Electroporation (EP) stands out as a promising non-viral plasmid delivery strategy, although achieving optimal transfection efficiency in vivo remains a challenge. A noteworthy advancement in the field of in vivo EP is the application of hyaluronidase, an enzyme with the capacity to degrade hyaluronic acid in the extracellular matrix, which thereby enhances DNA transfer efficiency by 2- to 3-fold. This paper focuses on elucidating the mechanism of hyaluronidase's impact on transfection efficiency. We demonstrate that hyaluronidase promotes a more uniform distribution of plasmid DNA (pDNA) within skeletal muscle. Additionally, our study investigates the effect of the timing of hyaluronidase pretreatment on EP efficiency by including time intervals of 0, 5, and 30 min between hyaluronidase treatment and the application of pulses. Serum levels of the pDNA-encoded transgene reveal a minimal influence of the hyaluronidase pretreatment time on the final serum protein levels following delivery in both mice and rabbit models. Leveraging bioimpedance measurements, we capture morphological changes in muscle induced by hyaluronidase treatment, which result in a varied pDNA distribution. Subsequently, these findings are employed to optimize EP electrical parameters following hyaluronidase treatment in animal models. This paper offers novel insights into the potential of hyaluronidase in enhancing the effectiveness of in vivo EP, as well as guides optimized electroporation strategies following hyaluronidase use. Gene therapy and its role in the medical field have evolved drastically in recent decades. Studies aim to define DNA-based medicine as well as encourage innovation and the further development of novel approaches. Gene therapy has been established as an alternative approach to treat a variety of diseases. Its range of mechanistic applicability is wide; gene therapy has the capacity to address the symptoms of disease, the body's ability to fight disease, and in some cases has the ability to cure disease, making it a more attractive intervention than some traditional approaches to treatment (i.e., medicine and surgery). Such versatility also suggests gene therapy has the potential to address a greater number of indications than conventional treatments. Many DNA-based therapies have shown promise in clinical trials, and several have been approved for use in humans. Whereas current treatment regimens for chronic disease often require frequent dosing, DNA-based therapies can produce robust and durable expression of therapeutic genes with fewer treatments. This benefit encourages the application of DNA-based gene therapy to manage chronic diseases, an area where improving efficiency of current treatments is urgent. Here, we provide an overview of two DNA-based gene therapies as well as their delivery methods: adeno associated virus (AAV)-based gene therapy and plasmid DNA (pDNA)-based gene therapy. We will focus on how these therapies have already been utilized to improve treatment of chronic disease, as well as how current literature supports the expansion of these therapies to treat additional chronic indications in the future. The thioether-functionalized 2-azabutadiene (iPrS)2C=C(H)-N=CPh2 L ligates to CdI2 and HgI2 to form the chelate compounds [CdI2{(iPrS)2C=C(H)-N=CPh2] (1) and [HgI2(iPrS)2C=C(H)-N=CPh2] (2). Their crystal structures were solved via X-ray diffraction. Both crystallize in the non-centrosymmetric space groups: monoclinic P21 (1) and orthorhombic P212121 (2), respectively. The closed-shell d10 metal centers are four-coordinated (two iodides and S and N coordinating atoms from the ligand L) in both complexes. The geometrical indexes τ indicate that a highly distorted trigonal pyramidal is adopted for 1 and a seesaw geometry for 2. The comparative nature of metal–ligand bonds is discussed on the basis of metric parameters and of QT-AIM (quantum theory of atoms in molecules) calculations. L was also treated with CuI to obtain the dinuclear species [LCu(μ2-I2)CuL] (3), in which the two Cu(I) centers are linked by a short metal–metal bond. I investigate the paradoxes associated with the Fédération Internationale de Football Association (FIFA) point-based ranking of national soccer teams. The ranking has been plagued with paradoxes that incentivize teams to avoid playing friendly matches, i.e., matches that are not part of any official FIFA tournament or preliminaries, and applying other counterintuitive strategies. The most spectacular paradox was the dramatic underrating of the hosts of major tournaments. For a long time, host teams, which were absent from preliminary matches, would play only friendly matches that awarded few points. Here, I present three models that estimate the magnitude of the resulting "host effect" at 14.2–16 positions. Such an estimate counteracts the intuition that a large investment in hosting a tournament should result in an improvement in the host team's standing. However, as discussed here, a given host's low ranking could decrease interest in the tournament, and likely result in a major loss of advertisement revenue. [ABSTRACT FROM AUTHOR]

The CuI- or Ag2CO3-catalyzed [3+2] cycloaddition of propargyl-substituted dihydroisoindolin- 1-one (3) with arylnitrile oxides 1a–d (Ar = Ph, p-MeC6H4, p-MeOC6H4, p-ClC6H4) produces in good yields novel 3,5-disubstituted isoxazoles 4 of the ethyl-2-benzyl-3-oxo-1-((3-arylisoxazol-5yl)methyl)- 2,3-dihydro-1H-isoindole-1-carboxylate type. With aryl azides 2a–d (Ar = Ph, p-MeC6H4, p-OMeC6H4, p-ClC6H4), a series of 1,4-disubstituted 1,2,3-triazoles 6 (ethyl-2-benzyl-3-oxo-1-((1-aryl-1H-1,2,3- triazol-4-yl)methyl)-2,3-dihydro-1H-isoindole-1-carboxylates) was obtained. The reactions proceed in a regioselective manner affording exclusively racemic adducts 4 and 6. Compared to the uncatalyzed cycloaddition, the yields are significantly improved in the presence of CuI as catalyst, without alteration of the selectivity. The regio- and stereochemistry of the cycloadducts has been corroborated by an X-ray diffraction study of 4a, and in the case of 6a by XH-correlation and HMBC spectra. [ABSTRACT FROM AUTHOR]

We study the application of Optimal Control Theory to Ion Cyclotron Resonance. We test the validity and the efficiency of this approach for the robust excitation of an ensemble of ions with a wide range of cyclotron frequencies. Optimal analytical solutions are derived in the case without any pulse constraint. A gradient-based numerical optimization algorithm is proposed to take into account limitation in the control intensity. The efficiency of optimal pulses is investigated as a function of control time, maximum amplitude and range of excited frequencies. A comparison with adiabatic and SWIFT pulses is done. On the basis of recent results in Nuclear Magnetic Resonance, this study highlights the potential usefulness of optimal control in Ion Cyclotron Resonance. [ABSTRACT FROM AUTHOR]

In the present study, the preparation of controlled-released leflunomide (LFD)-loaded skin patches was evaluated, utilizing the combination of chitosan (CS) nanoparticles (NPs) incorporated into suitable poly(l-lactic acid) (PLLA) or poly(lactic-co-glycolic acid) (PLGA) polyester matrices. Initially, LFD-loaded CS NPs of ~600 nm and a smooth surface were prepared, while strong inter-molecular interactions between the drug and the CS were unraveled. In the following step, the prepared LFD-loaded CS NPs were incorporated into PLLA or PLGA, and thin-film patches were prepared via spin-coating. Analysis of the prepared films showed that the incorporation of the drug-loaded CS NPs resulted in a significant increase in the drug's release rate and extent as compared to neat LFD-loaded polyester patches (i.e., prepared without the use of CS NPs). In-depth analysis of the prepared formulations showed that the amorphization of the drug within the matrix and the increased wetting properties of the prepared CS NPs were responsible for the improved thin-film patch characteristics. Mass spectrometry imaging is a powerful tool for analyzing the different kinds of molecules in tissue sections, but some substances cannot be measured easily, due to their physicochemical properties. In such cases, chemical derivatization could be applied to introduce the charge into the molecule and facilitate its detection. Here, we study cholesterol derivatization with betaine aldehyde from tissue slices and evaluate how different sample preparation methods influence the signal from the derivatization product. In this study, we have tested different solutions for betaine aldehyde, different approaches to betaine aldehyde deposition (number of layers, deposition nozzle height), and different MALDI matrices for its analysis. To modulate the biofunctionality of implantable medical devices commonly used in clinical practice, their surface modification with bioactive polymeric coatings is an attractive and successful emerging strategy. Biodegradable coatings based on poly(lactic acid-co-glycolic acid), PLGA, represent versatile and safe candidates for surface modification of implantable biomaterials and devices, providing additional tunable ability for topical delivery of desired therapeutic agents. In the present study, Ibuprofen-loaded PLGA coatings (PLGA/IBUP) were obtained by using the dip-coating and drop-casting combined protocol. The composite materials demonstrated long-term drug release under biologically simulated dynamic conditions. Reversible swelling phenomena of polymeric coatings occurred in the first two weeks of testing, accompanied by the gradual matrix degradation and slow release of the therapeutic agent. Irreversible degradation of PLGA coatings occurred after one month, due to copolymer's hydrolysis (evidenced by chemical and structural modifications). After 30 days of dynamic testing, the cumulative release of IBUP was ~250 µg/mL. Excellent cytocompatibility was revealed on human-derived macrophages, fibroblasts and keratinocytes. The results herein evidence the promising potential of PLGA/IBUP coatings to be used for surface modification of medical devices, such as metallic implants and wound dressings. In this study, the highly strained three-membered aziridine ring was successfully activated as the aziridinium ion by alkylation of the ring nitrogen with a methyl, ethyl or allyl group, which was followed by ring opening with external nucleophiles such as acetate and azide. Such alkylative aziridine ring opening provides an easy route for the synthesis of various N-alkylated amine-containing molecules with concomitant introduction of an external nucleophile at either its α- or β-position. This paper proposes and presents the chemical modification of linear hydroxyethers (LHE) with different molecular weights (380, 640, and 1830 g/mol) with the addition of three types of rubbers (polysulfide rubber (PSR), polychloroprene rubber (PCR), and styrene-butadiene rubber (SBR)). The main purpose of choosing this type of modification and the materials used was the possibility to use it in industrial settings. The modification process was conducted for a very wide range of modifier additions (rubber) per 100 g LHE. The materials obtained in the study were subjected to strength tests in order to determine the effect of the modification on functional properties. Mechanical properties of the modified materials were improved after the application of the modifier (rubber) to polyhydroxyether (up to certain modifier content). The most favorable changes in the tested materials were registered in the modification of LHE-1830 with PSR. In the case of LHE-380 and LHE-640 modified in cyclohexanol (CH) and chloroform (CF) solutions, an increase in the values of the tested properties was also obtained, but to a lesser extent than for LHE-1830. The largest changes were registered for LHE-1830 with PSR in CH solution: from 12.1 to 15.3 MPa for compressive strength tests, from 0.8 to 1.5 MPa for tensile testing, from 0.8 to 14.7 MPa for shear strength, and from 1% to 6.5% for the maximum elongation. Basic fibroblast growth factor (bFGF) is a crucial supplement for culture media of human pluripotent stem cells. However, bFGF is extremely unstable under cell culture conditions, which makes frequent (generally every day) medium refreshment requisite. We recently developed a water-floatable, bFGF-releasing membrane via a simple bFGF adsorption process following oxygen plasma treatment by utilizing a polyethylene nonwoven fabric as an adsorbent. This membrane allowed sustained release of bFGF while floating on medium, thereby keeping the bFGF concentration in the medium sufficient for maintaining human-induced pluripotent stem cells (iPSCs) in a proliferative and pluripotent state for as long as 3 days. In this study, lyophilization was applied to the membrane to stabilize bFGF. The sustained bFGF-releasing function of the membrane was kept unchanged even after lyophilization and subsequent cryopreservation at −30 °C for 3 months. The cryopreserved membrane supported proliferation and colony formation of human iPSCs while retaining their viability and pluripotency in a medium-change-free continuous culture for 3 days. The present bFGF-releasing membrane is ready-to-use, storable for at least 3 months, and obviates daily medium refreshment. Predominantly non-furanic commercial humins were used to prepare humin-based non-isocyanate polyurethane (NIPU) resins for wood panel adhesives. Pure humin-based NIPU resins and tannin–humin NIPU resins were prepared, the latter to upgrade the humins' performance. Species in the raw humins and species formed in the NIPU resins were identified by Matrix Assisted Laser Desorption Ionization Time of Flight (MALDI ToF) spectrometry and Fourier Transform Infrared (FTIR). Humins, fulvic acid and derivatives, humic acid and its fragments, some lignans present and furanic oligomers present formed NIPU linkages. Thermomechanical analysis (TMA) showed that as with other biomaterials-based NIPU resins, all these resins also showed two temperature peaks of curing, the first around 130 °C and the second around 220 °C. A decrease in the Modulus of Elasticity (MOE) between the two indicated that the first curing period corresponded to linear growth of the oligomers forming a physical entanglement network. This then disentangled, and the second corresponded to the formation of a chemical cross-linked network. This second peak was more evident for the tannin–humin NIPU resins. All the laboratory particleboard made and tested either bonded with pure humins or with tannin–humin NIPU adhesives satisfied well the internal bond strength requirements of the relevant standard for interior grade panels. Respiratory infections are a real threat for humans, and therefore the pig model is of interest for studies. As one of a case for studies, Actinobacillus pleuropneumoniae (APP) caused infections and still worries many pig breeders around the world. To better understand the influence of pathogenic effect of APP on a respiratory system—lungs and tracheobronchial lymph nodes (TBLN), we aimed to employ matrix-assisted laser desorption/ionization time-of-flight mass spectrometry imaging (MALDI-TOF MSI). In this study, six pigs were intranasally infected by APP and two were used as non-infected control, and 48 cryosections have been obtained. MALDI-TOF MSI and immunohistochemistry (IHC) were used to study spatial distribution of infectious markers, especially interleukins, in cryosections of porcine tissues of lungs (necrotic area, marginal zone) and tracheobronchial lymph nodes (TBLN) from pigs infected by APP. CD163, interleukin 1β (IL-1β) and a protegrin-4 precursor were successfully detected based on their tryptic fragments. CD163 and IL-1β were confirmed also by IHC. The protegrin-4 precursor was identified by MALDI-TOF/TOF directly on the tissue cryosections. CD163, IL-1β and protegrin-4 precursor were all significantly (p < 0.001) more expressed in necrotic areas of lungs infected by APP than in marginal zone, TBLN and in control lungs. The nuclei of cells may exhibit invaginations of the nuclear envelope under a variety of conditions. These invaginations form a branched network termed the nucleoplasmic reticulum (NR), which may be found in cells in pathological and physiological conditions. While an extensive NR is a hallmark of cellular senescence and shows associations with some cancers, very little is known about the formation of NR in physiological conditions, despite the presence of extensive nuclear invaginations in some cell types such as endometrial cells. Here we show that in these cells the NR is formed in response to reproductive hormones. We demonstrate that oestrogen and progesterone are sufficient to induce NR formation and that this process is reversible without cell division upon removal of the hormonal stimulus. Nascent lamins and phospholipids are incorporated into the invaginations suggesting that there is a dedicated machinery for its formation. This study is devoted to the degradation pathway (bio, photo degradation and photo/bio) of Poly(Lactic acid) PLA polymers by means of melt viscoelasticity. A comparison was made between three PLA polymers with different microstructures (L, D stereoisomers). Biodegradability was determined during composting by burying the polymer films in compost at 58 °C. Melt viscoelasticity was used to assess the molecular evolution of the materials during the composting process. Viscoelastic data were plotted in the complex plane. We used this methodology to check the kinetics of the molecular weight decrease during the initial stages of the degradation, through the evolution of Newtonian viscosity. After a few days in compost, the Newtonian viscosity decreased sharply, meaning that macromolecular chain scissions began at the beginning of the experiments. However, a double molar mass distribution was also observed on Cole–Cole plots, indicating that there is also a chain recombination mechanism competing with the chain scission mechanism. PLA hydrolysis was observed by infra-red spectroscopy, where acid characteristic peaks appeared and became more intense during experiments, confirming hydrolytic activity during the first step of biodegradation. During UV ageing, polymer materials undergo a deep molecular evolution. After photo-degradation, lower viscosities were measured during biodegradation, but no significant differences in composting were found. [ABSTRACT FROM AUTHOR]

In recent years, there has been an increasing interest in oncolytic adenoviral vectors as an alternative anticancer therapy. The induction of an immune response can be considered as a major limitation of this kind of application. Significant research efforts have been focused on the development of biodegradable polymer poly-gamma-glutamic acid (γ-PGA)-based nanoparticles used as a vector for effective and safe anticancer therapy, owing to their controlled and sustained-release properties, low toxicity, as well as biocompatibility with tissue and cells. This study aimed to introduce a specific destructive and antibody blind polymer-coated viral vector into cancer cells using γ-PGA and chitosan (CH). Adenovirus was successfully encapsulated into the biopolymer particles with an encapsulation efficiency of 92% and particle size of 485 nm using the ionic gelation method. Therapeutic agents or nanoparticles (NPs) that carry therapeutics can be directed specifically to cancerous cells by decorating their surfaces using targeting ligands. Moreover, in vitro neutralizing antibody response against viral capsid proteins can be somewhat reduced by encapsulating adenovirus into γ-PGA-CH NPs, as only 3.1% of the encapsulated adenovirus was detected by anti-adenovirus antibodies in the presented work compared to naked adenoviruses. The results obtained and the unique characteristics of the polymer established in this research could provide a reference for the coating and controlled release of viral vectors used in anticancer therapy. Malaria is an infectious disease caused by a protozoan parasite which is transmitted by female Anopheles mosquitoes around tropical and sub-tropical regions. Half of the world's population is at risk of being infected by malaria. This mainly includes children, pregnant women and people living with chronic diseases. The main factor that has contributed to the spread of this disease is the increase in the number of drug-resistant parasites. To overcome drug resistance, researchers have developed drug delivery systems from biodegradable polymers for the loading of antimalarials. The drug delivery systems were characterized by distinct features such as good biocompatibility, high percentage drug encapsulation, reduced drug toxicity and targeted drug delivery. Biopolymers have become increasingly popular as biostimulators of plant growth. One of them, oligo-alginate, is a molecule that regulates plant biological processes and may be used in horticultural practice as a plant growth regulator. Biostimulators are mainly used to improve plant tolerance to abiotic stresses, including salinity. The aim of the study was to assess the effects of salinity and oligo-alginate of various molecular masses on the growth and physiological activity of Eucomis autumnalis. The species is an ornamental and medicinal plant that has been used for a long time in the traditional medicine of South Africa. The bulbs of E. autumnalis were coated using depolymerized sodium alginate of molecular mass 32,000; 42,000, and 64,000 g mol−1. All of these oligo-alginates fractions stimulated plant growth, and the effect was the strongest for the fraction of 32,000 g mol−1. This fraction was then selected for the second stage of the study, when plants were exposed to salt stress evoked by the presence of 100 mM NaCl. We found that the oligo-alginate coating mitigated the negative effects of salinity. Plants treated with the oligomer and watered with NaCl showed smaller reduction in the weight of the above-ground parts and bulbs, pigment content and antioxidant activity as compared with those not treated with the oligo-alginate. The study demonstrated for the first time that low molecular mass oligo-alginate may be used as plant biostimulator that limits negative effects of salinity in E. autumnalis. [ABSTRACT FROM AUTHOR]

Background/Objectives: Poisonings among children are a major cause of morbidity and mortality worldwide and present a significant challenge for emergency medical services (EMS). The aim of this study was to analyze the types of substances causing poisonings and the intention of poisonings in children, providing detailed information on the most common causes of poisonings in different age groups. Methods : A retrospective study was conducted analyzing medical records of pediatric patients under the care of emergency medical services (EMS) in Wroclaw, Poland, between 2015 and 2017. The study included all patients under 18 years old diagnosed with poisoning. Data collected included age, sex, cause of poisoning, location of the incident, neurological status (GCS), and the type of healthcare facility. Results : The study included 484 patients, with a mean age of 13 years. The largest age group was 16-18 years (44%), and the majority were female (58%). The most common causes of poisonings were alcohol (29.3%), medications (26%), and intoxicants (24.8%). Over half of the incidents occurred at home (52.5%). Intentional poisonings constituted 75% of cases, particularly involving alcohol (38.6%), intoxicants (32%), and medications (26.7%). Accidental poisonings were mainly due to household chemicals (28.7%) and carbon monoxide (27%). The differences in causes and locations of poisonings were statistically significant ( p < 0.001). Conclusions : Our study showed that the most common causes of poisonings among children were alcohol, medications, and intoxicants. Poisonings most frequently occurred at home, especially with medications, while outside the home, alcohol and intoxicants were predominant. This study was not registered.

The modern construction industry is looking for new ecological materials (available, cheap, recyclable) that can successfully replace materials that are not environmentally friendly. Fibers of natural origin are materials that can improve the properties of gypsum composites. This is an important issue because synthetic fibers (hardly biodegradable-glass or polypropylene fibers) are commonly used to reinforce gypsum boards. Increasing the state of knowledge regarding the possibility of replacing synthetic fibers with natural fibers is another step towards creating more environmentally friendly building materials and determining their characteristics. This paper investigates the possibility of manufacturing fiber-gypsum composites based on natural gypsum (building gypsum) and hemp ( Cannabis sativa L.) fibers grown in Poland. The effect of introducing hemp fibers of different lengths and with varying proportions of mass (mass of gypsum to mass of fibers) into the gypsum matrix was investigated. The experimental data obtained indicate that adding hemp fibers to the gypsum matrix increases the static bending strength of the composites manufactured. The highest mechanical strength, at 4.19 N/mm 2 , was observed in fiber-gypsum composites with 4% hemp fiber content at 50 mm in length. A similar trend of increased strength was observed in longitudinal tension. Again, the composite variant with 4% fiber content within the gypsum matrix had the highest mechanical strength. Manufacturing fibers-gypsum composites with more than 4% hemp fiber content negatively affected the composites' strength. Mixing long (50 mm) hemp fibers with the gypsum matrix is technologically problematic, but tests have shown a positive effect on the mechanical properties of the refined composites. The article indicates the length and quantity limitations of hemp fibers on the basis of which fiber-gypsum composites were produced.

This paper presents the results of an experimental study designed to evaluate the effect of repeated stretching cycles on the electrical resistance change in a NiTi alloy wire. In particular, tests were carried out to determine the effect of the type of loading on resistance change in the investigated wires. Wires with a diameter of 100 μm were used in the research. The experiment was carried out on a dedicated test stand designed for this purpose. During the test, the samples were subjected to 40 identical tensile cycles. The electrical resistance, sample elongation, and tensile force during successive stretching cycles were measured. The conducted research demonstrated the impact of elongation and reorientation of the structure on the resistance change in NiTi alloy thin wires. The research included a comparison of the effect of two different types of loading on the electrical resistance change in the sample. During cyclic stretching of a NiTi alloy sample with constant displacement, a decrease in electrical resistance was observed after each successive stretching cycle. Alternatively, when stretching with a constant force, the value of electrical resistance increased. In both types of loads, the greatest change in resistance value was observed at the initial cycles.

This article presents a procedure for selecting optimal ceramic paste formulations dedicated to the 3D printing process using robocasting technology. This study investigated pastes with varying ceramic powder particle sizes and different proportions of additives, such as ceramic microspheres and nutshells. This selection process allowed for the classification of ceramic mixtures into those suitable and unsuitable for this additive manufacturing technique. Subsequently, the viscosity of the pastes was measured, and extrudability tests were performed to determine the force required for extrusion and evaluate the quality of the extruded material. In the final stage, the setting time of the ceramic pastes was assessed to establish the drying time of the printed elements. It was found that the length of the extruded band of ceramic paste was inversely proportional to the Al₂O₃ content. Moreover, the extrusion force for samples with varying ceramic powder particle sizes (MG1-MG5) ranged from 133 to 166 N, compared to 77 N for the base sample (BM1). The obtained results enable further development in robocasting additive technology, including the development of a rapid and effective method for validating ceramic pastes used in this process.

In the paper, a new design of a quasi-isotropic antenna for high-power electromagnetic (EM) field measurement is presented, along with its investigation into suitability. The measuring probe is intended for assessing pulsed microwaves, which cannot be measured by available meters due to the high value of electric field strength and short pulse duration. The measurement of such a strong field is required according to guidelines for protecting people against microwave fields, especially those emitted by radars. The proposed probe is based on the concept of dipole-diode detection. To enable high-power measurement, the receiving antenna is electrically "small," allowing diode detection within the diode square-law characteristics range. Additionally, the shortened dipole length minimizes the spatial integration error, which can be significant in the case of microwave measurement. To obtain the desired antenna polarization, a new dipole geometry was proposed. Fulfilling the requirement of measuring all incident EM field polarizations, the receiving antenna was based on three dipoles arranged within a specific "magic" angular arrangement, ensuring a suitable quasi-isotropic radiation pattern. The proposed probe can operate in a frequency range from 1 GHz to 12 GHz.

Due to variations in chemical composition and production processes, homonymous austenitic stainless steels can differ significantly regarding their initial microstructure, metastability, and thus, their fatigue behavior. Microstructural investigations and fatigue tests have been performed in order to evaluate this aspect. Three different batches and production forms of nominally one type of steel AISI 347 were investigated under monotonic tensile tests and cyclic loading under total strain and stress control in low and high cycle fatigue regimes, respectively. The deformation induced α'-martensite formation was investigated globally by means of in situ magnetic measurements and locally using optical light microscopy of color etching of micrographs. The investigation showed that the chemical composition and the different production processes influence the material behavior. In fatigue tests, a higher metastability and thus a higher level of deformation induced α'-martensite pronounced cyclic hardening, resulting in significantly greater endurable stresses in total strain-controlled tests and an increase in fatigue life in stress-controlled tests. For applications of non-destructive-testing, detailed knowledge of a component's metastability is required. In less metastable batches and for lower stress levels, α'-martensite primarily formed at the plasticization zone of a crack. Furthermore, the formation and nucleation points of α'-martensite were highly dependent on grain size and the presence of δ-ferrite. This study provides valuable insights into the different material behavior of three different batches with the same designation, i.e., AISI 347, due to different manufacturing processes and differences in the chemical composition, metastability, and microstructure.

Correct design of the sheet metal forming process requires knowledge of the friction phenomenon occurring in various areas of the drawpiece. Additionally, the friction at the drawbead is decisive to ensure that the sheet flows in the desired direction. This article presents the results of experimental tests enabling the determination of the coefficient of friction at the drawbead and using a specially designed tribometer. The test material was a DC04 carbon steel sheet. The tests were carried out for different orientations of the samples in relation to the sheet rolling direction, different drawbead heights, different lubrication conditions and different average roughnesses of the countersamples. According to the aim of this work, the Features Importance analysis, conducted using the Gradient-Boosted Regression Trees algorithm, was used to find the influence of several parameter features on the coefficient of friction. The advantage of gradient-boosted decision trees is their ability to analyze complex relationships in the data and protect against overfitting. Another advantage is that there is no need for prior data processing. According to the best of the authors' knowledge, the effectiveness of gradient-boosted decision trees in analyzing the friction occurring in the drawbead in sheet metal forming has not been previously studied. To improve the accuracy of the model, five MinLeafs were applied to the regression tree, together with 500 ensembles utilized for learning the previously learned nodes, noting that the MinLeaf indicates the minimum number of leaf node observations. The least-squares-boosting technique, often known as LSBoost, is used to train a group of regression trees. Features Importance analysis has shown that the friction conditions (dry friction of lubricated conditions) had the most significant influence on the coefficient of friction, at 56.98%, followed by the drawbead height, at 23.41%, and the sample width, at 11.95%. The average surface roughness of rollers and sample orientation have the smallest impact on the value of the coefficient of friction at 6.09% and 1.57%, respectively. The dispersion and deviation observed for the testing dataset from the experimental data indicate the model's ability to predict the values of the coefficient of friction at a coefficient of determination of R 2 = 0.972 and a mean-squared error of MSE = 0.000048. It was qualitatively found that in order to ensure the optimal (the lowest) coefficient of friction, it is necessary to control the friction conditions (use of lubricant) and the drawbead height.

Gastric cancer (GC) is the fifth most frequently diagnosed cancer and the fifth most common cause of cancer death in the world. Regarding the age at which the diagnosis was made, GC is divided into early-onset gastric cancer (EOGC-up to 45 years of age) and conventional GC (older than 45). EOGC constitutes approximately 10% of all GCs. Numerous reports indicate that EOGC is more aggressive than conventional GC and is often discovered at an advanced tumor stage, which has an impact on the five-year survival rate. The median survival rate for advanced-stage GC is very poor, amounting to less than 12 months. Risk factors for GC include family history, alcohol consumption, smoking, Helicobacter pylori , and Epstein-Barr virus infection. It has been shown that a proper diet and lifestyle can play a preventive role in GC. However, research indicates that risk factors for conventional GC are less correlated with EOGC. In addition, the unclear etiology of EOGC and the late diagnosis of this disease limit the possibilities of effective treatment. Genetic factors are considered a likely cause of EOGC, as young patients are less exposed to environmental carcinogens. Research characterizing GC in young patients is scarce. This comprehensive study presents all aspects: epidemiology, risk factors, new treatment strategies, and future directions.

Hemp ( Cannabis sativa L.) seeds are an interesting raw material for malting regarding its relatively high bioactive compounds concentration and proven advantageous properties in different food products and dietary supplements. In the first stage of the study, important seeds properties relevant to the malting process including moisture content, seed viability, and water absorption capacity were determined. However, a few parameters determining the seeds' usability for malt preparation, such as germination ability and water sensitivity, are different in comparison to typical malting raw materials such as barley or wheat. However, they make it possible to obtain high-quality hemp malt. In the next stage of research, spectroscopic and chromatographic analyses, including measurements of antioxidant activity and protein separation by SEC-HPLC, were conducted. The results showed that the malting process improved the total antioxidant potential of hemp seeds by 15%, leading to an increase in the concentration of lower molecular weight proteins and oligopeptides-below molecular mass of 10 kDa-responsible for this high antioxidant activity. The processing of hemp seeds reduced the phytate content while increasing phosphate fractions with fewer phosphate groups, which may have a beneficial effect on nutritional value. These results suggest that malting hemp seeds needs optimalization of the process but can increase its nutritional value as a promising raw material in the food industry.

Despite tremendous developments in the field of laboratory testing in transplantation, the rules of eligibility for corneal transplantation still do not include typing of human leukocyte antigens (HLAs) in the donor and recipient or detection of donor-specific antibodies (DSAs) in the patient. The standard use of diagnostic algorithms is due to the cornea belonging to immunologically privileged tissues, which usually determines the success of transplantation of this tissue. A medical problem is posed by patients at high risk of transplant rejection, in whom the immune privilege of the eye is abolished and the risk of transplant failure increases. Critical to the success of transplantation in patients at high risk of corneal rejection may be the selection of an HLA-matched donor and recipient, and the detection of existing and/or de novo emerging DSAs in the patient. Incorporating the assessment of these parameters into routine diagnostics may contribute to establishing immune risk stratification for transplant rejection and effective personalized therapy for patients.

Undercutting anchors are structural elements used in construction and geotechnics to stabilize both structures and soils. Their main applications include stabilizing slopes and embankments, reinforcing foundations, and providing support during tunnel construction and other underground works. The authors propose the use of these anchors in rock mass detachment technology. This article presents a comprehensive analysis of the mechanism behind rock mass detachment using an undercutting anchor. Particular attention is given to the influence of parameters such as the fracture energy of the medium and the coefficient of friction between the medium and the anchor head on the detachment process of rock elements during anchor expansion in the drilled hole. Numerical FEM analysis was employed to model the effect of changes in the shape and size of failure cones under varying simulation conditions. The discussed problem is crucial for evaluating the effectiveness of this anchor design under non-standard conditions, particularly in the unconventional destruction of rock media.

Rhabdomyosarcoma (RMS) of the biliary tract is a rare tumor in children, constituting 0.5-0.8% of all pediatric RMS. Still, it is the most common malignancy in this location in children. Due to its rarity and location, it may cause diagnostic and treatment difficulties. Above all, there are no therapeutic guidelines specific for this tumor location. The aim of the study was to present an analysis of our experience with the treatment of children with biliary tract rhabdomyosarcoma (RMS) and discuss clinical recommendations for this specific location published in the literature. A retrospective analysis of medical records of eight children with biliary tree RMS treated in one center between 1996-2022 was performed. Records of eight children, five boys and three girls aged 2 yrs 6 mo to 16 yrs 9 mo (median-6 yrs) were analyzed. All patients presented with jaundice as the first symptom. In two patients, initial diagnosis of a tumor was established. For the remaining six, the primary diagnoses were as follows: choledochal cyst-one, malformation of the biliary ducts-one, choledocholithiasis-one, cholangitis-three. In four patients, the extrahepatic bile ducts were involved; in four patients, both the intrahepatic and extrahepatic bile ducts were involved. Embryonal RMS was diagnosed in seven patients (three botryoides type). Alveolar RMS was found in one patient. Biopsy (three surgical, four during endoscopic retrograde cholangiopancreatography (ERCP)) was performed in seven patients. One child underwent primary partial tumor resection (R2). Seven patients received neoadjuvant chemotherapy, followed by delayed resection in five, including liver transplantation in one (five were R0). Two patients did not undergo surgery. Radiotherapy was administered in four patients (two in first-line treatment, two at relapse/progression). Six patients (75%) are alive with no evidence of disease, with follow-up ranging from 1.2 yrs to 27 yrs (median 11 yrs. and 4 mo.). Two patients died from disease, 2 y 9 mo and 3 y 7 mo from diagnosis. Children presenting with obstructive jaundice should be evaluated for biliary tract RMS. The treatment strategy should include biopsy and preoperative chemotherapy, followed by tumor resection and radiotherapy for residual disease and in case of relapse.

Background: Over the past years, the introduction of direct-acting antivirals (DAAs) revolutionized chronic hepatitis C treatment. We aimed to characterize and assess treatment efficacy in three specific groups of patients treated with DAAs: those with active solid malignant tumors (SMTs), hematological diseases (HDs) and hepatocellular carcinomas (HCCs)., Methods: A total of 203 patients with active oncological disease (SMT n = 61, HD = 67, HCC n = 74) during DAA treatment in 2015-2020 selected from the EpiTer-2 database were analyzed retrospectively and compared to 12,983 patients without any active malignancy., Results: Extrahepatic symptoms were more frequent in HD patients (17.2% vs. SMT = 10.3%, HCC = 8.2%, without = 7.8%, p = 0.004). HCC patients characterized with the highest ALT activity (81 IU/L vs. SMT = 59.5 IU/L, HD = 52 IU/L, without = 58 IU/L, p = 0.001) more often had F4 fibrosis as well (86.11% vs. SMT = 23.3%, HD = 28.8%, controls = 24.4%, p = 0.001). A significant majority of subjects in HCC, HD and SMT populations completed the full treatment plan (HCC = 91%; n = 67, HD = 97%; n = 65, SMT = 100%; n = 62). Concerning the treatment efficacy, the overall sustained virologic response, excluding non-virologic failures, was reported in 93.6% HD, 90.16% SMT and 80.6% in HCC patients., Conclusions: As presented in our study, DAA therapy has proven to be highly effective and safe in patients with active SMTs and HDs. However, therapy discontinuations resulting from liver disease progression remain to be the major concern in HCC patients.

Staphylococcus aureus infections present a significant threat to the global healthcare system. The increasing resistance to existing antibiotics and their limited efficacy underscores the urgent need to identify new antibacterial agents with low toxicity to effectively combat various S. aureus infections. Hence, in this study, we have screened T-muurolol for possible interactions with several S. aureus -specific bacterial proteins to establish its potential as an alternative antibacterial agent. Based on its binding affinity and interactions with amino acids, T-muurolol was identified as a potential inhibitor of S. aureus lipase, dihydrofolate reductase, penicillin-binding protein 2a, D-Ala:D-Ala ligase, and ribosome protection proteins tetracycline resistance determinant (RPP TetM), which indicates its potentiality against S. aureus and its multi-drug-resistant strains. Also, T-muurolol exhibited good antioxidant and anti-inflammatory activity by showing strong binding interactions with flavin adenine dinucleotide (FAD)-dependent nicotinamide adenine dinucleotide phosphate (NAD(P)H) oxidase, and cyclooxygenase-2. Consequently, molecular dynamics (MD) simulation and recalculating binding free energies elucidated its binding interaction stability with targeted proteins. Furthermore, quantum chemical structure analysis based on density functional theory (DFT) depicted a higher energy gap between the highest occupied molecular orbital and lowest unoccupied molecular orbital (E HOMO-LUMO ) with a lower chemical potential index, and moderate electrophilicity suggests its chemical hardness and stability and less polarizability and reactivity. Additionally, pharmacological parameters based on ADMET, Lipinski's rules, and bioactivity score validated it as a promising drug candidate with high activity toward ion channel modulators, nuclear receptor ligands, and enzyme inhibitors. In conclusion, the current findings suggest T-muurolol as a promising alternative antibacterial agent that might be a potential phytochemical-based drug against S. aureus . This study also suggests further clinical research before human application.

In end-stage heart failure, which is characterized by persistent or progressive ventricular dysfunction despite optimal medical therapy, a left ventricular assist device (LVAD) can be beneficial. Congestive heart failure provokes inflammatory and prothrombotic activation. The aim of this study was to evaluate the serum concentration of citrullinated histone 3 (CH3) representing neutrophil extracellular trap (NET) formation in patients referred for LVAD implantation. There were 10 patients with a median age of 61 (57-65) years enrolled in a prospective single-center analysis who underwent LVAD implantation. The CH3 plasma concentration was measured preoperatively and on the 1st and 7th postoperative days, followed by control measurements on the median (Q1-3) 88th (49-143) day. The preoperative CH3 concentration strongly correlated with brain natriuretic peptide (r = 0.879, p < 0.001). Significant differences in CH3 serum concentration were observed between pre- and postoperative measurements, including an increase on the first postoperative day ( p < 0.001), as well as a decrease on the seventh day ( p = 0.016) and in follow-up ( p < 0.001). CH3 concentration, as a marker of NET formation, decreases after LVAD implantation.

The aim of this study was to compare the head displacement of the KPSIT C50 dummy, representing a 50th percentile male, with the KPSIT C5 dummy, representing a 5th percentile female, during low-speed collisions. Low-speed collisions, such as those occurring in urban traffic jams, are increasingly common. The research was conducted on a dedicated educational workstation designed to measure forces in seat belts. This study is part of a comprehensive research project on crash tests involving both volunteers and physical KPSIT dummies. The tests were conducted at a speed of 20 km/h to simulate real-world low-speed collision scenarios. The findings demonstrate that using a sports bucket seat with four-point or five-point harnesses significantly reduces head displacement compared with standard car seats. Such seating configurations enhance safety by minimizing the risk of head injuries, which can occur when airbags do not deploy during low-speed collisions. Moreover, the study highlights that standard three-point seat belts allow for greater forward head movement, increasing the risk of head contact with the vehicle's interior during collisions at speeds too low to trigger airbag deployment.

This work presents a 2D radiochromic dosimeter for ultraviolet (UV) radiation measurements, based on cotton fabric volume-modified with nitroblue tetrazolium chloride (NBT) as a radiation-sensitive compound. The developed dosimeter is flexible, which allows it to adapt to various shapes and show a color change from yellowish to purple-brown during irradiation. The intensity of the color change depends on the type of UV radiation and is the highest for UVC (253.7 nm). It has been shown that the developed dosimeters (i) can be used for UVC radiation dose measurements in the range of up to 10 J/cm 2 ; (ii) can be measured in 2D using a flatbed scanner; and (iii) can have the obtained images after scanning be filtered with a medium filter to improve their quality by reducing noise from the fabric structure. The developed cotton-NBT dosimeters can measure UVC-absorbed radiation doses on objects of various shapes, and when combined with a dedicated computer software package and a data processing method, they form a comprehensive system for measuring dose distributions for objects with complex shapes. The developed system can also serve as a comprehensive method for assessing the quality and control of UV radiation sources used in various industrial processes.

The present study explores the synthesis and bio-safety evaluation of gadolinium-doped carbon quantum dots (GCQDs) as a potential dual-contrast agent for diagnostic imaging. GCQDs exhibit both fluorescent and magnetic properties, making them suitable for UV-Vis and magnetic resonance imaging (MRI). The synthesis of GCQDs was achieved via hydrothermal treatment, incorporating gadolinium into the carbon quantum dot matrix. The magnetic properties of GCQDs were analyzed, showing significantly enhanced values compared to gadobutrol, a common MRI contrast agent. However, synthesis constraints limit the gadolinium content achievable in nanodots. To assess the safety of GCQDs, their effects on the embryonic development of zebrafish ( Danio rerio ) were examined. Various concentrations of GCQDs were tested, observing mortality rates, hatchability, malformations, heartbeats, spontaneous movement, and GCQDs uptake. Dialysis studies indicated that gadolinium ions are incorporated into the internal structure of the carbon nanodots. Zebrafish toxicity tests revealed that while survival rates were comparable to control groups, hatchability decreased significantly with higher gadolinium concentrations in GCQDs. Fluorescence microscopy showed no statistical differences in the fluorescence intensity between groups. These findings suggest that GCQDs could serve as an effective dual-contrast agent, combining the optical imaging capabilities of CQDs with the enhanced MRI contrast provided by gadolinium. This study underscores the need for further research on the synthesis methods and biological interactions of GCQDs to ensure their safety and efficacy in medical applications.

This study focuses on the development of new amino-functionalized magnetic Fe 2 O 3 /SiO 2 nanocomposites with varying silicate shell ratios (1:0.5, 1:1, and 1:2) for the efficient elimination of Hg 2+ ions found in solutions. The Fe 2 O 3 /SiO 2 -NH 2 adsorbents were characterized for their structural, surface, and magnetic properties using various techniques, including Fourier transform infrared spectrum (FT-IR), powder X-ray diffraction (XRD), scanning electron microscopy (SEM), Braunauer-Emmett-Teller (BET), thermogravimetric analysis (TGA), zeta-potential, and particle size measurement. We investigated the adsorption circumstances, such as pH, dosage of the adsorbent, and duration of adsorption. The pH value that yielded the best results was determined to be 5.0. The Fe 2 O 3 /SiO 2 -NH 2 adsorbent with a silicate ratio of (1:2) exhibited the largest amount of adsorption capacity of 152.03 mg g -1 . This can be attributed to its significantly large specific surface area of 100.1 m 2 g -1 , which surpasses that of other adsorbents. The adsorbent with amino functionalization demonstrated a strong affinity for Hg 2+ ions due to the chemical interactions between the metal ions and the amino groups on the surface. The analysis of adsorption kinetics demonstrated that the adsorption outcomes adhere to the pseudo-second-order kinetic model. The study of adsorption isotherms revealed that the adsorption followed the Langmuir model, indicating that the adsorption of Hg 2+ ions with the adsorbent occurred as a monomolecular layer adsorption process. Furthermore, the thermodynamic analyses revealed that the adsorption of Hg 2+ ions using the adsorbent was characterized by a spontaneous and endothermic process. Additionally, the adsorbent has the ability to selectively extract mercury ions from a complex mixture of ions. The Fe 2 O 3 /SiO 2 -NH 2 nanocomposite, which is loaded with metal, can be easily recovered from a water solution due to its magnetic properties. Moreover, it can be regenerated effortlessly through acid treatment. This study highlights the potential use of amino-functionalized Fe 2 O 3 /SiO 2 magnetic nanoparticles as a highly efficient, reusable adsorbent for the removal of mercury ions from contaminated wastewater.

The infiltration of immune cells into the central nervous system mediates the development of autoimmune neuroinflammatory diseases. We previously showed that the loss of either Fabp5 or calnexin causes resistance to the induction of experimental autoimmune encephalomyelitis (EAE) in mice, an animal model of multiple sclerosis (MS). Here we show that brain endothelial cells lacking either Fabp5 or calnexin have an increased abundance of cell surface CD200 and soluble CD200 (sCD200) as well as decreased T-cell adhesion. In a tissue culture model of the blood-brain barrier, antagonizing the interaction of CD200 and sCD200 with T-cell CD200 receptor (CD200R1) via anti-CD200 blocking antibodies or the RNAi-mediated inhibition of CD200 production by endothelial cells increased T-cell adhesion and transmigration across monolayers of endothelial cells. Our findings demonstrate that sCD200 produced by brain endothelial cells regulates immune cell trafficking through the blood-brain barrier and is primarily responsible for preventing activated T-cells from entering the brain.

Introduction: Toll-like receptors (TLRs) play a vital role in the innate immune response, recognizing pathogens and initiating the inflammatory response. Research suggests that TLRs may also have a significant impact on the development and progression of cancers, including gastric cancer (GC). Understanding the role of individual TLRs in the immunopathogenesis of gastric cancer may provide new information necessary to develop more effective diagnostic and therapeutic methods., Aim of the Study: This study aimed to determine the role of selected TLR-2, -3, -4, and -9 in the immunopathogenesis of patients with newly diagnosed and untreated gastric cancer., Materials and Methods: The study included 60 newly diagnosed, untreated GC patients and 25 healthy volunteers. The research included analyses assessing the percentage of the tested TLRs on T and B lymphocyte subpopulations using multicolor flow cytometry and assessing their concentration in the serum of the examined patients using ELISA tests. The statistical analyses performed included a comparison of patients in individual stages of gastric cancer, an analysis of the most common clinical subtypes of gastric cancer, and a comparative analysis of differences in the gender of recruited patients., Results: Our studies showed different expression levels of TLR-2, -3, -4, and -9 on T and B lymphocyte subpopulations, as well as their different concentrations in patients' serum. Significant differences in the expression of these receptors were observed depending on the stage of gastric cancer and its clinical subtypes. These differences were also visible in the context of patient gender., Summary: The results of our studies suggest that TLR-2, -3, -4, and -9 may play an important role in the immunopathogenesis of gastric cancer. The differential expression of these receptors depending on the stage of the disease, clinical subtype, and gender of patients may have potential diagnostic and therapeutic significance. Further research is necessary to understand better the mechanisms of action of TLRs in gastric cancer and to apply this knowledge in clinical practice.

With the expansion of wireless mobile networks into both the daily lives of individuals as well as into the widely developing market of connected devices, communication is an increasingly attractive target for attackers. As the complexity of mobile cellular systems grows and the respective countermeasures are implemented to secure data transmissions, the attacks have become increasingly sophisticated on the one hand, but at the same time the system complexity can open up expanded opportunities for security and privacy breaches. After an in-depth summary of possible entry points to attacks to mobile networks, this paper first briefly reviews the basic principles of the physical layer implementation of 4G/5G systems, then gives an overview of possible attacks from a physical layer perspective. It also provides an overview of the software frameworks and hardware tool-software defined radios currently in use for experimenting with 4G/5G mobile networks, and it discusses their basic capabilities. In the final part, the paper summarizes the currently most promising families of techniques to detect illegitimate base stations-the machine-learning-based, localization-based, and behavior-based methods.

The study of phenomena related to the propagation of electromagnetic waves is a necessity with the development of mobile telephony networks. This paper deals with the problem of the quality of mobile telephony signals. The study uses a BTS scanner, dedicated to scanning the frequency spectrum and decoding information from base stations. The device is capable of scanning mobile networks from the second, third, fourth, and fifth generation (2G/3G/4G/5G) using a passive sensor. The article presents preliminary results of monitoring cellular network signals using a scanner mounted on an unmanned aircraft. The collected data call for the need to evaluate the signal parameters of the cellular network depending on the altitude of signal reception. This property emerges as very important in the case of areas with a high level of urbanization.

The characterization and modeling of the out-of-plane behavior of fiber-based materials is essential for understanding their mechanical properties and improving their performance in various applications, especially in the forming process. Despite this, research on paper and paperboard has mainly focused on its in-plane behavior rather than its out-of-plane behavior. However, for accurate material characterization and modeling, it is critical to consider the out-of-plane behavior. In particular, delamination occurs during forming processes such as creasing, folding, and deep drawing. In this study, three material models for paperboard are presented: a single all-material continuum model and two composite models using different cohesion methods. The two composite models decouple in-plane and out-of-plane behavior and consist of continuum models describing the behavior of individual layers and cohesive interface models connecting the layers. Material characterization experiments are performed to derive the model parameters and verify the models. The models are validated using three-point bending and bulge tests and show good agreement. A case study is also conducted on the application of the three models in the simulation of a deep drawing process with respect to wrinkle formation. By comparing the simulation results of wrinkle formation in the deep drawing process, the composite models, especially the cohesive interface composite model, show greater accuracy in replicating the experimental results, indicating that a single continuum model can also be used to represent wrinkles.

Fe-modified Cu catalysts with CeO 2 support, prepared by the impregnation method, were subjected to physicochemical analysis and catalytic tests in the steam reforming of methanol (SRM). Physicochemical studies of the catalysts were carried out using the XRF, TEM, STEM-EDS, XRD, TPR and nitrogen adsorption/desorption methods. XRD, TEM studies and catalytic tests of the catalysts were carried out at two reduction temperatures, 260 °C and 400 °C, to determine the relationship between the form and oxidation state of the active phase of the catalysts and the catalytic properties of these systems in the SRM. Additionally, the catalysts after the reaction were analysed for the changes in the structure and morphology using TEM methods. The presented results show that the composition of the catalysts, morphology, structure, form and oxidation state of the Cu and Fe active metals in the catalysts and the reaction temperature significantly impact their activity, selectivity and stability in the SRM process. The gradual deactivation of the studied catalysts under SRM conditions could result from the forming of carbon deposits and/or the gradual oxidation of the copper and iron phases under the reaction conditions.

Few studies have evaluated cytokeratin's (CK) staining patterns in atypical endometrial hyperplasia (AEH) coexisting with early-stage endometrial cancer (EC). We aimed to assess the staining patterns of selected CKs (CK7, CK19, CK20, CK AE1/AE3) in 74 patients with coexisting AEH and EC by independently analyzing both morphological variables. Specimens were collected from women with AEH and EC who underwent surgical interventions between 2012 and 2019 at the Department of Obstetrics and Gynecology of Vilnius University Hospital "Santaros Klinikos" in Vilnius, Lithuania. Immunostaining was also qualitatively classified as being heterogeneous or intense. The results revealed heterogeneous CK7 expression in all AEH cases and intense staining in 95.95% cases of AEH. The heterogeneous expression of CK7 was detected in all EC specimens. Intense CK7 expression was observed in 95.09% cases of EC G1 and in all G2 ECs. Heterogenous CK19 expression was present in all AEH specimens with intense staining in 92.42% of cases. Heterogeneous CK19 expression was observed in all EC samples with intense expression in 86.27% cases of EC G1 and 100% cases of EC G2. Interestingly, a significant relationship was found when comparing the heterogeneous expression of CK19 between AEH and well-differentiated EC. A significant difference was reported in the intense expression of CK AE1/AE3 ( p = 0.031; p = 0.029) between AEH and G2 ECs and in the intense expression of CK AE1/AE3 between G1 and G2 ECs. CK20 staining was not a characteristic feature for AEH and early-stage EC. CK staining is present either in AEH or in early-stage endometrioid-subtype EC in different manners. Heterogeneous CK19 expression was significantly more common in AEH than in EC. CK20 expression was not associated with either AEH nor early-stage EC. An intense expression of CK AE1/AE3 was mainly present in moderately differentiated ECs, whereas the intense reactivity of AE1/AE3 showed a significant difference in well to moderately differentiated uterine tumors. The clinical implication of CK staining may aid in the more accurate diagnosis of AEH and early-stage EC as well as detect micrometastases leading to better oncological outcomes.

The standard therapy for hormone-receptor-positive, human epidermal growth factor receptor 2-negative advanced breast cancer includes the use of cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) with endocrine therapy. The optimal post-CDK4/6i treatment sequence is unclear. This cohort study evaluated the initiation, characteristics, and outcomes of chemotherapy following CDK4/6i-based treatment. Among the 227 patients who began CDK4/6i therapy, 114 completed it. Seventy-nine female patients received further treatment, including 55 receiving chemotherapy. The average age was 60.1 years. Post-CDK4/6i chemotherapy was typically (69.1%) first-line due to an impending visceral crisis. The median progression-free survival (mPFS) was 3.0 months (range 0.5-18.9), and the median overall survival (mOS) was 8.3 months (0.5-26.1). The median OS from the end of CDK4/6i treatment was 12.4 months (1.5-26.8). In univariate analysis, neither mPFS nor mOS was associated with age, tumor grade, receptor status, Ki67 status, time from diagnosis to CDK4/6i cessation, therapy line, or CDK4/6i type. Dose reduction occurred in 12 patients (21.8%), and chemotherapy was ceased due to adverse events in 8 patients (14.6%). Chemotherapy showed limited benefit regardless of the regimen. The role of chemotherapy may evolve with broader CDK4/6i use in adjuvant treatment.

The continued global urbanization of the world is driving the development of the construction industry. In order to protect the environment, intensive research has been carried out in recent years on the development of sustainable materials and ecological construction methods. Scientific research often focuses on developing building materials that are renewable, energy-efficient, and have minimal impact on the environment throughout their life cycle. Therefore, this article presents research results aimed at developing a concrete mixture using cement with reduced CO 2 emissions. In the context of increasing ecological awareness and in line with European Union policy, the development of a mixture based on environmentally friendly cement is of key importance for the future development of the construction industry. The article compares the physical properties of two mixtures, their foaming possibilities, and the influence of the added polypropylene (PP) fibers on the strength properties of the produced composites. It was found that bending strength and compressive strength were highest in the material with silica fume and aluminum powder at 5.36 MPa and 28.76 MPa, respectively. Microscopic analysis revealed significant pore structure differences, with aluminum foamed samples having regular pores and hydrogen peroxide foamed samples having irregular pores. Optimizing aluminum powder and water content improved the materials' strength, crucial for maintaining usability and achieving effective 3D printing. The obtained results are important in the development of research focused on the optimization of 3D printing technology using concrete.

Resistant biofilms formed by Staphylococcus aureus on medical devices pose a constant medical threat. A promising alternative to tackle this problem is photodynamic inactivation (PDI). This study focuses on a polyurethane (PU) material with an antimicrobial surface consisting of a composite based on silicate, polycation, and erythrosine B (EryB). The composite was characterized using X-ray diffraction and spectroscopy methods. Anti-biofilm effectiveness was determined after PDI by calculation of CFU mL -1 . The liquid PU precursors penetrated a thin silicate film resulting in effective binding of the PU/silicate composite and the PU bulk phases. The incorporation of EryB into the composite matrix did not significantly alter the spectral properties or photoactivity of the dye. A green LED lamp and laser were used for PDI, while irradiation was performed for different periods. Preliminary experiments with EryB solutions on planktonic cells and biofilms optimized the conditions for PDI on the nanocomposite materials. Significant eradication of S. aureus biofilm on the composite surface was achieved by irradiation with an LED lamp and laser for 1.5 h and 10 min, respectively, resulting in a 10,000-fold reduction in biofilm growth. These results demonstrate potential for the development of antimicrobial polymer surfaces for modification of medical materials and devices.

Interesting alternatives to expensive biodegradable polymers are their composites with natural fillers. The addition of biochar to a blend of poly(lactic acid) (PLA) and poly(3-hydroxybutyrate- co -4-hydroxybutyrate) was studied, and the resulting materials were evaluated for their properties and changes during degradation. Introducing biochar as a filler brought a noticeable improvement in electrostatic properties. Surface resistivity decreased from 3.80 × 10 12 for the sample without biochar to 1.32 × 10 12 for the sample with 30% filler content. Degradation tests revealed distinct differences in the degradation profile for composites due to the presence of filler. Composites with a lower biochar content displayed curling crack edges during hydrolytic degradation, and when the filler content reached 20 wt%, PLA loss accelerated. This study suggests that biochar-based composites have potential to be used as sustainable materials with improved properties.
