Simple Summary: Monitoring animals' welfare is a tricky challenge. The responsiveness to stressors is subjective and its magnitude varies among species, breeds, and individuals. Small molecules named miRNAs obtained from body fluid samples are promising biomarkers to classify animal welfare. Under common farming conditions, we identified several miRNAs in the saliva of ewes. Based on the concentration and trend of individual salivary cortisol, one of the molecules historically recognized and used to define the stress level in vertebrates, we divided the animals into two clusters. The expression of two miRNAs involved in regulating genes related to the stress pathway resulted in significant differences between the two phenotypes, and their trends were correlated. These results have produced objective data to improve the knowledge about sheep physiology and the basis to develop new tools for welfare assessment. In the animal production field, the assessment of biomarkers that are useful in identifying individuals with different levels of vulnerability or resiliency to stress stimuli may help to introduce new strategies in animal farms and genetic research for the selection of animals. Farm procedures have an impact on animal welfare by activating the hypothalamic-pituitary-adrenal axis that induces a wide array of physiological responses. This adaptive system guarantees that the animal copes with environmental variations and it induces metabolic and molecular changes that can be quantified. MicroRNAs (miRNAs) play a key role in the regulation of homeostasis and emerging evidence has identified circulating miRNAs as promising biomarkers of stress-related disorders in animals. Based on a clustering analysis of salivary cortisol trends and levels, 20 ewes were classified into two different clusters. The introduction of a ram in the flock was identified as a common farm practice and reference time point to collect saliva samples. Sixteen miRNAs related to the adaptation response were selected. Among them, miR-16b, miR-21, miR-24, miR-26a, miR-27a, miR-99a, and miR-223 were amplified in saliva samples. Cluster 1 was characterized by a lower expression of miR-16b and miR-21 compared with Cluster 2 (p < 0.05). This study identified for the first time several miRNAs expressed in sheep saliva, pointing out significant differences in the expression patterns between the cortisol clusters. In addition, the trend analyses of these miRNAs resulted in clusters (p = 0.017), suggesting the possible cooperation of miR-16b and -21 in the integrated stress responses, as already demonstrated in other species as well. Other research to define the role of these miRNAs is needed, but the evaluation of the salivary miRNAs could support the selection of ewes for different profiles of response to sources of stressors common in the farm scenario. Simple Summary: Local chicken breeds Bionda Piemontese (BP) and Bianca di Saluzzo (BS) are promising for use in small-scale poultry farms, which adopt free-range and/or organic farming practice. However, in practical conditions, it can happen that animals are subjected to regrouping, creating a new flock by mixing different groups. Such situations may provoke a stressful response in birds, compromising their welfare. To evaluate the resilience of BP and BS breeds to adverse management practices (regrouping), the present study aimed to assess the birds' welfare status through a multifactorial approach. As a result, the practice of regrouping immediately compromised the welfare status and the productive performances in both BP and BS hens, but, in the following weeks, birds displayed a reliance, which help them adapt to the new stressful conditions. The aim of the present study was to investigate the ability of two Italian slow-growing poultry breeds (namely, Bionda Piemontese, BP, and Bianca di Saluzzo, BS) to cope with a stressful event, such as collective grouping, using a multifactorial approach. A total of 120 hens of BP and BS breeds were homogenously distributed, according to breed, in 12 pens (10 hens/pen; 6 pens/breed), from 18 to 49 weeks of age. At 50 weeks of age, hens were regrouped (Stressful Farm Practice, SFP), by removing separators, both in indoor and outdoor areas. At 49 weeks of age, 24 hens/breed were randomly selected for the evaluation of welfare (ethological and physiological) parameters, at different time points: T0: 1-week pre-SPF; T1: 1-week post-SFP; T2: 3-week post-SFP; T3: 5-week post-SFP. Egg production was recorded from 38 to 56 weeks of age. Grouping produced a social stress in both BS and BP-laying hens, which was expressed in terms of productive traits (reduction of oviposition), behavioral modification (worsening of plumage condition due to feather peaking and extension of the duration of the tonic immobility test) and physiological modification (increased heterophil/lymphocyte ratio and corticosterone metabolites in droppings). Both breeds reacted in a similar way; in fact, no differences were attributed to the breed. At the end of the observation period, the egg rate fully recovered, while the behavioral and physiological parameters partially recovered but failed to recover to those recorded prior to the stressful event. The phytoestrogen genistein (GEN) may interfere with permanent morphological changes in the brain circuits sensitive to estrogen. Due to the frequent use of soy milk in the neonatal diet, we aimed to study the effects of early GEN exposure on some physiological and reproductive parameters. Mice of both sexes from PND1 to PND8 were treated with GEN (50 mg/kg body weight, comparable to the exposure level in babies fed with soy-based formulas). When adult, we observed, in GEN-treated females, an advanced pubertal onset and an altered estrous cycle, and, in males, a decrease of testicle weight and fecal testosterone concentration. Furthermore, we observed an increase in body weight and altered plasma concentrations of metabolic hormones (leptin, ghrelin, triiodothyronine) limited to adult females. Exposure to GEN significantly altered kisspeptin and POMC immunoreactivity only in females and orexin immunoreactivity in both sexes. In conclusion, early postnatal exposure of mice to GEN determines long-term sex-specific organizational effects. It impairs the reproductive system and has an obesogenic effect only in females, which is probably due to the alterations of neuroendocrine circuits controlling metabolism; thus GEN, should be classified as a metabolism disrupting chemical. Simple Summary: Animal–visitor interactions are the experiences offered by zoos, sanctuaries, and other tourism facilities in which people can be very close, and even touch, wildlife. This proximity could damage animal welfare and be a risk for the health of both animals and visitors. Proximity, however, has a positive emotional impact on visitors, representing an excellent opportunity to communicate conservation and educational messages. We present a protocol to evaluate interaction activities, and describe its application in a "giraffe feeding" interaction evaluation. Behavioral observations and a risk assessment evaluated the impact on animals. A risk assessment related to both visitors and staff and a questionnaire investigated the risks for people and the emotional, educational, and conservation outcomes. An ethical analysis, using an ethical matrix and a checklist, integrated the results, and identified the possible ethical concerns of the interaction. Giraffes' behavioral freedom and welfare were safeguarded, and a positive emotional and conservation oriented impact was found, the only improvement that could be suggested, in case of restructuring of the facility, being the absence of hand washing facilities after the interaction. The protocol showed its potentiality to protect animal welfare and human health and to promote an ethical use of the interactions. Due to the popularity of wild animal–visitor interactions (AVIs), there is a need for an ethical assessment of their impact on animal welfare, education, and conservation. The protocol presented in this study is designed to evaluate such interactions on an integrated level, using a transparent analysis of all the aspects involved, including all the stakeholders and the potential conflicts of values. The protocol consists of a six-step process encompassing dedicated data acquisition and a specific ethical assessment. When the protocol was applied to assess a "giraffe feeding" interaction, steps devoted to data acquisition found that animal welfare risks were low, and that visitors described giraffes with emotionally linked descriptors more often after the interaction. The net promoter score, which refers to how likely visitors would recommend to a friend to join the animal–visitor interaction, was 74%. The subsequent ethical assessment, which consisted of a comparison of the results of the previous steps with an ethical matrix highlighting the ideal situation for all stakeholders' interests, allowed the overall identification of the ethical concerns entailed by the interaction. Reactive oxygen species (ROS) are produced as a natural byproduct of the normal metabolism of oxygen and play significant roles in cell signaling and homeostasis. Although ROS have been involved in pathological processes as diverse as cancer, cardiovascular disease, and aging, they may to exert an effect even in a physiological context. In the central nervous system, stem cells and hematopoietic stem cells are early progenitors that contain lower levels of ROS than their more mature progeny. These different concentrations have been reported to be crucial for maintaining stem cell function. Mammary gland remodeling has been proposed to be organized through the activation and regulation of cells with stemness, either considered real stem cells or primitive precursors. Given the state of oxidative stress in the mammary gland tissue induced by high milk production, in particular in highly productive dairy cows; several studies have focused on the relationship between adult mammary stem cells and the oxidative state of the gland. The oxidative state of the mammary gland appears to be involved in the initial development and metastasis of breast cancer through interference with mammary cancerous stem cells. Simple Summary: A pilot test of a six-step protocol to evaluate animal-visitor interactions was developed on a "giraffe feeding" program in a zoo. The steps devoted to animal welfare's assessment are presented in this paper. We observed the giraffes' behaviour, evaluated the suitability of the area in which the interactions took place, and assessed the intensity of various hazards for animal welfare. The long-term goal of this research project is to test a protocol for the overall evaluation of the quality of animal-visitor interactions in zoos and other facilities. Giraffes could choose whether to participate or not to the feeding interactions with visitors, and did not show any behaviour that suggested they did find the interaction to be a negative experience, so their welfare level was assessed as not to be adversely affected by the interaction with the visitors. In recent years, awareness of the controversial aspects connected with wild animal-visitor interactions (AVIs) in zoos and other facilities has increased due to cultural changes. Therefore, the need to apply transparent procedures to evaluate AVIs programs in zoos and similar facilities has also increased. This study presents results of animal welfare's assessment of a pilot test of a protocol based on six steps that aim to explore and assess the overall value of AVIs considering the impact both on animals and visitors. In the present paper, we discuss the multifaceted approach to animal welfare assessment during animal-visitor interactions, combining quantitative behavioural observations/analysis and a welfare risk-assessment procedure, which forms the basis of the six-step protocol. Pilot testing of said approach to animal welfare assessment involved giraffes (Giraffa camelopardalis) in an Italian zoo. No change in behaviour, suggestive of an increased welfare risk to the animals, was found. The risk analysis reported overall low risks for welfare, whereas enclosure analysis highlighted that the enclosure was suitable for allowing interactions without jeopardising animal welfare, mainly because it allowed animals to choose whether to interact or withdraw from interactions without decreasing the space available to them.
