The prevalence of apples as the most widely consumed fruit globally does not exempt them from storage-related issues, resulting in substantial harvest losses. A prominent concern is the development of rot due to various factors during storage. This research endeavors to examine the influence of agrotechnological methods on the longevity of apples and the incidence of rot throughout storage. Apple trees (Malus domestica Borkh. cv. Rubin) grafted on dwarfing rootstocks P60 were planted in 2010 in single rows with a spacing of 1.25 m between trees and 3.5 m between rows. Eight combinations of different growth control measures (manual, mechanical pruning, spraying, trunk cutting) were selected for the experiment. The implementation of mechanical pruning, in conjunction with trunk cutting and Ca-prohexadione spraying, as well as summer pruning, detrimentally impacted the shelf life of apples. Examination of the storage period revealed a loss of 33–40% of the crop due to rot. Conversely, manual pruning sustained a consistent level of phenolic compounds throughout the storage period. Other pruning methods resulted in a notable increase in phenolic compounds, ranging from 67% to a two-fold rise compared to the compounds present at harvest. However, the integration of mechanical pruning with subsequent manual pruning not only significantly augmented the yield of apples but also yielded a shelf life akin to that of manually pruned apples. The precise and automated diagnosis of apple leaf diseases is essential for maximizing apple yield and advancing agricultural development. Despite the widespread utilization of deep learning techniques, several challenges persist: (1) the presence of small disease spots on apple leaves poses difficulties for models to capture intricate features; (2) the high similarity among different types of apple leaf diseases complicates their differentiation; and (3) images with complex backgrounds often exhibit low contrast, thereby reducing classification accuracy. To tackle these challenges, we propose a three-residual fusion network known as MSCR-FuResNet (Fusion of Multi-scale Feature Extraction and Enhancements of Channels and Residual Blocks Net), which consists of three sub-networks: (1) enhancing detailed feature extraction through multi-scale feature extraction; (2) improving the discrimination of similar features by suppressing insignificant channels and pixels; and (3) increasing low-contrast feature extraction by modifying the activation function and residual blocks. The model was validated with a comprehensive dataset from public repositories, including Plant Village and Baidu Flying Paddle. Various data augmentation techniques were employed to address class imbalance. Experimental results demonstrate that the proposed model outperforms ResNet-50 with an accuracy of 97.27% on the constructed dataset, indicating significant advancements in apple leaf disease recognition. Microtubules undergo dynamic remodeling in response to diverse abiotic stress in plants. The plant-specific IQ67 DOMAIN (IQD) family proteins serve as microtubule-associated proteins, playing multifaceted roles in plant development and response to abiotic stress. However, the biological function of IQD genes in apple remains unclear. In this study, we conducted a comprehensive analysis of the Malus domestica genome, identifying 42 IQD genes distributed across 17 chromosomes and categorized them into four subgroups. Promoter analysis revealed the presence of stress-responsive elements. Subsequent expression analysis highlighted the significant upregulation of MdIQD17 and MdIQD28 in response to cold treatments, prompting their selection for further functional investigation. Subcellular localization studies confirmed the association of MdIQD17 and MdIQD28 with microtubules. Crucially, confocal microscopy and quantification revealed diminished microtubule depolymerization in cells transiently overexpressing MdIQD17 and MdIQD28 compared to wild-type cells during cold conditions. Apple cultivars 'Annurca' and 'Limoncella' are grown locally in the Campania region of Italy and are valued for their distinctive flavour and characteristics, including a high content of nutritionally important bioactive compounds. However, apples are typically stored chilled for several months before consumption, so it is important to assess if the valuable characteristics are still present after postharvest storage. Here, we compare the quality, nutritional parameters, and aroma of these two cultivars with two widely grown international cultivars, 'Golden Delicious' and 'Fuji', after 60 days of storage. The aroma profiles of all four apples were analysed using thermal desorption and gas chromatography–time-of-flight mass spectrometry. We show that the local cultivars are distinct from the international cultivars in their bioactive compound content and their antioxidant activity. 'Limoncella' shows high sugar content, which may be acting as a cryoprotectant during storage, and high total phenolics in the flesh, which is of nutritional interest. We identified 104 volatile organic compounds (VOCs) and showed that the overall aroma profile is distinct for each cultivar, containing 11 published odorant compounds. The 'Annurca' profile is uniquely low in esters. Seven VOCs retain good discrimination across the four cultivars and, together with the quality and nutritional data, separate the two local cultivars from the international cultivars by hierarchical clustering. Domesticated apple is a drought-sensitive species that spread from continental to Mediterranean temperate regions, where it can particularly experience prolonged water stress. One strategy to improve drought resistance in apple is engrafting on selected rootstocks. This study explores the potential of fast chlorophyll a fluorescence for the comparison of rootstock sensitivity to drought, looking for significant correlations with fruit productivity. The experiment was conducted in a field located in the coastal Po River Plain, Northern Italy, characterized by a loamy sandy soil, particularly prone to drought (86% sand). Mature plants of apple cv. Superchief® Sandidge engrafted on three different rootstocks (CIVP21pbr, MM106, M26) were monitored throughout the summer of 2021 and compared between irrigated and non-irrigated parcels, and at the end of the season, fruit production was evaluated. Despite soil water tension only reaching −13 kPa, the non-irrigated plants experienced a small but consistent loss of Photosystem II (PSII) activity and a lesser capacity of light energy conservation in the photosynthetic electron transport chain. The fruit weight correlated with PSII photochemical indexes recorded during early drought, particularly FV/FM and PIABS; a correlation emerged between fruit number per plant and median values of electron transport parameters, including PITOT. Although all rootstocks underwent a 40% loss of productivity, the fluorescence parameters revealed a graded susceptibility to drought, M26 > CIVP21pbr > MM106, which matched well with the plant vigour. Sucrose phosphate synthase (SPS) is an important link in the process of sugar metabolism. In addition, it is also involved in abiotic stresses in plants. In order to study the SPS gene family and its role in abiotic stress, we identified the MdSPS gene family members by bioinformatics methods such as correlation analysis, the HMM method, and the Clustering method, and analyzed the transient expression of MdSPS genes by quantitative real-time fluorescence analysis (qRT-PCR). The MdSPS gene family consists of a total of 19 members divided into three subfamilies distributed on 14 chromosomes in apples. The MdSPS gene family has 12 collinearity gene pairs, indicating significant duplication. Most members of this family contain a large number of plant hormone response elements, light-inducible elements, and abiotic stress response elements 2kb upstream of the promoter. Codon bias analysis shows that there are 28 high-frequency codons and no codons with strong preference in this family. Gene chip results showed that only MdSPS2, MdSPS3, MdSPS11, and MdSPS17 were up-regulated in roots, and they were all members of subfamily C. The qRT-PCR analysis showed that all members of this family responded significantly to drought stress, salt stress, and low temperature stress. Interestingly, the relative expression of MdSPS12 was significantly down-regulated under salt stress and low temperature stress. In addition, the expression of MdSPS3, MdSPS8, MdSPS11, and MdSPS17 was more than 20 fold higher than that of the control under drought stress, salt stress, and low temperature stress. 'Ruixue' apples were used as the test material to study the effect of 10 μM methyl jasmonate (MeJA) on the quality and cell wall metabolism of apples after 18 d of storage. The results showed that MeJA significantly decreased the respiratory rate, reduced the titratable acid content and maintained a high soluble solids content. MeJA has been shown to suppress the activities and gene expressions of WSP, CSP, ISP, and cellulose in contrast to the control group, thereby maintaining a lower cell permeability and higher exocarp firmness. MeJA significantly decreased the expression of MdACS, MdACO, MdPL, Mdgal, and MdPG genes in the apple exocarp when compared to the control group. In addition, the overexpression of MdPL18 increased the content of cell wall polysaccharides such as WSP and CSP, enhanced cell wall-degrading enzyme activities, and accelerated fruit ripening and softening, whereas silencing MdPL18 did the opposite. The effect of plant cultivar on the degree of mycorrhization and the benefits mediated by arbuscular mycorrhizal fungi (AMF) have been documented in many crops. In apple, a wide variety of rootstocks are commercially available; however, it is not clear whether some rootstock genotypes are more susceptible to mycorrhization than others and/or whether AMF species identity influences rootstock compatibility. This study addresses these questions by directly testing the ability/efficacy of four different AMF species (Rhizophagus irregularis, Septoglomus deserticola, Claroideoglomus claroideum or Claroideoglomus etunicatum) to colonize a variety of commercially available Geneva apple rootstock genotypes (G.11, G.41, G.210, G.969, and G.890). Briefly, micropropagated plantlets were inoculated with individual species of AMF or were not inoculated. The effects of the rootstock genotype/AMF interaction on mycorrhization, plant growth, and/or leaf nutrient concentrations were assessed. We found that both rootstock genotype and the identity of the AMF are significant sources of variation affecting the percentage of colonization. However, these factors largely operate independently in terms of the extent of root colonization. Among the AMF tested, C. etunicatum and R. irregularis represented the most compatible fungal partners, regardless of apple rootstock genotype. Among the rootstocks tested, semi-dwarfing rootstocks appeared to have an advantage over dwarfing rootstocks in regard to establishing and maintaining associations with AMF. Nutrient uptake and plant growth outcomes were also influenced in a rootstock genotype/AMF species-specific manner. This study investigated the properties of starch isolated from the unripe fruit of two apple cultivars (Malus domestica Borkh) grown in southern Poland (Central Europe). The chemical composition of both starches, molecular mass, their granulation, thermal characteristics, swelling characteristics, and rheological characteristics were studied. The starches differed significantly in ash, phosphorus, and protein content. The water-binding capacity at temperatures of 25–65 °C was similar, while differences of 20% appeared at higher temperatures. In contrast, a significant difference was found in the solubility of the two starches in the temperature range of 25–75 °C. The study showed that apple starches have a relatively low tendency to retrograde, with the enthalpy of gelatinization for starch from the Oliwka variety being 40% higher than that from the Pyros variety. However, the starches differed in the hardness of the gels formed, i.e., one variety formed soft gels with an internal structure resistant to external forces, while the other formed hard gels. The rhizosphere is a dynamic and highly interactive habitat where diverse microbial communities are established, and it plays crucial roles in plant health and disease dynamics. In this study, microbial communities and functional profiles in the rhizosphere of both asymptomatic and symptomatic apple trees were investigated through amplicon sequencing and shotgun metagenomics. The research was conducted at a location in the municipality of Cuauhtemoc, Chihuahua State, Mexico, and a total of 22 samples were collected, comprising 12 for amplicon sequencing and 10 for shotgun metagenomic sequencing. Symptomatic trees were identified based on reddish branches and internal necrosis in the trunk and root, while asymptomatic trees exhibited a healthy physiology. The findings showed that the dominant bacterial phyla included Proteobacteria, Actinobacteria, and Bacteroidetes, with prevalent genera such as Streptomyces, Pseudomonas, and Rhodanobacter. The fungal communities featured Ascomycota, Mortierellomycota, and Basidiomycota, which were dominated by Fusarium, Penicillium, and Mortierella. In the fungal communities, Mortierellomycota, notably abundant in asymptomatic trees, holds potential as a biocontrol agent, as seen in other studies on the suppression of Fusarium wilt disease. The application of shotgun metagenomic sequencing revealed significant differences in alpha and beta diversities in bacterial communities, suggesting a health-dependent change in species composition and abundance. Functional profile analysis highlighted enzymatic activities associated with lipid synthesis/degradation, amino acid biosynthesis, carbohydrate metabolism, and nucleotide synthesis, which have been documented to participate in symbiotic relationships between plants. These insights not only contribute to understanding the dynamics of rhizosphere microbial activity but also provide valuable perspectives on the potential application of microbial communities for tree health and implications for the management of apple orchards. Many viruses occur in apple (Malus domestica (Borkh.)), but no information is available on their seed transmissibility. Here, we report that six viruses infecting apple trees, namely, apple chlorotic leaf spot virus (ACLSV), apple green crinkle-associated virus (AGCaV), apple rubbery wood virus 2 (ARWV2), apple stem grooving virus (ASGV), apple stem pitting virus (ASPV), and citrus concave gum-associated virus (CCGaV) occur in seeds extracted from apple fruits produced by infected maternal trees. Reverse transcription polymerase chain reaction (RT-PCR) and quantitative RT-PCR (RT-qPCR) assays revealed the presence of these six viruses in untreated apple seeds with incidence rates ranging from 20% to 96%. Furthermore, ASPV was detected by RT-PCR in the flesh and peel of fruits produced by infected maternal trees, as well as from seeds extracted from apple fruits sold for fresh consumption. Finally, a large-scale seedling grow-out experiment failed to detect ACLSV, ASGV, or ASPV in over 1000 progeny derived from sodium hypochlorite surface sterilized seeds extracted from fruits produced by infected maternal trees, suggesting no detectable transmission via embryonic tissue. Fire blight, caused by Erwinia amylovora, is one of the most devastating apple diseases. The selection of cultivars of low susceptibility and the study of the genetic mechanisms of the disease play important roles in fire blight management. The susceptibility level to fire blight was evaluated in 102 accessions originating from Asturias, a cider-producing region located in the north of Spain with a wide apple germplasm. Evaluations took place under quarantine conditions using artificial inoculations of grafted plants. The results revealed wide variation in susceptibility responses and low-susceptible cultivars were identified. In addition, 91 cultivars were genotyped using the Affymetrix Axiom® Apple 480 K SNP array to conduct genome-wide association studies (GWAS). A statistically significant signal was detected on chromosome 10 using the multi-locus mixed model (MLMM). Two genes were identified as major putative candidate genes: a TIR-NBS-LRR class disease protein and a protein containing a development and cell death (DCD) domain. The outcomes of this study provide a promising source of information, particularly in the context of cider apples, and set a starting point for future genetic and breeding approaches. The impact of the ethylene inhibitor, 1-methylcyclopropene (1-MCP), on four apple cultivars (Braeburn, Fuji, Jazz and Golden Delicious) over 150 days of storage at 2 °C was assessed. Proton transfer reaction quadrupole mass spectrometry (PTR-QUAD-MS) was used to monitor changes in VOC composition, while texture analysis and X-ray microcomputer tomography (µ-CT) scanning were used to study microstructural changes. The application of 1-MCP on apples reduced VOC emissions, concurrently maintaining a firmer texture compared to the untreated apples at each time point. The µ-CT scanning revealed how changes in specific morphological characteristics such as anisotropy, connectivity and porosity, size and shape, as well as the interconnectivity of intracellular spaces (IS) influenced texture even when porosity was similar. Additionally, this study showed that the porosity and connectivity of IS were associated with VOC emission and increased simultaneously. This study highlights how the morphological parameters of an apple can help explain their ripening process during long-term storage and how their microstructure can influence the release of VOCs. This study investigated the composition of sugars, organic acids, individual and total phenolic compounds in the pulp, peel, leaves, and bark of a red-fleshed 'Baya Marisa' apple cultivar. As the fruit is known for its red pulp color, the study focused on comparing the profiles of primary and selected secondary metabolites in three sections along the equatorial fruit plane. The analyses were carried out using HPLC–MS system, and compositional differences were compared among fruit segments. Inner fruit segments accumulated higher levels of sorbitol and the sum of sugars and lower levels of citric acid. However, no differences in the sum of organic acids could be determined among segments. The phenolic composition differed among pulp (hydroxycinnamic acids > dihydrochalcones ≈ anthocyanins ≈ flavanols > flavonols), peel (flavanols > anthocyanins > dihydrochalcones > flavonols > hydroxycinnamic acids), leaves (flavonols > dihydrochalcones > hydroxycinnamic acids > anthocyanins), and bark (dihydrochalcones > flavonols > flavanols > anthocyanins > hydroxycinnamic acids). The greatest phenolic diversity was detected in bark (25), followed by leaves (18), peel (17) and finally, pulp (11). Three anthocyanins (cyanidin-3-O-galactoside > cyanidin-3-O-arabinoside > peonidin-3-O-galactoside) were determined in all 'Baya Marisa' tissues with their content highest in the peel. The innermost sections of the fruit were characterized by higher levels of dihydrochalcones and lower levels of most flavanols, flavonols and anthocyanins. These were predominant in the apple pulp nearest to the peel, with cyanidin-3-O-galactoside being the prevalent representative. Accumulation of anthocyanins in pulp is a rare trait in apples, and therefore, the distinct distribution and diversity of metabolites in this cultivar highlights its potential for high-nutrient products such as juices, apple chips or purees. Recent years have seen an increase in research focusing on the amelioration of apple pomace waste for use in the food and nutraceutical industries. Much of this work has concentrated on the characterisation of the polyphenol composition of apple pomace materials to determine their role in conferring nutritional and health benefits. Although apples contain substantial quantities of polymeric procyanidins (condensed tannins), this class of compounds has received limited attention in apple research. This study quantified the polymeric procyanidins in apple pomace extracts using a rapid, methyl-cellulose precipitation (MCP) approach for the first time. In addition, a non-targeted metabolomics approach was applied to determine the most abundant phenolic classes present. Polymeric procyanidins were found to be the most abundant type of polyphenol in apple pomace extracts and were generally oligomeric in nature. Multivariate statistical analysis revealed that the ferric-reducing antioxidant power (FRAP) was most strongly correlated with the polymeric procyanidin concentration. Noting that polymeric procyanidins may not cross the cell layer to exert antioxidant activity in vivo, their presence in apple pomace extracts may therefore overestimate the FRAP. This work highlights the importance of polymeric procyanidins in the phenolic diversity of apple pomaces, and it is proposed that in future studies, rapid MCP assays may be used for their quantification. Rapid apple decline is a phenomenon characterized by a weakening of young apple trees in high density orchards, often followed by their quick collapse. The nature of this phenomenon remains unclear. In this work, we investigated the root system architecture (RSA) of declining and non-declining apple trees in two orchards in New York State. High-density orchard A consisted of 4-year-old 'Honeycrisp' on 'Malling 9 Nic29', and conventional orchard B consisted of 8-year-old 'Fuji' on 'Budagovsky 9'. In both orchards, a negative correlation (−0.4–−0.6) was observed between RSA traits and decline symptoms, suggesting that declining trees have weaker root systems. Scion trunk diameter at the graft union, total root length, and the length of fine and coarse roots were significantly (p < 0.05) reduced in declining trees in both orchards. Additionally, internal trunk necrosis at, above, and below the graft union was observed in declining trees in orchard A but not in orchard B. Finally, latent viruses were not associated with decline, as their occurrence was documented in declining and non-declining trees in orchard A, but not in orchard B. Together, these results showed weakened root systems of declining trees, suggesting that these trees may experience deficiencies in water and nutrient uptake, although distinct RSA and trunk health traits between the two orchards were noticeable. Climate change has had a significant impact on apple phenology and fruit quality worldwide. Similarly, a decline in fruit quality has been observed in major apple-producing areas of Korea. It is predicted that the ideal cultivation areas for apples would need to shift toward higher latitudes due to these changes. Hence, the objective of this study was to assess the influence of climate change on apple cultivated in a higher-latitude region. To achieve this goal, we investigated the flowering and harvest times of apples. Additionally, we measured fruit weight, soluble solid content, and titratable acidity over a period of 20 years in a higher-latitude region in Korea. Subsequently, we examined the relationship between temperature, phenology, and fruit quality through the use of linear regression and correlation analysis methods. The 20-year meteorological data revealed a consistent rise in temperatures. Consistent with this trend, we found a significant advancement in budbreak and flowering dates for the two major apple cultivars grown in a higher latitude in Korea. Furthermore, the increase in temperatures has positively influenced fruit quality, indicating that climate change has an impact not only on phenology, but also on the quality of apples in Korea. Considering the projected gradual increase in temperature, our findings strongly support that higher-latitude regions in Korea have the potential to become optimal locations for apple cultivation. Apples represent a category of products frequently consumed by people, owing both to their beneficial effects on human health and to their antioxidant effects. Microwave (MW) treatment is a simple and fast method that can be used successfully in the food industry to obtain dry apple slices, rich in bioactive compounds and with a pleasant aspect. This study presents the effect of applying microwave treating to apple slices from two cultivars (Golden Delicious and Idared), for three, four, and five minutes, respectively, at a power of 450 W, in order to reduce the browning effect. For this purpose, the browning index (BI), chromatic parameters (CIE L*a*b*), total phenolic content (Folin-Ciocâlteu method), and antioxidant capacity (by Ferric Reducing Antioxidant Power (FRAP assay) were evaluated in the case of apple slices before and after MW treatment. Based on the results obtained, it can be argued that the microwave treatment results in a significant increase in the total phenolic content and enhances antioxidant capacity in the case of both apple cultivars. Apples from the Idared cultivar have a higher total phenolic content than apples from the Golden cultivar, and this concentration increased by 56.14% and 48.9%, respectively, after MW treatment. In terms of antioxidant capacity, Idared apples also recorded a higher value compared to Golden cultivars. According to the results of the multivariate analysis, there are variations between the two apple categories with regard to the phenolic content and the browning process; browning was inhibited at the five-minute exposure to microwaves. Commercial fruit production in Norway is located at around latitude 60° north, demanding a careful choice of adapted cultivars. The most comprehensive collection of apple genetic resources in Norway is being kept in the Norwegian Apple Collection (NAC) at the Njøs Fruit and Berry Centre (NJØS). The collection contains around 350 accessions and was recently genotyped with a single nucleotide polymorphism (SNP) array. Curated SNP data were used for the assessment of structure and diversity, pedigree confirmation, and core collection development. In the following SNP analysis, we identified several duplicates and parent-child relationships. Across the geographic regions represented, the collection was equally diverse. Different methods for analyzing population structure were applied. K-means clustering and a Bayesian modeling approach with prior assumptions of the data revealed five subpopulations associated with geographic breeding centers. The collection has a distinct genetic structure and low relatedness among the accessions; hence, two core collections with 100 accessions in each were created. These new core collections will allow breeders and researchers to use the NAC efficiently. Apple accessions, currently maintained within the two main ex situ collections in Bosnia and Herzegovina (B&H), have previously been genotyped using microsatellite markers. The obtained molecular data provided insight into mislabeled accessions and redundancies, as well as the overall genetic structure of the germplasm. The available dataset enabled the creation of a core collection consisting of 52 accessions. The reliability and usefulness of microsatellites has made this low-density marker system a norm in studies on apple germplasm. However, the increased access to medium- and high-density SNP arrays, developed specifically for apples, has opened new avenues of research into apple genetic resources. In this study, 45 apple genotypes consisting of 33 diploid core collection accessions from B&H and 12 international reference cultivars were genotyped using an Axiom® Apple 480 K SNP array in order to examine their genetic relationships, population structure and diversity, as well as to compare the obtained results with those calculated on previously reported SSR profiles. The SNPs displayed a better ability to differentiate apple accessions based on their origin, as well as to cluster them according to their pedigree. Calculating identity by descent revealed 16 pairings with first-degree relationships and uncovered the introgression of 'Delicious' and 'Golden Delicious' into the core collection. Triterpenoids are a group of secondary plant metabolites, with a remarkable pharmacological potential, occurring in the cuticular waxes of the aerial parts of plants. The aim of this study was to analyze triterpenoid variability in the fruits and leaves of three apple cultivars during the growing season and gain new insights into their health-promoting properties and fate during juice and purée production. The identification and quantification of the compounds of interest were conducted using gas chromatography coupled with mass spectrometry. The waxes of both matrices contained similar analytes; however, their quantitative patterns varied: triterpenic acids prevailed in the fruits, while higher contents of steroids and esterified forms were observed in the leaves. The total triterpenoid content per unit area was stable during the growing season; the percentage of esters increased in the later phases of growth. Antioxidative and anti-inflammatory properties were evaluated with a series of in vitro assays. Triterpenoids were found to be the main anti-inflammatory compounds in the apples, while their impact on antioxidant capacity was minor. The apples were processed on a lab scale to obtain juices and purées. Urban horticulture has to respond to several challenges, including reducing the growth vigor of the scion/rootstock combination, adapting the cultivated species/variety to the pedoclimatic conditions—more varied in recent years, and increasing tolerance to diseases and pests that cause extensive qualitative and quantitative damage to fruit production. For this experiment, 15 old apple varieties were chosen, all known and cultivated in the Banat area, which were grafted on the MM106 rootstock. Several parameters were analyzed that can be used for a relevant classification for planting them in limited spaces, such as in a private garden. Our research started 10 years ago and has already resulted in a doctoral thesis that extensively presents the data taken from the initial biological material and the first two years after grafting the trees; the research continued and is still ongoing in the experimental orchard. From the multitude of aspects studied, we have chosen to present some results related to the vigor of the scion/rootstock combination, the quality of the fruits, their content in acidity as well as the behavior with respect to diseases and pests—results that can be useful to those who want to grow old apple varieties in their own garden. Growth vigor was reduced in most varieties, but the highest influence of the rootstock on growth was observed in the 'Bănățenesc' variety, followed by 'Jonathan de Munte' and 'Caslere'. Grafting and agrotechniques had a positive influence on the fruit mass in all cultivars, except 'Crețesc', with an experimental average of 163.35 g, namely 60.56 g higher on average than that of the original biological material. After grafting, the SSC was between 10.05 °Brix in the 'Curcubătoase' variety and 18.48 °Brix in 'Parmen Auriu', with an experimental average of 14.96 °Brix. The issue of high concentration of salt in soil is not restricted to coastal areas, but also expands to cultivated lands, complicating, or even intercepting, the growth of plants. The objective of this paper is to study the effect of zeolite, compost and effective microorganisms (EM), seaweed extract, and ceramic powder on MM106 apple (Malus domestica Borkh.) plants in normal and saline conditions. More specifically, the weight of the dry matter of the plants, physiological parameters, proline, carbohydrate, carotenoid, phenolic, and flavonoid concentrations in leaf tissues and antioxidant capacity were determined. At the end of the experiment, it was ascertained that the plants of the treatments which included zeolite or EM exhibited the highest dry matter weight of the leaves in normal (5.07 g and 4.68 g, respectively) and saline conditions (4.14 g and 3.02 g, respectively), while the leaf dry weight in the control treatment was 4.37 g in the absence and 2.34 g in the presence of NaCl. Furthermore, these treatments resulted in significantly higher proline concentration in plant leaves under salinity with values of 5.63 in the EM treatment, 2.44 in the zeolite treatment, and 0.75 μmol/g of leaf fresh weight in the control. At the same time, the application of ceramic powder in combination with effective microorganisms led to the highest rate of photosynthesis in salinity conditions (12.8 μmol CO2/m2s), while the seaweed extract spraying was associated with low stomatal conductance in all treatments (0.09–0.13 mol H2O/m2s). Overall, the application of effective microorganisms appeared to associate more with plant vigor in both normal and salinity conditions. The complete netting of orchards is one strategy to protect fruit trees from pest and pathogen damage by reducing insect movement. When the cultivated trees were derived from genetic engineering (GE), reduced pollinator movement may also reduce outcrossing to cultivated or wild non-GE trees. We report on a field study over four years in a plot of apple tress supplied with insect side nets and covered with hail nets that were closed from shortly before flowering to harvest. A reduced number of arthropods in general, and large bees in particular, were recorded inside the netted plot compared with outside. However, wild bees colonized the plot before the net was closed and built up populations inside. An additional experiment demonstrated that small bees were able to cross the hail net. While the nets were effective in excluding large bees as active pollen vectors, the proportion of small bees acting as such remained unquantified. Furthermore, a companion study showed occasional cross-pollination events through the netting. For the field release of GE apple trees, acceptable levels of outcrossing thus need to be defined. There is a vast diversity of historic heirloom apple varieties around the world. Apples of various colors, sizes, tastes, uses, and ripening times can be found. In Slovakia, there has recently been a rising interest to preserve these apple varieties. The life of a tree is partially influenced by the growing conditions of the environment where the tree grows. In this paper, macroscopic and microscopic characteristics of four selected heirloom apple trees growing in Slovakia's Štiavnické vrchy were observed. Studying the microscopic structure of historical heirloom apple trees wood could reveal specific cell arrangements of tissues. This could help to specify the individual varieties. This study evaluated unripe fruits of ten apple cultivars removed during fruit thinning for their antioxidant activity and mineral content. The unripe fruits were collected from Chuncheon and Jeongseon located at Gangwon-Do of Korea. Antioxidant activities such as total phenolic content (TPC), total flavonoid content (TFC), DPPH (2,2-diphenyl-1-picrylhydrazyl) activity, FRAP (Ferric reducing antioxidant power), vitamin C and mineral contents were measured. In the unripe fruits obtained from Chuncheon and Jeongseon, TPC was in the range 8.97–81.4 and 7.11–42.15 mg GAE/g, TFC was in the range 9.38–33.81 and 6.83–19.24 mg QE/g, DPPH was in the range 27.17–82.58 and 29.73–73.24, FRAP was in the range 33.54–371.12 and 26.76–185.69 µM trolox /g, and Vitamin C was in the range 1.1–4.9 and 1.1–2.8 mg/AA/g, respectively. Among the cultivars tested, 'Hongro' and 'Honggeum' had consistently highest antioxidant activity, while 'Summer King,' 'Tsugaru,' and 'Arisoo' had the lowest value. 'Picnic' had the highest mineral contents expect P and K in Joengsoen, while 'Summer King' and 'Tsugaru' had the lowest value in both locations. These differences could be due to the genetic characteristics and/or their growth environments. Although the application of chemical fertilizers to crops promotes plant growth and yield, their continuous use affects soil heath and creates environmental pollution. On the other hand, plant biostimulants improve nutrients absorption, plant growth, yield and produce quality and are environment-friendly. Therefore, an experiment was conducted during 2021-22 to evaluate the effect of some biostimulants on the performance of the apple cv. Anna, planted in a sandy loam soil at Marsa Matruh governorate, Egypt. Ninety trees were randomly selected and sprayed with 4 or 6% moringa leaf extract (MLE), 0.3 or 0.4% seaweed extract (SWE), 0.1% or 0.2% Fulvic acid (FA), 4% MLE + 0.3% SWE + 0.1% FA (combination 1), or 6% MLE + 0.4% SWE + 0.2% FA (combination 2) before flowering, during full bloom and one month later and compared with a control (untreated trees). The results demonstrated that spraying MLE, SWE or FA or their combinations positively improved the vegetative growth, fruit set %, fruit yield and fruit physical and chemical characteristics as well as leaf nutritional status. The positive effect of MLE, SWE and FA was increased in parallel to an increase in the used concentration of each one of them. The present study aims to investigate the effectiveness of bioformulations based on endophytic fungi to control apple scab and Valsa canker disease in two orchards in the Aurès region (Algeria). In both orchards, the results showed that the treatment of senescent apple leaves by invert emulsions containing Trichoderma longibrachiatum and Chaetomium globosum harmed the ascogenesis of winter forms of Venturia inaequalis by reducing the number of ascospore-ejecting asci, the number of morphologically mature asci, and a considerable increase in the immature asci number. This antifungal activity was more essential in soil-incorporated leaves, showing the importance of the combination of treatments with cultural practices to efficiently control the apple scab disease. Furthermore, the disease incidence decreased by 52.63% and 50.68% in R'haouat and Bouhmama orchards, respectively. Moreover, the treatment of Valsa ceratosperma cankers with a biogel containing the endophytic yeast Metschnikowia sp. led to wound healing varying from 43.52% and 87.97% after 120 days but remained more considerable than conventional treatment with Folicur (tebuconazol). The phytohormone abscisic acid (ABA) plays an important role in the ability of plants to cope with drought stress. As core members of the ABA signaling pathway, protein phosphatase type 2Cs (PP2Cs) have been reported in many species. However, the functions of MdPP2Cs in apple (Malus domestica) are unclear. In this study, we identified two PP2C-encoding genes, MdPP2C24/37, with conserved PP2C catalytic domains, using sequence alignment. The nucleus-located MdPP2C24/37 genes were induced by ABA or mannitol in apple. Genetic analysis revealed that overexpression of MdPP2C24/37 in Arabidopsis thaliana led to plant insensitivity to ABA or mannitol treatment, in terms of inhibiting seed germination and overall seedling establishment. The expression of stress marker genes was upregulated in MdPP2C24/37 transgenic lines. At the same time, MdPP2C24/37 transgenic lines displayed inhibited ABA-mediated stomatal closure, which led to higher water loss rates. Moreover, when exposed to drought stress, chlorophyll levels decreased and MDA and H2O2 levels accumulated in the MdPP2C24/37 transgenic lines. Further, MdPP2C24/37 interacted with MdPYL2/12 in vitro and vivo. The results indicate that MdPP2C24/37 act as negative regulators in response to ABA-mediated drought resistance. Phosphate (P) deficiency severely limits the growth and production of plants. Adventitious root development plays an essential role in responding to low phosphorus stress for apple plants. However, the molecular mechanisms regulating adventitious root growth and development in response to low phosphorus stress have remained elusive. In this study, a mutation (C-T) in the coding region of the apple AUXIN/INDOLE-3-ACETIC ACID 27 (IAA27) gene was identified. MdIAA27T-overexpressing transgenic apple improved the tolerance to phosphorus deficiency, which grew longer and denser adventitious roots and presented higher phosphorous content than the control plants under low phosphorus conditions, while the overexpression of MdIAA27C displayed the opposite trend. Moreover, the heterologous overexpression of MdIAA27 in tobacco yielded the same results, supporting the aforementioned findings. In vitro and in vivo assays showed that MdIAA27 directly interacted with AUXIN RESPONSE FACTOR (ARF8), ARF26 and ARF27, which regulated Small Auxin-Up RNA 76 (MdSAUR76) and lateral organ boundaries domain 16 (MdLBD16) transcription. The mutation in IAA27 resulted in altered interaction modes, which in turn promoted the release of positive ARFs to upregulate SAUR76 and LBD16 expression in low phosphorus conditions. Domesticated international (standard) apple cultivars, together with resistant apple cultivars are the core of the Serbian apple production. Furthermore, autochthonous cultivars are characterized by a good adaptability to the local environmental conditions and represent a valuable source of genetic variability, as well as an important source of the gene pool for further breeding programs. Additionally, they show a higher phenolic content and a stronger antioxidant activity, in comparison to commercial cultivars. Therefore, they are more likely to be used as a functional food. The subjects of this study were seventeen samples of fruits and leaves from autochthonous apple cultivars, five international standard cultivars, and six resistant apple cultivars. The phenolic profile was determined using ultra-high performance liquid chromatography (UHPLC), coupled with a diode array detector and a TSQ Quantum Access Max triple-quadrupole mass spectrometer. A total of twenty compounds were quantified in the samples. Most of the analyzed phenolics were detected in higher amounts in the peel, compared to the mesocarp. The results of the multivariate analysis of variance (MANOVA) indicate that 5-O-caffeoylquinic acid is present in the highest amount in the mesocarp, while in the peel and leaves, quercetin-glycosides were detected in the highest amount. According to the MANOVA: phloretin, phlorizin, 5-O-caffeoylquinic acid, kaempferol, and p-coumaric acid are present in significantly higher levels in the autochthonous cultivars, compared to the standard and resistant ones (in both fruits and leaves). Increasing the reliability of fruit crops in unstable weather conditions of a temperate continental climate has become particularly relevant. This research was carried out based on a bioresource collection from the Russian Research Institute of Fruit Crop Breeding (VNIISPK BRC). Apple cultivars (diploids and triploids) of different maturation periods were studied. Triploid cultivars (3×) of summer maturation were developed using diploid gametes of the 'Papirovka tetraploid' cultivar as a donor (2-4-4-4×), triploids of winter maturation were developed using 13-6-106 (Suvorovetz: open pollination), and 'Wealthy tetraploid' (2-4-4-4×) or 'Giant Spy' (2-4-4-4×) were used as donors of diploid gametes. 'Antonovka' and 'Melba' were taken as control cultivars. The aim of this work was to evaluate the apple breeding cultivars according to the main economic and biological indicators and to determine the threshold of resistance to unfavorable conditions of the winter period for possible cultivation in specific conditions of a temperate continental climate. As a result of using the method of modeling damaging factors, apple cultivars that withstood not only the critical temperatures of January, but also especially dangerous frosts after thaws at the end of winter were identified, with the stability of vital tissues at the level of 'Antonovka' (a control cultivar), scab immune cultivars (RVi6) 'Ivanovskoye', 'Zdorovie' and 'Pamyati Hitrovo', and immune triploids (3×) 'Vavilovskoye', 'Aleksandr Boyko', Rozhdestvenskoye' and 'Academic Saveliev'. During the whole winter, the winter resistance of summer scab immune triploids, 'Zhilinskoye', 'Maslovskoye', 'Yablochny Spas' and 'Spasskoye', was at the level of 'Antonovka', but was higher than 'Melba'. The high precocity of scab-immune cultivars, 'Afrodita', 'Ivanovskoye', 'Veniaminovskoye', and 'Yubiley Moskvy', and triploids 'Patriot' and 'Rozhdestvenskoye', have been identified. Triploid cultivars are superior to diploid cultivars and control 'Antonovka' in fruit size. According to the complex of characteristics, promising apple cultivars were identified for planting in intensive orchards of the temperate continental climate zones, namely 'Avgusta' and 'Solnyshko' (summer cultivars), and 'Aleksandr Boyko', 'Vavilovskoye', 'Venyaminovskoye', 'Ivanovskoye' and 'Rozhdestvenskoye' (winter cultivars). In apple cultivation, dwarf rootstocks are chosen for dense planting and intensive cultivation, which is beneficial to production management. Dwarf rootstocks are widely used in apple production in China. However, the dwarfing mechanisms of dwarf interstock are still unclear. Here, M9 and SH40 were selected as the dwarf interstocks for potted Fuji apples. The key transcription factor MdWRKY24 was screened via transcriptional sequencing. The open reading frame sequence of the MdWRKY24 gene was 657 bp in length, encoded 218 amino acids, and was located on the cell membrane. The MdWRKY24-overexpressing Arabidopsis line showed a dwarf phenotype and delayed flowering. The DELLA protein RGA-like (RGL) gene is a repressor of the gibberellin signaling pathway. Yeast two-hybrid analysis revealed that MdWRKY24 could interact with MdRGL1/2/3. The results indicated that MdWRKY24 might affect plant dwarfing through the synergistic effect of MdRGL1/2/3. The protein Mal d 1 is responsible for most allergic reactions to apples (Malus domestica) in the northern hemisphere. Mal d 1 contains a cysteine residue on its surface, with its reactive side chain thiol exposed to the surrounding food matrix. We show that, in vitro, this cysteine residue is prone to spontaneous chemical modification by ascorbic acid (vitamin C). Using NMR spectroscopy and mass spectrometry, we characterize the chemical structure of the cysteine adduct and provide a three-dimensional structural model of the modified apple allergen. The S-ascorbylated cysteine partially masks a major IgE antibody binding site on the surface of Mal d 1, which attenuates IgE binding in sera of apple-allergic patients. Microbial food spoilage has been a major source of concern because it widens the global food production-to-consumption gap. Thus, in the current study, we aimed to isolate, characterize and identify bacteria associated with spoiled apple fruit in Al-Ahsa city, Saudi Arabia. MDK33, a bacterial strain isolated from rotten apple fruit, was characterized at the phenotypic and genotypic levels. Furthermore, the antibacterial effect of oregano essential oil (OEO) against MDK33 was investigated. MDK33 formed circular colonies with entire margins, and the cells were Gram-negative rods with no endospores. Biochemical characterizations of MDK33, as revealed by the Biolog Gen III MicroPlate test system, indicated that the strain utilized 66 (~70%) of the 94 diverse metabolites and did not utilize 29 (~30%). MDK33 grew well on a variety of substrates, including pectin, gelatin, Tween 40 and dextrin, at pH 5 and 6; tolerated salts up to 4% NaCl; and was resistant to multiple antibiotics. Furthermore, based on the sequences of the 16S rRNA gene, MDK33 was identified as Serratia liquefaciens at a 99.73% identity level. Koch's postulates were affirmed, confirming that S. liquefaciens MDK33 is the causative agent of apple rot disease in Al-Ahsa, Saudi Arabia. The minimum inhibitory concentration (MIC) of 0.23 mg/mL for oregano oil against S. liquefaciens MDK33 demonstrated remarkable antibacterial and antibiofilm activities. This is the first isolation of pathogenic S. liquefaciens as the causative agent of rot disease of apple fruit and its inhibition by oregano oil. Furthermore, the findings pave the way for oregano oil to be evaluated as a natural preservative to reduce post-harvest losses while maintaining high quality for sustainable food security in future studies. pH of xylem sap (pHx) was determined in three trees (Malus domestica (apple tree), Picea abies and Pinus cembra) in response to seasonal changes. Conifer trees from lowland (600 m) were compared to trees growing at the alpine timberline (1950 m a.s.l.). Xylem sap was extracted with a Scholander pressure bomb and pHx was measured with a pH microsensor. In all species, pHx changed markedly with season. In spring, pHx was acidic; during winter, the pHx was more alkaline. In apple trees, the pHx did not show a significant correlation with temperature but was rather affected by developmental stage. During flushing in spring, xylem sap acidification took place concomitant to the developmental stage "tight cluster", when foliar development enables a significant transpiration and a consequent movement of water in the xylem. The xylem sap of the two studied conifers showed a significantly larger seasonal alkalinisation (+2.1) than found in apple trees (+1.2) and was significantly more pronounced at the timberline. Xylem sap acidification took place before bud break. pHx had a significant negative correlation with soil temperatures and corresponded to already reported pHx of angiosperms. Aluminum (Al) is an important element in soil constitution. Previous studies have shown that high concentration of Al affects the normal growth of crops, resulting in crop yield reduction and inferior quality. Nevertheless, Al has also been referred to as a beneficial element, especially when used at low concentrations, but the cytological mechanism is not clear. Influences of low concentration AlCl3 on the pollen tube growth of apple (Malus domestica) and its possible cytological mechanism were investigated in this study. The results showed that 20 μM AlCl3 promoted pollen germination and tube elongation; 20 μM AlCl3 enhanced Ca2+ influx but did not affect [Ca2+]c of the pollen tube tip; and 20 μM AlCl3 decreased acid pectins in pollen tubes but increased esterified pectins and arabinan pectins in pollen tubes. Glomerella leaf spot (GLS), caused by the fungus Colletotrichum fructicola, is one of the most devastating apple diseases. Our previous study reported that the GLS resistance locus was defined on the chromosome 15 region. Here, we further found a single-nucleotide polymorphism (SNP) site (SNP7309212) in the GLS resistance that was able to distinguish resistant cultivars (lines) from susceptible ones. On the basis of the SNP site, we cloned a TNL gene from the GLS resistant locus and named it MdTNL1 (NCBI Accession Number: ON402514). This gene contains a toll/interleukin-1 receptor transmembrane domain (TIR), nucleotide-binding sites (NBS), and leucine-rich repeat (LRR) domain. Subcellular location indicated that MdTNL1 was expressed in the nucleus and cell membrane. Ectopic overexpression of MdTNL1 in Nicotiana benthamiana caused cell death. We further demonstrated allelic polymorphisms in MdTNL1. It is noteworthy that NBS and LRR domains of the MdTNL1 protein serve as the repository for generating allelic diversity. Quantitative real-time PCR (qRT-PCR) assay revealed that MdTNL1 was highly expressed in resistant apple cultivar 'Fuji' after inoculation with C. fructicola, whereas susceptible cultivar 'Golden Delicious' exhibited low expression after inoculation. Over-expression of MdTNL1-1 in susceptible apple fruits and leaves improved disease resistance, while in 'Orin' calli, silencing the MdTNL1-1 gene conversely decreased GLS resistance. In conclusion, we identified a GLS associated with SNP7309212 and demonstrated that a TIR-NBS-LRR gene MdTNL1-1 positively regulates GLS resistance in apple. [ABSTRACT FROM AUTHOR]

Bull's-eye rot, produced by Phlyctema vagabunda, is an important postharvest disease in apples. Current measures to control infection include synthetic fungicides, in addition to the application of copper hydroxide and potassium phosphite. However, growing public concern regarding fungicide residues in food has generated interest in developing non-chemical alternative control methods; biological control is one of the most promising alternatives. In this research, native endophytic yeasts were isolated and evaluated for the biocontrol of P. vagabunda in apples. The mechanisms of action involved were also determined. Our research found 2 isolates, Vishniacozyma victoriae EPL4.5 and EPL29.5, which exhibited biocontrol activity against P. vagabunda at 20 °C in apples, the incidence of bull's-eye rot was reduced by 39% and 61%, respectively, and the severity of the disease was decreased by 67% and 70%, respectively, when apples were inoculated with these yeasts 24 h before applying the pathogen. Plant MAPK cascade performs a critical role in the regulation of plant immunity and disease resistance. Although the function of MAPK cascade in immunity regulation is partially conserved between different species, the mechanism varies in different host and pathogen combinations. To date, the MAPK cascade function of woody plants in the regulation of disease resistance has seldom been reported. Here, we present evidence to show that apple MdMAPKKK1 performed an important role in the regulation of apple resistance to Botryosphaeria dothidea, the causal agent of apple ring rot. B. dothidea infection leads to enhanced MdMAPKKK1 expression and MAPK cascade activation, indicating that the MAPK cascade is involved in the defense against B. dothidea. MdMAPKKK1 overexpression-induced pathogen-independent cell death. MdMAPKKK1 silencing decreases the resistance of apple calli and fruits to B. dothidea. Further analysis indicates that MdMAPKKK1 can bind MdBSK1 and is likely phosphorylated by it. The MdBSK1-mediated phosphorylation of MdMAPKKK1 is important for resistance to B. dothidea. Early varieties with a short active vegetation period are not exposed to abiotic stress factors for instance drought, high temperature, etc., caused by global climate change, and biotic stress factors such as codling moth (Cydia pomonella) and apple scab (Venturia inequalis) that cause serious economic losses. Therefore, their places are extremely important regarding sustainable, economical and quality production. In this context, four early varieties of apple ('Vista Bella', 'Summer Red', 'Williams Pride' and 'Jersey Mac') grown in Eskisehir—Turkey where has continental climate were characterized in terms of horticultural characteristics. The difference observed in flowering and harvest times between years showed that average air temperatures are essential on phenological characteristics. Furthermore, high temperature accelerates development physiology of fruit. Major important characteristics, namely, fruit length, width, weight, soluble solid content, antioxidant activity, vitamin C and total phenolic content were found in the range of 39.19–50.88 mm, 51.48–60.81 mm, 80.59–86.16 g, 11.87–12.19%, 50.69–80.57%, 2.24–4.79 mg 100 mL−1 and 259.23–871.42 mg GAE (gallic acid equivalent) L−1, respectively. 'Summer Red' and 'Williams Pride' varieties were found to be good in terms of pomological characteristics, while 'Vista Bella' came to the fore with its chemical properties. On the other hand, 'Jersey Mac' has shown superior performance in terms of both pomological and chemical characteristics. Pomological characteristics are affected more by ecological differences, while chemical characteristics vary according to the changes in the pomological properties. The various fungal communities that adhere to apple fruit are influenced by agricultural practices. However, the effects of fruit bagging-based management practice on the fungal microbiota are still unknown, and little is known about the fungal communities of bagged apple fruit. We conducted a study using apple fruit grown in a conventionally managed orchard where pesticide use is an indispensable practice. Fungal communities were collected from the calyx-end and peel tissues of bagged and unbagged fruit and characterized using barcode-type next-generation sequencing. Fruit bagging had a stronger effect on fungal richness, abundance, and diversity of the fungal microbiota in comparison to non-bagging. In addition, bagging also impacted the compositional variation of the fungal communities inhabiting each fruit part. We observed that fruit bagging had a tendency to maintain ecological equilibrium since Ascomycota and Basidiomycota were more distributed in bagged fruit than in unbagged fruit. These fungal communities consist of beneficial fungi rather than potentially harmful fungi. Approximately 50 dominant taxa were detected in bagged fruit, for example, beneficial genera such as Articulospora, Bullera, Cryptococcus, Dioszegia, Erythrobasidium, and Sporobolomyces, as well as pathogenic genera such as Aureobasidium and Taphrina. These results suggested that fruit bagging could significantly increase fungal richness and promote healthy fungal communities, especially the harmless fungal communities, which might be helpful for protecting fruit from the effects of pathogens. Nitrogen is an integral element of foliar sprays during apple fruit formation. However, in red cultivars, the application may have adverse effects on color in the second stage of fruit development. The effect of a low-dose foliar application of urea was monitored on ‘Topaz’ apple pomometric (fruit mass, firmness, total solids, starch content, skin color) and biochemical traits (primary metabolites, phenolic compounds, and assimilatory pigments). Three applications (17 July, 27 July, and 18 August) of two different urea doses (N1 = 1.4 kg N ha-1; N4 = 4 kg N ha-1) and control (N0, only sprayed with water) were used. Low doses of urea did not affect flesh firmness, total soluble, solid, and starch content, but individual fruit mass was significantly higher in N1 treatment compared to the other two treatments. Significantly lower a* parameters and a lower content of anthocyanins were measured on apples subject to N4 treatment. Many secondary metabolites (phenolic acids, flavan-3-ols, flavonols, and chalcones) were also negatively affected by low-dose urea application. Conversely, urea treatments significantly increased total assimilatory pigments in apple skin. Even minor levels of nitrogen, applied directly on the leaf at later stages of red apple fruit development, negatively altered color traits and the composition of metabolites. [ABSTRACT FROM AUTHOR]

The apple (Malus domestica Borkh.) is one of the most widely cultivated temperate fruits globally, gaining scientific interest as a rich source of antioxidants with a demonstrated beneficial human health impact. Since a growing number of consumers are increasingly seeking safe and healthy food options, alternative fruit production systems such as organic farming, and their potential to provide safe and nutritious foods, have been gaining increasing attention in the last decades. The aim of the presented study was, therefore, to analyse and to compare the concentrations of selected health-promoting antioxidants, such as phenolic acids, flavonols, and vitamin C, in fruits of three apple cultivars (Champion, Gala, and Idared) grown in conventional and certified organic orchards in Poland. All analyses were performed using the high-performance liquid chromatography (HPLC) method. Organic apples tested within the study, compared to the conventionally grown ones, were characterised by significantly higher concentrations of phenolic acids (av. >31%) and flavonols (av. >66%) with the identified differences being consistent in all three cultivars and two seasons. The greatest production system effect was observed in the case of Idared. Significant cultivar and production season effects on the concentration of the measured fruit phenolics, with strong interactions between the two factors, were also identified. Vitamin C content in the fruits was strongly dependent on the year-to-year differences in the fruit growing conditions. The phenolic composition of skin and flesh from Malus domestica apples (Anna cultivar) and Prunus domestica plums (satsuma cultivar) commercial cultivars in Costa Rica, was studied using Ultra Performance Liquid Chromatography coupled with High Resolution Mass Spectrometry (UPLC-DAD-ESI-MS) on enriched-phenolic extracts, with particular emphasis in proanthocyanidin and flavonoids characterization. A total of 52 compounds were identified, including 21 proanthocyanidins ([(+)-catechin and (-)-epicatechin]) flavan-3-ols monomers, five procyanidin B-type dimers and two procyanidin A-type dimers, five procyanidin B-type trimers and two procyanidin A-type trimers, as well as one procyanidin B-type tetramer, two procyanidin B-type pentamers, and two flavan-3-ol gallates); 15 flavonoids (kaempferol, quercetin and naringenin derivatives); nine phenolic acids (protochatechuic, caffeoylquinic, and hydroxycinnamic acid derivatives); five hydroxychalcones (phloretin and 3-hydroxyphloretin derivatives); and two isoprenoid glycosides (vomifoliol derivatives). These findings constitute the first report of such a high number and diversity of compounds in skins of one single plum cultivar and of the presence of proanthocyanidin pentamers in apple skins. Also, it is the first time that such a large number of glycosylated flavonoids and proanthocyanidins are reported in skins and flesh of a single plum cultivar. In addition, total phenolic content (TPC) was measured with high values observed for all samples, especially for fruits skins with a TPC of 619.6 and 640.3 mg gallic acid equivalents/g extract respectively for apple and plum. Antioxidant potential using 2,2-diphenyl-1-picrylhidrazyl (DPPH) and oxygen radical absorbance capacity (ORAC) methods were evaluated, with results showing also high values for all samples, especially again for fruit skins with IC50 of 4.54 and 5.19 μg/mL (DPPH) and 16.8 and 14.6 mmol TE/g (ORAC) respectively for apple and plum, indicating the potential value of these extracts. Significant negative correlation was found for both apple and plum samples between TPC and DPPH antioxidant values, especially for plum fruits (R = -0.981, p < 0.05) as well as significant positive correlation between TPC and ORAC, also especially for plum fruits (R = 0.993, p < 0.05) and between both, DPPH and ORAC antioxidant methods (R = 0.994, p < 0.05).
