeding was made with Greek Salvia species. S. fruticosa and S. officinalis when used as seed parent were successfully crossed with S. pomifera ssp. pomifera, S. ringens and S. tomentosa, while when used as pollen parent it only succeeded between S. fruticosa and S. tomentosa. The growth of S. fruticosa and the four hybrids, S. officinalis × S. pomifera, S. officinalis × S. tomentosa, S. officinalis × S. ringens and S. fruticosa × S. ringens, selected for their ornamental traits, was evaluated under limited irrigation and modification of the substrate with attapulgite clay. The hybrids S. officinalis × S. ringens and S. officinalis × S. tomentosa developed a compact plant shape and most lateral shoots, desirable characteristics for potted plants and xeriscaping. All hybrids, especially S. officinalis × S. pomifera and S. officinalis × S. tomentosa, survived water stress better than S. fruticosa. Modification of the substrate with attapulgite, under limited irrigation, caused a decrease in the above ground/root biomass ratio in some hybrids and in S. fruticosa increased the dry weight of the root indicating increased drought resistance. Despite its importance as a component of urban green spaces, as far as we are aware no study has focused on plant diversity in urban green corridors (GCs). Therefore, this study aimed at: (i) characterizing tree and shrub communities in Lisbon's GCs and (ii) assessing whether GCs' users value trees and shrubs. We counted Lisbon's GCs users in the same places where we assessed the tree and shrub community. Along the nine GCs, we observed trees and shrubs belonging to 70 species, distributed across 35 families with most (≥50%) species and plants being trees, exotic, pollinated by insects, with fruit dispersion by animals, evergreen leaves, and producing dry fruits. Most GCs had a similar number of users (20–30 users h−1 survey−1) except for those of Central and Ribeirinho, which were more frequented (60 and 100 users h−1 survey−1, respectively). Most users (≥50%) were adults, walking accompanied, and performing leisure activities. Finally, the number of users was shown to be influenced by: (i) tree and shrub relative abundance, confirming that users preferred greener corridors; and (ii) function(s), showing that users preferred the most multifunctional GCs (i.e., GCs fulfilling ecological, cultural, and recreational functions). Multidrug resistance (MDR) is a major challenge in cancer treatment, and the breast cancer resistance protein (BCRP) is an important target in the search for new MDR-reversing drugs. With the aim of discovering new potential BCRP inhibitors, the crude extract of leaves of Eremophila galeata, a plant endemic to Australia, was investigated for inhibitory activity of parental (HT29par) as well as BCRP-overexpressing HT29 colon cancer cells resistant to the chemotherapeutic SN-38 (i.e., HT29SN38 cells). This identified a fraction, eluted with 40% acetonitrile on a solid-phase extraction column, which showed weak growth-inhibitory activity on HT29SN38 cells when administered alone, but exhibited concentration-dependent growth inhibition when administered in combination with SN-38. The major constituent in this fraction was isolated and found to be 5,3′,5′-trihydroxy-3,6,7,4′-tetramethoxyflavone (2), which at a concentration of 25 μg/mL potentiated the growth-inhibitory activity of SN-38 to a degree comparable to that of the known BCRP inhibitor Ko143 at 1 μM. A dye accumulation experiment suggested that 2 inhibits BCRP, and docking studies showed that 2 binds to the same BCRP site as SN-38. These results indicate that 2 acts synergistically with SN-38, with 2 being a BCRP efflux pump inhibitor while SN-38 inhibits topoisomerase-1. [ABSTRACT FROM AUTHOR]

Rhododendron (Ericaceae) extracts contain flavonoids, chromones, terpenoids, steroids, and essential oils and are used in traditional ethnobotanical medicine. However, little is known about the immunomodulatory activity of essential oils isolated from these plants. Thus, we isolated essential oils from the flowers and leaves of R. albiflorum (cascade azalea) and analyzed their chemical composition and innate immunomodulatory activity. Compositional analysis of flower (REOFl) versus leaf (REOLv) essential oils revealed significant differences. REOFl was comprised mainly of monoterpenes (92%), whereas sesquiterpenes were found in relatively low amounts. In contrast, REOLv was primarily composed of sesquiterpenes (90.9%), with a small number of monoterpenes. REOLv and its primary sesquiterpenes (viridiflorol, spathulenol, curzerene, and germacrone) induced intracellular Ca2+ mobilization in human neutrophils, C20 microglial cells, and HL60 cells transfected with N-formyl peptide receptor 1 (FPR1) or FPR2. On the other hand, pretreatment with these essential oils or component compounds inhibited agonist-induced Ca2+ mobilization and chemotaxis in human neutrophils and agonist-induced Ca2+ mobilization in microglial cells and FPR-transfected HL60 cells, indicating that the direct effect of these compounds on [Ca2+]i desensitized the cells to subsequent agonist activation. Reverse pharmacophore mapping suggested several potential kinase targets for these compounds; however, these targets were not supported by kinase binding assays. Our results provide a cellular and molecular basis to explain at least part of the beneficial immunotherapeutic properties of the R. albiflorum essential oils and suggest that essential oils from leaves of this plant may be effective in modulating some innate immune responses, possibly by inhibition of neutrophil migration. Simple Summary: Over 800 million hectares of arable lands are affected by salinity worldwide. All crops cannot grow in saline-alkali soils due to high salt content. However, halophytes are a special category of plants that grow in saline soils. Halophytes have potential economic value as grain, vegetable, fruit, medicine, animal feed, biofuel feedstocks, and in greening and coastal protection. These also provide possible directions for the development of saline-alkali land. In addition, we are also concerned about the coordinated and sustainable development of the protection and utilization of halophytes. Over 800 million hectares of arable lands are affected by salinity in the world. In China, saline-alkali soils account for 25% of farmland and are underutilized. One sustainable strategy to make better use of saline land is to plant halophytes, salt-tolerant plants that can survive and complete their life cycle in media containing more than 200 mM NaCl. Halophytes have potential economic value as grain, vegetable, fruit, medicine, animal feed, and biofuel feedstocks, and in greening and coastal protection. Therefore, the cultivation and protection of halophytes is very important. The current study investigates the content of sugars, organic acids, phenolic acids and flavonoids, as well as antioxidant and antimicrobial activity of Balkan-endemic Micromeria frivaldszkyana. Glucose was the most abundant sugar in the plant (2.77%), followed by fructose (1.18%) and galactose (0.82%). Eight organic acids were detected with quinic acid being in the highest content—556.3 mg/100 g DW. From the individual phenolic acids, rosmarinic acid was found in the most significant amounts (2040.1 ± 1.97 mg/100 g) and hesperidin was the major representative of flavonoids with content 131.2 ± 5.6 mg/100 g DW. The antioxidant activity of the plant was studied by six methods: 2,2′-diphenylpicrylhydrazyl (DPPH)—286.4 ± 10.43 mM TE/g, 2,2′azinobis (3)-ethylbenzthiazoline-6-sulfonic acid (ABTS)—358.4 ± 10.4 mM TE/g, ferric reducing antioxidant power (FRAP)—388.0 ± 32.4 mM TE/g, cupric reducing antioxidant capacity (CUPRAC)—905.6 ± 19.2 mM TE/g, Oxygen Radical Absorbance Capacity (ORAC)—3250.5 ± 208.1 µmol TE/g and Hydroxyl Radical Averting Capacity (HORAC)—306.1 ± 23.5 µmol GAE/g. In vitro antimicrobial activity against nine microorganism was evaluated but the extract displayed antimicrobial activity only against Listeria monocytogenes ATCC 19111 with inhibition zone diameter 9 mm and minimal inhibitory concentration (MIC) 10 mg/mL. Plant- and fungus-derived hepatotoxins are a major cause of disease and production losses in ruminants in Australia and around the world. Many are well studied and described in the literature; however, this is not the case for a number of hepatotoxicities with economic and animal welfare impacts, such as acute bovine liver disease (ABLD), brassica-associated liver disease (BALD) and Trema tomentosa, Argentipallium blandowskianum and Lythrum hyssopifolia toxicity. Additionally, significant overlap in the clinical presentation and pathology of these conditions can present a diagnostic challenge for veterinarians. This review summarizes the current and most recently published knowledge of common plant- and fungus-associated hepatotoxins affecting cattle in Australia, with a focus on the mechanisms of toxicity and distinguishing diagnostic features. Consolidation of the current understanding of hepatotoxic mechanisms in cattle provides insight into the potential mechanisms of lesser-known toxins, including cellular and subcellular targets and potential metabolic pathways. In the absence of specific etiological investigations, the study of epidemiological, clinical and pathological features of hepatotoxicity provides valuable insights into potential toxic mechanisms and is integral for the successful diagnosis and management of these conditions. Knowledge on alien species is needed nowadays to protect natural habitats and prevent ecological damage. The presence of new alien plant species in Italy is increasing every day. Calabria, its southernmost region, is not yet well known with regard to this aspect. Thanks to fieldwork, sampling, and observing many exotic plants in Calabria, here, we report new data on 34 alien taxa. In particular, we found three new taxa for Europe (Cascabela thevetia, Ipomoea setosa subsp. pavonii, and Tecoma stans), three new for Italy (Brugmansia aurea, Narcissus 'Cotinga', and Narcissus 'Erlicheer'), one new one for the Italian Peninsula (Luffa aegyptiaca), and 21 new taxa for Calabria (Allium cepa, Asparagus setaceus, Bassia scoparia, Beta vulgaris subsp. vulgaris, Bidens formosa, Casuarina equisetifolia, Cedrus atlantica, Chlorophytum comosum, Cucurbita maxima subsp. maxima, Dolichandra unguis-cati, Fagopyrum esculentum, Freesia alba, Juglans regia, Kalanchoë delagoënsis, Passiflora caerulea, Portulaca grandiflora, Prunus armeniaca, Prunus dulcis, Solanum tuberosum, Tradescantia sillamontana, and Washingtonia filifera). Furthermore, we provide the first geolocalized record of Araujia sericifera, the confirmation of Oxalis stricta, and propose a change of status for four taxa (Cenchrus setaceus, Salpichroa origanifolia, Sesbania punicea, and Nothoscordum gracile) for Calabria. The updated knowledge on the presence of new alien species in Calabria, in Italy and in Europe could allow for the prevention of other new entries and to eliminate this potential ecological threat to natural habitats. Nontuberculous mycobacteria (NTM) are the causative agent of severe chronic pulmonary diseases and is accountable for post-traumatic wound infections, lymphadenitis, endometritis, cutaneous, eye infections and disseminated diseases. These infections are extremely challenging to treat due to multidrug resistance, which encompasses the classical and existing antituberculosis agents. Hence, current studies are aimed to appraise the antimycobacterial activity of flavonoids against NTM, their capacity to synergize with pharmacological agents and their ability to block virulence. Flavonoids have potential antimycobacterial effects at minor quantities by themselves or in synergistic combinations. A cocktail of flavonoids used with existing antimycobacterial agents is a strategy to lessen side effects. The present review focuses on recent studies on naturally occurring flavonoids and their antimycobacterial effects, underlying mechanisms and synergistic effects in a cocktail with traditional agents. Correct color measurement by contact-type color measuring devices requires that the sample surface fully covers the head of the device, so their use on small samples remains a challenge. Here, we propose to use cardboard adaptors on the two aperture masks (3 and 8 mm diameter measuring area) of a broadly used portable spectrophotometer. Adaptors in black and white to reduce the measuring area by 50% and 70% were applied in this study. Representatives of the family Campanulaceae have been used to test the methodology, given the occurrence of small leaves. Our results show that, following colorimetric criteria, the only setting providing indistinguishable colors according to the perception of the human eye is the use of a 50%-reducing adaptor on the 3-mm aperture. In addition, statistical analysis suggests the use of the white adaptor. Our contribution offers a sound measurement technique to gather ecological information from the color of leaves, petals, and other small samples. [ABSTRACT FROM AUTHOR]

The leaves of Pyrola rotundifolia L. were extracted in the mixed solvent of methanol/acetone/water (2:2:1, v/v/v) and investigated for their phytochemical analysis and biological activity. Total phenolic and flavonoid contents were determined spectrophotometrically. A high content of phenols (208.35 mg GAE/g of dry extract), flavonoids (38.90 mg QE/g of dry extract) and gallotannins (722.91 GAE/g of dry extract) was obtained. Ultra-high performance liquid chromatography diode array detector tandem mass spectrometry (UHPLC–DAD–MS) allowed for the detection of 23 major peaks at 254 nm. The extract was analyzed for its antioxidant capacity using 2,2-diphenyl-1-picryl-hydrazyl (DPPH•) and 2,2′-azinobis[3-ethylbenzthiazoline]-6-sulfonic acid (ABTS•+) radical scavenging, metal chelating power and β-carotene-linoleic acid bleaching assays. The examined extract showed moderate radical scavenging and chelating activity, and good inhibiting ability of linoleic acid oxidation (EC50 = 0.05 mg/mL) in comparison to standards. The cytotoxic effect in increasing concentration on five types of leukemic cell lines was also investigated using trypan blue vital staining. It was found that the analyzed extract induced the apoptosis of all the tested cell lines. Our findings suggest that the leaves of P. rotundifolia are a source of valuable compounds providing protection against oxidative damage, hence their use in traditional medicine is justified. Despite the importance for scientific and conservation purposes, the knowledge of the Italian territory is far from exhaustive. New chorological data for 87 vascular taxa regarding the central-southern part of Italy and its two main islands (Sicilia and Sardegna) are presented. Among these taxa, Epilobium nummularifolium, Metrosideros excelsa, and Salvinia minima are recorded as casual aliens for the first time in Europe (excluding Azores and Madeira for M. excelsa), while Cyclamen balearicum and Polygala rupestris are reported for the first time and confirmed for Italian native flora, respectively. Furthermore, several taxa are new or confirmed at regional level. Thymus marschallianus Willd. is a Lamiaceae species spread in a large variety of habitats worldwide. The aim of the present research was to analyse two different samples belonging to this species, one obtained from the spontaneous flora and one from culture. The total polyphenols, flavonoids, and phenolic acid contents were spectrophotometrically determined. Qualitative and quantitative analysis of polyphenols was performed by an HPLC-DAD-ESI (+)-MS method. For the antibacterial assay, the well-diffusion and the broth microdilution methods were used. Analysis of polyphenols revealed for both samples the presence of flavonoids like luteolin, quercetin, apigenin and their derivatives, but also of rosmarinic acid and methyl-rosmarinate. Differences regarding the amount of these compounds were emphasized. Significantly larger amounts of flavonoids were found for the sample harvested in the spontaneous flora, while for the rosmarinic acid, larger amounts were found for the cultured sample. Both samples displayed promising antibacterial activity, particularly towards Gram positive organisms. T. marschallianus represents, therefore, a rich source of polyphenolic compounds that prove its promising potential as a medicinal species. [ABSTRACT FROM AUTHOR]

Plants in the Australian genus Eremophila (Scrophulariaceae) have attracted considerable recent attention for their antimicrobial compounds, which possess a wide range of chemical structures. As they are typically associated with the oily-waxy resin layer covering leaves and green branchlets, and Eremophila lucida is prominent among the species containing a pronounced sticky resin layer, this species was considered of interest for assessing its antibacterial constituents. The n-hexane fraction of the crude acetone extract of the leaves exhibited antibacterial activity against Staphylococcus aureus. Isolation led to the known compounds cembratriene, (3Z, 7E, 11Z)-15-hydroxycembra-3,7,11-trien-19-oic acid (1), the sesquiterpenoid, farnesal (2) and the viscidane diterpenoid, 5α-hydroxyviscida-3,14-dien-20-oic acid (3). Plant metabolites that have shown activity against bacteria and/or environmental fungi represent valuable leads for the identification and development of novel drugs against clinically important human pathogenic fungi. Plants from the genus Eremophila were highly valued in traditional Australian Aboriginal medicinal practices, and E. alternifolia was the most prized among them. As antibacterial activity of extracts from E. alternifolia has been documented, this study addresses the question whether there is also activity against infectious fungal human pathogens. Compounds from leaf-extracts were purified and identified by 1- and 2-D NMR. These were then tested by disk diffusion and broth microdilution assays against ten clinically and environmentally relevant yeast and mould species. The most potent activity was observed with the diterpene compound, 8,19-dihydroxyserrulat-14-ene against Cryptococcus gattii and Cryptococcus neoformans, with minimum inhibition concentrations (MIC) comparable to those of Amphotericin B. This compound also exhibited activity against six Candida species. Combined with previous studies showing an antibacterial effect, this finding could explain a broad antimicrobial effect from Eremophila extracts in their traditional medicinal usage. The discovery of potent antifungal compounds from Eremophila extracts is a promising development in the search for desperately needed antifungal compounds particularly for Cryptococcus infections. Utilizing bioassay- and TLC-guided column chromatography, fifteen secondary metabolites from Populus alba and eight compounds from Salix subserrata were isolated, including a novel plant metabolite salicyl ether and characterized using ultralviolet light (UV) absorbance, mass spectrometry (MS), 1H-, 13C-NMR (nuclear magnetic resonance), heteronuclear single quantum coherence spectroscopy (HSQC) and heteronuclear multiple bond correlation (HMBC). The extracts, their sub-fractions and the isolated compounds exhibited promising antioxidant activities in vitro in DPPH and FRAP assays. Also, the extracts of P. alba leaf (PL), shoots (PS), and S. subserrata leaf (SL) demonstrated substantial antioxidant activities in vivo in the multicellular model organism Caenorhabditis elegans. For the first time, the isolated secondary metabolites, aromadendrin, tremuloidin, salicin, isorhamnetin-3-O-β-d-rutinoside, gallocatechin, triandrin, and chrysoeriol-7-O-glucuronide were investigated. They exhibited substantial antioxidant activities in vivo. Salicin, isorhamnetin-3-O-β-d-rutinoside and gallocatechin, in particular, protected the worms against a lethal dose of the pro-oxidant juglone (80 µM), decreased the endogenous reactive oxygen species (ROS) level to 45.34%, 47.31%, 68.09% and reduced juglone- induced hsp-16.2::GFP (green fluorescence protein) expression to 79.62%, 70.17%, 26.77%, respectively. However, only gallocatechin induced higher levels of sod-3 expression. The striking rise of methicillin-resistant Staphylococcus aureus (MRSA) infections has become a serious threat to public health worldwide. In an effort to search for new anti-MRSA agents from natural products, a bioassay-guided phytochemical study was conducted on the semi-mangrove plant Myoporum bontioides A. Gray, which led to the isolation of two new sesquiterpene alkaloids (1 and 2) and six known furanosesquiterpenes (3–8). Their structures were elucidated on the basis of extensive analysis of their 1D, 2D NMR and mass spectroscopic data. These two new alkaloids (1 and 2) displayed potent anti-MRSA activity with MIC value of 6.25 μg/mL.
